    Suarez Contreras, C.; Diaz Palma, A.; Paz, M. D.

Hydrolysis of resistant cellulose of Onopordum nervosum Boiss (thistle) to reducing sugars in dilute sulfuric acid in glass ampoules and long residence times has been studied and kinetic parameters determined. The rate of hydrolysis is similar to that of the cellulose of Douglas fir, but comparatively the effect of the acid is more pronounced than temperature. From kinetic data it can be pre ducted the yield and since it can be obtained at least 45% of the potential glucose (48% as reducing sugars) at 190 degree centigree, 1,6% acid and 6,1 min. residence time, it indicates that the continuous acid hydrolysis of thistle may be a process of commercial interest. Hybridization is known to have a creative role in plant evolution. However, it can also have negative effects on parental species. Onopordum is a large genus whose species frequently hybridize. In the Southwest Iberian Peninsula, the rare O. hinojense co-occurs with the widely distributed O. nervosum, and hybrids between these two taxa have been described as O. × onubense. In this study we determine the extinction risk in a hybrid zone, both for hybrids and parentals, using analyses of morphological and cytogenetic traits as well as genetic markers and demographic models. To investigate the introgression process we used amplified fragment length polymorphism (AFLP) markers, Bayesian analyses and genome scan methods. Morphology, genome size and molecular markers confirmed homoploid hybridization and also indicated unidirectional backcrossing of F₁ hybrids with O. nervosum, which is likely to swamp O. hinojense, the parental with lower pollen size and a very low fruit set (8%). Genome scan methods revealed several loci significantly deviating from neutrality. Finally, our demographic modeling indicated that the higher fitness of O. nervosum threats the survival of O. hinojense by demographic swamping. Our study provides strong new evidence for a scenario of rapid extinction by unidirectional introgression and demographic swamping. The multifaceted approach used here sheds new light on the role of introgression in plant extinctions.

  4. Effect of gamma Irradiation on the acidic hydrolysis of free-hemi cellulose thistle; Efecto de la irradiacion gamma en la hidrolisis acida de cardo exento de pentosas

    Energy Technology Data Exchange (ETDEWEB)

    Suarez, C.; Paz, M. D.; Diaz, A.

The effect of gamma-irradiation on the subsequent acidic hydrolysis of free-hemi cellulose Onopordum Nervosum Boiss thistle Ls determined. Its shown the influence of gamma-irradiation on the yield of sugar obtained flora the batch wise hydrol isis of the call ulose (1% H{sub 2}SO{sub 4} and 180 degree centigree at increasing doses). At all irradiation levels studied, the rate of hydrolysis of thistle samples was higher than the rate of hydrolysis of the cellulose from paper treated similarly. The maximum over-all yield of sugar in the irradiated lignocellulose material was about 66o at 100 MRad, less than two times the yield obtainable from the control. The corresponding yield from papel was 53%, 2'3 times that of the control. Irradiation under 1% H{sub 2}SO{sub 4} doesn't enhance the yield anyway. (Author) 21 refs.

Positron annihilation lifetime spectroscopy (PALS) is applied to a series of bis(aniline)fluorene and bis(xylidine)fluorene-based cardo polyimide and bis(phenol)fluorene-based polysulfone membranes. It was found that favorable amounts of positronium (Ps, the positron-electron bound state) form in cardo polyimides with the 2,2-bis(3,4-dicarboxyphenyl) hexafluoropropane dianhydride (6FDA) moiety and bis(phenol)fluorene-based cardo polysulfone, but no Ps forms in most of the polyimides with pyromellitic dianhydride (PMDA) and 3,3',4,4'-biphenyltetracarboxylic dianhydride (BTDA) moieties. A bis(xylidine)fluorene-based polyimide membrane containing PMDA and BTDA moieties exhibits a little Ps formation but the ortho-positronium (o-Ps, the triplet state of Ps) lifetime of this membrane anomalously shortens with increasing temperature, which we attribute to chemical reaction of o-Ps. Correlation between the hole size (V(h)) deduced from the o-Ps lifetime and diffusion coefficients of O2 and N2 for polyimides with the 6FDA moiety and cardo polysulfone showing favorable Ps formation is discussed based on free volume theory of gas diffusion. It is suggested that o-Ps has a strong tendency to probe larger holes in rigid chain polymers with wide hole size distributions such as those containing cardo moieties, resulting in deviations from the previously reported correlations for common polymers such as polystyrene, polycarbonate, polysulfone, and so forth.

The effects of irradiation on the polyetherketone with cardo group(PEK-C) were studied. It was found that PEK-C can be crosslinked by irradiation under vacuum, while degradation reaction occurred in PEK-C at room temperature in the presence of air. Moreover, it was also found that Tg value of the crosslinked PEK-C at high temperature is higher than that at room temperature in the case of the same gel content, whose value is about 8 ℃ higher than that of unirradiated PEK-C. The gelation dose of PEK-C at 300 ℃ under vacuum is 1.S× 104Gy, which is about hundred times smaller than that at room temperature.

Full Text Available Cadmium nephrotoxicity is a serious environmental health problem as it will eventually end up with end stage renal disease. The pathobiochemical mechanism of this toxic heavy metal is related to oxidative stress. This study investigated whether Cleistocalyx nervosum var. paniala fruit extract (CNFE could protect the kidney against oxidative injury caused by cadmium. Initial analysis of the extract revealed antioxidant abilities and high levels of polyphenols, particularly catechin. Its potential renal benefits was further explored in rats treated with vehicle, CNFE, cadmium (2 mg/kg, and cadmium plus CNFE (0.5, 1, 2 g/kg for four weeks. Oxidative renal injury was developed after cadmium exposure as evidenced by blood urea nitrogen and creatinine retention, glomerular filtration reduction, renal structural damage, together with increased nitric oxide and malondialdehyde, but decreased antioxidant thiols, superoxide dismutase, and catalase in renal tissues. Cadmium-induced nephrotoxicity was diminished in rats supplemented with CNFE, particularly at the doses of 1 and 2 g/kg. It is concluded that CNFE is able to protect against the progression of cadmium nephrotoxicity, mostly via its antioxidant power. The results also point towards a promising role for this naturally-occurring antioxidant to combat other human disorders elicited by disruption of redox homeostasis.

Cleistocalyx nervosum var. paniala, an edible fruit found in Northern Thailand, contains high amounts of phenolic compounds with in vitro antioxidant activity. The aqueous extract of the ripe fruit was evaluated for its safety and beneficial effects using genotoxicity and toxicity tests. The C. nervosum extract was not only non-mutagenic in Salmonella typhimurium strains TA98 and TA100 in the presence and absence of metabolic activation, but exhibited also moderate antimutagenic effects against aflatoxin B1 and 2-amino-3,4-dimethylimidazo[4,5-f]quinoline-induced mutagenesis. Electrospray ionization-mass spectrometric analysis revealed the major anthocyanins, which included cyanidin-3,5-diglucoside, cyanidin-3-glucoside and cyanidin-5-glucoside. The administration of C. nervosum at concentration of 5,000 mg/kg bw did not induce acute toxicity in rats. A liver micronucleus test was performed to detect clastogenicity and anticlastogenicity. The extract in the dose of 1,000 mg/kg did not cause micronucleus formation in the liver of rats. Furthermore, in rats administered 100-1,000 mg/kg of the extract, no anticlastogenic effect against diethylnitrosamine-induced hepatic micronucleus formation was observed. These studies provide data concerning the safety and antimutagenic potency of an aqueous extract of C. nervosum fruit.

Full Text Available In this work, we intend to describe the reproductive dynamics of Sterna hirundinacea in an island from South Brazil. We studied the reproductive biology of this species in its natural environment and provide data on their growth, survival, and reproductive success in Ilha dos Cardos, Santa Catarina, South Brazil. Samplings were carried out daily on the island throughout the reproductive seasons of 2003, 2005, and 2006 and the different stages of development of the chicks were characterized according to age, length of the beak, and plumage characteristics. We provide a basic equation Lm=167.91 (1-e-0.062t--0.23 to determine the approximate age of individuals using their body mass. The main cause of chick mortality on the island was natural (63.17% in 2003, 81.41% in 2005, and 79.96% in 2006, whereas predation contributed to mortality in a proportion of 38.83% in 2003, 18.59% in 2005, and 20.04% in 2006. The absence in the area of the chicks’ main predator, Kelp gull (Larus dominicanus, the large number of chicks that reached the final stages of development, and their reproductive success demonstrate that Ilha dos Cardos is an important breeding site for the species in southern Brazil.

  13. Hole size distributions in cardo-based polymer membranes deduced from the lifetimes of ortho-positronium (United States)

    Kobayashi, Y.; Kinomura, A.; Kazama, S.; Inoue, K.; Toyama, T.; Nagai, Y.; Haraya, K.; Mohamed, H. F. M.; O'Rourke, B. E.; Oshima, N.; Suzuki, R.

To clarify the free volume size distributions of the cardo-based polymer membranes, where ortho-positronium (o-Ps) undergoes pick-off annihilation, the o-Ps lifetime distributions were analyzed by the LT9 programme. It was found that the cardo-based polysulfone membrane has much narrower o-Ps lifetime/hole size distributions than the cardo-based polyimide membranes with the 2,2-bis(3,4-dicarboxyphenyl)hexafluoropropane dianhydride (6FDA) moiety. Further, the lifetime/hole size distributions of the cardo-based polymer membranes are appreciably broadened with increasing temperature. This suggests that in these membranes there are holes not only of different sizes but also of different thermal expansion coefficients. It is also shown that in a membrane with a wider hole size distribution the average o-Ps lifetime tends to be longer than would be expected from the correlation between the o-Ps lifetime and the total free volume for common polymers.

The antioxidant activity of the extracts was assessed using the following methods: DPPH, ABTS+ both free radicals scavenging assays, and reducing assays. TP and TF were detected by spectrophotometric and HPLC methods. In former methods, the highest amount of TP content was ethy lacetate extract (EAE, expressed as gallic acid equivalents. The greatest TF content was in the n-butanol extract (BE, expressed as lutin equivalents. No significant difference was observed in the TP/TF content between these two extracts. The antioxidant activity and TP/TF content of three extracts seemed to follow the same trend. This implied that there is a good correlation between antioxidant activities and TP/TF content. But in HPLC methods, EAE contained the highest content of lutin and gallic acid, which decreased in the same order of EAE > BE > PE, the rank order of TP/TF content of EAE and BE were different according to antioxidant ability. The overall results showed that the EAE and BE were richer in phenolics and flavonoids than petroleum ether extract (PE, and may represent a good source of antioxidants. This study has determined oil, fatty acid (FA) and phytosterols content during the ripening of the Tunisian Onopordum acanthium L. seeds. In total, nine FAs and six phytosterols were identified. The main FAs were linoleic acid (0.18-8.06 mg/g of seed) followed by oleic acid (0.051-2.45 mg/g of seed), palmitic acid and stearic acid. Pentadecanoic acid was detected, for the first time, in unripe fruits and the two last stages of development were characterised by a relative abundance of erucic acid. Overall, β-sitosterol (34.5-77.79% of total sterols) was the major 4-desmethylsterols during maturation. The first episodes of growth were characterised by the best amounts of stigmasterol and campesterol, while stigmastanol and Δ7 sitosterol had quoted the semi-ripe and fully ripe fruits; however, cholesterol was absent. These findings are useful in understanding a potential new source of important natural compounds (Phytosterols and USFA) found in this fruit and when harvest should be undertaken to optimise desired FA and phytosterols content.

Silver and gold nanoparticles of Jasminum nervosum L. had unique optical properties such as broad absorbance band in the visible region of the electromagnetic spectrum. Characterization of the nanoparticles using UV spectrophotometer, Fourier transform infrared spectroscopy, X-ray diffraction, and transmission electron microscopy confirmed that the particles were silver (AgNPs) and gold (AuNPs) ranging between 4-22 and 2-20 nm with an average particles size of 9.4 and 10 nm, respectively. AgNPs and AuNPs of J. nervosum had high larvicidal activity on the filarial and arboviral vector, Culex quinquefasciatus, than the leaf aqueous extract. Observed lethal concentrations (LC50 and LC95) against the third instar larvae were 57.40 and 144.36 μg/ml for AgNPs and 82.62 and 254.68 μg/ml for AuNPs after 24 h treatment, respectively. The lethal time to kill 50% of C. quinquefasciatus larvae were 2.24 and 4.51 h at 150 μg/ml of AgNPs and AuNPs, respectively, while in the case of aqueous leaf extract of J. nervosum it was 9.44 h at 500 μg/ml (F 2,14 = 397.51, P < 0.0001). The principal component analysis plot presented differential clustering of the aqueous leaf extract, AgNP and AuNPs in relation to lethal dose and lethal time. It is concluded from the present findings that the biosynthesised AgNPs and AuNPs using leaf aqueous extract of J. nervosum could be an environmentally safer nanobiopesticide, and provided potential larvicidal effect on C. quinquefasciatus larvae which could be used for prevention of several dreadful diseases.

Full Text Available Sterna hirundinacea (Lesson, 1831 is a migratory seabird that breeds in the Pacific Coast (from Peru to Chile and along the Atlantic coast of South America from Espírito Santo (Brazil to Terra del Fuego (Argentina. This paper describes the reproductive success of South American Terns on Cardos Island, Florianopolis, Brazil in the breeding seasons of 2003, 2005 and 2006. The colony was formed in mid-May in 2003 and early April in other years, with the total number of nests ranging from 1,852 in 2006 to 2,486 in 2005. Hatching success was estimated at 76.39% in 2006, 62.73% in 2003 and 41.1% in 2005, the lowest value that could be attributed to predation by hawks Caracara plancus, lizards Tupinambis merianae and black vulture Coragyps atratus. The chicks hatched in July in 2003, and in June 2005 and 2006, and fledging success was 50.94%, 35.96 and 53.47% respectively. Cardos Island has been constantly used as a breeding site by South American Terns, and therefore represents an important area for conservation of this species. This success could be attributed to low pressure of Kelp gulls (Larus dominicanus, the main predator of seabirds along the Brazilian coast. Full Text Available Plants and plant extracts play a crucial role in the research into novel antineoplastic agents. Four sesquiterpene lactones, artecanin (1, 3β-chloro-4α,10α-dihydroxy-1α,2α-epoxy-5α,7αH-guaia-11(13-en-12,6α-olide (2, iso-seco-tanapartholide 3-O-methyl ether (3 and 4β,15-dihydro-3-dehydrozaluzanin C (4, were isolated from two traditionally used Asteraceae species (Onopordum acanthium and Artemisia asiatica. When tested for antiproliferative action on HL-60 leukemia cells, these compounds exhibited reasonable IC50 values in the range 3.6–13.5 μM. Treatment with the tested compounds resulted in a cell cycle disturbance characterized by increases in the G1 and G2/M populations, while there was a decrease in the S phase. Additionally, 1–3 elicited increases in the hypodiploid (subG1 population. The compounds elicited concentration-dependent chromatin condensation and disruption of the membrane integrity, as revealed by Hoechst 33258–propidium staining. Treatment for 24 h resulted in significant increases in activity of caspases-3 and -9, indicating that the tested sesquiterpenes induced the mitochondrial pathway of apoptosis. The proapoptotic properties of the sesquiterpene lactones were additionally demonstrated withannexin V staining. Compounds 1 and 2 increased the Bax/Bcl-2 expression and decreased the expressions of CDK1 and cyclin B2, as determined at the mRNA level by means of RT-PCR. These experimental results indicate that sesquiterpene lactones may be regarded as potential starting structures for the development of novel anticancer agents.

Sulfonated cardo poly(arylene ether sulfone)s (SPPA-PES) with various degrees of sulfonation (DS) were prepared by post-sulfonation of synthesized phenolphthalein anilide (PPA; N-phenyl-3,3'-bis(4-hydroxyphenyl)-1-isobenzopyrolidone) poly(arylene ether sulfone)s (PPA-PES) by using concentrated sulfuric acid. PPA-PES copolymers were synthesized by direct polycondensation of PPA with bis-(4-fluorophenyl)-sulfone and 4,4'-sulfonyldiphenol. The DS was varied with different mole ratios of PPA (24, 30, 40, 50 mol.%) in the polymer. The structure of the resulting SPPA-PES copolymers and the different contents of the sulfonated unit were studied by Fourier transform infrared (FT-IR) spectroscopy, {sup 1}H NMR spectroscopy, and thermogravimetric analysis (TGA). Sorption experiments were conducted to observe the interaction of sulfonated polymer with water. The ion exchange capacity (IEC) and proton conductivity of SPPA-PES were evaluated according to the increase of DS. The water uptake (WU) of the resulting SPPA-PES membranes was in the range of 20-72%, compared with 28% for Nafion 211 registered. Native or pretreated biomass from Onopordum nervosum Boiss, has been examined as candidate feedstock for cellulase production by two mutant strain of Trichoderma Ionqibrachiatum QM9414 and Rut C30. Batch cultivation methods were evaluated and compared with previous experiments using ball-milled, crystalline cellulose (Solka floc). Batch cultivation of T. Ionqibrachiatum Rut C30 on 5% (w/v) acid pretreated O. nervosum biomass yielded enzyme productivities and activities comparable to those obtained on Solka floc. However, the overall enzyme production performance was lower than on Solka floc at comparable cellulose concentrations. This fact may be due to the accumulation of pretreated by products and lignin in the ferment. (Author) 40 refs.

Full Text Available We assessed the effects of Cleistocalyx nervosum var paniala, Gynostemma pentaphyllum, Gynura procumbens, Houttuynia cordata, Hyptis suaveolens, Portulaca grandiflora, Phytolacca americana and Tradescantia spathacea on lymphocyte proliferation and the effects of C. nervosum, G. pentaphyllum, H. suaveolens and P. grandiflora on natural killer (NK cells activity. All of the extracts significantly stimulated human lymphocyte proliferative responses at various concentrations depending on each extract. The extracts of C. nervosum and H. suaveolens were significantly enhanced NK cells activity while those of G. pentaphyllum and P. grandiflora did not alter NK cells function. Our results suggested that the extracts of those plants have stimulating activity on human lymphocytes and could be clinically useful for modulating immune functions of the body.

  6. Structural insight into the substrate- and dioxygen-binding manner in the catalytic cycle of rieske nonheme iron oxygenase system, carbazole 1,9a-dioxygenase

Full Text Available Abstract Background Dihydroxylation of tandemly linked aromatic carbons in a cis-configuration, catalyzed by multicomponent oxygenase systems known as Rieske nonheme iron oxygenase systems (ROs, often constitute the initial step of aerobic degradation pathways for various aromatic compounds. Because such RO reactions inherently govern whether downstream degradation processes occur, novel oxygenation mechanisms involving oxygenase components of ROs (RO-Os is of great interest. Despite substantial progress in structural and physicochemical analyses, no consensus exists on the chemical steps in the catalytic cycles of ROs. Thus, determining whether conformational changes at the active site of RO-O occur by substrate and/or oxygen binding is important. Carbazole 1,9a-dioxygenase (CARDO, a RO member consists of catalytic terminal oxygenase (CARDO-O, ferredoxin (CARDO-F, and ferredoxin reductase. We have succeeded in determining the crystal structures of oxidized CARDO-O, oxidized CARDO-F, and both oxidized and reduced forms of the CARDO-O: CARDO-F binary complex. Results In the present study, we determined the crystal structures of the reduced carbazole (CAR-bound, dioxygen-bound, and both CAR- and dioxygen-bound CARDO-O: CARDO-F binary complex structures at 1.95, 1.85, and 2.00 Å resolution. These structures revealed the conformational changes that occur in the catalytic cycle. Structural comparison between complex structures in each step of the catalytic mechanism provides several implications, such as the order of substrate and dioxygen bindings, the iron-dioxygen species likely being Fe(III-(hydroperoxo, and the creation of room for dioxygen binding and the promotion of dioxygen binding in desirable fashion by preceding substrate binding. Conclusions The RO catalytic mechanism is proposed as follows: When the Rieske cluster is reduced, substrate binding induces several conformational changes (e.g., movements of the nonheme iron and the ligand

Full Text Available In this study, the total lipid content and sterol compositions were determined during the development of milk thistle seeds. The oil content increased to a maximum value of 36±1.7% and then declined to reach a value of 30.5±0.9% at full maturity. The sterol content of milk thistle seeds was affected by the ripening degree of the seeds. At the early stages of seed maturation, Δ7-stigmastenol was the most abundant sterol followed by β-sitosterol. However, at full maturity, β-sitosterol was the most predominant sterol (46.50±0.8%. As the seed developed, campesterol and stigmasterol amounts increased, while Δ7-avenasterol content decreased. It can be concluded that milk thistle seed oil has a characteristic sterol pattern comparable to the ones elucidated for olive oil and corn oil. The extracted oil from milk thistle seeds is rich in phytosterols and could be used in food preparation and human nutrition. In this study, a cardo poly(aryl ether ketone) ultrafiltration membrane containing an N-chloramine functional group (PEK-N-Cl membrane) was easily obtained via exposure of a cardo poly(aryl ether ketone) ultrafiltration membrane (PEK-NH membrane) to dilute sodium hypochlorite solution. The chlorination process did not harm membrane performance. In addition, the PEK-N-Cl membrane was stable in both air and water. The PEK-N-Cl membrane exhibited excellent antimicrobial properties against both Gram-negative and Gram-positive bacteria (i.e. E. coli and Bacillus subtilis, respectively). The PEK-N-Cl membrane provided 94.2% and 100% reduction of E. coli and Bacillus subtilis, respectively, within 30min of contact times. Moreover, nearly 100% of the E. coli was killed after 2h during the filtration process for the PEK-N-Cl membrane. In addition, the water flux decreased by 42% for the PEK-N-Cl membrane compared to 77.6% for the PEK-NH membrane after filtration of the E. coli solution and incubation on LB nutrient agar plate, indicating that the PEK-N-Cl membrane enhibits antifouling. Furthermore, the PEK-N-Cl membrane is recyclable via subsequent exposure to a sodium hypochlorite solution.

  18. Engineering single-molecule, nanoscale, and microscale bio-functional materials via click chemistry (United States)

    Daniele, Michael Angelo-Anthony

To expand the design envelope and supplement the materials library available to biomaterials scientists, the copper(I)-catalyzed azide-alkyne cycloaddition (CuCAAC) was explored as a route to design, synthesize and characterize bio-functional small-molecules, nanoparticles, and microfibers. In each engineered system, the use of click chemistry provided facile, bio-orthogonal control for materials synthesis; moreover, the results provided a methodology and more complete, fundamental understanding of the use of click chemistry as a tool for the synergy of biotechnology, polymer and materials science. Fluorophores with well-defined photophysical characteristics (ranging from UV to NIR fluorescence) were used as building blocks for small-molecule, fluorescent biosensors. Fluorophores were paired to exhibit fluorescence resonant energy transfer (FRET) and used to probe the metabolic activity of carbazole 1,9a-dioxygenase (CARDO). The FRET pair exhibited a significant variation in PL response with exposure to the lysate of Pseudomonas resinovorans CA10, an organism which can degrade variants of both the donor and acceptor fluorophores. Nanoparticle systems were modified via CuCAAC chemistry to carry affinity tags for CARDO and were subsequently utilized for affinity based bioseparation of CARDO from crude cell lysate. The enzymes were baited with an azide-modified carbazolyl-moiety attached to a poly(propargyl acrylate) nanoparticle. Magnetic nanocluster systems were also modified via CuCAAC chemistry to carry fluorescent imaging tags. The iron-oxide nanoclusters were coated with poly(acrylic acid-co-propargyl acrylate) to provide a clickable surface. Ultimately, alternate Cu-free click chemistries were utilized to produce biohybrid microfibers. The biohybrid microfibers were synthesized under benign photopolymerization conditions inside a microchannel, allowing the encapsulation of viable bacteria. Full Text Available BACKGROUND: Carbazole is a recalcitrant compound with a dioxin-like structure and possesses mutagenic and toxic activities. Bacteria respond to a xenobiotic by recruiting exogenous genes to establish a pathway to degrade the xenobiotic, which is necessary for their adaptation and survival. Usually, this process is mediated by mobile genetic elements such as plasmids, transposons, and insertion sequences. FINDINGS: The genes encoding the enzymes responsible for the degradation of carbazole to catechol via anthranilate were cloned, sequenced, and characterized from a carbazole-degrading Sphingomonas sp. strain XLDN2-5. The car gene cluster (carRAaBaBbCAc and fdr gene were accompanied on both sides by two copies of IS6100 elements, and organized as IS6100::ISSsp1-ORF1-carRAaBaBbCAc-ORF8-IS6100-fdr-IS6100. Carbazole was converted by carbazole 1,9a-dioxygenase (CARDO, CarAaAcFdr, meta-cleavage enzyme (CarBaBb, and hydrolase (CarC to anthranilate and 2-hydroxypenta-2,4-dienoate. The fdr gene encoded a novel ferredoxin reductase whose absence resulted in lower transformation activity of carbazole by CarAa and CarAc. The ant gene cluster (antRAcAdAbAa which was involved in the conversion of anthranilate to catechol was also sandwiched between two IS6100 elements as IS6100-antRAcAdAbAa-IS6100. Anthranilate 1,2-dioxygenase (ANTDO was composed of a reductase (AntAa, a ferredoxin (AntAb, and a two-subunit terminal oxygenase (AntAcAd. Reverse transcription-PCR results suggested that carAaBaBbCAc gene cluster, fdr, and antRAcAdAbAa gene cluster were induced when strain XLDN2-5 was exposed to carbazole. Expression of both CARDO and ANTDO in Escherichia coli required the presence of the natural reductases for full enzymatic activity. CONCLUSIONS/SIGNIFICANCE: We predict that IS6100 might play an important role in the establishment of carbazole-degrading pathway, which endows the host to adapt to novel compounds in the environment. The organization of the car

  5. Cascading effects of defaunation on the coexistence of two specialized insect seed predators. (United States)

Peguero, Guille; Muller-Landau, Helene C; Jansen, Patrick A; Wright, S Joseph

Identification of the mechanisms enabling stable coexistence of species with similar resource requirements is a central challenge in ecology. Such coexistence can be facilitated by species at higher trophic levels through complex multi-trophic interactions, a mechanism that could be compromised by ongoing defaunation. We investigated cascading effects of defaunation on Pachymerus cardo and Speciomerus giganteus, the specialized insect seed predators of the Neotropical palm Attalea butyracea, testing the hypothesis that vertebrate frugivores and granivores facilitate their coexistence. Laboratory experiments showed that the two seed parasitoid species differed strongly in their reproductive ecology. Pachymerus produced many small eggs that it deposited exclusively on the fruit exocarp (exterior). Speciomerus produced few large eggs that it deposited exclusively on the endocarp, which is normally exposed only after a vertebrate handles the fruit. When eggs of the two species were deposited on the same fruit, Pachymerus triumphed only when it had a long head start, and the loser always succumbed to intraguild predation. We collected field data on the fates of 6569 Attalea seeds across sites in central Panama with contrasting degrees of defaunation and wide variation in the abundance of vertebrate frugivores and granivores. Speciomerus dominated where vertebrate communities were intact, whereas Pachymerus dominated in defaunated sites. Variation in the relative abundance of Speciomerus across all 84 sampling sites was strongly positively related to the proportion of seeds attacked by rodents, an indicator of local vertebrate abundance.

In a number of recent articles in this journal F. Cardone and collaborators have claimed the observation of several striking nuclear phenomena which they attribute to "piezonuclear reactions". One such claim [Phys. Lett. A 373 (2009) 1956] is that subjecting a solution of 228Th to cavitation leads to a "transformation" of thorium nuclei that is 104 times faster than the normal nuclear decay for this isotope. In a "Comment" [Phys. Lett. A 373 (2009) 3795] to the thorium work, we have criticized the evidence provided for this claim. In a "Reply" [Phys. Lett. A 373 (2009) 3797] Cardone et al. answer only some minor points but avoid addressing the real issue. The information provided in their Reply displays a worrying lack of control of their experimental situation and the data they put forward as evidence for their claims. We point out several shortcomings and errors in the described experimental preparations, set-up and reporting, as well as in the data analysis. We conclude that the evidence presented by Cardo...

  7. At the Mercy of Gaia: Deep Ecologial Unrest and America's fall as Nature's Nation in Kingdom of the Spiders

    Jacob Lillemose

Full Text Available This paper looks as the animal horror genre as a way to discuss current notions of ecology in relation to a specific American idea of being "Nature's Nation". The central work for the discussion is the movie Kingdom of the Spiders (1977 by John Cardos, which depicts how a small Arizona town is taken over by a "swarm" of tarantulas. Without any obvious explanation the spiders slowly but steadily invade the town and start killing both other animals and humans until they have completely covered the town in their web. The paper connects the movie to a long tradition of fiction describing how nature turns on humans and reverses the power relation be-tween man and nature that is fundamental to modernity. Moreover, the paper connects the movie to Maurice Maeterlincks ideas of swarm communities as mani-fested by ants and termites to argue that these communities are ecologically superior to the the communities of man-made civilisation. Finally, the paper discusses Kingdom of the Spiders and animal horror in general in relation to recent ideas of non-human ecologies and critiques of anthropocentrism and makes the point that these works of fiction serve as both dramatic and philosophical visions of a world without humans.

Full Text Available ABSTRACT Seed predation is a natural phenomenon that can occur either before or after dispersal and can significantly reduce the economic value and reproductive potential of plants. The babassu palm (Attalea vitrivir, Arecaceae is important to rural communities that extract oil from its fruits for a wide variety of uses. We evaluated the predation and germination of A. vitrivirseeds in Pandeiros River Environmental Protection Area (EPA-Pandeiros in Minas Gerais State, Brazil. Sixty individual plants were evaluated to determine their fruiting patterns. Seed predation and germination were evaluated in the natural environment for eight months for fruits divided into two treatments: scarified and intact. Germination of fruits submitted to these same treatments was also evaluated under greenhouse conditions. Our results indicated that fruiting is continuous in this species and that fruit morphology does not influence either germination or predation. Likewise, fruit scarification did not influence seed germination. Pachymerus cardo (Coleoptera: Chrysomelidae preferentially oviposited on scarified fruits, but only after their dispersal. The predation rate in the natural environment was 14.6%. Germination was not observed under natural conditions, but reached 33.05% under greenhouse conditions.

Full Text Available In this paper I would like to highlight the signifi cant architectural regularity which characterizes the pre-Roman archeological remains registered in Carthage. Despite having been repeatedly interpreted from a preconceived logical and spatial point of view, these remains allow us to set out that the Tyrian foundation was established in the Carthaginian peninsula according to a model that is referred to as per strigas in archaeological literature because it was documented better at the Archaic Greek colonies in Sicily. To defend this idea, I will indicate which architectonic remains make up its material basic, paying special attention to the remains found underneath the crossroads of the decumanus maximus and the cardo x-east, and I will try to summarize the state of the current issue on the urban shape of Phoenician Carthage. I will then go over several documentated examples of orthogonal land division in urban spaces from the Ancient Near East. This information will provide insight on the possible technical and practical context of Phoenician societies in the 9th and 8th centuries BC and, more specifically, the socio-political context of the Carthaginian founders. Finally, in light of these elements, I will put forward a proposal of a reconstruction of the urban layout of Phoenician Carthage as a whole.

  11. Synthesis and characterization of polymers for light waveguide applications (United States)

    Li, Bo

The overall goal of this research was to prepare organo- soluble polymers that display low birefringence, low optical loss, and high thermal stability for use in light waveguide devices. Thus, two series of thermally stable polymers, i.e., aromatic polyimides and perfluorocyclobutane-containing poly(aryl ethers), were synthesized and characterized. The approach to the aromatic polyimides involved the synthesis of a new series of 4,4'-(9- fluorenylidene)dianilines containing large polarizable substituents. The diamines were polymerized with 2,2'-bis[4-(3,4- dicarboxyphenoxy)phenyl]propane dianhydride (BisA-DA) and 2,2'-bis(3,4- dicarboxyphenyl)hexafluoropropane dianhydride (6FDA) in refluxing m-cresol containing a catalytic amount of isoquinoline. The polyimides were soluble in common organic solvents such as N-methyl-2- pyrrolidinone (NMP), tetrahydrofuran (THF), chloroform (CHCl3), and cyclopentanone and could be solution cast into tough films. Thin films of the new cardo polyimides displayed birefringences that ranged from -0.0042 to 0.0074. In the second part of this work, a series of new difunctional and trifunctional trifluorovinylether-containing monomers was synthesized and polymerized via the thermal cyclodimerization of the vinyl groups. The number of carbon-hydrogen bonds was minimized in these systems in order to minimize the optical loss. A tough, transparent film was made from a polymer prepared from a trifunctional monomer, 1,1,1- trifluoro-2,2,2-tris(4-trifluorovinyloxyphenyl)ethane, which had a birefringence of 0.0008. The film showed negligible absorption at 1550 nm in the near-IR region. The polymer was also very thermally stable. Most of the linear polymers were soluble in common organic solvents such as NMP, THF, chloroform and cyclopentanone and could be solution cast into thin films, which displayed birefringences that ranged from -0.0005 to 0.0048.

Full Text Available The aim of this study was to model light interception and distribution in the mixed canopy of Common cocklebur (Xanthium stramarium with corn. An experiment was conducted in factorial arrangement on the basis of randomized complete blocks design with three replications in Gonabad in 2006-2007 and 2007-2008 seasons. The factors used in this experiment include corn density of 7.5, 8.5 and 9.5 plants per meter of row and density of Common cocklebur of zero, 2, 4, 6 and 8 plants per meter of row. INTERCOM model was used through replacing parabolic function with triangular function of leaf area density. Vertical distribution of the species' leaf area showed that corn has concentrated the most leaf area in layer of 80 to 100 cm while Common cocklebur has concentrated in 35-50 cm of canopy height. Model sensitivity analysis showed that leaf area index, species' height, height where maximum leaf area is seen (hm, and extinction coefficient have influence on light interception rate of any species. In both species, the distribution density of leaf area at the canopy length fit a triangular function, and the height in which maximum leaf area was observed was changed by change in density. There was a correlation between percentage of the radiation absorbed by the weed and percentage of corn seed yield loss (r² = 0.89. Ideal type of corn was determined until the stage of tasseling in competition with weed. This determination indicates that the corn needs more height and leaf area, as well as less extinction coefficient to successfully fight against the weed.O objetivo deste estudo foi desenvolver um modelo de intercepção de luz e distribuição na copa mista do cardo comum (Xanthium stramarium com o milho.

  15. Warionia (Asteraceae: a relict genus of Cichorieae?

Directory of Open Access Journals (Sweden)

    Katinas, Liliana

Full Text Available The genus Warionia, with its only species W. saharae, is endemic to the northwestern edge of the African Sahara desert. This is a somewhat thistle-like aromatic plant, with white latex, and fleshy, pinnately- partite leaves. Warionia is in many respects so different from any other genus of Asteraceae, that it has been tentatively placed in the tribes Cardueae, Cichorieae, Gundelieae, and Mutisieae. Until now, a comprehensive study of Warionia to have a complete context for discussing its taxonomic position is lacking. The general morphology, anatomy, palynology and chromosome number of W. saharae are investigated here, and the species is described and illustrated. Laticifers in leaves and stems indicate a relationship with Cichorieae, and are associated with the phloem, in contact with it or with the surrounding sclerenchyma sheath. The pollen features indicate a strong relation with Cardueae, namely the structure with Anthemoid pattern where the columellae are joined to the foot layer, the ectosexine with thin columellae, the endosexine with stout and ramified columellae, the conspicuous spines with globose bases and conspicuous apical channels, and the tectum surface very perforate. Chromosomal counts resulted in 2n = 34. The morphological and palynological evidence positions Warionia between the tribes Cardueae and Cichorieae suggesting that it could be a remnant of the ancestral stock that gave rise to both tribes.

  16. Carnaria: The Festival of the Goddess Carna

    Marjeta Šašel Kos

hinges has been questioned by many scholars, believing that the poet had confounded her with Cardea (cardo is the Latin for ‘door hinge’. To all appearances, however, no such goddess as Cardea had ever existed (a conclusion partly accepted by the scholarship, and Ovid’s lines offer no proof that Carna might not have been the goddess of hinges as well. Te idea could hardly have been Ovid’s own invention: what is more likely is that his contemporaries actually worshipped Carna also as a patron of doors, an office agreeing with her other characteristics. Her name, however, probably derives from caro, carnis, ‘flesh’, as an early etymology has claimed. Carna was an ancient Roman goddess: ‘Time has dimmed the tradition which sets forth how she acquired the powers she owns, but you shall learn it from my song’ (103–104. According to Ovid, the grove of Alernus near the Tiber, still held sacred in his own time, was the birthplace of a nymph, Cranaë. She managed to evade all suitors until she was caught by Janus, from whom she could not hide because he had eyes in the back of his head. In return for her maidenhood, he invested her with power over the hinges and gave her a twig of whitethorn (spina alba, which repels harm from doors. Particularly dangerous were nocturnal birds of prey, the striges which attacked nurseless infants in their cradles at night, rent their innards and sucked their blood. On one occasion, Carna thus rescued the five-day-old Proca (the future great-grandfather of Romulus and king in Alba Longa by touching the doorposts and threshold three times with a whitethorn twig, sprinkling the entrance with healing water, and sacrificing the innards of a suckling pig just two months old. Ovid begins by naming Carna and the Calends sacred to her and continues by using her ‘old’ name, Cranaë, but reverts to the name ‘Carna’ after the Proca episode, saying: ‘You ask why fat bacon is eaten on these [C]alends, and why beans are mixed with
