    Full Text Available Based on VPA-estimates of abundance, survey data and commercial catch statistics of Northeast Arctic Greenland halibut (Reinhardtius hippoglossoides Walbaum, the paper describes trends by year-class and age in the distribution of Greenland halibut between surveyed and not-surveyed areas. Changes in the distribution of I-group around 1990 to areas beyond the Svalbard surveys has previously been described and related to temperature changes in the Spitsbergen Current. This paper shows that this displaced distribution of the 1989-94 year-classes persisted up to age 7. The results indicate that the displacement was an extraordinary situation and other similar distribution shifts have not occurred during the last 30 years or more. Further, the shift co-occurred with extreme levels of the 137 year long time series of the index of the North-Atlantic Oscillation (NAO. The results are discussed in relation to stock management and climate change.

  13. Epitheliocystis in lake trout Salvelinus namaycush (Walbaum) is associated with a β-proteobacteria. (United States)

    Lake trout Salvelinus namaycush (Walbaum) raised for stocking experienced yearly (2011-13) winter epizootics of epitheliocystis. Affected fish were dispersed on the bottom of the tank, had decreased feed and fright response, and mortality often reached 40%. Peak mortality occurred within 3 weeks of the appearance of clinical signs, and outbreaks typically lasted 6 weeks. Affected fish had no gross lesions but histologically had branchial epithelial necrosis and lamellar hyperplasia, with small to large numbers of scattered epithelial cells containing 10- to 20-μm inclusions. A longitudinal study was undertaken of one annual outbreak, and lamellar hyperplasia was most closely associated with mortality. The number of inclusions was statistically greater (P < 0.05) before and during peak mortality, but inclusions were present in low numbers before clinical signs occurred. Results of histochemical staining, immunohistochemistry and transmission electron microscopy supported the presence of a β-proteobacteria rather than a Chlamydiales bacterium within inclusions. PCR primers to identify Chlamydiales did not give consistent results. However, the use of universal 16S rDNA bacterial primers in conjunction with laser capture microdissection of inclusions demonstrated that a β-proteobacteria was consistently associated with affected gills and is more likely the cause of the disease in lake trout. © 2015 John Wiley & Sons Ltd.

  14. Puffy Skin Disease Is an Emerging Transmissible Condition in Rainbow Trout Oncorhynchus mykiss Walbaum (United States)

    The transmission of puffy skin disease (PSD) to rainbow trout Oncorhynchus mykiss Walbaum was tested in the laboratory by conducting co-habitation challenges with puffy skin (PS)-affected fish (Trojans) collected from the field. Two separate challenges were conducted using Trojans sourced from two different sites and diploid (first trial) or triploid (second trial) naïve fish. PSD-specific clinical signs were observed in both groups of naïve fish, with 66% of the fish sampled during the challenges showing signs of varying severity. The first clinical features of PSD were presented as white oval skin patches on one or both flanks 15–21 days post-challenge (dpc). The extent of the lesions ranged from 10 to 90% of the body surface, depending on the severity of the lesion. Both the severity and number of affected fish increased during the challenge. Macroscopically, oedema of the skin and multifocal petechial haemorrhaging were observed towards the end of the trials. Abnormal fish behaviour consisting of “flashing” and excessive mucous production was noted from 15 dpc onwards. Fish with severe PSD lesions also displayed inappetence and associated emaciation. Rodlet cells were observed in 41% of the fresh skin scrapes analysed from the second trial. Histologically epidermal oedema was observed in 31% of the naive fish showing gross pathology, with additional 12% displaying epidermal hyperplasia, mostly observed at the end of the challenge. Other concomitant features of the PSD lesions in challenged fish were epithelial erosion and sloughing, and occasionally mild or focal inflammation. No consistent pathology of internal organs was observed. The parasites Ichthyophthirius multifiliis and Ichthyobodo necator were observed in skin samples of a proportion of naïve challenged fish and in Trojans but not in control fish.   16. Differences in Ichthyophonus prevalence and infection severity between upper Yukon River and Tanana River chinook salmon, Oncorhynchus tshawytscha (Walbaum), stocks (United States)

    Two genetically distinct populations of chinook salmon, Oncorhynchus tshawytscha (Walbaum), were simultaneously sampled at the confluence of the Yukon and Tanana rivers in 2003. Upper Yukon-Canadian fish had significantly higher infection prevalence as well as more severe infections (higher parasite density in heart tissue) than the lower Yukon-Tanana River fish. Both populations had migrated the same distance from the mouth of the Yukon River at the time of sampling but had significantly different distances remaining to swim before reaching their respective spawning grounds. Multiple working hypotheses are proposed to explain the differences between the two stocks: (1) the two genetically distinct populations have different inherent resistance to infection, (2) genetically influenced differences in feeding behaviour resulted in temporal and/or spatial differences in exposure, (3) physiological differences resulting from different degrees of sexual maturity influenced the course of disease, and (4) the most severely infected Tanana River fish either died en route or fatigued and were unable to complete their migration to the Tanana River, thus leaving a population of apparently healthier fish. ?? 2006 Blackwell Publishing Ltd.

  17. Impact of production technology on morphological lesions in the gills of commercial rainbow trout (Oncorhynchus mykiss, Walbaum 1792). (United States)

    The most popular rainbow trout (Oncorhynchus mykiss, Walbaum 1792) production technologies include both an extensive method with the flow through system (FTS) and an intensive method with the recirculating aquaculture system (RAS). Their impact on the fish was evaluated with a morphological assessment of the gills, as these are organs susceptible to environmental changes. Trout of 350 - 500 g body mass were caught for trial in spring and autumn, with 36 fish originating from 3 fish farms with the FTS system and an equal number from 3 RAS fish farms (n = 6). The fish were macroscopically examined and the gills were investigated microscopically (haematoxylin and eosin staining). Hypertrophy and hyperplasia were most commonly detected, amounting to 89% of all structural abnormalities. These lesions were slightly more common in the FTS, especially in autumn, whereas the changes to the blood vessels in the gills were observed more frequently in the rainbow trout from the RAS system than in the fish obtained from the FTS technology (the difference was statistically significant). The morphological lesions in the mucous cells of the gills were detected at a comparable severity regardless of the technology or production season. The predominantly low values of the histopathologic indices, which described the microscopic lesions in the gills of fish from the FTS and RAS systems, showed that the examined organ was most often free of lesions or demonstrated only minor morphological damage regardless of the production technology.

  18. Impacts of chloramine-T treatment on antioxidant enzyme activities and genotoxicity in rainbow trout, Oncorhynchus mykiss (Walbaum). (United States)

    Juvenile rainbow trout Oncorhynchus mykiss (Walbaum) were exposed to therapeutic, and higher concentrations of chloramine-T (Cl-T) to assess the effects of this chemical on the antioxidant enzyme system and genetic structure. Red blood cells acetylcholinesterase, ∆-aminolevulinic acid dehydratase, paraoxonase and liver glutathione S-transferase activity were increased at 10 and 20 mg L(-1) Cl-T-exposed fish, while they were decreased at 30 mg L(-1) Cl-T-exposed fish. On the other hand, liver catalase activity and liver protein levels increased at 10 mg L(-1) and decreased at 20 and 30 mg L(-1) concentrations of Cl-T. Liver super-oxide dismutase activity decreased at 10 mg L(-1) and 20 mg L(-1) Cl-T and increased at 30 mg L(-1) of Cl-T. Compared to control, comet assay indicated that Cl-T did not cause significant DNA damage to red blood cells of the fish. Results indicate that 10 or 20 mg L(-1) Cl-T can be safely used to prevent or treat external parasitic and bacterial infection of rainbow trout. © 2013 John Wiley & Sons Ltd.

  20. Taxonomy and host-specificity of Gyrodactylus aideni n. sp. and G. pleuronecti (Monogenea: Gyrodactylidae) from Pseudopleuronectes americanus (Walbaum) in Passamaquoddy Bay, New Brunswick, Canada. (United States)

    Wild and cultured winter flounder Pseudopleuronectes americanus (Walbaum) from Passamaquoddy Bay were surveyed for species of Gyrodactylus Nordmann, 1832. Two species were found: G. pleuronecti Cone, 1981 and G. aideni n. sp, both members of Malmberg's 'groenlandicus group'. Although the hard parts in the haptor are very similar in the two species, hamuli of G. aideni are consistently shorter than those of G. pleuronecti. The two species differed by 35 base pairs in the ITS 1, 5.8 and ITS 2 region. A BLAST search identified a variety of species of Gyrodactylus from marine fishes in the Atlantic Ocean as closest matches, indicating the 'groenlandicus group' is part of a major marine lineage within Gyrodactylus (sensu lato) that has successfully radiated among coastal percid, pleuronectid, cottid and anarhichadid fishes. Exposure experiments suggested that winter flounder is the primary host of both species of parasites and that three other pleuronectid species in the bay may potentially serve only as occasional transport hosts.

Her grupta her biri 101 gram başlangıç ağırlığındaki 20 balık vardı. Günlük yem tüketimi bir denklemle belirlendi: y=k*başlangıç BW*(1+kd, (d =deney gün sayısı, k = yem artışı oranı ve BW: Vücut ağırlığı. Sekiz besleme yoğunluğu için seçilen k değerleri 0.005 ve 0.02 aralığındaydı. Her besleme yoğunluğu için üç tekrar yapıldı. Her grup 3 kilogram yem tüketti. Deney süresi 229 gün ile 55 gün arasında değişti. Yem çevirim verimi, orta yoğunluktaki beslemede (0.010-0.0125 en yüksek değerdeydi ve yemleme yoğunluğu düştükçe artış gösterdi ve orta yoğunluktan aşağı seviyelerde azalışa geçti. Bu etki vücut ağırlık kazanımında da görüldü. Canlı ağırlık kazanımında protein konsantrasyonu yemleme yoğunluğu arttıkça düştü ancak canlı ağırlık yağ kazanımı arttı, enerji konsantrasyonu bu artışa bağlı olarak yükseldi.     We examined 1454 juvenile Chinook salmon, Oncorhynchus tshawytscha (Walbaum), captured in nearshore waters off the coasts of Washington and Oregon (USA) from 1999 to 2004 for infection by Renibacterium salmoninarum, Nanophyetus salmincola Chapin and skin metacercariae. The prevalence and intensities for each of these infections were established for both yearling and subyearling Chinook salmon. Two metrics of salmon growth, weight residuals and plasma levels of insulin-like growth factor-1, were determined for salmon infected with these pathogens/parasites, both individually and in combination, with uninfected fish used for comparison. Yearling Chinook salmon infected with R. salmoninarum had significantly reduced weight residuals. Chinook salmon infected with skin metacercariae alone did not have significantly reduced growth metrics. Dual infections were not associated with significantly more severe effects on the growth metrics than single infections; the number of triple infections was very low and precluded statistical comparison. Overall, these data suggest that infections by these organisms can be associated with reduced juvenile Chinook salmon growth. Because growth in the first year at sea has been linked to survival for some stocks of Chinook salmon, the infections may therefore play a role in regulating these populations in the Northeast Pacific Ocean. © 2014 John Wiley & Sons Ltd.

    Flavobacterium psychrophilum is the causative agent of bacterial coldwater disease (BCWD), which has a major impact on salmonid aquaculture globally. An Enterobacter species, C6-6, isolated from the gut of rainbow trout, Oncorhynchus mykiss (Walbaum), has been identified as a potential probiotic species providing protection against BCWD. This study examined the effects of alginate microencapsulation on the protective efficacy of C6-6 against BCWD in vivo when administered to rainbow trout fry orally or by intraperitoneal (IP) injection. Viable C6-6 bacteria were microencapsulated successfully, and this process (microencapsulation) did not significantly deteriorate its protective properties as compared to the administration of non-microencapsulated C6-6 bacteria. Both oral and IP delivery of C6-6 achieved significantly better protection than control treatments that did not contain C6-6 bacteria. The highest relative percent survival (RPS) resulted from IP delivery (71.4%) and was significantly greater than the highest oral RPS (38.6%). Successful intestinal colonization was not critical to protective effects of C6-6. The study showed that C6-6 administration, with or without encapsulation, was a viable choice for protecting fry from BCWD especially when administered intraperitoneally. © 2014 John Wiley & Sons Ltd.

    Full Text Available The present study was carried out to determine the effects of four rations on growth, chemical composition and digestibility of the Rainbow trout (Oncorhynchus mykiss Walbaum, 1792. Four test diets were formulated to have the same levels of protein (40% and energy (gross energy: around 4800 cal/g, however, these test diets were composed of basic diet with different carbohydrate (0, 3, 12 and 18% to lipid (0, 6, 15 and 18% ratios (Diet 1: 0/18; Diet 2: 3/15; Diet 3: 12/6 and Diet 4: 18/0. Fifty fish weighing 99.0 ± 0.65g were placed in each of the four experimental tanks and were fed with one of the experimental diets for 13 weeks consisting of two trial periods (Periods I and II. Growth parameters and chemical composition of fish flesh were monitored. Effects of the diets on apparent digestibility coefficients (ADC were also determined. At the end of the study, the fish growth was affected significantly (P0.05 changes in specific growth rate, although higher values were observed in fish fed with Diets 1 and 3. The experimental group fed with Diet 4 showed a lower (P0.05 changes in experimental groups fed with the diets. The percentage of water, protein and ash of fish flesh did not show any (P>0.05 change. However, the muscle lipid content of fish significantly (P<0.05 decreased as carbohydrate level increased. The ADC values were also affected (P<0.05 with carbohydrate and lipid levels in the diets. The ADC of lipid was lowest in the experimental group fed with Diet 1. The ADC values for dry matter, proteins and energy were lowest for fish fed on Diet 4, while for ash it was lowest for Diet 2. These results indicate that the best balance between the dietary lipid and carbohydrate levels was obtained on Diet 3, without lower growth.

  6. Evaluation of the suitability of a partially defatted black soldier fly (Hermetia illucens L.) larvae meal as ingredient for rainbow trout (Oncorhynchus mykiss Walbaum) diets. (United States)

    Two trials were performed to evaluate a partially defatted Hermetia illucens (HI) larvae meal as potential feed ingredient in rainbow trout (Oncorhynchus mykiss Walbaum) diets. In the first trial, 360 trout (178.9 ± 9.8 g of mean initial body weight) were randomly divided into three experimental groups (4 tanks/treatment, 30 fish/tank). The fish were fed for 78 days with isonitrogenous, isolipidic and isoenergetic diets containing increasing levels of HI, on as fed basis: 0% (HI0, control diet), 25% (HI25) and 50% (HI50) of fish meal substitution, corresponding to dietary inclusion levels of 0, 20% and 40%. In the second trial, 36 trout (4 tanks/treatment, 3 fish/tank) were used to evaluate the in vivo apparent digestibility coefficients (ADC) of the same diets used in the first trial. Survival, growth performance, condition factor, somatic indexes, and dorsal fillet physical quality parameters were not affected by diet. The highest dietary inclusion of HI larvae meal increased dry matter and ether extract contents of trout dorsal fillet. The use of HI larvae meal induced a decrease of valuable polyunsaturated fatty acids (PUFA) even if differences were only reported at the highest level of HI inclusion. The insect meal worsened the lipids health indexes of the same muscle. Dietary inclusion of insect meal did not alter the villus height of the fish. No differences were found among treatments in relation to ADC of ether extract and gross energy, while ADC of dry matter and crude protein were higher in HI25 if compared to HI50. The obtained results showed that a partially defatted HI larvae meal can be used as feed ingredient in trout diets up to 40% of inclusion level without impacting survival, growth performance, condition factor, somatic indexes, dorsal fillet physical quality parameters, and intestinal morphology of the fish.     Full Text Available A 120 days trial was conducted to investigate the effect of sea squirt (Halocynthia roretzi, Drasche 1884 tunic’s carotenoid to sea-reared rainbow trout (Oncorhynchus mykiss, Walbaum 1792 growth and muscle color. Sea-rearing was done at Tongyeong sea area, Korea. Three dietary treatments, namely control (C, CT, and AT, were administered to 6 groups of fish (n = 490-520. C was given basal diet which has contained 40 mg kg−1 of astaxanthin. A further inclusion of 10 mg kg−1 canthaxanthin was added to diet of CT, while AT’s feed was supplemented with 10 mg kg−1 H. roretzi tunics carotenoids extract. The result revealed that AT has the highest final weight (1119.2 ± 82.4 g compare to those of C (881.0 ± 121.2 g and CT (1068.2 ± 4.3 g. The specific growth rate (SGR of AT (1.0 ± 0.07%/day was significantly higher than C (0.7 ± 0.22%/day and CT (0.7 ± 0.25%/day while the feed conversion ratio (FCR were 1.5 ± 0.6, 1.4 ± 0.6, and 1.2 ± 0.1 for C, CT and AT respectively. The hepatosomatic index (HSI and Viscerosomatic index (VSI of all groups showed no significant difference (p > 0.05. The muscle color was also positively affected by the treatments, CT and AT were significantly different from C (p < 005. The initial muscle color score was 1.7 ± 0.0 and the final scores were 3.4 ± 0.2, 5.6 ± 0.1, and 5.7 ± 0.0 for C, CT, and AT respectively. Moreover, muscle carotenoids content of AT (8.5 ± 0.2 mg kg−1 was significantly higher (p < 0.05 than those of CT (6.9 ± 0.3 mg kg−1 and C (6.1 ± 0.2 mg kg−1.     Probiotics represent an alternative to chemotherapy and vaccination to control fish diseases, including lactococcosis caused by Lactococcus garvieae. The aims of this study were (i) to determine the in vitro probiotic properties of three bacteriocinogenic Lactococcus lactis subsp. cremoris of aquatic origin, (ii) to evaluate in vivo the ability of L. cremoris WA2-67 to protect rainbow trout (Oncorhynchus mykiss, Walbaum) against infection by L. garvieae, and (iii) to demonstrate the role of nisin Z (NisZ) production as an anti-infective mechanism. The three L. cremoris strains survived in freshwater at 18 °C for 7 days, withstood exposure to pH 3.0 and 10 % (v/v) rainbow trout bile, and showed different cell surface hydrophobicity (37.93-58.52 %). The wild-type NisZ-producer L. cremoris WA2-67 and its non-bacteriocinogenic mutant L. cremoris WA2-67 ∆nisZ were administered orally (10(6) CFU/g) to rainbow trout for 21 days and, subsequently, fish were challenged with L. garvieae CLG4 by the cohabitation method. The fish fed with the bacteriocinogenic strain L. cremoris WA2-67 reduced significantly (p LAB bacteriocin (NisZ) production as a mechanism to protect fish against bacterial infection. Our results suggest that the wild-type NisZ-producer strain L. cremoris WA2-67 could be used in fish farming to prevent lactococcosis in rainbow trout.

    Full Text Available The objective of our study is to test the efficacy of an extruded food on the growth performance of rainbow trout (Oncorhynchus mykiss Walbaum, 1792 at the fish farm Ain Aghbal in Azrou (Morocco. Generally, only the biochemical composition (percentages of moisture, proteins, lipids and carbohydrates and the (digestible and crude energies of the food are previously known, which is insufficient for the choice of the food in terms of growth performance and quality/environment. The rainbow trout breeding tanks used for this test are 222 days (15 June 2015 - 28 February 2016 are fed with water directly taken from the water of the Oum Er-Rbia sources according to an open circuit with a Renewal of water every half hour. The flow rate is 11.04m3/h. The initial breeding conditions, the density (Kg per volume 1.58kg/m3 and the initial charge with respect to the flow rate is 1.05 m3 / h. Fish are fed by ratios two to three times per day depending on the rearing stage. The frequency of sampling is biweekly where we measured the zootechnical performances of the fish and taken water samples for physicochemical analyzes in order to assess the quality of the waters of exit of the basins before their rejection in the Oum-Er-Rbia river. The results show that the test food shows a better growth of the rainbow trout and a conversion index not exceeding 0.9 for a size of 1043 g of trout.

    Full Text Available Results of stomach contents analyses of harp and hooded seals collected in West Greenland waters in the period 1986-1993 are reviewed, and compared with published data and circumstantial information from local hunters.  The diet of harp seals in this region is variable but consists mainly of pelagic crustaceans (Thysanoessa spp. and Parathemisto libellula and small fish species like capelin (Mallotus villosus, sandeel (Ammodytes spp., polar cod (Boreogadus saida and Arctic cod (Arctogadus glacialis. Species of importance for commercial fisheries in Greenland, such as Northern prawn (Pandalus borealis, Atlantic cod (Gadus morhua, and Greenland halibut (Reinhardtius hippoglossoides play a minor role in the diet of harp seals in this area. Variation in the diet of hooded seals is less well documented, but in addition to the species also taken by harp seals, larger demersal fishes like Greenland halibut, redfish (Sebastes spp., cod, and wolffish (Anarhichas minor are apparently important prey items.

  12. Susceptibility of juvenile sole Solea senegalensis to marine isolates of viral haemorrhagic septicaemia virus from wild and farmed fish. (United States)

    The susceptibility of sole Solea senegalensis to infection with 3 viral haemorrhagic septicaemia virus (VHSV) strains obtained from wild Greenland halibut Reinhardtius hippoglossoides and farmed turbot Psetta maxima was demonstrated. Fish were infected by an intraperitoneal (i.p.), immersion or cohabitational route, and maintained at 16 degrees C. Infection trials showed that VHSV isolates were pathogenic for sole fingerlings by i.p. injection and waterborne exposure causing moderate levels of mortality (10 to 55%). In addition, the mortality observed in fish cohabitating with i.p.-infected sole confirms horizontal transmission of the virus. However, the low rates of mortality registered in this challenge suggest that there is a low dissemination of virus by the i.p.-infected sole, which results in lower secondary challenge of the cohabitating fish. External signs of disease included haemorrhaging of the ventral area and ascitic fluid in the body cavity. Dead fish were tested for VHSV by both cell culture and RT-PCR assay, using pools of kidney and spleen from 10 individuals. Virus was recovered from most of the pools composed of dead fish. The results obtained in this study not only demonstrate the susceptibility of sole to the VHSV strains employed but also indicate that wild VHSV marine isolates represent a potential risk for sole aquaculture.

    Full Text Available Killer whale (Orcinus orca depredation (whales stealing or damaging fish caught on fishing gear adversely impacts demersal longline fisheries for sablefish (Anoplopoma fimbria, Pacific halibut (Hippoglossus stenolepis and Greenland turbot (Reinhardtius hippoglossoides in the Bering Sea, Aleutian Islands and Western Gulf of Alaska. These interactions increase direct costs and opportunity costs associated with catching fish and reduce the profitability of longline fishing in western Alaska. This study synthesizes National Marine Fisheries Service observer data, National Marine Fisheries Service sablefish longline survey and fishermen-collected depredation data to: 1 estimate the frequency of killer whale depredation on longline fisheries in Alaska; 2 estimate depredation-related catch per unit effort reductions; and 3 assess direct costs and opportunity costs incurred by longliners in western Alaska as a result of killer whale interactions. The percentage of commercial fishery sets affected by killer whales was highest in the Bering Sea fisheries for: sablefish (21.4%, Greenland turbot (9.9%, and Pacific halibut (6.9%. Average catch per unit effort reductions on depredated sets ranged from 35.1-69.3% for the observed longline fleet in all three management areas from 1998-2012 (p<0.001. To compensate for depredation, fishermen set additional gear to catch the same amount of fish, and this increased fuel costs by an additional 82% per depredated set (average $433 additional fuel per depredated set. In a separate analysis with six longline vessels in 2011 and 2012, killer whale depredation avoidance measures resulted in an average additional cost of $494 per depredated vessel-day for fuel and crew food. Opportunity costs of time lost by fishermen averaged $522 per additional vessel-day on the grounds. This assessment of killer whale depredation costs represents the most extensive economic evaluation of this issue in Alaska to date and will help

One component of the survey is mapping the upper pelagic zone using a trawl towed at several depths. However, the current technique with a single codend does not provide fine-scale spatial data needed to directly study species overlaps. An in-trawl camera system, Deep Vision, was mounted in front of the codend in order to acquire continuous images of all organisms passing. It was possible to identify and quantify of most young-of-the-year fish (e.g. Gadus morhua, Boreogadus saida and Reinhardtius hippoglossoides and zooplankton, including Ctenophora, which are usually damaged in the codend. The system showed potential for measuring the length of small organisms and also recorded the vertical and horizontal positions where individuals were imaged. Young-of-the-year fish were difficult to identify when passing the camera at maximum range and to quantify during high densities. In addition, a large number of fish with damaged opercula were observed passing the Deep Vision camera during heaving; suggesting individuals had become entangled in meshes farther forward in the trawl. This indicates that unknown numbers of fish are probably lost in forward sections of the trawl and that the heaving procedure may influence the number of fish entering the codend, with implications for abundance indices and understanding population dynamics. This study suggests modifications to the Deep Vision and the trawl to increase our understanding of the population dynamics.

    A Jaundice Syndrome occurs sporadically among sea-pen-farmed Chinook Salmon in British Columbia, the westernmost province of Canada. Affected salmon are easily identified by a distinctive yellow discolouration of the abdominal and periorbital regions. Through traditional diagnostics, no bacterial or viral agents were cultured from tissues of jaundiced Chinook Salmon; however, piscine reovirus (PRV) was identified via RT-rPCR in all 10 affected fish sampled. By histopathology, Jaundice Syndrome is an acute to peracute systemic disease, and the time from first clinical signs to death is likely Salmon, Sockeye Salmon and Atlantic Salmon, intraperitoneally inoculated with a PRV-positive organ homogenate from jaundiced Chinook Salmon, developed no gross or microscopic evidence of jaundice despite persistence of PRV for the 5-month holding period. The results from this study demonstrate that the Jaundice Syndrome was not transmissible by injection of material from infected fish and that PRV was not the sole aetiological factor for the condition. Additionally, these findings showed the Pacific coast strain of PRV, while transmissible, was of low pathogenicity for Atlantic Salmon, Chinook Salmon and Sockeye Salmon.

    Full Text Available Formaldehyde is commonly used as a disinfectant and also in a control of fish disease in aquaculture sector. However, this widespread use, can lead to environmental degradation and can cause negative effects on the treated fish with. In this study 250 mg/L (1 hour and 500 mg/L (45 min concentrations of formaldehyde were used. From the results of the histopathological findings degeneration was determined in the epithelial cells and pilar in the gill lamellae, lymphoid infiltration interlamellar necrosis and degeneration of the muscle tissue, dilatation in the liver, congestion in veins, degeneration in hepatocytes, damage in the blood vessels of fish which were treated with formaldehyde. In conclusion; formaldehyde was found to have a negative impact in histological examination in applied rainbow trout. Therefore, it was concluded that it should be used consciously and according the needs in aquaculture.

    60Co uptake from natural water, release and tissue distribution were investigated in the rainbow trout (Oncorhynchus mikiss), which is representative of the third order consumers of the freshwater trophic chains. The experiments were carried out on two groups of fingerling trout placed in 0.45 microm filtered river water, maintained at 12 +/- 0.5 degrees C, contaminated with about 30 Bq ml-1 of 60Co and renewed three times a week. After a 8-week exposure phase, the fish of one group were dissected to determine the contamination of the tissues and organs and the distribution of the accumulated 60Co. The fish of the other group were placed in non-contaminated water, renewed daily, to monitor radionuclide release. After a 42-day depuration phase, the specimens were dissected to study the tissue distribution of the residual 60Co.60Co accumulation from water by trout can be described by a one-compartment exponential model. The concentration factor, calculated from the ratio of the radionuclide concentration in filtered water and in the fish, reached a maximum value of 4.6 (w.w.) after 30 days exposure. After the 42-day depuration phase, the fish retained about 29% of the accumulated radionuclide. A single-compartment exponential model was fitted to the 60Co elimination data, and the corresponding radionuclide half-life was 21 days. At the end of the exposure phase, tissue contamination study showed 60Co accumulation by the gills, viscera (air bladder, heart and spleen) and kidneys, to be the highest. At the end of the depuration phase, the kidney was the most contaminated organ, followed by the viscera, head, gills, and liver. In both cases, 60Co concentration was by far the lowest in the muscle, which accounted for about 45% of the total body weight and only 20% of the total radionuclide body load.

  1. Development of a multiplex PCR for the genetic analysis of paddlefish (Polyodon spathula Walbaum,1792 populations

Paddlefish is commercially important species owing to its biological features and consumer characteristics, namely it produces valuable and delicious fish products, such as high quality meat and black caviar. Consequently, its cultivation under Ukrainian fish farm conditions and further realization in domestic and foreign markets are economically efficient. However, the paddlefish broodstock in Ukraine requires the efficient solution of increasing its productivity, identification and assessment of its genetic variation. Thus, the aim of our study was to develop and implement a multiplex PCR-analysis of paddlefish (Polyodon spathula for population-genetic monitoring of its artificial broodstocks in Ukraine. Methodology. A multiplex PCR was used for the study. The multiplex PCR development was performed for four microsatellite DNA markers: Psp12, Psp21, Psp26 and Psp28. Each investigated DNA loci, for which the multiplex PCR was optimized, was selected in such a way that the colored PCR products labeled with fluorescent dye did not overlap the length of the amplified fragments. Evaluation of the multiplex PCR effectiveness and processing of the data were performed by fragment analysis of DNA on the genetic analyzer ABI Prism 3130 (Applied Biosystem, USA. The size of the identified alleles was determined using the "Gene Mapper 3.7" program (Applied Biosystems, USA and LIZ-500 size standard (Applied Biosystems, USA. Results. Based on the results of capillary electrophoresis of multiplex PCR products, it was found that the amplified fragments for each of the four studied loci: Psp12, Psp21, Psp26 and Psp28 in one PCR reaction were within the expected size range. Data analysis on the electrophoregram demonstrated that Psp21 had the highest peak intensity at 611 fluorescent units (FU and the lowest peak intensity at 105 FU was observed for Psp26 locus. In the multiplex PCR after proper interpretation of the data we identified heterozygous allelic variants for Psp12 (218/220 bp, Psp26 (144/146 bp and Psp28 (256/258 bp. Herewith, the Psp21 locus (150/150 bp is identified in the homozygous state. Originality. The optimization was conducted and a multiplex PCR was developed for paddlefish genotyping using microsatellite DNA markers. Practical value. The obtained results can be used for paddlefish genotyping within selective-breeding programs for artificial reproduction of this fish species.

    Full Text Available Abstract Background Manta rays (Mobulidae, Mobula birostris and Mobula alfredi, are widely distributed in tropical and temperate waters. Still, little is known about their movements and their ecological interactions (e.g. behavior and diet. In Peru, M. birostris has only been reported along the northern shore within the Tropical East Pacific Marine Province. No official reports exist from central or south Peru within the Warm Temperate Southeastern Pacific Marine Province. Methods On December 22nd 2015, a rare sighting of a ~4 m disc width mobulid ray was recorded as video footage near the Palomino Islets, Lima, Peru. Results In situ observations and subsequent analysis of the footage confirmed that this single mobulid was a M. birostris with a melanistic color morph. Conclusions This sighting could be explained by the warm El Niño, primary (chl-a, and secondary (zooplankton productivity events during that same period. This represents the southernmost record of M. birostris in the southeastern Pacific (12°S and the first report of this species in the Warm Temperate Southeastern Pacific Marine Province and in the Humboldt Current Large Marine Ecosystem.

    Full Text Available In this study, anesthetics effects of spearmint (Menta piperita oil and lavandula (Lavandula angustifolia oil as alternative to clove (Eugenia caryophylatta oil were evaluated on rainbow trout (Oncorhynchus mykiss. Fish were exposed to different concentrations of the essential oils (30, 40, 50, 100, 150 and 200 mg l-1 for induction of anesthesia. Results showed that induction time decreased with increasing of the concentration of the clove oil and spearmint oil. However, recovery time increased with increasing of the concentration of this anesthetics. 40 and 50 mg l-1 doses of clove oil and 200 mg l-1 of mint oil were showed similar anesthetic effect. Lavandula oil was showed sedative effects on rainbow trout. These findings suggested that spearmint oil is useful anesthetics for rainbow trout.

  4. Movements of a deep-water fish: establishing marine fisheries management boundaries in coastal Arctic waters. (United States)

    Management boundaries that define populations or stocks of fish form the basis of fisheries planning. In the Arctic, decreasing sea ice extent is driving increasing fisheries development, highlighting the need for ecological data to inform management. In Cumberland Sound, southwest Baffin Island, an indigenous community fishery was established in 1987 targeting Greenland halibut (Reinhardtius hippoglossoides) through the ice. Following its development, the Cumberland Sound Management Boundary (CSMB) was designated and a total allowable catch (TAC) assigned to the fishery. The CSMB was based on a sink population of Greenland halibut resident in the northern section of the Sound. Recent fishing activities south of the CSMB, however, raised concerns over fish residency, the effectiveness of the CSMB and the sustainability of the community-based winter fishery. Through acoustic telemetry monitoring at depths between 400 and 1200 m, and environmental and fisheries data, this study examined the movement patterns of Greenland halibut relative to the CSMB, the biotic and abiotic factors driving fish movement and the dynamics of the winter fishery. Greenland halibut undertook clear seasonal movements between the southern and northern regions of the Sound driven by temperature, dissolved oxygen, and sea ice cover with most fish crossing the CSMB on an annual basis. Over the lifespan of the fishery, landfast ice cover initially declined and then became variable, limiting accessibility to favored fisher locations. Concomitantly, catch per unit effort declined, reflecting the effect of changing ice conditions on the location and effort of the fishery. Ultimately, these telemetry data revealed that fishers now target less productive sites outside of their favored areas and, with continued decreases in ice, the winter fishery might cease to exist.     Full Text Available Letrozole is a synthetic aromatase inhibitor and interfere in the committed step in the synthesis of endogenous estrogens from androgens. Also estrogens regulate the immune system in teleost. Changes of 17- β- esrtradiol (E2, serum immunoglobulin and lysozyme levels were measured using a method based on the ability of lysozyme to lyse the bacterium Micrococcus lysodeikticus, enzyme-linked immunosorbent assay (ELISA and ELISA respectively. Twelve broodstocks were injected weekly with 2.5 mg kg-1 letrozole (an endocrine disrupter component two months before spawning season and vaccinated intraperitoneally (i.p with a bacterin (inactivated L. garviae one month before spawning. Twelve broodstocks for vaccination and twelve female rainbow trout as control group were also immiunised (i.p with the bacterin and injected (i.p with PBS, respectively. In the group received 2.5 mg AI kg-1 per week, serum E2 levels were significantly lower than that of other groups. Total immunoglobulin level and lysozyme activity were significantly higher in the parents received 2.5 mg kg-1 per week and were immunized with 10-9 cells ml-1 Lactococcus garvieae  compared to the group which immunized with L. garvieae and the control (non- immunized. The present study, suggests that aromatase inhibitors such as letrozole may be a potential tool to regulate the synthesis of E2, is involved in the hormone- immune system interaction in rainbow trout.

    Full Text Available Rhinobatos percellens is one of three species of Rhinobatidae found on Brazilian shores and is one of the most abundant species on the shallow continental shelf of Paraná and Santa Catarina States. Specimens caught by artisanal fishery between July/2001 and March/2003 by fishing communities located on the frontier between two southern Brazilian States (Paraná and Santa Catarina had their stomach contents analyzed. According to the Alimentary Index (IAi, Decapoda (69% and Teleostei (22% were the main items consumed. Polychaeta, a common prey consumed by several benthonic fishes, was poorly represented in the feeding of R. percellens. This fact may be related to the availability of prey in the environment, or to the size of the guitarfish analyzed (ontogeny.     Full Text Available Rainbow trout at an average weight of 240 g were examined for the effect of dietary fructo–oligosaccharides in the diet on their growth and physiological state through selected biochemical parameters of the blood plasma. The prebiotic product Profeed® (experimental group, EG was administered on a continuous basis at a rate of g kg−1 of pellets for 105 days. The best growth performance for the EG was found in 42 days (363 ± 34.7 g vs. 340 ± 36.7 g, P = 0.003 and in 63 days (387 ± 35.6 g vs. 364 ± 42.3 g, P = 0.011. SGR of the fish from the EG was 0.69% and from the control group (CG was 0.70%. The feed conversion level was 0.82 in the EG and 0.86 in the CG. Survival rate was 99% (EG and 98% (CG. The results of the biochemical examination indicate significant differences in the creatinine (28 ± 5.5 vs. 22 ± 3.05 µmol L−1 and the sodium cation (157.9 1.66 vs. 155.7 ± 1.49 mmol L−1 level and in the catalytic concentration of alkaline phosphatase (5.18 ± 1.57 vs. 3.43 ± 0.78 µkat L−1. The positive results of the growth and biochemical tests as well as the favourable feed conversion suggest that it would be worthwhile to test higher concentrations of the Profeed® prebiotic product.

  10. Population Genetic Patterns of Threatened European Mudminnow (Umbra krameri Walbaum, 1792 in a Fragmented Landscape: Implications for Conservation Management.

    Full Text Available The European mudminnow (Umbra krameri is a Middle Danubian endemic fish species, which is characterised by isolated populations living mainly in artificial habitats in the centre of its range, in the Carpathian Basin. For their long term preservation, reliable information is needed about the structure of stocks and the level of isolation. The recent distribution pattern, and the population genetic structure within and among regions were investigated to designate the Evolutionary Significant, Conservation and Management Units (ESUs, CUs, MUs and to explore the conservation biological value of the shrinking populations. In total, eight microsatellite loci were studied in 404 specimens originating from eight regions. The results revealed a pronounced population structure, where strictly limited gene flow was detected among regions, as well as various strengths of connections within regions. Following the results of hierarchical structure analyses, two ESUs were supposed in the Carpathian Basin, corresponding to the Danube and Tisza catchments. Our results recommend designating the borders of CUs in an 80-90km range and 16 clusters should be set up as MUs for the 33 investigated populations. How these genetic findings can be used to better allocate conservation resources for the long term maintenance of the metapopulation structure of this threathened endemic fish is discussed.

  11. Effects of temperature on disease progression and swimming stamina in Ichthyophonus-infected rainbow trout, Oncorhynchus mykiss (Walbaum) (United States)

    Rainbow trout, Oncorhynchus mykiss, were infected with Ichthyophonus sp. and held at 10 ??C, 15 ??C and 20 ??C for 28 days to monitor mortality and disease progression. Infected fish demonstrated more rapid onset of disease, higher parasite load, more severe host tissue reaction and reduced mean-day-to-death at higher temperature. In a second experiment, Ichthyophonus-infected fish were reared at 15 ??C for 16 weeks then subjected to forced swimming at 10 ??C, 15 ??C and 20 ??C. Stamina improved significantly with increased temperature in uninfected fish; however, this was not observed for infected fish. The difference in performance between infected and uninfected fish became significant at 15 ??C (P = 0.02) and highly significant at 20 ??C (P = 0.005). These results have implications for changes in the ecology of fish diseases in the face of global warming and demonstrate the effects of higher temperature on the progression and severity of ichthyophoniasis as well as on swimming stamina, a critical fitness trait of salmonids. This study helps explain field observations showing the recent emergence of clinical ichthyophoniasis in Yukon River Chinook salmon later in their spawning migration when water temperatures were high, as well as the apparent failure of a substantial percentage of infected fish to successfully reach their natal spawning areas. ?? 2009 Blackwell Publishing Ltd.

  15. Infectious haematopoietic necrosis virus genogroup-specific virulence mechanisms in sockeye salmon, Oncorhynchus nerka (Walbaum), from Redfish Lake, Idaho (United States)

    Characterization of infectious haematopoietic necrosis virus (IHNV) field isolates from North America has established three main genogroups (U, M and L) that differ in host-specific virulence. In sockeye salmon, Oncorhynchus nerka, the U genogroup is highly virulent, whereas the M genogroup is nearly non-pathogenic. In this study, we sought to characterize the virus-host dynamics that contribute to genogroup-specific virulence in a captive stock of sockeye salmon from Redfish Lake in Idaho. Juvenile sockeye salmon were challenged by immersion and injection with either a representative U or M viral strain and sampled periodically until 14 days post-infection (p.i.). Fish challenged with each strain had positive viral titre by day 3, regardless of challenge route, but the fish exposed to the M genogroup virus had significantly lower virus titres than fish exposed to the U genogroup virus. Gene expression analysis by quantitative reverse transcriptase PCR was used to simultaneously assess viral load and host interferon (IFN) response in the anterior kidney. Viral load was significantly higher in the U-challenged fish relative to M-challenged fish. Both viruses induced expression of the IFN-stimulated genes (ISGs), but expression was usually significantly lower in the M-challenged group, particularly at later time points (7 and 14 days p.i.). However, ISG expression was comparable with 3 days post-immersion challenge despite a significant difference in viral load. Our data indicated that the M genogroup virus entered the host, replicated and spread in the sockeye salmon tissues, but to a lesser extent than the U genogroup. Both virus types induced a host IFN response, but the high virulence strain (U) continued to replicate in the presence of this response, whereas the low virulence strain (M) was cleared below detectable levels. We hypothesize that high virulence is associated with early in vivo replication allowing the virus to achieve a threshold level, which the host innate immune system cannot control. ?? 2009 Blackwell Publishing Ltd.

    Full Text Available Captive breeding programs of endangered fish species, such as the European mudminnow Umbra krameri, are essential for population restoration. To improve captive spawning and larvae rearing under controlled conditions, two experiments were carried out. In the first, the spawning substrate preference was tested in triplicate, where five different types of artificial surface were provided for mudminnow pairs:(isand, (iiartificial plants, (iiigravel, (ivsand + artificial plants and(vgravel + artificial plants. All fish preferred the gravel + artificial plant combination, which indicates that this type of surface could be the most appropriate for spawning in captivity. In the second trial, three feeding protocols were tested in triplicate under controlled conditions. In the first treatment fish were fed exclusively with Artemia nauplii; in the second treatment fish were fed with Artemiafor the first ten days then Artemia was gradually replaced with dry feed; for the third group the transition period started after 5 days of Artemia feeding. Although the survival rate of larvae could be maintained at a high level in some of the feeding protocols, a strong decrease in the growth rate was obvious in all diets containing dry food, which means that live food is essential for the first three weeks of mudminnow larvae rearing.

    Full Text Available En este artículo se analizan con parámetros de composición 58 puestas de trucha arcoiris obtenidas, manejadas y monitoreadas individualmente en desempeño desde la fertilización hasta finalizar la etapa de reabsorción de vesícula. Además de los valores reproductivos, el contenido de ácidos grasos en las ovas se determinó en fresco, mediante cromatografía de gases. En el perfil medio se destacan los ácidos palmítico (C16, oleico (C18:1n-9 y docosahexaenoico (C22:6n-3 como los más representativos, con casi el 60% del total y, en general, en concentraciones estables entre las hembras. Tanto para cada ácido determinado como para el conjunto de contenidos integrados de las series n-3 y n-6, y los colectivos de saturados (SAF, monoinsaturados (MUFA y poliinsaturados (PUFA, se define un patrón de composición similar al reportado para la especie en otros esquemas de manejo y bajo regímenes nutricionales variables. Se analizó la condición conservativa en la incorporación de ácidos grasos al huevo, y se discutió su utilidad como posibles definitorios de calidad, teniendo como referente la alta variabilidad registrada en la supervivencia al final del proceso de incubación.

    Winter flounder (Pseudopleuronectes americanus) once supported robust commercial and recreational fisheries in the New York (USA) region, but since the 1990s populations have been in decline. Available data show that settlement of young-of-the-year winter flounder has not declined as sharply as adult abundance, suggesting that juveniles are experiencing higher mortality following settlement. The recent increase of blue crab (Callinectes sapidus) abundance in the New York region raises the possibility that new sources of predation may be contributing to juvenile winter flounder mortality. To investigate this possibility we developed and validated a method to specifically detect winter flounder mitochondrial control region DNA sequences in the gut contents of blue crabs. A survey of 55 crabs collected from Shinnecock Bay (along the south shore of Long Island, New York) in July, August, and September of 2011 showed that 12 of 42 blue crabs (28.6%) from which PCR-amplifiable DNA was recovered had consumed winter flounder in the wild, empirically supporting the trophic link between these species that has been widely speculated to exist. This technique overcomes difficulties with visual identification of the often unrecognizable gut contents of decapod crustaceans, and modifications of this approach offer valuable tools to more broadly address their feeding habits on a wide variety of species.

Bu çalışmada 7000 prelarval gökkuşağı alabalığı yedi tanka eşit olarak bölünmüş ve bunlar sentetik hormonlar (3 ve 6 mg metiltestosteron -MT ve testosteron -T kg-1yem ve doğal hormonlar (50 ve 100 mg Tribulus terrestris -TT kg-1 içeren ticari alabalık yavru yemleri (Skretting, France ile keseli dönemden sonraki 90 gün boyunca beslenmiştir. 90 günlük ilk besleme periyodunun sonunda balıklar cinsiyetlerin makroskopik ve histolojik yöntemlerle belirlenebildiği döneme kadar hormon içermeyen ticari alabalık yemi ile beslenmiştir. Araştırma sonunda 50 ve100 mg/ kg Tribulus terrestris (TT içeren yemle beslenen balıkların sıra ile %55 ve 63’ü erkek iken, 3 ve 6 mg/ kg testosteron (T içeren yemle beslenen balıkların sıra ile %51 ve 53 oranında erkek olduğu bulunmuştur. Bu durum Tribulus terrestris’in testosterona göre alabalıkların doğrudan erkekleştirilmesinde daha etkin olduğunu fakat gruplar arasında fark bulunmamıştır. Ayrıca bu araştırma, Tribulus terrestris içeren (100 mg/kg mg/ kg yemde yemlerle beslenen alabalıkların diğer deneme gruplarındaki balıklara kıyasla daha yüksek canlı ağırlığa ulaştığını bildiren ilk çalışmadır. Kontrol ve deneme grupları arasında farklılaşmamış gonad yapıları bakımından önemli bir farklılık gözlenmemiştir. Sonuç olarak araştırmada, daha yüksek cinsiyet dönüşüm ve büyüme oranlarına ulaşmak için Tribulus terrestris’in daha yüksek konsantrasyonlarının (150, 200, 250 mg kg-1 etkilerini araştırılmalıdır. Ayrıca bu konsantrasyonların gökkuşağı alabalığında, diğer salmonid ve ticari türlerde kan testosteron seviyelerinin yanısıra gonad yapısına olan etkileri de belirlenmelidir

    This paper reports the effect of different concentrations of CO2/N2 gases on chemical/sensorial stability, color properties (L*, a*, and b* values), and microbiological properties of trout mince stored at 3 ± 1℃. The gas mixtures were used as follows; 60%CO2 + 40%N2 (B), 75%CO2 + 25%N2 (C), 40%CO2 + 60%N2 (D). Control samples (A) were packed under atmospheric air. Analyses were carried out on the 1st, 4th, 8th, 12th, 16th, 20th, 24th and 28th days of storage. Among three modified atmosphere gas compositions, the best preservation method and most suitable gas concentration for trout mince were found in group C including 75% CO2 The inconsumable day of trout mince packed with air (control) and MAPs (Modified Atmosphere Packaging) (group B, C, and D) were 12th, 16th, 24th, and 16th days, respectively. © The Author(s) 2015.

    Full Text Available The aim of this study was to describe the biometric characteristics of the European sardine (Sardina pilchardus catches and assess the current status of sardine stock in North Aegean Sea based on population characteristics and abundance trends. The stock was dominated by age groups 1 and 2, not exceeding age group 4. The sardine stock in this area was assessed through an Integrated Catch-at-Age model which implements a separable Virtual Population Analysis on catch at age data with weighted tuning indices. Sardine landings data derived from the commercial purse seine fishery over the period 2000-2008 were combined with the age structure of the stock as resulted from fisheries independent acoustic surveys. Sensitivity analysis of the impact of natural mortality values on stock assessment results was applied. Additionally forecast of the sardine population parameters and catches under different exploitation scenarios was implemented in a medium term basis. Results indicated that the North Aegean Sea sardine stock is considered fully exploited with the fishery operating close but over the empirical exploitation level for sustainability. Finally, the status of the sardine stock in N. Aegean Sea is discussed in relation to the sardine stocks from the western and the central Mediterranean basin.

To analyze the results of morphometric measurements and dates of aquaculture-biological characteristics of the young-of-the-year and age-1+ rainbow trout reared under industrial technology at instable conditions of the fish farm “Sloboda-Banyliv”. Methodology. The study has been performed at the industrial fish farm “Sloboda-Banyliv”, Chernivtsi region. The materials for the study were young-of-the-year and age-1+ rainbow trout obtained from the eggs of autumn-spawning form rainbow trout. The young-of-the-year were reared in a 216 m2 tank with stocking density of 255 ind./m2, age-1+ fish were reared in 108 m2 tank with a stocking density of 33 ind./m2 according to generally accepted methods in trout culture. Morphometric measurements of fish were performed according to I.F. Pravdin. Statistical processing of data was carried out in Microsoft Office Excel (2003. The analysis of values was done in the system of absolute values. The analyzed criteria of the measured parameters were their mean values and standard errors (M±m, deviation (σ, variability coefficient (Cv. Fish were fed with the artificial feed with high protein content manufactured by “Biomar” (Denmark. Findings. The studies on rainbow trout rearing under industrial conditions showed that fish body proportions did not change with age, however, the length of their fins decreased. The slenderness coefficient in age-1+ fish decreased insignificantly that is typical with increasing body depth. Despite instable rearing conditions, both young-of-the-year and age-1+ fish were characterized by moderate growth rate and high feed-conversion efficiency. Originality. For the first time, in conditions of Ukraine, a study on the formation of rainbow trout brood stocks in a fish farm with instable rearing conditions was performed with the use of the analysis of phenotypical and productive features. Practical value. The results of the performed work will provide an opportunity to form a rainbow trout brood stock based on characterized phenotypical and productive features that would have high productive and reproductive features in a fish farm with instable rearing conditions.

  8. Effect of dietary α-tocopherol + ascorbic acid, selenium, and iron on oxidative stress in sub-yearling Chinook salmon (Oncorhynchus tshawytscha Walbaum) (United States)

    A three-variable central composite design coupled with surface-response analysis was used to examine the effects of dietary ??-tocopherol + ascorbic acid (TOCAA), selenium (Se), and iron (Fe) on indices of oxidative stress in juvenile spring Chinook salmon. Each dietary factor was tested at five levels for a total of fifteen dietary combinations (diets). Oxidative damage in liver and kidney (lipid peroxidation, protein carbonyls) and erythrocytes (erythrocyte resistance to peroxidative lysis, ERPL) was determined after feeding experimental diets for 16 (early December) and 28 (early March) weeks. Only TOCAA influenced oxidative stress in this study, with most measures of oxidative damage decreasing (liver lipid peroxidation in December and March; ERPL in December; liver protein carbonyl in March) with increasing levels of TOCAA. We also observed a TOCAA-stimulated increase in susceptibility of erythrocytes to peroxidative lysis in March at the highest levels of TOCAA. The data suggest that under most circumstances a progressive decrease in oxidative stress occurs as dietary TOCAA increases, but higher TOCAA concentrations can stimulate oxidative damage in some situations. Higher levels of TOCAA in the diet were required in March than in December to achieve comparable levels of protection against oxidative damage, which may have been due to physiological changes associated with the parr-smolt transformation. Erythrocytes appeared to be more sensitive to variation in dietary levels of TOCAA than liver and kidney tissues. Using the March ERPL assay results as a baseline, a TOCAA level of approximately 350-600 mg/kg diet would provide adequate protection against lipid peroxidation under most circumstances in juvenile Chinook salmon. ?? 2008 The Authors.

    Rainbow trout (Oncorhynchus mykiss) intestinal strips (n = 10) were mounted in an isolated organ bath and the effect of incremental doses of compound 48/80 was recorded. Compound 48/80 induced concentration-related contraction in all the examined strips following a sigmoidal dose-response curve fit. Values for maximal contraction (E(max) , g cm(-2)), negative logarithm of the EC(50) (pD(2)), and hill slope were, respectively (mean±standard error), 12.88 ± 0.51, 1.88 ± 0.05, 1.49 ± 0.27. The histological modification induced on mast cells (MCs) due to compound 48/80 was characterized by mean of gray-levels and texture analysis. Significant differences were observed between gray-levels values (Linear mixed model, Pcompound 48/80-treated strips compared with MCs from untreated strips. Moreover, maximal intestinal contraction (due to compound 48/80) correlates positively and significantly (Pearson and Spearman correlations, Pcompound 48/80 induces the degranulation of trout intestinal MCs ex vivo, and that the aforementioned degranulation promotes a concentration-dependent intestinal contraction.     Full Text Available The purpose of this work was to investigate the size-relative effectiveness of clove oil as an anaesthetic for rainbow trout and goldfish. In total, 128 rainbow trout (Oncorhynchus mykiss (two groups of 20-23 and 30-33 cm mean fork length and 160 goldfish (Carassius auratus (four size groups of 1.5-2.5, 5-7, 11-15 and 20-25 cm were anaesthetized at different clove oil concentrations of 50, 100, 150 mg·l-1 for trouts and 75, 100, 150 mg·l-1 for goldfish. Rainbow trout exhibited total loss of balance and no response to external stimuli with shorter induction time as dosage increased (120.5 s, 64.4 s and 44.3 s, respectively. Goldfish exhibited total loss of balance and no response to external stimuli after induction time that varied with dosage used and body size of fish. The small fish (1.5-7 cm exhibited shorter induction time which ranged from 84.28 s at 75 mg·l-1 clove oil to 41.14 s at 150 mg·l-1 clove oil. The larger fish had a longer induction time inversely related to the dosage. Recovery time was longer than induction time in both species. Both species recovered within 6 min after anaesthesia at 150 mg·l-1 clove oil. Clove oil did not produce marked changes (P compared to the control. However, marked changes (P P > 0.05. For both fish species, clove oil was effective, producing minimum stress and zero mortalities, and can be recommended as an effective anaesthetic.

    Rainbow trout Oncorhynchus mykiss surviving proliferative kidney disease (PKD) are reported not to develop the disease upon re-exposure. However, the mechanisms involved in the immune response to re-exposure are unknown. We examined disease susceptibility and the immune response of naive 1+ rainbow trout when first exposed to Tetracapsuloides bryosalmonae in comparison with that of 1+ rainbow trout re-exposed to T. bryosalmonae. PKD pathogenesis, parasite burden and transcriptional signatures of the host immune response were assessed at 10, 25 and 40 d.p.e (days post-exposure). In addition, we evaluated the presence of IgM+ B cells in the blood and the posterior kidney. The exposure of 1+ rainbow trout to T. bryosalmonae for the first time resulted in 100% infection prevalence, high parasite burdens and severe clinical PKD, while re-exposed fish were either able to avoid reinfection completely or mount an earlier and more efficient adaptive-type immune response. This response was characterized by a greater amount of IgM+ B cells in the blood and elevated mRNA levels of secretory IgM in the posterior kidney which minimized pathogen burden and kidney inflammation. Our findings suggest that rainbow trout is able to develop immune protection against T. bryosalmonae. © 2017 John Wiley & Sons Ltd.

  14. Negligible risk associated with the movement of processed rainbow trout, Oncorhynchus mykiss (Walbaum), from an infectious haematopoietic necrosis virus (IHNV) endemic area (United States)

    To assess the risk of transmission of infectious haematopoietic necrosis virus (IHNV) associated with the movement of processed rainbow trout, Oncorhynchus mykiss, from an area where the virus is endemic, 240 freshly eviscerated fish (225-500 g) exhibiting spinal curvature or spinal compression types of deformities were tested for IHNV by virus isolation and polymerase chain reaction (PCR) techniques. Commercially produced rainbow trout, approximately 1-year-old, that exhibited spinal deformities were considered to have had a high likelihood of having survived an outbreak of IHN. Serological analysis of fish exhibiting spinal curvature or spinal compression types of deformities for anti-IHNV antibodies resulted, in 71 and 50% of the serum samples, respectively, with detectable neutralization activity suggesting previous infection with IHNV. A portion of the skin and muscle in the area of the deformity was collected, as well as brain tissue from each commercially processed fish. Tissue homogenates were tested for IHNV using the epithelioma papulosum cyprini (EPC) cell line pretreated with polyethylene glycol and the chinook salmon embryo (CHSE-214) cell line using standard methods. Nested, reverse transcriptase (RT)-PCR for the detection of IHNV used the central 1231 bp portion of the glycoprotein (G) challenge studies and is suggested as a mechanism responsible for virus clearance. These results provide scientific information that can be used to assess the risk associated with the movement of processed rainbow trout from an IHNV endemic area.

Materiales y métodos. Se utilizó el Índice peróxidos (VP y el índice anisidina (VA para detectar la presencia de productos de la oxidación en las raciones. Se realizó un análisis de varianzabajo un modelo de parcelas divididas en el tiempo. Cuando se encontraron diferencias (p<0.05 las medias fueron comparadas mediante la prueba de Tukey (5%. Resultados. Las raciones presentaron altos niveles de oxidación durante todo el experimento y hubo diferencias significativas entre tratamientos. La actividad SOD presentó niveles decrecientes a nivel hepático durante los dos períodos de exposición, sin embargo, en TGI se generó un incremento significativo de actividad SOD (175% en individuos sometidos a todoslos tratamientos. La actividad CAT presentó un alto nivel de correlación con la actividad SOD en todos los períodos de exposición y órganos. La actividad GPx presentó diferencias significativas para los dos períodos de exposición en hígado y al día 90 en TGI, indicandoalta sensibilidad de la enzima ante la ingestión de peróxidos. Conclusiones. La actividad GPx en TGI mostró coeficientes de correlación superiores a 0.95, sugiriendo que es buen indicador del estado oxidativo de las raciones.

    To investigate if aluminium (Al) modifies the rainbow trout response to radiation exposure and/or the induction of a radiation-induced bystander effect. Rainbow trout were exposed to 100 or 200 μg l(-1) Al (for 3 h), a 0.5 Gy X-ray dose or Al followed immediately by irradiation. The exposed fish were then swum with completely untreated bystander fish. A human reporter cell clonogenic assay was used to determine whether Al exposure modified the effects of irradiation on the skin and gills from directly exposed fish and also the radiation-induced bystander effect in untreated fish. Al exposure did not modify the response to direct irradiation by the skin, or the gill. Al did not modify the bystander effect in the skin. However Al did modify the bystander effect in the gill. Gills of bystander fish swum with fish exposed to 200 μg l(-1) Al, followed by irradiation, caused a greater reduction in HPV-G cell survival than was caused by irradiation only. Interestingly Al exposure only also caused a bystander effect (reduced HPV-G survival) in the gill. This study shows that, in a multiple stressor scenario, the communication of radiation-induced stress signals is modified on a tissue-specific basis by acute Al exposure. Aside from the implications this has for radiological protection this response may also have potential for environmental monitoring where detection of the bystander effect could act as an indicator of radiation exposure when direct exposure responses are not evident.

    This research demonstrates a significant increase in routine metabolic rate (RMR) following injection of a DNA vaccine concurrently with a polyvalent, oil-adjuvanted vaccine. The increase in RMR was transient and associated with increased activity of both the non-specific and specific immune responses. Rainbow trout (Oncorhynchus mykiss) were injected with a DNA vaccine (DV), a commercially available polyvalent, oil-adjuvanted vaccine (AV), or the two vaccines in combination and sampled at 203, 305, and 406 days (dd) post-vaccine injection (pvi) for RMR and key immune parameters (serum lysozyme activity, serum neutralization antibody titres). The RMR of fish that received both the DV and the AV was significantly higher at 203 dd pvi, compared to fish from all other treatment groups which included the control, the AV, and the DV groups. The increased RMR corresponded to elevated levels of serum lysozyme activity and an earlier seroconversion of virus-specific neutralizing antibodies. To determine if growth performance was affected by the transient increase in RMR, specific growth rate (SGR), percent daily weight gain (WG), and feed conversion ratio (FCR) were determined at 798, 1204, and 1610 dd pvi. Although fish in all three vaccine groups showed significant increases in SGR and WG at 798 and 1610 dd pvi compared to the control group, the overall weight of the fish was not different at the end of the experiment. In summary, this study shows that concurrent injection of a DV and an AV transiently increases the RMR of rainbow trout and changes the manner in which the immune response occurs, but does not affect the overall growth performance of the fish.     In fall 2013, anorexia, lethargy and mortalities up to 10-12,000 dead fish per week were observed in rainbow trout Oncorhynchus mykiss in three fresh water hatcheries (salinity 0-1 ‰) on the west coast of Norway. The fish (25-100 g) showed signs of circulatory failure with haemorrhages, ascites and anaemia. The histopathological findings comprised inflammation of the heart and red muscle and liver necrosis. The affected fish had a common origin. Disease and mortalities were also observed up to four months after sea water transfer. Microbiological examination did not reveal presence of any known pathogens. Based on histopathological similarities to heart and skeletal inflammation (HSMI) in Atlantic salmon, associated with piscine orthoreovirus (PRV), extended investigations to detect a virus within the family Reoviridae were conducted. By the use of primer sets targeting the PRV genome, a sequence with 85% identity to a part of segment S1 of PRV was obtained. Further analysis showed that the virus sequence could only be aligned with PRV and no other reoviruses both on amino acid and nucleotide level. Two PCR assays were developed for specific detection of the virus. High amounts of the virus were detected in diseased fish at all affected farms and low amounts were detected in low prevalence at the broodfish farms. Further investigations are needed to determine if the virus is associated with the new disease in rainbow trout and to further characterize the virus with respect to classification, relationship with PRV, virulence, pathology and epidemiology.

    Full Text Available Bu çalışma, Rize İli Güneysu Beldesindeki özel alabalık çiftliğinde yürütülmüştür. Gökkuşağı (Oncorhynchus mykiss alabalığı ile kaynak (Salvelinus fontinalis alabalığı anaçlarının yumurta büyüklüğü ile vücut büyüklüğü arasındaki ilişki araştırılmış ve yumurta verimine etkisi karşılaştırılmıştır. 3 ve 4 yaşındaki alabalıklar kuru yöntemle Aralık 2002, Ocak-Şubat 2003 tarihlerinde sağılmıştır. Anaç gökkuşağı alabalıkları ortalama 44,3±7,394 cm uzunluğunda, 1050,3±461,027 g ağırlığında olup, yumurta verimleri 3016,8±360,320 yumurta/birey belirlenmiştir. Yumurta çapı da 4,82±0,339 mm olarak bulunmuştur. Kaynak alabalıklarında ise damızlık boyu 33,9±3,900 cm ve ağırlığı 526,8±202,098 g olarak tespit edilmiştir. Yumurta verimleri de 1449,2±373,186 yumurta/birey ve yumurta çapları 4,28±0,261 mm olarak bulunmuştur.     The von Bertalanffy model of body growth is inappropriate for organisms whose growth is restricted to a seasonal period because it assumes that growth rate is invariant with time. Incorporation of a time-varying coefficient significantly improves the capability of the von Bertalanffy equation to describe changing body size of both the bivalve mollusc Macoma balthicain San Francisco Bay and the flathead sole, Hippoglossoides elassodon, in Washington state. This simple modification of the von Bertalanffy model should offer improved predictions of body growth for a variety of other aquatic animals.

    Yersinia ruckeri is the causative agent of enteric redmouth disease leading to significant losses in salmonid aquaculture worldwide. Little information is available on the pathogenesis of this disease. Basic steps in the establishment of an infection include attachment to the epithelium followed by invasion at the portal of entry. In this study, the interactions of Y. ruckeri with the gills and the gut of rainbow trout Oncorhynchus mykiss (Walbaum, 1792) were studied using standardized perfusion models. Virulent and avirulent Y ruckeri isolates appeared to adhere to and invade both tissues without significant differences. For the first time, the gill and gut perfusion models are shown to be suitable to study bacterial invasiveness.

    The purpose of this study is to assess the fishery improvement opportunities on the Box Canyon portion of the Pend Oreille River. This report contains the findings of the first year of the study. Chinook salmon (Oncorhynchus tshawytscha (Walbaum)) and steelhead (Oncorhynchus mykiss (Richardson)) were present in the Pend Oreille River prior to the construction of Grand Coulee Dam. The river also contained native cutthroat trout (Oncorhynchus clarki (Richardson)), bull trout (Salvelinus confluentus (Walbaum)) and mountain whitefish (Prosopium williamsoni (Girard)). Rainbow trout were planted in the river and some grew to lengths in excess of 30 inches. With the construction of Box Canyon Dam, in 1955, the most productive section of the river was inundated. Following the construction of the dam the trout fishery declined and the populations of spiny ray fish and rough fish increased. The objectives of the first year of the study were to determine the relative abundance of each species in the river and sloughs; the population levels in fish in the river and four selected tributaries; fish growth rates; the feeding habits and abundance of preferred prey; the migration patterns; and the total fishing pressure, catch per unit effort, and total harvest by conducting a year-round creel survey. 132 refs.

    This research was initiated in conjunction with a systematic, multiagency surveillance effort in the United States (U.S.) in response to reported findings of infectious salmon anaemia virus (ISAV) RNA in British Columbia, Canada. In the systematic surveillance study reported in a companion paper, tissues from various salmonids taken from Washington and Alaska were surveyed for ISAV RNA using the U.S.-approved diagnostic method, and samples were released for use in this present study only after testing negative. Here, we tested a subset of these samples for ISAV RNA with three additional published molecular assays, as well as for RNA from salmonid alphavirus (SAV), piscine myocarditis virus (PMCV) and piscine orthoreovirus (PRV). All samples (n = 2,252; 121 stock cohorts) tested negative for RNA from ISAV, PMCV, and SAV. In contrast, there were 25 stock cohorts from Washington and Alaska that had one or more individuals test positive for PRV RNA; prevalence within stocks varied and ranged from 2% to 73%. The overall prevalence of PRV RNA-positive individuals across the study was 3.4% (77 of 2,252 fish tested). Findings of PRV RNA were most common in coho (Oncorhynchus kisutch Walbaum) and Chinook (O. tshawytscha Walbaum) salmon.

    The objectives of this study were to examine levels of arsenic, cadmium, lead, mercury and selenium in edible tissue of seven species of marine fish collected from several Aleutian islands (in 2004) to determine: (1) interspecific differences, (2) locational differences (among Aleutian Islands), (3) size-related differences in any metal levels within a species, and (4) potential risk to the fish or to predators on the fish, including humans. We also compared metals levels to those of three other fish species previously examined in detail, as well as examining metals in the edible tissue of octopus (Octopus dofleini). Octopus did not have the highest levels of any metal. There were significant interspecific differences in all metal levels among the fish species, although the differences were less than an order of magnitude, except for arsenic (mean of 19,500 ppb in Flathead sole, Hippoglossoides elassodon). Significant intraisland variation occurred among the four sites on Amchitka, but there was not a consistent pattern. There were significant interisland differences for some metals and species. Mercury levels increased significantly with size for several species; lead increased significantly for only one fish species; and cadmium and selenium decreased significantly with size for halibut (Hippoglossus stenolepis). The Alaskan Department of Health and Social Services supports unrestricted consumption of most Alaskan fish species for all people, including pregnant women. Most mean metal concentrations were well below the levels known to adversely affect the fish themselves, or predators that consume them (including humans), except for mercury in three fish species (mean levels just below 0.3 ppm), and arsenic in two fish species.   6. Suppression of invasive lake trout in an isolated backcountry lake in Glacier National Park (United States)

    Fisheries managers have implemented suppression programmes to control non-native lake trout, Salvelinus namaycush (Walbaum), in several lakes throughout the western United States. This study determined the feasibility of experimentally suppressing lake trout using gillnets in an isolated backcountry lake in Glacier National Park, Montana, USA, for the conservation of threatened bull trout, Salvelinus confluentus (Suckley). The demographics of the lake trout population during suppression (2009–2013) were described, and those data were used to assess the effects of suppression scenarios on population growth rate (λ) using an age-structured population model. Model simulations indicated that the population was growing exponentially (λ = 1.23, 95% CI: 1.16–1.28) prior to suppression. However, suppression resulted in declining λ(0.61–0.79) for lake trout, which was concomitant with stable bull trout adult abundances. Continued suppression at or above observed exploitation levels is needed to ensure continued population declines.

As water velocity increased from 0 to 30 cm/sec area and distance traveled by juvenile bass 10 to 80 mm long decreased. However, as water velocity increased from 0 to 3 cm/sec the area and distance covered by larval bass increased. The presence of food increased the activity of larval bass, but decreased the activity of juveniles. Area ranged by striped bass at test velocities ranging from 0 to 30 cm/sec increased in proportion to body length. Juvenile striped bass tested at acclimation temperatures between 20 and 5/sup 0/C experienced a 30% reduction of activity. Activity was also reduced as temperature increased from 20 to 30/sup 0/C.

  8. Northeastern Chukchi Sea demersal fishes and associated environmental characteristics, 2009-2010 (United States)

    Three closely-spaced study areas in the northeastern Chukchi Sea off of Alaska provided a opportunity to examine demersal fish communities over a small spatial scale as part of a multidisciplinary program. During 2009 and 2010, fishes in the three study areas (Klondike, Burger, and Statoil) were sampled at 37 stations with a plumb staff beam trawl and a 3 m beam trawl; 70% of stations were sampled during all three cruises. Fish catches were dominated by small fishes (modeling of the data suggested that overall fish density, species richness, and density of Arctic staghorn sculpin (Gymnocanthus tricuspis) and Bering flounder (Hippoglossoides robustus) were higher in the more southerly Klondike study area than in the more northerly Burger and Statoil study areas. Arctic cod (Boreogadus saida) was abundant throughout the study region. Richness and density could be explained by the environmental variables that defined the overall study area. The Klondike study area was warmer and erosional in nature with higher proportions of gravel sediment. Other study areas were colder and more depositional in nature with muddier sediment and were characterized by high densities of megafaunal invertebrates such as brittle stars. There appeared to be a lack of ecological homogeneity across these three closely-spaced study areas of the Chukchi Sea.

  9. Isolation and characterization of 45 Polymorphie microsatellite loci of turbot ( Scophthalmus maximus) and cross-species amplification (United States)

    Turbot ( Scophthalmus maximus) is a flatfish species commercially important for aquaculture. In this study, we generated a microsatellite-enriched genomic DNA library for Scophthalmus maximus, and then isolated and characterized 45 microsatellite loci by genotyping 30 individuals. The observed number of alleles ranged from 2 to 19 with an average of 6.24, while the effective number of alleles ranged from 1.30 to 11.11 with an average of 3.66. The expected heterozygosities varied from 0.235 to 0.925 4 and Polymorphie information content ranged from 0.2044 to 0.903 3, with an average of 0.622. Twelve loci deviated significantly from Hardy-Weinberg equilibrium, and no significant linkage disequilibrium was observed between any pair of loci after Bonferroni correction. In cross-species amplification, five flatfish species ( Paralichthys lethostigma, Verasper moseri, platichthys stellatus, Hippoglossoides dubius and Cynoglossus semilaevis) showed at least one Polymorphie locus. These Polymorphie microsatellite loci should prove useful for Population analysis of turbot and other related species.

  10. Bilan des introductions de salmonidés dans les lacs et ruisseaux d'altitude des Hautes-Pyrénées

    Full Text Available Les introductions de Salmonidés ont été importantes au cours des 60 dernières années dans les lacs et ruisseaux d'altitude des Hautes-Pyrénées. Six espèces de Salmonidés ont été introduites dans des milieux qui, pour la plupart, étaient vierges de populations piscicoles : la truite commune (Salmo trutta L., la truite arc-en-ciel (Oncorhynchus mykiss Walbaum, l'omble de fontaine (Salvelinus fontinalis Mitchill, l'omble chevalier (Salvelinus alpinus L., le cristivomer (Salvelinus namaycush Walbaum et le splake (Salvelinus fontinalis x Salvelinus namaycush. Dans de très nombreux cas, ces introductions ont abouti à des acclimatations. En revanche, les naturalisations sont beaucoup plus rares. Seules les espèces lacustres (cristivomer et omble chevalier se sont naturalisées dans la majorité des lacs où elles ont été introduites. Les conditions de reproduction constituent le facteur clé permettant d'expliquer la naturalisation des espèces. En ruisseau, il faut y ajouter la compétition avec l'espèce indigène (la truite commune, la pression halieutique ainsi que les conditions hivernales très rigoureuses. Les incidences écologiques des introductions sur les populations de truites communes indigènes sont faibles. En revanche, elles ne sont pas négligeables pour les populations de batraciens. Cette politique d'introduction a largement participé au développement de l'halieutisme dans ces milieux d'altitude. En cela, les introductions ont parfaitement répondu aux objectifs halieutiques qu'on leur avait fixés. L'acquisition de connaissances sur l'ensemble de la chaîne pyrénéenne constitue aujourd'hui une étape incontournable pour une politique de gestion globale des introductions.

  11. Surveys of larval sealworm (Pseudoterranova decipiens infection in various fish species sampled from Nova Scotian waters between 1988 and 1996, with an assessment of examination procedures

    Full Text Available Between November 1988 and October 1996, >10,000 fish from the Breton Shelf, Sable Island Bank and the northeastern Gulf of Maine were examined for larval anisakines. Larval sealworm, Pseudoterranova decipiens, occurred in 30 of 39 species surveyed, including 8 new host records, Enchelyopus cimbrius, Lycodes reticulatus, Eumesogrammus praecisus, Lumpenus lumpretaeformis, Lumpenus maculatus, Cryptacanthodes maculatus, Artediellus atlanticus and Triglops murrayi. The parasite was most prevalent and abundant in mature demersal piscivores and benthic consumers. Sealworm densities (nr kg-1 host wt., however, were greatest in small benthophagous fish including mature E. cimbrius, A. atlanticus, T. murrayi and Aspidophoroides monopterygius, and juvenile Hippoglossoides platessoides. ANOVA revealed that geographical disparities in sealworm prevalence and abundance were highly significant in 14 of 20 species tested, although significant disparities between samples from each of the three areas were evident only in H. platessoides. Almost invariably, infection parameters were greatest in fish from Sable Island Bank. ANOVA also indicated that sealworm prevalence and/or abundance increased significantly in Sable Island Bank populations of Gadus morhua, H. platessoides, and seven other species between 1985-1986 and 1989-1990. Routine examinations, in which host flesh was sliced and candled, proved as efficacious as digestion in warm (35° C pepsin-HCl for detection of larval sealworm in the flesh of large frozen fish. Procedures employing fresh (iced samples, digestion at ambient temperature and microscopy are recommended, however, for surveys of small benthic consumers. Many of the sealworm infecting the latter hosts are tiny (2 to 10 mm in length nematodes, which escape detection by routine inspection, and may not survive in warm pepsin-HCl solution.

    Full Text Available This work studies final nutritional status and transcriptional responses of rainbow trout (Oncorhynchus mykiss Walbaum 1792 (28 g after a 10 week feeding experiment designed to elucidate the effect of adding a vitamin and mineral premix on growth, health, and nutritional endpoints. Juvenile fish were fed a either a diet supplemented with a vitamin and mineral premix (Diet S or the same diet without premix supplementation (Diet U. The analyzed micronutrient composition of diets differed accordingly. Pooled livers from 15 fish from each dietary group were used to create suppression subtractive hybridization (SSH cDNA libraries that were sequenced with 454 FLX GS Titanium Technology. In total 552 812 reads were sequenced from the two cDNA libraries. Ingenuity pathway analysis (IPA was then used to characterize the hepatic transcriptome of the two dietary groups of rainbow trout. In the present communication we discuss how selected micronutrients may affect the transcriptome at suboptimal status by directly impacting the cellular metabolism, functions, and structures, and by introducing respective compensatory mechanisms. Processes related to lipid metabolism, peptide hydrolysis, oxygen transportation, and growth development were mostly affected. Considering the transcriptomics data relative to changes in nutritional status from the feeding study and the background phenotypic outcome of growth performance and gill histopathology, the outcome of the transcriptional profiling are suggested to be mainly related to suboptimal pantothenic acid and vitamin C nutrition.

  13. Reciprocal Subsidies and Food Web Pathways Leading to Chum Salmon Fry in a Temperate Marine-Terrestrial Ecotone (United States)

    Stable isotope analysis was used to determine the relative proportions of terrestrial and marine subsidies of carbon to invertebrates along a tidal gradient (low-intertidal, mid-intertidal, high-intertidal, supralittoral) and to determine the relative importance of terrestrial carbon in food web pathways leading to chum salmon fry Oncorhynchus keta (Walbaum) in Howe Sound, British Columbia. We found a clear gradient in the proportion of terrestrially derived carbon along the tidal gradient ranging from 68% across all invertebrate taxa in the supralittoral to 25% in the high-intertidal, 20% in the mid-intertidal, and 12% in the low-intertidal. Stable isotope values of chum salmon fry indicated carbon contributions from both terrestrial and marine sources, with terrestrially derived carbon ranging from 12.8 to 61.5% in the muscle tissue of chum salmon fry (mean 30%). Our results provide evidence for reciprocal subsidies of marine and terrestrially derived carbon on beaches in the estuary and suggest that the vegetated supralittoral is an important trophic link in supplying terrestrial carbon to nearshore food webs.   14. Comparative molecular analysis of evolutionarily distant glyceraldehyde-3-phosphate dehydrogenase from Sardina pilchardus and Octopus vulgaris. (United States)

    The NAD(+)-dependent cytosolic glyceraldehyde-3-phosphate dehydrogenase (GAPDH, EC, which is recognized as a key to central carbon metabolism in glycolysis and gluconeogenesis and as an important allozymic polymorphic biomarker, was purified from muscles of two marine species: the skeletal muscle of Sardina pilchardus Walbaum (Teleost, Clupeida) and the incompressible arm muscle of Octopus vulgaris (Mollusca, Cephalopoda). Comparative biochemical studies have revealed that they differ in their subunit molecular masses and in pI values. Partial cDNA sequences corresponding to an internal region of the GapC genes from Sardina and Octopus were obtained by polymerase chain reaction using degenerate primers designed from highly conserved protein motifs. Alignments of the deduced amino acid sequences were used to establish the 3D structures of the active site of two enzymes as well as the phylogenetic relationships of the sardine and octopus enzymes. These two enzymes are the first two GAPDHs characterized so far from teleost fish and cephalopod, respectively. Interestingly, phylogenetic analyses indicated that the sardina GAPDH is in a cluster with the archetypical enzymes from other vertebrates, while the octopus GAPDH comes together with other molluscan sequences in a distant basal assembly closer to bacterial and fungal orthologs, thus suggesting their different evolutionary scenarios.

    Full Text Available Introduction: The goal of this study was to assess the influence of vaccination against enteric redmouth disease on oxidative stress biomarkers and antioxidant defence in the muscle tissue of rainbow trout (Oncorhynchus mykiss Walbaum vaccinated against Yersinia ruckeri in the first and second month after immunisation. Material and Methods: Healthy fish were vaccinated orally with inactivated whole cells of a virulent strain of Y. ruckeri. One and two months after immunisation the muscle samples were collected. Results: No significant difference was noted in lipid peroxidation level in either the first or second month after vaccination, while aldehydic and ketonic derivatives of oxidatively modified proteins (OMB in the vaccinated group were significantly lower in the second month compared to those in the first month after vaccination (P < 0.05. The content of ketonic derivatives of OMB in muscles in the first month after immunisation was higher compared to untreated control. All these culminated in a depletion of glutathione peroxidase (GPx activity and low level of total antioxidant capacity (TAC. Conclusion: Correlations between catalase activity and lipid peroxidation and TAC confirmed the pivotal role of catalase in antioxidant defence during immunisation. From a broader perspective, it is suggested that immunisation of fish with Yersinia vaccine is associated with induced free radical formation and oxidative stress. Free radicals would therefore be at least partially responsible for the induction of both humoral and cellular elements of the immunity and increased protective immunity against Y. ruckeri infection.

  16. In vitro antimicrobial activity of ethanolic extracts obtained from Ficus spp. leaves against the fish pathogen Aeromonas hydrophila

    Full Text Available The main goal of this study was to determine in vitro antimicrobial activity of ethanolic extracts obtained from the leaves of various Ficus species against Aeromonas hydrophila isolated locally from infected rainbow trout (Oncorhynchus mykiss Walbaum with the aim of providing scientific rationale for the use of the plant in the treatment of bacterial infections induced by Aeromonas spp. in fish. Antimicrobial susceptibility testing was done on Muller-Hinton agar with the disc diffusion method. In the present study, most ethanolic extracts proved effective against the A. hydrophila tested, with 10-12 mm inhibition zones observed. A. hydrophila demonstrated the highest susceptibility to F. pumila. Among various species of Ficus with moderate activity against A. hydrophila, the highest antibacterial activities were noted for F. benghalensis, F. benjamina, F. deltoidea, F. hispida, and F. lyrata. Thus, Ficus can be used as a natural antiseptic and antimicrobial agent in veterinary practice. Further investigations need to be conducted to isolate and identify the bioactive compounds that can then be subjected to detailed pharmacological studies and the development of clinical applications. The alarming rate of increasing resistance in bacterial pathogens in aquaculture environments means that medicinal plants with antibacterial properties are very important as natural resources of new active compounds.

    Full Text Available Streptococcus iniae is among the major pathogens of a large number of fish species cultured in fresh and marine recirculating and net pen production systems. Ten Iranian medicinal plants were assessed for their antimicrobial activity against Streptococcus iniae isolates obtained from diseased Oncorhynchus mykiss (Salmonidae; Walbaum, 1972 collected from fish farms in Iran. The antibacterial activity of ethanol extracts of Punica granatum, Quercus branti, Glycyrrhiza glabra and essential oils of Heracleum lasiopetalum, Satureja bachtiarica, Thymus daenensis, Myrtus communis, Echinophora platyloba, Kelussia odoratissima and Stachys lavandulifolia against Steptococcus iniae was evaluated by disc diffusion and serial dilution assays. Most of the extracts and essential oils showed a relatively high antibacterial activity against Streptococcus iniae. Of the plants studied, the most active extracts were those obtained from the essential oils of Satureja bachtiarica, Echinophora platyloba, Thymus daenensis and the ethanol extract of Quercus branti. Some of the extracts were active against Streptococcus iniae. Two essential oils showed lower MIC values; Heracleum lasiopetalum (78 μg/ml and Satureja bachtiarica (39 μg/ml. The essential oil of Satureja bachtiarica could be an important source of antibacterial compounds against the Streptococcus iniae isolated from rainbow trout.

    Full Text Available El objetivo del estudio fue demostrar el origen infeccioso de la mortandad de alevinos y de las lesiones presentadas en adultos de Oncorhynchus mykiss Walbaum 1792, "trucha arco iris", en la piscigranja "El Ingenio», empleando una metodología simple y efectiva. Se colectaron alevinos, ovas, adultos y muestras de agua de las pozas de alevinos. Las muestras se cultivaron sobre semillas de Cucurbita maxima ‘zapallo’, como sustrato, evidenciándose colonias típicas a los siete días. Las características microscópicas de las hilas correspondieron al patrón gráfico de Saprolegnia sp., lo que concuerda con la sintomatología observada en los adultos capturados. La presencia de este patógeno estaría relacionada con la elevada mortandad registrada en los alevinos (40%, probablemente por la importación de las ovas infectadas con el hongo. El método fue electivo, porque el sustrato empleado, favoreció el crecimiento del hongo, y es de fácil aplicación y bajo costo.

  19. Composition and location of simulated lake-shore redds influence incubation success in kokanee, Oncorhynchus nerka (United States)

    Methods for improving spawning habitat for lakeshore spawning kokanee, Oncorhynchus nerka (Walbaum), were explored by quantifying incubation success of embryos exposed to three substrate treatments in Lake Pend Oreille, Idaho, USA. Substrate treatments included no modification that used existing gravels in the lake (EXISTING), a cleaned substrate treatment where existing gravels were sifted in the water column to remove silt (CLEANED) and the addition of new, silt-free gravel (ADDED). Incubation success was evaluated using Whitlock-Vibert incubation boxes buried within each substrate treatment that contained recently fertilised embryos. Upon retrieval, live and dead sac fry and eyed eggs were enumerated to determine incubation success (sac fry and eyed eggs ?? 100/number of fertilised embryos). Incubation success varied significantly among locations and redd treatments. In general, incubation success among ADDED redds (0.0-13.0%) was significantly lower than that for EXISTING (1.4-61.0%) and CLEANED (0.4-62.5%) redds. Adding new gravel to spawning areas changed the morphometry of the gravel-water interface and probably exposed embryos to disturbance from wave action and reduced embryo survival. Moreover, efforts to improve spawning habitat for lakeshore spawning kokanee should consider water depth and location (e.g. protected shorelines) as important variables. Adding clean gravel to existing spawning areas may provide little benefit if water depth or lake-bottom morphometry are altered. ?? 2009 Blackwell Publishing Ltd.

  20. Testing the thermal-niche oxygen-squeeze hypothesis for estuarine striped bass (United States)

    In many stratified coastal ecosystems, conceptual and bioenergetics models predict seasonal reduction in quality and quantity of fish habitat due to high temperatures and hypoxia. We tested these predictions using acoustic telemetry of 2 to 4 kg striped bass (Morone saxatilis Walbaum) and high-resolution spatial water quality sampling in the Patuxent River, a sub-estuary of the Chesapeake Bay, during 2008 and 2009. Striped bass avoided hypoxic (dissolved oxygen ≤2 mg·l−1) subpycnocline waters, but frequently occupied habitats with high temperatures (>25 °C) in the summer months, as cooler habitats were typically not available. Using traditional concepts of the seasonal thermal-niche oxygen-squeeze, most of the Patuxent estuary would beconsidered unsuitable habitat for adult striped bass during summer. Application of a bioenergetics model revealed that habitats selected by striped bass during summer would support positive growth rates assuming fish could feed at one-half ofmaximum consumption. Occupancy of the estuary during summer by striped bass in this study was likely facilitated by sufficient prey and innate tolerance of high temperatures by sub-adult fish of the size range that we tagged. Our results help extend the thermalniche oxygen-squeeze hypothesis to native populations of striped bass in semi-enclosed coastal systems. Tolerance of for supraoptimal temperatures in our study supports recent suggestions by others that the thermal-niche concept for striped bass should be revised to include warmer temperatures.

  1. New observations on philometrid nematodes (Philometridae) in marine fishes from the Northern Gulf of Mexico and the Indian River Lagoon of Florida (Usa), with first description of the male of Caranginema americanum. (United States)

    The following 3 species of Philometra Costa, 1845 (Nematoda: Philometridae) were recorded from marine fishes off Florida: Caranginema americanum Moravec, Montoya-Mendoza and Salgado-Maldonado, 2008 from the subcutaneous tissue of the crevalle jack Caranx hippos (Linnaeus) (Carangidae); Philometra charlestonensis Moravec, de Buron, Baker and González-Solís, 2008 from the gonads (ovaries) of the scamp Mycteroperca phenax Jordan and Swain (Serranidae); and Philometra sp. (only subgravid females) from the gonads (ovaries) of the Atlantic needlefish Strongylura marina (Walbaum) (Belonidae). The male of C. americanum , the type species of Caranginema Moravec, Montoya-Mendoza, and Salgado-Maldonado, 2008 , is described for the first time. Its general morphology is similar to that of males of Philometra and Philometroides species. The males of C. americanum are mainly characterized by an elongate body, 3.13-3.28 mm long, a markedly elongate esophagus, and spicules and a gubernaculum 69-75 µm and 48-51 µm long, respectively. The present findings of C. americanum and P. charlestonensis represent new geographical records. The gonad-infecting Philometra sp. from S. marina probably belongs to an undescribed species.

  2. Bench-top validation testing of selected immunological and molecular Renibacterium salmoninarum diagnostic assays by comparison with quantitative bacteriological culture (United States)

    No gold standard assay exhibiting error-free classification of results has been identified for detection of Renibacterium salmoninarum, the causative agent of salmonid bacterial kidney disease. Validation of diagnostic assays for R. salmoninarum has been hindered by its unique characteristics and biology, and difficulties in locating suitable populations of reference test animals. Infection status of fish in test populations is often unknown, and it is commonly assumed that the assay yielding the most positive results has the highest diagnostic accuracy, without consideration of misclassification of results. In this research, quantification of R. salmoninarum in samples by bacteriological culture provided a standardized measure of viable bacteria to evaluate analytical performance characteristics (sensitivity, specificity and repeatability) of non-culture assays in three matrices (phosphate-buffered saline, ovarian fluid and kidney tissue). Non-culture assays included polyclonal enzyme-linked immunosorbent assay (ELISA), direct smear fluorescent antibody technique (FAT), membrane-filtration FAT, nested polymerase chain reaction (nested PCR) and three real-time quantitative PCR assays. Injection challenge of specific pathogen-free Chinook salmon, Oncorhynchus tshawytscha (Walbaum), with R. salmoninarum was used to estimate diagnostic sensitivity and specificity. Results did not identify a single assay demonstrating the highest analytical and diagnostic performance characteristics, but revealed strengths and weaknesses of each test.

  3. Skoulekia erythrini n. sp. (Digenea: Aporocotylidae): a parasite of Pagellus erythrinus (L.) (Perciformes: Sparidae) from the western Mediterranean with an amendment of the generic diagnosis. (United States)

    A new aporocotylid, Skoulekia erythrini n. sp., is described from the heart, cephalic kidney and gill blood vessels of the common pandora Pagellus erythrinus (L.) collected from several localities of the western Mediterranean off Spain. The new species differs from the type- and only species of the genus, S. meningialis Alama-Bermejo, Montero, Raga & Holzer, 2011 in possessing a symmetrical body (vs laterally curved), short tegumental spines (3-4 vs 7-10 µm) without hooked ends, diffuse (vs conspicuous) oesophageal gland-cells, a relatively longer oesophagus, a testis that is shorter in relation to body length and a much smaller seminal vesicle (17-34 × 10-26 vs 33-101 × 27-97 μm). Elongated ellipsoidal eggs of S. erythrini n. sp. were found trapped in gill vessels in histological sections. Phylogenetic analyses based on partial 28S rDNA and ITS2 sequences supported the placement of the new species within Skoulekia and the close relationships of this genus with Psettarium Goto & Ozaki, 1929 and Pearsonellum Overstreet & Køie, 1986. Skoulekia meningialis is described from a new sparid host, Diplodus puntazzo (Walbaum), collected off Santa Pola, Spain. The new morphological data for the two Skoulekia spp. and a re-examination of three paratypes of S. meningialis pinpointed features amending both the description of S. meningialis and the generic diagnosis of Skoulekia.

    Rainbow trout (Oncorhynchus mykiss Walbaum) and sea trout (Salmo trutta Linnaeus, 1758) show large karyotypic differences and their hybrid offspring is not viable due to unstable karyotype and chromosome fragmentation. However, gametes from these two species were used to induce gynogenetic development. Rainbow trout eggs activated by UV-irradiated sea trout sperm were subjected to high hydrostatic pressure (HHP) shock to prevent release of the 2nd polar body (early shock) or to inhibit the first cleavage (late shock) in order to produce diploid meiotic gynogenotes and gynogenetic doubled haploids (DHs), respectively. Cytogenetic analysis proved fish that development was induced by the sea trout spermatozoa were rainbow trout. In turn, molecular examination confirmed homozygosity of the gynogenetic DHs. Presumed appearance of the recessive alleles resulted in lower survival of the gynogenetic DH larvae (~25%) when compared to survival of the heterozygous (meiotic) gynogenotes (c. 50%). Our results proved that genomic incompatibilities between studied trout species result in the hybrid unviability. However, artificial gynogenesis including activation of rainbow trout eggs with UV-irradiated sea trout spermatozoa was successfully induced. As both species are unable to cross, application of the UV-irradiated sea trout spermatozoa to activate rainbow trout development assures only maternal inheritance with no contamination by the residues of the paternal chromosomes.

    Full Text Available Mean stream numerical density of the brown trout (Salmo trutta m. fario Linnaeus, 1758 and the rainbow trout (Onchorhynchus mykiss Walbaum, 1792 was 0.090 fish/m2 of which brown trout averaged 69% (72% in total biomass in 15 high-elevation New Mexico streams (1,661-2,560 m above sea level. Total trout density varied from 0.008/m2 in 1988 and 1989. Mean trout density ranged between 0.023-0.121 fish/m2 at site s open to public fishing. Considerably higher densities (0.142-0.409 fish/m2 were observed at sites closed for fishing. In the seven selected streams shared by both species, brown trout density exceeded rainbow trout density except at the two sites closed to fishing. Brown trout were stocked only as fingerlings (average 7,000 fish/stream/year while rainbow trout were stocked only in harvestable sizes (11,000 fish/stream/year. Reported total trout yield rates exceeded the total number of fish estimated to be in the stream by 1.01 to 11.63 in most small streams open to fishing. The proportional stock density (PSD ranged between O and 50 percent. Streams with low to moderate intensities of fishing had the highest PSD.

    Intense infections of a variant of Myxobolus procerus (Kudo, 1934) are described from trout-perch (Percopsis omiscomaycus (Walbaum)) collected in Duluth Harbor, Lake Superior, USA. This particular population of parasites has spores that are identical in shape (narrow pyriform) to those described for M. procerus except that they are significantly smaller (13-14.5 microm long versus 15-17 microm long). In contrast to what was originally described for M. procerus, the plasmodia develop primarily within red and white striated muscle fibres and only rarely among the subdermal connective tissue. Most plasmodia were at or near the same stage of development. Typical development involves growth within the fibre. The parasite eventually replaces the entire content of the host cell and appears to halt development before rupturing the outer cell membrane. The only obvious host response was an occasional cyst being invaded by a localized cellular infiltrate. Infected fish appeared of normal health and no grossly evident myoliquefaction was seen. The infections involved several hundred plasmodia per fish and the question of why such unusually high levels of infection would develop in hosts inhabiting a polluted habitat is raised. It is suggested that proliferation of a pollution tolerant oligochaete (the suspected alternate host) in the harbour and/or a compromised host immune system may have increased the probability of successful transmission and development in trout-perch living in the harbour.

    Full Text Available The aim of study was to determine the effects of exposure to the product DEZYNFEKTANT-CIP (Eng. - Disinfectant-CIP, which is formulated with peracetic acid and hydrogen peroxide, on oxidative stress biomarkers (lipid peroxidation (LPO levels and the carbonyl content of oxidatively modified proteins and antioxidant defenses (superoxide dismutase (SOD, catalase (CAT, glutathione reductase (GR, glutathione peroxidase (GPx, total antioxidant capacity in muscle, gill, hepatic, and cardiac tissues of rainbow trout, Oncorhynchus mykiss (Walbaum. LPO and carbonyl contents changed with tissue type. Exposure to Disinfectant-CIP led to a significant decrease in LPO in muscle tissues and carbonyl content in muscle and gill tissues. The inhibition of SOD and CAT activity in muscle, hepatic, and cardiac tissues was observed probably because of increased oxidative stress during disinfection; however, hepatic and cardiac GPx activity increased in an attempt to counteract oxidative stress. We suggest that oxidative stress during the oxidation of peracetic acid and hydrogen peroxide could be counteracted by the antioxidant system in trout tissues.   8. Persistent effects of wildfire and debris flows on the invertebrate prey base of rainbow trout in Idaho streams (United States)

    Wildfire and debris flows are important physical and ecological drivers in headwater streams of western North America. Past research has primarily examined short-term effects of these disturbances; less is known about longer-term impacts. We investigated wildfire effects on the invertebrate prey base for drift-feeding rainbow trout (Oncorhynchus mykiss, Walbaum) in Idaho headwater streams a decade after wildfire. Three stream types with different disturbance histories were examined: 1) unburned, 2) burned, and 3) burned followed by debris flows that reset channel morphology and riparian vegetation. The quantity of macroinvertebrate drift (biomass density) was more variable within than among disturbance categories. Average body weight and taxonomic richness of drift were significantly related to water temperature and influenced by disturbance history. During the autumn sampling period, the amount of terrestrial insects in rainbow trout diets varied with disturbance history and the amount of overhead canopy along the stream banks. Results indicate that there are detectable changes to macroinvertebrate drift and trout diet a decade after wildfire, and that these responses are better correlated with specific characteristics of the stream (water temperature, canopy cover) than with broad disturbance classes.

  9. Characteristics of Disease Spectrum in relation to Species, Serogroups, and Adhesion Ability of Motile Aeromonads in Fish

    Full Text Available An attempt was made to delineate the relationship between of Aeromonas species and/or serogroups and specific disease symptoms in common carp Cyprinus carpio L. and rainbow trout Oncorhynchus mykiss Walbaum. The adhesion of Aeromonas strains to various tissues in relation to disease spectrum was also tested. All strains of A. hydrophila caused skin ulcers as well as septicaemia in both carp and trout while the other strains were able to cause only skin ulcers or some specific internal lesions with or without septicaemia depending on which species and/or serogroup they represented. Disease symptoms depended also on fish species. It was found that adhesion intensity of Aeromonas strains tested was significantly higher to tissues, which were susceptible to infection with these strains. The results indicate that adhesion to various cells of the fish organism is principal marker to detect virulent Aeromonas strains. The findings presented in this study may be helpful in the appraisal of aeromonads disease risk and kind of the infection in particular fish farms by epizootiological studies or/and during routine fish examinations. They will also be useful to improve and facilitate diagnosis of bacterial fish disease.

  10. Trophic niche overlap between flatfishes in a nursery area on the Portuguese coast

    Full Text Available The diets and the trophic niche overlap between seven flatfish species were studied in a coastal nursery adjoining to the Tagus estuary (Portugal. Fish were sampled monthly, from March to November 1999, using a beach seine. Arnoglossus imperialis (Rafinesque, 1810, Arnoglossus laterna (Walbaum, 1792 and Arnoglossus thori Kyle, 1913, fed mainly on crustaceans. The diets of Buglossidium luteum (Risso, 1810 and Dicologoglossa cuneata (Moreau, 1881 were mainly composed of Bivalvia and Polychaeta, while for Scophthalmus rhombus (Linnaeus, 1758 the main food items were Mysidacea and Teleostei. The diet of Pegusa lascaris (Risso, 1810 was mainly composed by Cumacea, Bivalvia, Decapoda and Amphipoda. Based on diet similarities two main groups were identified: one composed of A. imperialis, A. laterna, A. thori and S. rhombus, and the other grouping B. luteum, P. lascaris and D. cuneata. For the most common flatfishes, a similar pattern of diet seasonal variation was found, such that Amphipoda presented higher indices values in the period from March to June, while from July to November, Decapoda were more abundant. Although high values of diet overlap were obtained among some of the species, the main items in the diet of flatfishes are probably the most abundant prey in this coastal area, which suggests a generalist and opportunistic utilization of these food resources. Furthermore, niche overlap between these species is probably minimized by differences in resource use in other niche dimensions, namely time and space.

Bill anomalies are generally thought to be genetic in origin and genetic changes in bill shape can occur rapidly in a population. The scythebill anomally demonstrates how quickly a drastic bill modification may occur. The crown color pattern anomaly is similar to distinctive markings found in other members of this genus [Queen Angelfish, H. ciliaris (Linnaeus and hybrid Townsend Angelfish H. ciliaris X H. bermudensis Goode] in the tropical western Atlantic. It suggests how quickly this pattern could have originated in the other species, and/or some propensity of this pattern in the genus. The distinct double-spot color pattern anomaly suggests how quickly new color patterns can originate in genus Hypoplectrus. This is important because species in this genus are distinguished almost solely on the basis of color pattern and speciation may be occurring rapidly. Anomalies should be recorded because they may give us some hints at the genetic origin of species characters and some could represent potentially inheritable characters. We suggest these potentially inheritable characters could be recognized and described when they first arise in an individual and before they become inherited by a population. Following these potentially inheritable characters could help to explain how such characters enter into a population.   12. Different computer-assisted sperm analysis (CASA) systems highly influence sperm motility parameters. (United States)

    In this study, we examined different computer-assisted sperm analysis (CASA) systems (CRISMAS, Hobson Sperm Tracker, and Image J CASA) on the exact same video recordings to evaluate the differences in sperm motility parameters related to the specific CASA used. To cover a wide range of sperm motility parameters, we chose 12-second video recordings at 25 and 50 Hz frame rates after sperm motility activation using three taxonomically distinct fish species (sterlet: Acipenser ruthenus L.; common carp: Cyprinus carpio L.; and rainbow trout: Oncorhynchus mykiss Walbaum) that are characterized by essential differences in sperm behavior during motility. Systematically higher values of velocity and beat cross frequency (BCF) were observed in video recordings obtained at 50 Hz frame frequency compared with 25 Hz for all three systems. Motility parameters were affected by the CASA and species used for analyses. Image J and CRISMAS calculated higher curvilinear velocity (VCL) values for rainbow trout and common carp at 25 Hz frequency compared with the Hobson Sperm Tracker, whereas at 50 Hz, a significant difference was observed only for rainbow trout sperm recordings. No significant difference was observed between the CASA systems for sterlet sperm motility at 25 and 50 Hz. Additional analysis of 1-second segments taken at three time points (1, 6, and 12 seconds of the recording) revealed a dramatic decrease in common carp and rainbow trout sperm speed. The motility parameters of sterlet spermatozoa did not change significantly during the 12-second motility period and should be considered as a suitable model for longer motility analyses. Our results indicated that the CASA used can affect motility results even when the same motility recordings are used. These results could be critically altered by the recording quality, time of analysis, and frame rate of camera, and could result in erroneous conclusions. Copyright © 2013 Elsevier Inc.   13. Transcriptomics of environmental acclimatization and survival in wild adult Pacific sockeye salmon (Oncorhynchus nerka) during spawning migration. (United States)

    Environmental shifts accompanying salmon spawning migrations from ocean feeding grounds to natal freshwater streams can be severe, with the underlying stress often cited as a cause of increased mortality. Here, a salmonid microarray was used to characterize changes in gene expression occurring between ocean and river habitats in gill and liver tissues of wild migrating sockeye salmon (Oncorhynchus nerka Walbaum) returning to spawn in the Fraser River, British Columbia, Canada. Expression profiles indicate that the transcriptome of migrating salmon is strongly affected by shifting abiotic and biotic conditions encountered along migration routes. Conspicuous shifts in gene expression associated with changing salinity, temperature, pathogen exposure and dissolved oxygen indicate that these environmental variables most strongly impact physiology during spawning migrations. Notably, transcriptional changes related to osmoregulation were largely preparatory and occurred well before salmon encountered freshwater. In the river environment, differential expression of genes linked with elevated temperatures indicated that thermal regimes within the Fraser River are approaching tolerance limits for adult salmon. To empirically correlate gene expression with survival, biopsy sampling of gill tissue and transcriptomic profiling were combined with telemetry. Many genes correlated with environmental variables were differentially expressed between premature mortalities and successful migrants. Parametric survival analyses demonstrated a broad-scale transcriptional regulator, cofactor required for Sp1 transcriptional activation (CRSP), to be significantly predictive of survival. As the environmental characteristics of salmon habitats continue to change, establishing how current environmental conditions influence salmon physiology under natural conditions is critical to conserving this ecologically and economically important fish species. © 2011 Blackwell Publishing Ltd.

  14. Interactions of highly and low virulent Flavobacterium columnare isolates with gill tissue in carp and rainbow trout. (United States)

    The interactions of Flavobacterium columnare isolates of different virulence with the gills of carp (Cyprinus carpio L.) and rainbow trout (Oncorhynchus mykiss Walbaum) were investigated. Both fish species were exposed to different high (HV) or low virulence (LV) isolates and sacrificed at seven predetermined times post-challenge. Histopathological and ultrastructural examination of carp and rainbow trout inoculated with the HV-isolate disclosed bacterial invasion and concomitant destruction of the gill tissue, gradually spreading from the filament tips towards the base, with outer membrane vesicles surrounding most bacterial cells. In carp, 5-10% of the fish inoculated with the LV-isolate became moribund and their gill tissue displayed the same features as described for the HV-isolate, albeit to a lesser degree. The bacterial numbers retrieved from the gill tissue were significantly higher for HV- compared to LV-isolate challenged carp and rainbow trout. TUNEL-stained and caspase-3-immunostained gill sections demonstrated significantly higher apoptotic cell counts in carp and rainbow trout challenged with the HV-isolate compared to control animals. Periodic acid-Schiff/alcian blue staining demonstrated a significantly higher total gill goblet cell count for HV- and LV-isolate challenged compared to control carp. Moreover, bacterial clusters were embedded in a neutral matrix while being encased by acid mucins, resembling biofilm formation. Eosinophilic granular cell counts were significantly higher in the HV-isolate compared to LV-isolate inoculated and control carp. The present data indicate a high colonization capacity, and the destructive and apoptotic-promoting features of the HV-isolate, and point towards important dynamic host mucin-F. columnare interactions warranting further research.

    Full Text Available Abstract Background Epidermal pseudotumours from Hippoglossoides dubius and Acanthogobius flavimanus in Japan and gill lesions in Limanda limanda from the UK have been shown to be caused by phylogenetically related protozoan parasites, known collectively as X-cells. However, the phylogenetic position of the X-cell group is not well supported within any of the existing protozoan phyla and they are currently thought to be members of the Alveolata. Ultrastructural features of X-cells in fish pseudotumours are somewhat limited and no typical environmental stages, such as spores or flagellated cells, have been observed. The life cycles for these parasites have not been demonstrated and it remains unknown how transmission to a new host occurs. In the present study, pseudobranchial pseudotumours from Atlantic cod, Gadus morhua, in Iceland and epidermal pseudotumours from the northern black flounder, Pseudopleuronectes obscurus, in Japan were used in experimental transmission studies to establish whether direct transmission of the parasite is achievable. In addition, X-cells from Atlantic cod were sequenced to confirm whether they are phylogenetically related to other X-cells and epidermal pseudotumours from the northern black flounder were analysed to establish whether the same parasite is responsible for infecting different flatfish species in Japan. Results Phylogenetic analyses of small subunit ribosomal DNA (SSU rDNA sequence data from Atlantic cod X-cells show that they are a related parasite that occupies a basal position to the clade containing other X-cell parasites. The X-cell parasite causing epidermal pseudotumours in P. obscurus is the same parasite that causes pseudotumours in H. dubius. Direct, fish to fish, transmission of the X-cell parasites used in this study, via oral feeding or injection, was not achieved.     Pharmaceuticals in the environment (PIE) are of importance since these compounds are designed to affect biological receptors/enzymes that are often conserved across vertebrate families. Across-species extrapolation of these therapeutic targets suggests potential for impacting amphibia and fish in the aquatic environment. Due to the scarcity of relevant ecotoxicological data, the long-tem impact of PIE remains a research question. Efficient use of mammalian data has been proposed to better understand and predict the potential for a given pharmaceutical to impact the environment. Using a model cardiovascular pharmaceutical (propranolol, a non-specific {beta}{sub 1}/{beta}{sub 2}-adrenergic antagonist), the hypothesis that mammalian data can be used to predict toxicity in fish was tested. Rainbow trout (Oncorhynchus mykiss (Walbaum)) have {beta}-adrenergic signalling mechanisms analogous to human cardiovascular receptors that respond to pharmacological doses of agonists and antagonists. Trout absorbed propranolol from water such that after 40 days of exposure, the linear relationship was [plasma] = 0.59[water] (n = 31, r = 0.96). Growth rate was affected only at very high aqueous concentrations (10-day {sup growth}NOEC = 1.0 and {sup growth}LOEC = 10 mg/l). Growth recovered with time (40-day {sup growth}NOEC = 10 mg/l), suggesting possible adaptation to the pharmaceutical, although the internal plasma concentration in trout exposed to 10 mg propranolol/l of water was higher than the mammalian therapeutic plasma concentration. Additional endpoints suggested subtle changes of liver and heart size at much lower concentrations may have occurred, although these were not concentration-related. Bu değişimler üzerine çok çeşitli araştırmalar yapılmış ve halen yapılmaktadır. Bu çalışma “Büyük Kanal Projesinin Aktif Hale Geçirilmesinin İzmir Körfezi Sularında Alt Besinsel Seviyeler Üzerine Etkileri (YDABAG 102Y116” adlı proje kapsamında yürütülmüş olup İzmir İç Körfezi’nde yaşayan balıkların yumurta ve larvalarının tür çeşitliliği, bolluğu, dağılımları ve mortaliteleri araştırılmıştır. İhtiyoplankton örnekleri 19.12.2002-26.12.2003 tarihleri arasında 15’er günlük periyotlarda 3 istasyondan toplanmış olup ağız çapı 51 cm, uzunluğu 2.5 m ve 200 μm göz açıklığında UNESCO-WP2 model plankton kepçesi ile vertikal olarak gerçekleştirilmiştir. Çalışma süresince toplam 9613.8 adet/m2 yumurta, 568.4 adet/m2 larva tespit edilmiştir. Sardina pilchardus (Walbaum, 1792, Engraulis encrasicolus (Linnaeus, 1758, Diplodus sp., Symphodus melops (Linnaeus, 1758, Callionymus pusillus Delaroche, 1809, Callionymus sp., Gobius niger Linnaeus, 1758, Gobius paganellus Linnaeus, 1758, Arnoglossus sp., Buglossidium luteum (Risso, 1810 türlerinin yumurta ve/veya larvaları tayin edilmiştir. E. encrasicolus türüne ait yumurta, prelarva ve postlarva dominanttır. Bu türün yumurtalarında %12.05 oranında mortalite belirlenmiştir
