    Full Text Available No Brasil, estima-se que as intoxicações por plantas tóxicas que contém monofluoroacetato de sódio (MFA causam a morte de aproximadamente 500.000 bovinosao ano. A inoculação ruminal de bactérias que degradam MFA tem sido proposta como uma forma de prevenir a intoxicação. O presente trabalho teve como objetivo avaliar, em caprinos, a resistência ao MFA presente em Amorimia septentrionalis, induzida por inoculação ruminal das bactérias Pigmentiphaga kullae e Ancylobacter dichloromethanicus. Doze caprinos, que nunca tiveram contato prévio com plantas que contêm MFA, foram divididos em dois grupos, com seis animais cada. No grupo 1, 60 mL de uma mistura das duas bactérias foi inoculada, diariamente, durante 10 dias em cada caprino. No grupo 2, os caprinos não receberam as bactérias. A partir do 10º dia de inoculação, A. septentrionalis foi administrada, diariamente, na dose de 5g/kg de peso vivo, sendo interrompida em cada animal após a observação dos primeiros sinais clínicos da intoxicação. Os caprinos do grupo 1 apresentaram sinais clínicos 5,83±2,56 dias após a administração da planta o que diferiu significativamente (p=0,037 dos caprinos do grupo 2, que apresentaram sinais clínicos aos 2,67±0,52 dias. A quantidade de planta ingerida pelos caprinos inoculados (28,83±12,97g/kg e os não inoculados (12,03±3,65g/kg para desencadear os sinais clínicos foi, também, estatisticamente diferente entre os grupos (p=0,025. Conclui-se que a administração intraruminal de Pigmentiphaga kullae e Ancylobacter dichloromethanicus induz resistência à intoxicação por plantas que contêm MFA.

    Two Gram-stain-negative bacterial strains, SAP-32T and SAP-36, were isolated from sap drawn from the Acer pictum from Mount Halla in Jeju, Republic of Korea. The organisms were strictly aerobic, non-sporulating, motile rods and showed growth at 10-30 °C, pH 7-8 and with 0-2 % NaCl. The major isoprenoid quinone was Q-8. The predominant fatty acids were C16 : 0, cyclo-C17 : 0, summed feature 3 and C18 : 0. The polar lipids contained phosphatidylcholine, phosphatidylethanolamine, phosphatidylglycerol, an unknown aminophosphoglycolipid, an unknown glycolipid, an unknown phospholipid and two unknown lipids. The DNA G+C content was 64.4 mol%. The results of phylogenetic analyses based on 16S rRNA gene sequences indicated that SAP-32T and SAP-36 formed a distinct cluster with members of the genus Pigmentiphaga within the family Alcaligenaceae. Both strains showed 16S rRNA gene sequence similarity of 100 % to each other. The closest relatives of the isolates were Pigmentiphaga daeguensis (97.08 % sequence similarity), Pigmentiphaga kullae (97.01 %) and Pigmentiphaga litoralis (96.73 %). On the basis of data from phenotypic, chemotaxonomic and phylogenetic analyses, SAP-32T (=KCTC 52619T=DSM 104039T) and SAP-36 (=KCTC 52620=DSM 104072) represent members of a novel species of the genus Pigmentiphaga, for which the name Pigmentiphaga aceris sp. nov. is proposed.

    Catalases are essential components of the cellular equipment used to cope with oxidative stress. The monofunctional catalase KatP was purified from Pigmentiphaga sp. using ammonium sulfate precipitation (ASP), diethylaminoethyl ion exchange chromatography (IEC), and hydrophobic interaction chromatography (HIC). The purified catalase formed polymer with an estimated monomer molecular mass of 54kDa, which were resolved by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and zymogram analysis. KatP exhibited a specific catalytic activity of 73,000U/mg, which was higher than that of catalase-1 of Comamonas terrigena N3H (55,900U/mg). Seven short tryptic fragments of this catalase were obtained by electrospray ionization quadrupole time-of-flight mass spectrometry (ESI-Q-TOF MS/MS), and the gene, katP, was cloned by PCR amplification and overexpressed in Escherichia coli BL21 (DE3). Based on the complete amino acid sequence, KatP was identified as a clade 3 monofunctional catalase. The specific activities of recombinant KatP for hydrogen peroxide (690,000U/mg) increased 9-fold over that of the parent strain. The Km and Vmax of recombinant KatP were 9.48mM and 81.2mol/minmg, respectively. The optimal pH and temperature for KatP were 7.0 and 37°C, respectively, and the enzyme displayed abroad pH-stable range of 4.0-11.0. The enzyme was inhibited by Zn(2+), Cu(2+), Cr(2+), and Mn(2+), whereas Fe(3+) and Mg(2+) stimulated KatP enzymatic activity. Interestingly, the catalase activity of recombinant KatP displayed high stability under different temperature and pH conditions, suggesting that KatP is a potential candidate for the production of catalase.

    Full Text Available Burkholderia sp. K24, formerly known as Acinetobacter lwoffii K24, is a soil bacterium capable of utilizing aniline as its sole carbon and nitrogen source. Genomic sequence analysis revealed that this bacterium possesses putative gene clusters for biodegradation of various monocyclic aromatic hydrocarbons (MAHs, including benzene, toluene, and xylene (BTX, as well as aniline. We verified the proposed MAH biodegradation pathways by dioxygenase activity assays, RT-PCR, and LC/MS-based quantitative proteomic analyses. This proteogenomic approach revealed four independent degradation pathways, all converging into the citric acid cycle. Aniline and p-hydroxybenzoate degradation pathways converged into the β-ketoadipate pathway. Benzoate and toluene were degraded through the benzoyl-CoA degradation pathway. The xylene isomers, i.e., o-, m-, and p-xylene, were degraded via the extradiol cleavage pathways. Salicylate was degraded through the gentisate degradation pathway. Our results show that Burkholderia sp. K24 possesses versatile biodegradation pathways, which may be employed for efficient bioremediation of aniline and BTX.

    Galaxy counts in the K band, (J-K)-colors, and apparent size distributions of faint galaxies in the Subaru Deep Field (SDF) down to K~24.5 were studied in detail. Special attention has been paid to take into account various selection effects including the cosmological dimming of surface brightness, to avoid any systematic bias which may be the origin of controversy in previously published results. We also tried to be very careful about systematic model uncertainties; we present a comprehensive surveys of these systematic uncertainties and dependence on various parameters. We found that the pure luminosity evolution (PLE) model is well consistent with all the SDF data down to K~22.5, without any evidence for number or size evolution in a low-density, Lambda-dominated flat universe which is now favored by various cosmological observations.     A membrane-anchored cytochrome c-550, which is highly expressed in obligately alkaliphilic Bacillus clarkii K24-1U, was purified and characterized. The protein contained a conspicuous sequence of Gly(22)-Asn(34), in comparison with the other Bacillus small cytochromes c. Analytical data indicated that the original and lipase-treated intermediate forms of cytochrome c-550 bind to fatty acids of C(15), C(16) and C(17) chain lengths and C(15) chain length, respectively, and it was considered that these fatty acids are bound to glycerol-Cys(18). Since there was a possibility that the presence of a diacylglycerol anchor contributed to the formation of dimeric states of this protein (20 and 17 kDa in SDS-PAGE), a C18M (Cys(18) --> Met)-cytochrome c-550 was constructed. The molecular mass of the C18M-cytochrome c-550 was determined as 15 and 10 kDa in SDS-PAGE and 23 kDa in blue native PAGE. The C18M-cytochrome c-550 bound with or without Triton X-100 formed a tetramer as the original cytochrome c-550 bound with Triton X-100, as determined by gel filtration. The midpoint redox potential of cytochrome c-550 as determined by redox titration was +83 mV, while that determined by cyclic voltammetric measurement was +7 mV. The above results indicate that cytochrome c-550 is a novel cytochrome c.

    Leucine-Rich Amelogenin Protein (LRAP) is a member of the amelogenin family of biomineralization proteins, proteins which play a critical role in enamel formation. Recent studies have revealed the structure and orientation of the N- and C-terminus of LRAP bound to hydroxyapatite (HAP), a surface used as an analog of enamel. The structure of one region, K24 to S28, was found to be sensitive to phosphorylation of S16, the only naturally observed site of serine phosphorylation in LRAP, suggesting that the residues from K24 to S28 may sit at a key region of structural flexibility and play a role in the protein’s function. In this work, we investigated the sensitivity of the structure and orientation of this region when bound to HAP as a function of several factors which may vary during enamel formation to influence structure: the ionic strength (0.05 M, 0.15 M, 0.2 M), the calcium concentration (0.07 mM and 0.4 mM), and the surface to which it is binding (HAP and carbonated apatite (CAP), a more direct mimic of enamel). A naturally occurring mutation found in amelogenin (T21I), was also investigated. The structure in the K24S28 region of the protein was found to be sensitive to these conditions, with the CAP surface and excess Ca2+ (8:1 [Ca2+]:[LRAP-K24S28(+P)]) resulting in a much tighter helix, while low ionic strength relaxed the helical structure. Higher ionic strength and the point mutation did not result in any structural change in this region. The distance of the backbone of K24 from the surface was most sensitive to excess Ca2+ and in the T21I-mutation. Collectively, these data suggest that the protein is able to accommodate structural changes while maintaining its interaction with the surface, and provides further evidence of the structural sensitivity of the K24 to S28 region, a sensitivity that may contribute to function in biomineralization.   15. Isolation and Identification of Sodium Fluoroacetate Degrading Bacteria from Caprine Rumen in Brazil

    Full Text Available The objective of this paper was to report the isolation of two fluoroacetate degrading bacteria from the rumen of goats. The animals were adult goats, males, crossbred, with rumen fistula, fed with hay, and native pasture. The rumen fluid was obtained through the rumen fistula and immediately was inoculated 100 μL in mineral medium added with 20 mmol L−1 sodium fluoroacetate (SF, incubated at 39°C in an orbital shaker. Pseudomonas fluorescens (strain DSM 8341 was used as positive control for fluoroacetate dehalogenase activity. Two isolates were identified by 16S rRNA gene sequencing as Pigmentiphaga kullae (ECPB08 and Ancylobacter dichloromethanicus (ECPB09. These bacteria degraded sodium fluoroacetate, releasing 20 mmol L−1 of fluoride ion after 32 hours of incubation in Brunner medium containing 20 mmol L−1 of SF. There are no previous reports of fluoroacetate dehalogenase activity for P. kullae and A. dichloromethanicus. Control measures to prevent plant intoxication, including use of fences, herbicides, or other methods of eliminating poisonous plants, have been unsuccessful to avoid poisoning by fluoroacetate containing plants in Brazil. In this way, P. kullae and A. dichloromethanicus may be used to colonize the rumen of susceptible animals to avoid intoxication by fluoroacetate containing plants.

    Allergic diseases belong to one of the most common diseases with steadily increasing incidence even among young children. There is an urgent need to identify a prognostic marker pointing to increased risk of allergy development enabling early preventive measures introduction. It has been shown that administration of selected probiotic strains or mixtures could prevent allergy development. In our study, we have tested the capacity of probiotic strain Escherichia coli O83:K24:H31 (E. coli O83) to promote dendritic cell (DC) maturation and polarisation of immune responses. Increased presence of activation marker CD83 was observed on DC stimulated by E. coli O83 and DC of newborns of allergic mothers have significantly more increased cell surface presence of CD83 in comparison to children of healthy mothers. Increased gene expression and secretion of IL-10 was detected in DC stimulated with E. coli O83 being higher in DC of newborns of healthy mothers in comparison to allergic ones. Generally, increased presence of intracellular cytokines (IL-4, IL-13, IFN-gamma, IL-17A, IL-22, IL-10) was detected in CD4+ T cells cocultured with DC of children of allergic mothers in comparison to healthy ones. E. coli O83 primed DC significantly increased IL-10 and IL-17A in CD4 T cells of newborns of healthy mothers in comparison to the levels detected in CD4 T cells cocultured with control non-stimulated DC. We can conclude E. coli O83 induces dendritic cell maturation and IL-10 production in DC. Newborns of allergic mothers have generally increased reactivity of both DC and CD4 T cells which together with decreased capacity of DC of newborns of allergic mothers to produce IL-10 could support inappropriate immune responses development after allergen encounter.     In accordance with DOE Regulations for Compliance with Floodplain/Wetlands Environmental Review Requirements, wetland surveys were conducted in selected areas within the K-25 Area of Responsibility during the summer of 1994. These areas are Mitchell Branch, Poplar Creek, the K-770 OU, Duct Island Peninsula, the Powerhouse area, and the K-25 South Corner. Previously surveyed areas included in this report are the main plant area of the K-25 Site, the K-901 OU, the AVLIS site, and the K-25 South Site. Wetland determinations were based on the USACE methodology. Forty-four separate wetland areas, ranging in size from 0.13 to 4.23 ha, were identified. Wetlands were identified in all of the areas surveyed with the exception of the interior of the Duct Island Peninsula and the main plant area of the K-25 Site. Wetlands perform functions such as floodflow alteration, sediment stabilization, sediment and toxicant retention, nutrient transformation, production export, and support of aquatic species and wildlife diversity and abundance. The forested, scrub-shrub, and emergent wetlands identified in the K-25 area perform some or all of these functions to varying degrees.

    A quantum statistical model of nuclear multifragmentation is proposed. The recurrence equation method used within the canonical ensemble makes the model solvable and transparent to physical assumptions and allows to get results without involving the Monte Carlo technique. The model exhibits the first-order phase transition. Quantum statistics effects are clearly seen on the microscopic level of occupation numbers but are almost washed out for global thermodynamic variables and the averaged observables studied. In the latter case, the recurrence relations for multiplicity distributions of both intermediate-mass and all fragments are derived and the specific changes in the shape of multiplicity distributions in the narrow region of the transition temperature is stressed. The temperature domain favorable to search for the HBT effect is noted.

    In this paper, the stability of disclination ring in nematic liquid crystals is studied. In the presence of saddle-splay elasticity (characterized by k24) the disclination ring has a universal equilibrium radius. Depending on the values of the saddle-splay constant k24, the universal equilibrium radius is altered. When k24 > 0.92k (m=1/2) and k24>0.88k (m = -1/2), the disclination will be a point rather than a ring, where k is the Frank elastic constant in the one-constant approximation.

  17. Diversity of bacteria carried by pinewood nematode in USA and phylogenetic comparison with isolates from other countries.

    Full Text Available Pine wilt disease (PWD is native to North America and has spread to Asia and Europe. Lately, mutualistic relationship has been suggested between the pinewood nematode (PWN, Bursaphelenchus xylophilus the causal nematode agent of PWD, and bacteria. In countries where PWN occurs, nematodes from diseased trees were reported to carry bacteria from several genera. However no data exists for the United States. The objective of this study was to evaluate the diversity of the bacterial community carried by B. xylophilus, isolated from different Pinus spp. with PWD in Nebraska, United States. The bacteria carried by PWN belonged to Gammaproteobacteria (79.9%, Betaproteobacteria (11.7%, Bacilli (5.0%, Alphaproteobacteria (1.7% and Flavobacteriia (1.7%. Strains from the genera Chryseobacterium and Pigmentiphaga were found associated with the nematode for the first time. These results were compared to results from similar studies conducted from other countries of three continents in order to assess the diversity of bacteria with associated with PWN. The isolates from the United States, Portugal and China belonged to 25 different genera and only strains from the genus Pseudomonas were found in nematodes from all countries. The strains from China were closely related to P. fluorescens and the strains isolated from Portugal and USA were phylogenetically related to P. mohnii and P. lutea. Nematodes from the different countries are associated with bacteria of different species, not supporting a relationship between PWN with a particular bacterial species. Moreover, the diversity of the bacteria carried by the pinewood nematode seems to be related to the geographic area and the Pinus species. The roles these bacteria play within the pine trees or when associated with the nematodes, might be independent of the presence of the nematode in the tree and only related on the bacteria's relationship with the tree.

    In many of the DNA-based stable-isotope probing (SIP) studies published to date in which soil communities were investigated, a single DNA extraction was performed on the soil sample, usually using a commercial DNA extraction kit, prior to recovering the 13C-labeled (heavy) DNA by density-gradient ultracentrifugation. Recent evidence suggests, however, that a single extraction of a soil sample may not lead to representative recovery of DNA from all of the organisms in the sample. To determine whether multiple DNA extractions would affect the DNA yield, the eubacterial 16S rRNA gene copy number, or the identification of anthracene-degrading bacteria, we performed seven successive DNA extractions on the same aliquot of contaminated soil either untreated or enriched with [U-13C]anthracene. Multiple extractions were necessary to maximize the DNA yield and 16S rRNA gene copy number from both untreated and anthracene-enriched soil samples. Sequences within the order Sphingomonadales, but unrelated to any previously described genus, dominated the 16S rRNA gene clone libraries derived from 13C-enriched DNA and were designated “anthracene group 1.” Sequences clustering with Variovorax spp., which were also highly represented, and sequences related to the genus Pigmentiphaga were newly associated with anthracene degradation. The bacterial groups collectively identified across all seven extracts were all recovered in the first extract, although quantitative PCR analysis of SIP-identified groups revealed quantitative differences in extraction patterns. These results suggest that performing multiple DNA extractions on soil samples improves the extractable DNA yield and the number of quantifiable eubacterial 16S rRNA gene copies but have little qualitative effect on the identification of the bacterial groups associated with the degradation of a given carbon source by SIP. Multiple extractions were necessary to maximize the DNA yield and 16S rRNA gene copy number from both untreated and anthracene-enriched soil samples. Sequences within the order Sphingomonadales, but unrelated to any previously described genus, dominated the 16S rRNA gene clone libraries derived from (13)C-enriched DNA and were designated "anthracene group 1." Sequences clustering with Variovorax spp., which were also highly represented, and sequences related to the genus Pigmentiphaga were newly associated with anthracene degradation. The bacterial groups collectively identified across all seven extracts were all recovered in the first extract, although quantitative PCR analysis of SIP-identified groups revealed quantitative differences in extraction patterns. These results suggest that performing multiple DNA extractions on soil samples improves the extractable DNA yield and the number of quantifiable eubacterial 16S rRNA gene copies but have little qualitative effect on the identification of the bacterial groups associated with the degradation of a given carbon source by SIP.

    Full Text Available Bu çalışmada, zeytin karasuyund a fenol ve lignin gibi bileşenlerin giderilmesine çalışıldı. I;eneylerde, elektrokimyasal ön arıtıma (20volt ve 20dak. tabi tutulan ve tutulmayan iki farklı karakterdeki atık su örnekleri kullanıldı. Adsorbent olarak, aktif karbon, ham ve aktiYe edilmiş sepiyolit mineralleri seçildi. Deneylerde, süre ve pH'mn giderime etkisi incelendi. Buna göre; ön arıtınn yapılan karasuda fenol ve lignin konsantrasyonu belirgin oranda azalma gösterdi. Ayrıca, her iki atıksu ö rneği için adsorpsiyon bir saatlik sürede dengeye ulaşırken., aktif karbon ile maksimum verim sağlandı. Bunun yanında aktif sepiyolitinde belirgin bir adsorpsiyon etkinliği gösterdiği ve aktif karbona alternatif olarak kulla nılabileceği görüldü. pH'nın gidcrime etkisi inc.elendiğinde, genel olarak ön arıtıma tabi tutulan karasuda düşük pH, ham suda ise yüksek pH 'da fen ol ve lignin gibi kirletici bileşenler in daha gazla giderildiği görü ldü.

    Oral microbiome impacts health and disease. T2DM and periodontitis are associated. Neem (Azadiracta indica) has antibacterial activity against oral microbiota. To characterize oral microbiota (OMB) in saliva samples of T2DM patients by Next generation sequencing. To analyze MCP-1 levels among the T2DM patients before and after a month of neem stick usage as a toothbrush. Blood and saliva samples were collected from adult T2DM patients before and after the neem stick usage. Metagenomic sequencing was performed on saliva samples targeting V6 region of 16s rRNA. Serum MCP-1 levels were determined using a quantitative sandwich Human MCP-1 standard ABTS development kit (Peprotech, USA). The profile of oral microbiota of T2DM patients (n=24) consists of Streptococcus (95.8%) counts ranging from 2644 to 27,214, Veillonella (72.2%, counts 25-19,709, Neisseria (87.5%) 453-33,445), Rothia (63.6%, 233-6734), Actinomycetes (25%, 161-3730), Fusobacterium (21%, 2252-21,334), and Pigmentiphaga (12.5% 3-16,644). Oral microbiota in healthy controls (n=10), consists of Streptococcus (26.1%), Veillonella (21.9%), Neisseria (16.9%), Haemophilus (10.7%), Actinomycetes (2.6%), Rothia (3.1%), Oribacterium (1.7%). Post neem samples showed drastic reduction in the load of bacteria which was statistically significant. The mean serum MCP-1 before the use of neem stick was 265.18±79.44 (range 141.6-980.5pg/ml) and dropped to 33.6±7.35 after a month of neem stick usage (P value>0.001). OMB of T2DM patients and healthy controls were similar, however bacterial loads were significantly higher in T2DM patients. Use of neem stick has a statistically significant reduction on bacterial loads and MCP-1 levels in T2DM patients. Copyright © 2017 Elsevier B.V. All rights reserved.

    The nonlinear dynamical equations of tilted support spring system with critical components were established under the action of semi-sinusoidal pulse, and the equations were solved using Runge-Kulla method. The shock response characteristics of the critical components were obtained and analyzed. To evaluate the shock, a new concept of shock response spectra was proposed. The ratio of the maximum shock response acceleration of the critical components to the peak pulse acceleration and the dimensionless pulse duration were employed as the basic parameters of the shock response spectra. Based on the numerical results, the effects of the peak pulse acceleration, the dimensionless pulse duration, the angle of the tilted support spring and the frequency ratio of the system on the shock response were discussed. It is shown that the effects of the angle of the tilted support spring and the frequency ratio of the system are particularly noticeable. By increasing frequency ratio of the system, the maximum shock response acceleration of the critical components can be obviously decreased, while the peak of the shock response of the critical components can be decreased by increasing mass ratio at low frequency ratio.%以考虑易损件的斜支承弹簧系统为研究对象,建立系统几何非线性动力学方程,利用龙格-库塔法对易损件冲击响应特性进行数值分析.以易损件加速度响应峰值与脉冲激励幅值之比为易损件在半正弦波脉冲激励作用下的响应指标,无量纲脉冲激励时间作为变量,构建易损件的冲击响应谱.通过数值分析讨论脉冲激励幅值、脉冲激励时间、系统支承角以及频率比等对易损件冲击响应的影响规律.研究表明,系统支撑角、频率比等对易损件冲击响应影响显著,增加频率比可使易损件加速度响应峰值明显降低,低频率比条件下增大质量比可抑制易损件加速度响应峰值.

As a result, if k24k(1-(1/2)ln(R1/rc)), for small particles the Saturn-ring defect is more stable, and contrarily for large particles the dipole de-fect is more stable. The results demonstrate that a transition from the Saturn-ring defect to the dipole defect is induced either by increasing the particle size, the saddle-splay elastic con-stant k24 or by decreasing k.

    Acetylation of nucleosome histones results in relaxation of DNA and its availability for the transcriptional regulators, and is generally associated with the enhancement of gene expression. Although it is well known that activation of a variety of pro-adaptive genes represents a key event in the development of brain hypoxic/ischemic tolerance, the role of epigenetic mechanisms, in particular histone acetylation, in this process is still unexplored. The aim of the present study was to investigate changes in acetylation of histones in vulnerable brain neurons using original well-standardized model of hypobaric hypoxia and preconditioning-induced tolerance of the brain. Using quantitative immunohistochemistry and Western blot, effects of severe injurious hypobaric hypoxia (SH, 180mm Hg, 3h) and neuroprotective preconditioning mode (three episodes of 360mm Hg for 2h spaced at 24h) on the levels of the acetylated proteins and acetylated H3 Lys24 (H3K24ac) in the neocortex and hippocampus of rats were studied. SH caused global repression of the acetylation processes in the neocortex (layers II-III, V) and hippocampus (CA1, CA3) by 3-24h, and this effect was prevented by the preconditioning. Moreover, hypoxic preconditioning remarkably increased the acetylation of H3K24 in response to SH in the brain areas examined. The preconditioning hypoxia without subsequent SH also stimulated acetylation processes in the neocortex and hippocampus. The moderately enhanced expression of the acetylated proteins in the preconditioned rats was maintained for 24h, whereas acetylation of H3K24 was intense but transient, peaked at 3h. The novel data obtained in the present study indicate that large activation of the acetylation processes, in particular acetylation of histones might be essential for the development of brain hypoxic tolerance.

    The conditions present during enamel crystallite development change dramatically as a function of time, including the pH, protein concentration, surface type and ionic strength. In this work, we investigate the role that two of these changing conditions, pH and ionic strength, have in modulating the interaction of amelogenin, LRAP, with hydroxyapatite (HAP). Using solid state NMR dipolar recoupling and chemical shift data, we investigate the structure, orientation and dynamics of three regions in the N-terminus of the protein, L15 to V19, V19 to L23 and K24 to S28. These regions are also near the only phosphorylated residue in the protein, pS16, therefore, changes in the LRAP-HAP interaction as a function of phosphorylation (LRAP(-P) vs. LRAP(+P)) were also investigated. All of the regions and conditions studies for the surface immobilized proteins showed restricted motion, with more mobility under all conditions for L15(+P) and K24(-P). The structure and orientation of the LRAP-HAP interaction in the N-terminus of the phosphorylated protein is very stable to changing solution conditions. From REDOR dipolar recoupling data, the structure and orientation in the region L15V19(+P) did not change significantly as a function of pH or ionic strength. The structure and orientation of the region V19L23(+P) were also stable to changes in pH, with the only significant change observed at high ionic strength, where the region becomes extended, suggesting this may be an important region in regulating mineral development. Chemical shift studies also suggest minimal changes in all three regions studied for both LRAP(-P) and LRAP(+P) as a function of pH or ionic strength. Phosphorylation also alters the LRAP-HAP interface. All of the three residues investigated (L15, V19, and K24) are closer to the surface in LRAP(+P), but K24S28 also changes structure as a result of phosphorylation, from a random coil to a largely helical structure, and V19L23 becomes more extended at high ionic

    Stapling peptides for inhibiting the p53/MDM2 interaction is a promising strategy for developing anti-cancer therapeutic leads. We evaluate double-click stapled peptides formed from p53-based diazidopeptides with different staple positions and azido amino acid side-chain lengths, determining the impact of these variations on MDM2 binding and cellular activity. We also demonstrate a K24R mutation, necessary for cellular activity in hydrocarbon-stapled p53 peptides, is not required for analogous 'double-click' peptides.

    We use Spitzer Space Telescope 24 {\\mu}m data to search for debris disks among 122 AFGKM stars from the \\sim 670 Myr clusters Hyades, Coma Ber, and Praesepe, utilizing a number of advances in data reduction and determining the intrinsic colors of main sequence stars. For our sample, the 1{\\sigma} dispersion about the main sequence V-K, K-[24] locus is approximately 3.1%. We identify seven debris disks at 10% or more (\\geq 3{\\sigma} confidence level) above the expected K-[24] for purely photospheric emission. The incidence of excesses of 10% or greater in our sample at this age is 5.7 +3.1/-1.7%. Combining with results from the literature, the rate is 7.8 +4.2/-2.1% for early- type (B9 - F4) stars and 2.7 +3.3/-1.7% for solar-like (F5 - K9) stars. Our primary sample has strict criteria for inclusion to allow comparison with other work; when we relax these criteria, three additional debris disks are detected.   5. The Effect of Molecular Anchoring and Curvature on Confined Liquid Crystals

    Nematic liquid crystals confined to curved geometries exhibit unique elastic and anchoring properties. One result of this study was the first simultaneous measurement of the azimuthal (W_phi) and polar (W_theta) anchoring strength and the saddle-splay surface elastic constant K_{24}. The technique confined 4^'-pentyl-4 -cyanobiphenyl (5CB) to submicrometer polyimide treated cylindrical cavities of polycarbonate (Nuclepore) membranes with concentric tangential anchoring conditions for deuterium nuclear magnetic resonance studies. A structural transition upon increasing cavity radius from a planar-bipolar (PB, R 0.5 mu m) structure resulted in the measurement of W _theta and W_phi (comparable in these systems), and of K _{24}/K. The effect of the bend elastic constant K _{33} on the escaped nematic director -field of 4^'-octyl-cyanobiphenyl (8CB) in the submicrometer cavities of Nuclepore membranes is significant near the nematic-smectic A transition. The project encompassed monitoring the anchoring angle, measuring the defect density, and estimating the values for W _theta and K_{24 }/K. The lack of pretransitional smectic layering was attributed to the roughness of the cavity wall. The effect of surface treatments on the nematic structures in the submicrometer cavities of alumina (Anopore) membranes was systematically studied by treating the cavity walls with aliphatic acids (C_{n} H_{2n+1}-COOH) of varying carbon number, n. A configuration transition upon decreasing n from a planar-polar (PP, n>7) to a parallel-axial (PA, n<7) structure indicated a discontinuous anchoring transition from homeotropic to planar anchoring at n = 7. Stable nematic director-fields of chiral nematics in Anopore membranes revealed a structural transition from a PA to a twisted PB (TPB) structure as the percentage of chiral additive increased. The TPB structure is the same as the PB structure in-plane, but the symmetry axis twists along the cylinder axis to accommodate the intrinsic twist

    We present maps of 14.4 deg^2 of the Ophiuchus dark clouds observed by the Spitzer Space Telescope Multiband Imaging Photometer for Spitzer (MIPS). These high quality maps depict both numerous point sources as well as extended dust emission within the star-forming and non-star-forming portions of these clouds. Using PSF-fitting photometry, we detect 5779 sources at 24 um and 81 sources at 70 um at the 10 sigma level of significance. Three hundred twenty-three candidate young stellar objects (YSOs) were identified according to their positions on the MIPS/2MASS K versus K$-$[24] color-magnitude diagrams as compared to 24 um detections in the SWIRE extragalactic survey. We find that more than half of the YSO candidates, and almost all the ones with protostellar Class I spectral energy distributions, are confined to the known cluster and aggregates.

    The development of a new synthesis method is still required for very fine oxide nanoparticles. In this study, a single-flow supercritical fluid system has been developed for the synthesis of highly crystalline nanosized oxide particles. Niobium oxide particles were synthesized by single-flow supercritical water treatment, batch-type supercritical water treatment and subcritical water treatment. Niobium pentoxide nanoparticles synthesized by single-flow supercritical water treatment at 673 K, 24.5 MPa, and 15 ml min-1 flow rate had a pseudohexagonal structure. The morphology of the nanoparticle was a rod, and it has a smaller particle size and larger crystallite size than those of the oxide particles synthesized by the other methods, because the particle growth and the decomposition of surfactant were rapidly suppressed in the single-flow supercritical water treatment. The nanosized niobium pentoxide is useful as a catalyst in harsh environments and as a precursor powder of lead-free piezoelectric materials.

  8. Interaction of Rahaliya-Ekhedhur groundwater with the aquifer rock, West Razzaza Lake, Central Iraq

    The groundwater of Dammam aquifer in Rahaliya-Ekhedhur area, West Razzaza, Iraq, was studied to identify the main hydrogeochemical processes and the groundwater-rock interaction. The results indicated that Na+ and SO4 2- are the dominant ions in the groundwater. The average contribution of cations in the aquifer is Na+ + K+ (24.7 %), Ca2+ (13.9 %), and Mg2+ (11.4 %), while anions contribution is SO4 2- (23.0 %), Cl- (20.7 %), and HCO3 - (6.3 %). The groundwater characterized by neutral to slightly alkaline hard water, excessively mineralized, and slightly brackish water type. Rock-water interaction processes are identified to include dissolution of carbonates, sulfates, halite, and clay minerals, leaching, and cation exchanges, with little impact of evaporation.

    We study the multi-instanton partition functions of the $\\Omega$-deformed $\\mathcal N =2^{*}$ $SU(2) $ gauge theory in the Nekrasov-Shatashvili (NS) limit. They depend on the deformation parameters $\\epsilon_{1}$, the scalar field expectation value $a$, and the hypermultiplet mass $m$. At fixed instanton number $k$, they are rational functions of $\\epsilon_{1}, a, m$ and we look for possible regularities that admit a parametrical description in the number of instantons. In each instanton sector, the contribution to the deformed Nekrasov prepotential has poles for "large" deformation parameters. To clarify the properties of these singularities we exploit Bethe/gauge correspondence and examine the special ratios $m/\\epsilon_{1}$ at which the associated spectral problem is $n$-gap.   10. Variation of Mid and Far-IR Luminosities among Early-Type Galaxies: Relation to Stellar Metallicity and Cold Dust

    The Hubble morphological sequence from early to late galaxies corresponds to an increasing rate of specific star formation. The Hubble sequence also follows a banana-shaped correlation between 24 and 70 micron luminosities, both normalized with the K-band luminosity. We show that this correlation is significantly tightened if galaxies with central AGN emission are removed, but the cosmic scatter of elliptical galaxies in both 24 and 70 micron luminosities remains significant along the correlation. We find that the 24 micron variation among ellipticals correlates with stellar metallicity, reflecting emission from hot dust in winds from asymptotic giant branch stars of varying metallicity. Infrared surface brightness variations in elliptical galaxies indicate that the K - 24 color profile is U-shaped for reasons that are unclear.     Development of rod photoreceptors in the mammalian retina is critically dependent on the basic motif-leucine zipper transcription factor NRL (neural retina leucine zipper). In the absence of NRL, photoreceptor precursors in mouse retina produce only cones that primarily express S-opsin. Conversely, ectopic expression of NRL in post-mitotic precursors leads to a rod-only retina. To explore the role of signaling molecules in modulating NRL function, we identified putative sites of post-translational modification in the NRL protein by in silico analysis. Here, we demonstrate the sumoylation of NRL in vivo and in vitro, with two small ubiquitin-like modifier (SUMO) molecules attached to the Lys-20 residue. NRL-K20R and NRL-K20R/K24R sumoylation mutants show reduced transcriptional activation of Nr2e3 and rhodopsin promoters (two direct targets of NRL) in reporter assays when compared with wild-type NRL. Consistent with this, in vivo electroporation of the NRL-K20R/K24R mutant into newborn Nrl(-/-) mouse retina leads to reduced Nr2e3 activation and only a partial rescue of the Nrl(-/-) phenotype in contrast to the wild-type NRL that is able to convert cones to rod photoreceptors. Although PIAS3 (protein inhibitor of activated STAT3), an E3-SUMO ligase implicated in photoreceptor differentiation, can be immunoprecipitated with NRL, there appears to be redundancy in E3 ligases, and PIAS3 does not seem to be essential for NRL sumoylation. Our studies suggest an important role of sumoylation in fine-tuning the activity of NRL and thereby incorporating yet another layer of control in gene regulatory networks involved in photoreceptor development and homeostasis.

  16. Association between 24-hour urine sodium and potassium excretion and diet quality in six-year-old children: a cross sectional study

    Full Text Available Abstract Background Limited data is available on sodium (Na and potassium (K intake in young children estimated by 24 hour (24h excretion in urine. The aim was to assess 24h urinary excretion of Na and K in six-year-old children and its relationship with diet quality. Methods The study population was a subsample of a national dietary survey, including six-year-old children living in the greater Reykjavik area (n=76. Three day weighed food records were used to estimate diet quality. Diet quality was defined as adherence to the Icelandic food based dietary guidelines. Na and K excretion was analyzed from 24h urine collections. PABA check was used to validate completeness of urine collections. The associations between Na and K excretion and diet quality were estimated by linear regression, adjusting for gender and energy intake. Results Valid urine collections and diet registrations were provided by 58 children. Na and K excretion was, mean (SD, 1.64 (0.54 g Na/24h (approx. 4.1 g salt/24h and 1.22 (0.43 g K/24h. In covariate adjusted models Na excretion decreased by 0.16 g Na/24h (95% CI: 0.31, 0.06 per 1-unit increase in diet quality score (score range: 1–4 while K excretion was increased by 0.18 g K/24h (95% CI: 0.06, 0.29. Conclusions Na intake, estimated by 24h urinary excretion was on average higher than recommended. Increased diet quality was associated with lower Na excretion and higher K excretion in six-year-old children.

    [目的]以血清凝血酶原前体蛋白(PIVK-Ⅱ)来研究晚发型维生素K缺乏性出血(late vitamin K deficiency bleeding,LVKDB)患儿及其母亲维生素K营养状况,观察不同剂量维生素K对PIVKA-Ⅱ的影响.[方法]来用前瞻性研究方法,18例自2002年1月~2003年12月在沧州市中心医院住院的LVKDB患儿及其母亲组成观察组,患儿又分为5 mg和10 mg治疗组.20例同期住院的急性上呼吸道感染患儿及其母亲组成对照组.各组分别化验PIVKA-Ⅱ水平,观察组患儿在应用维生素K24 h、3 d、7 d三个时间检测.PIVKA-Ⅱ采用酶联免疫吸附试验方法.[结果]①应用维生素K24 h、3 d、7 d观察组患儿PIVKA-Ⅱ水平高于对照组患儿(P<0.01);②观察组母亲PIVKA-Ⅱ水平与对照组母亲无差异(P>0.05);③观察组患儿维生素K 5 mg和10 mg治疗组血清PIVKA-Ⅱ水平无差异(P>0.05).[结论]LVKDB患儿维生素K严重缺乏,而其母亲不存在维生素K不足,在LVKDB患儿治疗中没有必要应用大剂量维生素K.

O número cromossômico de G. striatula nas duas localidades foi 2n=34, com fórmula cariotípica 2K=24M+10A. Em Gnamptogenys sp., o número cromossômico foi de 2n=46 (fêmea e n=23 (machos, com a fórmula cariotípica 2K=18M+28A. O número cromossômico de G. annulata foi 2n=68 com a fórmula cariotípica 2K= 6M+62A. In Gnamptogenys sp., the chromosomal number was 2n=46 (females and n=23 (males, and its karyotype formula was 2K=18M+28A. The chromosomal number of G. annulata was 2n=68 with the karyotype 2K= 6M + 62A. This study complements THOse carried out by our research group on cytogenetics of the poneromorph ants (sensu Bolton and would contribute to the better understanding of the ant evolution in this group considered primitive.

  19. High-resolution x-ray crystal structures of the villin headpiece subdomain, an ultrafast folding protein

    The 35-residue subdomain of the villin headpiece (HP35) is a small ultrafast folding protein that is being intensely studied by experiments, theory, and simulations. We have solved the x-ray structures of HP35 and its fastest folding mutant [K24 norleucine (nL)] to atomic resolution and compared their experimentally measured folding kinetics by using laser temperature jump. The structures, which are in different space groups, are almost identical to each other but differ significantly from previously solved NMR structures. Hence, the differences between the x-ray and NMR structures are probably not caused by lattice contacts or crystal/solution differences, but reflect the higher accuracy of the x-ray structures. The x-ray structures reveal important details of packing of the hydrophobic core and some additional features, such as cross-helical H bonds. Comparison of the x-ray structures indicates that the nL substitution produces only local perturbations. Consequently, the finding that the small stabilization by the mutation is completely reflected in an increased folding rate suggests that this region of the protein is as structured in the transition state as in the folded structure. It is therefore a target for engineering to increase the folding rate of the subdomain from ≈0.5 μs–1 for the nL mutant to the estimated theoretical speed limit of ≈3 μs–1. PMID:15894611

    An accident of misidentification has brought to light the interesting system HD 191588, a new RS CVn-type spectroscopic binary. A radial-velocity study of the primary star, the only seen component, carried out at the Observatoire de Haute-Provence with the Coravel instrument and subsequently at the Cambridge Observatories with a similar one, reveals two orbital motions: a short-period orbit (60 days) and a long-period one (about 4.5 years), so this star is a triple system.     The present work aimed to evaluate 16S rRNA, khe and rpoB gene sequencing for the identification of Klebsiella pneumoniae in comparison with phenotypic methods. Fifteen clinical isolates were examined, which were initially identified as K. pneumoniae subsp. pneumoniae using the automated VITEK 32 system in two hospitals in Enshi City, China. Their identity was further supported by conventional phenotypic methods on the basis of morphological and biochemical characteristics. Using Bayesian phylogenetic analyses and haplotypes network reconstruction, 13 isolates were identified as K. pneumoniae, whereas the other two isolates (K19, K24) were classified as Shigella sp. and Enterobacter sp., respectively. Of the three genes, 16S rRNA and khe gene could discriminate the clinical isolates at the genus level, whereas rpoB could discriminate Klebsiella at the species and even subspecies level. Overall, the gene tree based on rpoB is more compatible with the currently accepted classification of Klebsiella than those based on 16S rRNA and khe genes, showing that rpoB can be a powerful tool for identification of K. pneumoniae isolates. Above all, our study challenges the utility of khe as a species-specific marker for identification of K. pneumoniae.

    Mitochondrial p53 is involved in apoptosis and tumor suppression. However, its regulation is not well studied. Here, we show that TRAF6 E3 ligase is a crucial factor to restrict mitochondrial translocation of p53 and spontaneous apoptosis by promoting K63-linked ubiquitination of p53 at K24 in cytosol, and such ubiquitination limits the interaction between p53 and MCL-1/BAK. Genotoxic stress reduces this ubiquitination in cytosol by S13/T330 phosphorylation-dependent translocation of TRAF6 from cytosol to nucleus, where TRAF6 also facilitates the K63-linked ubiquitination of nuclear p53 and its transactivation by recruiting p300 for p53 acetylation. Functionally, K63-linked ubiquitination of p53 compromised p53-mediated apoptosis and tumor suppression. Colorectal cancer samples with WT p53 reveal that TRAF6 overexpression negatively correlates with apoptosis and predicts poor response to chemotherapy and radiotherapy. Together, our study identifies TRAF6 as a critical gatekeeper to restrict p53 mitochondrial translocation, and such mechanism may contribute to tumor development and drug resistance.

    We study a renormalization group flow of ABJM theory embedded into the warped A_8 geometry and explore the dependence of the vacuum structure on the parameters of the theory. This model has a product group gauge structure U(N)xU(n+l) and comes equipped with discrete parameters N, l and k, a continuous parameter b related to the ratio of the Yang-Mills coupling for the two gauge groups, and one dimensionful parameter gYM^2 setting the overall scale. A supersymmetric supergravity solution exists when Q=N-l(l-k)/2k-k/24 is positive and is interpretable as a RG flow from a Yang-Mills like UV fixed point to a superconformal IR fixed point with free energy of order Q^3/2. The fate of the theory when Q is taken to be negative is less clear. We explore the structure of the possible gravity solution for small negative Q by considering the linearized gravitational back reaction from adding a small number of anti-branes on the Q=0 background.   4. Stability of Disclinations in Nematic Liquid Crystals

    In the light of φ-mapping method and topological current theory, the stability of disclinations around a spherical particle in nematic liquid crystals is studied. We consider two different defect structures around a spherical particle: disclination ring and point defect at the north or south pole of the particle. We calculate the free energy of these different defects in the elastic theory. It is pointed out that the total Frank free energy density can be divided into two parts. One is the distorted energy density of director field around the disclinations. The other is the free energy density of disclinations themselves, which is shown to be concentrated at the defect and to be topologically quantized in the unit of (k -k24)π/2. It is shown that in the presence of saddle-splay elasticity a dipole (radial and hyperbolic hedgehog) configuration that accompanies a particle with strong homeotropic anchoring takes the structure of a small disclination ring, not a point defect.

  5. The K-band luminosity functions of cluster galaxies

    We derive the galaxy luminosity function in the $K_s$ band for galaxies in 24 clusters to provide a local reference for higher redshift studies and to analyse how and if the luminosity function varies according to environment and cluster properties. We use new, deep $K$ band imaging and match the photometry to available redshift information and to optical photometry from the SDSS or the UKST/POSS: $>80\\%$ of the galaxies to $K \\sim 14.5$ have measured redshifts. We derive composite luminosity functions, for the entire sample and for cluster subsamples . We consider the luminosity functions for red sequence and blue cloud galaxies. The full composite luminosity function has $K^*=12.79 \\pm 0.14$ ($M_K=-24.81$) and $\\alpha=-1.41 \\pm 0.10$. We find that $K^*$ is largely unaffected by the environment but that the slope $\\alpha$ increases towards lower mass clusters and clusters with Bautz-Morgan type $<$ II.     We present new observations of the quiescent giant molecular cloud GCM0.253+0.016 in the Galactic center, using the upgraded Karl G. Jansky Very Large Array. Observations were made at wavelengths near 1 cm, at K (24 to 26 GHz) and Ka (27 and 36 GHz) bands, with velocity resolutions of 1-3 km/s and spatial resolutions of ~0.1 pc, at the assumed 8.4 kpc distance of this cloud. The continuum observations of this cloud are the most sensitive yet made, and reveal previously undetected emission which we attribute primarily to free-free emission from external ionization of the cloud. In addition to the sensitive continuum map, we produce maps of 12 molecular lines: 8 transitions of NH3 -- (1,1),(2,2),(3,3),(4,4),(5,5),(6,6),(7,7) and (9,9), as well as the HC3N (3-2) and (4-3) lines, and CH3OH 4(-1) - 3(0) the latter of which is known to be a collisionally-excited maser.   8. An 8×8/4×4 Adaptive Hadamard Transform Based FME VLSI Architecture for 4K×2K H.264/AVC Encoder

    Fidelity Range Extension (FRExt) (i.e. High Profile) was added to the H.264/AVC recommendation in the second version. One of the features included in FRExt is the Adaptive Block-size Transform (ABT). In order to conform to the FRExt, a Fractional Motion Estimation (FME) architecture is proposed to support the 8×8/4×4 adaptive Hadamard Transform (8×8/4×4 AHT). The 8×8/4×4 AHT circuit contributes to higher throughput and encoding performance. In order to increase the utilization of SATD (Sum of Absolute Transformed Difference) Generator (SG) in unit time, the proposed architecture employs two 8-pel interpolators (IP) to time-share one SG. These two IPs can work in turn to provide the available data continuously to the SG, which increases the data throughput and significantly reduces the cycles that are needed to process one Macroblock. Furthermore, this architecture also exploits the linear feature of Hadamard Transform to generate the quarter-pel SATD. This method could help to shorten the long datapath in the second-step of two-iteration FME algorithm. Finally, experimental results show that this architecture could be used in the applications requiring different performances by adjusting the supported modes and operation frequency. It can support the real-time encoding of the seven-mode 4K×2K@24fps or six-mode 4K×2K@30fps video sequences.

    We have substantially extended the high-temperature and low-magnetic-field (and the related low-temperature and high-magnetic-field) bivariate expansions of the free energy for the conventional three-dimensional Ising model and for a variety of other spin systems generally assumed to belong to the same critical universality class. In particular, we have also derived the analogous expansions for the Ising models with spin s=1,3/2,.. and for the lattice euclidean scalar field theory with quartic self-interaction, on the simple cubic and the body-centered cubic lattices. Our bivariate high-temperature expansions, which extend through K^24, enable us to compute, through the same order, all higher derivatives of the free energy with respect to the field, namely all higher susceptibilities.   10. Ira as a pioneer in audiology: His contributions to the clinical measurement of hearing and hearing impairment

    Ira Hirsh's contributions to clinical science and research are diverse and significant. In fact, approximately one-third of the 100+ publications that Ira lists in his curriculum vitae (CV) are clinical in nature, dealing with various aspects of audiology, deafness, hearing aids, aural rehabilitation, and speech and language pathology. The majority of these citations, fully one-quarter of his publication list, addresses problems specific to the clinical measurement of hearing and hearing impairment. Undoubtedly, the most influential of these published works appears in his CV under the citation ``The Measurement of Hearing.'' The forward for this publication, his only textbook, was penned in June, 1952 (now precisely half a century past at the time of this session). The aims of this presentation are to (1) provide perspective on the fundamental importance of his virtually timeless text in shaping the fledgling discipline of audiology, and (2) celebrate Ira's many contributions to the profession and practice of audiology. [Preparation for this presentation was supported, in part, by a K24 career development award from NIDCD.

  11. Paleosecular variation of directions from lava flows of the Xalapa Volcanic Field, Veracruz, Mexico

    We collected 21 monogenetic type lava flows (316 specimens) in the Xalapa Volcanic Field, eastern part of the Mexican Volcanic Belt. Geochronological results of twelve Ar-Ar determinations range from late Pleistocene to Holocene. The ages fall into three groups, those older than 2.0 Ma, those between 0.25-0.40 Ma and those less than approximately 0.1 Ma. All the samples were demagnetized by thermal and AF treatment, showing mostly a single stable component of magnetization with unblocking temperature above 530°C and/or 40-60 mT. We calculate the mean direction (D= 359.70°, I=27.4°, k=24, 95=7.7°) and the VGP to compare and integrate with previous paleosecular variation analyses. The paleosecular variation parameter, upper and lower limit are: SF=14.6, SU=17.5 and SL=11.2, respectively. So, the VGP dispersion is consistent with the expected value of latitude-dependent variation of McFadden for the last 5My.

  12. Relevance of saddle-splay elasticity in complex nematic geometries.

    We demonstrate the relevance of saddle-splay elasticity in nematic liquid crystalline fluids in the context of complex surface anchoring conditions and the complex geometrical confinement. Specifically, nematic cells with patterns of surface anchoring and colloidal knots are shown as examples where saddle-splay free energy contribution can have a notable role which originates from nonhomogeneous surface anchoring and the varying surface curvature. Patterned nematic cells are shown to exhibit various (meta)stable configurations of nematic field, with relative (meta)stability depending on the saddle-splay. We show that for high enough values of saddle-splay elastic constant K24 a previously unstable conformation can be stabilised, more generally indicating that the saddle-splay can reverse or change the (meta)stability of various nematic structures affecting their phase diagrams. Furthermore, we investigate saddle-splay elasticity in the geometry of highly curved boundaries - the colloidal particle knots in nematic - where the local curvature of the particles induces complex spatial variations of the saddle-splay contributions. Finally, a nematic order parameter tensor based saddle-splay invariant is shown, which allows for the direct calculation of saddle-splay free energy from the Q-tensor, a possibility very relevant for multiple mesoscopic modelling approaches, such as Landau-de Gennes free energy modelling.

    The force transmission law of a prestressed anchor-rope and the groundsill structure in rock mass were studied.The foundation mechanical model of nonuniform linear unipoium load under the premise of the ground surface suffering nonuniform linear load was established and deduced by the superposition principle,and the calculating formula for the stress and displacement at the depth z in the foundation of the beam axis was deduced for the prestressed anchor-rope and groundsill structure.Taking the slope of K24+233-K27+89 of Xupu-Huaihua Expressway as a experimental region,the force transmission law of prestressed anchor-rope and groundsill structure in slope stability were studied.The results show that when the anchor cable is provided with pre-stress,the actual prestress in the prestressed cable anchor can be transferred to the strengthening geotechnical body through the ground beam,of which the transfer mode tends to decay from the inside to the outside in the form of approximate ellipse iso-surface with the endpoints.The intersection of the beam axis and the plane are perpendicular to the axis of the beam.The compressed strain in the peripheral rock of two ground beams tends to increase with the increase of the depth of the anchor,and then tends to decay gradually and finally tends to be 0 after reaching its peak value.This distribution form is in accordance with the theoretical form.%建立受非均布线形荷载作用的地基力学模型,应用叠加原理推导出预应力锚索-地梁结构中垂直于梁轴线平面内的地基中深度z处的应力和位移计算公式,为研究预应力锚索-地梁结构在岩体中的传力规律提供理论依据.以溆怀(湖南溆浦—怀化)高速公路K24+233~K27+89边坡为实险区域研究预应力锚索地梁结构在边坡稳定中的作用机理.研究结果表明:在对锚索施加预应力后,锚索预应力通过地梁传递给被加固的岩土体,在垂直于地梁轴线的平面内,其传递方式以

    Anelasticity of the austenitic steel 24Ni5Mo and the stainless steel 12Cr9Ni4Mo has been investigated in the austenitic state after quenching with respect to isothermal martensitic transformation during cooling and subsequent heating. Maxima of anelasticity due to isothermal transformation at {approx}200 K (24Ni5Mo, 0.002% C) and {approx}250 K (12Cr9Ni4Mo; 0.01% C) coincided well with C-curve noses obtained by methods based on magnetic properties and electric resistivity. Corresponding internal friction maxima were found to be dependent on cooling or heating rate, quenching temperature and the frequency of oscillation and may therefore be described using the Delorme approximation. The activation energy of isothermal martensitic transformation calculated from the lower part of the C-curves estimated using the Borgenstam-Hillert and Arrhenius methods (3-8 kJ/mol for 24Ni5Mo and 15-20 kJ/mol for 12Cr9Ni4Mo) are comparable with the energy of impurity-dislocation interaction ({approx}10 kJ/mol) and interpreted as too low to be caused by diffusion processes: the activation energy for carbon diffusion in austenitic steel 24Ni5Mo is found to be {approx}135 kJ/mol and {approx}145 kJ/mol for austenite in two-phase 12Cr9Ni4Mo steel. An estimation showed that the activation energy for the isothermal martensitic transformation for the 24Ni5Mo alloy with so-called binary martensitic kinetics was higher in the vicinity of the nose of the C-curve, became lower with a decrease in temperature range and approached zero in the vicinity of the athermal martensitic point. A similar effect was not observed in the 12Cr9Ni4Mo steel. (orig.)

  15. Thermal Properties, Thermal Shock, and Thermal Cycling Behavior of Lanthanum Zirconate-Based Thermal Barrier Coatings

    Lanthanum zirconate (La2Zr2O7) coatings are newly proposed thermal barrier coating (TBC) systems which exhibit lower thermal conductivity and potentially higher thermal stability compared to other traditional thermal barrier systems. In this work, La2Zr2O7 and 8 wt pct yttria stabilized zirconia (8YSZ) single-layer and double-layer TBC systems were deposited using the air plasma spray technique. Thermal properties of the coatings were measured. Furnace heat treatment and jet engine thermal shock tests were implemented to evaluate coating performance during thermal cycling. The measured average thermal conductivity of porous La2Zr2O7 coating ranged from 0.59 to 0.68 W/m/K in the temperature range of 297 K to 1172 K (24 °C to 899 °C), which was approximately 25 pct lower than that of porous 8YSZ (0.84 to 0.87 W/m/K) in the same temperature range. The coefficients of thermal expansion values of La2Zr2O7 were approximately 9 to 10 × 10-6/K from 400 K to 1600 K (127 °C to 1327 °C), which were about 10 pct lower than those of porous 8YSZ. The double-layer coating system consisting of the porous 8YSZ and La2Zr2O7 layers had better thermal shock resistance and thermal cycling performance than those of single-layer La2Zr2O7 coating and double-layer coating with dense 8YSZ and La2Zr2O7 coatings. This study suggests that porous 8YSZ coating can be employed as a buffer layer in La2Zr2O7-based TBC systems to improve the overall coating durability during service.

  16. Radionuclide distribution of Holocene sediments and its effects on the habitat of recent foraminifers: A case study from the Western Marmara Sea (Turkey)

    ABSTRACT Radionuclides cause radioactive contamination in aquatic environments just as other non-biodegradable pollutants, such as heavy metals, sink to the seafloor and accumulate in the sediments. These radioactive pollutants especially affect benthic foraminifera living on the sediment surface or in the sediments in the seafloor. Foraminifera were used as bioindicators to analyze the effect of radioactivity pollution on ecosystems. In this study, we have investigated natural and artificial radionuclide (232Th, 226Ra, 40K and 137Cs) distribution in sediment samples taken in the living areas of benthic foraminifera in the Western Marmara Sea by means of gamma spectrometry. Accordingly, 29 core samples taken in 2016 from depths of about 20-35 m close to the shores of the Marmara Sea were used. Core samples representing the pollution of the study area were collected at locations such as discharge points for domestic and industrial areas, port locations, and others. Other samples were taken from areas unaffected or less affected by pollution. The radionuclide concentration activity values in the sediment samples obtained from the locations, in Bq/kg, were 137Cs, 0.9-9.4; 232Th, 18.9-86; 226Ra, 10-50; 40K, 24.4-670. These values were compared with the United Nations Scientific Committee on the Effects of Atomic Radiation (UNSCEAR) data, and an environmental analysis was carried out. The 226Ra series, the 232Th series, and the 40K radionuclides accumulate naturally, and they are also increasing continuously due to anthropogenic pollution. Although the 226Ra values obtained throughout the study areas remained within normal limits according to the UNSCEAR values, the 40K and 232Th series values were found to be higher in almost all locations. According to these results, the main causes of radioactive pollution in the investigation area are agricultural and mining activities.     The Static X-ray Imager (SXI) is a diagnostic used at the National Ignition Facility (NIF) to measure the position of the X-rays produced by lasers hitting a gold foil target. The intensity distribution taken by the SXI camera during a NIF shot is used to determine how accurately NIF can aim laser beams. This is critical to proper NIF operation. Imagers are located at the top and the bottom of the NIF target chamber. The CCD chip is an X-ray sensitive silicon sensor, with a large format array (2k x 2k), 24 μm square pixels, and 15 μm thick. A multi-anode Manson X-ray source, operating up to 10kV and 10W, was used to characterize and calibrate the imagers. The output beam is heavily filtered to narrow the spectral beam width, giving a typical resolution E/ΔE≈10. The X-ray beam intensity was measured using an absolute photodiode that has accuracy better than 1% up to the Si K edge and better than 5% at higher energies. The X-ray beam provides full CCD illumination and is flat, within ±1% maximum to minimum. The spectral efficiency was measured at 10 energy bands ranging from 930 eV to 8470 eV. We observed an energy dependent pixel sensitivity variation that showed continuous change over a large portion of the CCD. The maximum sensitivity variation occurred at 8470 eV. The geometric pattern did not change at lower energies, but the maximum contrast decreased and was not observable below 4 keV. We were also able to observe debris, damage, and surface defects on the CCD chip. The Manson source is a powerful tool for characterizing the imaging errors of an X-ray CCD imager. These errors are quite different from those found in a visible CCD imager.

  19. The Padre Miguel Ignimbrite Suite, central Honduras: Paleomagnetism, geochronology, and tectonic implications

    The Padre Miguel Group in western Honduras is a silicic volcanic sequence that forms part of the Central American Miocene volcanic arc built on the Chortis continental fragment. We report new 40Ar/39Ar data of 16.1 ± 0.2 Ma and 14.42 ± 0.08 Ma for the Padre Miguel Group, and present paleomagnetic data for 36 cooling units (mainly ignimbrites) from localities north and northeast of the capital city of Tegucigalpa. These rocks are characterized by univectorial characteristic magnetization carried mainly by low-Ti titanomagnetites, or two component magnetizations with a minor secondary overprint. Dual polarity magnetizations suggest that it is a primary thermo-remanence and the obtained data scatter can be straightforwardly explained by paleosecular variation. The overall mean of 33 selected sites (14.39°N, 87.14°W) defines a paleomagnetic pole at lat = 80.0°N, lon = 142.9°E (K = 24.5, A95 = 5.2°). The overall mean compared with the North America expected direction indicates statistically insignificant rotation and inclination anomaly (R = - 4.1° ± 6.1° and an F = - 8.6° ± 11.3°). Restoring 255 km of sinistral slip in the Polochic-Motagua fault system, as suggested by Cayman Trough reconstructions since 15 Ma, brings the mid-Miocene arcs of southern Mexico (Oaxaca) and the Central America ignimbrite province into alignment; this is consistent with a derivation of the Chortis Block from southern Mexico. Our paleomagnetic and geochronological studies hence support models that explain the offset of the Miocene ignimbrite belt of Central America by post-Middle Miocene displacement of the Chortis Block relative to North America.

  20. Landsat TM image segmentation for delineating geological zone correlated vegetation stratification in the Kruger National Park, South Africa

    Image classification approaches are widely used in mapping vegetation on remotely sensed images. Vegetation assemblages are equivalent to habitats. Whereas sub-pixel classification approaches potentially can produce more realistic, homogenous habitat maps, pixel-based hard classifier approaches often result in non-homogenous habitat zones. This salt-and-pepper habitat mapping is particularly a challenge on images of savannas, given the characteristic patchy texture of scattered trees and grass. Image segmentation techniques offer possibilities for homogenous habitat classification. This study aimed at establishing the extent to which established, field surveyed and geology-related vegetation types in South Africa’s Kruger National Park (KNP) can be reproduced using image segmentation. Rain season Landsat TM images were used, selected to coincide with the peak in vegetation productivity, which was deemed the time of year when discrimination between key habitats in KNP is most likely to be successful. The multiresolution segmentation mode in eCognition 5.0 was employed, object classification accomplished using the nearest neighbour (NN) classifier, using object texture and training area mean values in the NN feature space. Compared to delineations of the vegetation types of KNP on a digital map of the vegetation zones that was tested, image segmentation successfully mapped the zones (overall accuracy 85.3%, K^ = 82.7%) despite slight shifts in the location of vegetation zone boundaries. Maximum likelihood classification (MLC) of the same images was only 37% accurate (K^ = 24.2%). Whereas the vegetation zones resulting from MLC were non-homogenous, with considerable spectral confusion among the vegetation zones, image segmentation produced more homogenous vegetation zones, comparably more useful for conservation management, because realistic and meaningful habitat maps are important in biodiversity conservation as input data upon which to base management decisions

    We present new observations of the quiescent giant molecular cloud GCM0.253+0.016 in the Galactic center, using the upgraded Karl G. Jansky Very Large Array. Observations were made at wavelengths near 1 cm, at the K (24–26 GHz) and Ka (27 and 36 GHz) bands, with velocity resolutions of 1–3 km s{sup −1} and spatial resolutions of ∼0.1 pc, at the assumed 8.4 kpc distance of this cloud. The continuum observations of this cloud are the most sensitive yet made, and reveal previously undetected emission which we attribute primarily to free–free emission from external ionization of the cloud. In addition to the sensitive continuum map, we produce maps of 12 molecular lines: 8 transitions of NH{sub 3}–(1, 1), (2, 2), (3, 3), (4, 4), (5, 5), (6, 6), (7, 7), and (9, 9), as well as the HC{sub 3}N (3–2) and (4–3) lines, and CH{sub 3}OH 4{sub −1}–3{sub 0}, the latter of which is known to be a collisionally excited maser. We identify 148 CH{sub 3}OH 4{sub −1}–3{sub 0} (36.2 GHz) sources, of which 68 have brightness temperatures in excess of the highest temperature measured for this cloud (400 K) and can be confirmed to be masers. The majority of these masers are concentrated in the southernmost part of the cloud. We find that neither these masers nor the continuum emission in this cloud provide strong evidence for ongoing star formation in excess of that previously inferred by the presence of an H{sub 2}O maser.

  2. Physicochemical action of potassium-magnesium citrate in nephrolithiasis

    Effect of potassium-magnesium citrate on urinary biochemistry and crystallization of stone-forming salts was compared with that of potassium citrate at same dose of potassium in five normal subjects and five patients with calcium nephrolithiasis. Compared to the placebo phase, urinary pH rose significantly from 6.06 +/- 0.27 to 6.48 +/- 0.36 (mean +/- SD, p less than 0.0167) during treatment with potassium citrate (50 mEq/day for 7 days) and to 6.68 +/- 0.31 during therapy with potassium-magnesium citrate (containing 49 mEq K, 24.5 mEq Mg, and 73.5 mEq citrate per day). Urinary pH was significantly higher during potassium-magnesium citrate than during potassium citrate therapy. Thus, the amount of undissociated uric acid declined from 118 +/- 61 mg/day during the placebo phase to 68 +/- 54 mg/day during potassium citrate treatment and, more prominently, to 41 +/- 46 mg/day during potassium-magnesium citrate therapy. Urinary magnesium rose significantly from 102 +/- 25 to 146 +/- 37 mg/day during potassium-magnesium citrate therapy but not during potassium citrate therapy. Urinary citrate rose more prominently during potassium-magnesium citrate therapy (to 1027 +/- 478 mg/day from 638 +/- 252 mg/day) than during potassium citrate treatment (to 932 +/- 297 mg/day). Consequently, urinary saturation (activity product) of calcium oxalate declined significantly (from 1.49 x 10(-8) to 1.03 x 10(-8) M2) during potassium-magnesium citrate therapy and marginally (to 1.14 x 10(-8) M2) during potassium citrate therapy.(ABSTRACT TRUNCATED AT 250 WORDS).

  3. A novel nonsense mutation in BBS4 gene identified in a Chinese family with Bardet-Biedl syndrome

    Background Bardet-Biedl syndrome (BBS) is a genetically heterogeneous disease,and information about BBS in Chinese populations is very limited.The purpose of the present study was to determine the genetic cause of BBS in a Chinese Han family.Methods Clinical data were recorded for the 4-year-old female proband and the available family members.The proband was screened for mutation by Sanger sequencing for a total of 142 exons of the 12 BBS-causing genes (BBS1-BBS12).The variants detected in the proband were further confirmed in the other family members.Results We identified a novel homozygous nonsense mutation (c.70A>T,p.K24X) in the BBS4 gene exon 2 in the proband.Such mutant allele was predicted to cause a premature truncation in the N-terminal of the BBS4 protein,and probably induced the nonsense-mediated decay of BBS4 messenger RNAs.The proband's parents and brother were heterozygous for the nonsense mutant allele.It was absent in 50 Chinese control subjects.An additional rare heterozygous missense single nucleotide polymorphism (SNP) named rs200718870 in BBS10 gene was also detected in the proband,her father and her brother.Some manifestations of the proband including atypical retinitis pigmentosa,choroidal sclerosis,high myopia,and early onset of obesity might be associated with this mutation in BBS4 gene.The proband's father also reported surgical removal of an extra finger during childhood.Conclusions The present study described a novel nonsense mutation in BBS4 gene in a Chinese family.This homozygous mutation was predicted to completely abolish the synthesis of the BBS4 protein.We also detected a rare heterozygous missense SNP in BBS10 gene in the family,but did not find sufficient evidence to support the triallelic inheritance.

  4. ‘Squeezing’ near-field thermal emission for ultra-efficient high-power thermophotovoltaic conversion

    We numerically demonstrate near-field planar ThermoPhotoVoltaic systems with very high efficiency and output power, at large vacuum gaps. Example performances include: at 1200 °K emitter temperature, output power density 2 W/cm2 with ~47% efficiency at 300 nm vacuum gap; at 2100 °K, 24 W/cm2 with ~57% efficiency at 200 nm gap; and, at 3000 °K, 115 W/cm2 with ~61% efficiency at 140 nm gap. Key to this striking performance is a novel photonic design forcing the emitter and cell single modes to cros resonantly couple and impedance-match just above the semiconductor bandgap, creating there a ‘squeezed’ narrowband near-field emission spectrum. Specifically, we employ surface-plasmon-polariton thermal emitters and silver-backed semiconductor-thin-film photovoltaic cells. The emitter planar plasmonic nature allows for high-power and stable high-temperature operation. Our simulations include modeling of free-carrier absorption in both cell electrodes and temperature dependence of the emitter properties. At high temperatures, the efficiency enhancement via resonant mode cross-coupling and matching can be extended to even higher power, by appropriately patterning the silver back electrode to enforce also an absorber effective surface-plasmon-polariton mode. Our proposed designs can therefore lead the way for mass-producible and low-cost ThermoPhotoVoltaic micro-generators and solar cells.

    We study the multi-instanton partition functions of the Ω-deformed N=2{sup ∗}SU(2) gauge theory in the Nekrasov-Shatashvili (NS) limit. They depend on the deformation parameters ϵ{sub 1}, the scalar field expectation value a, and the hypermultiplet mass m. At fixed instanton number k, they are rational functions of ϵ{sub 1},a,m and we look for possible regularities that admit a parametrical description in the number of instantons. In each instanton sector, the contribution to the deformed Nekrasov prepotential has poles for large deformation parameters. To clarify the properties of these singularities we exploit Bethe/gauge correspondence and examine the special ratios m/ϵ{sub 1} at which the associated spectral problem is n-gap. At these special points we illustrate several structural simplifications occurring in the partition functions. After discussing various tools to compute the prepotential, we analyze the non-perturbative corrections up to k=24 instantons and present various closed expressions for the coefficients of the singular terms. Both the regular and singular parts of the prepotential are resummed over all instantons and compared successfully with the exact prediction from the spectral theory of the Lamé equation, showing that the pole singularities are an artifact of the instanton expansion. The analysis is fully worked out in the 1-gap case, but the final pole cancellation is proved for a generic ratio m/ϵ{sub 1} relating it to the gap width of the Lamé equation.

    Dipole moments of proteins arise from helical dipoles, hydrogen bond networks and charged groups at the protein surface. High protein dipole moments were suggested to contribute to the electrostatic steering between redox partners in electron transport chains of respiration, photosynthesis and steroid biosynthesis, although so far experimental evidence for this hypothesis was missing. In order to probe this assumption, we changed the dipole moment of the electron transfer protein adrenodoxin and investigated the influence of this on protein-protein interactions and electron transfer. In bovine adrenodoxin, the [2Fe-2S] ferredoxin of the adrenal glands, a dipole moment of 803 Debye was calculated for a full-length adrenodoxin model based on the Adx(4-108) and the wild type adrenodoxin crystal structures. Large distances and asymmetric distribution of the charged residues in the molecule mainly determine the observed high value. In order to analyse the influence of the resulting inhomogeneous electric field on the biological function of this electron carrier the molecular dipole moment was systematically changed. Five recombinant adrenodoxin mutants with successively reduced dipole moment (from 600 to 200 Debye) were analysed for their redox properties, their binding affinities to the redox partner proteins and for their function during electron transfer-dependent steroid hydroxylation. None of the mutants, not even the quadruple mutant K6E/K22Q/K24Q/K98E with a dipole moment reduced by about 70% showed significant changes in the protein function as compared with the unmodified adrenodoxin demonstrating that neither the formation of the transient complex nor the biological activity of the electron transfer chain of the endocrine glands was affected. This is the first experimental evidence that the high dipole moment observed in electron transfer proteins is not involved in electrostatic steering among the proteins in the redox chain.

  8. Serum Renalase Levels Correlate with Disease Activity in Lupus Nephritis.

    Full Text Available Lupus nephritis (LN is among the most serious complications of systemic lupus erythematosus (SLE, which causes significant morbidity and mortality. Renalase is a novel, kidney-secreted cytokine-like protein that promotes cell survival. Here, we aimed to investigate the relationship of serum renalase levels with LN and its role in the disease progression of LN.For this cross-sectional study, 67 LN patients and 35 healthy controls were enrolled. Seventeen active LN patients who received standard therapies were followed up for six months. Disease activity was determined by the SLE Disease Activity-2000 (SLEDAI-2K scoring system and serum renalase amounts were determined by ELISA. Predictive value of renalase for disease activity was assessed. Furthermore, the expression of renalase in the kidneys of patients and macrophage infiltration was assessed by immunohistochemistry.Serum renalase amounts were significantly higher in LN patients than in healthy controls. Moreover, patients with proliferative LN had more elevated serum renalase levels than Class V LN patients. In proliferative LN patients, serum renalase levels were significantly higher in patients with active LN than those with inactive LN. Serum renalase levels were positively correlated with SLEDAI-2K, 24-h urine protein excretion, ds-DNA and ESR but inversely correlated with serum albumin and C3. Renalase amounts decreased significantly after six-months of standard therapy. The performance of renalase as a marker for diagnosis of active LN was 0.906 with a cutoff value of 66.67 μg/ml. We also observed that the amount of renalase was significantly higher in glomerular of proliferative LN along with the co-expression of macrophages.Serum renalase levels were correlated with disease activity in LN.   9. Poznań's program of cochlear and brainstem implantation: a general review.

    The cochlear implantation program in Poznań started in January 1994. Within 8 years, 151 patients were implanted (149 cochlear and 2 auditory brain-stem implants). The implanted group comprised 111 children and 40 adults. We have extended the lower age limit for cochlear implant candidates, and the youngest implanted patient was 18-months-old at the time of surgery. We have also implanted six multi-handicapped subjects. All patients were implanted with different nucleus devices (22, 24 M, 24 K, 24 Contour, 24 Double Array and 24 ABI). Insertion of the Nucleus 24 Contour electrode carrier requires a little larger cochleostomy in comparison with conventional straight array. After insertion, the intraoperative measurements are taken (impedance telemetry, electrically evoked stapedius reflex and NRT). The day after surgery, each patient undergoes an X-ray examination (the cochleogram or cochlear view). The calculated length of the electrode array insertion was 12.75-24.0 mm (mean: 22.05 mm) and the degree of electrode rotation was 180-540 degrees (mean: 292.82 degrees). After speech processor switch-on, we performed NRT measurements. Our results indicated that the NRT threshold was in 51.4% of the dynamic range within the group of examined patients. The adults' progress of rehabilitation was checked during individual sessions. The rehabilitation of children included two rehabilitation schedules: continuous ones and weekly rehabilitation camps. The progress of the recovery process of implanted children enabled them to change their school profile from hearing impaired to mainstream. Voice rehabilitation is often monitored by acoustic analysis using the Multi Dimensional Voice Program (MDVP).

    We numerically demonstrate near-field planar ThermoPhotoVoltaic systems with very high efficiency and output power, at large vacuum gaps. Example performances include: at 1200 °K emitter temperature, output power density 2 W/cm2 with ~47% efficiency at 300 nm vacuum gap; at 2100 °K, 24 W/cm2 with ~57% efficiency at 200 nm gap; and, at 3000 °K, 115 W/cm2 with ~61% efficiency at 140 nm gap.   11. VARIATIONS OF MID- AND FAR-INFRARED LUMINOSITIES AMONG EARLY-TYPE GALAXIES: RELATION TO STELLAR METALLICITY AND COLD DUST

    The Hubble morphological sequence from early to late galaxies corresponds to an increasing rate of specific star formation. The Hubble sequence also follows a banana-shaped correlation between 24 and 70 {mu}m luminosities, both normalized with the K-band luminosity. We show that this correlation is significantly tightened if galaxies with central active galactic nucleus (AGN) emission are removed, but the cosmic scatter of elliptical galaxies in both 24 and 70 {mu}m luminosities remains significant along the correlation. We find that the 24 {mu}m variation among ellipticals correlates with stellar metallicity, reflecting emission from hot dust in winds from asymptotic giant branch stars of varying metallicity. Infrared surface brightness variations in elliptical galaxies indicate that the K - 24 color profile is U-shaped for reasons that are unclear. In some elliptical galaxies, cold interstellar dust emitting at 70 and 160 {mu}m may arise from recent gas-rich mergers. However, we argue that most of the large range of 70 {mu}m luminosity in elliptical galaxies is due to dust transported from galactic cores by feedback events in (currently IR-quiet) AGNs. Cooler dusty gas naturally accumulates in the cores of elliptical galaxies due to dust-cooled local stellar mass loss and may accrete onto the central black hole, releasing energy. AGN-heated gas can transport dust in cores 5-10 kpc out into the hot gas atmospheres where it radiates extended 70 {mu}m emission but is eventually destroyed by sputtering. This, and some modest star formation, defines a cycle of dust creation and destruction. Elliptical galaxies evidently undergo large transient excursions in the banana plot in times comparable to the sputtering time or AGN duty cycle, 10 Myr. Normally regarded as passive, elliptical galaxies are the most active galaxies in the IR color-color correlation.

    Full Text Available High-resolution simulations of the chemical composition of the Arctic stratosphere during late spring 1997 and 2000 were performed with the Chemical Lagrangian Model of the Stratosphere (CLaMS. The simulations were performed for the entire northern hemisphere on two isentropic levels 450 K (~18 km and 585 K (~24 km. The spatial distribution and the lifetime of the vortex remnants formed after the vortex breakup in May 1997 display different behavior above and below 20 km. Above 20 km, vortex remnants propagate southward (up to 40°N and are "frozen in'' in the summer circulation without significant mixing. Below 20 km the southward propagation of the remnants is bounded by the subtropical jet. Their lifetime is shorter by a factor of 2 than that above 20 km, owing to significant stirring below this altitude. The behavior of vortex remnants formed in March 2000 is similar but, due to an earlier vortex breakup, dominated during the first 6 weeks after the vortex breakup by westerly winds, even above 20 km. Vortex remnants formed in May 1997 are characterized by large mixing ratios of HCl indicating negligible, halogen-induced ozone loss. In contrast, mid-latitude ozone loss in late boreal spring 2000 is dominated, until mid-April, by halogen-induced ozone destruction within the vortex remnants, and subsequent transport of the ozone-depleted polar air masses (dilution into the mid-latitudes. By varying the intensity of mixing in CLaMS, the impact of mixing on the formation of ClONO2 and ozone depletion is investigated. We find that the photochemical decomposition of HNO3 and not mixing with NOx-rich mid-latitude air is the main source of NOx within the vortex remnants in March and April 2000. Ozone depletion in the remnants is driven by ClOx photolytically formed from ClONO2. At the end of May 1997, the halogen-induced ozone deficit at 450 K poleward of 30°N amounts to ~12% with ~10% in the polar vortex and ~2% in well-isolated vortex remnants

    BACKGROUND. In humans, a single light exposure of 12 minutes and multiple-millisecond light exposures can shift the phase of the circadian pacemaker. We investigated the response of the human circadian pacemaker to a single 15-second or 2-minute light pulse administered during the biological night. METHODS. Twenty-six healthy individuals participated in a 9-day inpatient protocol that included assessment of dim light melatonin onset time (DLMO time) before and after exposure to a single 15-second (n = 8) or 2-minute (n = 12) pulse of bright light (9,500 lux; 4,100 K fluorescent) or control background dim light (lux; n = 6). Phase shifts were calculated as the difference in clock time between the two phase estimates. RESULTS. Both 15-second and 2-minute exposures induced phase delay shifts [median (± SD)] of –34.8 ± 47.2 minutes and –45.4 ± 28.4 minutes, respectively, that were significantly (P = 0.04) greater than the control condition (advance shift: +22.3 ± 51.3 minutes) but were not significantly different from each other. Comparisons with historic data collected under the same conditions confirmed a nonlinear relationship between exposure duration and the magnitude of phase shift. CONCLUSIONS. Our results underscore the exquisite sensitivity of the human pacemaker to even short-duration single exposures to light. These findings may have real-world implications for circadian disruption induced by exposure to brief light stimuli at night. TRIAL REGISTRATION.     Full Text Available Abstract Background Neonatal rats exposed to repetitive inflammatory pain have altered behaviors in young adulthood, partly ameliorated by Ketamine analgesia. We examined the relationships between protein expression, neuronal survival and plasticity in the neonatal rat brain, and correlated these changes with adult cognitive behavior. Methods Using Western immunoblot techniques, homogenates of cortical tissue were analyzed from neonatal rats 18–20 hours following repeated exposure to 4% formalin injections (F, N = 9, Ketamine (K, 2.5 mg/kg × 2, N = 9, Ketamine prior to formalin (KF, N = 9, or undisturbed controls (C, N = 9. Brain tissues from another cohort of rat pups (F = 11, K = 12, KF = 10, C = 15 were used for cellular staining with Fos immunohistochemistry or FluoroJade-B (FJB, followed by cell counting in eleven cortical and three hippocampal areas. Long-term cognitive testing using a delayed non-match to sample (DNMS paradigm in the 8-arm radial maze was performed in adult rats receiving the same treatments (F = 20, K = 24, KF = 21, C = 27 in the neonatal period. Results Greater cell death occurred in F vs. C, K, KF in parietal and retrosplenial areas, vs. K, KF in piriform, temporal, and occipital areas, vs. C, K in frontal and hindlimb areas. In retrosplenial cortex, less Fos expression occurred in F vs. C, KF. Cell death correlated inversely with Fos expression in piriform, retrosplenial, and occipital areas, but only in F. Cortical expression of glial fibrillary acidic protein (GFAP was elevated in F, K and KF vs. C. No significant differences occurred in Caspase-3, Bax, and Bcl-2 expression between groups, but cellular changes in cortical areas were significantly correlated with protein expression patterns.   15. Elemental characterization of particulate matter emitted from biomass burning: Wind tunnel derived source profiles for herbaceous and wood fuels

    Particulate matter emitted from wind tunnel simulations of biomass burning for five herbaceous crop residues (rice, wheat and barley straws, corn stover, and sugar cane trash) and four wood fuels (walnut and almond prunings and ponderosa pine and Douglas fir slash) was collected and analyzed for major elements and water soluble species. Primary constituents of the particulate matter were C, K, Cl, and S. Carbon accounted for roughly 50% of the herbaceous fuel PM and about 70% for the wood fuels. For the herbaceous fuels, particulate matter from rice straw in the size range below 10 μm aerodynamic diameter (PM10) had the highest concentrations of both K (24%) and Cl, (17%) and barley straw PM10 contained the highest sulfur content (4%). K, Cl, and S were present in the PM of the wood fuels at reduced levels with maximum concentrations of 6.5% (almond prunings), 3% (walnut prunings), and 2% (almond prunings), respectively. Analysis of water soluble species indicated that ionic forms of K, Cl, and S made up the majority of these elements from all fuels. Element balances showed K, Cl, S, and N to have the highest recovery factors (fraction of fuel element found in the particulate matter) in the PM of the elements analyzed. In general, chlorine was the most efficiently recovered element for the herbaceous fuels (10 to 35%), whereas sulfur recovery was greatest for the wood fuels (25 to 45%). Unique potassium to elemental carbon ratios of 0.20 and 0.95 were computed for particulate matter (PM10 K/C(e)) from herbaceous and wood fuels, respectively. Similarly, in the size class below 2.5 μm, high-temperature elemental carbon to bromine (PM2.5 C(eht)/Br) ratios of ˜7.5, 43, and 150 were found for the herbaceous fuels, orchard prunings, and forest slash, respectively. The molar ratios of particulate phase bromine to gas phase CO2 (PM10 Br/CO2) are of the same order of magnitude as gas phase CH3Br/CO2 reported by others.

  16. Deformation Along the Southeast Extension of the Lake Mead Fault System Evaluated with Paleomagnetic Data From Miocene Igneous Rocks, Hoover Dam area, Nevada and Arizona

    At and near Hoover Dam, southeast of Las Vegas, Cenozoic left-slip offset along the NE-SW trending Lake Mead fault system (LMFS) has resulted in the apparent rotation of structures and total displacement of up to 65 km. Defining any rotation of blocks within and near the LMFS is critical to assessing the kinematics of strike-slip faulting and attending extension. Paleomagnetic data from Miocene volcanic and some sedimentary rocks and intrusions (over 160 sites) deposited on Precambrian basement show that part of the Hoover Dam locality has experienced counterclockwise rotation . The middle Miocene (ca. 14.2 Ma)Tuff of Hoover Dam (THD)(sampled at over 90 sites) yields a well-grouped characteristic magnetization (ChRM); about 5 km south and east of the dam, gently east-dipping, north-striking rocks of the THD yield a corrected ChRM of moderate positive inclination and northwest declination (D=324.8°, I=27.4°, a95=10.7°, k=24, N=9 sites). Structural corrections, based on compaction fabrics in the THD are consistent with stratigraphic contacts. The anomalous shallow inclination for the THD ChRM implies that it was emplaced over a short period of time during a field instability. contact and conglomerate test results are interpreted to show that the THD ChRM is primary. Corrected data from north and west of the dam (D=289.7°, I=30.2°,a95=8.6°,k=32, N=10) are interpreted to indicate about 35° of counterclockwise rotation (R= -35.1°, delR= 12.4, F= -2.8°, delF = 10.8, relative to data from south of the dam) of crust across the dam site, consistent with progressive changes in strike of tilted fault blocks. The transition from apparently unrotated crust to rotated crust occurs over a zone about 1 km wide, where blocks of THD and older strata have been tilted up to 50°, probably concurrent with rotation.     The following accomplishments were made on task 4. Reproducibility of Catalyst Preparation: (1) Five slurry reactor tests were completed. Three tests were conducted using catalyst C (100 Fe/3 Cu/4 K/16 SiO{sub 2}) from three different batches (runs SB-2695, SB-2145 and SA-2715), and two tests were conducted with catalyst B (100 Fe/5 Cu/6 K/24 SiO{sub 2}) from two different preparation batches (runs SA-2615 and SB-2585). Performance of catalysts from different batches (activity, selectivity and deactivation rates) was similar to that of catalysts from the original batch (synthesized during DOE Contract DE- AC22-89PC89868). Thus, another major objective of the present contract, demonstration of reproducibility of catalyst preparation procedure and performance, has been accomplished. With these tests the work on Task 4 has been successfully completed. Two fixed bed reactor tests of catalysts B and C synthesized using potassium silicate solution as the source of potassium promoter were completed during this period (Task 5. The Effect of Source of Potassium and Basic Oxide Promoter). Activity of catalysts prepared using potassium silicate as the source of potassium promotion was somewhat higher, and their methane selectivities were higher than those of the corresponding catalysts prepared by incipient wetness impregnation using KHCO{sub 3} as the source of potassium promoter. However, these differences were not large, and may have been caused by experimental artifacts (e.g. existence of local hot spots in a reactor). A slurry reactor test (SA-2405) of catalyst with nominal composition 100 Fe/5 Cu/2 Ca/24 SiO{sub 2} was completed (Task 5). In general, the catalyst activity, space-time-yield, and hydrocarbon selectivities in this run during testing at:260{degrees}C, 2.17 MPa (300 psig), 2-2.6 Nl/g-cat/h and H{sub 2}CO=0.67 were quite good, and comparable to the best results obtained in our Laboratory.

  18. Imaging of nutrient elements in the leaves of Elsholtzia splendens by laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS).

    Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) was used for the quantitative imaging of nutrient elements (such as K, Mg, Mn, Cu, P, S and B) in the leaves of Elsholtzia splendens. The plant leaves were scanned directly with a focused Nd:YAG laser in the laser ablation chamber. The ablated material was transported with argon as carrier gas to a quadrupole-based ICP-MS (ICP-QMS), and the ion intensities of (39)K(+), (24)Mg(+), (55)Mn(+), (63)Cu(+), (31)P(+), (34)S(+) and (11)B(+) were measured by ICP-QMS to study the distribution of the elements of interest. The imaging technique using LA-ICP-MS on plant leaves does not require any sample preparation. Carbon ((13)C(+)) was used as an internal standard element to compensate for the difference in the amount of material ablated. Additional experiments were performed in order to study the influence of the water content of the analyzed leaves on the intensity signal of the analyte. For quantification purposes, standard reference material (NIST SRM 1515 Apple Leaves) was selected and doped with standard solutions of the analytes within the concentration range of 0.1-2000 mg L(-1). The synthetic laboratory standards together with the samples were measured by LA-ICP-MS. The shape and structure of the leaves was clearly given by LA-ICP-MS imaging of all the elements measured. The elemental distribution varied according to the element, but with a high content in the veins for all the elements investigated. Specifically, Cu was located uniformly in the mesophyll with a slightly higher concentration in the main vein. High ion intensity was measured for S with a high amount of this element in the veins similar to the images of the metals, whereas most of the B was detected at the tip of the leaf. With synthetic laboratory standard calibration, the concentrations of elements in the leaves measured by LA-ICP-MS were between 20 microg g(-1) for Cu and 14,000 microg g(-1) for K.

  19. The role of nitric oxide in Doxorubicin-induced cardiotoxicity: experimental study.

    Amaç: Bu çalışmada sıçanlarda doksorubisinin (DOX) tek başına ve nitrik oksit sentaz (NOS) inhibitörleri ile birlikte kullanımının kalp kası üzerinde yarattığı hasar araştırıldı.Gereç ve Yöntemler: Çalışmada 12 haftalık 24 adet erkek “Sprague Dawley” sıçanı (ortalama ağırlık 262±18 gr) rastgele dört gruba (n=6) dağıtıldı. Grup I kontrol grubu olarak ayırıldı. Grup II’ye haftada bir kez intraperitoneal (ip) 3 mg/kg, beş hafta DOX uygulandı. Grup III’e DOX enjeksiyonundan 30 dakika önce ip 30 mg/kg L-NAME (nonselektif NOS inhibitörü) 5 hafta uygulandı. Grup IV’e DOX enjeksiyonundan 30 dakika önce ip 3 mg/kg L-NIL (indüklenebilir NOS inhibitörü) 5 hafta uygulandı. Çalışma süresince sıçanlar haftada iki kez tartıldı. Altıncı haftada kalbin sol ventrikülünden alınan parçalardan elektron ve ışık mikroskobik inceleme ve lipit peroksidasyonu tayini (malondialdehid) yapıldı. Plazma lipit peroksidasyonunun belirlenmesi amacı ile sıçanlardan kan alındı.Bulgular: Grup II, Grup III ve Grup IV’te kilo kaybı gözlendi. DOX uygulanan grupta istatistiksel olarak anlamlı kilo kaybı saptandı. Miyokard hasarı bulguları tek başına DOX alan hayvanlarda kontrol grubuna oranla daha yüksek oranda izlendi. DOX’le beraber L-NAME ve L-NIL verilen sıçanlarda kardiyotoksitenin histopatolojik bulguları kontrol grubundan farklı değildi. Plazma malondialdehid düzeyi DOX grubunda (9,3± 3,4), diğer tüm gruplara göre daha yüksek olarak ölçüldü. Sonuç: Bizim çalışmamızda DOX’la beraber NOS inhibitörleri uygulandığında DOX’a bağlı kardiyak toksisite bulgularında anlamlı azalma olduğu gösterilmiştir.

    [目的]观察螺内酯对2型糖尿病大鼠肾功能的影响.[方法]48只大鼠用高脂高糖饲料喂养2个月后,腹腔注射 30 mg/kg链脲佐菌素制备糖尿病大鼠模型.随机分为糖尿病组 (DM组,n =24)、螺内酯治疗组(R组,n =24),另选同批普通饲养大鼠15只作为正常对照组 (C组,n=15),检测第16周末血清中血糖(FBG)、糖化血红蛋白(HbA1c)、钾(K+)、24 h尿白蛋白排泄率(UAER)、尿素氮(U-BUN)、肌酐(Cr)、甘油三酯(TG).并用He染色的方法观察16周后大鼠肾脏的病理学变化.[结果]第16周的血糖水平DM组和 R组相近,无显著差别( P >0.05),但DM组和 R组的血糖水平均显著高于A组( P <0.01);UAER水平DM组和 R组显著高于C组( P <0.01),且R组UAER水平显著低于DM组( P <0.01).[结论] 螺内酯能通过降低糖尿病大鼠UAER,改善糖尿病大鼠的肾功能从而发挥保护肾脏的作用.%[Objective] To observe the effect of spironolactone on renal function in rats with type 2 diabetes mellitus (T2DM). [Methods]The rat model of T2DM was prepared by intraperitoneal injection of streptozocin(STZ) 30mg/kg after feeding high-sugar and fat diet for 2 months. The rats were assigned to diabetic group(group DM, n =24) and spironolactone treatment groupCgroup R, n =24). Other 15 rats fed with normal diet were selected as normal control group (group C, n =15). Blood glucose(FBG) , glycosylated hemoglobin(HbA1c) , kalium(K+) , 24h urine albumin excretion rate(UAER) , urea nitrogen(U-BUN) , creatinine(Cr) and triglyceride(TG) at the end of 16th week were detected. HE staining was used to observe the pathological change of kidney in rats after 16 weeks. [Results] At the end of 16th week, BG level in group DM was similar to that in group R, and there was no significant difference( P >0. 05).   1. Thermal oxidation of Ge-implanted Si: Role of defects

    Thermal oxidation of Ge-implanted Si (SiGe) was carried out in dry O at 1073, 1173, and 1273 K for various times. Rutherford backscattering spectrometry in random and channeling geometry was used to characterize the SiO thickness and composition of the Si (dry oxidation) [3,4,8,9,13-17] or by bubbling N/O through HO (wet oxidation) [5-7,9-12,18]. In these studies SiGe thin films were obtained by different growth techniques such as chemical vapor deposition (CVD) [4-6,19,12], molecular beam epitaxy (MBE) [7,8,14,19,21-23], physical evaporation [3,18] as well as Ge ion implantation in Si [9-11,13,15-17]. Despite the great differences in the preparation of SiGe samples and oxidation procedures, the main features of SiGe thermal oxidation may be summarized by the following: Pure SiO was formed during oxidation: Ge atoms that were rejected from the growing silicon oxide piled up at the interface. This was observed in all cases, unless the temperature was low enough (⩽973 K)[24], or the oxidation pressure was high [6,19], or the Ge concentration, x, in the alloy satisfied x⩾0.5[7,8], or the oxidation time was very short [22,23]. All these conditions prevent Ge diffusion away from the reacting interface. The oxidation rate of SiGe in a wet atmosphere was enhanced in comparison to pure Si [4,5,9-12,18,25], while there was no enhancement in the dry O[9,16,22,25] (unless the sample was first pre-enriched with Ge to form approximately one monolayer of Ge at the interface [9]). Oxidation rate enhancement occurs during an initial linear regime of oxide growth [10]. SiGe oxidation rate enhancement has been explained by: (i) the weaker Si-Ge bond [11], (ii) Ge catalytic role for oxidation reaction [12], and (iii) changes in defect generation at the reacting interface [12]. It was shown for Ge ion implanted samples that the Deal and Grove model (DG model) for Si oxidation can still be applied with the linear B/ A constant modified to take into account enhanced oxidation

  2. 颈椎后纵韧带骨化症患者K线对两种颈后路手术疗效的影响%The influence of K-line on two posterior approaches for patient with ossification of posterior longitudinal ligament

    Objectives: To elevate the influence of K-line on the outcome of laminoplasty(Lam) vs laminec-tomy and instrumentation(LI) for patient with ossification of posterior longitudinal ligament(OPLL). Methods: 78 patients undergoing Lam(32) and LI(46) from January 2003 to February 2011 were included in this series. 54 cases with the ossification mass not beyond the line at the midpoint of spinal canal from C2 to C7 were grouped as K(+), while 24 cases with ossification mass exceeding the K-line were grouped as K(-). The pre-operative and follow-up JOA scores, and postoperative complications were recorded to analyze the influence of two approaches on the outcome. Results: The improvement rate of neurologic function(IR) after posterior approaches in cases of group K(+) and K(-) was 68.2% and 38.8% respectively, which showed significant difference (P<0.01). In group K(+), the IR of cases for Lam was 69.9%, and 65.6% for LI, which showed no significant difference (P>0.05). In group K(-), the IR for LI was 47.0%s and 33.9% for Lam, which showed significant difference(P<0.05). The onset was significantly earlier and the incidence of C5 palsy was significantly higher in cases with LI than cases with Lam(P<0.05). Conclusions: Preoperative K(+) predicates a better IR than K(-). For cases with K(-), LI provide better neurologic function recovery. LI indicate higher incidences of C5 palsy and the earlier onset of axial pain.%目的:评价颈椎后纵韧带骨化症(OPLL)患者的K线对颈后路单开门椎管扩大成形术(laminoplasty,Lam)和颈后路椎板切除减压融合内固定术(laminectomy and instrumentation,LI)疗效的影响.方法:回顾性分析2003年1月~2011年2月在我院接受手术治疗的78例OPLL患者,其中Lam治疗32例,LI治疗46例.将颈椎侧位片上C2和C7椎管中点连线未跨越骨化物的病例定为K(+),54例[K(+)组];跨越骨化物者定为K(-),24例[K(-)组].记录患者术前及末次随访时的JOA
