Evaluación de aislamientos de Bacillus thuringiensis tóxicos a Diatraea saccharalis (Lepodoctera: Pyralidae)

Resumen en español La caña de azúcar es uno de los cultivos de mayor importancia en Venezuela. En el país se cultivan alrededor de 120.000 hectáreas y uno de los problemas que enfrenta es el ataque de insectos-plaga, entre los que destaca Diatraea saccharalis. Una alternativa para el control de este insecto es mediante el uso de microorganismos tales como Bacillus thuringiensis, el agente de control biológico más utilizado contra insectos del orden Lepidoptera. Esta bacteria se caract (mas) eriza por la producción de un cristal conformado por proteínas insecticidas denominadas “Cry”. En este trabajo se evaluaron siete cepas nativas de Bacillus thuringiensis mediante bioensayos frente a Diatraea saccharalis y, utilizando geles de SDS-PAGE, se reconocieron proteínas de aproximadamente 130 kDa en cada una de las muestras. Estas proteínas derivan de una inclusión parasporal de forma bipiramidal típica de las proteínas del tipo Cry1. Los bioensayos señalaron a la cepa HD-1 como la más tóxica, con un 90 % de mortalidad en las larvas, seguida por la cepa UCLA-46 con 73 %. La cepa UCLA-41 mostró sólo 63 % de mortalidad, pero su efecto sobre el crecimiento de las larvas fue similar al de la cepa comercial HD-1.

Growth, structural, thermal, optical and electrical properties of CuIn5S8/ Crecimiento, propiedades estructurales, térmicas, ópticas y eléctricas del CuIn5S8

Resumen en español En el presente trabajo el compuesto CuIn5S8 es obtenido por fusión directa de la mezcla estequiométrica de los elementos químicos constituyentes. Los datos de la difracción de rayos X en polvo, indexados con el programa TREOR 90, muestran un cristal monofásico de estructura cúbica spinel, con grupo espacial Fd3m y parámetro de celda unidad de 10.69 Å. De acuerdo con las medidas de Análisis Térmico Diferencia (ATD) este cristal funde congruentemente alrededor de (mas) 1084ºC. Se utilizaron medidas de tramitancia para obtener el coeficiente de absorción a. A partir de este último se encontraron dos brechas de energía Eg a temperatura ambiente, una directa y otra indirecta, con valores de 1.41 y 1.25 eV, respectivamente. Por primera vez se reporta la reflectividad de este compuesto, a partir de ésta se confirmaron los valores de las brechas de energía obtenidos de las medidas de tramitancia. El estudio de las propiedades eléctricas muestra que la conductividad del CuIn5S8 es tipo n. Dos energías de activación fueron estimadas de los diagramas de Arrhenius de la resistividad y de la concentración de portadores, 56 meV y 0.16 eV, respectivamente. Resumen en inglés In the present work the compound CuIn5S8 is obtained by direct fusion of the estoichiometric mixture of the constituent chemical elements. The data of the X-ray powder diffraction, indexed with the program TREOR 90, show a single-phase crystal with cubic spinel structure, space group Fd3m and unit cell parameter of 10,69 Å. Differential Thermal Analysis (DTA) measurements show that this crystal melts congruently around 1084ºC. Transmittance measurements were used to obt (mas) ain the absorption coefficient a. From this last one two energy gaps Eg were found at room temperature, one direct and another indirect, with 1.41 and 1,25 eV, respectively. The reflectivity of this compound is reported for the first time and is used to confirm the energy gaps values obtained from transmittance measurements. The study of the electrical properties shows that the conductivity of the CuIn5S8 is n-type. Two activation energies were estimated from the Arrhenius plots of the resistivity and the carrier concentration, 56 meV and 0.16 eV, respectively.

Microestructura y polimorfismo en valvas recientes de Diplodon chilensis patagonicus (d’Orbigny 1835)/ Microstructure and polymorphism Oí Diplodon chilensis patagonicus (d’Orbigny 1835) recent shells

Resumen en español Gracias a su potencial como registro ambiental, las valvas del molusco Diplodon han despertado recientemente el interés de la comunidad científca. En este trabajo, por medio de microscopía electrónica de barrido (SEM) y espectroscopía Raman se investigó la microestructura y el polimorfsmo en valvas recientes de Diplodon chilensis patagonicus. Se encontró que la valva es completamente aragonítica y que la capa nacarada “externa" es la región más favorable para r (mas) ealizar mediciones de isótopos estables o elementos traza. Los análisis de microscopía confocal y SEM realizados demostraron que un incremento anual está compuesto de dos bandas: una de color claro, depositada durante los meses fríos y una de color oscuro, depositada durante los meses cálidos. Ambas bandas están formadas por nácar, compuesto de plaquetas poligonales de aproximadamente 5-7 µm de diámetro y 0.6-2 µm de espesor. Las plaquetas se encuentran ubicadas con el eje “c” del cristal (grupo cristalino: ortorrómbico) paralelo a la superfcie de la valva y están unidas entre sí por medio de material orgánico inter-cristalino, traslúcido a los electrones. El nácar depositado sobre la marca anual de crecimiento muestra plaquetas de espesores más delgados, coincidiendo con la hipótesis de un crecimiento desacelerado asociado a la época de reproducción. Como consecuencia, en esta zona la proporción de material orgánico es mayor que la de CaCO,, y explica la formación de una línea azul más nítida, cuando la muestra es teñida con Alcian blue Resumen en inglés Thanks to its potential as environmental archive, the val ves of the freshwater clam Diplodon recently gained the attention of the scientifc community. In this work, Scanning Electron Microscopy (SEM) and Raman Spectroscopy were used to investígate the material microstructure and polymorphism of recent Diplodon chilensis patagonicus shells. It was found that the shell is completely built of aragonite. In addition, the external nacreous layer shows to be the most appropri (mas) ate to do stable isotopes and trace elements measurements. Analyses with confocal microscopy and SEM demonstrated that an annual increment is composed of two bands: one of lighter colors deposited during the cold months and one of darker colors deposited during the summer time. Both bands are nacre, composed of polygonal aragonite platelets of approximately 5-7 µm diameter and 0.6-2 µm thickness. The platelets have its “c”-axis (crystal group: orthorhombic) parallel to the shell surface and are bound together with an inter-crystalline organic material, translucent to the electrons. The nacre deposited in the annual growth mark shows thinner platelets, coinciding with the hypothesis of a decelerated growth associated to the reproduction time. As consequence, the valve presents there higher organic to CaCO3, ratios and explains why the annual mark is dyed more sharply when using Alcian blue

Crecimiento y habilidad decolorante potencial de Hyphomycetes (Deuteromycetes) de Río Santiago sobre medio agarizado suplementado con cromóforos sintéticos/ Growth and potential decolorizing ability of Hyphomycetes (Deuteromycetes) from Río Santiago on agar medium supplemented with synthetic chromophores

Resumen en español Se analizó la capacidad de Dictyosporium triramosum, Minimidochium parvum y Tetraploa aristata, aisladas de materia orgánica colectada en Río Santiago (Provincia de Buenos Aires, Argentina), para crecer y decolorar medios agarizados y suplementados con diferentes cromóforos sintéticos al 0,01 % (p v-1). Cristal violeta y verde brillante redujeron el crecimiento de las 3 cepas revelando los mayores porcentajes de inhibición. Mientras que eosina y rosa de bengala no a (mas) fectaron a D. triramosum, valores superiores al 50 % de inhibición se observaron en M. parvum y T. aristata. Rojo congo y rojo neutral redujeron a D. triramosum y M. parvum en un 12-17 %, pero no a T. aristata. D. triramosum y T. aristata no resultaron afectados por azul de toluidina, mientras M. parvum fue inhibido por el colorante. Rojo de metilo sólo inhibió a M. parvum y T. aristata. Las 3 cepas probadas revelaron capacidad para decolorar el medio suplementado con azul de toluidina y rojo de metilo. D. triramosum decoloró además el medio suplementado con cristal violeta y rojo congo, y T. aristata el medio con cristal violeta y rojo neutral. Ninguno de los hongos estudiados decoloró los medios con eosina, rosa de bengala y verde brillante. Resumen en inglés The ability of Dictyosporium triramosum, Minimidochium parvum and Tetraploa aristata, isolated from organic matter collected in Río Santiago (Buenos Aires Province, Argentina), to grow and decolorize agar media supplemented with different synthetic chromophores at 0.01% (w v-1) was analyzed. Crystal violet and brilliant green reduced the growth of the three strains showing the highest inhibition percentages. While eosin and rose bengal did not affect D. triramosum, growt (mas) h inhibition values superior to 50 % were observed for M. parvum and T. aristata. Congo red and neutral red inhibited growth of both D. triramosum and M. parvum in a 12-17%, but those dyes did not reduced T. aristata growth. D. triramosum and T. aristata were not affected by the toluidine blue dye, while M. parvum was inhibited by that chromophore. Methyl red only inhibited to M. parvum and T. aristata. The 3 strains tested revealed ability to decolorize the medium supplemented with methyl red and toluidine blue. D. triramosum decolorized also the medium with congo red and crystal violet, and T. aristata produced also the decolorization of the agar medium with crystal violet and neutral red. None of the fungi studied decolorized the media with brilliant green, eosin and rose bengal.
