  1. Emergence of the Zoonotic Biliary Trematode Pseudamphistomum truncatum in Grey Seals (Halichoerus grypus) in the Baltic Sea

    The biliary trematode Pseudamphistomum truncatum parasitizes a wide range of fish-eating mammals, including humans. Here we report the emergence of this parasite in grey seals (Halichoerus grypus) in the Baltic Sea. One hundred eighty-three of 1 554 grey seals (11.9%) examined from 2002–2013 had detectable hepatobiliary trematode infection. Parasite identification was confirmed as P. truncatum by sequencing the ITS2 region of a pool of five to 10 trematodes from each of ten seals collected off the coast of seven different Swedish counties. The proportion of seals parasitized by P. truncatum increased significantly over time and with increasing age of seals. Males were 3.1 times more likely to be parasitized than females and animals killed in fishery interactions were less likely to be parasitized than animals found dead or hunted. There was no significant difference in parasitism of seals examined from the Gulf of Bothnia versus those examined from the Baltic Proper. Although the majority of infections were mild, P. truncatum can cause severe hepatobiliary disease and resulted in liver failure in at least one seal. Because cyprinid fish are the second intermediate host for opisthorchiid trematodes, diets of grey seals from the Baltic Sea were analysed regarding presence of cyprinids. The proportion of gastrointestinal tracts containing cyprinid remains was ten times higher in seals examined from 2008 to 2013 (12.2%) than those examined from 2002 to 2007 (1.2%) and coincided with a general increase of trematode parasitism in the host population. The emergence and relatively common occurrence of P. truncatum in grey seals signals the presence of this parasite in the Baltic Sea ecosystem and demonstrates how aquatic mammals can serve as excellent sentinels of marine ecosystem change.     Full Text Available Main objective of this study was to investigate the invasive activity of the liver fluke, Pseudamphistomom truncatum against the Caspian seal (Pusa caspica and was exemplified at the gross, light microscopy (LM and electron microscopy (EM levels.The study was done on a freshly dead Caspian Seal in the southern coast of Caspian Sea. The checked Caspian seal probably being died of canine distemper virus and was found host to numerous parasites of four helminth species.P. truncatum caused edematous foci on the surface of the liver with prominent fluid accumulation. Sections of the liver viewed with LM had multiple necrotic areas with extensive hemorrhaging and disorganized hepatic lobules. Granulocytes and invasion of connective tissue were prominent. Whole worms were visible with invasive pathways through the host tissue. Damage to both hepatic ducts and blood vessels were prominent. At the EM level, organelles within the impacted hepatocytes were disorganized as exemplified by the cristae of the mitochondria and the endoplasmic reticulum. Parasite eggs were scattered throughout the tissue.It was shown that this trematode can be very pathogenic to Caspian Seal and as this only mammal of Caspian Sea is an endangered species; this needs more investigation toward control or possible treatment of this helminth.

    Full Text Available A qualidade sanitária e o potencial fisiológico de sementes de soja são fatores determinantes para o estabelecimento de estandes adequados. Assim, objetivou-se avaliar o desempenho das sementes de soja e o crescimento das plantas após inoculação das sementes com Colletotrichum truncatum e tratamento fungicida via peliculização. As sementes foram inoculadas com C. truncatum utilizando técnica de restrição hídrica e, posteriormente, tratadas com fungicidas e polímero. Os tratamentos utilizados foram carbendazin, fludioxonil+mefenoxan, thiabendazol+thiram, tiofanato metílico, caboxin+thiram e sementes não tratadas. As sementes foram tratadas com e sem polímero. Foram realizadas avaliações de germinação das sementes, teste de frio, emergência de plântulas e sanidade das sementes por meio do Blotter test. As plantas, cultivadas em vasos, foram avaliadas pelo comprimento da parte aérea, da matéria seca de raízes e da parte aérea. O tratamento das sementes de soja com fludioxonil+mefenoxan e thiabendazole+thiram é eficiente no controle de Colletotrichum truncatum e melhora o desempenho fisiológico das sementes. O desempenho das sementes de soja e o crescimento das plantas são reduzidos quando as sementes são tratadas com tiofanato metílico. A peliculização não afeta o potencial fisiológico e a qualidade sanitária das sementes de soja.The sanitary and physiological qualities of soybean seeds are determining factors for the establishment of appropriate stands.     One hundred and five soybean germplasms of exotic and national origin were evaluated for their reaction to anthracnose under field condition in Bangladesh. In the field 36 materials were found to be free from infection of C. truncatum (highly resistant) while 19, 37, 3, 5 and 5 germplasms were graded as resistant, moderately resistant, moderately susceptible, susceptible and highly susceptible, respectively. Percentage of seed-borne infection by colletotrichum truncatum varied from one germplasm to another. In total 43 germplasms were completely free from seed-borne infection, whereas up to 5% infection was recorded in 25 samples, 6-30% in 32 samples and 31-36% infection was found in five samples.     To investigate the weight-reducing effect of Acer truncatum leave extract on alimentary obesity rats and its effect on fatty acid synthase (FAS). SPF-grade adult male Wistar rats were fed with high-fat diet and Acer truncatum leave extract (10, 30 and 100 mg/kg BW) was given by gavage once a day for 31 days. Body weight (BW), adipose weight and food consumption were recorded, and the activity of hepatic fatty acid synthase (FAS) was measured.   8. Phylogeny and variability of Colletotrichum truncatum associated with soybean anthracnose in Brazil.

    Fungal diseases are among the main factors limiting high yields of soybean crop. Colletotrichum isolates from soybean plants with anthracnose symptoms were studied from different regions and time periods in Brazil using molecular, morphological and pathogenic analyses. Bayesian phylogenetic inference of GAPDH, HIS3 and ITS-5.8S rDNA sequences, the morphologies of colony and conidia, and inoculation tests on seeds and seedlings were performed. All isolates clustered only with Colletotrichum truncatum species in three well-separated clusters. Intraspecific genetic diversity revealed 27 distinct haplotypes in 51 fungal isolates; some of which were identical to C. truncatum sequences from other regions around the world, while others were related to alternative hosts. Conidia were falcate, hyaline, unicellular and aseptate, formed in acervuli, with variable dimensions. Despite being pathogenic to seedlings by both inoculation methods, variation was observed in the aggressiveness of the tested isolates, which was not correlated with genetic variation. The identification of C. truncatum in the sampled isolates was evidenced as being the only causal agent of soybean anthracnose in Brazil until 2007, with relevant genetic, morphological and pathogenic variability as well as a broad geographical origin. The wide distribution of the predominant C. truncatum haplotype indicated the existence of a highly efficient mechanism of pathogen dispersal over long distances, reinforcing the role of seeds as the primary source of disease inoculum. The characterization and distribution of Colletotrichum species in soybean-producing regions in Brazil is fundamental for understanding the disease epidemiology and for ensuring effective control strategies against anthracnose. © 2016 The Society for Applied Microbiology.

    Colletotrichum truncatum, a major fungal phytopathogen, causes the anthracnose disease on an economically important spice crop chilli (Capsicum annuum), resulting in huge economic losses in tropical and sub-tropical countries. It follows a subcuticular intramural infection strategy on chilli with a short, asymptomatic, endophytic phase, which contrasts with the intracellular hemibiotrophic lifestyle adopted by most of the Colletotrichum species. However, little is known about the molecular determinants and the mechanism of pathogenicity in this fungus. A high quality whole genome sequence and gene annotation based on transcriptome data of an Indian isolate of C. truncatum from chilli has been obtained. Analysis of the genome sequence revealed a rich repertoire of pathogenicity genes in C. truncatum encoding secreted proteins, effectors, plant cell wall degrading enzymes, secondary metabolism associated proteins, with potential roles in the host-specific infection strategy, placing it next only to the Fusarium species. The size of genome assembly, number of predicted genes and some of the functional categories were similar to other sequenced Colletotrichum species. The comparative genomic analyses with other species and related fungi identified some unique genes and certain highly expanded gene families of CAZymes, proteases and secondary metabolism associated genes in the genome of C. truncatum. The draft genome assembly and functional annotation of potential pathogenicity genes of C. truncatum provide an important genomic resource for understanding the biology and lifestyle of this important phytopathogen and will pave the way for designing efficient disease control regimens.

  10. EST mining identifies proteins putatively secreted by the anthracnose pathogen Colletotrichum truncatum

    Full Text Available Abstract Background Colletotrichum truncatum is a haploid, hemibiotrophic, ascomycete fungal pathogen that causes anthracnose disease on many economically important leguminous crops. This pathogen exploits sequential biotrophic- and necrotrophic- infection strategies to colonize the host. Transition from biotrophy to a destructive necrotrophic phase called the biotrophy-necrotrophy switch is critical in symptom development. C. truncatum likely secretes an arsenal of proteins that are implicated in maintaining a compatible interaction with its host. Some of them might be transition specific. Results A directional cDNA library was constructed from mRNA isolated from infected Lens culinaris leaflet tissues displaying the biotrophy-necrotrophy switch of C. truncatum and 5000 expressed sequence tags (ESTs with an average read of > 600 bp from the 5-prime end were generated. Nearly 39% of the ESTs were predicted to encode proteins of fungal origin and among these, 162 ESTs were predicted to contain N-terminal signal peptides (SPs in their deduced open reading frames (ORFs. The 162 sequences could be assembled into 122 tentative unigenes comprising 32 contigs and 90 singletons. Sequence analyses of unigenes revealed four potential groups: hydrolases, cell envelope associated proteins (CEAPs, candidate effectors and other proteins. Eleven candidate effector genes were identified based on features common to characterized fungal effectors, i.e. they encode small, soluble (lack of transmembrane domain, cysteine-rich proteins with a putative SP. For a selected subset of CEAPs and candidate effectors, semiquantitative RT-PCR showed that these transcripts were either expressed constitutively in both in vitro and in planta or induced during plant infection.     The review summarizes the results of first genomic and transcriptomic investigations of the liver fluke Clonorchis sinensis (Opisthorchiidae, Trematoda). The studies mark the dawn of the genomic era for opisthorchiids, which cause severe hepatobiliary diseases in humans and animals. Their results aided in understanding the molecular mechanisms of adaptation to parasitism, parasite survival in mammalian biliary tracts, and genome dynamics in the individual development and the development of parasite-host relationships. Special attention is paid to the achievements in studying the codon usage bias and the roles of mobile genetic elements (MGEs) and small interfering RNAs (siRNAs). Interspecific comparisons at the genomic and transcriptomic levels revealed molecular differences, which may contribute to understanding the specialized niches and physiological needs of the respective species. The studies in C. sinensis provide a basis for further basic and applied research in liver flukes and, in particular, the development of efficient means to prevent, diagnose, and treat clonorchiasis.

    Brucella infection in seals was reported for the first time in 1994 around the coast of Scotland. Since then, marine mammal Brucella infections were found to be widely distributed in the northern hemisphere. Two Brucella species affect marine mammals: Brucella pinnipedialis in pinnipeds and Brucella ceti in cetaceans. We examined the livers of Baltic grey seals (Halichoerus grypus) from the Finnish coast (n=122) hunted, found dead, or killed as by-catch in fishing gear in 2013-15 as part of population health monitoring. We detected B. pinnipedialis in the livers of three grey seals. The bacterium was isolated from livers displaying parasitic cholangitis. We also detected Brucella DNA in liver flukes (Pseudamphistomum truncatum) obtained from a Brucella-infected grey seal, suggesting that flukes might be possible vectors of this pathogen in the marine environment.

    C. gloeosporioides sensu lato is one of the most economically important post-harvest diseases affecting papaya production worldwide. There is currently no information concerning the genetic structure or demographic history of this pathogen in any of the affected countries. Knowledge of molecular demographic parameters for different populations will improve our understanding of the biogeographic history as well as the evolutionary and adaptive potential of these pathogens. In this study, sequence data for ACT, GPDH, β-TUB and ITS gene regions were analyzed for C. gloeosporioides sensu lato and C. truncatum isolates infecting papaya in Trinidad and Mexico in order to determine the genetic structure and demographic history of these populations. The data indicated that Mexico is the ancestral C. gloeosporioides sensu lato population with asymmetrical migration to Trinidad. Mexico also had the larger effective population size but, both Mexico and Trinidad populations exhibited population expansion. Mexico also had greater nucleotide diversity and high levels of diversity for each gene. There was significant sub-division of the Trinidad and Mexico populations and low levels of genetic divergence among populations for three of the four gene regions; β-TUB was shown to be under positive selection. There were also dissimilar haplotype characteristics for both populations. Mutation may play a role in shaping the population structure of C. gloeosporioides sensu lato isolates from Trinidad and from Mexico, especially with respect to the ACT and GPDH gene regions. There was no evidence of gene flow between the C. truncatum populations and it is possible that the Mexico and Trinidad populations emerged independently of each other. The study revealed relevant information based on the genetic structure as well as the demographic history of two fungal pathogens infecting papaya, C. gloeosporioides sensu lato and C. truncatum, in Trinidad and Mexico.     Considering the potential of anthracnose to decrease soybean yield and the need to gain more information regarding its effect on soybean physiology, the present study performed an in-depth analysis of the photosynthetic performance of soybean leaflets challenged with Colletotrichum truncatum by combining chlorophyll a fluorescence images with gas-exchange measurements and photosynthetic pigment pools. There were no significant differences between non-inoculated and inoculated plants in leaf water potential, apparent hydraulic conductance, net CO 2 assimilation rate, stomatal conductance to water vapor and transpiration rate. For internal CO 2 concentration, significant difference between non-inoculated and inoculated plants occurred only at 36 h after inoculation. Reductions in the values of the chlorophyll a fluorescence parameters [initial fluorescence (F 0 ), maximal fluorescence (F m ), maximal photosystem II quantum yield (F v /F m ), quantum yield of regulated energy dissipation (Y(NPQ))] and increases in effective PS II quantum yield (Y(II)), quantum yield of non-regulated energy dissipation Y(NO) and photochemical quenching coefficient (q P ) were noticed on the necrotic vein tissue in contrast to the surrounding leaf tissue. It appears that the impact of the infection by C. truncatum on the photosynthetic performance of the leaflets was minimal considering the preference of the fungus to colonize the veins.     We assessed the influence of various carbon concentrations and carbon-to-nitrogen (C:N) ratios on Colletotrichum truncatum NRRL 13737 conidium formation in submerged cultures grown in a basal salts medium containing various amounts of glucose and Casamino Acids. Under the nutritional conditions tested, the highest conidium concentrations were produced in media with carbon concentrations of 4.0 to 15.3 g/liter. High carbon concentrations (20.4 to 40.8 g/liter) inhibited sporulation and enhanced the formation of microsclerotiumlike hyphal masses. At all the carbon concentrations tested, a culture grown in a medium with a C:N ratio of 15:1 produced more conidia than cultures grown in media with C:N ratios of 40:1 or 5:1. While glucose exhaustion was often coincident with conidium formation, cultures containing residual glucose sporulated and those with high carbon concentrations (>25 g/liter) exhausted glucose without sporulation. Nitrogen source studies showed that the levels of C. truncatum NRRL 13737 conidiation were similar for all protein hydrolysates tested. Reduced conidiation occurred when amino acid and inorganic nitrogen sources were used.     Pepper anthracnose, caused by Colletotrichum species complex is the most destructive disease of chilli (Capsicum annuum L.). miRNAs are key modulators of transcriptional and post- transcriptional expression of genes during defense responses. In the present study, we performed a comparative miRNA profiling of susceptible (Arka Lohit-AL) and resistant (Punjab Lal-PL) chilli cultivars to identify 35 differentially expressed miRNAs that could be classified as positive, negative or basal regulators of defense against C. truncatum, the most potent anthracnose pathogen. Interestingly, a novel microRNA can-miRn37a was significantly induced in PL but largely repressed in AL genotype post pathogen attack. Subsequent over-expression of can-miRn37a in AL showed enhanced resistance to anthracnose, as evidenced by decreased fungal growth and induced expression of defense-related genes. Consequently, the expression of its three target genes encoding the ethylene response factors (ERFs) was down-regulated in PL as well as in the over-expression lines of AL genotypes. The ability of these targets to be regulated by can-miRn37a was further confirmed by transient co-expression in Nicotiana benthamiana. Additionally, the virus-induced silencing of the three targets in the susceptible AL cultivar revealed their role in fungal colonization and induction of C. truncatum pathogenicity in chilli. Taken together, our study suggests that can-miRn37a provides a potential miRNA mediated approach of engineering anthracnose resistance in chilli by repressing ERFs and preventing fungal colonization.     To study the dynamic variations of the contents of total polyphenols, flvonoids and chlorogenic acid from Acer truncatum leaves in different months, and their inhibitory activities on fatty acid synthase. Spectrophotometry was used to determine the contents of total polyphenols, flavonoids and chlorogenic acid in extracts and the extracts' inhibitory effects were also investigated. All Leaves picked from May to November have inhibitory effect. But the contents of polyphenols in leaves of July appeared to be higher than other months', and consequently exhibited stronger inhibition against FAS. A positive correlation between the content of polyphenols in leaves extract and the inhibitory efficacy on FAS could be established.

    A two-year experiment exposing Acer truncatum Bunge seedlings to elevated ozone (O 3 ) concentrations above ambient air (AO) and drought stress (DS) was carried out using open-top chambers (OTCs) in a suburb of Beijing in north China in 2012–2013. The results suggested that AO and DS had both significantly reduced leaf mass area (LMA), stomatal conductance (Gs), light saturated photosynthetic rate (A sat ) as well as above and below ground biomass at the end of the experiment. It appeared that while drought stress mitigated the expression of foliar injury, LMA, leaf photosynthetic pigments, height growth and basal diameter, due to limited carbon fixation, the O 3 – induced reductions in A sat , Gs and total biomass were enhanced 23.7%. 15.5% and 8.1% respectively. These data suggest that when the whole plant was considered that drought under the conditions of this experiment did not protect the Shantung maple seedlings from the effects of O 3 . - Highlights: • The response of Acer truncatum Bunge to drought and ozone was investigated. • Drought could mitigate the foliage injury and leaf photosynthetic pigments. • The O 3 -induced reductions in Asat, Gs and total biomass were enhanced by drought. - Drought didn't protect Shantung maple from O 3 effects but rather cause more reductions in biomass

  3. Response Surface Methodology Optimization of Ultrasonic-Assisted Extraction of Acer Truncatum Leaves for Maximal Phenolic Yield and Antioxidant Activity.

    This study is the first to report the use of response surface methodology to improve phenolic yield and antioxidant activity of Acer truncatum leaves extracts (ATLs) obtained by ultrasonic-assisted extraction. The phenolic composition in ATLs extracted under the optimized conditions were characterized by UPLC-QTOF-MS/MS. Solvent and extraction time were selected based on preliminary experiments, and a four-factors-three-levels central composite design was conducted to optimize solvent concentration ( X ₁), material-to-liquid ratio ( X ₂), ultrasonic temperature ( X ₃) and power ( X ₄) for an optimal total phenol yield ( Y ₁) and DPPH• antioxidant activity ( Y ₂). The results showed that the optimal combination was ethanol:water ( v : v ) 66.21%, material-to-liquid ratio 1:15.31 g/mL, ultrasonic bath temperature 60 °C, power 267.30 W, and time 30 min with three extractions, giving a maximal total phenol yield of 7593.62 mg gallic acid equivalent/100 g d.w. and a maximal DPPH• antioxidant activity of 74,241.61 μmol Trolox equivalent/100 g d.w. Furthermore, 22 phenolics were first identified in ATL extract obtained under the optimized conditions, indicating that gallates, gallotannins, quercetin, myricetin and chlorogenic acid derivatives were the main phenolic components in ATL. What's more, a gallotannins pathway existing in ATL from gallic acid to penta- O -galloylglucoside was proposed. All these results provide practical information aiming at full utilization of phenolics in ATL, together with fundamental knowledge for further research.

    Polyphenols, including flavonoids, are the major components of the extracts from aceraceous plants. They possess remarkable antibacterial and antitumour activity. Our study focused on whether the inhibition of the bacterial type II fatty acid synthesis system is the mechanism for the antibacterial effect of the related plant polyphenols. Extracts obtained from the fallen leaves of the Shantung maple (Acer truncatum Bunge) using different solvents, and the related pure compound PGG (1,2,3,4,6-penta-O-galloyl-beta-D-glucose), potently inhibited the FabG (beta-oxoacyl-ACP reductase) steps in the fatty-acid-elongation cycle with the IC(50) values between 0.9 and 7.2 microg/ml. An ethyl acetate extract appeared to inhibit FabG reductase in a mixed manner with NADPH, as did PGG with NADPH, demonstrating that they interfered with the binding of the cofactor to the enzyme. Gram-positive and Gram-negative bacteria and some fungi were used to evaluate the antibacterial abilities of different extract samples. The experiments showed that a higher polyphenol content of the extracts led to a more potent inhibitory capacity against FabG, thus enhancing the antibacterial efficacy.

    The large population of feral cats in Denmark may potentially transmit pathogens to household cats and zoonotic parasites to humans. A total of 99 euthanized cats; feral cats (n=92) and household cats with outdoor access (n=7), were collected from March to May 2014 from the Zealand region, Denmark. The sedimentation and counting technique (SCT) was used to isolate helminths and coproscopy was done by concentration McMaster technique (c-McMaster). Overall, 90.1% of the cats were infected and a total of 10 species were recorded by SCT: 5 nematode species: Toxocara cati (84.8%), Ollulanus tricuspis (13.1%), Aonchotheca putorii (7.1%), Paersonema spp. (3.0%), Strongyloides spp. (1.0%); 3 cestodes: Hydatigera taeniaeformis (36.4%), Mesocestoides sp. (3.0%), Dipylidium caninum (1.0%); and 2 trematodes: Cryptocotyle spp. (5.1%) and Pseudamphistomum truncatum (1.0%). O. tricuspis was the second most common gastrointestinal nematode of cats but had the highest intensity of infection. For T. cati, prevalence and worm burden were significantly higher in feral than household cats. No juvenile cats were infected with H. taeniaeformis, and age thus had a significant effect on prevalence and worm burdens of this species. Rural cats had a higher prevalence and worm burden of A. putorii than urban cats. By c-McMaster, ascarid, capillarid, strongylid or taeniid type eggs were found in 77.9% of the cats while Cystoisospora felis was found in 2.1%. The sensitivity of the c-McMaster was 82.5% for T. cati but 26.5% for taeniid eggs, using the SCT as gold standard. A positive correlation between faecal egg counts and worm burdens was seen for T. cati, but not for taeniid eggs (assumed to be H. taeniaeformis). Coprological examination also detected the eggs of extraintestinal Capillariidae species including Eucoleus aerophilus and Eucoleus boehmi, but further necropsy studies are needed to confirm these findings.     Full Text Available Colletotrichum truncatum is the most common pathogen fungus associated with soybean anthracnose. Although the lignin-degrading enzyme laccase has been implicated in pathogenicity of a wide range of plant pathogenic fungi, its biological role in the Colletotrichum -soybean disease system is unknown. The extent of the infection in our country led us to examine laccase production in Argentinean Colletotrichum strains isolated from diseased soybean plants from different geographic locations. Ten strains (eight of them identified as C. truncatum , were screened for in vitro laccase production. Only six of the isolates, all of them C. truncatum , produced laccase activity when cultured on a defined medium based on pectin and asparagine as carbon and nitrogen sources, respectively. Strain BAFC 3102 (isolated from Chaco province, yielded the highest laccase titers (44 U/L in this medium. Denaturing polyacrylamide gel electrophoresis of extracellular culture fluids revealed one band with laccase activity (mol wt 67 kDa. CuSO 4 addition to media with either glucose or pectin as carbon sources increased up to 7-fold laccase production (280 U/L in the glucose medium, but the pattern of isoenzyme was not affected by culture age or medium composition.     Full Text Available Genetic diversity and differentiation of 50 Colletotrichum spp. isolates from legume crops studied through multigene loci, RAPD and ISSR analysis. DNA sequence comparisons by six genes (ITS, ACT, Tub2, CHS-1, GAPDH, and HIS3 verified species identity of C. truncatum, C. dematium and C. gloeosporiodes and identity C. capsici as a synonym of C. truncatum. Based on the matrix distance analysis of multigene sequences, the Colletotrichum species showed diverse degrees of intera and interspecific divergence (0.0 to 1.4% and (15.5–19.9, respectively. A multilocus molecular phylogenetic analysis clustered Colletotrichum spp. isolates into 3 well-defined clades, representing three distinct species; C. truncatum, C. dematium and C. gloeosporioides. The ISSR and RAPD and cluster analysis exhibited a high degree of variability among different isolates and permitted the grouping of isolates of Colletotrichum spp. into three distinct clusters. Distinct populations of Colletotrichum spp. isolates were genetically in accordance with host specificity and inconsistent with geographical origins. The large population of C. truncatum showed greater amounts of genetic diversity than smaller populations of C. dematium and C. gloeosporioides species. Results of ISSR and RAPD markers were congruent, but the effective maker ratio and the number of private alleles were greater in ISSR markers.

    Full Text Available Opisthorchiasis is a parasitic infection caused by the liver flukes of the Opisthorchiidae family. Both experimental and epidemiological data strongly support a role of these parasites in the etiology of the hepatobiliary pathologies and an increased risk of intrahepatic cholangiocarcinoma. Understanding a functional link between the infection and hepatobiliary pathologies requires a detailed description a host-parasite interaction on different levels of biological regulation including the metabolic response on the infection. The last one, however, remains practically undocumented. Here we are describing a host response on Opisthorchiidae infection using a metabolomics approach and present the first exploratory metabolomics study of an experimental model of O. felineus infection.We conducted a Nuclear Magnetic Resonance (NMR based longitudinal metabolomics study involving a cohort of 30 animals with two degrees of infection and a control group. An exploratory analysis shows that the most noticeable trend (30% of total variance in the data was related to the gender differences. Therefore further analysis was done of each gender group separately applying a multivariate extension of the ANOVA-ASCA (ANOVA simultaneous component analysis. We show that in the males the infection specific time trends are present in the main component (43.5% variance, while in the females it is presented only in the second component and covers 24% of the variance. We have selected and annotated 24 metabolites associated with the observed effects and provided a physiological interpretation of the findings.The first exploratory metabolomics study an experimental model of O. felineus infection is presented. Our data show that at early stage of infection a response of an organism unfolds in a gender specific manner.     Full Text Available Centrocestus formosanus es un parásito trematodo zoonótico originario de Asia asociado con muertes de peces principalmente de cultivo. 907 moluscos provenientes de estanques sembrados con tilapias, seleccionados uno por provincia fueron identificados al nivel taxonómico especifico. Se identificaron cuatro gastrópodos y un bivalvo: M. tuberculata, M. turricula, P. flagellata, H. cubensis y A. luteola. Se reporta, por primera vez, la presencia de dos especies de moluscos en Costa Rica. Se identificaron siete morfotipos de cercarias parasitando las cinco especies de moluscos encontradas. En la segunda exposición experimental se demostró que el morfotipo parapleurolofocercus encontrado en M. tuberculata concuerda con el hallazgo de C. formosanus en alevines de tilapia, después del examen clínico, anatomopatológico y parasitológico realizado a los alevines expuestos. Las metacercarias fueron extraídas del quiste utilizando microagujas y micropinzas lavadas en solución salina fisiológica (0.65%, fijadas en formol caliente al 4% y después esquematizadas con una cámara clara adaptada a un microscopio fotónico, estimándose una abundancia e intensidad media de 1018-1027 digeneos por branquia en cada pez parasitado, determinándose así el hospedador intermediario primario y secundario del parásito.     Amphimerus spp. flukes are known to infect mammals, but human infections have not been confirmed. Microscopy of fecal samples from 397 persons from Ecuador revealed Opisthorchiidae eggs in 71 (24%) persons. Light microscopy of adult worms and scanning electron microscopy of eggs were compatible with descriptions of Amphimerus spp. This pathogen was only observed in communities that consumed undercooked fish.

    Full Text Available A study was conducted in the subtropical area of Southern Brazil to determine the survival of pathogens in soybean residues under conventional and no-tillage cultivation systems from March to September of 1998 and 1999. The pathogens most frequently isolated were Colletotrichum truncatum, Phomopsis spp., Cercospora kikuchii, Fusarium spp., Macrophomina phaseolina, and Rhizoctonia solani. Other fungi isolated were Myrothecium roridum, Penicillium sp., Chaetomium sp., Epicoccum sp., Corynespora cassiicola and Trichoderma sp. The percent of survival of each pathogen varied according to the month and the year. Survival of C. truncatum, Phomopsis spp. and C. kikuchii were significantly reduced (pAvaliou-se a sobrevivência de patógenos em restos de soja, em sistema de semeadura direta e convencional, entre março e setembro de 1998 e 1999, em Londrina, PR. Os patógenos mais freqüentemente isolados foram Colletotrichum truncatum, Phomopsis spp., Cercospora kikuchii, Fusarium spp., Macrophomina phaseolina e Rhizoctonia solani. M. phaseolina e Fusarium spp. não foram afetados, ou foram favorecidos pelo enterro dos resíduos. A freqüência de isolamento de Fusarium spp. aumentou em resíduos enterrados no solo. A perda de biomassa mostrou redução de 44,4% no sistema convencional e 34,9% no sistema de semeadura direta, em 1998, quando a distribuição de chuvas foi mais regular.     The anthracnose caused by Colletotrichum species is an important disease that primarily causes fruit rot in pepper. Eighty-eight strains representing seven species of Colletotrichum were obtained from rotten pepper fruits in Sichuan Province, China, and characterized according to morphology and the glyceraldehyde-3-phosphate dehydrogenase (GAPDH) sequence. Fifty-two strains were chosen for identification by phylogenetic analyses of multi-locus sequences, including the nuclear ribosomal internal transcribed spacer (ITS) region and the β-tubulin (TUB2), actin (ACT), calmodulin (CAL) and GAPDH genes. Based on the combined datasets, the 88 strains were identified as Colletotrichum gloeosporioides, C. siamense, C. fructicola, C. truncatum, C. scovillei, and C. brevisporum, and one new species was detected, described as Colletotrichum sichuanensis. Notably, C. siamense and C. scovillei were recorded for the first time as the causes of anthracnose in peppers in China. In addition, with the exception of C. truncatum, this is the first report of all of the other Colletotrichum species studied in pepper from Sichuan. The fungal species were all non-host-specific, as the isolates were able to infect not only Capsicum spp. but also Pyrus pyrifolia in pathogenicity tests. These findings suggest that the fungal species associated with anthracnose in pepper may inoculate other hosts as initial inoculum.

  15. Detection of Rickettsia aeschlimannii and Rickettsia africae in ixodid ticks from Burkina Faso and Somali Region of Ethiopia by new real-time PCR assays.

    In the framework of cooperation for development projects in Burkina Faso and Ethiopia, we collected ixodid ticks from cattle, small ruminants and camels. We optimized new TaqMan Probe real-time PCR assays to detect Rickettsia aeschlimannii and Rickettsia africae OmpA gene in the collected samples. Rickettsia africae was identified in 75.0% Amblyomma variegatum (95%CI: 56.6-88.5), while R. aeschlimannii in 24.0% Hyalomma truncatum (95%CI: 9.4-45.1) and 50.0% H. rufipes (95%CI: 29.9-70.0) collected from cattle in different provinces throughout Burkina Faso. Ticks from the Libaan zone, Somali Region of Ethiopia, were also infected by R. africae (28.5% prevalence in Amblyomma gemma, 95%CI: 14.7-46.0) and R. aeschlimannii (27.0% H. truncatum, 95%CI: 5.0-62.9; 88.3% H. rufipes, 95%CI: 60.5-99.3). All tested ticks were adults. The developed diagnostic tools were highly sensitive and enabled us to rapidly classify R. aeschlimannii and R. africae, which were identified in Burkina Faso and in the Somali Region of Ethiopia for the first time. Further studies are needed to assess the zoonotic risk and prevalence of infection in local human populations, who have high contact rates with ticks and their animal hosts.     Full Text Available La antracnosis, causada por Colletotrichum spp., es una de la enfermedades poscosecha más importantes en papaya. En este estudio, frutas de papaya, híbrido Pococí, recolectados en plantaciones ubicadas en el cantón de Guácimo, provincia de Limón, presentaron 7 síntomas de antracnosis, luego de un período de almacenamiento. De cada síntoma se obtuvieron aislados de Colletotrichum en medio de cultivo PDA (papa-dextrosa-agar. Los aislamientos se clasificaron en 3 grupos según la similitud de los síntomas y la morfología de la colonia. El análisis de la secuencia de la región espaciadora interna transcrita (ITS del ADN ribosomal reveló que los aislamientos pertenecieron a 3 especies del género Colletotrichum: C. gloeosporioides sensu lato, C. magnum y C. capsici (C. truncatum. La identificación de las especies fue confirmada con imprimadores taxón específico para C. gloeosporioides e imprimadores especie-específicos para C. magnum. Las 3 especies identificadas mostraron ser patogénicas sobre frutas de papaya. Las especies C. magnum y C. capsici (C. truncatum se registran por primera vez en Costa Rica, como agentes causales de la antracnosis en papaya.

    A total of 264 scrub hares (Lepus saxatilis) were examined for ixodid ticks at various localities in the Kruger National Park, eastern Transvaal Lowveld. Thirteen tick species were recovered from these hares. The seasonal abundances of the immature stages of Amblyomma hebraeum, Amblyomma marmoreum, Hyalomma truncatum, Rhipicephalus appendiculatus, Rhipicephalus evertsi evertsi, Rhipicephalus simus and Rhipicephalus zambeziensis and all stages of a Rhipicephalus sp. (near R. pravus) were determined. Three scrub hares, examined in the north-western Transvaal Bushveld, were infested with five ixodid tick species. Ten hares examined in the eastern Transvaal Highveld harboured three species. A total of 15 ixodid tick species were recovered from the scrub hares examined in the three regions of the Transvaal. No haematozoa were found in blood smears made from the hares examined in the southern region of the Kruger National Park.

  18. Inter-individual and intragenomic variations in the ITS region of Clonorchis sinensis (Trematoda: Opisthorchiidae) from Russia and Vietnam.

    Here we examined the intraspecific genetic variability of Clonorchis sinensis from Russia and Vietnam using nuclear DNA sequences (the 5.8S gene and two internal transcribed spacers of the ribosomal cluster). Despite the low level of variability in the ITS1 region, this marker has revealed some features of C. sinensis across multiple geographic regions. The genetic diversity levels for the Russian and Vietnamese populations were similar (0.1 and 0.09%, respectively) but were significantly lower than the C. sinensis from China (0.31%). About half of the sequences of the Chinese (53%) and Korean (47%) populations and about a tenth of the Vietnamese (12%) and Russian (8%) sequences included a 5bp insertion. No sequences with nucleotide substitutions both upstream and downstream of the 5bp insertion were found within the whole data set. The population of northern China had both sequence variants (with substitutions either upstream or downstream of the insertion), while only one of these variants was presented at the other localities. The Vietnamese population had a higher frequency of intragenomic polymorphism than the Russian population (69% vs. 46% and 23% vs. 3% at the 114bp and 339bp positions, respectively).     Opisthorchis felineus or Siberian liver fluke is a trematode parasite (Opisthorchiidae) that infects the hepato-biliary system of humans and other mammals. Despite its public health significance, this wide-spread Eurasian species is one of the most poorly studied human liver flukes and nothing is known about its population genetic structure and demographic history. In this paper, we attempt to fill this gap for the first time and to explore the genetic diversity in O. felineus populations from Eastern Europe (Ukraine, European part of Russia), Northern Asia (Siberia) and Central Asia (Northern Kazakhstan). Analysis of marker DNA fragments from O. felineus mitochondrial cytochrome c oxidase subunit 1 and 3 (cox1, cox3) and nuclear rDNA internal transcribed spacer 1 (ITS1) sequences revealed that genetic diversity is very low across the large geographic range of this species. Microevolutionary processes in populations of trematodes may well be influenced by their peculiar biology. Nevertheless, we suggest that lack of population genetics structure observed in O. felineus can be primarily explained by the Pleistocene glacial events and subsequent sudden population growth from a very limited group of founders. Rapid range expansion of O. felineus through Asian and European territories after severe bottleneck points to a high dispersal potential of this trematode species. PMID:23634228

  2. Elucidation of biocontrol mechanisms of Trichoderma harzianum against different plant fungal pathogens: Universal yet host specific response.

    In the present study, different transcripts of Trichoderma harzianum ThHP-3 were evaluated for their response against four fungal pathogens Fusarium oxysporum, Colletotrichum capsici, Colletotrichum truncatum and Gloesercospora sorghi using RT-qPCR. The time course study of T. harzianum transcripts related to signal transduction, lytic enzymes, secondary metabolites and various transporters revealed variation in expression against four fungal pathogens. In a broader term, the transcripts were upregulated at various time intervals but the optimum expression of cyp3, abc, nrp, tga1, pmk, ech42 and glh20 varied with respect to host fungi. Additionally, the expression of transcripts related to transporters/cytochromes was also observed against Fusarium oxysporum after 96h whereas transcripts related to secondary metabolites and lytic enzymes showed significant difference in expression against Colletotrichum spp. from 72 to 96h.   3. Influence of He-Ne laser irradiation of soybean seeds on seed mycoflora, growth, nodulation, and resistance to Fusarium solani

    Laser irradiation of soybean seeds for 3 min caused a clear reduction in the number of seed-borne fungi which became more pronounced as the irradiation time was extended. Pretreatment of the seeds with methylene blue, methyl red and carmine enhanced the effect of laser. Rhizoctonia solani, Alternaria tenuissima, Cercospora kikuchii and Colletotrichum truncatum were completely eliminated when the seeds were pretreated with a dye and irradiated for 10 min. Seed germination was stimulated on exposure of the seed to 1-min irradiation. Chlorophyll a, chlorophyll b and carotenoid content of developed plants differed, depending on the irradiation dose and dye treatment of the seeds. The number and dry mass of nodules were mostly greater (as compared to the corresponding control), when the seeds irradiated for 1 or 3 min were pretreated with methyl red, chlorophenol red, crystal violet and methylene blue.   4. Differential Response of Extracellular Proteases of Trichoderma Harzianum Against Fungal Phytopathogens.

    In the present study, production of extracellular proteases by Trichoderma harzianum was evaluated based on the relative gene expression and spectrophotometric assay. The fungal isolates were grown in Czapek Dox Broth medium supplemented with deactivated mycelium of plant fungal pathogens such as Fusarium oxysporum, Colletotrichum capsici, Gloeocercospora sorghi, and Colletotrichum truncatum. The maximum protease activity was detected after 48 h of incubation against Colletotrichum spp. Similarly in qRT-PCR, the relative gene expression of four proteases varied from 48 to 96 h against host pathogens in a time-independent manner. Among proteases, statistically significant upregulation of asp, asp, and srp was observed against Colletotrichum spp., followed by F. oxysporum. But in the case of pepM22, maximum upregulation was observed against F. oxysporum. The variation in enzyme assay and qRT-PCR of proteases at different time intervals against various fungal phytopathogens could be due to the limitation of using casein as a substrate for all types of proteases or protease-encoding transcripts selected for qRT-PCR, which may not be true representative of total protease activity.

Meril were carried out aiming to control anthracnose (pod blight caused by fungus, Colletotrichum truncatum with five treatments represented by different fungicidal sprays against control receiving no spray with three replicates of each under field conditions during two consecutive years from 2012 to 2013. In 2012, the higher Percent Disease Control (PDC and Percent Yield Increase (PYI were estimated in plot treated with SAAF (Carbendazim 12% + Mancozeb 63% followed by Mancozeb fungicides. The mean Pod Infection (PI was low in plots treated with SAAF followed by Mancozeb. Almost similar trends of disease control were observed in 2013. The lower Percent Disease Index (PDI was 46.25% and mean PI was 29.67% with higher yield value of 2431.25 kg/ha obtained from the plots sprayed with SAAF then by Mancozeb. The results showed that, the combined treatment with fungicides, SAAF followed by Mancozeb were effective to control anthracnose or pod blight disease of soybean to increase the yield.

  6. Colletotrichum species causing anthracnose disease of chili in China.

    Anthracnose caused by Colletotrichum species is a serious disease of more than 30 plant genera. Several Colletotrichum species have been reported to infect chili in different countries. Although China is the largest chili-producing country, little is known about the species that have been infecting chili locally. Therefore, we collected samples of diseased chili from 29 provinces of China, from which 1285 strains were isolated. The morphological characters of all strains were observed and compared, and multi-locus phylogenetic analyses (ITS, ACT , CAL , CHS-1 , GAPDH , TUB2 , and HIS3 ) were performed on selected representative strains. Fifteen Colletotrichum species were identified, with C. fioriniae , C. fructicola , C. gloeosporioides , C. scovillei , and C. truncatum being prevalent. Three new species, C. conoides , C. grossum , and C. liaoningense , were recognised and described in this paper. Colletotrichum aenigma , C. cliviae , C. endophytica , C. hymenocallidis , C. incanum , C. karstii , and C. viniferum were reported for the first time from chili. Pathogenicity of all species isolated from chili was confirmed, except for C. endophytica . The current study improves the understanding of species causing anthracnose on chili and provides useful information for the effective control of the disease in China.

    Anthracnose caused by Colletotrichum is one of the most severe diseases that can afflict Camellia sinensis. However, research on the diversity and geographical distribution of Colletotrichum in China remain limited. In this study, 106 Colletotrichum isolates were collected from diseased leaves of Ca. sinensis cultivated in the 15 main tea production provinces in China. Multi-locus phylogenetic analysis coupled with morphological identification showed that the collected isolates belonged to 11 species, including 6 known species (C. camelliae, C. cliviae, C. fioriniae, C. fructicola, C. karstii, and C. siamense), 3 new record species (C. aenigma, C. endophytica, and C. truncatum), 1 novel species (C. wuxiense), and 1 indistinguishable strain, herein described as Colletotrichum sp. Of these species, C. camelliae and C. fructicola were the dominant species causing anthracnose in Ca. sinensis. In addition, our study provided further evidence that phylogenetic analysis using a combination of ApMat and GS sequences can be used to effectively resolve the taxonomic relationships within the C. gloeosporioides species complex. Finally, pathogenicity tests suggested that C. camelliae, C. aenigma, and C. endophytica are more invasive than other species after the inoculation of the leaves of Ca. sinensis.

  8. Oak Tree Canker Disease Supports Arthropod Diversity in a Natural Ecosystem

    Full Text Available Microorganisms have many roles in nature. They may act as decomposers that obtain nutrients from dead materials, while some are pathogens that cause diseases in animals, insects, and plants. Some are symbionts that enhance plant growth, such as arbuscular mycorrhizae and nitrogen fixation bacteria. However, roles of plant pathogens and diseases in natural ecosystems are still poorly understood. Thus, the current study addressed this deficiency by investigating possible roles of plant diseases in natural ecosystems, particularly, their positive effects on arthropod diversity. In this study, the model system was the oak tree (Quercus spp. and the canker disease caused by Annulohypoxylon truncatum, and its effects on arthropod diversity. The oak tree site contained 44 oak trees; 31 had canker disease symptoms while 13 were disease-free. A total of 370 individual arthropods were detected at the site during the survey period. The arthropods belonged to 25 species, 17 families, and seven orders. Interestingly, the cankered trees had significantly higher biodiversity and richness compared with the canker-free trees. This study clearly demonstrated that arthropod diversity was supported by the oak tree canker disease.

  9. The leaf phenophase of deciduous species altered by land pavements

    It has been widely reported that the urban environment alters leaf and flowering phenophases; however, it remains unclear if land pavement is correlated with these alterations. In this paper, two popular deciduous urban trees in northern China, ash (Fraxinus chinensis) and maple (Acer truncatum), were planted in pervious and impervious pavements at three spacings (0.5 m × 0.5 m, 1.0 m × 1.0 m, and 2.0 m × 2.0 m apart). The beginning and end dates of the processes of leaf budburst and senescence were recorded in spring and fall of 2015, respectively. The results show that leaf budburst and senescence were significantly advanced in pavement compared to non-pavement lands. The date of full leaf budburst was earlier by 0.7-9.3 days for ash and by 0.3-2.3 days for maple under pavements than non-pavements, respectively. As tree spacing increases, the advanced days of leaf budburst became longer. Our results clearly indicate that alteration of leaf phenophases is attributed to land pavement, which should be taken into consideration in urban planning and urban plant management.

    Centrocestusformosanus is a zoonotic trematode from Asia and has been mainly associated as cause of death of cultured fish. To identify pathogen trematode species in tilapia fry (Oreochromis niloticus) and to determine mollusks hosting these parasites, freshwater mollusks were collected from tilapia cultured ponds and experimental infections were carried out with tilapia fries and different mollusk species. A total of 907 freshwater mollusks were obtained from tilapia ponds and were identified to species level, four gastropods and one bivalve were determined: Melania tuberculata, Melanoides turricula, Pomacea flagellata, Haitia cubensis and Anodontiles luteola. For the first time, the presence of M. turricula and H. cubensis are reported in Costa Rica. Seven morphotypes of cercariae (Xifiodiocercaria, Equinostoma, Oftalmocercaria, Parapleurolofocercus, Cistocerca, Furcocercaria and Leptocercaria) parasitizing all five species of mollusks were found, all of distome type. Experimental exposure of tilapia fry to M. tuberculata demonstrated that the parapleurolofocercus morphotype found in the mollusk is in accordance with the finding of C. formosanus in tilapia fry. An abundance and mean intensity of 1018-1027 digeneans per gill in each exposed fish was determined. Centrocestus formosanus is reported for the first time in Costa Rica, for which the primary and secondary intermediate hosts were also determined.

  11. Epidemiology of fishborne trematodiasis in Kazakhstan.

    Fish borne trematodes are an important cause of morbidity in Kazakhstan. The number of human cases of opisthorchiidosis (infection with parasites of the family opisthorchiidae) reached a peak of 2521 recorded cases (17 cases per 100,000) in 2002 with a gradual decline to 1225 cases (7.4 cases per 100,000) in 2011. Most human cases are found in the north and north east part of Kazakhstan in areas drained by the Irtysh River and it tributaries. A further focus is found in the north west in the Ural river basin in the European part of Kazakhstan. The most common occupations of patients with opisthorchiidosis included the manual laborers, those employed in the home or unemployed. Necropsy investigations of village dogs in an endemic region revealed 37 of 51 (72%) village dogs infected with either Opisthorchis felineus or Methorchis bilis. Likewise an investigation of 242 cats consisting of strays, village, suburban and city cats revealed 79 (33%) animals infected with O. felineus. Higher prevalences were seen in strays and village cats compared to suburban cats. No urban cats, which lived in apartments, were found to be infected. Other important zoonoses included Echinococcus granulosus, detected in 2 of the 51 necropsied village dogs and E. multilocularis was found in 2 out of 124 necropsied stray cats. Investigations of locally caught fish revealed 10 of 107 (9%) roach (Rutilus rutilus), 49 of 68(72%) ide (Leuciscus idus) and 2 of 79 (2.5%) bream (Abramis brama) infested with trematode metacercariae. No metacercariae were found in 609 crucian carp (Carassius carassius), 35 tench (Tinca tinca), 79 carp (Cyprinus carpio), 46 perch (Perca fluviatilis) or 20 zander (Sander lucioperca).     Clonorchis sinensis (family Opisthorchiidae) is an important foodborne parasite that has a major socioeconomic impact on ~35 million people predominantly in China, Vietnam, Korea and the Russian Far East. In humans, infection with C. sinensis causes clonorchiasis, a complex hepatobiliary disease that can induce cholangiocarcinoma (CCA), a malignant cancer of the bile ducts. Central to understanding the epidemiology of this disease is knowledge of genetic variation within and among populations of this parasite. Although most published molecular studies seem to suggest that C. sinensis represents a single species, evidence of karyotypic variation within C. sinensis and cryptic species within a related opisthorchiid fluke (Opisthorchis viverrini) emphasise the importance of studying and comparing the genes and genomes of geographically distinct isolates of C. sinensis. Recently, we sequenced, assembled and characterised a draft nuclear genome of a C. sinensis isolate from Korea and compared it with a published draft genome of a Chinese isolate of this species using a bioinformatic workflow established for comparing draft genome assemblies and their gene annotations. We identified that 50.6% and 51.3% of the Korean and Chinese C. sinensis genomic scaffolds were syntenic, respectively. Within aligned syntenic blocks, the genomes had a high level of nucleotide identity (99.1%) and encoded 15 variable proteins likely to be involved in diverse biological processes. Here, we review current technical challenges of using draft genome assemblies to undertake comparative genomic analyses to quantify genetic variation between isolates of the same species. Using a workflow that overcomes these challenges, we report on a high-quality draft genome for C. sinensis from Korea and comparative genomic analyses, as a basis for future investigations of the genetic structures of C. sinensis populations, and discuss the biotechnological implications of these explorations.     Full Text Available Differentiation of the fish-borne trematodes belonging to the Opisthorchiidae, Heterophyidae and Lecithodendriidae is important from a clinical and epidemiological perspective, yet it is impossible to do using conventional coprological techniques, as the eggs are morphologically similar. Epidemiological investigation therefore currently relies on morphological examination of adult worms following expulsion chemotherapy. A PCR test capable of amplifying a segment of the internal transcribed spacer region of ribosomal DNA for the opisthorchiid and heterophyid flukes eggs taken directly from faeces was developed and evaluated in a rural community in central Thailand. The lowest quantity of DNA that could be amplified from individual adults of Opisthorchis viverrini, Clonorchis sinensis and Haplorchis taichui was estimated at 0.6 pg, 0.8 pg and 3 pg, respectively. The PCR was capable of detecting mixed infection with the aforementioned species of flukes under experimental conditions. A total of 11.6% of individuals in rural communities in Sanamchaikaet district, central Thailand, were positive for 'Opisthorchis-like' eggs in their faeces using conventional parasitological detection techniques. In comparison to microscopy, the PCR yielded a sensitivity and specificity of 71.0% and 76.7%, respectively. Analysis of the microscopy-positive PCR products revealed 64% and 23% of individuals to be infected with O. viverrini and C. sinensis, respectively. The remaining 13% (three individuals were identified as eggs of Didymozoidae, presumably being passed mechanically in the faeces following the ingestion of infected fishes. An immediate finding of this study is the identification and first report of a C. sinensis-endemic community in central Thailand. This extends the known range of this liver fluke in Southeast Asia.   14. First report and molecular identification of Opisthorchis viverrini infection in human communities from Lower Myanmar.

    Opisthorchis viverrini is endemic in the South East Asian region, especially in Cambodia, Lao People's Democratic Republic, Vietnam and Thailand, but there have been no previous records from Myanmar. During stool surveys of rural populations in three regions of Lower Myanmar, Opisthorchis-like eggs were found in 34 out of 364 (9.3%) participants by stool microscopy after using the modified formalin-ether concentration technique. DNA was extracted from these positive stool samples and a portion of the mitochondrial cytochrome c oxidase subunit I (cox1) gene was amplified using the polymerase chain reaction and then sequenced. DNA sequences, successfully obtained from 18 of 34 positive samples (Bago Region, n = 13; Mon State, n = 3; Yangon Region, n = 2), confirmed that the eggs were of O. viverrini. Sequences showed 99.7% identity with O. viverrini mitochondrial cox1 (GenBank accession no. JF739555) but 95%, 88.7%, 82.6% and 81.4% identities with those of Opisthorchis lobatus from Lao People's Democratic Republic (GenBank accession nos. HQ328539-HQ328541), Metorchis orientalis from China (KT239342), Clonorchis sinensis from China (JF729303) and Opisthorchis felineus from Russia (EU921260), respectively. When alignement with other Opisthorchiidae trematodes, 81% similarity with Metorchis bilis from Czech Republic (GenBank accession nos. KT740966, KT740969, KT740970) and Slovakia (GenBank accession nos. KT740971-KT740973), 84.6% similarity with Metorchis xanthosomus from Czech Republic (GenBank accession no. KT740974), 78.6% similarity with M. xanthosomus from Poland (GenBank accession no. KT740968) and 82.2% similarity with Euamphimerus pancreaticus from Czech Republic (GenBank accession no. KT740975) were revealed. This study demonstrated, for the first time, O. viverrini from rural people in Myanmar using molecular methods and is an urgent call for surveillance and control activities against opisthorchiasis in Myanmar.

    The Floristic and structural characterization of the main forest types in the Taraira Sierra, it was carried out to give to know the forests that exist there, since there were not preliminary studies on this ecosystem, and to enlarge the knowledge on the forests of the Colombian Guayana. For the composition study and it structures of the vegetation, three parcels of study of 0.05 hectares settled down, 10m x 50m each one; two in high Catinga and one in the Arbustiva; likewise they were traveled by different places of the Sierra to collect vegetable material. According to the floristic composition, to the covering analysis, and to the Sorensen index (Cordoba, 1995) they were defined three communities, the first of them in Catinga arbustiva, Clusia spathulaefolia - Retiniphyllum truncatum that is characterized to reach heights until of 10 meters, on sandy soils with inferior slope to 7%. In the high Catinga they were two communities, Micrandra sprucei - amazon Poecilanthe and Micrandra sprucei - Tovomita clarkii, which are characterized to present a superior stratum that reaches the 20 meters high.     Full Text Available Centrocestus formosanus es un parásito trematodo zoonótico originario de Asia asociado con muertes de peces principalmente de cultivo. 907 moluscos provenientes de estanques sembrados con tilapias, seleccionados uno por provincia fueron identificados al nivel taxonómico especifico. Se identificaron cuatro gastrópodos y un bivalvo: M. tuberculata, M. turricula, P. flagellata, H. cubensis y A. luteola. Se reporta, por primera vez, la presencia de dos especies de moluscos en Costa Rica. Se identificaron siete morfotipos de cercarias parasitando las cinco especies de moluscos encontradas. En la segunda exposición experimental se demostró que el morfotipo parapleurolofocercus encontrado en M. tuberculata concuerda con el hallazgo de C. formosanus en alevines de tilapia, después del examen clínico, anatomopatológico y parasitológico realizado a los alevines expuestos. Las metacercarias fueron extraídas del quiste utilizando microagujas y micropinzas lavadas en solución salina fisiológica (0.65%, fijadas en formol caliente al 4% y después esquematizadas con una cámara clara adaptada a un microscopio fotónico, estimándose una abundancia e intensidad media de 1018-1027 digeneos por branquia en cada pez parasitado, determinándose así el hospedador intermediario primario y secundario del parásito. En el presente trabajo se reporta por primera vez Centrocestus formosanus en Costa Rica.

    Full Text Available Opisthorchis felineus, O. viverrini, and Clonorchis sinensis (family Opisthorchiidae are parasitic flatworms that pose a serious threat to humans in some countries and cause opisthorchiasis/clonorchiasis. Chronic disease may lead to a risk of carcinogenesis in the biliary ducts. MicroRNAs (miRNAs are small noncoding RNAs that control gene expression at post-transcriptional level and are implicated in the regulation of various cellular processes during the parasite- host interplay. However, to date, the miRNAs of opisthorchiid flukes, in particular those essential for maintaining their complex biology and parasitic mode of existence, have not been satisfactorily described.Using a SOLiD deep sequencing-bioinformatic approach, we identified 43 novel and 18 conserved miRNAs for O. felineus (miracidia, metacercariae and adult worms, 20 novel and 16 conserved miRNAs for O. viverrini (adult worms, and 33 novel and 18 conserved miRNAs for C. sinensis (adult worms. The analysis of the data revealed differences in the expression level of conserved miRNAs among the three species and among three the developmental stages of O. felineus. Analysis of miRNA genes revealed two gene clusters, one cluster-like region and one intronic miRNA in the genome. The presence and structure of the two gene clusters were validated using a PCR-based approach in the three flukes.This study represents a comprehensive description of miRNAs in three members of the family Opistorchiidae, significantly expands our knowledge of miRNAs in multicellular parasites and provides a basis for understanding the structural and functional evolution of miRNAs in these metazoan parasites. Results of this study also provides novel resources for deeper understanding the complex parasite biology, for further research on the pathogenesis and molecular events of disease induced by the liver flukes.   18. Preliminary survey of ticks (Acari : Ixodidae on cattle in northern Sudan

    Full Text Available In a cross sectional survey conducted during the period June 2001 to July 2002, the geographical distribution of ticks on cattle in the Sudan was determined. Seventeen locations were surveyed from Northern, Central, Eastern, Western, Blue Nile and White Nile Provinces. Total body collections of ticks were made from 20 cattle at each location. Four tick genera and 11 species were identified. The tick species collected included Amblyomma lepidum, Amblyomma variegatum, Boophilus decoloratus, Hyalomma anatolicum anatolicum, Hyalomma dromedarii, Hyalomma impeltatum, Hyalomma marginatum rufipes, Hyalomma truncatum, Rhipicephalus evertsi evertsi, Rhipicephalus sanguineus group and Rhipicephalus simus simus. Major ecological changes have occurred due to extensive animal movement, deforestation, desertification and establishment of large mechanized agricultural schemes. These factors have certainly affected the distribution of ticks and tick-borne diseases in the Sudan. The absence of A. variegatum and A. lepidum in northern Sudan was not surprising, since these tick species are known to survive in humid areas and not in the desert and semi-desert areas of northern Sudan. The absence of B. annulatus in northern and central Sudan is in accordance with the finding that this tick species is restricted to the southern parts of the central Sudan. The presence of H. anatolicum anatolicum in Um Benin in relatively high abundance is an interesting finding. The present finding may indicate that the southern limit of this species has changed and moved southwards to latitude 13o N.   19. Ixodid ticks on cattle belonging to small-scale farmers at 4 communal grazing areas in South Africa

    Full Text Available Ixodid ticks were collected during the period September 1991 to August 1993 from cattle belonging to small-scale farmers utilising 4 communal grazing areas. Three of these were in North West Province and 1 in Mpumalanga province, South Africa. Ten tick species were collected in North West Province and 7 in Mpumalanga. The adults of Amblyomma hebraeum, Rhipicephalus appendiculatus and Rhipicephalus evertsi evertsi were most numerous in North West Province, while in Mpumalanga Boophilus decoloratus comprised more than 75% of the total population. Amblyomma hebraeum was present on all grazing areas, and heavy infestations of adults occurred during the period October to May on 1 of these. Few B. decoloratus were collected in North West Province, chiefly because the sampling method was inadequate, and most of these were present during early summer (October to December and late summer and autumn (March to May. The initially low population of B. decoloratus in Mpumalanga increased substantially towards the conclusion of the survey, probably because of the cessation of dipping. Boophilus microplus was present in small numbers on 2 grazing areas in the North West Province. Adult Hyalomma marginatum rufipes reached peak numbers from December to February and Hyalomma truncatum from February to April in the North West Province. Only H. marginatum rufipes was collected in Mpumalanga. Rhipicephalus appendiculatus was present on all the grazing areas, with most adults present from December to April. Most adult Rhipicephalus evertsi evertsi were collected from September to April and Rhipicephalus simus was present during the period October-April.

  20. Relationship between Leaf Surface Characteristics and Particle Capturing Capacities of Different Tree Species in Beijing

    Full Text Available Leaf surface is a multifunctional interface between a plant and its environment, which affects both ecological and biological processes. Leaf surface topography directly affects microhabitat availability and ability for deposition. In this study, atomic force microscopy (AFM and the resuspended particulate matter method were applied to evaluate the adsorptive capacity of the leaf surface. Patterns of particulate‐capturing capacities in different tree species and the effect of leaf surface features on these capacities were explored. Results indicated the following: (1 more total suspended particles (TSP per unit leaf area were captured by coniferous tree species than by broad‐leaved tree species in a particular order—i.e., Pinus tabuliformis > Pinus bungeana > Salix matsudana > Acer truncatum > Ginkgo biloba > Populus tomentosa; (2 Significant seasonal variation in particulate‐capturing capacities were determined. During the observation period, the broad‐leaved tree species capturing TSP and coarse particulate matter (PM10 clearly exhibited a ∩‐shape pattern— that is, increasing initially and later on decreasing; meanwhile, the ∩‐shape pattern was not clearly shown in P. tabuliformis and P. bungeana. However, no obvious patterns in the absorption of fine particulate matter (PM2.5 were found in the tested tree species; (3 The leaf surface topography, as observed by AFM and scanning electron microscopy, revealed that the broad‐leaved tree exhibits a good correlation between micro‐roughness of leaf surfaces and density of particles settling on leaf surfaces over time. However, the main factors affecting the adsorptive capacities of the leaves in coniferous trees are the number of stomata as well as the amount of epicuticular wax and the properties of the cuticle in different seasons.

    Full Text Available Soybean is one of the main crop in the world, because is a source of food products, for animals and humans, besides its use as biofuels. Although new technologies have been used in this crop, its productivity has been limited mainly by the occurrence of diseases, such as anthracnose (Colletotrichum truncatum and target spot (Corynespora cassiicola. Thus, the aim of this work was to evaluate the efficiency of fungicides for the control of anthracnose and target spot, applied to the leaves, in a commercial area in the north of Mato Grosso State, Brazil. The experimental design was randomized blocks with eight treatments and three repetitions. The treatments were: T1- Control; T2- 1st Application: Pyraclostrobin + Epoxiconazole, 2nd Application: fluxapyroxad + Pyraclostrobin; T3- 1st Application: Azoxystrobin, 2nd Application: Azoxystrobin; T4- 1st Application: Pyraclostrobin, 2nd Application: Pyraclostrobin + Epoxiconazol; T5- 1st Application: Fluxapyroxad + Pyraclostrobin, 2nd Application: Fluxapyroxad + Pyraclostrobin; T6- 1st Application: Trifloxystrobin + Prothioconazol, 2nd Application: Trifloxystrobin + Prothioconazol; T7- 1st Application: Picoxystrobin + Cyproconazol, 2nd Application: Picoxystrobin + Cyproconazol; T8- 1st Application: Tebuconazol, 2nd Application: Tebuconazol. The first application of fungicides was at soybean flowering and the second, 15 days after the first application.     Full Text Available A cross-sectional study was conducted to determine the prevalence and risk factors for ectoparasites infestation in sheep in three agro-ecological zones in central Oromia, Ethiopia, from October 2009 to April 2010. The study revealed that 637 (48.1% of the 1325 sheep examined were infested with one or more ectoparasites. The ectoparasites identified were Bovicola ovis (27.2%, Melophagus ovinus (16.4%, Ctenocephalides sp. (2.3%, Linognathus africanus (1.2%, Linognathus ovillus (0.3%, Sarcoptes sp. (1.2%, Amblyomma variegatum (4.4%, Rhipicephalus evertsi evertsi (1.9%, Rhipicephalus pravus (1.9%, Rhipicephalus (Boophilus decoloratus (1.1%, Rhipicephalus sanguineus (0.9%, Rhipicephalus praetextatus (1.1% and Hyalomma truncatum (1.6%. Statistically significant difference was observed in prevalence of B. ovis amongst study agroecological zones: highland 36.6%, midland 20.9% and lowland 14.0%. Significantly higher prevalence was recorded in highland agroecological zone. A significantly (OR = 0.041, p  0.05 was never recorded in the prevalence of all the identified species of ectoparasites between male and female sheep hosts. However, a significantly (p = 0.006 higher prevalence of B. ovis was recorded between young and adult sheep. The risk of B. ovis infestation was 1.45 times higher in young than the adult sheep. Furthermore, a significantly (p < 0.001 higher prevalence of M. ovinus, B. ovis and Sarcoptes sp. was found between sheep with poor and a good body condition. The ever increasing threat of ectoparasites on overall sheep productivity and tanning industry in Ethiopia warrants urgent control intervention. Further studies on the role of ectoparasites in transmission of diseases to sheep, zoonotic importance, comparative prevalence and load, and the importance of sheep as alternative hosts in different agroecological zones, breeds and management systems in Ethiopia are recommended so as to design applicable control programme in the country.

  3. How to help woody plants to overcome drought stress?-a control study of four tree species in Northwest China.

    Water is essential for plants and involves most physical and chemical processes within their lifecycles. Drought stress is a crucial limiting factor for plant growth and production. 48% of the land in China is arid and semi-arid, and non-irrigated land occupies approximately 51.9% of the total cultivated areas. Therefore, studies on plant drought resistant mechanisms have great significance for improving water use efficiency and thus increasing productivity of economical plants. Prior research has shown that the application of nitrogenous fertilizer affects the drought-resistant characteristics of plants. This study aimed to reveal the effect of nitrogenous fertilizer on physiological aspects and its impact on the drought resistance of four tree species (Robinia pseudoacacia L., Ligustrum lucidum Ait., Acer truncatum Bge. and Ulmus pumila L. ) in northwest China. Three levels of nitrogen fertilization (46% N based of urea adjusted to: 5g/15g soil, 15g/15g soil and 25g/15g soil) and an additional control study were applied to 2-year-old well-grown seedlings under drought conditions (30% field moisture capacity). Stomatal conductance, transpiration rate and net photosynthetic rate were measured by a LI-6400 photosynthesis system, while water use efficiency was calculated from net photosynthesis rate and transpiration rate. The results revealed that as the amount of urea applied was raised, stomatal conductance, transpiration rate and net photosynthetic rate decreased significantly, and thus water use efficiency significantly increased. It is therefore concluded that the application of nitrogenous fertilizer regulated physiological parameters by reducing stomata conductance to improve water use efficiency. In addition, among the four tree species, U. pumila had the maximum value of water use efficiency under the same drought condition. The outcome of this study provides a guided option for forest management in arid and semi-arid areas of northwest China.

  4. Prevalence of Theileria annulata in dairy cattle in Nyala, South Darfur State, Sudan

    Full Text Available Aim: This study was conducted in dairy cattle in Nyala, South Darfur State, during the period from June to September 2015, to study the prevalence of bovine tropical theileriosis. Materials and Methods: Apparently, healthy cattle of different age groups, different breeds, and from both sexes were randomly selected from seven locations. Three age groups of cattle were selected, group one <1 year old, group two 1-3 years old, and group three older than 3 years. These cattle were indigenous and cross (Friesian X zebu. A total of 150 blood samples were collected for blood smears, blood in EDTA tubes, and serum samples as well as ticks infesting cattle. Three diagnostic techniques were used such as blood smear, indirect fluorescent antibody test (IFAT, and polymerase chain reaction (PCR. Results: Of 150 samples, 11 (7.3%, 95% confidence interval [CI]: 9.1-5.5 were positive for Theileria spp. piroplasms in the blood smears, 70 (46.7%, 95% CI: 35.7-57.7 were positive for Theileria annulata antibodies in the IFAT, and of 100 samples, 39 (39%, 95% CI: 46.6-31.4 were positive for T. annulata using PCR. The prevalence of T. annulata was higher in indigenous breed than cross cattle by the three diagnostic techniques. The highest prevalence of T. annulata was recorded among cattle older than 3 years old. There were three genera and ten species of ticks found feeding on cattle. These were Rhipicephalus evertsi evertsi, Rhipicephalus decoloratus, Rhipicephalus annulatus, Hyalomma dromedrii, Hyalomma impeltatum, Hyalomma rufipes, Hyalomma anatolicum, Hyalomma truncatum, Amblyomma variegatum, and Amblyomma lepidum. Conclusion: The study concluded that tropical theileriosis is prevalent among dairy cattle in Nyala. H. anatolicum was found in very low numbers, suggesting other ticks may play a role in the transmission of the disease.     Hyalomma Koch, 1844 are ixodid ticks that infest mammals, birds and reptiles, to which 27 recognized species occur across the Afrotropical, Palearctic and Oriental regions. Despite their medical and veterinary importance, the evolutionary history of the group is enigmatic. To investigate various taxonomic hypotheses based on morphology, and also some of the mechanisms involved in the diversification of the genus, we sequenced and analysed data derived from two mtDNA fragments, three nuclear DNA genes and 47 morphological characters. Bayesian and Parsimony analyses based on the combined data (2242 characters for 84 taxa) provided maximum resolution and strongly supported the monophyly of Hyalomma and the subgenus Euhyalomma Filippova, 1984 (including H. punt Hoogstraal, Kaiser and Pedersen, 1969). A predicted close evolutionary association was found between morphologically similar H. dromedarii Koch, 1844, H. somalicum Tonelli Rondelli, 1935, H. impeltatum Schulze and Schlottke, 1929 and H. punt, and together they form a sister lineage to H. asiaticum Schulze and Schlottke, 1929, H. schulzei Olenev, 1931 and H. scupense Schulze, 1919. Congruent with morphological suggestions, H. anatolicum Koch, 1844, H. excavatum Koch, 1844 and H. lusitanicum Koch, 1844 form a clade and so also H. glabrum Delpy, 1949, H. marginatum Koch, 1844, H. turanicum Pomerantzev, 1946 and H. rufipes Koch, 1844. Wide scale continental sampling revealed cryptic divergences within African H. truncatum Koch, 1844 and H. rufipes and suggested that the taxonomy of these lineages is in need of a revision. The most basal lineages in Hyalomma represent taxa currently confined to Eurasia and molecular clock estimates suggest that members of the genus started to diverge approximately 36.25 million years ago (Mya).     Full Text Available A cross-sectional study was conducted to determine the prevalence and risk factors for ectoparasites infestation in sheep in three agro-ecological zones in central Oromia, Ethiopia, from October 2009 to April 2010. The study revealed that 637 (48.1% of the 1325 sheep examined were infested with one or more ectoparasites. The ectoparasites identified were Bovicola ovis (27.2%, Melophagus ovinus (16.4%, Ctenocephalides sp. (2.3%, Linognathus africanus (1.2%, Linognathus ovillus (0.3%, Sarcoptes sp.
