  1. Alternative Synthesis of 1,8-Difluoroanthracene via the Balz-Schiemann Reaction

    Full Text Available An alternative and improved method for the synthesis of 1,8-difluoroanthracene 5 is described. The precursor 1,8-difluoro-9,10-anthraquinone 4 was synthesized by precipitating the water-soluble 1,8-diazonium-9,10-anthraquinone fluoroborate 3 using isopropanol, before thermally decomposing it via the Balz-Schiemann reaction to yield 4 in the form of pure sublimed crystals. Compound 5 was later synthesized with 84% yield by reducing 4 in a Zn/NH4OH mixture. Using isopropanol to quantitatively precipitate water-soluble diazonium fluoroborate salts was also used to synthesize other fluoroanthraquinone derivatives with improved yields.

    Full Text Available Agota Kristof and Katalin Molnár, two Franco-Hungarian authors of the twentieth century, share the concern to use a language that can express a certain reality and truth. For that, they distance themselves from the normative French language and write in an incorrect language, more apt to represent their difference. However, while Kristof wants to reach maximum objectivity, Molnár writes in an exaggeratedly subjective way. This article examines the textual and extratextual position of these authors regarding the possibility of writing the truth, through the analysis of language in Kristof's trilogy and in La¬mour Dieu of Molnár. Agota Kristof et Katalin Molnár, deux auteures franco-hongroises du XXe siècle, partagent la même préoccupation d’utiliser un langage pouvant exprimer une certaine réalité et vérité. Pour cela, elles s’éloignent de la langue française normative et écrivent dans un langage incorrect, plus apte à représenter leur différence. Néanmoins, tandis que le but de Kristof est d’arriver à une objectivité totale, Molnár écrit d’une manière exagérément subjective. Dans cet article, nous examinons la posture textuelle et extratextuelle de ces auteures concernant la possibilité d’écrire la vérité, à travers l’analyse du langage dans la trilogie de Kristof et dans Lamour Dieu de Molnár.

    Full Text Available We point out that the fundamental irreversibility of Nature requires the introduction of a suitable measure for the distance from equilibrium. We show that entropy, which is widely held to be such a measure, suffers from the problem that it does not have a physical meaning, since it is introduced on the basis of mathematical arguments. As a consequence, the basic physics beyond irreversibility has remained obscure. We present here a simple but transparent physical approach for solving the problem of irreversibility. This approach shows that extropy, the fundamental thermodynamic variable introduced by Katalin Martinás, is the suitable measure for the distance from equilibrium, since it corresponds to the actual driver of irreversible processes. Since extropy explicitly contains in its definition all the general thermodynamic forces that drive irreversible processes, extropy is the suitable physical measure of irreversibility.

  11. Neuroleptic binding sites: specific labeling in mice with [18F]haloperidol, a potential tracer for positron emission tomography

    Haloperidol labeled with fluorine- 18 (T 1/2 . 110 min, positron emission 97%), prepared yielding .04 Ci/millimole by the Balz-Schiemann reaction, was evaluated in a murine model as a potential radiotracer for noninvasive determination, by positron-emission tomography, of regional concentrations of brain dopamine receptors in patients. As the haloperidol dose in mice was increased from 0.01 to 1000 micrograms/kg, the relative concentration of [ 18 F]haloperidol (microCi per g specimen/microCi per g of body mass), at one hour after injection decreased from 30 to 1.0 in the striatum and from 8.0 to 1.0 in the cerebellum. The striatal radioactivity, plotted as relative concentration against log of dose, decreased sigmoidally, presumably reflecting competition between labeled and unlabeled haloperidol for a single class of accessible binding sites. Because the cerebellum is relatively deficient in dopamine receptors, the observed decrease in cerebellar radioactivity may reflect a saturable component of haloperidol transport into brain. The high brain concentrations and the unexpectedly high striatum-to-cerebellum concentration ratios (greater than 4 at haloperidol doses less than or equal to 1 microgram/kg) suggest that [ 18 F]haloperidol warrants further investigation as a potential radiotracer for dopamine receptors

  12. Development of [11C]-α-aminoisobutyric acid and [18F]-haloperidol as substrate-specific radiotracers

    In light of the emergence of positron emission tomography (PET), two new substrate-specific radiotracers have been synthesized and evaluated as potential agents for the study of specific physiological processes: [1- 11 C]-α-aminoisobutyric acid ([1- 11 C]-AIB), a transport system-specific radiotracer for tumor localization and for the assessment of amino acid transport in vivo; and [ 18 F]-haloperidoll, a receptor-binding radiotracer for the assessment of brain dopamine receptors in vivo. AIB, a non-metabolized amino acid, accumulates in cells, particularly malignant cells, via the A type, or ''alanine-preferring'', amino acid transport system [1- 11 C]-AIB in normal and in treated and untreated tumor-bearing rats, in tumor-bearing dogs, and in a tumor-bearing patient indicate rapid blood clearance and, concomitantly, rapid tissue localization, with slow net redistribution. Generally, there was selective localization in the kidney, in the liver, and in the pancreas, as well as in various tumors. The canine tumors and the human tumor were well visualized by external scintigraphy, especially when utilizing PET. [ 18 F]-Haloperidol, a dopamine receptor-binding neuroleptic of the butyrophenone series widely used in the management of schizophrenia was prepared via the Balz-Schiemann reaction. As the haloperdol dose administered to mice was increased from 0.01 to 1000 μg/kg, the relative concentration (μCi found per gm tisue sample/μCi injected per gm body mass) of [ 18 F]-haloperidol at 1 hr decreased from 30 to 1.0 in the striatum and from 8.0 to 1.0 in the cerebellum. For the largest subgroup of breast cancers, hormone receptor-positive/human epidermal growth factor 2 (HER2-negative tumors, hormone treatment is the mainstay of therapy and is likely to result in significant improvement in disease outcomes. However, some of these cancers demonstrate de novo or acquired resistance to endocrine therapy. Despite intensive research to develop new strategies to enhance the efficacy of currently available treatment options for hormone receptor-positive breast cancer, progress has been slow, and there were few advances for a period of 10 years. In 2012, a new molecularly targeted therapeutic strategy, inhibition of mammalian target of rapamycin with everolimus, was introduced into clinical practice. Everolimus, in combination with a steroidal aromatase inhibitor, exemestane, resulted in an increase in progression-free survival, but not overall survival in patients with estrogen receptor (ER+ve advanced disease who had progressed on hormone therapy. In 2015, the first cyclin-dependent kinases 4/6 (CDK4/6 inhibitor, palbociclib, received accelerated US Food and Drug Administration approval for use in combination with letrozole for the treatment of postmenopausal ER+ve/HER2-ve advanced breast cancer as initial, endocrine-based therapy. The addition of palbociclib to endocrine therapy resulted in longer progression-free survival than letrozole alone. One year later, palbociclib received a new indication, use in combination with fulvestrant, in both premenopausal and postmenopausal females with advanced breast cancer of the same subtype with disease progression following endocrine therapy. Adding palbociclib to fulvestrant resulted in a significantly increased median progression-free survival compared to fulvestrant

    In the first part of this work different strategies for the design of sulfonated partially fluorinated poly(aryl)s are developed and synthetically realized. The applied concept is that partially fluorinated poly(aryl)s are distinguished from the nonfluorinated ones by an enhanced acidity. Moreover they possess higher bond dissociation energies of both the C-F bonds and any adjacent C-H bonds which should be associated with a gain in radical stability and thus in chemical and thermal stability. In order to investigate the influence of the chemical structure of (partially fluorinated) monomeric building blocks, homo-polymers with different structural units (with aromatic C-F bonds, C(CF3)2-bridged and/or CF3-substituted phenylene rings) are synthesized by polycondensation and structurally characterized (elemental analysis, NMR spectroscopy, gel permeation chromatography). Established organic reactions, such as the Balz-Schiemann reaction, Suzuki reaction and Ullmann's biaryl synthesis, are applied for the synthesis of the specific monomers. After sulfonation of the homo-polymers (ionically crosslinked) membranes are prepared and characterized in terms of suitability as polymer electrolyte membrane in fuel cells (ion-exchange capacity, proton conductivity, thermal and chemical stability, water uptake, dimensional change). Both the chemical nature of the monomers and their constitution in the ionomer are important for the properties of the resulting membranes. Therefore microphase-separated multiblock-co-ionomers based on hydrophilic (sulfonated) and hydrophobic (partially fluorinated) telechelic macromonomers are prepared and characterized. Both the influence of the block length and the chemical nature of the used monomers on the membrane properties are comparatively investigated. On the basis of the findings gained in this part of the work, the advantages and disadvantages of partially fluorinated ionomer membranes are analyzed and discussed. The second part of

The site of modification within a protein; their fixed or transient nature; the stoichiometry and potential crosstalk between modifications have been the focus of numerous large-scale studies. Most of this research is focused on PTMs involved in intracellular processes, such as phosphorylation, methylation, GlcNAcylation, acetylation, and ubiquitination. Of these, phosphorylation is the most studied due to the biological importance of this modification as well as the availability of tools for such studies. While the majority of protein phosphorylation occurs on intracellular proteins, phosphorylation of secreted proteins is well established. Well-characterized examples include the secreted milk protein, beta-casein, and the serum protein fetuin. Our current understanding of the biological role of extracellular phosphorylation, as well as knowledge regarding the process by which they are modified, is incomplete. A kinase known as FAM20 has been identified and is localized to the Golgi as well as secreted. It has been shown to phosphorylate extracellular proteins with a SXE motif.
