  1. Anti-Tribbles Pseudokinase 2 (TRIB2)-Immunization Modulates Hypocretin/Orexin Neuronal Functions.

    Recent findings showed that 16%-26% of narcolepsy patients were positive for anti-tribbles pseudokinase 2 (TRIB2) antibody, and the intracerebroventricular administration of immunoglobulin-G purified from anti-TRIB2 positive narcolepsy patients caused hypocretin/orexin neuron loss. We investigated the pathophysiological role of TRIB2 antibody using TRIB2-immunized rats and hypocretin/ataxin-3 transgenic (ataxin-3) mice. Plasma, cerebrospinal fluid (CSF), and hypothalamic tissues from TRIB2-immunized rats were collected. Anti-TRIB2 titers, hypocretin contents, mRNA expressions, the cell count of hypocretin neurons, and immunoreactivity of anti-TRIB2 antibodies on hypocretin neurons were investigated. The plasma from ataxin-3 mice was also used to determine the anti-TRIB2 antibody titer changes following the loss of hypocretin neurons. TRIB2 antibody titers increased in the plasma and CSF of TRIB2-immunized rats. The hypothalamic tissue immunostained with the sera from TRIB2-immunized rats revealed positive signals in the cytoplasm of hypcretin neurons. While no changes were found regarding hypothalamic hypocretin contents or cell counts, but there were significant decreases of the hypocretin mRNA level and release into the CSF. The plasma from over 26-week-old ataxin-3 mice, at the advanced stage of hypocretin cell destruction, showed positive reactions against TRIB2 antigen, and positive plasma also reacted with murine hypothalamic hypocretin neurons. Our results suggest that the general activation of the immune system modulates the functions of hypocretin neurons. The absence of a change in hypocretin cell populations suggested that factors other than anti-TRIB2 antibody play a part in the loss of hypocretin neurons in narcolepsy. The increased anti-TRIB2 antibody after the destruction of hypocretin neurons suggest that anti-TRIB2 antibody in narcolepsy patients is the consequence rather than the inciting cause of hypocretin cell destruction. © Sleep Research

  2. TRIB3 protein denotes a good prognosis in breast cancer patients and is associated with hypoxia sensitivity

    Background: Tribbles homolog 3 (TRIB3) is a pseudokinase involved in the regulation of several signaling pathways involved in cell survival and/or cell stress. Here, we determined the correlation between breast cancer prognosis and TRIB3 protein levels and established the role of TRIB3 in cell survival after hypoxia and/or radiotherapy. Material and methods: TRIB3 mRNA and protein were quantified in a new independent breast cancer patient cohort using QPCR and a new specific avian antibody against TRIB3. In addition, we used siRNA-mediated knockdown of TRIB3 in a colony-forming assay after hypoxia and radiotherapy. Results: TRIB3 mRNA and protein levels did not correlate in breast cancer cell lines or human breast cancer material. We validated our earlier finding that high TRIB3 mRNA denotes a poor prognosis, but found that high TRIB3 protein levels were associated with a good prognosis in breast cancer patients. We also show that knockdown of TRIB3 resulted in an increased survival under hypoxic conditions. Conclusion: Whereas mRNA levels of TRIB3 are related with a poor prognosis, TRIB3 protein is associated with a good prognosis in human breast cancer patients, possibly due to the fact that TRIB3 is involved in hypoxia tolerance.

  3. Skeletal Muscle TRIB3 Mediates Glucose Toxicity in Diabetes and High- Fat Diet–Induced Insulin Resistance

    In the current study, we used muscle-specific TRIB3 overexpressing (MOE) and knockout (MKO) mice to determine whether TRIB3 mediates glucose-induced insulin resistance in diabetes and whether alterations in TRIB3 expression as a function of nutrient availability have a regulatory role in metabolism. In streptozotocin diabetic mice, TRIB3 MOE exacerbated, whereas MKO prevented, glucose-induced insulin resistance and impaired glucose oxidation and defects in insulin signal transduction compared with wild-type (WT) mice, indicating that glucose-induced insulin resistance was dependent on TRIB3. In response to a high-fat diet, TRIB3 MOE mice exhibited greater weight gain and worse insulin resistance in vivo compared with WT mice, coupled with decreased AKT phosphorylation, increased inflammation and oxidative stress, and upregulation of lipid metabolic genes coupled with downregulation of glucose metabolic genes in skeletal muscle. These effects were prevented in the TRIB3 MKO mice relative to WT mice. In conclusion, TRIB3 has a pathophysiological role in diabetes and a physiological role in metabolism. Glucose-induced insulin resistance and insulin resistance due to diet-induced obesity both depend on muscle TRIB3. Under physiological conditions, muscle TRIB3 also influences energy expenditure and substrate metabolism, indicating that the decrease and increase in muscle TRIB3 under fasting and nutrient excess, respectively, are critical for metabolic homeostasis.   4. Trib1 Is Overexpressed in Systemic Lupus Erythematosus, While It Regulates Immunoglobulin Production in Murine B Cells

    Full Text Available Systemic lupus erythematosus (SLE is a severe and heterogeneous autoimmune disease with a complex genetic etiology, characterized by the production of various pathogenic autoantibodies, which participate in end-organ damages. The majority of human SLE occurs in adults as a polygenic disease, and clinical flares interspersed with silent phases of various lengths characterize the usual evolution of the disease in time. Trying to understand the mechanism of the different phenotypic traits of the disease, and considering the central role of B cells in SLE, we previously performed a detailed wide analysis of gene expression variation in B cells from quiescent SLE patients. This analysis pointed out an overexpression of TRIB1. TRIB1 is a pseudokinase that has been implicated in the development of leukemia and also metabolic disorders. It is hypothesized that Trib1 plays an adapter or scaffold function in signaling pathways, notably in MAPK pathways. Therefore, we planned to understand the functional significance of TRIB1 overexpression in B cells in SLE. We produced a new knock-in model with B-cell-specific overexpression of Trib1. We showed that overexpression of Trib1 specifically in B cells does not impact B cell development nor induce any development of SLE symptoms in the mice. By contrast, Trib1 has a negative regulatory function on the production of immunoglobulins, notably IgG1, but also on the production of autoantibodies in an induced model. We observed a decrease of Erk activation in BCR-stimulated Trib1 overexpressing B cells. Finally, we searched for Trib1 partners in B cells by proteomic analysis in order to explore the regulatory function of Trib1 in B cells. Interestingly, we find an interaction between Trib1 and CD72, a negative regulator of B cells whose deficiency in mice leads to the development of autoimmunity.   5. Regulation of Trib2 by an E2F1-C/EBPα feedback loop in AML cell proliferation.

    The loss of regulation of cell proliferation is a key event in leukemic transformation, and the oncogene tribbles (Trib)2 is emerging as a pivotal target of transcription factors in acute leukemias. Deregulation of the transcription factor E2F1, normally repressed by CCAAT enhancer-binding protein α (C\\/EBPα)-p42, occurs in acute myeloid leukemia (AML), resulting in the perturbation of cell cycle and apoptosis, emphasizing its importance in the molecular pathogenesis of AML. Here we show that E2F family members directly regulate Trib2 in leukemic cells and identify a feedback regulatory loop for E2F1, C\\/EBPα, and Trib2 in AML cell proliferation and survival. Further analyses revealed that E2F1-mediated Trib2 expression was repressed by C\\/EBPα-p42, and in normal granulocyte\\/macrophage progenitor cells, we detect C\\/EBPα bound to the Trib2 promoter. Pharmacological inhibition of the cell cycle or Trib2 knockdown resulted in a block in AML cell proliferation. Our work proposes a novel paradigm whereby E2F1 plays a key role in the regulation of Trib2 expression important for AML cell proliferation control. Importantly, we identify the contribution of dysregulated C\\/EBPα and E2F1 to elevated Trib2 expression and leukemic cell survival, which likely contributes to the initiation and maintenance of AML and may have significant implications for normal and malignant hematopoiesis.

    Tribbles homolog 3 (TRIB3) is a mammalian pseudokinase that is induced in neuronal cell cultures in response to cell death-inducing stresses, including neurotrophic factor deprivation. TRIB3 is an inhibitor of activating transcription factor 4 (ATF4), the central transcriptional regulator in the eukaryotic translation initiation factor 2α (eIF2α) phosphorylation pathway that is involved in the cellular stress response and behavioral processes. In this article, we study the expression of Trib3 in the mouse brain, characterize the brain morphology of mice with a genetic ablation of Trib3 and investigate whether Trib3 deficiency alters eIF2α-dependent cognitive abilities. Our data show that the consumption of a leucine-deficient diet induces Trib3 expression in the anterior piriform cortex, the brain region responsible for detecting essential amino acid intake imbalance. However, the aversive response to leucine-devoid diet does not differ in Trib3 knockout and wild type mice. Trib3 deletion also does not affect long-term spatial memory and reversal learning in the Morris water maze and auditory or contextual fear conditioning. During embryonic development, Trib3 expression increases in the brain and persists in the early postnatal stadium. Neuroanatomical characterization of mice lacking Trib3 revealed enlarged lateral ventricles. Thus, although the absence of Trib3 does not alter the eIF2α pathway-dependent cognitive functions of several areas of the brain, including the hippocampus, amygdala and anterior piriform cortex, Trib3 may serve a role in other central nervous system processes and molecular pathways.

    Despite the growing number of studies exhibited an association of diabetes mellitus (DM) and lung cancer progression, the concrete mechanism of DM aggravating lung cancer has not been elucidated. This study was to investigate whether and how high glucose (HG) contribute to the proliferation and migration of non-small cell lung cancer (NSCLC) cells in vitro. In the present study, we confirmed that HG promoted the proliferation and migration of NSCLC cells, and also induced an anti-apoptosis effect on NSCLC cells. Moreover, HG inhibited the expression of GAS5 in NSCLC cells but elevated the protein level of TRIB3. GAS5 overexpression promoted the degradation of TRIB3 protein by ubiquitination and inhibited the HG induced-proliferation, anti-apoptosis and migration of NSCLC cells. Importantly, TRIB3 overexpression reversed the effects of GAS5 on the HG-treated NSCLC cells. Taken together, down-regulated GAS5 by HG significantly enhanced the proliferation, anti-apoptosis and migration in NSCLC cells through TRIB3, thus promoting the carcinogenesis of NSCLC. ©2018 The Author(s).

    Porphyromonas endodontalis lipopolysaccharide (LPS) is considered to be correlated with the progression of bone resorption in periodontal and periapical diseases. Wnt5a has recently been implicated in inflammatory processes, but its role is unclear as a P. endodontalis LPS-induced mediator in osteoblasts. Tribbles homolog 3 (TRIB3) encodes a pseudokinase and has been linked to inflammation in certain situations. Here, we found that P. endodontalis LPS induced Wnt5a expression in a dose- and time-dependent manner and it also upregulated translocation, phosphorylation and transcriptional activity of nuclear factor-κB (NF-κB) in MC3T3-E1 cells. Bay 11-7082 blocked the translocation of NF-κB and Wnt5a expression induced by P. endodontalis LPS. Chromatin immunoprecipitation assay further established that induction of Wnt5a by P. endodontalis LPS was mediated through the NF-κB p65 subunit. Additionally, P. endodontalis LPS increased expression of TRIB3 in osteoblasts after 10 h simulated time. Overexpression of TRIB3 enhanced NF-κB phosphorylation and Wnt5a induction, whereas knockdown of TRIB3 inhibited NF-κB phosphorylation and Wnt5a expression in P. endodontalis LPS-stimulated osteoblasts. These results suggest that P. endodontalis LPS has the ability to promote the expression of Wnt5a in mouse osteoblasts, and this induction is mainly mediated by NF-κB pathway. TRIB3 seems to modulate the sustained expression of Wnt5a in osteoblasts stimulated by P. endodontalis LPS, as well as regulating NF-κB phosphorylation. © 2015 John Wiley & Sons A/S.     MicroRNAs (miRNAs) have significant roles in cell processes, including proliferation, apoptosis and stress responses. To investigate the involvement of miR-99 in the inhibition of HeLa cell proliferation, an miR-99 gene expression vector (pU6.1/miR-99), which overexpressed miR-99 in HeLa cells after transient transfection, was constructed. The expression of miR-99 was detected by qPCR. Cell proliferation and apoptosis were analyzed by cell viability, proliferation and apoptosis assays, as well as by electron microscopy. The results showed that overexpression of miR-99 in HeLa cells increased the HeLa cell mortality rate. Moreover, miR-99 overexpression was able to markedly inhibit HeLa cell proliferation according to the 3-(4, 5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. The cell apoptosis rate was significantly higher in pU6.1/miR-99-treated cells compared with that in the control cultures. Increases in intracellular electron density, as well as the proportion of nuclear plasma, blebbing phenomena and apoptotic bodies were observed in pU6.1/miR-99-treated cells compared with control cultures according to electron microscopy analysis. The Tribbles 2 (TRIB2) 3'-untranslated region was also observed to be targeted by miR-99 and the results further demonstrated that miR-99 was able to negatively regulate TRIB2 expression in HeLa cells The results indicate that miR-99 acts as a tumor suppressor gene in HeLa cells, establishing a theoretical basis for its application in cancer therapeutics.

(Orthoptera, Tettigonioidea, Phaneropteridae: second contribution to neotropical Phaneropterinae The new tribe Pycnopalpini is proposed, based on morphological similarity among the genera Hetaira, Pycnopalpa, Topana (Pycnopalpina n. subtr., Dolichocercus, Oxyprorella, Theia, and TheiellaTheia unicolor, Theia bifurcata n. sp., Theiella amazonica n. sp., Theiella andina n. sp., and D. santanderensis n. sp. from Colombia, and D. costaricensis n. sp. from Costa Rica. Hetaira (Atopana morona and Hetaira (Atopana varia are recorded from the Colombian Amazon, where both species seem to occur sympatrically. Theia peruviana n. comb. is also recorded from the Colombian Amazon, where it seems to occur sympatrically with T. bifurcata. The paper contains additional new combinations, and Oxyprorella gregoirea syn. n. is a new synonym for Insara bolivari. This contribution reconsiders the neotropical subfamily Phaneropterinae, aiming to establish the limits of the tribes and genera of this subfamily.

    ABSTRACT Hepatitis C virus (HCV) infection often causes chronic hepatitis, liver cirrhosis, and ultimately hepatocellular carcinoma. However, the mechanisms underlying HCV-induced liver pathogenesis are still not fully understood. By transcriptome sequencing (RNA-Seq) analysis, we recently identified host genes that were significantly differentially expressed in cell culture-grown HCV (HCVcc)-infected cells. Of these, tribbles homolog 3 (TRIB3) was selected for further characterization. TRIB3 was initially identified as a binding partner of protein kinase B (also known as Akt). TRIB3 blocks the phosphorylation of Akt and induces apoptosis under endoplasmic reticulum (ER) stress conditions. HCV has been shown to enhance Akt phosphorylation for its own propagation. In the present study, we demonstrated that both mRNA and protein levels of TRIB3 were increased in the context of HCV replication. We further showed that promoter activity of TRIB3 was increased by HCV-induced ER stress. Silencing of TRIB3 resulted in increased RNA and protein levels of HCV, whereas overexpression of TRIB3 decreased HCV replication. By employing an HCV pseudoparticle entry assay, we further showed that TRIB3 was a negative host factor involved in HCV entry. Both in vitro binding and immunoprecipitation assays demonstrated that HCV NS3 specifically interacted with TRIB3. Consequently, the association of TRIB3 and Akt was disrupted by HCV NS3, and thus, TRIB3-Akt signaling was impaired in HCV-infected cells. Moreover, HCV modulated TRIB3 to promote extracellular signal-regulated kinase (ERK) phosphorylation, activator protein 1 (AP-1) activity, and cell migration. Collectively, these data indicate that HCV exploits the TRIB3-Akt signaling pathway to promote persistent viral infection and may contribute to HCV-mediated pathogenesis.     The protein tribbles-1, encoded by the gene TRIB1, is increasingly recognized as a major regulator of multiple cellular and physiological processes in humans. Recent human genetic studies, as well as molecular biological approaches, have implicated this intriguing protein in the aetiology of multiple human diseases, including myeloid leukaemia, Crohn's disease, non-alcoholic fatty liver disease (NAFLD), dyslipidaemia and coronary artery disease (CAD). Genome-wide association studies (GWAS) have repeatedly identified variants at the genomic TRIB1 locus as being significantly associated with multiple plasma lipid traits and cardiovascular disease (CVD) in humans. The involvement of TRIB1 in hepatic lipid metabolism has been validated through viral-mediated hepatic overexpression of the gene in mice; increasing levels of TRIB1 decreased plasma lipids in a dose-dependent manner. Additional studies have implicated TRIB1 in the regulation of hepatic lipogenesis and NAFLD. The exact mechanisms of TRIB1 regulation of both plasma lipids and hepatic lipogenesis remain undetermined, although multiple signalling pathways and transcription factors have been implicated in tribbles-1 function. Recent reports have been aimed at developing TRIB1-based lipid therapeutics. In summary, tribbles-1 is an important modulator of human energy metabolism and metabolic syndromes and worthy of future studies aimed at investigating its potential as a therapeutic target. © 2015 Authors; published by Portland Press Limited.

    Methamphetamine (Meth) is a widely abused psychoactive drug that primarily damages the nervous system, notably causing dopaminergic neuronal apoptosis. CCAAT-enhancer binding protein (C/EBPβ) is a transcription factor and an important regulator of cell apoptosis and autophagy. Insulin-like growth factor binding protein (IGFBP5) is a proapoptotic factor that mediates Meth-induced neuronal apoptosis, and Trib3 (tribbles pseudokinase 3) is an endoplasmic reticulum (ER) stress-inducible gene involved in autophagic cell death through the mammalian target of rapamycin (mTOR) signaling pathway. To test the hypothesis that C/EBPβ is involved in Meth-induced IGFBP5-mediated neuronal apoptosis and Trib3-mediated neuronal autophagy, we measured the protein expression of C/EBPβ after Meth exposure and evaluated the effects of silencing C/EBPβ, IGFBP5, or Trib3 on Meth-induced apoptosis and autophagy in neuronal cells and in the rat striatum after intrastriatal Meth injection. We found that, at relatively high doses, Meth exposure increased C/EBPβ protein expression, which was accompanied by increased neuronal apoptosis and autophagy; triggered the IGFBP5-mediated, p53-up-regulated modulator of apoptosis (PUMA)-related mitochondrial apoptotic signaling pathway; and stimulated the Trib3-mediated ER stress signaling pathway through the Akt-mTOR signaling axis. We also found that autophagy is an early response to Meth-induced stress upstream of apoptosis and plays a detrimental role in Meth-induced neuronal cell death. These results suggest that Meth exposure induces C/EBPβ expression, which plays an essential role in the neuronal apoptosis and autophagy induced by relatively high doses of Meth; however, relatively low concentrations of Meth did not change the expression of C/EBPβ in vitro.     Highlights: • All manufacturing drawings of the first sector of VV have been completed. • Full scale mock-ups have been constructed to verify fabrication procedure. • Qualifications for welding and forming are done and for NDE are ongoing. • Manufacturing progress is around 40% for the sector and LPSE up to the end of 2015. - Abstract: Manufacturing design of Korean sectors and ports for the ITER Vacuum Vessel (VV) has been developed to comply with the tight tolerance and severe inspection requirements. The first VV sector and lower ports are being fabricated slowly under strict regulations after verification using several real scale mock-ups and qualifications for welding, forming and NDE. During three years after start of fabrication, manufacturing progress on four poloidal segments of the first sector is that (1) all inner shells were welded, (2) forgings for complicate components have been machined, (3) port stubs and poloidal T-ribs were assembled, and (4) machined components are welded on the inner shells by narrow-gap TIG welding and electron beam welding. The progress of lower ports is that (1) inner shells of stub extensions were bent and treated with heat, (2) T-ribs were fabricated and examined by qualified phased array UT, (3) supporting pads and gussets have been machined, and (4) inner shells are assembled with T-ribs and machined forgings. The progress rate of manufacturing is around 40% up to the end of 2015 for the first sector and lower port stub extensions.

    Vildagliptin promotes beta cell survival by inhibiting cell apoptosis. It has been suggested that chronic ER (endoplasmic reticulum) stress triggers beta cell apoptosis. The objective of the study is to explore whether the pro-survival effect of vildagliptin is associated with attenuation of endoplasmic reticulum stress in islets of db/db mice. Vildagliptin was orally administered to db/db mice for 6 weeks, followed by evaluation of beta cell apoptosis by caspase3 activity and TUNEL staining method. Endoplasmic reticulum stress markers were determined with quantitative RT-PCR, immunohistochemistry and immunoblot analysis. After 6 weeks of treatment, vildagliptin treatment increased plasma active GLP-1 levels (22.63±1.19 vs. 11.69±0.44, Pvildagliptin treatment down-regulated several genes related to endoplasmic reticulum stress including TRIB3 (tribbles homolog 3) (15.9±0.4 vs. 33.3±1.7, ×10⁻³, PVildagliptin promoted beta cell survival in db/db mice in association with down-regulating markers of endoplasmic reticulum stress including TRIB3, ATF-4 as well as CHOP.   12. Microarray and pathway analysis reveal distinct mechanisms underlying cannabinoid-mediated modulation of LPS-induced activation of BV-2 microglial cells.

    Full Text Available Cannabinoids are known to exert immunosuppressive activities. However, the mechanisms which contribute to these effects are unknown. Using lipopolysaccharide (LPS to activate BV-2 microglial cells, we examined how Δ(9-tetrahydrocannabinol (THC, the major psychoactive component of marijuana, and cannabidiol (CBD the non-psychoactive component, modulate the inflammatory response. Microarray analysis of genome-wide mRNA levels was performed using Illumina platform and the resulting expression patterns analyzed using the Ingenuity Pathway Analysis to identify functional subsets of genes, and the Ingenuity System Database to denote the gene networks regulated by CBD and THC. From the 5338 transcripts that were differentially expressed across treatments, 400 transcripts were found to be upregulated by LPS, 502 by CBD+LPS and 424 by THC+LPS, while 145 were downregulated by LPS, 297 by CBD+LPS and 149 by THC+LPS, by 2-fold or more (p≤0.005. Results clearly link the effects of CBD and THC to inflammatory signaling pathways and identify new cannabinoid targets in the MAPK pathway (Dusp1, Dusp8, Dusp2, cell cycle related (Cdkn2b, Gadd45a as well as JAK/STAT regulatory molecules (Socs3, Cish, Stat1. The impact of CBD on LPS-stimulated gene expression was greater than that of THC. We attribute this difference to the fact that CBD highly upregulated several genes encoding negative regulators of both NFκB and AP-1 transcriptional activities, such as Trib3 and Dusp1 known to be modulated through Nrf2 activation. The CBD-specific expression profile reflected changes associated with oxidative stress and glutathione depletion via Trib3 and expression of ATF4 target genes. Furthermore, the CBD affected genes were shown to be controlled by nuclear factors usually involved in regulation of stress response and inflammation, mainly via Nrf2/Hmox1 axis and the Nrf2/ATF4-Trib3 pathway.     This paper presents the development of a rainfall-triggered landslide module within an existing physically based spatially distributed ecohydrologic model. The model, tRIBS-VEGGIE (Triangulated Irregular Networks-based Real-time Integrated Basin Simulator and Vegetation Generator for Interactive Evolution), is capable of a sophisticated description of many hydrological processes; in particular, the soil moisture dynamics are resolved at a temporal and spatial resolution required to examine the triggering mechanisms of rainfall-induced landslides. The validity of the tRIBS-VEGGIE model to a tropical environment is shown with an evaluation of its performance against direct observations made within the study area of Luquillo Forest. The newly developed landslide module builds upon the previous version of the tRIBS landslide component. This new module utilizes a numerical solution to the Richards' equation (present in tRIBS-VEGGIE but not in tRIBS), which better represents the time evolution of soil moisture transport through the soil column. Moreover, the new landslide module utilizes an extended formulation of the factor of safety (FS) to correctly quantify the role of matric suction in slope stability and to account for unsaturated conditions in the evaluation of FS. The new modeling framework couples the capabilities of the detailed hydrologic model to describe soil moisture dynamics with the infinite slope model, creating a powerful tool for the assessment of rainfall-triggered landslide risk.

  17. Variants of Insulin-Signaling Inhibitor Genes in Type 2 Diabetes and Related Metabolic Abnormalities

    Full Text Available Insulin resistance has a central role in the pathogenesis of several metabolic diseases, including type 2 diabetes, obesity, glucose intolerance, metabolic syndrome, atherosclerosis, and cardiovascular diseases. Insulin resistance and related traits are likely to be caused by abnormalities in the genes encoding for proteins involved in the composite network of insulin-signaling; in this review we have focused our attention on genetic variants of insulin-signaling inhibitor molecules. These proteins interfere with different steps in insulin-signaling: ENPP1/PC-1 and the phosphatases PTP1B and PTPRF/LAR inhibit the insulin receptor activation; INPPL1/SHIP-2 hydrolyzes PI3-kinase products, hampering the phosphoinositide-mediated downstream signaling; and TRIB3 binds the serine-threonine kinase Akt, reducing its phosphorylation levels. While several variants have been described over the years for all these genes, solid evidence of an association with type 2 diabetes and related diseases seems to exist only for rs1044498 of the ENPP1 gene and for rs2295490 of the TRIB3 gene. However, overall the data recapitulated in this Review article may supply useful elements to interpret the results of novel, more technically advanced genetic studies; indeed it is becoming increasingly evident that genetic information on metabolic diseases should be interpreted taking into account the complex biological pathways underlying their pathogenesis.

  1. Improving the productive and reproductive efficiency in the new zealand rabbits under the egyptian conditions

    The present work was planned to study the effects of different alleviation techniques under both mild and hot conditions on productive and reproductive traits of growing female and male rabbits, mature bucks and mature does during pregnancy. In growing males and females, the results showed that the heat stress conditions caused significant decrease in body weight, daily body gain, feed consumption, as well as, significant decrease in most blood hematology, blood components and blood minerals, blood vitamins and hormonal levels, and increase in water intake, concentration of creatinine. In mature bucks, heat stress caused significant decrease in physical and chemical of semen characteristics and blood components. In pregnant does, heat stress caused significant decrease in DOE traits, as well as, significant decrease in most blood hematology, blood components and blood minerals, blood vitamins and hormonal levels, and increase in water intake.With regard to the effects of alleviation of heat load by drinking cool water during hot period or by injection of Tri-B two times weakly for growing male and female rabbits, as compared to without treated rabbits caused significant increase in growth performance, physiological traits and economic traits. In mature bucks, short treatment by injection of Tri-B induced increase in semen quality, i.e., ejaculate volume, sperm motility and sperm-cell concentration and decrease in dead sperm, sperm abnormalities and acrosomal abnormalities. In pregnant does, treatment by cool water during hot period improved daily body gain, litter size, litter weight, feed consumption and blood components and significant decrease in percentage of mortality rate.

Y15, a specific inhibitor of FAK activity, was used to inhibit the activity of FAK under normal gravity; Y15 decreased protein expression of TRIB3.   5. Dissecting Daily and Circadian Expression Rhythms of Clock-Controlled Genes in Human Blood.

    The identification and investigation of novel clock-controlled genes (CCGs) has been conducted thus far mainly in model organisms such as nocturnal rodents, with limited information in humans. Here, we aimed to characterize daily and circadian expression rhythms of CCGs in human peripheral blood during a sleep/sleep deprivation (S/SD) study and a constant routine (CR) study. Blood expression levels of 9 candidate CCGs (SREBF1, TRIB1, USF1, THRA1, SIRT1, STAT3, CAPRIN1, MKNK2, and ROCK2), were measured across 48 h in 12 participants in the S/SD study and across 33 h in 12 participants in the CR study. Statistically significant rhythms in expression were observed for STAT3, SREBF1, TRIB1, and THRA1 in samples from both the S/SD and the CR studies, indicating that their rhythmicity is driven by the endogenous clock. The MKNK2 gene was significantly rhythmic in the S/SD but not the CR study, which implies its exogenously driven rhythmic expression. In addition, we confirmed the circadian expression of PER1, PER3, and REV-ERBα in the CR study samples, while BMAL1 and HSPA1B were not significantly rhythmic in the CR samples; all 5 genes previously showed significant expression in the S/SD study samples. Overall, our results demonstrate that rhythmic expression patterns of clock and selected clock-controlled genes in human blood cells are in part determined by exogenous factors (sleep and fasting state) and in part by the endogenous circadian timing system. Knowledge of the exogenous and endogenous regulation of gene expression rhythms is needed prior to the selection of potential candidate marker genes for future applications in medical and forensic settings. © 2015 The Author(s).

    Full Text Available Se evaluaron tres bronces: SAE 67, nacional y americano usados típicamente como material de la chumacera en los molinos de caña de azúcar. Se realizaron pruebas desgaste por deslizamiento bajo la norma G77 usando bloques de bronce, anillos de acero SAE 1045 y aceite como lubricante. El tribómetro se calibró usando los anillos de acero SAE 4620 y se usó acero de herramienta SAE O1 para los bloques. Se realizaron metalografías, durezas y rugosidad de los materiales estudiados. El bronce nacional presentó menor coeficiente de fricción y el bronce SAE 67 presentó menor desgaste tanto en el bloque, como en el anillo.

    Human acute myeloblastic leukemia HL-60 cells become resistant to differentiation during long-term cultivation. After 150 passages, double minute chromosomes (dmins) found in early-passaged cells are replaced by large extrachromosomal elements (LEEs). In a DNA library derived from a purified fraction of LEEs, 12.6% (23/183) of clones were assigned to 8q24 and 9.2% (17/183) were assigned to 14q11 in the human genome. Fluorescence in situ hybridization (FISH) revealed a small aberrant chromosome, which had not been found in early-passaged cells, in addition to the purified LEEs. We determined that each LEE consisted of six discontinuous segments in a region that extended for 4.4Mb over the 8q24 locus. Five genes, namely, Myc (a proto-oncogene), NSMCE2 (for a SUMO ligase), CCDC26 (for a retinoic acid-dependent modulator of myeloid differentiation), TRIB1 (for a regulator of MAPK kinase) and LOC389637 (for a protein of unknown function), were encoded by the amplicon. Breaks in the chromosomal DNA within the amplicon were found in the NSMCE2 and CCDC26 genes. The discontinuous structure of the amplicon unit of the LEEs was identical with that of dmins in HL-60 early-passaged cells. The difference between them seemed, predominantly, to be the number (10-15 copies per LEE versus 2 or 3 copies per dmin) of constituent units. Expression of the Myc, NSMCE2, CCDC26 and LOC389637 and TRIB1 genes was constitutive in all lines of HL-60 cells and that of the first four genes was repressed during the terminal differentiation of early-passaged HL-60 cells. We also detected abnormal transcripts of CCDC26. Our results suggest that these genes were selected during the development of amplicons. They might be amplified and, sometimes, truncated to contribute to the maintenance of HL-60 cells in an undifferentiated state.

  9. Fine-Resolution Modeling of the Santa Cruz and San Pedro River Basins for Climate Change and Riparian System Studies

    This project is part of a multidisciplinary effort aimed at understanding the impacts of climate variability and change on the ecological services provided by riparian ecosystems in semiarid watersheds of the southwestern United States. Valuing the environmental and recreational services provided by these ecosystems in the future requires a numerical simulation approach to estimate streamflow in ungauged tributaries as well as diffuse and direct recharge to groundwater basins. In this work, we utilize a distributed hydrologic model known as the TIN-based Real-time Integrated Basin Simulator (tRIBS) in the upper Santa Cruz and San Pedro basins with the goal of generating simulated hydrological fields that will be coupled to a riparian groundwater model. With the distributed model, we will evaluate a set of climate change and population scenarios to quantify future conditions in these two river systems and their impacts on flood peaks, recharge events and low flows. Here, we present a model confidence building exercise based on high performance computing (HPC) runs of the tRIBS model in both basins during the period of 1990-2000. Distributed model simulations utilize best-available data across the US-Mexico border on topography, land cover and soils obtained from analysis of remotely-sensed imagery and government databases. Meteorological forcing over the historical period is obtained from a combination of sparse ground networks and weather radar rainfall estimates. We then focus on a comparison between simulation runs using ground-based forcing to cases where the Weather Research Forecast (WRF) model is used to specify the historical conditions. Two spatial resolutions are considered from the WRF model fields - a coarse (35-km) and a downscaled (10- km) forcing. Comparisons will focus on the distribution of precipitation, soil moisture, runoff generation and recharge and assess the value of the WRF coarse and downscaled products. Gustatory receptors (Grs) that regulate external signals in chemosensory organs have been found in many insects. Tribolium castaneum, a major pest of stored products, possesses over 200 Gr genes. An expanded repertoire of Gr genes appears to be required for diet recognition in species that are generalist feeders; however, it remains unclear whether T. castaneum recognizes a suite of chemicals common to many products or whether its feeding is activated by specific chemicals, and whether its Grs are involved in feeding behavior. It is difficult to determine the food preferences of T. castaneum based on dietary intake due to a lack of appropriate methodology. This study established a novel dietary intake estimation method using gypsum, designated the TribUTE (Tribolium Urges To Eat) assay. For this assay, T. castaneum adults were fed a gypsum block without added organic compounds. Sweet preference was determined by adding sweeteners and measuring the amount of gypsum in the excreta. Mannitol was the strongest activator of T. castaneum dietary intake. In a Xenopus oocyte expression, TcGr20 was found to be responsible for mannitol and sorbitol responses, but not for responses to other tested non-volatile compounds. The EC50 values of TcGr20 for mannitol and sorbitol were 72.6 mM and 90.6 mM, respectively, suggesting that TcGr20 is a feasible receptor for the recognition of mannitol at lower concentrations. We used RNAi and the TribUTE assay to examine whether TcGr20 expression was involved in mannitol recognition. The amounts of excreta in TcGr20 dsRNA-injected adults decreased significantly, despite the presence of mannitol, compared to control adults. Taken together, our results indicate that T. castaneum adults recognized mannitol/sorbitol using the TcGr20 receptor, thereby facilitating the dietary intake of these compounds.

  12. Phosphorus Export Model Development in a Terminal Lake Basin using Concentration-Streamflow Relationship

    Nutrient export to streams and lakes by anthropogenic activities can lead to eutrophication and degradation of surface water quality. In Devils Lake, ND, the only terminal lake in the Northern Great Plains, the algae boom is of great concern due to the recent increase in streamflow and consequent rise in phosphorus (P) export from prairie agricultural fields. However, to date, very few studies explored the concentration (c) -streamflow (q) relationship in the headwater catchments of the Devils Lake basin. A robust watershed-scale quantitative framework would aid understanding of the c-q relationship, simulating P concentration and load. In this study, we utilize c-q relationships to develop a simple model to estimate phosphorus concentration and export from two headwater catchments of different size (Mauvais Coulee: 1032 km2 and Trib 3: 160 km2) draining to Devils Lake. Our goal is to link the phosphorus export model with a physically based hydrologic model to identify major drivers of phosphorus export. USGS provided the streamflow measurements, and we collected water samples (filtered and unfiltered) three times daily during the spring snowmelt season (March 31, 2017- April 12, 2017) at the outlets of both headwater catchments. Our results indicate that most P is dissolved and very little is particulate, suggesting little export of fine-grained sediment from agricultural fields. Our preliminary analyses in the Mauvais Coulee catchment show a chemostatic c-q relationship in the rising limb of the hydrograph, while the recession limb shows a linear and positive c-q relationship. The poor correlation in the rising limb of the hydrograph suggests intense flushing of P by spring snowmelt runoff. Flushing then continues in the recession limb of the hydrograph, but at a more constant rate. The estimated total P load for the Mauvais Coulee basin is 193 kg/km2, consistent with other catchments of similar size across the Red River of the North basin to the east. We expect

    The oncogenic transcription factor TAL1/SCL is aberrantly expressed in over 40% of cases of human T cell acute lymphoblastic leukemia (T-ALL), emphasizing its importance in the molecular pathogenesis of T-ALL. Here we identify the core transcriptional regulatory circuit controlled by TAL1 and its regulatory partners HEB, E2A, LMO1/2, GATA3, and RUNX1. We show that TAL1 forms a positive interconnected autoregulatory loop with GATA3 and RUNX1 and that the TAL1 complex directly activates the MYB oncogene, forming a positive feed-forward regulatory loop that reinforces and stabilizes the TAL1-regulated oncogenic program. One of the critical downstream targets in this circuitry is the TRIB2 gene, which is oppositely regulated by TAL1 and E2A/HEB and is essential for the survival of T-ALL cells.     Full Text Available Se obtuvieron recubrimientos monocapa de CrC por evaporación por arco catódico (EAC. Se investigaron las propiedades tribológicas de los recubrimientos mediante pruebas de deslizamiento en seco usando un tribómetro de bola sobre disco. Las propiedades mecánicas se estudiaron por nanoindentación dinámica. Los valores de los coeficientes de fricción de los recubrimientos monocapa de CrC mostraron buena correlación con el contenido de carbono de estos recubrimientos, presentando el más bajo coeficiente de fricción el recubrimiento con el mayor contenido de carbono. Después de las pruebas de fricción, tanto los recubrimientos como las esferas de acero usadas como contracuerpos presentaron partículas de desgaste de color marrón que sugieren la presencia de productos de reacciones triboquímicas. Las mejores propiedades mecánicas fueron obtenidas para los recubrimientos CrC con el más bajo contenido de carbono.

    Full Text Available The present study reviews the taxonomy of Late Jurassic–Early Cretaceous oysters from the Northern and the Subpolar Urals (Western Siberia and northern East Siberia. Previous studies have documented 10 species from the genus Liostrea (L. delta, L. cucurbita, L. praeanabarensis, L. anabarensis, L. plastica, L. gibberosa, L. planoconvexa, L. siberica, L. uralensis, L. lyapinensis, and 3 species from the genus Gryphaea (G. borealis and 2 species in open nomenclature. Liostrea gibberosa, L. planoconvexa, L. uralensis, and L. cucurbita are transferred in this study to the genus Pernostrea. Furthermore, two new species of Pernostrea are described: P. mesezhnikovi sp. nov. and P.? robusta sp. nov. Liostrea siberica is transferred to the genus Praeexogyra. Liostrea praeanabarensis and L. anabarensis are attributed to the subgenus Boreiodeltoideum (genus Deltoideum as well as L. delta sensu Zakharov (1966 which is described here as new species Deltoideum (Boreiodeltoideum borealis sp. nov. The similar shell morphology of the genera Deltoideum and Pernostrea provides a basis to establish the new tribe Pernostreini trib. nov. in the subfamily Gryphaeinae.     Full Text Available El objetivo de este trabajo fue evaluar y caracterizar el comportamiento mecánico en desgaste del acero API 5L X65, revestido con niobio en comparación al desempeño del revestimiento de la aleación de inconel 625 empleados en la industria de petróleo y gas. El revestimiento de niobio fue obtenido por el proceso de aspersión térmica a plasma de arco no transferido y el revestimiento inconel 625 por soldadura con electrodo revestido. La resistencia al desgaste por abrasión fue evaluada según la norma Petrobras N-2568, en un tribómetro CTER, la rugosidad y el volumen de material desgastado se determinó a través de perfilometría y la dureza de los revestimientos por microscopia Vickers. Los revestimientos obtenidos fueron caracterizados respecto a su morfología por microscopia electrónica de barrido (MEB y microscopía óptica (MO. La mayor dureza del revestimiento con niobio obtenido puede haber contribuido a reducir la tasa de desgaste en comparación con el revestimiento de inconel 625.

    Insects and fungi have a long history of association in shared habitats. Fungus-feeding, or mycophagy, is remarkably widespread in beetles (Coleoptera) and appears to be a primitive feeding habit that preceded feeding on plant tissues. Numerous Mesozoic beetles belonging to extant fungus-associated families are known, but direct fossil evidence elucidating mycophagy in insects has remained elusive. Here, we report a remarkable genus and species, Vetuproteinus cretaceus gen. et sp. nov., belonging to a new tribe (Vetuproteinini trib. nov.) of the extant rove beetle subfamily Proteininae (Staphylinidae) in Mid-Cretaceous Burmese amber. The mouthparts of this beetle have a markedly enlarged protruding galea bearing an apparent spore brush, a specialized structure we infer was used to scrape spores off surfaces and direct them into the mouth, as in multiple modern spore-feeding beetles. Considering the long evolutionary history of Fungi, the Mid-Cretaceous beetles likely fed on ancient Basidiomycota and/or Ascomycota fungi or spore-producing organisms such as slime moulds (Myxomycetes). The discovery of the first Mesozoic proteinine illustrates the antiquity of the subfamily, and suggests that ancestral Proteininae were already diverse and widespread in Pangaea before the supercontinent broke up. © 2016 The Author(s).

  19. Construcción y puesta en funcionamiento de una máquina de cilindros cruzados para la evaluación del desgaste adhesivo

    Full Text Available En esta investigación se evaluó el desgaste adhesivo de cuatro metales (fundición gris, latón 60- 40, bronce fosforado y un acero AISI 1020, en un tribómetro de cilindros cruzados, bajo la norma ASTM G- 83-96. Para lograr dicho objetivo, se diseñó, construyó y puso en marcha una máquina de cilindros cruzados, la cual contiene dos probetas cilíndricas (una estática y una giratoria, que fueron maquinadas y pulidas con lijas hasta obtener una superficie con una rugosidad máxima de 0.34 mm. Con el volumen de metal perdido por cada material, se calculó su velocidad promedio de corrosión y se determinó el coeficiente de desgaste. Además, se realizaron ensayos a cada material, para determinar los perfiles de desgaste respectivos (Gramos de metal perdido vs Tiempo. En este trabajo se obtuvo un coeficiente de variación máximo, entre los cuatro metales, de 4.3%, indicando una buena correlación de los resultados obtenidos. Los coeficientes de desgaste se consideraron aceptables, puesto que sus valores corresponden y son del mismo orden a los establecidos en la literatura. Igualmente, los perfiles del desgaste mostraron las tres etapas del proceso, similares a los encontrados en otros trabajos realizados.

    Genome-wide association studies (GWAS have revealed numerous associations between many phenotypes and gene candidates. Frequently, however, further elucidation of gene function has not been achieved. A recent GWAS identified 69 candidate genes associated with elevated liver enzyme concentrations, which are clinical markers of liver disease. To investigate the role of these genes in liver homeostasis, we narrowed down this list to 12 genes based on zebrafish orthology, zebrafish liver expression and disease correlation. To assess the function of gene candidates during liver development, we assayed hepatic progenitors at 48 hours post fertilization (hpf and hepatocytes at 72 hpf using in situ hybridization following morpholino knockdown in zebrafish embryos. Knockdown of three genes (pnpla3, pklr and mapk10 decreased expression of hepatic progenitor cells, whereas knockdown of eight genes (pnpla3, cpn1, trib1, fads2, slc2a2, pklr, mapk10 and samm50 decreased cell-specific hepatocyte expression. We then induced liver injury in zebrafish embryos using acetaminophen exposure and observed changes in liver toxicity incidence in morphants. Prioritization of GWAS candidates and morpholino knockdown expedites the study of newly identified genes impacting liver development and represents a feasible method for initial assessment of candidate genes to instruct further mechanistic analyses. Our analysis can be extended to GWAS for additional disease-associated phenotypes.

    The anareolate New World subfamily Cladomorphinae Bradley & Galil, 1977 is reviewed and keys to the six tribes currently included are presented; these are: Cladomorphini Bradley & Galil, 1977, Cladoxerini Karny, 1923, Cranidiini Günther, 1953, Pterinoxylini n. trib., Hesperophasmatini Bradley & Galil, 1977 and Haplopodini Günther, 1953 rev. stat.. New diagnoses are presented for all these tribes and possible relationships within Cladomorphinae are discusssed. Morphology of the genitalia and egg-structures indicate Cladomorphinae as presently treated to be polyphyletic. Two subordinate groups are recognized within present Cladomorphinae, which differ considerably in numerous morphological characters of the insects and eggs. The first group and here regarded as Cladomorphinae sensu stricto is formed by the mostly South American Cladomorphini + Cranidiini + Cladoxerini, while the second group is formed by the predominantly Caribbean Hesperophasmatini + Pterinoxylini n. trib. + Haplopodini.        Members of the first group (= Cladomorphini sensu stricto) share the dorsally carinate basitarsus in which the two dorsal carinae are melted with another, increasingly elongated gonapophyses VIII of females which are noticeably longer than gonapophyses IX and lamellate as well as strongly displaced medioventral carina of the profemora. Cranidiini + Cladomorphini share the strongly elongated and filiform gonapophyses VIII and presence of gonoplacs in the females, specialized poculum of males and presence of a median line in the eggs. Cranidiini differs from all other tribes of Cladomorphinae by the entirely unarmed legs of both sexes, distinctly broadened and leaf-like body and prominent longitudinal keel of the mesosternum of females, prominently enlarged poculum and spinulose phallus of males as well as the conspicuous narrowing of the posteromedian gap of the internal micropylar plate of the eggs and noticeably separated median line.     Full Text Available Behavioral endophenotypes are determined by a multitude of counteracting but precisely balanced molecular and physiological mechanisms. In this study, we aim to identify potential novel molecular targets that contribute to the multigenic trait "anxiety". We used microarrays to investigate the gene expression profiles of different brain regions within the limbic system of mice which were selectively bred for either high (HAB or low (LAB anxiety-related behavior, and also show signs of comorbid depression-like behavior. We identified and confirmed sex-independent differences in the basal expression of 13 candidate genes, using tissue from the entire brain, including coronin 7 (Coro7, cathepsin B (Ctsb, muscleblind-like 1 (Mbnl1, metallothionein 1 (Mt1, solute carrier family 25 member 17 (Slc25a17, tribbles homolog 2 (Trib2, zinc finger protein 672 (Zfp672, syntaxin 3 (Stx3, ATP-binding cassette, sub-family A member 2 (Abca2, ectonucleotide pyrophosphatase/phosphodiesterase 5 (Enpp5, high mobility group nucleosomal binding domain 3 (Hmgn3 and pyruvate dehydrogenase beta (Pdhb. Additionally, we confirmed brain region-specific differences in the expression of synaptotagmin 4 (Syt4.Our identification of about 90 polymorphisms in Ctsb suggested that this gene might play a critical role in shaping our mouse model's behavioral endophenotypes. Indeed, the assessment of anxiety-related and depression-like behaviors of Ctsb knock-out mice revealed an increase in depression-like behavior in females. Altogether, our results suggest that Ctsb has significant effects on emotionality, irrespective of the tested mouse strain, making it a promising target for future pharmacotherapy.

  4. Additive Effects of Millimeter Waves and 2-Deoxyglucose Co-Exposure on the Human Keratinocyte Transcriptome.

    Millimeter Waves (MMW) will be used in the next-generation of high-speed wireless technologies, especially in future Ultra-Broadband small cells in 5G cellular networks. Therefore, their biocompatibilities must be evaluated prior to their massive deployment. Using a microarray-based approach, we analyzed modifications to the whole genome of a human keratinocyte model that was exposed at 60.4 GHz-MMW at an incident power density (IPD) of 20 mW/cm2 for 3 hours in athermic conditions. No keratinocyte transcriptome modifications were observed. We tested the effects of MMWs on cell metabolism by co-treating MMW-exposed cells with a glycolysis inhibitor, 2-deoxyglucose (2dG, 20 mM for 3 hours), and whole genome expression was evaluated along with the ATP content. We found that the 2dG treatment decreased the cellular ATP content and induced a high modification in the transcriptome (632 coding genes). The affected genes were associated with transcriptional repression, cellular communication and endoplasmic reticulum homeostasis. The MMW/2dG co-treatment did not alter the keratinocyte ATP content, but it did slightly alter the transcriptome, which reflected the capacity of MMW to interfere with the bioenergetic stress response. The RT-PCR-based validation confirmed 6 MMW-sensitive genes (SOCS3, SPRY2, TRIB1, FAM46A, CSRNP1 and PPP1R15A) during the 2dG treatment. These 6 genes encoded transcription factors or inhibitors of cytokine pathways, which raised questions regarding the potential impact of long-term or chronic MMW exposure on metabolically stressed cells.

    Full Text Available El acero AISI 4140 es un acero de baja aleación al cromo/molibdeno de elevada resistencia mecánica y buena tenacidad, el cual se utiliza para la fabricación de piezas de maquinaria como piñones, cigüeñales, ejes de levas, etc. Sin embargo, su resistencia al desgaste es bastante limitada y suele ser mejorada mediante el tratamiento superficial de la nitruración. El propósito de este trabajo consistió en mejorar las propiedades mecánicas e incrementar la resistencia al desgaste del acero 4140 mediante la aplicación de un tratamiento superficial DUPLEX consistente en la nitruración líquida (conocida como Teniffer seguido del depósito de un recubrimiento duro de nitruro de titanio (TiN. La microestructura cristalina fue evaluada mediante difracción de rayos X, la morfología y la composición química fue determinada por microscopía SEM y espectroscopía de energía dispersiva de rayos X (EDS, las propiedades tribológicas fueron analizadas mediante mediciones en un tribómetro de tipo esfera sobre disco y la microdureza se determinó por el método Vickers. Las muestras tratadas con el recubrimiento dúplex mostraron una resistencia al desgaste 4 veces mayor que la muestra en estado bonificado, lo cual demuestra la aplicación potencial que tiene el acero 4140 sometido a este tratamiento dúplex.

  6. A Participatory Modeling Application of a Distributed Hydrologic Model in Nuevo Leon, Mexico for the 2010 Hurricane Alex Flood Event

    A distributed hydrologic model can help bring consensus among diverse stakeholders in regional flood planning by producing quantifiable sets of alternative futures. This value is acute in areas with high uncertainties in hydrologic conditions and sparse observations. In this study, we conduct an application of the Triangulated Irregular Network (TIN)-based Real-time Integrated Basin Simulator (tRIBS) in the Santa Catarina basin of Nuevo Leon, Mexico, where Hurricane Alex in July 2010 led to catastrophic flooding of the capital city of Monterrey. Distributed model simulations utilize best-available information on the regional topography, land cover, and soils obtained from Mexican government agencies or analysis of remotely-sensed imagery from MODIS and ASTER. Furthermore, we developed meteorological forcing for the flood event based on multiple data sources, including three local gauge networks, satellite-based estimates from TRMM and PERSIANN, and the North American Land Data Assimilation System (NLDAS). Remotely-sensed data allowed us to quantify rainfall distributions in the upland, rural portions of the Santa Catarina that are sparsely populated and ungauged. Rural areas had significant contributions to the flood event and as a result were considered by stakeholders for flood control measures, including new reservoirs and upland vegetation management. Participatory modeling workshops with the stakeholders revealed a disconnect between urban and rural populations in regard to understanding the hydrologic conditions of the flood event and the effectiveness of existing and potential flood control measures. Despite these challenges, the use of the distributed flood forecasts developed within this participatory framework facilitated building consensus among diverse stakeholders and exploring alternative futures in the basin.

    Full Text Available Certain concepts concerning EPO/EPOR action modes have been challenged by in vivo studies: Bcl-x levels are elevated in maturing erythroblasts, but not in their progenitors; truncated EPOR alleles that lack a major p85/PI3K recruitment site nonetheless promote polycythemia; and Erk1 disruption unexpectedly bolsters erythropoiesis. To discover novel EPO/EPOR action routes, global transcriptome analyses presently are applied to interrogate EPO/EPOR effects on primary bone marrow-derived CFUe-like progenitors. Overall, 160 EPO/EPOR target transcripts were significantly modulated 2-to 21.8-fold. A unique set of EPO-regulated survival factors included Lyl1, Gas5, Pim3, Pim1, Bim, Trib3 and Serpina 3g. EPO/EPOR-modulated cell cycle mediators included Cdc25a, Btg3, Cyclin-d2, p27-kip1, Cyclin-g2 and CyclinB1-IP-1. EPO regulation of signal transduction factors was also interestingly complex. For example, not only Socs3 plus Socs2 but also Spred2, Spred1 and Eaf1 were EPO-induced as negative-feedback components. Socs2, plus five additional targets, further proved to comprise new EPOR/Jak2/Stat5 response genes (which are important for erythropoiesis during anemia. Among receptors, an atypical TNF-receptor Tnfr-sf13c was up-modulated >5-fold by EPO. Functionally, Tnfr-sf13c ligation proved to both promote proerythroblast survival, and substantially enhance erythroblast formation. The EPOR therefore engages a sophisticated set of transcriptome response circuits, with Tnfr-sf13c deployed as one novel positive regulator of proerythroblast formation.

  9. A genome-wide association study for body weight in Japanese Thoroughbred racehorses clarifies candidate regions on chromosomes 3, 9, 15, and 18

    ABSTRACT Body weight is an important trait to confirm growth and development in humans and animals. In Thoroughbred racehorses, it is measured in the postnatal, training, and racing periods to evaluate growth and training degrees. The body weight of mature Thoroughbred racehorses generally ranges from 400 to 600 kg, and this broad range is likely influenced by environmental and genetic factors. Therefore, a genome-wide association study (GWAS) using the Equine SNP70 BeadChip was performed to identify the genomic regions associated with body weight in Japanese Thoroughbred racehorses using 851 individuals. The average body weight of these horses was 473.9 kg (standard deviation: 28.0) at the age of 3, and GWAS identified statistically significant SNPs on chromosomes 3 (BIEC2_808466, P=2.32E-14), 9 (BIEC2_1105503, P=1.03E-7), 15 (BIEC2_322669, P=9.50E-6), and 18 (BIEC2_417274, P=1.44E-14), which were associated with body weight as a quantitative trait. The genomic regions on chromosomes 3, 9, 15, and 18 included ligand-dependent nuclear receptor compressor-like protein (LCORL), zinc finger and AT hook domain containing (ZFAT), tribbles pseudokinase 2 (TRIB2), and myostatin (MSTN), respectively, as candidate genes. LCORL and ZFAT are associated with withers height in horses, whereas MSTN affects muscle mass. Thus, the genomic regions identified in this study seem to affect the body weight of Thoroughbred racehorses. Although this information is useful for breeding and growth management of the horses, the production of genetically modified animals and gene doping (abuse/misuse of gene therapy) should be prohibited to maintain horse racing integrity.   10. Estudio de la resistencia a la abrasión de capas vitrocerámicas depositadas sobre acero

    Full Text Available Wear test has been completed on two types of layered materials: a non-reinforced enamel plate and a ceramic reinforced enamel plate. The latter has been tested in two forms: with 10 wt. % and 20 wt. % ceramic particles reinforcement. For comparison the same test has also been completed on polished steel. Wear resistance was measure using a tribometer, equipped with a chromium steel sphere of 6 mm diameter. The sphere-plate surface contact area increases with distance covered, which in the polished steel provokes exponential increments in wear values. This exponential behaviour was not observed in the enamel plates due to their greater wear resistance. The reinforcement of the enamel plates with ceramic particles increases the wear resistance. A higher percentage of ceramic reinforcement results in a greater wear resistance of the plates.

    Se han realizado ensayos de desgaste, tanto en placas esmaltadas sin refuerzo como en esmaltes con distinto porcentaje de carga cerámica (10 y 20% en peso, y se han comparado frente al del acero base pulido. Las medidas de resistencia a la abrasión se han obtenido utilizado un tribómetro con bola de acero al cromo de 6 mm de diámetro. El aumento de la superficie de contacto bola-placa a medida que aumenta la distancia recorrida, provoca aumentos exponenciales en los desgastes en el caso del acero pulido. Sin embargo, en el caso de las chapas esmaltadas no se observa este comportamiento, debido a la mayor resistencia al desgaste de las mismas. Cuando las capas esmaltadas están reforzadas con cargas cerámicas, la resistencia al desgaste aumenta con el porcentaje de refuerzo cerámico.

  11. Distributed Hydrologic Modeling of Semiarid Basins in Arizona: A Platform for Land Cover and Climate Change Assessments

    Watershed management is challenged by rising concerns over climate change and its potential to interact with land cover alterations to impact regional water supplies and hydrologic processes. The inability to conduct experimental manipulations that address climate and land cover change at watershed scales limits the capacity of water managers to make decisions to protect future supplies. As a result, spatially-explicit, physically-based models possess value for predicting the possible consequences on watershed hydrology. In this study, we apply a distributed watershed model, the Triangulated Irregular Network (TIN)-based Real-time Integrated Basin Simulator (tRIBS), to the Beaver Creek basin in Arizona. This sub-basin of the Verde River is representative of the regional topography, land cover, soils distribution and availability of hydrologic data in forested regions of northern Arizona. As such, it can serve as a demonstration study in the broader region to illustrate the utility of distributed models for change assessment studies. Through a model application to summertime conditions, we compare the hydrologic response from three sources of meteorological input: (1) an available network of ground-based stations, (2) weather radar rainfall estimates, and (3) the North American Land Data Assimilation System (NLDAS). Comparisons focus on analysis of spatiotemporal distributions of precipitation, soil moisture, runoff generation, evapotranspiration and recharge from the root zone at high resolution for an assessment of sustainable water supplies for agricultural and domestic purposes. We also present a preliminary analysis of the impact of vegetation change arising from historical treatments in the Beaver Creek to inform the hydrologic consequences in the form of soil moisture and evapotranspiration patterns with differing degrees of proposed forest thinning.   12. Obesity challenges the hepatoprotective function of the integrated stress response to asparaginase exposure in mice.

    Obesity increases risk for liver toxicity by the anti-leukemic agent asparaginase, but the mechanism is unknown. Asparaginase activates the integrated stress response (ISR) via sensing amino acid depletion by the eukaryotic initiation factor 2 (eIF2) kinase GCN2. The goal of this work was to discern the impact of obesity, alone versus alongside genetic disruption of the ISR, on mechanisms of liver protection during chronic asparaginase exposure in mice. Following diet-induced obesity, biochemical analysis of livers revealed that asparaginase provoked hepatic steatosis that coincided with activation of another eIF2 kinase PKR-like endoplasmic reticulum kinase (PERK), a major ISR transducer to ER stress. Genetic loss of Gcn2 intensified hepatic PERK activation to asparaginase, yet surprisingly, mRNA levels of key ISR gene targets such as Atf5 and Trib3 failed to increase. Instead, mechanistic target of rapamycin complex 1 (mTORC1) signal transduction was unleashed, and this coincided with liver dysfunction reflected by a failure to maintain hydrogen sulfide production or apolipoprotein B100 (ApoB100) expression. In contrast, obese mice lacking hepatic activating transcription factor 4 ( Atf4 ) showed an exaggerated ISR and greater loss of endogenous hydrogen sulfide but normal inhibition of mTORC1 and maintenance of ApoB100 during asparaginase exposure. In both genetic mouse models, expression and phosphorylation of Sestrin2, an ATF4 gene target, was increased by asparaginase, suggesting mTORC1 inhibition during asparaginase exposure is not driven via eIF2-ATF4-Sestrin2. In conclusion, obesity promotes a maladaptive ISR during asparaginase exposure. GCN2 functions to repress mTORC1 activity and maintain ApoB100 protein levels independently of Atf4 expression, whereas hydrogen sulfide production is promoted via GCN2-ATF4 pathway. © 2017 by The American Society for Biochemistry and Molecular Biology, Inc.

  13. Effect of Downscaled Forcings and Soil Texture Properties on Hyperresolution Hydrologic Simulations in a Regional Basin in Northwest Mexico

    Hyper-resolution ( 10 km) scales. In this study, we address some of the challenges by applying a parallel version of the Triangulated Irregular Network (TIN)-based Real Time Integrated Basin Simulator (tRIBS) to the Rio Sonora Basin (RSB) in northwest Mexico. The RSB is a large, semiarid watershed ( 21,000 km2) characterized by complex topography and a strong seasonality in vegetation conditions, due to the North American monsoon. We conducted simulations at an average spatial resolution of 88 m over a decadal (2004-2013) period using spatially-distributed forcings from remotely-sensed and reanalysis products. Meteorological forcings were derived from the North American Land Data Assimilation System (NLDAS) at the original resolution of 12 km and were downscaled at 1 km with techniques accounting for terrain effects. Two grids of soil properties were created from different sources, including: (i) CONABIO (Comisión Nacional para el Conocimiento y Uso de la Biodiversidad) at 6 km resolution; and (ii) ISRIC (International Soil Reference Information Centre) at 250 m. Time-varying vegetation parameters were derived from the Moderate Resolution Imaging Spectroradiometer (MODIS) composite products. The model was first calibrated and validated through distributed soil moisture data from a network of 20 soil moisture stations during the monsoon season. Next, hydrologic simulations were conducted with five different combinations of coarse and downscaled forcings and soil properties. Outputs in the different configurations were then compared with independent observations of soil moisture, and with estimates of land surface temperature (1 km, daily) and evapotranspiration (1 km, monthly) from MODIS. This study is expected to support the community involved in hyper-resolution hydrologic modeling by identifying the crucial factors that, if available at higher resolution, lead to the largest improvement of the simulation prognostic capability.

Los aceites vegetales son ampliamente usados en muchas formulaciones de grasas. En este trabajo se determinaron las propiedades fisicoquímicas y tribológicas de una grasa lubricante fabricada con aceite vegetal de Jatropha curcas L y jabón de litio (JC1G5. Alfa tocoferol acetato y grafito fueron usados como aditivos antioxidante y de extrema presión respectivamente. La grasa JC1G5 mostró un color negro, una temperatura de goteo de 140 oC, apropiadas propiedades de corrosión al cobre y un grado de consistencia NLGI Grado1. El efecto de la lubricación de la grasa JC1G5 fue probado, empleando una máquina de cuatro bolas y un tribómetro timken.   15. Mechanism-based biomarker gene sets for glutathione depletion-related hepatotoxicity in rats

    Chemical-induced glutathione depletion is thought to be caused by two types of toxicological mechanisms: PHO-type glutathione depletion [glutathione conjugated with chemicals such as phorone (PHO) or diethyl maleate (DEM)], and BSO-type glutathione depletion [i.e., glutathione synthesis inhibited by chemicals such as L-buthionine-sulfoximine (BSO)]. In order to identify mechanism-based biomarker gene sets for glutathione depletion in rat liver, male SD rats were treated with various chemicals including PHO (40, 120 and 400 mg/kg), DEM (80, 240 and 800 mg/kg), BSO (150, 450 and 1500 mg/kg), and bromobenzene (BBZ, 10, 100 and 300 mg/kg). Liver samples were taken 3, 6, 9 and 24 h after administration and examined for hepatic glutathione content, physiological and pathological changes, and gene expression changes using Affymetrix GeneChip Arrays. To identify differentially expressed probe sets in response to glutathione depletion, we focused on the following two courses of events for the two types of mechanisms of glutathione depletion: a) gene expression changes occurring simultaneously in response to glutathione depletion, and b) gene expression changes after glutathione was depleted. The gene expression profiles of the identified probe sets for the two types of glutathione depletion differed markedly at times during and after glutathione depletion, whereas Srxn1 was markedly increased for both types as glutathione was depleted, suggesting that Srxn1 is a key molecule in oxidative stress related to glutathione. The extracted probe sets were refined and verified using various compounds including 13 additional positive or negative compounds, and they established two useful marker sets. One contained three probe sets (Akr7a3, Trib3 and Gstp1) that could detect conjugation-type glutathione depletors any time within 24 h after dosing, and the other contained 14 probe sets that could detect glutathione depletors by any mechanism.     Full Text Available Los recubrimientos de níquel químico (NQ constituyen un tratamiento superficial de altas prestaciones funcionales. La incorporación de micropartículas de teflón (PTFE da lugar a la formación de un "composite" (NQ-PTFE consistente en partículas de teflón (PTFE de tamaño micrométrico, uniformemente embebidas en una matriz de NQ. De esta manera, el recubrimiento resultante combina las propiedades de dureza del NQ con las excelentes propiedades de lubricación del teflón. En el presente trabajo se presentan los resultados de la caracterización tribológica de diferentes recubrimientos con la estructura sustrato NQ/NQ-PTFE depositados sobre acero al carbono, con una proporción de teflón presente en el NQ-PTFE de alrededor del 20 %. Los ensayos se realizaron en un tribómetro de bola sobre disco contra bola de alúmina, mediante los cuales se determinó el coeficiente de fricción. Se discuten los resultados de los recubrimientos NQ-PTFE obtenidos en distintas condiciones del baño químico y se comparan con los obtenidos para recubrimientos simples de NQ.Electroless nickel coating (EN is a highly functional engineered finish. A "composite" coating (EN-PTFE of electroless nickel and PTFE (Teflon is formed when PTFE micro-particles are uniformly dispersed throughout the coating. The resulting coating combines the corrosion resistance and hardness of EN with the excellent release properties and lubricity of PTFE. In this work we present the results of the tribological characterization of different coatings with a PTFE content of about 20 % and a structure Substrate EN-PTFE deposited on steel. The tribological characterization was conducted in a ball-on-disk tribometer using an alumina ball from which the friction coefficients were determined. The results for different ENPTFE coatings are discussed and compared to those obtained for simple EN coatings.

  18. Deep Percolation in Arid Piedmont Slopes: Multiple Lines of Evidence Show How Land Use Change and Ecohydrological Properties Affect Groundwater Recharge

    A critical hydrologic process in arid regions is the contribution of episodic streamflow in ephemeral channels to groundwater recharge. This process has traditionally been studied in channels that drain large watersheds (10s to 100s km2). In this study, we aim to characterize the provision of the ecosystem services of surface and groundwater supply in a first-order watershed (4.6 ha) in an arid piedmont slope of the Jornada Experimental Range (JER). We use an observational and modeling approach to estimate deep percolation. During a 6 year study period, we observed 428 mm of percolation (P) and 39 mm of runoff (Q); ratios of P to rainfall (R) of P/R = 0.27 and Q/R = 0.02. Utilizing an instrument network and site measurements, we determine that percolation occurs primarily inside channel reaches when these receive runoff from upland hillslopes and find that a monthly rainfall threshold of 62 mm is needed for significant percolation to be generated. In order to quantify the mechanisms leading to this threshold response, we develop a channel transmission loss module for the TIN-based Real-time Integrated Basin Simulator (tRIBS) and test the model thoroughly against the available observations over the study period. For these purposes, we make use of image classifications from Unmanned Aerial Vehicle flights, a ground-based phenocam, and species-level measurements to parameterize vegetation processes in the model. We then conduct an extensive set of sensitivity experiments to determine the relative roles of channel, soil, and vegetation properties on modifying the relation between monthly rainfall and percolation. Additionally, we test how the observed vegetation transitions in the JER over the last 150 years affect the deep percolation and runoff estimates. By quantifying mechanisms through which vegetation changes affect water resource provision, this work provides new insights on the ecohydrological controls on the water yield of arid piedmont slopes.

    Highlights: ► Nobiletin-mediated alterations of gene expression were examined with DNA microarrays. ► Three organ-derived cell lines were treated with 100 μM nobiletin for 24 h. ► In all cell lines, 3 endoplasmic reticulum stress-responsive genes were up-regulated. ► Some cell cycle-regulating and oxidative stress-promoting genes were down-regulated. ► These alterations may contribute to nobiletin-mediated biological effects. -- Abstract: Nobiletin, a polymethoxylated flavonoid that is highly contained in the peels of citrus fruits, exerts a wide variety of beneficial effects, including anti-proliferative effects in cancer cells, repressive effects in hyperlipidemia and hyperglycemia, and ameliorative effects in dementia at in vitro and in vivo levels. In the present study, to further understand the mechanisms of these actions of nobiletin, the nobiletin-mediated alterations of gene expression in three organ-derived cell lines – 3Y1 rat fibroblasts, HuH-7 human hepatocarcinoma cells, and SK-N-SH human neuroblastoma cells – were first examined with DNA microarrays. In all three cell lines, treatments with nobiletin (100 μM) for 24 h resulted in more than 200% increases in the expression levels of five genes, including the endoplasmic reticulum stress-responsive genes Ddit3, Trib3, and Asns, and in less than 50% decreases in the expression levels of seven genes, including the cell cycle-regulating genes Ccna2, Ccne2, and E2f8 and the oxidative stress-promoting gene Txnip. It was also confirmed that in each nobiletin-treated cell line, the levels of the DDIT3 (DNA-damage-inducible transcript 3, also known as CHOP and GADD153) and ASNS (asparagine synthetase) proteins were increased, while the level of the TXNIP (thioredoxin-interacting protein, also known as VDUP1 and TBP-2) protein was decreased.     Developmental exposure to cuprizone (CPZ), a demyelinating agent, impairs intermediate-stage neurogenesis in the hippocampal dentate gyrus of rat offspring. To investigate the possibility of alterations in adult neurogenesis following postpubertal exposure to CPZ in a framework of general toxicity studies, CPZ was orally administered to 5-week-old male rats at 0, 120, or 600 mg/kg body weight/day for 28 days. In the subgranular zone (SGZ), 600 mg/kg CPZ increased the number of cleaved caspase-3 + apoptotic cells. At ≥ 120 mg/kg, the number of SGZ cells immunoreactive for TBR2, doublecortin, or PCNA was decreased, while that for SOX2 was increased. In the granule cell layer, CPZ at ≥ 120 mg/kg decreased the number of postmitotic granule cells immunoreactive for NEUN, CHRNA7, ARC or FOS. In the dentate hilus, CPZ at ≥ 120 mg/kg decreased phosphorylated TRKB + interneurons, although the number of reelin + interneurons was unchanged. At 600 mg/kg, mRNA levels of Bdnf and Chrna7 were decreased, while those of Casp4, Casp12 and Trib3 were increased in the dentate gyrus. These data suggest that CPZ in a scheme of 28-day toxicity study causes endoplasmic reticulum stress-mediated apoptosis of granule cell lineages, resulting in aberrations of intermediate neurogenesis and late-stage neurogenesis and following suppression of immediate early gene-mediated neuronal plasticity. Suppression of BDNF signals to interneurons caused by decreased cholinergic signaling may play a role in these effects of CPZ.   1. Genetic influences on hand osteoarthritis in Finnish women--a replication study of candidate genes.

    Full Text Available OBJECTIVES: Our aims were to replicate some previously reported associations of single nucleotide polymorphisms (SNPs in five genes (A2BP1, COG5, GDF5, HFE, ESR1 with hand osteoarthritis (OA, and to examine whether genes (BCAP29, DIO2, DUS4L, DVWA, HLA, PTGS2, PARD3B, TGFB1 and TRIB1 associated with OA at other joint sites were associated with hand OA among Finnish women. DESIGN: We examined the bilateral hand radiographs of 542 occupationally active Finnish female dentists and teachers aged 45 to 63 and classified them according to the presence of OA by using reference images. Data regarding finger joint pain and other risk factors were collected using a questionnaire. We defined two hand OA phenotypes: radiographic OA in at least three joints (ROA and symptomatic DIP OA. The genotypes were determined by PCR-based methods. In statistical analysis, we used SNPStats software, the chi-square test and logistic regression. RESULTS: Of the SNPs, rs716508 in A2BP1 was associated with ROA (OR = 0.7, 95% CI 0.5-0.9 and rs1800470 in TGFB1 with symptomatic DIP OA (1.8, 1.2-2.9. We found an interaction between ESR1 (rs9340799 and occupation: teachers with the minor allele were at an increased risk of symptomatic DIP OA (2.8, 1.3-6.5. We saw no association among the dentists. We also found that the carriage of the COG5 rs3757713 C allele increased the risk of ROA only among women with the BCAP29 rs10953541 CC genotype (2.6; 1.1-6.1. There was also a suggestive interaction between the HFE rs179945 and the ESR1 rs9340799, and the carriage of the minor allele of either of these SNPs was associated with an increased risk of symptomatic DIP OA (2.1, 1.3-2.5. CONCLUSIONS: Our results support the earlier findings of A2BP1 and TBGF1 being OA susceptibility genes and provide evidence of a possible gene-gene interaction in the genetic influence on hand OA predisposition.

    Full Text Available The tribological behaviour of various Si3N4 based materials, including Si3N4/SiC composites, has been analysed from room temperature up to 700ºC, and for sliding speeds between 0.5 and 2 m/s. Materials have been prepared by hot pressing at 1750ºC in nitrogen atmosphere, varying α-Si3N4 phase content, microstructure coarseness and shape and size of the SiC particles. Wear tests were performed in self-mated pairs using a pin on disc tribometer, without lubrication and at a fixed load of 5 N. Pin and disc worn surfaces were observed by scanning electron microscopy (SEM-EDS. Debris generated during tests was analysed by SEM-EDS and X-ray diffraction (XRD. Results showed friction, f, and wear coefficients, K, above 0.3 y 10-6 mm3N-1m-1, respectively. Wear mechanism depended on the microstructure and the mechanical properties of the materials.Se ha analizado el comportamiento tribológico de materiales de Si3N4 y de compuestos Si3N4/SiC, desde temperatura ambiente hasta 700ºC y en el rango de velocidades 0.5-2 m/s. Los materiales se han preparado mediante prensado en caliente a 1750ºC en atmósfera de nitrógeno, variando el contenido en fase α-Si3N4, el tamaño medio de grano, y la morfología de las partículas de SiC adicionadas. Los ensayos se han realizado en pares homólogos utilizando un tribómetro del tipo punta sobre disco, sin lubricación y a una carga constante de 5 N. Las superficies de desgaste de las puntas y los discos se observaron en un microscopio electrónico de barrido con capacidad analítica (MEB-EDX. Los residuos generados durante los ensayos se analizaron mediante MEB-EDX y difracción de rayos X (DRX. Los resultados obtenidos en los ensayos daban valores de coeficiente de fricción, f, y de desgaste, K, superiores a 0.3 y 10-6 mm3N-1m-1, respectivamente. El mecanismo de desgaste predominante dependía de la microestructura y de las propiedades mecánicas del material ensayado.

  4. Identifying Genetic Differences Between Dongxiang Blue-Shelled and White Leghorn Chickens Using Sequencing Data

    Full Text Available The Dongxiang Blue-shelled chicken is one of the most valuable Chinese indigenous poultry breeds. However, compared to the Italian native White Leghorn, although this Chinese breed possesses numerous favorable characteristics, it also exhibits lower growth performance and fertility. Here, we utilized genotyping sequencing data obtained via genome reduction on a sequencing platform to detect 100,114 single nucleotide polymorphisms and perform further biological analysis and functional annotation. We employed cross-population extended haplotype homozygosity, eigenvector decomposition combined with genome-wide association studies (EigenGWAS, and efficient mixed-model association expedited methods to detect areas of the genome that are potential selected regions (PSR in both chicken breeds, and performed gene ontology (GO enrichment and quantitative trait loci (QTL analyses annotating using the Kyoto Encyclopedia of Genes and Genomes. The results of this study revealed a total of 2424 outlier loci (p-value <0.01, of which 2144 occur in the White Leghorn breed and 280 occur in the Dongxiang Blue-shelled chicken. These correspond to 327 and 94 PSRs containing 297 and 54 genes, respectively. The most significantly selected genes in Blue-shelled chicken are TMEM141 and CLIC3, while the SLCO1B3 gene, related to eggshell color, was identified via EigenGWAS. We show that the White Leghorn genes JARID2, RBMS3, GPC3, TRIB2, ROBO1, SAMSN1, OSBP2, and IGFALS are involved in immunity, reproduction, and growth, and thus might represent footprints of the selection process. In contrast, we identified six significantly enriched pathways in the Dongxiang Blue-shelled chicken that are related to amino acid and lipid metabolism as well as signal transduction. Our results also reveal the presence of a GO term associated with cell metabolism that occurs mainly in the White Leghorn breed, while the most significant QTL regions mapped to the Chicken QTL Database (GG_4

    Hoy es un día muy especial por haberme permitido acercarme a esta insigne Corporación, y a pesar de mis escasos méritos, recibirme como Miembro Honorario de la misma.

    Me doy cuenta de que entro a la Academia Nacional de Medicina que fue establecida por la Ley 71 de 1890, gracias al deseo expreso de los doctores Jorge Holguín, Adriano Tribín, Enrique de Narváez y Miguel Angel Peñarredonda de transformar la Sociedad nacida en el año de 1873 para el estudio y adelanto de las ciencias médicas y naturales, solidarizar al cuerpo médico y darle unidad al ejercicio profesional. Me parece sentir las voces gentiles de augustos caballeros como Manuel Plata Azuero, catedrático de Terapéutica y hábil parlamentario; Nicolás Osorio, eximio investigador y Decano de la Universidad; Liborio Zerda, extraordinario naturalista, apóstol de la educación; Leoncio Barreto, catedrático, farmaceuta y obstetra de gran prestigio; Evaristo García, catedrático polifacético y profundo humanista; y el joven médico bugueño Abraham Aparicio, más tarde Concejal y Alcalde de esta maravillosa ciudad de Bogotá.     Full Text Available In this study an analysis of the behavior of dry wear coefficient of a Metal Matrix Composite (MMC in 316 stainless steel reinforced with particles of titanium carbide (TiC according to ASTM G 99 standards, in a pin-on-disk test. In this research it is tested the effect of the percentage of reinforcement in the MMC manufactured with 3, 6 y 9% (vol. of TiC, in samples compacted at 800 MPa, generating different values of grain size, hardness and density, they are sintered with abnormal glow discharge, at a temperature of 1200 °C ±5 °C, with a protection atmosphere H2 – N2 and a permanence time of 30 minutes. According to the results obtained it is concluded that the best condition for the MMC manufacturing, in relation to the reinforcement percentage, is the one obtained when the mixture contains 6% of TiC compacted at 800 MPa. In these conditions, it was obtained: achieving smaller grain size, the greater hardness and the lowest coefficient of friction.     Nutritional strategies can decrease saturated fatty acids (SFAs) and increase health beneficial fatty acids (FAs) in bovine milk. The pathways/genes involved in these processes are not properly defined. Next-generation RNA-sequencing was used to investigate the bovine mammary gland transcriptome following supplemental feeding with 5% linseed oil (LSO) or 5% safflower oil (SFO). Holstein cows in mid-lactation were fed a control diet for 28 days (control period) followed by supplementation with 5% LSO (12 cows) or 5% SFO (12 cows) for 28 days (treatment period). Milk and mammary gland biopsies were sampled on days-14 (control period), +7 and +28 (treatment period). Milk was used to measure fat(FP)/protein(PP) percentages and individual FAs while RNA was subjected to sequencing. Milk FP was decreased by 30.38% (LSO) or 32.42% (SFO) while PP was unaffected (LSO) or increased (SFO). Several beneficial FAs were increased by LSO (C18:1n11t, CLA:10t12c, CLA:9c11t, C20:3n3, C20:5n3, C22:5n3) and SFO (C18:1n11t, CLA:10t12c, C20:1c11, C20:2, C20:3n3) while several SFAs (C4:0, C6:0, C8:0, C14:0, C16:0, C17:0, C24:0) were decreased by both treatments (P < 0.05). 1006 (460 up- and 546 down-regulated) and 199 (127 up- and 72 down-regulated) genes were significantly differentially regulated (DE) by LSO and SFO, respectively. Top regulated genes (≥ 2 fold change) by both treatments (FBP2, UCP2, TIEG2, ANGPTL4, ALDH1L2) are potential candidate genes for milk fat traits. Involvement of SCP2, PDK4, NQO1, F2RL1, DBI, CPT1A, CNTFR, CALB1, ACADVL, SPTLC3, PIK3CG, PIGZ, ADORA2B, TRIB3, HPGD, IGFBP2 and TXN in FA/lipid metabolism in dairy cows is being reported for the first time. Functional analysis indicated similar and different top enriched functions for DE genes. DE genes were predicted to significantly decrease synthesis of FA/lipid by both treatments and FA metabolism by LSO.     Cualquier aproximación que se quiera hacer a la historia de la práctica del infanticidio en las diferentes sociedades humanas a lo largo de los tiempos debe intentar ver ese acto a la luz de las relaciones entre la producción y la reproducción de cada sociedad en particular, entendiendo que todas las sociedades buscan a la larga, conciente o inconscientemente, un equilibrio entre los recursos de que disponen y el tamaño de la población y tratan de evitar saturar la capacidad de sustentación que ofrecen su entorno natural y su sistema de producción. (1

    Todas las sociedades humanas de la misma forma que se han preocupado por mantener e incrementar la producción de sus alimentos y bienes también se han preocupado por regular -estabilizando, aumentando o disminuyendo- su población.

    Con base en este referente teórico, he querido hacer unas breves consideraciones alrededor del infanticidio y del abandono de los niños a lo largo de nuestra prehistoria e historia hasta antes del siglo XX.

    ¿En esta perspectiva, las sociedades prehispánicas de la actual Colombia, ¿cómo regularon el crecimiento de su población? Ellas lo hicieron principalmente a través de cinco grandes estrategias: la lactancia materna prolongada; la abstinencia sexual, el aborto, el infanticidio, la discriminación de las niñas y mujeres y de una sexta, al parecer con menos impacto poblacional, por sus costos de vidas de varones jóvenes, como fueron los frecuentes y casi crónicos conflicto bélicos ínter – étnicos.     ón de ferrocarriles, de líneas telegráficas, de puentes y otras obras públicas, extensa legislación sobre expropiaciones, sobre pensiones, sobre gobierno de indios salvajes, sobre creación y financiación de lazaretos y sobre reformas a los procedimientos judiciales (ésta última con 25 títulos divididos en sus correspondientes capítulos y una extensión de 82 páginas. El presupuesto nacional apenas excedía los 20 millones de pesos y los auxilios del gobierno a los hospitales de las ciudades más grandes fluctuaban entre los 1.200 y los 3.000 pesos anuales. Llaman la atención, sin embargo, por su precisión y concisión los 9 artículos que forman la Ley de creación de la Academia;luego de fijar el número y la condición de miembros activos honorarios y correspondientes dice, en su artículo 50. que “la Academia dará al Gobierno los informes que se le pidan sobre puntos relacionados con las ciencias médicas y naturales y pasará al fin de cada año una relación de sus trabajos”; establece que la Academia dará dos premios anuales a los mejores trabajos que se le presenten sobre Medicina Nacional y dice que “el Gobierno proveerá a la Academia de un local adecuado para sus reuniones y formación de biblioteca, museo y conservación de sus archivos”, proveyéndola también, en su artículo 90. de un auxilio de 3.000 pesos anuales. La ley aparece firmada por don Jorge Holguín, presidente del Senado, y por don Adriano Tribín, presidente de la Cámara de Representantes.

    Qué contraste entre esa Colombia rural, pobretona y casi bucólica con la que nos tocaría vivir 100 años después. Cesaron las guerras civiles pero dieron paso a una guerra no declarada pero casi interminable entre diversas facciones no necesariamente políticas y que hace que el homicidio, la violencia y el trauma sean quizás la primera causa de mortalidad y morbilidad en los colombianos. La justicia y las leyes han dado paso a una maraña de procedimientos que por venales
