  1. Inhibition of spring viraemia of carp virus replication in an Epithelioma papulosum cyprini cell line by RNAi.

    Spring viraemia of carp virus (SVCV) is an aetiological agent of a serious disease affecting carp farms in Europe and is a member of the Rhabdoviridae family of viruses. The genome of SVCV codes for five proteins: nucleoprotein (N), phosphoprotein (P), matrix protein (M), glycoprotein (G) and RNA-dependent RNA polymerase (L). RNA-mediated interference (RNAi) by small interfering RNAs (siRNAs) is a powerful tool to inhibit gene transcription and is used to study genes important for viral replication. In previous studies regarding another member of Rhabdoviridae, siRNA inhibition of the rabies virus nucleoprotein gene provided in vitro and in vivo protection against rabies. In this study, synthetic siRNA molecules were designed to target SVCV-N and SVCV-P transcripts to inhibit SVCV replication and were tested in an epithelioma papulosum cyprini (EPC) cell line. Inhibition of gene transcription was measured by real-time quantitative reverse-transcription PCR (RT-qPCR). The efficacy of using siRNA for inhibition of viral replication was analysed by RT-qPCR measurement of a reporter gene (glycoprotein) expression and by virus endpoint titration. Inhibition of nucleoprotein and phosphoprotein gene expression by siRNA reduced SVCV replication. However, use of tandem siRNAs that target phosphoprotein and nucleoprotein worked best at reducing SVCV replication. © 2014 The Authors. Journal of Fish Diseases published by John Wiley & Sons Ltd.

This number was refined to 40 cases de novo BCE in children and teenagers.     Thirty patients with basal cell epithelioma received high energy electron beam therapy. They were irradiated with a dose ranging from 4,800 rad (24 fractions, 35 days) to 12,000 rad (40 fractions, 57 days). Tumors disappeared in all cases. These were no disease-related deaths; in one patient there was recurrence after 2 years. We conclude that radiotherapy with high energy electron beam is very effective in the treatment of basal cell epithelioma.

    A 81-year-old male had perioral radiodermatitis of 50 years' duration which was associated with Bowen's disease and basal cell epitheliomas since age 59 years. One of those basal cell epitheliomas treated with topical 5-FU and bleomycin ointments increased to form an ulcer of 25 x 15mm in size nearby the right side of nose, accompanying with a fistule to the oral cavity while he hesitated to visit the hospital. 5-FU was intralesionally injected into the tumor. After the injections in total dose of 6,550mg the ulcer got epithelized and the biopsy could not reveal the tumor cell. The case proves the effectiveness of intralesional injection of 5-FU for basal cell epithelioma which avoids the surgical excision.

    Full Text Available Basal cell epithelioma is one of the most frequent malignant skin neoplasms. Angiogenesis plays an important part in the development of basal cell epithelioma. The article presents a review of the angiogenesis of this tumor with the help the immunohistochemistry analysis using CD31 and CD34 markers. The authors established a reliable relationship between the number of vessels expressing CD31 and those expressing CD34 in the superficial and nodular forms of the tumor as well as superficial, multi-center, nodular and infiltrative basal cell epitheliomas. A comparison of the number of vessels expressing CD31 and CD34 markers in different histological types made revealed a trend to their growth in the infiltrative type vs. superficial, multi-center and nodular ones.

    Full Text Available Calcifying epithelioma, a benign tumor derived from the hair apparatus and consisted of hair matrix cells, is relatively prevalent in females. We report a case of right preauricular calcifying epithelioma that was incidentally detected at the examination of multiple facial fractures and became an old lesion without symptoms for 40 years. The patient who was a 42-year-old male visited our department for the first time in October 2011 with a chief complaint of multiple facial fractures. Radiographic imaging demonstrated fracture lines at the anterior and posterior walls of the left maxillary sinus and zygomatic arch and revealed a mass at a right preauricular area. The extraction was performed under general anaesthesia. No recurrence has been observed 15 months after surgery. We also reviewed the literature of calcifying epithelioma.

    A woman 63 years old had received an unknown amount of roentgen rediation from a physician for spondylitis tuberculosa of the thoracic vertebrae fifty years ago. About five years ago two small brownish black tumors appeared on her back and gradually increased. Within the past month ulcer and bleeding in these tumors. The Patient presented a chronic radiodermatitis (so-called roentgen-skin) of the interscapular space. In addition, two tumors were present in the upper and lower parts of the roentgen-skin. The upper tumor revealed adenoid basal cell epithelioma and the lower tumor was pigmented solid basal cell epithelioma. A view on the development of basal cell epitheliomas on the roentgen-skin was assumed as follows: When epidermal and adnexal cells suffer from a certain damage by X-ray, especially soft X-ray, these cells may become modulated cells which have the same functions as adult immature pluripotential cells. From these modulated cells, basal cell epitheliomas may develop. (author)

    This report deals with a basal cell epithelioma, partially adenoid and partially morphea-like in structure, which despite intensive X-ray treatment relapsed constantly and which finally developed into an ulcus terebrans. Approximately 13 years after the primary tumor had developed (located on the left wing of the nose) both a lymphogenic and a hematogenic formation of metastases occurred with a subsequent exitus letalis 4 months later. Besides the metastases of the skin, there were multiple metastases in the lymph nodes, vertebral column, ribs, spleen, liver, stomach, pleura, and peritoneum as well as in the myocard of both ventricles and in the perimysium of the skeletal muscles. Their histological structure was similar to a partly adenoid, partily morphea-like basal cell epithelioma. The possible influence of X-ray treatment on the tumor tissue in way of benignity or malignancy is discussed in view of relevant literature on this topic.     Full Text Available Aim. To describe clinical manifestations of rare eyelid tumor (epithelioma Malherbe and to improve differential diagnosis of benign eyelid tumors in children. Patients and methods. We observed 8 children aged 3,5‑8 years (sex ratio was 1:1. In all cases, examination, palpation, surgical excision of the tumor with histological examination were performed. Results. Trichilemmoma, or pilomatricoma, was suggested from clinical manifestations. Epithelioma Malherbe was diagnosed by histology only. Microscopically, the tumor is surrounded by a capsule which includes two cell types. Peripheral basophilic cells are small cells with poor cytoplasm, indistinct borders, and deeply basophilic nucleus. Central shadow cells have a distinct border and a central unstained area. Islands of small basaloid epithelial cells with squamous cell focuses and cornification are embedded in the stroma. Epithelial lesions are often necrotized. Epithelial mass is surrounded by granulations with giant cells. Osseous trabeculae are often adjacent to necrotic lesions. Further follow-up revealed no complications or recurrences. Conclusions. Our observations and literature data suggest that epithelioma Malherbe is occured in 1.3 % of benign eyelid tumors in childern. Tumor growth is slow and non-invasive.

A 43-year-old Japanese male presented with chronic radiodermatitis on both dorsal surfaces of his fingers, following a long-term irradiation by Grenz rays for the treatment of psoriasis vulgaris. At the same site, he developed squamous cell carcinoma and Bowen's disease 7 years after the last irradiation. Three years later, he noticed a black papule on the dorsal surface of his right middle finger. After a histological examination, this papule diagnosed to be basal cell epithelioma. (author)

    Full Text Available The present paper deals with a clinico-pathological analysis of 172 cases of basal cell epithelioma representing 28% of the total skin cancer case. 1.44% of the total malignant tumours, 1% of the total tumours and 0.24% of the hospital admissions. M:F ratio was 1.26:1. The mean age was 54.06 years with the maximum number of cases in the fifth decade., Most (94% of the lesions were seen on the head and neck region. Histopathologically it showed solid (60.5%, adenoid cystic (15,7%, keratotic (9.3%, pigmented (6.4%, basi-squamous (3.5% and morphoea patterns (2.3%. Perineural lymphatic invasion was seen in 52% of the adenoid cystic basal call epithelioma. Amyloid (12.8% and amyloid like deposits (29.65% were seen in 42% of the case. Amyloid deposits were seen as lichen amyloidosis in 10.46% cases and in or around the tumour cell masses in 16.8% cases. It appeared as structureless homogeneous eosinophilic material and showed congophilia with apple green birefringence on polarising microscopy. It was found to be KMN04 sensitive (AA type protein- secondary amyloidosis. Amyloid like deposits appeared as less homogeneous,, basophilic, fractured material which showed non-specific congophilia with apple- green birefringence and stained with acid orcein stain indicating degenerated collagen (elastoid-degeneration.

    The therapeutic results are presented in cases of basocellular epithelioma treated by three methods. The best and most radical results were obtained by the surgical method, followed in the order of effectiveness by radiotherapy (45-55 kV) in a total dose of 4500-6000 R. In case of contraindications to these methods local chemotherapy was applied which was associated with a high proportion of failures (28.6%).

    Bone scintigraphy has been considerably improved by the use of technetium 99m-labelled organic phosphates: the physical characteristics are almost ideal for detection and the extemporaneous labelling of the product is convenient. This study on 75 patients has proved the value of bone scintigraphy in evaluating the bone invasion of tumoral lesions affecting the face and skull. An 88% agreement was found for scintigraphy, radiography giving 81%. The technique allows a fast assessment of whole-body bone invasion from one examination, with no extra injection of radioactive product, and it is therefore possible to orient the radiological enquiry without having to X-ray the whole skeleton, which reduces the amount of irradiation delivered to the organism. From these observations it may be concluded that scintigraphy is a simple, harmless method, with many advantages, but its limits must be known and it must always be interpreted as a function of other clinical and radiological data.     A 32-year-old woman in whom basal cell epithelioma (BCE) arose on the P-32 irradiated area is reported. At the age of 2, the patient had received superficial irradiation with P-32 for hemangioma simplex on the right precordial site. A black, ill-defined mass has occurred and gradually increased 3 years earlier. Chronic radiodermatitis characterized by teleangiectasis and skin atrophy was seen in the area surrounding the mass, which corresonded to that exposed to P-32. Histopathological examination leads to the diagnosis of BCE developing in part of chronic radiodermatitis. Review of the literature shows 3 cases of squamous cell carcinoma of the skin resulting from P-32 therapy. This is the first report on a patient with BCE resulting from irradiation with P-32.     A 32-year-old woman in whom basal cell epithelioma (BCE) arose on the P-32 irradiated area is reported. At the age of 2, the patient had received superficial irradiation with P-32 for hemangioma simplex on the right precordial site. A black, ill-defined mass has occurred and gradually increased 3 years earlier. Chronic radiodermatitis characterized by teleangiectasis and skin atrophy was seen in the area surrounding the mass, which corresonded to that exposed to P-32. Histopathological examination leads to the diagnosis of BCE developing in part of chronic radiodermatitis. Review of the literature shows 3 cases of squamous cell carcinoma of the skin resulting from P-32 therapy. This is the first report on a patient with BCE resulting from irradiation with P-32. (N.K.)

  10. Isolation and characterization of a rhabdovirus from starry flounder (Platichthys stellatus) collected from the northern portion of Puget Sound, Washington, USA

    The initial characterization of a rhabdovirus isolated from a single, asymptomatic starry flounder (Platichthys stellatus) collected during a viral survey of marine fishes from the northern portion of Puget Sound, Washington, USA, is reported. Virions were bullet-shaped and approximately 100 nm long and 50 nm wide, contained a lipid envelope, remained stable for at least 14 days at temperatures ranging from -80 to 5 degrees C and grew optimally at 15 degrees C in cultures of epithelioma papulosum cyprini (EPC) cells. The cytopathic effect on EPC cell monolayers was characterized by raised foci containing rounded masses of cells. Pyknotic and dark-staining nuclei that also showed signs of karyorrhexis were observed following haematoxylin and eosin, May-Grunwald Giemsa and acridine orange staining. PAGE of the structural proteins and PCR assays using primers specific for other known fish rhabdoviruses, including Infectious hematopoietic necrosis virus, Viral hemorrhagic septicemia virus, Spring viremia of carp virus, and Hirame rhabdovirus, indicated that the new virus, tentatively termed starry flounder rhabdovirus (SFRV), was previously undescribed in marine fishes from this region. In addition, sequence analysis of 2678 nt of the amino portion of the viral polymerase gene indicated that SFRV was genetically distinct from other members of the family Rhabdoviridae for which sequence data are available. Detection of this virus during a limited viral survey of wild fishes emphasizes the void of knowledge regarding the diversity of viruses that naturally infect marine fish species in the North Pacific Ocean.

    The bystander response has been documented in cell lines and cell cultures derived from aquatic species over the past several years. However, little work has been undertaken to identify a similar bystander response in tissue explant cultures from fish. In this study, indirect effects of ionizing gamma radiation on tissue explant cultures of fish were investigated. Tissue explants in culture were exposed to 0.5 Gy and 5 Gy gamma radiation from a 60Co teletherapy unit. A bystander response in Epithelioma papulosum cyprini (EPC) cells exposed to gamma-irradiated tissue conditioned medium from rainbow trout explants was investigated, and the effects on cell survival were quantified by the clonogenic survival assay. Dichlorofluorescein and rhodamine 123 fluorescent dyes were used to identify alterations in reactive oxygen species (ROS) and mitochondrial membrane potential (MMP), respectively. Results indicate a different response for the three tissue types investigated. Clonogenic assay results vary from a decrease in cell survival (gill) to no effect (skin) to a stimulatory effect (spleen). Results from fluorescence assays of ROS and MMP show similarities to clonogenic assay results. This study identifies a useful model for further studies relating to the bystander effect in aquatic organisms in vivo and ex vivo.

  12. Genotoxic and teratogenic potential of marine sediment extracts investigated with comet assay and zebrafish test

    Organic extracts of marine sediments from the North Sea and the Baltic Sea were investigated with two toxicity assays. The comet assay based on the fish cell line Epithelioma papulosum cyprini (EPC) was applied to determine the genotoxic potential; zebrafish embryos (Danio rerio) were used to quantify the teratogenic potential of the samples. EC 50 values were calculated from dose-response curves for both test systems. Highest teratogenic and genotoxic effects normalised to total organic carbon (TOC) content were detected in sediment samples of different origins. Polychlorinated biphenyls (PCBs) and polycyclic aromatic hydrocarbons (PAHs) are not likely to be the causes of the observed effects, as demonstrated by a two-step fractionation procedure of selected extracts. The toxic potential was more pronounced in fractions having polarity higher than those possessed by PAHs and PCBs.     Full Text Available Vaccination is an important strategy for the control and prevention of infectious pancreatic necrosis (IPN in farmed Atlantic salmon (Salmo salar in the post-smolt stage in sea-water. In this study, a heterologous gene expression system, based on a replicon construct of salmonid alphavirus (SAV, was used for in vitro and in vivo expression of IPN virus proteins. The large open reading frame of segment A, encoding the polyprotein NH2-pVP2-VP4-VP3-COOH, as well as pVP2, were cloned and expressed by the SAV replicon in Chinook salmon embryo cells (CHSE-214 and epithelioma papulosum cyprini (EPC cells. The replicon constructs pSAV/polyprotein (pSAV/PP and pSAV/pVP2 were used to immunize Atlantic salmon (Salmo salar by a single intramuscular injection and tested in a subsequent IPN virus (IPNV challenge trial. A low to moderate protection against IPN was observed in fish immunized with the replicon vaccine that encoded the pSAV/PP, while the pSAV/pVP2 construct was not found to induce protection.

  14. Contribution of scintigraphy for somatostatin receptors in case of non-iodine-fixating metastases of thyroid epithelioma

    The objective of this work was to evaluate the sensitivity and importance of scintigraphy of somatostatin receptors (SSR) in the detection of non iodine-fixating metastases of thyroid epithelioma. We have studied retrospectively 7 patients with a detectable thyroglobulin under hormonal hindrance, amounting after cessation of treatment up to 72 - 1500 μ g/l and a normal post-irradiation therapy (3.7 GBq of 131 I) scintigraphy. These patients (4 F / 3 M), aged from 27 to 74 years (average age, 54 years), were treated by total and ganglionic curettage for papillary cancer of thyroid. Their evolutive surveillance extended from 1 to 24 years (on average, 8.4 years) and they received curative cumulated 131 I activities of 13 to 26 GBq. Each of them benefited by an osseous scintigraphy, of a cervico-thoraco-abdominal TDM and of a 111 In Pentetreotide SSR. The SSR was positive for 4/7 patients showing cervico-mediastinal fixations, the localizations being not detected by TDM. The osseous scintigraphy revealed once to be positive showing a costal lesion already detected by SSR. Cervical non-palpable adenopathies were detected by TDM for one patient. The anatomical-pathological analysis confirmed their thyroid origin. For other two patients the TDM showed suspect pulmonary nodules. These cervical and pulmonary localization were not detected by SSR. The SSR + TDM couple was positive for 6/7 patients. In conclusion, in this series the sensitivity of SSR was estimated to be 57%.     A non-specific cytotoxic cell assay for fish is presented that is based on the release of the activated fluorochrome calcein AM from lysed carp epithelioma papulosum cyprini (EPC) cells. To establish the suitability of treating EPC cells with calcein AM the uptake and spontaneous release of the calcein AM by the EPC cells was evaluated. Incubation of 5 microM calcein AM in culture medium with 1x10(5)EPC cells well(-1)for a minimum of 3 h provided sufficient labelling. Spontaneous release of fluorescence from the labelled EPC cells during 10 h of post labelling incubation ranged from 30 to 39% of the total observed fluorescence. Cytotoxic activity of trout leucocytes was evaluated at three leucocyte to target cell ratios (10:1, 2:1 and 1:1) following incubation (4, 6, 8, and 10 h) with calcein AM-labelled EPC cells at 15 degrees C. In some instances, the monoclonal antibody specific for the NCC surface receptor NCCRP-1 (MAb5C.6) was included in the cultures.   16. A new bacilliform fathead minnow rhabdovirus that produces syncytia in tissue culture.

    A pathogenic bacilliform virus 130-180 nm in length and 31-47 nm in diameter was isolated from moribund fathead minnows (Pimephales promelas) exhibiting hemorrhages in their eyes and skin. A cytopathic effect of multifocal syncytia was observed in the epithelioma papulosum cyprini cell line after a 48 h incubation at 20 degrees C. A similar cytopathic effect was also observed in other cell lines tested, but not in bluegill fry, koi fin, or Chinook salmon embryo cells. The filterable agent was inactivated by exposure to 50 degrees C for 10 min, 20% ether, 2 and 50% chloroform, pH 3, and pH 10, was unaffected by 5'-iodo-2 deoxyuridine, and appeared bacilliform and occasionally bullet-shaped by electron microscopy. These results are consistent with those of rhabdoviruses. Immunodot blots performed with antisera against selected fish rhabdoviruses, an aquareovirus, and a birnavirus were all negative. River's postulates were fulfilled in fathead minnows, but the agent did not replicate or cause disease in other cyprinids or salmonids during challenge experiments. Hepatic, splenic, and renal lesions were observed during histological analysis of diseased fish from viral challenges and from the original case. Structural proteins resolved via SDS-PAGE had molecular weights similar to those reported in lyssaviruses of the family Rhabdoviridae; however, syncytia formation is not a typical cytopathic effect of rhabdoviruses. This virus, has tentatively been named the fathead minnow rhabdovirus (FHMRV) and is most similar to the members of the family Rhabdoviridae, but atypical properties like syncytia formation may justify the assignment to a novel taxon.

    Full Text Available Our purpose was demographic clinical and pathological aspect of BCE in patients seen in Razi Hospital, during a six-month period (75.8.12 to 76.2.12. Results: From the total 20000 patients, 103 cases of BCE were detected. (0.5%. The male female ratio was 1.71 BCE was more frequent in sixth decade. 40.8% of patients were fair skin (Type II, 54.4% tawny (Type III and 4.9% brown (Type IV, V. 15.5% of patients had a past history of freckles and history of radiotherapy in childhood was present in 41.7% 89.3% had no history of acne and seborrhea. The scalp was the most common site of BCE. The most common clinical type was nodular BCE and solid BCE was the dominant histological feature. Conclusion: BCE was more common in male and fair skin patients with dry skin. In those having history of radiotherapy of the scalp, lesions were seen mostly on the scalp, forehead and neck: pigmented BCE was predominant in this group.

    Summary Pour étudier l'ulcère de Marjolin, tous les dossiers des patients qui se sont présentés pendant une période quinquennale à un Service de Chirurgie Plastique et des Brûlés au Maroc ayant un cancer sur cicatrice de brûlure, objectivé par un examen histopathologique, ont été inclus dans une fiche de recueil de données comprenant des paramètres liés à l'identité du malade, à l'inventaire préthérapeutique, au traitement et à l'évolution du cas. Les Auteurs, après avoir présenté les données des cinq patients inclus dans l'étude, considèrent les problèmes posés par les épithéliomas spinocellulaires sur cicatrice de brûlure, qui sont des affections graves et rares. La dégénérescence des cicatrices de brûlures est une évolution dramatique, causée par la négligence. Le traitement préventif par exérèse systématique de toute lésion suspecte doit être fortement souligné puisqu'il garantit la guérison quasi certaine. PMID:21991185

    Tripartite motif-containing protein 32 (TRIM32) belongs to the tripartite motif (TRIM) family, which consists of a large number of proteins containing a RING (Really Interesting New Gene) domain, one or two B-box domains, and coiled coil motif followed by different C-terminal domains. The TRIM family is known to be implicated in multiple cellular functions, including antiviral activity. However, it is presently unknown whether TRIM32 of common carp (Cyprinus carpio) has the antiviral effect. In this study, the sequence, expression, and antiviral function of TRIM32 homolog from common carp were analyzed. The full-length coding sequence region of trim32 was cloned from common carp. The results showed that the expression of TRIM32 (mRNA) was highest in the brain, remained stably expressed during embryonic development, and significantly increased following spring viraemia of carp virus (SVCV) infection. Transient overexpression of TRIM32 in affected Epithelioma papulosum cyprinid cells led to significant decrease of SVCV production as compared to the control group. These results suggested a potentially important role of common carp TRIM32 in enhancing host immune response during SVCV infection both in vivo and in vitro. PMID:27735853

    Radiolesions of the pelvic bones (excluding the femur) were observed in 29 patients after irradiation therapy for cervix uteri cancer. Three regions can be affected corresponding to the areas irradiated: the sacrum and internal part of the iliac wings, the pubis, and the cotyles. In most cases the lesions appeared after 1 to 4 years. Their radiological appearances are characteristic, the principal sign being irregular bone condensation in a demineralized bone, without true cavitation. The only lytic lesions observed were in the pubis. Calcification of soft tissues may occur and fractures are frequent. There is a slow progression of the lesions over long periods.   4. Description of an as yet unclassified DNA virus from diseased Cyprinus carpio species.

    Numerous deaths of koi and common carp (Cyprinus carpio) were observed on many farms throughout Israel, resulting in severe financial losses. The lethal viral disease observed is highly contagious and extremely virulent, but morbidity and mortality are restricted to koi and common carp populations. Diseased fish exhibit fatigue and gasping movements in shallow water. Infected fish had interstitial nephritis and gill necrosis as well as petechial hemorrhages in the liver and other symptoms that were not consistent with viral disease, suggesting a secondary infection. Here we report the isolation of carp nephritis and gill necrosis virus (CNGV), which is the etiologic agent of this disease. The virus propagates and induces severe cytopathic effects by 5 days postinfection in fresh koi or carp fin cell cultures (KFC and CFC, respectively), but not in epithelioma papillosum cyprini cells. The virus harvested from KFC cultures induced the same clinical signs, with a mortality of 75 to 95%, upon inoculation into naive koi and common carp. Using PCR, we provide final proof that the isolated virus is indeed the etiologic agent of food and ornamental carp mortalities in fish husbandry. Electron microscopy revealed viral cores with icosahedral morphology of 100 to 110 nm that resembled herpesviruses. Electron micrographs of purified pelleted CNGV sections, together with viral sensitivities to ether and Triton X-100, suggested that it is an enveloped virus. However, the genome of the isolated virus is a double-stranded DNA (dsDNA) molecule of 270 to 290 kbp, which is larger than known herpesviruses. The viral DNA seems highly divergent and bears only small fragments (16 to 45 bp) that are similar to the genomes of several DNA viruses. Nevertheless, amino acid sequences encoded by CNGV DNA fragments bear similarities primarily to members of the Poxviridae and Herpesviridae and to other large dsDNA viruses.   5. Identification of DreI as an antiviral factor regulated by RLR signaling pathway.

    Full Text Available BACKGROUND: Retinoic acid-inducible gene I (RIG-I-like receptors (RLRs had been demonstrated to prime interferon (IFN response against viral infection via the conserved RLR signaling in fish, and a novel fish-specific gene, the grass carp reovirus (GCRV-induced gene 2 (Gig2, had been suggested to play important role in host antiviral response. METHODOLOGY/PRINCIPAL FINDINGS: In this study, we cloned and characterized zebrafish Gig2 homolog (named Danio rerio Gig2-I, DreI, and revealed its antiviral role and expressional regulation signaling pathway. RT-PCR, Western blot and promoter activity assay indicate that DreI can be induced by poly I:C, spring viremia of carp virus (SVCV and recombinant IFN (rIFN, showing that DreI is a typical ISG. Using the pivotal signaling molecules of RLR pathway, including RIG-I, MDA5 and IRF3 from crucian carp, it is found that DreI expression is regulated by RLR cascade and IRF3 plays an important role in this regulation. Furthermore, promoter mutation assay confirms that the IFN-stimulated regulatory elements (ISRE in the 5' flanking region of DreI is essential for its induction. Finally, overexpression of DreI leads to establish a strong antiviral state against SVCV and Rana grylio virus (RGV infection in EPC (Epithelioma papulosum cyprinid cells. CONCLUSIONS/SIGNIFICANCE: These data indicate that DreI is an antiviral protein, which is regulated by RLR signaling pathway.

  6. Identification and characterization of a novel gene differentially expressed in zebrafish cross-subfamily cloned embryos

    Full Text Available Abstract Background Cross-species nuclear transfer has been shown to be a potent approach to retain the genetic viability of a certain species near extinction. However, most embryos produced by cross-species nuclear transfer were compromised because that they were unable to develop to later stages. Gene expression analysis of cross-species cloned embryos will yield new insights into the regulatory mechanisms involved in cross-species nuclear transfer and embryonic development. Results A novel gene, K31, was identified as an up-regulated gene in fish cross-subfamily cloned embryos using SSH approach and RACE method. K31 complete cDNA sequence is 1106 base pairs (bp in length, with a 342 bp open reading frame (ORF encoding a putative protein of 113 amino acids (aa. Comparative analysis revealed no homologous known gene in zebrafish and other species database. K31 protein contains a putative transmembrane helix and five putative phosphorylation sites but without a signal peptide. Expression pattern analysis by real time RT-PCR and whole-mount in situ hybridization (WISH shows that it has the characteristics of constitutively expressed gene. Sub-cellular localization assay shows that K31 protein can not penetrate the nuclei. Interestingly, over-expression of K31 gene can cause lethality in the epithelioma papulosum cyprinid (EPC cells in cell culture, which gave hint to the inefficient reprogramming events occurred in cloned embryos. Conclusion Taken together, our findings indicated that K31 gene is a novel gene differentially expressed in fish cross-subfamily cloned embryos and over-expression of K31 gene can cause lethality of cultured fish cells. To our knowledge, this is the first report on the determination of novel genes involved in nucleo-cytoplasmic interaction of fish cross-subfamily cloned embryos.

    In 21 patients with a variety of skin tumors (squamous cell carcinomas, malignant melanomas, basal cell epitheliomas and mycosis fungoides) or precancerous lesions (Bowen's disease, actinic keratosis, junctional nevus cell nevus) the radioactive phosphorus uptake test demonstrates a sign ficantly increased concentration of P 32 in those tumors. There were no false negative tests. The possibility of differentiation of malignant melanoma from benign nevus cell nevus and the early recognition of cutaneous metastases is described.     Schizothorax o'connori and Oxygymnocypris stewartii are two endangered endemic Tibetan fishes that thrive in the Lhasa River at an average altitude over 4000 m. During artificial reproduction of endemic Tibetan fishes, the juvenile fish of S. o'connori and O. stewartii experienced mass mortality event. The causative agent is diagnosed to be a ciliate parasite, Chilodonella piscicola (syn. C. cyprini), which is common in various fishes. Here, we supplemented its description based on the morphological and molecular data. The body of C. piscicola is oval, 30-60 × 25-40 μm in vivo. Cyrtos is hook-like, composed of 9-10 toothed nematodesmal rods. Somatic kineties usually contain seven right kineties and nine left kineties. Two parallel circumoral kineties revolve round the cyrtos, and one preoral kinety extends to the anterior end of the fourth left-most kinety. Terminal fragment kinety is linear and on the top left of dorsal side. Sequence alignments revealed that the present SSU rDNA and ITS1-5.8S-ITS2 sequences are both most similar to the sequences of C. uncinata with the similarities of 98.2 and 99.5%. The phylogenetic analyses showed that C. piscicola is sister to other Chilodonella species, whereas C. cyprini (FJ873805) cluster with Tetrahymena species. Molecular analysis shows that the ITS1-5.8S-ITS2 sequence of C. cyprini in GenBank is unreliable. Our study extended the host range of C. piscicola and supplemented and revised the molecular data. Besides, as far as we know, this is the first record of C. piscicola in Tibetan plateau.

    A 12-year-old female miniature schnauzer and a 12-year-old neutered female cocker spaniel each had a grey-yellow nodular lesion arising from the gingiva. Microscopical examination showed that both nodules were composed of varied proportions of sebocytes and basal-type reserve cells with few ducts lined by stratified squamous epithelium. Based on the histopathological findings, the cases were diagnosed as sebaceous adenoma and sebaceous epithelioma, respectively. In man, the occurrence of sebaceous neoplasms in the oral cavity has been reported to some extent, but these lesions are very rare in animals. To the best of our knowledge, this is the first report of intra-oral sebaceous neoplasms in dogs. Intra-oral sebaceous epithelioma has never been reported in animals. Copyright © 2017 Elsevier Ltd.     Full Text Available A 21-year-old woman presented with complaints of skin lesions on her face, palms and soles. On examination, 8 to 10 well-defined, pigmented nodules with raised and pearly borders were seen on the face. Multiple pits were present on the palms and soles. Biopsy of both lesions revealed the presence of basal cell epitheliomas. The patient also gave history of removal of a keratocyst of mandible 10 years back. The case was diagnosed as Gorlin′s syndrome. It is atypical due to the low number of basal cell epitheliomas, the occurrence of only a single odontogenic cyst and the absence of other features usually associated with this condition

    Full Text Available Abstract Background Outbreaks with mass mortality among common carp Cyprinus carpio carpio and koi Cyprinus carpio koi have occurred worldwide since 1998. The herpes-like virus isolated from diseased fish is different from Herpesvirus cyprini and channel catfish virus and was accordingly designated koi herpesvirus (KHV. Diagnosis of KHV infection based on viral isolation and current PCR assays has a limited sensitivity and therefore new tools for the diagnosis of KHV infections are necessary. Results A robust and sensitive PCR assay based on a defined gene sequence of KHV was developed to improve the diagnosis of KHV infection. From a KHV genomic library, a hypothetical thymidine kinase gene (TK was identified, subcloned and expressed as a recombinant protein. Preliminary characterization of the recombinant TK showed that it has a kinase activity using dTTP but not dCTP as a substrate. A PCR assay based on primers selected from the defined DNA sequence of the TK gene was developed and resulted in a 409 bp amplified fragment. The TK based PCR assay did not amplify the DNAs of other fish herpesviruses such as Herpesvirus cyprini (CHV and the channel catfish virus (CCV. The TK based PCR assay was specific for the detection of KHV and was able to detect as little as 10 fentograms of KHV DNA corresponding to 30 virions. The TK based PCR was compared to previously described PCR assays and to viral culture in diseased fish and was shown to be the most sensitive method of diagnosis of KHV infection. Conclusion The TK based PCR assay developed in this work was shown to be specific for the detection of KHV. The TK based PCR assay was more sensitive for the detection of KHV than previously described PCR assays; it was as sensitive as virus isolation which is the golden standard method for KHV diagnosis and was able to detect as little as 10 fentograms of KHV DNA corresponding to 30 virions.

    Full Text Available Pilomatrixoma, also known as calcifying epithelioma of Malherbe, is a rare benign skin tumor arising from hair follicle stem cells. The most common localization is the head and neck region. Female/male ratio is 3/2. It shows deep subcutaneous placement and occurs in the first two decades of life. Its diameter ranges from 0.5 cm to 3 cm. Multiple lesions are rarely seen. Histopathologically it is characterized by basoloid and ghost cells. Perforating type is a rare clinical variant. Treatment is surgical excision. Our case is presented to draw attention to a rare clinical variant of pilomatrixioma.

But it was in 1960, Gorlin-Goltz established the association of basal cell epithelioma, jaw cyst and bifid ribs, a combination which is now frequently known as Gorlin-Goltz syndrome as well as Nevoid Basal Cell Carcinoma Syndrome (NBCCS. NBCCS is inherited as an autosomal dominant trait with high penetrance and variable expressivity. NBCCS is characterized by variety of cutaneous, dental, osseous, opthalmic, neurologic and sexual abnormalities. One such case of Gorlin-Goltz syndrome is reported here with good illustrations.

The overall prevalence of BCC is 3 times higher than that of SCC, but this can vary when looking at specific locations such as the hand, where SCC is much more common than BCC. Carcinoma (or epithelioma cuniculatum is a rare variant of SCC. It was originally described as a verrucous carcinoma of the soles. Exceptionally, it can arise in other parts of the skin. We report a rare case of carcinoma cuniculatum of the right thenar region with bone and lymph node involvement.

    The Authors try to show the possibility to combine radical excision with minimal invasiveness in the surgery of penile cancer. The focal point of every therapeutic decision is correct clinical staging. Unfortunately there's some confusion in the two international staging systems (TNM and Jackson's classification). In fact it's not clear the anatomical difference between epithelioma of the glans infiltrating corpus spongiosum and subcoronary epithelioma of the shaft infiltrating the corpora cavernosa. It's obvious that the infiltration of the corpora cavernosa is a far more aggressive oncological manifestation than that of tumour infiltrating the corpus spongiosum. So we consider Jackson's classification more congenial. In terms of surgery this anatomical independence makes it easy to consider the corpora cavernosa as a distinct entity, so they remain perfectly functional when separated from the glandulo-spongio-urethral unit with its vasculo-nervous bundle. This makes conservation of the erectile function, when clinical staging show us that the tumour is not infiltrating the corpora cavernosa. The Authors show their results, which seem to be rather good.

    A review is presented of patient with rhinoescleroma, corresponding to the first years of operation of the then National Institute of Radium. It is a 35 year-old patient, the current illness it began before three years with a frequent nasal cold and difficulty of the step of the air until arriving to the total obstruction; additionally, a mass appears inside the nose, with painful ulceration that extends, first to the lip, to the hard palate and the place of installation of the teeth, and then to the part previous of the palatine vault, with fall of the teeth and communication with the skin. The patient also refers loss of the audition of the left side. The tumor to the physical exam measures 6 centimeters of diameter and of height, and there is extension with ulceration of the vestibular area. The rhinoscopy shows obstruction with ulceration and purulent secretion. The x-rays of July 9 1935 show partial destruction of the maxillary superiors that includes the alveolar edges, the palatine apophysis and the nasal spine. In the clinical history, it reports two biopsies; the biopsy of the nose concludes that it is a rhinoescleroma with atypical epithelioma and the biopsy gingival says that it is an incipient epithelioma spine-cellular

    Peripheral primitive neuroectodermic tumor (PNET) or peripheral neuro epithelioma is a malignant neoplasia consisting of round small blue cells, which derive from the neural crest. Amongst malignant tumors it is one the least differentiated histological types. Ewings sarcoma and PNET are morphologically impossible to differentiate; they are also very similar pathologically in terms of their imagining appearance. Due to these similarities we will consider them as a single entity. These can appear in infants, but are more common in adolescents. Treatment includes hemo therapy,surgery and radiotherapy. We present a case of an 8 year old female patient, diagnosed with Ewing Sarcoma, confirmed by histopathology and immunohistochemistry, we will also present a brief review of the literature.

    Twenty-one primary bone tumours of the hand in children from 8 paediatric hospitals are reported. Osteochondromas and enchondromas were not included. Our material consisted of 16 patients with common tumours (3 Ewing's sarcoma, 5 aneurysmal bone cyst, 6 osteoid osteoma and 2 epithelioma) and 5 patients with uncommon tumours (osteoma, simple bone cyst, haemangiopericytoma, capillary angiomatous tumour and benign ossifying fibroma or osteoblastoma). The X-ray diagnosis of the common tumours should have high concordance with histology, whereas that of uncommon tumours in much more difficult and uncertain. The characteristic features of Ewing's sarcoma are stressed as all our children with this tumour had a delayed diagnosis and a fatal outcome. Differential diagnosis with other short tubular bone lesions of the hand - specifically osteomyelitis - is discussed and the posibilities of microscopic diagnosis are stressed. (orig.)

    Three years following an irradiation by the Betatron's electron beam of an epithelioma in left parieto occipital area of the scalp in a female patient aged 77, early suffering from high blood pressure, a fatal pseudo-tumoral brain necrosis occurs presenting as a rapidly increasing from of Wernicke's aphasia. The necropsy shows intense radionecrosis lesions of the brain and the bone, free of any parenchymatous malignant proliferation note-wortly for the striking density of microvascular changes as previously described in radiation therapy. The case observed some years ago, allows to definite again the limits doses of the extracranial irradiations now estimated at 1760 rets.     Among a total of 65,268 Nagasaki atomic bomb survivors recorded in the Scientific Data Center of Atomic Bomb Disaster, Nagasaki University School of Medicine, 140 cases with skin cancer were collected from 31 hospitals in Nagasaki City from 1961 through 1987. Subsequently, these cases of skin cancer in Nagasaki atomic bomb survivors were statistically analyzed in relation to the estimated distance from the hypocenter by age, sex, histology and latent period. The results were as follows: (1) A high correlation was observed between the incidence of skin cancer and the distance from the hypocenter. (2) The incidence of skin cancer in Nagasaki atomic bomb survivors now appears to be increasing in relation to exposure distance. (3) Among 140 cases, basal cell epithelioma was observed in 67 cases (47.9%) and squamous cell carcinoma in 43 cases (30.7%). (author).

    A review was made on 412 cases of postirradiation skin malignancies reported in Japan up to March 1988. The ratio of male to female was 2:1. Histologically, squamous cell carcinoma occupied 60% of all cases. The incidence of sarcoma has recently been increased. Sixty percent of all skin malignancies resulted from irradiation for benign diseases. Radiotherapy has recently become the treatment of choice for malignancy. The incidence of malignancy resulting from occupational exposure has remained unchanged. The latency period before the development of radiation-induced malignancy varied in the following order with cause or primary disease: occupation>benign tumors>malignant tumors; and it varied with histology in the following order: basal cell epithelioma>squamous cell carcinoma>sarcoma. Malignant tumors treated with large doses of high energy photon beams were likely to develop sarcomas in a relatively short latency period of time. (N.K.).

    Full Text Available Niger is a tropical country where leishmaniasis is endemic. The first case was reported in 1911. Leishmania major is practically the pathogen found in Niger, a country lying between 8° and 20° north latitudes from the Atlantic to the Chadian border. We report a case of cutaneous leishmaniasis in the type of ulcerative lesion, characterized by its localization at the internal angle of the right eye posing a diagnostic problem. The diagnosis of leishmaniasis was made by parasitological examination. Anatomipathologic examination eliminated cutaneous tuberculosis, pyogenic granuloma, molluscum contagiosum and basal cell epithelioma. Metronidazole management has accelerated healing. Thus in a tropical country, in front of any chronic, painless ulcerative lesion and resistant to all therapeutics, the diagnosis of cutaneous leishmaniasis must be evoked, for a consequent management after confirmation.

    Data from 140 A-bomb survivors with skin cancer were analyzed with the purpose of elucidating the relationship between atomic bombing and skin cancer. The incidence of skin cancer was significantly correlated with the distance from the hypocenter (p<0.01), regardless of sex. Basal cell epithelioma was the most predominant, followed by squamous cell carcinoma. Histology of skin cancer seemed independent of the distance. Since 1965, the incidence of skin cancer has been increased with aging in A-bomb survivors exposed at le2500 m from the hypocenter. It has been significantly higher since 1975 in the le2500 m group than in the ge3000 m group. (N.K.).

  12. Clinical outcome of 19 patients with nasopharyngeal cancer. A review of neo-adjuvant chemotherapy

    We clinically examined 19 cases of nasopharyngeal cancer in which primary care was administered in the Department of Otolaryngology, Tokyo Metropolitan Fuchu Hospital between September 1990 and August 1999. The subjects consisted of 11 males and 8 females. Histophathological study revealed 17 cases of WHO type III tumors (14 cases were poorly differentiated squamous cell carcinoma and 3 cases were lymph-epithelioma). The accumulated 5-year survival rate by the Kaplan-Meier method was 50% in T1, 75% in T2, 0% in T4, and 36% overall. Neo-adjuvant chemotherapy was administered in 15 cases and distant metastasis appeared in 3 cases (21%) after definitive radiotherapy. The biological characteristic of WHO type III tumors is a tendency towards early metastasis, and we speculated that this might be the cause of the lower level of effectiveness of the neo-adjuvant chemotherapy in these cases. At present, this therapy is not effective and further improvement is required. (author)

    Three years following an irradiation by the Betatron's electron beam of an epithelioma in left parieto occipital area of the scalp in a female patient aged 77, early suffering from high blood pressure, a fatal pseudo-tumoral brain necrosis occurs presenting as a rapidly increasing from of Wernicke's aphasia. The necropsy shows intense radionecrosis lesions of the brain and the bone, free of any parenchymatous malignant proliferation note-wortly for the striking density of microvascular changes as previously described in radiation therapy. The case observed some years ago, allows to definite again the limits doses of the extracranial irradiations now estimated at 1760 rets. That is the 'Nominal Standard Dose' (NSD) measured by rets and taking into account the number of seances (N) and the duration of irradiation (T) which would be to take the place of 'the total dose' (D) (rads). These dosimetric criteria themselves must be adjusted to the age and the vascular features of each patient.

  16. Repair of DNA in xeroderma pigmentosum conjunctiva

    Xeroderma pigmentosum (XP) is an autosomal recessive disease with tumor formation on sun-exposed areas of the skin and eyes. Cells from most XP patients are deficient in repairing DNA damaged by ultraviolet (uv) light as shown by a reduced rate of tritiated thymidine (3HTdR) incorporation during their DNA repair synthesis. We have studied such repair synthesis in conjunctival cells from an XP patient with a conjunctival epithelioma and from normal cadaver conjunctiva. Cultured conjunctival cells were irradiated with uv light and then incubated with 3HTdR. Autoradiograms were prepared and showed that uv radiation induced a considerably slower rate of DNA repair synthesis in the XP cells than in normal cells. Many of the ocular abnormalities of XP, including tumor formation, may be the result of this defective DNA repair process

    Full Text Available Aquatic macrophyte biomass, diaspore bank distribution and their relationship to spatial variability of depth, nutrients (nitrite, nitrate, ammonium and soluble reactive phosphorus as well as sediment granulometry in an athalassohaline lake have been studied during one wet hydrological year. The results indicate that species growing in the lake show different spatial distribution patterns throughout the lake. Indirect gradient analysis (canonical analysis results showed a first axis defined as a function of Ulva flexuosa Wulfen biomass which is, in turn, positively correlated with interstitial ammonium and Soluble Reactive Phosphorus (SRP. The second axis was mainly established due to Lamprothamnium papulosum (Wallr. J. Groves biomass which correlated positively to depth and negatively to interstitial ammonium and SRP. These results revealed a NESW eutrophic gradient allowing the U. Flexuosa biomass proliferation. This phenomenon could increase the shadow effect over the rest of the macrophytes inhabiting this shallow lake. Moreover, the eutrophic harmful effect on the macrophyte physiology and over the diaspore bank could have important consequences in the survival of such important populations. The results reported in this study show the need for studies as the base to select sampling points for monitoring this wetland.

  18. Nodular goiter after occupational accidental exposure to radiation

    Full text: Purpose: During a control of the safety measures at the Radiology Service in a local hospital in Buenos Aires, a lack of an appropriate shielding in the X-ray equipment was detected. Methods and Materials: Therefore studies were performed in the group of physicians and technicians exposed and in an age and sex matched group of non-exposed physicians and technicians. Check-ups included thyroid sonography, circulating thyroid hormones, TSH and thyroid antibodies. The exposed group included 9 females and 5 males, while the non-exposed group comprised 8 females and 5 males. Results: No thyroid abnormalities were found in the non-exposed patients. On the contrary in the exposed cohort in 11 out of 14 patients nodular goiter was diagnosed, while an additional patient presented diffuse goiter. In 5 out of the 12 patients with goiter, nodule growth or the appearance of new nodules was observed along the follow-up. Four patients presented changes in thyroid function: two had elevated basal TSH with increased titters of anti TPO antibodies in one, while another showed an abnormal TRH-TSH test. A fourth patient had low total T 4 values. Other abnormalities included: sperm abnormalities in 2 out of the 5 males, dry-eye syndrome in other two, mammary nodules in one female and a basocellular epithelioma in another one. Conclusions: accidental irradiation leads to thyroid abnormalities, such as the appearance of nodules and alterations in thyroid function. (author)

    Objective was to study the clinical and histopathologic spectrum of pilomatricoma, the benign tumor of hair matrix. Retrospective review of 27 cases of pilomatricoma (Calcifying epithelioma of Malherbe) reported at Bahrain Defence Force Hospital from 1993 - 1999. Most of the cases were confused clinically with sebaceous cysts. Seventy eight per cent of the cases occurred below the age of 30 years. Female to male ratio was 5:4. Head, neck and upper limb were the most common sites for pilomatricoma. The size of the tumors ranged from between 4-35 mm in diameter. Tumors were encapsulated and solid composed of shadow and basophilic cells, and stroma containing varying amounts of calcification, ossification and inflammatory cells. Pilomatricomas have a wide variety of clinical characteristics and are often misdiagnosed with other skin conditions. They should be considered along with other benign and malignant conditions in the clinical differential diagnosis of solitary firm skin nodules especially those, which occur in the head, neck and upper limb.     Full Text Available The fowlpox, also known as contagiosum epithelioma, is caused by a poxvirus of the genus Avipoxvirus and affects both domestic and wild birds. The disease has two forms cutaneous and diphtheria. In March 2008 the sector of rehabilitation of wild birds from Mangual das Garças Park, located in Belém, Pará attended a duck (Dendrocygna autumnalis that had nodular lesions in regions of the wing devoid of feathers. The lesions consisted of two large cutaneous tumor-like nodules that measured 4,2 x 3,8 cm and 2,8 x 2,2 cm in length and width, respectively. The nodules were surgically removed, fixed in 10% formalin, processed and stained with hematoxylin-eosin. Histopathology showed acanthosis and expressive hyperkeratosis, some cells showed spongiosis. The confirmatory diagnosis of fowlpox was established by the pathognomonic sign of the presence of large intracytoplasmic eosinophilic inclusion corpuscles (Bollinger bodies in epithelial cells. This is the first report of poxvirus infection in a wild bird in the State of Pará, Brazil.

    The Gorlin-Goltz syndrome is a dominant autosomic disorder characterized by cancerigenic predisposition and multiple development defects, apparently without reproductive compromise. The complex is characterized by four primary symptoms, which include nevoid basal cell epitheliomas malignantly prone, keratocystic jaw, skeletal abnormalities and intracranial calcifications. Apparently, reproductive problems reported had been rarely associated with this syndrome. We present the case of a patient with clinic stigmatae of Gorlin-Goltz syndrome, who had a characteristic progress as seen in the literature; he was the fifth product of a 43 year-old female (father was 48 years old); who at birth disclosed right eye microftalmy, bilateral cryptorchidism surgically treated at age of six. At puberty, an odontogenic cyst of the jaw was noted and enucleated. He also showed facial nevi in neck, thorax and abdomen. When he was admitted being 14 years old in our clinic, he had recurrent bilateral cryptorchidism, sexual immatturity and infertility. It is important to take into consideration Gorlin-Goltz stigmatae in cases of hypogonadism in order to recognize a further genetic influence.

  2. Incidence of skin cancer in Nagasaki atomic bomb survivors based on DS86 dosimetry system, 1958-1985

    The incidence of skin cancer during the period 1958-1985 was examined in the population registered in the life span study extension (LSSE) and the adult health study (AHS). Among 25,942 A-bomb survivors in whom DS86 was available, skin cancer was confirmed in 47 A-bomb survivors. These A-bomb survivors consisted of 24 males and 23 females. According to DS86 dosimetry system, ten A-bomb survivors had been exposed to 0.50 Gy or more. The most common histology was basal cell epithelioma (n=25), followed by malignant melanoma (n=4) and basosquamous cell carcinoma and sweat gland carcinoma (one each). In the group of 0.50 Gy or more, the incidence of occurrence of skin cancer was 20.8/100,000 population per year (PY) for the LSSE population and 22.8/100,000 PY for the AHS population. In the group of 0.01-0.49 Gy, it was 6.8/100,000 PY for the LSSE population and 12.8/100,000 PY for the AHS population. It was significantly associated with higher exposure doses.     External radiation therapy, or teletherapy, is the use of ionizing radiation to destroy cancer cells. Clinical use of ionizing radiation as treatment for cancer began with the discovery of x-rays in 1895, the identification of natural radioactivity (radium) in 1896, and the first reported cure of cancer, a basal cell epithelioma, induced by radiation in 1899. Initially, radiation was administered as a single large dose and produced severe, life-threatening side effects. The basis for the use of ionizing radiation in daily increments for a period of weeks was provided by Regaud in 1922; ten years later, Coutard clinically developed the method of dose fractionation, which remains in use today. Although the use of ionizing radiation as a treatment is over eighty years old, only in recent years have advancements in its clinical application been based on research related to the biologic effect of radiation on human cells. To effectively care for the patient prior to, during, and at the completion of external radiation therapy, the nurse must know the physical and biologic basis of external radiation therapy and its clinical application.

  4. Radiation therapy for malignant lid tumor; Effectiveness of racket-shaped lead protector

    The case of a 42-year-old man with Meibomian gland carcinoma in his right lower lid is reported. The tumor found in the nasal part of the lower lid, was 12 mm x 13 mm in size. First, surgical resection was performed. The pathological diagnosis of the frozen section was 'undifferentiated basal cell epithelioma'. Second, cryotherapy was performed all over the cut surface. Later, the permanent section was pathologically diagnosed as 'undifferentiated Meibomian gland carcinoma'. Total 50 Gy irradiation therapy was therefore performed using a 9 Mev Linac electron beam, 25 x 20 mm field, with a lead protector for the cornea and lens. A lead contact lens did not afford good results because it was too easily shifted on the cornea, owing to its weight. Therefore, we made a racket-shaped lead protector. Fixed well with tape, this protector afforded good protective effect. Three years after treatment, the patient has good visual function, with no recurrence. This racket-shaped lead protector is thought to be useful in radiation therapy for malignant lid tumors. (author).

    In this work four original observations of more or less long-delayed cancers induced by ionizing radiations are compared with 34 other cases in the literature, after which an attempt is made to establish a general and prognostic synthesis of the results; the indications to emerge are as follows: - Ionizing radiation-induced cancers are very rare, especially when compared with the extensive therapeutic use made of X-rays; - The probability of radio-cancer formation, though no figures are given in the many papers consulted, seems nevertheless to be higher in cases of benign lesion irradiation; - Induced cancers have been observed after treatments with all types of radiation, whether or not the lesion is tumoral or cancerous, whatever the patient's age at the time of irradiations; - As a general rule these neoplasms appear after a variable latency period but usually from the 6th post-radiotherapy year onwards, with a greater frequency range between 6 and 12 years; - These induced cancers are generally epitheliomas or sarcomas, the latter being noticeably more predominant than in the case of spontaneous cancers.     The effects produced by fast neutrons (0-100 Gy) on chromatin structure were analyzed by (i) [ 1 H]-NMR spectroscopy, (ii) time resolved spectroscopy, and (iii) fluorescence resonance energy transfer (FRET). Two types of chromatin were tested: (i) a chromatin from a normal tissue (liver of Wistar rats) and (ii) a chromatin from a tumoral tissue (Guerin limphotrope epithelioma, a rat solid tumor). The fast-neutron action on chromatin determines greater values of the [ 1 H]-NMR transverse relaxation time, indicating a more injured structure. Time-resolved fluorescence measurements show that the relative contribution of the excited state lifetime of bound ethidium bromide to chromatin DNA diminishes with increasing irradiation doses. This reflects the damage that occurs in DNA structure: production of single- and double-strand breaks due to sugar and base modifications. By the FRET method, the distance between dansyl chloride and acridine orange coupled at chromatin was determined. This distance increases upon fast-neutron action. The radiosensitivity of the tumor tissue chromatin seems higher than that of the normal tissue chromatin, probably because of its higher (loose) euchromatin/(compact) heterochromatin ratio. As the values of the physical parameters analyzed are specific for a determined dose, the establishment of these parameters may constitute a criterion for the microdosimetry of chromatin radiolesions produced by fast neutrons.   8. Effects of retinoids on ultraviolet-induced carcinogenesis

    The evidence for effects of the retinoids on UV-induced carcinogenesis is sparse. Clinical observations indicate that topical RA can cause significant regression of premalignant actinic keratoses. Also there is some evidence that this agent can cause dissolution of some basal cell epitheliomas. However this latter effect does not appear to be of therapeutic value. Systemic retinoids are of little value in the treatment of premalignant and malignant cutaneous lesions though 13-cis-retinoic acid might be of use in the basal cell nevus syndrome. Examination of the influence of the retinoids on photocarcinogenesis essentially has been confined to RA and animal experimentation. RA in nontoxic concentrations can both stimulate and inhibit photocarcinogenesis depending upon the circumstances of the study. The mechanisms of these responses are not clear. Influences on DNA synthesis directly and/or indirectly or on immune responses may be involved in both effects. Preliminary studies with oral 13-cis-retinoic acid have not demonstrated any effects to date on UV-induced skin cancer formation

  9. Calicoblastic neoplasms in Acropora palmata, with a review of reports on anomalies of growth and form in corals.

    Colonies of the elkhorn coral (Acropora palmata), possessing raised, whitened, irregularly shaped skeletal protuberances, were discovered at Carysfort Reef and Grecian Rocks, Key Largo National Marine Sanctuary, Key Largo, FL. These lesions exhibited relatively rapid growth and spread along the branches, as polyps were overlaid by coenosteal skeletal material. Histopathological examinations of the soft tissues surrounding and extending into the skeletal masses revealed proliferation of gastrovascular canals and associated calicoblastic epidermis, with loss of normal polyp structures and zooxanthellae. The slightly atypical tumor calicoblasts were cuboidal to columnar, resembling those found in the rapidly growing apical tips. Stable carbon isotope ratios of skeletal samples revealed that the tumor skeleton was isotopically lighter than the skeleton in the normal or apical track regions, indicative of higher tissue metabolic rates and lack of carbon isotope fractionation by zooxanthellae. This condition appears to be a neoplasm of the coral, and it is proposed that it be classified "calicoblastic epithelioma." Several different types of abnormal skeletal deposition, possibly the result of neoplastic processes, have been reported to occur in stony corals and are here reviewed.

    A 60 year-old woman with a scalp epithelioma underwent radiotherapy, the dose being 57 Gray. A first epileptic seizure occurred twenty months later. Neurological examination revealed signs of left hemisphere involvement. γEG, angiography, CT scans, demonstrated a pseudotumoral avascular process. On account of the localisation, the patient being right-handed, no surgical procedure was performed. In spite of corticotherapy and anticonvulsive treatment, seizures recurred and neurological signs slowly progressed. The patient died, 22 months after the first seizure, of an associated disseminated carcinoma with cachexia. Neuropathological examination showed a massive lesion presenting all the features of delayed radionecrosis in the left hemisphere: situated mainly in the white matter; numerous vascular abnormalities; wide-spread demyelination; disappearance of oligoglial cells. The Authors recall the clinical and anatomical aspects of this condition for which the only successful treatment is surgical removal when location and size of the lesion permit.     In 1882, Philippe Gaucher described a 32-year-old woman with massive splenomegaly and unusually large cells in the spleen, which he called a "primary epithelioma of the spleen." The systemic nature and inheritance of the disease and its variants involving the viscera and CNS were described over the next century. The delineation of the causal enzymatic defects, genetics, molecular pathology, and genomics have provided pathogenic insights into the phenotypic spectrum and the bases for development of specific therapies for what is now known as Gaucher disease. As a prototype, the clinically and economically successful intracellular enzyme therapy provided the impetus for the expansion of similar research and therapeutic developments for other lysosomal storage diseases (LSDs) and orphan diseases, including Fabry, Pompe, and Niemann-Pick diseases, as well as several mucopolysaccharidoses. Continuing studies of such LSDs, which occur as a group in more than 7000 live births, have revealed the complex molecular interdigitation with the autophagy and apoptotic pathways and proteostasis and the impact of disruptions of the lysosomal/autophagy and proteostasis systems on more common diseases has been recognized. Examples include age-related neurodegenerative diseases (eg, Parkinson disease and Gaucher disease), idiopathic hypertrophic myocardiopathies, stroke and renal failure (eg, Fabry disease), and Nonalcoholic Fatty Liver Disease/Nonalcoholic SteatoHepatitis (NAFLD/NASH) and atherosclerosis (eg, lysosomal acid lipase deficiencies). Although perceived as rare, the availability of treatment and the impact of the LSDs on more common diseases require their integration into routine clinical practice.

    Inhaled tobacco smoke comes in direct contact with few organs such as mouth, lungs, and stomach. Cigarette smoke (CS) in lungs has been extensively studied. However, limited data exist on its effect on skin, and there are no long-term experimental studies suggesting toxic effects on skin. Even though it is generally accepted that CS is among the main factors of skin aging, the number of experimental studies showing this aging effect is limited. We hereby studied the effect of long-term exposure to CS on the skin of hairless mice in combination with or without ultraviolet (UV) light. In addition, we investigated potential skin protection by a potent antioxidant namely procyanidine-rich French maritime pine bark extract (PBE) pycnogenol. Male and female hairless SKH-2 mice were exposed for 10 months to tobacco smoke and/or UV light in vivo, and their effects on skin were investigated. Some biophysical parameters such as development of erythema, transepidermal water loss (TEWL), and skin elasticity were measured. The results show that UV and CS may be acting synergistically, as shown by the enhanced TEWL, erythema values, epitheliomas, and squamous cell carcinomas (SCCs) observed, whereas PBE seems to protect skin against SCC.

  15. Radiation therapy for malignant lid tumor

    The case of a 42-year-old man with Meibomian gland carcinoma in his right lower lid is reported. The tumor found in the nasal part of the lower lid, was 12 mm x 13 mm in size. First, surgical resection was performed. The pathological diagnosis of the frozen section was 'undifferentiated basal cell epithelioma'. Second, cryotherapy was performed all over the cut surface. Later, the permanent section was pathologically diagnosed as 'undifferentiated Meibomian gland carcinoma'. Total 50 Gy irradiation therapy was therefore performed using a 9 Mev Linac electron beam, 25 x 20 mm field, with a lead protector for the cornea and lens. A lead contact lens did not afford good results because it was too easily shifted on the cornea, owing to its weight. Therefore, we made a racket-shaped lead protector. Fixed well with tape, this protector afforded good protective effect. Three years after treatment, the patient has good visual function, with no recurrence. This racket-shaped lead protector is thought to be useful in radiation therapy for malignant lid tumors. (author)

    Full Text Available Abstract Nevoid basal cell carcinoma syndrome (NBCCS, also known as Gorlin syndrome, is a hereditary condition characterized by a wide range of developmental abnormalities and a predisposition to neoplasms. The estimated prevalence varies from 1/57,000 to 1/256,000, with a male-to-female ratio of 1:1. Main clinical manifestations include multiple basal cell carcinomas (BCCs, odontogenic keratocysts of the jaws, hyperkeratosis of palms and soles, skeletal abnormalities, intracranial ectopic calcifications, and facial dysmorphism (macrocephaly, cleft lip/palate and severe eye anomalies. Intellectual deficit is present in up to 5% of cases. BCCs (varying clinically from flesh-colored papules to ulcerating plaques and in diameter from 1 to 10 mm are most commonly located on the face, back and chest. The number of BBCs varies from a few to several thousand. Recurrent jaw cysts occur in 90% of patients. Skeletal abnormalities (affecting the shape of the ribs, vertebral column bones, and the skull are frequent. Ocular, genitourinary and cardiovascular disorders may occur. About 5–10% of NBCCS patients develop the brain malignancy medulloblastoma, which may be a potential cause of early death. NBCCS is caused by mutations in the PTCH1 gene and is transmitted as an autosomal dominant trait with complete penetrance and variable expressivity. Clinical diagnosis relies on specific criteria. Gene mutation analysis confirms the diagnosis. Genetic counseling is mandatory. Antenatal diagnosis is feasible by means of ultrasound scans and analysis of DNA extracted from fetal cells (obtained by amniocentesis or chorionic villus sampling. Main differential diagnoses include Bazex syndrome, trichoepithelioma papulosum multiplex and Torre's syndrome (Muir-Torre's syndrome. Management requires a multidisciplinary approach.     Full Text Available Charophyte oospores and their features (size, shape, colour, outer wall ornamentation, etc. are able to persist in seed banks of aquatic ecosystems. Thus, these characteristics can be used for taxonomic species identification, thereby providing knowledge about the specific composition of the charophyte community in a habitat that currently lacks, or has greatly altered, submerged vegetation. Several years ago, l’Albufera de València lost the charophyte meadows that covered the lake bottom in the past, which had never been studied in detail. Several sediment cores were extracted from the lagoon at three sites and the oospores and gyrogonites were isolated and characterized. The oospore/gyrogonite densities in the sediment layers were estimated. Ten different taxa have been identified: six belonging to genus Chara [Ch. vulgaris, Ch. Hispida var. major, Ch. fragilis (Ch. globularis, Ch. Hispida var. baltica, Ch. aspera, Ch. tomentosa]; one Nitella species (N. hyalina, two Tolypella species (T. glomerata and T. hispanica and one Lamprothamnium species (L. papulosum. The latter, which indicates the period when water salinity was high in the lagoon, was found in the deepest sediment layers. An increase in the oospore abundance and diversity was observed in line with sediment depth. This indicates, on the one hand, that the lagoon went through different ecological stages (with important changes in salinity, mainly and, on the other hand, the eutrophication process that the lagoon has been undergoing for at least the last 45 years, has progressively affected the abundance and diversity of the charophyte community.

    In recent years, due to the development of nanotechnology new horizons in treatment and diagnosis of cancer open up. Development of nano-systems for simultaneous transfer of active substances and imaging of tumor regions gathers an important amount of scientific interest. This new category of nano-systems is called Theranostics. Theranostics methods can provide multiple benefits by inserting nanoparticles into the patient and using photodynamic therapy and pave the way for personalized medicine. The objective of this paper is to study the use and application of Theranostics in the diagnosis and treatment of cancer, in order to achieve personalized anticancer treatment. For this purpose, investigation of existing literature has been conducted using electronic databases, PubMed, Google Scholar and IEEE Xplore. In addition, there was a secondary research phase, using paper citations found during the first research phase. It has to be pointed out that nanoparticles are the basis of Theranostics, since, due to their properties, they provide the ability to display accurate imaging and provide diagnosis along with simultaneous treatment of diseases. Theranostics methods may be applied in treatment of esophageal cancer, prostate cancer, breast cancer, in treatment of actinic keratosis, actinic cheilitis and Bowen's disease and in treatment of basal cell epithelioma and macular degeneration. As a result, application of Theranostics can provide multiple benefits by inserting nanoparticles into the patient. This method is currently encountering many challenges, but continuation of research on the field is necessary not only for the improvement of the medical field and the healthcare techniques, but also for the creation of new treatment methods for patients with diseases that are incurable until now.

Presentamos un caso clínico de tricoepitelioma con deformidad severa de la cara en el que se realizó tratamiento radical con restitución de unidades estéticas mediante colgajos locales. Consideramos que los casos agresivos o deformantes deben ser tratados de forma radical, eliminando todo el espesor de la piel para obtener resultados satisfactorios.Trichoepithelioma, first described by Brooke in 1892, also known as cystic adenoid epithelioma, is a dominantly inherited genodermatosis originated in the follicle bulb. There are two clinical forms, solitary and multiple. Some authors advocate not to treat them, but multiple trichoepithelioma presents a significant cosmetic problem and partial resection can lead to recurrence and excessive scarring, some cases with an aggressive behavior. We present a clinical case of multiple tricoepithelioma with severe deformity of the face in which we realized radical resection with restitution of anatomical units by local flaps. We consider that the multiple cases should be excised with total skin to avoid recurrence and obtain acceptable results.

    Full Text Available Arsenicosis is a multisystem disorder, with virtually no system spared from its vicious claw; though its predominant manifestations are linked to cutaneous involvement. Cutaneous effects take the form of pigmentary changes, hyperkeratosis, and skin cancers (Bowen′s disease, squamous cell carcinoma, and basal cell epithelioma. Peripheral vascular disease (blackfoot disease, hypertension, ischemic heart disease, noncirrhotic portal hypertension, hepatomegaly, peripheral neuropathy, respiratory and renal involvement, bad obstetrical outcome, hematological disturbances, and diabetes mellitus are among the other clinical features linked to arsenic toxicity. The effects are mediated principally by the trivalent form of arsenic (arsenite, which by its ability to bind with sulfhydryl groups present in various essential compounds leads to inactivation and derangement of body function. Though the toxicities are mostly linked to the trivalent state, arsenic is consumed mainly in its pentavalent form (arsenate, and reduction of arsenate to arsenite is mediated through glutathione. Body attempts to detoxify the agent via repeated oxidative methylation and reduction reaction, leading to the generation of methylated metabolites, which are excreted in the urine. Understandably the detoxification/bio-inactivation process is not a complete defense against the vicious metalloid, and it can cause chromosomal aberration, impairment of DNA repair process, alteration in the activity of tumor suppressor gene, etc., leading to genotoxicity and carcinogenicity. Arsenic causes apoptosis via free radical generation, and the cutaneous toxicity is linked to its effect on various cytokines (e.g., IL-8, TGF-β, TNF-α, GM-CSF, growth factors, and transcription factors.     Full Text Available Background and Design: Photodynamic therapy (FDT is a photochemotherapy modality which is used frequently and effectively in the treatment of actinic keratosis, Bowen disease and basal cell carcinomas. This study was performed to determine cure rates, cosmetic outcome and recurrence rates after aminolevulinic acid (ALA-based photodynamic therapy for skin lesions showing complete response to treatment procedure. Material and Method: Sixty-eight patients (27 females and 41 males with 78 lesions were included in the study. Among them, 25 were actinic keratosis (AK, 8 were actinic cheilitis (AC, 30 were basal cell carcinomas (BCC, 3 were Bowen disease, 10 were intraepidermal epithelioma (IEE, one lesion was parapsoriasis and one lesion was verruca plantaris. Six to 8 hours after topical administration of ALA (20%, the lesions were exposed to light from a broad-band light source. Skin biopsy specimens were obtained from 74 lesions for histopathological control. Results: At the end of the second month of treatment, fifty-six (72% of seventy-eight lesions showed complete clinical response, whereas fourty-seven of 74 lesions (63.5% exhibited complete histopathological clearance. A total of 9 recurrences (16% was observed during a median follow-up of 36 months. Recurrence rates were 3 (14% in AK, 1 (17% in AC, 1 (8% in superficial BCC, 3 (75% nodular BCC and 1 (12.5% in IEE. Cosmetic outcome was excellent and good in 42 lesions (89%, fair in 3 lesions (6% and poor in 2 lesions (5%. Conclusion: Topical photodynamic therapy is a noninvasive, effective and cosmetic modality of treatment in the selected skin lesions, as an alternative to the conventional procedures.

    Cancer is relatively rare disease among children and adolescents. The incidence of solid tumors other than CNS is less than 2/100,000. Tumors of the chest wall can arise either from the somatic tissue or ribs. These are rare, so either institutional reviews or multi institutional studies should determine optimal therapeutic management. Of the bony chest wall, Ewing's sarcoma or the family of tumor (peripheral neuro epithelioma, Askin tumor), are the most common. These lesions are lytic and have associated large extra pleural component. This large extra pleural component often necessitates major chest wall resection (3 or more ribs), and when lower ribs are involved, this entails resection of portion of diaphragm. Despite this resection, survival in the early 1970 was 10-20%. Since 1970 multi agent chemotherapy has increased survival rates. of importance, however, is these regimens have caused significant reduction of these extra pleural components so that major chest wall resections have become a rarity. With improved survival and decreased morbidity preoperative chemotherapy followed by surgery is now the accepted modality of treatment. Another major advantage of this regimen is that potential radiation therapy may be obviated. The most common chest wall lesion is rhabdomyosarcoma. In the IRS study of 1620 RMS patients, in 141 (9%) the primary lesion was in the chest wall. these are primarily alveolar histology. when lesions were superficial, wide local excision with supplemental radiation therapy was associated with low morbidity and good overall survival. however, a majority have significant intra- thoracic components. in these circumstances the resectability rate is less than 30% and the survival poor. Other lesions include non rhabdomyosarcomas, eosinophilic granuloma, chondrosarcoma, and osteomyelitis.     Before the development of mechanical suture machines, we often used the York Mason transrectal approach technique for resection of villous tumors of the rectum and small carcinomas.Three female patients who underwent this procedure in 1979, 1983 and 1986 were followed up to the present. In two of the cases (An, 28 years, and Eg, 68 years), small epitheliomas were resected (4 and 3 cm). Eg underwent radiotherapy, as she had a Malpighian tumor, and a year later plastia of the anus.Evolution was good and 14 year later a small perineal tumor appeared which was resected on may 23 1997 and turned out to be a fibrosarcoma with little cellular activity (the patient is now 81 years old).The third patient(MZ; 70 years old) had a great villous tumor which was resected in late 1986,received and was re operated with York Mason approach four years later (october 1990)At the beginning of 1994 a new received was resected by means of endoscopy, but bled profusely and has to undergo urgent surgery carrying out a perineal amputation of the rectum which was left open (Lisfranc).Histological study showed it was a tubular and villous adenocarcinoma with high mucosecretant activity.In March of the same year the perineum was closed and the sigmoidal stump was resected below the colostomy. In late 1996 an encapsulated tumor appeared with subcutaneous situation in the posterior perineum; this was resected in november 19, 1996 with local anesthesia.Microscopical findings also showed an adenocarcinoma with great production of mucine.The patient is now 80 years old and she has not relapsed.The three patients are alive; two of them have continent anus.They have been operated on for 20, 14 and 11 years

  4. Modulation of tumour spectra in C 57 black mice following fast-neutron versus Co irradiation at low dosage

    A total of 7109 C 57 Black mice of either sex were either sham-, neutron- or cobalt-irradiated at the age of 33+-3 days. The dose range extended from 3.2 to 47.2 rads for neutrons and from 18 to 141 rads for γ rays. This experiment was aimed at assessing relative biological effectiveness (RBE) and its possible dose dependence from radiation mortality and morbidity data, using actuarial statistics. The primary goal was to define the spontaneous or basic tumour spectrum in control animals of either sex, which included a major proportion of lymphocytic lymphomas, followed by reticulum cell lymphomas. A variety of other tumours were observed, among them various adenocarcinomas, squamous cell carcinomas and leiomyosarcomas of the gastrointestinal tract, liver and spleen hemangiomas, micro- and macrofollicular hyperplasia as well as papillary adenomas of the thyroid, fibromas and fibrosarcomas of connective tissue, muscles and bones, myomas and fibrosarcomas of uterus, luteomas and cysts of the ovary, papillary and transitional cell carcinomas of kidneys and an occasional mammary adenocarcinoma or skin epithelioma. Modulation of the basic tumour pattern by neutrons or cobalt was primarily operative on the reticular tissue. Age-specific incidence rates for combined mature and immature lymphocytic lymphomas decreased following neutron or cobalt irradiation at higher dose levels. In reticulum cell sarcomas, however, the tumour type evidently shifted almost entirely from the mature to the immature type. Furthermore, all rates increased markedly following irradiation at all dose levels. One major finding in all groups was the non-linearity of tumour response. Furthermore, it appeared that the control as well as the radiation-modulated tumour spectra were likely to proceed from intercompetitive stochastic processes rather than from some probabilistic random distribution.     Enteroviruses have major clinical and public health importance and are one of the leading causes of aseptic meningitis. There are many diseases with similar clinical symptoms and cerebrospinal fluid (CSF) findings of aseptic meningitis, thus virus isolation and identification is crucial for definitive diagnosis. Virological diagnosis is nonetheless important to distinguish between induced meningitis and other treatable causes of disease with a similar clinical picture. A total of 249 samples obtained from 246 cases (age range: 0-15 years), prediagnosed as aseptic meningitis, were sent to Virology Laboratory of Refik Saydam Hygiene Center. The patients were followed at Department of Pediatric Infectious Diseases in the Social Security Hospital, Ankara, Turkey, between 2001 and 2004. Stool (n: 180), CSF (n: 54) and throat swab (n: 15) samples have been inoculated to RD (rhabdomyosarcoma), Hep-2 (human epithelioma) and L20B (transgenic mice) cell lines, and followed up for the presence of cytopathic effects. A total of 95 enterovirus strains were isolated from 85 (34.6%) cases, and serotyped by using RIVM (National Institute of Public and the Environment, Nederlands) antisera with microneutralization method. As a result, the most frequently isolated types were found as echovirus type 30 (n: 24) and coxsackievirus type B (n: 19), which were most frequently isolated between July to October. This is the first report from Turkey for aseptic meningitis cases due to echovirus type 25 (n:3), 18 (n:2), 14 (n:1), 13 (n:4), 11 (n:6), 9 (n:1), 6 (n:9), 5 (n:1), 4 (n:1) and coxsackievirus type A9 (n:1).

    A single overlay plaque assay was designed and evaluated for infectious hematopoietic necrosis virus. Epithelioma papillosum carpio cells were grown in normal atmosphere with tris(hydroxymethyl)aminomethane- or HEPES (N-2-hydroxyethylpiperazine-N'-2-ethanesulfonic acid)-buffered media. Plaques were larger and formed more quickly on 1- to 3-day-old cell monolayers than on older monolayers. Cell culture medium with a 10% addition of fetal calf serum (MEM 10) or without serum (MEM 0) were the most efficient virus diluents. Dilution with phosphate-buffered saline, saline, normal broth, or deionized water reduced plaque numbers. Variations in the pH (7.0 to 8.0) of a MEM 0 diluent did not affect plaque numbers. Increasing the volume of viral inoculum above 0.15 ml (15- by 60-mm plate) decreased plaquing efficiency. Significantly more plaques occurred under gum tragacanth and methylcellulose than under agar or agarose overlays. Varying the pH (6.8 to 7.4) of methylcellulose overlays did not significantly change plaque numbers. More plaques formed under the thicker overlays of both methylcellulose and gum tragacanth. Tris(hydroxymethyl)aminomethane and HEPES performed equally well, buffering either medium or overlay. Plaque numbers were reduced when cells were rinsed after virus adsorption or less than 1 h was allowed for adsorption. Variation in adsorption time between 60 and 180 min did not change plaque numbers. The mean plaque formation time was 7 days at 16 degrees C. The viral dose response was linear when the standardized assay was used.

    The aim of the present paper is to evaluate cancer survival in patients resident in the municipality of Florence according to different deprivation levels. We used data from the Tuscan Cancer Registry and data from the national census 2001. We used a deprivation index, measured as a continue variable, classified in tertiles according to the distribution of the resident population. We compared more deprived patients (category 3) vs less deprived ones (category 1-2). 10-year relative survival has been computed for patients diagnosed with 27 different cancer sites during 1997-2002, for different deprivation categories. Cancer sites were split into three groups of the same dimension, on the basis of 10-year survival (bad, intermediate and good prognosis). For each category the relative excess risk of death (RER) for most deprived patients has been computed using a Generalized Liner Model. We evaluated also the effect of marital status, classified as married and non-married. We analysed 14 549 invasive cancer cases (out of skin epithelioma). Overall bad prognosis cancers did not show any RER of dying for most deprived patients. For intermediate prognosis cancers RER was 1.13 (1.02 ; 1.24). A excess occurs in the most disadvantaged tertile for tumors diagnosed under 50 years. For good prognosis cancers the RER was 1.06 (0.89 ; 1.26). We found a relative excess of mortality for non-married vs married. In the area of Florence there is an effect of deprivation level of survival for median-better prognosis cancers, for tumours diagnosed under 50 years and for unmarried people compared to unmarried ones.

De estas zonas reviste importancia por su cercanía a órganos vitales el canto interno ocular, sobre todo en lesiones con extensa invasión local, por la disyuntiva del compromiso ocular y la decisión de su conservación o no. Comentamos nuestra experiencia en el tratamiento de lesiones infiltrativas de canto interno, así como la secuencia de estudio y las opciones terapéuticas.Basal cell epithelioma is the most common malignant tumor affecting the skin. It is mainly located in sun-exposed areas of the body surface due to solar actinic radiation. The facial skin is the most exposed, and the one with the highest incidence of these tumors. The dermis normally acts as a barrier against deepening, a fact which does not occur in certain areas. Among these critical sites the inner canthus region has a critical interest due to its proximity to vital organs with doubts about eye involvement and its preservation. We comment our experience in the management of infiltrative tumors of the inner canthus, diagnostic steps and therapeutical alternatives.

    Full Text Available A disease in the koi carp (Cyprinus carpio koi and the common carp (Cyprinus carpio carpio, caused by the herpesvirus and accompanied by a high mortality rate, has spread across numerous fish ponds all over the world since 1998, resulting in massive mortality and significant financial losses. The herpesvirus-like virus, called the koi herpesvirus (KHV has been isolated and identified from the koi and the common carp in the course of the incidences of massive mortalities. The first appearance of a disease with a high mortality in the common and the koi carp caused by the koi herpesvirus (KHV was described in 1998 in Israel and the United States of America (USA. Since that time, a large number of cases of outbreaks of this disease have been confirmed throughout the world, including the USA, Israel, and a large number of European countries. The deaths occurred seasonally, in late spring or early autumn, when the water temperature was from 18-28ºC. The most important factor of the environment that affects the occurrence and gravity of this disease is the water temperature. This disease is currently considered one of the factors that present the biggest threat to populations of the common and the koi carp. Diseased fish are disoriented, their movements uncoordinated, their breathing rapid, gills swollen, and they have local skin lesions.     Full Text Available Se presenta un caso con diagnóstico de queratosis actínica en el área de salud "Elpidio Berovides" del municipio La Lisa. Se tiene como interés demostrar la no correlación clínico-histopatológica. Se trata de un paciente que acude a la consulta portador de una lesión en piel de 30 años de evolución, asintomático. Se indicó estudio histopatológico, y se tomaron muestras de piel en sitios diferentes de la lesión en 2 ocasiones: primera muestra con diagnóstico de epitelioma basocelular sólido, y la segunda muestra se tomó en 3 sitios diferentes de la lesión con diagnóstico definitivo de queratosis actínica. Se presenta el caso porque, a pesar de lo frecuente de la enfermedad, no es usual encontrarla en la localización en que la presenta este paciente, y menos aún en la raza negra. No debemos restarle importancia, por encontrarnos en un país tropical con un clima muy cálido y soleado.A case with diagnosis of actinic keratosis in the health area of "Elpidio Berovides", in La Lisa municipality, is presented. It is our interest to demonstrate the non-clinicohistopathological correlation. This asymptomatic patient, carrier of a skin lesion of 30 years of evolution, was seen at the physician's office. A histopathological study was indicated and samples of the skin were taken twice in different sites of the lesion: the first sample with diagnosis of solid basocellular epithelioma, and the second sample was taken in 3 different sites of the lesion with final diagnosis of actinic keratosis. Although this is a common disease, this case is presented because it is not usual to find it in the localization where this patient had it, and even less in black people. We should not understimate it, since we live in a tropical country with a very hot and sunny climate.

  11. The discrimination of some dermal dischromias using the ESR and NAA

    The information provided by the electron spin resonance (ESR) and neutron activation analysis (NAA) seem to complete each other most of time. ESR is applied in biology, due to its significant sensitivity and selectivity in the detection of paramagnetic states of free radicals implied in many physiological processes. Many enzymatic systems, photosynthesis intermediates or electrons transporting chain species are paramagnetic. On the other hand, free radicals are involved in the protection against ionic radiation, in immunity reactions and pathological processes including the oncogenesis. ESR spectroscopy informs about the quantity, interactions and dynamics of paramagnetic centres of the systems through spectral parameters as: the intensity, the g factor, the form and the width of line, hyperfine splitting, etc. NAA allows simultaneous determination of the concentration of essential microelements in tissues involved in the major biological pathways. In the present work we have tested the possibility of using the ESR and NAA investigations techniques combined with the factorial statistic analysis of the data, in the aim to discriminate some dermal dischromias of human skin. Having in view that the hyper- and hipochromia are due to an excess and deficit in melanin, respectively, we used ESR to observe the radical species from the skin samples. We studied the skin samples that represent different dischromias (malign melanoma, epithelioma, pigmentary nevus, vitiligo) in comparison with normal skin. On the same samples we have determined by NAA the concentrations of elements (Cu, Na, Mg, Mn, and Al) which could be involved in the mechanisms of producing and releasing the melanin. The signal ESR intensities and widths and the NAA elemental concentration values of the skin samples have been processed by a STATCF program to develop differences between dischromic and normal skin.     Full Text Available Após terem sido apreciadas, à luz de fatos experimentais, as principais causas determinantes das multiplicações celulares normais e aquelas responsabilidades pelas multiplicações anormais, encontradas nos tumores blastomatosos, foram analisados os tumores animais que reconhecidamente são causados por virus tais como: o sarcoma e o epitelioma das galinhas; o papiloma e o mixoma dos coelhos; os carcinomas de ratos e rãs, e finalmente, o papiloma oral e o linfo-sarcoma venéreo dos cães. Em seguida, foram focalizadas as várias propriedades em comum existentes entre os vírus indutores dêsses tumores como: capacidade de determinar multiplicações anormais nos tecidos atacados; elevada especificidade para as espécies sensíveis; existência de diminuição do poder infeccioso à medida que aumenta a capacidade de estimular a formação de tumores e, as afinidades existentes entre êles e os gens, principalmente àquelas referentes à sensibilidade aos raios X.   13. The Exploration of Virtual Non-contrast CT Spectral Imaging in Detection of Renal Lesions%能谱CT虚拟平扫在肾脏占位病变的应用探索

    Objective To evaluate whether the virtual non-contrast (VNC) spectral CT imaging can replace the true non-contrast(TNC) CT imaging to diagnose renal lesions.Methods From December 2013 to October 2015,54 patients pathologically diagnosed with renal lesions underwent gemstone spectral imaging (GSI) including pre-contrast,arterial and venous phase acquisitions.The quality of VNC images and TNC images were evaluated on a five-point scale by two radiologists who were specialized in abdominal CT diagnosis.Interobserver agreement with regards to image quality was evaluated by Cohen's kappa analysis.The contrast-to-noise ratio (CNR) of lesion-normal renal parenchyma on the TNC and VNC images were calculated,and intergroup differences were comparied by one-way ANOVA analysis.The diagnostic accuracy of the TNC images and VNCs were calculated and compared by chi-square test.Results Among the 72 renal lesions of the 54 patients,there were hypoattenuating cysts (n =24),suprarenal epithelioma (n =15),metastatic tumors (n =14),hyperattenuating cysts (n =11) and nephrapostasis (n =8).Interobserver agreement with regards to image quality was excellently coincident (Kappa >0.80).There was no significant difference among the image quality of the TNC images,arterial and venous phase VNC images.The mean CNR of lesion-normal renal parenchyma in the TNC,arterial and venous phase VNC images were 0.51 ±0.16,0.77 ± 0.21,0.65 ± 0.18,respectively.The mean CNR of arterial or venous phase VNC was significantly higher than that of TNC imaging (P < 0.05).There was no significant difference of diagnostic accuracy between the TNC images and VNCs.Conclusion The VNC spectral CT imaging may seem as a surrogate for the conventional non-contrast scan in detection of renal lesions to some degree.%目的 探讨在肾脏占位性病变诊断方面,能谱CT虚拟平扫(VNC)能否代替传统CT真实平扫(TNC).方法 回顾性分析本院2013年12月至2015年10月行常规肾脏
