Salicylate Treatment Improves Age-Associated Vascular Endothelial Dysfunction: Potential Role of Nuclear Factor kB and Forkhead Box O Phosphorylation

We hypothesized that I kappa B kinase (IKK)-mediated nuclear factor kappa B and forkhead BoxO3a phosphorylation will be associated with age-related endothelial dysfunction. Endothelium-dependent dilation and aortic protein expression/phosphorylation were determined in young and old male B6D2F1 mice and old mice treated with the IKK inhibitor, salicylate. IKK activation was greater in old mice and was associated with greater nitrotyrosine and cytokines. Endothelium-dependent dilation, nitric oxide (NO), and endothelial NO synthase phosphorylation were lower in old mice. Endothelium-dependent dilation and NO bioavailability were restored by a superoxide dismutase mimetic. The research project started in April, 2008 before adoption of the cabinet report of the Federal Government to the German strategy for Adaptation to Climate Change. The German Strategy for Adaptation emphasizes the meaning of information about requirements for adaptation and the active involvement of all social groups in the other process of the strategy. For this the research project has made concrete proposals for the information, communication and participation of social groups. These proposals are based in the essentials on an interest analysis about interviews with stakeholders and the economy to their demands and expectations to the German strategy for Adaptation. Besides we have researched international examples for the adaptation to the climate change for the public relations. For the interactive development of the Internet platform www.anpassung.net concrete proposals were developed. (orig.)

Morusin induces apoptosis and suppresses NF-#kappa#B activity in human colorectal cancer HT-29 cells

Morusin is a pure compound isolated from root bark of Morusaustralis (Moraceae). In this study, we demonstrated that morusin significantly inhibited the growth and clonogenicity of human colorectal cancer HT-29 cells. Apoptosis induced by morusin was characterized by accumulation of cells at the sub-G_1 phase, fragmentation of DNA, and condensation of chromatin. Morusin also inhibited the phosphorylation of IKK-#alpha#, IKK-#beta# and I#kappa#B-#alpha#, increased expression of I#kappa#B-#alpha#, and suppressed nuclear translocation of NF-#kappa#B and its DNA binding activity. Dephosphorylation of NF-#kappa#B upstream regulators PI3K, Akt and PDK1 was also displayed. In addition, activation of caspase-8, change of mitochondrial membrane potential, release of cytochrome c and Smac/DIABLO, and activation of caspase-9 and -3 were observed at the early time point. An overview is given of the interest of the Dutch government, several organizations and consumers in the quality of the indoor environment. Main aspects, sub-aspects and indicators for the indoor environment of dwellings are listed: site selection, thermal comfort, air quality, light, view, privacy, noise, safety, and radiation. A novel small-molecule inhibitor of NF-#kappa#B signaling

The inducible transcription factor NF-#kappa#B regulates divergent signaling pathways including inflammatory response and cancer development. Selective inhibitors for NF-#kappa#B signaling are potentially useful for treatment of inflammation and cancer. NF-#kappa#B is canonically activated by preferential disposal of its inhibitory protein; I#kappa#B, which suppresses the nuclear translocation of NF-#kappa#B. I#kappa#B#alpha# (a major member of I#kappa#B family proteins) is phosphorylated with an I#kappa#B kinase (IKK) and subsequently polyubiquitylated by SCF"#beta#"T"r"C"P"1 ubiquitin-ligase in the presence of E1 and E2 prior to proteasomal degradation. Here, we describe a novel inhibitor termed GS143, which suppressed I#kappa#B#alpha# ubiquitylation, but not I#kappa#B#alpha# phosphorylation, MDM2-directed p53 ubiquitylation, and proteasome activity in vitro. This report focuses on the question whether a further increase in environmental taxes can contribute to achieving a sustainable economy in which depletion and climate risks should be reduced to an acceptable level for 2050. A three-dimensional steady numerical computation was made for a component model of fin and tube type of compact heat exchangers situated in a uniform flow. In this study, in order to find a clue to design high-efficient heat exchangers, numerical computation was made in the case where two fences are attached on the plate fin surface and their geometrical effects such as their locations, their heights and their attack angles on flow and heat transfer characteristics were investigated especially in the low Reynolds number range. The effect of a hole, open in the plate fin behind the fence, on heat transfer and flow structures was also examined. This hole simulates the hole of the plate fin to be produced in practical application, when a fence is produced by punching from the original plain fin plate. Norway is a member of Annex 29, 'Ground-Source Heat Pump Systems Overcoming Technical and Market Barriers' (2004-2006), organized under the umbrella of the International Energy Agency (IEA) and the IEA Heat Pump Programme (HPP). The 7 participating countries are Austria (Operating Agent), Canada, Japan, Norway, Spain, Sweden and the USA. The Norwegian participation is financed by ENOVA SF, and SINTEF Energy Research is responsible for planning and carrying out the Norwegian activities. This report provides a status for ground-source heat pump (GSHP) systems in Norway with regard to state-of-the-art technology, installation examples, geological data, costs and market opportunities. A Norwegian Internet home page for ground-source heat pump systems (www.energy.sintef.no/prosjekt/Annex29) is also presented.
