  1. Structural insight into dynamic bypass of the major cisplatin-DNA adduct by Y-family polymerase Dpo4

    Y-family DNA polymerases bypass Pt-GG, the cisplatin-DNA double-base lesion, contributing to the cisplatin resistance in tumour cells. To reveal the mechanism, we determined three structures of the Y-family DNA polymerase, Dpo4, in complex with Pt-GG DNA. The crystallographic snapshots show three stages of lesion bypass: the nucleotide insertions opposite the 3{prime}G (first insertion) and 5{prime}G (second insertion) of Pt-GG, and the primer extension beyond the lesion site. We observed a dynamic process, in which the lesion was converted from an open and angular conformation at the first insertion to a depressed and nearly parallel conformation at the subsequent reaction stages to fit into the active site of Dpo4. The DNA translocation-coupled conformational change may account for additional inhibition on the second insertion reaction. The structures illustrate that Pt-GG disturbs the replicating base pair in the active site, which reduces the catalytic efficiency and fidelity. The in vivo relevance of Dpo4-mediated Pt-GG bypass was addressed by a dpo-4 knockout strain of Sulfolobus solfataricus, which exhibits enhanced sensitivity to cisplatin and proteomic alterations consistent with genomic stress.

  2. Isotopic effect in phase transitions of (NH4)2HPO4 and (ND4)2DPO4

    Specific heat and dielectric constant measurements at low frequency, have shown two transitions in the ionic systems (NH4)2HPO4 and (ND4)2DPO4 bellow 300 K. For (NH4)2HPO4, the transition are observed at 174 K and 246 K, while (ND4)2DPO4, they are observed at 147 K and 229 K. We have also found a shift of the transition temperatures to smaller values when the hydrogen is replaced by deuterium. The specific heat anomalies associated with these transitions are reversible in successive thermal cycles (heating and cooling) and we did not detect latent heat through them. Furthermore, we have detected anomalies in the dielectric constant in the same transition points. We have attributed these transition phases to reorientations of the tetrahedra of NH4 and ND4, so that the activation energy Ea for these process that we associated with the thermal energy KBTt required for the transition, is inversely related to the mass of the hydrogen isotope

  3. MsDpo4—a DinB Homolog from Mycobacterium smegmatis—Is an Error-Prone DNA Polymerase That Can Promote G:T and T:G Mismatches

    Full Text Available Error-prone DNA synthesis in prokaryotes imparts plasticity to the genome to allow for evolution in unfavorable environmental conditions, and this phenomenon is termed adaptive mutagenesis. At a molecular level, adaptive mutagenesis is mediated by upregulating the expression of specialized error-prone DNA polymerases that generally belong to the Y-family, such as the polypeptide product of the dinB gene in case of E. coli. However, unlike E. coli, it has been seen that expression of the homologs of dinB in Mycobacterium tuberculosis are not upregulated under conditions of stress. These studies suggest that DinB homologs in Mycobacteria might not be able to promote mismatches and participate in adaptive mutagenesis. We show that a representative homolog from Mycobacterium smegmatis (MsDpo4 can carry out template-dependent nucleotide incorporation and therefore is a DNA polymerase. In addition, it is seen that MsDpo4 is also capable of misincorporation with a significant ability to promote G:T and T:G mismatches. The frequency of misincorporation for these two mismatches is similar to that exhibited by archaeal and prokaryotic homologs. Overall, our data show that MsDpo4 has the capacity to facilitate transition mutations and can potentially impart plasticity to the genome.

  4. How Y-Family DNA polymerase IV is more accurate than Dpo4 at dCTP insertion opposite an N2-dG adduct of benzo[a]pyrene. (United States)

    To bypass DNA damage, cells have Y-Family DNA polymerases (DNAPs). One Y-Family-class includes DNAP κ and DNAP IV, which accurately insert dCTP opposite N(2)-dG adducts, including from the carcinogen benzo[a]pyrene (BP). Another class includes DNAP η and DNAP V, which insert accurately opposite UV-damage, but inaccurately opposite BP-N(2)-dG. To investigate structural differences between Y-Family-classes, regions are swapped between DNAP IV (a κ/IV-class-member) and Dpo4 (a η/V-class-member); the kinetic consequences are evaluated via primer-extension studies with a BP-N(2)-dG-containing template. Four key structural elements are revealed. (1) Y-Family DNAPs have discreet non-covalent contacts between their little finger-domain (LF-Domain) and their catalytic core-domain (CC-Domain), which we call "non-covalent bridges" (NCBs). Arg37 and Arg38 in DNAP IV's CC-Domain near the active site form a non-covalent bridge (AS-NCB) by interacting with Glu251 and Asp252, respectively, in DNAP IV's LF-Domain. Without these interactions dATP/dGTP/dTTP misinsertions increase. DNAP IV's AS-NCB suppresses misinsertions better than Dpo4's equivalent AS-NCB. (2) DNAP IV also suppresses dATP/dGTP/dTTP misinsertions via a second non-covalent bridge, which is ∼8Å from the active site (Distal-NCB). Dpo4 has no Distal-NCB, rendering it inferior at dATP/dGTP/dTTP suppression. (3) dCTP insertion is facilitated by the larger minor groove opening near the active site in DNAP IV versus Dpo4, which is sensible given that Watson/Crick-like [dCTP:BP-N(2)-dG] pairing requires the BP-moiety to be in the minor groove. (4) Compared to Dpo4, DNAP IV has a smaller major groove opening, which suppresses dGTP misinsertion, implying BP-N(2)-dG bulk in the major groove during Hoogsteen syn-adduct-dG:dGTP pairing.   5. Engineering of Bacteriophages Y2::dpoL1-C and Y2::luxAB for Efficient Control and Rapid Detection of the Fire Blight Pathogen, Erwinia amylovora. (United States)

    Erwinia amylovora is the causative agent of fire blight, a devastating plant disease affecting members of the Rosaceae Alternatives to antibiotics for control of fire blight symptoms and outbreaks are highly desirable, due to increasing drug resistance and tight regulatory restrictions. Moreover, the available diagnostic methods either lack sensitivity, lack speed, or are unable to discriminate between live and dead bacteria. Owing to their extreme biological specificity, bacteriophages are promising alternatives for both aims. In this study, the virulent broad-host-range E. amylovora virus Y2 was engineered to enhance its killing activity and for use as a luciferase reporter phage, respectively. Toward these aims, a depolymerase gene of E. amylovora virus L1 ( dpoL1-C ) or a bacterial luxAB fusion was introduced into the genome of Y2 by homologous recombination. The genes were placed downstream of the major capsid protein orf68 , under the control of the native promoter. The modifications did not affect viability of infectivity of the recombinant viruses. Phage Y2:: dpoL1-C demonstrated synergistic activity between the depolymerase degrading the exopolysaccharide capsule and phage infection, which greatly enhanced bacterial killing. It also significantly reduced the ability of E. amylovora to colonize the surface of detached flowers. The reporter phage Y2:: luxAB transduced bacterial luciferase into host cells and induced synthesis of large amounts of a LuxAB luciferase fusion. After the addition of aldehyde substrate, bioluminescence could be readily monitored, and this enabled rapid and specific detection of low numbers of viable bacteria, without enrichment, both in vitro and in plant material.     Sulfolobus solfataricus DNA polymerase IV (Dpo4), a prototype Y-family DNA polymerase, contains a unique little finger domain besides a catalytic core. Here, we report the chemical shift assignments for the backbone nitrogens, α and β carbons, and amide protons of the little finger domain of Dpo4. This work and our published backbone assignment for the catalytic core provide the basis for investigating the conformational dynamics of Dpo4 during catalysis using solution NMR spectroscopy.

  9. Identification of an Unfolding Intermediate for a DNA Lesion Bypass Polymerase (United States)

    Sulfolobus solfataricusDNA Polymerase IV (Dpo4), a prototype Y-family DNA polymerase, has been well characterized biochemically and biophysically at 37 °C or lower temperatures. However, the physiological temperature of the hyperthermophile S. solfataricus is approximately 80 °C. With such a large discrepancy in temperature, the in vivo relevance of these in vitro studies of Dpo4 has been questioned. Here, we employed circular dichroism spectroscopy and fluorescence-based thermal scanning to investigate the secondary structural changes of Dpo4 over a temperature range from 26 to 119 °C. Dpo4 was shown to display a high melting temperature characteristic of hyperthermophiles. Unexpectedly, the Little Finger domain of Dpo4, which is only found in the Y-family DNA polymerases, was shown to be more thermostable than the polymerase core. More interestingly, Dpo4 exhibited a three-state cooperative unfolding profile with an unfolding intermediate. The linker region between the Little Finger and Thumb domains of Dpo4 was found to be a source of structural instability. Through site-directed mutagenesis, the interactions between the residues in the linker region and the Palm domain were identified to play a critical role in the formation of the unfolding intermediate. Notably, the secondary structure of Dpo4 was not altered when the temperature was increased from 26 to 87.5 °C. Thus, in addition to providing structural insights into the thermal stability and an unfolding intermediate of Dpo4, our work also validated the relevance of the in vitro studies of Dpo4 performed at temperatures significantly lower than 80 °C.     The diffuse pulmonary ossification (DPO) is a rare disease characterized by metaplastic formation of bony tissue in the lung parenchyma. Generally it is associated with other disorders as mitral stenosis and interstitial fibrosis. Sometimes the DPO is idiopathic. We report the case of a 49-year-old man who presented with cough and hemoptysis. The radiological findings suggested an interstitial lung disease. The fiber optic bronchoscopy was normal. The pulmonary function tests showed a mild airway obstruction. The bacteriological and serological studies for tuberculosis, mycosis, and collagen-vascular disease were negatives. An open lung biopsy was performed and the pathologic findings were interpreted as diffuse pulmonary ossification (DPO). Any other disease was found; so, in this case the DPO was classified as idiopathic

  12. Economic dispatch optimization algorithm based on particle diffusion

    Highlights: • A dispatch model that considers fuel, emissions control and wind power cost is built. • An optimization algorithm named diffusion particle optimization (DPO) is proposed. • DPO was used to analyze the impact of wind power risk and emissions on dispatch. - Abstract: Due to the widespread installation of emissions control equipment in fossil fuel-fired power plants, the cost of emissions control needs to be considered, together with the plant fuel cost, in providing economic power dispatch of those units to the grid. On the other hand, while using wind power decreases the overall power generation cost for the power grid, it poses a risk to a traditional grid, because of its inherent stochastic characteristics. Therefore, an economic dispatch optimization model needs to consider all of the fuel cost, emissions control cost and wind power cost for each of the generating unit conforming the fleet that meets the required grid power demand. In this study, an optimization algorithm referred as diffusion particle optimization (DPO) is proposed to solve such complex optimization problem. In this algorithm, Brownian motion theory is used to guide the movement of particles so that the particles can search for an optimal solution over the entire definition region. Several benchmark functions and power grid system data were used to test the performance of DPO, and compared to traditional algorithms used for economic dispatch optimization, such as, particle swarm optimization and artificial bee colony algorithm. It was found that DPO has less probability to be trapped in local optimums. According to results of different power systems DPO was able to find economic dispatch solutions with lower costs. DPO was also used to analyze the impact of wind power risk and fossil unit emissions coefficients on power dispatch. The result are encouraging for the use of DPO as a dynamic tool for economic dispatch of the power grid.

  13. [Cointegration test and variance decomposition for the relationship between economy and environment based on material flow analysis in Tangshan City Hebei China]. (United States)


    The material flow account of Tangshan City was established by material flow analysis (MFA) method to analyze the periodical characteristics of material input and output in the operation of economy-environment system, and the impact of material input and output intensities on economic development. Using econometric model, the long-term interaction mechanism and relationship among the indexes of gross domestic product (GDP) , direct material input (DMI), domestic processed output (DPO) were investigated after unit root hypothesis test, Johansen cointegration test, vector error correction model, impulse response function and variance decomposition. The results showed that during 1992-2011, DMI and DPO both increased, and the growth rate of DMI was higher than that of DPO. The input intensity of DMI increased, while the intensity of DPO fell in volatility. Long-term stable cointegration relationship existed between GDP, DMI and DPO. Their interaction relationship showed a trend from fluctuation to gradual ste adiness. DMI and DPO had strong, positive impacts on economic development in short-term, but the economy-environment system gradually weakened these effects by short-term dynamically adjusting indicators inside and outside of the system. Ultimately, the system showed a long-term equilibrium relationship. The effect of economic scale on economy was gradually increasing. After decomposing the contribution of each index to GDP, it was found that DMI's contribution grew, GDP's contribution declined, DPO's contribution changed little. On the whole, the economic development of Tangshan City has followed the traditional production path of resource-based city, mostly depending on the material input which caused high energy consumption and serous environmental pollution.

    Full Text Available Protein-DNA recognition is a central biological process that governs the life of cells. A protein will often undergo a conformational transition to form the functional complex with its target DNA. The protein conformational dynamics are expected to contribute to the stability and specificity of DNA recognition and therefore may control the functional activity of the protein-DNA complex. Understanding how the conformational dynamics influences the protein-DNA recognition is still challenging. Here, we developed a two-basin structure-based model to explore functional dynamics in Sulfolobus solfataricus DNA Y-family polymerase IV (DPO4 during its binding to DNA. With explicit consideration of non-specific and specific interactions between DPO4 and DNA, we found that DPO4-DNA recognition is comprised of first 3D diffusion, then a short-range adjustment sliding on DNA and finally specific binding. Interestingly, we found that DPO4 is under a conformational equilibrium between multiple states during the binding process and the distributions of the conformations vary at different binding stages. By modulating the strength of the electrostatic interactions, the flexibility of the linker, and the conformational dynamics in DPO4, we drew a clear picture on how DPO4 dynamically regulates the DNA recognition. We argue that the unique features of flexibility and conformational dynamics in DPO4-DNA recognition have direct implications for low-fidelity translesion DNA synthesis, most of which is found to be accomplished by the Y-family DNA polymerases. Our results help complete the description of the DNA synthesis process for the Y-family polymerases. Furthermore, the methods developed here can be widely applied for future investigations on how various proteins recognize and bind specific DNA substrates.

  15. Investigation of aluminum and iron deposition on metaplastic bones in three patients with diffuse pulmonary ossification. (United States)

    Diffuse pulmonary ossification (DPO) is a rare pulmonary lesion. DPO is typically detected at autopsy rather than premortem. Recently, however, several cases were diagnosed antemortem using computed tomography, high-resolution computed tomography, or video-assisted thoracic surgery. In the present study, we evaluated DPO at autopsy from two patients with post-myocardial infarction (cases 1 and 3) and one patient with duodenal cancer (case 2). Multiple metaplastic bones (nodular in case 1 and 3 or dendriform in case 2) were detected in these three cases. In an attempt to detect aluminum and iron deposition in these metaplastic bones, histochemical investigations were performed. The two nodular types of one and three cases were positive for aluminum and iron, but the dendriform type of case 2 was positive only for aluminum. The depositions occurred in a linear pattern along the calcifying front. It is of great interest that these deposition patterns were similar to those of bones from three previously reported DPO cases and from the bones of hemodialysis patients. It is suggested that these abnormal metal depositions in the calcifying front might disturb the normal mineralization processes of the metaplastic bones, although no morphological abnormality was detected, except for dense black color of calcifying front lines. Further investigations are needed in more patients with DPO to obtain more information on this topic.

  16. Chronology of early embryonic development and embryo uterine migration in alpacas. (United States)

    The objectives were to: (1) describe the chronology of early embryonic development from ovulation to entry into the uterus; and (2) to determine the timing of embryo migration to the left uterine horn when ovulation occurred from the right ovary. The experiment was conducted in Peru. Females (n = 132) were randomly assigned to 15 experimental groups. All females were mated to an intact male, given 50 μg GnRH im (Cystorelin) and ovulation time determined by transrectal ultrasonography, conducted every 6 hours, starting 24 hours postmating. Animals were slaughtered at a specific intervals postovulation and reproductive tracts were recovered and subjected to oviductal and uterine flushing for females slaughtered between 1 and 6 days postovulation (dpo; Day 0 = ovulation) and uterine flushing for females slaughtered from 7 to 15 dpo for recovery of oocytes/embryos. Season of mating did not influence the interval from mating to ovulation (winter: 29 ± 6 hours vs. summer: 30 ± 6 hours; P = 0.49). Ovulation rates for females mated during winter and summer were 92% versus 100%, respectively (P = 0.05). Fertilization rates for winter and summer mated females were 72% and 82% (P = 0.29). Unfertilized ova were not retained in the uterine tube. All embryos collected were in the uterine tube ipsilateral to the side of ovulation between 1 and 5 dpo. Embryos reached the uterus on 6 dpo. Embryos began to elongate on 9 dpo; at this time, 83% of embryos derived from right-ovary ovulations were collected from the left uterine horn. Embryos occupied the entire uterine cavity by 10 dpo. In conclusion, we characterized early embryo development and location of embryo during its early developmental stages in alpaca. This was apparently the first report regarding chronology of embryo development and migration to the left horn in alpaca which merits further investigation regarding its role in maternal recognition of pregnancy.   18. Visible light cure characteristics of a cycloaliphatic polyester dimethacrylate alternative oligomer to bisGMA. (United States)

    Objective : The goal of this study was to characterize the light curing characteristics of a new oligomer PEM-665 designed to be used as an alternative monomer to BisGMA. Materials and methods : PEM-665 (P) and BisGMA (B) solutions were prepared with triethylene glycol dimethacrylate (T) diluent in different weight proportions (70/30 and 50/50). Solutions containing 70% P and 30% T were designated as 70PT, 70%B and 30%T as 70BT, 50%P and 50%T as 50PT and 50%B and 50%T as 50BT. The initiators were CQ (EDMAB was used as amine accelerator for CQ) and DPO in 1% concentration. Eight solutions were prepared in a factorial design: 70PT/DPO; 70PT/CQ; 50PT/DPO; 50PT/CQ; 70BT/DPO; 70BT/CQ; 50BT/DPO; 50BT/CQ. BISCO VIP visible light was used to cure the monomer solutions using 30 s exposure time and 400 W power setting. TA Instruments Differential Scanning Calorimeter (DSC 2910) was used to determine the heat of cure (J/g) during polymerization at 37 °C, from which molar heat of cure (kJ/mole) and %Conversion values were estimated. Results : Range of mean values as a function monomer selections were: heat of cure (J/g): 161.7 for 70PT/DPO system to 198.6 for 50BT/CQ system; molar heat of cure (kJ/mole): 67.3 for 70BT/DPO to 78.86 for 50PT/CQ; % conversion: 59.9 for 70BT/DPO to 70.3 for 50PT/CQ. Analysis of variance and Tukey HSD pairwise contrast showed statistically significant differences between % conversion means of PEM and BisGMA mixtures, with PEM mixtures showing significantly higher mean values. Conclusions : The results suggest that PEM-665 is a promising candidate material for dental polymer applications.

  20. Gamma irradiation of mushrooms, preliminary studies: effect on O-diphenyl oxidase activity and amino acid content

    Mushrooms are a valuable food raw materials because of their nutritional and taste values. Post-harvest ripening, chemical composition (94% water) and possible microbial contamination decrease not only organoleptic and nutritional value, but also the shelf-life. As an objective method of evaluation of irradiated mushrooms we adopted activity determination of o-diphenyl oxidase (o-DPO) which is responsible for discoloration of the edible mushrooms and altered qualitative and quantitative content of amino acids. It was observed that doses up to 2 kGy did not cause any increase in the activity of o-DPO; irradiation also did not affect the taste. Mushrooms irradiated with doses up to 4 kGy were of good quality after 5 days of storage at 4 C, while the control samples (unirradiated) after the same time were considerably changed, probably due too post-harvest ripening. Immediately after exposure the activity of o-DPO increased in proportion to the dose used. During subsequent storage, however, no increase in o-DPO activity was observed. Irradiation used in the range from 0.2 to 0.4 kGy did not affect the nutritional value of the raw material.   3. Mechanistic Basis for the Bypass of a Bulky DNA Adduct Catalyzed by a Y-Family DNA Polymerase (United States)

    1-Nitropyrene (1-NP), an environmental pollutant, induces DNA damage in vivo and is considered to be carcinogenic. The DNA adducts formed by the 1-NP metabolites stall replicative DNA polymerases but are presumably bypassed by error-prone Y-family DNA polymerases at the expense of replication fidelity and efficiency in vivo. Our running start assays confirmed that a site-specifically placed 8-(deoxyguanosin-N2-yl)-1-aminopyrene (dG1,8), one of the DNA adducts derived from 1-NP, can be bypassed by Sulfolobus solfataricus DNA polymerase IV (Dpo4), although this representative Y-family enzyme was paused strongly by the lesion. Pre-steady-state kinetic assays were employed to determine the low nucleotide incorporation fidelity and establish a minimal kinetic mechanism for the dG1,8 bypass by Dpo4. To reveal a structural basis for dCTP incorporation opposite dG1,8, we solved the crystal structures of the complexes of Dpo4 and DNA containing a templating dG1,8 lesion in the absence or presence of dCTP. The Dpo4·DNA-dG1,8 binary structure shows that the aminopyrene moiety of the lesion stacks against the primer/template junction pair, while its dG moiety projected into the cleft between the Finger and Little Finger domains of Dpo4. In the Dpo4·DNA-dG1,8·dCTP ternary structure, the aminopyrene moiety of the dG1,8 lesion, is sandwiched between the nascent and junction base pairs, while its base is present in the major groove. Moreover, dCTP forms a Watson–Crick base pair with dG, two nucleotides upstream from the dG1,8 site, creating a complex for “-2” frameshift mutation.   4. Some quantitative indicators of postovulatory aging and its effect on larval and juvenile development of Atlantic salmon (Salmo salar). (United States)

    Modern out-of-season egg production in Atlantic salmon (Salmo salar) increases the risk of postovulatory aging (POA) of oocytes. Postovulatory aging is known to influence oocyte quality in salmonids, but reliable tests for POA are lacking in Atlantic salmon egg production. To address this problem, we have collected oocytes from the same 20 Atlantic salmon females sequentially in approximately 1-week intervals, from the start of ovulation until 28 days postovulation (dpo), to determine the effect of natural retention of matured oocytes in body coelomic cavity on further performance of embryos and juveniles produced from those oocytes. Also, we investigated oocyte water hardening and several coelomic fluid parameters as potential quantitative indicators of POA. Oocyte quality decreased significantly from 22 dpo onward, as inferred from decrease in fertilization success and survival of embryos, alevins, and juveniles and increase in alevin and juvenile deformity rates. The occurrence of head deformities was significantly related to postovulatory age of oocytes. Coelomic fluid pH decreased significantly at 28 dpo and correlated positively with fertilization rates (r = 0.45), normal eyed embryo rates (r = 0.67), and alevin relative survival rates (r = 0.63) and negatively correlated with total alevin deformity rates (r = -0.59). Oocyte weight gain at 60 minutes decreased significantly at 28 dpo and correlated negatively with total alevin deformities and the occurrence of cranial nodules (r = -0.99). Generally, quality of ovulated oocytes remained stable for the first 2 weeks after ovulation. Later on, POA negatively influenced Atlantic salmon embryo, alevin, and juvenile performance. For the first time, we show a long-term effect of POA on salmonid juvenile performance. Standardized pH measurements of coelomic fluid could potentially improve embryo and juvenile production by identifying low-quality oocytes at an early stage during the production. Copyright © 2015

  5. Mechanistic Investigation of the Bypass of a Bulky Aromatic DNA Adduct Catalyzed by a Y-family DNA Polymerase (United States)

    3-Nitrobenzanthrone (3-NBA), a nitropolyaromatic hydrocarbon (NitroPAH) pollutant in diesel exhaust, is a potent mutagen and carcinogen. After metabolic activation, the primary metabolites of 3-NBA react with DNA to form dG and dA adducts. One of the three major adducts identified is N-(2’-deoxyguanosin-8-yl)-3-aminobenzanthrone (dGC8-N-ABA). This bulky adduct likely stalls replicative DNA polymerases but can be traversed by lesion bypass polymerases in vivo. Here, we employed running start assays to show that a site-specifically placed dGC8-N-ABA is bypassed in vitro by Sulfolobus solfataricus DNA polymerase IV (Dpo4), a model Y-family DNA polymerase. However, the nucleotide incorporation rate of Dpo4 was significantly reduced opposite both the lesion and the template position immediately downstream from the lesion site, leading to two strong pause sites. To investigate the kinetic effect of dGC8-N-ABA on polymerization, we utilized pre-steady-state kinetic methods to determine the kinetic parameters for individual nucleotide incorporations upstream, opposite, and downstream from the dGC8-N-ABA lesion. Relative to the replication of the corresponding undamaged DNA template, both nucleotide incorporation efficiency and fidelity of Dpo4 were considerably decreased during dGC8-N-ABA lesion bypass and the subsequent extension step. The lower nucleotide incorporation efficiency caused by the lesion is a result of a significantly reduced dNTP incorporation rate constant and modestly weaker dNTP binding affinity. At both pause sites, nucleotide incorporation followed biphasic kinetics with a fast and a slow phase and their rates varied with nucleotide concentration. In contrast, only the fast phase was observed with undamaged DNA. A kinetic mechanism was proposed for the bypass of dGC8-N-ABA bypass catalyzed by Dpo4. PMID:25048879

    In July 1994, the Executive Director for Operations of the US Nuclear Regulatory Commission (NRC) appointed a Special Review Panel to assess the Differing Professional View or Opinion (DPV/DPO) process, including its effectiveness, how well it is understood by employees, and the organizational climate for having such views aired and properly decided. An additional area within this review was to address the effectiveness of the DPO procedures as they pertain to public access and confidentiality. Further, the Panel was charged with the review of the submittals completed since the last review to identify employees who made significant contributions to the agency or to the public health and safety but had not been adequately recognized for this contribution. The report presents the Special Review Panel's evaluation of the NRC's current process for dealing with Differing Professional Views or Opinions. Provided in this report are the results of an employee opinion survey on the process; highlights and suggestions from interviews with individuals who had submitted a Differing Professional View or Opinion, as well as with agency managers directly involved with the Differing Professional Views or Opinions process; and the Special Review Panel's recommendations for improving the DPV/DPO process

  8. Sensitivity of the ureter to radiation before and after ureterojejuno-implantation

    The investigation refers to dexhal ureterosigmoidostomies, carried out on 15 dogs. The implanted right ureters and the left ureters left in the bladders of 5 dogs were then treated with radiation of a total focal dose of 60 Gy. The investigation referred to the comparison of different ureter modalities and their sensibility to radiation in respect to statements on the various sorts of therapy strategies. The test animals which has been only operated on were killed 68 d.p.o at earliest and 295 d.p.o. The survival time of the 5 animals which has been operated on and irradiated was 156-269 d.p.o. and 22-59 d after the last irradiation, respectively. While the non-altered ureters remained without pathological findings, there were anastomotic stenoses with ascending infections of the upper urinary tract in 40% of the ureters only implanted into the sigma and another increase in the anastomotic stenoses (80%) was observed in the ureters which had been implanted and irradiated with a total focal dose of 60 Gy. It was also shown by means of X-ray method, macro and micropathology, that those ureters which had only been irradiated tolerated 30 and 60 Gy, resp., well and that the implanted ureters with subradical total focal dose of 30 Gy showed exact passage conditions in the i.v. urogram.   9. Assessing environmental risks for high intensity agriculture using the material flow analysis method--a case study of the Dongting Lake basin in South Central China. (United States)

    This study primarily examined the assessment of environmental risk in high intensity agricultural areas. Dongting Lake basin was taken as a case study, which is one of the major grain producing areas in China. Using data obtained from 1989 to 2012, we applied Material Flow Analysis (MFA) to show the material consumption, pollutant output and production storage in the agricultural-environmental system and assessed the environmental risk index on the basis of the MFA results. The results predicted that the status of the environmental quality of the Dongting Lake area is unsatisfactory for the foreseeable future. The direct material input (DMI) declined by 13.9%, the domestic processed output (DPO) increased by 28.21%, the intensity of material consumption (IMC) decreased by 36.7%, the intensity of material discharge (IMD) increased by 10%, the material productivity (MP) increased by 27 times, the environmental efficiency (EE) increased by 15.31 times, and the material storage (PAS) increased by 0.23%. The DMI and DPO was higher at rural places on the edge of cities, whereas the risk of urban agriculture has arisen due to the higher increasing rate of DMI and DPO in cities compared with the counties. The composite environmental risk index increased from 0.33 to 0.96, indicating that the total environmental risk changed gradually but seriously during the 24 years assessed. The driving factors that affect environmental risk in high intensity agriculture can be divided into five classes: social, economic, human, natural and disruptive incidents. This study discussed a number of effective measures for protecting the environment while ensuring food production yields. Additional research in other areas and certain improvements of this method in future studies may be necessary to develop a more effective method of managing and controlling agricultural-environmental interactions.

  10. Global conformational dynamics of a Y-family DNA polymerase during catalysis.

    Full Text Available Replicative DNA polymerases are stalled by damaged DNA while the newly discovered Y-family DNA polymerases are recruited to rescue these stalled replication forks, thereby enhancing cell survival. The Y-family DNA polymerases, characterized by low fidelity and processivity, are able to bypass different classes of DNA lesions. A variety of kinetic and structural studies have established a minimal reaction pathway common to all DNA polymerases, although the conformational intermediates are not well defined. Furthermore, the identification of the rate-limiting step of nucleotide incorporation catalyzed by any DNA polymerase has been a matter of long debate. By monitoring time-dependent fluorescence resonance energy transfer (FRET signal changes at multiple sites in each domain and DNA during catalysis, we present here a real-time picture of the global conformational transitions of a model Y-family enzyme: DNA polymerase IV (Dpo4 from Sulfolobus solfataricus. Our results provide evidence for a hypothetical DNA translocation event followed by a rapid protein conformational change prior to catalysis and a subsequent slow, post-chemistry protein conformational change. Surprisingly, the DNA translocation step was induced by the binding of a correct nucleotide. Moreover, we have determined the directions, rates, and activation energy barriers of the protein conformational transitions, which indicated that the four domains of Dpo4 moved in a synchronized manner. These results showed conclusively that a pre-chemistry conformational change associated with domain movements was too fast to be the rate-limiting step. Rather, the rearrangement of active site residues limited the rate of correct nucleotide incorporation. Collectively, the conformational dynamics of Dpo4 offer insights into how the inter-domain movements are related to enzymatic function and their concerted interactions with other proteins at the replication fork.

    Full Text Available Abstract Background The main hypothesis of this study is that oral heme iron polypeptide (HIP; Proferrin® ES administration will more effectively augment iron stores in erythropoietic stimulatory agent (ESA-treated peritoneal dialysis (PD patients than conventional oral iron supplementation (Ferrogradumet®. Methods Inclusion criteria are peritoneal dialysis patients treated with darbepoietin alpha (DPO; Aranesp®, Amgen for ≥ 1 month. Patients will be randomized 1:1 to receive either slow-release ferrous sulphate (1 tablet twice daily; control or HIP (1 tablet twice daily for a period of 6 months. The study will follow an open-label design but outcome assessors will be blinded to study treatment. During the 6-month study period, haemoglobin levels will be measured monthly and iron studies (including transferring saturation [TSAT] measurements will be performed bi-monthly. The primary outcome measure will be the difference in TSAT levels between the 2 groups at the end of the 6 month study period, adjusted for baseline values using analysis of covariance (ANCOVA. Secondary outcome measures will include serum ferritin concentration, haemoglobin level, DPO dosage, Key's index (DPO dosage divided by haemoglobin concentration, and occurrence of adverse events (especially gastrointestinal adverse events. Oenococcus oeni (O. oeni, as the one of most important microorganisms in wine industry and the most resistant lactic acid bacteria (LAB species to ethanol, has not been investigated regarding the selection of stable reference genes for RT-qPCR normalization under ethanol stress conditions. In this study, nine candidate reference genes (proC, dnaG, rpoA, ldhD, ddlA, rrs, gyrA, gyrB, and dpoIII were analyzed to determine the most stable reference genes for RT-qPCR in O. oeni SD-2a under different ethanol stress conditions (8, 12, and 16% (v/v ethanol. The transcript stabilities of these genes were evaluated using the algorithms geNorm, NormFinder, and BestKeeper. The results showed that dnaG and dpoIII were selected as the best reference genes across all experimental ethanol conditions. Considering single stress experimental modes, dpoIII and dnaG would be suitable to normalize expression level for 8% ethanol shock treatment, while the combination of gyrA, gyrB, and rrs would be suitable for 12% ethanol shock treatment. proC and gyrB revealed the most stable expression in 16% ethanol shock treatment. This study selected and validated for the first time the reference genes for RT-qPCR normalization in O. oeni SD-2a under ethanol stress conditions.

    Full Text Available Abstract Background Many changes in gene expression occur in distal stumps of injured nerves but the transcriptional control of these events is poorly understood. We have examined the expression of the transcription factors ATF3 and c-Jun by non-neuronal cells during Wallerian degeneration following injury to sciatic nerves, dorsal roots and optic nerves of rats and mice, using immunohistochemistry and in situ hybridization. Results Following sciatic nerve injury – transection or transection and reanastomosis – ATF3 was strongly upregulated by endoneurial, but not perineurial cells, of the distal stumps of the nerves by 1 day post operation (dpo and remained strongly expressed in the endoneurium at 30 dpo when axonal regeneration was prevented. Most ATF3+ cells were immunoreactive for the Schwann cell marker, S100. When the nerve was transected and reanastomosed, allowing regeneration of axons, most ATF3 expression had been downregulated by 30 dpo. ATF3 expression was weaker in the proximal stumps of the injured nerves than in the distal stumps and present in fewer cells at all times after injury. ATF3 was upregulated by endoneurial cells in the distal stumps of injured neonatal rat sciatic nerves, but more weakly than in adult animals. ATF3 expression in transected sciatic nerves of mice was similar to that in rats. Following dorsal root injury in adult rats, ATF3 was upregulated in the part of the root between the lesion and the spinal cord (containing Schwann cells, beginning at 1 dpo, but not in the dorsal root entry zone or in the degenerating dorsal column of the spinal cord. Following optic nerve crush in adult rats, ATF3 was found in some cells at the injury site and small numbers of cells within the optic nerve displayed weak immunoreactivity.     Full Text Available PURPOSE: To evaluate the effects of fat embolism in organs such as lung and liver. METHODS: Twenty rabbits underwent autologous intramuscular fat grafting in the right thigh after liposuction. The groups were determined according to the postoperative day that occurred in euthanasia: 60, 90 and 120 day. Then, lung and liver were excised and sent to the histopathology laboratory for analysis in search of late injury secondary to a prior event of fat embolism in the tissues. RESULTS: The results showed a change in the macroscopic sample with discoloration of the liver tissue heterogeneously. There were no changes consistent with embolic effect under the microscope. CONCLUSION: The option of performing a technique of fat grafting with a less traumatic surgical procedure can be considered protective against embolic events, with no impact to late embolic events on the tissues.OBJETIVO: Avaliar a repercussão da embolia gordurosa em órgãos como pulmão e fígado. MÉTODOS: Vinte coelhos foram submetidos à enxertia autóloga intramuscular de gordura em coxa direita após lipoaspiração. Os grupos foram determinados conforme os dias pós-operatórios (DPO em que ocorreu a eutanásia: 60 DPO, 90DPO, 120 DPO. Em seguida, o pulmão e o fígado foram ressecados e encaminhados ao laboratório de histopatologia para análise em busca de lesão tardia secundária a evento de embolia gordurosa prévia nos tecidos. RESULTADOS: Foi evidenciada uma alteração na amostra a analise macroscópica com alteração da coloração do tecido hepático de forma heterogênea. Não houve alterações compatíveis com repercussão de processo embólico à microscopia. CONCLUSÃO: A opção pela realização de uma técnica de lipoenxertia menos traumática e com pequeno tempo cirúrgico pode ser considerada protetora para eventos embólicos, não havendo repercussão embólica a tardiamente.

    Full Text Available Phytoplasmas and Xylella spp. are bacteria that cause many economically important plant diseases worldwide. TaqMan probe-based quantitative real-time polymerase chain reaction (qPCR assays have been utilized to universally detect phytoplasmas or Xylella fastidiosa. To develop a superior universal qPCR method, we used a dual priming oligonucleotide (DPO with two annealing sites as a reverse primer to target the well-conserved bacterial 16S rDNA. The new qPCR assays universally detected various species of phytoplasmas and subspecies of X. fastidiosa as well as Xylella taiwanensis, and generally showed superior threshold cycle values when amplifying specific or non-specific products compared to current universal qPCR assays. The proposed qPCR assays were integrated to develop a multiplex qPCR assay that simultaneously detected phytoplasmas, Xylella spp., and an internal plant DNA positive control within 1 hour. This assay could detect a minimum of ten bacterial cells and was compatible with crude extractions used in the rapid screening of various plants. The amplicons were of sufficient lengths to be directly sequenced for preliminary identification, and the primers could be used in universal conventional PCR assays. Additionally, reverse DPO primers can be utilized to improve other probe-based qPCR assays.

    Full Text Available Pesquisa avaliativa sobre a gestão municipal do Programa Nacional de Alimentação Escolar (PNAE nos dez maiores municípios de Santa Catarina, Brasil. A matriz de indicadores contempla duas dimensões: político-organizacional (DPO, com as subdimensões recursos, atuação intersetorial e controle social; e técnico-operacional (DTO, com as subdimensões eficácia alimentar e nutricional, monitoramento alimentar e nutricional, e atuação pedagógica para a alimentação saudável. Foram elencados 22 indicadores e os dados foram coletados por meio de entrevistas com nutricionistas do PNAE. A DPO resultou em 60% dos casos regulares e 40% ruins, e a DTO obteve 90% dos casos ruins, consequência do baixo desempenho no monitoramento e na atuação pedagógica. Embora os casos avaliados tenham resultados pouco satisfatórios, a avaliação oferece dados importantes sobre a gestão do PNAE. Ressalta-se a importância de avaliações com esse caráter como possibilidade de alertar os gestores para a revisão e qualificação das ações.

No 3° DPO a lesão do grupo experimental apresentou menor tecido necrótico, menor esfacelo e maior tecido de granulação em relação ao grupo controle positivo. No 7° e 10º DPO, a lesão do grupo experimental apresentou menor esfacelo em relação ao grupo controle positivo. A análise microscópica das lesões revelou no 5º DPO maior fibroplasia no grupo experimental em relação aos grupos controles; no 14º DPO ocorreu menor neovascularização no grupo experimental e maior formação de folículos pilosos no grupo controle negativo. O extrato hidroalcoólico de S. oleraceus propiciou reparo tecidual obedecendo a padrões fisiológicos normais e confirmando os dados empíricos de sua utilização.

    Full Text Available Identifying etiologies of acute febrile illnesses (AFI is challenging due to non-specific presentation and limited availability of diagnostics. Prospective AFI studies provide a methodology to describe the syndrome by age and etiology, findings that can be used to develop case definitions and multiplexed diagnostics to optimize management. We conducted a 3-year prospective AFI study in Puerto Rico. Patients with fever ≤7 days were offered enrollment, and clinical data and specimens were collected at enrollment and upon discharge or follow-up. Blood and oro-nasopharyngeal specimens were tested by RT-PCR and immunodiagnostic methods for infection with dengue viruses (DENV 1-4, chikungunya virus (CHIKV, influenza A and B viruses (FLU A/B, 12 other respiratory viruses (ORV, enterovirus, Leptospira spp., and Burkholderia pseudomallei. Clinical presentation and laboratory findings of participants infected with DENV were compared to those infected with CHIKV, FLU A/B, and ORV. Clinical predictors of laboratory-positive dengue compared to all other AFI etiologies were determined by age and day post-illness onset (DPO at presentation. Of 8,996 participants enrolled from May 7, 2012 through May 6, 2015, more than half (54.8%, 4,930 had a pathogen detected. Pathogens most frequently detected were CHIKV (1,635, 18.2%, FLU A/B (1,074, 11.9%, DENV 1-4 (970, 10.8%, and ORV (904, 10.3%. Participants with DENV infection presented later and a higher proportion were hospitalized than those with other diagnoses (46.7% versus 27.3% with ORV, 18.8% with FLU A/B, and 11.2% with CHIKV.     Y-Family DNA polymerases are known to bypass DNA lesions in vitro and in vivo. Sulfolobus solfataricus DNA polymerase (Dpo4) was chosen as a model Y-family enzyme for investigating the mechanism of DNA synthesis in single crystals. Crystals of Dpo4 in complexes with DNA (the binary complex) in the presence or absence of an incoming nucleotide were analyzed by Raman microscopy. (13)C- and (15)N-labeled d*CTP, or unlabeled dCTP, were soaked into the binary crystals with G as the templating base. In the presence of the catalytic metal ions, Mg(2+) and Mn(2+), nucleotide incorporation was detected by the disappearance of the triphosphate band of dCTP and the retention of *C modes in the crystal following soaking out of noncovalently bound C(or *C)TP. The addition of the second coded base, thymine, was observed by adding cognate dTTP to the crystal following a single d*CTP addition. Adding these two bases caused visible damage to the crystal that was possibly caused by protein and/or DNA conformational change within the crystal. When d*CTP is soaked into the Dpo4 crystal in the absence of Mn(2+) or Mg(2+), the primer extension reaction did not occur; instead, a ternary protein·template·d*CTP complex was formed. In the Raman difference spectra of both binary and ternary complexes, in addition to the modes of d(*C)CTP, features caused by ring modes from the template/primer bases being perturbed and from the DNA backbone appear, as well as features from perturbed peptide and amino acid side chain modes. These effects are more pronounced in the ternary complex than in the binary complex. Using standardized Raman intensities followed as a function of time, the C(*C)TP population in the crystal was maximal at ∼20 min. These remained unchanged in the ternary complex but declined in the binary complexes as chain incorporation occurred.

    Full Text Available PURPOSE: To study the influence of albumin on changes of liver function in the extrahepatic biliary obstruction through an experimental model in rats. METHODS: Sixty rats were divided into four groups: Group C (Control: 6 animals. Group M (Fictitious Operation: 18 rats underwent laparotomy and handling of the bile ducts; Groups O (extrahepatic biliary obstruction and A (Treated with 2% albumin: 18 animals in each group underwent ligation of the ductus liver; The animals in groups M, O and A were divided into three subgroups of 6 animals each to be killed in the 7, 14 and 21 days postoperative (POD. Blood was drawn for determination of total bilirubin (TB, indirect bilirubin (IB, direct bilirubin (DB, alkaline phosphatase (ALP, aspartate aminotransferase (AST and alanine aminotransferase (ALT. RESULTS: On POD 7, BI levels were 4.5 mg / dl in group O and 2.1 mg / dl in group A (p = 0.025. On the 14th POD, the levels of PA were 1185.2 U / l in the group and O 458.3 U / l in group A (p = 0.004. ALT levels were 101.7 U / l in the group O and 75.7 U / l in group A (= 0.037.     DNA replication in cells is performed in the presence of four dNTPs and four rNTPs. In this study, we re-evaluated the fidelity of DNA polymerases using the general misincorporation frequency consisting of three incorrect dNTPs and four rNTPs but not using the traditional special misincorporation frequency with only the three incorrect dNTPs. We analyzed both the general and special misincorporation frequencies of nucleotide incorporation opposite dG, rG, or 8-oxoG by Pseudomonas aeruginosa phage 1 (PaP1) DNA polymerase Gp90 or Sulfolobus solfataricus DNA polymerase Dpo4. Both misincorporation frequencies of other DNA polymerases published were also summarized and analyzed. The general misincorporation frequency is obviously higher than the special misincorporation frequency for many DNA polymerases, indicating the real fidelity of a DNA polymerase should be evaluated using the general misincorporation frequency.     In order to study the flow of water in an underground oil reservoir, tritiated water was injected in a well and, subsequently, tritium was determined in the water produced by the surrounding wells. Since the specific radioactivity of the water produced by the more remote wells appeared to be rather low, partly owing to dilution of the tritium water by the water present in the formation, enrichment methods were used, in order to increase counting sensitivity. Three methods of enrichment were examined: exchange with ethanol, conversion to toluene, and electrolysis. The latter method proved to be the most useful. The tritium was determined with a liquid scintillation counter; the scintillator consisted of a toluene-ethanol (2:1) mixture with DPO and POPOP, the water sample being dissolved in this liquid. Some statistical problems in connexion with the minimum detectable specific activity and the reduction of background are discussed briefly.   10. Toxicity of bovicin HC5 against mammalian cell lines and the role of cholesterol in bacteriocin activity. (United States)

    Bacteriocins are ribosomally synthesized antimicrobial peptides produced by Bacteria and some Archaea. The assessment of the toxic potential of antimicrobial peptides is important in order to apply these peptides on an industrial scale. The aim of the present study was to investigate the in vitro cytotoxic and haemolytic potential of bovicin HC5, as well as to determine whether cholesterol influences bacteriocin activity on model membranes. Nisin, for which the mechanism of action is well described, was used as a reference peptide in our assays. The viability of three distinct eukaryotic cell lines treated with bovicin HC5 or nisin was analysed by using the MTT assay and cellular morphological changes were determined by light microscopy. The haemolytic potential was evaluated by using the haemoglobin liberation assay and the role of cholesterol on bacteriocin activity was examined by using model membranes composed of DOPC (1,2-dioleoyl-sn-glycero-3-phosphocholine) and DPoPC (1,2-dipalmitoyl-sn-glycero-3-phosphocholine). The IC(50) of bovicin HC5 and nisin against Vero cells was 65.42 and 13.48 µM, respectively. When the MTT assay was performed with MCF-7 and HepG2 cells, the IC(50) obtained for bovicin HC5 was 279.39 and 289.30 µM, respectively, while for nisin these values were 105.46 and 112.25 µM. The haemolytic activity of bovicin HC5 against eukaryotic cells was always lower than that determined for nisin. The presence of cholesterol did not influence the activity of either bacteriocin on DOPC model membranes, but nisin showed reduced carboxyfluorescein leakage in DPoPC membranes containing cholesterol. In conclusion, bovicin HC5 only exerted cytotoxic effects at concentrations that were greater than the concentration needed for its biological activity, and the presence of cholesterol did not affect its interaction with model membranes.

    Full Text Available OBJETIVO: Estudar o efeito da correção volêmica com diferentes tipos de solução, na mucosa do intestino delgado de ratos. MÉTODOS: Foram utilizados 120 ratos Wistar (Rattus norvegicus albinus, machos, adultos, com peso individual entre 310 e 410g, oriundos do Instituto Evandro Chagas de Belém do Pará, submetidos a período de adaptação por 15 dias, recebendo água e ração ad libitum, durante todo o experimento. Os animais foram distribuídos em: Grupo Padrão (P, Grupo Choque (C, Grupo Solução Fisiológica (SF e Grupo Solução Hipertônica (SH, com 30 animais cada. Estes foram divididos em subgrupos com 10 animais cada, de acordo com o dia de pós-operatório (DPO previsto para a eutanásia dos animais, (1masculine, 3masculine ou 7masculine DPO, sendo após esta, colhido material para realização de teste de absorvância pelo MTT em todos os animais. RESULTADOS: O grupo SF apresentou menores índices de viabilidade celular comparado aos grupos SH e C (p<0.05. CONCLUSÃO: A correção volêmica com solução de cloreto de sódio a 7.5% levou a manutenção de maior quantidade de células viáveis, no intestino delgado em ratos no 7masculine dia do experimento.

    Full Text Available OBJETIVO: Comparar os efeitos dos exercícios de respiração profunda (ERP e espirômetro de incentivo a fluxo (EI em pacientes submetidos a cirurgia de revascularização do miocárdio (CRVM por meio das seguintes variáveis: capacidade vital forçada - CVF, volume expiratório forçado de primeiro segundo - VEF1, pressões respiratórias máximas e saturação de oxigênio. MÉTODOS: Trinta e seis pacientes foram submetidos a aplicação de ventilação não-invasiva por dois períodos de 30 minutos durante as primeiras 24 horas pós-extubação e distribuídos randomicamente em dois grupos, a saber: ERP (n=18 e EI (n=18, sendo as variáveis espirométricas avaliadas no pré-operatório e sétimo dia pós-operatório (DPO e a força da musculatura respiratória e saturação de oxigênio no pré-operatório, primeiro, segundo e sétimo DPO. RESULTADOS: Os grupos foram considerados homogêneos em relação às variáveis demográficas e cirúrgicas. Observouse queda dos valores referentes à CVF e VEF1 entre o préoperatório e o sétimo DPO, porém sem diferença estatística entre os grupos. Houve queda das pressões respiratórias máximas no primeiro DPO, mas com restabelecimento gradual e parcial até o sétimo DPO, também sem diferença estatística entre os grupos. A saturação de oxigênio foi a única variável restabelecida no sétimo DPO, mas também sem diferença estatística entre os grupos.     Full Text Available PURPOSE: To evaluate the effects of S-methylisothiourea hemisulfate (SMT on the healing of colonic anastomosis in rats. METHODS: Sixty rats Wistar were distributed into two groups of 30 animals: experimental (E and control C. The animals of experimental group received intraperitoneal SMT at 50mg/kg/dose every 12 hours for 72 hours. The control group received intraperitoneal saline at the same volume of SMT. The rats were subdivided into subgroups groups of 10 for euthanasia on the third, seventh, and 14th postoperative days (POD. We evaluated clinical and weight evolution, breaking strength and histopathology; also, a blood sample was collected for serum dosage of nitrite/nitrate. RESULTS: There was more vascular neoformation (p=0.006 and granulation (p=0.002 in the E3 group, and more mononuclear infiltrates in the C3 group (p=0.041. There was also more edema in the C14 group (p=0.008. There was no statistically significant difference in breaking strength, nitrite/nitrate dosage, and the remaining histopathological parameters.     Consensus mechanism is the core of the cryptocurrency system which is based on the block chain technology. This paper analyzes and compares the three typical consensus mechanisms to provide references and suggestions for the design of consensus mechanism for block chain developers. The basic idea, operation process and advantages and disadvantages of the three mainstream consensus mechanisms, PoW, PoS and DPoS, are analyzed. Then the characteristics of each consensus mechanism are summarized from the aspects of fairness, running cost and sustainability. And then we analyze and compare them from the credit to the degree of decentralization, authorization and other aspects, consider the consensus needs under the different application scenarios.     Full Text Available Abstract Background To gain insight into the branching order of the five main lineages currently recognized in the green algal class Chlorophyceae and to expand our understanding of chloroplast genome evolution, we have undertaken the sequencing of chloroplast DNA (cpDNA from representative taxa. The complete cpDNA sequences previously reported for Chlamydomonas (Chlamydomonadales, Scenedesmus (Sphaeropleales, and Stigeoclonium (Chaetophorales revealed tremendous variability in their architecture, the retention of only few ancestral gene clusters, and derived clusters shared by Chlamydomonas and Scenedesmus. Unexpectedly, our recent phylogenies inferred from these cpDNAs and the partial sequences of three other chlorophycean cpDNAs disclosed two major clades, one uniting the Chlamydomonadales and Sphaeropleales (CS clade and the other uniting the Oedogoniales, Chaetophorales and Chaetopeltidales (OCC clade. Although molecular signatures provided strong support for this dichotomy and for the branching of the Oedogoniales as the earliest-diverging lineage of the OCC clade, more data are required to validate these phylogenies. We describe here the complete cpDNA sequence of Oedogonium cardiacum (Oedogoniales. Results Like its three chlorophycean homologues, the 196,547-bp Oedogonium chloroplast genome displays a distinctive architecture. This genome is one of the most compact among photosynthetic chlorophytes. It has an atypical quadripartite structure, is intron-rich (17 group I and 4 group II introns, and displays 99 different conserved genes and four long open reading frames (ORFs, three of which are clustered in the spacious inverted repeat of 35,493 bp. Intriguingly, two of these ORFs (int and dpoB revealed high similarities to genes not usually found in cpDNA.   17. Synthesis, structure, and properties of a series of chiral tweezer-diamine complexes consisting of an achiral zinc(II) bisporphyrin host and chiral diamine guest: induction and rationalization of supramolecular chirality. (United States)

    We report here the synthesis, structure, and spectroscopic properties of a series of supramolecular chiral 1:1 tweezer-diamine complexes consisting of an achiral Zn(II) bisporphyrin (Zn2DPO) host and five different chiral diamine guests, namely, (R)-diaminopropane (DAP), (1S,2S)-diaminocyclohexane (CHDA), (S)-phenylpropane diamine (PPDA), (S)-phenyl ethylenediamine (PEDA), and (1R,2R)-diphenylethylene diamine (DPEA). The solid-state structures are preserved in solution, as reflected in their (1)H NMR spectra, which also revealed the remarkably large upfield shifts of the NH2 guest protons with the order Zn2DPO·DAP > Zn2DPO·CHDA > Zn2DPO·PPDA> Zn2DPO·PEDA ≫ Zn2DPO·DPEA, which happens to be the order of binding constants of the respective diamines with Zn2DPO. As the bulk of the substituent at the chiral center of the guest ligand increases, the Zn-Nax distance of the tweezer-diamine complex also increases, which eventually lowers the binding of the guest ligand toward the host. Also, the angle between the two porphyrin rings gradually increases with increasing bulk of the guest in order to accommodate the guest within the bisporphyrin cavity with minimal steric clash. The notably high amplitude bisignate CD signal response by Zn2DPO·DAP, Zn2DPO·CHDA, and Zn2DPO·PPDA can be ascribed to the complex's high stability and the formation of a unidirectional screw as observed in the X-ray structures of the complexes. A relatively lower value of CD amplitude shown by Zn2DPO·PEDA is due to the lower stability of the complex. The projection of the diamine binding sites of the chiral guest would make the two porphyrin macrocycles oriented in either a clockwise or anticlockwise direction in order to minimize host-guest steric clash.   18. Quantitative Analysis of the Mutagenic Potential of 1-Aminopyrene-DNA Adduct Bypass Catalyzed by Y-Family DNA Polymerases (United States)

    N- (deoxyguanosin-8-yl)-1-aminopyrene (dGAP) is the predominant nitro polyaromatic hydrocarbon product generated from the air pollutant 1-nitropyrene reacting with DNA. Previous studies have shown that dGAP induces genetic mutations in bacterial and mammalian cells. One potential source of these mutations is the error-prone bypass of dGAP lesions catalyzed by the low-fidelity Y-family DNA polymerases. To provide a comparative analysis of the mutagenic potential of the translesion DNA synthesis (TLS) of dGAP, we employed short oligonucleotide sequencing assays (SOSAs) with the model Y-family DNA polymerase from Sulfolobus solfataricus, DNA Polymerase IV (Dpo4), and the human Y-family DNA polymerases eta (hPolη), kappa (hPolκ), and iota (hPolι). Relative to undamaged DNA, all four enzymes generated far more mutations (base deletions, insertions, and substitutions) with a DNA template containing a site-specifically placed dGAP. Opposite dGAP and at an immediate downstream template position, the most frequent mutations made by the three human enzymes were base deletions and the most frequent base substitutions were dAs for all enzymes. Based on the SOSA data, Dpo4 was the least error-prone Y-family DNA polymerase among the four enzymes during the TLS of dGAP. Among the three human Y-family enzymes, hPolκ made the fewest mutations at all template positions except opposite the lesion site. hPolκ was significantly less error-prone than hPolι and hPolη during the extension of dGAP bypass products. Interestingly, the most frequent mutations created by hPolι at all template positions were base deletions. Although hRev1, the fourth human Y-family enzyme, could not extend dGAP bypass products in our standing start assays, it preferentially incorporated dCTP opposite the bulky lesion. Collectively, these mutagenic profiles suggest that hPolkk and hRev1 are the most suitable human Y-family DNA polymerases to perform TLS of dGAP in humans. PMID:22917544

    Identifying etiologies of acute febrile illnesses (AFI) is challenging due to non-specific presentation and limited availability of diagnostics. Prospective AFI studies provide a methodology to describe the syndrome by age and etiology, findings that can be used to develop case definitions and multiplexed diagnostics to optimize management. We conducted a 3-year prospective AFI study in Puerto Rico. Patients with fever ≤7 days were offered enrollment, and clinical data and specimens were collected at enrollment and upon discharge or follow-up. Blood and oro-nasopharyngeal specimens were tested by RT-PCR and immunodiagnostic methods for infection with dengue viruses (DENV) 1-4, chikungunya virus (CHIKV), influenza A and B viruses (FLU A/B), 12 other respiratory viruses (ORV), enterovirus, Leptospira spp., and Burkholderia pseudomallei. Clinical presentation and laboratory findings of participants infected with DENV were compared to those infected with CHIKV, FLU A/B, and ORV. Clinical predictors of laboratory-positive dengue compared to all other AFI etiologies were determined by age and day post-illness onset (DPO) at presentation. Of 8,996 participants enrolled from May 7, 2012 through May 6, 2015, more than half (54.8%, 4,930) had a pathogen detected. Pathogens most frequently detected were CHIKV (1,635, 18.2%), FLU A/B (1,074, 11.9%), DENV 1-4 (970, 10.8%), and ORV (904, 10.3%). Participants with DENV infection presented later and a higher proportion were hospitalized than those with other diagnoses (46.7% versus 27.3% with ORV, 18.8% with FLU A/B, and 11.2% with CHIKV). Predictors of dengue in participants presenting dengue in participants presenting 3-5 DPO were leukopenia, thrombocytopenia, facial/neck erythema, nausea, eye pain, signs of poor circulation, and diarrhea; presence of rhinorrhea, cough, and red conjunctiva predicted non-dengue AFI.   1. Outcome prediction in gastroschisis - The gastroschisis prognostic score (GPS) revisited. (United States)

    The GPS enables risk stratification for gastroschisis and helps discriminate low from high morbidity groups. The purpose of this study was to revalidate GPS's characterization of a high morbidity group and to quantify relationships between the GPS and outcomes. With REB approval, complete survivor data from a national gastroschisis registry was collected. GPS bowel injury scoring was revalidated excluding the initial inception/validation cohorts (>2011). Length of stay (LOS), 1st enteral feed days (dFPO), TPN days (dTPN), and aggregate complications (COMP) were compared between low and high morbidity risk groups. Mathematical relationships between outcomes and integer increases in GPS were explored using the entire cohort (2005-present). Median (range) LOS, dPO, and dTPN for the entire cohort (n=849) was 36 (26,62), 13 (9,18), and 27 (20,46) days, respectively. High-risk patients (GPS≥2; n=80) experienced significantly worse outcomes than low risk patients (n=263). Each integer increase in GPS was associated with increases in LOS and dTPN by 16.9 and 12.7days, respectively (pGPS effectively discriminates low from high morbidity risk groups. Within the high risk group, integer increases in GPS produce quantitatively differentiated outcomes which may guide initial counseling and resource allocation. IIb. Copyright © 2017 Elsevier Inc.   2. Economy-wide material input/output and dematerialization analysis of Jilin Province (China). (United States)

    In this paper, both direct material input (DMI) and domestic processed output (DPO) of Jilin Province in 1990-2006 were calculated and then based on these two indexes, a dematerialization model was established. The main results are summarized as follows: (1) both direct material input and domestic processed output increase at a steady rate during 1990-2006, with average annual growth rates of 4.19% and 2.77%, respectively. (2) The average contribution rate of material input to economic growth is 44%, indicating that the economic growth is visibly extensive. (3) During the studied period, accumulative quantity of material input dematerialization is 11,543 x 10(4) t and quantity of waste dematerialization is 5,987 x10(4) t. Moreover, dematerialization gaps are positive, suggesting that the potential of dematerialization has been well fulfilled. (4) In most years of the analyzed period, especially 2003-2006, the economic system of Jilin Province represents an unsustainable state. The accelerated economic growth relies mostly on excessive resources consumption after the Revitalization Strategy of Northeast China was launched.

El control, en su estrecho vínculo con la planeación, se ha visto obligado a evolucionar en la misma medida en que han evolucionado los sistemas de planificación hasta llegar a la Dirección Estratégica. En esta evolución la Dirección Estratégica se ha nutrido de elementos importantes de modelos de planeación precedentes, como son la DPO y la Planeación Estratégica. Resulta evidente que esta misma situación le ocurra al control estratégico, apareciendo en su concepción elementos de sistemas de control tradicionales o de gestión, que incorporan aquellos que marcan sus diferencias y superioridad respecto a los anteriores.

    Highlights: • We report the new fabrication approach for producing a self- assembled Mn dpoed Ge quantum dots (QDs) on SiO{sub 2} thin film with a Curie temperature above room temperature. These magnetic QDs are crystalline, monodisperse and have a well-defined shape and a controlled size. The investigation opens new routes for elaboration of self-assembled magnetic nanocrystals - Abstract: In this study, we demonstrate an original elaboration route for producing a Mn-doped Ge self-assembled quantum dots on SiO{sub 2} thin layer for MOS structure. These magnetic quantum dots are elaborated using dewetting phenomenon at solid state by Ultra-High Vacuum (UHV) annealing at high temperature of an amorphous Ge:Mn (Mn: 40%) nanolayer deposed at very low temperature by high-precision Solid Source Molecular Beam Epitaxy on SiO{sub 2} thin film. The size of quantum dots is controlled with nanometer scale precision by varying the nominal thickness of amorphous film initially deposed. The magnetic properties of the quantum-dots layer have been investigated by superconducting quantum interference device (SQUID) magnetometry. Atomic force microscopy (AFM), x-ray energy dispersive spectroscopy (XEDS) and transmission electron microscopy (TEM) were used to examine the nanostructure of these materials. Obtained results indicate that GeMn QDs are crystalline, monodisperse and exhibit a ferromagnetic behavior with a Curie temperature (TC) above room temperature. They could be integrated into spintronic technology.

  5. Shaker-related voltage-gated K+ channel expression and vasomotor function in human coronary resistance arteries. (United States)

    K V channels are important regulators of vascular tone, but the identity of specific K V channels involved and their regulation in disease remain less well understood. We determined the expression of K V 1 channel subunits and their role in cAMP-mediated dilation in coronary resistance arteries from subjects with and without CAD. HCAs from patients with and without CAD were assessed for mRNA and protein expression of K V 1 channel subunits with molecular techniques and for vasodilator response with isolated arterial myography. Assays of mRNA transcripts, membrane protein expression, and vascular cell-specific localization revealed abundant expression of K V 1.5 in vascular smooth muscle cells of non-CAD HCAs. Isoproterenol and forskolin, two distinct cAMP-mediated vasodilators, induced potent dilation of non-CAD arterioles, which was inhibited by both the general K V blocker 4-AP and the selective K V 1.5 blocker DPO-1. The cAMP-mediated dilation was reduced in CAD and was accompanied by a loss of or reduced contribution of 4-AP-sensitive K V channels. K V 1.5, as a major 4-AP-sensitive K V 1 channel expressed in coronary VSMCs, mediates cAMP-mediated dilation in non-CAD arterioles. The cAMP-mediated dilation is reduced in CAD coronary arterioles, which is associated with impaired 4-AP-sensitive K V channel function. © 2017 John Wiley & Sons Ltd.

  6. Tumour oxygen dynamics measured simultaneously by near-infrared spectroscopy and 19F magnetic resonance imaging in rats

    Simultaneous near-infrared spectroscopy (NIRS) and magnetic resonance imaging (MRI) were used to investigate the correlation between tumour vascular oxygenation and tissue oxygen tension dynamics in rat breast 13762NF tumours with respect to hyperoxic gas breathing. NIRS directly detected global variations in the oxygenated haemoglobin concentration (Δ[HbO 2 ]) within tumours and oxygen tension (pO 2 ) maps were achieved using 19 F MRI of the reporter molecule hexafluorobenzene. Multiple correlations were examined between rates and magnitudes of vascular (Δ[HbO 2 ]) and tissue (pO 2 ) responses. Significant correlations were found between response to oxygen and carbogen breathing using either modality. Comparison of results for the two methods showed a correlation between the vascular perfusion rate ratio and the mean pO 2 values (R 2 > 0.7). The initial rates of increase of Δ[HbO 2 ] and the slope of dynamic pO 2 response, d(pO 2 )/dt, of well-oxygenated voxels in response to hyperoxic challenge were also correlated. These results demonstrate the feasibility of simultaneous measurements using NIRS and MRI. As expected, the rate of pO 2 response to oxygen is primarily dependent upon the well perfused rather than poorly perfused vasculature

  7. Disability inclusion in higher education in Uganda: Status and strategies. (United States)

    Uganda has embraced inclusive education and evidently committed itself to bringing about disability inclusion at every level of education. Both legal and non-legal frameworks have been adopted and arguably are in line with the intent of the Convention on the Rights of Persons with Disabilities (CRPD) on education. The CRPD, in Article 24, requires states to attain a right to education for persons with disabilities without discrimination and on the basis of equal opportunities at all levels of education. Despite Uganda's robust disability legal and policy framework on education, there is evidence of exclusion and discrimination of students with disabilities in the higher education institutions. The main objective of this article is to explore the status of disability inclusion in higher education and strategies for its realisation, using evidence from Emong's study, workshop proceedings where the authors facilitated and additional individual interviews with four students with disabilities by the authors. The results show that there are discrimination and exclusion tendencies in matters related to admissions, access to lectures, assessment and examinations, access to library services, halls of residence and other disability support services. The article recommends that institutional policies and guidelines on support services for students with disabilities and special needs in higher education be developed, data on students with disabilities collected to help planning, collaboration between Disabled Peoples Organisations (DPO's) strengthened to ensure disability inclusion and the establishment of disability support centres.

  8. Biodistribution of radioactive organocations and cationic technetium complexes - implications to the uptake mechanism of potential heart-affine radiopharmaceuticals

    The biodistribution of various 14 C labeled organocations (ammonium, phosphonium) and cationic technetium complexes (TcCl 2 DMPE 2 ,TcDMPE 3 + ) was studied in rats to find potential relations between the chemical composition, structure, molecular size, lipophilicity and heart uptake. All of the radioactive cations were distributed in a similar manner, but with different biokinetics. Molecules containing several electronegative atoms in their ligand structure, e.g. nitrogen or oxygen which enhance polarity, were rapidly accumulated, but washed out again with resultant transient and diminished heart uptake. More highly lipophilic materials, by contrast, showed more extensive accumulation and persistent retention in the heart. The effect of human serum albumin on heart uptake was studied in a perfused heart model. Strong albumin binding prevented extensive heart uptake. This undesirable effect was neutralized by adding detergents to the perfusion bath. Evidence from these animal experiments was considered in the development of a new heart-seeking technetium compound (Tc-DPO), which is characterized by rapid accumulation in the human heart and substantially delayed washout. Heart images with minimal overlap of the inferoposterior wall and the right liver lobe were available in no more than 10 minutes p.i. in both planar and SPECT modes. A deficit in the inferior region was much more clearly defined than on the corresponding Thallium image. (Author)

  9. Structural, optical and morphological studies of Cd2+ doping in CH3NH3PbI3 perovskite semiconductor at Pb2+ site for photovoltaic applications (United States)

    Doping of semiconductors in a controlled mannner have paramount technological importance as far as the optical and electronic properties of the devices are concerned. Hybrid organic-inorganic perovskites (HOPs) as intrinsic semiconductors have sensational properties required for both the solar photovoltaics and perovskite light emitting diodes. However, undoped and complexity in the dpoing process of HOPs have limited their exploitation in the field of elcronics. In this papper we present the synthesis of HOP semiconductor (CH3NH3PbI3) doped in Pb2+ position by Cd2+. We studied the effect of the incorporation of Cd2+ into the crystalline structure and analysed the changes in the properties like crystal structure, optical absorption and the surface morphology. The structure of HOPs confirmed by X-ray diffraction analysis is tetragonal perovskite type. It can be found that the crystallinity of the samples was enhanced with the doping concentration as the intensity of diffraction peaks were observed to increase with doping. The absorption spectra as obtained from UV-Visible spectrophotometry and Tauc plot analysis indicated that the band gap observed (1.73 eV) is direct type and gets reduced to 1.67 eV with the doping concentration. The red shift may be due to the increase in the size of nanocrystalline material with doping.

This system can detect the cauliflower mosaic virus 35S (CaMV 35S), terminator of nopaline synthase gene (NOS), figwort mosaic virus 35S (FMV 35S) promoter, neomycin phosphotransferaseII (NPTII), Bt Cry 1Ab, phosphinothricin acetyltransferase genes (bar) and Streptomyces viridochromogenes (pat) simultaneously, which covers more than 90% of all authorized GMO species worldwide. This system exhibits a high tolerance to annealing temperatures, high specificity and a limit of detection equal to conventional PCR. A total of 214 samples from markets, national entry-exit agencies, the Institute for Reference Materials and Measurement (IRMM) and the American Oil Chemists' Society (AOCS) were also tested for applicability.     Full Text Available A Celulose Nipo-Brasileira é uma das maiores produtoras de celulose kraft branqueada de eucalipto no Brasil. Produz 860.000 tsa/ano em duas linhas, que são equipadas com digestores contínuos. Ambas as linhas fabricam polpa ECF (Elemental Chlorine Free com as seqüências Dhot(EOPD(EPD e D(EOPDP, respectivamente, na linha 1 e 2. A fábrica tem tratamento do efluente com lodo ativado com dois tanques de aeração com capacidade para 20.000 m³, equipados com aeradores superficiais seguidos por quatro clarificadores secundários (dois para cada reator. Nas últimas décadas, a fábrica tem otimizado e vem mudando seus processos, a fim de melhorar a preservação ambiental. Com o objetivo de reduzir o volume de efluente, DQO e carga de AOX, a seqüência Ahot(EOPD(PO proposta foi avaliada em testes laboratoriais, com reciclagem de filtrado parcial. Este artigo propôs a reciclagem de filtrado, que reduz o volume de efluente da fábrica em 9 m³/tsa (tonelada secada ao ar, isto é, mais ou menos 50% do total. O filtrado recuperado é parcialmente desviado para o ciclo de recuperação e para o estágio de deslignificação oxigênio. A reutilização do filtrado Ahot no ciclo de recuperação é para substituir os filtrados, atualmente usados para lavar lama de cal e "dregs". O impacto dos NPEs no ciclo de cálcio não foi significante. Essa estratégia permitiu uma recuperação de carga alcalina de 12 kg NaOH/tsa de polpa, que, do contrário, seria perdida. A branqueabilidade da polpa e a sua qualidade não foram afetadas significativamente.   12. A phase I study of dexosome immunotherapy in patients with advanced non-small cell lung cancer

    Full Text Available Abstract Background There is a continued need to develop more effective cancer immunotherapy strategies. Exosomes, cell-derived lipid vesicles that express high levels of a narrow spectrum of cell proteins represent a novel platform for delivering high levels of antigen in conjunction with costimulatory molecules. We performed this study to test the safety, feasibility and efficacy of autologous dendritic cell (DC-derived exosomes (DEX loaded with the MAGE tumor antigens in patients with non-small cell lung cancer (NSCLC. Methods This Phase I study enrolled HLA A2+ patients with pre-treated Stage IIIb (N = 4 and IV (N = 9 NSCLC with tumor expression of MAGE-A3 or A4. Patients underwent leukapheresis to generate DC from which DEX were produced and loaded with MAGE-A3, -A4, -A10, and MAGE-3DPO4 peptides. Patients received 4 doses of DEX at weekly intervals. Results Thirteen patients were enrolled and 9 completed therapy. Three formulations of DEX were evaluated; all were well tolerated with only grade 1–2 adverse events related to the use of DEX (injection site reactions (N = 8, flu like illness (N = 1, and peripheral arm pain (N = 1. The time from the first dose of DEX until disease progression was 30 to 429+ days. Three patients had disease progression before the first DEX dose. Survival of patients after the first DEX dose was 52–665+ days. DTH reactivity against MAGE peptides was detected in 3/9 patients. Immune responses were detected in patients as follows: MAGE-specific T cell responses in 1/3, increased NK lytic activity in 2/4. Conclusion Production of the DEX vaccine was feasible and DEX therapy was well tolerated in patients with advanced NSCLC.     Full text: Mutations of genes Dpo (dehiscing pods) and A (anthocyanin synthesis) played a role in pea domestication. A number of other genes were important in cultivar development for 3 types of usage (dry seeds, green vegetable types, fodder), e.g. fn, fna, le, p, v, fas and af. New genes (induced and spontaneous), are important for present ideotypes and are registered by the Pisum Genetics Association (PGA). Comparison of a pea variety ideotype with the variation available in gene banks shows that breeders need 'new' features. In mutation induction experiments, genotype, mutagen and method of treatment (e.g. combined or fractionated doses) are varied for broadening the mutation spectrum and selecting more genes of agronomic value. New genes are genetically analysed. In Poland, some mutant varieties with the gene afila were registered, controlling lodging by a shorter stem and a higher number of internodes. Really non-lodging pea varieties could strongly increase seed yield. But the probability of detecting a major gene for lodging resistance is low. Therefore, mutant genes with smaller influence on plant architecture are sought, to combine their effect by crossing. Promising seem to be the genes rogue, reductus and arthritic as well as a number of mutant genes not yet genetically identified. The gene det for terminal inflorescence - similarly to Vicia faba - changes plant development. Utilisation of assimilates and ripening should be better. Improvement of harvest index should give higher seed yield. A number of genes controlling disease resistance are well known (eg. Fw, Fnw, En, mo and sbm). Important in mass screening of resistance are closely linked gene markers. Pea gene banks collect respective lines, but mutants induced in highly productive cultivars would be better. Within the frame of a system-defined approach the research identifies and describes the individual- typological features of the phenomena interesting to us. The works that focus on the study of self-realization are subjected to the analysis, as the result it is noted that in this field there are still gaps in the understanding of this issue. Thus, the study from the perspective of the influence of professional identity on the successful self-realization of the personality is recognized as relevant. In the framework of this study the following assumptions are put forward: there is a relationship between the professional identity and successful self- realization of the personality; the level of formation of professional identity has an impact on the success of personality self-realization; the professional identity not fully formed acts as a barrier for creativity, activity, internality and constructive self- realization; fully formed professional identity provides a high success rate of self-realization. The purpose of this study was to investigate the features of self- realization and professional identity of psychologists-teachers. For the study, we used “Multidimensional questionnaire of personality self-realization” (MQPS by S.I. Kudinov, for the study of professional identity the following methods were used: “Professional identity-marginalism” questionnaire by E.P. Ermolaeva and Methods of measuring professional identity (MMPI by L.B. Schneider. The results obtained were subjected to quantitative analysis.   15. Neutron diffraction study of crystal structures of deuterated glycinium phosphite in paraelectric and ferroelectric phases

    Crystal structure of deuterated glycinium phosphite was studied in the paraelectric (P) phase at 348 K and in the ferroelectric (F) phase at 223 K by means of the single crystal neutron diffraction. Deuteration rate is estimated to be 0.939 by the least-squares refinement. In the P phase, quasi-one-dimensional hydrogen bond chains are built by mutually linking the DPO 3 2- anions through two different types of hydrogen bonds with the bond angles of 179.2 and 171.6deg. Two independent deuterons within the hydrogen bonds forming the chains are disordered over two sites separated by 0.545 and 0.539A. In the F phase, they order at a position nearly equal to one of two sites related by the disorder in the P phase. With the ordering of the deuterons, the P-O bonds with covalently bonded deuteron elongate, and those without covalently bonded deuteron reduce their lengths to some extend from the values determined in the P phase. Two oxygens involved in the hydrogen bond with the bond angle 179.2deg exhibits especially large displacements in the F phase. This suggests strongly an importance of this hydrogen bond in the polarization appearance and in the ferroelectric transition. Comparison with results of non-deuterated salt indicates that only the hydrogen bonds forming the chains show significant isotope shift. In particular, the hydrogen bond with the bond angle 179.2deg exhibits the most pronounced shift on the angle parameter defined by the angle between the line connecting two sites of disordered proton or deuteron and the line connecting two oxygens involved in the hydrogen bond.   16. Vernakalant hydrochloride for the treatment of atrial fibrillation. (United States)

    Atrial fibrillation (AF) is the most common arrhythmia encountered in clinical practice. Rhythm control strategy for AF is limited by drug toxicity and side effects, and recent trials have shown that this strategy is not superior to a rate control one. New antiarrhythmic drugs, free of undesired effects, would enhance rhythm control, with the possibility of sinus rhythm restoration and maintenance. A promising find in the search for new antiarrhythmic therapies is atrial-tissue specific ion channels. The findings that the ultrarapid delayed rectifier (I(Kur)) and the inwardly rectifying, acetylcholine-regulated current (I(K-Ach)) exist in atrial but not ventricular tissue increase the probability that atrioselective drugs without ventricular proarrhythmic toxicity can be developed for treatment of patients with AF. There are also other potential targets for atrial-selective therapy: transient outward current (I(to)), rapidly and slowly activating delayed rectifier currents (I(Kr) and I(Ks)), atrial sodium current (I(Na)) and atrially expressed connexins. New drugs under development with promising atrial-selectivity include: tertiapin, NIP-142, NIP-141, JTV-519, AVE0118, AVE1231, DPO-1, AZD7009 and many others. Among such new agents, vernakalant hydrochloride is currently in an advanced phase of development and has already been evaluated in clinical trials. In this overview, we describe the history and current state of developmental process of the drug, as well as its mechanism of action and influence on electrophysiological parameters. Vernakalant seems to be effective in terminating recent-onset AF, but is not efficacious in long-lasting AF and atrial flutter. The drug may be relatively free of proarrhythmic effects, and exerts a protective effect on ventricular tissue and ventricular repolarization. It is expected that the intravenous formulation will soon be approved for the pharmacological termination of recent-onset AF.

    Full Text Available This article is based on the pilot study "Workplaces for All" conducted in Macedonia in 2006. Another article by Prof. Risto Petrov, based on a part of the same study was published in Journal of Special Education and Rehabilitation January-June No. 1-2, Skopje, 2006; 1-123.The study was a qualitative research project con­ducted in co-operation between Macedonian and Norwegian scientists and representatives from a Macedonian disabled people's organisation (DPO. The main objective was to get research based knowledge about working conditions among people with disabilities in Macedonia in order to prepare for further research, and to raise awareness about the importance of joining forces to improve the rights and working conditions for people with dis­abilities.The pilot study included qualitative interviews of employers and employees at seven protective com­panies. The study has shown the advantage of hav­ing partners from different organisations working together in a research project. The findings do not unveil any irregularities or mistreatment of this quite vulnerable group of employees.The pilot study has produced research based docu­mentation about employment and working condi­tions among people with disabilities in Macedonia. Even though the study found the conditions quite positive, it unveiled several challenges that should be looked deeper into. Among these are the use of assistive technology and adaptations, the social in­clusion both in the company and in the local society and the regulations and process of receiving bene­fits by the protective companies.

Masalah yang terjadi adalah adanya defect sol sandal mudah mengelupas yang berakibat pada kualitas sandal dan salah satu penghambat visi dan misi perusahaan. Penelitian ini dibagi dalam 5 tahap. Tahap pertama adalah Define, yang bertujuan untuk mengidentifikasi masalah yang terjadi yang berasal dari kebutuhan konsumen berdasarkan data DCA dan OQC defect di CV. SANCU. Tahap kedua adalah Measure yaitu mengukur kondisi level performance proses saat ini (Z-Level. Pada tahap ini dilakukan validasi sistem pengukuran  dengan  menghitung  Dpo,Dpmo,Yield  dan  Sigma  level.  Tahap  ketiga  adalah Analyze yaitu menentukan faktor-faktor penyebab terjadinya sol sandal mengelupas dan sekaligus menentukan vital faktor. Hasil dari pengujian menunjukkan bahwa vital faktor adalah waktu penempelan.Tahap keempat adalah improvement yaitu melakukan perbaikan dengan memberikan timer diarea lokasi penempelan. Control yaitu kegiatan melakukan pemantauan dari hasil perbaikan yang telah di implementasikan Kondisi   setelah   perbaikan   menghasilkan   perubahan   yang   signifikan   terhadap kapabilitas performance pada sandal, yaitu perubahan nilai sigma level setelah perbaikan 4.74s dari yang sebelum perbaikan adalah 4.26 s. Dari nilai tersebut dapat disimpulkan bahwa metode waktu penempelan dan kontrol pada performance sandal Cv. Sancu sudah membaik.

  19. Structural basis for proficient incorporation of dTTP opposite O6-methylguanine by human DNA polymerase iota. (United States)

    O(6)-methylguanine (O(6)-methylG) is highly mutagenic and is commonly found in DNA exposed to methylating agents, even physiological ones (e.g. S-adenosylmethionine). The efficiency of a truncated, catalytic DNA polymerase ι core enzyme was determined for nucleoside triphosphate incorporation opposite O(6)-methylG, using steady-state kinetic analyses. The results presented here corroborate previous work from this laboratory using full-length pol ι, which showed that dTTP incorporation occurs with high efficiency opposite O(6)-methylG. Misincorporation of dTTP opposite O(6)-methylG occurred with ∼6-fold higher efficiency than incorporation of dCTP. Crystal structures of the truncated form of pol ι with O(6)-methylG as the template base and incoming dCTP or dTTP were solved and showed that O(6)-methylG is rotated into the syn conformation in the pol ι active site and that dTTP misincorporation by pol ι is the result of Hoogsteen base pairing with the adduct. Both dCTP and dTTP base paired with the Hoogsteen edge of O(6)-methylG. A single, short hydrogen bond formed between the N3 atom of dTTP and the N7 atom of O(6)-methylG. Protonation of the N3 atom of dCTP and bifurcation of the N3 hydrogen between the N7 and O(6) atoms of O(6)-methylG allow base pairing of the lesion with dCTP. We conclude that differences in the Hoogsteen hydrogen bonding between nucleotides is the main factor in the preferential selectivity of dTTP opposite O(6)-methylG by human pol ι, in contrast to the mispairing modes observed previously for O(6)-methylG in the structures of the model DNA polymerases Sulfolobus solfataricus Dpo4 and Bacillus stearothermophilus DNA polymerase I.

  20. Mechanisms of Insertion of dCTP and dTTP Opposite the DNA Lesion O6-Methyl-2′-deoxyguanosine by Human DNA Polymerase η* (United States)

    O6-Methyl-2′-deoxyguanosine (O6-MeG) is a ubiquitous DNA lesion, formed not only by xenobiotic carcinogens but also by the endogenous methylating agent S-adenosylmethionine. It can introduce mutations during DNA replication, with different DNA polymerases displaying different ratios of correct or incorrect incorporation opposite this nucleoside. Of the “translesion” Y-family human DNA polymerases (hpols), hpol η is most efficient in incorporating equal numbers of correct and incorrect C and T bases. However, the mechanistic basis for this specific yet indiscriminate activity is not known. To explore this question, we report biochemical and structural analysis of the catalytic core of hpol η. Activity assays showed the truncated form displayed similar misincorporation properties as the full-length enzyme, incorporating C and T equally and extending from both. X-ray crystal structures of both dC and dT paired with O6-MeG were solved in both insertion and extension modes. The structures revealed a Watson-Crick-like pairing between O6-MeG and 2"-deoxythymidine-5"-[(α, β)-imido]triphosphate (approximating dT) at both the insertion and extension stages with formation of two H-bonds. Conversely, both the structures with O6- MeG opposite dCTP and dC display sheared configuration of base pairs but to different degrees, with formation of two bifurcated H-bonds and two single H-bonds in the structures trapped in the insertion and extension states, respectively. The structural data are consistent with the observed tendency of hpol η to insert both dC and dT opposite the O6-MeG lesion with similar efficiencies. Comparison of the hpol η active site configurations with either O6-MeG:dC or O6-MeG:dT bound compared with the corresponding situations in structures of complexes of Sulfolobus solfataricus Dpo4, a bypass pol that favors C relative to T by a factor of ∼4, helps rationalize the more error-prone synthesis opposite the lesion by hpol η.     Full Text Available Abstract Background DC derived-exosomes are nanomeric vesicles harboring functional MHC/peptide complexes capable of promoting T cell immune responses and tumor rejection. Here we report the feasability and safety of the first Phase I clinical trial using autologous exosomes pulsed with MAGE 3 peptides for the immunization of stage III/IV melanoma patients. Secondary endpoints were the monitoring of T cell responses and the clinical outcome. Patients and methods Exosomes were purified from day 7 autologous monocyte derived-DC cultures. Fifteen patients fullfilling the inclusion criteria (stage IIIB and IV, HLA-A1+, or -B35+ and HLA-DPO4+ leukocyte phenotype, tumor expressing MAGE3 antigen were enrolled from 2000 to 2002 and received four exosome vaccinations. Two dose levels of either MHC class II molecules (0.13 versus 0.40 × 1014 molecules or peptides (10 versus 100 μg/ml were tested. Evaluations were performed before and 2 weeks after immunization. A continuation treatment was performed in 4 cases of non progression. Results The GMP process allowed to harvest about 5 × 1014 exosomal MHC class II molecules allowing inclusion of all 15 patients. There was no grade II toxicity and the maximal tolerated dose was not achieved. One patient exhibited a partial response according to the RECIST criteria. This HLA-B35+/A2+ patient vaccinated with A1/B35 defined CTL epitopes developed halo of depigmentation around naevi, a MART1-specific HLA-A2 restricted T cell response in the tumor bed associated with progressive loss of HLA-A2 and HLA-BC molecules on tumor cells during therapy with exosomes. In addition, one minor, two stable and one mixed responses were observed in skin and lymph node sites. MAGE3 specific CD4+ and CD8+ T cell responses could not be detected in peripheral blood. Conclusion The first exosome Phase I trial highlighted the feasibility of large scale exosome production and the safety of exosome administration.

    ■The expression of miR-1 is increased in lungs from the Hyp/Su5416 PAH rat model. ■PASMC from this animal model are more depolarised and show decreased expression and activity of Kv1.5. ■miR-1 directly targets Kv1.5 channels, reduces Kv1.5 activity and induces membrane depolarization. ■Antagomir-1 prevents Kv1.5 channel downregulation and the depolarization induced by hypoxia/Su5416 exposition. Impairment of voltage-dependent potassium channel (Kv) plays a central role in the development of cardiovascular diseases, including pulmonary arterial hypertension (PAH). MicroRNAs (miRNAs) are non-coding RNAs that regulate gene expression by binding to the 3'-UTR region of specific mRNAs. The aim of this study was to analyze the effects of miR-1 on Kv channel function in pulmonary arteries (PA). Kv channel activity was studied in PA from healthy animals transfected with miR-1 or scrambled-miR. Kv currents were studied using the whole-cell configuration of patch-clamp technique. The characterization of the Kv1.5 currents was performed with the selective inhibitor DPO-1. miR-1 expression was increased and Kv1.5 channels were decreased in lungs from a rat model of PAH induced by hypoxia and Su5416. miR-1 transfection increased cell capacitance, reduced Kv1.5 currents and induced membrane depolarization in isolated pulmonary artery smooth muscle cells (PASMCs). Luciferase reporter assay indicated that KCNA5, which encodes Kv1.5 channels, is a direct target gene of miR-1. Incubation of PA with Su5416 and hypoxia (3% O 2 ) increased miR-1 and induced a decline in Kv1.5 currents, which was prevented by antagomiR-1. In conclusion, these data indicate that miR-1 induces PASMC hypertrophy and reduces the activity and expression of Kv channels, suggesting a pathophysiological role in PAH.   3. Uso de Eculizumab na síndrome hemolítica urêmica atípica após transplante renal

    Full Text Available Descrever uso do Eculizumab na síndrome hemolítica urêmica atípica (SHUa após transplante renal. Paciente de 16 anos, com diagnóstico de doença renal crônica desde 2010, decorrente de SHUa, submetida à hemodiálise. Transplante renal por doador falecido: fevereiro de 2012. Apresentou boa evolução clínica até 14º DPO, quando iniciou quadro de febre, oligúria, piora da função renal [creatinina sérica (CRs: 4,0 mg/dl] e sinais de hemólise [plaquetas: 110.000 mm3; hemoglobina (Hb: 4,5 g/dL; LDH: 3366 U/L]. Biópsia do enxerto: microangiopatia trombótica. Realizado manejo com hemoderivados (plasma fresco e plasmaférese, com melhora da função renal (CRs: 1,46 mg/dl. Uma semana após esta intercorrência, reapresentou quadro de febre, anemia, sinais de hemólise e ITU, então manejados com ciprofloxacina, pulsoterapia com metilprednisolona e transfusão de plasma (plaquetas: 43.000 mm3; Hb: 6,0 mg/dl, reticulócitos: 1,3%, haptoglobina < 5,8 mg/dl, LDH: 1181 U/L. Após piora clínica, iniciada terapêutica com Eculizumab, 900 mg a cada cinco dias durante duas semanas. Evoluiu com boa resposta clínica, caracterizada pela melhora da função renal, normalização hematológica (plaquetas: 160.000 mm3; Hb: 11,4 g/dL e alta hospitalar em cinco dias. Desde então, mantém uso de Eculizumab 900 mg de 15/15 dias, com quadro renal e hematológico estável.     Antibiotics resistance in Helicobacter pylori (H. pylori) is the major factor for eradication failure. Molecular tests including fluorescence in situ hybridization, PCR-restriction fragment length polymorphism, and dual priming oligonucleotide-PCR (DPO-PCR) play critical roles in the detection of antibiotic susceptibility; however, limited knowledge is known about application of multiple genetic analysis system (MGAS) in the area of H. pylori identification and antibiotics resistance detection.The aim of this study is to determine the antibiotics resistance using different molecular tests and evaluate the treatment outcomes of E-test-based genotypic resistance.A total of 297 patients with dyspepsia complaint were recruited for gastroscopies. Ninety patients with H. pylori culture positive were randomly divided into 2 groups (test group and control group). E-test, general PCR, and MGAS assay were performed in test group. Patients in control group were treated with empirical therapy (rabeprazole + bismuth potassium citrate + amoxicillin [AMX] + clarithromycin [CLR]), whereas patients in test group received quadruple therapy based on E-test results twice daily for 14 consecutive days. The eradication effect of H. pylori was confirmed by C-urea breath test after at least 4 weeks when treatment was finished.Rapid urease test showed 46.5% (128/297) patients with H. pylori infection, whereas 30.3% (90/297) patients were H. pylori culture positive. E-test showed that H. pylori primary resistance rate to CLR, AMX, metronidazole, tetracycline, and levofloxacin (LVX) was 40.0% (18/45), 4.4% (2/45), 53.3% (24/45), 0% (0/45), and 55.6% (25/45), respectively. In addition, there are many multidrug resistant (MDR) phenotypes, and the MDR strains have higher minimum inhibitory concentration than their single-drug resistant counterparts.     Full Text Available JUSTIFICATIVA E OBJETIVOS: A deficiência do fator XI é uma doença hematológica rara na população. A hemofilia C (deficiência do fator XI ocorre em ambos os sexos e normalmente não apresenta qualquer sintomatologia, podendo manifestar-se apenas como hemorragia pós-cirúrgica. É uma doença autossômica recessiva, homozigótica ou heterozigótica, e sua gravidade depende dos níveis de fator XI. O objetivo desse relato foi apresentar a estratégia anestésica em paciente portadora de hemofilia C. RELATO DO CASO: Paciente com 32 anos, gesta I/para 0, 39 semanas de gestação programada para cesariana eletiva. Paciente portadora de deficiência de fator XI. Exame clínico e laboratorial sem alterações. Conforme orientação do hematologista, no dia da cesárea a paciente usou prometazina 25 mg; hidrocortisona 500 mg, devido a reações transfusionais prévias, e plasma 10 num total de 700 mL. Após 2 horas foi submetida ao bloqueio subaracnóideo sob monitorização de rotina. Hidratação com RL 2000 mL. Procedimento anestésico-cirúrgico sem intercorrências. A paciente evoluiu no pós-operatório sem intercorrências, sendo que no 3º DPO fez uso de plasma fresco congelado (PFC com o objetivo de evitar sangramento pós cirúrgico tardio. CONCLUSÕES: O objetivo do caso foi apresentar o protocolo anestésico para pacientes portadores de hemofilia C e alertar para a necessidade de investigação em caso de antecedente de sangramento pós-operatório, quando um estudo da coagulação deve ser realizado antes de qualquer procedimento invasivo e, se um TTPA prolongado for encontrado, torna-se imperativo pesquisar a deficiência desse fator.JUSTIFICATIVA Y OBJETIVOS: La discapacidad del factor XI es una enfermedad hematológica rara en la población.     Full Text Available INTRODUÇÃO: Após a cirurgia cardíaca, os pacientes apresentam limitação na força muscular respiratória, o que favorece a instalação de complicações pulmonares. OBJETIVO: Analisar a eficácia da estimulação elétrica nervosa transcutânea sobre o processo doloroso e força muscular respiratória em pacientes submetidos à cirurgia de revascularização do miocárdio (CRM. MÉTODOS: Foram inclusos pacientes em pós-operatório de CRM por meio de esternotomia, com uso de circulação extracorpórea, anestesia geral, sem estar sob efeito de bloqueio neuromuscular, uso de drenos de tórax e mediastino, extubados até 6 horas pós-procedimento e apresentando índice igual ou superior a três na escala analógica visual da dor (EVA, estando no primeiro dia de pós-operatório (1º DPO. Foram recrutados 20 pacientes, divididos em dois grupos, sem predomínio de sexo: Grupo Controle (n=10, que recebeu terapia analgésica mais fisioterapia; e Grupo TENS, que recebeu terapia analgésica, fisioterapia e TENS. A TENS foi aplicada por 30 minutos, três vezes ao dia, num intervalo de 3 horas cada aplicação. RESULTADOS: Para o grau de dor, houve uma média inicial e final, respectivamente, de 7,0 e 1,0 para o Grupo TENS e 7,0 e 8,0 para o Grupo Controle. Para a Pimáx, a média inicial e final foi de, respectivamente, -102,5 cmH2O e -141,17 cmH2O para o Grupo TENS e -97,0 cmH2O e -100,3 cmH2O para o Controle. Quanto a Pemáx, a média inicial e final foi de, respectivamente, 63 cmH2O e 125 cmH2O para o Grupo TENS e 55,3 cmH2O e 53,2 cmH2O para o Grupo Controle.   8. Investigating the possibility of the CONSERT instrument operating as a bi-static RADAR sounder during the seperation, descent and landing phase of the ROSETTA mission (United States)

    The main scientific objective of the Comet Nucleus Sounding Experiment by Radiowave Transmission (CONSERT) is to determine the dielectric properties of comet 67P/Chuyurmov-Gerasimenko's nucleus. This will be achieved by performing a sounding of the comet's core between the lander "Philae" launched on the comet's surface and the orbiter "Rosetta". For the sounding the lander will receive, process and retransmit the radio signal emitted by the CONSERT instrument aboard the orbiter. With data measured during the first science phase, a three-dimensional model of the material distribution with regard to the complex dielectric permittivity of the comet's nucleus is to be reconstructed. In order to increase the scientific outcome of the experiment and to collect data beneficial for the main scientific objective, it may be considered to operate the CONSERT instrument as a bi-static RADAR sounder during the non mission-critical parts of the separation, descent and landing (SDL) phase, i.e. when the lander is launched onto the comet's surface, of the ROSETTA mission. The data measured during this phase will be mainly echoes from the comet's surface and first meters of subsurface. Based on this data, we intent to create an initial dielectric permittivity mapping of the comet's surface at and around the landing site In order to estimate the performance of the instrument in this special operational mode, simulations of a sounding in SDL configuration were performed. The simulations are based on a hybrid method-of-moments physical-optics (EFIE-DPO) approach for large dielectric bodies with consideration of the behavior of the instrument's antennas and coupling with the spacecraft as well as polarization effects. The simulated results are furthermore processed in a system-level-instrument-simulator to include effects such as a realistic sounding signal, pulse-compression and analog digital conversion in the estimation of the sounding capabilities. Os sensores de molhamento no vinhedo foram instalados no topo das plantas com um ângulo de inclinação de 45º e com a face superior do sensor voltada para sudoeste, com três repetições. Os quatro modelos de estimativa da DPM foram: número de horas com umidade relativa do ar acima de 90% (NHUR > 90%, depressão do ponto de orvalho (DPO, árvore de classificação e regressão (CART e o de Penman-Monteith (PM. O modelo CART estimou melhor a DPM no gramado, apresentando uma boa precisão (R² = 0,82 e uma ótima exatidão (d = 0,94, resultando num bom índice de confiabilidade (c = 0,85. Esta estimativa também apresentou uma boa correlação com a DPM medida no interior do vinhedo, com uma precisão razoável (R² = 0,87 e uma ótima exatidão (d = 0,96, resultando num ótimo índice de confiabilidade (c = 0,93, o que permite concluir que é possível estimar a DPM no vinhedo de 'Niagara Rosada' por meio de dados medidos ou estimados na estação meteorológica padrão.

As peças foram coradas com Hematoxilina e Eosina e Von Kossa e analisadas por um patologista que desconhecia os grupos. RESULTADOS: Não foram observadas alterações nas estruturas renais. Nas amostras hepáticas foram observados achados histológicos como a degeneração hidrópica no grupo de animais mortos no sétimo DPO (p= 0,029. CONCLUSÃO: O pneumoperitônio com dióxido de carbono não afetou a morfologia renal e causou a degeneração hidrópica no fígado dos animais mortos no 7º dia pós-operatório.

Esta política se acompaña de otros discursos ligados a las TIC y a los medios: sociedad de la información, educación virtual, entornos virtuales de aprendizaje y ambientes hipermedia, entre otros.Ahora bien, no se ha de reconocer este el espacio pertinente para explicar, promover o difundir las políticas del gobierno de turno en materia de educación. Pero sí se considera conveniente hacer visible estos dos escenarios que reflejan el discurso pedagógico oficial dominante, a propósito de varios de los artículos que conforman esta nueva edición de Pedagogía y Saberes: el hecho de recibir un número significativo de trabajos académicos que abordanestas dos temáticas muestra el “efecto” de ese discurso en los agentes y en las agencias educativas.El concepto de discurso pedagógico oficial, Dpo –es decir, el discurso oficial de la educación– se entiende aquí desde la perspectiva ofrecida por Basil Bernstein. Para el sociólogo y lingüista británico este discurso opera como el aparato legal de legitimación y regulacióndel universo escolar. Lo reconoce como una práctica política del Estado y como una realización del poder y del control que ejerce. Asimismo, no se trata de un espacio de producción autónoma sino de una apropiación y, a su vez, reubicación ideológica de otros discursos.
