    To construct the prokaryotic expression vector of phytoplasma immunodominant membrane protein A (IdpA) in prokaryotic cell, express and purify the IdpA and prepare its antiserum. With the recombinant plasmid pMD18-T-IdpA as templates, IdpA gene was amplified by PCR and cloned into prokaryotic expression vector pET-28a(+) by endonuclease reaction and T4 DNA ligase reaction. Then the recombinant plasmid pET-28a(+)-IdpA was transformed into E.coli BL21 (DE3). After confirmed by PCR and double enzyme digestion, the recombinant protein IdpA was expressed under IPTG induction and purified. The purified product was used to immunize BALB/c mice to prepare its antiserum. IdpA-specific mouse antiserum was identified by ELISA and Westerrn blotting. The prokaryotic vectors of pET-28a(+)-IdpA were constructed successfully and the recombinant protein IdpA was induced to express stably in the E.coli BL21. The purity of IdpA was up to over 90%. In the BALB/c mice immunized by the purified IdpA, the titre of IdpA-specific antiserum was as high as 1:320 000. The recombinant protein IdpA was expressed successfully in E.coli and the IdpA-specific antiserum was prepared.

    Full Text Available O objetivo foi analisar a influência da barragem de Tucuruí no desempenho da pesca artesanal, desenvolvida a jusante. Partindo-se da hipótese de que o desequilíbrio do rio Tocantins repercutiu de forma intensa sobre a vida das comunidades humanas locais, afetando o desempenho da atividade pesqueira, procurou-se dar resposta para a seguinte questão: qual a influência da barragem de Tucuruí, estado do Pará, sobre o desempenho da pesca artesanal desenvolvida a jusante? Os dados utilizados são de origem primária e secundária. Utilizou-se o modelo de análise fatorial exploratória (AFE, para construir o Índice de Desempenho da Pesca Artesanal (IDPA. Os resultados mostram que 21% dos pescadores têm baixo desempenho e 79%, desempenho intermediário, fato que aponta para a difícil situação enfrentada pelos trabalhadores da pesca ao longo do tempo.

    Full Text Available Central nervous system (CNS tuberculosis (TB is the most severe form of extra-pulmonary TB and disproportionately affects young children where the developing brain has a unique host response. New Zealand white rabbits were infected with Mycobacterium tuberculosis via subarachnoid inoculation at postnatal day 4-8 and evaluated until 4-6 weeks post-infection. Control and infected rabbit kits were assessed for the development of neurological deficits, bacterial burden, and postmortem microbiologic and pathologic changes. The presence of meningitis and tuberculomas was demonstrated histologically and by in vivo magnetic resonance imaging (MRI. The extent of microglial activation was quantified by in vitro immunohistochemistry as well as non-invasive in vivo imaging of activated microglia/macrophages with positron emission tomography (PET. Subarachnoid infection induced characteristic leptomeningeal and perivascular inflammation and TB lesions with central necrosis, a cellular rim and numerous bacilli on pathologic examination. Meningeal and rim enhancement was visible on MRI. An intense microglial activation was noted in M. tuberculosis-infected animals in the white matter and around the TB lesions, as evidenced by a significant increase in uptake of the tracer 124I-DPA-713, which is specific for activated microglia/macrophages, and confirmed by quantification of Iba-1 immunohistochemistry. Neurobehavioral analyses demonstrated signs similar to those noted in children with delayed maturation and development of neurological deficits resulting in significantly worse composite behavior scores in M. tuberculosis-infected animals. We have established a rabbit model that mimics features of TB meningitis in young children. This model could provide a platform for evaluating novel therapies, including host-directed therapies, against TB meningitis relevant to a young child's developing brain.

  5. Geometric Reconstruction of Bedrock and Overlying Recent Deposits In An Intra-mountain Basin: The Clusone Basin (southern Alps, Italy)

    Regione Lombardia (Direzione Generale Territorio e Urbanistica) and the National Research Council (CNR-IDPA Milano) acquired seismic reflection profiles in the Clu- sone basin (Middle Val Seriana, Southern Alps). In the study area, the bedrock is rep- resented by late Triassic carbonate units (Formazione di Castro, Dolomia Principale and coeval basinal facies, bordered northward by an important alpine fault) covered by a large amount of recent deposits that covers an area of more than 10 km2, with a maximum thickness of more than two hundreds meters, as documented by available well data. The aim of the seismic prospecting was to identify the sediments layering and the rock basement depth. The acquisition parameters were as follows: group in- terval 10 m; shot interval 5 m; geophone frequency 14 Hz; sample rate 1 ms; record length 2 s, energy source hydrapulse. The cable, with 120 channels, remained dur- ing all the experiment allowing reflection/refraction events acquisition. The data were processed by a standard procedure using PROMAX and SUNT5 processing codes. The statics were calculated starting from the refracted first arrivals using a two layer inversion based on least square optimisation. Standard seismic reflection processing was applied to obtain reflection images and it was integrated with seismic refraction data inversion. Seismic profiles allow to reconstruct both the main reflectors in the recent deposits and the geometry of the bedrock. The first results document a complex history in the drainage patterns of the Clusone basin, allowing to identify, in an intra- mountain basin, drainage directions that in some cases are different from the ones that can be observed today. The integration of well data and seismic profiles in this study of an intra-mountain basin allows on one side the identification of the bedrock geome- tries and, on the other, gives constrains for the reconstruction of the geomorphologic evolution of a sector of a mountain chain.

    were conducted on the Kilimanjaro Northern Ice Field (NIF3) ice core at the Institute for the Dynamics of Environmental Processes-CNR (IDPA-CNR) resulting in concentrations up to 96 pg/mL, demonstrating the repeatability of the results and the applicability of the method to tropical ice. These results allow for the creation of a comprehensive decadal to centennial history of African savanna fires over the past 4000 years using levoglucosan as an atmospheric molecular tracer for biomass burning. We integrate a record of organic compounds with traditional ice core analyses of ionic species and dust to expand the current understanding of the climate system with implications for the global carbon cycle.
