Full Text Available A pinha, Annona squamosa L., é uma frutífera tropical da família anonácea, cujo mercado tem-se ampliado a cada ano, sendo cultivada expressivamente na região Nordeste, onde pequenos produtores a têm como principal fonte de renda. Entretanto, problemas causados pelas duas pragas-chave, Cerconota anonella (Sepp.,1830 (Lepidoptera: Oecophoridae e Bephratelloides pomorum (Fab.,1808 (Hymenoptera: Eurytomidae, têm limitado a produção e, consequentemente, a comercialização dos frutos. No intuito de minimizar essas perdas, um experimento de campo foi realizado em Maceió, Estado de Alagoas, Brasil, para avaliar diferentes formas de controle para estas pragas. O experimento foi conduzido no delineamento em blocos casualizados, com oito tratamentos e quatro repetições. Cada repetição correspondeu a quatro frutos, totalizando dezesseis frutos por tratamento. Os tratamentos foram: frutos sem proteção (testemunha; saco de papel branco impermeável aberto; saco plástico microperfurado; saco de TNT (tecido não tecido branco aberto; saco de TNT vermelho aberto; gaiola de arame revestida com tecido voile; inseticida Profenofós (12g/L-1 + Cipermetrina (1,2 g/L-1 e caulim (10 g/ 100 mL-1. Foram avaliadas as seguintes variáveis nos frutos: números de orifícios causados pelas pragas, peso, comprimento, diâmetro, percentagem de frutos colhidos e o custo do tratamento por unidade. Os melhores resultados foram obtidos com o saco de TNT vermelho aberto, obtendo-se 87,50% de frutos comercializáveis. O saco plástico microperfurado teve o menor custo, porém sua fragilidade impede a reutilização nas safras seguintes. Assim, indica-se o saco de TNT vermelho aberto como o mais econômico e eficiente.

Selenoprotein P Inhibits Radiation-Induced Late Reactive Oxygen Species Accumulation and Normal Cell Injury

Purpose: Radiation is a common mode of cancer therapy whose outcome is often limited because of normal tissue toxicity. We have shown previously that the accumulation of radiation-induced late reactive oxygen species (ROS) precedes cell death, suggesting that metabolic oxidative stress could regulate cellular radiation response. The purpose of this study was to investigate whether selenoprotein P (SEPP1), a major supplier of selenium to tissues and an antioxidant, regulates late ROS accumulation and toxicity in irradiated normal human fibroblasts (NHFs). Methods and Materials: Flow cytometry analysis of cell viability, cell cycle phase distribution, and dihydroethidium oxidation, along with clonogenic assays, were used to measure oxidative stress and toxicity. Human antioxidant mechanisms array and quantitative real-time polymerase chain reaction assays were used to measure gene expression during late ROS accumulation in irradiated NHFs. Sodium selenite addition and SEPP1 overexpression were used to determine the causality of SEPP1 regulating late ROS accumulation and toxicity in irradiated NHFs. Results: Irradiated NHFs showed late ROS accumulation (4.5-fold increase from control; P<.05) that occurs after activation of the cell cycle checkpoint pathways and precedes cell death. The mRNA levels of CuZn- and Mn-superoxide dismutase, catalase, peroxiredoxin 3, and thioredoxin reductase 1 increased approximately 2- to 3-fold, whereas mRNA levels of cold shock domain containing E1 and SEPP1 increased more than 6-fold (P<.05). The addition of sodium selenite before the radiation treatment suppressed toxicity (45%; P<.05). SEPP1 overexpression suppressed radiation-induced late ROS accumulation (35%; P<.05) and protected NHFs from radiation-induced toxicity (58%; P<.05). Conclusion: SEPP1 mitigates radiation-induced late ROS accumulation and normal cell injury.

Selenium (Se) supplementation has been used to help prevent the progression of Graves' ophthalmopathy (GO) and autoimmune thyroid diseases (AITD) patients. We investigated Se serum and selenoprotein P (SePP) levels in Graves' disease (GD) with and without GO, Hashimoto's thyroiditis (HT) patients and in 27 control individuals (C). We studied 54 female and 19 male patients: 19 with GD without GO, 21 GD with GO, 14 with HT and 19 with HT+LT4. Se values were measured using graphite furnace atomic absorption spectrophotometry. Serum SePP levels were measured by ELISA. Median Se levels were similar among all groups; GD patients: 54.2 (46.5-61.1 μg/L), GO: 53.6 (43.5-60.0 μg/L), HT: 51.9 (44.6-58.5 μg/L), HT+LT4 54.4 (44-63.4) and C group patients: 56.0 (52.4-61.5 μg/L); P = 0.48. However, serum SePP was lower in GO patients: 0.30 (0.15-1.05 μg/mL) and in HT patients: 0.35 (0.2-1.17 μg/mL) compared to C group patients: 1.00 (0.564.21 μg/mL) as well as to GD patients: 1.19 (0.62-2.5 μg/mL) and HT+LT4 patients: 0.7 (0,25-1.95); P = 0.002. Linear regression analysis showed a significant relationship between SePP and TPOAb values (r = 0.445, R2 = 0.293; P < 0.0001). Multiple regression analysis found no independent variables related to Se or SePP. A serum Se concentration was lower than in some other countries, but not significantly among AITD patients. The low serum SePP levels in GO and HT patients seems to express inflammatory reactions with a subsequent increase in Se-dependent protein consumption remains unclear.

Seasonal Water Transport in the Atmosphere of Mars: Applications of a Mars General Circulation Model Using Mars Global Surveyor Data

This is a Final Report for a Joint Research Interchange (JRI) between NASA Ames Research Center and San Jose State University, Department of Meteorology. We present below a summary of progress made during the duration of this JRI. The focus of this JRI has been to investigate seasonal water vapor transport in the atmosphere of Mars and its effects on the planet's present climate. To this end, the primary task has been to adapt a new dynamical processor for the adiabatic tendencies of the atmospheric circulation into the NASA Ames Mars general circulation model (MGCM). Using identical boundary and initial conditions, several comparative tests between the new and old MGCMs have been performed and the nature of the simulated circulations have been diagnosed. With confidence that the updated version of the Ames MGCM produces quite similar mean and eddy circulation statistics, the new climate model is well poised as a tool to pursue fundamental questions related to the spatial and seasonal variations of atmospheric water vapor on Mars, and to explore exchanges of water with non-atmospheric reservoirs and transport within its atmosphere. In particular, the role of surface sources and sinks can be explored, the range of water-vapor saturation altitudes can be investigated, and plausible precipitation mechanisms can be studied, for a range of atmospheric dust loadings. Such future investigations can contribute to a comprehensive study of surface inventories, exchange mechanisms, and the relative importance of atmospheric transport Mars' water cycle. A listing of presentations made and manuscripts submitted during the course of this project is provided.

Stochastic energy procurement of large electricity consumer considering photovoltaic, wind-turbine, micro-turbines, energy storage system in the presence of demand response program

The EPSs contain power market (PM), bilateral contracts (BCs), micro-turbines (MTs), and renewable energy sources (RESs). Moreover, the RESs include photovoltaic (PV) systems and wind-turbines (WT). The ESS and DRP are incorporated in the SEPP by the LEC’s decision-maker to reduce the expected energy procurement cost (EEPC). Meanwhile, the uncertainty models of market price, load and RES output power are considered in the SEPP formulation. The error of forecasting of market price, load, temperature and radiation of PV systems are modeled using the normal distribution for generating the related scenarios. Also, the weibull distribution is used to generate variable wind speed scenarios for WT output power uncertainty modeling. Furthermore, the fast forward selection based on Kantorovich distance approach is used for the scenarios reduction. Finally, the influences of ESS and DRP on EEPC are investigated, and four case studies are used to illustrate the capability of the proposed SEPP. Plasma Selenium Biomarkers in Low Income Black and White Americans from the Southeastern United States

Biomarkers of selenium are necessary for assessing selenium status in humans, since soil variation hinders estimation of selenium intake from foods. In this study, we measured the concentration of plasma selenium, selenoprotein P (SEPP1), and glutathione peroxidase (GPX3) activity and their interindividual differences in 383 low-income blacks and whites selected from a stratified random sample of adults aged 40–79 years, who were participating in a long-term cohort study in the southeastern United States (US). We assessed the utility of these biomarkers to determine differences in selenium status and their association with demographic, socio-economic, dietary, and other indicators. Dietary selenium intake was assessed using a validated food frequency questionnaire designed for the cohort, matched with region-specific food selenium content, and compared with the US Recommended Dietary Allowances (RDA) set at 55 µg/day. We found that SEPP1, a sensitive biomarker of selenium nutritional status, was significantly lower among blacks than whites (mean 4.4±1.1 vs. 4.7±1.0 mg/L, p = 0.006), with blacks less than half as likely to have highest vs. lowest quartile SEPP1 concentration (Odds Ratio (OR) 0.4, 95% Confidence Interval (CI) 0.2–0.8). The trend in a similar direction was observed for plasma selenium among blacks and whites, (mean 115±15.1 vs. 118±17.7 µg/L, p = 0.08), while GPX3 activity did not differ between blacks and whites (136±33.3 vs. 132±33.5 U/L, p = 0.320). Levels of the three biomarkers were not correlated with estimated dietary selenium intake, except for SEPP1 among 10% of participants with the lowest selenium intake (≤57 µg/day). The findings suggest that SEPP1 may be an effective biomarker of selenium status and disease risk in adults and that low selenium status may disproportionately affect black and white cohort participants. Full Text Available Abstract Background Selenium (Se status in non-deficient subjects is typically assessed by the Se contents of plasma/serum. That pool comprises two functional, specific selenoprotein components and at least one non-functional, non-specific components which respond differently to changes in Se intake. A more informative means of characterizing Se status in non-deficient individuals is needed. Methods Multiple biomarkers of Se status (plasma Se, serum selenoprotein P [SEPP1], plasma glutathione peroxidase activity [GPX3], buccal cell Se, urinary Se were evaluated in relation to selenoprotein genotypes (GPX1, GPX3, SEPP1, SEP15, dietary Se intake, and parameters of single-carbon metabolism in a cohort of healthy, non-Se-deficient men (n = 106 and women (n = 155. Conclusions Plasma Se concentration was 142.0 ± 23.5 ng/ml, with GPX3 and serum-derived SEPP1 calculated to comprise 20% and 34%, respectively, of that total. The balance, comprised of non-specific components, accounted for virtually all of the interindividual variation in total plasma Se. Buccal cell Se was associated with age and plasma homocysteine (hCys, but not plasma Se. SEPP1 showed a quadratic relationship with body mass index, peaking at BMI 25-30. Urinary Se was greater in women than men, and was associated with metabolic body weight (kg0.75, plasma folate, vitamin B12 and hCys (negatively. One GPX1 genotype (679T/T was associated with significantly lower plasma Se levels than other allelic variants. Selenium intake, estimated from food frequency questionnaires, did not predict Se status as indicated by any biomarker. These results show that genotype, methyl-group status and BMI contribute to variation in Se biomarkers in Se-adequate individuals.

Full Text Available Clinical trials have suggested a protective effect of selenium supplementation on the risk of esophageal cancer, which may be mediated through the antioxidant activity of selenoenzymes. We investigated whether serum selenium concentrations, selenoenzyme activity, oxidative stress and genetic variation in selenoenzymes were associated with the risk of neoplastic progression to esophageal adenocarcinoma (EA and two intermediate endpoints, aneuploidy and tetraploidy. In this prospective cohort study, during an average follow-up of 7.3 years, 47 EA cases, 41 aneuploidy cases and 51 tetraploidy cases accrued among 361 participants from the Seattle Barrett's Esophagus Research Study who were free of EA at the time of blood draw and had at least one follow-up visit. Development to EA was assessed histologically and aneuploidy and tetraploidy by DNA content flow cytometry. Serum selenium concentrations were measured using atomic absorption spectrometry, activity of glutathione peroxidase (GPX 1 and GPX3 by substrate-specific coupled test procedures, selenoprotein P (SEPP1 concentrations and protein carbonyl content by ELISA method and malondialdehyde concentrations by HPLC. Genetic variants in GPX1-4 and SEPP1 were genotyped. Serum selenium was not associated with the risk of neoplastic progression to EA, aneuploidy or tetraploidy (P for trend = 0.25 to 0.85. SEPP1 concentrations were positively associated with the risk of EA [hazard ratio (HR = 3.95, 95% confidence intervals (CI = 1.42-10.97 comparing the third tertile with the first] and with aneuploidy (HR = 6.53, 95% CI = 1.31-32.58, but not selenoenzyme activity or oxidative stress markers. No genetic variants, overall, were associated with the risk of neoplastic progression to EA (global p = 0.12-0.69. Our results do not support a protective effect of selenium on risk of neoplastic progression to EA. Our study is the first to report positive associations of plasma SEPP1

Full Text Available Nato il 10 aprile 1924 a Lahage (nell’Alta Garonna, in Francia, Louis Blajan si è spento lo scorso 10 febbraio 2010 a Mont-de-Marsan.Dopo aver frequentato il Lycée Pierre de Fermat di Tolosa, nel 1948 Louis completa gli studi presso la Scuola di Veterinaria di Tolosa. Nel 1949, si diploma all’Institut d’élevage et de médecine vétérinaire des pays tropicaux (IEMVT e dal 1949 al 1952 è nel territorio d’oltremare francese del Mali. Tornato in Francia, dal 1953 al 1968 Louis esercita come veterinario di Stato presso il Ministero dell’Agricoltura, responsabile per l’afta epizootica, la peste suina e la malattia di Newcastle, nonché come Presidente del Comitato consultivo per le malattie infettive. In seguito, viene nominato Ispettore capo veterinario e Responsabile per la politica di controllo delle frontiere.Nel 1952, Louis sposa Janine, moglie affezionata che lo appoggia con ardore in ogni fase della sua carriera e perde la vita in un incidente un mese prima che il marito vada in pensione nel 1990.Dal 1968 al 1977, Louis è Direttore tecnico di Cofranimex (Compagnie Française pour l’importation et l’exportation des animaux reproducteurs et leur semence e dal 1977 al 1978 riveste il ruolo di Direttore dell’Association pour le développement des techniques de l’élevage français (ADETEF.Louis entra a lavorare nell’Office International des Épizooties (oggi noto come Organizzazione Mondiale per la Sanità Animale o ‘OIE’ dove, dal 1978 al 1980, occupa la posizione di Direttore del Dipartimento tecnico. Due anni più tardi, nel 1980, viene eletto Direttore Generale, coronando il sogno di ogni professionista della medicina veterinaria e raggiungendo l’apice della propria carriera. In a population-based case-control study, we explored the associations between 42 polymorphisms in seven genes in this region and non-small cell lung cancer (NSCLC) risk among Caucasian (364 cases; 380 controls) and African American (95 cases; 103 controls) women. Two TERT region SNPs, rs2075786 and rs2853677, conferred an increased risk of developing NSCLC, especially among African American women, and TERT-rs2735940 was associated with a decreased risk of lung cancer among African Americans. Five of the 20 GHR polymorphisms and SEPP1-rs6413428 were associated with a marginally increased risk of NSCLC among Caucasians. Random forest analysis reinforced the importance of GHR among Caucasians and identified AMACR, TERT, and GHR among African Americans, which were also significant using gene-based risk scores. Smoking-SNP interactions were explored, and haplotype in TERT and GHR associated with NSCLC risk were identified. The roles of TERT, GHR, AMACR and SEPP1 genes in lung carcinogenesis warrant further exploration

Full Text Available In this paper the continuous dynamic model of Verhulst is used. It had been initially elaborated to describe the population growth of biological species with food restriction and physical space, in order to synthesize a new distributed power control algorithm (DPCA, applicable in direct sequence code division multiple access (DS-CDMA systems. The discretization of the corresponding differential equation is accomplished via numeric integration Euler method (NIE. The properties of the proposed recursive algorithm, as Euclidian distance from optimum vector after convergence, convergence speed, normalized mean squared error (NSE, average power consumption per user and implementation complexity, are investigated through simulations. The simulation results are confronted with the other DPCAs: the classic algorithm derived by Foschini and Miljanic and the sigmoidal by Uykan and Koivo. With estimate errors, the proposed DPCA showed smaller discrepancy from the optimum power vector allocation after convergence and better convergence. Additionally, the Gerchgorin Circles theory (GC is applied for the feasibility of the power allocation problem.

ABSTRACT Patients with inflammatory bowel disease are often deficient in micronutrients such as selenium and have an increased risk of colon cancer. We tested whether the selenium transport protein, selenoprotein P, could modify colitis-associated cancer. Mercury is a potent toxicant of concern to both the general public and occupationally exposed workers (e.g., dentists). Recent studies suggest that several genes mediating the toxicokinetics of mercury are polymorphic in humans and may influence inter-individual variability in mercury accumulation. This work hypothesizes that polymorphisms in key glutathione synthesizing enzyme, glutathione s-transferase, and selenoprotein genes underlie inter-individual differences in mercury body burden as assessed by analytical mercury measurement in urine and hair, biomarkers of elemental mercury and methylmercury, respectively. Urine and hair samples were collected from a population of dental professionals (n = 515), and total mercury content was measured. Average urine (1.06 {+-} 1.24 ug/L) and hair mercury levels (0.49 {+-} 0.63 ug/g) were similar to national U.S. population averages. Taqman assays were used to genotype DNA from buccal swab samples at 15 polymorphic sites in genes implicated in mercury metabolism. Linear regression modeling assessed the ability of polymorphisms to modify the relationship between mercury biomarker levels and exposure sources (e.g., amalgams, fish consumption). Five polymorphisms were significantly associated with urine mercury levels (GSTT1 deletion), hair mercury levels (GSTP1-105, GSTP1-114, GSS 5 Prime ), or both (SEPP1 3 Prime UTR). Glacial–interglacial dynamics of Antarctic firn columns: comparison between simulations and ice core air-δ15N measurements

Full Text Available Correct estimation of the firn lock-in depth is essential for correctly linking gas and ice chronologies in ice core studies. Here, two approaches to constrain the firn depth evolution in Antarctica are presented over the last deglaciation: outputs of a firn densification model, and measurements of δ15N of N2 in air trapped in ice core, assuming that δ15N is only affected by gravitational fractionation in the firn column. Since the firn densification process is largely governed by surface temperature and accumulation rate, we have investigated four ice cores drilled in coastal (Berkner Island, BI, and James Ross Island, JRI and semi-coastal (TALDICE and EPICA Dronning Maud Land, EDML Antarctic regions. Combined with available ice core air-δ15N measurements from the EPICA Dome C (EDC site, the studied regions encompass a large range of surface accumulation rates and temperature conditions. Our δ15N profiles reveal a heterogeneous response of the firn structure to glacial–interglacial climatic changes. While firn densification simulations correctly predict TALDICE δ15N variations, they systematically fail to capture the large millennial-scale δ15N variations measured at BI and the δ15N glacial levels measured at JRI and EDML – a mismatch previously reported for central East Antarctic ice cores. New constraints of the EDML gas–ice depth offset during the Laschamp event (~41 ka and the last deglaciation do not favour the hypothesis of a large convective zone within the firn as the explanation of the glacial firn model–δ15N data mismatch for this site. While we could not conduct an in-depth study of the influence of impurities in snow for firnification from the existing datasets, our detailed comparison between the δ15N profiles and firn model simulations under different temperature and accumulation rate scenarios suggests that the role of accumulation rate may have been underestimated in the current description of firnification

While many lesbian, gay, bisexual, transgender, and queer (LGBTQ) students are able to resiliently navigate their public school education many others experience harsh school climates and negative health and educational outcomes. Harassment and bullying of LGBTQ students in school environments have been linked to numerous negative psychological and academic outcomes for students diverse in sexual orientation and/or gender identity. Preparing teacher candidates (TCs) to respond effectively to harassment and bullying of students and to create inclusive curriculum has been recommended to improve outcomes for students. Yet the development of these teaching practices has not been pursued broadly in educator preparation programs (EPPs) or specifically in science EPPs (SEPPs). This dissertation broadens the notion of diversity traditionally attended to in EPPs through three studies. The first study is a holistic single-case study of an LGBTQ-inclusive EPP. It focused on the following three research questions: What were the contextual features that surrounded the LGBTQ-inclusive EPP? What were the specific elements of LGBTQ inclusion in the EPP? And, what were the strengths and weaknesses of the LGBTQ-inclusive EPP? This study drew primarily from data collected from interviews with faculty and administrators in a large post-baccalaureate 5th year preparation for licensure program. Document analysis was used to triangulate and expand upon the data collected during the interviews. A framework for analyzing LGBTQ inclusion across the components of an EPP was developed as part of this study. This study has direct implications for the particular EPP, but also clarifies research needs around LGBTQ inclusion in secondary EPPs. While little has research exists about LGBTQ inclusion in EPPs, far less has been attempted and understood in the discipline of secondary life science. Mass spectrometry is increasingly used in research on membrane lipid homeostasis, both in analyses of the steady state lipidome at the level of molecular lipid species, and in pulse-chase approaches employing stable isotope-labeled lipid precursors addressing the dynamics of lipid metabolism. Here my experience with, and view on mass spectrometry-based lipid analysis is presented, with emphasis on aspects of quantification of membrane lipid composition of the yeast Saccharomyces cerevisiae. Selenium (Se) incorporated in selenoproteins as selenocysteine and supports various important cellular and organismal functions. We recently reported that chicken brain exhibited high priority for Se supply and retention under conditions of dietary Se deficiency and supernutrition Li et al. (2012) . However, the selenotranscriptome expressions and their response to Se status in chicken central nervous system (CNS) are unclear. To better understand the relationship of Se homeostasis and selenoproteins expression in chicken CNS, 1day-old HyLine White chickens were fed a low Se diet (Se-L, 0.028mg/g) supplemented with 4 levels of dietary Se (0 to 5.0mgSe/kg) as Na 2 SeO 3 for 8weeks. Then chickens were dissected for getting the CNS, which included cerebral cortex, cerebellum, thalamus, bulbus cinereus and marrow. The expressions of selenoproteome which have 24 selenoproteins were detected by the quantitative real-time PCR array. The concept of a selenoprotein hierarchy was developed and the hierarchy of different regions in chicken CNS was existence, especially cerebral cortex and bulbus cinereus. The expression of selenoproteins has a hierarch while changing Se content, and Selenoprotein T (Selt), Selenoprotein K (Selk), Selenoprotein W (Selw), Selenoprotein U (Selu), Glutathione peroxidase 3 (Gpx3), Glutathione peroxidase 4 (Gpx4), Selenoprotein P (Sepp1), Selenoprotein O (Selo), Selenoprotein 15 (Sel15), Selenoprotein N (Seln), Glutathione peroxidase 2 (Gpx2) and Selenoprotein P 2 (Sepp2) take more necessary function in the chicken CNS. Therefore, we hypothesize that hierarchy of regulated the transcriptions of selenoproteome makes an important role of CNS Se metabolism and transport in birds. Copyright © 2017 Elsevier Inc. Background: Selenomethionine, which is the principal dietary form of selenium, is metabolized by the liver to selenide, which is the form of the element required for the synthesis of selenoproteins. The liver synthesizes selenium-rich selenoprotein P (SEPP1) and secretes it into the plasma to supply extrahepatic tissues with selenium. Objectives: We conducted a randomized controlled trial to determine whether cirrhosis is associated with functional selenium deficiency (the lack of selenium for the process of selenoprotein synthesis even though selenium intake is not limited) and, if it is, whether the deficiency is associated with impairment of selenomethionine metabolism. Design: Patients with Child-Pugh (C-P) classes A, B, and C (mild, moderate, and severe, respectively) cirrhosis were supplemented with a placebo or supranutritional amounts of selenium as selenate (200 or 400 μg/d) or as selenomethionine (200 μg/d) for 4 wk. Plasma SEPP1 concentration and glutathione peroxidase (GPX) activity, the latter due largely to the selenoprotein GPX3 secreted by the kidneys, were measured before and after supplementation. Results: GPX activity was increased more by both doses of selenate than by the placebo in C-P class B patients. The activity was not increased more by selenomethionine supplementation than by the placebo in C-P class B patients. Plasma selenium was increased more by 400 μg Se as selenate than by the placebo in C-P class C patients. Within the groups who responded to selenate, there was a considerable variation in responses. Conclusion: These results indicate that severe cirrhosis causes mild functional selenium deficiency in some patients that is associated with impaired metabolism of selenomethionine. The JSTAMP-Works/NV packaged the automatic healing function of CAD and had much more new capabilities such as prediction of 3D trimming lines for flanging or hemming, remote control of solver execution for multi-stage forming processes and shape evaluation between FEM and CAD. On the other way, a multi-stage multi-purpose inverse FEM solver HYSTAMP is developed and will be soon put into market, which is approved to be very fast, quite accurate and robust. Lastly, authors will give some application examples of user defined ductile damage subroutine in LS-DYNA for the estimation of material failure and springback in metal forming simulation.

Structure- and cell-specific effects of imidoselenocarbamates on selenoprotein expression and activity in liver cells in culture.

The essential micronutrient selenium (Se) exerts its biological effects mainly through selenoproteins thereby affecting a number of physiological pathways including intracellular redox control, stress response and cancer cell proliferation. Besides affecting selenoprotein expression, some selenocompounds have been synthesized and analyzed in order to serve as chemotherapeutic substances preferentially targeting cancer cells. This promising chemotherapeutic potential has recently been verified for a particular imidoselenocarbamate in a mouse tumor model. In the present study we tested the effects of this and a number of related Se-methyl- and Se-benzyl-imidoselenocarbamates on selenoprotein expression in nontransformed and hepatic carcinoma cells in culture. Most of the Se-benzyl-imidoselenocarbamates strongly stimulated selenoprotein P (SePP) secretion while the Se-methyl-imidoselenocarbamates elicited less pronounced effects in hepatocarcinoma HepG2 cells. However, most of the Se-methyl-imidoselenocarbamates increased glutathione peroxidase (GPx) activity and decreased thioredoxin reductase (TXNRD) activity in parallel, while the majority of the Se-benzyl-imidoselenocarbamates were without a respective effect in HepG2 cells. Performing inhibitor assays in vitro, GPx activity was unaffected by the imidoselenocarbamates. In contrast, most of the Se-methyl-imidoselenocarbamates inhibited TXNRD activity in vitro in line with the results in HepG2 cells. Both classes of imidoselenocarbamates strongly induced selenoprotein S (SELS) expression without a respective increase in ER stress or unfolded protein response which are known inducers of SELS biosynthesis. Notably, many of these effects were cancer cell-specific, and not observed in nontransformed AML12 hepatocytes. Our results indicate that these novel selenocompounds affect expression and activity of crucial selenoenzymes in a compound- and cell-specific way in hepatocytes. Especially the Se

We previously found that the anti-cancer drug methylglyoxal bis(guanylhydrazone) (mitoguazone) depresses carnitine-dependent oxidation of long-chain fatty acids in cultured mouse leukaemia cells [Nikula, Alhonen-Hongisto, Seppänen & Jänne (1984) Biochem. Biophys. Res. Commun. 120, 9-14]. We have now investigated whether carnitine also influences the development of the well-known mitochondrial damage produced by the drug in L1210 leukaemia cells. Palmitate oxidation was distinctly inhibited in tumour cells exposed to 5 microM-methylglyoxal bis(guanylhydrazone) for only 7 h. Electron-microscopic examination of the drug-exposed cells revealed that more than half of the mitochondria were severely damaged. Similar exposure of the leukaemia cells to the drug in the presence of carnitine not only abolished the inhibition of fatty acid oxidation but almost completely prevented the drug-induced mitochondrial damage. The protection provided by carnitine appeared to depend on the intracellular concentration of methylglyoxal bis(guanylhydrazone), since the mitochondria-sparing effect disappeared at higher drug concentrations.

Full Text Available The permaculture includes landscape shaping, terrace formation, establishment of raised planes and hilly beds. Sepp Holzer’s permaculture has been practices since 1962, having its followers in Columbia, Thailand, Brazil, the USA and Scotland, etc. Important specifics of the Fukuoka method consists of the requirement that the agricultural farm is close to nature, without any attempts to conquer or improve it, whence the method’s name “natural”, i.e. real or natural. In his book entitled “The One-Straw Revolution” the author examines the issues of natural agriculture. The Biodynamic French Intensive method of vegetable gardening combines two gardening traditions: biodynamics, created in the 1920s by Rudolph Steiner, and French intensive gardening, a method practiced by 19th century farmers on the outskirts of Paris. British horticulturist Alan Chadwick bought these methods to the United States in the 1960s and coined the term biointensive. This style of organic gardening focuses on sustainable, high productivity using minimal space.

Micro-Pressure Sensors for Future Mars Missions

The joint research interchange effort was directed at the following principal areas: u further development of NASA-Ames' Mars Micro-meteorology mission concept as a viable NASA space mission especially with regard to the science and instrument specifications u interaction with the flight team from NASA's New Millennium 'Deep-Space 2' (DS-2) mission with regard to selection and design of micro-pressure sensors for Mars u further development of micro-pressure sensors suitable for Mars The research work undertaken in the course of the Joint Research Interchange should be placed in the context of an ongoing planetary exploration objective to characterize the climate system on Mars. In particular, a network of small probes globally-distributed on the surface of the planet has often been cited as the only way to address this particular science goal. A team from NASA Ames has proposed such a mission called the Micrometeorology mission, or 'Micro-met' for short. Surface pressure data are all that are required, in principle, to calculate the Martian atmospheric circulation, provided that simultaneous orbital measurements of the atmosphere are also obtained. Consequently, in the proposed Micro-met mission a large number of landers would measure barometric pressure at various locations around Mars, each equipped with a micro-pressure sensor. Much of the time on the JRI was therefore spent working with the engineers and scientists concerned with Micro-met to develop this particular mission concept into a more realistic proposition.

An estimate of seasonality and intensity of infection with gastrointestinal nematodes in sheep and goats . in West Java

Each 3 months for 12 months worm-free male sheep (5 and goats (5 about 5 months of age were ~ $Jri~tgldI o% ef- farmer, and managed as part of their flock for 2 months . Animals were then returned to the laboratory and maintained on "`~^taaan-ftwAiet in elevated slatted pens for 3 weeks prior to slaughter. In all trials sheep had higher faecal egg counts than goats . Egg counts were significantly lower during the late dry-early wet season due mainly to lower burdens of Oesophagostomum spp. than at other times of the year. The predominant genera recovered from faecal larval cultures were Haemonchus and Trichostrongylus . At post mortem more than 94 percent of animals were infected with Trichostrongylus colubriformis, T. axei, Haemonchus contortus, Oesophagostomum columbianwn and Strongyloides papillosus . Other species found, in descending order of occurrence, were Cooperia curticei, Trichuris ovis, Bunostornum trigonocephalum, Oesophagostomumn asperum, Capillaria bovis and Gaigena pachycelis. It was concluded that intensity of exposure of both sheep and goats to H contortus, T. axei and C. curticei was similar throughout the year, but that availability of infectioe larvae of T. colubriformis was higher during the dry than the wet season and vise versa for O. columbianum . Sheep had higher burdens of T. Colubrzformis than goats but similar numbers of other species.

We report the clinical and radiological outcome of 86 revisions of cemented hip arthroplasties using JRI-Furlong hydroxyapatite-ceramic-coated acetabular and femoral components. The acetabular component was revised in 62 hips and the femoral component in all hips. The mean follow-up was 12.6 years and no patient was lost to follow-up. The mean age of the patients was 71.2 years. The mean Harris hip and Oxford scores were 82 (59 to 96) and 23.4 (14 to 40), respectively. The mean Charnley modification of the Merle d'Aubigné and Postel score was 5 (3 to 6) for pain, 4.9 (3 to 6) for movement and 4.4 (3 to 6) for mobility. Migration of the acetabular component was seen in two hips and the mean acetabular inclination was 42.6 degrees. The mean linear polyethylene wear was 0.05 mm/year. The mean subsidence of the femoral component was 1.9 mm and stress shielding was seen in 23 (28%) with bony ingrowth in 76 (94%). Heterotopic ossification was seen in 12 hips (15%). There were three re-revisions, two for deep sepsis and one for recurrent dislocation and there were no re-revisions for aseptic loosening. The mean EuroQol EQ-5D description scores and health thermometer scores were 0.69 (0.51 to 0.89) and 79 (54 to 95), respectively. With an end-point of definite or probable loosening, the probability of survival at 12 years was 93.9% and 95.6% for the acetabular and femoral components, respectively. Overall survival at 12 years, with removal or further revision of either component for any reason as the end-point, was 92.3%. Our study supports the continued use of this arthroplasty and documents the durability of hydroxyapatite-ceramic-coated components.

Full Text Available Neurocognitive impairment among children born preterm may arise from complex interactions between genes and the intra-uterine environment.(1 To characterize the transcriptomic profiles of chorioamniotic membranes in preterm neonates with and without neurocognitive impairment via microarrays and (2 to determine if neonates with neurocognitive impairment can be identified at birth.A retrospective case-control study was conducted to examine the chorioamniotic transcriptome of gestational-age matched very preterm neonates with and without neurocognitive impairment at 18-24 months' corrected-age defined by a Bayley-III Cognitive Composite Score 1.5; 2 Gene ontology analysis indicated enrichment of 19 biological processes and 3 molecular functions; 3PADOG identified 4 significantly perturbed KEGG pathways: oxidative phosphorylation, Parkinson's disease, Alzheimer's disease and Huntington's disease (q-value <0.1; 4 48 of 90 selected differentially expressed genes were confirmed by qRT-PCR, including genes implicated in energy metabolism, neuronal signaling, vascular permeability and response to injury (e.g., up-regulation of SEPP1, APOE, DAB2, CD163, CXCL12, VWF; down-regulation of HAND1, OSR1(p<0.05; and 5 a multi-gene model predicted 18-24 month neurocognitive impairment (using the ratios of OSR1/VWF and HAND1/VWF at birth in a larger, independent set (sensitivity = 74%, at specificity = 83%.Gene expression patterns in the chorioamniotic membranes link neurocognitive impairment in preterm infants to neurodegenerative disease pathways and might be used to predict neurocognitive impairment. Further prospective studies are needed.

HIPPI: highly accurate protein family classification with ensembles of HMMs

Full Text Available Abstract Background Given a new biological sequence, detecting membership in a known family is a basic step in many bioinformatics analyses, with applications to protein structure and function prediction and metagenomic taxon identification and abundance profiling, among others. Yet family identification of sequences that are distantly related to sequences in public databases or that are fragmentary remains one of the more difficult analytical problems in bioinformatics. Results We present a new technique for family identification called HIPPI (Hierarchical Profile Hidden Markov Models for Protein family Identification. HIPPI uses a novel technique to represent a multiple sequence alignment for a given protein family or superfamily by an ensemble of profile hidden Markov models computed using HMMER. An evaluation of HIPPI on the Pfam database shows that HIPPI has better overall precision and recall than blastp, HMMER, and pipelines based on HHsearch, and maintains good accuracy even for fragmentary query sequences and for protein families with low average pairwise sequence identity, both conditions where other methods degrade in accuracy. Conclusion HIPPI provides accurate protein family identification and is robust to difficult model conditions. Our results, combined with observations from previous studies, show that ensembles of profile Hidden Markov models can better represent multiple sequence alignments than a single profile Hidden Markov model, and thus can improve downstream analyses for various bioinformatic tasks. Further research is needed to determine the best practices for building the ensemble of profile Hidden Markov models. HIPPI is available on GitHub at https://github.com/smirarab/sepp .

Full Text Available This article suggests some pause for reflection amongst intellectual property lawyers, and for serious consideration of the words of an internationally-renowned IP law expert: "Possessing a right does not mean that it is a good idea to enforce it always, and to the hilt. Discretion may be nine parts of possession". It provides some prominent, recent examples of trademark bullying or overly-aggressive enforcement in the IP law context. These examples are mainly from other jurisdictions but they are directly relevant to some of the IP law challenges present in South Africa at the moment. The article further examines why lawyers and rights' holders engage in trademark bullying (why it's done, and start to deal briefly with some of the legal implications. A future article is to examine the legal aspects of trademark bullying in much more detail and considers its legitimacy within the context of IP law, more generally, and some other areas of law, more specifically.

Efficacy of insecticides in fruit borer control and residues on sugar apple fruit

Full Text Available ABSTRACT Bahia is the Brazilian state with the largest production of sugar apple fruits (Annona squamosa L., and fruit borer (Cerconota anonella, Sepp. 1830 is a key crop pest. Insecticides are the main strategy for pest control even though there are no pesticides registered for this crop. This study aimed to assess the efficacy of insecticides to control fruit borer and determine the levels of insecticide residues in sugar apple fruits aiming at requesting the extension of authorization to use insecticide products in this crop. The experiment was conducted in an eight-year-old irrigated orchard (2 × 4 m located in Anagé, Bahia, Brazil. The experimental design was a randomized block design with 10 treatments (three insecticides with three doses and a control with water and 5 replications. Each plot was composed of four plants but only the two central ones were assessed. Insecticides and doses (g a.i. 100 L−1 water were Bacillus thuringiensis: 0.8, 1.7, and 2.5; triflumuron: 2.4, 3.6, and 4.8; and imidacloprid: 4.0, 10.0, and 16.0. Nine sprayings were carried out at fortnightly intervals with a costal sprayer with constant pressure, JA-2 nozzle, and with jet directed to the fruits. Ten assessments were performed in order to observe fruit borer presence in 30 previously marked fruits per plot. Imidacloprid, at the highest studied dose, was the only effective treatment. Analyses of imidacloprid residues, at 21 and 30 days after the highest dose application, indicated levels higher than the maximum limit allowed. Insecticides under the conditions tested do not meet the norms for requesting the extension of authorization to use insecticides for citrus in sugar apple fruits.

Non-Dipole Features of the Geomagnetic Field May Persist for Millions of Years

Here we present paleointensity results from within the South Atlantic Anomaly (SAA), which is a large non-dipole feature of the geomagnetic field. Within the area of the SAA, anomalous declinations, inclinations, and intensities are observed. Our results suggest that the SAA has been present for at least 5 Ma. This is orders-of-magnitude greater than any previous estimate, and suggests that some non-dipole features do not `average out' over geologic time, which is a fundamental assumption in all paleodirectional studies. The SAA has been steadily growing in size since the first magnetic measurements were made in the South Atlantic, and it is widely believed to have appeared 400 years ago. Recent studies from South Africa (Tarduno et al. (2015)) and Tristan da Cunha (Shah et al. (2016)) have suggested that the SAA has persisted for 1 ka and 96 ka respectively. We conducted paleointensity (PI) experiments on basaltic lavas from James Ross Island, on the Antarctic Peninsula. This large shield volcano has been erupting regularly over the last 6+ Ma (dated via Ar/Ar geochronology), and therefore contains the most complete volcanostratigraphic record in the south Atlantic. Our PI experiments used the Thellier-Thellier method, the IZZI protocol, and the same selection criteria as the Lawrence et al. (2009) study of Ross Island lavas (near McMurdo Station), which is the only comparable PI study on the Antarctic continent. We determined an average paleointensity at JRI of 13.8±5.2 μT, which is far lower than what we would expect from a dipole field (55 μT). In addition, this is far lower than the current value over James Ross Island of 36 μT. These results support the following conclusions: The time-averaged field model of Juarez et al. (1998) and Tauxe et al. (2013) is strongly favored by our PI data. The SAA has persisted over James Ross Island for at least 5 Ma, and has not drifted significantly over that time. Memory and mood dysfunction are the key symptoms of Gulf war illness (GWI), a lingering multi-symptom ailment afflicting >200,000 veterans who served in the Persian Gulf War-1. Research probing the source of the disease has demonstrated that concomitant exposures to anti-nerve gas agent pyridostigmine bromide (PB), pesticides, and war-related stress are among the chief causes of GWI. Indeed, exposures to GWI-related chemicals (GWIR-Cs) and mild stress in animal models cause memory and mood impairments alongside reduced neurogenesis and chronic low-level inflammation in the hippocampus. In the current study, we examined whether exposure to GWIR-Cs and stress causes chronic changes in the expression of genes related to increased oxidative stress, mitochondrial dysfunction, and inflammation in the hippocampus. We also investigated whether GWI is linked with chronically increased activation of Nrf2 (a master regulator of antioxidant response) in the hippocampus, and inflammation and enhanced oxidative stress at the systemic level. Adult male rats were exposed daily to low-doses of PB and pesticides (DEET and permethrin), in combination with 5 min of restraint stress for 4 weeks. Analysis of the hippocampus performed 6 months after the exposure revealed increased expression of many genes related to oxidative stress response and/or antioxidant activity (Hmox1, Sepp1, and Srxn1), reactive oxygen species metabolism (Fmo2, Sod2, and Ucp2) and oxygen transport (Ift172 and Slc38a1). Furthermore, multiple genes relevant to mitochondrial respiration (Atp6a1, Cox6a1, Cox7a2L, Ndufs7, Ndufv1, Lhpp, Slc25a10, and Ucp1) and neuroinflammation (Nfkb1, Bcl6, Csf2, IL6, Mapk1, Mapk3, Ngf, N-pac, and Prkaca) were up-regulated, alongside 73–88% reduction in the expression of anti-inflammatory genes IL4 and IL10, and nuclear translocation and increased expression of Nrf2 protein. Memory and mood dysfunction are the key symptoms of Gulf war illness (GWI), a lingering multi-symptom ailment afflicting >200,000 veterans who served in the Persian Gulf War-1. Research probing the source of the disease has demonstrated that concomitant exposures to anti-nerve gas agent pyridostigmine bromide (PB), pesticides, and war-related stress are among the chief causes of GWI. Indeed, exposures to GWI-related chemicals (GWIR-Cs) and mild stress in animal models cause memory and mood impairments alongside reduced neurogenesis and chronic low-level inflammation in the hippocampus. Full Text Available O gênero Lepidagathis apresenta distribuição pantropical com cerca de 100 espécies. No Brasil ocorrem 16 espécies, a maioria nas regiões Centro - Oeste e Sudeste. O estudo foi realizado em sub - bosque de remanescente florestal do município de Tangará da Serra - MT e teve como objetivo analisar a fenologia de floração, descrever a morfologia e biologia floral, verificar os visitantes florais e avaliar o sistema e o sucesso reprodutivo por meio de polinizações manuais. Lepidagathis sessilifolia apresenta inflorescências espiciformes, terminais, com cálice de cor rósea vistosa e corola de coloração branco-rósea. A floração ficou restrita aos meses de março a abril, durante a estação chuvosa. A senescência floral ocorreu após 24 ou 48 horas. A viabilidade dos grãos de pólen foi elevada (92,5%. O único polinizador observado visitando as flores de L. sessilifolia foi a abelha Partamona nhambiquara (Apidae - Meliponini. O sistema reprodutivo misto da espécie é caracterizado pela formação de frutos por meio de agamospermia, autopolinização e polinização cruzada. Esse sistema reprodutivo flexível é vantajoso, pois, garante a manutenção da espécie na área de estudo mesmo na ausência de polinizadores.The genus Lepidagathis has pantropical distribution and about 100 species, 16 occurring in Brazil, mostly in the Midwest and Southeast regions. The research was carried out to evaluate in the understory of a forest remnant in Tangará da Serra County - MT to examine the flowering phenology, describe the floral morphology and biology and verify the floral visitors to evaluate the system and reproductive success through hand pollination tests. Lepidagathis sessilifolia shows spiciform and terminal inflorescence with calyx of pink color and corolla whitish-pink. Flowering was restricted from March to April, during the rainy season. Floral senescence occurred after 24 or 48 hours. The viability of pollen grains was high (92.5%. The effective pollinator of L. sessilifolia was Partamona nhambiquara (Apidae - Meliponini. The mixed mating system of the species is characterized by the formation of fruits by agamospermy, self and cross-pollination, spontaneous self-pollination, agamospermy and outcrossing. This flexible reproductive system is advantageous because it ensures the maintenance of species in the study area even in the absence of pollinators.

Full Text Available This paper presents an expert-based operationalization strategy to measure the degree of democracy in the Argentine provinces. Starting with a mainstream and “thick” definition of regime type, I assess each of its aspects using a subjective or perception-based approach that taps the knowledge of experts on the politics of each province. I present and justify the methodological design of the resulting Survey of Experts on Provincial Politics (SEPP and conduct a preliminary analysis of its results. Some aspects of the provincial regimes appear to be clearly democratic, while others are mixed or even leaning towards authoritarianism. Moreover, some show little interprovincial variance, while others vary considerably from province to province. An analysis of the central tendency and dispersion of the survey items allows for a general description of the Argentine provincial regimes. Inclusion is the most democratic dimension, while the effectiveness of institutional constraints on the power of the Executive is the most deficient. Electoral contestation is generally free of traditional forms of fraud, but incumbents often command far more campaign resources and media attention than do their challengers. Physical repression is rare, but opponents in some provinces face subtler forms of punishment. While the survey does not uncover any clear cases of subnational authoritarianism, stricto sensu, provincial regimes do vary significantly from basically democratic to clearly hybrid. This 120-page book is a concise, yet comprehensive, clearly-written and well-illustrated monograph that covers the subject matter from basic principles through design, construction and calibration details to the principal applications. The book will be useful, as a primer, to the uninitiated and, as a reference book to the practitioner involved with transformer-type ratio devices. The length of the book and the style of presentation will not overburden any informed reader. The described techniques and the cited references are primarily from the work at the National Research Council, Canada (NRC). Any omissions, however, are not serious with respect to coverage of the subject matter, since most of the development work has been done at NRC. The role of transformers and transformer-like devices for establishing accurate voltage and current ratios has been recognized for over half a century. Transformer techniques were much explored and developed in the fifties and sixties for accuracy levels suitable for standards laboratories. Three-winding voltage transformers were developed for scaling of impedances in connection with the calculable Thompson Lampard capacitor; three-winding current transformers or current comparators were initially explored for the calibration of current transformers and later for specialized impedance measurements. Extensive development of the current comparator and its applications has been and is still being conducted at the NRC by a team that was started and, until his retirement, led by N L Kusters. The team is now led by W J M Moore. He and P N Miljanic, the authors of this book, have had the principal roles in the development of the current comparator. It is fortunate for the field of metrology that considerabe resources and a talented group of researchers were available to do this development along with mechanisms that were available to transfer this technology to a private sector instrument manufacturer and, thus, disseminate it world wide

SMART-1 leaves Earth on a long journey to the Moon

The European Space Agency’s SMART-1 was one of three payloads on Ariane Flight 162. The generic Ariane-5 lifted off from the Guiana Space Centre, Europe’s spaceport at Kourou, French Guiana, at 2014 hrs local time (2314 hrs GMT) on 27 September (01:14 Central European Summer time on 28 September). 42 minutes after launch, SMART-1 as last of the three satellites had been successfully released into a geostationary transfer orbit (654 x 35 885 km, inclined at 7 degrees to the Equator). While the other two satellites are due to manoeuvre towards geostationary orbit, the 367 kg SMART-1 will begin a much longer journey to a target ten times more distant than the geostationary orbit: the Moon. “Europe can be proud”, said ESA Director General Jean-Jacques Dordain, after witnessing the launch from ESA’s ESOC space operations centre in Darmstadt, Germany, “we have set course for the Moon again. And this is only the beginning: we are preparing to reach much further”. The spacecraft has deployed its solar arrays and is currently undergoing initial checkout of its systems under control from ESA/ESOC. This checkout will continue until 4 October and will include with the initial firing of SMART-1’s innovative ion engine. By ion drive to the Moon “Science and technology go hand in hand in this exciting mission to the Moon. The Earth and Moon have over 4 thousand million years of shared history, so knowing the Moon better will help scientists in Europe and all over the world to better understand our planet and will give them valuable new hints on how to better safeguard it” said ESA Director of Science David Southwood, following the launch from Kourou.
