  5. Directed neuronal differentiation of human embryonic stem cells

    Full Text Available Abstract Background We have developed a culture system for the efficient and directed differentiation of human embryonic stem cells (HESCs to neural precursors and neurons. HESC were maintained by manual passaging and were differentiated to a morphologically distinct OCT-4+/SSEA-4- monolayer cell type prior to the derivation of embryoid bodies. Embryoid bodies were grown in suspension in serum free conditions, in the presence of 50% conditioned medium from the human hepatocarcinoma cell line HepG2 (MedII. Results A neural precursor population was observed within HESC derived serum free embryoid bodies cultured in MedII conditioned medium, around 7–10 days after derivation. The neural precursors were organized into rosettes comprised of a central cavity surrounded by ring of cells, 4 to 8 cells in width. The central cells within rosettes were proliferating, as indicated by the presence of condensed mitotic chromosomes and by phosphoHistone H3 immunostaining. When plated and maintained in adherent culture, the rosettes of neural precursors were surrounded by large interwoven networks of neurites. Immunostaining demonstrated the expression of nestin in rosettes and associated non-neuronal cell types, and a radial expression of Map-2 in rosettes. Differentiated neurons expressed the markers Map-2 and Neurofilament H, and a subpopulation of the neurons expressed tyrosine hydroxylase, a marker for dopaminergic neurons. Conclusion This novel directed differentiation approach led to the efficient derivation of neuronal cultures from HESCs, including the differentiation of tyrosine hydroxylase expressing neurons. HESC were morphologically differentiated to a monolayer OCT-4+ cell type, which was used to derive embryoid bodies directly into serum free conditions. Exposure to the MedII conditioned medium enhanced the derivation of neural precursors, the first example of the effect of this conditioned medium on HESC.

    The alpha-scattering experiment aboard soft-landing Surveyor VI has provided a chemical analysis of the surface of the moon in Sinus Medii. The preliminary results indicate that, within experimental errors, the composition is the same as that found by Surveyor V in Mare Tranquillitatis. This finding suggests that large portions of the lunar maria resemble basalt in composition.

    In the paper is shown the case of the patient of the CVI life, age of 40 years. It was about the vascular lesion left of the temperoparienal MRA shew the amputation of amputation of the temporal branches art. cerebri medii from the left side. The patient 6 years before she suffered of CVI infract myocard after which she recovered well. After the performed neurologic and physiatric treatment it came to the strength of the rude motor strength with the normalization of the speech.

    Full Text Available All surgeons must bear in mind the existence of muscular variations when performing common tendon transfers. Presence of additional bellies and tendons of existing muscles or presence of additional muscles in unusual locations might misguide a surgeon, during surgery and also during diagnosis. In the present paper we are reporting four cases of variations encountered during the study of extensor muscles of the forearm in 100 limbs. In Case 1, additional bellies of extensor carpi radialis longus and extensor carpi radialis brevis and multiple tendons of insertion of abductor pollicis longus were observed in a single limb. In Case 2, an additional belly of the abductor pollicis longus was observed. In Case 3, a short muscle on the dorsum of the hand going to the index finger [extensor indicis brevis (EIB] was found in addition to the normal extensor indicis (EI. It was also observed that some of the most superficial fleshy fibers of EIB were getting inserted into the tendon of EI. In Case 4, a rare incidence of extensor digiti medii proprius was observed. Further, the related literature is reviewed and the clinical and surgical importance of these muscular variations in diagnosis and proper planning of treatment is discussed.

    Full Text Available Este elaborat şi argumentat teoretic un algoritm eficient pentru determinarea strategiilor optime staţionare în proble-mele stocastice de control optimal discret cu perioada de dirijare infinită, definite pe reţele decizionale cu multiple clase recurente, în care este aplicat criteriul de optimizare a combinaţiei convexe a costurilor medii în clasele recurente. Sunt examinate probleme în care costurile de tranziţie între stările sistemului dinamic şi probabilităţile de tranziţie, definite în stările necontrolabile, sunt constante independente de timp. Algoritmul elaborat este bazat pe modelul de programare liniară pentru determinarea strategiilor optime în problemele de control definite pe reţele decizionale perfecte [3,4].AN ALGORITHM FOR DETERMINING STATIONARY OPTIMAL STRATEGIES FOR STOCHASTIC DISCRETE OPTIMAL CONTROL PROBLEMS DEFINED ON NETWORKS WITH MULTIPLE RECURRENT CLASSESAn efficient algorithm for determining optimal stationary strategies for the stochastic discrete optimal control problems with infinite time horizon is developed and theoretically justified. The problems are defined on decision networks with multiple recurrent classes. The average costs convex combination optimization criterion is applied. We examine problems in which the costs of transitions between the states of the dynamic system and transition probabilities, defined on the uncontrollable states, are constants independent on time. The algorithm is based on the linear programming model developed for determining optimal strategies in control problems defined on perfect decision networks [3,4].

  14. Slow neutrons and secondary gamma ray distributions in concrete shields followed by reflecting layers

    Slow neutrons and secondary gamma ray distributions in concrete shields with and without a reflecting layer behind layer behind the concrete shield have been investigated first in case of using a bare reactor beam and then on using a B-4 C filtered beam. The total and capture secondary gamma ray coefficient (B gamma and B gamma C ), the ratio of the reflected thermal neutron (gamma) the ratio of the secondary gamma rays caused by reflected neutrons to those caused transmitted neutrons (Th I gamma/F I gamma) and the effect of inserting a blocking layer (a B-4 C layer) between the concrete shield and the reflector on the suppression of the produced secondary gamma rays have been investigated. It was found that the presence of the reflector layer behind the concrete shield reflects some thermal neutrons back to the concrete shields and so it increases the number of thermal neutrons at the interface between the concrete shield and the reflector. Also the capture secondary gamma rays was increased at the interface between the two medii due to the capture of the reflected thermal neutrons in the concrete shields. It was shown that B-gamma is higher than and that B g amma B gamma C and I gamma T h/ I gamma i f for the different concrete types is higher in case of using the graphite reflector than that in using either water or paraffin reflectors. Putting a blocking layer (B 4 C layer) between the concrete shield and the reflector decreases the produced secondary gamma rays due to the absorption of the reflected thermal neutrons. 17 figs

    Full Text Available Small and medium agricultural producers of the Republic of Moldova focus on developing highvalue agricultural sector, which provides the greatest profits and, due to this fact, could become an important source of increasing income in the rural sector. This paper presents the results of economic calculations (performed by the authors related to the amount of investments and terms of investment recovery (calculated per 1 hectare for 47 high value agricultural crops (fruits and vegetables, as well as to the economic results of the operational activity of growing these crops (sales incomes, sale costs, gross profit, economic profitability. The authors developed cost budgets and their components for each crop. The presented calculation data will enable local entrepreneurs to select the correct crops based on the economic indicators and to deliberately invest the financial resources. Rezumat. Producătorii agricoli mici şi medii din Republica Moldova sunt orientaţi spre dezvoltarea sectorului agricol de valoare înaltă, care oferă cele mai mari profituri şi care, datorită acestui fapt, ar putea deveni o sursă importantă de majorare a veniturilor în sectorul rural. În această lucrare sunt prezentate rezultatele calculelor economice (efectuate de autori privind suma investiţiilor necesare si termenele de recuperare a investiţiilor (în calcul la 1 ha pentru 47 de culturi de valoare înaltă (fructe şi legume şi estimările privind rezultatele economice din activitatea operaţională la cultivarea acestor culturi (venituri din vânzări, costul vânzărilor, profitul brut, rentabilitatea economică. Autorii au elaborat pentru fiecare cultură bugetul costurilor şi componenţa lor. Datele de calcul prezentate vor permite antreprenorilor autohtoni să selecteze corect culturile în baza indicilor economici şi să investească in mod chibzuit resursele financiare.

    Full Text Available Bee colonies are exposed to a wide variety of actions of the agricultural ecosystem and to a multitude of variations of the environment throughout the year. All these factors can affect the bees’ microbial balance. This study aimed to determine the microflora of bee families before the wintering period. The bees for the research were randomly collected at two apiaries of 17 and 24 bee families. Under laboratory conditions, insemination was performed on common, selective and differential growth media using material of the bees’ intestines. As a result of the investigations, an intensive growth of bacterial colonies was noticed on all microbial culture media. They were predominantly formed by different forms of bacteria such as streptococci, staphylococci, E. coli, Salmonellaand microscopic fungi. Most colonies of microorganisms developed on Endo and Peptonate Agar media, being represented by the microbial colonies of E. coli and streptococci, respectively. Rezumat. Coloniile de albine pe tot parcursul anului sunt expuse la o varietate largă de acțiuni ale ecosiste-melor agricole și la o multitudine de variații ale mediului, care le pot afecta echilibrul microbian. Studiul de faţă îşi propune de a stabili varietatea microflorei familiilor de albine înaintea perioadei de iernat. Albinele pentru cer-cetare au fost colectate în mod aleatoriu de la două stupine, cu 17 şi 24 de familii de albine. În condiții de labora-tor, din intestinele albinelor au fost efectuate însămânțări pe medii nutritive obișnuite, selective și diferențiale. În rezultatul investigațiilor s-a stabilit o creștere intensivă de colonii bacteriene pe toate mediile microbiene nutritive. Acestea au fost constituite preponderent din formele bacteriene precum streptococi, stafilococi, E. coli, salmoneleși fungi microscopici. Cele mai multe colonii de microorganisme s-au dezvoltat pe mediile Endo și Agarul pepto-nat, fiind reprezentate de coloniile

    Full Text Available The article presents the results of field testing of three waste types: wine yeast, vinasse and grain mash derived from grape processing and ethyl alcohol production. The results proved that the administration of two doses of wine yeast (13 and 26 t/ha, of vinasse (300 and 600 m3 /ha and of grain mash (50 and 100 m3 /ha contributed to the significant increase of the organic matter content, mobile phosphorus and changeable potassium in the arable soil layer. The use of wine yeast annually ensured a significant increase of grape yields by 1.2 – 2.2 t/ha or by 12 – 22 % more than those obtained from the unfertilized control variant, where only 10.2 t/ha were harvested. The vinasse had a significant impact on the grapevines productivity as well. The grape harvest constituted on average 0.9 – 1.0 t/ha or by 9 – 10 % more than in the control variant. The grain mash contributed to obtain an average increase of the yield of 993-1292 kg/ha of grain units or by 45 -58 % more compared to the unfertilized variant. Rezumat. În articol sunt prezentate rezultatele testării în câmp a 3 deşeuri: drojdii de vin, vinasă şi borhot de cereale, provenite de la prelucrarea strugurilor şi producerea alcoolului etilic. Rezultatele au demonstrat că administrarea, în două doze, a drojdiilor de vin (13 şi 26 t/ha, a vinasei (300 şi 600 m3 /ha şi a borhotului de cereale (50 şi 100 m3 /ha a favorizat majorarea semnificativă a conţinutului de materie organică, fosfor mobil şi potasiu schimbabil în stratul arabil de sol. Aplicarea drojdiilor de vin anual a asigurat un spor semnificativ de struguri de 1,2-2,2 t/ha sau cu 12-22% mai mult decât la martorul nefertilizat, unde s-a recoltat 10,2 t/ha. Acţiune semnificativă asupra productivităţii plantelor viţei-de-vie a avut-o şi vinasa. Sporul de recoltă în medie a constituit 0,9-1,0 t/ha sau cu 9-10 % mai mult decât la martor. Borhotul de cereale a contribuit la obţinerea unor sporuri medii

Cercetările s-au efectuat pe parcursul anilor 2015–2016. Pe parcursul cercetărilor s-a studiat cantitatea de inflorescenţe până la rarire şi după, cantitatea de fructe pe pom şi la 100 de inflorescenţe, producţia, greutatea şi diametrul medii ale unui fruct. S-a stabilit că tratarea cu Geramid New a diminuat cu 41,9-51,5% cantitatea de inflorescenţe legate, ceea ce a permis şi o amplasare mai raţională a fructelor într-o inflorescenţă. Cea mai mare producţie medie de fructe a fost înregistrată în varianta tratată cu Geramid New în doza de 1,5 l/ha – 50,69 t/ha ori cu 85,2% mai mult în comparaţie cu varianta martor.

  20. The Design of Two Nano-Rovers for Lunar Surface Exploration in the Context of the Google Lunar X Prize

    This paper summarizes a study performed by ten students at the Delft University of Technology on a lunar exploration vehicle suited for competing in the Google Lunar X Prize1. The design philosophy aimed at a quick and simple design process, to comply with the mission constraints. This is achieved by using conventional technology and performing the mission with two identical rovers, increasing reliability and simplicity of systems. Both rovers are however capable of operating independently. The required subsystems have been designed for survival and operation on the lunar surface for an estimated mission lifetime of five days. This preliminary study shows that it is possible for two nano-rovers to perform the basic exploration tasks. The mission has been devised such that after launch the rovers endure a 160 hour voyage to the Moon after which they will land on Sinus Medii with a dedicated lunar transfer/lander vehicle. The mission outline itself has the two nano-rovers travelling in the same direction, moving simultaneously. This mission characteristic allows a quick take-over of the required tasks by the second rover in case of one rover breakdown. The main structure of the rovers will consist of Aluminium 2219 T851, due to its good thermal properties and high hardness. Because of the small dimensions of the rovers, the vehicles will use rigid caterpillar tracks as locomotion system. The track systems are sealed from lunar dust using closed track to prevent interference with the mechanisms. This also prevents any damage to the electronics inside the tracks. For the movement speed a velocity of 0.055 m/s has been determined. This is about 90% of the maximum rover velocity, allowing direct control from Earth. The rovers are operated by a direct control loop, involving the mission control center. In order to direct the rovers safely, a continuous video link with the Earth is necessary to assess its immediate surroundings. Two forward pointing navigational cameras
