  1. Kinetics of saccharose fermentation by Kombucha

    Full Text Available Kinetics of saccharose fermentation by Kombucha is not yet well defined due to lack of knowledge of reaction mechanisms taking place during this process. In this research kinetics of saccharose fermentation by Kombucha was analysed using the suggested empirical model. The data were obtained on 1.5 g L-1 of black tea, with 66.47 g L-1 of saccharose and using 10% (v/v or 15% (v/v of Kombucha. Total number of viable cells was as follows: approximately 5x105 of yeast cells per mL of the inoculum and approximately 2x106 of bacteria cells per mL of the inoculum. The samples were analysed after 0, 3, 4, 5, 6, 7 and 10 days. Their pH values and contents of saccharose, glucose, fructose, total acids and ethanol were determined. A saccharose concentration model was defined as sigmoidal function at 22oC and 30oC, and with 10% (v/v and 15% (v/v of inoculum quantity. Determination coefficients of the functions were very high (R2>0.99. Reaction rates were calculated as first derivatives of Boltzmann’s functions. No simple correlation between rate of reaction and independent variables (temperature and inoculum concentration was found.     Multiple (ten times and more) increase in intensities of separate reflections and of lauegram reflections from organic single crystals of saccharose (C sub 1 2H sub 2 2O sub 1 1) was observed under influence of certain temperature gradient. On the base of the present experiment and the data of our previous woks we show that the controllable reflection process has a common nature and the intensity of the diffracted beam under external influences does not depend on the total number of electrons per unit volume of the unit cell of the single crystal.

  6. Structural Features of Carbons Produced Using Glucose, Lactose, and Saccharose

    Glucose, lactose, and saccharose were used as precursors to prepare chars at 400 °C then activated at 800 °C or 1000 °C in closed vessels with controlled amounts of oxygen penetrating through nanopores in the vessel walls. There are correlations between the porosity, amounts of residual O- and H-containing functionalities, and electroconductivity of amorphous carbons studied. The pore size distributions calculated using the nitrogen adsorption isotherms and TEM images show that all carbons are mainly nanoporous with certain contribution of narrow mesopores (at pore half-width x < 5 nm). Oxidizing activation by oxygen penetrating into the closed vessels with chars through nanopores can more strongly change the outer layers of char particles than the inner pores. Therefore, despite relatively great burn-off degree, the textural characteristics are relatively low for activated carbons.

    Full Text Available INTRODUÇÃO: A ingestão adequada de folato é essencial durante a embriogênese, e sua deficiência está associada à ocorrência de defeitos no fechamento do tubo neural. OBJETIVO: Determinar se a sacarose é um bom veículo para a suplementação de folato em camundongos. MATERIAL E MÉTODOS: Quarenta camundongos Swiss fêmeas foram divididos nos grupos: C: ração comercial + água ad libitum; DS: ração balanceada isenta de folato + folato adicionado à sacarose diluída na água por 14 dias; D/DS: ração balanceada isenta de folato + água com sacarose sem folato por 14 dias seguida de ração balanceada isenta de folato + folato adicionado à sacarose diluída na água por mais 14 dias; D: ração balanceada isenta de folato + água com sacarose sem folato por 14 dias. Os animais de todos os grupos experimentais receberam ração balanceada isenta de folato + folato adicionado à sacarose diluída na água durante os três dias do acasalamento e nos 15 dias restantes até o sacrifício. RESULTADOS: Os animais dos grupos D e D/DS apresentaram alopecia, palidez ocular e adinamia enquanto consumiam água com sacarose sem folato, sinais que foram revertidos quando receberam folato adicionado à sacarose diluída na água. Não houve diferença entre os grupos em relação a prenhez, implantes, fetos vivos, reabsorção, morte fetal tardia, nível sérico de folato e contagem de hemácias ao final do experimento, não tendo sido observadas anomalias congênitas em nenhum dos grupos. CONCLUSÃO: A sacarose é um meio adequado para a suplementação de folato na dieta.Adequate folate intake is essential during embryogenesis and its deficiency is associated with neural tube defects.     Full Text Available Glucose-oxidase (GOD, suffers conformational change during freeze-drying. In order to determine the protection level granted by amorphous matrices (AM of saccharose, maltose, trehalose and their combinations, the thermal inactivation constants (K D of GOD trapped in these systems were determined. For its evaluation, GOD samples were balanced at different water activities and heated up to 30, 50 and 70 ºC. The best AM found for GOD stability was saccharose-trehalose (5/10% p/v. The K D values (K D.10-4 at a w = 0.0 were 3 at 30 ºC and 6 at 70 ºC. For non-protected GOD under the same conditions these values were 48 at 30 ºC and 257 at 70 ºC.

    Tests conducted with rats demonstrated that rations carrying saccharose cause a rise in the pre-beta-lipoproteids, blood triglycerides, total lipids and triglycerides in the aorta, as well as an accelerated biosynthesis of the latter in the liver and the fatty tissue. The effect of the saccharose making part of an isocaloric ration depends upon the quality of the diet as a whole. In rats receiving saccharose in a ration with a reduced amount of fat (11% bythe calorific value) there takes place an accelerated biosynthesis of phospholipids with no evidence of fatty degeneration of the liver. Animals receiving saccharose in a ration with a physiological level of fat (26% by caloricity) demonstrated a higher content of beta-lipoproteids in the blood, of total lipids and tryglycerides in the liver with lacking acceleration of the phospholipids biosynthesis in the latter.

    Silver nanoparticles are well known for their catalytic and antimicrobial properties. In their production, the modified Tollens' process using saccharides as reduction agents is very popular. In this paper, the possibility of silver nanoparticles reduction by fructose, glucose, galactose, mannose, maltose, lactose and saccharose is shown. The size of successfully prepared nanoparticles was 16-70 nm depending on the saccharide type. The influence of NaOH and NH3 presence in reaction mixture on size of nanoparticles was described. Surprisingly good results were obtained using saccharose that is, however, known as non-reducing disaccharide.

    Mulberry pekmez can be adulterated in different ways either during the production process or after production is completed. To identify these adulterations, stable carbon isotope ratio analysis (SCIRA) was performed on the model examples prepared by adding saccharose syrup (SS), glucose syrup (GS) and high fructose corn syrup (HFCS) into two different pure mulberry pekmez samples in the ratios of 0%, 10%, 30% and 50%. The δ(13)C ratio of the pure mulberry pekmez was determined as -26.60‰ on average, the saccharose syrup as -24.80‰, the glucose syrup as -11.20‰ and the high-fructose corn syrup as -11.40‰. In identifying the adulteration made to pekmez, especially with the high-fructose corn syrup, which is obtained from corn starch, and with the glucose syrup, the δ(13)C ratio comes into prominence. However it remains impossible identify the adulterations made with the saccharose, which is obtained from beet sugar, or invert sugar syrups. Copyright © 2014 Elsevier Ltd. All rights reserved.

    Lactose and saccharose have the same molecular formula; however, the arrangement of their atoms is different. A major difference between lactose and saccharose with regard to digestion and processing is that it is not uncommon for individuals to be lactose intolerant (around two thirds of the population has a limited ability to digest lactose after infancy), but it is rather unlikely to be saccharose intolerant. The pharmaceutical industry uses lactose and saccharose as inactive ingredients of drugs to help form tablets because of their excellent compressibility properties. Some patients with severe lactose intolerance may experience symptoms of many allergic reactions after taking medicine that contains this substance. People who are specifically "allergic" to lactose (not just lactose intolerant) should not use tablets containing this ingredient. Fourier Transform Infrared (FTIR) spectroscopy has a unique chemical fingerprinting capability and plays a significant important role in the identification and characterization of analyzed samples and hence has been widely used in pharmaceutical science. However, a typical FTIR spectrum collected from tablets contains a myriad of valuable information hidden in a family of tiny peaks. Powerful multivariate spectral data processing can transform FTIR spectroscopy into an ideal tool for high volume, rapid screening and characterization of even minor tablet components. In this paper a method for distinction between FTIR spectra collected for tablets with or without lactose is presented. The results seem to indicate that the success of identifying one component in FTIR spectra collected for pharmaceutical composition (that is tablet) is largely dependent on the choice of the chemometric technique applied.

    The feeding of rats for a space of 30 days on diets with elevated content of starch or saccharose (71 per cent of the total calorific value) was followed by an accelerated synthesis and secretion into the blood of pre-beta-lipoproteins. The ration with succharose, however, produced a much greater effect on the rate of synthesis of pre-beta-lipoproteins than did the one containing starch. The action of both carbohydrate-rich rations was noted to be attended by a growing rate of the apoproteids and glycerides exchange in the low-density lipoproteins. The feature distinguishing the effect produced by the ration with saccharose, as compared with that containing starch, was an accelerated etherification of cholesterol in the blood.

    Sugars and organic acids of tomatoes, variety Original Fruehrot, in different stages of development were examined by gas chromatography after silylation. Changes in the carbohydrate and organic acid content of half-ripe and ripe tomatoes were determined after treatment with different radiation doses (5, 15, 30, 60, 250 and 500 krad). Fructose, glucose, saccharose and malic acid could be detected in the tomato samples. The saccharose and malic acid contents of half-ripe tomatoes decreased as a result of irradiation with doses above 250 krad. In ripe tomatoes, the malic acid content fell most markedly under the influence of irradiation. The sugar content tended to decrease as a function of storage time, except for samples treated with 15 krad which usually delayed decomposition of the components tested. The effect of irradiation was more extensive in half-ripe tomatoes than in ripe ones.

    Isopropyl-thio-galactoside (IPTG), an inducer of 3-galactosidase, makes it possible to synthesise this enzyme in vitro with the subcellular structure (P{sub 1}). The enzyme is isolated from the bacteria Escherichia coli K 12 which are inductive but not induced. The incorporation of radioactive amino-acids, which is stimulated by the presence of an inducer, was studied during the course of the enzyme synthesis. Saccharose suppresses the induction of {beta}-galactosidase. The presence of a specific inhibitor in the structure studied is considered.     Full Text Available The recovery of native potato varieties is a current priority in Venezuela, given their value as a genetic resource of high nutritional quality. Unfortunately, native potato varieties were progressively replaced by exotic commercial, varieties. To date, the limited existing germplasm of native potato varieties is impoverished and of low quality, due to its continual reutilization in crop cycles. Nevertheless, efforts can be made to recover and standardize the production of quality propagules under adequate sanitary conditions. The aim of this study was to assay the effect of varying concentrations of nitrates and saccharose in tissue culture media of three Venezuelan varieties "Cucuba", "Arbolona Negra" and "Rosada". Unimodal segments were planted and incubated using a photoperiod of 16 h light (76 μmol photon m-2 s-1 at 19 ± 1 ºC. The experiment was designed following a standard factor analysis, consisting of three treatments, parting from the basic Murashige & Skoog (MS medium and data were submitted to an multifactor ANOVA. Our findings indicate significant statistical differences amongst all of the treatments assayed, confirming that all of the varieties reached maximum physiological response under increasing concentrations of nitrates. Such was the case with 1.98 g L-1 ammonium nitrate and 2.28 g L-1 potassium nitrate using a concentration of saccharose 20g L-1. Mean shoot and root lengths under optimal concentrations were 5.82 cm and 3.0 cm, respectively. In contrast, MS basic culture media represented the treatment of least growth induction; yielding un rooted shoots of a mean length of 2.94 cm. Of these three native varieties, "Cucuba" proved to have the highest growth rates. All of the Vitroplantlets were then transferred to liquid MS media, with a saccharose concentration of 8 %, originating microtubers after 90 days. We conclude that these findings may be of use for massive in vitro production of native potato varieties.

    The phytochemical investigation of Sabicea brasiliensis roots led to the isolation of 5-O-caffeoylquinic acid, 3,5- and 4,5-O-dicaffeoylquinics acids, scopoletin, ursolic acid, a mixture of β-sitosterol, stigmasterol and campesterol, daucosterol and saccharose. The structures of the isolated compounds were assigned on the basis of one- and two-dimensional NMR spectroscopic methods and by comparison with literature data. The anti-inflammatory and antioxidant activities of the crude methanolic extract and its fractions were analyzed. (author)

    Developmental centered care (DC) is focused on sensorineural and emotional development of the newborns. In Spain we have had information on the application of DC since 1999, but the extent of actual implementation is unknown. To determine the level of implementation of DC in Spanish neonatal units where more than 50 infants weighing under 1500g were cared for in 2012. A comparison was made with previous data published in 2006. A descriptive observational cross-sectional study was performed using a survey with seven questions as in the 2006 questionnaire. The survey was sent to 27 units. The response rate was 81% in 2012 versus 96% in 2006. Noise control measures were introduced in 73% of units in 2012 versus 11% in 2006 (P<.01). The use of saccharose was 50% in 2012 versus 46% in 2006 (P=.6). Parents free entry was 82% in 2012 versus 11% in 2006 (P<.01). Kangaroo care was used without restriction by 82% in 2012 compared to 31% in 2006 (P<.01). The implementation of the DC in Spain has improved. There is still room for improvement in areas, such as the use of saccharose or noise control.     Full Text Available The article presents an investigation on the effectiveness of nanostructured titanium-10 wt% 45S5 Bioglass-1 wt% Ag composite foams as a novel class of antibacterial materials for medical applications. The Ti-based composite foams were prepared by the combination of mechanical alloying and a “space-holder” sintering process. In the first step, the Ti-10 wt% 45S5 Bioglass-1 wt% Ag powder synthesized by mechanical alloying and annealing mixed with 1.0 mm diameter of saccharose crystals was finally compacted in the form of pellets. In the next step, the saccharose crystals were dissolved in water, leaving open spaces surrounded by metallic-bioceramic scaffold. The sintering of the scaffold leads to foam formation. It was found that 1:1 Ti-10 wt% 45S5 Bioglass-1 wt% Ag/sugar ratio leads to porosities of about 70% with pore diameter of about 0.3–1.1 mm. The microstructure, corrosion resistance in Ringer’s solution of the produced foams were investigated. The value of the compression strength for the Ti-10 wt% 45S5 Bioglass-1 wt% Ag foam with 70% porosity was 1.5 MPa and the Young’s modulus was 34 MPa. Silver modified Ti-10 wt% 45S5 Bioglass composites possess excellent antibacterial activities against Staphylococcus aureus. Porous Ti-10 wt% 45S5 Bioglass-1 wt% foam could be a possible candidate for medical implants applications.

    During the last years the Brazilian floriculture expanded and it is expected to achieve a prominent role in near future. The national territory has many favorable regions, with adequate climate for the development of commercially important cultures. Flower exportation is not expressive yet, but the country has conditions to become a great exotic tropical plants exporter. On the other hand, developed countries, as USA and Japan, have a rigorous phyto sanitary inspection to prevent the introduction of new plagues and diseases through fresh products. Ionizing radiation is considered a reliable disinfestation method to control numerous fruit and flower plagues. The aim of of this work was to verify the tolerance of some Compositae family flowers to ionizing radiation and the effects to sucrose action in protecting the flowers against the gamma-radiation induced damages. This was done by measuring biochemical and physiological parameters as a function of time after irradiation. The chrysanthemum flowers were sensitive to gamma radiation, but when the inflorescences were supplied after irradiation with preservative solution containing 2% sucrose, protection against the damaging effects of radiation was achieved. The dose of 750 Gy, considered appropriate for disinfestation purpose, did not modify the protein and lipid levels, nor plasma, tonoplast and mitochondria membrane ATPase or mitochondria cytochrome-c oxidase activities. The ethylene and carbonic gas rate production increased soon after the irradiation, but decreased one day later. The decrease of the microsomal membrane fluidity and the increase of the chrysanthemum were the most sensitive parameters to measure the irradiation treatment changes. The sucrose supply was able to maintain the irradiated flowers membrane fluidity level close to the unirradiated control. Rhodante manglesii Lindl and Helichrysum bracteatum Andr. were tolerant up to 1 KGy, thus being adequate to be disinfected by gamma radiation. (author)

  18. Extracellular enzymatic activities and physiological profiles of yeasts colonizing fruit trees.

    Yeasts form a significant and diverse part of the phyllosphere microbiota. Some yeasts that inhabit plants have been found to exhibit extracellular enzymatic activities. The aim of the present study was to investigate the ability of yeasts isolated from leaves, fruits, and blossoms of fruit trees cultivated in Southwest Slovakia to produce extracellular enzymes, and to discover whether the yeasts originating from these plant organs differ from each other in their physiological properties. In total, 92 strains belonging to 29 different species were tested for: extracellular protease, β-glucosidase, lipase, and polygalacturonase activities; fermentation abilities; the assimilation of xylose, saccharose and alcohols (methanol, ethanol, glycerol); and for growth in a medium with 33% glucose. The black yeast Aureobasidium pullulans showed the largest spectrum of activities of all the species tested. Almost 70% of the strains tested demonstrated some enzymatic activity, and more than 90% utilized one of the carbon compounds tested. Intraspecies variations were found for the species of the genera Cryptococcus and Pseudozyma. Interspecies differences of strains exhibiting some enzymatic activities and utilizing alcohols were also noted. The largest proportion of the yeasts exhibited β-glucosidase activity and assimilated alcohols independently of their origin. The highest number of strains positive for all activities tested was found among the yeasts associated with leaves. Yeasts isolated from blossoms assimilated saccharose and D-xylose the most frequently of all the yeasts tested.     Aim: the aim of this work is to study the influence of medium density on the CT or external source attenuation corrected images, by simulation on a phantom, with various positron emission tomographs. Material and method: a series of experiments on a cylindrical phantom filled with water marked with [18 F]-FDG, containing six vials filled per pair with mediums of different densities or solutions of KI, CaCl 2 and saccharose with various densities, was carried out under comparable conditions on three different tomographs. In only one of the vials of each pair, an identical radioactivity of [18 F]-FDG was added, three to five fold the surrounding activity. The reconstructions and attenuation corrections suggested by the manufacturers, were carried out under the usual conditions of each site. The activity of each structure was estimated by the methods of profiles and regions of interest, on the non attenuation corrected images (N.A.C.), the images corrected by CT (C.T.A.C.), and/or external source (G.P.A.C.). Results: with all three tomographs, the activities estimated on the N.A.C. images present an inverse correlation to the medium density (important absorption by dense material). On C.T.A.C. images, we observed with only two of the three tomographs, an overestimation of the activity in the 'radioactive' vials, depending on the medium mean Z number and density (over correction), and a artifactual 'activity' in the denser 'cold' vial (incorrect attenuation correction. The dense saccharose solutions, with non elevated Z number, do not affect the CT attenuation correction. (authors)

  3. Combined effect of water activity, radiation dose, and temperature during irradiation on the survival of three Salmonella serotypes

    Stationary-phase cells of S. typhimurium, S. thompson and S. senftenberg were irradiated at room temperature with 10 MeV electrons, either in water or in glucose, glyzerine, NaCl, CaCl 2 , or LiCl solutions of different concentrations. Furhter experimental parameters, i.e. the temperatures O 0 C and 50 0 C, were investigated in samples whose water activity had been adjusted to certain asub(W) values. Using the sorption isotherm method, different water activities were obtained in egg powder at temperatures of 5 0 C, 25 0 C, and 45 0 C. In all experiments, the survival of Salmonellae was investigated by determining the number of surface germs. In saccharose, glucose, glyzerine, and NaCl, the radioresistance of Salmonellae increases with decreasing water activity. There is a linear relation at different temperatures between D 10 values and asub(W) values in saccharose and glucose. In glyzerine solutions, the D 10 values show a steep rise with decreasing asub(W) = 1.0 and 0.9 and a less steep rise below 0.9. Below asub(W) = 0.5, they do not change any further. At asub(W) values below 0,90, CaCl 2 and LiCl show a strong cytotoxic effect, independent of irradiation. The higher the temperature on irradiation, the lower will be the radioresistance of Salmonellae. Irradiation at a temperature of 50 0 C showed a synergistic effect. The radioresistance of Salmonellae in egg powder increased with decreasing asub(W) value of the egg powder. When egg powder was irradiated at 45 0 C, there was a strong synergistic effect which is important for practical applications of irradiation in egg powder sterilisation.     The wine products showed different profiles of carbohydrates, organic acids, phenolic compounds, and minerals. Especially, saccharose (max. 0.21 g/L, rhamnose (max. 0.45 g/L, fructose (max. 1.9 g/L, and phosphate (max 1.4 g/L quantities were extremely high in some wines. The results also showed that yeast fermentation in winemaking agitated high production of lactic (max 5.7 g/L and tartaric (max 1.7 g/L acids. The red wines processed by cold maceration and natural fermentation gave similar profiles. Only one of the Pinot Noir wines entirely differentiated from the others with comparison of carbohydrates and organic acids.

    Full Text Available Abstract This paper proposes to study the sedimentation characteristics of anaerobic sludge, by determining the settling velocity of sludge granules with the Griffith Tube. This is a simple, low-cost method, suitable for use in full-scale treatment plants. The settling characteristics of sludge from two laboratory-scale UASB reactors fed with saccharose and different concentrations of sodium oleate and sodium stereate were evaluated. Addition of fatty acids caused a gradual destabilization of the system, affecting overall performance. The sedimentation profile changed after addition of fatty acids to the synthetic substrate, decreased sedimentation velocity and increased granule diameter. This behaviour was attributed to the adsorption of fatty acids onto the granules, modifying the diameter, shape and density of these bioparticles.

    Full Text Available Immobilized yeast invertase was applied for treatment of alcoholic beverages with the aim of transformation of higher alcohols into alkylfructosides. Gas-liquid chromatography of treated water-alcoholic medium containing 3.0 mg/l isoamyl alcohol and 4% saccharose by immobilized invertase had shown the convertion of 40% isoamyl alcohol, which amounts to 1.8 mg/l absolute alcohol. Other parameters remained at the previous level. The high level of enzyme activity was observed when the initial concentration of sucrose in the reaction mixture attained 4.0-12.5%. Tasting of treated samples indicated the improvement of quality and degustational properties of beverages, they had softer and more harmonious taste and aroma in comparison with control sample and finished Vodka, which completed the cycle of technological processing.

    A model system which permits observing for a long time and fixing interaction of fungi with a radiation source has been created on the basis of an isolated "hot" particle, deficient mineral medium (saccharose content 60 mg/l) and suspension of fungal conidia. Five species (six strains) of micromycetes isolated from radionuclide-contaminated soils and fifteen "hot" particles have been tested. It has been found out for the first time that Cladosporium cladosporioides and Penicillium roseo-purpureum are able actively overgrow "hot" particles whose radioactivity did not exceed 3.1-1.0(-7) Ci by gamma-spectrum and to destroy them 50-150 days later. Certain changes in morphology of fungi-destructors of "hot" particles are revealed. A problem on ecological significance of the found phenomenon is discussed.

    Ionizing radiation effect on DNA replication on fibroblasts of a healthy donor and a patient with Down's syndrome either by direct 3 H-thymidine inclusion into DNA, or by analysis of the sizes of daughter DNA moleculs at the state of stable distribution in acid saccharose, gradients was studied. Gamma-radiation doses (5-10 Gy) suppressing DNA synthesis in normal fibroblasts practically had no effect on DNA synthesisin fibroblasts of a patient with Down's syndrome. Radioresistant DNA synthesis in Down's syndrome is conditioned by a far less supression of replicon initiation as compared with the one in normal cells. So, it is stated that in Down's disease there is no delay in DNA synthesis by ionizing radiation that enables the normal cells to repair DNA damages before replication renewal

    Full Text Available The objective of this study was to determine the physicochemical characteristics and thermophysical properties of cocoa hoeny. The cocoa honey had the following physicalchemical characteristics: pH (2.76, titratable acidity (0.73 %, moisture (87,22 %, soluble solids (14,03 °Brix, reducing sugar (10,2 % in glucose, non-reducing sugar (4,06 % in saccharose and ash (0,23 %. With respect to the thermophysical properties were determined the specific heat, density, thermal diffusivity and the dynamic viscosity as a function of temperature. The empirical models for each property were obtained. It was found that the temperature directly affects the cocoa liquor properties. The data are important for the development, adaptation and optimization of equipment for more efficient processing of cocoa honey, since the information on this subject is unknown.

    In the fulfillment of this study, pineapple of the spanish red variety, cut in 1.0 - 1.5 cm slices, were used. The product was sunken in a saccharose syrup with the addition of sodium methabisulphite and potassium sorbate and packed in polyethylene bags and latter irradiated with 0.2 and KGy doses. The behavior of potassium sorbate with two addition levels is similar during storage at room temperature for the studied irradiation doses. The degradation process of sodium methabisulphite takes place almost totally at one month storage. Unlike of potassium sorbate there exist no meaning differences between sulfur dioxide in irradiated and nonirradiated samples.It is concluded that the irradiation process cause an increasing in the degradation of potassium sorbate basically at the beginning of the storage, while seems no exert some effect on the sodium methabisulphite degradation. We described the methods of sampling and cultivation as well as diagnostic criteria. Pasteurella multocida was diagnosed on the basis of characteristic cultural and tinctorial properties and the facts that all strains produced indole and induced ornithine decarboxilation, glucose, saccharose and manitole fermentation. Isolates produced neither urease, nor fermented lactose and maltose. Further classification to subspecies multocida was based on the fact that all investigated isolates fermented trechalose, xylose and sorbitol the traits which are diagnostically significant for the species. Patients deny any contact with farm animals or pets, which indicates a possible aerosol transport and animal-human as well as human-human infection. We consider that this organism should be paid more attention by dentist, oral surgeons and microbiologists.

    In the fulfillment of this study, pineapple of the spanish red variety, cut in 1.0 - 1.5 cm slices, were used. The product was sunken in a saccharose syrup with the addition of sodium methabisulphite and potassium sorbate and packed in polyethylene bags and latter irradiated with 0.2 and KGy doses. The behavior of potassium sorbate with two addition levels is similar during storage at room temperature for the studied irradiation doses. The degradation process of sodium methabisulphite takes place almost totally at one month storage. Unlike of potassium sorbate there exist no meaning differences between sulfur dioxide in irradiated and nonirradiated samples.It is concluded that the irradiation process cause an increasing in the degradation of potassium sorbate basically at the beginning of the storage, while seems no exert some effect on the sodium methabisulphite degradation.     Full Text Available O objetivo deste trabalho foi avaliar o efeito da adição de condimentos na carne de coelho, sobre a estabilidade térmica de suas proteínas miofibrilares. Retalhos de 24 músculos Longissimus dorsis foram moídos e misturados, para a obtenção de um lote homogêneo. Foram adicionados, em alíquotas dessa carne: 1,0; 2,0 ou 3,0% de sal; 2,5; 5,0 ou 7,5% de sacarose e 1,5; 3,0 ou 4,5% de ácido acético 3% em solução aquosa, e uma mistura perfazendo 3% de sal, 5% de sacarose e 3% da mesma solução de ácido acético. Após homogeneização manual, as amostras foram mantidas em repouso sob refrigeração, por 30 minutos. Alíquotas da ordem de 10mg foram usadas para análise térmica e o restante, para análises de pH, proteínas e umidade. As análises térmicas foram realizadas em um calorímetro diferencial de varredura DSC-TA2010, a 10ºC/min, entre 0 e 100ºC. As curvas de DSC da carne pura apresentaram endotermas nas temperaturas de (Td 58,4(0,7ºC (miosina e 78,2(0,2ºC (actina, envolvendo entalpia (DHd de 17,5J/g de proteínas. O sal e o ácido acético desestabilizaram as proteínas, provocando redução da Td da miosina e actina, contrariamente à sacarose, que apresentou efeito estabilizante. A adição da mistura dos condimentos desestabilizou as proteínas miofibrilares. Apenas o ácido, adicionado acima de 1,5%, provocou desnaturação das proteínas.The objective of this work was to evaluate the effect of some additives over the myofibrillar protein thermal stability of rabbit meat. Fuji, fisiologicamente maduras e super maduras, foram fatiadas na forma de palitos, concentradas osmoticamente e desidratadas, objetivando-se avaliar suas características físicas, químicas e sensoriais. A concentração osmótica foi com soluções a 50% de sacarose, maltodextrina de milho (Mor-rex 1920 e glicose de milho (Excell 1040, sendo os dois últimos produzidos pela Refinações de Milho Brasil. Os palitos de maçãs foram imersos nas soluções concentradoras, por seis horas, drenados e desidratados a 70º C até 10% de umidade pelo controle de peso, seguindo-se armazenamento ambiental em vidros. As avaliações da matéria-prima e produto final foram de umidade, sólidos solúveis totais (º Brix, açúcares, pH, acidez titulável, aceitação pelo consumidor e rendimento. A avaliação estatística dos dados seguiu o delineamento inteiramente ao acaso, com médias submetidas a comparações múltiplas através do teste de Duncan ao nível de 5% de probabilidade. A composição da maçã "in natura" foi similar a encontrada na literatura. Os agentes de concentração osmótica passaram a fazer parte da composição dos produtos desidratados, alterando as características físicas e químicas em ambos estádios de maturação. O rendimento melhorou nos três tratamentos, sendo o melhor com sacarose que, também, resultou na preferência dos consumidores que optaram por "gosto muito", independentemente dos estádios de maturação das maçãs. Although there are many studies regarding phenolic compounds in wines, there is a lack of information about antioxidants and phenolic content in South American wines. In this study, 35 South American red wines from four different countries, vintages 2004–2013, purchased at retail stores in La Paz, Bolivia, were studied. Resveratrol content, total antioxidant capacity (TAC by the 2,2′-azino-bis(3-ethylbenzotiazoline-6-sulfonic acid (ABTS and ferric-reducing antioxidant power (FRAP methods, total phenolic content (TPH, total flavonoids (TF, and main saccharides were assessed using the well-established spectrophotometric and high-performance liquid chromatography methods. The results ranged from 4 to 24 mmol/L for TAC determined by ABTS method and 14 to 43 mmol/L for TAC determined by FRAP method, 1600 to 3500 mg gallic acid equivalents/L for TPH, and 2 to 6 mmol catechin equivalents /L for TF. The resveratrol content ranged from 0.1 to 8 mg/L. Saccharides, glucose, and fructose content ranged from 0.4 to 10 g/L, 1.4 to 8.6 g/L, and 0.2 to 12 g/L, respectively. There was a high correlation among the different methods. The results showed that some wines growing at high altitude (>1500 meters above the sea level have higher amounts of TAC and phenolic content, including resveratrol, while non-varietal wines showed the lowest values.     Degree of microbiological decontamination, organoleptic and physico-chemical properties of natural honeys were investigated after radiation treatment. Seven kinds of honeys were irradiated with the beams of 10 MeV electrons from a 10 kW linear accelerator "Elektronika 10-10" at the dose 10 kGy. It was shown, that after irradiation, the total count of aerobic and anaerobic bacteria and moulds decrease by 99%. The antibiotic value in investigated honeys increased in turn from 1.67 to 2.67 after irradiation. Such factors and parameters of investigated honeys as their consistency, content of water and saccharose, acidity, the diastase and 5-HMF values were not changed significantly after irradiation. Decontamination by irradiation is a process which allows us to obtain high microbiological purity of honeys. It is especially needed, when honeys are used in surgical treatment of injuries and in nutrition of babies with food deficiency.

    This report presents the results of an investigation into the effects of ionizing radiation on eukaryotic cells, aimed at revealing the molecular mechanisms leading to cell inactivation as a result of ionizing radiation. The quantitative determination of radiation-induced double strand breaks (DSB) is done via sedimentation of the DNA released from the cells in a neutral saccharose gradient in a preparative ultracentrifuge. The 'experimental mass spectrum' of DNA molecules thus obtained, the mean number of DSB per cell is calculated using a special computer program which simulates the stochastic induction of DSB in the DNA of non-irradiated cells and links the 'simulated' mass spectrum with the 'experimental' one on the basis of the least square fit.     The components of the food related to the "deliciousness" are usually evaluated by componential analysis. The component content and type of components in the food are determined by this analysis. However, componential analysis is not able to analyze measurements in detail, and the measurement is time consuming. We propose a method to measure the two-dimensional distribution of the component in food using a near infrared ray (IR) image. The advantage of our method is to be able to visualize the invisible components. Many components in food have characteristics such as absorption and reflection of light in the IR range. The component content is measured using subtraction between two wavelengths of near IR light. In this paper, we describe a method to measure the component of food using near IR image processing, and we show an application to visualize the saccharose in the pumpkin.

  2. A simple method for preparing artificial larval diet of the West Indian sweetpotato weevil, Euscepes postfasciatus (Fairmaire) (Coleoptera: Curculionidae)

    The method for preparing ordinary larval artificial diet for Euscepes postfasciatus (old diet) was complicated and time consuming. Some ingredients (casein, saccharose, salt mixture, etc.) of the diet were added to boiled agar solution, others (vitamin mixture, sweetpotato powder, etc.) were added after the solution was cooled to 55degC. To simplify the diet preparation, we combined all ingredients before mixing with water, and then boiled the solution (new diet). There were no significant differences of survival rate (from egg hatching to adult eclosion) and right elytron length between the weevils reared on the old and new diets, but the development period (from egg to adult) of the weevils fed the new diet was significantly (1.3 days) longer than that of those fed the old diet. Preparation time of the new diet was half that of the old diet. These results suggest that simplified diet preparation can be introduced into the mass-rearing of E. postfasciatus

    A nonpathogenic bacterial strain Bacillus amyloliquefaciens TUL 308 synthesized minor 2,3-butanediol (2,3-BD) amounts from glucose, fructose, sucrose, and glycerol, and efficiently produced the diol from molasses and hydrolysates of food processing residues. Batch fermentations yielded 16.53, 10.72, and 5 g/L 2,3-BD from enzymatic hydrolysates of apple pomace, dried sugar beet pulp, and potato pulp (at initial concentrations equivalent to 45, 20, and 30 g/L glucose, respectively), and 25.3 g/L 2,3-BD from molasses (at its initial concentration equivalent to 60 g/L saccharose). Fed-batch fermentations in the molasses-based medium with four feedings with either glucose or sucrose (in doses increasing their concentration by 25 g/L) resulted in around twice higher maximum 2,3-BD concentration (of about 60 and 50 g/L, respectively). The GRAS Bacillus strain is an efficient 2,3-BD producer from food industry byproducts.

    Full Text Available The duration of dormancy varies significantly among cultivars, but even short dormancy can limit usage of potato microtubers for seed production. The aim of the research was to test efficacy of dormancy breaking by treatment with an aqueous solution of ethyl alcohol, saccharose, gibberellic acid and kinetin (ethanol treatment in comparison to treatment with aqueous solution of thiourea, daminozide and gibberellic acid (standard treatment. Prolonging the period of microtuber production at the in vitro stage significantly favored the short-ening of the dormancy and facilitated its breaking. While the standard treatment had the strongest effect, the ethanol treatment was slightly less efficient. The statistically significant differences were only observed during the first 13 days after the microtuber treatment. After that time, efficacy of ethanol and standard treatments was similar to control treatment with water. The investigated treatments had no effect on the natural decrease of ABA level in microtubers.

    Full Text Available This study was conducted to determine the composition of sugars and organic acids in root exudates of Miscanthus × Giganteus and to find out if microorganisms of the rhizospheric soil are limited by mi­ne­ral nutrients. The following sugars and organic acids were determined in root exudates of this plant: glucose, saccharose, and acids such as succinic, propionic, citric, tartaric, malic, oxalic, ascorbic, acetic and fumaric. Respiration of soil from rhizosphere of Miscanthus × Giganteus was found to be limited by N, K and Ca. Respiration rate after application of mineral compounds increased in following orther: nitrate > calcium > potassium > ammonium, giving approx. 165, 99, 52 and 31 % increase compared to control. Further research is necessary to determine the role of plant nutrients from the point of their limitations for rhizosphere microorganisms, to broader very rare knowledges in this topic, especially for polluted soils to stimulate efficiency of phytoremediations.

    Full Text Available Oncidium varicosum is a native Brazilian orchid popularly known as ‘Golden Shower´ because of its very ramified inflorescence and many yellow flowers. The carboydrate type and concentration are important in promoting plantlet development of in vitro orchids. The present study was carried out to asses the effect of different carbohydrate sources and concentrations on the in vitro growth of O. varicosum plantlets. Murashige e Skoog culture medium was used modified with half concentration of the macronutrients. The plantlets, derived from seeds that were already established in vitro and 0.8 + 0.2 cm in height, were inoculated in the culture media containing the following carbohydrate sources: saccharine, maltose and glucose, at concentrations of 0, 10, 20, 30, 60 and 90 g.L-1. The following variables were analyzed 8 months later: canopy height, number of roots, greatest root length, pseudobulb diameter and fresh weight. A completely randomized block experimental design was used with five replications per treatment. Analysis of variance and the Tukey test (5% were performed to compare the means. It was concluded that 60 g.L-1 saccharose was the best treatment for all the parameters assessed. The sugars 30 g.L-1 glucose and 60 g.L-1 maltose were also suitable, but presented lower pseudobulb diameter and lower fresh weight when compared to 60 g.L-1saccharose.O trabalho teve o objetivo de avaliar a influência do ácido indol-butírico sobre o enraizamento de estacas de acerola (Malpighia emarginata D.     Lactase non-persistence (LNP), or primary hypolactasia, is a genetic condition that mediates lactose malabsorption and can cause lactose intolerance. Here we report the prevalence of lactose intolerance in a double-blind placebo study. The LCT C>T-13910 variant was genotyped by RT-PCR in 121 volunteers and lactose malabsorption was assessed using the hydrogen breath test (HBT) after consuming 25 g of lactose. Lactose intolerance was assessed by scoring symptoms (SS) using a standardized questionnaire following challenge with a lactose solution or saccharose placebo. The LNP genotype was observed in 57% of the volunteers, among whom 87% were HBT⁺. In the HBT⁺ group the median SS was 9 and in the HBT⁻ group the median SS was 3 (p lactose intolerance was defined as the presence of an SS ≥ 6 points after subtracting the placebo effect and 34% of the study population met this definition. The LNP genotype was present in more than half of subjects evaluated and the observed prevalence of lactose intolerance was 34%. © 2014 S. Karger AG, Basel.

    Full Text Available The impacts of AMD pollution on biological systems are mostly severe and the problem may persist from many decadesto thousands of years. Consequently AMD prior to being released into the environment must be treated to meet government standardsfor the amount of metal and non-metal ions contained in the water. One of the best available technologies for the removal of metals fromAMD is precipitation as metal sulphides. SRB applications for AMD treatment involve a few principal stages. The first stageis the cultivation of SRB i.e. the bacterial sulphate reduction. At the laboratory conditions the sodium lactate is the energetic substratefor the growth of bacteria. Its price is not economic for the application in the practice and is needed investigate the alternativesubstitutes. The aim of this work was the cultivation of SRB using the selected energetic substrates such as: calcium lactate, ethanol,saccharose, glucose and whey. Experimental studies confirm that in the regard to the amount of reduced sulphates the calcium lactateand ethanol are the best alternative substrates for the bacterial sulphate-reduction.

    Full Text Available A uva Isabel da Serra Gaúcha geralmente não alcança teor de açúcar suficiente para produzir vinho equilibrado e, em determinados casos, para atingir 10% v/v de álcool, concentração mínima para vinho de mesa estabelecida pela legislação brasileira. O objetivo do presente trabalho foi avaliar o efeito de diferentes produtos utilizados para correção do mosto na composição química do vinho Isabel. O experimento constou de seis tratamentos: testemunha (sem correção, sacarose, açúcar mascavo, mosto concentrado, álcool vínico e glicose de milho. Os vinhos foram elaborados em microvinificações, com seis repetições. As amostras foram avaliadas através das análises clássicas, efetuadas por métodos físico-químicos, e dos elementos minerais por absorção atômica. A sacarose foi o produto mais adequado para a correção do mosto, pois não incorporou componentes estranhos, não alterou a relação álcool em peso/extrato seco reduzido e apresentou elevado rendimento alcoólico. O álcool vínico pode ser utilizado desde que seja um produto genuíno de procedência garantida, preferencialmente obtido de vinho branco.The cv.     Full Text Available The goal of this study was to develop a symbiotic lacteous drink, evaluate its physicochemical and sensory characteristics, and verify the viability of Lactobacillus acidophilus in the drink. The milk serum-based drink consisted of 50% milk serum containing 10% saccharose, 25% powdered milk, 15% yacon pulp, and cultures of Lactobacillus acidophilus-La 5E and Bifidobacterium bifidum BB12. It was stored for up to 21 days under refrigeration. Coliforms and Salmonella sp were not detected, and L. acidophilus and B. bifidum provided satisfactory probiotic counts up to 21 days of storage under refrigeration. Lactobacillus acidophilus resistance to gastric acids and bile salts was detected only up to seven days of storage when evaluated in vitro. Sensory analysis and purchase viability were evaluated by consumers at 0, 7, 14, and 21 days of storage. Based on the analytical results and consumer evaluation, the drink was acceptable up to seven days of storage.

    The aqueous-ethanolic extract of Tephrosia purpurea seeds is currently exploited in the cosmetic industry as a natural ingredient of skin lotions. The aim of this study was to chemically characterize this ingredient by combining centrifugal partition extraction (CPE) as a fractionation tool with two complementary identification approaches involving dereplication and computer-assisted structure elucidation. Following two rapid fractionations of the crude extract (2 g), seven major compounds namely, caffeic acid, quercetin-3-O-rutinoside, ethyl galactoside, ciceritol, stachyose, saccharose, and citric acid, were unambiguously identified within the CPE-generated simplified mixtures by a recently developed (13)C NMR-based dereplication method. The structures of four additional compounds, patuletin-3-O-rutinoside, kaempferol-3-O-rutinoside, guaiacylglycerol 8-vanillic acid ether, and 2-methyl-2-glucopyranosyloxypropanoic acid, were automatically elucidated by using the Logic for Structure Determination program based on the interpretation of 2D NMR (HSQC, HMBC, and COSY) connectivity data. As more than 80% of the crude extract mass was characterized without need for tedious and labor-intensive multistep purification procedures, the identification tools involved in this work constitute a promising strategy for an efficient and time-saving chemical profiling of natural extracts.

    Full Text Available The aims of liophilization (freeze-drying of lactic acid bacteria are to preserve pure cultures or to prepare starters for the dairy industry. In both cases, the choice of the cryoprotectant is very important. In this work, samples of Bifidobacterium breve A71 and Bifidobacterium bifidum BbTD were freeze-dried in a new cryoprotective medium containing lactose, gelatine and glycerol (medium B. The reference medium contained saccharose, gelatine and skim milk (medium A. Before liophilization, the eutectic points of both media were determined, because the products must be cooled to a temperature below its freezing point. The success of the cryoprotectants was estimated in terms of the number of surviving organisms after lyophilization. Bifidobacterium breve A71 and Bifidobacterium bifidum BbTD freeze-dried in media A and B showed high survival rates. Bifidobacterium breve A71 showed a greater percentage survival in combination with medium B than with medium A. These results could be utilized in the manufacture of Bifidobacterium breve A71 as a starter in the diary industry because it is a human isolate which, except for acidification, has probiotic activity.

    The nature of carbohydrates that accumulate in the cells of photosynthetic green sulphur bacteria of Chlorobium limicola Ya-2002 has been investigated. It is shown by infra-red spectrometry, that carbohydrates accumulated in the cells of bacteria are identical (by 90-95%) to glycogen of the bull liver. Exogenous glucose, saccharose, maltose, did not stimulate formation of glycogen. Growth of glycogen level in the cells of bacteria was observed at addition of acetate or piruvate in the conditions of bacteria cultivation in the light and in the presence CO2 and H2S in the environment. Washed cells of C. limicola Ya-2002 did not use glucose of the environment neither in the conditions of illumination nor in darkness, however acetate and piruvate are actively used in the light. During incubation of the washed cells in darkness the level of glycogen fell down approximately three times. Its amount during cells incubation in the light did not change. The decline of glycogen level in cells during their incubation in darkness was accompanied by piling up of carbonic acids in the environment acetate prevailing among them.

Materials and methods: Rheological characteristics were determined on a Reotest device by the 2 nd method of viscometry Results and discussion. Reasonability of ß-galactosidase use for milk lactose hydrolyze during the production of canned products with sugar was proved in the previous works. This technology gives possibility to increase the quality of condensed canned foods, to reduce sugar concentration till 50 %, to increase dietary properties. Due to the reducing of saccharose mass part till 22 and 31 % the products had a liquid consistency that’s why was a necessity to increase the viscosity properties of condensed products. One of method to increase the product viscosity is inoculation of stabilization systems. Reasonability of the usage of stabilization system Bivicioc 1L was proved. The researches of viscosity determination in whole hydrolyzed sweetened condensed milk were shown in the work. Relations of viscosity of whole hydrolyzed condensed milk to the deformation rate were presented. Conclusions Viscosity indices of experimental samples in the fresh produced products and during storage are determined and justified.

    Full Text Available The physiological repair of osteochondral lesions requires the development of a scaffold that is compatible with the structure of the damaged tissue, cartilage and bone. The aim of this study was to evaluate the biological performance of a PLDLA/PCL-T (90/10 scaffold for repairing osteochondral defects in rabbits. Polymeric scaffolds containing saccharose (75% w/v were obtained by solvent casting and then implanted in the medial knee condyles of 12 New Zealand rabbits after osteochondral damage with a trephine metallic drill (diameter: 3.3 mm in both medial femoral condyles. Each rabbit received the same treatment, i.e., the polymeric scaffold was implanted on the right side while no material was implanted on the left side (control. Four and 12 weeks later histological examination revealed bone neoformation in the implant group, with the presence of hyaline cartilage and mesenchymal tissue. In contrast, the control group showed bone neoformation with necrosis, exacerbated superficial fibrosis, inflammation and cracks in the neoformed tissue. These findings indicate that the PLDLA/PCL-T scaffold was biocompatible and protected the condyles by stabilizing the lesion and allowing subchondral bone tissue and hyaline cartilage formation.

    In the present study, we assessed the agro-industrial performance of 22 sugarcane genotypes adaptable to edaphoclimatic conditions in production microregions in the State of Pernambuco, Brazil, and we recommended the commercial cultivation of select genotypes. The variables analyzed were as follows: sucrose percentage in cane juice, tonnage of saccharose per hectare (TPH), sugarcane tonnage per hectare (TCH), fiber, solid soluble contents, total recoverable sugar tonnage (ATR), and total recoverable sugar tonnage per hectare (ATR t/ha). A randomized block design with 4 repeats was used. Combined variance of the experiments, genetic parameter estimates, and environment stratification were analyzed. Phenotypic adaptability and stability were analyzed using the Annicchiarico and Wricke methods and analysis of variance. Genetic gain was estimated using the classic index and sum of ranks. Genotype selection was efficient for TPH, TCH, and ATR t/ha. Genotypes presented a great potential for improvement and a similar response pattern in Litoral Norte and Mata Sul microregions for TPH and TCH and Litoral Norte and Litoral Sul microregions for ATR t/ha. Genotypes SP78-4764, RB813804, and SP79-101 showed better productivity and phenotypic adaptability and stability, according to the Wricke and Annicchiarico methods. These genotypes can be recommended for cultivation in the sugarcane belt in the State of Pernambuco.

    The process by which organic matter decomposes deep underground to form petroleum and its underlying kerogen matrix has so far remained a no man's land to theoreticians, largely because of the geological (Myears) timescale associated with the process. Using reactive molecular dynamics and an accelerated simulation framework, the replica exchange molecular dynamics method, we simulate the full transformation of cellulose into kerogen and its associated fluid phase under prevailing geological conditions. We observe in sequence the fragmentation of the cellulose crystal and production of water, the development of an unsaturated aliphatic macromolecular phase and its aromatization. The composition of the solid residue along the maturation pathway strictly follows what is observed for natural type III kerogen and for artificially matured samples under confined conditions. After expulsion of the fluid phase, the obtained microporous kerogen possesses the structure, texture, density, porosity and stiffness observed for mature type III kerogen and a microporous carbon obtained by saccharose pyrolysis at low temperature. As expected for this variety of precursor, the main resulting hydrocarbon is methane. The present work thus demonstrates that molecular simulations can now be used to assess, almost quantitatively, such complex chemical processes as petrogenesis in fossil reservoirs and, more generally, the possible conversion of any natural product into bio-sourced materials and/or fuel.

  19. Performance of Chicks Submitted to Fasting Post-Hatching and with Maltodextrine Supplementing to Diet as Hydrating and Energetic Additive

    Full Text Available ABSTRACT This study aimed to evaluate the effect of maltodextrin on performance of chicks passed through four fasting periods. A completely randomized design was performed, consisting of three treatments (1 - control, 2 - 4% saccharose; 3-4% maltodextrin 20 and four fasting periods after hatching (0, 12, 24 and 36 hours with four replications, totaling 1920 birds of both sexes. The studied variables were: water intake in the first 12 hours, average food intake, average body weight and real feed conversion. Data was submitted to analysis of variance and means were compared with each other by Dunnett test at 5% significance. The use of additives and imposing of different fasting periods did not influence the performance at 42 days old. Therefore, fasted chicks consumed significantly more water. At 7 days old, chicks fasted showed higher body weight and higher feed intake, however, the viability did not suffer any influence, the weight remained higher after 21days with the fast imposition and there was no influence on other variables. There was no effect of fasting on broiler chiken's performance at 42 days old. The inclusion of sucrose and maltodextrin to drinking water stimulated the chick´s water intake and provided higher viability after seven days old.

    We compared phenotypic characteristics of Lactococcus lactis subsp. lactis derived from different sources including the intestinal tract of marine fish and freshwater fish, and cheese starter culture. In the phylogenetic analysis based on partial 16S rRNA gene nucleotide sequences (1371 bp), freshwater fish-, marine fish- and cheese starter culture-derived strains were identical to that of L. lactis subsp. lactis previously reported. Fermentation profiles determined using the API 50 CH system were similar except for fermentation of several sugars including l-arabinose, mannitol, amygdalin, saccharose, trehalose, inulin and gluconate. The strains did have distinct levels of halotolerance: marine fish-derived strains > cheese starter-derived strain > freshwater fish-derived isolate. Lactococcus lactis subsp. lactis showed extensive diversity in phenotypic adaptation to various environments. The phenotypic properties of these strains suggested that L. lactis subsp. lactis strains from fish intestine have additional functions compared with the cheese starter-derived strain that has previously described. The unique phenotypic traits of the fish intestinal tract-derived L. lactis subsp. lactis might make them useful as a probiotics in aquaculture, and contribute to the development of functional foods and novel food additives, since the strains derived from fish intestines might have additional functions such as antibacterial activity.

    In this study, response surface methodology (RSM) based on Box-Behnken design (BBD) was employed to optimize the aqueous extraction of crude polysaccharides from Tunisian cyanobacteria Phormidium versicolor (NCC 466). The optimal extraction conditions with an extraction yield of 21.56±0.92% were as follows: extraction temperature at 81.05°C, extraction time of 3.99h, and water to raw material ratio of 21.52mLg -1 . Crude Phormidium versicolor polysaccharides (CPv-PS) are found to be a hetero-sulfated-anionic polysaccharides that contained carbohydrate (79.37±1.58%), protein (0.45±0.11%), uronic acids (4.37±0.19%) and sulfate (6.83±0.28%). The carbohydrate fraction was composed of arabinose, xylose, ribose, rhamnose, N-acetyl glucosamine, galactose, glucose, mannose, glucuronic acid and saccharose with corresponding mole percentages of 2.41, 14.58, 2.18, 6.23, 7.04, 28.21, 26.04, 3.02, 0.86 and 5.07, respectively. Evaluation of the antioxidant activity in vitro suggested that CPv-PS strongly scavenged radicals, prevented bleaching of β-carotene and reduced activity. The prepared porous PBI membranes had high proton conductivity and high content of acid doping at room temperature with 15.7 mol H 3 PO 4 per PBI repeat unit, much higher than pure PBI membrane at the same condition. Further, the performance and stability of the porous PBI membrane in high-temperature proton-exchange-membrane fuel cells was tested. It was found that the cell performance remained stable during 200 h stability test under a constant current discharge of 0.5 A cm −2 except for the last fifty hours.   3. Primary prevention of gestational diabetes mellitus and large-for-gestational-age newborns by lifestyle counseling: a cluster-randomized controlled trial.

    Full Text Available BACKGROUND: Our objective was to examine whether gestational diabetes mellitus (GDM or newborns' high birthweight can be prevented by lifestyle counseling in pregnant women at high risk of GDM. METHOD AND FINDINGS: We conducted a cluster-randomized trial, the NELLI study, in 14 municipalities in Finland, where 2,271 women were screened by oral glucose tolerance test (OGTT at 8-12 wk gestation. Euglycemic (n = 399 women with at least one GDM risk factor (body mass index [BMI] ≥ 25 kg/m(2, glucose intolerance or newborn's macrosomia (≥ 4,500 g in any earlier pregnancy, family history of diabetes, age ≥ 40 y were included. The intervention included individual intensified counseling on physical activity and diet and weight gain at five antenatal visits. Primary outcomes were incidence of GDM as assessed by OGTT (maternal outcome and newborns' birthweight adjusted for gestational age (neonatal outcome. Secondary outcomes were maternal weight gain and the need for insulin treatment during pregnancy. Adherence to the intervention was evaluated on the basis of changes in physical activity (weekly metabolic equivalent task (MET minutes and diet (intake of total fat, saturated and polyunsaturated fatty acids, saccharose, and fiber. Liophilization is the most propr form of these products for long-term maintenance, but this procedure is affected by unstabilizing agent that results in destruction of membrane, release of content and change in size and microbial contamination; hence for prevention of the adverse effects, the protective role of sugars such as: Maltose, Fructose, Glucose, Galactose, Saccharose and Lactose were studied. For this purpose, after preparation of liposomal suspention, categorized in for duplicate groups and concentrations of 25, 50, 100 percent of these sugars were added to those. On the basis of color and consistency of products, the best method of freezing is as application of absolute alcohol and then chilling in-70 oc for 16 h. In survey of protective substances concentrations 0.7, 1.4, 2.8, and 5.6 percent of the mentioned sugars were used for calculating of leakage percent (Upon on the ratio of optical density of treated samples to untreated. In this study, released maltose had highest effect. Level of fusion and aggregation had any significant difference between pre and post lyophilized samples in centrifugation with 10000 rpm.     Full Text Available Enzymatic synthesis of neoglucoconjugates by purified α-glucosidase from cockroach Periplaneta americana (Linnaeus. Cockroach Periplaneta americana (Linnaeus contains in his digestive tract an acid (pH 5,0 and mesophile (50°C α-glucosidase. This enzyme, purified to homogeneity, hydrolyses highly maltose, sucrose and p-nitrophenyl-α-Dglucopyranoside. The ability of α-glucosidase from cockroach purified to homogeneity to catalyse transglucosylation reactions was tested using maltose and saccharose as glucosyl donors and 2-phenylethanol and phenol as acceptors. The experimental conditions were optimized in relation to the time course of the reaction, pH and concentrations of glucosyl donors and acceptors. The yields in transglucosylation reactions at 37 °C were very high and could attain 67% and 48% with 2-phenylethanol and phenol respectively as glucosyl acceptors. This α-glucosidase hydrolyzed the products formed. It seems that the products formed were the phenylethyl-α-D-glucoside and phenyl-α-D-glucoside. These results suggest that α- glucosidase from cockroach is an exoglucosidase which catalyse the splitting of the α-glucosyl residue from the non reducing terminal of the substrate to liberate α-glucose. This comportment indicates that this enzyme operated by a mechanism involving the retention of the anomeric configuration. On the basis of this work, α-glucosidase from P. americana appears to be a valuable tool for the preparation of α-neoglucoconjugates.

  6. Physical activity, nutritional status, and dietary habits of students of a medical university.

    Nutritional habits and physical activity influence the health status of young adults. In this study, we engaged a group of 151 students from a medical university (90 female and 61 male subjects). Anthropometric parameters, dietary habits (a 7-day dietary recall), and level of physical activity were measured. It was found that the daily food rations of female (F) and male (M) students were improperly balanced and characterized by high amount of total and animal protein, phosphorus, vitamin A, cholesterol, and insufficient intake of carbohydrates, dietary fiber, and vitamin C. Female subjects consumed low amounts of total fat and calcium. The intake of protein (total and animal), fat, phosphorus, and cholesterol correlated with higher body mass. The physical activity of the students was found to be higher than the average physical activity of the European Union populations, and a general tendency of lowering level of physical activity with age was observed. Students with the highest level of physical activity (MET > 1500) consumed lower amounts of simple carbohydrates (galactose and saccharose) when compared to students with lower physical activity (MET habits should be modified to prevent the development of diet-dependent diseases. Various forms of physical activity should be proposed to students and they should be encouraged to participate in high level of physical activity so as to promote good health status.

    The microwave-assisted solvothermal synthesis of C-doped TiO2 and ZnO hybrid materials was performed. Saccharose, titanium isopropoxide and zinc acetate were used as organic and inorganic sources for the synthesis. The influence of temperature and reaction time on the textural and optoelectronic properties of the hybrid materials was verified. Carbon quantum-dots of TiO2 and ZnO nanostructured spheres were obtained in a second pot by controlled calcination steps of the precursor hybrid materials. A carefully characterization by adsorption-desorption N2 isotherms, XRD, XPS, SEM, UV-vis/DR and electro- and photo-electrochemistry properties of the carbon quantum-dots TiO2 and ZnO spheres was performed. The photoelectrochemical activity of TiO2-C and ZnO-C films proved to be dependent on the conditions of synthesis. It was found a red-shift in the energy band gap of the semiconductors with values of 3.02 eV and 3.13 eV for the TiO2-C and ZnO-C, respectively, clearly lower than those on bare semiconductors, which is associated with the C-doping effect. From the photo-electrochemistry characterization of C-doped TiO2 and ZnO films can be concluded that the present materials have potential applications as photoelectrodes for quantum-dots sensitized solar cells.

The treatments were: T1- control; T2- pyraclostrobin 0.2 g L-1; T3- boscalid 0.075 g L-1, T4- pyraclostrobin 0.2 g L-1 + boscalid 0.075 g L-1, T5- IBA + GA3 + kinetin 375 mg L-1, T6- GA4+7 + benzylaminopurine 100 mg L-1 and T7- plant extract 100 mg L-1. The carbohydrate accumulation curve was accomplished with 5 samples, at 20-day intervals between evaluations, the 1st evaluation being carried out at 30 days after transplantation, on the day of the first treatment application. At each sampling the plants were separated in stem, leaves and fruits, of which the contents of total soluble sugars, reducing sugars and saccharose were evaluated. The effects of the treatments on chlorophyll content and gas exchanges were also evaluated. The experimental design was completely randomized, with 4 repetitions and 6 destructive evaluations during the development, with 1 plant per experimental unit for each sampling.     Full Text Available Abstarct Shiitake mushroom [Lentinula edodes(Berk Singer/Pegler] has the second production class among the most important edible mushrooms. For a long time this mushroom has draw attention due to its unique flavor and taste and also therapeutic properties. ِDue to the importance of this mushroom in the world and also the effect of substrate on the production of this medicinal mushroom, a research were conducted in randomized complete block design with 6 replications on 4 different substrate formulations including: 1 sawdust, wheat bran, millet; 2 sawdust, wheat bran, molasses, chalk, calcium super phosphate; 3sawdust, wheat bran, tea waste; 4sawdust, saccharose, citric acid, chalk and calcium carbonate on L.edodes production for determining the suitable substarte on early fruiting, yield, biological effeciency, average mushroom numbers and weights per block. Results showed that substrate formulation 1 caused earlier fruiting than others (58 days from incubation time, this formulation also had the highest yield (112/3 g/log and biological effeciency (35/09% between the others formula. mashroom number was the most in substrate formulation 1 (16/67 n/log and the highest mushroom wieght was obtained from formulation 4 (14/33 g/per mushroom. there wasn’t any significant differences at (p≤0.05 between substrate formulation 1 and 2.   10. [Appreciation of selenium concentration in blood and tissues of male rat as a result of diet ingredients changes and its supplementation with chosen group B vitamins].

    The influence of diet ingredients and its supplementation with chosen B group vitamins on concentration of selenium in blood serum and tissues and activity of glutathione peroxidase in blood and liver of male rats was examined in the conducted experiment. The animals, aged 5 months, were divided into three groups and fed ad libitum with granulated mixes. Group I with basic mix containing among other things full grains, Group II with modified mix in which full grains were exchanged for wheat flour and in part with saccharose and Group III with modified mix supplemented in excess with vitamins B1, B2, B6 and PP. The experiment was conducted for six weeks during which the amount of consumed feeding stuff was calculated currently and once a week body mass of the animals was checked. When the experiment was finished the activity of GSH-Px was determined by spectrophotometric method in blood and liver whereas concentration of selenium in blood serum, muscles and in liver by fluorometric method. It was ascertained that the change of diet ingredients and its supplementation with chosen group B vitamins was in favour of lowering the amount of selenium in the examined tissues, and the decrease was not only the result of lower amount of the consumed element, but also of its increased usage, forced by the changes taking place under the influence of diet components and its supplementation.

    The aim of this work is to study the addition of oligosaccharides and glycolipids in lamellar phases of the cationic surfactant DDAB (di-dodecyl-dimethyl-ammonium bromide). Two steps have been followed: the determination of phases prisms and the thermodynamic interpretation in terms of molecular interactions. In order to characterize these systems, two new experimental small angle scattering methods have been perfected: 1) a neutron scattering contrast variation method which allows to study the adsorption of aqueous solution in bilayers and 2) a capillary concentration gradient method to establish directly and quantitatively the phases diagrams of ternary systems by X rays scattering. It has been pointed out that the oligosaccharides induce a depletion attractive force on the lamellar-lamellar equilibrium of the DDAB when they are excluded of the most concentrated phase. For the two studied glycolipids: 2-O lauroyl-saccharose and N-lauroyl N-nonyl lactitol, the ternary phase diagrams water-DDAB-glycolipid have been established in terms of temperature. Critical points at ambient temperature have been given. The osmotic pressure in concentrated lamellar phases has been measured. It has been shown that glycolipids increase the hydration repulsion at short distance and that the electrostatic repulsion is outstanding and unchanged at high distance if there is at less 1 mole percent of ionic surfactant. In a dilute solution, glycolipids decrease the maximum swelling of lamellar phases, with a competition between the lamellar phase and the micellae dilute phase for water. (O.M.)

  12. Investigations on the question of radiation-induced intracellular ion translocations in the liver of rats

    Radiation-induced ion translocations in cellular and/or subcellular units can be detected in the course of the examination of whole organs. The present paper attempts to study this problem in the liver of rats. The size and ion composition of the extracellular space and its subunits (interstice, intravascular space, biliary discharge system) must therefore be known in order to obtain purely cellular electrolyte values. They can be determined with the aid of dilution methods and by applying Donnan's law. For measuring radiation-induced ion translocations in nuclei and mitochondria, they must be isolated in an aqueous phase. Processing in 0.25 mol l/sup -1/ saccharose yields the best results with regard to ion losses, since the vitality of the membranes is largely preserved when using this method. During suspension, some of the nuclei and mitochondria obtained in sacharose solution show considerable ion translocations, depending on the composition of the medium. Irradiation up to 10 kR showed that relatively low doses (100 R) already cause further changes of the electrolyte concentrations of nuclei and mitochondria. In-vivo irradiation of the liver also caused definite potassium-sodium translocations in the nucleus and the mitochondria at doses as low as 100 R. These intracellular electrolyte translocations cannot be detected in examinations of the whole organ. All in all, it is shown that intracellular ion translocations can be found at relatively low radiation doses already. The present paper discusses the possible methods by which they can be detected.

  13. Decomposition of thermally unstable substances in film evaporators

    It is widely known that film evaporators are considered to permit really gentle evaporation of heat-sensitive substances. Nevertheless, decomposition of such substance still occurs to an extent depending upon the design and operation of the evaporator. In the following a distinction is made between evaporators with films not generated mechanically, namely the long tube evaporator (lTE) or climbing film evaporator, the falling film evaporator (FFE) and the multiple phase helical tube (MPT) or helical coil evaporators (TFE). Figs 1 and 2 illustrate the mode of operation. A theory of the decomposition of thermally unstable substances in these evaporators is briefly outlined and compared with measurements. Such a theory cannot be developed without any experimental checks; on the other hand, meausrements urgently need a theoretical basis if only to establish what actually has to be measured. All experiments are made with a system of readily adjustable decomposability, namely with aqueous solutions of saccharose; the thermal inversion of this compound can be controlled by addition of various amounts or concentrations of hydrochloric acid. In the absence of any catalysis by hydrochloric acid, the decomposition rates within in the temperature interval studied (60-130/sup 0/C) are so low that the experiments would take much too long and determination of the concentration differences (generally by polarimetric methods) would be very complicated. Such slight effects would also be very unfavourable for comparison with theory. (orig.)

  14. Effect of the oxygen balance on ignition and detonation properties of liquid explosive mixtures

    The objective is to compare the ignition and detonation properties of various liquid high explosives having negative up to positive oxygen balance (OB): nitromethane (OB < 0), saccharose and hydrogen peroxide based mixture (quasi nil OB), hydrogen peroxide with more than 90% purity (OB > 0). The decomposition kinetic rates and the equations of state (EOS) for the liquid mixtures and detonation products (DP) are the input data for a detonation model. EOS are theoretically determined using the Woolfolk et al. universal liquid polar shock law and thermochemical computations for DP. The decomposition kinetic rate laws are determined to reproduce the shock to detonation transition for the mixtures submitted to planar plate impacts. Such a model is not sufficient to compute open field explosions. The aerial overpressure is well reproduced in the first few microseconds, however, after it becomes worse at large expansion of the fireball and the impulse is underestimated. The problem of the DP EOS alone is that it takes only the detonation into account, the secondary combustion DP – air is not considered. To solve this problem a secondary combustion model has been developed to take the OB effect into account. The detonation model has been validated on planar plate impact experiments. The secondary combustion parameters were deduced from thermochemical computations. The whole model has been used to predict the effects of the oxygen balance on open air blast effects of spherical charges.

    Full Text Available Homemade salamis may have their food safety guaranteed by means of the addition of curing salt and starter cultures, without loosing their traditional manufacturing recipes brought to Brazil by the Italian immigration in the early 20th century. In this work, the influence of curing salt and of a starter culture, composed of Lactobacillus and Staphylococcus, over the sensory and microbiological characteristics of Italian type salamis, containing 3% lactose and 0.5% saccharose, was evaluated. The starter culture and the curing salt inhibited the development of Staphylococcus aureus and of coliforms, and the salamis added with curing salt presented better color attributes.Os salames produzidos artesanalmente podem ter sua segurança alimentar garantida pela adição de sal de cura e de culturas iniciadoras sem perda das receitas tradicionais trazidas pela imigração italiana do início do século vinte. Neste trabalho, a partir da produção de salames com 3% de lactose e 0,5% de sacarose, avaliou-se a influência do sal de cura e da cultura iniciadora, composta de Lactobacillus e Staphylococcus, sobre as características sensoriais e microbiológicas dos salames. A cultura iniciadora e o sal de cura inibiram o crescimento de Staphylococcus aureus e de coliformes totais, sendo que os salames adicionados de sal de cura apresentaram uma melhor coloração.

    Full Text Available Oncidium varicosum is a native Brazilian orchid popularly known as ‘Golden Shower´ because of its very ramified inflorescence and many yellow flowers. The carboydrate type and concentration are important in promoting plantlet development of in vitro orchids. The present study was carried out to asses the effect of different carbohydrate sources and concentrations on the in vitro growth of O. varicosum plantlets. Murashige e Skoog culture medium was used modified with half concentration of the macronutrients. The plantlets, derived from seeds that were already established in vitro and 0.8 + 0.2 cm in height, were inoculated in the culture media containing the following carbohydrate sources: saccharine, maltose and glucose, at concentrations of 0, 10, 20, 30, 60 and 90 g.L-1. The following variables were analyzed 8 months later: canopy height, number of roots, greatest root length, pseudobulb diameter and fresh weight. A completely randomized block experimental design was used with five replications per treatment. Analysis of variance and the Tukey test (5% were performed to compare the means. It was concluded that 60 g.L-1 saccharose was the best treatment for all the parameters assessed. The sugars 30 g.L-1 glucose and 60 g.L-1 maltose were also suitable, but presented lower pseudobulb diameter and lower fresh weight when compared to 60 g.L-1saccharose.Oncidium varicosum é uma orquídea nativa do Brasil conhecida popularmente como “chuva de ouro”, devido a sua inflorescência muito ramificada e com inúmeras flores amarelas. The highest conversion of jatropha oil to biodiesel at 75.2% was achieved using re-crystallized HT from its mixed oxides when reaction was carried out at 65 °C with a methanol:jatropha oil molar ratio of 12:1, a reaction time of 6 h and a catalyst loading of 4 wt.%.     The aim of this study was to compare the efficacy of treating osmotic diarrhea and dehydration in calves with hypertonic saline solution (HSS) IV, isotonic electrolyte solution (IES) PO, and a combination of these 2 solutions (HSS + IES). Eighteen male calves 8-30 days of age were used to evaluate the efficacy of 3 methods of fluid therapy after induction of osmotic diarrhea and dehydration. The diarrhea and dehydration were induced by administration of saccharose, spironolactone, and hydrochlorothiazide for 48 hours. The animals were randomly divided into 3 experimental groups: Group 1: 7.2% hypertonic saline solution-HSS (5 mL/kg IV); Group 2: oral isotonic electrolyte solution IES (60 mL/kg PO); or Group 3: HSS+IES. Clinical signs and laboratory finding observed 48 hours post-induction (Time 0) included diarrhea, dehydration, lethargy, and metabolic acidosis. Calves treated with HSS + IES experienced decreases in hematocrit, total protein concentration, albumin concentration, urea nitrogen concentration, and plasma volume as well as increases in blood pH, blood bicarbonate concentration, and central venous pressure between 1 and 3 hours post-treatment. These findings also were observed in animals treated with IES, however, at a slower rate than in the HSS + IES-treated animals. Animals treated with HSS continued to display signs of dehydration, lethargy, and metabolic acidosis 24 hours post-treatment. Treatment with a combination of HSS and IES produced rapid and sustainable correction of hypovolemia and metabolic acidosis in calves with noninfections diarrhea and dehydration.     Full Text Available Nineteen plant extracts obtained from plants from the Brazilian Amazon showed activity against planktonic Streptococcus mutans, an important bacterium involved in the first steps of biofilm formation and the subsequent initiation of several oral diseases. Objective: Our goal was to verify whether plant extracts that showed activity against planktonic S. mutans could prevent the organization of or even disrupt a single-species biofilm made by the same bacteria. Material and Methods: Plant extracts were tested on a single-bacteria biofilm prepared using the Zürich method. Each plant extract was tested at a concentration 5 times higher than its minimum inhibitory concentration (MIC. Discs of hydroxyapatite were submersed overnight in brain-heart infusion broth enriched with saccharose 5%, which provided sufficient time for biofilm formation. The discs were then submersed in extract solutions for one minute, three times per day, for two subsequent days. The discs were then washed with saline three times, at ten seconds each, after each treatment. Supports were allowed to remain in the enriched medium for one additional night. At the end of the process, the bacteria were removed from the discs by vortexing and were counted. Results: Only two of 19 plant extracts showed activity in the present assay: EB1779, obtained from Dioscorea altissima, and EB1673, obtained from Annona hypoglauca. Although the antibacterial activity of the plant extracts was first observed against planktonic S. mutans, influence over biofilm formation was not necessarily observed in the biofilm model. The present results motivate us to find new natural products to be used in dentistry.

The linden-added lemonade showed the highest value (597.9 mg/kg for ascorbic acid content, whereas the lemon verbena-added lemonade showed the lowest value (486.04 mg/kg. The amount of saccharose was changed by 11.07-11.58%, the glucose by 1.11-1.62% and the fructose between 11.07-11.58% in the beverages. K, Na, Mg and P values in the lemonades were determined in the range of 178.83-210.98 mg/kg, 33.75-39.13 mg/kg, 22.37-27.89 mg/kg and 7.22-10.04 mg/kg, respectively. The total phenolic content in the samples changed between 315.11-397.57 mg GAE/100 g. When the samples were analyzed for antioxidant activity, FRAP (17.13-26.79 µmol trolox/mL and ABTS (16.91-25.38 µmol trolox/mL methods gave higher results than the DPPH (14.88-17.72 µmol trolox/mL method. Linden-, ginger- and peppermint-added lemonades, which had been found to be rich in total phenolic compounds and ascorbic acid, showed higher antioxidant activity than the control sample. Overall, the most preferred lemonades were heather- and ginger-added beverages and the control sample. Mate-added lemonade was the least preferred.

    After three to seven days following to fractionated total body X-ray irradiation (TBI) (four expositions with doses of 0.3 to 5.0 cGy per fraction at intervals of 24 hours), a maximum 50 percent stimulation of the semiconservative DNA synthesis (SDS) of spleen cells was measured in vitro. This was not dependent of the fact if an acute high-dose (400 and/or 800 cGy) unique irradiation was applied after the fractionated TBI at the moment of stimulation. A significant increase of {sup 3}H-thymidine incorporation into the DNA of bone marrow and thymus cells was only found when doses of 1.25 cGy per fraction had been used. After fractionated TBI with doses of {ge}5 cGy per fraction, an increase of DNA synthesis resistant to hydroxyurea ('unprogrammed' DNA synthesis, UDS) was demonstrated in spleen cells. The UV-simulated UDS decreased proportionately. The sedimentation of thymus, spleen, and bone marrow nucleoids in a neutral saccharose gradient gave no evidence of an increased DNA repair capacity after fractionated TBI. Whereas the SDS stimulation by fractionated TBI with small doses can be explained by a modified proliferation behavior of exposed cells, the UDS behavior of spleen cells after considerably higher radiation doses suggests regenerative processes correlated with an increased number of cells resistant to hydroxyurea and cells presenting an UV repair deficiency. These findings can be considered to be a further proof of the assumed immune-stimulating effect of small radiation doses. (orig.).

    To assess the impact of prenatal exposure to Maillard reaction products (MRPs) -rich diet and postnatal Coca-Cola consumption on metabolic status of female rats. Diet rich in MRPs and consumption of saccharose/fructose sweetened soft drinks is presumed to impose increased risk of development of cardiometabolic afflictions, such as obesity or insulin resistance. At the first day of pregnancy, 9 female Wistar rats were randomized into two groups, pair-fed either with standard rat chow (MRP-) or MRPs-rich diet (MRP+). Offspring from each group of mothers was divided into two groups and given either water (Cola-) or Coca-Cola (Cola+) for drinking ad libitum for 18 days. Oral glucose tolerance test was performed, and circulating markers of inflammation, oxidative stress, glucose and lipid metabolism were assessed. MRP+ groups had higher weight gain, significantly so in the MRP+/Cola- vs MRP-/Cola-. Both prenatal and postnatal intervention increased carboxymethyllysine levels and semicarbazide-sensitive amine oxidase activity, both significantly higher in MRP+/Cola + than in MRP-/Cola-. Total antioxidant capacity was lower in MRP+ groups, with significant decrease in MRP+/Cola + vs MRP-/Cola+. Rats drinking Coca-Cola had higher insulin, homeostatic model assessment of insulin resistance, heart rate, advanced oxidation of protein products, triacylglycerols, and oxidative stress markers measured as thiobarbituric acid reactive substances compared to rats drinking water, with no visible effect of MRPs-rich diet. Metabolic status of rats was affected both by prenatal and postnatal dietary intervention. Our results suggest that combined effect of prenatal MRPs load and postnatal Coca-Cola drinking may play a role in development of metabolic disorders in later life.

  5. Production of Microbial Transglutaminase on Media Made from Sugar Cane Molasses and Glycerol

    Full Text Available Transglutaminase is an enzyme that catalyses an acyl transfer reaction between γ-carboxamide groups of glutaminyl residues and lysine residues in proteins. Due to this property, this enzyme is used for enhancing textural properties of protein-rich food. The transglutaminase used as food additive is obtained by microorganisms, mainly by Streptoverticillium ladakanum. On the other hand, sugar cane molasses is a viscous liquid rich in noncrystallized carbohydrates (saccharose, glucose and fructose. In this work, the feasibility of using sugar cane molasses as a carbon source for the production of microbial transglutaminase by Streptoverticillium ladakanum NRRL 3191 has been studied. Carbon sources including sugar cane molasses (60 g of total sugars per L, glycerol (60 g/L and their mixture in a ratio of 1:1 (30 g/L of each were evaluated. Time course of microbial growth, transglutaminase activity and carbon source consumption were determined every 24 h during 120 h of fermentations at three agitation speeds (200, 300 or 400 rpm. The results showed that with the increase in agitation speed, the biomass concentration increased up to 8.39 g/L in the medium containing sugar cane molasses alone or the mixture of molasses and glycerol. The highest transglutaminase activity was obtained at 400 rpm in the medium containing a mixture of molasses and glycerol, reaching 0.460 U/mL, while in the medium containing sugar cane molasses alone, the activity was 0.240 U/mL, and using glycerol alone it was 0.250 U/mL. These results show that sugar cane molasses is a suitable medium for transglutaminase production when it is combined with glycerol.

  6. Sweet sorghum: A new raw material source for the sugar industry

    The primary uses of sweet sorghum have been as a syrup for human consumption and as a livestock feed. More recently, interest in sweet sorghum has risen because of its potential use as a source of sugar and/or as a raw material for the production of energy and for different biotechnological processes. However, before sweet sorghums may be utilized as a source for both sugar and energy, adequate and adapted genotypes must be identified. The objective of this study is to measure the agronomic performance of different lines and cultivars of sweet sorghum for cold tolerance yield of biomasse, yield of sugar and sugar quality. Of the more than 1000 entries from 20 different world-wide origins, 614 accessions have been cultivated for evaluation purposes in the last 2 years. Among the tested material 18 genotypes exhibited more than 90 tons biomass (FM) per hectare in 1985 and 32 types in 1986. The number of accessions which produced more than 90 th FM/ha in both years was 10, one type produced more than 11 tons FM/ha. The biomasse production of one accession achieved 169 t FM/ha in 1986. The highest sugar content in FM mounted to 9,2% in 1985 and 11,1% in 1986. Five genotypes produced more than 10 tons sugar per hectare, the highest sugar yield was 12 t/ha. In average of the high sugar yielding accessions, more than 50% of the sugar consists of saccharose, 28% of glucose and 19% of fructose. The corresponding quantity of alcohol which can be produced amounts to 7 000 l/ha. (orig.)

  7. Preparation and investigation of diamond-like carbon stripper foils by filtered cathodic vacuum arc

    Thin diamond-like carbon (DLC) stripper foils ∼5μg/cm 2 in thickness were produced and evaluated as heavy-ion strippers for the Beijing HI-13 Tandem Accelerator. The DLC layers ∼4μg/cm 2 in thickness were produced by the filtered cathodic vacuum arc technology onto glass slides coated with betaine–saccharose as releasing agent, which were previously covered with evaporated carbon layers ∼1μg/cm 2 in thickness by the controlled ac arc-discharge method. Irradiation lifetimes of the DLC stripper foils were tested using the heavy-ion beams at the terminal of the Beijing HI-13 Tandem Accelerator, and compared with those of the standard carbon stripper foils made by the combined dc and ac arc-discharge method. The measurements indicate that the DLC stripper foils outlast the standard combined dc and ac arc-discharge carbon stripper foils by a factor of at least 13 and 4 for the 197 Au − (∼9MeV, ∼1μA) and 63 Cu − (∼9MeV, ∼1μA) ion beams, respectively. The structure and properties of the DLC foils deposited onto silicon substrates by the filtered cathodic vacuum arc technology were also evaluated and analyzed by scanning electron microscopy, Raman spectroscopy and X-ray photoelectron spectroscopy. The scanning electron microscopy images show that the DLC foils contain hardly droplets through the double 90° filters. The X-ray photoelectron spectrum indicates that sp 3 bonds of the DLC foils exceed 70%. The integral intensity ratio of the D peak to the G peak (I D /I G ) measured by the Raman spectroscopy is 0.78

    During the last years the Brazilian floriculture expanded and it is expected to achieve a prominent role in near future. The national territory has many favorable regions, with adequate climate for the development of commercially important cultures. Flower exportation is not expressive yet, but the country has conditions to become a great exotic tropical plants exporter. On the other hand, developed countries, as USA and Japan, have a rigorous phyto sanitary inspection to prevent the introduction of new plagues and diseases through fresh products. Ionizing radiation is considered a reliable disinfestation method to control numerous fruit and flower plagues. The aim of of this work was to verify the tolerance of some Compositae family flowers to ionizing radiation and the effects to sucrose action in protecting the flowers against the gamma-radiation induced damages. This was done by measuring biochemical and physiological parameters as a function of time after irradiation. The chrysanthemum flowers were sensitive to gamma radiation, but when the inflorescences were supplied after irradiation with preservative solution containing 2% sucrose, protection against the damaging effects of radiation was achieved. The dose of 750 Gy, considered appropriate for disinfestation purpose, did not modify the protein and lipid levels, nor plasma, tonoplast and mitochondria membrane ATPase or mitochondria cytochrome-c oxidase activities. The ethylene and carbonic gas rate production increased soon after the irradiation, but decreased one day later. The decrease of the microsomal membrane fluidity and the increase of the chrysanthemum were the most sensitive parameters to measure the irradiation treatment changes. The sucrose supply was able to maintain the irradiated flowers membrane fluidity level close to the unirradiated control. Rhodante manglesii Lindl and Helichrysum bracteatum Andr. were tolerant up to 1 KGy, thus being adequate to be disinfected by gamma radiation. (author)

    The influence of diet content and its supplementation with chosen group of B vitamins on the intake of feeding stuff increase, changes of body mass, accumulation of fat tissue, lipids and lipoproteins concentration in the blood of female rats were under research. The animals, aged 5 months, were divided into three groups (8 persons each) and fed ad libitum with granulated Labofeed B type mix. Group I with the basic mix containing among other things whole grain, Group II with a modified mix, where whole grain was replaced by wheat flour and saccharose and Group III with modified mix supplemented in excess with chosen vitamins of B group. This experiment took 6 weeks during which the amount of consumed feed was currently evaluated, and the body mass was controlled weekly. After finishing the experiment in the obtained serum the concentration of triacylglycerols, complete cholesterol with enzyme method and the content of cholesterol fractions with electrophoretic separation method were determined. Analysis of fat content in muscles and livers was conducted and the amount of round the bodily organ fat was determined. It was ascertained that change of the content of the feed and its supplementation with the chosen B group vitamins did not influence in a substantial way its intake and the increase of body mass, however it had influenced substantially, in animals fed with the modified feed the accumulation of round the organ fat and in supplemented the intramuscular fat. Analysis of the results enabled the ascertainment that the diet supplementation with chosen ingredients of the B group vitamins corrects the negative effect of accumulation of the visceral fat tissue as a result of the change of its contents, caused substantial increase in the concentration of triacylglycerols, complete cholesterol and its fractions VLDL- and LDL- with simultaneous decrease of the concentration of cholesterol HDL- fractions.

    Full text: Besides ''in vivo'' methods, the Radiation Genetics Section of CENA/USP is also using ''in vitro'' methods for mutation breeding to obtain resistance to Panama disease caused by Fusarium oxysporum f.sp. cubense in the banana cultivar ''Maca''. A protocol has been established for the ''in vitro'' development of shoot tips, obtained from plants in the field or already cultivated under {sup i}n vitro'' conditions. For both cases, only one culture medium was used during all steps of ''in vitro'' cultivation. New buds were formed and these buds grew and developed to form roots. The medium was composed of macro and micro nutrients, with added Morel vitamines, BAP (5 mg/l), saccharose (30 g/l) and agar (6.5 g/l), at pH 5.7. Cultures were allowed to grow in a controlled environment at 27 deg. C and 16 h illumination. Shoot tips which originated from ''in vitro'' plantlets, were cut longitudinally down the middle. This was done to avoid a tendency of regeneration of the original tissue instead of the formation of new lateral buds. To resolve the chimerism resulting when mutagenic treatment is applied to shoot tips, there is a need of vegetative propagation of new lateral buds. Selection can then be done at M{sub 1}V{sub 4} generation. Once the protocol was established, the gamma ray sensitivity was determined. The dose that produced a 50% decrease in the number of new lateral buds was around 40 Gy and this dose will be utilised. The methodology was completed by soil inoculation with Fusarium of young plants 15 cm in height, obtained from ''in vitro'' cultures. After 3 weeks all inoculated plants showed symptoms of wilt, demonstrating the possibility of screening. The method is now being utilised on a large scale in an attempt to induce a resistant mutant.   11. Severe selective magnesium malabsorption: tests of tolerance of oral magnesium supplements.

    Since his birth, we have been monitoring a 12-year-old boy suffering from selective severe magnesium malabsorption. Our essential problem is to prepare a form of galena with acceptable taste, tolerated by the digestive tract and well absorbed; also, the carrier compound must not cause short- or long-term side effects. An additional factor is the steadily increasing need for magnesium from 1 mmol/kg.d at 1 year to 14 mmol/kg.d at present age (345 mg/kg.d). The galena forms currently on sale were, with the exception of lactate and pyrollidone carboxylate, immediately rejected since they contain insufficient Mg2+. Following short trials resulting in diarrhoea, the other two preparations were also rejected. We then constituted - and also abandoned - our own galena compounds: aspartate (bitterness), aspartate + glycerophosphate (GLP) (bitterness), glutamate + GLP ('Chinese restaurant syndrome' and fear of the long term toxic effect of the glutamate), gluconate (excessive volume: 11/1 proportion with Mg2+). A recent test featuring GLP of Mg 40 g + cocoa butter 40 g + cocoa 10 g, brought about vomiting and diarrhoea, and was not adequately absorbed. The best tolerated formula is: Mg GLP 21.33 g; saccharose 6 g; aspartam 1 g; gelatin 0.5 g; citric acid, conserving agent, fruity aroma; water: qs 100 g. Such composition yields a caramel cream absorbed in five small portions, at a daily quantity of 375 g (80 g GLP Mg, 10 g Mg2+). Vitamin B6, which promotes intestinal absorption of magnesium, must be given separately in tablet form at a dose of 1 g/d, since it causes nausea if it is included in the Mg preparation.(ABSTRACT TRUNCATED AT 250 WORDS)

    Full Text Available O ciclamato é usado como adoçante artificial não calórico em diversos alimentos e bebidas, sendo 30 vezes mais doce que a sacarose sem o sabor amargo da sacarina. Aparece na composição dos produtos como ciclamato de sódio, ciclamato de cálcio e ácido ciclâmico. O ciclamato e a ciclohexilamina, seu principal metabólito, atravessam a barreira placentária em humanos e desse modo podem ser expostos ao feto. O rim de ratos pode ser afetado por elevadas doses de ciclamato de sódio. Estudos sobre efeitos do ciclamato de sódio na espécie humana são necessários, pois, além de poder substituir a sacarose - prejudicial em casos de diabetes ou quando o controle e a redução do peso corporal são essenciais para a saúde dos pacientes - não propicia desenvolvimento de cárie dentária.Cyclamate is used as an artificial non-caloric sweetener in a variety of foods and beverages, being 30 times as sweet as sugar without the bitter after-taste of saccharin. It is present in the formula of products such as sodium and calcium cyclamates and cyclamic acid. Cyclamate and cyclohexylamine, its principal metabolite, can cross the human placenta exposing the fetus. It has been demonstrated in rats that the kidney may be adversely affected by high doses of cyclamate. Studies on the effects of sodium cyclamate in the human species are necessary, because in addition to replacing saccharose - harmful in individuals with diabetes or patients in whom weight reduction and control are essential for health - it does not cause dental caries.

  14. Effect of yeast storage temperature and flour composition on fermentative activities of baker's yeast

    Full Text Available Baker's yeast is a set of living cells of Saccharomyces cerevisiae. It contains around 70-72% of water, 42-45% of proteins, around 40% of carbohydrates, around 7.5% of lipids (based on dry matter, and vitamin B-complex. On the basis of yeast cell analysis it can be concluded that yeast is a complex biological system which changes in time. The intensity of the changes depends on temperature. Yeast sample was stored at 4°C i 24°C for 12 days. During storage at 4°C, the content of total carbohydrates decreased from 48.81% to 37.50% (dry matter, whereas carbohydrate loss ranged from 40.81% to 29.28% at 24°C. The content of trehalose was 12.33% in the yeast sample stored at 4°C and 0.24% at 24°C. Loss of fermentative activity was 81.76% in the sample stored at 24°C for 12 days. The composition of five samples of 1st category flour was investigated. It was found that flours containing more reducing sugars and maltose enable higher fermentation activities. The flours with higher ash content (in the range 0.5-0.94% had higher contents of phytic acid. Higher ash and phytic contents in flour increased the yeast fermentative efficiency. In bakery industry, a range of ingredients has been applied to improve the product's quality such as surface active substances (emulsifiers, enzymes, sugars and fats. In the paper, the effect of some ingredients added to dough (margarine, saccharose, sodium chloride and malted barley on the yeast fermentative activity was studied. The mentioned ingredients were added to dough at different doses: 0.5, 1.0, 1.5 and 2.0%, flour basis. It was found that the investigated ingredients affected the fermentative activity of yeast and improved the bread quality.

    Full Text Available A comparative study has been done about the use of three methods: Microscopic, Dissolution of Phases and X-Ray Diffraction, for the determination of the mineralogical composition of clinker, XRD appears (just as it is shown in this paper as a complicated method which does not guarantee a good determination of C3A. The consecutive dissolution of phases whith methanol and salicylic acid and saccharose is a simple, cheap and reliable alternative method. The dissolution of phases with salicylic acid is a good method to determine the whole of silicates and liquid phase. Optic microscopy (reflected light is a method of determining bi and tricalcium silicates but the results so given are unreliable. Varations which could improve these methods are indicated.

    Se hace un estudio comparativo del empleo de tres métodos, microscópico, disolución de fases y difracción de rayos X, en la determinación de la composición mineralógica del clínker. Se revela DRX (tal y como se desarrolla en este trabajo como un método poco fiable en la determinación del C3A y además es complicada. La disolución sucesiva de fases con metanol-salicílico y sacarosa es una alternativa sencilla, económica y fiable. La disolución de fases con salicílico es un buen método en la determinación total de silicatos y fase líquida. La microscopía óptica de luz reflejada es un método para la determinación de los silicatos bi y tricálcicos, aunque da resultados poco fiables. Se indican variaciones que mejoran los métodos.

  16. Effects of dietary counselling on food habits and dietary intake of Finnish pregnant women at increased risk for gestational diabetes - a secondary analysis of a cluster-randomized controlled trial.

    The incidence of gestational diabetes mellitus (GDM) is increasing and GDM might be prevented by improving diet. Few interventions have assessed the effects of dietary counselling on dietary intake of pregnant women. This study examined the effects of dietary counselling on food habits and dietary intake of Finnish pregnant women as secondary outcomes of a trial primarily aiming at preventing GDM. A cluster-randomized controlled trial was conducted in 14 municipalities in Finland, including 399 pregnant women at increased risk for developing GDM. The intervention consisted of dietary counselling focusing on dietary fat, fibre and saccharose intake at four routine maternity clinic visits. Usual counselling practices were continued in the usual care municipalities. A validated 181-item food frequency questionnaire was used to assess changes in diet from baseline to 26-28 and 36-37 weeks gestation. The data were analysed using multilevel mixed-effects linear regression models. By 36-37 weeks gestation, the intervention had beneficial effects on total intake of vegetables, fruits and berries (coefficient for between-group difference in change 61.6 g day(-1), 95% confidence interval 25.7-97.6), the proportions of high-fibre bread of all bread (7.2% units, 2.5-11.9), low-fat cheeses of all cheeses (10.7% units, 2.6-18.9) and vegetable fats of all dietary fats (6.1% -units, 2.0-10.3), and the intake of saturated fatty acids (-0.67 energy-%-units, -1.16 to -0.19), polyunsaturated fatty acids (0.38 energy-%-units, 0.18-0.58), linoleic acid (764 mg day(-1), 173-1354) and fibre (2.07 g day(-1) , 0.39-3.75). The intervention improved diet towards the recommendations in pregnant women at increased risk for GDM suggesting the counselling methods could be implemented in maternity care. © 2012 John Wiley & Sons Ltd.

    Aseptically collected sandstone and soil samples from the antarctic Dry Valleys were inoculated into oligotrophic media and incubated under low light intensities. A total of 41 Gram-negative isolates were obtained with reddish colonies spreading on agar. A sandstone isolate and four soil strains were characterized further. They were nearly identical in morphological, physiological, biochemical and chemotaxonomic properties. They produced large amounts of extracellular polymer and utilized for growth: glucose, saccharose, mannitol, sorbitol, L-aspartate, malate and acetate, but not D-ribose, adonitol, DL-alanine, glutamate, glycolate, lactate or succinate. All strains hydrolyzed gelatin, starch, casein, xylan, Tweens 80 or 60 and dead or living yeast cells, but not cellulose or pectin. Nitrate was not reduced, ethanol was not oxidized and acid was not produced from maltose, mannitol or dulcitol. Ammonia was not produced from peptone. They were strictly aerobic. Major fatty acids were n 16:1 d 9, n 16:1 d 11, n 17:1 d 11, and i 15:0. The strains contained the quinone MK-7 and phosphatidylethanolamine as the main phospholipid. The base ratio ranged from 55 to 61 mol% G+C. A 16S rRNA sequence analysis of strains AA-688 and AA-718 showed these to be identical and to represent a special phylogenetic group within the Cytophaga/Flavobacterium/Bacteroides major line of descent. Three soil strains labeled "Taxeobacter" Txc1, Txg1, and Txo1 (Reichenbach, 1992) belonged to the same group but had lower sequence similarities (<95%). Some of their characteristics were different from those of the antarctic strains: the utilization of C-compounds, hydrolysis of polymers, temperature tolerances, major fatty acids and base ratios. Txc1 and Txg1 may later have to be considered as members of this group, possibly on the species level, while Txo1 could represent a different related genus.   19. Evaluation of the fermentation of high gravity thick sugar beet juice worts for efficient bioethanol production

    Background Sugar beet and intermediates of sugar beet processing are considered to be very attractive feedstock for ethanol production due to their content of fermentable sugars. In particular, the processing of the intermediates into ethanol is considerably facilitated because it does not require pretreatment or enzymatic treatment in contrast to production from starch raw materials. Moreover, the advantage of thick juice is high solid substance and saccharose content which eliminates problems with the storability of this feedstock. Results The objective of this study were to investigate bioethanol production from thick juice worts and the effects of their concentration, the type of mineral supplement, as well as the dose of yeast inoculum on fermentation dynamics and ethanol yield. The obtained results show that to ensure efficient ethanolic fermentation of high gravity thick juice worts, one needs to use a yeast strain with high ethanol tolerance and a large amount of inoculum. The highest ethanol yield (94.9 ± 2.8% of the theoretical yield) and sugars intake of 96.5 ± 2.9% were obtained after the fermentation of wort with an extract content of 250 g/kg supplemented with diammonium hydrogen phosphate (0.3 g/L of wort) and inoculated with 2 g of Ethanol Red dry yeast per L of wort. An increase in extract content in the fermentation medium from 250 g/L to 280 g/kg resulted in decreased efficiency of the process. Also the distillates originating from worts with an extract content of 250 g/kg were characterized by lower acetaldehyde concentration than those obtained from worts with an extract content of 280 g/kg. Conclusions Under the favorable conditions determined in our experiments, 38.9 ± 1.2 L of 100% (v/v) ethyl alcohol can be produced from 100 kg of thick juice.     Full Text Available Rationale: Most cases of listeriosis are caused by the pathogenic Listeria monocytogenes. Some cases of isolation of L.  innocua with pathogenicity factors from foods have been published, as well as on the cases of the disease in humans caused by this species. Aim: To assess biological properties including potential pathogenicity of L.  innocua, isolated from food and environmental objects. Materials and methods: We performed microbiological study of L. innocua cultures isolated from foods (n = 35 and environmental objects (n = 15 on the territory of Primorye Territory (Russian Federation, as well as assessment of their sensitivity to antibiotics. Results: The studied L.  innocua cultures showed stable phenotypic features of their biological properties, such as morphology, typical colony growth on the medium with characteristic odor of fermented milk, blue or blue-green luminescence induced by inclined light, presence of catalase activity and absence of the oxidase activity. Only 38 ± 6.9% of L.  innocua demonstrated movements at T 22 °С. L.  innocua cultures did not ferment mannitol (100% of cultures; they degraded ramnose to its acid without gas (70 ± 6.5% and degraded xylose (42.8 ± 7%. Listeria isolated from vegetables and environmental objects could ferment ramnose (92.8 ± 7.2% of the studied cultures and xylose (28.5 ± 12.5% more frequently than L. innocua isolated from meat and fish foods. L.  innocua demonstrated variable biochemical activities towards mannose (92 ± 3.8%, saccharose (85.7 ± 7.8% and melesitose (76.2 ± 9.5%.     To determine whether (1) rapid consumption of 1 L of apple juice increases blood antioxidant capacity, measured as ferric-reducing ability of plasma (FRAP) and serum 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activity, and (2) apple polyphenols or fructose-induced elevation of plasma uric acid contributes to post-juice increase of blood antioxidant activity. The study involved 12 (mean age 32 ± 5 years, mean body weight 73 ± 7 kg) healthy nonsmoking subjects. Tested subjects consumed 1 L of clear apple juice and then FRAP; serum DPPH-scavenging activity, serum uric acid, and total plasma phenolics and quercetin levels were measured just before juice ingestion and 1, 2.5, and 4 hours after ingestion. This was repeated 3 times with 4-day intervals, but volunteers drank either 1 L of clear apple juice without polyphenols (placebo), or 1 L of cloudy apple juice (positive control), or 1 L of water (negative control) at the time. All juices had similar content of sugars (i.e., saccharose, glucose, and fructose) and precisely defined composition of phenolics and antioxidant activity. Consumption of all 3 juices transiently increased FRAP and serum DPPH-scavenging activity, with peak values at 1 hour post-juice ingestion. This was paralleled by the rise of serum uric acid, but no significant changes in plasma total phenolics and quercetin levels were observed after all dietary interventions. At the same time, no substantial differences were found between juices (especially between clear apple juice and clear apple juice without polyphenols) concerning the measured variables. A strong significant correlation was noted instead between serum uric acid and plasma antioxidant activity at all analyzed time points, before and after juice ingestion. Plasma total phenolics and quercetin levels were not associated with FRAP and serum DPPH radical-scavenging activity. Twenty three isolates from genera Arthrobotrys, Aspergillus, Coniothyrium, Cunninghamella, Cylindrocarpon, Fusarium, Monacrosporium, Mortierella, Paecilomyces, Penicillium, Sclerotinia, Trichoderma and Verticillium, presented toxic activity against wheat coleoptile, though only isolates of Cylindrocarpon magnusianum (Sacc. Wollenw., Fusarium moniliforme Shelden, Mortierella sp., Paecilomyces lilacinus (Thom. Samson and Penicillium sp., presented phytotoxic effects in the lettuce seeds assay. Only one isolate of F. moniliforme was inactive in both assays, what suggests that this fungus is not able to produce phytotoxins.

  5. Quantitative assessment of nutrition in patients with polycystic ovary syndrome (PCOS)

    PCOS (polycystic ovary syndrome) is called a pathology of the XX century and affects at least 10-15% women of childbearing age. The therapy involves pharmacotherapy of hormonal imbalance, as well as the change of lifestyle, including the diet. Performing the quantitative assessment of components of diets of women with PCOS, comparing the results with current dietary standards for Polish people and defining dietary requirements for the patients. The study was performed on 54 women of childbearing age (average age 26.03± 5.52) with PCOS syndrome diagnosed according to on the Rotterdam criteria. Anthropometric measurements of the patients were made and BMI and WHR calculated. Quantitative assessment of women’s diets was performed based on the analysis of 3-day food diaries and food records taken from the previous 24h with the interview method. The data were introduced to a dietary software DIETA 5.0, calculating the average intake of the energy, nutrients, vitamins, minerals, cholesterol and dietary fibre. The obtained results were compared to Polish dietary guidelines. Examined group was characterized by increased waist circumference (98.71± 13.6 cm) and an average WHR was 0.92± 0.08. An increased average value of BMI was also shown (28.91± 5.54 kg/m2). The patients consumed, on average, 1952.5±472.7 kcal daily, and the risk of insufficient intake of protein was determined in 36.7% of examined women. The highest risk of deficiency in minerals in women with PCOS was related to calcium (634 mg), potassium (3493 mg) and magnesium (250.1 mg), whereas with reference to vitamins deficiency as much as 70% of tested women were at risk of insufficient intake of folic acid, 36.7% of them - vitamin C, and 26.7% - vitamin B12.     Contemporarily, food production without food additives is very rare. Increasingly often, however, scientific works report on adverse effects of specified, single food additives on the body. Data is, in turn, lacking on the synergistic effect of a mixture of different food additives on body functions and its main metabolic pathways. The objective of this study, an animal model, was to evaluate if and in what way the compound of chosen and most frequently used and consumed food additives, along with the change of diet composition to processed, purified, influence the selected markers of protein metabolism. The animals were divided into four groups, which were fed with compound of feed pellets: group I and II with basic compound, group III and IV with modified compound in which part of the full grain was replaced by isocalorie wheat flour type 500 and saccharose. Animals from groups I and III received tap water, which was standing for some time, to drink. Animals from groups II and IV received solution of chosen additives to food and next they were given water to drink. The amount of given food additives was evaluated by taking into consideration their consumption by people recalculated to 1 kg of their body mass. The experiment spanned for 7 weeks. It was ascertained that the applied additives caused significant changes in total protein concentration and its fractions: albumin, alpha1-globulin, alpha2-globulin, beta-globulin and gamma-globulin in the blood serum of the animals under research, which can indicate and contribute to disclosure of creation of undesirable food reaction, especially when recommended levels of consumption of those additives are being exceeded.     This study was aimed at investigating the effects of low-dose gamma irradiation on shelf-life extension and phyto sanitary safety of minimally processed red beet with basis on physicochemical; microbiological; chemical and sensory analyses. The samples (Beta vulgaris ssp. vulgaris L.), Early Wonder cultivar, were cultivated in the experimental area of the Horticulture Sector of the Departamento de Fitotecnia of the Instituto de Agronomia, Universidade Federal Rural do Rio de Janeiro, Seropedica, RJ, as part of two experiments performed during the second semester of 2005. In each experiment, 1200 plants (40 per linear meter of terrain) were sowed. Physicochemical analyses (fresh mass; length; average diameter and total soluble solids) of the edible part of the red beets (their tuberous roots) were periodically carried out in order to assess the most appropriate time for harvest by monitoring the development of the plants. Those times were found to be 104 and 73 days after transplanting, respectively. The harvested edible part of the roots were minimally processed and separated in two groups: (1) gamma irradiated (with doses of 0.5; 1.0 and 1.5 kGy) and (2) non-irradiated (control). All samples were stored at 8 degree C. Microbiological analyses were performed during the storage period (22 and 21 days, for experiments 1 and 2, respectively) in order to evaluate the phyto sanitary quality of the samples (Salmonella sp.; coliforms and total count of mesophilic aerobic and lactic acid bacteria). The samples irradiated with 1.0 and 1.5 kGy were found to remain appropriate for consumption for 21 days, as compared to only 7 days for the control.     Extraction of U from technological solution excludes a loss of valuable metal and pollution of the environment by heavy and radioactive element. For the purpose, nontraditional method of extraction using capacity of the microorganism sells to accumulate metal ions (cation) has been applied. Aim of this work is to study of U sorption from technological solutions by different microorganisms and to search more effective bio sorbents among microorganisms- actinomycetes, fungal culture (Acremonium, Aspergillus) by using INAA. INAA is more available and cheap method for us also it has high enough sensitivity and allows to avoid exposing the bacteria to radioactive metals. Atomic absorptive analyzer is more suitable for the determination of impurities in solutions, but it is expensive and not available for us. Actinomycetes were reproduced on starch-ammoniac agar medium: NaNO 3 - 1 g, actinomycetes MgSO 4 - 1 g, K 2 HPO 4 -1 g, starch -10 g, agar -20 g, distilled water - 1000 ml, pH = 6.8 - 7.0 on horizontal shaker (180 rpm) at temperature 28-30 degree for 1-3 days. Fungi were reproduced in liquid medium: NaNO 3 - 2 g, K 2 HPO 4 -1 g, KCl - 0.5 g, MgSO 4 - 0.5 g, FeSO 4 - 0.01 g, saccharose -20.0 g, tap water -1000 ml, pH = 7.0-7.2 on horizontal shaker (180 rpm) at temperature 28-30 degree for 1-3 days. After reproduction strains actinomycetes and fungi were separated by centrifuging (6000 rpm) and washed many times by distilled water. Strains of actinomycetes and fungi (1 g of each) were placed in 1 litre of the U technological solution (70.0 mg/l) and mixed on horizontal shaker (180 rpm). Interaction between strains and solution lasted 5, 10, 15, 30 and 60 min. After interaction the content of U not sorbed by strains (residual content) was measured from the over-sedimentary solution (decantate). 1 ml solution (decantate) was dropped on strip of ash free filter paper.     Full Text Available The effectiveness of a trap for triatomines baited with yeast cultures has been previously demonstrated for Triatoma infestans in laboratory assays. We report here results from laboratory assays testing yeast traps for Triatoma sordida, Triatoma brasiliensis, Triatoma pseudomaculata, and Panstrongylus megistus. All assays were conducted in an open experimental arena 100 cm X 100 cm, with two traps placed at opposite sides of the arena. One of the traps contained a yeast culture, and the other trap contained a saccharose solution as a control. Two series of experiments were done, one without a refuge for the insects and one with a refuge. The results obtained clearly demonstrated that the yeast-baited traps were effective in the laboratory in capturing both T. sordida and P. megistus. For T. sordida, yeast-baited traps captured significantly more bugs than did the control traps (t test P value = 0.03. For P. megistus, when a refuge was provided during the assay, yeast-baited traps also captured significantly more bugs than did the control traps (t test P value = 0.006. In the experiments with T. brasiliensis and T. pseudomaculata, both traps captured some insects, but the yeast traps captured many fewer bugs than was true with the T. sordida and P. megistus bugs. These results indicate that, in the laboratory, yeast traps can capture considerable numbers of T. sordida and P. megistus in one night.     Full Text Available Resumen: Se evaluó la viabilidad del polen de berenjena (Solanum melongena L. mediante el método de germinación in vitro. Botones florales de la variedad Lila criolla con características de pre-antesis fueron colectados de las 07:00 a las 08:00 horas. Los granos de polen fueron extraídos con un vibrador eléctrico y rehidratados en cámara húmeda durante dos horas a temperatura de 25 ºC. Posteriormente, para la germinación en condiciones in vitro, los granos fueron dispersados, utilizando un pincel, en un medio de cultivo constituido por 100 g de sacarosa (C12H22O11, 500 mg de nitrato de calcio [Ca (NO32 4H2O], 120 mg de sulfato de magnesio (MgSO4, 100 mg de nitrato de potasio (KNO3 y 120 mg de ácido bórico (H3BO3 disueltos en 1.000 mL de agua destilada. Seguidamente, se agregaron 10 g de agar y el pH fue ajustado a 6,0. El polen fresco fue incubado durante ocho horas con lecturas cada dos horas. Los resultados indican que el método es confiable para cuantificar la viabilidad de granos de polen, ya que después de ocho horas de incubación se logró un 79% de germinación, 0,50 mm de longitud del tubo polínico y 0,0532 mm de diámetro del mismo. Por lo tanto, el uso de polen con ocho horas de almacenamiento es favorable para la producción de semilla híbrida a través de la hibridación artificial, por haber registrado un aumento de germinación de 0,4942% con efecto cuadrático, por cada hora de incubación.Abstract: We assessed the viability of pollen of eggplant (Solanum melongena L. using in vitro germination method.     the presence of Giardia cysts; 447 faecal samples were taken from 232 persons coming from 5 cities situated in Wielkopolska, and 1736 faecal samples were obtained from 123 animal species, including: 266 faecal samples from 113 species of animals kept in the Zoological Garden in Poznań, 1286 samples from 4 species of breeding animals, 118 samples from dogs, and 66 samples from 5 species of wild animals. Faecal samples were taken from animals coming from 25 places in Wielkopolska. Moreover, seven isolates of G. intestinalis were used in the studies, which were obtained from various species of hosts and kept in an axenic in vitro culture. Microscopic, molecular and bio-informative methods were used in the studies. From each faecal sample fresh smears were made in a 0.6% solution of physiological salt and in Lugol's solution, as well as a permanent smear stained with trichrome was made. Moreover, the following molecular techniques were implemented in the studies: DNA extraction and purification, the PCR technique (two molecular markers), electrophoresis and visualisation of PCR products, and sequencing. A fragment of the beta-giardin gene was used as a molecular marker in order to define the genotype and subgenotype of Giardia. Only in the case of genotyping of two Giardia isolates obtained from Peromyscus eremicus another molecular marker (SSU rRNA)was additionally used. Some widely available computer programmes (Chromas, CAP 3, BioEdit, BLASTn, MEGA version 4.0) were utilised in the analysis of the sequence of the beta-giardin gene fragment and in the phylogenetic analysis. The culture of Giardia trophozoites was established to compare the sequence of the partial beta-giardin gene from cysts and trophozoites. Concentration and purification of Giardia cysts in the saccharose gradient, and the excystation technique were applied in the studies to obtaining an axenic in vitro culture. In this study, Giardia cysts were found in 12 faecal samples obtained from 3 persons and
