  1. Pigmentiphaga aceris sp. nov., isolated from tree sap.

    Two Gram-stain-negative bacterial strains, SAP-32T and SAP-36, were isolated from sap drawn from the Acer pictum from Mount Halla in Jeju, Republic of Korea. The organisms were strictly aerobic, non-sporulating, motile rods and showed growth at 10-30 °C, pH 7-8 and with 0-2 % NaCl. The major isoprenoid quinone was Q-8. The predominant fatty acids were C16 : 0, cyclo-C17 : 0, summed feature 3 and C18 : 0. The polar lipids contained phosphatidylcholine, phosphatidylethanolamine, phosphatidylglycerol, an unknown aminophosphoglycolipid, an unknown glycolipid, an unknown phospholipid and two unknown lipids. The DNA G+C content was 64.4 mol%. The results of phylogenetic analyses based on 16S rRNA gene sequences indicated that SAP-32T and SAP-36 formed a distinct cluster with members of the genus Pigmentiphaga within the family Alcaligenaceae. Both strains showed 16S rRNA gene sequence similarity of 100 % to each other. The closest relatives of the isolates were Pigmentiphaga daeguensis (97.08 % sequence similarity), Pigmentiphaga kullae (97.01 %) and Pigmentiphaga litoralis (96.73 %). On the basis of data from phenotypic, chemotaxonomic and phylogenetic analyses, SAP-32T (=KCTC 52619T=DSM 104039T) and SAP-36 (=KCTC 52620=DSM 104072) represent members of a novel species of the genus Pigmentiphaga, for which the name Pigmentiphaga aceris sp. nov. is proposed.

  3. Purification, cloning, expression, and biochemical characterization of a monofunctional catalase, KatP, from Pigmentiphaga sp. DL-8.

    Catalases are essential components of the cellular equipment used to cope with oxidative stress. The monofunctional catalase KatP was purified from Pigmentiphaga sp. using ammonium sulfate precipitation (ASP), diethylaminoethyl ion exchange chromatography (IEC), and hydrophobic interaction chromatography (HIC). The purified catalase formed polymer with an estimated monomer molecular mass of 54kDa, which were resolved by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and zymogram analysis. KatP exhibited a specific catalytic activity of 73,000U/mg, which was higher than that of catalase-1 of Comamonas terrigena N3H (55,900U/mg). Seven short tryptic fragments of this catalase were obtained by electrospray ionization quadrupole time-of-flight mass spectrometry (ESI-Q-TOF MS/MS), and the gene, katP, was cloned by PCR amplification and overexpressed in Escherichia coli BL21 (DE3). Based on the complete amino acid sequence, KatP was identified as a clade 3 monofunctional catalase. The specific activities of recombinant KatP for hydrogen peroxide (690,000U/mg) increased 9-fold over that of the parent strain. The Km and Vmax of recombinant KatP were 9.48mM and 81.2mol/minmg, respectively. The optimal pH and temperature for KatP were 7.0 and 37°C, respectively, and the enzyme displayed abroad pH-stable range of 4.0-11.0. The enzyme was inhibited by Zn(2+), Cu(2+), Cr(2+), and Mn(2+), whereas Fe(3+) and Mg(2+) stimulated KatP enzymatic activity. Interestingly, the catalase activity of recombinant KatP displayed high stability under different temperature and pH conditions, suggesting that KatP is a potential candidate for the production of catalase.     Full Text Available ‘‘To distinguish true from false love: Manly and unmanly bodies in Martha Sandwall-Bergström’s Kulla-Gulla series.’’ The protagonist Gulla in Martha Sandwall-Bergström’s (1913-2000 Kulla-Gulla series (1945-51 encounters ’’true’’ and ’’false’’ love through two men, Tomas Tomasson and Ivan Malma. The hypothesis of this article is that true love is placed in a manly body and false love in an unmanly body. Tomas’ and Ivan’s manliness and unmanliness are constructed through characteristics which are marked by different power categories, such as gender, age, class, ‘‘race’’ and functionality. Therefore an intersectional perspective is used. I find that Tomas and Ivan have similar social positions except for their different classes. Tomas’ body is portrayed as manly, mobile and warm, whereas Ivan’s body is characterized as unmanly, immobile and cold. Tomas evokes love in Gulla, whereas Ivan arouses lust as well as ambivalence. Tomas’ manly body functions as a signal for true love and guides the reader as well as Gulla in distinguishing his true from Ivan’s false love. When Gulla chooses Tomas, she is defined as an adult.

  5. Experimental investigation on the spiral trepanning of K24 superalloy with femtosecond laser

    Film cooling holes are crucial for improving the performance of the aviation engine. In the paper, the processing of the film cooling holes on K24 superalloy by femtosecond laser is investigated. By comparing the three different drilling methods, the spiral trepanning method is chosen, and all the drilling experiments are carried out in this way. The experimental results show that the drilling of femtosecond laser pulses has distinct merits against that of the traditional long pulse laser, which can realize the "cold" processing with less recasting layer and less crack. The influence of each process parameter on roundness and taper, which are the important parameters to measure the quality of holes, is analyzed in detail, and the method to decrease it is proposed. To further reduce the recasting layer, the processing quality of the inner wall of the micro hole is investigated by scanning electron microscopy (SEM) equipped with energy disperse spectroscopy (EDS), the mechanism of the femtosecond laser interaction with K24 superalloy is further revealed. The investigation to the film hole machining by femtosecond laser has important practical significance.

    Full Text Available Burkholderia sp. K24, formerly known as Acinetobacter lwoffii K24, is a soil bacterium capable of utilizing aniline as its sole carbon and nitrogen source. Genomic sequence analysis revealed that this bacterium possesses putative gene clusters for biodegradation of various monocyclic aromatic hydrocarbons (MAHs, including benzene, toluene, and xylene (BTX, as well as aniline. We verified the proposed MAH biodegradation pathways by dioxygenase activity assays, RT-PCR, and LC/MS-based quantitative proteomic analyses. This proteogenomic approach revealed four independent degradation pathways, all converging into the citric acid cycle. Aniline and p-hydroxybenzoate degradation pathways converged into the β-ketoadipate pathway. Benzoate and toluene were degraded through the benzoyl-CoA degradation pathway. The xylene isomers, i.e., o-, m-, and p-xylene, were degraded via the extradiol cleavage pathways. Salicylate was degraded through the gentisate degradation pathway. Our results show that Burkholderia sp. K24 possesses versatile biodegradation pathways, which may be employed for efficient bioremediation of aniline and BTX.

  10. Isolation and Identification of Sodium Fluoroacetate Degrading Bacteria from Caprine Rumen in Brazil

    Full Text Available The objective of this paper was to report the isolation of two fluoroacetate degrading bacteria from the rumen of goats. The animals were adult goats, males, crossbred, with rumen fistula, fed with hay, and native pasture. The rumen fluid was obtained through the rumen fistula and immediately was inoculated 100 μL in mineral medium added with 20 mmol L−1 sodium fluoroacetate (SF, incubated at 39°C in an orbital shaker. Pseudomonas fluorescens (strain DSM 8341 was used as positive control for fluoroacetate dehalogenase activity. Two isolates were identified by 16S rRNA gene sequencing as Pigmentiphaga kullae (ECPB08 and Ancylobacter dichloromethanicus (ECPB09. These bacteria degraded sodium fluoroacetate, releasing 20 mmol L−1 of fluoride ion after 32 hours of incubation in Brunner medium containing 20 mmol L−1 of SF. There are no previous reports of fluoroacetate dehalogenase activity for P. kullae and A. dichloromethanicus. Control measures to prevent plant intoxication, including use of fences, herbicides, or other methods of eliminating poisonous plants, have been unsuccessful to avoid poisoning by fluoroacetate containing plants in Brazil. In this way, P. kullae and A. dichloromethanicus may be used to colonize the rumen of susceptible animals to avoid intoxication by fluoroacetate containing plants.

    Allergic diseases belong to one of the most common diseases with steadily increasing incidence even among young children. There is an urgent need to identify a prognostic marker pointing to increased risk of allergy development enabling early preventive measures introduction. It has been shown that administration of selected probiotic strains or mixtures could prevent allergy development. In our study, we have tested the capacity of probiotic strain Escherichia coli O83:K24:H31 (E. coli O83) to promote dendritic cell (DC) maturation and polarisation of immune responses. Increased presence of activation marker CD83 was observed on DC stimulated by E. coli O83 and DC of newborns of allergic mothers have significantly more increased cell surface presence of CD83 in comparison to children of healthy mothers. Increased gene expression and secretion of IL-10 was detected in DC stimulated with E. coli O83 being higher in DC of newborns of healthy mothers in comparison to allergic ones. Generally, increased presence of intracellular cytokines (IL-4, IL-13, IFN-gamma, IL-17A, IL-22, IL-10) was detected in CD4+ T cells cocultured with DC of children of allergic mothers in comparison to healthy ones. E. coli O83 primed DC significantly increased IL-10 and IL-17A in CD4 T cells of newborns of healthy mothers in comparison to the levels detected in CD4 T cells cocultured with control non-stimulated DC. We can conclude E. coli O83 induces dendritic cell maturation and IL-10 production in DC. Newborns of allergic mothers have generally increased reactivity of both DC and CD4 T cells which together with decreased capacity of DC of newborns of allergic mothers to produce IL-10 could support inappropriate immune responses development after allergen encounter.     In accordance with DOE Regulations for Compliance with Floodplain/Wetlands Environmental Review Requirements, wetland surveys were conducted in selected areas within the K-25 Area of Responsibility during the summer of 1994. These areas are Mitchell Branch, Poplar Creek, the K-770 OU, Duct Island Peninsula, the Powerhouse area, and the K-25 South Corner. Previously surveyed areas included in this report are the main plant area of the K-25 Site, the K-901 OU, the AVLIS site, and the K-25 South Site. Wetland determinations were based on the USACE methodology. Forty-four separate wetland areas, ranging in size from 0.13 to 4.23 ha, were identified. Wetlands were identified in all of the areas surveyed with the exception of the interior of the Duct Island Peninsula and the main plant area of the K-25 Site. Wetlands perform functions such as floodflow alteration, sediment stabilization, sediment and toxicant retention, nutrient transformation, production export, and support of aquatic species and wildlife diversity and abundance. The forested, scrub-shrub, and emergent wetlands identified in the K-25 area perform some or all of these functions to varying degrees.

    The whole-genome sequence of the multiply antibiotic resistant Acinetobacter baumannii isolate RCH51 belonging to sequence type ST103 (Institut Pasteur scheme) revealed that the set of genes at the capsule locus, KL24, includes four genes predicted to direct the synthesis of 3-acetamido-3,6-dideoxy-d-galactose (d-Fuc3NAc), and this sugar was found in the capsular polysaccharide (CPS). One of these genes, fdtE, encodes a novel bifunctional protein with an N-terminal FdtA 3,4-ketoisomerase domain and a C-terminal acetyltransferase domain. KL24 lacks a gene encoding a Wzy polymerase to link the oligosaccharide K units to form the CPS found associated with isolate RCH51, and a wzy gene was found in a small genomic island (GI) near the cpn60 gene. This GI is in precisely the same location as another GI carrying wzy and atr genes recently found in several A. baumannii isolates, but it does not otherwise resemble it. The CPS isolated from RCH51, studied by sugar analysis and 1D and 2D 1H and 13C NMR spectroscopy, revealed that the K unit has a branched pentasaccharide structure made up of Gal, GalNAc and GlcNAc residues with d-Fuc3NAc as a side branch, and the K units are linked via a β-d-GlcpNAc-(1→3)-β-d-Galp linkage formed by the Wzy encoded by the GI. The functions of the glycosyltransferases encoded by KL24 were assigned to formation of specific bonds. A correspondence between the order of the genes in KL24 and other KL and the order of the linkages they form was noted, and this may be useful in future predictions of glycosyltransferase specificities.

    Full Text Available Pine wilt disease (PWD is native to North America and has spread to Asia and Europe. Lately, mutualistic relationship has been suggested between the pinewood nematode (PWN, Bursaphelenchus xylophilus the causal nematode agent of PWD, and bacteria. In countries where PWN occurs, nematodes from diseased trees were reported to carry bacteria from several genera. However no data exists for the United States. The objective of this study was to evaluate the diversity of the bacterial community carried by B. xylophilus, isolated from different Pinus spp. with PWD in Nebraska, United States. The bacteria carried by PWN belonged to Gammaproteobacteria (79.9%, Betaproteobacteria (11.7%, Bacilli (5.0%, Alphaproteobacteria (1.7% and Flavobacteriia (1.7%. Strains from the genera Chryseobacterium and Pigmentiphaga were found associated with the nematode for the first time. These results were compared to results from similar studies conducted from other countries of three continents in order to assess the diversity of bacteria with associated with PWN. The isolates from the United States, Portugal and China belonged to 25 different genera and only strains from the genus Pseudomonas were found in nematodes from all countries. The strains from China were closely related to P. fluorescens and the strains isolated from Portugal and USA were phylogenetically related to P. mohnii and P. lutea. Nematodes from the different countries are associated with bacteria of different species, not supporting a relationship between PWN with a particular bacterial species. Moreover, the diversity of the bacteria carried by the pinewood nematode seems to be related to the geographic area and the Pinus species. The roles these bacteria play within the pine trees or when associated with the nematodes, might be independent of the presence of the nematode in the tree and only related on the bacteria's relationship with the tree.

    A Gram-negative, short rod-shaped bacterial strain, YC06271T, was isolated from the vermicompost (VC) collected at Masan, Korea and its taxonomic position was investigated by a polyphasic taxonomic approach. Strain YC06271T grew optimally at 28-30 degrees C and at pH 7.0-9.0. The 16S rRNA gene sequence of strain YC06271T was most closely related to members of the genera Bordetella (96.4-95.8 %), Achromobacter (96.0-95.7 %), Alcaligenes (96.0-94.2 %), Pusillimonas noertemannii (95.9 %), Pigmentiphaga (95.8-95.5 %) and less than 95.5 % similarity with the members of the other genera of the family Alcaligenaceae. Strain YC06271T contained ubiquinone-8 (Q-8) as the major respiratory quinone system and putrescine as the major polyamine. The major fatty acids of strain YC06271T were C16:1omega7c and/or C15:0 iso 2-OH and C16:0. The major polar lipids were phosphatidylglycerol and phosphatidylethanolamine. The G+C content of the genomic DNA was 55.4 mol%. Phylogenetic analysis, biochemical, chemotaxonomic and phenotypic characteristics strongly supported the differentiation of strain YC06271T from the validly published genera of the family Alcaligenaceae. Therefore, it is proposed that strain YC06271T represents a novel species within a novel genus, with the name Eiseniicola composti gen. nov., sp. nov. The type strain is YC06271T (= KCTC 22250T = DSM 21045T).

    Host-associated microbiotas of vertebrates are diverse and complex communities that contribute to host health. In particular, for amphibians, cutaneous microbial communities likely play a significant role in pathogen defense; however, our ecological understanding of these communities is still in its infancy. Here, we take advantage of the fully endemic and locally species-rich amphibian fauna of Madagascar to investigate the factors structuring amphibian skin microbiota on a large scale. Using amplicon-based sequencing, we evaluate how multiple host species traits and site factors affect host bacterial diversity and community structure. Madagascar is home to over 400 native frog species, all of which are endemic to the island; more than 100 different species are known to occur in sympatry within multiple rainforest sites. We intensively sampled frog skin bacterial communities, from over 800 amphibians from 89 species across 30 sites in Madagascar during three field visits, and found that skin bacterial communities differed strongly from those of the surrounding environment. Richness of bacterial operational taxonomic units (OTUs) and phylogenetic diversity differed among host ecomorphs, with arboreal frogs exhibiting lower richness and diversity than terrestrial and aquatic frogs. Host ecomorphology was the strongest factor influencing microbial community structure, with host phylogeny and site parameters (latitude and elevation) explaining less but significant portions of the observed variation. Correlation analysis and topological congruency analyses revealed little to no phylosymbiosis for amphibian skin microbiota. Despite the observed geographic variation and low phylosymbiosis, we found particular OTUs that were differentially abundant between particular ecomorphs.     Full Text Available Little is known about physiological factors that affect the sense of olfaction in dogs. The objectives of this study were to describe the canine nasal and oral microbiota in detection dogs. We sought to determine the bacterial composition of the nasal and oral microbiota of a diverse population of detection canines. Nasal and oral swabs were collected from healthy dogs (n = 81 from four locations-Alabama, Georgia, California, and Texas. Nasal and oral swabs were also collected from a second cohort of detection canines belonging to three different detection job categories: explosive detection dogs (SP-E; n = 22, patrol and narcotics detection dogs (P-NDD; n = 15, and vapor wake dogs (VWD-E; n = 9. To understand if the nasal and oral microbiota of detection canines were variable, sample collection was repeated after 7 weeks in a subset of dogs. DNA was extracted from the swabs and used for 454-pyrosequencing of the16S rRNA genes. Nasal samples had a significantly lower diversity than oral samples (P<0.01. Actinobacteria and Proteobacteria were higher in nasal samples, while Bacteroidetes, Firmicutes, Fusobacteria, and Tenericutes were higher in oral samples. Bacterial diversity was not significantly different based on the detection job. No significant difference in beta diversity was observed in the nasal samples based on the detection job.     Moessbauer spectroscopy was used to investigate the effect of heating the Fischer-Tropsch catalyst 100 Fe/5 Cu/4.2 K/24 SiO 2 in two different atmospheres while ramping the temperature of the catalyst from room temperature to 280 C in 5.5 h prior to pretreatment of the catalyst. Preheating in H 2 /CO = 0.7 gave rise to an iron (Fe 2+ ) silicate, while preheating in helium resulted in the formation of ε'-carbide Fe 2.2 C. Iron oxides and χ-carbide Fe 5 C 2 were also formed in both preheat treatments. (orig.)

    Calculations using multiblock computational technologies and a model of shear-stress transport modified with allowance for the curvature of streamlines in turbulent airflow were performed at a zero angle of attack for a semicircular airfoil containing one or two surface vortex cells with slot suction. The results showed evidence of stabilization of a nearly undetached flow and attainment of an extremal lift of C y = 5.2 and a lift-to-drag ratio of K = 24 with allowance for energy losses for suction in the vortex cells.

    In Digital Cinema, the video compression must be as transparent as possible to provide the best image quality to the audience. The goal of compression is to simplify transport, storing, distribution and projection of films. For all those tasks, equipments need to be developed. It is thus mandatory to reduce the complexity of the equipments by imposing limitations in the specifications. In this sense, the DCI has fixed the maximum bitrate for a compressed stream to 250 Mbps independently from the input format (4K/24fps, 2K/48fps or 2K/24fps). The work described in this paper This parameter is discussed in this paper because it is not consistent to double/quadruple the input rate without increasing the output rate. The work presented in this paper is intended to define quantization steps ensuring the visually lossless compression. Two steps are followed first to evaluate the effect of each subband separately and then to fin the scaling ratio. The obtained results show that it is necessary to increase the bitrate limit for cinema material in order to achieve the visually lossless.

    This paper describes a PC add on MCA card for nuclear spectroscopy applications. It consists of a 8K nuclear ADC, control logic and 8K * 24 bit dual ported histogram memory. The integral card assembly plugs into a single I/O expansion slot of the IBM-PC XT/AT or any compatible PC. This together with the data acquisition and processing software package, developed in Turbo C transforms the PC into a powerful Multichannel Analyzer. In the design of the MCA card, the PC interface, sequencers and control logic is implemented using programmable logic devices in order to achieve compact, efficient and reliable design. The dual ported 8K * 24 bit histogram memory is implemented using conventional static memory with external arbitration logic. Built-in ADC is designed around commercially available monolithic successive approximation type ADC chip. The low differential nonlinearity is achieved using an innovative interpolation technique along with Gatti sliding technique. The data acquisition and processing package developed in Turbo C, includes interrupt driven spectrum display driver and a pull down menu based user interface. Data processing features include spectrum smoothing, auto peak search and energy calibration. Separate memory buffers are used for data acquisition and data processing.     Full Text Available One of the most important criteria in breeding process of perennial grasses is maturity. Cultivars with different maturity play a very important role in utilization of perennial grasses, by providing the ability to create a mixture of different aspects utilization and time. The first grass species in Serbia whose breeding program involved this criterion was cocksfoot (Dactylis glomerata L.. In general cocksfoot is early to medium-early in maturity in comparison with other grasses and legumes, and that is mayor problem since in the optimum phase for cutting, cocksfoot is often earlier then other species in mixtures. As a result of this work, in the previous period, two cultivars of different maturity were released, Kruševačka 24 (K-24 and Kruševačka 25 (K-25. K-24 is medium and K-25 is late in maturity. New material is adapted to local agro-ecological conditions and productive in the same time. In breeding process of both cultivars initial material originated from autochthonous populations collected in eastern and central Serbia. Material from the wild flora is selected based on medium and late maturity which is already adapted and has good productivity. We applied the standard method of phenotypic recurrent selection with the creation of synthetic varieties by polycross.

    The present paper describes the development of a high throughput octal alpha spectrometry system specially developed for the estimation of low levels of actinides in bioassay and environmental samples. The system processes simultaneously the outputs coming from eight independent detectors. It can be configured to simultaneously record low level alpha and gamma spectra. The high throughput is achieved by using a prioritised multiplexer router. The prioritised multiplexing and routing coupled with fast 8K ADC (conversion time 20 μsec) allow simultaneous acquisition of multiple spectra without any significant loss in counts. The dual (8K, 24bit) port memory facilitates easy online viewing of spectrum buildup. A menu driven user friendly software makes the operating system convenient to use. A specially developed software provides built-in routines for processing the spectra and estimating the isotopic activity.   1. In Silico Measurement of Elastic Moduli of Nematic Liquid Crystals

    Experiments on confined droplets of the nematic liquid crystal 5CB have questioned long-established bounds imposed on the elastic free energy of nematic systems. This elasticity, which derives from molecular alignment within nematic systems, is quantified through a set of moduli which can be difficult to measure experimentally and, in some cases, can only be probed indirectly. This is particularly true of the surfacelike saddle-splay elastic term, for which the available experimental data indicate values on the cusp of stability, often with large uncertainties. Here, we demonstrate that all nematic elastic moduli, including the saddle-splay elastic constant k24, may be calculated directly from atomistic molecular simulations. Importantly, results obtained through in silico measurements of the 5CB elastic properties demonstrate unambiguously that saddle-splay elasticity alone is unable to describe the observed confined morphologies.

    Reactor thermal neutron irradiation of biological matrices induces high levels of intense gamma-ray or bremsstrahlung radiation from 82 Br, 42 K, 24 Na, and 32 P, that interfere with the determination of As, Cd, Cu and Mo by INAA. Central nervous system (CNS) tissue samples from subjects with Alzheimer's disease (AD), amyotrophic lateral sclerosis (ALS), and controls were analyzed using a simple RNAA procedure involving a rapid two-step solvent extraction procedure to determine these four elements. Significant increases (p ≤ 0.05) in concentrations of Cd and Mo were observed for brain of AD subjects compared to controls, but significant imbalances were not observed for ALS subjects.     The stability of nematic structures within a cylindrical capillary whose wall exhibits a homeotropic boundary condition is studied. The structures are obtained numerically from Euler-Lagrange equations resulting from the minimization of the Frank free energy functional. Stability diagrams are presented showing dependence on elastic properties, surface anchoring, and external transversal field strength. Emphasis is given to the effects of the saddle-splay elastic constant (K 24 ), which plays an important role in the weak anchoring regime. A new structure---the planar polar structure with two line defects---is predicted. It is shown that it is stable in a finite interval of the external field strength in the strong anchoring regime

    Full Text Available Abstract Background Limited data is available on sodium (Na and potassium (K intake in young children estimated by 24 hour (24h excretion in urine. The aim was to assess 24h urinary excretion of Na and K in six-year-old children and its relationship with diet quality. Methods The study population was a subsample of a national dietary survey, including six-year-old children living in the greater Reykjavik area (n=76. Three day weighed food records were used to estimate diet quality. Diet quality was defined as adherence to the Icelandic food based dietary guidelines. Na and K excretion was analyzed from 24h urine collections. PABA check was used to validate completeness of urine collections. The associations between Na and K excretion and diet quality were estimated by linear regression, adjusting for gender and energy intake. Results Valid urine collections and diet registrations were provided by 58 children. Na and K excretion was, mean (SD, 1.64 (0.54 g Na/24h (approx. 4.1 g salt/24h and 1.22 (0.43 g K/24h. In covariate adjusted models Na excretion decreased by 0.16 g Na/24h (95% CI: 0.31, 0.06 per 1-unit increase in diet quality score (score range: 1–4 while K excretion was increased by 0.18 g K/24h (95% CI: 0.06, 0.29. Conclusions Na intake, estimated by 24h urinary excretion was on average higher than recommended. Increased diet quality was associated with lower Na excretion and higher K excretion in six-year-old children.

  5. Selective cytotoxicity of the antibacterial peptide ABP-dHC-Cecropin A and its analog towards leukemia cells.

    Some cationic antibacterial peptides, with typical amphiphilic α-helical conformations in a membrane-mimicking environment, exhibit anticancer properties as a result of a similar mechanism of action towards both bacteria and cancer cells. We previously reported the cDNA sequence of the antimicrobial peptide ABP-dHC-Cecropin A precursor cloned from drury (Hyphantria cunea) (dHC). In the present study, we synthesized and structurally characterized ABP-dHC-Cecropin A and its analog, ABP-dHC-Cecropin A-K(24). Circular dichroism spectroscopy showed that ABP-dHC-Cecropin A and its analog adopt a well-defined α-helical structure in a 50% trifluorethanol solution. The cytotoxicity and cell selectivity of these peptides were further examined in three leukemia cell lines and two non-cancerous cell lines. The MTT assay indicated both of these peptides have a concentration-dependent cytotoxic effect in leukemia cells, although the observed cytotoxicity was greater with ABP-dHC-Cecropin A-K(24) treatment, whereas they were not cytotoxic towards the non-cancerous cell lines. Moreover, ABP-dHC-Cecropin A and its analog had a lower hemolytic effect in human red blood cells. Together, these results suggest the peptides are selectively cytotoxic towards leukemia cells. Confocal laser scanning microscopy determined that the peptides were concentrated at the surface of the leukemia cells, and changes in the cell membrane were determined with a permeability assay, which suggested that the anticancer activity of ABP-dHC-Cecropin A and its analog is a result of its presence at the leukemia cell membrane. ABP-dHC-Cecropin A and its analog may represent a novel anticancer agent for leukemia therapy, considering its cancer cell selectivity and relatively low cytotoxicity in normal cells.     Full Text Available Cucumber (Cucumis sativus L is an important member of the Cucurbitaceae family. Production of healthy nursery is necessary for high-quality production of this crop in greenhouses and in fields. With the idea of minimizing the use of pesticides and mineral fertilizers to preserve soil quality, we investigated the effects of plant growth promoting bacteria (PGPB on growth promotion and protection of cucumber plants from phytopathogenic fungi. The effects of Pseudomonas spp. strains with different antifungal activities and Bacillus sp. Q10 strain with PGP activity were tested on cucumber plants. Antagonistic activity of Pseudomonas spp. against the growth of several phytopathogenic fungi isolated from cucumber: F. acuminatum, B. cinerea and A. niger, was observed. The influences of overnight cultures, supernatants and heat-stable antifungal factors were tested on the phytopathogenic fungi in vitro. Pseudomonas sp. K35 and K24 strains were more effective than P. chlororaphis Q16 and Pseudomonas sp. K27, showing 70-80% of fungal growth inhibition regardless of culture or fraction applied. The good antagonists that belong to pseudomonads and Bacillus sp. Q10 strain were used as mixtures for estimation of plant growth and health promoting effects on cucumber plants. Growth dynamics differed depending on the applied strain of Pseudomonas sp. The M3 treatment (a mixture of Bacillus sp. Q10 and P. chlororaphis Q16 stimulated the initial phase of growth, while M4 (a mixture of Bacillus sp. Q10 and Pseudomonas sp. K24 resulted in the maximal height at the final measurement. Significant differences in leaf and plant weight (M4, and leaf weight (M5, containing K35 strain were found after the treatments. No significant differences in chlorophyll and NBI level were observed in any of the tested combinations. The obtained results suggested that M3 was suitable for stimulation of the early phase of cucumber growth, while the mixtures M4 and M5 improved plant

The chromosomal number of G. annulata was 2n=68 with the karyotype 2K= 6M + 62A. This study complements THOse carried out by our research group on cytogenetics of the poneromorph ants (sensu Bolton and would contribute to the better understanding of the ant evolution in this group considered primitive.

Differences in evolution of the absorption band and field enhancement upon Au nanoparticle adsorption are shown. The experimental results are found to be in good agreement with theoretical studies based on the finite-difference time-domain method and rigorous coupled-wave analysis.     Samples from the Islamic holy water Zamzam in Mecca, Saudi Arabia and the famous mineral water of Hot Springs, in Hot Springs, Arkansas were analyzed for trace elements content by thermal neutron activation analysis. For Zamzam the concentration of 37 S, 49 Ca, 38 Cl, 31 Si, 42 K, 24 Na and 82 Br were found, respectively, to be 3, 107, 11, 12, 4, 14, and 9 ppm; and that for Hot Springs Sample, replacing 82 Br with 27 Mg, are 2, 44, 2, 10, 1, 4, and 5 ppm. The experimental limit of detection for pure standards of the nuclides 27 Mg, 128 I, 64 Cu, and 56 Mn were found to be 8, 8x10 - 3, 6x10 - 2, and 2x10 - 4 μg, respectively. These nuclides were not detected in Zamzam, therefore, it was concluded that in Zamzam the concentration levels of the nuclides 27 Mg, 128 I, 64 Cu, and 56 Mn were below that of the limit of detection of pure standards.   10. Genetic diversity of K-antigen gene clusters of Escherichia coli and their molecular typing using a suspension array.

    Capsular polysaccharides (CPSs), or K-antigens, are the major surface antigens of Escherichia coli. More than 80 serologically unique K-antigens are classified into 4 groups (Groups 1-4) of capsules. Groups 1 and 4 contain the Wzy-dependent polymerization pathway and the gene clusters are in the order galF to gnd; Groups 2 and 3 contain the ABC-transporter-dependent pathway and the gene clusters consist of 3 regions, regions 1, 2 and 3. Little is known about the variations among the gene clusters. In this study, 9 serotypes of K-antigen gene clusters (K2ab, K11, K20, K24, K38, K84, K92, K96, and K102) were sequenced and correlated with their CPS chemical structures. On the basis of sequence data, a K-antigen-specific suspension array that detects 10 distinct CPSs, including the above 9 CPSs plus K30, was developed. This is the first report to catalog the genetic features of E. coli K-antigen variations and to develop a suspension array for their molecular typing. The method has a number of advantages over traditional bacteriophage and serum agglutination methods and lays the foundation for straightforward identification and detection of additional K-antigens in the future.

  11. Stability of Disclinations in Nematic Liquid Crystals

    In the light of φ-mapping method and topological current theory, the stability of disclinations around a spherical particle in nematic liquid crystals is studied. We consider two different defect structures around a spherical particle: disclination ring and point defect at the north or south pole of the particle. We calculate the free energy of these different defects in the elastic theory. It is pointed out that the total Frank free energy density can be divided into two parts. One is the distorted energy density of director field around the disclinations. The other is the free energy density of disclinations themselves, which is shown to be concentrated at the defect and to be topologically quantized in the unit of (k-k 24 )π/2. It is shown that in the presence of saddle-splay elasticity a dipole (radial and hyperbolic hedgehog) configuration that accompanies a particle with strong homeotropic anchoring takes the structure of a small disclination ring, not a point defect.

    Plastic deformation mechanisms under cyclic or monotonous solicitations, are analysed in function of Inconel X750 initial macrostructure. Two heat treated Inconel (first one is treated at 1366 K one hour, air cooled, aged at 977 K 20 hours, and air cooled, the second alloy is aged at 1158 K 24 hours, air cooled, aged at 977 K 20 hours, and air cooled), are characterized respectively by a fine and uniform precipitation of the γ' phase (approximative formulae: Ni 3 (Al,Ti)), and by a bimodal distribution of γ' precipitates. In both alloys, dislocations pairs (characteristic of a shearing by antiphase wall creation) are observed, and the crossing mechanism of the γ' precipitates by creation of overstructure pile defects is the same. But, glissile loops dislocations are less numerous than dislocations pairs in the first alloy, involving denser bands structure for this alloy (dislocations loops are always observed around γ' precipitates).     The response to the extract of the egyptian wild herb Calendula Micrantha, with the Molluscicidal activity, was examined in the unicellular no bacterium Synechococcus sp. strain PCC 6301. growth and chlorophyll of the cells were only slightly affected by low plant extract concentrations but were drastically reduced by high concentration. the rate of protein synthesis progressively decreased by increasing extract concentration. the cells preferentially induced the synthesis of a limited number of polypeptides in response to the treatment. Among the induced polypeptides were those with apparent molecular weights of 161 K (161.000), 96.7 K, 93.4 K, 69.9 K, 59 K, 49 K, 45 K, 35 K, 32.4 K, 28 K, 24 K, 21.7 K, 18 K and 16 K based on their mobilities in gel electrophoresis. these initial studies suggest that the plant extract exerted certain stress which stimulated alteration in the pattern of protein synthesis in Synechococcus sp. some of induced polypeptides are similar to that known to occur in other stresses specially heat shock stress. 3 figs

  14. A memory module for experimental data handling

    A compact CAMAC memory module for experimental data handling was developed to eliminate the need of direct memory access in computer controlled measurements. When using autonomous controllers it also makes measurements more independent of the program and enlarges the available space for programs in the memory of the micro-computer. The memory module has three modes of operation: an increment-, a list- and a fifo mode. This is achieved by connecting the main parts, being: the memory (MEM), the fifo buffer (FIFO), the address buffer (BUF), two counters (AUX and ADDR) and a readout register (ROR), by an internal 24-bit databus. The time needed for databus operations is 1 μs, for measuring cycles as well as for CAMAC cycles. The FIFO provides temporary data storage during CAMAC cycles and separates the memory part from the application part. The memory is variable from 1 to 64K (24 bits) by using different types of memory chips. The application part, which forms 1/3 of the module, will be specially designed for each application and is added to the memory chian internal connector. The memory unit will be used in Mössbauer experiments and in thermal neutron scattering experiments.

    We describe 99 (experimental and certain quasi-experimental) U.S.-based trials, reported or published since 1988, of behavioral and social interventions that measured prespecified behavioral and biologic outcomes and aimed to reduce risk for HIV infection. Studies identified through June 1998 by the HIV/AIDS Prevention Research Synthesis project were grouped into 4 risk behavior areas: drug-related (k [number of studies] = 48), heterosexual youth (k = 24), heterosexual adult (k = 17), and same-sex (k = 10). We compared the studies in the 4 areas by variables key to the development, evaluation, and transfer of interventions. Participants comprised injection drug users (43% of studies), drug users out of treatment (29%), African Americans (18%), clinic patients (18%), youth in schools (10%), and drug users in treatment (10%). Most studies were randomized (85%), provided another intervention to the control or comparison groups (71%), and evaluated behavioral interventions (92%). On average, interventions were conducted in 5 sessions (total, 8 hours) during 3 months. The theoretical basis of the intervention was not noted in 57% of the reports. At least one variable from each of the 3 outcome classifications was measured in 8% of the studies: behavioral, biologic, and psychosocial. Distinct profiles exist for the 4 risk areas. Addressing gaps in research and reporting would be helpful for analytical and program activities. This sizable portfolio of evaluated interventions contributes to effectiveness reviews and to considerations of transfer to program practice.

    Ira Hirsh's contributions to clinical science and research are diverse and significant. In fact, approximately one-third of the 100+ publications that Ira lists in his curriculum vitae (CV) are clinical in nature, dealing with various aspects of audiology, deafness, hearing aids, aural rehabilitation, and speech and language pathology. The majority of these citations, fully one-quarter of his publication list, addresses problems specific to the clinical measurement of hearing and hearing impairment. Undoubtedly, the most influential of these published works appears in his CV under the citation ``The Measurement of Hearing.'' The forward for this publication, his only textbook, was penned in June, 1952 (now precisely half a century past at the time of this session). The aims of this presentation are to (1) provide perspective on the fundamental importance of his virtually timeless text in shaping the fledgling discipline of audiology, and (2) celebrate Ira's many contributions to the profession and practice of audiology. [Preparation for this presentation was supported, in part, by a K24 career development award from NIDCD.

  17. Interferometric diameters of five evolved intermediate-mass planet-hosting stars measured with PAVO at the CHARA Array

    Debate over the planet occurrence rates around intermediate-mass stars has hinged on the accurate determination of masses of evolved stars, and has been exacerbated by a paucity of reliable, directly-measured fundamental properties for these stars. We present long-baseline optical interferometry of five evolved intermediate-mass (˜ 1.5 M⊙) planet-hosting stars using the PAVO beam combiner at the CHARA Array, which we combine with bolometric flux measurements and parallaxes to determine their radii and effective temperatures. We measured the radii and effective temperatures of 6 Lyncis (5.12±0.16 R⊙, 4949±58 K), 24 Sextantis (5.49±0.18 R⊙, 4908±65 K), κ Coronae Borealis (4.77±0.07 R⊙, 4870±47 K), HR 6817 (4.45±0.08 R⊙, 5013±59 K), and HR 8641 (4.91±0.12 R⊙, 4950±68 K). We find disagreements of typically 15 % in angular diameter and ˜ 200 K in temperature compared to interferometric measurements in the literature, yet good agreement with spectroscopic and photometric temperatures, concluding that the previous interferometric measurements may have been affected by systematic errors exceeding their formal uncertainties. Modelling based on BaSTI isochrones using various sets of asteroseismic, spectroscopic, and interferometric constraints tends to favour slightly (˜ 15 %) lower masses than generally reported in the literature.

    Full Text Available Biodiesel is a fuel derived from vegetable oil or animal fats that can be used as an additive to or entirely replace conventional petroleum diesel fuel. In most cases, biodiesel is mixed with conventional diesel because of the higher cost of biodiesel, product availability and engine compatibility issues. In Indonesia, the decree No. 3675K/24/DJM/2006 regarding the quality and specification of diesel oil type Solsr 48 and Solar 51 has been issued this decree regulates the use of FAME (fatty acid methyl ester up to the maximum of 10 percent of the volume of automotive diesel fuel with which it is to be blended. The cost to measure the properlles of fuel is expensive and time consuming, therefore it is important to develop a simple method to predict those blending properties. This paper presents the development of a simple calculation method for the validation of blend palm biodiesel-mineral diesel specification (density, viscosity, cetane number and lubricity which has been measured in the author's previous study The result shows that the lubricity and viscosity shows a higher average error value (difference value between calculation and measurement result of 1.66% and 1.35%, whereas density and cetane number shows lower average error values of 0.06 and 0.6%. The average error value less than 2% is still acceptable.

  20. Molecular dynamics simulations of Gay-Berne nematic liquid crystal: Elastic properties from direct correlation functions

    We report NVT and NPT molecular dynamics simulations of a Gay-Berne nematic liquid crystal using generalization of recently proposed algorithm by Toxvaerd [Phys. Rev. E47, 343, 1993]. On the basis of these simulations the Oseen-Zoher-Frank elastic constants K 11 , K 22 and K 33 as well as the surface constants K 13 and K 24 have been calculated within the framework of the direct correlation function approach of Lipkin et al. [J. Chem. Phys. 82, 472 (1985)]. The angular coefficients of the direct pair correlation function, which enter the final formulas, have been determined from the computer simulation data for the pair correlation function of the nematic by combining the Ornstein-Zernike relation and the Wienier-Hopf factorization scheme. The unoriented nematic approximation has been assumed when constructing the reference, isotropic state of Lipkin et al. By an extensive study of the model over a wide range of temperatures, densities and pressures a very detailed information has been provided about elastic behaviour of the Gay-Berne nematic. Interestingly, it is found that the results for the surface elastic constants are qualitatively different than those obtained with the help of analytical approximations for the isotropic, direct pair correlation function. For example, the values of the surface elastic constants are negative and an order of magnitude smaller than the bulk elasticity. (author). 30 refs, 9 figs

    A compact Analog to Digital Converter system suitable for high resolution γ-ray analysers has been developed based on the ADADC84 12-bit converter from Analog Devices. The converter was equipped with a peak detector and a stretcher, and with a memory card providing the sliding scale circuits, the lower threshold, the channel number identification and the zero suppression. The conversion time is 10 μs and the differential non linearity is less than ±1% for a 12-bit resolution. The converter consists of a 12-bit spectroscopy analog-to-digital converter (ADC) while the memory card includes a 8K 24-bit buffer memory. The two cards are plugged into a slot of an IBM PC AT and using an emulation software converts the micro-computer into a full-featured pulse height analyser. In data acquisition mode, the cards can operate independently, making the computer free for other tasks. The software offers acquisition control, visualization, data handling functions and various types of result presentation

  3. Radiation safety aspects of a 30 MeV proton cyclotron

    High current accelerators are increasingly used in nuclear medicine, power industry, material properties, material damage and astrophysical studies, etc. In the present work we have assessed the direct and transmitted neutron dose, build up and decay of air activity in the vault and soil activity for a 30 MeV 350 μA proton cyclotron. The transmitted dose equivalent H through ordinary concrete shield of different thickness has been estimated using the two different sets of values of the attenuation coefficient T eff . It is observed that while the two sets of H values differ by 25-26%, the required shield thickness is around 2.1 m. in both the cases to bring down the dose to the ICRP specified limit of 1 μSv/hr. Activity induced in the air due to 13 N and 15 O has been estimated. It has been found that for a target vault of 4m.x4m.x4m. dimension the activity concentration goes above the DAC value within a few seconds of commencement of operation even with 12 air changes per hour. A theoretical study of the radioactivity that may be induced in the soil indicates formation of 40 K, 24 Na, 56 Mn, 59 Fe, 27 Mg, 60 Co, 59,63,65 Ni, 64,66 Cu, 65,69 Zn radioisotopes. (author)

  4. Flat Field Anomalies in an X-ray CCD Camera Measured Using a Manson X-ray Source (HTPD 08 paper)

    The Static X-ray Imager (SXI) is a diagnostic used at the National Ignition Facility (NIF) to measure the position of the X-rays produced by lasers hitting a gold foil target. The intensity distribution taken by the SXI camera during a NIF shot is used to determine how accurately NIF can aim laser beams. This is critical to proper NIF operation. Imagers are located at the top and the bottom of the NIF target chamber. The CCD chip is an X-ray sensitive silicon sensor, with a large format array (2k x 2k), 24 (micro)m square pixels, and 15 (micro)m thick. A multi-anode Manson X-ray source, operating up to 10kV and 10W, was used to characterize and calibrate the imagers. The output beam is heavily filtered to narrow the spectral beam width, giving a typical resolution E/ΔE ∼ 10. The X-ray beam intensity was measured using an absolute photodiode that has accuracy better than 1% up to the Si K edge and better than 5% at higher energies. The X-ray beam provides full CCD illumination and is flat, within ±1% maximum to minimum. The spectral efficiency was measured at 10 energy bands ranging from 930 eV to 8470 eV. We observed an energy dependent pixel sensitivity variation that showed continuous change over a large portion of the CCD. The maximum sensitivity variation occurred at 8470 eV. The geometric pattern did not change at lower energies, but the maximum contrast decreased and was not observable below 4 keV. We were also able to observe debris, damage, and surface defects on the CCD chip. The Manson source is a powerful tool for characterizing the imaging errors of an X-ray CCD imager.     We study the multi-instanton partition functions of the Ω-deformed N=2{sup ∗}SU(2) gauge theory in the Nekrasov-Shatashvili (NS) limit. They depend on the deformation parameters ϵ{sub 1}, the scalar field expectation value a, and the hypermultiplet mass m. At fixed instanton number k, they are rational functions of ϵ{sub 1},a,m and we look for possible regularities that admit a parametrical description in the number of instantons. In each instanton sector, the contribution to the deformed Nekrasov prepotential has poles for large deformation parameters. To clarify the properties of these singularities we exploit Bethe/gauge correspondence and examine the special ratios m/ϵ{sub 1} at which the associated spectral problem is n-gap. At these special points we illustrate several structural simplifications occurring in the partition functions. After discussing various tools to compute the prepotential, we analyze the non-perturbative corrections up to k=24 instantons and present various closed expressions for the coefficients of the singular terms. Both the regular and singular parts of the prepotential are resummed over all instantons and compared successfully with the exact prediction from the spectral theory of the Lamé equation, showing that the pole singularities are an artifact of the instanton expansion. The analysis is fully worked out in the 1-gap case, but the final pole cancellation is proved for a generic ratio m/ϵ{sub 1} relating it to the gap width of the Lamé equation.

  6. Calibration of a 4π-γ well-type ionization chamber system for measuring of the radionuclides activity

    The calibration of a 4π well-type ionization Chamber System installed at the Laboratorio de Metrologia Nuclear, of the Instituto de Energia Atomica of Sao Paulo used for of the activity determination of radioactive solutions is descrided. The determination can be performed by two methods: 1) Direct Method, comparing the ionization Chamber response for solutions of unknown activity against that obtained with a solution which is standardized by the Absolute 4πβγ Coincidence Method. By this method the following radionuclides are standardized: 241 Am, 139 Ce, 198 Au, 22 Na, 134 Cs, 54 Mn, 60 Co, 42 K, 24 Na. In this case, the accuracy achieved is about 0.2 to 0,4%. 2) Indirect Method, by means of curves of relative beta or gama efficiency, which were determined in this work. This method can be applied for those radionuclides not included in the direct method. In this case, the accuracy depends on the gama energy range of the curves and on the accuracy of the absolute gama intensities, taken from the literature. In general the uncertainty is greater than the direct method, but values of 0,2% can be achieved in favourable cases. The upper and lower limits of Activity that can be measured depend on the radionuclide. These limits are from a few micro-curies to many mili-curies, which are satisfactory for most purposes. The sample preparation is simple and the time spent in the measurement is, in general, restricted to a few minutes.   7. Dynamics and control of Ebola virus transmission in Montserrado, Liberia: a mathematical modeling analysis

    Background A substantial scale-up in public health response is needed to control the unprecedented Ebola virus disease (EVD) epidemic in West Africa. Current international commitments seek to expand intervention capacity in three areas: new EVD Treatment Centers (ETCs); case ascertainment through contact tracing; and household protective kit allocation. Methods We developed a transmission model of Ebola virus that we fitted to reported EVD cases and deaths in Montserrado County, Liberia. We used this model to evaluate effectiveness of expanding ETCs, improving case ascertainment, and allocating protective kits for controlling the outbreak in Montserrado. Findings We estimated the basic reproductive number for EVD in Montserrado to be 2·49 [2·38–2·60]. We expect that allocating 4,800 additional ETC beds and increasing case ascertainment fivefold in November can avert 77312 [68400–85870] cases relative to the status quo by 15 December. Complementing these measures with protective kit allocation increases the expectation as high as 97940 [90096–105606] cases. If deployed by 15 October, equivalent interventions would have been expected to avert 137432 [129736–145874] cases. If delayed to 15 November, we expect the interventions will at best avert 53957 [49963–60490] cases. Interpretation The number of ETC beds needed to effectively control EVD in Montserrado substantially exceeds the total pledged by the United States to West Africa. Accelerated case ascertainment is required to maximize effectiveness of expanding ETC capacity. Distributing protective kits can further augment EVD prevention. Our findings highlight the rapidly closing window of opportunity for controlling the outbreak and averting a catastrophic toll of EVD cases and deaths.     Full Text Available Air temperature changes were applied to a regional model of permafrost probability under equilibrium conditions for an area of nearly 0.5 × 106 km2 in the southern Yukon and northwestern British Columbia, Canada. Associated environmental changes, including snow cover and vegetation, were not considered in the modelling. Permafrost extent increases from 58% of the area (present day: 1971–2000 to 76% under a −1 K cooling scenario, whereas warming scenarios decrease the percentage of permafrost area exponentially to 38% (+ 1 K, 24% (+ 2 K, 17% (+ 3 K, 12% (+ 4 K and 9% (+ 5 K of the area. The morphology of permafrost gain/loss under these scenarios is controlled by the surface lapse rate (SLR, i.e. air temperature elevation gradient, which varies across the region below treeline. Areas that are maritime exhibit SLRs characteristically similar above and below treeline resulting in low probabilities of permafrost in valley bottoms. When warming scenarios are applied, a loss front moves to upper elevations (simple unidirectional spatial loss. Areas where SLRs are gently negative below treeline and normal above treeline exhibit a loss front moving up-mountain at different rates according to two separate SLRs (complex unidirectional spatial loss. Areas that display high continentally exhibit bidirectional spatial loss in which the loss front moves up-mountain above treeline and down-mountain below treeline. The parts of the region most affected by changes in MAAT (mean annual air temperature have SLRs close to 0 K km−1 and extensive discontinuous permafrost, whereas the least sensitive in terms of areal loss are sites above the treeline where permafrost presence is strongly elevation dependent.

  9. Syzygium jambos Displayed Antibacterial and Antibiotic-Modulating Activities against Resistant Phenotypes

    Full Text Available The present study was designed to evaluate the antibacterial activities of methanol extracts of bark and leaves of Syzygium jambos, as well as their synergistic effects with selected antibiotics against drug-resistant Gram-positive and Gram-negative bacteria. The crude extracts were subjected to qualitative phytochemical screening; broth microdilution method was used for antibacterial assays. Phytochemical studies indicate that leaves and bark extracts contained polyphenols, anthraquinones, tannins, and steroids. Extract of the leaves was active against all the 26 strains of Staphylococcus aureus and all the 21 strains of Gram-negative bacteria tested, within the minimum inhibitory concentration (MIC range of 32–512 μg/mL. The lowest MIC value of 32 μg/mL was obtained with extract of the leaves against Staphylococcus aureus MRSA9 strain. In Gram-negative bacteria, the lowest MIC value of 64 μg/mL was also obtained against Enterobacter aerogenes EA294 and Klebsiella pneumoniae K24 strains. Against S. aureus strains, antibiotic-modulating activity of extracts at MIC/2 towards more than 70% of the tested strains was obtained when leaves and bark extracts were tested in association with chloramphenicol (CHL. This was also the case when leaves extract was combined with CHL, kanamycin (KAN, tetracycline (TET, and erythromycin (ERY and when bark extract was combined with ciprofloxacin (CIP, TET, and ERY against Gram-negative bacteria. In conclusion, this study demonstrated that Syzygium jambos has antibacterial and antibiotic-modulating activities.

    Dipole moments of proteins arise from helical dipoles, hydrogen bond networks and charged groups at the protein surface. High protein dipole moments were suggested to contribute to the electrostatic steering between redox partners in electron transport chains of respiration, photosynthesis and steroid biosynthesis, although so far experimental evidence for this hypothesis was missing. In order to probe this assumption, we changed the dipole moment of the electron transfer protein adrenodoxin and investigated the influence of this on protein-protein interactions and electron transfer. In bovine adrenodoxin, the [2Fe-2S] ferredoxin of the adrenal glands, a dipole moment of 803 Debye was calculated for a full-length adrenodoxin model based on the Adx(4-108) and the wild type adrenodoxin crystal structures. Large distances and asymmetric distribution of the charged residues in the molecule mainly determine the observed high value. In order to analyse the influence of the resulting inhomogeneous electric field on the biological function of this electron carrier the molecular dipole moment was systematically changed. Five recombinant adrenodoxin mutants with successively reduced dipole moment (from 600 to 200 Debye) were analysed for their redox properties, their binding affinities to the redox partner proteins and for their function during electron transfer-dependent steroid hydroxylation. None of the mutants, not even the quadruple mutant K6E/K22Q/K24Q/K98E with a dipole moment reduced by about 70% showed significant changes in the protein function as compared with the unmodified adrenodoxin demonstrating that neither the formation of the transient complex nor the biological activity of the electron transfer chain of the endocrine glands was affected. This is the first experimental evidence that the high dipole moment observed in electron transfer proteins is not involved in electrostatic steering among the proteins in the redox chain.

  11. Serum Renalase Levels Correlate with Disease Activity in Lupus Nephritis.

    Full Text Available Lupus nephritis (LN is among the most serious complications of systemic lupus erythematosus (SLE, which causes significant morbidity and mortality. Renalase is a novel, kidney-secreted cytokine-like protein that promotes cell survival. Here, we aimed to investigate the relationship of serum renalase levels with LN and its role in the disease progression of LN.For this cross-sectional study, 67 LN patients and 35 healthy controls were enrolled. Seventeen active LN patients who received standard therapies were followed up for six months. Disease activity was determined by the SLE Disease Activity-2000 (SLEDAI-2K scoring system and serum renalase amounts were determined by ELISA. Predictive value of renalase for disease activity was assessed. Furthermore, the expression of renalase in the kidneys of patients and macrophage infiltration was assessed by immunohistochemistry.Serum renalase amounts were significantly higher in LN patients than in healthy controls. Moreover, patients with proliferative LN had more elevated serum renalase levels than Class V LN patients. In proliferative LN patients, serum renalase levels were significantly higher in patients with active LN than those with inactive LN. Serum renalase levels were positively correlated with SLEDAI-2K, 24-h urine protein excretion, ds-DNA and ESR but inversely correlated with serum albumin and C3. Renalase amounts decreased significantly after six-months of standard therapy. The performance of renalase as a marker for diagnosis of active LN was 0.906 with a cutoff value of 66.67 μg/ml. We also observed that the amount of renalase was significantly higher in glomerular of proliferative LN along with the co-expression of macrophages.Serum renalase levels were correlated with disease activity in LN.     Full Text Available Intrinsically disordered proteins (IDPs are frequently associated with human diseases such as cancers, and about one-fourth of disease-associated missense mutations have been mapped into predicted disordered regions. Understanding how these mutations affect the structure-function relationship of IDPs is a formidable task that requires detailed characterization of the disordered conformational ensembles. Implicit solvent coupled with enhanced sampling has been proposed to provide a balance between accuracy and efficiency necessary for systematic and comparative assessments of the effects of mutations as well as post-translational modifications on IDP structure and interaction. Here, we utilize a recently developed replica exchange with guided annealing enhanced sampling technique to calculate well-converged atomistic conformational ensembles of the intrinsically disordered transactivation domain (TAD of tumor suppressor p53 and several cancer-associated mutants in implicit solvent. The simulations are critically assessed by quantitative comparisons with several types of experimental data that provide structural information on both secondary and tertiary levels. The results show that the calculated ensembles reproduce local structural features of wild-type p53-TAD and the effects of K24N mutation quantitatively. On the tertiary level, the simulated ensembles are overly compact, even though they appear to recapitulate the overall features of transient long-range contacts qualitatively. A key finding is that, while p53-TAD and its cancer mutants sample a similar set of conformational states, cancer mutants could introduce both local and long-range structural modulations to potentially perturb the balance of p53 binding to various regulatory proteins and further alter how this balance is regulated by multisite phosphorylation of p53-TAD.   13. The Padre Miguel Ignimbrite Suite, central Honduras: Paleomagnetism, geochronology, and tectonic implications

    The Padre Miguel Group in western Honduras is a silicic volcanic sequence that forms part of the Central American Miocene volcanic arc built on the Chortis continental fragment. We report new 40Ar/39Ar data of 16.1 ± 0.2 Ma and 14.42 ± 0.08 Ma for the Padre Miguel Group, and present paleomagnetic data for 36 cooling units (mainly ignimbrites) from localities north and northeast of the capital city of Tegucigalpa. These rocks are characterized by univectorial characteristic magnetization carried mainly by low-Ti titanomagnetites, or two component magnetizations with a minor secondary overprint. Dual polarity magnetizations suggest that it is a primary thermo-remanence and the obtained data scatter can be straightforwardly explained by paleosecular variation. The overall mean of 33 selected sites (14.39°N, 87.14°W) defines a paleomagnetic pole at lat = 80.0°N, lon = 142.9°E (K = 24.5, A95 = 5.2°). The overall mean compared with the North America expected direction indicates statistically insignificant rotation and inclination anomaly (R = - 4.1° ± 6.1° and an F = - 8.6° ± 11.3°). Restoring 255 km of sinistral slip in the Polochic-Motagua fault system, as suggested by Cayman Trough reconstructions since 15 Ma, brings the mid-Miocene arcs of southern Mexico (Oaxaca) and the Central America ignimbrite province into alignment; this is consistent with a derivation of the Chortis Block from southern Mexico. Our paleomagnetic and geochronological studies hence support models that explain the offset of the Miocene ignimbrite belt of Central America by post-Middle Miocene displacement of the Chortis Block relative to North America.

    A standard model of experimental sepsis was elaborated at rats in dependence on a gamma irradiation dose and a time interval between irradiation and application infectious agents E. coli O 83:K 24:H 31. For a development of experimental sepsis was proved that it is not decisive in these laboratory animals when infectious agents is i.v. or i.p. applicated. Such amount of organisms (1-20.10 7-8 ) was applicated in particular not to develop sepsis in non-irradiated laboratory animals. Laboratory animals were irradiated with sublethal doses and approximately. LD 50-30 doses of gamma radiation. The laboratory animals were treated only in experiments and qinolone drug ofloxacin was used in the treatment of experimental sepsis. Ofloxacin perorally administrated in the dose of 40 mg/kg in an hour after application of infectious agents and its administration for 5 days in the 24-h intervals confirmed in fact 100% therapeutic effectiveness in irradiated experimental animals. In non-treated experimental groups, animals died in 24-28 hours interval after application of infectious agents and sepsis was a cause of death. In treated experimental groups, animal survived by day 30 after irradiation with sublethal doses or died during the period typical for a bone marrow syndrome of acute radiation injury after irradiation with lethal doses of gamma radiation. Acquired experimental outcomes may suggest that fluorochinolone chemotherapeuticals in the respect of their essential pharmacokinetic properties will be used for a prevention of infectious complications in acute radiation injury.   15. The effect of a probiotic Escherichia coli strain on regulatory T-cells in six year-old children.

    Probiotics are believed to prevent or reduce allergy development but the mechanism of their beneficial effect is still poorly understood. Immune characteristics of regulatory T cells (Tregs) in peripheral blood of perinatally probiotic-supplemented children of allergic mothers (51 children), non-supplemented children of allergic mothers (42 children), and non-supplemented children of healthy mothers (28 children) were compared at the age of 6-7 years. A first dose of a probiotic Escherichia coli strain (E. coli O83:K24:H31) was administered within 2 days after the birth and then 12 times during the first months of life and children were followed longitudinally. Proportion and functional properties of Tregs were estimated by flow cytometry in relation to the children's allergy status. Proportion of Tregs in the peripheral blood of children suffering from allergy tends to be higher whereas median of fluorescence intensity (MFI) of FoxP3 was significantly decreased in allergic group. Intracellular presence of regulatory cytokine interleukin (IL)-10 was also lower in allergic children. Immune functions of Tregs reflected by both MFI of FoxP3 and IL-10 in the group of probiotic-supplemented children of allergic mothers were nearly comparable with children of healthy mothers while probiotic non-supplemented children of allergic mothers have decreased immune function of Tregs. Supplementation by probiotic E. coli strain decreases allergy incidence in high-risk children. In contrast to our expectation, proportion of Tregs has not been increased in probiotic supplemented children. Beneficial effect of probiotics on newborn immature immune system could be, at least partially, explained by the modulating immune function of Tregs. In summary, we detected increased proportion of Tregs in peripheral blood of allergic children, their functional properties were decreased in comparison with the Tregs of healthy children.   16. Radionuclide distribution of Holocene sediments and its effects on the habitat of recent foraminifers: A case study from the Western Marmara Sea (Turkey)

    ABSTRACT Radionuclides cause radioactive contamination in aquatic environments just as other non-biodegradable pollutants, such as heavy metals, sink to the seafloor and accumulate in the sediments. These radioactive pollutants especially affect benthic foraminifera living on the sediment surface or in the sediments in the seafloor. Foraminifera were used as bioindicators to analyze the effect of radioactivity pollution on ecosystems. In this study, we have investigated natural and artificial radionuclide (232Th, 226Ra, 40K and 137Cs) distribution in sediment samples taken in the living areas of benthic foraminifera in the Western Marmara Sea by means of gamma spectrometry. Accordingly, 29 core samples taken in 2016 from depths of about 20-35 m close to the shores of the Marmara Sea were used. Core samples representing the pollution of the study area were collected at locations such as discharge points for domestic and industrial areas, port locations, and others. Other samples were taken from areas unaffected or less affected by pollution. The radionuclide concentration activity values in the sediment samples obtained from the locations, in Bq/kg, were 137Cs, 0.9-9.4; 232Th, 18.9-86; 226Ra, 10-50; 40K, 24.4-670. These values were compared with the United Nations Scientific Committee on the Effects of Atomic Radiation (UNSCEAR) data, and an environmental analysis was carried out. The 226Ra series, the 232Th series, and the 40K radionuclides accumulate naturally, and they are also increasing continuously due to anthropogenic pollution. Although the 226Ra values obtained throughout the study areas remained within normal limits according to the UNSCEAR values, the 40K and 232Th series values were found to be higher in almost all locations. According to these results, the main causes of radioactive pollution in the investigation area are agricultural and mining activities.   17. Flat Field Anomalies in an X-Ray CCD Camera Measured Using a Manson X-Ray Source

    The Static X-ray Imager (SXI) is a diagnostic used at the National Ignition Facility (NIF) to measure the position of the X-rays produced by lasers hitting a gold foil target. It determines how accurately NIF can point the laser beams and is critical to proper NIF operation. Imagers are located at the top and the bottom of the NIF target chamber. The CCD chip is an X-ray sensitive silicon sensor, with a large format array (2k x 2k), 24 (micro)m square pixels, and 15 (micro)m thick. A multi-anode Manson X-ray source, operating up to 10kV and 2mA, was used to characterize and calibrate the imagers. The output beam is heavily filtered to narrow the spectral beam width, giving a typical resolution E/ΔE ∼ 12. The X-ray beam intensity was measured using an absolute photodiode that has accuracy better than 1% up to the Si K edge and better than 5% at higher energies. The X-ray beam provides full CCD illumination and is flat, within ±1.5% maximum to minimum. The spectral efficiency was measured at 10 energy bands ranging from 930 eV to 8470 eV. The efficiency pattern follows the properties of Si. The maximum quantum efficiency is 0.71. We observed an energy dependent pixel sensitivity variation that showed continuous change over a large portion of the CCD. The maximum sensitivity variation was >8% at 8470 eV. The geometric pattern did not change at lower energies, but the maximum contrast decreased and was less than the measurement uncertainty below 4 keV. We were also able to observe debris on the CCD chip. The debris showed maximum contrast at the lowest energy used, 930 eV, and disappeared by 4 keV. The Manson source is a powerful tool for characterizing the imaging errors of an X-ray CCD imager. These errors are quite different from those found in a visible CCD imager

    Potential cost effectiveness of endovascular aneurysm repair (EVAR) compared with open aortic repair (OAR) is offset by the use of intraoperative adjuncts (components) or late reinterventions. Anatomic severity grade (ASG) can be used preoperatively to assess abdominal aortic aneurysms, and provide a quantitative measure of anatomic complexity. The hypothesis of this study is that ASG is directly related to the use of intraoperative adjuncts and cost of aortic repair. Patients who undergo elective OAR and EVAR for abdominal aortic aneurysms were identified over a consecutive 3-year period. ASG scores were calculated manually using three-dimensional reconstruction software by two blinded reviewers. Statistical analysis of cost data was performed using a log transformation. Regression analyses, with a continuous or dichotomous outcome, used a generalized estimating equations approach with the sandwich estimator, being robust with respect to deviations from model assumptions. One hundred forty patients were identified for analysis, n = 33 OAR and n = 107 EVAR. The mean total cost (± standard deviation) for OAR was per thousand (k) $38.3 ± 49.3, length of stay (LOS) 13.5 ± 14.2 days, ASG score 18.13 ± 3.78; for EVAR, mean total cost was k $24.7 ± 13.0 (P = .016), LOS 3.0 ± 4.4 days (P = .012), ASG score 15.9 ± 4.13 (P = .010). In patients who underwent EVAR, 25.2% required intraoperative adjuncts, and analysis of this group revealed a mean total cost of k $31.5 ± 15.9, ASG score 18.48 ± 3.72, and LOS 3.9 ± 4.5, which were significantly greater compared with cases without adjunctive procedures. An ASG score of ≥15 correlated with an increased propensity for requirement of intraoperative adjuncts; odds ratio, 5.75 (95% confidence interval, 1.82-18.19). ASG >15 was also associated with chronic kidney disease, end stage renal disease, hypertension, female sex, increased cost, and use of adjunctive procedures.     Color and pH of meat measured 24 h post mortem are common selection objectives in pig breeding programs. Several amino acid substitutions in PRKAG3 have been associated with various meat quality traits. In our previous study ASGA0070625, a SNP next to PRKAG3, had the most significant association with meat quality traits in the Finnish Yorkshire. However, the known amino acid substitutions, including I199V, did not show any association. The aims of this study were to characterize further variation in PRKAG3 and its promoter region, and to test the association between these variants and the pH and color of pork meat. The data comprised of 220 Finnish Landrace and 230 Finnish Yorkshire artificial insemination boars with progeny information. We sequenced the coding and promoter region of PRKAG3 in these and in three additional wild boars. Genotypes from our previous genome-wide scans were also included in the data. Association between SNPs or haplotypes and meat quality traits (deregressed estimates of breeding values from Finnish national breeding value estimation for pH, color lightness and redness measured from loin or ham) was tested using a linear regression model. Sequencing revealed several novel amino acid substitutions in PRKAG3, including K24E, I41V, K131R, and P134L. Linkage disequilibrium was strong among the novel variants, SNPs in the promoter region and ASGA0070625, especially for the Yorkshire. The strongest associations were observed between ASGA0070625 and the SNPs in the promoter region and pH measured from loin in the Yorkshire and between I199V and pH measured from ham in the Landrace. In contrast, ASGA0070625 was not significantly associated with meat quality traits in the Landrace and I199V not in the Yorkshire. Haplotype analysis showed a significant association between a haplotype consisting of 199I and 24E alleles (or g.-157C or g.-58A alleles in the promoter region) and pH measured from loin and ham in both breeds (P-values varied from 1.72

    Study has been done on rabbit to find out the effect of giving sheep serum post vaccination with irradiated gastric worm challenged with infective H. contortus larva. Forty eight local rabbit divided into 3 groups : K, VI and V2, each groups: consists of 4 rabbit, by using 4 times repetition. K group which only inoculated 10,000L3 H. contortus infective. V1 = group which twice serum inoculated with 21 days interval, inoculation dose were 0,3 ml serum each, and challenged with 10.000 L3 H.contortus infective. V2 = group which 3 times serum inoculation with 21 days interval, inoculation dose 0,3 ml serum each and challenged with 10.000 L3 H.contortus infective. Design model used with Completely Random Design (RAL), Duncan was used as inter treatment test. Parameter observed are total protein fraction, Larva L4 content in gaster, cumulative weight gain, and death percentage rabbits. Result of study showed that on average parameter total protein fraction on K + 2,0086 ± 0,2353 mg/dl, V1 = 3,2781 ± 0,9227 mg/dl and V2 = 5,5035 ± 1,6794 mg/d. Average parameter L4 death showed on K = 10, V1 = 8, and V2 = 4, while L4 alive on K = 3, V1 + 1 and V2 = 0. Commutative Average parameter PCV value on K = 19,91 ± 1,24%, V1= 27,31 ± 1,22%, and V2 35,71 ± 0,56 %. Average parameter weight gain per week on K = 24,46 ± 0,74 g, V1 = 26,04 ± 2.23 g, and V2 = 28,93 ± 1,67 g. Percentage average parameter death rate on K = 37,5 ± 14,46%, V1=18,75 ± 23,90% and V2 = 0± 0%.   1. Preclinical and Clinical Resistance Profile of EDP-239, a Novel Hepatitis C Virus NS5A Inhibitor.

    EDP-239, a potent and selective hepatitis C virus (HCV) nonstructural protein 5A (NS5A) inhibitor developed for the treatment of HCV infection, has been investigated in vitro and in vivo This study sought to characterize genotypic changes in the HCV NS5A sequence of genotype 1 (GT1) replicons and to compare those changes to GT1 viral RNA mutations isolated from clinical trial patients. Resistance selection experiments in vitro using a subgenomic replicon identified resistance-associated mutations (RAMs) at GT1a NS5A amino acid positions 24, 28, 30, 31, and 93 that confer various degrees of resistance to EDP-239. Key RAMs were similarly identified in GT1b NS5A at amino acid positions 31 and 93. Mutations F36L in GT1a and A92V in GT1b do not confer resistance to EDP-239 individually but were found to enhance the resistance of GT1a K24R and GT1b Y93H. RAMs were identified in GT1 patients at baseline or after dosing with EDP-239 that were similar to those detected in vitro Baseline RAMs identified at NS5A position 93 in GT1, or positions 28 or 30 in GT1a only, correlated with a reduced treatment response. RAMs at additional positions were also detected and may have contributed to reduced EDP-239 efficacy. The most common GT1a and GT1b RAMs found to persist up to weeks 12, 24, or 48 were those at NS5A positions 28, 30, 31, 58 (GT1a only), and 93. Those RAMs persisting at the highest frequencies up to weeks 24 or 48 were L31M and Q30H/R for GT1a and L31M and Y93H for GT1b.   2. The impact of biotic/abiotic interfaces in mineral nutrient cycling: A study of soils of the Santa Cruz chronosequence, California

    Biotic/abiotic interactions between soil mineral nutrients and annual grassland vegetation are characterized for five soils in a marine terrace chronosequence near Santa Cruz, California. A Mediterranean climate, with wet winters and dry summers, controls the annual cycle of plant growth and litter decomposition, resulting in net above-ground productivities of 280-600gm -2yr -1. The biotic/abiotic (A/B) interface separates seasonally reversible nutrient gradients, reflecting biological cycling in the shallower soils, from downward chemical weathering gradients in the deeper soils. The A/B interface is pedologically defined by argillic clay horizons centered at soil depths of about one meter which intensify with soil age. Below these horizons, elevated solute Na/Ca, Mg/Ca and Sr/Ca ratios reflect plagioclase and smectite weathering along pore water flow paths. Above the A/B interface, lower cation ratios denote temporal variability due to seasonal plant nutrient uptake and litter leaching. Potassium and Ca exhibit no seasonal variability beneath the A/B interface, indicating closed nutrient cycling within the root zone, whereas Mg variability below the A/B interface denotes downward leakage resulting from higher inputs of marine aerosols and lower plant nutrient requirements.The fraction of a mineral nutrient annually cycled through the plants, compared to that lost from pore water discharge, is defined their respective fluxes F j,plants=q j,plants/(q j,plants+q j,discharge) with average values for K and Ca (F K,plants=0.99; F Ca,plants=0.93) much higher than for Mg and Na (F Mg,plants 0.64; F Na,plants=0.28). The discrimination against Rb and Sr by plants is described by fractionation factors (K Sr/Ca=0.86; K Rb/K=0.83) which are used in Rayleigh fractionation-mixing calculations to fit seasonal patterns in solute K and Ca cycling.     Full Text Available High-resolution simulations of the chemical composition of the Arctic stratosphere during late spring 1997 and 2000 were performed with the Chemical Lagrangian Model of the Stratosphere (CLaMS. The simulations were performed for the entire northern hemisphere on two isentropic levels 450 K (~18 km and 585 K (~24 km. The spatial distribution and the lifetime of the vortex remnants formed after the vortex breakup in May 1997 display different behavior above and below 20 km. Above 20 km, vortex remnants propagate southward (up to 40°N and are "frozen in'' in the summer circulation without significant mixing. Below 20 km the southward propagation of the remnants is bounded by the subtropical jet. Their lifetime is shorter by a factor of 2 than that above 20 km, owing to significant stirring below this altitude. The behavior of vortex remnants formed in March 2000 is similar but, due to an earlier vortex breakup, dominated during the first 6 weeks after the vortex breakup by westerly winds, even above 20 km. Vortex remnants formed in May 1997 are characterized by large mixing ratios of HCl indicating negligible, halogen-induced ozone loss. In contrast, mid-latitude ozone loss in late boreal spring 2000 is dominated, until mid-April, by halogen-induced ozone destruction within the vortex remnants, and subsequent transport of the ozone-depleted polar air masses (dilution into the mid-latitudes. By varying the intensity of mixing in CLaMS, the impact of mixing on the formation of ClONO2 and ozone depletion is investigated. We find that the photochemical decomposition of HNO3 and not mixing with NOx-rich mid-latitude air is the main source of NOx within the vortex remnants in March and April 2000. Ozone depletion in the remnants is driven by ClOx photolytically formed from ClONO2. At the end of May 1997, the halogen-induced ozone deficit at 450 K poleward of 30°N amounts to ~12% with ~10% in the polar vortex and ~2% in well-isolated vortex remnants

    Full Text Available Abstract Background Neonatal rats exposed to repetitive inflammatory pain have altered behaviors in young adulthood, partly ameliorated by Ketamine analgesia. We examined the relationships between protein expression, neuronal survival and plasticity in the neonatal rat brain, and correlated these changes with adult cognitive behavior. Methods Using Western immunoblot techniques, homogenates of cortical tissue were analyzed from neonatal rats 18–20 hours following repeated exposure to 4% formalin injections (F, N = 9, Ketamine (K, 2.5 mg/kg × 2, N = 9, Ketamine prior to formalin (KF, N = 9, or undisturbed controls (C, N = 9. Brain tissues from another cohort of rat pups (F = 11, K = 12, KF = 10, C = 15 were used for cellular staining with Fos immunohistochemistry or FluoroJade-B (FJB, followed by cell counting in eleven cortical and three hippocampal areas. Long-term cognitive testing using a delayed non-match to sample (DNMS paradigm in the 8-arm radial maze was performed in adult rats receiving the same treatments (F = 20, K = 24, KF = 21, C = 27 in the neonatal period. Results Greater cell death occurred in F vs. C, K, KF in parietal and retrosplenial areas, vs. K, KF in piriform, temporal, and occipital areas, vs. C, K in frontal and hindlimb areas. In retrosplenial cortex, less Fos expression occurred in F vs. C, KF. Cell death correlated inversely with Fos expression in piriform, retrosplenial, and occipital areas, but only in F. Cortical expression of glial fibrillary acidic protein (GFAP was elevated in F, K and KF vs. C. No significant differences occurred in Caspase-3, Bax, and Bcl-2 expression between groups, but cellular changes in cortical areas were significantly correlated with protein expression patterns.   5. Certification measurement of the cadmium, copper and lead contents in rice using isotope dilution inductively coupled plasma mass spectrometry

    This paper describes the certification of the Cd, Cu and Pb amount contents in a rice material, that were used as reference values for round 19 of the international measurement evaluation programme (IMEP). The protocol developed in this study was based on isotope dilution associated to inductively coupled plasma mass spectrometry (ID-ICP-MS). A multiple spiking approach was applied to reduce significantly the number of analytical steps. For the decomposition of the sample, three different microwave assisted digestion procedures were tested and compared. The use of hydrofluoric acid was found necessary to ensure full solubilisation and complete isotopic equilibration. Estimation of the combined uncertainty attached to the measurement results was performed according to the ISO guidelines. Contributions from the correction for moisture content, sample homogeneity, procedural blank, spike impurities, spectral interferences, instrumental background and dead-time effects were evaluated. SI-traceable values with less than 2% combined uncertainty (k = 2) were obtained for Cd, Cu and Pb, respectively, (14.39 ± 0.21) x 10 -6 mol kg -1 , (44.31 ± 0.42) x 10 -6 mol kg -1 and (2.034 ± 0.034) x 10 -6 mol kg -1 . The rice powder was observed to be highly hygroscopic and a second series of isotope dilution mass spectrometry measurements was carried out on samples in equilibrium with the ambient moisture conditions ('saturated' samples). The Comite Consultatif pour la Quantite de Matiere (CCQM) requested this approach for the participation to the key comparison 24 on the same rice test material. The excellent agreement for Cd between the IMEP-19 reference value, the value submitted by the institute for reference materials and measurements (IRMM) to CCQM-K24 and the corresponding reference value obtained as the arithmetic mean from the results of the 11 participating National Measurement Institutes (less than 0.27% difference) further validated this work.     Recognition is growing that social anxiety disorder (SAnD) is a chronic and disabling disorder, and data from early trials demonstrate that medication may be effective in its treatment. This systematic review is an update of an earlier review of pharmacotherapy of SAnD. To assess the effects of pharmacotherapy for social anxiety disorder in adults and identify which factors (methodological or clinical) predict response to treatment. We searched the Cochrane Common Mental Disorders Controlled Trials Register (CCMDCTR-Studies and CCMDCTR-References) to 17 August 2015. The CCMDCTR contains reports of relevant RCTs from MEDLINE (1950-), Embase (1974-), PsycINFO (1967-) and CENTRAL (all years). We scanned the reference lists of articles for additional studies. We updated the search in August 2017 and placed additional studies in Awaiting Classification, these will be incorporated in the next version of the review, as appropriate. We restricted studies to randomised controlled trials (RCTs) of pharmacotherapy versus placebo in the treatment of SAnD in adults. Two authors (TW and JI) assessed trials for eligibility and inclusion for this review update. We extracted descriptive, methodological and outcome information from each trial, contacting investigators for missing information where necessary. We calculated summary statistics for continuous and dichotomous variables (if provided) and undertook subgroup and sensitivity analyses. We included 66 RCTs in the review (> 24 weeks; 11,597 participants; age range 18 to 70 years) and 63 in the meta-analysis. For the primary outcome of treatment response, we found very low-quality evidence of treatment response for selective serotonin reuptake inhibitors (SSRIs) compared with placebo (number of studies (k) = 24, risk ratio (RR) 1.65; 95% confidence interval (CI) 1.48 to 1.85, N = 4984).     Full Text Available Amaç:Hekimlik pratiğine dair meslek örgütüne yansıyan ihlal iddialarını ve belirleyenlerini incelemektir. Yöntem:Araştırma, Bursa Tabip Odası (BTO Yönetim Kurulu’nun 1991-2014 yılları arasındaki tüm kayıtlarının, Onur Kurulu dosyalarının ve bilgisayar kayıtlarının taranmasıyla gerçekleştirilmiştir.Bulgular:İncelenen dönemde BTO’ya yapılan 593 başvuruda toplam 623 ihlal iddiası bulunmaktadır. İhlal iddialarının %46.2’si Onur Kurulu’na iletilmiş ve %29.5’ine yaptırım uygulanmasına karar verilmiştir. Başvurular en çok hasta, hasta yakını ya da hastanın vekili tarafından yapılmıştır (%34.1. Hakkında başvuru yapılan hekimlerin %75.7’si uzman,  %23.3’ü pratisyen, %1.0’i asistan hekimdir. En sık üç ihlal iddiası sırasıyla; tanıtım kurallarının ihlali, haksız kazanç/rekabete aykırılık (%24.8, tıbbi uygulama hatası (%21.4 ve gerçek dışı belge/rapor düzenlemektir (%18.2. ‘Gerçek dışı belge/rapor düzenlemek’, ‘Hasta haklarına saygısızlık’ ve ‘Aracılık, yönlendirme, çıkar ilişkisi’ eylemlerinin Sağlıkta Dönüşüm Programı (SDP sonrasında anlamlı derecede arttığı saptanmıştır. Zorunlu Mesleki Sorumluluk Sigortası sonrasında tıbbi uygulama hatası iddialarının sayısında ve bu iddia sonucunda yaptırıma uğratılan hekim sayısında anlamlı bir artış saptanmamıştır. Niedoczynność tarczycy, najczęściej będąca skutkiem przewlekłego autoimmunologicznego zapalenia, jest jedną z najczęstszych patologii gruczołu tarczowego i dotyczy około 2% populacji. Niedobór hormonów tarczycy manifestuje się zwykle zmniejszeniem napędu życiowego, stanami depresyjnymi, pogorszeniem sprawności intelektualnej, nietolerancją niższych temperatur. Patologia ta wiąże się ze zwiększonym ryzykiem  nadwagi lub otyłości, wtórnie do zwolnionego metabolizmu, a także jest czynnikiem sprzyjającym hiperlipidemii, co wiąże się ze wzrostem ryzyka sercowo-naczyniowego. Celem pracy była ocena występowania nadwagi i otyłości w grupie pacjentów z niedoczynnością tarczycy. Materiał i metody. W badaniu wzięło udział 101 pacjentów (77K, 24M, w wieku średnio 43 lat z rozpoznaną niedoczynnością tarczycy. W toku badań zastosowano metodę sondażu diagnostycznego z wykorzystaniem autorskiego kwestionariusza ankiety oraz metodę pomiaru parametrów antropometrycznych, w tym BMI, obwodu talii i bioder. Wyniki. Wśród badanych pacjentów nadwagę (BMI 25-29,9 miało 39,6% badanych, otyłość I stopnia (BMI 30-34,9 26,7% pacjentów, otyłość II stopnia (BMI 35-29,9- 10,9%. Wartości BMI≥40 cechujące otyłość III stopnia stwierdzono u 2% badanych. Średnia wartość BMI wyniosła 28,9, przy czym pacjenci w wieku 18-25 lat istotnie częściej cechowali się prawidłową masą ciała niż badani w grupie wiekowej ≥65 lat (p=0,0003, podobnie pacjenci wieku 26-40 lat w porównaniu z badanymi ≥65 lat (p=0,001.  Wnioski. Nadwaga i otyłość dotyczą większości pacjentów z niedoczynnością tarczycy, a ryzyko to rośnie wraz z wiekiem. Wartym rozważenia jest
