Resumen en español Antecedentes. Los organismos vivos responden al estrés aumentando la síntesis de proteínas. El estrés por choque térmico ha sido el más estudiado y las proteínas que se inducen, se han denominado genéricamente como proteínas de choque térmico. Objetivo.En este trabajo se establecieron las condiciones óptimas de producción de la proteína HSC70, expresada en E.coli BL21 (DE3) y de los anticuerpos policlonales que permitan identificarla. Material y métodos. Med (mas) iante varios ensayos se establecieron las concentraciones óptimas del agente inductor isopropil- -D-thiogalacto­piranosido (PTG), del inóculo bacteriano, de empleo de los plásmidos de expresión pET-3a y pET-28a (+); y el método más eficiente para la recuperación de las formas soluble o insoluble de la proteína y de anticuerpos policlonales que la identifiquen. Resultados. Encontramos que el inóculo de cinco colonias con IPTG (2mM) en tubos con cinco mililitros de medio modificado e incubadas por 24 horas a 37°C con agitación constante (200 r.p.m) y tratamiento de sonicación, produce el mejor rendimiento de HSC70. La calidad de la proteína inducida se estableció mediante “Western blotting”. Conclusión. La proteína recombinante así obtenida permitió, generar anticuerpos policlonales que a su vez permiten detectar la proteína HSC70 natural en la membrana citoplasmática de diferentes células por inmunofluorescencia, en ELISA, en Western Blot y en pruebas de bloqueo de infección de rotavirus. Resumen en inglés Background. Live organisms respond against stressing conditions raising the production of stress proteins. The heat shock stress is a very well kwon condition that produces heat shock proteins. Objective. In this work we determined the optimum conditions to produce HSC70 protein in E. coli BL12 (DE3) under stress conditions, and the best way to obtain its polyclonal antibodies. Materials and methods. The best concen­trations for isopropyl- ß -D-thiogalacto­pyra­noside (mas) (IPTG), bacterial inoculum, type of vector plasmid used pET-3a and pET28a (+) and recovery method of soluble and insoluble forms of HSC70 protein and its polyclonal antibodies were established. Results .Five-colony bacteria inoculum, 2 mM IPTG and sonication treatment were found as best conditions with either expression plasmid. We also produced polyclonal antibodies against the HSC70 protein by inoculating rabbits with this purified protein. Conclusions.Specificity and titer of antiserum was determined by ELISA, Western blotting and immunofluorescence assays. Antiserum was also tested by its inhibition ability of rotavirus infection.

Resumen en español INTRODUCCIÓN: la inducción de las proteínas de choque térmico constituyen un mecanismo homeostático que protege a las células del efecto destructivo del calor u otras condiciones de estrés ambiental, paralelamente, ellas cumplen importantes funciones celulares. La proteína de choque térmico de 20 kDa se reportó recientemente en Leishmania amazonensis. OBJETIVO: describir la metodología utilizada para realizar el clonaje de las proteínas de choque térmico, lo (mas) que permitió acometer estudios de algunas propiedades biológicas. MÉTODOS: la región codificante del gen hsp20 se amplificó mediante la reacción en cadena de la polimerasa con cebadores adecuados. El producto amplificado se clonó inicialmente en el vector pCR2.1 (Invitrogen) y después en el vector de expresión en procariotas pET-28b (Novagen), para obtener proteína recombinante. De manera paralela, el mismo fragmento se clonó en el vector de expresión en eucariotas pcDNA3 (Invitrogen) para obtener un posible preparado vacunal de ADN. Se realizó la secuenciación nucleotídica de los clones obtenidos, con la finalidad de verificar su fidelidad. RESULTADOS: se obtuvieron plásmidos recombinantes que codifican la HSP20 de Leishmania, y permiten la obtención de proteína recombinante y de ADN en forma masiva. CONCLUSIONES: ambos plásmidos fueron útiles para estudiar algunas de las propiedades biológicas de esta proteína. Este acercamiento puede ser de interés en otros trabajos de esta índole y constituir una guía metodológica. Resumen en inglés INTRODUCTION: the induction of heat shock proteins is a homeostatic mechanism that protects cells from the deleterious effects of thermal and other environmental stresses. In addition, they have important cell functions. The 20kDa heat shock protein in Leishmania amazonensis was recently reported. OBJECTIVE: to describe the methodology used for cloning of heat shock proteins, which allowed the study of some biological properties. METHODS: the hsp20 gene coding region was (mas) amplified by polymerase chain reaction using adequate primers. The amplified product was initially cloned in pCR2.1 vector (Invitrogen) and then in pET-28b vector (Novagen), to obtain recombinant protein. The same fragment was cloned also in the eukariote expression vector pcDNA3 (Invitrogen). The nucleotidic sequencing of the different clones was made, in order to verify their fidelity. RESULTS: the recombinant plasmids that encode HSP20 protein in Leishmania and allow obtaining massively recombinant protein and DNA were produced. CONCLUSIONS: both plasmids were useful to study some of the biological properties of this protein. This approach could be useful for similar research and represent a suitable methodological guideline.

Resumen en inglés Background: The expression of heat shock proteins (HSP70) in tumor cells or virus infected cells is important for the induction of specific cellular immune response. They are implicated in transport of immunodominants peptides in the endoplasmic reticulum, activation of antigen presenting cells and cross priming of CD8 T cells. Aim: To analyze the expression of HSP70 protein in its constitutive (HSP73) and inducible forms (HSP72) in Hodgkin's lymphoma (HL), infected or no (mas) t by Epstein Barr virus (EBV) and to assess its relationship with pathological subtype, clinical stages and treatment response. Material and methods: The analysis of HSP73 and HSP72 was done by immunoperoxidase on routinely processed paraffin sections with prior antigen retrieval. Results: Sixty seven cases were studied. The expression of HSP73 and HSP72 was detected in 19.4 and 17.9% of samples respectively. The infiltrating lymphocytes expressed HSP72 in 58% of cases. The pathological subtypes with the higher expression in lymphocytes were mixed cellularity and nodular sclerosis. No differences in HSP70 expression were observed, according to clinical stage, treatment response or the presence of EBV. Conclusions: The expression of HSP72 on lymphocytes suggests that this protein plays an important role in the induction and amplification of anti-tumor immune response (Rev Méd Chile 2003; 131: 1375-81)

Resumen en español Introducción: los rotavirus son la principal causa de gastroenteritis en niños menores de cinco años. Hasta el momento se desconoce cómo entran estos virus a la célula huésped. La proteína de choque térmico, HSC70 se ha visto involucrada en la entrada del virus, sin embargo no se han relacionado bien todos los sitios de unión entre las proteínas virales del rotavirus y la HSC70. Objetivos: el propósito de este trabajo fue determinar cuáles proteínas virales d (mas) el rotavirus son responsables de la unión de la proteína de choque térmico, HSC70. Material y métodos: se realizaron ensayos de coinmunoprecipitación utilizando lisados de células infectadas así como proteínas virales recombinantes. Estos ensayos se revelaron mediante SDS-PAGE y Western blot utilizando anticuerpos policlonales que reconocen las proteínas del rotavirus. Resultados. En los experimentos realizados se encontró que la proteína viral VP6 coinmunoprecipita con HSC70 en las células MA104 durante la infección con rotavirus. La VP6, producida con vaccinia recombinante en MA104, también coinmunoprecipitó con HSC70. Conclusión. La proteína VP6 de rotavirus interacciona con la proteína HSC70 en lisados de células MA104 infectadas con rotavirus. La unión de la VP6 recombinante a la HSC70 sugiere que la interacción de estas proteínas es independiente de otras proteínas virales. Resumen en inglés Introduction: Rotaviruses are the main cause of gastroenteritis in children under five years old. At this moment is unknown how these viruses entry to host cell. The heat shock cognate protein HSC70 has been involved in rotavirus cell entry. However, the relationship between rotavirus proteins and HSC70 has not been established. Objectives: the purpose of this work was to determine wich rotaviruses proteins interact with HSC70. Material and methods: coimmunoprecipitation (mas) assays were done on a rotavirus infected supernatant and recombinant viral proteins. These assays were revealed by SDS-PAGE and Western blot using policlonal antibodys against rotavirus structural proteins. Results: data show that viral protein VP6 coimmunoprecipated with HSC70 in MA104 cells during infection. Recombinant vaccinia of VP6 produced in MA104 cells also coimmunoprecipitated with HSC70. Conclusions: these results suggest the interaction between VP6 and HSC70 during rotavirus infection. Furthermore, this union may be independent of other rotaviruses proteins.

Resumen en español Antecedentes. El osteoblasto tiene la capacidad de estimular la neoformación del tejido óseo, mediante la síntesis de proteínas tales como las proteínas morfogenéticas, factores de crecimiento y proteínas colágenas y no colágenas. A su vez, indirectamente controla los procesos de reabsorción sintetizando otra serie de proteínas estimuladoras de la actividad osteoclastogénica. Objetivo. El propósito de este trabajo de investigación fue analizar la expresión (mas) de la proteína de choque térmico HSC70 en un cultivo primario de osteoblastos, después de someterlo a una fuerza tensil mediante centrifugación, determinando el patrón de proteínas mediante análisis electroforético. Material y métodos. A partir de cráneos de ratones lactantes ICR se obtuvieron cultivos de células con características morfológicas tipo fibroblastoide, con prolongaciones alargadas y citoplasmas ligeramente cuboidales que sugiere un cultivo enriquecido de osteoblastos; los cuales fueron sometidos a centrifugación y se analizó por inmunocitoquímica, electroforesis y Western blot la expresión diferencial de la proteína HSC70. Resultados.La técnica de electroforesis SDS-PAGE unidimensional no permitió determinar diferencias en el patrón de corrido entre osteoblastos tratados respecto a los no tratados con la fuerza tensil. Sin embargo, la proteína HSC70 se sobre-expresa en osteoblastos sometidos a centrifugación analizada mediante la técnica de Western blot e inmuno­histoquímica. Conclusión.Aunque la electroforesis no determinó diferencias en el patrón de corrido, la proteína de estrés HSC70 se sobre-expresó en osteoblastos sometidos a centrifugación analizada mediante la técnica de Western blot e inmunocitoquímica, sugiriendo que es importante en este tipo de estrés. Resumen en inglés Background. Osteoblasts can stimulate new bone formation by protein synthesis, such as, bone morphogenetic proteins, growth factors and collagen and non-collagenous proteins. As well, they controls reabsortion processes, because they can synthesize proteins to promote osteoclastogenic activity, when necessary. Objetive. The objetive of this work was analyze the expression of the heat shock cognate protein HSC70 in a primary culture of murine osteoblasts, after putting und (mas) er a tensil force by centrifugation and determining the protein pattern by electrophoretic analysis. Materials and methods. From skulls of newborn mice ICR, cultures of cells were obtained with morphologic characteristics fibroblasts-like type, extended prolongations and cytoplasms slightly cuboidal and plump, it suggests an enriched culture of osteoblasts. Results. The technique of unidimensional electroforesis SDS-PAGE did not allow determine differences in the run pattern between osteoblasts treated with tensil force and control cells. Nevertheless, protein HSC70 overexpresion in osteoblasts was confirmed by Western blot and inmunocyto­chemistry on-are expressed. Conclusions. Electroforesis did not determine differences between control cell and cell submited to centrifugation. Stress protein HSC70 was expressed in osteoblasts submitted to centrifugación analyzed by Western's blot technology suggesting a important roll of this stres type.

Resumen en inglés Ischemia-reperfusion (IR) liver injury is associated with temporary clamping of hepatoduodenal ligament during liver surgery, hypoperfusion shock and graft failure after liver transplantation. Mechanisms of IR liver injury include: i) loss of calcium homeostasis, ii) reactive oxygen and nitrogen species generation, iii) changes in microcirculation, iv) Kupffer cell activation, and (v) complement activation. Pre-exposure of the liver to transient ischemia increases the tol (mas) erance to IR injury, a phenomenon known as hepatic ischemic preconditioning (IP). IP involves: i) recovery of the energy supply and calcium, sodium and pH homeostasis, ii) enhancement in the antioxidant potential, and iii) expression of multiple stress-response proteins, including acute phase proteins, heat shock proteins, and heme oxygenase. These observations and preliminary studies in humans give a rationale for the assessment of IP in minimizing or preventing IR injury during surgery and non surgical conditions of tissue hypoperfusion

Evaluación de la capacidad protectora de la alfa-cristalina en el endotelio vascular

Resumen en español El endotelio vascular es un órgano de vital importancia para el mantenimiento de la función circulatoria. Su disfunción se asocia con enfermedades vasculares de alta incidencia. La búsqueda de agentes que reviertan o atenúen los efectos del daño oxidativo sobre el endotelio constituye una prioridad. Se utilizó la línea celular H5V para evaluar el daño producido por el isoprostano iPF2-III, producto de la peroxidación del ácido araquidónico por la acción de lo (mas) s radicales libres, y la capacidad protectora de la a-cristalina frente a esta agresión. La supervivencia de los cultivos celulares se evaluó por el ensayo MTT. Se obtuvo una disminución de la supervivencia en los cultivos tratados con iPF2-III a concentraciones mayores de 300 nmol durante 18 h de exposición. El tratamiento con alfa-cristalina disminuyó el efecto de iPF2-III en 20 %. Este trabajo propuso el efecto de las proteínas de shock término como protectoras endoteliales Resumen en inglés The vascular endothelium is a vital organ for keeping the circulatory function; its dysfunction is associated with vascular diseases of high incidence. The search for agents that reverse or mitigate the effects of the oxidative damage on the endothelium is a high priority. H5V cell line was used to evaluate the damage caused by iPF2-III isoprostane, a product of the free-radical catalized peroxidation of arachidonic acid, and the alpha crystallin´s protective capacity to (mas) this attack. The survival of cell cultures was assessed through the MTT assay. There was reduced survival in cultures treated with iPF2-III isoprostane at concentrations higher than 300 nmol for 18 hrs of exposure. The therapy with alpha crystallin lowered the iPF2-III effect by 20%. This paper showed the effect of heat-shock proteins as endothelial protective agents
