  1. Duration of hydrothermal treatment and peeling of 'Murcott' tangor

    Full Text Available Hydrothermal treatment facilitates the peeling of 'Pera' sweet orange fruit and does not alter its quality. The aim of this work was to adapt the technology of peeling for the use of hydrothermal treatment in 'Murcott' tangor and to evaluate its influence in the CO2 production and the physicochemical, microbiologic and sensorial characteristics of fruits. The peeling time, the yield of marketable fruits and the internal temperature of fruits during the treatment were also evaluated. The hydrothermal treatment consisted of placing the fruits in a water-bath at 50 ºC for 5, 10, 15, 20, 25 and 30 min. Fruits were peeled by first opening a gap in the peduncle region with a knife and then manually removing the flavedo and albedo. Fruits were stored at 5 ºC for six days. Hydrothermal treatment caused changes in the fruits' CO2 production for only the first few hours after processing. Internal fruit temperature after 30 min of treatment reached 35 ºC. There were no changes in the physicochemical and microbiologic characteristics of the fruits. The treatment did not change the flavor, improved the fruits' appearance, decreased the peeling time of the treated fruits by 57 % and increased the yield of marketable fruits. In conclusion, the hydrothermal treatment accomplished from 5 to 30 min at 50 ºC can be used as part of the peeling process for 'Murcott' tangor.

    Full Text Available This study aimed at evaluating compositional changes in the quality of 'Ortanique' tangor after coating with the carnauba-based waxes Aruá Tropical® or Star Light®. The storage conditions studied simulated those of local marketing (22 ± 2 °C, 60 ± 5% RH. Non-destructive analysis, mass loss, peel color, and sensory evaluation, were performed upon coating and every three days up to the fifteenth day of storage. Destructive analysis, peel moisture content, chlorophyll of the peel, pulp color, juice content, soluble solids (SS, titratable acidity (TA, pH, and soluble solids to titratable acidity ratio, were performed upon coating and every four days up to the sixteenth day of storage. The assay was conducted using an entirely randomized design, with three replications (destructive analyses or ten replications (non-destructive analyses, in a split plot scheme. Wax-coating, especially Aruá Tropical®, maintained fruit freshness by reducing mass loss and peel dehydration and retaining green color. Peel moisture content, chlorophyll content, and juice content had lower rates in the wax coated fruits. Puncture force, soluble solids, titratable acidity, pH, and soluble solids to titratable acidity ratio varied vary little over the course of storage. Sensory evaluation showed that the application of Aruá Tropical keeps 'Ortanique' tangor fresher for 6 days longer for commercialization.

    Full Text Available O objetivo deste trabalho foi avaliar a diversidade genética em uma população de 148 híbridos de tangor 'Murcott' (Citrus reticulata Blanco x C. sinensis L. Osbeck e laranja 'Pêra' (C. sinensis L. Osbeck obtidos por polinização controlada, pelo uso de marcadores fAFLP e RAPD. Marcadores polimórficos (416 marcadores fAFLP e 33 RAPD foram utilizados para avaliar a similaridade genética entre os híbridos, calculada com o coeficiente Jaccard pelo método UPGMA. A consistência de cada agrupamento foi determinada pelo programa BOOD. Houve alta similaridade genética entre os parentais. A laranja 'Pêra' apresentou maior número (132 de loci em heterozigose em relação ao tangor 'Murcott' (105, corroborando a teoria de origem híbrida para a laranja-doce. Observaram-se dois grupos distintos de plantas, e um deles abrangeu 80% dos híbridos com maior similaridade com a laranja 'Pêra'. A análise bootstrap não revelou consistência estatística entre esses grupos. Marcadores fAFLP são mais eficientes na avaliação do polimorfismo, sendo indicados para seleção de indivíduos híbridos mais próximos a um dos parentais.The objective of this work was to evaluate the genetic diversity in a population of 148 hybrids of 'Murcott' tangor (Citrus reticulata Blanco x C. sinensis L. Osbeck and 'Pêra' sweet orange (C. sinensis L. Osbeck, obtained by controlled polination, using fAFLP and RAPD markers.     Somatic hybrid plants were produced by protoplast fusion of navel orange and 'Murcott' tangor. Hybridity of the plants was confirmed by the restriction endonuclease analysis of nuclear ribosomal DNA. All of the plants (16 clones) were normal, uniform, and had the amphidiploid chromosome number of 36 (2n=2x=18 for each parent). The cpDNA analysis showed that each of the 16 somatic hybrids contained either one parental chloroplast genome or the other. In all cases, the mitochondrial genomes of the regenerated somatic hybrids were of the navel orange type.

    Full Text Available O valor de comercialização no mercado de fruta fresca e a maior resistência a doenças, como o declínio e a clorose variegada dos citros, comparado às variedades de laranjas, estimularam a produção de tangerinas e de 'Murcott' na última década. Por outro lado, faltam informações seguras para o diagnóstico das necessidades de adubação para esses cítricos. Assim, o presente trabalho foi planejado com os objetivos de estudar a demanda por nutrientes e estabelecer doses de fertilizantes para maximizar a produtividade e a qualidade dos frutos de tangor 'Murcott' e definir critérios de diagnóstico da análise de folhas para o manejo nutricional dessas plantas. O experimento foi desenvolvido durante seis anos, num pomar comercial da variedade Murcott sobre limoeiro 'Cravo', com 4 anos de idade. Os tratamentos foram arranjados no delineamento fatorial fracionado, do tipo ½ (4³ e constituídos por níveis de N (30; 100; 170 e 240 kg N ha-1, de P (20; 80; 140 e 200 kg ha-1 de P2O5 e de K (30; 110; 190 e 270 kg ha-1 de K2O. O N aumentou a produção média do período, enquanto o efeito do K foi negativo. Não houve efeito significativo para doses de P. A produção máxima de frutos foi obtida com as doses 155; 20 e 30 kg ha-1, respectivamente, de N, P2O5 e K2O, enquanto, para otimizar o tamanho dos frutos, foi necessário elevar a doses de K para 270 kg ha-1 de K2O.     Full Text Available Polpas de frutas podem ser utilizadas para elaboração de blends com características diferentes das matérias-primas originais, tendo por objetivo a melhoria das características sensoriais e nutricionais dos componentes isolados ou apenas a criação de novos produtos, cuja principal característica seja a diferenciação em relação aos produtos obtidos na natureza. Este trabalho foi realizado como o objetivo de se avaliar as características químicas e físico-químicas de blends elaborados a partir de polpa de tangor “Ortanique” e polpa de beterraba, com diferentes proporções de cada polpa. Os blends foram caracterizados quanto ao pH, o teor de sólidos solúveis totais, acidez total titulável, cinzas, teor de água, açúcares redutores, açúcares totais, açúcares não redutores e cor. A partir dos resultados obtidos observou-se que os blends de tangor e beterraba resultaram em amostras mais escuras com a elevação do teor de polpa de beterraba. O aumento na proporção de polpa de beterraba influenciou o pH, a acidez total titulável, o teor de água e os açúcares redutores. Aumentou com maior intensidade o teor de cinzas, os açúcares totais, os açúcares não redutores e o índice intensidade de vermelho, e reduziu a luminosidade.Palavras chave: hortaliças, frutas, Beta vulgaris, citrus

    Full Text Available A Clorose Variegada dos Citros (CVC, causada pela bactéria Xylella fastidiosa, é uma séria ameaça à citricultura brasileira, constituindo-se, atualmente, numa das principais doenças dos citros no Brasil. O objetivo desse trabalho foi avaliar cultivares de citros introduzidas quanto à suscetibilidade ou resistência à CVC, em condições de campo. O trabalho foi conduzido em Bebedouro-SP. Os materiais genéticos estudados foram cultivares de tangerinas e híbridos (tangores e tangelos introduzidas de bancos de germoplasma da Itália, Portugal, Espanha e Córsega. O trabalho foi constituído por 56 parcelas, com três plantas das quais uma foi inoculada, empregando-se o método de encostia, utilizando-se de mudas previamente infectadas como fontes da bactéria. Foram avaliados os sintomas da doença mediante observação visual através de notas e teste de PCR, específico para Xylella fastidiosa. Os materiais genéticos que se apresentaram positivos ao PCR, mas não apresentaram sintomas, e os que foram negativos ao PCR, possuem um potencial de utilização em programas de melhoramento genético visando à resistência e/ou à tolerância à doença.The Citrus Variegated Chlorosis (CVC, caused by the bacteria Xylella fastidiosa, is a serious threat to the Brazilian citriculture, being, actually, one of the mains diseases to the citrus in Brazil. The objective of this work was evaluate citrus cultivars, introduced according to the susceptibility or resistance to CVC, in field conditions. The research was carried in Bebedouro city - São Paulo state - Brazil. The genotypes studied were cultivars of mandarines and hybrids (tangors and tangelos, introduced from the germplasm collections of Italy, Portugal, Spain and Corsica.     Full Text Available O presente ensaio foi conduzido em cultivo comercial do tangor 'Murcote' e teve por objetivo avaliar a atuação dos biorreguladores 2,4-D (auxina, NAA (auxina, GA3 (giberelina e BA (citocinina, na redução do número de sementes, sem afetar a qualidade dos frutos cítricos. Os tratamentos foram: Testemunha; 10 e 20 mg.L-1 de 2,4-D; 100, 150 e 200 mg.L-1 de NAA, 100 e 200 mg.L-1 de GA3 e 20 e 40 mg.L-1 de BA. Verificou-se que nenhum dos reguladores vegetais influenciou na qualidade dos frutos de tangor 'murcote', sem redução de peso, tamanho e teor de sólidos solúveis totais. Já em relação ao número de sementes, nenhum dos reguladores vegetais foi efetivo na redução de sementes inviáveis, porém mostraram efeito na redução de sementes viáveis, conseqüentemente com redução do número total de sementes nos frutos, quando tratados com NAA a 100 e 200 mg.L-1 juntamente com GA3 a 100 mg.L-1, com redução de 30 % do total de sementes.The present experiment was conducted in a commercial tangor 'Murcote' citrus grove in Pratania, São Paulo State, Brazil and had the objective to evaluate the effects of, 2,4-D (auxin, NAA (auxin, GA3 (gibberellin and BA (cytokinin, on the reduction of seed number, without modifications on citrus fruit quality. The treatments sprayed were as follow: control (water; 10 and 20 mg.L-1 of 2,4-D; 100, 150 and 200 mg.L-1 of NAA; 100 and 200 mg.L-1 of GA3 ; 20 and 40 mg.L-1 of BA. The results showed that none of plant growth regulators influenced fruit quality, without weight reduction, diameter or ºBrix. In relation to seed number, none of the plant growth regulators were effective on reduction of seed number, however the reduced of viable seed number and total seed number of fruits, specially with the treatment of 100 and 200 mg.L-1 of NAA and 100 mg.L-1 of GA3, that showed a reduction of 30% of total seed of tangor murcott fruits.

    Full Text Available O tangor 'Murcote' apresenta necessidade de desbaste de frutos devido a alternância de produção, caracterizado por anos de excessiva produção intercalado com anos de baixa produção, evitando assim, a diminuição da qualidade dos frutos. O presente trabalho teve por objetivo avaliar a eficiência de reguladores vegetais, a auxina ANA (ácido naftalenacético e ethephon (etileno no desbaste químico de frutos de tangor 'Murcote' aplicado 40 dias após o pleno florescimento. O experimento foi conduzido em Pratânia, SP, onde plantas de 5 anos de idade, enxertadas sobre o limoeiro 'Cravo', foram pulverizadas com ANA a 0, 100, 200, 300 e 400 mg L-1 e com ethephon a 200, 300 e 400 mg L-1, ambos em solução aquosa juntamente com adjuvante não iônico a 0,05%. A contagem dos frutos foi realizada previamente aos tratamentos em 2 ramos marcados por planta. O ANA não interferiu significativamente no desbaste de frutos, com porcentagens de queda variando entre 7 a 14%, enquanto que as pulverizações com ethephon mostraram maior eficiência no desbaste de frutos, principalmente na dose de 400 mg L-1, promovendo 66,6% de queda de frutos, sem contudo induzir a abscisão foliar. As doses inferiores de ethephon também promoveram desbaste de frutos da ordem de 40%. As porcentagens de queda de frutos foram pequenas, para plantas pulverizadas com ANA, enquanto que a aplicação de ethephon promoveu maior eficiência no desbaste de frutos.Alternate bearing is an important caracteristic of some mandarins that reduces fruit quality and yield in tangor Murcott, and to reduce this effect is necessary to perform fruit thinning to avoid high and low production year by year.     Effect of red light irradiation on skin color development and carotenoid composition of stylar end of Miyauchi iyo (citrus iyo hort. ex Tanaka) tangor fruit was examined during storage. Both increase in a and a/b value and decrease in b value were enhanced by the irradiation. The a/b values of the fruit exposed were higher than those of control during the whole storage period. HPLC analyses also revealed the increase in total carotenoids content, of which the carotenoids that were tentatively named B and F most greatly accumulated. Similar tendencies were observed at equator and stem end portions. These results indicate that red light irradiation is involved in not only acceleration of overall color development but also enhancement of red color pigmentation by influencing a certain specific pathway of carotenoid biosynthesis

    The mandarin horticultural group is an important component of world citrus production for the fresh fruit market. This group formerly classified as C. reticulata is highly polymorphic and recent molecular studies have suggested that numerous cultivated mandarins were introgressed by C. maxima (the pummelos). C. maxima and C. reticulata are also the ancestors of sweet and sour oranges, grapefruit, and therefore of all the "small citrus" modern varieties (mandarins, tangors, tangelos) derived from sexual hybridization between these horticultural groups. Recently, NGS technologies have greatly modified how plant evolution and genomic structure are analyzed, moving from phylogenetics to phylogenomics. The objective of this work was to develop a workflow for phylogenomic inference from Genotyping By Sequencing (GBS) data and to analyze the interspecific admixture along the nine citrus chromosomes for horticultural groups and recent varieties resulting from the combination of the C. reticulata and C. maxima gene pools. A GBS library was established from 55 citrus varieties, using the ApekI restriction enzyme and selective PCR to improve the read depth. Diagnostic polymorphisms (DPs) of C. reticulata/C. maxima differentiation were identified and used to decipher the phylogenomic structure of the 55 varieties. The GBS approach was powerful and revealed 30,289 SNPs and 8,794 Indels with 12.6% of missing data. 11,133 DPs were selected covering the nine chromosomes with a higher density in genic regions. GBS combined with the detection of DPs was powerful for deciphering the "phylogenomic karyotypes" of cultivars derived from admixture of the two ancestral species after a limited number of interspecific recombinations. All the mandarins, mandarin hybrids, tangelos and tangors analyzed displayed introgression of C. maxima in different parts of the genome.     Full Text Available O bloqueio de eventos dependentes da sinalização do etileno pode afetar de maneira positiva ou negativa a qualidade de frutos tropicais após o armazenamento refrigerado. Dessa forma, os objetivos do presente trabalho foram estudar o envolvimento do etileno no desenvolvimento de injúrias de frio em tangor 'Murcott' e avaliar as respostas envolvidas no processo de resistência às injúrias. Os frutos foram expostos a 500nL L-1 de 1-metilciclopropeno (1-MCP durante 12 horas ou imersos em soluções contendo 2000nL L-1 de ethephon ou ácido salicílico durante cinco minutos antes de serem armazenados a 1°C, por 90 dias. Como controle, parte dos frutos foi armazenada a 1°C. O tratamento de frutos com ethephon ou ácido salicílico antecipou e intensificou as injúrias de frio. Por outro lado, a inibição do etileno pelo 1-MCP retardou o surgimento dos sintomas e resultou em menor índice de injúrias e percentual de frutos podres ao final do armazenamento. A atividade da superóxido dismutase (SOD foi intensificada aos 45 dias, contudo em menor intensidade nos frutos tratados com ácido salicílico. Nas avaliações subsequentes, houve decréscimo na atividade da SOD em todos os tratamentos, porém aos 90 dias a intensidade manteve-se levemente superior à observada nos primeiros 30 dias de armazenamento. Os teores de putrescina (Put e espermina (Spm, no flavedo dos frutos, não sofreram significativa alteração durante o armazenamento.     Full Text Available Degreening treatment has been applied in traditional countries exporters and consumers of fresh citrus to improve rind color of fruit cultivated in regions that no shown low temperature during maturation. In Brazil, this technique is performed empirically, without control of application conditions and fruit shown low extern quality. This study was carried out with the objective to determinate conditions to the technique of postharvest degreening of ‘Murcott’ orange, involving the application of exogenous ethylene. The concentrations 5 and 10 mL L-1 of ethylene was applied inside the chamber for 96 and 120 hours at 25oC and 90% RH. After treatments fruit were stored at 5°C during for 30 days, and after that, plus 3 days at 25ºC. Fruit were evaluated in four periods: after harvest; after degreening treatment, after cold storage and after 3 days at 25°C (simulated marketing. The variable analyzed were: skin color, soluble solids (SS, titratable acidity (TA, ratio (SS/TA, ascorbic acid, percentage of juice, chlorophyll and carotenoids content, chlorophyllase activity, respiratory rate and ethylene production. The treatments did not affect the soluble solids, titratable acidity, ratio, percentage of juice and ascorbic acid content. Fruit treated with ethylene showed increase of color index concomitant by higher chlorophyllase activity and lower chlorophyll content. There were no differences in the carotenoids content in the periods evaluated. Fruit treated with ethylene at both concentrations had higher ethylene production, whereas the dose of 10 mL L-1 favored the incidence of decay. Ethylene 5 mL L-1 for 96 hours is enough to trigger the process of degreening ‘ Murcott’ tangor and can be used to improve skin color of this cultivar.

Esta doença causa desfolha grave, queda de frutos e manchas nas frutas. O objetivo deste trabalho foi estabelecer o melhor fungicida e a melhor dose para o controle da mancha marrom de alternaria. O delineamento experimental foi de parcelas subdivididas em blocos, com 10 tratamentos principais e 3 doses (subparcelas, com 5 repetições. Foram feitas 5 aplicações, com intervalo de 15 dias. Os tratamentos foram: azoxystrobin, pyraclostrobin, trifloxystrobin, trifloxystrobin + tebuconazole (2 aplicações seguido de 3 aplicações de mancozeb, difenoconazole, trifloxystrobin + propiconazole, iprodione, trifloxystrobin + propineb (2 aplicações seguido de 3 aplicações de oxicloreto de cobre, oxicloreto de cobre + óleo e testemunha. Simultaneamente foram feitas avaliações de incidência e número de lesões por folha. Ao surgimento dos frutos foram avaliadas a incidência em frutos e a produtividade em Kg/ha. Todos os tratamentos foram superiores à testemunha quanto a produtividade.     Full Text Available O objetivo deste trabalho foi obter plantas autotetraplóides de tangerina 'Ponkan', laranja 'Pêra-de-abril' e tangor 'Murcott', que serão usadas em cruzamentos com cultivares diplóides, visando à obtenção de indivíduos triplóides sem sementes. Utilizou-se o método de cultivo in vitro de segmentos de epicótilo em meio com colchicina (0,025, 0,05 e 0,1%, por diversos períodos (1, 2, 3, 7 e 14 dias, com subseqüente regeneração de brotações em meio sem a presença do alcalóide. As brotações foram microenxertadas in vitro e aclimatizadas em estufas. A determinação do nível de ploidia das plantas foi realizada por citometria de fluxo. A colchicina demonstrou ser tóxica aos explantes das três variedades, ocasionando redução significativa no número médio de brotações adventícias e aumento na porcentagem de explantes não-responsivos, em comparação com o controle. Entre as quatro plantas de laranja e uma de tangor obtidas, duas plantas de laranja e a de tangor, demonstraram ser autotetraplóides, apresentando folhas com maior espessura, arredondadas e coloração verde intensa. O método utilizado na duplicação cromossômica, com uso de colchicina, é eficiente em produzir plantas autotetraplóides de citros.The objective of this work was to obtain autotetraploid plants of 'Murcott' tangor, 'Ponkan' mandarin and 'Pêra-de-abril' sweet orange to be used in crosses with diploid cultivars, aiming to produce triploid seedless hybrids. Aumentar o número de variedades, assim como ampliar o período de safra são fatores importantes tendo em vista que trarão vantagens ao consumidor e ao produtor desse grupo de citros. No período de 2002 a 2005, foram estudadas as características da variedade Fremont e comparadas com as tangerinas 'Ponkan' e 'Clementina Nules', no município de Capão Bonito-SP. Pode-se concluir, dos resultados obtidos, que as plantas da tangerina Fremont são de porte reduzido (semi-anãs; os frutos apresentam características excepcionais para consumo in natura, maturação de meia-estação e período de colheita, que se estende por até três meses, sendo dessa maneira boa opção para plantios visando à entressafra da 'Ponkan'.Commercial grown tangerines variety are mainly restricted to a few options such as Ponkan tangerine and Murcott tangor. The increasing number of varieties as well as the harvest extending season is important, potentially benefiting consumers and producers of this kind of citrus. Fremont characteristics were compared to Ponkan and Clementina Nules tangerines at Capão Bonito County in São Paulo State from 2002 to 2005. By the results, it was possible to conclude that Fremont trees were semi-dwarfed, the fruits had excellent characteristics for in natura consumption and a harvesting season enlargement of three months, therefore appearing to be a good alternative for orchards, aiming at Ponkan' off-season period.

    Full Text Available The spatial distribution of larvae of Phyllocnistis citrella Stainton (Lepidoptera: Gracillariidae, the citrus leaf miner, in leaves, shoots, and tree crowns of Montenegrina tangerine (Citrus deliciosa Tenore and Murcott tangor (Citrus sinensis L. Osbeck x Citrus reticulata Blanco was determined. Fortnightly samplings from July/2001 to June/2003, in orchards located in Montenegro (29º 68’ S and 51º 46’ W, RS, Brazil, were carried out. At each sampling occasion eighth shoots randomly selected were collected. The spatial distribution pattern of P. citrella larvae between tree crowns and in the shoots was aggregated in most sampling occasions in both citrus species.     Full Text Available A ampliação da oferta de tangerinas tanto no mercado interno como no externo é limitada pela grande quantidade de sementes nos frutos das principais cultivares, danos na casca resultante de doenças e pela pequena produção nas épocas mais quentes do ano. O presente trabalho teve como objetivo caracterizar a qualidade dos frutos e a época de maturação de 46 tangerinas e híbridos em condições de campo na Estação Experimental de Citricultura de Bebedouro (EECB, Bebedouro-SP. As variedades de tangerinas e híbridos (tangores e tangelos foram introduzidas de bancos de germoplasma da Itália, Portugal, Espanha e França, e a pesquisa desenvolveu-se durante os anos agrícolas de 2001/2002 e 2002/2003. Foram avaliadas características externas de qualidade, como altura e diâmetro dos frutos, massa dos frutos, facilidade em descascar com a mão e características internas como: número de sementes, teor de sólidos solúveis totais, acidez total titulável, rendimento em suco, "ratio" e rendimento em polpa. Duas cultivares apresentaram frutos de boa qualidade e época de maturação precoce, podendo ser disponibilizadas no mercado, num período de escassez deste tipo de citros, e serem alternativas à tangerina-'Cravo'.     Phlorin (3,5-dihydroxyphenyl beta-D-glucopyranoside), an orange peel marker, has been searched in 45 species and varieties of Citrus. The phlorin content was determined by high-pressure liquid chromatography in juices and aqueous peel extracts of these different fruits. The phlorin content in C. reticulata peel extract varies from 0 to 1012 mg L(-)(1) with a mean of 162 mg L(-)(1). On the contrary, phlorin was not found in mandarin and clementine juices except for mandarin "Commune" and "Beauty" (33 and 30 mg L(-)(1), respectively). In the 14 species of oranges and varieties, phlorin was detected in juices and peel extracts with a mean of 22 and 492 mg L(-)(1), respectively, while for grapefruits, means were 108 mg L(-)(1) in juices and 982 mg L(-)(1) for peel extracts. Tangors and tangelos which are hybrids (C. reticulata x C. sinensis and C. reticulata x C. paradisi, respectively) are characterized by the systematic presence of phlorin in peels (mean: 406 and 659 mg L(-)(1), respectively) while in juices its presence could be variable (0-73 mg L(-)(1)). These heterogeneity and values may be explained by the genetic variability of these hybrids and the phlorin content of their parentage group.

    Flavonoids are polyphenolic compounds in fruits, vegetables, grains, etc with antioxidant capacity and anti-inflamatory, anticoagulant, antiallergic proprieties that contributes to human health. The effect of the pasteurization and concentration processes on the citric juices can influence the composition of the flavanones. The flavanones composition in the orange juice (a mixture of Pera and Valencia and of the tangor Murcott was determined by HPLC. The flavanones (narirutin and hesperidin were extracted with methanol, heated in 55°C for 15 minutes and separated from the insoluble solids by centrifugation. The separation was done in C18 column (5μm, 250x4.6mm using the isocratic system of acetonitrile mixture: water, acetic acid (21:75:4v/v/v in a flow of 0.6L/min, detention of 280nm and quantification by external pattern. The effect of the pasteurization and concentration on the citric juice did not affect significantly the amount of flavones in the samples of murcott and orange juice (mixture Pera and Valencia.

    To investigate the level of boron nutrient in citrus and its impact factors, a total of 954 citrus leaf samples and 302 soil samples were collected from representative orchards in the 12 main citrus production counties in the Three Gorges Reservoir region of Chongqing to determine the boron content in citrus leaves, as well as the relationships between leaf boron content with soil available boron content, soil pH value, cultivar, rootstock and the age of tree. Results indicated that the leaf samples from 41.6% orchards (tree did affect the leaf boron content. The leaves from the orchards with soil pH of 4.5-6.4 demonstrated significantly higher boron contents than with the soil pH of 6.5-8.5. The leaf boron contents in the different cultivars was ranged as Satsuma mandarin > pomelo > valencia orange > sweet orange > tangor > navel orange. The citrus on trifoliate orange and sour pomelo rootstocks had significantly higher leaf boron contents than on Carrizo citrange and red tangerine rootstocks. Compared with the adult citrus trees (above 8 year-old), 6.6% more of leaf samples of younger trees (3 to 8 year-old) contained boron contents in the optimum range (35-100 mg x kg(-1)).

    ABSTRACT Xylella fastidiosa is the causal agent of citrus variegated chlorosis (CVC), a destructive disease of sweet orange cultivars in Brazil. Polymerase chain reaction (PCR)-based assays constitute the principal diagnostic method for detection of these bacteria. In this work, we established a real-time quantitative PCR (QPCR) assay to quantify X. fastidiosa in naturally and artificially infected citrus. The X. fastidiosa cell number detected in the leaves increased according to the age of the leaf, and bacteria were not detected in the upper midrib section in young leaves, indicating temporal and spatial distribution patterns of bacteria, respectively. In addition, the X. fastidiosa cell number quantified in leaves of 'Pera' orange and 'Murcott' tangor reflected the susceptible and resistant status of these citrus cultivars. None of the 12 endophytic citrus bacteria or the four strains of X. fastidiosa nonpathogenic to citrus that were tested showed an increase in the fluorescence signal during QPCR. In contrast, all 10 CVC-causing strains exhibited an increase in fluorescence signal, thus indicating the specificity of this QPCR assay. Our QPCR provides a powerful tool for studies of different aspects of the Xylella-citrus interactions, and can be incorporated into breeding programs in order to select CVC-resistant plants more quickly.

    Full Text Available The objective of this study was to produce citrus somatic asymmetric hybrids by fusing gamma-irradiated protoplasts with iodoacetamide-treated protoplasts. Protoplasts were isolated from embryogenic suspension cells of grapefruit (Citrus paradisi Macfad. cultivars Ruby Red and Flame, sweet oranges (C. sinensis Osbeck 'Itaboraí', 'Natal', Valencia', and 'Succari', from 'Satsuma' (C. unshiu Marcow. and 'Changsha' mandarin (C. reticulata Blanco and 'Murcott' tangor (C. reticulata x C. sinensis. Donor protoplasts were exposed to gamma rays and receptor protoplasts were treated with 3 mmol L-1 iodoacetamide (IOA, and then they were fused for asymmetric hybridization. Asymmetric embryos were germinated, and the resulting shoots were either grafted onto sour orange, rough lemon or 'Swingle' (C. paradisi x Poncirus trifoliata x 'Sunki' mandarin rootstock seedlings, or rooted after dipping their bases in indol-butyric acid (IBA solution. The products were later acclimatized to greenhouse conditions. Ploidy was analyzed by flow cytometry, and hybridity was confirmed by amplified fragment length polymorphism (AFLP analysis of plantlet DNAsamples. The best treatment was the donor-recipient fusion combination of 80 Gy-irradiated 'Ruby Red' protoplasts with 20 min IOA-treated 'Succari' protoplasts. Tetraploid and aneuploid plants were produced.     ABSTRACT The genetic inheritance of resistance to leprosis, the most important viral disease of citrus in Brazil, was characterized through the phenotypic assessment of 143 hybrids resulting from crosses between tangor 'Murcott' (Citrus sinensis x C. reticulata) and sweet orange 'Pêra' (C. sinensis), considered to be resistant and susceptible to the disease, respectively. All plants were grafted onto Rangpur lime (C. limonia) and inoculated with Citrus leprosis virus, cytoplasmic type through the infestation with viruliferous mites, Brevipalpus phoenicis. The experiments were arranged in a completely randomized block design with 10 replicates. Incidence and severity of the disease in leaves and stems as well as plant growth parameters (plant height and stem diameter) were recorded for 3 years after the infestation with the viruliferous mites. The average values of all variables were analyzed using principal component analysis, discriminant factorial analysis, estimation of the clonal repeatability coefficients, and frequency of the distributions of the average values for each measured variable. The principal component analysis resulted in the identification of at least two groups with resistance and susceptibility to leprosis, respectively. About 99% of all hybrids were correctly classified according to the discriminant factorial analysis. The broad-sense heritability coefficients for characteristics associated with incidence and severity of leprosis ranged from 0.88 to 0.96. The data suggest that the inheritance of resistance to leprosis may be controlled by only a few genes.

  9. Genetic variability in the endophytic fungus Guignardia citricarpa isolated from citrus plants

    Full Text Available During some phases of of their life-cycle endophytic fungi colonize plants asymptomatically being found most frequently inside the aerial part of plant tissues. After surface disinfection of apparently healthy leaves from three varieties of mandarin orange and one tangor, and after incubation on appropriate culture medium, 407 fungal isolates were obtained, giving a total infection frequency of 81%. No fungal growth was observed from disinfected seeds, indicating that fungi are probably not transmitted via seeds. Of the fungal isolates, 27% belonged to the genus Guignardia, with 12 isolates being identified as Guignardia citricarpa Kiely, which is described as a citrus pathogen. The isolates were variable in respect to the presence of sexual structures and growth rates. Most of the isolates produces mature asci, supporting the hypothesis that they are nonpathogenic endophytes, which recently were identified as G. mangiferae. High intraspecific genetic variability (an average similarity coefficient of 0.6 was detected using random amplified polymorphic DNA (RAPD markers generated by seven different primers. The highest similarity coefficient (0.9 was between isolates P15 and M86 and the smallest (0.22 between isolates P15 and C145. These results did not allow us to establish an association between genetic similarity of the fungal isolates and the citrus varieties from which they were obtained.

    The objective of this study was to produce citrus somatic asymmetric hybrids by fusing gamma.irradiated protoplasts with iodoacetamide-treated protoplasts. Protoplasts were isolated from embryogenic suspension cells of grapefruit (Citrus paradisi Macfad.) cultivars Ruby Red and Flame, sweet oranges (C. sinensis Osbeck) 'Itaborai', 'Natal', Valencia', and 'Succari', from 'Satsuma' (C. unshiu Marcow.) and 'Changsha' mandarin (C. reticulata Blanco) and 'Murcott' tangor (C. reticulata x C. sinensis). Donor protoplasts were exposed to gamma rays and receptor protoplasts were treated with 3 mmol L{sup -1} iodoacetamide (IOA), and then they were fused for asymmetric hybridization. Asymmetric embryos were germinated, and the resulting shoots were either grafted onto sour orange, rough lemon or 'Swingle' (C. paradisi x Poncirus trifoliata) x 'Sunki' mandarin rootstock seedlings, or rooted after dipping their bases in indol.butyric acid (IBA) solution. The products were later acclimatized to greenhouse conditions. Ploidy was analyzed by flow cytometry, and hybridity was confirmed by amplified fragment length polymorphism (AFLP) analysis of plantlet DNA samples. The best treatment was the donor-recipient fusion combination of 80 Gy.irradiated 'Ruby Red' protoplasts with 20 min IOA.treated 'Succari' protoplasts. Tetraploid and aneuploid plants were produced.   11. Protein changes in the albedo of citrus fruits on postharvesting storage.

    In this work, major protein changes in the albedo of the fruit peel of Murcott tangor (tangerine x sweet orange) during postharvest ageing were studied through 2D PAGE. Protein content in matured on-tree fruits and in fruits stored in nonstressing [99% relative humidity (RH) and 25 degrees C], cold (99% RH and 4 degrees C), and drought (60% RH and 25 degrees C) conditions was initially determined. Protein identification through MS/MS determinations revealed in all samples analyzed the occurrence of manganese superoxide dismutase (Mn SOD), actin, ATP synthase beta subunit (ATPase), citrus salt-stress associated protein (CitSap), ascorbate peroxidase (APX), translationally controlled tumor protein (TCTP), and a cysteine proteinase (CP) of the papain family. The latter protein was identified in two different gel spots, with different molecular mass, suggesting the simultaneous presence of the proteinase precursor and its active form. While Mn SOD, actin, ATPase, and CitSap were unchanged in the assayed conditions, TCTP and APX were downregulated during the postharvest ageing process. Ageing-induced APX repression was also reversed by drought. CP contents in albedo, which were similar in on- and off-tree fruits, were strongly dependent upon cold storage. The active/total CP protein ratio significantly increased after cold exposure. This proteomic survey indicates that major changes in protein content in the albedo of the peel of postharvest stored citrus fruits are apparently related to the activation of programmed cell death (PCD).

    Genome wide selection (GWS) is essential for the genetic improvement of perennial species such as Citrus because of its ability to increase gain per unit time and to enable the efficient selection of characteristics with low heritability. This study assessed GWS efficiency in a population of Citrus and compared it with selection based on phenotypic data. A total of 180 individual trees from a cross between Pera sweet orange (Citrus sinensis Osbeck) and Murcott tangor (Citrus sinensis Osbeck x Citrus reticulata Blanco) were evaluated for 10 characteristics related to fruit quality. The hybrids were genotyped using 5287 DArT_seq TM (diversity arrays technology) molecular markers and their effects on phenotypes were predicted using the random regression - best linear unbiased predictor (rr-BLUP) method. The predictive ability, prediction bias, and accuracy of GWS were estimated to verify its effectiveness for phenotype prediction. The proportion of genetic variance explained by the markers was also computed. The heritability of the traits, as determined by markers, was 16-28%. The predictive ability of these markers ranged from 0.53 to 0.64, and the regression coefficients between predicted and observed phenotypes were close to unity. Over 35% of the genetic variance was accounted for by the markers. Accuracy estimates with GWS were lower than those obtained by phenotypic analysis; however, GWS was superior in terms of genetic gain per unit time. Thus, GWS may be useful for Citrus breeding as it can predict phenotypes early and accurately, and reduce the length of the selection cycle. This study demonstrates the feasibility of genomic selection in Citrus.

  13. Nuclear Species-Diagnostic SNP Markers Mined from 454 Amplicon Sequencing Reveal Admixture Genomic Structure of Modern Citrus Varieties

    Most cultivated Citrus species originated from interspecific hybridisation between four ancestral taxa (C. reticulata, C. maxima, C. medica, and C. micrantha) with limited further interspecific recombination due to vegetative propagation. This evolution resulted in admixture genomes with frequent interspecific heterozygosity. Moreover, a major part of the phenotypic diversity of edible citrus results from the initial differentiation between these taxa. Deciphering the phylogenomic structure of citrus germplasm is therefore essential for an efficient utilization of citrus biodiversity in breeding schemes. The objective of this work was to develop a set of species-diagnostic single nucleotide polymorphism (SNP) markers for the four Citrus ancestral taxa covering the nine chromosomes, and to use these markers to infer the phylogenomic structure of secondary species and modern cultivars. Species-diagnostic SNPs were mined from 454 amplicon sequencing of 57 gene fragments from 26 genotypes of the four basic taxa. Of the 1,053 SNPs mined from 28,507 kb sequence, 273 were found to be highly diagnostic for a single basic taxon. Species-diagnostic SNP markers (105) were used to analyse the admixture structure of varieties and rootstocks. This revealed C. maxima introgressions in most of the old and in all recent selections of mandarins, and suggested that C. reticulata × C. maxima reticulation and introgression processes were important in edible mandarin domestication. The large range of phylogenomic constitutions between C. reticulata and C. maxima revealed in mandarins, tangelos, tangors, sweet oranges, sour oranges, grapefruits, and orangelos is favourable for genetic association studies based on phylogenomic structures of the germplasm. Inferred admixture structures were in agreement with previous hypotheses regarding the origin of several secondary species and also revealed the probable origin of several acid citrus varieties.   14. Isolation and identification of citrus psorosis virus Egyptian isolate (CPsV-EG).

    Citrus psorosis ophiovirus (CPsV), is considered to be of the most serious and deter mental virus pathogen's citrus species trees in Egypt. CPsV-EG was isolated from infected citrus grapefruit (C. paradisi Macf.) at Agric. Res. Centre (ARC). The grapefruit which used for CPsV-EG isolate was found to be free from CTV, CEVd and Spiroplasma citri where as gave -ve results with DTBIA, tissue print hybridization and Diene's stain respectively. CPsV-EG was detected on the basis of biological indexing by graft inoculation which gave oak leaf pattern (OLP) on Dweet tangor and serological assay by DAS-ELISA using Mab specific CPsV. CPsV-EG was reacted with variable responses on 16 host plants belonging to 6 families. Only 8 host plants are susceptible and showed visible external symptoms which appeared as local, systemic and local followed by systemic infections. CPsV-EG isolate was transmitted from infected citrus to citrus by syringe and grafting and herbaceous plants by forefinger inoculation and syringe. The woody indicators and rootstocks were differed in response to CPsV-EG isolate which appeared as no-response, response, sensitivity and hypersensitivity. The serological characters represented as the antigenic determinants of CPsV-EG isolate related to monoclonal antibodies specific CPsV strain where as appeared precipitation reaction by DAS-ELISA and DTBIA. The partial fragment of RNA3 (coat protein gene) of CPsV-EG (-1140bp and -571bp) was amplified by reverse transcription-polymerase chain reaction (RT-PCR) from grapefruit tissues using two sets primers specific CPsV (CPV3 and CPV4) and (PS66 and PS65) respectively. The virus under study was identified as CPsV-EG isolate according to biological, serological and molecular characters.

    Full Text Available Este trabalho teve como objetivos verificar e identificar lepidópteros minadores em plantas de crescimento espontâneo, presentes em pomar de citros, e verificar se o "minador-das-folhas-dos-citros", Phyllocnistis citrella (Lepidóptera: Gracillariidae, ocorre nessas plantas espontâneas. O trabalho foi conduzido em um pomar de tangoreiro 'Murcott', em Montenegro- RS. Realizaram-se amostragens quinzenais, de maio de 2003 a maio de 2004, coletando-se, em cada ocasião, todas as plantas e/ou ramos com minas, contidas na área delimitada por um aro de 0,28 m2, que era jogado nas linhas e nas entrelinhas de 30 árvores sorteadas. As plantas hospedeiras dos minadores foram coletadas para identificação. Registraram-se 11 espécies de lepidópteros minadores, distribuídas em seis famílias, coletadas em 15 espécies de plantas hospedeiras de nove famílias botânicas. A comunidade de plantas de crescimento espontâneo, na área amostrada, hospedou uma vasta diversidade de lepidópteros minadores, incluindo até possíveis novas famílias, porém não hospedaram P. citrella.This study aimed to verify and to identify Lepidopterous leafminers in plants of spontaneous growth present in citrus orchard and to verify the occurrence of "citrus leafminer", Phyllocnistis citrella (Lepidoptera: Gracillariidae, on those plants of spontaneous growth.     Full Text Available As moscas-das-frutas são as principais pragas da fruticultura mundial. Consideradas chaves para a produção de citros, torna-se necessário o seu monitoramento, visando a evitar os danos diretos. O experimento teve como objetivos conhecer a variação populacional de Anastrepha fraterculus e a relação de sua população com danos em pomares orgânicos de Citrus sinensis, cultivar Céu e de C. sinensis x Citrus reticulata tangor 'Murcott'. Os dados foram coletados em 2003 e 2004 durante o período de maturação dos frutos, na região do vale do Caí, RS, Brasil. O número de moscas-das-frutas foi registrado, semanalmente, por meio de armadilhas McPhail, contendo suco de uva, a 25%. Danos aos frutos foram determinados pela razão entre frutos sadios e frutos danificados pela mosca. Registros meteorológicos de temperatura, umidade relativa e precipitação pluviométrica foram obtidos, em estação meteorológica distante 30 km das áreas experimentais. Verificou-se que, em condições ideais de precipitação pluvial, maiores foram as populações de A. fraterculus, espécie predominante na região. A população estimada capaz de causar danos aos frutos variou de acordo com o cultivar, sendo a laranjeira 'Céu' a mais susceptível. Os maiores picos populacionais ocorrem na fase de mudança de coloração dos frutos. Porém, na fase de maturação, as moscas causaram os maiores danos, dada a intolerância dos frutos ao ataque. Conclui-se que a infestação dos frutos de citros por A. fraterculus está relacionada com espécie e cultivar e com fatores climáticos, principalmente com a precipitação pluvial. O monitoramento constante da população de mosca-das-frutas é importante na determinação da infestação na colheita.

    Full Text Available Cold storage allows citrus budstick conservation for several months, although gradual bud viability loss and abscission of leaf petioles is observed. This study aimed to maintain bud viability reducing leaf petiole abscission in budsticks of 'Baianinha' and 'Valencia' sweet oranges (C. sinensis Osbeck, 'Tahiti' lime (C. latifolia Tanaka, and 'Murcott' tangor (C. sinensis × C. reticulata, using NAA, GA3, 2,4-D, and low temperature storage. After a 15-minutes-immersion in distilled water and NAA and GA3 solutions at concentrations of 10, 100, 500 and 1000 µmol L-1, and 2,4-D solutions at concentrations of 1, 10, 50 and 100 µmol L-1, the shoots were stored in polyethylene bags at 5 ± 1ºC for 74 days. Percentage of leaf petiole abscission, fresh and dry matter losses, bud viability, and growth flush were evaluated. NAA and 2,4-D had the highest efficiency for petiole abscission inhibition, reaching almost 99% of success. The packaging system and shoot storage under low temperature efficiently reduced fresh and dry matter losses. Bud viability of 'Baianinha', 'Valencia' and 'Murcott' shoots was not reduced by storage, maintaining above 95% of grafting success.
