    In Japan with its rapidly aging society, fundamental technologies are required for the development of artificial nerves substituting for or supporting damaged ones, and ultimately constituting artificial neurons based on the knowledge of the brain functions at the molecular and cellular levels. This study defines the fundamental technologies which would be required for the development in the area, and further, evaluates the potential of the technologies to develop the novel industry. The brain function is closely related to the activity in neuronal circuits. In order to repair injured nerves and to develop the advanced technologies of electronics for helping impaired neuronal functions, the most important and urgent is to understand how to work the neuronal circuit system in the brain. Based on these viewpoints, new methodological approaches would make it possible to relieve neural impairment in the sensory input system and the motor system by the use of electronic circuits. They also would improve rehabilitation after injury, and treat neurodegenerative diseases such as Parkinson`s disease.     Researches are conducted about the present and future of the research and development of elementary technological details for elucidating, from the viewpoint of the level of molecules or cells, the functions of neurons which are the basic elements of the cranial nerve circuit and for applying the results to cranial nerve cell engineering. It is then concluded that the most important and urgent is to 'develop basic technologies for understanding the functions of neural circuits so that a remedial step may be taken in the presence of a defect in a neural circuit and that electronic devices may be applied to help neural functions.' It is agreed that the most immediate and ripple-effective is to 'establish the technology of intelligent connection between electronic devices and neural circuits for the reconstruction of motor functions.' It is also concluded that, upon achievement of the above two goals, new basic technologies will become available for the moderation of problems in the sensor input and motor output systems by means of electronic circuits, for the improvement on efficiency of rehabilitation with the aid of electronics, for the cure of denatured nerves or a damaged spinal cord, for the study of neural circuits, etc., and that a new industrial series will be created through the utilization of the above-mentioned details.     For forming cell tissues alternative to bio-tissues, this research targets the technology for forming bio-tissue modules by cultivating 3-D cell tissues from various kinds of cells. In fiscal 1999, research was made on various issues of element technologies necessary for forming module structure of bio-tissues, and study was also made on the application of 3-D cell tissue module engineering to industrial fields. Survey was made on element technologies and solutions supporting such engineering, and in addition, on progressive conditions of cell cultivation techniques, and concrete techniques for expressing cell functions. Research was made on a cell behavior under physicochemical stimulus environment to study the optimum environment for cell multiplication and function expression. Further research was made on state analysis of cells, in particular, fast precise measurement techniques of activation or malignant of cells and secretion of toxic substances by physical/optical analytical evaluation techniques such as photo-CT and spectroscopic analysis, and biochemical analysis techniques using bio-sensors. Study was also made on successful development and application cases of practical bio-artificial organs in western countries. (NEDO)

    For the formation of cellular tissues to replace bionic tissues, researches were conducted about technologies of forming bionic tissue modules by culturing various kinds of cells. As for the materials and methods for constructing cellular tissues, researches were conducted about the trends of research and development of 3-dimensional tissue culturing matrices and materials for micromanipulation. As for the development of technologies for the functionalization of 3-dimensionally structured cells, research and study were conducted about the technology of 3-dimensional cell structure organization through application of physical stimulation, the biochemical technology of differentiation inducing, and the differentiation inducing technology for hetero tissue culturing. As for the development of technologies for evaluation using 3-dimensionally structured cells, light CT (computer tomography), analysis and evaluation using spectroscopy and the like, feasibility of the biochemical analysis of the cell state using biosensors, technologies for measuring the secretion of carcinogenic and toxic substances, etc., were studied.     Full Text Available The paper explores the development of Confucian thought in Japan, showing that Confucianism, with its key concept of the Way, remained the foundation of Japanese intellectual circles even as it underwent numerous transformations and reinterpretations. The universal Way as a natural Principle was transformed into a product of the sages within the framework of the kogaku school, whereas the kokugaku school applied the concept of the Way to the Japanese Shinto tradition. The adherents of the rangaku school, on the other hand, adopted Western science and technology in the Confucian intellectual universe. The new nation-state built by the Meiji revolutionaries thus became a mixture of all these intellectual currents. The Meiji state was a modern, industrial and technologically advanced state that identified itself with the imperial system and native Shinto tradition while being held together institutionally by a firm Confucian ideology.

    Intracellular signaling from receptor tyrosine kindles in mammalian cells results in activation of a signal cascade that includes the guanine nucleotide binding protein Ras and the protein kinases Raf, MEK [Mitogen activated protein kindle (MAPK) or Extracellular signal regulated kinase (ERK) kinase] and MAPK. MAPK activation that is dependent on the coupling of Ras and Raf was reconstituted in yeast. Yeast genes were isolated that, when overexpressed, enhanced the function of Raf. One of them is identical to BMH 1, which encodes a protein similar to members of the mammalian 14-3-3 family. Bacterially synthesized mammalian 14-3-3 protein stimulated the activity of Raf prepared from yeast cells expressing c-Raf-1. Thus, the 14-3-3 protein may participate in or be required for activation of Raf. 9 refs., 2 figs.

    The relationship between environmental control and biotechnology is discussed, and purification of moats and lakes is taken up as a concrete example of introducing biological waste water treatment technology to ecotechnology. In addition, an overall waste disposal system connecting cities and agricultural districts is investigated. After the purification of water and waste problem solution, etc. are discussed, the importance of creating ecopolis and ecotown is described. The 3 concepts of microbial ecotechnology (high densification of organism, field control, interaction between organisms) are used to explain biological treatment technology. Attempts for direct purification and ecotechnology are described quoting a few examples. If waste produced in cities and ligneous materials or charcoal obtained from forests are mixed for treatment, effective waste treatment or new resources production may be possible. It seems to be possible to create ideal ecopolis and ecotown by applying the doctrine of irrigation, drainage and reclamation engineering and the technology established by now. 35 refs., 1 fig., 2 tabs.

    It has been reported that hydroxyapatite (HA), which is the main inorganic component of teeth and bones of homo sapiens and used for biomaterials such as artificial tooth roots, adsorbs viruses such as influenza viruses. In this article, the history of development up to now of HA and its adsorption mechanism of protein, virus, etc., are introduced. HA was applied for chromatography in 1956 becoming one of the separating and refining methods of protein and nucleic acid, then after the development of spherical porous HA, it has become applied for high speed liquid chromatography (HPLC). Also by means of a column filled with HA granules, T-cells have been able to be purified in a short time from lymphocyte which was separated from the blood of homo sapiens. Recently it has also been reported that HA granules can adsorb influenza viruses, Japanese encephalitis viruses, polio viruses and hepatitis B viruses, and a cold-preventative mask based upon this report is now on sale. 11 refs., 7 figs.

    Developments are made on an optical imager (ASTER) used to collect mainly images of land areas and an infrared sounder (IMG) to measure vertical air temperature distribution and vertical concentration distribution of specific gases, as satellite mounted sensors for earth observation. All the sensor characteristics of the ASTER comprising a visible near infrared radiometer, short wave infrared radiometer and thermal infrared radiometer are required to be capable of providing measurement, evaluation and assurance at the required accuracies during the entire life time. A problem to be solved is how to combine the on-ground calibration prior to launching, on-satellite calibration, and calibration between the test site and the sensors. The IMG is a Fourier transform spectroscopic infrared sounder, which is demanded of a high wave resolution over extended periods of time as well as a high radiation measuring capability. Also required are the level elevation of analysis algorithms to solve inverse problems from the observed radiation spectra, and the data base with high accuracy. 19 refs., 4 figs., 4 tabs.

    On January 22nd, 2011(JST), KOUNOTORI2 (H-II Transfer Vehicle: HTV2) was successfully launched from Tanegashima Space Center toward the International Space Station (ISS) and two new JAXA payload racks, Kobairo rack and MSPR (Multi-purpose Small Payload Rack) were transferred to ISS/KIBO (Japanese Experiment Module: JEM). In addition to Saibo rack and Ryutai rack which are already in operation in KIBO, in total 4 Japanese experiment payload racks start operations in KIBO. Then KIBO payload operations embark on a new realm, full utilization phase. While the number and variety of microgravity experiments become increasing, simultaneous operation constraints should be considered to achieve multitask payload operations in ISS/KIBO and ever more complicated cooperative operations between crewmember and flight control team/science team are required. Especially for g-jitter improvement in ISS/KIBO, we have greatly advanced cooperative operations with crewmember in the recent increment based on the microgravity data analysis results. In this paper, newly operating Japanese experiment payloads characteristics and some methods to improve g-jitter environment are introduced from the front line of KIBO payload operations.

    This paper proposes a draft plan for global civil engineering that takes on the earth. The paddy field environmental system in Japan has been a high-degree ecological system from viewpoints of material and energy circulation, coexistence and symbiosis with living organisms, and social frameworks. The capital of pre-war Manchuria, Xin jing, was represented by large-scale green park zones with artificial lakes utilized for recreational activities and used for basic infrastructures for the city. With regard to the use of balanced water tanks, corals were cultivated successfully, achieving purification with use of natural power without using mechanical purifying devices. As referenced, the Japanese civil engineering technologies have a number of features that can contribute internationally to global environmental problems. Japan has accumulated civil engineering experiences that harmonize with diverse nature in a small land. It has a history of maintaining high-level sustained agriculture with the paddy field ecological system, and having converged the modern civil engineering technologies used in Xin jing into the city plans of environment harmonizing type.     For developing intelligent material, animal test alternative model, bio-cell analysis equipment, self-controlling bio-reactor and medical material, development of functional cells was studied by cell proliferation regulation technology. In fiscal 1996, the expression analysis and separation technology of specific gene for cell proliferation, and the intracellular regulation technology were surveyed from the viewpoint of intracellular regulation. The cell proliferation regulation technology by specific regulating material of cells, extracellular matrix, coculture system and embryonic cell was surveyed from the viewpoint of extracellular regulation. In addition, based on these survey results, new cell culture/analysis technology, new bio-material, artificial organ system, energy saving bio-reactor, environment purification microorganism, and animal test alternative model were surveyed as applications to industrial basic technologies from a long-term viewpoint.     The Accreditation Examination Committee of engineering education was established in 1997 in the Japan Engineering Education Society, and the International Universal Engineer Review Board was established in the Japan Engineering Education Society and the Japan Engineering Society in 1998. A series of the activities was connected to the establishment of 'Japan Accreditation Board for Engineering Education' (JABEE). To adapt to the movements, the Educational Program Review Board in the Engineering was installed in 1997 mainly composed of directors of engineering departments of 8 national universities.     Cellular and tissue engineering for bioartificial organs need lots of information about extra-cellular matrices and cytokines network which play important roles for the regulation of cellular function. In order to develop high performance artificial livers, we made establishment of extracellular matrix (ECM) engineering by the combination of genetic engineering and polymer engineering. We designed a superglycoprotein-like polymer (PVLA) as a specific model ligand to asialoglycoprotein receptors (ASGP-R) on hepatocytes. Hepatocytes morphology was remarkably affected by PVLA concentration for coating on polystyrene dishes. In addition, hepatic functions such as differentiation and proliferation were closely related with cell morphology. Especially, round hepatocytes on the polystyrene surface coated with high concentration of PVLA were found to reconstruct 3D liver-like tissue stimulated by EGF. It turned out that the adsorptive states of PVLA polymeric micells can switch the function of EGF to be motogenic or mitogenic. The mechanisms of 3D-tissue formation on two types of substrata mediated by ASGP-R were examined. The proliferation and cell death (apoptosis) in hepatitis of hepatocytes were also examined in the viewpoint of cytokine network such as TNF - {alpha} and interferon-{gamma}. The highly efficient gene-transfection into hepatocytes was also successfully made using PVLA as ECM for cell attachment. As one of the trials to clarify liver cells society formation the mechanism of the final differentiation of kupffer cells and the role of HGF in embryonic construction of liver were successfully examined.     Advanced research and its applicability were surveyed to apply the advanced functional cells to industry. The basic target was set to develop, produce, control and utilize the functional cells, such as intelligent materials and self-regulation bioreactors. The regulation factors regarding apotosis, which is a process of cell suicide programmed within the cell itself of multicellular organisms, cell cycle and aging/ageless were investigated. Furthermore, the function of regulatory factors was investigated at the protein level. Injection of factors regulating cellular function and tissue engineering required for the regulation of cell proliferation were investigated. Tissue engineering is considered to be the intracellular regulation by gene transduction and the extracellular regulation by culture methods, such as coculture. Analysis methods for cell proliferation and function of living cells were investigated using the probes recognizing molecular structure.     Use of microbial enzymes have been widely extended as an effective means for asymmetric synthesis. However, the asymmetric selectivity often decreases due to competitive catalysis among plural enzymes in a microbe. The author has been studied development of methods for control of the stereo-selectivity using subtle difference of enzyme characteristics. When Michaelis constant (Km) differs between two enzymes, one enzyme of lower Km becomes active with decrease in concentration of substrate, expressing its stereo-selectivity. Reduction of {alpha}-ketoesters in water by bread yeast (Saccharomyces cerevisiae) yields products of S-configuration, whereas those of R-configuration are obtained in an organic solvent in the presence of small amount of water. This is because reaction field of the yeast is in water and because R-configuration enzyme of lower Km works for substrate whose concentration in water has decreased due to two phase partition of organic solvent and water system. Further, use of difference of decrease in enzyme activity by inhibitors in stereo-selective synthesis of {alpha}-hydroxyketones (I) from {alpha}-diketone and use of difference of thermal endurance in improvement of formation ratio among I, are also introduced. 6 refs., 3 figs., 2 tabs.

    This paper describes the following matters as one example of structural coloring of non-motional nature on the coloring mechanism in wings of Morpho sulkouskyi butterfly: The structure of scales on butterfly wings may be modelled into such a structure of a right angle frame of reference having a cyclic arrangement (with an interval of 0.7 [mu]m) of a zigzagged protrusion in the 'x' direction called a ridge (with a height of 1.8 [mu]m and a width of 0.54 [mu]m, the shape not changing in the 'y' direction), and having a plurality of protrusions of a ridge form called lamella in the 'z' (height) direction. A simple multi-layered thin film model was assumed that has lamella layers and air layers laminated alternately in the 'z' direction. The model was used to calculate reflection spectra on vertically incident light based on thickness and refractive index of material layers determined by a structural analysis of the scales. The result was considerably close to the reflection spectra of scales measured by using a microscopic spectrophotometer when the number of layers is set to six. The paper describes briefly examples of motility structural coloring (skin cells of rainbow color in tropical fish). 7 refs., 12 figs.

    Considerations were given from a viewpoint of assessing toxicity to human bodies on methods of assessing water pollutants and organisms, and applicability of cytotoxity test using cultured cells to water quality assessment. Biological assessment systems used for water environment may use tests using multicellular organisms, cells, or organelles in cells. The organism assessment method is intended mainly for assessing ecological effects, and a suitable method must be selected upon extrapolating it to human bodies. A toxicity parameter used most frequently in a cytotoxity test is the cell revival rate, and life and death are determined from liberation of enzymes in cells, or with color rejection tests and incorporation tests. There are a number of test specimens of raw tap water and its chlorine treatment condensate that show no mutagenicity but cytotoxity. Efficiencies of removal by means of mild chlorine treatment, fast filtration, and activated carbon adsorption vary greatly with cytotoxity and mutagenicity. Introducing the cytotoxity test is expected of further contributing to improving safety in water quality. 24 refs., 1 fig., 7 tabs.

    The author of this article is a professor of Department of Mechanical Engineering of Massachusetts Institute of Technology (MIT) and the director general of Laboratory for Information Systems and Technology of the institute. At Department of Mechanical Engineering of Engineering Faculty of world-famous MIT, Laboratory for Information Systems and Technology was established in 1994 and in the same year, the curricula of the department were greatly revised after 30 years for its enforcement from the new semester starting from September, 1995. These two occasions are suggestive for guessing the future of mechanical engineering, hence its aim and meaning are introduced.     Trend of robotics and mechatronics in 1989, Japanese and foreign, are reviewed in the respective fields. In the field of manipulation technology, development of micro-manipulation, simulation by neural network, motion control technology in space, and development of whole-arm manipulator were advanced. Studies on locomotive function of wheels. legs, and crawler types in auto-locomotive robots were progressed. On sensor technology, combined technologies with control engineering and sensor information were noteworthy.     Nonoxide glasses whose main constituent are chalcogen elements like S, Se, or Te etc. show a lot of various properties, for instance, high infrared transmittancy and semi-conductivity which are already well known. Additionally, the optical properties change a lot along with the phase transition's happening between crystal and noncrystal under comparative low temperature. Further, it is also observed that the glasses containing proper cation appear high ion-conductivity.     Optical division using chiral fixing phase for polysaccharide derivative based HPLC has been exhibiting superior capability as an analysis means. It now has transcended the framework of analysis as a result of the encounter with the SMB process, and is completing itself as an optically active body manufacturing means. This paper introduces the following examples of applying the chiral fixing phase for HPLC using polysaccharide derivatives as an unconformity identifier to the unconformity identifying mechanism and mass synthesis: 1) aromatic groups and carbamate groups as substitution groups are arranged regularly along a polysaccharide main chain having a regular and unconformable stereostructure, and a certain `unconformity identification field` is formed: 2) hydrogen bonding and dipole-to-dipole interactions take place between solutes in the ester groups and carbamate groups, and such an interaction act as stacking of aromatic rings among those having aromatic rings; and as a result, interactions in 2) occur in the `unconformity identification field` in 1), where the unconformity identification is executed. This paper describes the pseudo migration bed (SMB) process developed by UOP Corporation as a method for mass-synthesizing fillers for the polysaccharide derivative based HPLC. 6 refs., 3 figs.

    Development was attempted on organic-inorganic complexes as nonlinear optical materials. The objective is to develop elements with SHG activity (generating second harmonics) to change a visible ray into an ultraviolet ray by inserting P-nitroaniline (pNA) into such solid acids having strong static electric field as zeolite, laminar compounds, and heteropolyacid, and utilizing the static electric field and the molecule coordination field of inorganic compounds. Used for the pNA{center_dot}Bronsted acid composite are HBr, HCl, HNO3, and H2SO4. The pNA that has a center of symmetry in the crystalline structure and does not generate the second harmonics shows SHG when it is formed into a composite with HBr. No SHG is recognized in other acid composites. This difference is due to the difference in the crystalline structure. The SHG activity was generated when pNA is loaded onto alumina and silica as solid oxides. When zeolites are used as carriers, no SHG was observed. A composite of pNA{center_dot}AlCl3 has high SHG intensity, which reached a maximum when the pNA/AlCl3 ratio was two. This composite showed four new peaks that belong to the SHG activity phase. 8 refs., 2 figs., 2 tabs.

    Study was made on the effect of exposure to diesel emissions on inflammatory cells in a rat lungs. Four kinds of exposure gases with different contents of NO2 and particulate were prepared by diluting diesel emissions. Rats were exposed to diluted diesel emissions for 24 months, and inflammatory cells were detected morphologically in light microscopic and TEM specimens. As a result, particle-laden- alveolar macrophages increased dose- and time-dependently into the submucosa of intrapulmonary bronchioles, alveolar spaces and interstitume of alveolar walls, and bronchoassociated lymphatic tissues. Mast cells infiltrated into the interspaces of epithelial cells in airways. In the submucosa of the terminal bronchioles and the interstitume of alveolar walls, some sorts of inflammatory cells such as mast cells, plasma cells, neutrophils and lymphocytes infiltrated, and some cells showed cell-to-cell contacts. However, the airways were rarely injured by infiltration of inflammatory cells except for a fibrotic change. 2 refs., 2 figs., 2 tabs.

    Investigations were given on nanocrystals of CuCl and CdTe with regard to their nonlinear optical mechanism. The experiment used a femto-second pump probe spectroscope. The experiment on CuCl nanocrystals revealed the following facts: in the case where one photon is absorbed into one nanocrystal, cascade mitigation occurs to the pair of electrons and holes, and exciters; and in the case where two photons are absorbed into one nanocrystal, exciter molecules are made via the pair of electrons and holes and the exciters. Thus, it was found that the optical nonlinearity occurs when more than two photons are absorbed into one nanocrystal, and inter-exciter interactions and formation of the exciter molecules are the physical causes thereof. The experiment on CdTe nanocrystals indicated that electrons and holes produced by laser beam are distributed instantaneously between the size-quantized discrete levels, and that temperature in the electron system drops with lapse of time. 9 refs., 6 figs.

For the systematization of cities, it is necessary to process and control the intelligent information by holding integrated and transverse information in common, as to logistics, lifeline, communication, monitoring, and control. For the communication engineering, the centralized systems are acceleratingly to be converted into the distributed network systems. For the mechanical engineering, intelligent control and robot technology are required. For the architectural engineering, a concept exceeding the conventional antiseismic structure idea is investigated. It is necessary to develop a new information technology providing an intelligent social infrastructures by merging the information networks and the physical world seamlessly.   9. Assessing Pearl Quality Using Reflectance UV-Vis Spectroscopy: Does the Same Donor Produce Consistent Pearl Quality?

    Full Text Available Two groups of commercial quality (“acceptable” pearls produced using two donors, and a group of “acceptable” pearls from other donors were analyzed using reflectance UV-Vis spectrophotometry. Three pearls with different colors produced by the same donor showed different absorption spectra. Cream and gold colored pearls showed a wide absorption from 320 to about 460 nm, while there was just slight reflectance around 400 nm by the white pearl with a pink overtone. Cream and gold pearls reached a reflectance peak at 560 to 590 nm, while the white pearl with pink overtone showed slightly wider absorption in this region. Both cream and gold pearls showed an absorption peak after the reflectance peak, at about 700 nm for the cream pearl and 750 nm for the gold pearl. Two other pearls produced by the same donor (white with cream overtone and cream with various overtones showed similar spectra, which differed in their intensity. One of these pearls had very high lustre and its spectrum showed a much higher percentage reflectance than the second pearl with inferior lustre. This result may indicate that reflectance is a useful quantitative indicator of pearl lustre. The spectra of two white pearls resulting from different donors with the same color nacre (silver showed a reflectance at 260 nm, followed by absorption at 280 nm and another reflectance peak at 340 nm. After this peak the spectra for these pearls remained flat until a slight absorption peak around 700 nm. Throughout the visible region, all white pearls used in this study showed similar reflectance spectra although there were differences in reflectance intensity. Unlike the spectral results from white pearls, the results from yellow and gold pearls varied according to color saturation of the pearl. The results of this study show that similarities between absorption and reflectance spectra of cultured pearls resulting from the same saibo donor are negligible and could not be detected

    This is a proceedings of the 55th Meeting on Cryogenics and Superconductivity. As for wires, made public were Bi-system 2223, Y-system NbTi, NbSn, Nb3Al, etc. As to oxide superconductors, some of them are promising. The study of those characteristics and the developmental application to the electric power field such as coils and cables were introduced. Beside wires, multilayer film superconductors were also introduced. With relation to fundamentals and theories, became topics were pinning characteristics, NbTi proximity effects, magnetic flux and visualization, stability, etc. About the application, large refrigerators are discussed in the refrigeration field in addition to GM/pulse tube refrigerators. Concerning cooling technology, made public were the development of a floating railroad use 80K cooling system, etc. Also studied were power leads, superconducting magnet, control action characteristics, etc. SMES (superconducting magnetic energy storage) also became a topic, and the element coil energization experiment, stability and mechanical properties were described. The paper also touched on accelerators, large superconducting devices such as LHD (large helical device) and ITER (international thermal-nuclear experimental reactor) as well as generators and current limiters. The cryogenetic engineering was also reported such as fracture toughness and superfluidity of cryostats and structural materials for extremely low temperatures.

    The shape of osteoblast, gene appearance, gene of rice blast, cellular fusion of plants, gravity acceptance mechanism of unicellular organisms, and physiological and immunity functions of mice were investigated under the microgravity condition. The influence of gravity on the vital reaction and the influence of microgravity on the crystallization of vital substances were also investigated. For the observation of osteoblast, the fluorescence dye reacted with Ca was well taken in the cells. The microgravity affected the stability of rice blast, but hardly affected the protoplast culture of mushroom. The reaction of ciliate against the gravity related to the specific gravity difference between cells and outer liquid. The level of adrenaline in blood of mice increased during the drop. The moving speed of trigger waves of chemical parallel slit formed at the BZ reaction under the microgravity became 60% to 80% of that on the ground. In the case of crystallization at the deposition agent concentration of 1% to 4%, the turbidity showing the degree of crystallization changed complicatedly.     The project aims to establish basic technologies for the mass culture of cells by utilizing the features of animal cells, which will enable a large, stable supply of fine chemicals such as highly functional physiologically active proteins whose industrial production the conventional technique of synthesis fails to realize. For this purpose, efforts are exerted to select cells which are capable of producing target substances, to breed highly productive strains, to develop a serumless medium that satisfies demands for an all-purpose feature and heat sterilization, and to develop technologies of high-density culture and effective refining, etc. Some DNA (deoxyribonucleic acid) recombination technologies are also resorted to. As the result, a high-density culture of 2 times 10{sup 8} cells/ml has become available. Production is now feasible of various differentiation-inducing elements such as monoclonal antibodies with anti-HB virus activity, DIF, etc. Furthermore, cooperation with the Tsukuba University leads to the discovery of a novel vasopressor peptide (endothelin). The heat sterilization accepting serumless medium, which is put to practical use as the first of the kind in the world under this research and development project, contributes to the advancement of basic studies about cell biology.     As part of validation processes of in vitro toxicity assays for the risk assessment of environmental hazards, we applied an in vitro toxicity test using two human cell lines, neuroblastoma NB-1 cells and glioblastoma U-87 MG cells, to the assessment of organochlorine compounds detected in the water environment. The in vitro toxicity assay using NB-1 cells was calibrated by testing reference chemicals proposed by MEIC (Multicenter Evaluation of In Vitro Cytotoxicity), an international program for the validation of in vitro cytotoxicity assays. Beforehand, an assay using cells in frozen stock without subcultivation was examined by comparing IC50 values with the ordinary assay using subcultured cells. IC50 values for MEIC reference chemicals from the former assay showed good correlation with those from the latter assay, suggesting that the assay using cells in frozen stock can be used at least for the assessment of basal cytotoxicity. IC50 values for ten organochlorine compounds frequently detected in the sediment samples from contaminated rivers, p-chloroaniline, 3,4-dichloroaniline, p-dichlorobenzene, o-dichlorobenzene, Tris (2-chloroethyl)-phosphate, 2,5-dichlorophenol, 2,5-dichloroanisol, Triclosan and Triclocarban, were obtained with the in vitro assays and compared with their LD50 values in rats. No significant correlation, however, was seen between the IC50 and LD50 values, indicating that further improvement of in vitro toxicity assays is necessary for the application to the risk assessment of environmental hazards. 7 refs., 4 figs., 1 tab.

    In order to clarify the relationship between the heat transfer rate and the expansion bed in a group of horizontal pipes in a freeboard region (an area of the heat-transfer pipe exposed above the height of static particle bed from the beginning) in a cold model of the fluidized bed, particle behavior was measured using an optical measuring method. The light axis position was set higher than the heat-transfer as X {sub p} in a direction perpendicular from the distributor, and the static bed height was set to L {sub c}. The frequency of particles and particle lumps coming to presence between the light axes is termed V {prime}{sub p}(time-averaged dimensionless amount of the optical probe output). The V {prime}{sub p} decreases with an increase in the flow velocity, and, when the difference between the probe tip and the static bed height, X {sub p}{minus} L {sub c} is small, it shows the minimum value at a certain flow velocity and then rises again. The root mean square value of the probe output, V {prime}{sub f} increased with an increase in the flow velocity, reached its maximum, then decreased to the minimum, and rose again. The flow velocity that takes the maximum heat transfer rate can be identified from the relationship among the dimensionless amount of the maximum expansion bed height and the average expansion bed height, the dimensionless height of X {sub p} when V {prime}{sub p} and V {prime}{sub f} obtained at each X {sub p} show the extreme values, and the dimensionless height of the heat-transfer pipes when the average transfer rate takes the maximum value. 6 refs., 5 figs.

    Researches are conducted for showing some guidelines on the system and method of the intelligent social infrastructure technology (ISIT) as a 21st century type systematization technology, and a report is submitted on the result. Discussed in the report are the technological system and image of ISIT, the research and development setup and administrative tasks involving intelligent social infrastructure construction, and the systematization and operation of the ISIT. Mentioned as important element technologies are the infrastructure and information/communications technologies for security and disaster prevention, information technology and automatization technology for maintenance, and intelligent materials enabling disaster prediction and detection. Stated in connection with the future development of ISIT are information/communication technologies for system control, monitoring, and normal/urgent dual use. Outstanding among the achievements are Professor Morio Okazaki's 'Study of manipulation of transfer of heat and substance in porous solid bodies' (winner of prize from The Society of Chemical Engineers, Japan), and Professor Tomoyuki Inui's 'Study of fuel synthesis using porous crystalline catalysts' (winner of prize from The Japan Institute of Energy).     Research is conducted concerning the possibility of anti-gene engineering basic to the application of DNA (deoxyribonucleic acid) drugs capable of serving as functional DNAs to the control of the expression of industrially useful substance producing genes or of anomalous genes and concerning the possibility of novel industry creation on the strength of the said engineering. The specific research items are described below. Technical seeds are investigated relating to the tissue- and cell-specific drug delivery system for the expression of the molecular device function of the DNA drug. Concerning molecular target technologies such as the anti-sense method, possibilities are studied of utilizing the currently available technical seeds for the eventual creation of novel industries. Concerning novel designing methods utilizing genome information such as SNPs (single nucleotide polymorphisms), investigations are conducted to determine if they would help novel technology development and novel material development. The domestic state is surveyed in relation to DNA drugs, and possibilities are investigated of novel substance production and novel medicine creation with the aid of anti-gene engineering. (NEDO)

    Since light can be transformed into electricity, electrolysis of water using light is possible naturally, and photosynthesis in the nature is based on this principle. The reaction field in which electrochemical energy necessary for decomposition of water is generated using light at the first process of photosynthesis is a biofilm less than 10nm thick which is called thylakoid film. In this article, the effects are roughly explained which have been made for constructing an artificial system corresponding to the thylakoid film. Accumulation of knowledge of the molecular level is necessary for the construction of the artificial system and the three dimensional structure concerning the reaction center of photosynthetic bacteria has been determined. At the reaction center, oxidation species of electron donor and reduction species of electron acceptor are generated by charge separation which succeeds electron excitation by light energy. In order to lead the above process to materialistic production, efforts are being made for construction of a reaction center model compound for artificial photosynthesis from both physical chemistry and organic chemistry and the positive results have steadily been obtained in the development of photocatalysis of a semiconductor. 9 figs.

    This research project was designed to develop the advanced measurement system for higher mental activities. Specifically, the following five issues were investigated: 1) the measurements of the mental activity on situations with ecological validity. 2) the construction of systematized measurement systems. 3) the development for integrated multiple measurement indices. 4) the development for comprehensive algorithm models. 5) the revision of concept for measurement of the mental activity. The following six sub-projects were run. 1) we constructed the system for high-speed seiral stimulus presentation on large-scale panel, and developed the technique for psychophysical measurements on dynamic characteristics of visual attention. 2) We developed the pseudo-habitat apparatus for measurement of possible involvement of brain acetylcholine in learning/memory function in rats. 3) We developed the computer-assisted system for measurement of achievement of long-term English-words home learning in high school students. 4) We developed the multi-phasic measurement system for concurrent recordings of eye-movement, behavior, response-time and correct responses during performance of cognitive tasks. 5) We constructed the standard audio-visual database for species-specific behavioral patterns of inter-individual behaviors such as aggressive and sexual behaviors in mice. 6 we developed the audio-visual recording system for measurement and analysis of conversation for social interaction in class rooms.     The project aims to establish basic technologies for the mass culture of cells which will enable a large, stable supply of fine chemical products whose industrial production the conventional technique of synthesis has failed to realize. In the 1st phase, cell stains were selected, a serumless medium was developed, basic culturing conditions were established, and substances production rates were improved, and the 2nd phase stands succeeding these 1st phase achievements. Toward the ultimate goal of establishing basic technologies for serumless high-density cultures for industrial use and technologies for isolating and refining useful substances, a small-scale optimum culture technique and a serumless culture technique are developed. Serumless cultures are developed for strains of man's lymphoid cells, cells deriving from bone marrow, epithelial cells, and hybridomas of man and mouse, all these sampled during the 1st phase endeavors. Success is attained in growing each cell strain on a scale of 1-10L to increase to 10{sup 7} cells/ml or more in population density.     The aim is to add value to the chemical industry by establishing technologies for realizing a large and stable supply of fine chemical products whose industrial production is difficult when conventional techniques are applied, the products including various useful substances yielded by animal cells including those of man. To fulfill the aim, efforts are exerted to complete basic technologies involving the mass culture of cells through conducting researches into animal biofunctions. This report covers the endeavors made in fiscal 1987 under the above-named project created in fiscal 1981.     The Yoshino river is one of the most prominent rivers in Japan, representing the Shikoku district, with the trunk length of 194km and the basin of 3,750km{sup 2} extending in 4 prefectures in the district. The upstream area includes areas where the average annual rainfalls exceed 3,000mm, being famous as a high-rain zone. The basic water level stream is 24,000m{sup 3} s which is one of the largest in Japan. Schistosity develops with the Sambagawa metamorphic rocks, which tend to be easily weathered. As a result, there are many landslide spots distributed in the Sambagawa belt. The Shikoku district as a whole has one fifth of the landslide dangerous areas of Japan. Countermeasure activities against landslides were initiated in 1982 in Tokushima and Kochi prefectures. Outlines of landslides in those areas, underground structure, activity of landsliding, countermeasure activity against landslides, and the result of the activity are introduced. 12 figs.

    This paper describes the surveys in fiscal 1999 on establishing industrial technology strategies in the human life engineering field. The human life engineering is an engineering to identify scientifically humans and their living characteristics, and apply them to products, systems, and environment building. Market size for the human living related industries is more than two trillion yen, whose annual growth rate is 9%. Discussions were given on problems that hinder technological innovations in this field. What lacks is the comprehensive strategic conception, and insufficient is the structural approach by related organizations. Technological systems for designing have not been established, and technological foundations relative to human characteristics and living characteristics are not well arranged. Methods to evaluate research achievements have not been established, and the works on proposing international standards are weak. Insufficiency in human resource nurturing is also an issue. As the future prospect and strategies, such fields as psychology, recognition, and sensitivity are important for the seeds technologies, and so are the measurement of supportive technologies, and information processing. The human living engineering aims at supporting safety, freedom from care, and high-quality life, realizing industrial and technological competitiveness of the world top groups, and forming societies with activity and high quality.     The research and development efforts are made for super-selective localized diagnosis and treatment and simultaneous observation of the treatment results at the catheter edge point, based on the molecular biology and gene engineering methods, and the FY 2000 results are reported. The program for investigating the catheter hardware designs include the diagrammatic evaluation of a multi-lumen multi-functional laparoscope and pancreatoscope for localized diagnosis and medical treatment. The program for development of diagnostic technology leads to development of double staining of cancer cells and macrophage, which allows the in vivo analysis of the cell-to-cell interactions. The program for studying the technology related to affective genes which can be diagnosed and treated by a catheter includes cloning of genes for therapy of chronic glomerular nephritis and adrenal neoplasm, and those for liver regeneration. The program for method of local delivery of genes includes studies on therapy of metastatic cancer of lymph node using liposome and adenovirus as a gene carrier, and on pulse laser delivery for local and efficient delivery of the genes.     In order to type the sausage-shaped bacteria forming A-type sulfur-turf, cell length distributions and physics-chemical conditions of hot spring waters were investigated on twelve samples collected from all over Japan. The frequency distributions of the cell length of eight samples were bimodal, while the other four samples were unimodal. In seven samples with bimodal distributions, two types (large and small) of the sausage-shaped bacteria could be differentiated. The cell length of the large-type was between 10.1 and 31.9{mu}m, while that of the small-type ranged 2.2 to 6.6{mu}m. The pH of seven hot spring waters were between 6 and 8, and the two types (large and small) formed together sulfur-turf. In contrast, pH of the three hot springs were over 8, and the two types of the sausage-shaped bacteria could not be detected in the sulfur-turf. Therefore, it is reasonable to set a third type of the sausage-shaped bacteria which prefers high-pH (over 8) and low calcium condition. The cell lengths of the third type were in the range of 5.5 to 8.6{mu}m, which correspond to the sausage-shaped bacteria of medium size. 20 refs., 5 figs., 2 tabs.

    21.9km of the route between Doi and Iyosaijo of the Shikoku Jukan (Transverse) expressway was chosen along the central tectonic line which is the largest active fault in Japan. Countermeasure works for slope faces were carried out attaching importance to ground anchor and reinforced earth works, and the section was completed on March 29, 1991. In the section, longitudinal alignment, plain alignment and road structure were reexamined to determine the center line so as to avoid landslide areas as far as possible, and much cost curtailment was realized for the landslide countermeasure works although there were many slope face collapses during excavation. The collapsed soil amounted to about 50 to 600m{sup 3} for one collapsed spot, with 1 to 2m average thickness of the slid layers. Reinforced earth work method was found to be effective for small to medium scale slope face countermeasure works. PC anchor or other landslide countermeasure works were required for large scale landslides which have the depths of slide exceeding 2m. 5 refs., 7 figs.

    The basic data are collected by the experiments, in order to optimize the {sup 13}C-MRS analyzer for non-invasion type brain metabolism measurement. The tests are conducted for normal aged volunteers and volunteer patients of, e.g., dementia as the subjects. Each subject is orally dosed with glucose marked with {sup 13}C at the 1-site, and the metabolized {sup 13}C glucose signals from two points in the back of the head are observed as the MR spectra for a total of 3 hours. The time-series data of the {sup 13}C glutamic acid and glutamine signals are analyzed, to determine metabolism speed of the TCA circuits, among others, which provides important information for understanding the brain cell functions. It is 0.33{mu}mol/g/minute as the average of the 5 subjects of normal aged volunteers, which is on a level with that of the young volunteers. On the other hand, it is 0.26{mu}mol/g/minute as the average of the 4 subjects of volunteer patients of dementia, although there is no difference of statistical significance between these values. These results, therefore, suggest that the patients of dementia tend to have deteriorated brain metabolism than the healthy persons. (NEDO)

    The research and development efforts are made for the fundamental technology necessary for visualizing higher order information of the organs and tissues at high precision and non-invasively, and for supporting treatment of the malignant tumor and cerebrovascular diseases, while keeping the functions of the affected organs intact, and the FY 1999 results are reported. The program for the visualizing techniques includes designs of the pulse sequence for a 1H/13C-MRSI analyzer which observes proton and carbon by magnetic resonance spectrometric imaging (MRSI), and starts its programming. The program for the real-time analysis of tissue dynamics starts the designs of the coil for pelvis receiving the signals for monitoring dynamics of the abdominal organs at superhigh field. The program for systems for supporting treatment while keeping the functions of the affected organs intact starts development of the MR-matching heating system, for which the applications are designed, and measures against eddy current and resistance of the system as a whole to noise are studied. Development of the high-precision supersonic wave and far-infrared ray irradiation systems is started, in order to establish the treatment aided by radiant ray while keeping functions of the affected organs intact. The program for radiotherapy and ultrasonic therapy of malignant tumors in the body stems includes development of high-accuracy irradiation system aided by real-time three-dimensional body motion monitoring system.     The basic researches on minimally invasive diagnostic/treatment systems are conducted for circulatory system diseases, and the FY 2000 results are reported. The program for developing the heart surgery supporting manipulator includes development of the manipulator of 7 degree of freedom and fail-safe mechanisms to be incorporated in the patient-side system, and research and development of the heart motion compensation type robot system. The program for developing the diagnosis/treatment system aided by intravascular optical analysis includes development of intravascular endoscopy by the aid of LED emitting blue color of high brightness, and automatic analyzer for the in vivo vascular endothelial cell functions. The program for the minimally invasive diagnostic system includes development of superimposing system for integrating the images by the NOGA system and cine-coronary angiography. The other R and D items include artificial vascular systems to be put in the blood vessels, adhesives for a living body, suture instruments for fine blood vessels, heart surgery supporting system, based on the infrared spectroscopy, endoscopic system for the cranical bones, arterialization method, and gene-aided treatment.     Research and development was carried out for the establishment and commercialization of the technical seeds of a newly proposed 'parallel grinding type aspheric surface grinding method,' based on the results of studies of ultra-accuracy grinding and aspheric surface machining technologies conducted at Tohoku University. The new method is characterized in that a work point on the work piece faces the grinding point on the grindstone cross section on the one-to-one basis because the grinding point travels on the grindstone cross section. Accordingly, the method is quite suitable for corrective grinding for eliminating only the error in case a geometrical error occurs on the wrought surface. For this purpose, measurement of the machine surface on board the machine is indispensable. Hence: a study of corrective grinding methods under an on-board measuring system. As the result, the initially intended geometrical accuracy level was attained in the machining of axis-symmetric aspheric glass lenses and aspheric metal molds, and coarseness on the finished surface in the grinding of glass lenses was reduced to 0.3{mu}m using a diamond grindstone. In conclusion, machining accuracy and finished surface coarseness were remarkably improved, and this enables machining without the need of polishing and demonstrates that there are possibilities of commercialization.     As materials for next generation lithography use optical device using short wavelength light sources such as F{sub 2} and Ar{sub 2}, the development was proceeded with of fluoride monocrystal materials and 12-inch class large/high quality monocrystal production technology. Studies were made in the following five fields: 1) proposal/design of new materials and the heightening of performance: 2) establishment of the large/high quality crystal production method; 3) evaluation of optical properties and elucidation of micro-defect formation mechanism; 4) comprehensive investigational research; 5) study of the evaluation technology by vacuum ultraviolet area pulse light. In 1), for the development of the optimum materials, a lot of materials were tried to be monocrystallized, and the permeability was estimated by measuring the reflectance in the vacuum ultraviolet area. As to LiCaAlF{sub 6}, monocrystal with 1-inch diameter was made by the Bridgman method. In 2), studies were made of conditions for large crystal growth by the pull method, large crystal growth by the Bridgman method, and the structure of production equipment for crystals with larger diameter.     On the basis of the basement technology such as the cloning technology of biosynthesis enzyme gene groups of secondary metabolite of plant and the identification technology of the expression protein enzyme function in the heterologous expression system, the development was proceeded with of the technology to produce useful substances using the biosynthesis function of plant components. Studies were made in the following four fields: 1) cloning of genes of useful biosynthetic plants and functional elucidation/application of the related genes; 2) analytical technology of genes and proteome; 3) development of plant genome database; 4) comprehensive investigational study. In FY 2000, in 1), the cloning was conducted of biosynthetic enzyme genes of oxidation resistant polyphenol, bioactive triterpenoid saponin, etc., and the function of a part of the enzyme proteins was elucidated. In 2), as the basic technology for improving producibility of secondary electrophoresis, fabricated was the electrophoresis chip so structured that slide glasses on which micro-channels were two-dimensionally fabricated were joined.     The fundamental research is conducted for developing the microelectrode on which a synthetic oligonucletide probe is immobilized, and for establishing the system capable of detecting the hybrid formation simply and quickly, in order to develop the advanced DNA chips for diagnosis. The project results include development of the prototype array electrode with 25 1mm-diameter gold electrodes uniformly arranged at intervals of 4.5mm; development of the electrochemical activity analyzer for multi-electrode systems, showing the performance almost on a level with that of the existing electrochemical analyzer for the single-electrode systems; establishment of the gene databases; development of the method which can produce a sufficient quantity of nucleic acid for DNA chip analysis by studying the method of preparing the nucleic acid from the blood serum, preparing RNA from a trace quantity of the living liver sample and amplifying the genes, wherein the nucleic acid is produced while its profile before the amplification is kept intact; and establishment of the method for detecting the hepatitis B virus by combining the electrochemical detection of DNA by a non-immobilized probe with the PCR method.
