    The case of a mentally disturbed young man is described, whose identity could not be revealed over more than four months. It is compared to Kaspar Hauser, similarities and differences are highlighted. The "Kaspar Hauser of Stuttgart" is retrospectively revealed as a young man with experiences of severe deprivation. The missing case history and independent history were an obstacle in the treatment, albeit not fundamental, advantages being reached with a relation-oriented approach. Informations obtained after clarification of the identity of the young man suggest a multiplication of unfavourable circumstances, leading to sub-optimal treatment and finally the "Kaspar-Hauser-Situation". Contributing were his belonging to the less integrated group of migrants in Germany, as well as severe deprivation in the family and a generally sub-optimal treatment of mentally disturbed young patients.

    In 1828 a mysterious young man appeared in Nürnberg, Germany, who was barely able to speak or walk but could write down his name, Kaspar Hauser. He quickly became the centre of social interest but also the victim of intrigue. His appearance, his origin and assassination in 1833 were, and still are, the source of much debate. The most widely accepted theory postulates that Kaspar Hauser was the son of Grand Duke Carl von Baden and his wife Stephanie de Beauharnais, an adopted daughter of Napoleon Bonaparte. To check this theory, DNA analysis was performed on the clothes most likely worn by Kaspar Hauser when he was stabbed on December 14th, 1833. A suitable bloodstain from the underpants was divided and analysed independently by the Institute of Legal Medicine, University of Munich (ILM) and the Forensic Science Service Laboratory, Birmingham (FSS). Mitochondrial DNA (mtDNA) was sequenced from the bloodstain and from blood samples obtained from two living maternal relatives of Stephanie de Beauharnais. The sequence from the bloodstained clothing differed from the sequence found in both reference blood samples at seven confirmed positions. This proves that the bloodstain does not originate from a son of Stephanie de Beauharnais. Thus, it is becoming clear that Kaspar Hauser was not the Prince of Baden.

    Full Text Available The topos of the "Wild Child" occupies an important place in the mythic and literary imagination of the West. The European climax of a long line of wild children, Kaspar Hauser was a nineteenth-century German foundling whose fate has inspired a host of novels, dramas, novellas, poems, songs, and movies, even an opera and a ballet. It has been treated by Paul Verlaine, R. M. Rilke, and Klaus Mann, by the Dada poet Hans Arp, by the dramatist Peter Handke, and by the filmmaker Werner Herzog. This article offers a brief historical sketch of Hauser's life before discussing a key aspect of the literary Kaspar Hauser reception: the motif of the foundling as a latter-day messiah. By examining a set of twentieth-century texts that develop this motif in an exemplary manner—Jakob Wassermann's novel Caspar Hauser or The Inertia of the Heart , Georg Trakl's Kaspar Hauser Song , the anthroposophist Kaspar Hauser poems by Emma Krell-Werth, and David Constantine's Caspar Hauser: A Poem in Nine Cantos —we can gain insight into our century's desire for redemption and its ethical ramifications.

    Full Text Available A partir de uma abordagem histórico-cultural em Psicologia, este trabalho analisa o percurso de desenvolvimento de Kaspar Hauser, um personagem real e enigmático que, quando encontrado em Nuremberg, em 1928, com supostamente 15 anos, não sabia falar, nem andar e não se comportava como humano. Até hoje o seu enigma persiste: apesar de muitas hipóteses e suspeitas, não se descobriu sua origem. Apoiando-se em estudos de Vygotsky e Luria, que indicam que a percepção depende, sobretudo, da práxis social, necessária para gestar o referencial cultural de apreensão da realidade, a autora analisa como se articulam linguagem e pensamento no desenvolvimento cognitivo de Kaspar Hauser e como ele concebe o mundo que o cerca, tendo sido privado dos filtros e estereótipos culturais que condicionam a percepção e o conhecimento.Starting from a historical-cultural approach in Psychology, this work analyzes the development of Kaspar Hauser, a real and enigmatic character that didn't know how to speak, nor to walk and didn't behave as a human being when he was found in Nuremberg, in 1928, at supposedly age 15. Still today, his enigma remains: in spite of a lot of hypotheses and suspicions we have not yet discovered its origins. Leaning on studies of Vygotsky and Luria, which indicate that the perception depends, above all, on the social practice which is necessary to produce the cultural reference for the apprehension of reality, the author analyzes how language and thought are articulated in Kaspar Hauser's cognitive development and how he conceives the world that surrounds him, having been deprived from the filters and cultural stereotypes that stipulate the perception and knowledge.

    Full Text Available A história de Kaspar Hauser foi retratada em filme pelo autor Werner Herzog. Este filme apresenta uma criança selvagem, encarcerada, sem nenhum contato verbal ou social até por volta de seus 16 anos. Esta criança, quando levada a uma comunidade alemã, passa a ser objeto de curiosidade e estudo, devido a não conseguir se expressar, conceituar, raciocinar, ou até mesmo diferenciar sonho de realidade. Durante sua convivência nesta comunidade, pôde ser observado o seu desenvolvimento na linguagem e sua socialização. Este artigo reflete sobre o desenvolvimento linguístico e social de Kaspar Hauser, sob o viés do sócio-interacionismo vygotskiano. Destaca-se que, a interação com outro indivíduo foi essêncial para o desenvolvimento e aprendizagem do protagonista do filme.

    In 1661, Kaspar Schott published his comprehensive textbook "Cursus mathematicus" in Würzburg for the first time, his "Encyclopedia of all mathematical sciences". It was so successful that it was published again in 1674 and 1677. In its 28 books, Schott gave an introduction for beginners in 22 mathematical disciplines by means of 533 figures and numerous tables. He wanted to avoid the shortness and the unintelligibility of his predecessors Alsted and Hérigone. He cited or recommended far more than hundred authors, among them Protestants like Michael Stifel and Johannes Kepler, but also Catholics like Nicolaus Copernicus. The paper gives a survey of this work and explains especially interesting aspects: The dedication to the German emperor Leopold I., Athanasius Kircher's letter of recommendation as well as Schott's classification of sciences, explanations regarding geometry, astronomy, and algebra.

    The death of Kaspar Hauser is described taking present medicolegal and criminological knowledge into account, the main question being whether the injury was self-inflicted, homicidal or suicidal. In spite of a critical retrospective analysis this question cannot be answered with sufficient certainty even from our modern perspective. It seems unlikely that the stab to the chest was inflicted exclusively for the purpose of self-damage, but both a suicidal stab and a homicidal act (assassination) cannot be definitely ruled out.

    The feral children literature has frequently been cited for relevance to understanding historical antecedents of autism. Kaspar Hauser, who appeared in Nuremberg, Germany in 1828, is one of these children, raised under conditions of extreme deprivation. His case history and gradual acquisition of language after age 17 years are summarized. There is strong evidence that he was the prince of Baden, abducted from his cradle in 1812. Findings of postmortem examination, conducted after his assassination, are discussed. Hauser's postadolescent recovery of language contradicts the notion of a "critical period" for language development.

    Full Text Available The text on Sami shamanism in Caspar Peucer’s Commentarius de praecipuis divinationum generibus (Wittenberg 1560: Critical edition, with translation and commentary. Among the sources dealing with the shamanistic skills of the Sami (formerly Lapponian population, a certain text by Kaspar Peucer has so far been little known. This man of extreme learning was the son-in-law of Philip Melanchthon and a Professor at the University of Wittenberg. A true polyhistor, well versed in Medicine, Geography, Astrology, Theology, etc., Peucer included in his chef-d’oeuvre on divination an elaborate description of the shamanism of the so-called Pilappii. The present article offers a critical edition of this text, based on the editions of Wittenberg 1560 (A, 1572 (B, 1580 (C, as well as Zerbst 1591 (D and Frankfurt 1593 (E. In addition to translations into North Sami and Norwegian (see Appendix, some contextualisation is offered, which can be summarised as follows: A similar testimony on shamanism is found in the Historia de gentibus Septentrionalibus by Olaus Magnus (Rome 1555. However, that text is not elaborate enough to prove that Kaspar Peucer has copied his description from him. It is more likely that some student among the considerable number of Swedes, Finns and Norwegians that were immatriculated at Wittenberg University in the years following the Reformation, presented this account to Peucer. Many details in the account make it strikingly similar to Sami folk narratives that have been assembled several centuries later. For example, the description of maritime Sami by Anders Larsen (1870–1949, the Sami book by Johan Turi (published 1910 and Sami songs (joik that were collected by Jacob Fellman in the 1820’s can be compared with Peucer’s account. Peucer himself, however, categorised the shamanism of the Sami as a form of theomanteia, i.e. a form of magic which he considered to originate not from the true God, but from the Devil.

    Full Text Available Robots have been used in a variety of education, therapy or entertainment contexts. This paper introduces the novel application of using humanoid robots for robot-mediated interviews. An experimental study examines how children's responses towards the humanoid robot KASPAR in an interview context differ in comparison to their interaction with a human in a similar setting. Twenty-one children aged between 7 and 9 took part in this study. Each child participated in two interviews, one with an adult and one with a humanoid robot. Measures include the behavioural coding of the children's behaviour during the interviews and questionnaire data. The questions in these interviews focused on a special event that had recently taken place in the school. The results reveal that the children interacted with KASPAR very similar to how they interacted with a human interviewer. The quantitative behaviour analysis reveal that the most notable difference between the interviews with KASPAR and the human were the duration of the interviews, the eye gaze directed towards the different interviewers, and the response time of the interviewers. These results are discussed in light of future work towards developing KASPAR as an 'interviewer' for young children in application areas where a robot may have advantages over a human interviewer, e.g. in police, social services, or healthcare applications.

    This paper is an account of a four-times-a-week analysis with a very deprived, feral-like child. The author, who has come to understand feral children as very deprived children, turns to an historical account written in 1826 by Anselm von Feuerbach, the jurist who was asked to make enquiries into the identity of a savage and mysterious boy, Kaspar Hauser. The author describes how she has made use of the observations and thoughts of Anselm von Feurbach in her attempts to process her own thoughts and reflections around the unfolding clinical material and how the exploration of the differences and similarities between Casper and Kaspar Hauser helped her to gradually grasp the psychological essence of the patient. The question of an appropriate reading of the clinical material for theoretical purposes remains open.

    Full Text Available Philosopher, poet and Belgian French speaking writer, Véronique Bergen reaches, with her book Kaspar Hauser (2006 (rewarded with the great literary triennial prize of the city of Tournai in 2007, an extraordinary mastery, as well by the way she depicts the complexity of the character as the way she describes European history at the beginning of the 19th century. She does it by not only diving into the frenzied relations to power or into the intimate history of several characters full of contrasts, but also by weaving all her book from the Kaspar character, from the issues of language and symbolic – and especially from the ground water of the poetic use of the language and of the world representation. By doing so, she also confirms that French speaking fictions are rooted in an in-between of the language and the History.

    The merger announcement between DeWaal Pharmaceuticals and BioHealth Labs was front-page news. Pictures of CEO Steve Lindell and chairman Kaspar van de Velde had appeared in newspapers around the world. Two months later, the press had moved on to a new story, and the hard labor of integration loomed. Steve had worked tirelessly to clear regulatory hurdles, and all signs pointed toward approval in the near future. Now Steve was feeling pressure to attack the real challenge of the merger: bringing together two very different cultures as quickly and efficiently as possible. DeWaal was an established drug-maker based in the Netherlands, and BioHealth, headquartered just north of New York City, had in recent years become competitive at the highest tier of the market. The first step in integrating the two companies was to select the top layers of management for the new company. At the moment, there were some 120 people on two continents for about 65 senior-level jobs. Steve's urgency was not without cause: talented people from both sides were jumping ship, and BioHealth's stock price had dipped 20% after the initial euphoria over the deal had worn off. Complicating matters was confusion over who was really in charge: Steve wanted to take leadership and move ahead rapidly, but he was often disarmed by Kaspar's charming persuasiveness. As the two men attempt to work through the important personnel issues during a lunch meeting, they quickly hit a roadblock. How can they come to agreement about who goes and who stays?     Chloroplast DNA is a primary source of molecular variations for phylogenetic analysis of photosynthetic eukaryotes. However, the sequencing and analysis of multiple chloroplastic regions is difficult to apply to large collections or large samples of natural populations. The objective of our work was to demonstrate that a molecular taxonomic key based on easy, scalable and low-cost genotyping method should be developed from a set of Single Nucleotide Polymorphisms (SNPs) diagnostic of well-established clades. It was applied to the Aurantioideae subfamily, the largest group of the Rutaceae family that includes the cultivated citrus species. The publicly available nucleotide sequences of eight plastid genomic regions were compared for 79 accessions of the Aurantioideae subfamily to search for SNPs revealing taxonomic differentiation at the inter-tribe, inter-subtribe, inter-genus and interspecific levels. Diagnostic SNPs (DSNPs) were found for 46 of the 54 clade levels analysed. Forty DSNPs were selected to develop KASPar markers and their taxonomic value was tested by genotyping 108 accessions of the Aurantioideae subfamily. Twenty-seven markers diagnostic of 24 clades were validated and they displayed a very high rate of transferability in the Aurantioideae subfamily (only 1.2 % of missing data on average). The UPGMA from the validated markers produced a cladistic organisation that was highly coherent with the previous phylogenetic analysis based on the sequence data of the eight plasmid regions. In particular, the monophyletic origin of the "true citrus" genera plus Oxanthera was validated. However, some clarification remains necessary regarding the organisation of the other wild species of the Citreae tribe.   13. [The proof of paternity. An andrological-forensic challenge in historical perspective].

    For centuries, difficulties have occurred in determining unresolved paternities. In addition to the modern standard methods, such as the examination of DNA or serological proof, expert opinion on fertility once played an important role. The andrological difference between incapability to fertilise and the inability to participate in sexual intercourse was also distinguished historically. Of special significance was the discovery of spermatozoa by the medical student Johan Ham in 1677 and their further investigation by Antoni van Leeuwenhoek.Recently, modern DNA methods have also been applied for historical investigations. Illustrious examples are the DNA analysis in the case of Kaspar Hauser of Ansbach and the dispute about Thomas Jefferson, President of the U.S., fathering a child by one of his slaves. In this discourse, a medicinal-forensic review of the development of expert opinion, illustrated with historical case studies, is given.

    This book explores the relations between physical parameters of extrasolar planets and their respective parent stars. Planetary parameters are often directly dependent upon their stellar counterparts. In addition, the star is almost always the only visible component of the system and contains most of the system mass. Consequently, the parent star heavily influences every aspect of planetary physics and astrophysics. Drs. Kaspar von Braun and Tabetha Boyajian use direct methods to characterize exoplanet host starts that minimize the number of assumptions needed to be made in the process. The book provides a background on interferometric techniques for stellar diameter measurements, illustrates the authors' approach on using additional data to fully characterize the stars, provides a comprehensive update on the current state of the field, and examines in detail a number of historically significant and well-studied exoplanetary systems.

    Full Text Available Fictional autobiographies of a Belgian writer, Véronique Bergen, describing fates of outstanding characters albeit marginalised by history (Kaspar Hauser, Ludwig II of Bavaria, Ulrike Meinhof, may be interpreted as a metaphor of a contemporary man’s fate, a man caught, often against his will, in a course of political or social events. The characters described here adopt an active resistance stand against life, the stand defi ned by Bergen in her essay Résistances philosophiques, and manifested by striving to create one’s own, coherent world and a new language.     Full Text Available Quinoa ( Willd. is an important seed crop throughout the Andean region of South America. It is important as a regional food security crop for millions of impoverished rural inhabitants of the Andean Altiplano (high plains. Efforts to improve the crop have led to an increased focus on genetic research. We report the identification of 14,178 putative single nucleotide polymorphisms (SNPs using a genomic reduction protocol as well as the development of 511 functional SNP assays. The SNP assays are based on KASPar genotyping chemistry and were detected using the Fluidigm dynamic array platform. A diversity screen of 113 quinoa accessions showed that the minor allele frequency (MAF of the SNPs ranged from 0.02 to 0.50, with an average MAF of 0.28. Structure analysis of the quinoa diversity panel uncovered the two major subgroups corresponding to the Andean and coastal quinoa ecotypes. Linkage mapping of the SNPs in two recombinant inbred line populations produced an integrated linkage map consisting of 29 linkage groups with 20 large linkage groups, spanning 1404 cM with a marker density of 3.1 cM per SNP marker. The SNPs identified here represent important genomic tools needed in emerging plant breeding programs for advanced genetic analysis of agronomic traits in quinoa.

    Leaf rust (Puccinia triticina Eriks. & Henn.) is a major disease affecting durum wheat production. The Lr14a-resistant gene present in the durum wheat cv. Creso and its derivative cv. Colosseo is one of the best characterized leaf-rust resistance sources deployed in durum wheat breeding. Lr14a has been mapped close to the simple sequence repeat markers gwm146, gwm344 and wmc10 in the distal portion of the chromosome arm 7BL, a gene-dense region. The objectives of this study were: (1) to enrich the Lr14a region with single nucleotide polymorphisms (SNPs) and high-resolution melting (HRM)-based markers developed from conserved ortholog set (COS) genes and from sequenced Diversity Array Technology (DArT(®)) markers; (2) to further investigate the gene content and colinearity of this region with the Brachypodium and rice genomes. Ten new COS-SNP and five HRM markers were mapped within an 8.0 cM interval spanning Lr14a. Two HRM markers pinpointed the locus in an interval of HRM designed for agarose gel electrophoresis/KASPar(®) assays and high-resolution melting analysis, respectively, as well as the double-marker combinations ubw14/ubw18, ubw14/ubw35 and wPt-4038-HRM-ubw35 will be useful for germplasm haplotyping and for molecular-assisted breeding.

    Full Text Available Autosomal dominant polycystic kidney disease (ADPKD is the most common heritable kidney disease and is characterized by bilateral renal cysts. Hypertension is a frequent cause of chronic kidney disease (CKD and mortality in patients with ADPKD. The aldosterone synthase gene polymorphisms of the renin-angiotensin-aldosterone system have been extensively studied as hypertension candidate genes. The present study is aimed to investigate the potential modifier effect of CYP11B2 gene on the progression of CKD in ADPKD. One hundred and two ADPKD patients and 106 healthy controls were recruited based on Ravine inclusion and exclusion criteria. The three tag-SNPs within CYP11B2 gene (rs3802230, rs4543, and rs4544 were genotyped using FRET-based KASPar method. Cochran-Armitage trend test was used to assess the potential associations between these polymorphisms and CKD stages. Mantel- Haenszel stratified analysis was used to explore confounding and interaction effects of these polymorphisms. Of the three tag-SNPs genotyped, rs4544 polymorphism was monomorphic and rs3802230 deviated Hardy-Weinberg equilibrium. The CYP11B2 tag-SNPs did not show significant association with ADPKD or CKD. Further, these polymorphisms did not exhibit confounding effect on the relationship between CKD progression and hypertension. Our results suggest that aldosterone synthase gene is not a major susceptibility gene for progression of CKD in South Indian ADPKD patients.

  7. Associations between RET tagSNPs and their haplotypes and susceptibility, clinical severity, and thyroid function in patients with differentiated thyroid cancer.

    Full Text Available It is unclear whether common genetic variants of the RET proto-oncogene contribute to disease susceptibility, clinical severity, and thyroid function in differentiated thyroid cancer (DTC.A total of 300 DTC patients and 252 healthy controls were enrolled in this study. Seven RET tagging single nucleotide polymorphisms were genotyped using the KASPar platform.Subgroup analysis showed that concomitant thyroid benign diseases were less likely to occur in DTC subjects with the rs1799939 AG or AG plus AA genotypes (odds ratio (OR = 1.93 and 1.88, P = 0.009 and 0.011, respectively. A rare haplotype, CGGATAA, was associated statistically with a reduced risk of DTC (OR = 0.18, P = 0.001. Concerning the aggressive features of DTC, higher level of N stage was more likely to occur in subjects carrying the wild-type genotypes at rs1800860 site (for dominant model: OR = 0.48, P = 0.008. Another rare haplotype, CAAGCGT, conferred increased risk for the occurrence of distant metastasis (OR = 7.57, P = 0.009. Notably, higher thyroid stimulating hormone levels and lower parathyroid hormone levels were found in patients with rs2075912, rs2565200, and rs2742240 heterozygotes and rare homozygotes; similar results were observed between PTH levels and rs1800858.This study provided useful information on RET variants that should be subjected to further study.

    Full Text Available To estimate genetic diversity within and between 10 interfertile Cicer species (94 genotypes from the primary, secondary and tertiary gene pool, we analysed 5,257 DArT markers and 651 KASPar SNP markers. Based on successful allele calling in the tertiary gene pool, 2,763 DArT and 624 SNP markers that are polymorphic between genotypes from the gene pools were analyzed further. STRUCTURE analyses were consistent with 3 cultivated populations, representing kabuli, desi and pea-shaped seed types, with substantial admixture among these groups, while two wild populations were observed using DArT markers. AMOVA was used to partition variance among hierarchical sets of landraces and wild species at both the geographical and species level, with 61% of the variation found between species, and 39% within species. Molecular variance among the wild species was high (39% compared to the variation present in cultivated material (10%. Observed heterozygosity was higher in wild species than the cultivated species for each linkage group. Our results support the Fertile Crescent both as the center of domestication and diversification of chickpea. The collection used in the present study covers all the three regions of historical chickpea cultivation, with the highest diversity in the Fertile Crescent region. Shared alleles between different gene pools suggest the possibility of gene flow among these species or incomplete lineage sorting and could indicate complicated patterns of divergence and fusion of wild chickpea taxa in the past.

    Full Text Available Introduction: Autosomal dominant polycystic kidney disease (ADPKD is characterized by the presence of numerous cysts in the kidney and manifest with various renal and extra-renal complications leading to ESRD. Endothelin may contribute to various renal and extra-renal manifestations pointing to genetic and environmental modifying factors that alter the risk of developing chronic kidney disease (CKD in ADPKD. In the present study we investigated six genes coding for endothelin 1 (EDN1 tagging-single nucleotide polymorphisms (tag-SNPs to unravel the EDN1 gene modifier effect for renal disease progression in ADPKD. Materials and Methods: The tag-SNPs were genotyped using FRET-based KASPar method in 108 ADPKD patients and 119 healthy subjects. Cochran-Armitage trend test was used to determine the association between ADPKD and EDN1 tag-SNPs. Multivariate logistic regression analysis was performed to assess the effect of tag-SNPs on CKD progression. The relationship between different CKD stages and hypertension and their interaction Mantel-Haenszel stratified analysis was performed. Results: All loci are polymorphic and followed Hardy-Weinberg equilibrium. Distribution of EDN1 genotypes and haplotypes in control and ADPKD is not statistically significant. Five SNPs covering 3.4 kb forming single LD block, but the LD was not strong between SNPs. The EDN1 genotypes are not contributing to the CKD advancement among the ADPKD patients. Conclusion: These results suggest that the EDN1 gene is not a major modifier of CKD advancement among ADPKD patients.

    Full Text Available The platelet endothelial cell adhesion molecule 1 (PECAM-1 plays an important role in many inflammatory processes, including the development of atherosclerosis. Polymorphism rs668 of the PECAM-1 gene (373C/G is functional, and it was reported to be associated with increased serum levels of PECAM-1. We investigated the association between the rs668 polymorphism of PECAM-1 and subclinical markers of carotid atherosclerosis in subjects with type 2 diabetes mellitus (T2DM. Five hundred and ninety-five T2DM subjects and 200 control subjects were enrolled. The carotid intima-media thickness (CIMT and plaque characteristics (presence and structure were assessed ultrasonographically. Biochemical analyses were performed using standard biochemical methods. Geno-typing of the PECAM-1 gene polymorphism (rs668 was performed using KASPar assays. The control examinations were performed 3.8 ± 0.5 years after the initial examination. Higher CIMT was found in patients with T2DM in comparison with subjects without T2DM. Statistically sig-nificantly faster progression of the atherosclerotic markers was shown in subjects with T2DM in comparison with the control group. When adjusted to other risk factors, the rs668 GG genotype was associated with an increased risk of carotid plaques in subjects with T2DM. We concluded that our study demonstrated a minor effect of the rs668 PECAM-1 on markers of carotid atherosclerosis in subjects with T2DM.

  12. Drum-mate: interaction dynamics and gestures in human-humanoid drumming experiments

    This article investigates the role of interaction kinesics in human-robot interaction (HRI). We adopted a bottom-up, synthetic approach towards interactive competencies in robots using simple, minimal computational models underlying the robot's interaction dynamics. We present two empirical, exploratory studies investigating a drumming experience with a humanoid robot (KASPAR) and a human. In the first experiment, the turn-taking behaviour of the humanoid is deterministic and the non-verbal gestures of the robot accompany its drumming to assess the impact of non-verbal gestures on the interaction. The second experiment studies a computational framework that facilitates emergent turn-taking dynamics, whereby the particular dynamics of turn-taking emerge from the social interaction between the human and the humanoid. The results from the HRI experiments are presented and analysed qualitatively (in terms of the participants' subjective experiences) and quantitatively (concerning the drumming performance of the human-robot pair). The results point out a trade-off between the subjective evaluation of the drumming experience from the perspective of the participants and the objective evaluation of the drumming performance. A certain number of gestures was preferred as a motivational factor in the interaction. The participants preferred the models underlying the robot's turn-taking which enable the robot and human to interact more and provide turn-taking closer to 'natural' human-human conversations, despite differences in objective measures of drumming behaviour. The results are consistent with the temporal behaviour matching hypothesis previously proposed in the literature which concerns the effect that the participants adapt their own interaction dynamics to the robot's.

  14. Genetic polymorphisms in homologous recombination repair genes in healthy Slovenian population and their influence on DNA damage

    Homologous recombination (HR) repair is an important mechanism involved in repairing double-strand breaks in DNA and for maintaining genomic stability. Polymorphisms in genes coding for enzymes involved in this pathway may influence the capacity for DNA repair. The aim of this study was to select tag single nucleotide polymorphisms (SNPs) in specific genes involved in HR repair, to determine their allele frequencies in a healthy Slovenian population and their influence on DNA damage detected with comet assay. In total 373 individuals were genotyped for nine tag SNPs in three genes: XRCC3 722C>T, XRCC3 -316A>G, RAD51 -98G>C, RAD51 -61G>T, RAD51 1522T>G, NBS1 553G>C, NBS1 1197A>G, NBS1 37117C>T and NBS1 3474A>C using competitive allele-specific amplification (KASPar assay). Comet assay was performed in a subgroup of 26 individuals to determine the influence of selected SNPs on DNA damage. We observed that age significantly affected genotype frequencies distribution of XRCC3 -316A>G (P = 0.039) in healthy male blood donors. XRCC3 722C>T (P = 0.005), RAD51 -61G>T (P = 0.023) and NBS1 553G>C (P = 0.008) had a statistically significant influence on DNA damage. XRCC3 722C>T, RAD51 -61G>T and NBS1 553G>C polymorphisms significantly affect the repair of damaged DNA and may be of clinical importance as they are common in Slovenian population

  15. SNP discovery in the bovine milk transcriptome using RNA-Seq technology.

    High-throughput sequencing of RNA (RNA-Seq) was developed primarily to analyze global gene expression in different tissues. However, it also is an efficient way to discover coding SNPs. The objective of this study was to perform a SNP discovery analysis in the milk transcriptome using RNA-Seq. Seven milk samples from Holstein cows were analyzed by sequencing cDNAs using the Illumina Genome Analyzer system. We detected 19,175 genes expressed in milk samples corresponding to approximately 70% of the total number of genes analyzed. The SNP detection analysis revealed 100,734 SNPs in Holstein samples, and a large number of those corresponded to differences between the Holstein breed and the Hereford bovine genome assembly Btau4.0. The number of polymorphic SNPs within Holstein cows was 33,045. The accuracy of RNA-Seq SNP discovery was tested by comparing SNPs detected in a set of 42 candidate genes expressed in milk that had been resequenced earlier using Sanger sequencing technology. Seventy of 86 SNPs were detected using both RNA-Seq and Sanger sequencing technologies. The KASPar Genotyping System was used to validate unique SNPs found by RNA-Seq but not observed by Sanger technology. Our results confirm that analyzing the transcriptome using RNA-Seq technology is an efficient and cost-effective method to identify SNPs in transcribed regions. This study creates guidelines to maximize the accuracy of SNP discovery and prevention of false-positive SNP detection, and provides more than 33,000 SNPs located in coding regions of genes expressed during lactation that can be used to develop genotyping platforms to perform marker-trait association studies in Holstein cattle.

    Transcription factors are very diverse family of proteins involved in activating or repressing the transcription of a gene at a given time. Several studies using animal models demonstrated the role of transcription factor genes in craniofacial development. We aimed to investigate the association of IRF6 intron-6 polymorphism in the non-syndromic cleft lip with or without Palate in a south Indian population. 173 unrelated nonsyndromic cleft lip with or without Palate patients and 176 controls without clefts patients were genotyped for IRF6 rs2235375 variant by allele-specific amplification using the KASPar single nucleotide polymorphism genotyping system. The association between interferon regulatory factor-6 gene intron-6 dbSNP208032210:g.G>C (rs2235375) single nucleotide polymorphism and non-syndromic cleft lip with or without palate risk was investigated by chi-square test. There were significant differences in genotype or allele frequencies of rs2235375 single nucleotide polymorphism between controls and cases with non-syndromic cleft lip with or without palate. IRF6 rs2235375 variant was significantly associated with increased risk of non-syndromic cleft lip with or without palate in co-dominant, dominant (OR: 1.19; 95% CI 1.03-2.51; p=0.034) and allelic models (OR: 1.40; 95% CI 1.04-1.90; p=0.028). When subset analysis was applied significantly increased risk was observed in cleft palate only group (OR dominant: 4.33; 95% CI 1.44-12.97; p=0.005). These results suggest that IRF6 rs2235375 SNP play a major role in the pathogenesis and risk of developing non-syndromic cleft lip with or without palate.     Autosomal dominant polycystic kidney disease (ADPKD) is an inherited systemic disorder, characterized by the fluid filled cysts in the kidneys leading to end stage renal failure in later years of life. Hypertension is one of the major factors independently contributing to the chronic kidney disease (CKD) progression. The renin-angiotensin aldosterone system (RAAS) genes have been extensively studied as hypertension candidate genes. The aim of the present study was to investigate the role of angiotensin converting enzyme tagging - single nucleotide polymorphisms (ACE tag-SNPs) in progression of CKD in patients with ADPKD. m0 ethods: In the present study six ACE tagSNPs (angiotensin converting enzyme tag single nucleotide polymorphisms) and insertion/deletion (I/D) in 102 ADPKD patients and 106 control subjects were investigated. The tagSNPs were genotyped using FRET-based KASPar method and ACE ID by polymerase chain reaction (PCR) and electrophoresis. Genotypes and haplotypes were compared between ADPKD patients and controls. Univariate and multivariate logistic regression analyses were performed to assess the effect of genotypes and hypertension on CKD advancement. Mantel-Haenszel (M-H) stratified analysis was performed to study the relationship between different CKD stages and hypertension and their interaction. All loci were polymorphic and except rs4293 SNP the remaining loci followed Hardy-Weinberg equilibrium. Distribution of ACE genotypes and haplotypes in controls and ADPKD patients was not significant. A significant linkage disequilibrium (LD) was observed between SNPs forming two LD blocks. The univariate analysis revealed that the age, hypertension, family history of diabetes and ACE rs4362 contributed to the advancement of CKD. The results suggest that the ACE genotypes are effect modifiers of the relationship between hypertension and CKD advancement among the ADPKD patients.

    The famous German poet Johann Wolfgang Goethe (1749 - 1832) was a man of an outstanding interest for the Earth sciences. In the Czech geological dictionary his own biography remembers his frequent visits to the famous West Bohemian health resorts. In this region he was focusing his attention to the geological history, petrography and mineralogy, genesis of mineral water springs etc. Some of his studies were published. His geological points of view were not always correct (as seen from a recent knowledge) but his efforts to deepen studies of this territory cannot be forgotten. - In his rich correspondence with the count Kaspar Maria Sternberg (1761 - 1838) - founder of the (nowadays) National Museum in Prague - the author of this article has recently discovered in the Prague archives a letter written just one week before the death of the poet. It is a confession of his deep relation especially to the region if West Bohemia where he found lot of enjoyment and new knowledge in the course of numerous visits and stays. - Goethe had the largest private collection of minerals in all of Europe (17800 rock samples). A mineral goethite has been named after him. - The Czech composer Václav Jan Tomášek (1774 - 1850) describing his visit paid to Goethe in Cheb (Eger) in 1822 remembers also mineralogical interest of the poet and his excursions to the region for collecting local minerals. The main reason for personal contact in this case was the art (Tomášek composed songs using Goethe's poems). But Tomášek described also his frequent talks on science with the famous Swedish chemist Jöns Jacob Berzelius (1779 - 1848) in Karlsbad (1822). From other sources a common stay of Berzelius, Goethe and Sternberg in Marienbad (also 1822) is reported.

Acidophiles that thrive in these conditions must overcome significant challenges, including an environment with proton concentrations at pH 3 or below. The role of sulfur metabolism in the archaeon “Ferroplasma acidarmanus” strain fer1's ability to thrive in this environment was investigated due to its growth-dependent production of methanethiol, a volatile organic sulfur compound. Two putative sequences for methionine-γ-lyase (EC 4.4.1.11, an enzyme known to carry out α, γ-elimination on L-methionine to produce methanethiol, were identified in fer1. Bioinformatic analyses identified a conserved pyridoxal-5'-phosphate (PLP binding domain and a partially conserved catalytic domain in both putative sequences. Detection of PLP-dependent and L-methionine-dependent production of α-keto compounds and thiol groups in fer1 confirmed the presence of methionine-γ-lyase activity. Further, fer1 lysate was capable of processing related substrates, including D-methionine, L-cysteine, L-cystathionine, and L/D-homocysteine. When the two putative fer1 methionine-γ-lyase gene-coded proteins were expressed in Escherichia coli cells, one sequence demonstrated an ability to carry out α, γ-elimination activity, while the other exhibited γ-replacement activity. These fer1 methionine-γ-lyases also exhibited optimum pH, substrate specificity, and catalytic preferences that are different from methionine-γ-lyases from other organisms.     The 2017 ABoVE Airborne Campaign (AAC) was one of the largest airborne experiments ever conducted by NASA's Earth Science Division. It involved nine aircraft in 17 deployments - more than 100 flights - between April and October and sampled over 4 million km2in Alaska and northwestern Canada. Many of these flights were coordinated with detailed, same-day ground-based measurements to link field-based, process-level studies with geospatial data products derived from satellite remote sensing. A major goal of the 2017 AAC was to collect data that spanned the critical intermediate space and time scales that are essential for a comprehensive understanding of scaling issues across the ABoVE Study Domain and extrapolation to the pan-Arctic. Additionally, the 2017 AAC provided unique opportunities to validate satellite and airborne remote sensing data for northern high latitude ecosystems, develop and advance fundamental remote sensing science, and explore scientific insights from innovative sensor combinations. The 2017 AAC science strategy coupled domain-wide sampling with L-band and P-band synthetic aperture radar (SAR), imaging spectroscopy (AVIRIS-NG), full waveform lidar (LVIS) and atmospheric carbon dioxide and methane with more spatially and temporally focused studies using Ka-band SAR (Ka-SPAR) and solar induced chlorophyll fluorescence (CFIS). Additional measurements were coordinated with the NEON Airborne Observing Platform, the ASCENDS instrument development suite, and the ATOM EV-S2 investigation.   2. Gene-based single nucleotide polymorphism markers for genetic and association mapping in common bean.

    In common bean, expressed sequence tags (ESTs) are an underestimated source of gene-based markers such as insertion-deletions (Indels) or single-nucleotide polymorphisms (SNPs). However, due to the nature of these conserved sequences, detection of markers is difficult and portrays low levels of polymorphism. Therefore, development of intron-spanning EST-SNP markers can be a valuable resource for genetic experiments such as genetic mapping and association studies. In this study, a total of 313 new gene-based markers were developed at target genes. Intronic variation was deeply explored in order to capture more polymorphism. Introns were putatively identified after comparing the common bean ESTs with the soybean genome, and the primers were designed over intron-flanking regions. The intronic regions were evaluated for parental polymorphisms using the single strand conformational polymorphism (SSCP) technique and Sequenom MassARRAY system. A total of 53 new marker loci were placed on an integrated molecular map in the DOR364 × G19833 recombinant inbred line (RIL) population. The new linkage map was used to build a consensus map, merging the linkage maps of the BAT93 × JALO EEP558 and DOR364 × BAT477 populations. A total of 1,060 markers were mapped, with a total map length of 2,041 cM across 11 linkage groups. As a second application of the generated resource, a diversity panel with 93 genotypes was evaluated with 173 SNP markers using the MassARRAY-platform and KASPar technology. These results were coupled with previous SSR evaluations and drought tolerance assays carried out on the same individuals. This agglomerative dataset was examined, in order to discover marker-trait associations, using general linear model (GLM) and mixed linear model (MLM).   3. Genetic risk analysis of coronary artery disease in Pakistani subjects using a genetic risk score of 21 variants.

    Conventional coronary artery disease (CAD) risk factors like age, gender, blood lipids, hypertension and smoking have been the basis of CAD risk prediction algorithms, but provide only modest discrimination. Genetic risk score (GRS) may provide improved discrimination over and above conventional risk factors. Here we analyzed the genetic risk of CAD in subjects from Pakistan, using a GRS of 21 variants in 18 genes and examined whether the GRS is associated with blood lipid levels. 625 (405 cases and 220 controls) subjects were genotyped for variants, NOS3 rs1799983, SMAD3 rs17228212, APOB rs1042031, LPA rs3798220, LPA rs10455872, SORT1 rs646776, APOE rs429358, GLUL rs10911021, FTO rs9939609, MIA3 rs17465637, CDKN2Ars10757274, DAB2IP rs7025486, CXCL12 rs1746048, ACE rs4341, APOA5 rs662799, CETP rs708272, MRAS rs9818870, LPL rs328, LPL rs1801177, PCSK9 rs11591147 and APOE rs7412 by TaqMan and KASPar allele discrimination techniques. Individually, the single SNPs were not associated with CAD except APOB rs1042031 and FTO rs993969 (p = 0.01 and 0.009 respectively). However, the combined GRS of 21 SNPs was significantly higher in cases than controls (19.37 ± 2.56 vs. 18.47 ± 2.45, p = 2.9 × 10 -5 ), and compared to the bottom quintile, CAD risk in the top quintile of the GRS was 2.96 (95% CI 1.71-5.13). Atherogenic blood lipids showed significant positive association with GRS. The GRS was quantitatively associated with CAD risk and showed association with blood lipid levels, suggesting that the mechanism of these variants is likely to be, in part at least, through creating an atherogenic lipid profile in subjects carrying high numbers of risk alleles.   4. Genetic variants in PARP1 (rs3219090) and IRF4 (rs12203592) genes associated with melanoma susceptibility in a Spanish population

    Few high penetrance genes are known in Malignant Melanoma (MM), however, the involvement of low-penetrance genes such as MC1R, OCA2, ASIP, SLC45A2 and TYR has been observed. Lately, genome-wide association studies (GWAS) have been the ideal strategy to identify new common, low-penetrance susceptibility loci. In this case–control study, we try to validate in our population nine melanoma associated markers selected from published GWAS in melanoma predisposition. We genotyped the 9 markers corresponding to 8 genes (PARP1, MX2, ATM, CCND1, NADSYN1, CASP8, IRF4 and CYP2R1) in 566 cases and 347 controls from a Spanish population using KASPar probes. Genotypes were analyzed by logistic regression and adjusted by phenotypic characteristics. We confirm the protective role in MM of the rs3219090 located on the PARP1 gene (p-value 0.027). Additionally, this SNP was also associated with eye color (p-value 0.002). A second polymorphism, rs12203592, located on the IRF4 gene was associated with protection to develop MM for the dominant model (p-value 0.037). We have also observed an association of this SNP with both lentigines (p-value 0.014) and light eye color (p-value 3.76 × 10 -4 ). Furthermore, we detected a novel association with rs1485993, located on the CCND1 gene, and dark eye color (p-value 4.96 × 10 -4 ). Finally, rs1801516, located on the ATM gene, showed a trend towards a protective role in MM similar to the one firstly described in a GWAS study. To our knowledge, this is the first time that these SNPs have been associated with MM in a Spanish population. We confirmed the proposed role of rs3219090, located on the PARP1 gene, and rs12203592, located on the IRF4 gene, as protective to MM along the same lines as have previous genome-wide associated works. Finally, we have seen associations between IRF4, PARP1, and CCND1 and phenotypic characteristics, confirming previous results for the IRF4 gene and presenting novel data for the last two, suggesting that

A total of 75 patients with venous leg ulcers extensively covered with fibrin and necrotic tissue were randomized to a PA-containing hydrogel or an amorphous hydrogel. Wounds were treated for 14 days and costs were estimated from the German payer's perspective. Medical costs included study treatment, wound treatment supply, and labor time. The clinical benefit was expressed as the number of patients with wounds >50% covered with granulation tissue within 14 days. The incremental cost-effectiveness ratio (ICER was expressed as the additional cost spent with >50% granulation tissue per day per patient within 14 days of leg ulcer care.Results: Because of individual pricing of wound dressings in hospitals, cost data were derived from the outpatient sector. A total of 33 patients were treated using the PA-based hydrogel and 37 patients using the amorphous hydrogel. The estimated total direct costs per patient and per 14 days of therapy were €306 for both treatment groups. However, with the PA-based hydrogel, 2.5 additional days with wounds covered >50% with granulation tissues were gained within 14 days of leg ulcer care compared to the comparator. The ICER was €0 per additional day spent with >50% granulation tissue.Conclusion: Although there were a greater number of dressing changes in the PA-based hydrogel treatment, the total treatment cost for 14 days of leg ulcer care was the same for both

  6. SNP Discovery for mapping alien introgressions in wheat

    Background Monitoring alien introgressions in crop plants is difficult due to the lack of genetic and molecular mapping information on the wild crop relatives. The tertiary gene pool of wheat is a very important source of genetic variability for wheat improvement against biotic and abiotic stresses. By exploring the 5Mg short arm (5MgS) of Aegilops geniculata, we can apply chromosome genomics for the discovery of SNP markers and their use for monitoring alien introgressions in wheat (Triticum aestivum L). Results The short arm of chromosome 5Mg of Ae. geniculata Roth (syn. Ae. ovata L.; 2n = 4x = 28, UgUgMgMg) was flow-sorted from a wheat line in which it is maintained as a telocentric chromosome. DNA of the sorted arm was amplified and sequenced using an Illumina Hiseq 2000 with ~45x coverage. The sequence data was used for SNP discovery against wheat homoeologous group-5 assemblies. A total of 2,178 unique, 5MgS-specific SNPs were discovered. Randomly selected samples of 59 5MgS-specific SNPs were tested (44 by KASPar assay and 15 by Sanger sequencing) and 84% were validated. Of the selected SNPs, 97% mapped to a chromosome 5Mg addition to wheat (the source of t5MgS), and 94% to 5Mg introgressed from a different accession of Ae. geniculata substituting for chromosome 5D of wheat. The validated SNPs also identified chromosome segments of 5MgS origin in a set of T5D-5Mg translocation lines; eight SNPs (25%) mapped to TA5601 [T5DL · 5DS-5MgS(0.75)] and three (8%) to TA5602 [T5DL · 5DS-5MgS (0.95)]. SNPs (gsnp_5ms83 and gsnp_5ms94), tagging chromosome T5DL · 5DS-5MgS(0.95) with the smallest introgression carrying resistance to leaf rust (Lr57) and stripe rust (Yr40), were validated in two released germplasm lines with Lr57 and Yr40 genes.     Full Text Available Abstract Background Variations within the FOXA family have been studied for a putative contribution to the risk of type 2 diabetes (T2D, and recently the minor T-allele of FOXA2 rs1209523 was reported to associate with decreased fasting plasma glucose levels in a study using a weighted false discovery rate control procedure to enhance the statistical power of genome wide association studies in detecting associations between low-frequency variants and a given trait. Thus, the primary aim of this study was to investigate whether the minor T-allele of rs1205923 in FOXA2 associated with 1 decreased fasting plasma glucose and 2 a lower risk of developing T2D. Secondly, we investigated whether rs1205923 in FOXA2 associated with other glucose-related phenotypes. Methods The variant was genotyped in Danish individuals from four different study populations using KASPar® PCR SNP genotyping system. We examined for associations of the FOXA2 genotype with fasting plasma glucose and estimates of insulin release and insulin sensitivity following an oral glucose tolerance test in 6,162 Danish individuals from the population-based Inter99 study while association with T2D risk was assessed in 10,196 Danish individuals including four different study populations. Results The FOXA2 rs1209523 was not associated with fasting plasma glucose (effect size (β = -0.03 mmol/l (95%CI: -0.07; 0.01, p = 0.2 in glucose-tolerant individuals from the general Danish population. Furthermore, when employing a case-control setting the variant showed no association with T2D (odds ratio (OR = 0.82 (95%CI: 0.62-1.07, p = 0.1 among Danish individuals. However, when we performed the analysis in a subset of 6,022 non-obese individuals (BMI 2 an association with T2D was observed (OR = 0.68 (95%CI: 0.49-0.94, p = 0.02. To elucidate potential underlying phenotypes we aimed to investigate whether these variants associated with: 1 quantitative metabolic traits, 2 anthropometric measures (waist circumference (WC, waist-hip ratio, and BMI, or 3 type 2 diabetes, and central and general overweight and obesity.The four variants were genotyped in Danish individuals using KASPar®. Quantitative metabolic traits were examined in a population-based sample (n = 6,038 and WC and BMI were furthermore analyzed in a combined study sample (n = 13,507. Case-control studies of diabetes and adiposity included 15,326 individuals. The major G-allele of LYPLAL1 rs2605100 associated with increased fasting serum triglyceride concentrations (per allele effect (β = 3%(1;5(95%CI, p(additive = 2.7×10(-3, an association driven by the male gender (p(interaction = 0.02. The same allele associated with increased fasting serum insulin concentrations (β = 3%(1;5, p(additive = 2.5×10(-3 and increased insulin resistance (HOMA-IR (β = 4%(1;6, p(additive = 1.5×10(-3. The minor G-allele of rs10146997 in NRXN3 associated with increased WC among women (β = 0.55cm (0.20;0.89, p(additive = 1.7×10(-3, p(interaction = 1.0×10(-3, but showed no associations with obesity related metabolic traits. The MSRA rs545854 and TFAP2B rs987237 showed nominal associations with central obesity; however, no underlying metabolic phenotypes became obvious, when investigating quantitative metabolic traits.     Financing new nuclear power plants in a deregulated market was the issue addressed by the 2010 conference of the Swiss Association for Energy Economics. Stefan Hirschberg of the Paul Scherrer Institute first gave an overview of the internal and external costs of nuclear and other electricity supply technologies. Vincent Rits of the Prognos company presented a worldwide review of the current status of nuclear energy for power production and its prospects, showing that despite the construction of plants at new locations the total installed power will not be significantly different from today's figure, i.e. of the order of 400 GW. This even if the plants built in the 70es and 80es are systematically replaced at the end of their life time by new ones in the next decades. Boris Zuercher of the Avenir Suisse 'think tank' raised the question: 'Are nuclear power plants too big to fail?'. Using arguments from the lessons recently learned from the worldwide financial crisis, he concluded his analysis by pointing out the fact that electricity utilities in Switzerland have already been bailed out by the state, because they are owned by the regional governments and the big cities, even though they are operating as private companies in a (soon completely) deregulated market. Risk management is missing, so Zuercher, and the risks are covered by the state. Benoit Gaillochet of Axa Private Equity presented an appraisal of investments in nuclear power projects by a global financial player. For him, a number of key issues need to be addressed in order to make nuclear projects eligible for infrastructure investors; in particular, the long-term economic viability of these projects is critical. Kaspar Mueller from the Ellipson company critically evaluated future investments in nuclear energy from the financial point of view.     Full Text Available The paper gives an overview of a network of authors who have been active since the end of the 20th century, mainly (but not only in Tartu. They can be characterized as having an experimental attitude and maintaining distance from the literary mainstream, but at the same time they do not try to be consciously polemical or novatory, because partly they repeat the techniques of the 20th century avant-garde and, what is maybe more important, they sidestep the dialectics of innovating or altering the literary canon. The network is not strictly organized; it is rather a rhizome that has some denser nodes (literary journals and mailing lists. There have been different attempts to name the phenomenon: Tartu avant-garde, microcosmic literature, cognitive realism, Y-literature, "eksp" and so on. I have chosen "paranoiac-critique" as the term that best captures the common characteristics of these authors and their artistic practices (some of the authors are active not only as text writers, but also as conceptual artists.The paper then gives a list, with commentary, of the most important groups and individuals of that movement. In the role of "predecessors" are the group 14NÜ (Paavo Matsin, Mait Laas, Marianne Ravi et al., which also experimented with the format of the book, and a company in Tartu that developed a specific actionist stylistics called "mugiv" (Marko Kompus, Mehis Heinsaar, Martiini, Kaspar et al.. The sovereign figure in the experimental network is Valdur Mikita, who has also academically researched the problems of creativity; he is also a theoretician and metapoet. An important institution for the network has been the literary journal Vihik (initiator Berk Vaher; the later editor of the journal Jaak Tomberg has also participated in activities of the Tartu Theatre Laboratory (managed by Andreas W, which has experimented with "technological theatre".
