This study pays attention to the relation between individual care and residents` activities in the social welfare institutions for the elderly (nursing homes) and aims to clarify the influence of building environment (nursing home as dwelling place) on individual care. Behaviors of care staffs and residents are surveyed in four nursing homes. The results of the analysis of the survey are as follows: The care staff`s activities are influenced not only by physical environment and also by the residents` activities. A single bed room is more effective than a multi-bed room for individual care. The small care unit is effective in building plan if the number of staffs is enough. The personal care activities which constitute the greater part of the care activities depend on the location of the residents. It is important to pay attention not only to arrange the common space but also how to use the common space. Spatial structure is required the consideration of the relative position of not only care rooms and resident rooms but also of care rooms and the space used by residents. 7 refs., 14 figs., 7 tabs.

Taking up the home automation (HA) as the information facility of residence, business and technical trend of HA system together with its present and future condition are described. The service functions of HA system used for multiple dwelling houses are home telephone function, security function, control function and visual communication function. Besides, development concept of dormitory control system and service functions which are attractive to people like owner, manager and residents are described. As for its trend in foreign countries, though in France HA system under government guidance was applied in the 1980`s, it was not successful and market related to HA system was held in check. Since 1990`s, due to government policy for its introduction, this system is getting popular in the residences of disabled person, advanced age person and so forth. As for the development of HA system, more improvement not only indoor the home but also comfort, safety and so forth linking to the outdoor system is necessary. 6 figs.

Surveying Fusarium resistance in wheat with easy applicable molecular markers such as simple sequence repeats (SSRs) is a prerequest for molecular breeding. Expressed sequence tags (ESTs) are one of the main sources for development of new SSR candidates. Therefore, 18.292 publicly available wheat ESTs were mined and genotyping of newly developed 55 EST-SSR derived primer pairs produced clear fragments in ten wheat cultivars carrying different levels of Fusarium resistance. Among the proved markers, 23 polymorphic EST-SSRs were obtained and related alleles were mostly found on B and D genome. Based on the fragment profiling and similarity analysis, a 327bp amplicon, which was a product of contig 1207 (chromosome 5BL), was detected only in Fusarium head blight (FHB) resistant cultivars (CM82036 and Sumai) and the amino acid sequences showed a similarity to pathogen related proteins. Another FHB resistance related EST-SSR, Contig 556 (chromosome 1BL) produced a 151bp fragment in Sumai and was associated to wax2-like protein. A polymorphic 204bp fragment, derived from Contig 578 (chromosome 1DL), was generated from root rot (FRR) resistant cultivars (2-49; Altay2000 and Sunco). A total of 98 alleles were displayed with an average of 1.8 alleles per locus and the polymorphic information content (PIC) ranged from 0.11 to 0.78. Dendrogram tree with two main and five sub-groups were displayed the highest genetic relationship between FRR resistant cultivars (2-49 and Altay2000), FRR sensitive cultivars (Seri82 and Scout66) and FHB resistant cultivars (CM82036 and Sumai). Thus, exploitation of these candidate EST-SSRs may help to genotype other wheat sources for Fusarium resistance. Copyright © 2017 Elsevier Ltd. All rights reserved.

Full Text Available The field experiment was carried out in 2010–2012 at the Dłoń Agricultural Research Station, the Poznań University of Life Sciences, Poland. The study was designed to evaluate the degree of infection by powdery mildew, brown rust, and septoria leaf blotch in 61 spring wheat genotypes differing in their resistance to Fusarium ssp. The vast majority of spring wheat genotypes in the collection of gene resources in the USA defined as resistant to Fusarium ssp. confirmed their resistance under Polish climatic conditions. The B .graminis infection rate of genotypes that are considered to be resistant to Fusarium head blight was high. The resistance ranged from 7 for Sumai 3 (PL2 up to 8.8 for Ning 8331 (in a 9-point scale. Most of the genotypes (56.5% were infected by Puccinia recondita at a level of 1–3 (in a 9-point scale. The genotypes of Sumai 3 exhibited high resistance to septoria leaf blotch, amounting to 1–2 in a 9-point scale; the resistance of Frontana ranged from 1 to 3.5, while the genotypes of Ning were infected by Mycosphaerella graminicola at 5–6.

Full Text Available A previous study provided an in-depth understanding of molecular population genetics of European and Asian wheat gene pools using a sequenced 3.1-Mb contig (ctg954 on chromosome 3BS. This region is believed to carry the Fhb1 gene for response to Fusarium head blight. In this study, 266 wheat accessions were evaluated in three environments for Type II FHB response based on the single floret inoculation method. Hierarchical clustering (UPGMA based on a Manhattan dissimilarity matrix divided the accessions into eight groups according to five FHB-related traits which have a high correlation between them; Group VIII comprised six accessions with FHB response levels similar to variety Sumai 3. Based on the compressed mixed linear model (MLM, association analysis between five FHB-related traits and 42 molecular markers along the 3.1-Mb region revealed 12 significant association signals at a threshold of P0.1 and P0.05 within each HapB at r(2>0.1 and P<0.001 showed significant differences between the Hap carried by FHB resistant resources, such as Sumai 3 and Wangshuibai, and susceptible genotypes in HapB3 and HapB6. These results suggest that Fhb1 is located within HapB6, with the possibility that another gene is located at or near HapB3. SSR markers and Haps detected in this study will be helpful in further understanding the genetic basis of FHB resistance, and provide useful information for marker-assisted selection of Fhb1 in wheat breeding.

Full Text Available Deoxynivalenol (DON is the most prevalent trichothecene in Europe and its occurrence is associated with infections of Fusarium graminearum and F. culmorum, causal agents of Fusarium head blight (FHB on wheat. Resistance to FHB is a complex character and high variability occurs in the relationship between DON content and FHB incidence. DON conjugation to glucose (DON-3-glucoside, D3G is the primary plant mechanism for resistance towards DON accumulation. Although this mechanism has been already described in bread wheat and barley, no data are reported so far about durum wheat, a key cereal in the pasta production chain. To address this issue, the ability of durum wheat to detoxify and convert deoxynivalenol into D3G was studied under greenhouse controlled conditions. Four durum wheat varieties (Svevo, Claudio, Kofa and Neodur were assessed for DON-D3G conversion; Sumai 3, a bread wheat variety carrying a major QTL for FHB resistance (QFhs.ndsu-3B, was used as a positive control. Data reported hereby clearly demonstrate the ability of durum wheat to convert deoxynivalenol into its conjugated form, D3G.

An in vitro spike culture method was optimized to evaluate Fusarium head blight (FHB) resistance in wheat (Triticum aestivum) and used to screen a population of ethyl methane sulfonate treated spike culture-derived variants (SCDV). Of the 134 SCDV evaluated, the disease severity score of 47 of the variants was ≤30%. Single nucleotide polymorphisms (SNP) in the UDP-glucosyltransferase (UGT) genes, TaUGT-2B, TaUGT-3B, and TaUGT-EST, differed between AC Nanda (an FHB-susceptible wheat variety) and Sumai-3 (an FHB-resistant wheat cultivar). SNP at 450 and 1,558 bp from the translation initiation site in TaUGT-2B and TaUGT-3B, respectively were negatively correlated with FHB severity in the SCDV population, whereas the SNP in TaUGT-EST was not associated with FHB severity. Fusarium graminearum strain M7-07-1 induced early expression of TaUGT-2B and TaUGT-3B in FHB-resistant SCDV lines, which were associated with deoxynivalenol accumulation and reduced FHB disease progression. At 8 days after inoculation, deoxynivalenol concentration varied from 767 ppm in FHB-resistant variants to 2,576 ppm in FHB-susceptible variants. The FHB-resistant SCDV identified can be used as new sources of FHB resistance in wheat improvement programs.

Pectin methyl esterase (PME) genes code for enzymes that are involved in structural modifications of the plant cell wall during plant growth and development. They are also involved in plant-pathogen interaction. PME genes belong to a multigene family and in this study we report the first comprehensive analysis of the PME gene family in bread wheat (Triticum aestivum L.). Like in other species, the members of the TaPME family are dispersed throughout the genome and their encoded products retain the typical structural features of PMEs. qRT-PCR analysis showed variation in the expression pattern of TaPME genes in different tissues and revealed that these genes are mainly expressed in flowering spikes. In our attempt to identify putative TaPME genes involved in wheat defense, we revealed a strong variation in the expression of the TaPME following Fusarium graminearum infection, the causal agent of Fusarium head blight (FHB). Particularly interesting was the finding that the expression profile of some PME genes was markedly different between the FHB-resistant wheat cultivar Sumai3 and the FHB-susceptible cultivar Bobwhite, suggesting a possible involvement of these PME genes in FHB resistance. Moreover, the expression analysis of the TaPME genes during F. graminearum progression within the spike revealed those genes that responded more promptly to pathogen invasion. We have developed a high efficiency L-lactate fermentation process employing the homofermentative lactic acid bacterium, Lactococcus lactis IO-1 with a pH-dependent substrate feed system in order to consume the substrate completely and maintain high activity of cell growth, production, and substrate consumption. Furthermore a pH-dependent substrate feed system was combined with turbidity control consisting of a laser probe and micro filter module for cell-recycling. With this system, we achieve a high efficiency L-lactate continuous culture. We are able to construct high efficiency bioconversion systems from xylose to P(3HB) via L-lactate and other organic acids because Lactococcus lactis IO-1 can utilize not only glucose but also xylose as a carbon source. Thus, we have developed a new substrate feed system which is able to automatically control substrate concentration in a broth. A new batch fermentative system for production of biodegradable plastic material, poly hydroxybutyrate [P(3HB)] from culture broth of Lactococcus lactis IO-1 is vealized. (author)

This is No.93 report of National Institute for Research in Inorganic Materials. Single crystal growth of YB66 was investigated to develop the single crystal of YB66 as a spectral material for synchrotron emitted soft X-ray. The emitted light is white light including from visible radiation to hard X-ray. Usually, it is used as homogeneous light through spectra. There are K-absorption edges of Mg and Si in the region ranging from 1 to 2 keV, which is significant for material science. There has been no proper spectral elemental device for application of the emitted spectra. The YB66 is the most suitable for this purpose. For the single crystal growth of high crystalline YB66, high frequency indirect heating floating method has been developed. For the growth furnace, a mechanism has been developed, by which pressurized gas atmosphere can be sealed with magnetic fluid. At the same time, the growth axis can be driven in high accuracy. From evaluation of the elemental device, energy resolution of 0.5{times}10{sup -3} was obtained as expected. By using this spectral device, accurate measurements of XAFS and EXAFS can be conducted with excellent operability for K-absorption edges of Mg and Si. 15 refs., 54 figs., 1 tab.

For the construction of houses that effectively utilize natural energy and impose less load on environments, an example of house building respecting environmental symbiosis is described. Reported herein are a heating system and the measurements of thermal environment in winter. In winter, the floor is heated by warm water from solar heat collectors circulating in pipes buried in downstair underfloor RC foundation. For the verification of the heating effect, comparison was made between the heater-provided southeast (SE) room and heater-unprovided northeast (NE) room, both downstairs. Warm water circulated in the underfloor pipes only in the daytime, the pipe surface recording the maximum of 32{degree}C in the daytime and the minimum of 17{degree}C early in the morning. At midnight, the SE room was warmer than the NE room by 2{degree}C. The slab surface in the SE room recorded 28{degree}C at the maximum and 17{degree}C at the minimum, but the temperature remained constant at 10{degree}C in the NE room. The rubble temperature was 17.5-20{degree}C in the SE room and approximately 11{degree}C in the NE room. It was noted that the SE room floor slab was capable of heat accumulation. The resident is a devotee of environmental symbiosis with mindset for enhancing natural energy utilization, burning an oil heater and a fireplace but intermittently. 1 ref., 12 figs., 1 tab.
