    Two vulnerability analysis techniques, Diversion Path Analysis (DPA) and Matrix Analysis of the Insider Threat (MAIT), were applied by EG and G Idaho, Inc. Safeguards and Security to the same item accountable SNM storage area at INEL. Technical and cost data for each methodology were collected and compared. A recommendation that MAIT be utilized for future vulnerability analyses of item accountable SNM storage and use areas operated by EG and G Idaho for DOE-ID resulted. Unclassified results of the two techniques and MAIT/DPA technical and cost comparisons will be presented which show that MAIT can be used for vulnerability analyses to comply with Department of Energy (DOE) requirements

    Full Text Available Mucosal-associated invariant T (MAIT cells display two evolutionarily conserved features: an invariant T cell receptor (TCRalpha (iTCRalpha chain and restriction by the nonpolymorphic class Ib major histocompatibility complex (MHC molecule, MHC-related molecule 1 (MR1. MR1 expression on thymus epithelial cells is not necessary for MAIT cell development but their accumulation in the gut requires MR1 expressing B cells and commensal flora. MAIT cell development is poorly known, as these cells have not been found in the thymus so far. Herein, complementary human and mouse experiments using an anti-humanValpha7.2 antibody and MAIT cell-specific iTCRalpha and TCRbeta transgenic mice in different genetic backgrounds show that MAIT cell development is a stepwise process, with an intra-thymic selection followed by peripheral expansion. Mouse MAIT cells are selected in an MR1-dependent manner both in fetal thymic organ culture and in double iTCRalpha and TCRbeta transgenic RAG knockout mice. In the latter mice, MAIT cells do not expand in the periphery unless B cells are added back by adoptive transfer, showing that B cells are not required for the initial thymic selection step but for the peripheral accumulation. In humans, contrary to natural killer T (NKT cells, MAIT cells display a naïve phenotype in the thymus as well as in cord blood where they are in low numbers. After birth, MAIT cells acquire a memory phenotype and expand dramatically, up to 1%-4% of blood T cells. Finally, in contrast with NKT cells, human MAIT cell development is independent of the molecular adaptor SAP. Interestingly, mouse MAIT cells display a naïve phenotype and do not express the ZBTB16 transcription factor, which, in contrast, is expressed by NKT cells and the memory human MAIT cells found in the periphery after birth.     Full Text Available Dengue virus (DENV and Zika virus (ZIKV are members of the Flaviviridae and are predominantly transmitted via mosquito bites. Both viruses are responsible for a growing number of infections in tropical and subtropical regions. DENV infection can cause lethargy with severe morbidity and dengue shock syndrome leading to death in some cases. ZIKV is now linked with Guillain-Barré syndrome and fetal malformations including microcephaly and developmental disorders (congenital Zika syndrome. The protective and pathogenic roles played by the immune response in these infections is unknown. Mucosal-associated invariant T (MAIT cells are a population of innate T cells with potent anti-bacterial activity. MAIT cells have also been postulated to play a role in the immune response to viral infections. In this study, we evaluated MAIT cell frequency, phenotype, and function in samples from subjects with acute and convalescent DENV infection. We found that in acute DENV infection, MAIT cells had elevated co-expression of the activation markers CD38 and HLA-DR and had a poor IFNγ response following bacterial stimulation. Furthermore, we found that MAIT cells can produce IFNγ in response to in vitro infection with ZIKV. This MAIT cell response was independent of MR1, but dependent on IL-12 and IL-18. Our results suggest that MAIT cells may play an important role in the immune response to Flavivirus infections.

    HTLV-1 infection is associated with several inflammatory disorders, including the neurodegenerative condition HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP). It is unclear why a minority of infected subjects develop HAM/TSP. The cellular immune response has been implicated in the development of inflammatory alterations in these patients; however the pathogenic mechanisms for disease progression remain unclear. Furthermore, HTLV-1-infected individuals have an increase incidence of Mycobacterium tuberculosis (Mtb) infection, suggesting that immunological defect are associated with HTLV-1 infection. Evidence suggests an important role for Mucosal-associated invariant T (MAIT) cells in the early control of Mtb infection. Chronic viral infections like HIV and HCV have been associated with decreased frequency and functionality of MAIT cells. We hypothesized that HTLV-1 infection is associated with similar perturbations in MAIT cells. We investigated MAIT cell frequency, phenotype, and function by flow cytometry in a cohort of 10 asymptomatic and 10 HAM/TSP HTLV-1 infected patients. We found that MAIT cells from HTLV-1-infected subjects were reduced and showed high co-expression of the activation markers CD38 and HLA-DR but normal levels of CCR6 and CD127. MAIT cells had a lower expression of the transcription factor PLZF in HAM/TSP patients. Unlike Tax-specific CD8+T cells, which are hyperfunctional, MAIT cells from HTLV-1-infected subjects had a poor IFNγ response following antigen stimulation.     Full Text Available Mucosal-associated invariant T (MAIT cells are characterized by the combined expression of the semi-invariant T cell receptor (TCR Vα7.2, the lectin receptor CD161, as well as IL-18R, and play an important role in antibacterial host defense of the gut. The current study characterized CD161(+ MAIT and CD161-TCRVα7.2(+ T cell subsets within a large cohort of HIV patients with emphasis on patients with slow disease progression and elite controllers. Mononuclear cells from blood and lymph node samples as well as plasma from 63 patients and 26 healthy donors were analyzed by multicolor flow cytometry and ELISA for IL-18, sCD14 and sCD163. Additionally, MAIT cells were analyzed after in vitro stimulation with different cytokines and/or fixed E.coli. Reduced numbers of CD161(+ MAIT cells during HIV infection were detectable in the blood and lymph nodes of all patient groups, including elite controllers. CD161+ MAIT cell numbers did not recover even after successful antiretroviral treatment. The loss of CD161(+ MAIT cells was correlated with higher levels of MAIT cell activation; an increased frequency of the CD161-TCRVα7.2(+T cell subset in HIV infection was observed. In vitro stimulation of MAIT cells with IL-18 and IL-12, IL-7 and fixed E.coli also resulted in a rapid and additive reduction of the MAIT cell frequency defined by CD161, IL-18R and CCR6. In summary, the irreversible reduction of the CD161(+ MAIT cell subset seems to be an early event in HIV infection that is independent of later stages of the disease. This loss appears to be at least partially due to the distinctive vulnerability of MAIT cells to the pronounced stimulation by microbial products and cytokines during HIV-infection.

  14. MAIT, MR1, microbes and riboflavin: a paradigm for the co-evolution of invariant TCRs and restricting MHCI-like molecules?

    MAIT cells express an invariant TCR that recognizes non-peptidic microbial antigens presented by the non-polymorphic MHCI-like molecule, MR1. We briefly describe how the antigens recognized by MAIT cells are generated from an unstable precursor of the riboflavin (Vitamin B2) biosynthesis pathway, as well as the main features of MAIT cells in comparison with other related T cell subsets. In silico analysis of bacterial genomes shows that the riboflavin biosynthesis pathway is highly prevalent in all groups of Prokaryotes with, however, notable exceptions. We discuss the putative functions and the evolution of the MAIT/MR1 couple: it appeared in the ancestors of mammals and is highly conserved across this group, but was independently lost in three orders. We describe the four instances of known invariant TCR and MHC-I-like molecules encountered in Vertebrates. Both T cells bearing semi-invariant TCR and the associated, evolutionarily conserved MHC-I related molecules have been found in mammals or in amphibians, which suggests that other MHC1-like/invariant TCR couples might be present in other classes of Vertebrates to detect generic microbial compounds. This allows us to discuss how the recognition of riboflavin precursor derivatives by the MAIT TCR may be a way to detect invasive microbes in specific organs, and may epitomize other invariant T cell systems across vertebrates.

    Full Text Available Superantigens (SAgs are potent exotoxins secreted by Staphylococcus aureus and Streptococcus pyogenes. They target a large fraction of T cell pools to set in motion a "cytokine storm" with severe and sometimes life-threatening consequences typically encountered in toxic shock syndrome (TSS. Given the rapidity with which TSS develops, designing timely and truly targeted therapies for this syndrome requires identification of key mediators of the cytokine storm's initial wave. Equally important, early host responses to SAgs can be accompanied or followed by a state of immunosuppression, which in turn jeopardizes the host's ability to combat and clear infections. Unlike in mouse models, the mechanisms underlying SAg-associated immunosuppression in humans are ill-defined. In this work, we have identified a population of innate-like T cells, called mucosa-associated invariant T (MAIT cells, as the most powerful source of pro-inflammatory cytokines after exposure to SAgs. We have utilized primary human peripheral blood and hepatic mononuclear cells, mouse MAIT hybridoma lines, HLA-DR4-transgenic mice, MAIThighHLA-DR4+ bone marrow chimeras, and humanized NOD-scid IL-2Rγnull mice to demonstrate for the first time that: i mouse and human MAIT cells are hyperresponsive to SAgs, typified by staphylococcal enterotoxin B (SEB; ii the human MAIT cell response to SEB is rapid and far greater in magnitude than that launched by unfractionated conventional T, invariant natural killer T (iNKT or γδ T cells, and is characterized by production of interferon (IFN-γ, tumor necrosis factor (TNF-α and interleukin (IL-2, but not IL-17A; iii high-affinity MHC class II interaction with SAgs, but not MHC-related protein 1 (MR1 participation, is required for MAIT cell activation; iv MAIT cell responses to SEB can occur in a T cell receptor (TCR Vβ-specific manner but are largely contributed by IL-12 and IL-18; v as MAIT cells are primed by SAgs, they also begin to

    Mucosal-associated invariant T (MAIT) cells are a T-cell subset that expresses a conserved TRAV1-2 (Vα7.2) T-cell receptor (TCR) chain and the surface marker CD161. They are involved in the defence against microbes as they recognise small organic molecules of microbial origin that are presented by the non-classical MHC molecule 1 (MR1). MAIT cells express a semi-restricted TCR α chain with TRAV1-2 preferentially linked to TRAJ33, TRAJ12, or TRAJ20 which pairs with a limited set of β chains. To investigate the TCR repertoire of human CD161(hi)TRAV1-2(+) T cells in depth we analysed the α and β chains of this T-cell subset by next generation sequencing. Concomitantly we analysed 132 paired α and β chains from single cells to assess the αβ pairing preferences. We found that the CD161(hi)TRAV1-2(+) TCR repertoire in addition to the typical MAIT TCRs further contains polyclonal elements reminiscent of classical αβ T cells.     Mucosal-associated invariant T (MAIT) cells contribute to protection against certain microorganism infections. The aims of this study were to examine the levels of MAIT cells in pulmonary tuberculosis (TB) and nontuberculous mycobacteria (NTM) lung disease patients, to evaluate the clinical relevance of MAIT cell levels, and to investigate the functions of MAIT cells. Patients with pulmonary TB (n = 35), NTM (n = 29), and healthy controls (n = 75) were enrolled in the study. MAIT cell levels and functions were measured by flow cytometry. Circluating MAIT cell levels were found to be reduced in TB and NTM patients. MAIT cell deficiency reflects a variety of clinical conditions. In particular, MAIT cell numbers were significantly correlated with sputum AFB positivity, extent of disease, hemoglobin levels, lymphocyte counts, CRP and ESR levels. MAIT cells in TB patients failed to produce interferon-γ irrespective of the mode of stimulation, whereas NTM patients displayed a defect in MR1-dependent signaling pathway. Notably, an elevated expression of programmed death-1 was also associated with MAIT cell deficiency in TB.     Mucosal-associated invariant T (MAIT) cells contribute to protection against certain microorganism infections. However, little is known about the role of MAIT cells in Orientia tsutsugamushi infection. Hence, the aims of this study were to examine the level and function of MAIT cells in patients with scrub typhus and to evaluate the clinical relevance of MAIT cell levels. Thirty-eight patients with scrub typhus and 53 health control subjects were enrolled in the study. The patients were further divided into subgroups according to disease severity. MAIT cell level and function in the peripheral blood were measured by flow cytometry. Circulating MAIT cell levels were found to be significantly reduced in scrub typhus patients. MAIT cell deficiency reflects a variety of clinical conditions. In particular, MAT cell levels reflect disease severity. MAIT cells in scrub typhus patients displayed impaired tumor necrosis factor (TNF)-α production, which was restored during the remission phase. In addition, the impaired production of TNF-α by MAIT cells was associated with elevated CD69 expression. This study shows that circulating MAIT cells are activated, numerically deficient, and functionally impaired in TNF-α production in patients with scrub typhus. These abnormalities possibly contribute to immune system dysregulation in scrub typhus infection.

  6. Maximal exercise increases mucosal associated invariant T cell frequency and number in healthy young men.

    Mucosal associated invariant T (MAIT) cells have properties of the innate and acquired immune systems. While the response to vigorous exercise has been established for most leukocytes, MAIT cells have not been investigated. Therefore, the purpose was to determine if MAIT cell lymphocytosis occurs with acute maximal aerobic exercise and if this response is influenced by exercise duration, cardiovascular fitness, or body composition. Twenty healthy young males with moderate fitness levels performed an extended graded exercise test until volitional fatigue. Peripheral blood mononuclear cells were isolated from venous blood obtained prior and immediately after exercise and were labeled to identify specific T cell populations using flow cytometry. The percentage of MAIT cells relative to total T cells significantly increased from 3.0 to 3.8% and absolute MAIT cell counts increased by 2.2-fold following maximal exercise. MAIT cell subpopulation proportions were unchanged with exercise. Within cytotoxic T lymphocytes (CTL), MAIT cells consisted of 8% of these cells and this remained constant after exercise. MAIT cell counts and changes with exercise were not affected by body composition, VO 2peak , or exercise duration. Maximal exercise doubled MAIT cell numbers and showed preferential mobilization within total T cells but the response was not influenced by fitness levels, exercise duration, or body composition. These results suggest that acute exercise could be used to offset MAIT cell deficiencies observed with certain pathologies. MAIT cells also make up a substantial proportion of CTLs, which may have implications for cytotoxicity assays using these cells.

    Full Text Available Mucosal-associated invariant T (MAIT cells, defined as CD161++TCR iVα7.2+ T cells, play an important role in the innate defense against bacterial infections, and their functionality is impaired in chronic viral infections. Here, we investigated the frequency and functional role of MAIT cells in chronic hepatitis B virus (HBV infection. The peripheral CD3+CD161++TCR iVα7.2+ MAIT cells in chronic HBV-infected patients and healthy controls were phenotypically characterized based on CD57, PD-1, TIM-3, and CTLA-4, as well as HLA-DR and CD38 expression. The frequency of MAIT cells was significantly decreased among chronic HBV-infected individuals as compared to controls. Expression of CD57, PD-1, CTLA-4, as well as HLA-DR and CD38 on MAIT cells was significantly elevated in chronic HBV-infected individuals relative to controls. The percentage of T cell receptor (TCR iVα7.2+ CD161+ MAIT cells did not correlate with HBV viral load but inversely with HLA-DR on CD4+ T cells and MAIT cells and with CD57 on CD8+ T cells suggesting that decrease of MAIT cells may not be attributed to direct infection by HBV but driven by HBV-induced chronic immune activation. The percentage and expression levels of PD-1 as well as CTLA-4 on MAIT cells inversely correlated with plasma HBV-DNA levels, which may suggest either a role for MAIT cells in the control of HBV infection or the effect of HBV replication in the liver on MAIT cell phenotype. We report that decrease of TCR iVα7.2+ MAIT cells in the peripheral blood and their functions were seemingly impaired in chronic HBV-infected patients likely because of the increased expression of PD-1.

    Full Text Available Mucosal-associated invariant T cells (MAITs are innate-like T cells that play a pivotal role in the host defense against infectious diseases, and are also implicated in autoimmune diseases, metabolic diseases, and cancer. Recent studies have shown that induced pluripotent stem cells (iPSCs derived from MAITs selectively redifferentiate into MAITs without altering their antigen specificity. Such a selective differentiation is a prerequisite for the use of MAITs in cell therapy and/or regenerative medicine. However, the molecular mechanisms underlying this phenomenon remain unclear. Here, we performed methylome and transcriptome analyses of MAITs during the course of differentiation from iPSCs. Our multi-omics analyses revealed that recombination-activating genes (RAG1 and RAG2 and DNA nucleotidylexotransferase (DNTT were highly methylated with their expression being repressed throughout differentiation. Since these genes are essential for V(DJ recombination of the T cell receptor (TCR locus, this indicates that nascent MAITs are kept from further rearrangement that may alter their antigen specificity. Importantly, we found that the repression of RAGs was assured in two layers: one by the modulation of transcription factors for RAGs, and the other by DNA methylation at the RAG loci. Together, our study provides a possible explanation for the unaltered antigen specificity in the selective differentiation of MAITs from iPSCs.

    Full Text Available Mucosal-associated invariant T (MAIT cells, a novel subpopulation of innate-like T cells that express an invariant T cell receptor (TCRα chain and a diverse TCRβ chain, can recognize a distinct set of small molecules, vitamin B metabolites, derived from some bacteria, fungi but not viruses, in the context of an evolutionarily conserved major histocompatibility complex-related molecule 1 (MR1. This implies that MAIT cells may play unique and important roles in host immunity. Although viral antigens are not recognized by this limited TCR repertoire, MAIT cells are known to be activated in a TCR-independent mechanism during some viral infections, such as hepatitis C virus and influenza virus. In this article, we will review recent works in MAIT cell antigen recognition, activation and the role MAIT cells may play in the process of bacterial and viral infections and pathogenesis of non-infectious diseases.

    Full Text Available Mucosal-associated invariant T (MAIT cells are unconventional CD3+CD161high T lymphocytes that recognize vitamin B2 (riboflavin biosynthesis precursor derivatives presented by the MHC-I related protein, MR1. In humans, their T cell receptor is composed of a Vα7.2-Jα33/20/12 chain, combined with a restricted set of Vβ chains. MAIT cells are very abundant in the liver (up to 40% of resident T cells and in mucosal tissues, such as the lung and gut. In adult peripheral blood, they represent up to 10% of circulating T cells, whereas they are very few in cord blood. This large number of MAIT cells in the adult likely results from their gradual expansion with age following repeated encounters with riboflavin-producing microbes. Upon recognition of MR1 ligands, MAIT cells have the capacity to rapidly eliminate bacterially infected cells through the production of inflammatory cytokines (IFNγ, TNFα, and IL-17 and cytotoxic effector molecules (perforin and granzyme B. Thus, MAIT cells may play a crucial role in antimicrobial defense, in particular at mucosal sites. In addition, MAIT cells have been implicated in diseases of non-microbial etiology, including autoimmunity and other inflammatory diseases. Although their participation in various clinical settings has received increased attention in adults, data in children are scarce. Due to their innate-like characteristics, MAIT cells might be particularly important to control microbial infections in the young age, when long-term protective adaptive immunity is not fully developed. Herein, we review the data showing how MAIT cells may control microbial infections and how they discriminate pathogens from commensals, with a focus on models relevant for childhood infections.

    Mucosa-associated invariant T (MAIT) cells are a subset of innate-like T lymphocytes known for their ability to respond to MHC-related protein 1 (MR1)-restricted stimuli and select cytokine signals. They are abundant in humans and especially enriched in mucosal layers, common sites of neoplastic transformation. MAIT cells have been found within primary and metastatic tumors. However, whether they promote malignancy or contribute to anticancer immunity is unclear. On the one hand, MAIT cells produce IL-17A in certain locations and under certain circumstances, which could in turn facilitate neoangiogenesis, intratumoral accumulation of immunosuppressive cell populations, and cancer progression. On the other hand, they can express a potent arsenal of cytotoxic effector molecules, NKG2D and IFN-γ, all of which have established roles in cancer immune surveillance. In this review, we highlight MAIT cells' characteristics as they might pertain to cancer initiation, progression, or control. We discuss recent findings, including our own, that link MAIT cells to cancer, with a focus on colorectal carcinoma, as well as some of the outstanding questions in this active area of research. Finally, we provide a hypothetical picture in which MAIT cells constitute attractive targets in cancer immunotherapy.

  17. Intra-Hepatic Depletion of Mucosal-Associated Invariant T Cells in Hepatitis C Virus-Induced Liver Inflammation.

    Chronic hepatitis affects phenotypes of innate and adaptive immune cells. Mucosal-associated invariant T (MAIT) cells are enriched in the liver as compared with the blood, respond to intra-hepatic cytokines, and (via the semi-invariant T-cell receptor) to bacteria translocated from the gut. Little is known about the role of MAIT cells in livers of patients with chronic hepatitis C virus (HCV) infection and their fate after antiviral therapy. We collected blood samples from 42 patients with chronic HCV infection who achieved a sustained virologic response after 12 weeks of treatment with sofosbuvir and velpatasvir. Mononuclear cells were isolated from blood before treatment, at weeks 4 and 12 during treatment, and 24 weeks after the end of treatment. Liver biopsies were collected from 37 of the patients prior to and at week 4 of treatment. Mononuclear cells from 56 blood donors and 10 livers that were not suitable for transplantation were used as controls. Liver samples were assessed histologically for inflammation and fibrosis. Mononuclear cells from liver and blood were studied by flow cytometry and analyzed for responses to cytokine and bacterial stimulation. The frequency of MAIT cells among T cells was significantly lower in blood and liver samples of patients with HCV infection than of controls (median, 1.31% vs 2.32% for blood samples, P = .0048; and median, 4.34% vs 13.40% for liver samples, P = .001). There was an inverse correlation between the frequency of MAIT cells in the liver and histologically determined levels of liver inflammation (r = -.5437, P = .0006) and fibrosis (r = -.5829, P = .0002). MAIT cells from the liver had higher levels of activation and cytotoxicity than MAIT cells from blood (P liver inflammation and MAIT cell activation and cytotoxicity, and increased the MAIT cell frequency among intra-hepatic but not blood T cells.     Full Text Available Mucosal-associated invariant T (MAIT cells are innate-like T cells abundant in humans that can be activated in a TCR-independent manner by inflammatory and antiviral cytokines. In humans, the capacity for TCR-independent activation is functionally linked to a transcriptional program that can be identified by the expression of the C-type lectin receptor, CD161. In addition to MAIT cells, it has been demonstrated that a subset of γδT cells expresses CD161 and can be activated by TCR-independent cytokine stimulation. In this study, we sought to clarify the nature of cytokine-responsive human γδT cells. We could link CD161 expression on Vδ2+ versus Vδ1+ γδT cells to the observation that Vδ2+ γδT cells, but not Vδ1+ γδT cells, robustly produced IFN-γ upon stimulation with a variety of cytokine combinations. Interestingly, both CD161+ and CD161− Vδ2+ γδT cells responded to these stimuli, with increased functionality within the CD161+ subset. This innate-like responsiveness corresponded to high expression of PLZF and IL-18Rα, analogous to MAIT cells. Vδ2+ γδT cells in human duodenum and liver maintained a CD161+ IL-18Rα+ phenotype and produced IFN-γ in response to IL-12 and IL-18 stimulation. In contrast to MAIT cells, we could not detect IL-17A production but observed higher steady-state expression of Granzyme B by Vδ2+ γδT cells. Finally, we investigated the frequency and functionality of γδT cells in the context of chronic hepatitis C virus infection, as MAIT cells are reduced in frequency in this disease. By contrast, Vδ2+ γδT cells were maintained in frequency and displayed unimpaired IFN-γ production in response to cytokine stimulation. In sum, human Vδ2+ γδT cells are a functionally distinct population of cytokine-responsive innate-like T cells that is abundant in blood and tissues with similarities to human MAIT cells.

    A variety of innate subsets of lymphoid cells such as natural killer (NK) cells, several populations of innate lymphoid cells (ILCs), and mucosal-associated invariant T (MAIT) cells as innate-like T lymphocytes are involved in asthma and may have important effector functions in asthmatic immune responses. In the present study, we investigated whether NK cells, ILCs, and MAIT cells in the peripheral blood of patients with asthma would be associated with clinical asthma parameters. We recruited 75 adult patients with mild to severe asthma. The peripheral blood mononuclear cells in peripheral venous blood samples from the patients were purified and stained with different combinations of appropriate antibodies. The cells were analyzed by flow cytometry. The percentage of activated (i.e., CD69 + ) NK cells in the total NK cell population was negatively correlated with FEV 1 % which is calculated by the forced expiratory volume in 1 s (FEV 1 )/the forced vital capacity (FVC). The percentages of CD69 + ILC1s and ILC2s were negatively correlated with FEV 1 % and %FEV 1 . The percentage of CD69 + ILC3s was positively correlated with BMI, and the percentage of CD69 + MAIT cells was negatively correlated with FEV 1 %. Moreover, the percentage of CD69 + NK cells, ILC1s, ILC2s, ILC3s, and MAIT cells were positively correlated with each other. For the first time, our data showed that activated NK cells, ILC1s, ILC2s, ILC3s, and MAIT cells were positively correlated with each other and may be associated with airflow limitation in patients with asthma. In this study, disease specific biomarkers were sought in blood samples from patients to facilitate objective diagnoses of FMS, and distinguish it from RA and SpA.Peripheral blood mononuclear cells (PBMCs from patients and healthy donors (HD were subjected to multicolor flow cytometric analysis. The percentage of mucosal-associated invariant T (MAIT cells in PBMCs and the mean fluorescent intensity (MFI of cell surface antigen expression in MAIT cells were analyzed.There was a decrease in the MAIT cell population in FMS, RA, and SpA compared with HD.     Full Text Available The maternal part of the placenta, the decidua, consists of maternal immune cells, decidual stromal cells, and extravillous fetal trophoblasts. In a successful pregnancy, these cell compartments interact to provide an intricate balance between fetal tolerance and antimicrobial defense. These processes are still poorly characterized in the two anatomically different decidual tissues, basalis and parietalis. We examined immune cells from decidua basalis and parietalis from term placentas (n=15 with flow cytometry. By using multivariate discriminant analysis, we found a clear separation between the two decidual compartments based on the 81 investigated parameters. Decidua parietalis lymphocytes displayed a more activated phenotype with a higher expression of coinhibitory markers than those isolated from basalis and contained higher frequencies of T regulatory cells. Decidua basalis contained higher proportions of monocytes, B cells, and mucosal-associated invariant T (MAIT cells. The basalis B cells were more immature, and parietalis MAIT cells showed a more activated phenotype. Conventional T cells, NK cells, and MAIT cells from both compartments potently responded with the production of interferon-γ and/or cytotoxic molecules in response to stimulation. To conclude, leukocytes in decidua basalis and parietalis displayed remarkable phenotypic disparities, indicating that the corresponding stromal microenvironments provide different immunoregulatory signals.

    Full Text Available Non-conventional T lymphocytes constitute a special arm of the immune system and act as sentinels against pathogens at mucosal surfaces. These non-conventional T cells (including mucosal-associated invariant T [MAIT] cells, gamma delta [γδ] T cells, and natural killer T [NKT] cells display several innate cell-like features and are rapidly activated by the recognition of conserved, stress-induced, self, and microbial ligands. Here, we review the role of non-conventional T cells during respiratory infections, with a particular focus on the encapsulated extracellular pathogen Streptococcus pneumoniae, the leading cause of bacterial pneumonia worldwide. We consider whether MAIT cells, γδ T cells, and NKT cells might offer opportunities for preventing and/or treating human pneumococcus infections.

    KAERI has been developing the KALIMER-600 core design with a breakeven fissile conversion ratio. The core is loaded with a ternary metallic fuel (TRU-U-10Zr), and the breakeven characteristics are achieved without any blanket assembly. As an alternative plan, a KALIMER-600 burner core design has been also performed. In the early stage of the development of a fast reactor, the main purpose is an economical use of a uranium resource but nowadays in addition to the maximum utilization of a uranium resource, the burning of a high level radioactive waste is taken as an additional interest for the harmony of the environment. In way of constructing the commercial size reactor which has the power level ranging from 800 MWe to 1600 MWe, the demonstration reactor which has the power level ranging from 200 MWe to 600 MWe was usually constructed for the midterm stage to commercial size reactor. In this paper, a 300 MWe burner core design was performed with purpose of demonstration reactor for KALIMER-600 burner of 600 MWe.     In this paper the AM-induced evolution of the design process for small satellites is investigated, leading to the identification of optimal design strategies and the definition of a new MAIT concept. A review of the open literature is presented and some introductory concepts are exposed to highlight the effect of the introduction of AM technologies in the development of new satellites systems. In particular, an innovative structural configuration for the CubeSat class of satellites is proposed, with the ultimate goal of minimizing system complexity via parts reduction and the integration of subsystems through an innovative assembly configuration, as an example to be considered for larger satellites.

    NOMAD, the “Nadir and Occultation for MArs Discovery” spectrometer suite has been selected by ESA and NASA to be part of the payload of the ExoMars Trace Gas Orbiter mission 2016. This instrument suite will conduct a spectroscopic survey of Mars’ atmosphere in the UV, visible and IR regions covering the 0.2-0.65 and 2.2-4.3 μm spectral ranges. NOMAD’s observation modes include solar occultation, nadir and limb observations. The NOMAD instrument is composed of 3 channels: a solar occultation only channel (SO) operating in the infrared wavelength domain, a second infrared channel capable of observing nadir, solar occultation and limb observations (LNO), and an ultraviolet/visible channel (UVIS) that can work in all observation modes. The spectral resolution of SO and LNO surpasses previous surveys in the infrared by more than one order of magnitude. NOMAD offers an integrated instrument combination of a flight-proven concept (SO is a copy of SOIR on Venus Express), and innovations based on existing and proven instrumentation (LNO is based on SOIR/VEX and UVIS has heritage from the ExoMars lander), that will provide mapping and vertical profile information at high spatio-temporal resolution. The three channels have each their own ILS and optical bench, but share the same single interface to the S/C. The NOMAD flight model is due for delivery to ESA in January 2015. We will present results so far of the manufacturing, assembly and especially testing of the various components. The UV CCDs have been characterised in thermal-vacuum; optical fibres have been studied with UV exposure to look at transmission degradation; the IR AOTFs have been tested for their transfer functions; lifetime and vibration testing has been carried out on the flip mirror mechanism. These are all vital inputs to the scientific results from NOMAD.     Innate lymphoid cells (ILCs) are positioned in tissues perinatally, constitutively express receptors responsive to their organ microenvironments, and perform an arsenal of effector functions that overlap those of adaptive CD4 + T cells. Based on knowledge regarding subsets of invariant-like lymphocytes (e.g., natural killer T [NKT] cells, γδ T cells, mucosal-associated invariant T [MAIT] cells, etc.) and fetally derived macrophages, we hypothesize that immune cells established during the perinatal period-including, but not limited to, ILCs-serve intimate roles in tissue that go beyond classical understanding of the immune system in microbial host defense. In this Perspective, we propose mechanisms by which the establishment of ILCs and the tissue lymphoid niche during early development may have consequences much later in life. Although definitive answers require better tools, efforts to achieve deeper understanding of ILC biology across the mammalian lifespan have the potential to lift the veil on the unknown breadth of immune cell functions.     Full Text Available Innate T cells are a heterogeneous group of αβ and γδ T cells that respond rapidly (<2 hours upon activation. These innate T cells also share a non MHC class I or II restriction requirement for antigen recognition. Three major populations within the innate T cell group are recognized, namely Invariant NKT cells (iNKT; Mucosal associated invariant T cells (MAIT and gamma delta T cells. These cells recognize foreign/self-lipid presented by non-classical MHC molecules, such as CD1d, MR1 and CD1a.They are activated during the early stages of bacterial infection and act as a bridge between the innate and adaptive immune systems. In this review we focus on the functional properties of these 3 innate T cell populations and how they are purposed for antimicrobial defense. Furthermore we address the mechanisms through which their effector functions are targeted for bacterial control and compare this in human and murine systems. Lastly we speculate on future roles of these cell types in therapeutic settings such as vaccination.

    Full Text Available The T cell co-receptor CD8αβ enhances T cell sensitivity to antigen, however studies indicate CD8αα has the converse effect and acts as a co-repressor. Using a combination of Thymic Leukaemia antigen (TL tetramer, which directly binds CD8αα, anti-CD161 and anti-Vα7.2 antibodies we have been able for the first time to clearly define CD8αα expression on human CD8 T cells subsets. In healthy controls CD8αα is most highly expressed by CD161 bright (CD161++ mucosal associated invariant T (MAIT cells, with CD8αα expression highly restricted to the TCR Vα7.2+ cells of this subset. We also identified CD8αα-expressing populations within the CD161 mid (CD161+ and negative (CD161- non-MAIT CD8 T cell subsets and show TL-tetramer binding to correlate with expression of CD8β at low levels in the context of maintained CD8α expression (CD8α+CD8βlow. In addition, we found CD161-CD8α+CD8βlow populations to be significantly expanded in the peripheral blood of HIV-1 and hepatitis B (mean of 47% and 40% of CD161- T cells respectively infected individuals. Such CD8αα expressing T cells are an effector-memory population (CD45RA-, CCR7-, CD62L- that express markers of activation and maturation (HLA-DR+, CD28-, CD27-, CD57+ and are functionally distinct, expressing greater levels of TNF-α and IFN-γ on stimulation and perforin at rest than their CD8α+CD8βhigh counterparts. Antigen-specific T cells in HLA-B*4201+HIV-1 infected patients are found within both the CD161-CD8α+CD8βhigh and CD161-CD8α+CD8βlow populations. Overall we have clearly defined CD8αα expressing human T cell subsets using the TL-tetramer, and have demonstrated CD161-CD8α+CD8βlow populations, highly expanded in disease settings, to co-express CD8αβ and CD8αα. Co-expression of CD8αα on CD8αβ T cells may impact on their overall function in-vivo and contribute to the distinctive phenotype of highly differentiated populations in HBV and HIV-1 infection.

    The X-ray telescope concept for XEUS is based on an innovative high performance and light weight Silicon Pore Optics technology. The XEUS telescope is segmented into 16 radial, thermostable petals providing the rigid optical bench structure of the stand alone XRay High Precision Tandem Optics. A fully representative Form Fit Function (FFF) Model of one petal is currently under development to demonstrate the outstanding lightweight telescope capabilities with high optically effective area. Starting from the envisaged system performance the related tolerance budgets were derived. These petals are made from ceramics, i.e. CeSiC. The structural and thermal performance of the petal shall be reported. The stepwise alignment and integration procedure on petal level shall be described. The functional performance and environmental test verification plan of the Form Fit Function Model and the test set ups are described in this paper. In parallel to the running development activities the programmatic and technical issues wrt. the FM telescope MAIT with currently 1488 Tandem Optics are under investigation. Remote controlled robot supported assembly, simultaneous active alignment and verification testing and decentralised time effective integration procedures shall be illustrated.

  14. Study on integrated TRU multi-recycling in sodium cooled fast reactor CDFR

    In view of recently proposed closed fuel cycle strategy which would recycle the integrated transuranics (TRU) from PWR spent fuel in the fast reactors, the neutronics characteristics of TRU recycled in China Demonstration Fast Reactor (CDFR) are studied in this paper. The results show that loading integrated TRU to substitute pure Pu as driver fuel will mainly make the influence on sodium void worth and negligible effects on other parameters, and hence TRU recycling in CDFR is feasible from viewpoint of core neutronics. If TRU is multi-recycled, the variation of TRU composition depends on fuel types and the ratio of TRU and U when recycling. It is indicated that, when TRU is multi-recycled in CDFR with MOX fuel, the minor actinides (MA) fraction in TRU will firstly decrease to ∼7.24% (minimum) within 8 TRU recycle times and then slowly increase to ∼7.7% after 20 TRU recycle times; while when TRU is multi-recycled in CDFR with metal fuel (TRU-U-10Zr), the MA fraction in TRU will gradually approach to an equilibrium state with the MA fraction of ∼3.8%, demonstrating better MA transmutation effect in metal fuel core. No matter 7.7 or 3.8%, they are both lower than ∼10% in PWR spent fuel with burnup of 45 GWd/tU, which presents satisfying effect of MA amount controlling for TRU multi-recycling strategy. On the other hand, the corresponding recycling parameters such as TRU heat release and neutron emission rate are also much lower in metal fuel than those in MOX fuel. Moreover, TRU recycled in metal fuel will bring greater fissile Pu isotopes equilibrium fraction due to better breeding capability of metal fuel. Finally, it could be summarized that integrated TRU multi-recycling in fast reactor can make contributions to both breeding and transmutation, and such strategy is a prospective closed fuel cycle manner to achieve the object of effective control of cumulated MA amount and sustainable development of nuclear energy.

    Results of investigaytion have been discussed ipto the effect of certain temperature-force factors on regularities in the formation of stretch zones during the crack initiation static and transition zones at crack jumps in the process of cyclic loading. The 15Kh2NMFA pressure vessel steel has been investigated. The steel fracture toughnesKub(Ic) has been determined testing s the specimens for excentric stretching or a bending through an angle. It has been shown that transition zones in a front of fatique cracks at the jump beginning and end are formed through the shift mechanism owing to the material separation along the maximum failure zone contour, i.e. along the plastic zone contour in a crack vertex. This is the mait difference of regularities in the formation of the transition zones during fatique crack jumps from stretching zones formed through the break-away mechanism of crack vertex bluntness during its static move. It is noted that a final conclusion on the mechanism of transition zone formation during fartique crack jumps allows one to perform systematic investigation into the plastic zone configuration in a fatique crack verteX and stereofractographic measurement of two identically conjugate jump surfaces on opposite fractures of the same samples

    For decades, proteins were thought to be the sole or at least the dominant source of antigens for T cells. Studies in the 1990s demonstrated that CD1 proteins and mycobacterial lipids form specific targets of human αβ T cells. The molecular basis by which T-cell receptors (TCRs) recognize CD1-lipid complexes is now well understood. Many types of mycobacterial lipids function as antigens in the CD1 system, and new studies done with CD1 tetramers identify T-cell populations in the blood of tuberculosis patients. In human populations, a fundamental difference between the CD1 and major histocompatibility complex systems is that all humans express nearly identical CD1 proteins. Correspondingly, human CD1 responsive T cells show evidence of conserved TCRs. In addition to natural killer T cells and mucosal-associated invariant T (MAIT cells), conserved TCRs define other subsets of human T cells, including germline-encoded mycolyl-reactive (GEM) T cells. The simple immunogenetics of the CD1 system and new investigative tools to measure T-cell responses in humans now creates a situation in which known lipid antigens can be developed as immunodiagnostic and immunotherapeutic reagents for tuberculosis disease. © 2015 John Wiley & Sons A/S.     The liver is a central immunological organ with a high exposure to circulating antigens and endotoxins from the gut microbiota, particularly enriched for innate immune cells (macrophages, innate lymphoid cells, mucosal-associated invariant T (MAIT) cells). In homeostasis, many mechanisms ensure suppression of immune responses, resulting in tolerance. Tolerance is also relevant for chronic persistence of hepatotropic viruses or allograft acceptance after liver transplantation. The liver can rapidly activate immunity in response to infections or tissue damage. Depending on the underlying liver disease, such as viral hepatitis, cholestasis or NASH, different triggers mediate immune-cell activation. Conserved mechanisms such as molecular danger patterns (alarmins), Toll-like receptor signalling or inflammasome activation initiate inflammatory responses in the liver. The inflammatory activation of hepatic stellate and Kupffer cells results in the chemokine-mediated infiltration of neutrophils, monocytes, natural killer (NK) and natural killer T (NKT) cells. The ultimate outcome of the intrahepatic immune response (for example, fibrosis or resolution) depends on the functional diversity of macrophages and dendritic cells, but also on the balance between pro-inflammatory and anti-inflammatory T-cell populations. As reviewed here, tremendous progress has helped to understand the fine-tuning of immune responses in the liver from homeostasis to disease, indicating promising targets for future therapies in acute and chronic liver diseases.

    This scientific-technical report is the result of the research work carried out by Maite Villadoniga Garcia, to obtain the Master's Final Project, this has been developed at the Unit of Soil Degradation in the Department of Environment at CIEMAT. Two main reasons are important for the study of the vegetation in Almaden, firstly to ascertain the state in which they find the different landscapes surrounding the mine and secondly, it would be interesting to know the potential applications for recovery areas, degraded by this type of mining. This report presents the initial analysis of a natural area where there has not been activity related to mining of mercury, but their lithologic conditions and proximity to the mine of Almaden (within 3 km) make it a space in which they are both elevated mercury concentrations in soil (1.25 to 35.55 mg kg - 1) and plant species (Quercus ilex: 0.10-2.29 mg kg - 1; Pseudovernia sp. 1.10-21.06 mg kg - 1). We can conclude after this study that the natural vegetation of the area if they had been affected by years of mining, although not find any obvious signs of damage to these ecosystems and that different factors affect the uptake of mercury by of different species.     Full Text Available Bicuspid valve disease is associated with the development of thoracic aortic aneurysm. The molecular mechanisms underlying this association still need to be clarified. Here, we evaluated the circulating levels of T and B lymphocyte subsets associated with the development of vascular diseases in patients with bicuspid aortic valve or tricuspid aortic valve with and without thoracic aortic aneurysm. We unveiled that the circulating levels of the MAIT, CD4+IL−17A+, and NKT T cell subsets were significantly reduced in bicuspid valve disease cases, when compared to tricuspid aortic valve cases in either the presence or the absence of thoracic aortic aneurysm. Among patients with tricuspid aortic valve, these cells were higher in those also affected by thoracic aortic aneurysm. Similar data were obtained by examining CD19+ B cells, naïve B cells (IgD+CD27−, memory unswitched B cells (IgD+CD27+, memory switched B cells (IgD−CD27+, and double-negative B cells (DN (IgD−CD27−. These cells resulted to be lower in subjects with bicuspid valve disease with respect to patients with tricuspid aortic valve. In whole, our data indicate that patients with bicuspid valve disease show a quantitative reduction of T and B lymphocyte cell subsets. Future studies are encouraged to understand the molecular mechanisms underlying this observation and its pathophysiological significance.

    Full Text Available The biography of Julija Beniuševičiūtė-Žymantie­nė – Žemaitė (1845–1921, one of the greatest Lithu­anian Realists, has got a broad accent of feminist ac­tivity. Some short stories written by Žemaitė express feminist ideas with publicistic pathos (In Town, „Mieste“, 1905, Mister Zigmantas, „Ponas Zigman­tas“, and others. In her short story Eve („Ieva“ Že­maitė presents an interesting ironic interpretation of the biblical archetype. Žemaitė’s best literary visions of women’s infelicity varies the topos of a quiet / silent / silenced woman and interestingly imply some gap between feminist ideology and literary meaning. In the short stories The Daughter-in-Law („Marti“, 1896, They Didn’t Have a Good Mother („Neturėjo geros motinos“, 1895 the reason of misfortunes se­ems to be deeper than just women being oppressed by men in the patriarchal society. The patriarchal op­pression of women is constructed as a metonymy of the philosophical evil. The short story Sutkai (1897 may be interpreted as a literary vision of a feminine utopia: woman’s identity here is defined rather as wholeness than “otherness”.

    Full Text Available Lactobacilli are probiotic commensal bacteria and potent modulators of immunity. When present in the gut or supplemented as probiotics, they beneficially modulate ex vivo immune responsiveness. Further, factors derived from several lactobacilli strains act immune regulato-ry in vitro. In contrast, Staphylococcus aureus (S. aureus is known to induce excessive T cell activation. In this study we aimed to investigate S. aureus-induced activation of human muco-sal associated invariant T cells (MAIT cells, γδ T cells, NK cells, as well as of conventional CD4+ and CD8+ T cells in vitro. Further, we investigated if lactobacilli-derived factors could modulate their activation.PBMC were cultured with S. aureus 161:2 cell free supernatant (CFS, staphylococcal en-terotoxin A or CD3/CD28-beads alone or in combination with Lactobacillus rhamnosus (L. rhamnosus GG-CFS or Lactobacillus reuteri (L. reuteri DSM 17938-CFS, and activation of T and NK cells was evaluated. S. aureus-CFS induced IFN-γ and CD107a expression as well as proliferation. Co-stimulation with lactobacilli-CFS dampened lymphocyte activation in all cell types analysed. Pre-incubation with lactobacilli-CFS was enough to reduce subsequent activation and the ab-sence of APC or APC-derived IL-10 did not prevent lactobacilli-mediated dampening. Final-ly, lactate selectively dampened activation of unconventional T cells and NK cells. In summary, we show that molecules present in the lactobacilli-CFS are able to directly dampen in vitro activation of conventional and unconventional T cells and of NK cells. This study provides novel insights on the immune modulatory nature of probiotic lactobacilli and suggests a role for lactobacilli in modulation of induced T and NK cell activation.

    This first monograph of the Ifri program on European Governance and Geopolitics of Energy is devoted to the control of carbon dioxide emissions within the European Union. Since it is almost unanimously accepted that Greenhouse Gas emissions constitute the main cause of the observed increase of the world average temperature, the system implemented by the European Union to limit and decrease the CO{sub 2} emissions is a significant pillar of the EU energy policy, the two others being the acceptance by the Member States of long-term commitments (for instance on the future share of renewable energy sources in their energy balance sheet) and the establishment of an internal market for electricity and gas. Though simple in principle, the European Union Greenhouse Gas Emission Trading Scheme (EU ETS) is in fact rather complex, and only experts really understand its merits and its deficiencies. These deficiencies are real and will have to be corrected in the future for the system to be effective. At this moment, when the 2005-2007 trial phase of the EU ETS is ending, the monograph has the purpose to stimulate the discussion between experts and to enable all those interested in the topic to understand the issues and to take part in the public debates on the subject.     Full Text Available OBJETIVO. No presente estudo propusemo-nos a avaliar, por meio da microscopia de luz, os aspectos morfológicos e morfométricos dos cardiomiócitos do ventrículo esquerdo de ratas albinas durante a prenhez. MÉTODOS. Acasalamos doze ratas virgens que foram dividas ao acaso em quatro grupos, de acordo com a idade gestacional. Os animais correspondentes a cada grupo foram sacrificados ao 1o(G-A, 7o(G-B, 14o(G-C e 21o(G-D dias de prenhez, sendo coletados fragmentos do terço médio do ventrículo esquerdo, os quais após processamento apropriado, permitiram observação adequada à microscopia de luz. A cariometria foi realizada mensurando-se os diâmetros maiores e menores dos cardiomiócitos com o auxílio de um tambor rotativo modelo K 8 X adaptado a um microscópio de luz. RESULTADOS. O estudo em nível da microscopia de luz praticamente não mostrou alterações com o decorrer da prenhez. No entanto, a morfometria revelou que os volumes dos cardiomiócitos estão aumentados no 14o dia da prenhez, mostrando-se estatisticamente significante quando comparado aos demais grupos estudados. Assim, nossos resultados demonstraram haver hipertrofia ventricular esquerda durante a gestação. CONCLUSÃO. Durante a gestação há um processo dinâmico reversível de remodelação ventricular em conseqüência das alterações adaptativas gravídicas.PURPOSE. On the other hand, lipid profile modification after a cardiovascular event related to acute coronary syndrome (ACS has been recognized. The prevalence of conventional risk factors and the lipid profile at the time of admission in patients with ACS and significant CAD (stenosis ≥50% determined through coronary angiography is not well described. Methods: We studied 3,447 patients with a diagnosis of ACS and significant CAD with stenosis ≥50%, as shown on angiography. We recorded the presence of conventional risk factors, including smoking, hypertension, dyslipidemia, and diabetes. In addition, we analyzed the lipid profiles within the first 24 hours of admission. We analyzed the studied population and compared findings according to sex.Results: Most patients (81.7% were male. ST-elevation myocardial infarction was present in 51.3% of patients, and non-ST-elevation acute coronary syndrome was present in 48.7%. The most frequent risk factor was smoking, which was present in 68% of patients, followed by hypertension (57.8%, dyslipidemia (47.5%, and diabetes (37.7%. In women, the most frequent risk factors were hypertension, diabetes, and dyslipidemia, whereas in men, smoking was the most frequent.     In the decades following India's independence from British rule in 1947, the West's image of India was summarized in three simple cliches: the world's largest democracy, an impoverished continent, and economic growth hampered by a fussy bureaucracy and the caste system, all in a context of a particular religion. These cliches are perhaps one of the reasons that the success of India's green revolution was recognized so late, a revolution that allowed the country to develop its agricultural sector and to feed its population. Since the 1990's, the easing of planning constraints have liberated the Indian economy and allowed it to embark on a more significant path of growth. New cliches have begun to replace the old: India will become a second China and, lagging by 10 to 20 years, will follow the same trajectory, with its development marked more by services and the use of renewable energy. However, these trends will not prevent primary energy demand from exploding. On the contrary, India faces difficult choices on how it increases clean, secure, affordable energy to all its citizens. Many of the choices are the same as found elsewhere, but on a scale matched only by China. The IFRI European Governance and Geopolitics of Energy Project intends this study to deepen public understanding of the magnitude of India's challenges. Various aspects of the serious energy problems are studied throughout this monograph. The authors have written freely on these matters without attempting to reconcile their different viewpoints. The first chapter, by Maite Jaureguy-Naudin and Jacques Lesourne, presents an overview of India's present and future energy system. The authors follow a prudent but realistic view of India's future.     In the decades following India's independence from British rule in 1947, the West's image of India was summarized in three simple cliches: the world's largest democracy, an impoverished continent, and economic growth hampered by a fussy bureaucracy and the caste system, all in a context of a particular religion. These cliches are perhaps one of the reasons that the success of India's green revolution was recognized so late, a revolution that allowed the country to develop its agricultural sector and to feed its population. Since the 1990's, the easing of planning constraints have liberated the Indian economy and allowed it to embark on a more significant path of growth. New cliches have begun to replace the old: India will become a second China and, lagging by 10 to 20 years, will follow the same trajectory, with its development marked more by services and the use of renewable energy. However, these trends will not prevent primary energy demand from exploding. On the contrary, India faces difficult choices on how it increases clean, secure, affordable energy to all its citizens. Many of the choices are the same as found elsewhere, but on a scale matched only by China. The IFRI European Governance and Geopolitics of Energy Project intends this study to deepen public understanding of the magnitude of India's challenges. Various aspects of the serious energy problems are studied throughout this monograph. The authors have written freely on these matters without attempting to reconcile their different viewpoints. The first chapter, by Maite Jaureguy-Naudin and Jacques Lesourne, presents an overview of India's present and future energy system. The authors follow a prudent but realistic view of India's future. The second chapter, by Jean-Joseph Boillot, a French expert on India who has published several books and articles on this subject, and Nicolas Autheman, research fellow, describes in greater detail the specifics of India

    Full Text Available The paper gives an overview of a network of authors who have been active since the end of the 20th century, mainly (but not only in Tartu. They can be characterized as having an experimental attitude and maintaining distance from the literary mainstream, but at the same time they do not try to be consciously polemical or novatory, because partly they repeat the techniques of the 20th century avant-garde and, what is maybe more important, they sidestep the dialectics of innovating or altering the literary canon. The network is not strictly organized; it is rather a rhizome that has some denser nodes (literary journals and mailing lists. There have been different attempts to name the phenomenon: Tartu avant-garde, microcosmic literature, cognitive realism, Y-literature, "eksp" and so on. I have chosen "paranoiac-critique" as the term that best captures the common characteristics of these authors and their artistic practices (some of the authors are active not only as text writers, but also as conceptual artists.The paper then gives a list, with commentary, of the most important groups and individuals of that movement. In the role of "predecessors" are the group 14NÜ (Paavo Matsin, Mait Laas, Marianne Ravi et al., which also experimented with the format of the book, and a company in Tartu that developed a specific actionist stylistics called "mugiv" (Marko Kompus, Mehis Heinsaar, Martiini, Kaspar et al.. The sovereign figure in the experimental network is Valdur Mikita, who has also academically researched the problems of creativity; he is also a theoretician and metapoet. An important institution for the network has been the literary journal Vihik (initiator Berk Vaher; the later editor of the journal Jaak Tomberg has also participated in activities of the Tartu Theatre Laboratory (managed by Andreas W, which has experimented with "technological theatre".     Full Text Available In this article I discuss autobiographical texts which focus on children and childhood in late 19th century Estonia. Childhood memories as well as other autobiographical material became popular in Estonia in the 1920s-1930s, when most of the studied works--the memoirs by Anna Haava, Mait Metsanurk, Jaan Lattik, Jaan Vahtra, Friedebert Tuglas, August Kitzberg and Marta Sillaots--were written. Some texts come from the 19th century (e.g. Lilli Suburg’s autobiographical works or early 20th century (e.g. manuscripts by Hans Leoke, and Johannes Kõrv. Childhood as described in these autobiographical texts covers a period of circa 1850-1900, and the majority of the authors come from the families of South-Estonian peasants or manorial servants. In addition to being written in Estonian and having the same theme, they were all also written by authors of fiction for children or by people who had close contact with children, such as schoolteachers. The article offers a novel approach in the Estonian context by presenting a typology of childhood stories and looking at childhood recollections as an important part of childhood studies. The researchers of childhood investigate how society understands and values children and childhood, what children’s everyday life is like, what possibilities there are for development and if there exists a specific children’s culture in society (such as clothing, food, language, leisure activities, or independent creative work. Childhood studies as a separate discipline does not exist in Estonia, although some important works have been published by educational scholars and art historians. The autobiographical texts under discussion show that in the late 19th century, the majority of Estonian children lived in the countryside in patriarchal families, and childhood was short because children had to help their parents with farmwork quite early, at the age of six.
