The relation between transmucosal fluid movement and its effect on absorption and exsorption of imipramine was studied with the in-situ single-pass perfusion technique in rats. Dinoprost (prostaglandin F2 alpha, PGF2 alpha) caused a dose-related inhibition of both absorption and secretion of water across the intestinal membrane. When PGF2 alpha was infused at a rate of 5 mumols kg-1 h-1, the absorption rate of water decreased from 51.7 to 21.5 mL h-1 and the secretion rate decreased from 48.9 to 26.8 mL h-1. Net water flux changed from net water absorption (0.9 mL h-1) to net water secretion (5.33 mL h-1) by infusion of PGF2 alpha. However, absorption and exsorption of imipramine were little affected by infusion of PGF2 alpha. The absorption rates of imipramine were 3.03 and 2.36 mg h-1 in the absence and presence of PGF2 alpha, respectively. Furthermore, the average amounts of imipramine exsorbed into the intestinal lumen in 2 h were 7.82 and 8.10% in the absence and presence of PGF2 alpha, respectively. Infusion of PGF2 alpha also enhanced motility of the small intestine compared with the control. From these results, it appears that PGF2 alpha has no effect on the absorption and exsorption of imipramine across the intestinal membrane although it is reasonable to use PGF2 alpha in the case of patients with overdoses of drugs which decrease gastrointestinal motility.

The authors evaluated the usefulness of prostaglandin F2 alpha in inducing second trimester abortions in two consecutive groups of patients. Progressive and fractionated intracervical instillation of 10 ml of Tylose gel containing 10 mg of Prostine F2 alpha is the least traumatic method of uterine evacuation in the majority of cases (18 out of 24 cases). Advantages of this method consist of the ease with which it can be performed, that it may be repeated on subsequent days, and that there have been no serious complications. In case of failure, the authors recommend using an intraamnionic injection (usually performed only once) of 40 mg of Prostine F2 alpha diluted in 20 ml of normal saline solution. This more involved method requires an experienced physician and ultrasonography, but is very effective (31 cases out of 3). Exceptionally, this can be repeated after a several day interval if the initial injection fails (2 out of 3). These methods avoid recourse to surgery (except in the case of uterine scarring) in the induction of second trimester abortions.

Full Text Available This paper presents results of investigations of gonadoreline application (14-16, 17-20 and 21-25 days post partum influence, followed by prostaglanin F2 α ten days later, on the reproductive parameters of dairy cows with difficult calving and endometritis. The study was carried out on a total of 92 Holstein-Friesian cows. Fifty of them treated with hormones, while 42 animals served as controls, receiving placebo injections. Analysis of the results in cows with postpartal endometrits revealed that mean anoestrus period, open days period and intervals between calving were significantly lower in cows that underwent hormonal treatment, when compared to the controls. The period from gonadoreline application, followed by prostaglandin injection, to conception was shorter by 44, 28 and 14 days respectively when compared to the control groups. The total conception rate from the first three artificial inseminations was higher in the experimental groups in comparison to the controls. .

Gilts were treated during midgestation with prostaglandin (PG) F to study the efficacy of different treatment regimens on induction of abortion and to determine the adverse consequences of PGF-induced abortion in swine. In study 1, pregnant purebred Duroc gilts (60 to 90 days of gestation) were given (IM) 500 micrograms of cloprostenol (n = 12), 20 mg of dinoprost tromethamine (n = 11), or 10 mg of dinoprost tromethamine repeated 12 hours later by an additional 10 mg of dinoprost tromethamine (n = 11). The percentage of gilts that aborted and percentage of aborted gilts that returned to estrus for each treatment group were as follows: cloprostenol, 91.7% and 100%, respectively; 20 mg of dinoprost tromethamine, 36.4% and 25.0%, respectively; and 10 + 10 mg of dinoprost tromethamine, 100% and 90.9%, respectively. Treatment with cloprostenol and with 10 + 10 mg of dinoprost tromethamine caused more gilts to abort (P less than 0.01) than did treatment with 20 mg of dinoprost tromethamine. Gilts that did not abort were given a second treatment with 10 + 10 mg of dinoprost tromethamine. When the abortions by gilts initially treated with 500 micrograms of cloprostenol or 10 + 10 mg of dinoprost tromethamine were combined with those re-treated with 10 + 10 mg of dinoprost tromethamine, 32 of 33 (97.0%) gilts aborted, and 30 of the 32 (93.8%) aborted gilts returned to estrus.(ABSTRACT TRUNCATED AT 250 WORDS)

The results of the cervical priming with a Dinoprost-containing gel and a Gemeprost-containing vaginal suppository were compared in 68 patients, who required termination of pregnancy beyond 14 weeks because of a severe maternal disease or a fetal abnormality. The priming consisted of either an intracervical application of Dinoprost (500 micrograms) in a tylose-gel in 6-8 hour intervals or a retrocervical application of Gemeprost (1 mg) as a vaginal suppository in 12 hour intervals. Although no significant parameter variances were found in the selected patient groups, abortion was induced in 75% of cases within 24 hours, in 89% within 36 hours using Gemeprost. Mean induction time for Gemeprost was 19.5 hours. Using Dinoprost only 19% of patients had an abortion within 24 hours (44% within 36 hours, respectively), mean induction time was significantly longer (38.8 hours, p < 0.005). These differences remained unchanged, when patients who had a prior caesarean section were not evaluated. Using Gemeprost the additional systemic administration of Sulprost was necessary in 21% of cases, using Dinoprost, in 50% of cases. Severe complications did not occur and minor side effects such as nausea or vomiting were observed in single cases. These results demonstrate that Gemeprost can be used in cervical priming even after 14 weeks of pregnancy and that the longer application interval of 12 hours results in a reduction of side effects without a decrease in efficacy.

Natural lambing in sheep in Ethiopia occurs throughout the year in a scattered manner negatively affecting survival and growth rates of the lambs born during the unfavorable season of the year. Thus, controlling the time of mating artificially using exogenous source of hormones is considered as one of the ways to mitigated problems related to haphazard lambing. To this end, an experiment was conducted to evaluate efficacy of prostaglandin-based estrus synchronization protocol in local and crossbred ewes. A total of 160 ewes (80 local and 80 crossbreds) which lambed at least once and aged 3-5 years were used. Lutalyse® (dinoprost tromethamine sterile solution equivalent to 5 mg dinoprost per ml) and its analog, Synchromate® (cloprostenol sodium equivalent to 0.250 mg cloprostenol per ml), were tested at different doses. The treatments used were intramuscular injection of (1) 2.50 ml of Lutalyse® (12.5 mg dinoprost tromethamine), (2) 2 ml of Lutalyse® (10.0 mg dinoprost tromethamine), (3) 1 ml of Synchromate® (0.25 mg of cloprostenol Sodium), and (4) 0.8 ml of Synchromate® (0.20 mg of cloprostenol Sodium). Forty ewes (20 local and 20 crossbreds) were allocated per treatment. Following injection of the respective hormones, rams of known fertility were introduced into the flock for the duration of 96 h at the ratio of one ram to 10 ewes. All estrus synchronization protocols except treatment 4 (0.8 ml of Synchromate®) induced estrus (heat) in majority (55-65%) of local and crossbred ewes within 96 h post-hormone injection. The time interval from hormone administration to onset of estrus was also more or less similar for all treatment groups except for treatment group 4 which showed heat quicker. The highest lambing rate was recorded in local ewes (84.62% (11/13) treated with 2.5 ml of Lutalyse®, whereas the least was obtained in crossbreds (33.33% (3/9) treated with 0.8 ml Synchromate®. The level of progesterone produced by luteal cell culture was measured at day 9th and 11thincubation. The result showed concentration of PGF2? cell product of seminal vesicle cell culture wassignificantly higher (P < 0.05 compared to endometrial cell culture. There was no significant difference(P>0.05 in luteolytic ability between PGF2? cell culture product and dinoprost. An efficient and trans-diastereoselective Rh(I)-catalyzed 1,4-conjugate addition reaction of alkenylboronic acids and a homochiral (R)-4-silyloxycyclopentenone useful for the synthesis of derivatives of prostaglandins E and F is described for the first time. The reaction functions under mild conditions and is particularly rapid (≤6 h) under low power (50 W) microwave irradiation at 30 °C in MeOH in the presence of a catalytic amount of KOH. Under these conditions, 3 mol % of [RhCl(COD)]2 is typically required to produce high yields. The method also functions without microwave irradiation at 3 °C in the presence of a stoichiometric amount of KOH. Under these conditions, only 1.5 mol % of [RhCl(COD)]2 is needed, but the reaction is considerably slower. The method accepts a range of aryl- and alkyl-substituted alkenylboronic acids, and its utility has been demonstrated by the synthesis of PGF2α (dinoprost) and tafluprost.

The effect of propylene glycol drenching on ovarian and hormonal dynamics was studied in heifers. Five cycling heifers were used twice (as control and treatment) with crossover design. After the confirmation of ovulation (day 0), the heifers in the treatment group received propylene glycol on days 6, 7 and 8 as an oral drench (250 ml of 90% propylene glycol). On day 10, prostaglandin F2α (PGF2α), 15 mg per head of dinoprost, was administered intramuscularly to induce luteal regression followed by the follicular phase and then propylene glycol was again administered twice daily (500 ml/day) on days 10, 11 and 12. Palpation per rectum and ovarian ultrasonography were performed every other day from days 0 to 10, and daily after PGF2α administration until the subsequent ovulation (second ovulation) for analysis of follicular and luteal dynamics. Blood samples were also collected every other day from days 0 to 10, and then at 6 h intervals after PGF2α administration until the second ovulation. For the samples taken at 6-h intervals after PGF2α administration, the concentrations of glucose showed clear daily fluctuations in both groups. Changes in the plasma concentration of glucose in the treatment group were significantly (P propylene glycol drenching in heifers. Full Text Available Aim: The current study was done to study the therapeutic effect of natural prostaglandin in treatment of canine pyometra. Materials and Methods: Seven bitches were treated with natural PGF2 á i.e. dinoprost tromethamine at the dose rate of 100 μg/kg body weight subcutaneously once daily for 7 days with supportive therapies. The physiological, haematological and biochemical parameters were studied before (0th day and after treatment (8th day. Therapeutic efficacy was assessed in terms of return of abnormal parameters to either normal or near normal value as compared to the untreated control group, intensity of side effects and post treatment reproductive status. Results: All physiological, haematological and biochemical parameters in the seven treated bitches returned to normal range at the end of treatment. The intensity of side effects was quite severe in the treatment group. Six bitches came to estrus within 2 months of treatment and out of them four conceived on subsequent mating. In rest three bitches there was recurrence of pyometra within 4 months of treatment. Conclusion: Though conception rate of recovered bitches is decreased when compared with that of normal healthy bitches still this treatment protocol can be used successfully in treatment of canine pyometra to conserve the breeding capability of bitches. Full Text Available ABSTRACT: This study evaluated the presence of vaginitis and the bacterial load associated with different intravaginal implants in ewes. Twenty-four Dorper and crossbred ewes were allocated into three groups and received intravaginal implant containing 0.3g progesterone (CIDR(r, 60mg MAP or sponges without progesterone (CONTROL for six days. Then, CIDR and MAP treated-ewes received 12.5mg dinoprost and 300IU eCG. Vaginal mucus samples were collected at four times: before device insertion, at the day of its removal, 24 and 48 hours after. The samples were cultured and the colonies were counted (CFU/mL and identified. The results obtained from the counting of CFU mL-1 were submitted to Kruskal-Wallis H test, with P0.05 in comparison with MAP and CIDR-treated ewes. During the occurrence of vaginitis, the predominant isolates belonged to the coliform group, mainly Escherichia coli (72.7%. Such infection was not determined by the members of the vaginal microbiota that were present before implant insertion and normal microbiota was restored between 24 to 48 hours after insert removal.

The objective was to determine serum progesterone concentrations, ovarian responses, and pregnancy rate in sheep inseminated following a short-term protocol (6 days of treatment) with a previously used controlled internal drug release-G (CIDR-G) device. In experiment 1, 30 ewes were put on a short-term protocol using a CIDR-G of first use (new devices, N = 10), second use (previously used for 6 days, N = 10), or third use (previously used twice for 6 days, N = 10). All ewes were given prostaglandin F(2α) (10 mg dinoprost) and eCG (300 IU) im at device withdrawal. Mean serum progesterone concentrations were greater for ewes treated with new versus reused devices (P synchronization and ovulation, with lower serum progesterone concentrations for reused devices. Three times used CIDR-G yielded a pregnancy rate >70%, which tended to be lower than that obtained with new devices, adding evidence of the detrimental effect of low serum progesterone concentrations on fertility in sheep.

Animals were kept under grazing conditions in a farm at Mamborê, Paraná state. Cows were submitted to two sessions of multiple ovulation 80 days apart. At a random stage of estrus cycle (Day 0 donors received an intravaginal device with 1.9 g of progesterone (CIDR; Pfizer; Brasil. Twenty four hours later (Day 1 cows were injected with 2.5 mg/ IM of Estradiol Benzoate (Estrogin, Farmavet, Brasil. Between Day 5 and Day 9, donors received decreasing doses of 250 UI of FSH (Pluset; Calier, Brasil. Full Text Available In randomized block design, 58 indigenous black mountain goats were examined for the effect of different hormonal treatments in inducing oestrus on selected biochemical characteristics of blood serum that were (aspartate transaminase, AST. Alanine transaminase ALT, Akaline phosphatase, ALP, total protein, albumin and total cholesterol. The animals were randomly assigned into four groups according to their treatment. The control group (C consisted of 10 females whereas the rest of the groups, each consisted of 12 females. The treatments included a double PGF2α (Dinoprost tromethamine intramuscular injection (5 mg at a time interval of 11 days plus an intramuscular injection of pregnant mare serum (PMSG (400I. Uand 600I. U. two days before second injection for (treatments, TI, T2 respectively. Treatment (T3 was as that of T1 except PMSG was not injected. The success of dental implants depends on optimal osseointegration, which is affected by many factors, such as systemic drug administration. In clinical practice, non-steroidal anti-inflammatory drugs(NSAID) are prescribed to deal with chronic inflammation or pain caused by some joint disorders or fractures and to control discomfort after implant surgery. This study evaluated the effect of different doses of eCG (control, 300 or 400 IU) administered at progesterone (P4) device removal in suckled Bos taurus beef cows undergoing a timed artificial insemination (TAI) protocol. A total of 966 cows received a P4 insert and 2.0 mg intramuscular estradiol benzoate at the onset of the synchronization. After 9 days, P4 insert was removed, and 12.5 mg of dinoprost tromethamine and 1 mg of estradiol cypionate were administered, followed by TAI 48 hours later. Then, the cows received one of three treatments as follows: control (n = 323), 300 (n = 326), or 400 IU of eCG (n = 317). A subset (n = 435) of cows in anestrus had their ovaries evaluated using ultrasound at the time of P4 removal and at TAI. Data were analyzed by orthogonal contrasts (C): C1 (eCG effect) and C2 (eCG dose effect). Estrous occurrence (control = 53.7%, 300 IU = 70.6%, and 400 IU = 77.0%) and pregnancy per artificial insemination (control = 29.7%, 300 IU = 44.8%, and 400 IU = 47.6%) were improved by eCG treatment (C1; P = 0.0004 and P 0.15). In conclusion, the eCG treatment administered at the time of P4 removal increased the occurrence of estrus, the larger follicles at TAI, and pregnancy per artificial insemination of suckled B taurus beef cows. Despite the greater occurrence of estrus in noncyclic cows receiving 400 IU of eCG, both eCG doses (300 and 400 IU) were equally efficient to improve pregnancy to artificial insemination.

Introduction: We undertook a prospective cohort study to compare the effectiveness and safety of 50 µg misoprostol versus 3 mg dinoprostone in two vaginal doses 6 hours apart, followed if necessary by oxytocin for labor induction in low-risk post-term (> 40 weeks) pregnancies with unfavorable cervix (Bishop score ≤ 6). Methods: Labor induction and subsequent management were conducted using a standardized protocol. The primary outcome of the study was labor induction rate. Secondary outcomes included mode of delivery, time interval from induction to delivery, maternal complications and neonatal outcome. Results: 107 patients received misoprostol (Group A) and 99 patients received dinoprostone (Group B). Compared with group A, more women in Group B needed a second vaginal dose of prostaglandin or oxytocin infusion in order to proceed to labor (21.5 vs. 43.4 %; p = 0.01). Misoprostol alone as a single or double vaginal dose was more effective than dinoprostone alone in inducing labor without oxytocin administration (85.0 vs. 50.4 %; p = 0.04). Overall, the rate of successful induction of labor did not differ between groups (91.6 vs. 85.8 %; p = 0.75). Vaginal delivery, operative vaginal delivery and Caesarean section rates were not significantly different. Full Text Available The aim of this study was to evaluate ovarian antral follicle populations (OAFP of Nelore and Girolando breed heifers (12–18 months old and cows (24–60 months old. Animals were assigned to four groups: (1 Nelore cows (n = 18, (2 Girolando cows (n = 20, (3 Nelore heifers (n = 7, and (4 Girolando heifers (n = 7. Cows were treated to synchronize follicular wave emergence by implantation of an intravaginal device containing 1.9 g of progesterone, as well as intramuscular administration of 2 mg of estradiol benzoate and 25 mg of dinoprost. This synchronization treatment was administered at a random day of the estrous cycle of each cow, designated D0. Intravaginal devices were removed on D7, and on D11, OAFP counts were performed by transvaginal ovarian ultrasound. For each cow, all follicles ?3 mm in diameter were counted in both ovaries and counts were performed three times at 35-day intervals. Counts were also obtained from heifers, but these animals were not treated for synchronization of follicular wave emergence. Analysis of variance (ANOVA with Tukey’s test and Pearson’s correlation test were used to compare mean OAFPs between counts as well as mean OAFPs between breed and age groups. No differences were observed in mean OAFPs between Nelore and Girolando cows (30.9 vs. 26.7, respectively; P > 0.05 or heifers (16.2 vs. 18.1, respectively; P > 0.05. However, within each breed, there were differences in mean OAFPs between heifers and cows (for Nelore cattle: 16.2 and 30.9, respectively; for Girolando cattle: 18.1 and 26.7, respectively; both P < 0.05. The objective of this study was to evaluate the effects of early postpartum PGF two alpha treatment on reproductive performance in dairy cows with calving and puerperal traits. A total of 363 Holstein cows (128 primiparous and 235 multiparous) were selected based on the presence of at least one of calving and puerperal traits (dystocia, retained placenta, twin, abortion, and postpartum uterine infections) and were assigned to two groups (treatment and control) irrespective of presence or absence of luteal tissue. Cows in the treatment group were treated twice with 25 mg dinoprost 8 h apart on day 20 postpartum, and for the control group saline placebo was administered. As it was speculated that the timing of a second dose would mimic the release of endogenous PGF2α from the uterus, our hypothesis was that two doses of PGF2α 8 h apart may increase the duration of elevated plasma prostaglandin F2α metabolite concentration in these cows. Recorded reproductive variables included days to first estrus, days to first AI, first service conception rate, pregnancy by 150 days in milk, service per conception, open days, and the percentage of repeat breeder animals. The data were analyzed using SPSS (Version 15) (IBM North America, New York, NY, USA) and Minitab (Version 14) (Minitab, State College, PA, USA). Although early postpartum PGF2α treatment had no effect on days to first estrus (36.7 days vs. 34.9 days, P = 0.056) and days to first AI (70.5 days vs. 72.2 days, P = 0.537), it increased first service conception rate (47.1% vs. 27.6%, P remarkable in primiparous cows (64.7% vs. 25%, P service per conception (1.92 vs. 2.72, P < 0.001) and the mean open days (112 days vs. 144 days, P < 0.001), and increased pregnancy by 150 days in milk (DIM) (80% vs. 66%, P = 0.004). Ovulation to the first GnRH (G1) treatment of the Ovsynch protocol improves synchronization rate and pregnancies per AI (P/AI). Elevated progesterone (P4) concentrations at G1 decrease the ovulatory response by decreasing the magnitude of the GnRH-induced LH surge. Therefore, our objective was to evaluate the effect of temporarily decreasing P4 concentrations before initiation of an Ovsynch protocol on ovulatory response to G1 and P/AI. Lactating Holstein cows (n=800) at 53±3 (herd A) or 51±3 (herd B) d in milk (DIM) were synchronized using a modified Double-Ovsynch protocol [pre-Ovsynch protocol (d 0, GnRH; d 7, PGF2α; d 10, GnRH) followed 7 d later by an Ovsynch-56 protocol (d 0, G1; d 7, PGF2α; d 8, PGF2α; d 9.5, GnRH)] to receive first timed artificial insemination (TAI; 80±3 DIM) 16h after the last GnRH treatment. Cows were randomly assigned to receive 12.5mg of PGF2α (a half-dose of dinoprost tromethamine) 2 d before G1 (low-P4) or serve as untreated controls (high-P4). Overall, high-P4 cows had greater P4 concentrations at G1 compared with low-P4 cows (3.0 vs. 1.3ng/mL, respectively). Ovulatory response to G1 was greater for low-P4 than high-P4 cows [81.1 vs. 60.3%, respectively]. Premature luteal regression during the second Ovsynch protocol did not differ between treatments [15.0% vs. 10.7%; for low-P4 vs. high-P4 cows, respectively]. Overall, P/AI did not differ between treatments 32 d after TAI [56.3 vs. 52.9%, for low-P4 vs. high-P4 cows, respectively] or 67 d after AI [50.5 vs. 47.6%, for low-P4 vs. high-P4 cows, respectively]. Pregnancy loss from 32 to 67 d after TAI did not differ between treatments [9.9 vs. 9.3%, for low-P4 vs. high-P4 cows, respectively]. Overall, cows that ovulated to G1 had more P/AI than cows that did not ovulate [58.2 vs. 41.8%, respectively]. Our hypothesis was that increasing the length of an estradiol and progesterone (P4) timed artificial insemination (TAI) protocol would improve pregnancy per artificial insemination (P/AI). Lactating Holstein cows (n=759) yielding 31 ± 0.30 kg of milk/d with a detectable corpus luteum (CL) at d -11 were randomly assigned to receive TAI (d 0) following 1 of 2 treatments: (8d) d -10 = controlled internal drug release (CIDR) and 2.0mg of estradiol benzoate, d -3 = PGF2α(25mg of dinoprost tromethamine), d -2 = CIDR removal and 1.0mg of estradiol cypionate, d 0 = TAI; or (9 d) d -11 = CIDR and estradiol benzoate, d -4 = PGF2α, d -2 CIDR removal and estradiol cypionate, d 0 TAI. Cows were considered to have their estrous cycle synchronized in response to the protocol by the absence of a CL at artificial insemination (d 0) and presence of a CL on d 7. Pregnancy diagnoses were performed on d 32 and 60. The ovulatory follicle diameter at TAI (d 0) did not differ between treatments (14.7 ± 0.39 vs. 15.0 ± 0.40 mm for 8 and 9 d, respectively). The 9 d cows tended to have greater P4 concentrations on d 7 in synchronized cows (3.14 ± 0.18 ng/mL) than the 8d cows (3.05 ± 0.18 ng/mL). Although the P/AI at d 32 [45 (175/385) vs. 43.9% (166/374) for 8d and 9 d, respectively] and 60 [38.1 (150/385) vs. 40.4% (154/374) for 8d and 9 d, respectively] was not different, the 9 d cows had lower pregnancy losses [7.6% (12/166)] than 8d cows [14.7% (25/175)]. The cows in the 9 d program were more likely to be detected in estrus [72.0% (269/374)] compared with 8d cows [62% (240/385)]. Expression of estrus improved synchronization [97.4 (489/501) vs. 81% (202/248)], P4 concentrations at d 7 (3.22 ± 0.16 vs. 2.77 ± 0.17 ng/mL), P/AI at d 32 [51.2 (252/489) vs. 39.4% (81/202)], P/AI at d 60 [46.3 (230/489) vs. 31.1% (66/202)], and decreased pregnancy loss [9.3 (22/252) vs. 19.8% (15/81)] compared with cows that did not show estrus, respectively.
