    This article reports the molecular cloning, protein expression and purification, crystallization and preliminary X-ray crystallographic analysis of the vibriobactin synthetase VibE from V. cholerae. Vibriobactin synthetases (VibABCDEFH) catalyze the biosynthesis of vibriobactin in the pathogenic bacterium Vibrio cholerae. VibE, a vibriobactin-specific 2,3-dihydroxybenzoate-AMP ligase, plays a critical role in the transfer of 2,3-dihydroxybenzoate to the aryl carrier protein domain of holo VibB. Here, the cloning, protein expression and purification, crystallization and preliminary X-ray crystallographic analysis of VibE from V. cholerae are reported. The VibE crystal diffracted to 2.3 Å resolution. The crystal belonged to space group P2 1 , with unit-cell parameters a = 56.471, b = 45.927, c = 77.014 Å, β = 95.895°. There is one protein molecule in the asymmetric unit, with a corresponding Matthews coefficient of 1.63 Å 3 Da −1 and solvent content of 24.41%

    Full Text Available Filamentous fungi that thrive on plant biomass are the major producers of hydrolytic enzymes used to decompose lignocellulose for biofuel production. Although induction of cellulases is regulated at the transcriptional level, how filamentous fungi sense and signal carbon-limited conditions to coordinate cell metabolism and regulate cellulolytic enzyme production is not well characterized. By screening a transcription factor deletion set in the filamentous fungus Neurospora crassa for mutants unable to grow on cellulosic materials, we identified a role for the transcription factor, VIB1, as essential for cellulose utilization. VIB1 does not directly regulate hydrolytic enzyme gene expression or function in cellulosic inducer signaling/processing, but affects the expression level of an essential regulator of hydrolytic enzyme genes, CLR2. Transcriptional profiling of a Δvib-1 mutant suggests that it has an improper expression of genes functioning in metabolism and energy and a deregulation of carbon catabolite repression (CCR. By characterizing new genes, we demonstrate that the transcription factor, COL26, is critical for intracellular glucose sensing/metabolism and plays a role in CCR by negatively regulating cre-1 expression. Deletion of the major player in CCR, cre-1, or a deletion of col-26, did not rescue the growth of Δvib-1 on cellulose. However, the synergistic effect of the Δcre-1; Δcol-26 mutations circumvented the requirement of VIB1 for cellulase gene expression, enzyme secretion and cellulose deconstruction. Our findings support a function of VIB1 in repressing both glucose signaling and CCR under carbon-limited conditions, thus enabling a proper cellular response for plant biomass deconstruction and utilization.

    The hydrolysis of biomass to simple sugars used for the production of biofuels in biorefineries requires the action of cellulolytic enzyme mixtures. During the last 50 years, the ascomycete Trichoderma reesei , the main source of industrial cellulase and hemicellulase cocktails, has been subjected to several rounds of classical mutagenesis with the aim to obtain higher production levels. During these random genetic events, strains unable to produce cellulases were generated. Here, whole genome sequencing and transcriptomic analyses of the cellulase-negative strain QM9978 were used for the identification of mutations underlying this cellulase-negative phenotype. Sequence comparison of the cellulase-negative strain QM9978 to the reference strain QM6a identified a total of 43 mutations, of which 33 were located either close to or in coding regions. From those, we identified 23 single-nucleotide variants, nine InDels, and one translocation. The translocation occurred between chromosomes V and VII, is located upstream of the putative transcription factor vib1 , and abolishes its expression in QM9978 as detected during the transcriptomic analyses. Ectopic expression of vib1 under the control of its native promoter as well as overexpression of vib1 under the control of a strong constitutive promoter restored cellulase expression in QM9978, thus confirming that the translocation event is the reason for the cellulase-negative phenotype. Gene deletion of vib1 in the moderate producer strain QM9414 and in the high producer strain Rut-C30 reduced cellulase expression in both cases. Overexpression of vib1 in QM9414 and Rut-C30 had no effect on cellulase production, most likely because vib1 is already expressed at an optimal level under normal conditions. We were able to establish a link between a chromosomal translocation in QM9978 and the cellulase-negative phenotype of the strain.   4. Carbide-forming groups IVB-VIB metals: a new territory in the periodic table for CVD growth of graphene.

    Early transition metals, especially groups IVB-VIB metals, can form stable carbides, which are known to exhibit excellent "noble-metal-like" catalytic activities. We demonstrate herein the applications of groups IVB-VIB metals in graphene growth using atmospheric pressure chemical vapor deposition technique. Similar to the extensively studied Cu, Ni, and noble metals, these transition-metal foils facilitate the catalytic growth of single- to few-layer graphene. The most attractive advantage over the existing catalysts is their perfect control of layer thickness and uniformity with highly flexible experimental conditions by in situ converting the dissolved carbons into stable carbides to fully suppress the upward segregation/precipitation effect. The growth performance of graphene on these transition metals can be well explained by the periodic physicochemical properties of elements. Our work has disclosed a new territory of catalysts in the periodic table for graphene growth and is expected to trigger more interest in graphene research.

  6. Vib--rotational energy distributions and relaxation processes in pulsed HF chemical lasers

    The rate equations governing the temporal evolution of photon densities and level populations in pulsed F+H 2 →HF+H chemical lasers are solved for different initial conditions. The rate equations are solved simultaneously for all relevant vibrational--rotational levels and vibrational--rotational P-branch transitions. Rotational equilibrium is not assumed. Approximate expressions for the detailed state-to-state rate constants corresponding to the various energy transfer processes (V--V, V--R,T, R--R,T) coupling the vib--rotational levels are formulated on the basis of experimental data, approximate theories, and qualitative considerations. The main findings are as follows: At low pressures, R--T transfer cannot compete with the stimulated emission, and the laser output largely reflects the nonequilibrium energy distribution in the pumping reaction. The various transitions reach threshold and decay almost independently and simultaneous lasing on several lines takes place. When a buffer gas is added in excess to the reacting mixture, the enhanced rotational relaxation leads to nearly single-line operation and to the J shift in lasing. Laser efficiency is higher at high inert gas pressures owing to a better extraction of the internal energy from partially inverted populations. V--V exchange enhances lasing from upper vibrational levels but reduces the total pulse intensity. V--R,T processes reduce the efficiency but do not substantially modify the spectral output distribution. The photon yield ranges between 0.4 and 1.4 photons/HF molecule depending on the initial conditions. Comparison with experimental data, when available, is fair

  10. Metastable Structural Phases of Metals in Columns IVB to Vib, and Rows 4 TO 6 OF the Periodic Table

    Total energy calculations as a function of strain along the direction have been carried out for the bcc metals V, Nb, Ta, Cr, Mo and W, and the hcp metals Ti, Zr and Hf, all in the block of the periodic table defined by columns IVB to VIB, and rows 4 to 6. Since strain along the direction corresponds to variation of the c lattice constant with respect to the a lattice constant, the total energy per unit cell has being calculated as a function of the c/a ratio. The highly accurate FP-LAPW (Full Potential Linearized Augmented Plane Wave) band structure method in the DFT (Density Functional Theory) formalism has been used for the calculations. In all cases except for the hcp column IVB elements, Zr, Hf and Ti, a metastable state was predicted from the calculations. Electronic properties are computed for all structures and are correlated with electrical and mechanical properties of metastable phases that have been observed experimentally. Properties of metastable phases, which were predicted in this work but which as of yet have not been observed experimentally, have also been predicted. Special attention is paid to the phases of tantalum and calculated transport properties are used to show that the observed high resistivity of the beta phase of tantalum relative to the alpha bcc phase cannot be explained solely by simple tetragonal distortions of the bcc phase.

  11. A molecular beam study of the one, two, and three photon photodissociation mechanism of the group VIB (Cr,Mo,W) hexacarbonyls at 248 nm

    Photodissociation of the group VIB (Cr,Mo,W) hexacarbonyls has been studied at 248 nm using molecular beam photofragment spectroscopy. One, two and three photon processes have been observed. Analysis of the product velocity distributions shows that the photodissociation mechanism consists of sequential CO eliminations with the nth photon channel best described as the single photon photodissociation of the stable products of the n-1st photon channel. The product translational energy distribution for the first CO elimination step is quantitatively similar for all three hexacarbonyls and characteristic of a repulsive translational energy release. The product translational energy distributions of all subsequent CO elimination steps are accurately described by a simple, microcanonical model. Qualitative molecular orbital considerations suggest that the large product translational energy observed in the first CO elimination step results from a repulsive σ interaction between the closed shell CO ligand and an excited molecular orbital which has a significant admixture of metal (n+1)p z , (n+1)s and nd z 2 orbitals. This repulsive interaction is absent in the remaining CO elimination steps because there are vacancies in the coordination shell along the z axis

    During the administration of timed bilateral alternate vibration to homonymous leg or trunk muscles during quiet upright stance, Parkinsonian (PD) patients undergo cyclic antero-posterior and medio-lateral transfers of the centre of foot pressure. This event might be potentially exploited for improving gait in these patients. Here, we tested this hypothesis by applying alternate muscle vibration during walking in PD. Fifteen patients and 15 healthy subjects walked on an instrumented walkway under four conditions: no vibration (no-Vib), and vibration of tibialis anterior (TA-Vib), soleus (Sol-Vib) and erector spinae (ES-Vib) muscles of both sides. Trains of vibration (internal frequency 100 Hz) were delivered to right and left side at alternating frequency of 10% above preferred step cadence. During vibration, stride length, cadence and velocity increased in both patients and healthy subjects, significantly so for ES-Vib. Stance and swing time tended to decrease. Width of support base increased with Sol-Vib or TA-Vib, but was unaffected by ES-Vib. Alternate ES vibration enhances gait velocity in PD. The stronger effect of ES over leg muscle vibration might depend on the relevance of the proprioceptive inflow from the trunk muscles and on the absence of adverse effects on the support base width. Trunk control is defective in PD. The effect of timed vibratory stimulation on gait suggests the potential use of trunk proprioceptive stimulation for tuning the central pattern generators for locomotion in PD.     High-frequency, low-magnitude vibration enhances bone formation ostensibly by mimicking normal postural muscle activity. We tested this hypothesis by examining whether daily exposure to low-magnitude vibration (VIB) would maintain bone in a muscle disuse model with botulinum toxin type A (BTX). Female 16-18 wk old BALB/c mice (N = 36) were assigned to BTX-VIB, BTX-SHAM, VIB, or SHAM. BTX mice were injected with BTX (20 µL; 1 U/100 g body mass) into the left hindlimb posterior musculature. All mice were anaesthetized for 20 min/d, 5 d/wk, for 3 wk, and the left leg mounted to a holder. Through the holder, VIB mice received 45 Hz, ± 0.6 g sinusoidal acceleration without weight bearing. SHAM mice received no vibration. At baseline and 3 wk, muscle cross-sectional area (MCSA) and tibial bone properties (epiphysis, metaphysis and diaphysis) were assessed by in vivo micro-CT. Bone volume fraction in the metaphysis decreased 12 ± 9% and 7 ± 6% in BTX-VIB and BTX-SHAM, but increased in the VIB and SHAM. There were no differences in dynamic histomorphometry outcomes between BTX-VIB and BTX nor between VIB and SHAM. Thus, vibration did not prevent bone loss induced by a rapid decline in muscle activity nor produce an anabolic effect in normal mice. The daily loading duration was shorter than would be expected from postural muscle activity, and may have been insufficient to prevent bone loss. Based on the approach used in this study, vibration does not prevent bone loss in the absence of muscle activity induced by BTX.

  15. Tendon vibration attenuates superficial venous vessel response of the resting limb during static arm exercise

    Full Text Available Abstract Background The superficial vein of the resting limb constricts sympathetically during exercise. Central command is the one of the neural mechanisms that controls the cardiovascular response to exercise. However, it is not clear whether central command contributes to venous vessel response during exercise. Tendon vibration during static elbow flexion causes primary muscle spindle afferents, such that a lower central command is required to achieve a given force without altering muscle force. The purpose of this study was therefore to investigate whether a reduction in central command during static exercise with tendon vibration influences the superficial venous vessel response in the resting limb. Methods Eleven subjects performed static elbow flexion at 35% of maximal voluntary contraction with (EX + VIB and without (EX vibration of the biceps brachii tendon. The heart rate, mean arterial pressure, and rating of perceived exertion (RPE in overall and exercising muscle were measured. The cross-sectional area (CSAvein and blood velocity of the basilic vein in the resting upper arm were assessed by ultrasound, and blood flow (BFvein was calculated using both variables. Results Muscle tension during exercise was similar between EX and EX + VIB. However, RPEs at EX + VIB were lower than those at EX (P P vein in the resting limb at EX decreased during exercise from baseline (P vein at EX + VIB did not change during exercise. CSAvein during exercise at EX was smaller than that at EX + VIB (P vein did not change during the protocol under either condition. The decreases in circulatory response and RPEs during EX + VIB, despite identical muscle tension, showed that activation of central command was less during EX + VIB than during EX. Abolishment of the decrease in CSAvein during exercise at EX + VIB may thus have been caused by a lower level of central command at EX + VIB rather than EX.   16. Whole-Body-Vibration Training and Balance in Recreational Athletes With Chronic Ankle Instability.

      Deficits in the propioceptive system of the ankle contribute to chronic ankle instability (CAI). Recently, whole-body-vibration (WBV) training has been introduced as a preventive and rehabilitative tool.   To evaluate how a 6-week WBV training program on an unstable surface affected balance and body composition in recreational athletes with CAI.   Randomized controlled clinical trial.   Research laboratory.   Fifty recreational athletes with self-reported CAI were randomly assigned to a vibration (VIB), nonvibration (NVIB), or control group.   The VIB and NVIB groups performed unilateral balance training on a BOSU 3 times weekly for 6 weeks. The VIB group trained on a vibration platform, and the NVIB group trained on the floor.   We assessed balance using the Biodex Balance System and the Star Excursion Balance Test (SEBT). Body composition was measured by dual-energy x-ray absorptiometry.   After 6 weeks of training, improvements on the Biodex Balance System occurred only on the Overall Stability Index ( P = .01) and Anterior-Posterior Stability Index ( P = .03) in the VIB group. We observed better performance in the medial ( P = .008) and posterolateral ( P = .04) directions and composite score of the SEBT in the VIB group ( P = .01) and in the medial ( P Balance System, whereas the VIB and NVIB groups displayed better performance on the SEBT.

O objetivo deste trabalho foi estimar a velocidade de infiltração da água e os teores de matéria orgânica do solo em sistema de consórcio milho e forrageiras. O experimento foi realizado em delineamento de blocos casualizados, com três repetições. Foram estudados os efeitos de cinco tratamentos submetidos ao manejo de plantio direto: 1 Milho Solteiro (Zea mays L. ; 2 Braquiária Humidícola (Brachiaria humidicola; 3 Consórcio Milho e Braquiária Humidícola; 4 Capim Aruana (Panicum maximum cv. Aruana; 5 Consórcio Milho e Capim Aruana. Utilizou-se, para determinação da velocidade de infiltração básica (VIB, o método dos infiltrômetros de cilindros concêntricos. Outros atributos físicos do solo e a matéria orgânica do solo foram avaliados buscando-se associá-los aos valores de VIB encontrados. Os resultados revelaram que sob plantio direto os sistemas apresentaram elevados valores de VIB. O sistema Milho Solteiro apresentou a maior VIB e a inserção de forrageiras nos sistemas provocou redução da VIB; todos os sistemas apresentaram diminuição dos teores de matéria orgânica em profundidade.Infiltration is a process by which water penetrates through the soil by its surface, and its dynamics can be influenced by several factors, among them, the soil management system adopted.   19. Use of Valtrac™-Secured Intracolonic Bypass in Laparoscopic Rectal Cancer Resection

    Abstract The occurrence of anastomotic leakage (AL) remains a major concern in the early postoperative stage. Because of the relatively high morbidity and mortality of AL in patients with laparoscopic low rectal cancer who receive an anterior resection, a fecal diverting method is usually introduced. The Valtrac™-secured intracolonic bypass (VIB) was used in open rectal resection, and played a role of protecting the anastomotic site. This study was designed to assess the efficacy and safety of the VIB in protecting laparoscopic low rectal anastomosis and to compare the efficacy and complications of VIB with those of loop ileostomy (LI). Medical records of the 43 patients with rectal cancer who underwent elective laparoscopic low anterior resection and received VIB procedure or LI between May 2011 and May 2013 were retrospectively analyzed, including the patients’ demographics, clinical features, and operative data. Twenty-four patients received a VIB and 19 patients a LI procedure. Most of the demographics and clinical features of the groups, including Dukes stages, were similar. However, the median distance of the tumor edge from the anus verge in the VIB group was significantly longer (7.5 cm; inter-quartile range [IQR] 7.0–9.5 cm) than that of the L1 group (6.0 cm; IQR 6.0–7.0 cm). None of the patients developed clinical AL. The comparisons between the LI and the VIB groups were adjusted for the significant differences in the tumor level of the groups. These diverse investigations on the function of the insect brain use gene expression patterns that can be visualized and provide the means for manipulating groups of neurons as a common ground. To take advantage of these patterns one needs to know their typical anatomy. Results This paper describes the Virtual Insect Brain (VIB) protocol, a script suite for the quantitative assessment, comparison, and presentation of neuroanatomical data. It is based on the 3D-reconstruction and visualization software Amira, version 3.x (Mercury Inc.) [1]. Besides its backbone, a standardization procedure which aligns individual 3D images (series of virtual sections obtained by confocal microscopy) to a common coordinate system and computes average intensities for each voxel (volume pixel) the VIB protocol provides an elaborate data management system for data administration. The VIB protocol facilitates direct comparison of gene expression patterns and describes their interindividual variability. It provides volumetry of brain regions and helps to characterize the phenotypes of brain structure mutants. Using the VIB protocol does not require any programming skills since all operations are carried out at an intuitively usable graphical user interface. Although the VIB protocol has been developed for the standardization of Drosophila neuroanatomy, the program structure can be used for the standardization of other 3D structures as well. Conclusion Standardizing brains and gene expression patterns is a new approach to biological shape and its variability.   4. Raman study of vibrational dephasing in liquid CH3CN and CD3CN

    The Raman line shapes of the ν 1 (a 1 ) C--H and C--D fundamentals in liquid acetonitrile and acetonitrile-d 3 have been measured as a function of pressure up to 4 kbar within the temperature interval 30--120 degreeC. Densities have also been determined. From the isotropic component of the vibrational Raman band shape the vibrational relaxation times have been obtained as a function of temperature and pressure (density). The experimental results can be summarized as follows: (i) as T increases at constant density rho, the vibrational relaxation rate (tau/sub vib/) -1 increases; (ii) at constant T as density is raised tau/sub vib/ -1 increases; (iii) at constant pressure the T increase produces higher tau/sub vib/ -1 , however, the change is more pronounced for the CD 3 CN liquid. Isotopic dilution studies of the CH 3 CN/CD 3 CN mixtures shows no significant effect on (tau/sub vib/ -1 ). The experimental data are interpreted in terms of the Kubo stochastic line shape theory and the dephasing model of Fischer and Laubereau. The results based on Kubo formalism indicate that dephasing is the dominant relaxation mechanism and that the modulation is fast. The isolated binary collision model proposed by Fischer and Laubereau for vibrational dephasing reproduces the essential features of the density and temperature dependence of the (tau/sub vib/) -1 and suggests that pure dephasing is the dominant broadening mechanism for the isotropic line shapes studied.     This article aims at a quantitative assessment of the performances of a panel of exchange-correlation functionals, including semilocal (BLYP and PBE), global hybrids (B3LYP, PBE0, M06, BHandHLYP, M06-2X, and M06-HF), and range-separated hybrids (CAM-B3LYP, LC-ωPBE, LC-BLYP, ωB97X, and ωB97X-D), in predicting the vibrationally resolved absorption spectra of BF 2 -carrying compounds. To this end, for 19 difluoroborates as examples, we use, as a metric, the vibrational reorganization energy (λ vib ) that can be determined based on the computationally efficient linear coupling model (a.k.a. vertical gradient method). The reference values of λ vib were determined by employing the CC2 method combined with the cc-pVTZ basis set for a representative subset of molecules. To validate the performances of CC2, comparisons with experimental data have been carried out as well. This study shows that the vibrational reorganization energy, involving Huang-Rhys factors and normal-mode frequencies, can indeed be used to quantify the reliability of functionals in the calculations of the vibrational fine structure of absorption bands, i.e., an accurate prediction of the vibrational reorganization energy leads to absorption band shapes better fitting the selected reference. The CAM-B3LYP, M06-2X, ωB97X-D, ωB97X, and BHandHLYP functionals all deliver vibrational reorganization energies with absolute relative errors smaller than 20% compared to CC2, whereas 10% accuracy can be achieved with the first three functionals. Indeed, the set of examined exchange-correlation functionals can be divided into three groups: (i) BLYP, B3LYP, PBE, PBE0, and M06 yield inaccurate band shapes (λ vib,TDDFT poor band topologies (λ vib,TDDFT > λ vib,CC2 ). This study also demonstrates that λ vib can be reliably estimated using the CC2 model and the relatively small cc-pVDZ basis set.     The excess vibrational entropy (Δ S {vib/ex}) of several silicate solid solutions are found to be linearly correlated with the differences in end-member volumes (Δ V i ) and end-member bulk moduli (Δκ i ). If a substitution produces both, larger and elastically stiffer polyhedra, then the substituted ion will find itself in a strong enlarged structure. The frequency of its vibration is decreased because of the increase in bond lengths. Lowering of frequencies produces larger heat capacities, which give rise to positive excess vibrational entropies. If a substitution produces larger but elastically softer polyhedra, then increase and decrease of mean bond lengths may be similar in magnitude and their effect on the vibrational entropy tends to be compensated. The empirical relationship between Δ S {vib/ex}, Δ V i and Δκ i , as described by Δ S {vib/ex} = (Δ V i + mΔκ i ) f, was calibrated on six silicate solid solutions (analbite-sanidine, pyrope-grossular, forsterite-fayalite, analbite-anorthite, anorthite-sanidine, CaTs-diopside) yielding m = 0.0246 and f = 2.926. It allows the prediction of Δ S {vib/ex} behaviour of a solid solution based on its volume and bulk moduli end-member data.

    A new series of ionic liquid functionalized copolymers for anion exchange membranes (AEM) is prepared. Poly(butylvinylimidazolium)(b-VIB) is copolymerized with para-methyl styrene (p-MS) by the radical polymerization formed block copolymers b-VIB/p-MS, which is crosslinked with poly(diphenylether bibenzimidazole) (DPEBI) providing the desired materials b-VIB/p-MS/DPEBI. Structures are characterized via H1NMR, FTIR spectra and elemental analysis. The b-VIB blocks offer the anion conduction function while DPEBI moieties contribute to enhancing other properties. The prepared membranes display chloride conductivity as high as 19.5 mS/cm at 25 °C and 69.2 mS/cm at 100 °C-higher than that of the commercial membrane tokuyuama A201-. Their hydroxide conductivity reaches 35.7 Scm-1 at 25 °C and 73.1 Scm-1 at 100 °C. The membranes showed a linear Arrhenius behavior in the anion conduction, low activation energies and distinguished nanophase separation of hydrophilic/hydrophobic regions by the transmission electron microscopy (TEM) studies. Thermal investigations using TGA and DSC confirm that the membranes are stable up to 250 °C. Particularly, drastically alkaline stability due to no decrease in the hydroxide conductivity after 168 h of treatment with 2M KOH.

    Whole-body vibration has been suggested for the prevention of muscle mass loss and muscle wasting as an attractive measure for disuse atrophy. This study examined the effects of daily intermittent whole-body vibration and weight bearing during hindlimb suspension on capillary number and muscle atrophy in rat skeletal muscles. Sixty male Wistar rats were randomly divided into four groups: control (CONT), hindlimb suspension (HS), HS + weight bearing (WB), and HS + whole-body vibration (VIB) (n = 15 each). Hindlimb suspension was applied for 2 weeks in HS, HS + WB, and HS + VIB groups. During suspension, rats in HS + VIB group were placed daily on a vibrating whole-body vibration platform for 20 min. In HS + WB group, suspension was interrupted for 20 min/day, allowing weight bearing. Untreated rats were used as controls. Soleus muscle wet weights and muscle fiber cross-sectional areas (CSA) significantly decreased in HS, HS + WB, and HS + VIB groups compared with CONT group. Both muscle weights and CSA were significantly greater in HS + WB and HS + VIB groups compared with HS group. Capillary numbers (represented by capillary-to-muscle fiber ratio) were significantly smaller in all hindlimb suspension-treated groups compared with CONT group. However, a reduction in capillary number by unloading hindlimbs was partially prevented by whole-body vibration. These findings were supported by examining mRNA for angiogenic-related factors. Expression levels of a pro-angiogenic factor, vascular endothelial growth factor-A mRNA, were significantly lower in all hindlimb suspension-treated groups compared with CONT group. There were no differences among hindlimb suspension-treated groups. Expression levels of an anti-angiogenic factor, CD36 (receptor for thrombospondin-1) mRNA, were significantly higher in all hindlimb suspension-treated groups compared with CONT group.   11. Development of high performance negative ion sources and accelerators for MeV class neutral beam injectors

    Operation of accelerator at low pressure is an essential requirement to reduce stripping loss of the negative ions, which in turn results in high efficiency of the NB systems. For this purpose, a vacuum insulated beam source (VIBS) has been developed at JAERI, which reduces the gas pressure in the accelerator by enhanced gas conductance through the accelerator. The VIBS achieves the high voltage insulation of 1 MV by immersing the whole structure of accelerator in vacuum with long (∼ 1.8 m) insulation distance. Results of the voltage holding test using a long vacuum gap of 1.8 m indicate that a transition from vacuum discharge to gas discharge occurs at around 0.2 Pa m in the long vacuum gap. So far, the VIBS succeeded in acceleration of 20 mA (H - ) beam up to 970 keV for 1 s. The high voltage holding capability of the VIBS was drastically improved by installing a new large stress ring, which reduces electric field concentration at the triple junction of the accelerator column. At present the VIBS sustains 1 MV stably for more than 1200 s. Acceleration of ampere class H- beams at high current density is to be started soon to demonstrate ITER relevant beam optics. Operation of negative ion source at low pressure is also essential to reduce the stripping loss. However, it was not so easy to attain high current density H - ions at low pressure, since destruction cross section of the negative ions becomes large if the electron temperature is > 1 eV, in low pressure discharge. Using strong magnetic filter to lower the electron temperature, and putting higher arc discharge power to compensate reduction of plasma density through the filter, an H - ion beam of 310 A/m 2 was extracted at very low pressure of 0.1Pa. This satisfies the ITER requirement of current density at 1/3 of the ITER design pressure (0.3 Pa). (author)

  12. The acute effects of stretching with vibration on dynamic flexibility in young female gymnasts.

    While stretching with vibration has been shown to improve static flexibility; the effect of stretching with vibration on dynamic flexibility is not well known. The purpose of this study was to examine the effectiveness of stretching with vibration on acute dynamic flexibility and jump height in novice and advanced competitive female gymnasts during a split jump. Female gymnast (n=27, age: 11.5 ± 1.7 years, Junior Olympic levels 5-10) participated in this cross-over study. Dynamic flexibility during gymnastic split jumps were video recorded and analyzed with Dartfish software. All participants completed both randomized stretching protocols with either the vibration platform turned on (VIB) (frequency of 30 Hz and 2 mm amplitude) or off (NoVIB) separated by 48 h. Participants performed 4 sets of three stretches on the vibration platform. Each stretch was held for 30 s with 5 s rest for a total of 7 min of stretch. Split jump flexibility decreased significantly from pre to post measurement in both VIB (-5.8°±5.9°) (p<0.001) and NoVIB (-2.6°±6.1°) (p=0.041) conditions (adjusted for gymnast level). This effect was greatest in lower skill level gymnasts (p=0.003), while the highest skill level gymnasts showed no significant decrease in the split jump (p=0.105). Jump height was not significantly different between conditions (p=0.892) or within groups (p=0.880). An acute session of static stretching with or without vibration immediately before performance does not alter jump height. Stretching with vibration immediately prior to gymnastics competition decreases split jump flexibility in lower level gymnasts more than upper level gymnasts.

    To prevent falls in the elderly, especially those with low bone density, is it necessary to maintain muscle coordination and balance. The aim of this study was to examine the effect of classical balance training (BAL) and whole-body vibration training (VIB) on postural control in post-menopausal women with low bone density. Sixty-eight subjects began the study and 57 completed the nine-month intervention program. All subjects performed resistive exercise and were randomized to either the BAL- (N=31) or VIB-group (N=26). The BAL-group performed progressive balance and coordination training and the VIB-group underwent, in total, four minutes of vibration (depending on exercise; 24-26Hz and 4-8mm range) on the Galileo Fitness. Every month, the performance of a single leg stance task on a standard unstable surface (Posturomed) was tested. At baseline and end of the study only, single leg stance, Romberg-stance, semi-tandem-stance and tandem-stance were tested on a ground reaction force platform (Leonardo). The velocity of movement on the Posturomed improved by 28.3 (36.1%) (ppostural control in post-menopausal women with low bone density.     We investigated the atomistic mechanism of He–He and He–metal interactions in bcc transition metals (V, Nb, Ta, Cr, Mo, W, and Fe) using first-principles methods. We calculated formation energy and binding energy of He–He pair as function of distance within the host lattices. The strengths of He–He attraction in Cr, Mo, W, and Fe (0.37–1.11 eV) are significantly stronger than those in V, Nb, and Ta (0.06–0.17 eV). Such strong attractions mean that He atoms would spontaneously aggregate inside perfect Cr, Mo, W, and Fe host lattices in absence of defects like vacancies. The most stable configuration of He–He pair is <100> dumbbell in groups VB metals, whereas it adopts close <110> configuration in Cr, Mo, and Fe, and close <111> configuration in W. Overall speaking, the He–He equilibrium distances of 1.51–1.55 Å in the group VIB metals are shorter than 1.65–1.70 Å in the group VB metals. Moreover, the presence of interstitial He significantly facilitates vacancy formation and this effect is more pronounced in the group VIB metals. The present calculations help understand the He-metal/He–He interaction mechanism and make a prediction that He is easier to form He cluster and bubbles in the groups VIB metals and Fe.

    Low energy electron loss spectroscopy (ELS) study was performed on 1T-HfSe/sub 2/ (group IVB metal compound) and 2H-NbSe/sub 2/ (group VB metal compound) by using incident electron energies of 30-250 eV. From the loss data in the second derivative form, maxima in density-of-states in the conduction band of the compounds were deduced through the information on the filled core states by X-ray photoelectron spectroscopy. The conduction band of the transition-metal dichalcogenides could be divided into two parts. The results are discussed in relation to the previous work on WS/sub 2/ (group VIB metal compound), and also to proposals based on band calculations and experimental studies on the transition-metal dichalcogenides with constituent metals of group IVB, VB and VIB.

    A diagnostic method has been evaluated for measuring the relative vibrational ground-state population of molecular hydrogen and deuterium. It is based on the analysis of the diagonal Fulcher bands · 3 Π u →a 3 Σ g + ) and the Franck-Condon principle of excitation. The validity of the underlying assumptions was verified by experiments in microwave discharges and the method is recommended for application in divertor plasmas in controlled fusion experiments. By attributing a vibrational temperature T vib to the ground-state electronic level (X 1 Σ g + ) and assuming population via the Franck-Condon principle, the upper Fulcher state vibrational distribution can be derived theoretically with T vib as parameter. Comparison with experimentally derived upper-state population gives the corresponding T vib of the ground state. The Franck-Condon factors for the · 3 Π 1 Σ g + and · 3 Π u →a 3 Σ g + transitions have been calculated for both hydrogen and deuterium from molecular constants using the FCFRKR code. The method has been applied to low pressure H 2 /He and D 2 /He microwave plasmas, showing good agreement of experimentally and theoretically derived upper Fulcher state vibrational distributions. The vibrational temperatures range from 3200 K to 6800 K for H 2 and 2600 K to 4000 K for D 2 · depending on molecular density, pressure and electron temperature, but indicating nearly the same vibrational population for H 2 and D 2 for comparable plasma conditions.     Vibrational energy relaxation (VER) of diatomics following collisions with the surrounding medium is an important elementary process for modeling high-temperature gas flow. VER is characterized by two parameters: the vibrational relaxation time τ vib and the state relaxation rates. Here the vibrational relaxation of CO(ν=0←ν=1) in Ar is considered for validating a computational approach to determine the vibrational relaxation time parameter (pτ vib ) using an accurate, fully dimensional potential energy surface. For lower temperatures, comparison with experimental data shows very good agreement whereas at higher temperatures (up to 25 000 K), comparisons with an empirically modified model due to Park confirm its validity for CO in Ar. Additionally, the calculations provide insight into the importance of Δν>1 transitions that are ignored in typical applications of the Landau-Teller framework.

    Full Text Available Este trabalho verificou se a velocidade de infiltração básica de água (VIB pode ser um indicador da qualidade do sistema poroso do solo cultivado com diferentes espécies vegetais. Foi realizado no Instituto Federal de Educação, Ciência e Tecnologia de Mato Grosso – campus Confresa, em um Argissolo Vermelho- Amarelo de textura arenosa. O delineamento experimental foi o de blocos casualizados com quatro tratamentos constituídos pelos cultivos de Brachiaria ruziziensis (braquiária, Pennisetum americanum (milheto, Phaseolus vulgaris L. (feijão e Sorghum bicolor L. Moench (sorgo em quatro repetições totalizando 16 parcelas. A ideia do cultivo das referidas espécies vegetais foi promover alteração natural do calibre dos poros do solo por meio dos seus sistemas radiculares e mineralização dos resíduos, viabilizando o estudo da velocidade de infiltração básica de água no solo como indicador da qualidade do calibre de poros. A partir de 90 dias após semeadura do milho avaliou-se a densidade do solo, volume de macroporos (Ma, microporos (Mi, porosidade total (VTP, resistência do solo à penetração (Rsp e VIB. As profundidades do solo consideradas foram 0-0,10 m e 0,10 a 0,20 m. Os dados numéricos foram submetidos a análise de variância e teste de significância de “F” ao nível de 5% de probabilidade. Características significativas tiveram suas médias comparadas pelo método Tukey. Entre as características citadas obtiveram-se os coeficientes de correlação lineares simples de Pearson. Nas condições experimentais avaliadas foi possível utilizar a VIB como indicador da qualidade do seu sistema poroso, embora tenha sido necessário realizar associações indiretas com as características avaliadas. Dessa forma, constatou-se tendência do aumento da VIB com aumento da Ma.

    With the childhood obesity epidemic, efficient methods of exercise are sought to improve health. We tested whether whole body vibration (WBV) exercise can positively affect bone metabolism and improve insulin/glucose dynamics in sedentary overweight Latino boys. Twenty Latino boys 8-10 years of age were randomly assigned to either a control (CON) or 3 days/wk WBV exercise (VIB) for 10-wk. Significant increases in BMC (4.5 ± 3.2%; p=0.01) and BMD (1.3 ± 1.3%; pBMC significantly increased (2.0 ± 2.2%; p=0.02), with no change in BMD (0.8 ± 1.3%; p=0.11). There were no significant between group changes in BMC or BMD. No significant change was observed for osteocalcin and (collagen type I C-telopeptide) CTx for the VIB group. However, osteocalcin showed a decreasing trend (p=0.09) and CTx significantly increased (pBMC and BMD by decreasing bone resorption in overweight Latino boys.

    Full Text Available We aimed to study whether short-duration vibration exercise or football sessions of two different durations acutely changed plasma markers of bone turnover and muscle strain. Inactive premenopausal women (n=56 were randomized to complete a single bout of short (FG15 or long duration (FG60 small sided football or low magnitude whole body vibration training (VIB. Procollagen type 1 amino-terminal propeptide (P1NP was increased during exercise for FG15 (51.6±23.0 to 56.5±22.5 μg·L−1, mean ± SD, P0.05. An increase in osteocalcin was observed 48 h after exercise (P<0.05, which did not differ between exercise groups. C-terminal telopeptide of type 1 collagen was not affected by exercise. Blood lactate concentration increased during exercise for FG15 (0.6±0.2 to 3.4±1.2 mM and FG60 (0.6±0.2 to 3.3±2.0 mM, but not for VIB (0.6±0.2 to 0.8±0.4 mM (P<0.05. Plasma creatine kinase increased by 55±63% and 137±119% 48 h after FG15 and FG60 (P<0.05, but not after VIB (26±54%, NS. In contrast to the minor elevation in osteocalcin in response to a single session of vibration exercise, both short and longer durations of small sided football acutely increased plasma P1NP, osteocalcin, and creatine kinase. This may contribute to favorable effects of chronic training on musculoskeletal health.

    Full Text Available Purpose: The purpose of this study was to investigate the effects of general and specific warm-up protocols on rate of force development (RFD, relative RFD (rRFD, ground reaction force (GRF and relative ground reaction force (rGRF during an isometric mid-thigh pull (IMTP, after WBV exposure. Methods: Fifteen healthy recreationally trained males  (age: 24.1 ± 2.3 yrs, height: 72.9 ± 7.8 cm; mass: 86.9 ± 8.3 completed five protocols: baseline, isometric vibration (iVib, isometric no vibration (iNV, dynamic vibration (dVib and dynamic no vibration (dNV. The baseline was completed without any warm-up prior to the IMTP. The intervention protocols had the same prescription of 4 sets of 30-second bouts of quarter squats (dynamic [DQS] and isometric [IQS] on the WBV platform with or without vibration. Following a one-minute rest period after each protocol, participants completed three maximal IMTPs. Results: Repeated measures ANOVA with a Bonferroni post hoc demonstrated that RFD in dNV (7657.8 ± 2292.5 N/s was significantly greater than iVib (7156.4 ± 2170.0 N/s. However, the other experimental trials for RFD demonstrated no significant differences (p>0.05. There were also no significant differences for rRFD, GRF or rGRF between protocols. Conclusion: These results demonstrate that a dynamic warm-up without WBV elicits greater RFD than an isometric warm-up with WBV prior to a maximal isometric exercise.     Full Text Available Objetivando estimar a velocidade de infiltração básica (VIB, avaliou-se a associação desta com outras propriedades físicas das camadas superficial e subsuperficial de um Latossolo Roxo e um Podzólico Vermelho-Amarelo, ambos ocorrentes no campus da Universidade Federal de Lavras. Para tanto, delineou-se um plano amostral consistindo de uma rede cujas malhas apresentaram espaçamentos variáveis. Nos pontos de interseção das malhas determinou-se a VIB e, posteriormente, coletaram-se amostras com estrutura deformada e indeformada nas camadas de 0-20 e 60-80 cm. Com essas amostras determinou-se a textura, densidade do solo e de partículas, porosidade total, macro e microporosidade e condutividade hidráulica do solo saturado. Após análises estatísticas de correlação e regressão, verificou-se que as propriedades físicas da camada de 0-20 cm do Latossolo Roxo e 60-80 cm do Podzólico Vermelho-Amarelo associaram-se melhor com a VIB. Tanto a condutividade hidráulica do solo saturado quanto a densidade do solo se mostraram adequadas para estimar a VIB desses solos.With the objective of estimating the soil basic infiltration velocity (BIV, a study was conducted to evaluate the degree of association between BIV and other physical properties of the superficial and subsuperficial layers of a Dusky-Red Latosol (Oxisol and of a Red-Yellow Podzolic soil (Ultisol, both located at the campus of the Universidade Federal de Lavras, Minas Gerais State, Brazil. The sampling was performed with systematic spacing and the points constituted a grid, with variable spacing.     Foam roller is a device used as a massage intervention for rehabilitation and fitness performance. To examine the effects on the ankle dorsiflexion mobility of the foam roller as well as the combination of foam roller and vibration applied to the ankle plantarflexors muscles, and to observe the possible cross-effect. Thirty-eight undergraduate students participated in the study (19 males and 19 females). This study investigated. Three conditions (3 sets of 20 s) were performed in a randomized order (independent variables): 1) foam roller (Roller), 2) foam roller and vibration (Roller+VIB), and 3) no foam roller or vibration (Control). to determine whether of foam roller with or without vibration would benefit ankle dorsiflexion mobility. Ankle dorsiflexion ROM and plantar flexor were measured in both legs before and immediately after the treatment. A cross-effect was found in the non-stimulated leg. There was a significant effect on ankle mobility of Roller and Roller+VIB conditions (6% and 7%, pFoam roller massage and vibration stimulus' foam roller massage increase ankle mobility producing a cross-effect.

    A system for exploration of active-faults by seismic reflection profiling method was introduced at Earthquake Research Institute, University of Tokyo. A test-run was conducted to check the performance of this system at Ranzan, Saitama Prefecture. This paper describes the confirmed performance of mini-VIB as a wide band frequency seismic source, the quality of data obtained using a digital data acquisition system, and problems for data processing of fault exploration in the future. For the test-run at Ranzan, two-dimensional exploration was conducted by the quasi-three-dimensional data acquisition method using three geophones of 8 Hz, 28 Hz, and 40 Hz, simply arranged in parallel on the measurement line. Using an active seismic vibrator, mini-VIB, data acquisition of faults in the wide band frequency was achieved, which would result in the highly accurate imaging. Operation of data acquisition and processing systems is easy, and the system can be also used as a kind of black box. The existing methods are to be used sufficiently as a tool for imaging of faults. Further research for accumulating experience may become necessary toward the extension of the system expected in the future. 5 refs., 6 figs.

    Article VI.B.2 of the Standard Contract for Disposal of Spent Nuclear Fuel and/or High-Level Radioactive Waste (10 CFR Part 961, Standard Contract) provides that spent nuclear fuel shall be subject to verification prior to acceptance by the Department of Energy. As part of the overall process for scheduling deliveries of spent nuclear fuel, the Standard Contract requires contract holders to submit detailed descriptions of the spent nuclear fuel they intend to deliver. Thus, the provision for verification in the Standard contract allows the Department the opportunity to ensure that the spent nuclear fuel intended for deliver is consistent with the description provided, and that the material is properly loaded, packaged, marked, and labeled. The Department is currently evaluating the verification requirements for spent nuclear fuel that it will establish pursuant to Article VI.B.2 of the Standard Contract. The Department recognizes that there may be significant overlap between these verification requirements and the requirements established for material control and accountability (MC ampersand A). Therefore, the Department may recommend modifications to the process established in the Standard Contract to ensure that the verification and MC ampersand A functions and activities are fully integrated

    Neuromuscular electrical stimulation (NMES) is commonly used to activate skeletal muscles and reverse muscle atrophy in clinical populations. Clinical recommendations for NMES suggest the use of short pulse widths (100-200 μs) and low-to-moderate pulse frequencies (30-50 Hz). However, this type of NMES causes rapid muscle fatigue due to the (non-physiological) high stimulation intensities and non-orderly recruitment of motor units. The use of both wide pulse widths (1000 μs) and tendon vibration might optimize motor unit activation through spinal reflex pathways and thus delay the onset of muscle fatigue, increasing muscle force and mass. Thus, the objective of this study was to examine the acute effects of patellar tendon vibration superimposed onto wide-pulse width (1000 μs) knee extensor electrical stimulation (NMES, 30 Hz) on peak muscle force, total impulse before "muscle fatigue", and the post-exercise recovery of muscle function. Tendon vibration (Vib), NMES (STIM) or NMES superimposed onto vibration (STIM + Vib) were applied in separate sessions to 16 healthy adults. Total torque-time integral (TTI), maximal voluntary contraction torque (MVIC) and indirect measures of muscle damage were tested before, immediately after, 1 h and 48 h after each stimulus. TTI increased (145.0 ± 127.7%) in STIM only for "positive responders" to the tendon vibration (8/16 subjects), but decreased in "negative responders" (-43.5 ± 25.7%). MVIC (-8.7%) and rectus femoris electromyography (RF EMG) (-16.7%) decreased after STIM (group effect) for at least 1 h, but not after STIM + Vib. No changes were detected in indirect markers of muscle damage in any condition. Tendon vibration superimposed onto wide-pulse width NMES increased TTI only in 8 of 16 subjects, but reduced voluntary force loss (fatigue) ubiquitously. Negative responders to tendon vibration may derive greater benefit from wide-pulse width NMES alone.

    Full Text Available In the race towards two-dimensional electronic and optoelectronic devices, semiconducting transition metal dichalcogenides (TMDCs from group VIB have been intensively studied in recent years due to the indirect to direct band-gap transition from bulk to the monolayer. However, new materials still need to be explored. For example, semiconducting TMDCs from group IVB have been predicted to have larger mobilities than their counterparts from group VIB in the monolayer limit. In this work we report the mechanical exfoliation of ZrX2 (X = S, Se from bulk down to the monolayer and we study the dimensionality dependence of the Raman spectra in ambient conditions. We observe Raman signal from bulk to few layers and no shift in the peak positions is found when decreasing the dimensionality. While a Raman signal can be observed from bulk to a bilayer for ZrS2, we could only detect signal down to five layers for flakes of ZrSe2. These results show the possibility of obtaining atomically thin layers of ZrX2 by mechanical exfoliation and represent one of the first steps towards the investigation of the properties of these materials, still unexplored in the two-dimensional limit.

  8. A novel bacteriocin-like substance (BLIS) from a pathogenic strain of Vibrio harveyi.

    Inter-strain and inter-species inhibition mediated by a bacteriocin-like inhibitory substance (BLIS) from a pathogenic Vibrio harveyi strain VIB 571 was demonstrated against four isolates of the same species, and one culture each of a Vibrio sp., Vibrio fischeri, Vibrio gazogenes and Vibrio parahaemolyticus. The crude BLIS, which was obtained by ammonium-sulphate precipitation of the cell-free supernatant of a 72 h broth culture of strain VIB 571, was inactivated by lipase, proteinase K, pepsin, trypsin, pronase E, SDS and incubation at > or =60 degrees C for 10 min. The activity was stable between pH 2-11 for at least 5 h. Anion-exchange chromatography, gel filtration, SDS-PAGE and two-dimensional gel electrophoresis revealed the presence of a single major peak, comprising a protein with a pI of approximately 5.4 and a molecular mass of approximately 32 kDa. The N-terminal amino acid sequence of the protein comprised Asp-Glu-Tyr-Ile-Ser-X-Asn-Lys-X-Ser-Ser-Ala-Asp-Ile (with X representing cysteine or modified amino acid residues). A similarity search based on the matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) generated peptide masses and the N-terminal sequence did not yield any significant matches.

    Full Text Available The production of virulence factors by many pathogenic microorganisms depends on the intercellular communication system called quorum sensing (QS, which involves the production and release of signal molecules known as autoinducers. Based on this, new-therapeutic strategies have emerged for the treatment of a variety of infections, such as the enzymatic degradation of signalling molecules, known as quorum quenching (QQ. In this study, we present the screening of QQ activity amongst 450 strains isolated from a bivalve hatchery in Granada (Spain, and the selection of the strain PQQ-42, which degrades a wide range of N-acylhomoserine lactones (AHLs. The selected strain, identified as Alteromonas stellipolaris, degraded the accumulation of AHLs and reduced the production of protease and chitinase and swimming motility of a Vibrio species in co-cultivation experiments in vitro. In the bio-control experiment, strain PQQ-42 significantly reduced the pathogenicity of V. mediterranei VibC-Oc-097 upon the coral Oculina patagonica showing a lower degree of tissue damage (29.25±14.63 % in its presence, compared to when the coral was infected with V. mediterranei VibC-Oc-097 alone (77.53±13.22 %. Our results suggest that this AHL-degrading bacterium may have biotechnological applications in aquaculture.

    Heterokaryon formation is an essential step in asexual recombination in Fusarium oxysporum. Filamentous fungi have an elaborate nonself recognition machinery to prevent formation and proliferation of heterokaryotic cells, called heterokaryon incompatibility (HI). In F. oxysporum the regulation of this machinery is not well understood. In Neurospora crassa, Vib-1, a putative transcription factor of the p53-like Ndt80 family of transcription factors, has been identified as global regulator of HI. In this study we investigated the role of the F. oxysporum homolog of Vib-1, called Suf, in vegetative hyphal and conidial anastomosis tube (CAT) fusion and HI. We identified a novel function for an Ndt80 homolog as a nutrient-dependent regulator of anastomosis. Strains carrying the SUF deletion mutation display a hyper-fusion phenotype during vegetative growth as well as germling development. In addition, conidial paring of incompatible SUF deletion strains led to more heterokaryon formation, which is independent of suppression of HI. Our data provides further proof for the divergence in the functions of different members Ndt80 family. We propose that Ndt80 homologs mediate responses to nutrient quality and quantity, with specific responses varying between species. Copyright © 2016 Elsevier Inc. All rights reserved.

    Full Text Available Cytochrome c oxidase (COX is the terminal enzyme of the electron transport chain and catalyzes the transfer of electrons from cytochrome c to oxygen. COX consists of 14 subunits, three and eleven encoded, respectively, by the mitochondrial and nuclear DNA. Tissue- and condition-specific isoforms have only been reported for COX but not for the other oxidative phosphorylation complexes, suggesting a fundamental requirement to fine-tune and regulate the essentially irreversible reaction catalyzed by COX. This article briefly discusses the assembly of COX in mammals and then reviews the functions of the six nuclear-encoded COX subunits that are expressed as isoforms in specialized tissues including those of the liver, heart and skeletal muscle, lung, and testes: COX IV-1, COX IV-2, NDUFA4, NDUFA4L2, COX VIaL, COX VIaH, COX VIb-1, COX VIb-2, COX VIIaH, COX VIIaL, COX VIIaR, COX VIIIH/L, and COX VIII-3. We propose a model in which the isoforms mediate the interconnected regulation of COX by (1 adjusting basal enzyme activity to mitochondrial capacity of a given tissue; (2 allosteric regulation to adjust energy production to need; (3 altering proton pumping efficiency under certain conditions, contributing to thermogenesis; (4 providing a platform for tissue-specific signaling; (5 stabilizing the COX dimer; and (6 modulating supercomplex formation.

    Full Text Available Tremendous effort has been expended over the past two and a half decades to understand many aspects of camelid heavy chain antibodies, from their biology, evolution, and immunogenetics to their potential applications in various fields of research and medicine. In this article, I present a historical perspective on the development of camelid single-domain antibodies (sdAbs or VHHs, also widely known as nanobodies since their discovery and discuss the advantages and disadvantages of these unique molecules in various areas of research, industry, and medicine. Commercialization of camelid sdAbs exploded in 2001 with a flurry of patents issued to the Vrije Universiteit Brussel (VUB and later taken on by the Vlaams Interuniversitair Instituut voor Biotechnologie (VIB and, after 2002, the VIB-founded spin-off company, Ablynx. While entrepreneurial spirit has certainly catalyzed the exploration of nanobodies as marketable products, IP restrictions may be partially responsible for the relatively long time span between the discovery of these biomolecules and their entry into the pharmaceutical market. It is now anticipated that the first VHH-based antibody drug, Caplacizumab, a bivalent anti-vWF antibody for treating rare blood clotting disorders, may be approved and commercialized in 2018 or shortly thereafter. This elusive first approval, along with the expiry of key patents, may substantially alter the scientific and biomedical landscape surrounding camelid sdAbs and pave the way for their emergence as mainstream biotherapeutics.

The emission lines are attributed to free helium atoms and molecules desorbing from clusters in electronically excited states. With increasing cluster size the intensity of molecular transitions becomes dominant.     The refolding mechanism of the homodimeric two-domain 3-isopropylmalate dehydrogenase (IPMDH) from the organisms adapted to different temperatures, Thermus thermophilus (Tt), Escherichia coli (Ec), and Vibrio sp. I5 (Vib), is described. In all three cases, instead of a self-template mechanism, the high extent of symmetry and cooperativity in folding of subunits and domains have been concluded from the following experimental findings: The complex time course of refolding, monitored by Trp fluorescence, consists of a fast (the rate constant varies as 16.5, 25.0, and 11.7 min-1 in the order of Tt, Ec, and Vib IPMDHs) and a slow (the rate constants are 0.11, 0.80, and 0.23 min-1 for the three different species) first-order process. However, a burst increase of Trp fluorescence anisotropy to the value of the native states indicates that in all three cases the association of the two polypeptide chains occurs at the beginning of refolding. This dimeric species binds the substrate IPM, but the native-like interactions of the tertiary and quaternary structures are only formed during the slow phase of refolding, accompanied by further increase of protein fluorescence and appearance of FRET between Trp side chain(s) and the bound NADH. Joining the contacting arms of each subunit also takes place exclusively during this slow phase. To monitor refolding of each domain within the intact molecule of T. thermophilus IPMDH, Trp's (located in separate domains) were systematically replaced with Phe's. The refolding processes of the mutants were followed by measuring changes in Trp fluorescence and in FRET between the particular Trp and NADH. The high similarity of time courses (both in biphasicity and in their rates) strongly suggests cooperative folding of the domains during formation of the native three-dimensional structure of IPMDH.

  3. Evolution and functional insights of different ancestral orthologous clades of chitin synthase genes in the fungal tree of life

    Full Text Available Chitin synthases (CHSs are key enzymes in the biosynthesis of chitin, an important structural component of fungal cell walls that can trigger innate immune responses in host plants and animals. Members of CHS gene family perform various functions in fungal cellular processes. Previous studies focused primarily on classifying diverse CHSs into different classes, regardless of their functional diversification, or on characterizing their functions in individual fungal species. A complete and systematic comparative analysis of CHS genes based on their orthologous relationships will be valuable for elucidating the evolution and functions of different CHS genes in fungi. Here, we identified and compared members of the CHS gene family across the fungal tree of life, including 18 divergent fungal lineages. Phylogenetic analysis revealed that the fungal CHS gene family is comprised of at least 10 ancestral orthologous clades, which have undergone multiple independent duplications and losses in different fungal lineages during evolution. Interestingly, one of these CHS clades (class III was expanded in plant or animal pathogenic fungi belonging to different fungal lineages. Two clades (classes VIb and VIc identified for the first time in this study occurred mainly in plant pathogenic fungi from Sordariomycetes and Dothideomycetes. Moreover, members of classes III and VIb were specifically up-regulated during plant infection, suggesting important roles in pathogenesis. In addition, CHS-associated networks conserved among plant pathogenic fungi are involved in various biological processes, including sexual reproduction and plant infection. We also identified specificity-determining sites, many of which are located at or adjacent to important structural and functional sites that are potentially responsible for functional divergence of different CHS classes. Em condições de intensidade de precipitação abaixo da velocidade básica do solo (VIB o microrrelevo superficial comportou-se de maneira estável, apresentando uma superfície espelhada crescente (acúmulo de água sobre o solo, ao longo dos primeiros 120 min de ensaio e, em condições de intensidade de precipitação acima da velocidade básica (VIB observou-se que a superfície espelhada estabilizava-se em menos de 30 min, apresentando, posteriormente, a formação de canais preferenciais de escoamento de água sobre o solo (erosão laminar.This paper aims at the development of a new methodology for quantification of water over a soil surface, as a function of the precipitation intensity from a sprinkler irrigation system. The research was carried out at the experimental station belonging to Rural Engineering Department of the University of São Paulo (ESALQ, on a soil often used for traditional agricultural practices. The results obtained showed the existence of a relationship between the precipitation intensity over the area and the microrelief of the soil. Under conditions of low intensity, the microrelief stayed stable throughout the formation of puddles on the soil surface. Under conditions of high intensity, runoff was observed over the area, with the formation of small preferential water flow channels among the puddles, starting the laminar erosion process of the soil.

    Coolant temperature coefficients for the Mark 22 charge published previously are non-conservative because of the neglect of a significant mechanism which has a positive contribution to reactivity. Even after correcting for this effect, dynamic tests made on a Mark VIB charge in the early 60's suggest the results are still non-conservative. This memorandum takes both of these sources of information into account in making a best estimate of the prompt (coolant plus metal) temperature coefficient. Although no safety issues arise from this work (the overall temperature coefficient still strongly contributes to reactor stability), it is obviously desirable to use best estimates for prompt coefficients in limits and other calculations

    Here we describe NanoPack, a set of tools developed for visualization and processing of long read sequencing data from Oxford Nanopore Technologies and Pacific Biosciences. The NanoPack tools are written in Python3 and released under the GNU GPL3.0 License.   7. Prediction of two-dimensional diluted magnetic semiconductors: Doped monolayer MoS2 systems

    Using first-principles calculations, we propose a two-dimensional diluted magnetic semiconductor: monolayer MoS2 doped by transition metals. Doping of transition metal atoms from the IIIB to VIB groups results in nonmagnetic states, since the number of valence electrons is smaller or equal to that of Mo. Doping of atoms from the VIIB to IIB groups becomes energetically less and less favorable. Magnetism is observed for Mn, Fe, Co, Zn, Cd, and Hg doping, while for the other dopants from these groups it is suppressed by Jahn-Teller distortions. Analysis of the binding energies and magnetic properties indicates that (Mo,X)S2 (X=Mn, Fe, Co, and Zn) are promising systems to explore two-dimensional diluted magnetic semiconductors.

Se-II ' and S-II were found to be isostructural and to belong to the tetragonal space group I4 1 /acd, which is made up of 16 atoms in the unit cell. The structure consisted of unique spiral chains with both 4 1 and 4 3 screws. The results confirmed that the structure sequence of the pressure-induced phase transitions for the group VIb elements depended on the initial molecular form.     Insertion reactions are some of the most common and important reactions in organometallic chemistry. Aside from being mechanistically interesting, they have proven useful in organic synthesis and have been postulated in catalytic cycles. A subset of the broad group of insertion reactions is the insertion of carbon monoxides of group VIB metal carbonyl complexes into early-transition-metal alkyl, aryl and hydride bonds. Although the coupling of metal alkylidenes with metal carbonyl complexes is rare in monometallic systems, such coupling is frequently postulated in heterogeneous Fischer-Tropsch systems to yield surface-bound ketene fragments. The formation of bound ketene in bimetallic and cluster complexes has been documented. Herein the authors report the coupling of titanocene methylidene with Cr(CO)/sub 6/ to yield a titanocene ketene complex.

    In view of the inability of the present theory of superconductivity to make reliable predictions for the magnitude of Tsub(c) it seems useful to search for empirical relationships between the composition of the compound and the Tsub(c) value. Table I gives a list of the available Tsub(c) data for transition metals (TM) having from 3 to 9 outer electrons and Tsub(c) data for non-transition elements (NTE) of groups IIB, IIIB and IVB, including data for amorphous (Am) structures and structures (marked by triangles) obtained at high pressures. The analogous metals have therefore the same structure. In Tables II to IV the Tsub(c) data are presented for analogous compounds of NTE from IB - VIB group. Conclusions are presented. (author)

    The synthesis, characterization, and some analytical applications of a new triazine resin are described. Separation of group IB, IIB, VIB, and VIIB metal ions from group VIII metal ions is achieved by this PDT-4 resin. Calcium(II) and magnesium(II) are taken up at pH = 6, 0.1 M acetate and are eluted at pH = 6, 0.1 M sodium nitrate. Copper(II) is retained at pH = 6, 0.1 M acetate and pH = 1 hydrochloric acid and is eluted subsequently by 5 M perchloric acid. Molybdenum(VI) is sorbed selectively from 0.1 N sulfuric acid or hydrochloric acid and is eluted in a tight band by 0.1 N sodium hydroxide. Numerous rapid column chromatographic separations are reported using this new resin, including analysis of NBS standard samples

    The history of separation of metal complexes by gas chromatography is reviewed. The greatest success has been achieved with metal halides or metal β-diketonates, particularly with chelates of the small elements, Be and Al, and the extremely inert chromium diketonates. Many problems still remain concerning the development of suitable chromatographic techniques for many chelates; however, the technique of synergic solvent extraction, mixed-ligand complex chromatography, the synthesis of new heteroatomic ligands, and the advent of new liquid phases are discussed as means to increase the number of unique techniques for separation and analysis of metals. Methods are suggested and references are cited for chromatographic analysis of elements in the following groups: alkali metals, alkaline earths, Group IIIA, IVB, VB, VIB, VIIB, VIII, IB, and IIB.     Newcastle disease (ND), caused by virulent strains of avian paramyxovirus type 1 (APMV-1), is considered throughout the world as one of the most important animal diseases. For over three decades now, there has been a continuing panzootic caused by a variant virulent APMV-1 strain, so-called pigeon paramyxovirus type 1 (PPMV-1), primarily in racing pigeons, which has also spread to wild birds and poultry. PPMV-1 isolations have been made in Great Britain every year since 1983. In this study, we have completed a comparative phylogenetic analysis based on a 374 nucleotide section of the fusion protein gene of 63 isolates of PPMV-1 that were isolated over a 26-year period; 43 of these were sequenced for this study. Phylogenetic analysis of these sequences revealed that all were closely related and placed in the genetic sublineage 4b (VIb), subdivision 4biif. © 2012 Crown copyright.

    Eurasian collared doves (Streptopelia decaocto) have expanded their range across the United States since their introduction several decades ago. Recent mortality events in Eurasian collared doves in Arizona and Montana, USA, during the winter of 2009-2010 were the result of pigeon paramyxovirus (PPMV), a novel disease agent. The first instance of mortality by this emerging infectious disease in this species occurred in Florida in 2001 with subsequent disease events in 2006 and 2008. Full diagnostic necropsies were performed on carcasses from the three states. PPMV was identified by RT-PCR and virus isolation and was sequenced to the VIb genotype of avian paramyxovirus-1 (APMV). Other APMVs are common in a variety of free-ranging birds, but concern is warranted because of the potential for commingling of this species with native birds, virus evolution, and threats to domestic poultry. Improved surveillance for wildlife mortality events and efforts to prevent introduction of non-native animals could reduce the threat of introducing new pathogens.

  17. Root finding in the complex plane for seismo-acoustic propagation scenarios with Green's function solutions.

    A normal mode solution to the ocean acoustic problem of the Pekeris waveguide with an elastic bottom using a Green's function formulation for a compressional wave point source is considered. Analytic solutions to these types of waveguide propagation problems are strongly dependent on the eigenvalues of the problem; these eigenvalues represent horizontal wavenumbers, corresponding to propagating modes of energy. The eigenvalues arise as singularities in the inverse Hankel transform integral and are specified by roots to a characteristic equation. These roots manifest themselves as poles in the inverse transform integral and can be both subtle and difficult to determine. Following methods previously developed [S. Ivansson et al., J. Sound Vib. 161 (1993)], a root finding routine has been implemented using the argument principle. Using the roots to the characteristic equation in the Green's function formulation, full-field solutions are calculated for scenarios where an acoustic source lies in either the water column or elastic half space. Solutions are benchmarked against laboratory data and existing numerical solutions.

    A normal coordinate analysis has been carried out on four compounds having a similar ring structure with different side chain substitutions, which are xanthine, caffeine, theophylline, and theobromine. Xanthine is chemically known as 2,6-dihydroxy purine. Caffeine, theophylline and theobromine are methylated xanthines. Considering the methyl groups as point mass, the number of normal modes of vibrations can be distributed as Γ vib=27 A'+12 A″ based on C s point group symmetry associated with the structures. In the present work 15 A' and 12 A″ normal modes are considered. A new set of orthonormal symmetry co-ordinates have been constructed. Wilson's F- G matrix method has been adopted for the normal coordinate analysis. A satisfactory vibrational band assignment has been made by employing the FTIR and FT Raman spectra of the compounds. The potential energy distribution is calculated with the arrived values of the force constants and hence the agreement of the frequency assignment has been checked.

    Full Text Available O objetivo deste estudo é contribuir para o conhecimento que auxilie o profissional de enfermagem na identificação dos tipos mais comuns de formações venosas da região da fossa cubital e, ainda, enfocar a importância de estar sempre atento aos casos pouco comuns como o aqui relatado. Através de uma revisão bibliográfica, constatamos que as formações venosas dessa região podem ser classificadas em 5 tipos mais comuns, sendo o tipo II o mais frequente. Constatamos ainda, que a VICo é o local de punção mais indicado, seguido pela VIB. Descrevemos também uma variação anatômica, onde observamos ausência de comunicação entre VC e VB no nível da fossa cubital e VIA drenando na VB, estando presente a VCA.

    o-carboranylmethyl ethers of steroids were synthesized by insertion of steroidal 2-propynyloxy derivatives into 6,9-bis(acetonitrile)decarborane. This reaction provided compounds with an estrane and androstane skeleton, potentially useful in boron neutron capture therapy of hormone-sensitive forms of cancer: 17β-o-carboranylmethyl ether of estradiol IXb (yield 14%) and 3β- and 17β-carboranylmethyl ethers of androstenediol Vb and VIIb (yield 12% and 13%, respectively). Jones oxidation gave the carboranyl derivative of androsten-17-one VIb in 75% yield. As shown by a study of insertion of 3β-(2-propynyloxy)cholest-5-ene (IVa), the low yields of the insertion reaction cannot be increased by change in the reaction conditions. The relative binding affinity of compound IXb to the estrogen receptor from rat uterine and human breast tumor cytosol was 3.0 and 0.29%, respectively, of that of estradiol.     An electrode having increased output with slower degradation is formed of a film applied to a beta-alumina solid electrolyte (BASE). The film comprises a refractory first metal M.sup.1 such as a platinum group metal, suitably platinum or rhodium, capable of forming a liquid or a strong surface adsorption phase with sodium at the operating temperature of an alkali metal thermoelectric converter (AMTEC) and a second refractory metal insoluble in sodium or the NaM.sup.1 liquid phase such as a Group IVB, VB or VIB metal, suitably tungsten, molybdenum, tantalum or niobium. The liquid phase or surface film provides fast transport through the electrode while the insoluble refractory metal provides a structural matrix for the electrode during operation. A trilayer structure that is stable and not subject to deadhesion comprises a first, thin layer of tungsten, an intermediate co-deposited layer of tungsten-platinum and a thin surface layer of platinum.

    Meta-heuristic multi-response optimization methods are widely in use to solve multi-objective problems to obtain Pareto optimal solutions during optimization. This work focuses on optimal multi-response evaluation of process parameters in generating responses like surface roughness (Ra), surface hardness (H) and tool vibration displacement amplitude (Vib) while performing operations like tangential and orthogonal turn-mill processes on A-axis Computer Numerical Control vertical milling center. Process parameters like tool speed, feed rate and depth of cut are considered as process parameters machined over brass material under dry condition with high speed steel end milling cutters using Taguchi design of experiments (DOE). Meta-heuristic like Dragonfly algorithm is used to optimize the multi-objectives like ‘Ra’, ‘H’ and ‘Vib’ to identify the optimal multi-response process parameters combination. Later, the results thus obtained from multi-objective dragonfly algorithm (MODA) are compared with another multi-response optimization technique Viz. Grey relational analysis (GRA).

Western blot analysis, using antibodies specific to Drosophila CcO subunits IV, Va, Vb, VIb, VIc, VIIc, and VIII, indicated that the abundance these polypeptides decreased, ranging from 11% to 40%, during aging. These and previous results suggest that CcO is a specific intra-mitochondrial site of age-related deterioration, which may have a broad impact on mitochondrial physiology.

    Full Text Available This paper shows the results of a study conducted on five different categories of vehicles in a specific test site. The aim was to investigate how the effect of the test site discontinuity determines variations of comfort related to the increase in speed and to the five selected road vehicles of different classes. Measurements were obtained by combining data relating to vibrations in the three reference axes, detected through a vibration dosimeter (VIB-008, and geolocation data (latitude, longitude, and speed identified by the GPS inside a smartphone. This procedure, through the synchronization between dosimeter and GPS location, has been helpful in postprocessing to eliminate any measurement anomalies generated by the operator. After the survey campaign it was determined that a formulation allows defining a Comfort Index (CI depending on velocity and five vehicles of different classes. This study showed that the presence of speed bumps, in the test site investigated, appears to be uncomfortable even at speeds well below those required by the Highway Code.

    The basic material of the above-mentioned layer consists of pure aluminium oxide or essentially aluminium oxide. To improve this protective layer metal oxides from the groups IIA, IIIA, IIIB, VB, VIB, VIIB or VIII of the periodic system are added to its basic material before the said protective coating is applied. In this way a corundum structure is formed in the case of aluminium oxide. Gallium oxide, vanadium oxide, chromium oxide or iron oxide are particularly suited for the correlation of such a corundum structure. The formation of the corundum structure increases the resistance of the protective coating to the corrosive effects of vanadium pentoxide and sodium sulfate. By the addition of a specific quantity of magnesium oxide it is possible not only to stimulate the formation of corundum but also to reduce the increase in grain size in the case of the aluminium oxide. The other metallic oxides are especially favorable to the formation of the corundum structure, so that preferably magnesium oxide is to be added to these metallic oxides in order to reduce the increase in grain size.     Full Text Available In the Celtic Oppidum near Čarnok (Bačka, Vojvodina, Serbia remains of four mud-plastered granarybaskets were discovered. They are all dated to the period Gomolava VI-B (first half of the 1st century B.C.. Hulled barley and broomcorn millet were stored in the granary-baskets. The mesh of the granary-baskets was composed of young branches of oak tree (Quercus, common English elm tree (Ulmus cf. campestris L., poplar / willow (Populus / Salix, purgin buckthorn (Rhamnus cf. cathartica L., wayfaring tree (Viburnum cf. lantana L., spindle tree (Euonymus cf. europaeus L., barberry (Berberis vulgaris L. and a Pomoidae-tree. At least four different tree taxa were used for basket making. Construction of two-thousand-year old mud-plastered granary-baskets from the Pannonian plain is very similar to that of traditional granaries in some present-day villages in Africa. Additionally, one non-plastered basket was also discovered at Oppidum. It was build out of maple (Acer, barberry, buckthorn (Rhamnus and branches of a pomaceous fruits tree (Pomoidae. .

  10. Promotion of hydrogen entry into iron from NaOH solution by iron-oxygen species

    This study was carried out to explain reasons of the enhanced hydrogen entry into iron at low polarisations. Hydrogen permeation rate (HPR) through a 35-μm thick iron membrane was studied with the electrochemical technique in 0.1 M NaOH at 25 o C. A rotating split-ring disk electrode was used to detect soluble Fe(II) species. Enhanced hydrogen entry (HPR peaks) was observed at low cathodic and low anodic polarisations during voltammetric cycling, and also during galvanostatic anodic polarisation applied after cathodic charging. HPR peaks occurred at potentials from about -1.2 to -0.9 V (NHE) which were more cathodic than the potentials of thermodynamic stability of Fe(OH) 2 or Fe 3 O 4 , and of the formation of soluble Fe(II) species. It has been suggested that the enhanced hydrogen entry is associated with the presence of FeOH ad . In this species oxygen is bound with hydrogen (oxo-hydride), hence it can be supposed that the mechanism of its promoting effect can be similar to that of hydrides of other elements of the VIb group

The solution of the problem in the presence of wake is obtained by using Greens' function method, Fourier transform, the Wiener-Hopf technique and the modified stationary phase method. Expressions for the total far field for the trailing edge (wake present) situation are given. It is observed that the field produced by the Kutta-Joukowski condition will be substantially in excess of the field when this condition is ignored. Finally, a simple procedure is devised to calculate the inverse temporal Fourier transform. The solution for the leading edge situation can be obtained if the wake, and consequently a Kutta-Joukowski edge condition, is ignored. This can also be seen from the numerical results.

    Osmotic swelling of NIH3T3 mouse fibroblasts activates a bromoenol lactone (BEL)-sensitive taurine efflux, pointing to the involvement of a Ca2+-independent phospholipase A2 (iPLA2) (Lambert IH. J Membr Biol 192: 19-32, 2003). We report that taurine efflux from NIH3T3 cells was not only increased...... by cell swelling but also decreased by cell shrinkage. Arachidonic acid release to the cell exterior was similarly decreased by shrinkage yet not detectably increased by swelling. NIH3T3 cells were found to express cytosolic calcium-dependent cPLA2-IVA, cPLA2-IVB, cPLA2-IVC, iPLA2-VIA, iPLA2-VIB......, and secretory sPLA2-V.   15. Trajectory study of supercollision relaxation in highly vibrationally excited pyrazine and CO2.

    Classical trajectory calculations were performed to simulate state-resolved energy transfer experiments of highly vibrationally excited pyrazine (E(vib) = 37,900 cm(-1)) and CO(2), which were conducted using a high-resolution transient infrared absorption spectrometer. The goal here is to use classical trajectories to simulate the supercollision energy transfer pathway wherein large amounts of energy are transferred in single collisions in order to compare with experimental results. In the trajectory calculations, Newton's laws of motion are used for the molecular motion, isolated molecules are treated as collections of harmonic oscillators, and intermolecular potentials are formed by pairwise Lennard-Jones potentials. The calculations qualitatively reproduce the observed energy partitioning in the scattered CO(2) molecules and show that the relative partitioning between bath rotation and translation is dependent on the moment of inertia of the bath molecule. The simulations show that the low-frequency modes of the vibrationally excited pyrazine contribute most to the strong collisions. The majority of collisions lead to small DeltaE values and primarily involve single encounters between the energy donor and acceptor. The large DeltaE exchanges result from both single impulsive encounters and chattering collisions that involve multiple encounters.

    First principles calculations are used to study the preferential occupation of ternary alloying additions into the binary Ti-Al phases, namely, γ-TiAl, α{sub 2}-Ti{sub 3}Al, β{sub o}-TiAl, and B19-TiAl. While the early transition metals (TMs, group IVB, VB, and VIB elements) prefer to substitute for Ti atoms in the γ-, α{sub 2}-, and B19-phases, they preferentially occupy Al sites in the β{sub o}-TiAl. Si is, in this context, an anomaly, as it prefers to sit on the Al sublattice for all four phases. B and C are shown to prefer octahedral Ti-rich interstitial positions instead of substitutional incorporation. The site preference energy is linked with the alloying-induced changes of energy of formation, hence alloying-related (de)stabilisation of the phases. We further show that the phase-stabilisation effect of early TMs on β{sub o}-phase has a different origin depending on their valency. Finally, an extensive comparison of our predictions with available theoretical and experimental data (which is, however, limited mostly to the γ-phase) shows a consistent picture.

    Full Text Available As práticas alimentares da população brasileira vêm se alterando nas últimas décadas e a publicidade é um dos fatores que contribuem para esta situação. O objetivo deste trabalho foi avaliar o conteúdo das propagandas de alimentos veiculadas na televisão e dirigidas a crianças, sob a ótica da regulação. As propagandas foram gravadas em fitas VHS de duas grandes emissoras de televisão, no período de férias escolares. Foram utilizadas técnicas de análise de conteúdo e definidas oito categorias de análise a partir do referencial teórico sobre as práticas alimentares e seus determinantes. Para subsidiar a análise utilizou-se a proposta de Regulamento Técnico da Anvisa. Foram contabilizadas 1018 propagandas, das quais foram selecionadas as 132 (12,9% que anunciavam alimentos dirigidos para as crianças, sendo identificados somente 12 produtos diferentes. De acordo com o Regulamento proposto, todas as propagandas analisadas infringiam pelo menos três artigos, entre os quais se destaca o 4º, Incisos III e VIb. É nítida a urgência do setor público de regular o conteúdo das propagandas de alimentos infantis, cujo consumo pode ser prejudicial à saúde devido à influência que exercem na decisão pela compra, tanto por parte das próprias crianças, quanto dos pais.The eating habits of the Brazilian population have been changing in recent decades and publicity is one of the factors contributing to this situation.     A complex of special properties of materials and a threshold of their working temperatures which is frequently limited 500-550°C define the level of modern technological development and, first of all, for nuclear-industrial plants. The alloys on the basis of iron are the most widespread kind of constructional materials, and therefore an increase of their special properties (high-temperature strength, corrosion stability, durability and other characteristics) is of great value. The ionic beams with a low energy (300-1,500 eV) which can change the structure and the properties of a target, in particular iron and its alloys were used for the solution of these problems in the present work. In this work theoretical and experimental results of the research of the process of impact interaction of the ionic beams with iron are also given. The properties of a iron surface with the help of modern methods of physical and chemical analyse are investigated. The opportunities to produce new surface properties of iron using the ionic beams are shown. On the basis of the results obtained the supplementary work directions are analyzed.

  1. Impact of Alloying on Stacking Fault Energies in γ-TiAl

    Full Text Available Microstructure and mechanical properties are key parameters influencing the performance of structural multi-phase alloys such as those based on intermetallic TiAl compounds. There, the main constituent, a γ -TiAl phase, is derived from a face-centered cubic structure. Consequently, the dissociation of dislocations and generation of stacking faults (SFs are important factors contributing to the overall deformation behavior, as well as mechanical properties, such as tensile/creep strength and, most importantly, fracture elongation below the brittle-to-ductile transition temperature. In this work, SFs on the { 111 plane in γ -TiAl are revisited by means of ab initio calculations, finding their energies in agreement with previous reports. Subsequently, stacking fault energies are evaluated for eight ternary additions, namely group IVB–VIB elements, together with Ti off-stoichiometry. It is found that the energies of superlattice intrinsic SFs, anti-phase boundaries (APBs, as well as complex SFs decrease by 20–40% with respect to values in stoichiometric γ -TiAl once an alloying element X is present in the fault plane having thus a composition of Ti-50Al-12.5X. In addition, Mo, Ti and V stabilize the APB on the (111 plane, which is intrinsically unstable at 0 K in stoichiometric γ -TiAl.

  2. Potential virulence factors of bacteria associated with tail fan necrosis in the spiny lobster, Jasus edwardsii.

    Tail fan necrosis (TFN) is a common condition found in commercially exploited spiny lobsters that greatly diminishes their commercial value. Bacteria possessing proteolytic, chitinolytic and lipolytic capabilities were associated with TFN in spiny lobsters, Jasus edwardsii. In this study, 69 bacterial isolates exhibiting all the three enzymatic capabilities from the haemolymph and tail fans of J. edwardsii with and without TFN were further characterized and compared, including morphology, biofilm formation, antimicrobial activity, antimicrobial resistance, and production of siderophores, melanin and ammonia. The genomic patterns of the most common Vibrio crassostreae isolates were also compared between TFN-affected and unaffected lobsters. Biofilm formation was stronger in bacterial isolates from both haemolymph and tail fans of TFN-affected lobsters compared to those from the unaffected lobsters, while melanin production and siderophore production were stronger in the isolates from tail fans of lobsters with TFN. By contrast, the other characteristics of isolates were similar in lobsters with and without TFN. The Vib. crassostreae isolates from the affected lobsters had similar genomic patterns. Overall, the results indicate that in addition to proteolytic, chitinolytic and lipolytic activities, the bacteria associated with TFN commonly have enhanced activity of important virulence factors, including biofilm formation, melanin production and siderophore production. © 2018 John Wiley & Sons Ltd.

    Based on the complete genome sequence of pigeon-origin Newcastle disease virus strain JS/07/04/ Pi(genotype VIb), nine overlapped fragments covering its full-length genome were amplified by RT-PCR. The fragments were connected sequentially and then inserted into the transcription vector TVT7/R resulting in the TVT/071204 which contained the full genome of strain JS/07/04/Pi. The TVT/071204 was co-transfected with three helper plasmids pCI-NP, pCI-P and pCI-L into the BSR cells, and the transfected cells and culture supernatant were inoculated into 9-day-old SPF embryonated eggs 60 h post-transfection. The HA and HI tests were conducted following the death of embryonated eggs. The results showed that the allantoic fluids obtained were HA positive and the HA could be inhibited by anti-NDV serum which indicated that the strain JS/07/04/Pi was rescued successfully. The rescued virus rNDV/071204 showed similar growth kinetics to its parental virus in CEF. The successful recovery of this strain would contribute to the understanding of the host-specificity of pigeon-origin NDV and to the development of the novel vaccines against the NDV infection in pigeons.

  4. Expression of a serine protease (motopsin PRSS12) mRNA in the mouse brain: in situ hybridization histochemical study.

    Serine proteases are considered to play several important roles in the brain. In an attempt to find novel brain-specific serine proteases (BSSPs), motopsin (PRSS-12) was cloned from a mouse brain cDNA library by polymerase chain reaction (PCR). Northern blot analysis demonstrated that the postnatal 10-day mouse brain contained the most amount of motopsin mRNA. At this developmental stage, in situ hybridization histochemistry showed that motopsin mRNA was specifically expressed in the following regions: cerebral cortical layers II/III, V and VIb, endopiriform cortex and the limbic system, particularly in the CA1 region of the hippocampal formation. In addition, in the brainstem, the oculomotor nucleus, trochlear nucleus, mecencephalic and motor nuclei of trigeminal nerve (N), abducens nucleus, facial nucleus, nucleus of the raphe pontis, dorsoral motor nucleus of vagal N, hypoglossal nucleus and ambiguus nucleus showed motopsin mRNA expression. Expression was also found in the anterior horn of the spinal cord. The above findings strongly suggest that neurons in almost all motor nuclei, particularly in the brainstem and spinal cord, express motopsin mRNA, and that motopsin seems to have a close relation to the functional role of efferent neurons.     The synthesis and crystal chemistry of Np, Pu, Am and Cm transuranium element chalcogenides are described. From plutonium, transuranium element chalcogenides exhibit the same crystal structure as their rare-earth homologues. The variations of the lattice constants of these compounds in terms of the atomic number are characterized by the lack of the 5f contraction and are interpreted by a localization of the 5f electrons depending upon the considered transuranium element, the nature of the ligand and the crystal structure. To compare the degree of magnitude of the 5f electron delocalization in various compounds, a delocalization scale is proposed based on a comparison between the molar volumes of actinide and isostructural lanthanide compounds. This scale provides a delocalization coefficient for each compound under study. Examination of these coefficients shows that the 5f electrons, in series of actinide compounds, become localized when going from neptunium to curium and that the delocalization process does not only depend upon overlaps between 5f-6d orbitals of neighbouring actinide atoms; the delocalization coefficients show the existence of a secondary delocalization effect due to overlaps between the p anion and f actinide orbitals which are more important for the Vb anion group (N, P, As, Sb) than for the Vib one (S,Se,Te) [fr

  6. Spectroanalytical investigations on inductively coupled N2/Ar and Ar/Ar high frequency plasmas

    In order to improve the detection limits of trace elements in corrosion products of metallic materials, the inductively coupled plasma excitation source (ICP) was applied for spectroscopic analysis. Besides optimizing the working conditions for the mentioned materials, the fundamental research clearing the excitation processes in ICP was carried out. Basicly, two plasma systems were investigated: the nitrogen cooled N 2 /Ar- and pure Ar/Ar-plasma. The computed detection limits for 8 chosen elements are between 0.1 and 50 μg ml -1 in both plasmas. The advantage of ion lines was clearly present; in N 2 /Ar-plasma it was larger than in Ar/Ar-plasma. The excitation temperatures measured with help of ArI, FeI and ZnI lines rise with increasing power and decreasing distance from the induction coil. The distribution of Zn excitation temperature in N 2 /Ar-plasma as well as the measured N + 2 rotational and CN vibrational temperatures indicate, that the toroidal structure of Ar/Ar-plasma is not analogue to the N 2 /Ar-plasma. The values of the various excitation temperatures (Ar, Fe, Zn) and the differences between the excitation, vibration, rotation and ionization temperatures (Tsub(i) > Tsub(n) = Tsub(vib) > Tsub(rot)) indicate an absence of thermal equilibrium in the concerned system.     Predictive computation of the nonlinear dynamical responses of gap-supported tubes subjected to flow excitation has been the subject of very active research. Nevertheless, experimental results are still very important, for validation of the theoretical predictions as well as for asserting the integrity of field components. Because carefully instrumented test tubes and tube-supports are seldom possible, due to space limitations and to the severe environment conditions, there is a need for robust techniques capable of extracting, from the actual vibratory response data, information that is relevant for asserting the components integrity. The dynamical contact/impact (vibro-impact) forces are of paramount significance, as are the tube/support gaps. Following our previous studies in this area using wave-propagation techniques (De Araujo, Antunes, and Piteau, 1998, 'Remote Identification of Impact Forces on Loosely Supported Tubes: Part 1-Basic Theory and Experiments', J. Sound Vib., 215, pp. 1015-1041; Antunes, Paulino, and Piteau, 1998, 'Remote Identification of Impact Forces on Loosely Supported Tubes: Part 2-Complex Vibro-Impact Motions', J.   8. Phylogenetic analysis of some Newcastle disease virus isolates from the Sudan

    Full Text Available A reverse transcription-polymerase chain reaction (RT-PCR was used to amplify 1412 bp of the fusion protein gene (F gene of four Newcastle disease virus (NDV isolates; two velogenic (TY-1/90 and DIK-90 and two lentogenic isolates (Dongla 88/1 and GD.S.1. Following sequencing, nucleotide sequences were annotated and 894 bp were compared phylogenetically with those from strains previously reported in the Sudan and the virus strains published on the GenBank. It could be demonstrated that TY-1/90 and DIK-90 strains belong to the genotype VI of NDV and are in close genetic relationship to sub- genotype VIb. TY-1/90 and DIK-90 strains were observed to be genetically unrelated to the earlier Sudanese isolates of 1970/80s and the late of 2000s suggesting a different origin. The close genetic relationship to the European and African pigeon paramyxovirus type 1 (PPMV-1 suggests a common ancestor. Dongola, GD.S.1 strains were classified into genotype II that comprises non-pathogenic lentogenic NDV strains. The present genetic classification of NDV isolates of the Sudan provides valuable information on genotypes of NDV. Further molecular epidemiological investigations of the recent outbreaks of Newcastle disease in the Sudan are needed in order to improve the efficiency of control strategies and vaccine development.

    Recent developments in Optical Coherence Tomography (OCT) allow measurements of cochlear motions through the bony cochlear wall without holes at spatial resolutions approaching about 10 µm. Measurements to date have been made with custom OCT systems with long development times. We present measurements made with a commercial OCT system driven by custom software (VibOCT) that facilitates near real-time frequency response measurements. The 905-nm wavelength laser and high-speed (100 kHz) camera provide higher axial resolution (3 µm in air) and temporal resolution than previous studies and a sub-nanometer noise floor in air. We gathered anatomical images of the gerbil cochlear apex in vivo at higher resolution than available previously, sufficient to resolve individual outer hair cells, pillar cells, tunnel of Corti and inner sulcus regions. Images from the 3rd apical turn show a bulging of Reissners membrane in vivo that flattened post-mortem with a concomitant reduction in the distance between the Henson cell border and the stria vascularis wall. Vibrometry of the organ of Corti shows a low-pass characteristic in-vivo and post-mortem with a traveling wave-like phase delay similar to a recent study rather than the sharp tuning seen more basally. This system can provide valuable information on cochlear function, which is also useful for the development of detailed cochlear models of the passive and active gerbil apex.

    Low-intensity laser irradiation (LILI) has been shown to stimulate cellular functions leading to increased adenosine triphosphate (ATP) synthesis. This study was undertaken to evaluate the effect of LILI on genes involved in the mitochondrial electron transport chain (ETC, complexes I-IV) and oxidative phosphorylation (ATP synthase). Four human skin fibroblast cell models were used in this study: normal non-irradiated cells were used as controls while wounded, diabetic wounded, and ischemic cells were irradiated. Cells were irradiated with a 660 nm diode laser with a fluence of 5 J/cm(2) and gene expression determined by quantitative real-time reverse transcription (RT) polymerase chain reaction (PCR). LILI upregulated cytochrome c oxidase subunit VIb polypeptide 2 (COX6B2), cytochrome c oxidase subunit VIc (COX6C), and pyrophosphatase (inorganic) 1 (PPA1) in diabetic wounded cells; COX6C, ATP synthase, H+transporting, mitochondrial Fo complex, subunit B1 (ATP5F1), nicotinamide adenine dinucleotide (NADH) dehydrogenase (ubiquinone) 1 alpha subcomplex, 11 (NDUFA11), and NADH dehydrogenase (ubiquinone) Fe-S protein 7 (NDUFS7) in wounded cells; and ATPase, H+/K+ exchanging, beta polypeptide (ATP4B), and ATP synthase, H+ transporting, mitochondrial Fo complex, subunit C2 (subunit 9) (ATP5G2) in ischemic cells. LILI at 660 nm stimulates the upregulation of genes coding for subunits of enzymes involved in complexes I and IV and ATP synthase.

    During recent years, transition metal dichalcogenides of groups IVB, VB and VIB have received considerable attention because of the great diversity in their transport properties. 2H-WSe 2 (Tungsten diselenide) is an interesting member of the transition metal dichalcogenide (TMDC's) family and known to be a semiconductor useful for photovoltaic and optoelectronic applications. The anisotropy usually observed in this diamagnetic semiconductor material is a result of the sandwich structure of Se-W-Se layers interacting with each other, loosely bonded by the weak Van der Waals forces. Recent efforts in studying the influence of the anisotropic electrical and optical properties of this layered-type transition metal dichalcogenides have been implemented by doping the samples with different alkali group elements. Unfortunately, little work is reported on doping of metals in WSe 2. Therefore, it is proposed in this work to carry out a systematic growth of single crystals of WSe 2 by doping it with copper in different proportions i.e. Cu xWSe 2 ( x=0, 0.5, 1.0) by direct vapour transport technique. Transport properties like low and high temperature resistivity measurements, high pressure resistivity, Seebeck coefficient measurements at low temperature and Hall Effect at room temperature were studied in detail on all these samples. These measurements show that tungsten diselenide single crystals are p-type whereas doped with copper makes it n-type in nature. The results obtained and their implications are discussed in this paper.

  12. Determination of bond energies by mass spectrometry. Some transition metal carbonyls

    Two groups of transition metal carbonyls have been studied, M(CO) 6 and M(CO) 5 CS complexes of the Group VIB metals and M 2 (CO) 10 complexes of the Group VIIB metals. Results for the hexacarbonyl complexes indicate that the measured fragmentation energies are in error by 0.25 +- 0.02 eV per CO produced. This is attributed to excitation of CO to the first vibrational state. Least-squares dissociation energies calculated from corrected data for M(CO) 5 CS complexes indicate that the M--CS bond is 3 to 4 times stronger than the M--CO bonds. Substitution of CS for CO in going from M(CO) 6 to M(CO) 5 CS weakens the remaining M--CO bonds by an average of 0.2 eV. Previously unreported MnTc(CO) 10 and TcRe(CO) 10 are prepared by halide substitution of Tc(CO) 5 Br and Re(CO) 5 Br with Mn(CO) 5 - and Tc(CO) 5 - , respectively. In the positive ion, metal and mixed-metal decacarbonyls are considered as (CO) 5 M + --M(CO) 5 complexes possessing five strong and five weak M--CO bonds. For Mn 2 (CO) 10 and Re 2 (CO) 10 , M + --M dissociation energies are 3.0 +- 0.1 and 4.0 +- 0.3 eV, respectively. These energies are 2.5 times greater than those reported for homolytic cleavage to M(CO) 5 + and M(CO) 5

Film elemental composition and phase content is thoroughly characterized with time-of-flight elastic recoil detection analysis (ToF-E ERDA), Rutherford backscattering spectrometry (RBS), and x-ray diffraction.     Full Text Available As a result of the growing demand for highly customized and individual products, companies need to enable flexible and intelligent manufacturing. Cyber-physical production systems (CPPS will act autonomously in the future in an interlinked production and enable such flexibility. However, German mid-sized plant manufacturers rarely use virtual technologies for design and validation in order to design CPPS. The research project Virtual Commissioning with Smart Hybrid Prototyping (VIB-SHP investigated the usage of virtual technologies for manufacturing systems and CPPS design. Aspects of asynchronous communicating, intelligent- and autonomous-acting production equipment in an immersive validation environment, have been investigated. To enable manufacturing system designers to validate CPPS, a software framework for virtual prototyping has been developed. A mechatronic construction kit for production system design integrates discipline-specific models and manages them in a product lifecycle management (PLM solution. With this construction kit manufacturing designers are able to apply virtual technologies and the validation of communication processes with the help of behavior models. The presented approach resolves the sequential design process for the development of mechanical, electrical, and software elements and ensures the consistency of these models. With the help of a bill of material (BOM- and signal-based alignment of the discipline-specific models in an integrated mechatronic product model, the communication of the design status and changes are improved. The re-use of already-specified and -designed modules enable quick behavior modeling, code evaluation, as well as interaction with the virtualized assembly system in an immersive environment.

  16. Potassium, rubidium, strontium, thorium and uranium of the cretaceous plutons in the Kitakami Mountains

    The Cretaceous plutonic rocks in the Kitakami Mountains, 110 to 125 Ma in K-Ar ages on biotite, etc., can be petrographically and petrochemically classified into seven groups; Zones I, II, III, IV, V, VIa and VIb. Some of them are arranged in the east to west direction. The plutons of Zones II and V are mainly felsic. They are of comparatively big dimension reaching 600 square kilometers. The plutons of other zones are composed of felsic and mafic rocks. They occur on a small scale. The plutonic rocks have been chemically analysed for CaO, K 2 O, Rb, Sr, Th, and U. In the mafic rocks of CaO > 8.2 percent, the contents of CaO and Na 2 O are nearly equal in each zone throughout the Kitakami Mountains. K 2 O, Rb, Th, and U contents, however, generally increase westward. Sr also does. In the westernmost zone, Zone IV, the mafic rocks resemble shoshonite concerning their major elements. Some of the plutons, especially in Zones II and V, are zonally divided into marginal and central facies. The two facies are different in their K 2 O, Rb, etc. contents, and K/Rb and other ratios. In the marginal facies K 2 O and Rb are richer and K/Rb is smaller than those in the central facies. This fact suggests the different processes of magmatic differentiation in the two facies. Intrusion of the marginal facies have successively followed by that of the central facies. (author)

  17. The discovery of a highly polarized bipolar nebula

    During a search for the optical counterparts of IRAS sources whose flux peaks at 25 microns, a small faint bipolar nebula was discovered in Monoceros at the position of IRAS 07131-0147. The CCD images display the object's considerable structure. The central star seems relatively free of closeby nebulosity: the two lobes have a bow-tie structure with those parts nearest to the star consisting of series of small knots. The outer parts of the lobes seem to be made up of filaments streaming away from knots. On the basis of its optical spectrum, the central star was classified as a M5-6 giant. In the IRAS color classification scheme of Van der Veen and Habing (1988), the central star is VIb which indicates that there are distinct hot and cold components of circumstellar dust and that the mass loss process may have temporarily abated. Therefore, it is proposed that the object is in the post main sequence stage of evolution and is a protoplanetary nebulae. Young protoplanetary nebulae have totally obscured central stars illuminating reflective lobes whereas older ones such as M2-9 have lobes seen in emission from gas ionized by the central hot star which is clearly visible. Since the central object of IRAS07131-0147 is a relatively unobscured late type star and the lobes are seen only by reflection, it is suggested that this nebula is a protoplanetary nebula in an evolutionary stage intermediate between that of CRL2688 and M2-9

  18. BLSSpeller: exhaustive comparative discovery of conserved cis-regulatory elements.

    The accurate discovery and annotation of regulatory elements remains a challenging problem. The growing number of sequenced genomes creates new opportunities for comparative approaches to motif discovery. Putative binding sites are then considered to be functional if they are conserved in orthologous promoter sequences of multiple related species. Existing methods for comparative motif discovery usually rely on pregenerated multiple sequence alignments, which are difficult to obtain for more diverged species such as plants. As a consequence, misaligned regulatory elements often remain undetected. We present a novel algorithm that supports both alignment-free and alignment-based motif discovery in the promoter sequences of related species. Putative motifs are exhaustively enumerated as words over the IUPAC alphabet and screened for conservation using the branch length score. Additionally, a confidence score is established in a genome-wide fashion. In order to take advantage of a cloud computing infrastructure, the MapReduce programming model is adopted. The method is applied to four monocotyledon plant species and it is shown that high-scoring motifs are significantly enriched for open chromatin regions in Oryza sativa and for transcription factor binding sites inferred through protein-binding microarrays in O.sativa and Zea mays. Furthermore, the method is shown to recover experimentally profiled ga2ox1-like KN1 binding sites in Z.mays. BLSSpeller was written in Java.     Full Text Available  In the isoperabelic calorimeter at 1870 are determined partial and integral enthalpies of mixture of liquid alloys of systems Fe – Ni – O - Me and Fe – Ni – O – Me – Si, where Me – IVb-, Vb-, VIb-metals. The basis of alloy was served invars with the contents of oxygen by 0,06 %. Established, that the melts of systems Fe – Ni – O – IVb- (Vb--metals are characterized by strong interparticle interplay. The components of the maiden portions IVb- (Vb--metals in Fe – Ni – O melts are accompanied by very large exothermal effects (from – 400 up to – 1000 kJ/mol, which one considerably surpass those in double melts Fe(Ni – Me. The subsequent portions IVb- (Vb--metals caused smaller allocation of a heat (in limits from – 100 up to – 30 kJ/mol, that is conditioned by a decrease of the contents of dissolved oxygen. The partial enthalpies of mixture of molybdenum and tungsten in melts Fe – Ni – O are close to those in a nickel, and for a chromium exceed them. Is rotined, that D` HSi in liquid alloys Fe – Ni – O – Mo (– 450 kJ/mol considerably surpass that are characteristic for a nickel (– 50 kJ/mol. It is explained by interplay of silicon with the stayed dissolved oxygen in initial melts of a system Fe – Ni – O – Mo.

  1. Thermodynamic scaling behavior in genechips

    Full Text Available Abstract Background Affymetrix Genechips are characterized by probe pairs, a perfect match (PM and a mismatch (MM probe differing by a single nucleotide. Most of the data preprocessing algorithms neglect MM signals, as it was shown that MMs cannot be used as estimators of the non-specific hybridization as originally proposed by Affymetrix. The aim of this paper is to study in detail on a large number of experiments the behavior of the average PM/MM ratio. This is taken as an indicator of the quality of the hybridization and, when compared between different chip series, of the quality of the chip design. Results About 250 different GeneChip hybridizations performed at the VIB Microarray Facility for Homo sapiens, Drosophila melanogaster, and Arabidopsis thaliana were analyzed. The investigation of such a large set of data from the same source minimizes systematic experimental variations that may arise from differences in protocols or from different laboratories. The PM/MM ratios are derived theoretically from thermodynamic laws and a link is made with the sequence of PM and MM probe, more specifically with their central nucleotide triplets. Conclusion The PM/MM ratios subdivided according to the different central nucleotides triplets follow qualitatively those deduced from the hybridization free energies in solution. It is shown also that the PM and MM histograms are related by a simple scale transformation, in agreement with what is to be expected from hybridization thermodynamics. Different quantitative behavior is observed on the different chip organisms analyzed, suggesting that some organism chips have superior probe design compared to others.

  2. Reference binding energies of transition metal carbides by core-level x-ray photoelectron spectroscopy free from Ar+ etching artefacts

    We report x-ray photoelectron spectroscopy (XPS) core level binding energies (BE's) for the widely-applicable groups IVb-VIb transition metal carbides (TMCs) TiC, VC, CrC, ZrC, NbC, MoC, HfC, TaC, and WC. Thin film samples are grown in the same deposition system, by dc magnetron co-sputtering from graphite and respective elemental metal targets in Ar atmosphere. To remove surface contaminations resulting from exposure to air during sample transfer from the growth chamber into the XPS system, layers are either (i) Ar+ ion-etched or (ii) UHV-annealed in situ prior to XPS analyses. High resolution XPS spectra reveal that even gentle etching affects the shape of core level signals, as well as BE values, which are systematically offset by 0.2-0.5 eV towards lower BE. These destructive effects of Ar+ ion etch become more pronounced with increasing the metal atom mass due to an increasing carbon-to-metal sputter yield ratio. Systematic analysis reveals that for each row in the periodic table (3d, 4d, and 5d) C 1s BE increases from left to right indicative of a decreased charge transfer from TM to C atoms, hence bond weakening. Moreover, C 1s BE decreases linearly with increasing carbide/metal melting point ratio. Spectra reported here, acquired from a consistent set of samples in the same instrument, should serve as a reference for true deconvolution of complex XPS cases, including multinary carbides, nitrides, and carbonitrides.

  3. Topological side-chain classification of beta-turns: ideal motifs for peptidomimetic development.

    Beta-turns are important topological motifs for biological recognition of proteins and peptides. Organic molecules that sample the side chain positions of beta-turns have shown broad binding capacity to multiple different receptors, for example benzodiazepines. Beta-turns have traditionally been classified into various types based on the backbone dihedral angles (phi2, psi2, phi3 and psi3). Indeed, 57-68% of beta-turns are currently classified into 8 different backbone families (Type I, Type II, Type I', Type II', Type VIII, Type VIa1, Type VIa2 and Type VIb and Type IV which represents unclassified beta-turns). Although this classification of beta-turns has been useful, the resulting beta-turn types are not ideal for the design of beta-turn mimetics as they do not reflect topological features of the recognition elements, the side chains. To overcome this, we have extracted beta-turns from a data set of non-homologous and high-resolution protein crystal structures. The side chain positions, as defined by C(alpha)-C(beta) vectors, of these turns have been clustered using the kth nearest neighbor clustering and filtered nearest centroid sorting algorithms. Nine clusters were obtained that cluster 90% of the data, and the average intra-cluster RMSD of the four C(alpha)-C(beta) vectors is 0.36. The nine clusters therefore represent the topology of the side chain scaffold architecture of the vast majority of beta-turns. The mean structures of the nine clusters are useful for the development of beta-turn mimetics and as biological descriptors for focusing combinatorial chemistry towards biologically relevant topological space.

    Highlights: • Mo-doped tungsten bronzes were hydrothermally synthesized at 80 °C. • Samples transformed from hexagonal to monoclinic structure with Mo-content increase. • 1.5% Mo-doped samples show the best near-infrared shielding performance. • High Mo-doping weakens localized surface plasmon resonance (LSPR) absorption and thus NIR shielding performance. • Small polaron absorption seems to be less affected by Mo-doping. - Abstract: Both Mo and W belong to VIB-sub-group, and possess similar ionic radii, electronegativity and oxide lattice configuration. Herein, Mo-doped (0–80 at.%) tungsten bronzes, M{sub x}WO{sub 3}, were hydrothermally prepared to systematically explore the influence of Mo-doping on their micro-structure and optical performance. The products adopted a hexagonal structure within 6 at.% Mo-doping, and transformed into a monoclinic phase with higher Mo-doping content. Further tests suggested that 1.5 at.% Mo-doping is beneficial for the formation of pure hexagonal phase and uniform nano-rod morphology. Optical measures showed that all samples exhibited high and comparable visible transmittance (70–80%), but a very different near infrared (NIR) shielding ability. The sample doped with 1.5 at.% Mo demonstrated the best NIR shielding ability with a transmittance minimum of 20% at 1300 nm. Further increase of Mo-doping dosage remarkably deteriorated NIR shielding ability by depressing the absorption of localized surface plasmon resonance (LSPR). However, the optical absorption from small-polaron was less influenced by the introduction of Mo. As a result, Mo-doping caused an evident blue shift of the infrared absorption peaks from 1350 to 750 nm.

    Full Text Available The study aims at developing a nation character and culture values-based interactive English textbook and examining its effectivity for the elementary school students in Malang. The development and experiment designs are used and the subjects  consist of experts and target. The expert is an expert in instructional materials and multimedia, while the target is students of the sixth grade Elementary School Dinoyo IV Malang which totals 40 students. The true experiment design, pretest-posttest control group is used  by random sampling technique to obtain experiment and control groups, each of which consists of 20 students. The instruments are questionnaire, interview, and test. Based on the data analysis, it was found that the interactive English textbook fulfilled good criteria of textbook in either material coverage and selection or multimedia application. The positive responses of the students and teachers indicated good quality of textbook in improving students’ competences in English. Through statistical test, it was found t result (5.93  was bigger than t table (1.310 in level of significance 0.01 and (1.697 in level of significance 0.05. It means that Ha which states “The students of class VIB Elementary School Dinoyo IV Malang who were taught using the nation character and culture values-based interactive English textbook, had better English competences than those of class VIA who were taught using manual English textbook” was accepted, while H0 was rejected. Therefore, it can be concluded that the  use of the textbook is very effective in improving the students English competences of Elementary School in Malang.

    Full Text Available A identificação de zonas de manejo de uma unidade de conservação é uma peça-chave para o sucesso de sua gestão. Esse zoneamento, proposto no plano de manejo, baseia-se nos atributos dos ecossistemas inseridos nos limites e entorno da unidade de conservação. No entanto, a proposição de zonas geralmente é um processo mais empírico que técnico. Isso ocorre devido à carência de ferramentas capazes de unir atributos ambientais e valores biológicos dispersos na paisagem. Aqui é apresentada uma ferramenta de análise que identifica regiões de diferentes graus de relevância biológica com base no cruzamento de camadas de informação espacial (declividade, altimetria, classes de vegetação, com um índice denominado Valor de Importância Biológica (VIB produzido pela riqueza e grau de ameaça da fauna em diferentes pontos amostrais. Os registros pontuais de fauna foram obtidos durante uma Avaliação Ecológica Rápida dos Parques Nacionais da Serra da Bodoquena e Chapada dos Guimarães para seu plano de manejo. A importância das diferentes camadas de informação (biológica e espacial foi ponderada utilizando ferramentas de GIS. Este método representou eficientemente as características da paisagem, ajudou a delimitar a zona de amortecimento e indicou a relevância de cada fitofisionomia das Unidades de Conservação na conservação da biodiversidade.

    The authors would like to thank the discussers for their interest in the paper. The discussers raise several objections to the original work; namely, that the state-space equations of motion were derived incorrectly, thus rendering the original results incorrect by association. In actuality, the error in the original state-space equations (Eq. (16) in the original work) is typographic only. This, in conjunction with a typographic error in Table A1, prevented the discussers from replicating a subset of the original results (though the discussers were able to replicate many of the results presented in the original work, despite the presumed error in the equations). These typographic errors are rectified here. In addition, results are presented here corresponding to the solution that would have been obtained had the state-space equations been incorrect in the manner presumed by the discussers. Finally, the discussers state that the results presented in the original work do not adhere to physical principles because the steady-state solution in one case indicates contact even though the linear response to imbalance is less than the radial clearance. This seeming discrepancy is also addressed here.

We doped the transition metal atoms from the IIIB to VIB groups in nonmagnetic 1T-HfS{sub 2}. Numerical results show that the pristine 1T-HfS{sub 2} is a semiconductor with indirect gaps of 1.250 eV. Magnetism can be observed for V, Cr, Mn, Fe, Co, and Cu doping. The polarized charges mainly arise from the localized 3d electrons of the TM atom. The strong p–d hybridization was found between the 3d orbitals of TM and 3p orbitals of S. The substituted 1T-HfS{sub 2} can be a metal, semiconductor or half-metal. Analysis of the band structure and magnetic properties indicates that TM-doped HfS{sub 2} (TM = V, Fe, Cu) are promising systems to explore two-dimensional diluted magnetic semiconductors. The formation energy calculations also indicate that it is energetically favorable and relatively easier to incorporate transition metal atom into the HfS{sub 2} under S-rich experimental conditions. In contrast, V-doped HfS{sub 2} has relatively wide half-metallic gap and low formation energy. So V-doped 1T-HfS{sub 2} is ideal for spin injection, which is important for application in semiconductor spintronics.

  10. Assessing genetic diversity among Brettanomyces yeasts by DNA fingerprinting and whole-genome sequencing.

    Brettanomyces yeasts, with the species Brettanomyces (Dekkera) bruxellensis being the most important one, are generally reported to be spoilage yeasts in the beer and wine industry due to the production of phenolic off flavors. However, B. bruxellensis is also known to be a beneficial contributor in certain fermentation processes, such as the production of certain specialty beers. Nevertheless, despite its economic importance, Brettanomyces yeasts remain poorly understood at the genetic and genomic levels. In this study, the genetic relationship between more than 50 Brettanomyces strains from all presently known species and from several sources was studied using a combination of DNA fingerprinting techniques. This revealed an intriguing correlation between the B. bruxellensis fingerprints and the respective isolation source. To further explore this relationship, we sequenced a (beneficial) beer isolate of B. bruxellensis (VIB X9085; ST05.12/22) and compared its genome sequence with the genome sequences of two wine spoilage strains (AWRI 1499 and CBS 2499). ST05.12/22 was found to be substantially different from both wine strains, especially at the level of single nucleotide polymorphisms (SNPs). In addition, there were major differences in the genome structures between the strains investigated, including the presence of large duplications and deletions. Gene content analysis revealed the presence of 20 genes which were present in both wine strains but absent in the beer strain, including many genes involved in carbon and nitrogen metabolism, and vice versa, no genes that were missing in both AWRI 1499 and CBS 2499 were found in ST05.12/22. Together, this study provides tools to discriminate Brettanomyces strains and provides a first glimpse at the genetic diversity and genome plasticity of B. bruxellensis.     A cytosolic 84-kDa group VIA phospholipase A(2) (iPLA(2)beta) that does not require Ca(2+) for catalysis has been cloned from several sources, including rat and human pancreatic islet beta-cells and murine P388D1 cells. Many potential iPLA(2)beta functions have been proposed, including a signaling role in beta-cell insulin secretion and a role in generating lysophosphatidylcholine acceptors for arachidonic acid incorporation into P388D1 cell phosphatidylcholine (PC). Proposals for iPLA(2)beta function rest in part on effects of inhibiting iPLA(2)beta activity with a bromoenol lactone (BEL) suicide substrate, but BEL also inhibits phosphatidate phosphohydrolase-1 and a group VIB phospholipase A(2). Manipulation of iPLA(2)beta expression by molecular biologic means is an alternative approach to study iPLA(2)beta functions, and we have used a retroviral construct containing iPLA(2)beta cDNA to prepare two INS-1 insulinoma cell clonal lines that stably overexpress iPLA(2)beta. Compared with parental INS-1 cells or cells transfected with empty vector, both iPLA(2)beta-overexpressing lines exhibit amplified insulin secretory responses to glucose and cAMP-elevating agents, and BEL substantially attenuates stimulated secretion. Electrospray ionization mass spectrometric analyses of arachidonic acid incorporation into INS-1 cell PC indicate that neither overexpression nor inhibition of iPLA(2)beta affects the rate or extent of this process in INS-1 cells. Immunocytofluorescence studies with antibodies directed against iPLA(2)beta indicate that cAMP-elevating agents increase perinuclear fluorescence in INS-1 cells, suggesting that iPLA(2)beta associates with nuclei. These studies are more consistent with a signaling than with a housekeeping role for iPLA(2)beta in insulin-secreting beta-cells.

  12. Spectroscopy of vibrationally hot molecules: Hydrogen cyanide and acetylene

    An efficient formula for calculating nuclear spin statistical weights is presented. New experimental methods to distinguish electric and magnetic multipole transitions are proposed and used to prove that the formaldehyde A - X 0-0 transition is a magnetic dipole transition. HIgh resolution vacuum ultraviolet studies of the A → X fluorescence excitation spectrum of hydrogen cyanide (HCN) have: (i) determined that only the (0,1,0) vibrational level of the HCN A-state has a sufficiently long fluorescence lifetime to be suitable for Stimulated Emission Pumping (SEP) studies; and (ii) measured the electric dipole moment of the A-state. Several transitions in the hydrogen cyanide A → X SEP spectrum are shown to be due to the axis-switching mechanism. From a Franck-Condon plot of the intensities and a comparison between sums of predicted rotational constants and sums of observed rotational constants, all of the remaining transitions in the SEP spectrum can be securly assigned. Two weak resonances; a 2:3 CH:CN stretch Fermi resonance and a 6:2 bend:CN stretch resonance appear in the SEP spectrum. Excitation of the CH stretching vibration is predicted and shown to be entirely absent, apart from resonances, in the HCN SEP spectrum. A → X SEP spectra of acetylene (HCCH) near E VIB = 7,000 cm -1 display a wealth of strong and fully assignable anharmonic resonances and forbidden rotational transitions. It is proved that Darling-Dennison resonance between the cis and trans bending vibrations is the crucial first step in a series of anharmonic resonances which can transfer nearly all the vibrational energy out of the initial CC stretch/trans-bend excitation at high vibrational energy. Secondary steps in the vibrational energy flow are vibrational-l-resonance and the '2345' Fermi resonance. For short times, the vibrational energy redistribution obeys very restrictive rules

  13. Ab initio anharmonic vibrational frequency predictions for linear proton-bound complexes OC-H(+)-CO and N(2)-H(+)-N(2).

    Ab initio anharmonic transition frequencies are calculated for strongly coupled (i) asymmetric and (ii) symmetric proton stretching modes in the X-H(+)-X linear ionic hydrogen bonded complexes for OCHCO(+) and N(2)HN(2)(+). The optimized potential surface is calculated in these two coordinates for each molecular ion at CCSD(T)/aug-cc-pVnZ (n = 2-4) levels and extrapolated to the complete-basis-set limit (CBS). Slices through both 2D surfaces reveal a relatively soft potential in the asymmetric proton stretching coordinate at near equilibrium geometries, which rapidly becomes a double minimum potential with increasing symmetric proton acceptor center of mass separation. Eigenvalues are obtained by solution of the 2D Schrödinger equation with potential/kinetic energy coupling explicity taken into account, converged in a distributed Gaussian basis set as a function of grid density. The asymmetric proton stretch fundamental frequency for N(2)HN(2)(+) is predicted at 848 cm(-1), with strong negative anharmonicity in the progression characteristic of a shallow "particle in a box" potential. The corresponding proton stretch fundamental for OCHCO(+) is anomalously low at 386 cm(-1), but with a strong alternation in the vibrational spacing due to the presence of a shallow D(infinityh) transition state barrier (Delta = 398 cm(-1)) between the two equivalent minimum geometries. Calculation of a 2D dipole moment surface and transition matrix elements reveals surprisingly strong combination and difference bands with appreciable intensity throughout the 300-1500 cm(-1) region. Corrected for zero point (DeltaZPE) and thermal vibrational excitation (DeltaE(vib)) at 300 K, the single and double dissociation energies in these complexes are in excellent agreement with thermochemical gas phase ion data.

  14. Numerical simulation of the generation mechanism of axisymmetric supersonic jet screech tones

    In this paper an axisymmetric computational aeroacoustic procedure is developed to investigate the generation mechanism of axisymmetric supersonic jet screech tones. The axisymmetric Navier-Stokes equations and the two equations standard k-ɛ turbulence model modified by Turpin and Troyes ["Validation of a two-equation turbulence model for axisymmetric reacting and non-reaction flows," AIAA Paper No. 2000-3463 (2000)] are solved in the generalized curvilinear coordinate system. A generalized wall function is applied in the nozzle exit wall region. The dispersion-relation-preserving scheme is applied for space discretization. The 2N storage low-dissipation and low-dispersion Runge-Kutta scheme is employed for time integration. Much attention is paid to far-field boundary conditions and turbulence model. The underexpanded axisymmetric supersonic jet screech tones are simulated over the Mach number from 1.05 to 1.2. Numerical results are presented and compared with the experimental data by other researchers. The simulated wavelengths of A0, A1, A2, and B modes and part of simulated amplitudes agree very well with the measurement data by Ponton and Seiner ["The effects of nozzle exit lip thickness on plume resonance," J. Sound Vib. 154, 531 (1992)]. In particular, the phenomena of modes jumping have been captured correctly although the numerical procedure has to be improved to predict the amplitudes of supersonic jet screech tones more accurately. Furthermore, the phenomena of shock motions are analyzed. The predicted splitting and combination of shock cells are similar with the experimental observations of Panda ["Shock oscillation in underexpanded screeching jets," J. Fluid. Mech. 363, 173 (1998)]. Finally, the receptivity process is numerically studied and analyzed. It is shown that the receptivity zone is associated with the initial thin shear layer, and the incoming and reflected sound waves.

  15. Using the integral equations method to model a 2G racetrack coil with anisotropic critical current dependence

    Second-generation (2G) superconducting wires have already proved their potential in several applications. These materials have a highly nonlinear behavior that turns an optimized engineering project into a challenge. Between several numerical techniques that can be used to perform this task, the integral equations (IE) method stands out for avoiding mesh problems by representing the 2G wire cross-sectional area by a line. While most applications need to be represented in a 3D geometry, the IE is limited to longitudinal or axisymmetric models. This work demonstrates that a complex 3D geometry can be modeled by several coupled simulations using the IE method. In order to prove this statement, the proposed technique was used to simulate a 2G racetrack coil considering the self-field magnitude (B) and incidence angle (θ) on the tape. The J c characteristic was modeled in terms of parallel and normal to the tape plane magnetic field components (J c(B ∥ , B ⊥)) obtained from a V-I(B, θ) characterization of a tape segment. This result was implemented using commercial software with both A-V (vector magnetic potential and scalar voltage potential) and IE coupled simulations solved by finite elements. This solution bypasses the meshing problem due to the tapes slim geometry, considering each turn a single 1D model, all magnetically interacting in two 2D models. The simulations results are in good agreement to what was both expected and observed in the literature. The simulation is compared to the measured V-I characteristic for a single pancake racetrack coil built with same geometry as its simulation models, and a theoretical study demonstrates the possibilities of the proposed tool for analyzing a racetrack coil current density and electric field behavior in each of its turns.

  16. Infiltração de água no solo em um latossolo vermelho da região sudoeste dos cerrados com diferentes sistemas de uso e manejo

    Full Text Available Na região do Cerrado, as atividades agrícolas têm provocado impactos negativos, principalmente, na qualidade do solo e da água. Tanto em sistemas de exploração agrícola ou de pecuária, com diferentes manejos, a degradação física do solo é uma das consequências da redução da produtividade. O objetivo deste trabalho foi verificar a infiltração de água no solo sob influência de diferentes sistemas de manejo em um Latossolo Vermelho distrófico na região dos Cerrados. Os tratamentos foram os seguintes sistemas de manejo: a solo sob vegetação nativa; b solo com implantação de lavoura de soja durante um ano, seguido de três anos de pastagens; c solo com implantação de lavoura de soja durante quatro anos, seguidos por quatro anos de pastagem; d solo com lavoura de soja contínua em sistema de plantio direto; e e solo com pastagem continuamente. Foram avaliadas, no período de 1995 a 2002, a velocidade de infiltração básica (VIB e a infiltração de água acumulada no solo, utilizando o método de duplos anéis concêntricos. Os usos do solo com sistema de lavoura contínua, pecuária contínua e integração lavoura-pecuária tiveram a velocidade de infiltração básica e infiltração acumulada reduzidas em relação ao solo com vegetação nativa na região do Cerrado. Entre os sistemas estudados, o sistema de integração lavoura-pecuária foi o que apresentou valores mais próximos aos do solo sob vegetação nativa.

  18. A Chimeric NaV1.8 Channel Expression System Based on HEK293T Cell Line

    Full Text Available Among the nine voltage-gated sodium channel (NaV subtypes, NaV1.8 is an attractive therapeutic target for pain. The heterologous expression of recombinant NaV1.8 currents is of particular importance for its electrophysiological and pharmacological studies. However, NaV1.8 expresses no or low-level functional currents when transiently transfected into non-neuronal cell lines. The present study aims to explore the molecular determinants limiting its functional expression and accordingly establish a functional NaV1.8 expression system. We conducted screening analysis of the NaV1.8 intracellular loops by constructing NaV chimeric channels and confirmed that the NaV1.8 C-terminus was the only limiting factor. Replacing this sequence with that of NaV1.4, NaV1.5, or NaV1.7 constructed functional channels (NaV1.8/1.4L5, NaV1.8/1.5L5, and NaV1.8/1.7L5, respectively, which expressed high-level NaV1.8-like currents in HEK293T cells. The chimeric channel NaV1.8/1.7L5 displayed much faster inactivation of its macroscopic currents than NaV1.8/1.4L5 and NaV1.8/1.5L5, and it was the most similar to wild-type NaV1.8 expressed in ND7/23 cells. Its currents were very stable during repetitive depolarizations, while its repriming kinetic was different from wild-type NaV1.8. Most importantly, NaV1.8/1.7L5 pharmacologically resembled wild-type NaV1.8 as revealed by testing their susceptibility to two NaV1.8 selective antagonists, APETx-2 and MrVIB. NaV chimeras study showed that at least the domain 2 and domain 4 of NaV1.8 were involved in binding with APETx-2. Our study provided new insights into the function of NaV1.8 intracellular loops, as well as a reliable and convenient expression system which could be useful in NaV1.8 studies.

    Neuromuscular electrical stimulation (NMES) is commonly used in skeletal muscles in people with spinal cord injury (SCI) with the aim of increasing muscle recruitment and thus muscle force production. NMES has been conventionally used in clinical practice as functional electrical stimulation (FES), using low levels of evoked force that cannot optimally stimulate muscular strength and mass improvements, and thus trigger musculoskeletal changes in paralysed muscles. The use of high intensity intermittent NMES training using wide-pulse width and moderate-intensity as a strength training tool could be a promising method to increase muscle force production in people with SCI. However, this type of protocol has not been clinically adopted because it may generate rapid muscle fatigue and thus prevent the performance of repeated high-intensity muscular contractions in paralysed muscles. Moreover, superimposing patellar tendon vibration onto the wide-pulse width NMES has been shown to elicit further increases in impulse or, at least, reduce the rate of fatigue in repeated contractions in able-bodied populations, but there is a lack of evidence to support this argument in people with SCI. Nine people with SCI received two NMES protocols with and without superimposing patellar tendon vibration on different days (i.e. STIM and STIM+vib), which consisted of repeated 30 Hz trains of 58 wide-pulse width (1000 μs) symmetric biphasic pulses (0.033-s inter-pulse interval; 2 s stimulation train; 2-s inter-train interval) being delivered to the dominant quadriceps femoris. Starting torque was 20% of maximal doublet-twitch torque and stimulations continued until torque declined to 50% of the starting torque. Total knee extensor impulse was calculated as the primary outcome variable.     Full Text Available Na região do Cerrado, as atividades agrícolas têm provocado impactos negativos, principalmente, na qualidade do solo e da água. Tanto em sistemas de exploração agrícola ou de pecuária, com diferentes manejos, a degradação física do solo é uma das consequências da redução da produtividade. O objetivo deste trabalho foi verificar a infiltração de água no solo sob influência de diferentes sistemas de manejo em um Latossolo Vermelho distrófico na região dos Cerrados. Os tratamentos foram os seguintes sistemas de manejo: a solo sob vegetação nativa; b solo com implantação de lavoura de soja durante um ano, seguido de três anos de pastagens; c solo com implantação de lavoura de soja durante quatro anos, seguidos por quatro anos de pastagem; d solo com lavoura de soja contínua em sistema de plantio direto; e e solo com pastagem continuamente. Foram avaliadas, no período de 1995 a 2002, a velocidade de infiltração básica (VIB e a infiltração de água acumulada no solo, utilizando o método de duplos anéis concêntricos. Os usos do solo com sistema de lavoura contínua, pecuária contínua e integração lavoura-pecuária tiveram a velocidade de infiltração básica e infiltração acumulada reduzidas em relação ao solo com vegetação nativa na região do Cerrado.     Chapter 1: Introduction. A brief introduction to supercritical fluids is given, illustrating why supercritical fluids are unique solvents and why there is a wide application of supercritical fluids in industry and laboratories. Potential ways for solvation in supercritical fluids to affect reactivity are briefly reviewed. A general introduction to the photochemistry of organometallic complexes is also given. Chapter 2: Time resolved vibrational spectroscopy. Time resolved resonance Raman is introduced and compared with Time-resolved infrared spectroscopy (TRIR). The different approaches of TRIR, including microsecond, nanosecond, and ultrafast (picosecond and femtosecond) systems are discussed. The advantages and disadvantages of these systems are also compared. The TRIR apparatus using an IR diode laser used for work in this thesis are described in detail. Experimental procedures for supercritical fluid TRIR experiments are described with emphasis on handling the IR cell for supercritical fluids and preparation of supercritical fluid solutions. Chapter 3: Photochemistry of group VIB hexacarbonyl compounds in supercritical noble gases and CO 2 solutions. A systematic TRIR study of the photolysis of M(CO) 6 in supercritical Ar, Kr, Xe, and CO 2 and the observation of M(CO) 5 L (M = Cr, Mo, and W; L = Ar (W only), Kr, Xe, and CO 2 ) is described. The second-order rate constants for the reaction of M(CO) 5 L with CO have been evaluated and the reactivity for each metal is Kr > Xe ∼ CO 2 . For M(CO) 5 Kr, M(CO) 5 Xe, or M(CO) 5 (CO 2 ), the reactivity is Cr ∼ Mo > W. In supercritical Kr doped with either Xe or CO 2 , the M(CO) 5 moiety interacts with Xe or CO 2 in preference to Kr. The effect of solvent density on the rate of the reaction of W(CO) 5 (CO 2 ) with CO has been investigated. The reaction of W(CO) 5 (CO 2 ) with CO in scCO 2 is predominantly a dissociative process. Após 441 dias, estas mensurações foram realizadas novamente, sendo acrescidas de outras análises: COT, N total, N-NH4+, N-NO3-, porosidade e resistência à penetração. Nas plantas, foram mensurados altura, diâmetro à altura do peito e matéria seca de tronco. Verificou-se que o trânsito do forwarder aumentou a densidade, microporosidade e a resistência do solo à penetração e reduziu o diâmetro médio geométrico, a porosidade total, a macroporosidade e a infiltração de água no solo. Não houve efeito da compactação sobre a produção de matéria seca de tronco e altura das plantas. A maior parte dos efeitos da compactação foi manifestada por apenas duas passadas do forwarder. Os efeitos da compactação ainda permaneciam no solo após 441 dias do trânsito do forwarder.
