Australia?s nationally consistent framework for gene technology regulation is underpinned by the Gene Technology Act 2000, administered by an independent decision-maker, the Gene Technology Regulator. The object of the Act is ?to protect the health and safety of people, and to protect the environment, by identifying risks posed by or as a result of gene technology, and by managing those risks through regulating certain dealings with genetically modified organisms?. Marketing and trade impacts are outside the scope of assessments required by the Act. Since 2001, seven licences have been issued for the commercial cultivation of genetically modified (GM) cotton with insect resistance and/or herbicide tolerance. Licences have also been issued for 32 GM cotton field trials with a broader range ...
Kong-Ming Wu (Corresponding author) The application of recombinant DNA technology has resulted in many insect-resistant varieties by genetic engineering (GE). Crops expressing Cry toxins derived from Bacillusthuringiensis (Bt) have been planted worldwide, and are an effective tool for pest control. However, one ecological concern regarding the potential effects of insect-resistant GE plants on non-target organisms (NTOs) has been continually debated. In the present study, we briefly summarize the data regarding the development and commercial use of transgenic Bt varieties, elaborate on the procedure and methods for assessing the non-target effects of insect-resistant GE plants, and synthetically analyze the related research results, mostly those published between 2005 and 2010. A mass of laboratory and field studies have shown that the currently available Bt crops have no direct detrimental effects on NTOs due to their narrow spectrum of ...
Full text: Pink bollworm (PBW), Pectinophora gossypiella (Saunders), an introduced pest from Mexico, first occurred in United States cotton production in Texas in 1917. Unacceptable economic losses have occurred. The development of PBW sterile moth release technology, gossyplure sex pheromone behavioral control, cotton plant cultural control to reduce overwintered PBW populations, and the transfer of the insect toxin protein gene into cotton from Bacillusthuringiensis kurstaki (Berliner) have provided an effective integrated pest management (IPM) system with PBW eradication potential. Sterile insect release was considered a potential option for PBW population suppression in the early 1960s. Research on the isolated island of St. Croix, (US Virgin Islands) demonstrated its validity. Reductions of larval infestations in bolls following sterile moth releases began when ratios of released PBW sterile male to native male moths ...
The report presents details of the distributions of air pollution in the Berlin area and the developments of the past 20 years. Highly polluted areas are indicated in which air pollution reduction measures are particularly urgent. The report therefore intends not only to inform but also to point out the deficiencies of air pollution abatement measures, so that political consequences can be derived and measures can be taken. In all, data were recorded by 39 measuring stations in West Berlin, 16 in East Berlin, and 7 in Potsdam and Frankfurt/Oder. In most stations, sulphur dioxide and airborne dust were measured; 14 stations in West Berlin and one in East Berlin measured carbon monoxide and nitric oxides; 8 stations in West Berlin recorded ozone concentrations. This study was conducted to investigate the antibacterial effect of BSAP-254 on Bacillus cereus with the induced stress proteins. The BSAP-254 is an antimicrobial peptide isolated from soybean-fermenting bacteria, Bacillus subtilis SC-8. It had a narrow spectrum of activity against B. cereus group. The growth inhibitory effect of BSAP-254 (50??g/mL) reduced the population of B. cereus from >108 to 104 colony-forming units per milliliter within 30?min. In B. cereus exposed to BSAP-254, 14 intracellular proteins were differentially expressed as determined by 2-DE coupled with MS. Of the differentially expressed proteins identified, the stress protein GroEL, which is heat shock protein, was induced in B. cereus exposed to antibacterial peptide.

In this study, bacterial strains were investigated in order to determine their heavy metal tolerance. The bacterial strains were identified as Bacillus cereus and Bacillus pumilus. In the batch system, the effects of operating variables such as solution pH, initial metal concentration, contact time, and adsorbent dosage were investigated. Both isolates were highly resistance to copper and lead in comparison with the control strain examined. The adsorption capacities of B. cereus and B. pumilus were found to be 22.1mg/g and 28.06mg/g, respectively. The biosorption follows pseudo-second order kinetics and the isotherm fits well to the Langmuir isotherm model. In column experiments, the biosorption was fitted well by the Thomas model. The breakthrough and exhaustion capacity of each biosorben...

A broad range of persistent organic pollutants (POPs) and selected heavy metals has been analysed in sediment cores of North-East Germany. The pollutants analysed include polychlorinated dioxins and furans (PCDD/Fs), polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs), several pesticides (DDT, HCH, CBz) and its metabolites as well as selected heavy metals. The sediment cores were sampled at five locations, reflecting a range of anthropogenic influences and background contamination: Arkona Basin (AK) representing a remote marine site, Lake Bugsin (BS) a background location only with atmospheric deposition, Lake Quenz (QS) close to the industrial city of Brandenburg, Teltowkanal (TK) in the suburban-industrial zone of Berlin and Lake White (WS) in the centre of Berlin. The age of the lower part of the AK, BS and TK cores were dated at 100-150 years. Results of selected pollutants (PCDD/Fs) have been presented earlier, ...
Calculations in "7Li have been carried out for the differential coherent elastic scattering cross-section of thermal neutrons at a fixed scattering angle #theta#_0(148"o) and varying the energy of incident neutrons (or corresponding d-spacings). The results are reported, as variations of the ratio #SIGMA#_0"c"o"h (#theta#_0, d)/#SIGMA#_0"c"o"h (#theta#_0) with d-spacing, at atmospheric pressure and at two temperatures, 80 and 20 K. The calculations at 80 K correspond to BCC lithium whereas at 20 K all the possible crystallographic structures of polytype lithium (BCC, 9R, HCP and FCC) were considered. On comparing our results with the time-of-flight (TOF) measurements of Berliner et al. (Physical Review, B40, 12086, 1989) we find that up to 80 K lithium-7 does not undergo any martenistic transformation whereas the structural state at 20 K can be described by the co-existence of the original BCC phase with 9R, HCP and FCC closed packed structures. Bacillus licheniformis ?-glutamyltranspeptidase (BlGGT) undergoes an autocatalytic process to generate 44.9 and 21.7?kDa subunits; however, a mutant protein (T399A) loses completely the processing ability and mainly exists as a precursor. For a comprehensive understanding of their structural features, the biophysical properties of these two proteins were investigated by circular dichroism and fluorescence spectroscopy. Tryptophan fluorescence and circular dichroism spectra were nearly identical for BlGGT and T399A, but unfolding analyses revealed that these two proteins had a different sensitivity towards temperature- and guanidine hydrochloride (GdnHCl)-induced denaturation. BlGGT and the unprocessed T399A displayed T m values of 61.4?C and 68.1?C, respectively, and thermal unfolding of b...
The production of {alpha}-amylase by Bacillus licheniformis M27 in submerged fermentation was completely inhibited due to catabolic repression in medium containing 1% glucose. In contrast, the enzyme production in a solid state fermentation system was 19,550 units/ml extract even when the medium contained 15% glucose. The peak in enzyme titre was, however, shifted from 48 to 72 h. The ability of the solid state fermentation system to significantly overcome catabolic repression was not known earlier and is probably conferred by various physico-chemical factors and culture conditions specific to the system. The French agency of environment and energy mastery (Ademe) and Deutsche Energie Agentur (DENA) organised in Berlin on November 22 and 23, 2001, the first seminar and forum of ideas and exchanges on the implementation of energy efficiency and renewable energy policies in France and in Germany. On this occasion, Ademe asked Energie-Cites - an association of European cities with a long practical experience in 'good practice' exchanges - to prepare a publication describing the experience of about ten French cities or local energy agencies in energy management issues. The case studies presented in this document concern the following cities: Besancon, Dole, Dunkerque, Lille, Montpellier, and the local energy agencies of Clermont-Ferrand (Aduhme), Grenoble, Montreuil-Vincennes, Mulhouse and Rennes. Data from a detailed evaluation of environment conditions in three U.S. Army field stations are presented. Three field stations were evaluated: Kunia, Augsburg, and Berlin. Results from a questionnaire administered to over 600 people at all sites in three job types (operator, analyst, and administrative/other) are given which indicate major concerns with conditions such as temperature, lighting, space, furniture, equipment functioning, and general environmental quality. Physical data obtained from measurements of over 270 work stations indicated the presence of cold temperatures, low light levels, reduced VDT screen contrast, glare, and distracting noises. The report provides the basic data that support the concerns expressed by Headquarters; namely, that field-station personnel perform their jobs under conditions likely to impair their effectiveness. Suggestions for improving conditions are given in a companion report by Rubin and Collins.

The aim of the study was to establish by means of a measuring programme how much energy for space heating can be saved by the installed hybrid system. Further, experience with the systems was to be gathered under practical use conditions. As user behaviour can influence space heating energy consumption to an extent having several times the effect of the contribution of the hybrid system, a fact that is also reported in literature, it is not sufficient to measure only space heating energy consumption. So, a unit in one of the terraced houses was scrupulously measured. In the three other terraced houses, only space heating energy consumption was measured.- This final report contains not only a description of the concepts of the buildings and hybrid system but also the energy consumptions measured, and detailed analyses of the budgets. The paper presents fundamental studies on natural biosorption to algae of heavy metals. The investigations were carried out by TU Berlin University in a special research project (Sfb 193, 'Biologische Behandlung industrieller und gewerblicher Abwaesser'). The chlorophyceae C. vulgaris was used as reference alga, and 38 further algae were analzyed with regard to their sorption characteristics for lead, cadmium, nickel and zinc. The most effective algae were the chlorophyceae C. salina, the cyanophyceae S. hofmani and L. taylorii. The biosorption characteristics of C. salina and L. taylorii were investigated in detail. L. taylorii was found to be exploitable successfully in a technical process, so the investigations were extended to include copper. Knowledge about the chemical processes of biosorption is important for any attempt to modify the biosorption characteristics. Experiments on the incorporation of additional functional groups in the cell ...
This study was focused on the possibility to inactivate main food pathogens, their spores and biofilms on the surface of packaging material polyolefine by Na-chlorophyllin (Na-Chl)-based photosensitization and to compare efficiency of this treatment with conventional antimicrobials. Data indicate that Bacillus cereus and Listeria monocytogenes were effectively inactivated (7 log) by Na-Chl (7.5x10-7M)-based photosensitization in vitro and on the surface of packaging. Meanwhile to achieve adequate inactivation of thermo-resistant strains, spores or biofilms the higher Na-Chl concentration and longer illumination times had to be used. Comparison of different surface decontamination treatments reveal that photosensitization is much more effective against B. cereus and L. monocytogenes attache...
Abstract Microbiological quality and biophenol content evolution was studied in minimally processed Thassos cv table olives by hot air dehydration under mild conditions (40C, 24-h, aw-=-0.893) and storage under characteristic packaging conditions (vacuum, 100% N2 and air) at 4 and 20C over a period of 180 days. No salt was used in the production line or packaging. The undesirable microorganisms (Enterobacteriaceae, Staphylococcus aureus, Bacillus, Clostridium) were undetectable until the end of the storage period. Also, modified atmospheres prevented fungal growth at both temperatures apart from the samples stored in air, in which Penicillium and Aspergillus spp. were identified. At 20C, a coexistence of mesophilic bacteria and yeasts occurred. At 4C, yeasts were the predominant microflora...

We describe a simple one-pot thermal decomposition method for the production of a stable colloidal suspension of narrowly dispersed superparamagnetic Fe3O4-Ag core-shell nanostructures. These biocompatible nanostructures are highly toxic to microorganisms. Antimicrobial activity studies were carried out on both Gram negative (Escherichia coli and Proteus vulgaris) and Gram positive (Bacillus megaterium and Staphylococcus aureus) bacterial strains. Efforts have been made to understand the underlying molecular mechanism of such antibacterial actions. The effect of the core-shell nanostructures on Gram negative strains was found to be better than that observed for silver nanoparticles. The minimum inhibitory concentration (MIC) values of these nanostructures were found to be considerably lowe...
A number of aerobic species capable of decolorizing some of the dyes in a textile mill effluent were isolated. One of the isolates was able to decolorize Terasil black dye under aerobic conditions in the presence of an exogenous carbon source after 5 days. Glucose or starch (%1 ea) are essential for decolorization but the process proceeds faster in the presence of 0.5% yeast extract. Results of the BOD5 show that the untreated effluent samples have a low BOD value, whereas treated samples show an initial increase in BOD up to 15 days followed by a decrease after 20 days. FT-IR and GC-MS data also reveal that the initial components in the untreated effluent disappear after 20 days of treatment, confirming biodegradation of the dye. Phytotoxicity tests on the untreated effluent samples using...
The role of the HCR system in the repair of prelethal lesions induced by UV light, #gamma# radiation and alkylating agents was studied in the Bacillus subtilis SPP1 phage, its heat sensitive mutants (N3, N73 nad ts_1) and corresponding infectious DNA. The survival of phages and their transfecting DNA after treatment with UV light is substantially higher in hcr"+ cells than in hcr cells, the differences being more striking in intact phages than in their transfecting DNA's. Repair inhibitors reduce survival in hcr"+ cells: caffeine lowers the survival of UV-irradiated phage SPP1 in exponentially growing hcr"+ cells but has no effect on its survival in competent hcr"+ cells; acriflavin and ethidium bromide decrease the survival of the UV-irradiated SPP1 phage in both exponentially growing and competent hcr"+ cells to the level of survival observed in hcr cells; moreover, ethidium bromide lowers the number of infective centres in hcr"+ cells of the UV-irradiated DNA of ...
We studied the interaction of Pu(VI) with Pseudomonas stutzeri ATCC 17588 and Bacillus sphaericus ATCC 14577, representatives of the main aerobic groups of soil bacteria present in the upper soil layers. The accumulation studies have shown that these soil bacteria accumulate high amounts of Pu(VI). The sorption efficiency toward Pu(VI) decreased with increasing biomass concentration due to increased agglomeration of the bacteria resulting in a decreased total surface area and number of available complexing groups. Spores of Bacillus sphaericus showed a higher biosorption than the vegetative cells at low biomass concentration which decreased significantly with increasing biomass concentration. At higher biomass concentrations (> 0.7 g/L), the vegetative cells of both strains and the spores of B. sphaericus showed comparable sorption efficiencies. Correlates between immunological parameters and protection against Bacillus anthracis infection in animals vaccinated with protective antigen (PA)-based vaccines could provide surrogate markers to evaluate the putative protective efficiency of immunization in humans. In previous studies we demonstrated that neutralizing antibody levels serve as correlates for protection in guinea pigs (S. Reuveny et al., Infect. Immun. 69:2888-2893, 2001; H. Marcus et al., Infect. Immun. 72:3471-3477, 2004). In this study we evaluated similar correlates for protection by active and passive immunization of New Zealand White rabbits. Full immunization and partial immunization were achieved by single and multiple injections of standard and diluted doses of a PA-based vaccine. Passive immunization was carried out by injection of immune sera from rabbits vaccinated with PA-based vaccine prior to challenge with B. anthracis spores. New tetradentate N(2)O(2) donor Schiff bases and their mononuclear Co(II), Ni(II), Cu(II), and Pd(II) complexes were synthesized and characterized extensively by IR, (1)H-, (13)C-NMR, mass, ESR, conductivity measurements, elemental and thermal analysis. Specifically the magnetic and electronic spectral measurements demonstrate the octahedral structures of cobalt(II), nickel(II) complexes and square planar geometries of copper(II), palladium(II) complexes. All the ligands and complexes were screened for their in vitro antibacterial activity against two gram-positive bacteria (Bacillus subtilis, Staphylococcus aureus) and two gram-negative bacteria (Escherichia coli, Klebsiella pneumonia). In this study, Pd(II) complexes exhibited potent antibacterial activity against B. subtilis, S. aureus whereas other metal complexes also exerted good activity towards all tested strains even than standard drugs streptomycin and ampicillin. The adequacy of the nasopharyngeal airway has been found to be related to craniofacial development. Obstruction of the airway by adenoid tissue, nasal septal deviation or abnormal morphology of the area is associated with characteristic changes in craniofacial morphology such as long anterior face height, facial retrognathism, and a steep inclination of mandibular plane often with a high vaulted palate and crossbite. Some studies have shown the changes to be reversible after adenoidectomy which improves nasal airway patency and a control mechanism for facial growth has been proposed to account for the relationships between airway adequacy, craniofacial morphology and craniocervical postural relationships. It is therefore important to be able to measure nasal respiratory resistance so that the effect of operative procedures in the area such as rapid maxillary expansion (RME) can be determined. Nasal respiratory resistance (NRR) is a measure of airway adequacy. It can be recorded by ...
Ab initio calculations of the electronic structure were performed for the silver palladium oxide, Ag_2PdO_2, by the full potential mixed linearized augmented plane wave and augmented plane wave plus local orbitals method (mixed LAPW/APW+lo) within the density functional theory and using the generalized gradient approximation (GGA96, Phys. Rev. Lett. 77 (1996) 3865) for the exchange-correlation potential. The lattice parameters were calculated from the ground-state total energy versus volume curve, and are in good agreement with the available experimental results. Our calculations of band structure predicted that the compound Ag_2PdO_2 has indirect band gap, and the Pd 4d states strongly hybridize with the O 2p states all over the valence bands and conduction bands. The valence bands are composed mainly of Ag, Pd 4d states and O 2p states, while the conduction bands consist mainly of the Pd 4d states and O 2p states. In addition, focusing on the calculation precision of the band gap, ...
For evaporation crystallization of extremely corrosive FGD scrubber solutions which - depending on the process - can lead all the way to a product suitable for depositing, an economic equipment concept based on metallic construction materials was to be investigated. In a parameter study, electrochemical test methods were used to determine the influence of the pH value, the Fe{sup 3+}, F and So{sub 2} contents in a solution containing 200 g/l CaCl{sub 2}+30 g/l NaCl at 80deg C on the resistance to pitting corrosion of stainless steels and NiCrMo alloys. In addition, electrochemical and wet corrosion tests (e.g. crevice corrosion) were conducted using original FGD scrubber solutions with 43-50% dry substance from the power plants Reuter-West, Oberhavel and Rudow of the BEWAG, Berlin. The results were compared to those of standardized and non-standardized short-term tests for determining the critical pitting corrosion and crevice corrosion temperatures for the various ...
Adenine DNA glycosylase catalyzes the glycolytic removal of adenine from the promutagenic A {center_dot} oxoG base pair in DNA. The general features of DNA recognition by an adenine DNA glycosylase, Bacillus stearothermophilus MutY, have previously been revealed via the X-ray structure of a catalytically inactive mutant protein bound to an A:oxoG-containing DNA duplex. Although the structure revealed the substrate adenine to be, as expected, extruded from the DNA helix and inserted into an extrahelical active site pocket on the enzyme, the substrate adenine engaged in no direct contacts with active site residues. This feature was paradoxical, because other glycosylases have been observed to engage their substrates primarily through direct contacts. The lack of direct contacts in the case of MutY suggested that either MutY uses a distinctive logic for substrate recognition or that the X-ray structure had captured a noncatalytically competent state in lesion ...
pT181 is a naturally-occurring 4437 basepair (bp) plasmid isolated from Staphylococcus aureus which encodes inducible resistance to tetracycline (Tc). The DNA sequence data has identified three open reading frames (ORFs). The largest ORF B, has been found to be responsible for the Tc resistance phenotype of pT181. Since most Tc resistance systems appear to be regulated by an effector protein and a repressor protein, several Bal 31 deletion mutants of pT181 were constructed and analyzed in an effort to identify the elements involved in Tc resistance. Two transcomplementing groups of mutants were identified within the tet gene. The mechanism of Tc resistance was studied by assaying the accumulation of (7-/sup 3/H) Tc by Tc sensitive cells, and uninduced and induced pT181-containing cells. A sharp decrease in accumulation of the drug after an initial increase was observed in Tc induced pT181-containing cells. In vivo labeling of Bacillus subtilis minicells containing ...
