    This paper describes an investigation of the tensile capacity of in-situ cast U-bar loop connections between precast concrete elements. The basic idea is to introduce a small precast cylindrical dowel of fiber reinforced mortar that fits into the bend diameter of the overlapping U...... that use of a precast fiber reinforced dowel performs at a slightly lower load level, as compared to a connection grouted solely with regular mortar and reinforced with the same amount of transverse reinforcement.     We study the d-bar - u-bar asymmetry of the proton in a model approach in which hadronic fluctuations of the nucleon are generated through gluon splitting and recombination mechanisms. Within this framework, it is shown that the d -u asymmetry of the proton is consistently described by including only nucleon fluctuations to |πN> and |πΔ> bound states. Predictions of the model closely agree with the recent experimental data of the E-866/Nu Sea Collaboration. (author)

    Full Text Available Karl Popper once criticizes the positivistic paradigm by means of what he calls the theory of falsification. To him, the positivistic paradigm constitutes a failure simply because it cannot distinguish between science and prude science, that is, between empirical method and non-empirical method. In the scientific knowledge, empirical science, mathematics and logic must be differentiated, something that the positivistic paradigm has failed to do. This paradigm furthermore, did not have a clear picture what logic-based science is, empirical knowledge and psychology of knowledge. It also fails to properly understand knowledge that springs from tradition, emotion, authority and so forth. All positivistic paradigms such as that of Karl Max’s Historicism, Freud’s theory of Psycho Analysis, Adler’s theory of Individual Analysis and Newton’s theory of Gravitation fall into this trap of failure.

  20. New results from Fermilab E866 (NuSea) for d-bar/u-bar

    The Fermilab dimuon experiment 866/NuSea measured Drell-Yan yields from an 800 GeV/c proton beam incident on liquid hydrogen and deuterium targets. Over 370,000 Drell-Yan muon pairs were recorded. From these data, the ratio of anti-down (d-bar) to anti-up (u-bar) quark distributions in the proton sea is determined over a wide range in Bjorken-x. A strong x dependence is observed in the ratio d-bar/u-bar, showing substantial enhancement of d-bar with respect to u-bar for x < 0.2. The results presented here for the full data sets confirm previously published results from E866 and are compared with parametrizations of parton distribution functions calculated both before and after the publication of the high-mass E866 data

    The practical peak voltage (PPV) has been adopted as the reference measuring quantity for the x-ray tube voltage. However, the majority of commercial kV-meter models measure the average peak, U-bar{sub P}, the average, U-bar, the effective, U{sub eff} or the maximum peak, U{sub P} tube voltage. This work proposed a method for determination of the PPV from measurements with a kV-meter that measures the average U-bar or the average peak, U-bar{sub p} voltage. The kV-meter reading can be converted to the PPV by applying appropriate calibration coefficients and conversion factors. The average peak k{sub PPV,kVp} and the average k{sub PPV,Uav} conversion factors were calculated from virtual voltage waveforms for conventional diagnostic radiology (50-150 kV) and mammography (22-35 kV) tube voltages and for voltage ripples from 0% to 100%. Regression equation and coefficients provide the appropriate conversion factors at any given tube voltage and ripple. The influence of voltage waveform irregularities, like 'spikes' and pulse amplitude variations, on the conversion factors was investigated and discussed. The proposed method and the conversion factors were tested using six commercial kV-meters at several x-ray units. The deviations between the reference and the calculated - according to the proposed method - PPV values were less than 2%. Practical aspects on the voltage ripple measurement were addressed and discussed. The proposed method provides a rigorous base to determine the PPV with kV-meters from U-bar{sub p} and U-bar measurement. Users can benefit, since all kV-meters, irrespective of their measuring quantity, can be used to determine the PPV, complying with the IEC standard requirements.

    QCD one-loop corrections to the full gauge invariant set of electroweak diagrams describing the hadronic process $e^+ e^- \\to u~\\bar d~s~\\bar c$ are computed. Four-jet shape variables for $WW$ events are studied at next-to-leading order and the effects of QCD corrections on the determination of the $W$--mass in the hadronic channel at Lep 2 and NLC is discussed. We compare the exact calculation with a ``naive''approach to strong radiative corrections which has been widely used in the literature.

    The eminent mathematical physicist Sir Hermann Bondi once said: “There is no more to science than its method, and there is no more to its method than Popper has said.” Indeed, many regard Sir Karl Raimund Popper the greatest philosopher of science in our generation. Much of what Popper “has said” refers to physics, but physicists, generally speaking, have little knowledge of what he has said. True, Popper's philosophy of science and, in particular, his realistic interpretation of quantum mechanics deviates considerably from the generally accepted doctrine. But as Popper, rightly I think, points out, it is precisely the proliferation of divergent theories which promotes the growth of scientific knowledge; it would be a danger for physics if physicists were dogmatically tied to a single theory or would not test their theory against alternatives. It is for this purpose that, on the occasion of the nonagenarian celebration of Popper's birthday, the present essay has been written.

    Full Text Available This article outlines the development of nineteenth-century theatre from a sociological viewpoint, in particular, focusing on the coming of age of the Wiener Volkstheater under the direction of Raimund and Nestroy, as well as with the staging of works by playwrights as different from each other as Grabbe, Grillparzer and Büchner. All three of them, from different angles, tackled the historical genre as well as the problem of theatre seen in a new light. However, it was not their plays that were successful during the period of the Restoration but rather grand scale historical dramas on familiar themes by mostly forgotten playwrights, often translations and adaptations of French and Classical works. In the decades that followed the Revolution of 1848, in which fiction was the dominant literary genre, Hebbel renovated tragedy as form, whereas Wagner went on to reform theatre at all levels.

  16. Parton degrees of freedom from the path-integral formalism

    We formulate the hadronic tensor W μν of deep inelastic scattering in the path-integral formalism. It is shown that there are 3 gauge invariant and topologically distinct contributions. In addition to the valence contribution, there are two sources for the sea--one in the connected insertion and the other in the disconnected insertion. The operator product expansion is carried out in this formalism. The operator rescaling and mixing reveal that the connected sea partons evolve the same way as the valence; i.e., their evolution is decoupled from the disconnected sea and the gluon distribution functions. We explore the phenomenological consequences of this classification in terms of the small x behavior, Gottfried sum rule violation, and flavor dependence. In particular, we point out that in the nucleon u(bar sign) and d(bar sign) partons have both connected and disconnected sea contributions, whereas the s(bar sign) parton has only the disconnected sea contribution. This difference between u(bar sign)+d(bar sign) and s(bar sign), as far as we know, has not been taken into account in the fitting of parton distribution functions to experiments.   17. Hypotrochoids in conformal restriction systems and Virasoro descendants

    A conformal restriction system is a commutative, associative, unital algebra equipped with a representation of the groupoid of univalent conformal maps on connected open sets of the Riemann sphere, along with a family of linear functionals on subalgebras, satisfying a set of properties including conformal invariance and a type of restriction. This embodies some expected properties of expectation values in conformal loop ensembles CLE κ (at least for 8/3 iθ and w. We find that it has an expansion in positive powers of u and u-bar , and that the coefficients of pure u ( u-bar ) powers are holomorphic in w ( w-bar ). We identify these coefficients (the ‘hypotrochoid fields’) with certain Virasoro descendants of the identity field in conformal field theory, thereby showing that they form part of a vertex operator algebraic structure. This largely generalizes works by the author (in CLE), and the author with his collaborators Riva and Cardy (in SLE 8/3 and other restriction measures), where the case of the ellipse, at the order u 2 , led to the stress–energy tensor of CFT. The derivation uses in an essential way the Virasoro vertex operator algebra structure of conformal derivatives established recently by the author. The results suggest in particular the exact evaluation of CLE expectations of products of hypotrochoid fields as well as nontrivial relations amongst them through the vertex operator algebra, and further shed light onto the relationship between CLE and CFT.     The semi-inclusive difference asymmetry A^{h+ - h-} for hadrons of opposite charge has been measured by the COMPASS experiment at CERN. The data were collected in the years 2002-2004 using a 160 GeV polarised muon beam scattered off a large polarised 6-LiD target and cover the range 0.006 < x < 0.7 and 1 < Q^2 < 100 (GeV/c)^2. In leading order QCD (LO) the asymmetry A_d^{h+ - h-} measures the valence quark polarisation and provides an evaluation of the first moment of Delta u_v + Delta d_v which is found to be equal to 0.40 +- 0.07 (stat.) +- 0.05 (syst.) over the measured range of x at Q^2 = 10 (GeV/c)^2. When combined with the first moment of g_1^d previously measured on the same data, this result favours a non-symmetric polarisation of light quarks Delta u-bar = -Delta d-bar at a confidence level of two standard deviations, in contrast to the often assumed symmetric scenario Delta u-bar = Delta d-bar = Delta s-bar = Delta s.

    The complete tree level cross section for e + e - → e - ν-bar e ud-barγ is computed and discussed in comparison with the cross sections for e + e - → e - V-bar e u d-bar and e + e - → u-bar d u d-bar. Event generators based on the GRACE package for the non-radiative and radiative case are presented. Special interest is brought to the effect of the non-resonant diagrams overlooked so far in other studies. Their contribution to the total cross section is presented for the LEP II energy range for future linear colliders (√s = 500 GeV). (author)

  20. Identified Hadron Production and Light Quark Fragmentation in Z{sup 0} Decays

    We have measured the differential cross sections for the production of {pi}{sup +}, K{sup +}, K{sup 0}, K*{sup 0}, {phi}, p, {Lambda} and their corresponding antiparticles in separate samples of flavor-tagged Z{sup 0} {yields} light-flavor (u{bar u}, d{bar d}, or s{bar s}), Z{sup 0} {yields} b{bar b} events. Clear flavor dependences are observed, and the results are compared with the predictions of three fragmentation models. We have also performed a direct measurement of A{sub s}, the parity-violating coupling of the Z{sup 0} to strange quarks, by measuring the left-right-forward-backward production asymmetry in polar angle of the tagged s quark. Our preliminary result is A{sub s} = 0.82 {+-} 0.10(stat.) {+-} 0.07(syst.).

    The measurements were performed using the CASAC (Center for Astrochemical Studies Absorption Cell) spectrometer at the Max-Planck-Institut fuer extraterrestrische Physik. Full details of the experimental apparatus can be found in Bizzocchi et al. (2017A&A...602A..34B). The instrument employs the frequency modulation technique in the mm and submm range and is equipped with a glow discharge cell for production of unstable species. Measurements were carried out at room temperature to avoid condensation of the precursors. Dithioformic acid was produced by a 40mA DC discharge (~0.8kV) from a 1:1 mixture of CS2 and H2, diluted in Ar buffer gas. The total pressure in the cell was 20-30mTorr (27-40ubar). (4 data files).

    New data on structure functions from deep-inelastic scattering provide new information on parton distributions, particularly in the 0.01 2 data from the New Muon Collaboration (NMC) and its implications for other processes, and the evidence for SU(2) symmetry breaking in the light quark sea. We show that although good fits can be obtained with or without this symmetry breaking, more physically reasonable parton distributions are obtained if we allow d-bar > u-bar at small x. With the inclusion of the latest deep-inelastic data we find α s (M Z ) = 0.111 -0.005 +0.004 . We also show how W, Z and Drell-Yan production at p-barp colliders can give information on parton distributions. (Author)

    This paper concerns the in-plane shear strength of connections between precast concrete wall elements reinforced with looped high-strength wire ropes. The looped wire ropes are pre-installed in so-called ‘wire boxes’ which function as shear keys. Although only a small amount of research...... on the shear strength of such connections can be found in the literature, this type of connection is increasingly being used because wire ropes are much more construction-friendly than traditional U-bars. A rigid plastic upper bound model for the shear strength of wall connections reinforced with looped wire...... ropes that are pre-installed in wire boxes is presented along with test results on the shear strength of connections with double-wire boxes. It is shown that the plastic solution agrees well with both the obtained test results and results from previously conducted tests....

    Full Text Available To what extent and in what ways did the intellectual climate of Austria’s often ethnolinguistically heterogeneous borderlands contribute to the formation, institutionalization and diffusion of emerging social scientific discourses during the final decades of the 19th century? Investigating the intellectual exchange between two early proponents of folklore studies (Volkskunde—the Slavonian-German-Jewish Friedrich Salomon Krauss (1859–1938 and Bukovinian-German Raimund Friedrich Kaindl (1866–1930—this paper argues that imperial peripheries, while traditionally overlooked as sites of knowledge production, in fact played a pivotal role in the development of an important brand of “progressive” social scientific research, one defined by a critical stance toward the prevailing historicist paradigms of the time. These self-described “social democrats of scholarship” collaborated, both formally and informally, on a number of related theoretical projects aimed at disrupting the exclusionary narratives of the academic establishment and re-focusing scholarly attention on the sociological, rather than historical, character of ethnonational difference. In this way, the nationalities question spurred, both in the center and at the margins of the monarchy, the development of new sciences of nationality intended to sustain Austria’s imperial structure.

    Full Text Available Instead of using the term Biedermeier to label the literary period between Romanticism and Realism, the terms “literature of the Restoration period” or “pre-March period literature” are sometimes used. Biedermeier designates the period between 1815 (Congress of Vienna and 1850 (the aftermath of the revolutionary years 1848/49. Paul Kluckholm was the first to use the term “biedermeier” (in 1927 after it had lost its pejorative overtones at the end of the 19th century and, at the beginning of the 20th, taken on a meaning designating the lifestyle and fashion of the first half of the 19th century. The Biedermeier epoch does not have a specific aesthetic or theoretical programme, which led to a stylistic discontinuity: a naïve concatenation of various forms of expression, mixing of genres, and underestimating the formal aspects of a work of art — putting emphasis on “detailed descriptions”, frequent reflections. This allowed for an unprecedented proliferation of trivial fiction issued in a multitude of almanacs, pocket editions, and numerous “family” magazines. Drama became an important genre, mainly the Viennese people play and conversational plays (Raimund, Nestroy, Bauernfeld.

    Over the last three decades, the philosophy of Karl Raimund Popper has had a strong influence on the field of systematic biology. Unequivocally, no other philosopher's work has had such an influence during this formative period in systematics. Much, but not all, of the early discourse on Popper and systematics dealt with the philosophical basis of systematics as a science. More recently Popper's work has been discussed in the systematics literature in relation to specific methodologies such as parsimony and maximum likelihood. In this paper, we provide the reader with a concise summary of Popper's ideas relevant to systematics, review the systematic literature invoking or declining Popper's importance to the field, and make a recommendation for the future course of philosophical thinking in systematics. We try to make clear various authors' interpretations of Popper's work and how those interpretations have impacted systematic thought. Although the reader may come away from this review with a clearer idea of Popper's relevance or lack thereof, our primary hope is that the reader will be compelled to question him- or herself about the philosophical basis of the systematic work that he or she does, and to delve into the literature herein cited. We begin by presenting a synopsis of Popper's philosophical views to allow those views to be placed in the context of systematics.

    Decoupling the chiral dynamics in the canonical approach to the WZNW model requires an extended phase space that includes left and right monodromy variables M and M-bar. Earlier work on the subject, which traced back the quantum group symmetry of the model to the Lie-Poisson symmetry of the chiral symplectic form, left some open questions: How to reconcile the necessity to set M M-bar -1 = 1 (in order to recover the monodromy invariance of the local 2D group valued field g = uu-bar) with the fact the M and M-bar obey different exchange relations? What is the status of the quantum symmetry in the 2D theory in which the chiral fields u(x-t) and u-bar(x+t) commute? Is there a consistent operator formalism in the chiral (and the extended 2D) theory in the continuum limit? We propose a constructive affirmative answer to these questions for G = SU(2) by presenting the quantum field u and u-bar as sums of products of chiral vertex operators and q Bose creation and annihilation operators.   8. QCD tests with SLD and polarized beams

    The author presents a measurement of the strong coupling {alpha}{sub s} derived from multijet rates using data collected by the SLD experiment at SLAC and find that {alpha}{sub s}(M{sub Z}{sup 2}) = 0.118 {+-} 0.002(stat.) {+-} 0.003(syst.) {+-} 0.010(theory). He presents tests of the flavor independence of strong interactions via preliminary measurements of the ratios {alpha}{sub s}(b)/{alpha}{sub s}(udsc) and {alpha}{sub s}(uds)/{alpha}{sub s}(bc). In addition, the group has measured the difference in charged particle multiplicity between Z{sup 0} {yields} b{bar b} and Z{sup 0} {yields} u{bar u}, d{bar d}, s{bar s} events, and find that it supports the prediction of perturbative QCD that the multiplicity difference be independent of center-of-mass energy. Finally, the group has made a preliminary study of jet polarization using the jet handedness technique.

  9. Contribution to the study of transverse turbulent diffusion in streams

    In this research our objective is to study the turbulent diffusion in a water flow, in particular the transverse diffusion. According to formulae reviewed in literature the diffusion coefficients (K) may be expressed as a function of several velocity parameters. A synthetic formula depending on a macro-scale length of turbulence L x and a turbulent intensity √(u' 2 ) is proposed: K = β.L x .U-bar.(√(u' 2 /U-bar)) 2 . In order to validate this expression we performed two in situ experiments (one in a wide river and the other in an irrigation canal) with a double set of measurements: turbulent velocities and concentrations of a diffusing tracer. The first set gives us usable data in our formula. The results, compared with values available in literature, give a good agreement. Moreover it appears that it is possible to roughly divide the data in two groups according to (1) the cross section shape and (2) the bed roughness. The second set allows us to evaluate a global turbulent mixing coefficient. The coefficients calculated by the two methods are in accordance so our formula is validated. Nevertheless some problems appear because of what is called secondary currents and coherent structures as those seen above bed cracks in the Garonne river. Those phenomenon may play a major part upon turbulent diffusion in real streams. Although they were made conspicuous by an analysis of transverse velocity component, it has not been yet possible to quantify their effects. (author) [fr

  10. A Preliminary Direct Measurement of the Parity-Violating Coupling of the Z{sup 0} to Strange Quarks, A{sub s}

    We present an updated direct measurement of the parity-violating coupling of the Z{sup 0} to strange quarks, A{sub s}, derived from the full SLD data sample of approximately 550,000 hadronic decays of Z{sup 0} bosons produced with a polarized electron beam and recorded by the SLD experiment at SLAC between 1993 and 1998. Z{sup 0} {r_arrow} s{bar s} events are tagged by the presence in each event hemisphere of a high-momentum K{sup {+-}}, K{sub s} or {Lambda}{sup 0}/{bar {Lambda}}{sup 0} identified using the Cherenkov Ring Imaging Detector and/or a mass tag. The CCD vertex detector is used to suppress the background from heavy-flavor events. The strangeness of the tagged particle is used to sign the event thrust axis in the direction of the initial s quark. The coupling A{sub s} is obtained directly from a measurement of the left-right-forward-backward production asymmetry in polar angle of the tagged s quark. The background from u{bar u} and d{bar d} events is measured from the data, as is the analyzing power of the method for s{bar s} events. We measure: A{sub s} = 0.85 {+-} 0.06(stat.) {+-} 0.07(syst.)(preliminary).

    A multivariate likelihood method to measure electroweak couplings with the Drell-Yan process at the LHC is presented. The process is described by the dilepton rapidity, invariant mass, and decay angle distributions. The decay angle ambiguity due to the unknown assignment of the scattered constituent quark and antiquark to the two protons in a collision is resolved statistically using correlations between the observables. The method is applied to a sample of dimuon events from proton-proton collisions at sqrt(s) = 7 TeV collected by the CMS experiment at the LHC, corresponding to an integrated luminosity of 1.1 inverse femtobarns. From the dominant u-ubar, d-dbar to gamma*/Z to opposite sign dimuons process, the effective weak mixing angle parameter is measured to be sin^2(theta[eff]) = 0.2287 +/- 0.0020 (stat.) +/- 0.0025 (syst.). This result is consistent with measurements from other processes, as expected within the standard model.

    The purpose of pipe rupture studies in JAERI is to perform the model tests on pipe whip, restraint behavior, jet impingement and jet thrust force, and to establish the computational method for analyzing these phenomena. This report describes the experimental results of pipe whip on the pipe specimens of 4 inch in diameter under BWR condition on which the pressure is 6.77 MPa and the temperature is 285 0 C. The pipe specimens were 114.3 mm (4 inch) in diameter and 8.6 mm in thickness and 4500 mm in length. Four pipe whip restraints used in the tests were the U-bar type of 8 mm in diameter and fabricated from type 304 stainless steel. The experimental parameter was the clearance (30, 50 and 100 mm). The dynamic strain behavior of the pipe specimen and the restraints was investigated by strain gages and their residual deformation was obtained by measuring marking points provided on their surface. The Pressure-time history in the pipe specimens was also obtained by pressure gages. The maximum pipe strain is caused near the restraints and increases with increase of the clearance. The experimental results of pipe whip tests indicate the effectiveness of pipe whip restraints. The ratio of absorbed strain energy of the pipe specimen to that of the restraints is nearly constant for different clearances at the overhang length of 400 mm. (author)

    Double-spin asymmetries for production of charged pions and kaons in semi-inclusive deep-inelastic muon scattering have been measured by the COMPASS experiment at CERN. The data, obtained by scattering a 160 GeV muon beam off a longitudinally polarised NH_3 target, cover a range of the Bjorken variable x between 0.004 and 0.7. A leading order evaluation of the helicity distributions for the three lightest quarks and antiquark flavours derived from these asymmetries and from our previous deuteron data is presented. The resulting values of the sea quark distributions are small and do not show any sizable dependence on x in the range of the measurements. No significant difference is observed between the strange and antistrange helicity distributions, both compatible with zero. The integrated value of the flavour asymmetry of the helicity distribution of the light-quark sea, \\Delta u-bar - \\Delta d-bar, is found to be slightly positive, about 1.5 standard deviations away from zero.

    Full Text Available The title “I have called you by your number” is paraphrased from the book of Isaiah. In Torah the change of name of the main character suggests the turning point of his/her fate. In the article I concentrate on the gesture of assuming pseudonym-anagrams by a few poets – it is viable to read it as an attempt at self-determination, and as a painful dividing line, and as a form of a mask, and an analogue of a perturbed fate. My reflection concentrates on texts by Sachs, Domin, Celan, Amichai, Améry with a motif of numbers (the turned over analogues of the names – I find a context in the poem by Wisława Szymborska Still; and in the performance by Raimund Hoghe, staged only once, just in Poland, entitled site specific; the project 80064 by Artur Żmijewski or works by Roman Opałka. The promise written in the Book of Isaiah of liberating Israel, which was supposed to be a work of God’s omnipotence, God talking to Jacob – who is an allegory of Israel: “Don’t be afraid, for I have redeemed you; I have called you by your name, you are mine. When you pass through the waters, I will be with you; and through the rivers, they shall not overflow you: when you walk through the fire, you shall not be burned, neither shall the flame kindle on you. For I am Yahweh your God”, in Auschwitz turned into the antithesis, and hence a threat that has a completely different sender than a good father: Be afraid, or else nobody will redeem you. I am calling you by the number, you are no one’s. You will go to the fire and a flame will consume you. You are alone.

  17. Trapping, percolation, and anomalous diffusion of particles in a two-dimensional random field

    The authors analyze the advection of particles in a velocity field with Hamiltonian H(x,y) = bar V 1 y-bar V 2 x + W 1 (y) - W 2 (x), where W i , i=1,2, are random functions with stationary, independent increments. In the absence of molecular diffusion, the particle dynamics are sensitive to the streamline topology, which depends on the mean-to-fluctuations ratio p=max(|bar V 1 |/bar U;|bar V 2 |/bar U), with bar U = [|W' 1 | 2 ] 1/2 = rms fluctuations. The model is exactly solvable for p≥1 and well suited for Monte Carlo simulations for all p. Statistics are considered of streamlines for p=0, deriving power laws for the escape probability and the length of escaping trajectories for a box of size L much-gt 1. Also obtained is a characterization of the statistical topography of the Hamiltonian. The large-scale transport is studied of advected particles with p > 0. For 0 -v/2 [x(t) - (x(t))] and t -v/2 [y(t) - (y(t))]. The large-scale motions are Fickian (v=1) or superdiffusive (v=3/2) with a non-Gaussian coarse-grained probability, according to the direction of the mean velocity relative to the underlying lattice. These results are obtained analytically for p≥1 and extended to the regime 0 1 , bar V 2 ) for which stagnation regions in the flow exist. The results are compared with existing predictions on the topology of streamlines based on percolation theory and with mean-field calculations of effective diffusivities. 29 refs., 15 figs., 7 tabs

    Using a luminosity of 210.7 fb{sup -1} integrated at the {upsilon}(4S) resonance with BaBar experiment, we exclusively reconstruct the B{sup {+-}} to DK{sup *{+-}} decay for constraining the angle {gamma} of the unitarity triangle. Following the method described by Gronau, London et Wyler, the D meson is reconstructed in CP eigenstates: K{sup +}K{sup -}, {pi}{sup +}{pi}{sup -}, et K{sub s}{pi}{sup 0}, K{sub s}{phi}, K{sub s}{omega}. The interference between amplitudes b-bar {yields} c-bar u s-bar and b-bar {yields} u-bar c s-bar, which contribute to the final states, is the origin of the sensitivity on {gamma}. The technical difficulty comes from the suppression of the second amplitude compared to the first one, which we expect a ratio of the order of 0.1. The small ratio of r{sub B} limits the impact on the {gamma} measurement. Physical observables for this method have been measured: A{sub +} = -0,08 {+-} 0,19 (stat.) {+-} 0,08 (syst.); A{sub -} = -0,26 {+-} 0,40 (stat.) {+-} 0,12 (syst.); R{sub +} = 1,96 {+-} 0,40 (stat.) {+-} 0,11 (syst.); and R{sub -} = 0,65 {+-} 0,26 (stat.) {+-} 0,08 (syst.).   19. New Parameters for Higher Accuracy in the Computation of Binding Free Energy Differences upon Alanine Scanning Mutagenesis on Protein-Protein Interfaces.

    Knowing how proteins make stable complexes enables the development of inhibitors to preclude protein-protein (P:P) binding. The identification of the specific interfacial residues that mostly contribute to protein binding, denominated as hot spots, is thus critical. Here, we refine an in silico alanine scanning mutagenesis protocol, based on a residue-dependent dielectric constant version of the Molecular Mechanics/Poisson-Boltzmann Surface Area method. We have used a large data set of structurally diverse P:P complexes to redefine the residue-dependent dielectric constants used in the determination of binding free energies. The accuracy of the method was validated through comparison with experimental data, considering the per-residue P:P binding free energy (ΔΔG binding ) differences upon alanine mutation. Different protocols were tested, i.e., a geometry optimization protocol and three molecular dynamics (MD) protocols: (1) one using explicit water molecules, (2) another with an implicit solvation model, and (3) a third where we have carried out an accelerated MD with explicit water molecules. Using a set of protein dielectric constants (within the range from 1 to 20) we showed that the dielectric constants of 7 for nonpolar and polar residues and 11 for charged residues (and histidine) provide optimal ΔΔG binding predictions. An overall mean unsigned error (MUE) of 1.4 kcal mol -1 relative to the experiment was achieved in 210 mutations only with geometry optimization, which was further reduced with MD simulations (MUE of 1.1 kcal mol -1 for the MD employing explicit solvent). This recalibrated method allows for a better computational identification of hot spots, avoiding expensive and time-consuming experiments or thermodynamic integration/ free energy perturbation/ uBAR calculations, and will hopefully help new drug discovery campaigns in their quest of searching spots of interest for binding small drug-like molecules at P:P interfaces.

Adenosine and dipyridamole are the most commonly used agents to induce coronary arterial vasodilation for myocardial perfusion imaging. Both cause frequent undesirable side-effects. Because of its ultrashort half-life, adenosine must be administered by constant intravenous infusion during the examination. A key feature of an ideal A2A agonist for myocardial perfusion imaging studies would be an optimal level and duration of hyperemic response. Drugs with a longer half-time and more selective A2A adenosine receptor agonism, such as regadenoson, should theoretically result in a similar degree of coronary vasodilation with fewer or less severe side-effects than non-selective, ultrashort-lasting adenosine receptor stimulation. The available preclinical and clinical data suggest that regadenoson is a highly subtype-selective, potent, low-affinity A2A adenosine receptor agonist that holds promise for future use as a coronary vasodilator in myocardial perfusion imaging studies. Infusion of regadenoson achieves maximum coronary hyperemia that is equivalent to adenosine. After a single bolus infusion over 10 s, hyperemia is maintained significantly longer (approximately 2–5 min than with adenosine, which should facilitate radionuclide distribution for myocardial perfusion imaging studies. In comparison with the clinically competitive A2A adenosine receptor agonist binodenoson, regadenoson has a several-fold shorter duration of action, although the magnitude of hyperemic response is comparable between the two.
