La mutación adaptativa: Polémicas y mecanismos

Resumen en español La variabilidad genómica, que es una propiedad intrínseca de los organismos, juega un rol esencial en el proceso evolutivo. Se analizan algunos de los procesos generadores de variabilidad genómica, con particular énfasis en el mecanismo de la mutagénesis que tiene lugar en las bacterias en las condiciones de estrés de fase estacionaria. Este proceso requiere la presencia de roturas en el ADN, la participación de proteínas de recombinación y reparación del ADN y de polimerasas específicas, y la inducción de respuestas celulares al estrés. Resumen en inglés Genomic variation plays an essential role in evolution. Some of the processes underlying genomic variability are being analyzed, with particular emphasis on the mechanism of mutagenesis taking place with bacteria under stationary-phase stress conditions. This process involves the presence of DNA nicks or breaks, the participation of recombination-repair proteins and specific polymerases, and the induction of cellular stress responses.

Desarrollo tecnológico y estudio de estabilidad del colirio de idoxiuridina 0,1 %/ Technological development and study of 0.1 % Idoxuridine (eyedrops)

Resumen en español La idoxiuridina, que se asemeja a la timidina, inhibe la timidílico fosforilasa y las polimerasas específicas del ADN que son necesarias para la incorporación de la timidina en el ADN del virus. La idoxiuridina se incorpora en el ADN defectuoso que incapacita al virus, para infectar o destruir tejidos o para reproducirse. El objetivo del presente trabajo consistió en el desarrollo tecnológico de una formulación de idoxiuridina, para su administración por vía ocula (mas) r, que cumple con todos los requisitos de calidad exigidos para el producto, como: características organolépticas, pH, valoración y esterilidad, con un tiempo de vida útil de 2 años, a temperatura de 2 a 8 oC, y que mantuvo invariables sus características físicas, químicas y microbiológicas durante el estudio de estabilidad realizado mediante el estudio de estabilidad acelerado a 25 oC y vida de estante de 2 a 8 oC, por espacio de 6 meses y de 24 meses, respectivamente. Resumen en inglés Idoxyuridine just like the Thymidine, inhibits thymidyl-phophorylase and DNA-specific polymerases, needed for Thymidine incorporation in virus-DNA. Idoxyuridine, added to defective DNA, renders unfit virus to infect or destroys tissues or for its reproduction. Aim of present paper was achieve technological development of a Idoxyuridine-formulation administered by ocular route, which meet all quality criteria called for the product, e.g. organoleptic features, pH, assessme (mas) nt and sterility, a 2 years useful lifetime, temperature of 2-8 °C, and with its invariable physical, chemical and microbiologic features over stability study performed by means of accelerated stability study to 25 °C, and support-life of 2-8 °C during 6 months and 24 months, respectively.

Strategies for introducing non-radioactive labels during the automated sequence analysis of nucleic acids.

Resumen en inglés Rapid, large-scale genome as well as diagnostic DNA sequencing projects are, at present, dependent on the use of sensitive automated sequencers that rely on the detection of fluorescent signals. This emission is not an intrinsic property of the biomolecules but is a property of an optical label that must be incorporated before, during or after the sequence specific reaction. The choice of strategy, that is to be reviewed here, is a function of both the chemistry and the e (mas) nzymology of the system as well as the nature of the fractionation and the physical parameters of the detection. One may differentiate beween the labelling of the primer, incorporation of the label during elongation or base specific termination with labelled terminators. In reviewing the development of each of these strategies, the conclusion is reached that, whereas labelled primers have universal applicability, the current generation of fluorescent terminators have in, conjunction with appropriate DNA polymerases, attained a refinement that strongly favours their use. However, since they are not available for all commercial sequencing systems, the alternative procedures are not merely of historic interest but are an essential component in DNA sequencing protocols. The possibility of automated direct sequence analysis of RNA has not been ignored completely
