'The long-term goal of this project is to develop antibodies and antibody-based methods for detection and recovery of polynuclear aromatic hydrocarbons (PAHs) and PAH adducts that are potential biomarkers in environmental and biological samples. The inherent cross-reactivity will be exploited by pattern recognition methods. Dr. Karu''s laboratory uses new haptens representing key PAHs to derive recombinant Fab (rFab) and single-chain Fv (scFv) antibodies from hybridoma lines and combinatorial phage display libraries. Computational models of the haptens and combining sites made by Dr. Roberts''s group are used to guide antibody engineering by mutagenesis. Dr. Li''s laboratory develops enzyme immunoassays (EIAs), sensors, and immunoaffinity methods that make use of the novel haptens and antibodies for practical analytical applications in support of DOE''s mission. This report summarizes work completed in one and one-half years of a 3-year project, with close collaboration between the three research groups. Dr. Alexander Karu''s laboratory: the authors proceeded with the two strategies described in the original proposal. Site-directed mutagenesis was used to correct differences in the rFab N-terminal amino acids that were introduced by the degenerate PCR primers used for gene amplification. The binding constants of the rFabs with the corrected sequences will be compared with those of the parent MAbs, and should be very similar. The 4D5 and 10C10 heavy and light chain sequences are being moved to the pCOMB3H phagemid vector to facilitate selection of new engineered mutants.'

There is an increasing movement against use of antibiotic growth promoters in animal feed. Prebiotic supplementation is a potential alternative to enhance the host's natural defense through modulation of gut microbiota. In the present study, the effect of mannan oligosaccharide (MOS) and virginiamycin (VIRG) on cecal microbial ecology and intestinal morphology of broiler chickens raised under suboptimal conditions was evaluated. MOS and VIRG induced different bacterial community structures, as revealed by denaturing gradient gel electrophoresis of 16S rDNA. The antibiotic treatment reduced cecal microbial diversity while the community equitability increased. A higher bacterial diversity was observed in the cecum of MOS-supplemented birds. Quantitative polymerase chain reaction results indicated that MOS changed the cecal microbiota in favor of the Firmicutes population but not the Bacteroidetes population. No difference was observed in total bacterial counts among treatments. MOS promoted the growth of Lactobacillus spp. and Bifidobacterium spp. in the cecum and increased villus height and goblet cell numbers in the ileum and jejunum. These results provide a deeper insight into the microbial ecological changes after supplementation of MOS prebiotic in poultry diets.

Agrobacterium-mediated gene transfer is the method of choice for many plant biotechnology laboratories; however, large-scale use of this organism in conifer transformation has been limited by difficult propagation of explant material, selection efficiencies and low transformation frequency. We have analyzed co-cultivation conditions and different disarmed strains of Agrobacterium to improve transformation. Additional copies of virulence genes were added to three common disarmed strains. These extra virulence genes included either a constitutively active virG or extra copies of virG and virB, both from pTiBo542. In experiments with Norway spruce, we increased transformation efficiencies 1000-fold from initial experiments where little or no transient expression was detected. Over 100 transformed lines expressing the marker gene beta-glucuronidase (GUS) were generated from rapidly dividing embryogenic suspension-cultured cells co-cultivated with Agrobacterium. GUS activity was used to monitor transient expression and to further test lines selected on kanamycin-containing medium. In loblolly pine, transient expression increased 10-fold utilizing modified Agrobacterium strains. Agrobacterium-mediated gene transfer is a useful technique for large-scale generation of transgenic Norway spruce and may prove useful for other conifer species.

Isolation and Characterization of Agrobacterium Strains from Soil: A Laboratory Capstone Experience

Full Text Available In this investigation, the students’ goal was to isolate and characterize Agrobacterium strains from soil. Following selection and enrichment on 1A-t medium, putative Agrobacterium isolates were characterized by Gram stain reaction and biochemical tests. Isolates were further evaluated using polymerase chain reaction (PCR with different primer sets designed to amplify specific regions of bacterial deoxyribonucleic acid (DNA. Primer sets included AGRH to identify isolates that were members of the Rhizobiaceae, BIOVAR1 primers to identify members of Agrobacterium biovar group I, and a third set, VIRG, to determine presence of virG (only present in pathogenic Agrobacterium strains. During the investigation, students applied previously learned techniques including serial dilution, use of selective/differential media, staining protocols, biochemical analysis, molecular analysis via PCR, and electrophoresis. Students also gained practical experience using photo documentation to record data for an eventual mock journal publication of the capstone laboratory experience. Pre- and post-evaluation of class content knowledge related to the techniques, protocols, and learning objectives of these laboratories revealed significant learning gains in the content areas of Agrobacterium–plant interactions (p ≤ 0.001 and molecular biology (p ≤ 0.01. The capstone journal assignment served as the assessment tool to evaluate mastery and application of laboratory technique, the ability to accurately collect and evaluate data, and critical thinking skills associated with experimental troubleshooting and extrapolation. Analysis of journal reports following the capstone experience showed significant improvement in assignment scores (p ≤ 0.0001 and attainment of capstone experience learning outcomes.

The effects of 2 levels of mannanoligosaccharide (MOS) in feed were compared with antibiotic growth promoters on growth performance, intestinal morphology, cecal and litter microbial populations, and carcass parameters in broilers raised in a sanitary environment. Dietary treatments included: 1) antibiotic growth promoter-free diet (control), 2) VIRG (diet 1 + 16.5 mg/kg of virginiamycin), 3) BACT (diet 1 + 55 mg/kg of bacitracin), 4) LMOS (diet 1 + 0.2% MOS), and 5) HMOS (diet 1 + 0.5% MOS). Birds were randomly assigned to 3 replicate pens/treatment (n = 55/pen). Body weight and feed intake were recorded weekly throughout 38 d. At d 14, 24, and 34, a 1-cm segment of duodenum, jejunum, and ileum was used in morphological analysis (n = 9 birds/d per treatment). At the same bird ages, cecal contents were assayed for lactobacilli, bifidobacteria, Salmonella, Campylobacter, and Escherichia coli, whereas litter was analyzed for Salmonella, Campylobacter, and E. coli. Carcass yields (breast fillet and tenders, thigh, drumstick, and wing) were determined at d 38. Body weight, feed conversion, and carcass yields did not differ among treatments. In contrast to birds fed VIRG or BACT, LMOS and HMOS consistently increased (P litter from all treatments were free of Salmonella. At d 14 and 24, cecal E. coli and Campylobacter counts were not different among treatments. In comparison to birds fed control, at d 34, BACT, LMOS, and HMOS significantly reduced (P Litter bacterial counts were not altered by dietary treatments. In conclusion, under conditions of this study, MOS conferred intestinal health benefits to chickens by improving its morphological development and microbial ecology. But, there were no additional benefits of the higher MOS dosage.

Full Text Available The paper presents In Vino Veritas – IVV, a hybrid, pervasive and ubiquitous game, developed in Bento Gonçalves, located in Serra Gaúcha, Southern Brazil. The goal is to discuss the cultural, educational and wine tourism potential of the IVV, in a context of innovation, including learning in the areas Enology, Gastronomy and History of Italian immigration. The conception and development methodology were inspired by the concept of Living Lab (William J. Mitchell and the cartographic method of intervention-research (Virgínia Kastrup As instruments were used participant observation, photographic records, audio, text, digital video and interviews. The data produced were analyzed and interpreted from the Actor-Network Theory (Bruno Latour. The results point to the innovation initiatives such as the IVV might contribute in the field of culture, education and wine tourism, articulated through a narrative able to engage tourists in experiences of knowledge, mixing geographical and digital spaces, valuing local culture.

Full Text Available As Geórgicas (37-30 a.C. constituem um poema sobre a vida rural e as diversas atividades agrícolas: o cultivo dos cereais, a arboricultura, em especial o cultivo da vinha, a criação dos rebanhos e a apicultura. Cada um dos quatro livros que compõem a obra descreve uma atividade específica, por meio do discurso eminentemente didático. Ao longo da obra, porém, notam-se diversas passagens digressivas, em que o poeta narra sobre outros temas, em geral, de cunho religioso, mitológico e filosófico. Uma destas passagens, conhecida como Laudes Italiae, versos 136 a 176 do II canto, constitui um verdadeiro hino de louvor às terras italianas. Intentamos verificar a relevância de alguns dos temas tratados nos referidos versos das Geórgicas, considerando, sobretudo, o contexto de produção da obra, e buscando compreender o seu significado dentro deste poema didático.

Full Text Available Histidine kinases serve as critical environmental sensing modules, and despite their designation as simple two-component modules, their functional roles are remarkably diverse. In Agrobacterium tumefaciens pathogenesis, VirA serves with VirG as the initiating sensor/transcriptional activator for inter-kingdom gene transfer and transformation of higher plants. Through responses to three separate signal inputs, low pH, sugars, and phenols, A. tumefaciens commits to pathogenesis in virtually all flowering plants. However, how these three signals are integrated to regulate the response and why these signals might be diagnostic for susceptible cells across such a broad host-range remains poorly understood. Using a homology model of the VirA linker region, we provide evidence for coordinated long-range transmission of inputs perceived both outside and inside the cell through the creation of targeted VirA truncations. Further, our evidence is consistent with signal inputs weakening associations between VirA domains to position the active site histidine for phosphate transfer. This mechanism requires long-range regulation of inter-domain stability and the transmission of input signals through a common integrating domain for VirA signal transduction.

Full Text Available Este trabalho discute os desdobramentos de uma oficina de leitura com internos de uma prisão no Rio de Janeiro. A oficina se insere no quadro de trabalhos que usam a arte como instrumento de produção de subjetividade e transformação social, como política cognitiva e existencial. Para a compreensão da instituição-prisão, são usadas as ideias de Erving Goffman, Michel Foucault e Loïc Wacquant. Sobre os estudos de produção de subjetividade em oficinas de leitura, o texto toma como referencial Gilles Deleuze, Felix Guattari e Virgínia Kastrup. Foram realizados encontros semanais com um grupo heterogêneo quanto à escolaridade, para a prática da leitura em conjunto. São apresentados relatos dos encontros, para demonstrar a dinâmica do grupo e a multiplicidade dos efeitos produzidos. Full Text Available The red rice in Brazil is cultivated mainly in the Northwester region and practically unknown for major of the Brazilian population. This culture have a great importance for small farmers, but needs more studies. The objective of this research was evaluate agronomics traits, the seed physiological quality and the crude protein content of selected cultivars of red rice, Vermelho Pequeno and Vermelho Virgínia, from cultivation and selection in the UFRRJ, contrasting with the commercial cultivars, Caiapó and BRS Primavera, of white rice. The experiment was installed in November of 2004, in Seropédica, RJ, in the experimental design of randomized blocks with four repetitions. It was evaluated: plant height; harvest index; number of viable tillers/ m²; number of spikiest/panicle; percentage of fertile spikelets/panicle; weight of a thousand spikelets; seed water content; weight of 1000 spikelets; productivity; germination; first counting of the germination test and crude protein content. The Vermelho Pequeno cultivar presented higher productivity of grain, higher tillering capacity, higher spikelets production, medium size, index of harvest, crude protein content and quality seed to similar to that of commercial cultivars.

Full Text Available Giorgio Agamben, ao elaborar o livro Signatura Rerum (2008,  afirma ser possível investigar o passado, para chegar ao paradigma, definido como um conhecimento analógico. Agamben disserta também sobre a assinatura, a linguagem do mundo, da qual se percebe o espírito interior das coisas em sua exterioridade, além de definir a arqueologia filosófica, momento em que os saberes e os discursos são constituídos. Na esteira de Michel Foucault, o filósofo italiano argumenta que a “arqueologia é a ciência das ruínas, prática, que indaga a história não pela origem, mas como a emergência do fenômeno”. (AGAMBEM, 2008, p. 121.   Neste caso, da obra literária emergem fenômenos e indícios, a exemplo da Divina Comédia, de cujo poema surgem personagens da mitologia grega, filósofos e poetas, dentre os quais, o mantuano Virgílio, e outras personagens. Essas singularidades, identificadas na Comédia, são objeto para um possível entrecruzamento com o conceito de assinatura, de Agamben.

Long breeding cycles make cultivar development a lengthy process in deciduous fruit species. Gene transfer is, accordingly, a goal with significant commercial value. In many plant species, especially in woody plants, a prerequisite for genetic engineering is the ability to regenerate plants from transformed cells. Development of single cell regeneration is the first step towards exploration of gene transfer techniques. In this investigation media for plum and apple leaf disk regeneration were developed. Transformation experiments were performed. The vector EHA105 containing the gus-intron gene was found to be effective for gene transfer. Induction of the virG genes with aceto-syringone did not enhance transformation. Cefotaxime that was supplemented in the plum selection medium to suppress the Agrobacterium vector seriously inhibited leaf disk regeneration. However, in applies it was not detrimental. With further apple transformation experiments, factors such as preculturing, age of leaves, sucrose and cefotaxime concentrations did not increase the transformation efficiency of the marker gene. The harpin protein, essential for the pathogenicity of Pseudomonas syringae pv. syringae which incites bacterial canker of stone fruit, ws amplified and cloned into an expression vector. The fusion protein was purified. This will be used in future studies to elucidate the host-pathogen interaction, and to identify antibacterial genes. (author)

Full Text Available Esta pesquisa objetivou analisar a potencialidade terapêutica de encontros realizados pela pesquisadora com crianças de três a seis anos no contexto de uma creche. O referencial teórico utilizado foi a Abordagem Centrada na Pessoa, fundamentado na proposta terapêutica de Virgínia Mae Axline, denominada Ludoterapia Centrada na Criança. As próprias crianças geraram a estrutura dos encontros, estabelecendo com a pesquisadora uma relação que guarda semelhanças com o plantão psicológico. Trata-se de um estudo fenomenológico que adotou a construção de narrativas baseadas nos encontros terapêuticos realizados na instituição. As narrativas possibilitaram uma análise fenomenológica da relação intersubjetiva vivida nos encontros. Os resultados demonstraram a capacidade das crianças em: assumir a iniciativa pela procura espontânea por uma relação de ajuda psicológica; comunicar sentimentos a respeito de si mesmas e de seus problemas; compreender e colaborar para a estruturação da relação de ajuda psicológica.

Incorporation of star measurements for the determination of orbit and attitude parameters of a geosynchronous satellite: An iterative application of linear regression

Currently on NOAA/NESS's VIRGS system at the World Weather Building star images are being ingested on a daily basis. The image coordinates of the star locations are measured and stored. Subsequently, the information is used to determine the attitude, the misalignment angles between the spin axis and the principal axis of the satellite, and the precession rate and direction. This is done for both the 'East' and 'West' operational geosynchronous satellites. This orientation information is then combined with image measurements of earth based landmarks to determine the orbit of each satellite. The method for determining the orbit is simple. For each landmark measurement one determines a nominal position vector for the satellite by extending a ray from the landmark's position towards the satellite and intersecting the ray with a sphere with center coinciding with the Earth's center and with radius equal to the nominal height for a geosynchronous satellite. The apparent motion of the satellite around the Earth's center is then approximated with a Keplerian model. In turn the variations of the satellite's height, as a function of time found by using this model, are used to redetermine the successive satellite positions by again using the Earth based landmark measurements and intersecting rays from these landmarks with the newly determined spheres. This process is performed iteratively until convergence is achieved. Only three iterations are required.

Para isso, depois de situar Plínio do ponto de vista humano, intelectual e histórico em sua própria época, passamos a examinar como o autor fez a abordagem do assunto dos elefantes e de vários seres fabulosos (como dragões e basiliscos na mesma parcela da obra citada. No primeiro caso, assim como o Virgílio do livro IV das Geórgicas (que esse poeta dedicou à apicultura, Plínio desejou mostrar a sociedade animal focalizada como algo, ao mesmo tempo, muito próximo e distante da cultura e dos sentimentos humanos. Quanto à abordagem pliniana de seres que hoje diríamos “monstruosos” por sua desmesura, forte hibridização ou capacidade de lesar bastante os homens ou outras criaturas, nota-se que o autor antigo parece realizá-la com a naturalidade de alguém ciente do caráter altamente inventivo das forças geradoras atuantes no mundo. Nesse movimento de deter-se sobre os aspectos bizarros do mundo, Plínio foi posteriormente seguido de perto por Jorge Luis Borges, autor de O livro dos seres imaginários.

Avaliação das Previsões do Modelo Eta na Região da Serra do Mar (Estado de São Paulo, Brasil

Full Text Available Comparações entre as previsões do modelo Eta-5km e observações nas Plataformas de Coleta de Dados meteorológicas em Cunha, Curucutu, Itanhaém, Paraibuna, Picinguaba, Santa Virgínia e São José dos Campos, localizadas na região da Serra do Mar (SP, são realizadas para o ano de 2008. A temperatura a 2 m, pressão atmosférica ao nível da estação, ventos a 10 m e precipitação são avaliados para as previsões de 24, 48 e 72 horas. Os resultados mostraram que a pressão atmosférica ao nível da estação foi subestimada (superestimada em Paraibuna e Picinguaba (Cunha e Curucutu, devido a diferenças entre a altitude do modelo e a altitude real da estação, porém os ciclos diurnos foram bem previstos, apresentando dois máximos (0 e 12 Z e dois mínimos (6 e 18 Z, como observados. Para a pressão atmosférica a performance do modelo é superior na previsão de 48-h. O ciclo diurno da temperatura do ar foi muito bem previsto em todas as localidades. Os valores de correlação temporal entre as previsões e observações foram bem elevados, oscilando entre 73 e 91%. Para algumas localidades verificou-se que ocorre superestimativa da temperatura do ar (cerca de 3ºC em Curucutu e Santa Virgínia e 2ºC em Itanhaém, e que tal erro é sistemático. A previsão de temperatura do ar com 24-h de antecedência foi superior às demais. De forma geral o modelo apresentou uma tendência de subestimar (superestimar a freqüência de ocorrência de ventos calmos cuja velocidade é inferior a 1 m.s-1 (fortes – superior a 7 m.s-1. A direção dos ventos é a variável mais difícil de prever devido provavelmente às diferenças entre a topografia do modelo e a topografia real, contudo nota-se que o modelo produz o giro característico na direção ao longo do dia, provocado pelo aquecimento diurno. Full Text Available The aim of this paper is to apply, as a way of interpretation of Virgil’s Fourth Bucolic, verses 18-25, the idea that one of his “inspiration” sources could have been the visual and iconic universe in which the poet flourished. Taking into account the argumentation about different types of ekphraseis, we try to define what we call “implicit ekphrasis”, that is, a kind of ekphrasis not produced by isolated artistic objects, but another one based on concepts, in order to apply it to our verses commentary. The results could allow us to say that the icons which, at Rome, symbolized aeternitas, aurea aetas, felicitas temporum, tempora anni, etc., permit the poet (among other inspiration sources to build his message of a better time through the puer nascens’s metaphor.

The virA and virG gene products are required for the regulation of the vir regulon on the tumor-inducing (Ti) plasmid of Agrobacterium tumefaciens. VirA is a membrane-associated protein which is homologous to the sensor molecules of other two-component regulatory systems. The authors overproduced truncated VirA proteins in Escherichia coli be deleting different lengths of the 5'-coding region of the virA gene and placing these genes under lacZ control. These proteins were purified from polyacrylamide gels and renatured. The renatured proteins became radiolabeled when they were incubated with [γ- 32 P]ATP but not with [γ- 32 P]GTP or [α- 32 P]ATP, which suggests an ATP γ-phosphate-specific autophosphorylation. The smallest VirA protein, which retained only the C-terminal half of the protein, gave the strongest autophosphorylation signal, which demonstrates that the C-terminal domain has the autophosphorylation site. The phosphorylated amino acid was identified as phosphohistidine, and a highly conserved histidine was found in all of the VirA homologs. When this histidine was changed to glutamine, which cannot be phosphorylated, the resulting VirA protein lost both its ability to autophosphorylate and its biological function as a vir gene regulator. Full Text Available This paper aims to approach, even though in a briefly way, the subject of invention in the training of teachers. In order to do this, we will make a brief overview on Virgínia Kastrup contributions about this issue. One of the questions is the time - through Henri Bergson’s duration concept and its using in training theme. From this point, we will try to deconstruct a view about the identity reference in contemporaneity toward to multiple and open ways of existence. In the inventive movements of the processuality, the time is considered as an expression of human being individuation. It means to last refer to an immediacy of being, of being time, which are modulable and mutant, always in a creation process. In a movement of building a body which leaves an identity reference – that is opposite of self-transformation – necessarily we report us to the relation between the idea of time as duration and the individuation principle by Gilberto Simondon. With these concepts and their possible resonance in the discussion about formative politics, we can develop a view over the invisible to training of teachers and over the learners and another ways of operating in the world. The invention as a lever direct us to a field of possibilities to conceive and experience another time (and space which is producer of subjectivity, singularities and differences.

Host and bacterial proteins that repress recruitment of LC3 to Shigella early during infection.

Full Text Available Shigella spp. are intracytosolic gram-negative pathogens that cause disease by invasion and spread through the colonic mucosa, utilizing host cytoskeletal components to form propulsive actin tails. We have previously identified the host factor Toca-1 as being recruited to intracellular S. flexneri and being required for efficient bacterial actin tail formation. We show that at early times during infection (40 min., the type three-secreted effector protein IcsB recruits Toca-1 to intracellular bacteria and that recruitment of Toca-1 is associated with repression of recruitment of LC3, as well as with repression of recruitment of the autophagy marker NDP52, around these intracellular bacteria. LC3 is best characterized as a marker of autophagosomes, but also marks phagosomal membranes in the process LC3-associated phagocytosis. IcsB has previously been demonstrated to be required for S. flexneri evasion of autophagy at late times during infection (4-6 hr by inhibiting binding of the autophagy protein Atg5 to the Shigella surface protein IcsA (VirG. Our results suggest that IcsB and Toca-1 modulation of LC3 recruitment restricts LC3-associated phagocytosis and/or LC3 recruitment to vacuolar membrane remnants. Together with published results, our findings suggest that IcsB inhibits innate immune responses in two distinct ways, first, by inhibiting LC3-associated phagocytosis and/or LC3 recruitment to vacuolar membrane remnants early during infection, and second, by inhibiting autophagy late during infection.

Water beetles in mountainous regions in southeastern Brazil

Full Text Available Inventories provide information on the state of biodiversity at a site or for a geographic region. Species inventories are the basis for systematic study and critical to ecology, biogeography and identification of biological indicators and key species. They also provide key information for assessments of environmental change, for natural resource conservation or recovery of degraded ecosystems. Thus, inventories play a key role in planning strategies for conservation and sustainable use. This study aimed to inventory the fauna of water beetles, larvae and adults, in two mountainous regions in the state of São Paulo, in Serra da Mantiqueira (Parque Estadual de Campos do Jordão and Pindamonhangaba region and in Serra do Mar (Santa Virgínia and Picinguaba Divisions as well as to generate information about the habitats used by the different genera recorded. Specimens were collected in lotic and lentic systems, between the years 2005 to 2010. In total 14,492 specimens were collected and 16 families and 50 genera of Coleoptera were identified. This study in mountainous regions showed a significant portion of the faunal composition of South America and the state of São Paulo. The composition of the fauna, in terms of richness and abundance by family, indicated the predominance of Elmidae, followed by Hydrophilidae and Dytiscidae. Despite the diversity found, the results of estimated richness indicated the need for additional sampling effort for both regions, since the curves of estimated richness did not reach an asymptote, suggesting that new species can be found in future surveys.

Unauthorised Biographies: An Analysis Under the Perspective of Robert Alexy's External Theory

Full Text Available The present paperwork aimed to analyse whether or not the publication of a biography should require the prior authorisation from the subject  or their family. As there is, in this case, a collision between personality rights and freedom of speech, the proportionality test was used to solve such conflict. The first step was to define biography as a non-ficctional genre. The paperwork then aimed to define the theory of the principles, the external theory of the restriction of fundamental rights and the proportionality test developed by jurist Robert Alexy, which are largely used in Brazil by authors like Virgílio Afonso da Silva, Ingo Wolfgang Sarlet and Daniel Sarmento. It was observed that when fundamental rights collide, the proportionality must be verified and as a result there is a conditional precedence relation between the rights. In such case, the ceding right will be restricted by the preciding one. The external theory is a recognised manner of verifying whether the restriction of a fundamental right is legit or if it is a violation. The need for prior authorisation, based on the personality protection, restricts freedom of speech, on the other hand, its exemption is based on the right to freedom of speech. By applying the aforementioned method, a particular rule was developed and the conditions for it to be applicable: freedom of speech prevails over personality rights in given conditions (public figure, over suitable means, no expectation for privacy in the publication, diligence in the verification of truth or social and journalistic interest over the information

Differential regulation of caspase-1 activation, pyroptosis, and autophagy via Ipaf and ASC in Shigella-infected macrophages.

Full Text Available Shigella infection, the cause of bacillary dysentery, induces caspase-1 activation and cell death in macrophages, but the precise mechanisms of this activation remain poorly understood. We demonstrate here that caspase-1 activation and IL-1beta processing induced by Shigella are mediated through Ipaf, a cytosolic pattern-recognition receptor of the nucleotide-binding oligomerization domain (NOD-like receptor (NLR family, and the adaptor protein apoptosis-associated speck-like protein containing a C-terminal caspase recruitment domain (ASC. We also show that Ipaf was critical for pyroptosis, a specialized form of caspase-1-dependent cell death induced in macrophages by bacterial infection, whereas ASC was dispensable. Unlike that observed in Salmonella and Legionella, caspase-1 activation induced by Shigella infection was independent of flagellin. Notably, infection of macrophages with Shigella induced autophagy, which was dramatically increased by the absence of caspase-1 or Ipaf, but not ASC. Autophagy induced by Shigella required an intact bacterial type III secretion system but not VirG protein, a bacterial factor required for autophagy in epithelial-infected cells. Treatment of macrophages with 3-methyladenine, an inhibitor of autophagy, enhanced pyroptosis induced by Shigella infection, suggesting that autophagy protects infected macrophages from pyroptosis. Thus, Ipaf plays a critical role in caspase-1 activation induced by Shigella independently of flagellin. Furthermore, the absence of Ipaf or caspase-1, but not ASC, regulates pyroptosis and the induction of autophagy in Shigella-infected macrophages, providing a novel function for NLR proteins in bacterial-host interactions.

We report the first community-based evaluation of Shigella flexneri 2a strain SC602, a live, oral vaccine strain attenuated by deletion of the icsA (virG) plasmid virulence gene, given at 10(4) CFU. The primary objectives of this trial were to determine the safety and immunogenicity of the vaccine and to determine the duration of colonization. Four of 34 volunteers experienced transient fevers, and three reported diarrhea during the first 3 days of the study. Half of the volunteers mounted a positive serum immunoglobulin A (IgA) response to S. flexneri lipopolysaccharide. All but one of the volunteers excreted the vaccine in their stools for 1 to 33 days, and this excretion was often intermittent. Data from the community-based study were supplemented with an inpatient trial in which three volunteers received 10(3) and nine received 10(4) CFU. All volunteers who received 10(3) CFU excreted SC602 and had an IgA antibody-secreting cell response. Two of these had a serum IgA response. Six of the nine volunteers who received 10(4) CFU excreted SC602. One vaccinee had a transient fever and two met the definition of diarrhea. Six volunteers that received 10(4) CFU had an antibody-secreting cell response, and four had a serum IgA response. SC602 has now been tested at 10(4) CFU in a total of 58 volunteers. The cumulative results of these clinical trials, reported here and previously (Coster et al., Infect. Immun. 67:3437-3443, 1999), have demonstrated that SC602 is a substantially attenuated candidate vaccine that can evoke protection against the most severe symptoms of shigellosis in a stringent human challenge model of disease.

As variedades comerciais podem ser reunidas em três grupos - Virgínia, Spanish e Valência - de acôrdo com a distribuição das gemas vegetativas e reprodutivas e também com o número de sementes por fruto. Nêste trabalho é apresentado um estudo da morfologia, duração e fertilização da flor, mostrando que no amendoim não existe a suposta distinção de flôres férteis e estéreis. Também a formação do fruto é descrita, mostrando a interessante característica desta planta, que é ter flôres aéreas e frutos subterrâneos.This work is based mostly on the descriptions given in Smith's paper "Arachis hypogxa L. Aerial flower and Subterranean Fruit" (2 and in the symposium "The Peanut. The unpredictable Legume" (3, and also on some of the author's observations. All cultivated peanut varieties belong to the species Arachis hypogxa L.; other species of the genus are not used for commercial production and may be of interest only for breeding purposes. Commercial peanut varieties can be grouped into one of three types : Virginia, Spanish, and Valencia. This grouping is done according to the distribution of vegetative and reproductive buds on the plant, and to the number of seeds per fruit. Studies were made on flower morphology, its duration and fertilization ; they indicated that all peanut flowers are potentially fertile and cannot, therefore, be classed into fertile and infertile types. A description of how the aerial flowers produce subterranean fruits is given.

Full Text Available O artigo faz um resumo do relatório de três projetos pelos quais buscamos entender a educação no período que convencionou-se denominar de nacional-desenvolvimentista. Os projetos permitiram entender a educação num contexto marcado por novo ciclo de desenvolvimento do modo capitalista de produção que, estando em plena “era dos impérios”, e que bem poderia ser denominada “era do capitalismo monopolista e financeiro”, num contexto marcado pela Guerra Fria e pelo Plano Marshall, teve como particularidade o fortalecimento das ideologias nacionalistas e desenvolvimentistas. Os projetos ajudaram a aprofundar o entendimento dos mecanismos usados pela UNESCO no Brasil, particularmente no Estado da Bahia onde, através do “Projeto Colúmbia”, Anísio Teixeira e o grupo de intelectuais que lhe eram próximos implementou um amplo leque de iniciativas que expressavam as políticas liberais hegemônicas; o mesmo também se deu no Estado de São Paulo onde teve atuação destacada Fernando de Azevedo, Florestan Fernandes, Octávio Ianni, Fernando Henrique Cardoso, Oracy Nogueira, Virgínia L. Bicudo, Aniela Ginsberg e outros, implementando outros projetos e articulações. O texto foi estruturado em quatro partes que, indo do geral para o particular, busca didaticamente explicar a educação no contexto de transformações internacionais do modo capitalista de produção no pós II GUERRA MUNDIAL; em seguida dedica ao entendimento de como essas transformações se deram no Brasil onde, pela articulação da ideologia nacional-desenvolvimentista; na terceira parte encontra-se sintetizado o longo debate sobre a educação e o ensino que aqui ocorreu nesse período; finalmente adentra-se no entendimento das concepções e idéias pedagógicas que animaram os debates e embates ocorridos, bem como as várias ações no campo educacional levadas a cabo nesse período que, rigorosamente, mantém suas bandeiras atuais até os dias de hoje.

Full Text Available Aunque los gasterópodos son uno de los grupos más abundantes en los manglares del Caribe, poco se sabe del ámbito de especies particulares debido al enfoque comunitario de la mayoría de los estudios. El ámbito de las poblaciones de L. angulifera y N. virginea fue estudiado en la Ecorregión Darién, Caribe colombiano. El muestreo se realizó entre junio y agosto 2009, se recolectaron 3 963 individuos de ambas especies, a los cuales se les midió la talla de la concha. La Ecorregión presentó diferencias espaciales en el ámbito de la salinidad del agua a nivel superficial, la cobertura de manglar y el ámbito de las poblaciones. En la parte externa de la Ecorregión, L. angulifera presentó el mayor porcentaje de aparición (84.8% en comparación a N. virginea (15.2%. En el interior N. virginea presentó el mayor porcentaje (84.6%, mientras L. angulifera apareció solo en bahía Turbo (15.4%. Se encontraron diferencias en el ámbito de tallas promedio de las conchas, donde L. angulifera varió entre 6 y 22mm y N. virginea entre 6 y 12mm dentro del Golfo. Se confirma que L. angulifera es una especie de carácter estenohalina y marina, mientras que N. virginea es eurihalina y estuarina.

Os dados foram analisados por meio de software estatístico. RESULTADOS: Os problemas de sono mais comum entre os idosos foram: dormir menos de 4 horas por noite (84,6%, sonolência diurna (71,2%, despertares noturnos (71,2%, uso de medicamentos para dormir (21,2%. A análise de associação mostrou-se significativa com as variáveis, idade (p=0,05, presença de morbidade (p=0,02, tabagismo (p=0,01 e dor ao mover-se (p=0,05. CONCLUSÃO: As demandas para a necessidade de dormir/repousar nos idosos estiveram associadas significativamente a fatores socioeconômicos e condições de saúde.

Full Text Available Aunque los gasterópodos son uno de los grupos más abundantes en los manglares del Caribe, poco se sabe del ámbito de especies particulares debido al enfoque comunitario de la mayoría de los estudios. El ámbito de las poblaciones de L. angulifera y N. virginea fue estudiado en la Ecorregión Darién, Caribe colombiano. El muestreo se realizó entre junio y agosto 2009, se recolectaron 3 963 individuos de ambas especies, a los cuales se les midió la talla de la concha. La Ecorregión presentó diferencias espaciales en el ámbito de la salinidad del agua a nivel superficial, la cobertura de manglar y el ámbito de las poblaciones. En la parte externa de la Ecorregión, L. angulifera presentó el mayor porcentaje de aparición (84.8% en comparación a N. virginea (15.2%. En el interior N. virginea presentó el mayor porcentaje (84.6%, mientras L. angulifera apareció solo en bahía Turbo (15.4%. Se encontraron diferencias en el ámbito de tallas promedio de las conchas, donde L. angulifera varió entre 6 y 22mm y N. virginea entre 6 y 12mm dentro del Golfo. Se confirma que L. angulifera es una especie de carácter estenohalina y marina, mientras que N. virginea es eurihalina y estuarina.Distribution of the mangrove gastropods Neritina virginea (Neritidae and Littoraria angulifera (Littorinidae within the Colombian Caribbean Darién Ecoregion. Gastropods are one of the most abundant groups within the Caribbean mangroves, however, little is known about the distribution of particular species at a regional scale. With this aim, we studied the geographic distribution of Littoraria (Littorinopsis angulifera and Neritina (Vitta virginea within the Darién Ecoregion in the Caribbean coast of Colombia, from 77 sampling stations along 609km between the Colombian-Panamá border and Córdoba State, Colombia. The fieldwork was conducted in June-August 2009, and a total of 3 963 individuals of both species were hand-picked from the ground, prop-roots and trunks along 50m transects, and shell sizes were measured. The description of geographic patterns considered surface water salinity, mangrove cover and gastropod distribution within the Gulf of Urabá. In the outer-most part of the Gulf, L. angulifera was present in 84.8% of the stations, while N. virginea was only present in 15.2% of the stations. In this part, mangroves areas were patchily distributed, and the gastropods (mainly L. angulifera were found on woody debris along the supralittoral zone in sandy shores. In the inner-most part, in contrast, N. virginea occurred in 84.6% of the stations, mostly in estuaries, deltas and river margins, while L. angulifera only appeared in Turbo Bay (15.4%. Mean shell size also exhibited a clear geographic pattern: size range was 6-22mm in L. angulifera, and 6-12mm in N. virginea. L. angulifera was found in open-water stations with water salinities >10PSU, but it was absent in sites with lower salinities like the Atrato River Delta and other small rivers. Its presence on coastal woody debris suggests that despite of the recruitment of small individuals from the nearshore stock of larvae, populations are unable to establish due to the absence of mangroves protection. Oppositely, N. virginea was found under estuarine conditions on mangrove roots and ground. Our results confirm that L. angulifera is an esteno-tolerant marine species, and N. virginea is an eury-tolerant estuarine species, thus their geographic distribution is strongly shaped by the large freshwater discharge of the Atrato River. We hypothesize that absence or limited distribution of gastropods in various areas of the Darién Ecoregion may be further explained by the poor conservation state of mangroves.

Full Text Available Prezados Leitores, Nesta edição especial, a Encontros Bibli publica os trabalhos apresentados no 3º Simpósio Brasileiro de Comunicação Científica (SBCC, realizado na Universidade Federal de Santa Catarina (UFSC, em Florianópolis, nos dias 5 e 6 de junho de 2012. É uma grande satisfação estabelecer parceria com este evento, pois o SBCC é um importante fórum que agrupa especialistas, promove discussões e o compartilhamento de informações relacionadas à área de comunicação científica. Nesta edição de 2012, o evento concentrou-senos temas de acesso aberto, acesso livre e preservação digital, com foco em Perspectivas em Acesso Aberto – Cenários para 2020. O trabalho que abre esta edição fundamentou a palestra da professora pesquisadora Elena Maceviciute da Universidade de Boras - Suécia, e também da Universidade Vilnius - Lituânia, e destaca diversos questionamentos e alternativas para a preservação digital. statements.Recent linguistic studies provide some new results regarding the word karst.The paper repeats some well known facts about the origin of the term karst but at the same time it gives some new results and interpretations.The name of the plateau in the background of the Trieste Bay (the Adriatic Sea) which the Slovenes call Kras,Italians Carso and Germans Karst is of pre-roman origin and Latinised into Carsus.The original name had the base * Karus-( Ptolemy wrote Kαpouσαδíωóρε) from the root * kar- meaning rock,stone.From the Latinised form Carsus developed Slovene,Italian and German names according to the rules of their languages.From the Central Europe the easiest and practically the only way leading to the Mediterranean was the road from Vienna to the port of Trieste.Travellers across the Karst described this unusual country and its natural phenomena,mostly with negative connotation and subsequently other limestone landscapes were compared to Karst.In 1830 F.J.H.Hohenwart wrote that "karst is not on the Karst only,but it stretches from the plain of Friuli to the Greek islands".Most of the descriptions of the Karst from that period were published in German language and this is the reason why the German form of the name became the international karstological term,the karst.

Full Text Available Objetiva, a partir das pesquisas aplicada (prototipagem, explicativa e bibliográfica, e de abordagem qualitativa, apresentar uma proposta de catálogo bibliográfico 2.0 na perspectiva dos sistemas gerenciadores de conteúdo (CMS, em consonância com os preceitos da Web 2.0 e com as ações de representação da informação da biblioteconomia (catalogação e indexação. Resulta em um catálogo bibliográfico 2.0 desenvolvido no CMS WordPress mediante o uso do template Fashionista, e dos plug-ins Book Review Library, que possibilita a realização de ações de representação da informação; e Rating-Widget: Star Rating, que permite avaliar os itens do catálogo. Conclui-se que a proposta apresenta um ambiente dinâmico e intuitivo que permite e incentiva a participação ativa dos usuários mediante avaliação dos registros, sugestão de livros cujos gêneros sejam similares e rotulação livre, diferentemente das propostas dos tradicionais catálogos públicos de acesso em linha, sem participação do usuário, dificuldades na navegação e insatisfação nos processos de busca.
