  1. Transesterification using the cross-linked enzyme aggregate of Photobacterium lipolyticum lipase M37.

    Biodiesel is methyl and ethyl esters of long-chain fatty acids produced from vegetable oils or animal fats. Lipase enzymes have occasionally been used for the production of this biofuel. Recently, biodiesel production using immobilized lipase has received increased attention. Through enhanced stability and reusability, immobilized lipase can contribute to the reduction of the costs inherent to biodiesel production. In this study, methanol-tolerant lipase M37 from Photobacterium lipolyticum was immobilized using the cross-linked enzyme aggregate (CLEA) method. Lipase M37 has a high lysine content (9.7%) in its protein sequence. Most lysine residues are located evenly over the surface of the protein, except for the lid structure region, which makes the CLEA preparation yield quite high (~93%). CLEA M37 evidences an optimal temperature of 30oC, and an optimal pH of 9-10. It was stable up to 50°C and in a pH range of 4.0-11.0. Both soluble M37 and CLEA M37 were stable in the presence of high concentrations of methanol, ethanol, 1-propanol, and nbutanol. That is, their activities were maintained at solvent concentrations above 10% (v/v). CLEA M37 could produce biodiesel from olive oil and alcohols such as methanol and ethanol. Additionally, CLEA M37 generated biodiesel via both 2-step methanol feeding procedures. Considering its physical stability and reusability, CLEA M37 may potentially be used as a catalyst in organic synthesis, including the biodiesel production reaction.

    We are conducting a transit survey of the open cluster M37 using the Megacam instrument on the 6.5 m Multiple-Mirror Telescope. We have obtained ~4500 images of this cluster over 18.5 nights and have achieved the precision necessary to detect planets smaller than Saturn. In this presentation we provide an overview of the project, describe the ongoing data reduction/analysis and present some of our preliminary results.

    Algae are referred to as a third-generation biomass for ethanol production. However, salinity treatment is a problem that needs to be solved, because algal hydrolysates often contain high salt. Here, we isolated the salt-tolerant ethanol-producing yeast Citeromyces matritensis M37 from the east coast of Miura Peninsula in Japan. This yeast grew under osmotic stress conditions (20% NaCl or 60% glucose). It produced 6.55 g/L ethanol from YPD medium containing 15% NaCl after 48 h, and the ethanol accumulation was observed even at 20% NaCl. Using salted Undaria pinnatifida (wakame), we obtained 6.33 g/L glucose from approx. 150 g/L of the salted wakame powder with acidic and heat pretreatment followed by enzymatic saccharification, and the ethanol production reached 2.58 g/L for C. matritensis M37. The ethanol concentration was 1.4 times higher compared with that using the salt-tolerant ethanol-producing yeast Zygosaccharomyces rouxii S11.

Finally, we determined that the increased acarbose production was related to the high activity of glutamate dehydrogenase and glucose 6-phosphate dehydrogenase.

    Full Text Available A thermostable solvent-tolerant lipase was isolated from Pseudomonas mendocina M-37. The lipase production medium was optimized for cost-effective production. Olive oil as a carbon source, and glycine as a nitrogen source were selected as the best for maximum lipase production. Medium optimization led to 3.75-fold increase in the lipase production. The extracellular lipase was purified 42.2-fold to homogeneity by precipitation using polyethyleneglycol, ultrafiltration and hydrophobic interaction chromatography. Its molecular mass, determined with sodium dodecyl sulphate polyacrylamide gel electrophoresis, was 32 kDa. The enzyme was further immobilized on microcrystalline cellulose. The lipase showed an optimal water activity of 0.53 for both, acidolysis and interesterification reactions. Six- to sevenfold increase in synthetic activity of immobilized lipase was observed when interesterification activity of 0.139 IU/mg and transesterification activity of 0.181 IU/mg, respectively, were obtained. This is the first report on Pseudomonas mendocina lipase with synthetic activity immobilized on microcrystalline cellulose.

    Three phase I trials of the rhesus rotavirus (RRV)-based quadrivalent vaccine [composed of serotype 3 (RRV), and serotypes 1 (D x RRV), 2 (DS1 x RRV), and 4 (ST3 x RRV) human rotavirus x RRV reassortants] and the M37 (nursery strain) rotavirus vaccine candidates were conducted in an attempt to find a safe and optimally antigenic formulation. Infants 10-20 weeks old received, in trial I, 1) the quadrivalent vaccine as two separate bivalent doses (1 x 10(4) PFU each of D x RRV and RRV, followed 4 weeks later by 1 x 10(4) PFU each of DS1 x RRV and ST3 x RRV) or 2) placebo; in trial II, 1) one dose of quadrivalent vaccine (10(4) PFU of each component), or 2) two doses of quadrivalent vaccine, or 3) a 10(4) PFU dose of M37 vaccine, or 4) M37 vaccine followed by the quadrivalent vaccine, or 5) placebo; in trial III, 1) a dose of a higher-titered quadrivalent vaccine (10(5) PFU of each component), or 2) two doses of higher titered quadrivalent vaccine, or 3) a dose of higher titered M37 vaccine (10(5) PFU) or 4) two doses of M37 vaccine (10(5) PFU), or 5) M37 vaccine (10(5) PFU) followed by the higher titered quadrivalent vaccine, or 6) placebo. A mild, transient fever during the first week postvaccination was associated with the bivalent or quadrivalent vaccines but not with the M37 vaccine.     Photobacterium lipolyticum M37 lipase (LipM37) was immobilized on the surface of intracellular polyhydroxybutyrate (PHB) granules in Escherichia coli. LipM37 was genetically fused to Cupriavidus necator PHA synthase (PhaC Cn ), and the engineered PHB operon containing the lip M37 -phaC Cn successfully mediated the accumulation of PHB granules (85 wt.%) inside E. coli cells. The PHB granules were isolated from the crude cell extract, and the immobilized LipM37 was comparable with the free form of LipM37 except for a favorable increase in thermostability. The immobilized LipM37 was used to synthesize oleic acid methyl ester (biodiesel) and oleic acid dodecyl ester (wax ester), and yielded 98.0 % conversion in esterification of oleic acid and dodecanol. It was suggested that the LipM37-PhaCCn fusion protein successfully exhibited bifunctional activities in E. coli and that in situ immobilization of lipase to the intracellular PHB could be a promising approach for expanding the biocatalytic toolbox for industrial chemical synthesis.

  16. Photobacterium lipolyticum sp. nov., a bacterium with lipolytic activity isolated from the Yellow Sea in Korea.

    A Gram-negative, motile, non-spore-forming, pleomorphic and lipolytic bacterial strain, M37T, was isolated from an intertidal sediment of the Yellow Sea in Korea. This organism grew optimally at 25-28 degrees C and in the presence of 1-2 % NaCl. It did not grow without NaCl or in the presence of more than 6 % NaCl. Strain M37T was characterized chemotaxonomically by having Q-8 as the predominant respiratory lipoquinone and C(16 : 1)omega7c and/or iso-C(15 : 0) 2-OH and C(16 : 0) as the major fatty acids. The DNA G+C content was 47 mol%. Phylogenetic analyses based on 16S rRNA gene sequences placed strain M37T within the clade comprising Photobacterium species, forming a coherent cluster with the type strains of Photobacterium profundum and Photobacterium indicum (16S rRNA gene similarity levels of 97.5-98.0 %). The mean DNA-DNA relatedness levels between strain M37T and P. profundum JCM 10084T and P. indicum DSM 5151T were in the range 12-15 %. Similarities between 16S rRNA gene sequences of strain M37T and those of the type strains of the other Photobacterium species ranged from 93.9 % (with Photobacterium fischeri) to 96.2 % (with Photobacterium phosphoreum). On the basis of phenotypic properties and phylogenetic and genomic distinctiveness, strain M37T (=KCTC 10562BPT=DSM 16190T) should be placed in the genus Photobacterium as a novel species, for which the name Photobacterium lipolyticum sp. nov. is proposed.

    Biodiesel, an alternative fuel, is generated via the transesterification reaction of vegetable oil or animal oil with alcohol. Currently, many reports have noted that microbial lipases might be utilized for the production of biodiesel. Among them, immobilized Candida antarctica lipase B (Novozym435) is frequently utilized for its biocatalytic efficiency and availability. However, as the enzyme is unstable in a medium containing high concentrations of methanol, a multi-stepwise methanol supply is required for the efficient production of biodiesel. Photobacterium lipolyticum lipase (M37) was determined to be quite stable in a medium containing a high concentration of methanol. The enzyme activity was maintained for longer than 48 h without any loss at a methanol concentration of 10%. In an effort to evaluate enzyme performance in the production of biodiesel, we have compared M37 lipase and Novozym435 in the biodiesel production reaction using fresh or waste oil and methanol. In the 3-stepwise methanol feeding method generally conducted for Novozym435 in biodiesel production, the M37 lipase showed a similar or superior conversion yield to Novozym435. However, the M37 lipase evidenced significantly higher conversion yields in the 2 and 1 step methanol feeding reactions. Particularly in the 1 step process using 10% of methanol where almost no conversion was detected by Novozym435, the biodiesel yield achieved with M37 lipase reached a level of up to 70% of the possible maximum yield. Consequently, this methanol-tolerant lipase, M37, has been shown to be a suitable enzyme for use in the biodiesel production process.

    Avalanche fluctuations set a limit to the energy and position resolutions that can be reached by gaseous detectors. This paper presents a method based on a laser test-bench to measure the absolute gain and the relative gain variance of a Micro-Pattern Gaseous Detector from its single-electron response. A Micromegas detector was operated with three binary gas mixtures, composed of 5% isobutane as a quencher, with argon, neon or helium, at atmospheric pressure. The anode signals were read out by low-noise, high-gain Cremat CR-110 charge preamplifiers to enable single-electron detection down to gain of 5× 10{sup 3} for the first time. The argon mixture shows the lowest gain at a given amplification field together with the lowest breakdown limit, which is at a gain of 2×10{sup 4} an order of magnitude lower than that of neon or helium. For each gas, the relative gain variance f is almost unchanged in the range of amplification field studied. It was found that f is twice higher (f∼0.6) in argon than in the two other mixtures. This hierarchy of gain and relative gain variance agrees with predictions of analytic models, based on gas ionisation yields, and a Monte-Carlo model included in the simulation software Magboltz version 10.1.

    The study examines over-indebted fathers after separation/divorce with respect to health-related lifestyles/state of health. A cross-sectional survey was conducted among over-indebted persons in Germany 2006/2007 (response rate 39.7%). Fathers in complete families (n = 73; M = 37 years) and single mothers (n = 72; M = 33.5 years) served as comparison groups to the separated fathers (n = 59; M = 37 years). Risks of depressive mood, unhealthy nutrition, and hypertension were higher compared to fathers in complete families. Tobacco use and low health awareness proved to be higher than among both comparison groups. The findings indicate problematic health-related behavior and health status among separated over-indebted fathers, and suggest recognizing them as a high risk group within social work.

    Full Text Available The current study sought to increase our understanding of the factors involved in the early vocabulary development of Australian Indigenous children. Data from the Longitudinal Study of Indigenous Children were available for 573 Indigenous children (291 boys who spoke English (M=37.0 months, SD=5.4 months, at wave 3. Data were also available for 86 children (51 boys who spoke an Indigenous language (M=37.1 months, SD=6.0 months, at wave 3. As hypothesised, higher levels of parent-child book reading and having more children’s books in the home were associated with better English vocabulary development. Oral storytelling in Indigenous language was a significant predictor of the size of children’s Indigenous vocabulary.

    Atmospheric and room temperature plasma (ARTP) was first employed to generate mutants of Actinomyces JN537 for improving acarbose production. To obtain higher acarbose producing strains, the method of screening the strains for susceptibility to penicillin was used after treatment with ARTP. The rationale for the strategy was that mutants showing penicillin susceptibility were likely to be high acarbose producers, as their ability to synthesize cell walls was weak which might enhance metabolic flux to the pathway of acarbose biosynthesis. Acarbose yield of the mutant strain M37 increased by 62.5 % than that of the original strain. The contents of monosaccharides and amino acids of the cell wall of M37 were lower than that of the original strain. The acarbose production ability in mutant strain remained relatively stable after 10 generations. This work provides a promising strategy for obtaining high acarbose-yield strains by combination of ARTP mutation method and efficient screening technique.

    Full Text Available This study compares Italian mothers’ and Ecuadorian immigrant mothers’ daily family routines. Thirty mothers took part in this investigation: 15 Italians (M = 37.5 years, with ages ranging from 30 to 42 years and 15 Ecuadorians (M = 36.1 years, with ages ranging from 18 to 46 years and having lived in Italy for an average of 15 years, with children ranging in age from 4 to 8 years. The objective is to highlight the differences and similarities between the daily routines and family relations that characterize the lives of each of the two groups of participants. The instruments employed in service of these aims were in-depth interviews and diaries. The results showed similarities regarding the routines of Italian and Ecuadorian families. However, a relevant difference emerged regarding their perceptions of social support. Specifically, Italians receive more social support from their parents than do Ecuadorians, who are supported more by other relatives or by friends.

    This study was a prospective 2-year longitudinal investigation of associations between negative maternal parenting and disruptive child behavior across the preschool to school transition. Our main goals were to 1) determine the direction of association between early maternal negativity and child disruptive behaviors across this important developmental transition and 2) examine whether there would be different patterns of associations for boys and girls. Participants were 235 children (111 girls; T1; M= 37.7 months, T2; M=63.4 months) and their mothers and teachers. Observational and multi-informant ratings of child disruptive behavior showed differential patterns of stability and associations with measures of parenting risk. Results indicated bidirectional and interactive contributions of externalizing behavior and negative parenting across time. Results also indicated that risk mechanisms operate similarly for both sexes. Findings support transactional models of disruptive child behavior that highlight the joint contributions of parents and children. PMID:19533326

    Patients A and M developed allergic symptoms when working in a greenhouse with paprika plants and freesia plants, respectively. The possible involvement of an IgE-mediated mechanism was investigated with the skin prick test, radioallergosorbent test (RAST) and the histamine release test (HRT). Paprika flower, leaf and stem extract released 58, 47 and 43% of the total amount of histamine from washed leukocytes of patient A. In serum A IgE antibodies against paprika leaves and flowers could be demonstrated by RAST (11% binding of 125I-anti-IgE added). Freesia flower and stem extract released 46 and 43% histamine, respectively, from washed leukocytes of patient M. In the RAST, specific IgE antibodies against freesia flowers and stems were found in serum M (37% binding of 125I-anti-IgE added).

    Serum paraoxonase 1 (PON1) was purified from bovine serum using hydrophobic interaction chromotography on Sepharose 4B-coupled l-tyrosine 1-naphthylamine gel, and monitored by sodium dodecyl sulfate polyacrylamide gel electrophoresis. Paraoxonase enzyme was immobilized using different ratios of glutaraldehyde and the maximum activity was observed with 7% glutaraldehyde. The effects of inhibition by Mn(+2), Co(+2) and Cu(+2) heavy metals on the immobilized and free enzyme activities were studied. At the optimum pH and temperature, the K(m) and V(max) kinetic values for bovine serum paraoxonase and immobilized paraoxonase towards paraoxon substrate were determined as 0.296 × 10(-3) M & 37.04 EU vs. 0.727-10(-3) M & 36.36 EU, respectively.

    Full Text Available O estudo objetivou descrever as estrat��gias de enfrentamento adotadas por pessoas portadoras de HIV/Aids e identificar associações entre modalidades de enfrentamento e variáveis sociodemográficas (sexo, escolaridade e situação conjugal e médico-clínica (condição sintomática e assintomática. A amostra de conveniência foi composta de 241 pessoas soropositivas, 161 (66,8% homens, de 20 a 64 anos (M=37,4; DP=7,8, 169 (70,1% sintomáticas. A coleta de dados incluiu questionários estruturados e a Escala Modos de Enfrentamento de Problemas (EMEP, com aplicação individual assistida e preenchimento da EMEP sem a presença do pesquisador. Os resultados indicaram a variabilidade das estratégias de enfrentamento, com predomínio de enfrentamento focalizado no problema. Em mulheres e pessoas com níveis mais baixos de escolaridade predominaram enfrentamento focalizado na emoção e busca de práticas religiosas. Os resultados fornecem subsídios para a intervenção psicológica em se tratando de pessoas soropositivas.     Full Text Available In the present study, a total of 353 nodule-associated bacteria were isolated from 220 legume plant samples belonging to Cicer arietinum (85, Glycine max (74, Vigna radiata (21 and Cajanus cajan (40. A total of 224 bacteria were identified as fast-growing Rhizobium spp. on the basis of differential staining (Gram staining and carbol fuchsin staining and biochemical tests. All the isolates were tested for indole acetic acid production (IAA, phosphate solubilization and siderophore production on plate assay. To examine the effect of volatile organic metabolites (VOM and water soluble soil components (WSSC on nodule bacteria, culture conditions were optimized by observing the effects of various parameters such as pH, salt content and temperatures on the growth of bacteria. Selected rhizobia were subjected to random amplified polymorphic DNA (RAPD and amplified ribosomal DNA restriction analysis (ARDRA analysis to identify their species. On the basis of RAPD and ARDRA, 10 isolates were identified as Rhizobium meliloti. In this study, Rhizobium GO4, G16, G20, G77, S43, S81, M07, M37, A15 and A55 were observed as the best candidates among the tested bacteria and can be further used as potent bioinoculants.

    More than 100 million individuals worldwide are exposed to arsenic-contaminated water, making the investigation of arsenic mobility in aquatic systems of utmost importance. Iron (hydr)oxides play a key role in preventing arsenic release in aquifers and soils due to their strong arsenic sorption and are even used to remove arsenic in water treatment. Neutrophilic Fe(II)-oxidizing bacteria produce Fe(III) minerals and therefore have the potential to affect arsenic mobility. In the present study, we demonstrate that the metabolism of anaerobic nitrate-reducing and phototrophic Fe(II)-oxidizing bacteria is not significantly affected by arsenate concentrations of up to 500 muM (37.5 mg/L). Even in the presence of the more toxic arsenic species, arsenite, cell metabolism was significantly impaired only at the highest arsenite concentration (500 muM) for one of the Fe(II)-oxidizers. All Fe(II)-oxidizing bacteria tested effectively immobilized arsenic during Fe(II) oxidation (>96%), lowering the remaining dissolved arsenic concentrations to values close to or even lower than the current drinking water limit of 10 microg/L. Since the minerals formed by these bacteria included highly crystalline Fe(III) minerals that are hardly reducible by Fe(III)-reducing bacteria, stimulation of arsenic immobilization by Fe(II)-oxidizing bacteria can potentially support water treatment systems or even be applied as an effective remediation strategy.

    Respiratory syncytial virus (RSV) is the most common cause of bronchiolitis in infants and young children. A small percentage of these individuals develop severe and even fatal disease. To better understand the pathogenesis of severe disease and develop therapies unique to the less-developed infant immune system, a model of infant disease is needed. The neonatal lamb pulmonary development and physiology is similar to that of infants, and sheep are susceptible to ovine, bovine, or human strains of RSV. RSV grown in Vero (African green monkey) cells has a truncated attachment G glycoprotein as compared to that grown in HEp-2 cells. We hypothesized that the virus grown in HEp-2 cells would cause more severe clinical symptoms and cause more severe pathology. To confirm the hypothesis, lambs were inoculated simultaneously by two different delivery methods (intranasal and nebulized inoculation) with either Vero-grown or HEp-2-grown RSV Memphis 37 (M37) strain of virus to compare viral infection and disease symptoms. Lambs infected with HEp-2 cell-derived virus by either intranasal or nebulization inoculation had significantly higher levels of viral RNA in lungs as well as greater clinical disease including both gross and histopathologic lesions compared to lambs similarly inoculated with Vero-grown virus. Thus, our results provide convincing in vivo evidence for differences in viral infectivity that corroborate previous in vitro mechanistic studies demonstrating differences in the G glycoprotein expression by RSV grown in Vero cells.

    Cannabis use rates continue to rise in the United States and currently cannabis is among the most widely used substances in the world. Cannabis use is associated with several mental health problems, low educational attainment, low income, and underemployment. The current study explored the tendency to experience negative affect (negative affectivity) as a factor accounting for the association between perceived distress tolerance and problems related to the use of cannabis. Participants included 203 urban adult daily cannabis users (29.2% female, M=37.7years, 63% African American). Results indicated that there was a significant indirect effect of distress tolerance via negative affectivity in terms of cannabis use problems (b=-0.58, 95%CI [-1.14, -0.21]), cannabis withdrawal (b=-0.65, 95%CI [-1.36, -0.21]), self-efficacy for quitting (b=-0.83, 95%CI [-1.85, -0.22]), and perceived barriers for cannabis cessation (b=-0.71, 95%CI [-1.51, -0.24]). The present data provide novel empirical evidence suggesting negative affectivity may help explain the relation between perceived distress tolerance and an array of clinically significant cannabis use processes. Intervention programming for daily cannabis users may benefit from targeting negative affectivity to facilitate change in cannabis use processes among users who tend to perceive that they are less capable of tolerating distress.     Effects of a 24-week strength training performed twice weekly (24 ST) (combined with explosive exercises) followed by either a 3-week detraining (3 DT) and a 21-week re-strength-training (21 RST) (experiment A) or by a 24-week detraining (24 DT) (experiment B) on neural activation of the agonist and antagonist leg extensors, muscle cross-sectional area (CSA) of the quadriceps femoris, maximal isometric and one repetition maximum (1-RM) strength and jumping (J) and walking (W) performances were examined. A group of middle-aged (M, 37-44 years, n = 12) and elderly (E, 62-77, n = 10) and another group of M (35-45, n = 7) and E (63-78, n = 7) served as subjects. In experiment A, the 1-RM increased substantially during 24 ST in M (27%, Pelderly during the initial training phases. Neural adaptation seemed to play a greater role than muscle hypertrophy. Short-term detraining led to only minor changes, while prolonged detraining resulted in muscle atrophy and decreased voluntary strength, but explosive jumping and walking actions in both age groups appeared to remain elevated for quite a long time by compensatory types of physical activities when performed on a regular basis.

    Full Text Available : In this study, 24 healthy Kangal dogs, whose fundamental duty is flock guard, were used in three different regions (1950 m altitude of Karasar, 1225 m altitude of Divriği and 918 m altitude of Çiğdemli of Sivas province (homeland for Kangal dogs. Mean pulse, respiration and body temperature values of the Kangal dogs at rest and 10 minutes post-exercise were respectively; 93.00 and 113.96/m; 22.58 and 35.38/m; 37.60 and 37.77ºC. Male Kangals had higher body temperature than did females only after exercise (P<0.05. According to the evaluation made in three different altitudes of the study fields, the differences between the groups were statistically significant for all variables (P<0.001. This study shows that the physiology of Kangal dogs, which drew international attention and were taken to many different regions of the world, may be affected by the new living areas. Although it is not known how physiological changes alter Kangals in future this should be investigated with further studies

    To evaluate the involvement of the bursa located next to the head of the 5th metatarsal bone in patients with psoriatic arthritis (PsA) in comparison with the other seronegative spondyloarthritis (SpA). All patients with PsA seen during a period of 24 months were enrolled. The control group included healthy subjects and patients with the other SpA. All subjects underwent clinical and ultrasound (US) examination of the lateral surface of the 5th metatarsal. 150 PsA patients (88 M; 62 F), 172 SpA (107 M; 65 F), and 95 healthy controls (58 M; 37 F) were evaluated. Based on clinical and US evaluation, bursitis was diagnosed in 17/150 (11.3%) PsA patients but in none of the SpA (P bursa of the 5th metatarsophalangeal joint appears to be involved in PsA more frequently than by chance. If confirmed by other studies, this finding could be considered as a distinctive clinical sign of PsA, useful for differential diagnosis with the other SpA. In asymptomatic patients, US proved to be more sensitive in the detection of bursitis.

  17. Co-digestion of bovine slaughterhouse wastes, cow manure, various crops and municipal solid waste at thermophilic conditions: a comparison with specific case running at mesophilic conditions.

    A co-digestion process was evaluated when mixing different ratios of agro-industrial residues, i.e. bovine slaughterhouse waste (SB); cow manure (M); various crop residues (VC); and municipal solid waste (MSW) by anaerobic batch digestion under thermophilic conditions (55 °C). A selected study case at mesophilic condition (37 °C) was also investigated. The performance of the co-digestion was evaluated by kinetics (k(0)). The best kinetic results were obtained under thermophilic operation when a mixture of 22% w/w SB, 22% w/w M, 45% w/w VC and 11% w/w MSW was co-digested, which showed a proper combination of high values in r(s)CH(4) and k(0) (0.066 Nm(3)CH(4)/kgVS*d, 0.336 d(-1)) during the anaerobic process. The effect of temperature on methane yield (Y(CH4)), specific methane rate (r(s)CH(4)) and k(0) was also analyzed for a specific study case; there a mixture of 25% w/w of SB, 37.5% w/w of M, 37.5% of VC and 0% of MSW was used. Response variables were severely affected by mesophilic conditions, diminishing to at least 45% of the thermophilic values obtained for a similar mixture. The effect of temperature suggested that thermophilic conditions are suitable to treat these residues.

    Full Text Available While the vertical structure of cold fronts has been studied using various methods, previous research has shown that traditional methods of observing meteorological phenomena (such as pencil-beam radars in PPI/volumetric mode are not well-suited for resolving small-scale cold front phenomena, due to relatively low spatiotemporal resolution. Additionally, non-simultaneous elevation sampling within a vertical cross-section can lead to errors in analysis, as differential vertical advection cannot be distinguished from temporal evolution. In this study, a cold front from 19 September 2015 is analyzed using the Atmospheric Imaging Radar (AIR. The AIR transmits a 20-degree fan beam in elevation, and digital beamforming is used on receive to generate simultaneous receive beams. This mobile, X-band, phased-array radar offers temporal sampling on the order of 1 s (while in RHI mode, range sampling of 30 m (37.5 m native resolution, and continuous, arbitrarily oversampled data in the vertical dimension. Here, 0.5-degree sampling is used in elevation (1-degree native resolution. This study is the first in which a cold front has been studied via imaging radar. The ability of the AIR to obtain simultaneous RHIs at high temporal sampling rates without mechanical steering allows for analysis of features such as Kelvin-Helmholtz instabilities and feeder flow.

  19. Positive Impact on Physical Activity and Health Behaviour Changes of a 15-Week Family Focused Intervention Program: “Juniors for Seniors”

    Full Text Available The promotion of physical activity (PA in children and their parents requires effective planning and sometimes even interventions. This study shows the effect of PA during a 15-week intervention program “Junior for Seniors” by applying a socioecological model to the interpretation of the data. This comprehensive approach emphasizes the fact that health promotion should focus not only on intrapersonal factors but also on the multilevel factors that might be determinants and modulators of increased PA. In 2015, 24 children (“juniors,” 14 girls and 10 boys, aged M=7.96±0.69 and 22 parents (“seniors,” 14 mothers aged M=38.86±2.96 and 8 fathers aged M=37.38±2.97 were voluntarily enrolled in a study spread across three primary schools in the city of Poznań, Poland. The effectiveness of the intervention was determined according to postintervention behavioural changes in PA in comparison to preintervention levels, as reported by the parents and children. Overall, the study found increases in PA levels and reductions in sedentary time. Although the changes are modest, there are some unrecognized benefits of the intervention which may have occurred, such as improved sport and motor skills, more frequent family social behaviours (walks, meals, and visiting relatives, or simply improved quality of “do-together” leisure time PA.

    Full Text Available The purpose of this study is to develop and validate a dissolution test for fluconazole, an antifungal used for the treatment of superficial, cutaneous, and cutaneomucous infections caused by Candida species, in capsules dosage form. Techniques by HPLC and UV first derivative spectrophotometry (UV-FDS were selected for quantitative evaluation. In the development of release profile, several conditions were evaluated. Dissolution test parameters were considered appropriate when a most discriminative release profile for fluconazole capsules was yielded. Dissolution test conditions for fluconazole capsules were 900 mL of HCl 0.1 M, 37 ± 0.5 °C using baskets with 50 rpm for 30 min of test. The developed HPLC and UV-FDS methods for the antifungal evaluation were selective and met requirements for an appropriate and validated method, according to ICH and USP requirements. Both methods can be useful in the registration process of new drugs or their renewal. For routine analysis application cost, simplicity, equipment, solvents, speed, and application to large or small workloads should be observed.

  1. Engagement and Adherence With ezPARENT, an mHealth Parent-Training Program Promoting Child Well-Being.

    Mobile health (mHealth) interventions use mobile technology (tablets and smartphones) delivery platforms for interventions to improve health outcomes. Despite growing acceptance, there is little understanding of how consumers engage with and adhere to mHealth interventions. This study analyzes usage data from the intervention arm ( n = 42) of a randomized clinical trial testing the efficacy of the ezPARENT program and provides recommendations for using engagement and adherence metrics. Engagement was measured by parent usage (duration, frequency, and activity) of ezPARENT and adherence using an adherence index (the sum of individual modules completed, number of visits to ezPARENT, and maximum time between visits). Parents spent M = 37.15 min per module and had M = 13.55 program visits in the 3-month intervention period. Parents visited the program over a period of M = 69.5 days and completed 82% of the modules. These data provide support that parents will use intervention programs delivered digitally; engagement and adherence metrics are useful in evaluating program uptake.

    MALDI imaging mass spectrometry is a highly sensitive and selective tool used to visualize biomolecules in tissue. However, identification of detected proteins remains a difficult task. Indirect identification strategies have been limited by insufficient mass accuracy to confidently link ion images to proteomics data. Here, we demonstrate the capabilities of MALDI FTICR MS for imaging intact proteins. MALDI FTICR IMS provides an unprecedented combination of mass resolving power (~75,000 at m/z 5000) and accuracy (identification based on correlation with LC-MS/MS proteomics data. Analysis of rat brain tissue was performed as a proof-of-concept highlighting the capabilities of this approach by imaging and identifying a number of proteins including N-terminally acetylated thymosin β4 ( m/z 4,963.502, 0.6ppm) and ATP synthase subunit ɛ ( m/z 5,636.074, -2.3ppm). MALDI FTICR IMS was also used to differentiate a series of oxidation products of S100A8 ( m/z 10,164.03, -2.1ppm), a subunit of the heterodimer calprotectin, in kidney tissue from mice infected with Staphylococcus aureus. S100A8 - M37O/C42O3 ( m/z 10228.00, -2.6ppm) was found to co-localize with bacterial microcolonies at the center of infectious foci. The ability of MALDI FTICR IMS to distinguish S100A8 modifications is critical to understanding calprotectin's roll in nutritional immunity.

  3. Divergent electrophysiologic profile of fluconazole and voriconazole in an experimental whole-heart model of proarrhythmia.

    In several case reports a prolongation of the QT-interval and even proarrhythmic effects of fluconazole and voriconazole were reported. The aim of the present study was to investigate if application of fluconazole or voriconazole has the potential to provoke polymorphic ventricular tachycardia in a sensitive model of proarrhythmia. In female rabbits, fluconazole (10, 30 and 50 µM, n=6) and voriconazole (10, 30 and 50 µM, n=6) were infused after obtaining baseline data. Eight endocardial and epicardial monophasic action potentials and a simultaneously recorded 12-lead ECG showed a significant QT prolongation after application of fluconazole as compared with baseline (10 µM:+12 ms, 30 µM:+22 ms, 50 µM:+37 ms; Pfluconazole induced a significant increase (30 µM:+15 ms, 50 µM:+16 ms; Pfluconazole led to the reproducible induction of EADs in 4 of 6 hearts and polymorphic ventricular tachycardia in 3 of 6 hearts (36 episodes). In the present study, voriconazole demonstrated a safe electrophysiologic profile despite significant QT prolongation. In contrast, fluconazole led to a more marked prolongation of myocardial repolarization combined with a more marked increase of dispersion of repolarization. These results imply that application of fluconazole might be torsadogenic and the QT-interval should be closely monitored. Copyright © 2016 Elsevier B.V. All rights reserved.

  4. Return to work following mild traumatic brain injury.

    To examine factors relating to return to work (RTW) following mild traumatic brain injury (mTBI). One hundred and nine patients (Age: M = 37.4 years, SD = 13.2; 52.3% women) who sustained an mTBI. Inception cohort design with questionnaires and neuropsychological testing completed approximately 3 to 4 weeks postinjury. Emergency Department of Tampere University Hospital, Finland. Self-report (postconcussion symptoms, depression, fatigue, and general health) and neurocognitive measures (attention and memory). The cumulative RTW rates were as follows: 1 week = 46.8%, 2 weeks = 59.6%, 3 weeks = 67.0%, 4 weeks = 70.6%, 2 months = 91.7%, and 1 year = 97.2%. Four variables were significant predictors of the number of days to RTW: age, multiple bodily injuries, intracranial abnormality at the day of injury, and fatigue ratings (all P work fewer than 30 days after injury (n = 82, 75.2%) versus more than 30 days (n = 27, 24.8%) did not differ on demographic or neuropsychological variables. The vast majority of this cohort returned to work within 2 months. Predictors of slower RTW included age, multiple bodily injuries, intracranial abnormality at the day of injury, and fatigue.

  5. Levels of "toxic" and "essential" metals in samples of bovine milk from various dairy farms in Calabria, Italy.

    The aim of this research was to evaluate the possible contamination by heavy metals of milk from cows bred on various farms in Calabria. The concentrations of heavy metals were determined in 40 samples of bovine milk from the various dairy farms. Each sample, homogenized and powdered, was mineralized in a microwave oven. Quantitative analyses of Cd, Cr, Cu, Pb and Se were performed using an atomic absorption spectrophotometer with graphite furnace; As was analyzed by hot vapor generation technique and Zn with the flame method. As regards toxic heavy metals, highest values are those of As (g.m. 37.90 microg/kg w.w.) and Pb (g.m. 1.32 microg/kg w.w.), while lowest concentrations are those of Cd (g.m. 0.02 microg/kg w.w). With regard to essential metals, Zn (g.m. 2016 microg/kg w.w) and Se (g.m. 13.24 microg/kg w.w.) showed the highest concentrations, followed by Cr (g.m. 2.03 microg/kg w.w.) and Cu (g.m. 1.98 microg/kg w.w.). Further investigations of the levels of heavy metals (As) in a greater number of milk samples from various zones of Calabria are necessary, both to examine this problem from the clinical epidemiological point of view and to identify the possible causes of milk contamination.

  6. The relationship of depressive symptoms, self-esteem, and sexual behaviors in a predominantly Hispanic sample of men who have sex with men.

    Despite public health campaigns and safer sex messages, many men who have sex with men (MSM) continue to participate in high-risk sexual behaviors, which may make them vulnerable to HIV infection and sexually transmitted infections. The purpose of this study was to determine the relationship of depressive symptoms, self-esteem, and sexual behaviors in a predominantly Hispanic sample of MSM. This correlational study sampled 205 MSM (M = 37 years of age, SD = +/-8) representing the diverse ethnic composition of South Florida. This sample consisted of ethnic minorities (79%) with a large number of foreign-born men (69%). Participants completed measures of depressive symptoms, self-esteem, and sexual behaviors. Results indicated that higher levels of depressive symptoms and higher levels of self-esteem had a statistically significant relationship to lower levels of safer sexual behaviors. Lower income, lower educational level, and preference for Spanish language were associated with higher levels of depressive symptoms; lower income was associated with lower levels of self-esteem; and foreign birth and a preference for Spanish language were associated with lower levels of safer sex behaviors. Higher levels of depressive symptoms and higher levels of self-esteem were associated with high-risk sexual behaviors in this sample of MSM. Further research needs to be directed at culturally specific mental health and HIV prevention strategies for these vulnerable MSM.

  7. The shuttle walk test: a new approach to functional walking capacity measurements for patients after stroke?

    To determine the construct validity, test-retest reliability, and measurement error of the shuttle walk test (SWT) for patients after stroke. Clinimetric study. Three rehabilitation centers in the Netherlands. A sample of patients after stroke (N=75; mean age ± SD, 58.8±9.8y) who are capable of walking without physical assistance. Patients were excluded if they had sustained a subarachnoid hemorrhage or a stroke in the cerebellum or brainstem, or had any other conditions that limited their walking capacity more than the current stroke, or had sensory aphasia. Not applicable. Construct validity (6-minute walk test [6MWT]) and test-retest reliability of the SWT were assessed. Measurement error was determined with the standard error of measurement (SEM), limits of agreement, and smallest detectable differences (SDDs). Construct validity was confirmed by high significant correlations (r(p)≥.65, Pwalking distance in favor of the 6MWT. Test-retest reliability was good (intraclass correlation coefficient model 2,1 [ICC(2,1)]=.961 [.936-.977]). SEM was 6.0%, and the SDDs for individual and group were 302.0m (37%) and 38.7m (5%), respectively. The SWT is a valid and reliable measure and therefore a feasible instrument to determine functional walking capacity of patients after stroke, especially in high-speed walkers.   8. Responses of Blood Glucose, Insulin, Glucagon, and Fatty Acids to Intraruminal Infusion of Propionate in Hanwoo

    Full Text Available This study was carried out to investigate the effects of intraruminal infusion of propionate on ruminal fermentation characteristics and blood hormones and metabolites in Hanwoo (Korean cattle steers. Four Hanwoo steers (average body wt. 270 kg, 13 month of age equipped with rumen cannula were infused into rumens with 0.0 M (Water, C, 0.5 M (37 g/L, T1, 1.0 M (74 g/L, T2 and 1.5 M (111 g/L, T3 of propionate for 1 hour per day and allotted by 4×4 Latin square design. On the 5th day of infusion, samples of rumen and blood were collected at 0, 60, 120, 180, and 300 min after intraruminal infusion of propionate. The concentrations of serum glucose and plasma glucagon were not affected (p>0.05 by intraruminal infusion of propionate. The serum insulin concentration at 60 min after infusion was significantly (p<0.05 higher in T3 than in C, while the concentration of non-esterified fatty acid (NEFA at 60 and 180 min after infusion was significantly (p<0.05 lower in the propionate treatments than in C. Hence, intraruminal infusion of propionate stimulates the secretion of insulin, and decreases serum NEFA concentration rather than the change of serum glucose concentration.

The sample consists of 365 adults aged 18-77 years (M = 37.70, SD = 12.64. Participants were administered two measures of procrastination, the IPS and the Decisional Procrastination Questionnaire, as well as the Big Five Inventory, and the Satisfaction With Life Scale. First, the factor and replication analysis revealed that the internal structure of the scale is clearly one-dimensional, supporting the idea that IPS seems to measure general procrastination as a single trait. Second, the internal consistency is satisfactory as is the temporal stability of the IPS scores. Third, the correlations encountered between the IPS scores and other measures of procrastination, personality traits and satisfaction with life are all in the expected direction and magnitude. Finally, consistent with previous research, procrastination is related to age, with the youngest being the most procrastinating group. This study represents the first attempt in adapting and validating the IPS measure of procrastination into Spanish. Results suggest that the Spanish version of the IPS offers valid and reliable scores when applied to adult population.

    Full Text Available Background The purpose of this study was to investigate the relationship between psychological entitlement (active, passive, and revenge, narcissism and two types of organizational behaviors in employees. Interactions between narcissism and psychological entitlement as predictors of organizational citizenship behaviors (OCB and counterproductive work behaviors (CWB were explored. Predictors were analyzed in an effort to determine whether psychological entitlement plays a more destructive role among narcissistic employees than among non-narcissistic ones and whether the effects of narcissism on OCB and CWB are mediated by entitlement. Participants and procedure Data were obtained from 100 employees (34% men aged 22 to 59 years (M = 37.00, SD = 9.30 from public and private companies. Participants were asked to complete the Organizational Citizenship Behavior Scale (OCBS, the Counterproductive Organizational Behaviors Scale (COBS, the Entitlement Questionnaire, and the Narcissistic Personality Inventory (NPI. Results Positive aspects of entitlement were positively associated with OCB only among narcissistic employees, and active entitlement mediated the effects of narcissism on CWB. Active entitlement was negatively related to CWB. Negative aspects of entitlement were negatively related to OCB and unrelated to CWB. Conclusions This study provides evidence for positive (healthy aspects of entitlement for organizations. The unexpected interaction between narcissism and entitlement in predicting higher levels of OCB suggests that among narcissistic employees, healthy aspects of entitlement are desirable and profitable for an organization. Psychological entitlement was observed to be an important predictor of organizational behaviors beyond narcissism itself.

    Investigations of self-acceptance are critical to understanding the development and maintenance of psychological health. However, valid and reliable instruments for measuring self-acceptance in persons with early blindness have yet to be developed. The current research describes three studies designed to develop and validate the Self-acceptance Scale for Persons with Early Blindness (SAS-EB). In Study 1, we developed the initial item pool. Thirty-three items were generated, based on data from specialized literature and from 2 focus groups. Items were organized in a three-factor structure, theoretically predicted for SAS-EB - (1) body acceptance, (2) self-protection from social stigmas, and (3) feeling and believing in one's capacities. In Study 2, information obtained from a panel of 9 experts and 22 persons with early blindness representing the target population was used to refine the initial item pool, generating a new pool of 27 items. In Study 3, 318 persons with early blindness (141 women and 177 men), between 18 and 60 years of age (M = 37.74 years, SD = 12.37) answered the new pool of 27 items. After the elimination of 9 items using confirmatory factor analysis, we confirmed the theoretical three-factor structure of the SAS-EB. Study 3 also provided support for the scale's internal consistency and construct validity. Finally, the psychometric properties of the SAS-EB, its utility, and its limitations are discussed along with considerations for future research.

    Six runs, each nominally at one week's duration, M-34 through M-39, were completed; this concludes the PDU experimental program for Project Lignite. The first three runs concluded the series to study the effects of different lignites on plant operability and product yields. Runs M-37 and M-38 were made using two Montana subbituminous coals. It was concluded that coals with the highest ash content, both lignite and subbituminous coals, gave greater overall conversion to gaseous and liquid organic products. The subbituminous coals gave lower gas yields and lower overall conversion while producing similar liquid yields when compared with lignite. Reactor solids build up rapidly during initial operations, then level off and remain constant at 29 pounds per cubic foot of reactor volume as a run proceeds. Reactor solids evidently are composed of clay minerals and large particles of pyridine insoluble organic material when processing either lignites or subbituminous coals; in addition calcium/magnesium carbonate crystals (calcite/dolomite) are present in the case of lignite. Long term, trouble-free operation of the deashing unit was not accomplished. Integration of the unit for several hours of combined operation of the process was achieved, and the product from the deashing unit contained as low as 0.6% ash and pyridine insoluble material. Solids buildup in the solids settling tower was a continuing problem.

  14. Measuring parenting practices and family functioning with brief and simple instruments: validation of the Spanish version of the PAFAS.

    A set of instruments with different response formats is usually used to assess parenting practices in clinical settings and in research studies. These complex protocols can be problematic for parents with low-literacy levels. The Parenting and Family Adjustment Scales (PAFAS) is a brief, easy to read instrument that has been developed to address these concerns. The English version of this instrument suggested that it has good internal consistency (range from .70 to .96), as well as satisfactory construct and predictive validity. The aim of the present study was to explore the validity and reliability of the Spanish version of the PAFAS. A sample of 174 Spanish-speaking parents (85% mothers; M = 37 years old; SD = 9.1) from Panama in Central America completed the instrument alongside the Parenting Scale and the Depression Anxiety Stress Scale (DASS-21). Psychometric evaluations revealed that the measure had satisfactory construct and concurrent validity as well as good internal consistency (values >.60 for all subscales) and test-retest reliability (ICC >.60 for all subscales). The PAFAS shows promise as a brief outcome measure to assess parenting practices and family functioning with Spanish-speaking parents. Potential uses of the measure and implications for further validation with diverse samples are discussed.

Tomaram parte deste estudo, 25 profissionais de educação física, com idade entre 27 e 59 anos (M = 37,28. Os entrevistados atuavam em programas de intervenção sócio educacional através do esporte situados no interior de duas favelas do município do Rio de Janeiro. Os professores responderam a uma entrevista semi-estruturada com questões abertas sobre suas experiências com o esporte (benefícios e avaliação, suas orientações sobre os objetivos dos programas, sobre os alunos e sobre os benefícios ou prejuízos do esporte, tanto em termos pessoais quanto entre os participantes dos projetos. Os dados revelam as seguintes evidências: a os professores não conseguem perceber em seus alunos potenciais para crescerem no esporte; b não acreditam no esporte como meio de formação pessoal e profissional; c não acreditam que as condições materiais e de infra-estrutura sejam suficientes para a formação dos alunos; d não percebem em si competência para contribuir na formação dos alunos; e não estão comprometidos com o processo de formação dos alunos.     Full Text Available O efeito de diferentes espaçamentos entrelinhas e entre plantas, foi avaliado nas características produtivas e na conservação pós-colheita de bulbos de cebola. O experimento foi conduzido de abril a novembro de 1998, na Embrapa Semi-Árido, em Petrolina (PE. O delineamento experimental utilizado foi de blocos ao acaso, no esquema fatorial 2x3, compreendendo dois espaçamentos entrelinhas (0,10 e 0,15 m e três espaçamentos entre plantas (0,10; 0,20 e 0,30 m, sendo utilizada a cultivar Franciscana IPA-10 com quatro repetições. O espaçamento entrelinhas com 0,10 m alcançou maior produtividade comercial (39,30 t/ha comparativamente a 0,15 m (37,67 t/ha. Ao se analisar o espaçamento entre plantas verificou-se uma redução linear com o aumento do espaçamento, tendo o espaçamento com 0,10 m propiciado o maior rendimento com 42,88 t/ha. O menor rendimento de bulbos não comerciais foi proporcionado pelos espaçamentos de 0,29 e 0,26 m entre plantas, respectivamente, para os espaçamentos entrelinhas de 0,10 e 0,15 m. O peso fresco dos bulbos aumentou linearmente à medida que se aumentaram os espaçamentos entre plantas. Maior porcentagem de bulbos pequenos e médios foi verificada nos menores espaçamentos. Entretanto, à medida que se incrementou o espaçamento entre plantas ocorreu maior perda de peso pós-colheita.The effect of different planting spacing was observed on the yield characteristics and the postharvest conservation of onion bulbs.     Music is used in healthcare to promote physical and psychological well-being. As clinical applications of music continue to expand, there is a growing need to understand the biological mechanisms by which music influences health. Here we explore the neurochemistry and social flow of group singing. Four participants from a vocal jazz ensemble were conveniently sampled to sing together in two separate performances: pre-composed and improvised. Concentrations of plasma oxytocin and adrenocorticotropic hormone (ACTH) were measured before and after each singing condition to assess levels of social affiliation, engagement and arousal. A validated assessment of flow state was administered after each singing condition to assess participants' absorption in the task. The feasibility of the research methods were assessed and initial neurochemical data was generated on group singing. Mean scores of the flow state scale indicated that participants experienced flow in both the pre-composed (M = 37.06) and improvised singing conditions (M = 34.25), with no significant difference between conditions. ACTH concentrations decreased in both conditions, significantly so in the pre-composed singing condition, which may have contributed to the social flow experience. Mean plasma oxytocin levels increased only in response to improvised singing, with no significant difference between improvised and pre-composed singing conditions observed. The results indicate that group singing reduces stress and arousal, as measured by ACTH, and induces social flow in participants. The effects of pre-composed and improvised group singing on oxytocin are less clear. Higher levels of plasma oxytocin in the improvised condition may perhaps be attributed to the social effects of improvising musically with others. Further research with a larger sample size is warranted.

  18. Development of three Drosophila melanogaster strains with different sensitivity to volatile anesthetics.

    The mechanisms of action for volatile anesthetics remain unknown for centuries partly owing to the insufficient or ineffective research models. We designed this study to develop three strains derived from a wild-type Drosophila melanogaster with different sensitivities to volatile anesthetics, which may ultimately facilitate molecular and genetic studies of the mechanism involved. Median effective doses (ED(50)) of sevoflurane in seven-day-old virgin female and male wild-type Drosophila melanogaster were determined. The sensitive males and females of percentile 6 - 10 were cultured for breeding sensitive offspring (S(1)). So did median ones of percentile 48 - 52 for breeding median offspring (M(1)), resistant ones of percentile 91 - 95 for breeding resistant offspring (R(1)). Process was repeated through 31 generations, in the 37th generation, S(37), M(37) and R(37) were used to determine ED(50) for enflurane, isoflurane, sevoflurane, desflurane, halothane, methoxyflurane, chloroform and trichloroethylene, then ED(50) values were correlated with minimum alveolar concentration (MAC) values in human. From a wild-type Drosophila melanogaster we were able to breed three strains with high, median and low sevoflurane requirements. The ratio of sevoflurane requirements of three strains were 1.20:1.00:0.53 for females and 1.22:1.00:0.72 for males. Strains sensitive, median and resistant to sevoflurane were also sensitive, median and resistant to other volatile anesthetics. For eight anesthetics, ED(50) values in three strains correlated directly with MAC values in human. Three Drosophila melanogaster strains with high, median and low sensitivity to volatile anesthetics, but with same hereditary background were developed. The ED(50) are directly correlated with MAC in human for eight volatile anesthetics.

  19. Group hypnosis vs. relaxation for smoking cessation in adults: a cluster-randomised controlled trial.

    Despite the popularity of hypnotherapy for smoking cessation, the efficacy of this method is unclear. We aimed to investigate the efficacy of a single-session of group hypnotherapy for smoking cessation compared to relaxation in Swiss adult smokers. This was a cluster-randomised, parallel-group, controlled trial. A single session of hypnosis or relaxation for smoking cessation was delivered to groups of smokers (median size = 11). Participants were 223 smokers consuming ≥ 5 cigarettes per day, willing to quit and not using cessation aids (47.1% females, M = 37.5 years [SD = 11.8], 86.1% Swiss). Nicotine withdrawal, smoking abstinence self-efficacy, and adverse reactions were assessed at a 2-week follow-up. The main outcome, self-reported 30-day point prevalence of smoking abstinence, was assessed at a 6-month follow up. Abstinence was validated through salivary analysis. Secondary outcomes included number of cigarettes smoked per day, smoking abstinence self-efficacy, and nicotine withdrawal. At the 6-month follow up, 14.7% in the hypnosis group and 17.8% in the relaxation group were abstinent. The intervention had no effect on smoking status (p = .73) or on the number of cigarettes smoked per day (p = .56). Smoking abstinence self-efficacy did not differ between the interventions (p = .14) at the 2-week follow-up, but non-smokers in the hypnosis group experienced reduced withdrawal (p = .02). Both interventions produced few adverse reactions (p = .81). A single session of group hypnotherapy does not appear to be more effective for smoking cessation than a group relaxation session.   20. Resting energy expenditure prediction in recreational athletes of 18-35 years: confirmation of Cunningham equation and an improved weight-based alternative.

    Resting energy expenditure (REE) is expected to be higher in athletes because of their relatively high fat free mass (FFM). Therefore, REE predictive equation for recreational athletes may be required. The aim of this study was to validate existing REE predictive equations and to develop a new recreational athlete specific equation. 90 (53 M, 37 F) adult athletes, exercising on average 9.1 ± 5.0 hours a week and 5.0 ± 1.8 times a week, were included. REE was measured using indirect calorimetry (Vmax Encore n29), FFM and FM were measured using air displacement plethysmography. Multiple linear regression analysis was used to develop a new FFM-based and weight-based REE predictive equation. The percentage accurate predictions (within 10% of measured REE), percentage bias, root mean square error and limits of agreement were calculated. Results: The Cunningham equation and the new weight-based equation REE(kJ / d) = 49.940* weight(kg) + 2459.053* height(m) - 34.014* age(y) + 799.257* sex(M = 1,F = 0) + 122.502 and the new FFM-based equation REE(kJ / d) = 95.272*FFM(kg) + 2026.161 performed equally well. De Lorenzo's equation predicted REE less accurate, but better than the other generally used REE predictive equations. Harris-Benedict, WHO, Schofield, Mifflin and Owen all showed less than 50% accuracy. For a population of (Dutch) recreational athletes, the REE can accurately be predicted with the existing Cunningham equation. Since body composition measurement is not always possible, and other generally used equations fail, the new weight-based equation is advised for use in sports nutrition.

    Increased teammate familiarity in emergency medical services (EMS) promotes development of positive teamwork and protects against workplace injury. Measures were collected using archival shift records, workplace injury data, and cross-sectional surveys from a nationally representative sample of 14 EMS agencies employing paramedics, prehospital nurses, and other EMS clinicians. One thousand EMS clinicians were selected at random to complete a teamwork survey for each of their recent partnerships and tested the hypothesized role of teamwork as a mediator in the relationship between teammate familiarity and injury with the PROCESS macro. We received 2566 completed surveys from 333 clinicians, of which 297 were retained. Mean participation was 40.5% (standard deviation [SD] = 20.5%) across EMS agencies. Survey respondents were primarily white (93.8%), male (67.3%), and ranged between 21-62 years of age (M = 37.4, SD = 9.7). Seventeen percent were prehospital nurses. Respondents worked a mean of 3 shifts with recent teammates in the 8 weeks preceding the survey (M = 3.06, SD = 4.4). We examined data at the team level, which suggest positive views of teamwork (M = 5.92, SD = 0.69). Our hypothesis that increased teammate familiarity protects against adverse safety outcomes through development of positive teamwork was not supported. Teamwork factor Partner Adaptability and Backup Behavior is a likely mediator (odds ratio = 1.03, P = .05). When dyad familiarity is high and there are high levels of backup behavior, the likelihood of injury is increased. The relationship between teammate familiarity and outcomes is complex.   2. Refining the Jurassic Magnetic Quiet Zone

    We present a coherent marine magnetic reversal record from the Pacific to refine the Jurassic Quiet Zone (JQZ). Our definitive magnetic anomaly record consists of high-resolution sea surface, mid-water (3-km level deep-towed), and near-bottom profiles (0.1-km to the seafloor) with the magnetic source layer constrained by gravity anomaly data and reflection and refraction seismics, all of which are newly collected during TN272 and SKQ2014S2 cruises. All magnetic anomaly data were corrected diurnal variations and the present-day ambient geomagnetic field. In comparing our three-level JQZ magnetic anomaly profiles with previous work in the Japanese lineations, we confirm a globally coherent anomaly sequence in the JQZ from M29 to M42, including the distinctive amplitude envelope decreasing back in time from M19 to M38, with a minimum at M41, and then increasing back in time. A strong similarity in the M37/M38 polarity attributes found both in magnetostratigraphic and marine magnetic records suggest that rapid magnetic reversals were occurring during the M38 time in the JQZ. Seismic and gravity profiles from the Hawaiian JQZ seafloor show late-stage Cretaceous volcanism thickening crust by up to 150% with extra melt emplaced at the Moho, and numerous sills and volcanic cones in the sediment and on the seafloor. The region of thickest crust in the Hawaiian lineation corridor coincides with the region of the lowest JQZ anomaly amplitudes, very similar to the Low Amplitude Zone of Japanese lineation sequence, suggesting that the JQZ anomaly character can represent changes in geomagnetic field intensity over time but is free of the effects of Cretaceous volcanic overprint. We conducted inversion modeling to establish polarity block models and to estimate reversal rates. Reversal rates are the highest during periods with the lowest anomaly amplitudes, indicating a unique period of geomagnetic field behavior in the Earth's history.

  3. Prader-Willi Syndrome Genetic Subtypes and Clinical Neuropsychiatric Diagnoses in Residential Care Adults.

    The historical diagnosis of Prader-Willi syndrome (PWS), a complex genetic disorder, in adults by clinical presentation rather than genetic testing has limited genetic subtype-specific psychometric investigations and treatment. Genetic testing and clinical psychiatric evaluation using DSM-IV-TR criteria were undertaken on 72 adult residents (34M; 38F) from the Prader-Willi Homes of Oconomowoc (PWHO), a specialty PWS group home system. Methylation specific-multiplex ligation probe amplification and high-resolution microarrays were analyzed for methylation status, 15q11-q13 deletions and maternal uniparental disomy 15 (mUPD15). Seventy (33M; 37F) of 72 residents were genetically confirmed and 36 (51%) had Type I or Type II deletions; 29 (42%) with mUPD15 and 5 (7%) with imprinting defects from three separate families. Psychiatric comorbidities were classified as anxiety disorder (38%), excoriation (skin picking) (33%), intermittent explosive disorder [(30%-predominantly among males at 45% compared with females at 16% (OR=4.3, 95%CI 1.4-13.1, p < 0.008)] and psychotic features (23%). Psychiatric diagnoses did not differ between mUPD15 vs deletion, but a greater number of psychiatric diagnoses were observed for larger Type I (4.3) vs smaller Type II (3.6) deletions when age was controlled (F = 5.0, p < 0.04). Adults with PWS presented with uniformly higher rates of psychiatric comorbidities which differed by genetic subtype with gender-specific trends. This article is protected by copyright.   6. User identified positive outcome expectancies of electronic cigarette use: A concept mapping study.

    Electronic cigarette (ECIG) use is growing in popularity, but little is known about the perceived positive outcomes of ECIG use. This study used concept mapping (CM) to examine positive ECIG outcome expectancies. Sixty-three past 30-day ECIG users (38.1% female) between the ages of 18 and 64 (M = 37.8, SD = 13.3) completed a CM module. In an online program, participants provided statements that completed a prompt: "A specific positive, enjoyable, or exciting effect (i.e., physical or psychological) that I have experienced WHILE USING or IMMEDIATELY AFTER USING an electronic cigarette/electronic vaping device is. . . ." Participants (n = 35) sorted 123 statements into "piles" of similar content and rated (n = 43) each statement on a 7-point scale (1 = Definitely NOT a positive effect to 7 = Definitely a positive effect). A cluster map was created using data from the sorting task, and analysis indicated a 7 cluster model of positive ECIG use outcome expectancies: Therapeutic/Affect Regulation, High/Euphoria, Sensation Enjoyment, Perceived Health Effects, Benefits of Decreased Cigarette Use, Convenience, and Social Impacts. The Perceived Health Effects cluster was rated highest, although all mean ratings were greater than 4.69. Mean cluster ratings were compared, and females, younger adults, past 30-day cigarette smokers, users of more "advanced" ECIG devices, and nonlifetime (less than 100 lifetime cigarettes) participants rated certain clusters higher than comparison groups (ps < 0.05). ECIG users associate positive outcomes with ECIG use. ECIG outcome expectancies may affect product appeal and tobacco use behaviors and should be examined further to inform regulatory policies.   7. Land-based infrared imagery for marine mammal detection

    A land-based infrared (IR) camera is used to detect endangered Southern Resident killer whales in Puget Sound, Washington, USA. The observations are motivated by a proposed tidal energy pilot project, which will be required to monitor for environmental effects. Potential monitoring methods also include visual observation, passive acoustics, and active acoustics. The effectiveness of observations in the infrared spectrum is compared to observations in the visible spectrum to assess the viability of infrared imagery for cetacean detection and classification. Imagery was obtained at Lime Kiln Park, Washington from 7/6/10-7/9/10 using a FLIR Thermovision A40M infrared camera (7.5-14μm, 37°HFOV, 320x240 pixels) under ideal atmospheric conditions (clear skies, calm seas, and wind speed 0-4 m/s). Whales were detected during both day (9 detections) and night (75 detections) at distances ranging from 42 to 162 m. The temperature contrast between dorsal fins and the sea surface ranged from 0.5 to 4.6 °C. Differences in emissivity from sea surface to dorsal fin are shown to aid detection at high incidence angles (near grazing). A comparison to theory is presented, and observed deviations from theory are investigated. A guide for infrared camera selection based on site geometry and desired target size is presented, with specific considerations regarding marine mammal detection. Atmospheric conditions required to use visible and infrared cameras for marine mammal detection are established and compared with 2008 meteorological data for the proposed tidal energy site. Using conservative assumptions, infrared observations are predicted to provide a 74% increase in hours of possible detection, compared with visual observations.

  8. The neurochemistry and social flow of singing: bonding and oxytocin

    Music is used in healthcare to promote physical and psychological well-being. As clinical applications of music continue to expand, there is a growing need to understand the biological mechanisms by which music influences health. Here we explore the neurochemistry and social flow of group singing. Four participants from a vocal jazz ensemble were conveniently sampled to sing together in two separate performances: pre-composed and improvised. Concentrations of plasma oxytocin and adrenocorticotropic hormone (ACTH) were measured before and after each singing condition to assess levels of social affiliation, engagement and arousal. A validated assessment of flow state was administered after each singing condition to assess participants' absorption in the task. The feasibility of the research methods were assessed and initial neurochemical data was generated on group singing. Mean scores of the flow state scale indicated that participants experienced flow in both the pre-composed (M = 37.06) and improvised singing conditions (M = 34.25), with no significant difference between conditions. ACTH concentrations decreased in both conditions, significantly so in the pre-composed singing condition, which may have contributed to the social flow experience. Mean plasma oxytocin levels increased only in response to improvised singing, with no significant difference between improvised and pre-composed singing conditions observed. The results indicate that group singing reduces stress and arousal, as measured by ACTH, and induces social flow in participants. The effects of pre-composed and improvised group singing on oxytocin are less clear. Higher levels of plasma oxytocin in the improvised condition may perhaps be attributed to the social effects of improvising musically with others.     Full Text Available O artigo tem por objetivo rastrear sintomatologia de transtorno de estresse pós-traumático (TEPT em profissionais que prestaram ajuda humanitária à população haitiana, após o terremoto de 2010. Estudo transversal. A sintomatologia de TEPT foi avaliada pela Escala Impacto do Evento - Revisada (IES-R. Os participantes foram 32 brasileiros (idade m = 37.58 +/- 7.01, 22 estadosunidenses (idade m = 33.67 +/- 8.03 e 12 equatorianos (idade m = 44.80 +/- 15.88 e não apresentaram sintomatologia de TEPT. A relação entre as variáveis experiência prévia em situação de desastre e escore total da IES-R [F(2 = 4.34, p = 0.017] bem como experiência prévia em situação de desastre e subescala intrusão [F(2 = 3.94, p = 0.024] foram significantes nos modelos de regressão linear. Experiência prévia se mostrou preditor significante para escore total da IES-R (p < 0,05. Os resultados demonstraram que vivências atuais podem ser potencializadas pelas memórias de experiências anteriores, aumentando a probabilidade de desenvolvimento de TEPT. Portanto, o cuidado com a saúde mental dos profissionais deve favorecer a precoce identificação do fator de risco experiência prévia, não permitindo que a iniciativa voluntária se sobreponha aos critérios seletivos e aos cuidados específicos.

  10. [Appearance of anti-red blood cell antibodies in 80 years old and over transfused patients: result of 3 years haemovigilance survey].

    Today most transfusions are given to people over 70. In order to evaluate the production and the circumstances of the appearance of red blood cells (RBC) allo-antibodies (Ab), a three-year study was performed in transfused patients aged 80 and over. Based on the Adverse Event Reports (AER) on RBC Ab from 2007 to 2009 in the Rhône-Alpes area, the prevalence and specificity of the RBC Ab, the type of blood component involved, the imputability and the previous transfusion and obstetrical history were studied. Of 2,169 AER, 240 (11.1%) related to the appearance of RBC Ab; they included 150 females (F) patients (62.5%) and 90 males (M) (37.5%). The Rhesus (RH) blood group was most involved (75 AER) and anti-E was the most frequent Ab (52 cases; 69.3%). Packed RBC were predominantly involved (233 cases; 97,1%). Absolute imputability could be established in 120 cases only (50.0%). Previous transfusion history was observed in 85 F patients (56.7%) and 52M (57.8%). Pretransfusion Ab was noted in 18 F patients (12.0%) and five M (5.6%). Seventy-three F patients (48.7%) had had a pregnancy but the number of unknown data was high (71 F patients; 47.3%). In the transfused patient population aged 80 and over, RBC Ab are common and in most cases are due to RBC transfusions. On the contrary, pretransfusion RBC Ab are not frequent.   11. Waist circumference does not predict circulating adiponectin levels in sub-Saharan women

    Full Text Available Abstract Background Because of previously reported ethnic differences in determinants and markers of obesity and related metabolic disorders, we sought to investigate circulating levels of adiponectin and their correlates in a sub-Saharan African (sSA population. Subjects and Methods We studied 70 non-diabetic volunteers (33M/37F living in Yaoundé, Cameroon, aged 24–69 yr, with BMI 20–42 kg/m2. In all participants we measured waist circumference and total body fat by bioimpedance, and obtained a fasting venous blood sample for measurement of plasma glucose, serum insulin and adiponectin concentrations. We performed a euglycaemic hyperinsulinaemic clamp in 1/4 subjects, and HOMAIR was used as surrogate of fasting insulin sensitivity index since it best correlates to clamp measurements. Results Males had lower adiponectin levels than females (8.8 ± 4.3 vs. 11.8 ± 5.5 μg/L. There was no significant correlation between adiponectin and total body fat (rs = -0.03; NS, whereas adiponectin was inversely correlated with waist circumference (rs = -0.39; p = 0.001. Adiponectin correlated negatively with insulin resistance (rs = -0.35; p = 0.01. In a regression analysis using fasting adiponectin concentration as the dependent variable, and age, HOMAIR, waist circumference, and fat mass as predictors, waist circumference (β = -3.30; p = 0.002, fat mass (β = -2.68; p = 0.01, and insulin resistance (β = -2.38; p = 0.02 but not age (β = 1.11; p = 0.27 were independent predictors of adiponectin. When considering gender, these relations persisted with the exception of waist circumference in females. Conclusion Adiponectin correlates in this study population are comparable to those observed in Caucasians with the exception of waist circumference in women. The metabolic significance of waist circumference is therefore questioned in sSA women.

  12. Online psychophysics: reaction time effects in cognitive experiments.

    Using the Internet to acquire behavioral data is currently on the rise. However, very basic questions regarding the feasibility of online psychophysics are still open. Here, we aimed to replicate five well-known paradigms in experimental psychology (Stroop, Flanker, visual search, masked priming, attentional blink) in three settings (classical "lab", "web-in-lab", "web") to account for possible changes in technology and environment. Lab and web-in-lab data were both acquired in an in-lab setting with lab using "Gold Standard" methods, while web-in-lab used web technology. This allowed for a direct comparison of potential differences in acquisition software. To account for additional environmental differences, the web technology experiments were published online to participate from home (setting web), thereby keeping the software and experimental design identical and only changing the environmental setting. Our main results are: First, we found an expected fixed additive timing offset when using web technology (M = 37 ms, SD = 8.14) and recording online (M = 87 ms, SD = 16.04) in comparison to lab data. Second, all task-specific effects were reproduced except for the priming paradigm, which couldn't be replicated in any setting. Third, there were no differences in error rates, which are independent of the timing offset. This finding further supports the assumption of data equality over all settings. Fourth, we found that browser type might be influencing absolute reaction times. Together, these results contribute to the slowly but steadily growing literature that online psychophysics is a suitable complement - or even substitute - to lab data acquisition.

  13. Neuroimaging and clinical predictors of fatigue in Parkinson disease.

    Fatigue is disabling in Parkinson disease. It is often associated with other non-motor symptoms, but little is known about its underlying pathophysiology. To investigate neuroimaging (using dopaminergic and cholinergic PET) and clinical factors associated with fatigue severity in PD. 133 PD subjects (96M/37F) completed the Fatigue Severity Scale, Movement Disorders Society-Sponsored Revision of the Unified PD Rating Scale (MDS-UPDRS), Hoehn-Yahr staging, validated scales for depression, anxiety, apathy, sleep, and cognition, and underwent [(11)C]methyl-4-piperidinyl propionate (PMP) acetylcholinesterase (AChE) and [(11)C]dihydrotetrabenazine (DTBZ) monoaminergic PET imaging. We explored contributions to PD fatigue using separate regression models based either on neuroimaging parameters or clinicometric scales. In a neuroimaging regression model, neither striatal DTBZ uptake nor AChE PMP uptake were predictors of fatigue in PD. In a post-hoc neuroimaging regression model, stratifying the total cohort into mild vs. moderate-to-severe PD, striatal DTBZ uptake was a significant predictor of fatigue in mild but not moderate-to-severe PD. In a clinicometric regression model, higher Beck Depression Inventory-somatic subscore, higher levodopa dose equivalents and younger age were all significant predictors of fatigue in PD, but the MDS-UPDRS non-motor experiences of daily living score was the best predictor overall. Cholinergic uptake was not a predictor of fatigue in PD, but nigrostriatal dopaminergic denervation predicted fatigue in mild disease. Total non-motor symptom burden, somatic affective symptoms, levodopa dose equivalents, and younger age were independent clinical predictors of fatigue. Copyright © 2015 Elsevier Ltd.   14. A simple parameterization for the height of maximum ozone heating rate

    It is well-known that the height of the maximum ozone heating rate is much higher than the height of the maximum ozone concentration in the stratosphere. However, it lacks an analytical expression to explain it. A simple theoretical model has been proposed to calculate the height of maximum ozone heating rate and further understand this phenomenon. Strong absorption of ozone causes the incoming solar flux to be largely attenuated before reaching the location of the maximum ozone concentration. By comparing with the exact radiative transfer calculations, the heights of the maximum ozone heating rate produced by the theoretical model are generally very close to the true values. When the cosine of solar zenith angle μ0 = 1.0 , in US Standard atmosphere, the heights of the maximum ozone heating rate by the theoretical model are 41.4 km in the band 0.204-0.233 μm, 47.9 km in the band 0.233-0.270 μm, 44.5 km in the band 0.270-0.286 μm, 37.1 km in the band 0.286-0.303 μm, and 30.2 km in the band 0.303-0.323 μm, respectively. The location of the maximum ozone heating rate is sensitive to the solar spectral range. In band 1, the heights of the maximum ozone heating rate by the theoretical model are 52.3 km for μ0 = 0.1 , 47.1 km for μ0 = 0.3 , 44.6 km for μ0 = 0.5 , 43.1 km for μ0 = 0.7 , 41.9 km for μ0 = 0.9 , 41.4 km for μ0 = 1.0 in US Standard atmosphere, respectively. This model also illustrates that the location of the maximum ozone heating rate is sensitive to the solar zenith angle.

  15. Updating phylogeny of mitochondrial DNA macrohaplogroup m in India: dispersal of modern human in South Asian corridor.

    Full Text Available To construct maternal phylogeny and prehistoric dispersals of modern human being in the Indian sub continent, a diverse subset of 641 complete mitochondrial DNA (mtDNA genomes belonging to macrohaplogroup M was chosen from a total collection of 2,783 control-region sequences, sampled from 26 selected tribal populations of India. On the basis of complete mtDNA sequencing, we identified 12 new haplogroups--M53 to M64; redefined/ascertained and characterized haplogroups M2, M3, M4, M5, M6, M8'C'Z, M9, M10, M11, M12-G, D, M18, M30, M33, M35, M37, M38, M39, M40, M41, M43, M45 and M49, which were previously described by control and/or coding-region polymorphisms. Our results indicate that the mtDNA lineages reported in the present study (except East Asian lineages M8'C'Z, M9, M10, M11, M12-G, D are restricted to Indian region.The deep rooted lineages of macrohaplogroup 'M' suggest in-situ origin of these haplogroups in India. Most of these deep rooting lineages are represented by multiple ethnic/linguist groups of India. Hierarchical analysis of molecular variation (AMOVA shows substantial subdivisions among the tribes of India (Fst = 0.16164. The current Indian mtDNA gene pool was shaped by the initial settlers and was galvanized by minor events of gene flow from the east and west to the restricted zones. Northeast Indian mtDNA pool harbors region specific lineages, other Indian lineages and East Asian lineages. We also suggest the establishment of an East Asian gene in North East India through admixture rather than replacement.

  16. Unique barriers and needs in weight management for obese women with fibromyalgia.

    The aim of this study was to identify barriers, needs, and preferences of weight management intervention for women with fibromyalgia (FM). Obesity appears in higher rates in women with fibromyalgia compared to the population at large, and no study to date has taken a qualitative approach to better understand how these women view weight management in relation to their disease and vice versa. We designed a qualitative interview study with women patients with FM and obesity. Women (N = 15) were recruited by their participation in a fibromyalgia treatment program (FTP) within the year prior. The women approached for the study met the following inclusion criteria: confirmed diagnosis of FM, age between 30 and 60 years (M = 51 ± 6.27), and body mass index (BMI) ≥ 30 (M = 37.88 ± 4.87). Patients completed questionnaire data prior to their participation in focus groups (N = 3), including weight loss history, physical activity data, the Revised Fibromyalgia Impact Questionnaire (FIQR), and the Patient Health Questionnaire 9-item (PHQ-9). Three focus group interviews were conducted to collect qualitative data. Consistent themes were revealed within and between groups. Patients expressed the complex relationships between FM symptoms, daily responsibilities, and weight management. Weight was viewed as an emotionally laden topic requiring compassionate delivery of programming from an empathetic leader who is knowledgeable about fibromyalgia. Patients view themselves as complex and different, requiring a specifically tailored weight management program for women with FM. Women with FM identify unique barriers to weight management, including the complex interrelationships between symptoms of FM and health behaviors, such as diet and exercise. They prefer a weight management program for women with FM that consists of an in-person, group-based approach with a leader but are open to a tailored conventional weight management program.   17. Identification of a Functional TPH1 Polymorphism Associated with Irritable Bowel Syndrome Bowel Habit Subtypes

    Background & Aims Alterations in 5-hydroxytryptamine (5-HT) signaling have been implicated as a factor contributing to the altered bowel habit of irritable bowel syndrome (IBS) patients. Tryptophan hydroxylase 1 (TPH1) is the rate–limiting enzyme in enterochromaffin cell 5-HT biosynthesis. We hypothesized that genetic variants affecting TPH1 gene expression might alter intestinal 5-HT bioavailability and subsequently the propensity for distinct bowel habit subtypes in IBS. In this study, we assessed the only common TPH1 proximal promoter variant (-347C/A; rs7130929) and its association with bowel habit predominance in IBS. Methods Electrophoretic mobility shift assays were performed to assess whether the -347C/A allele variant affects the DNA-binding of nuclear factors. Genotype distribution was determined for 422 IBS patients subtyped using Rome III criteria and for 495 healthy controls recruited from two university medical centers. Association with bowel habit was tested using a multinomial logistic regression model controlling for race, anxiety, depression, and study site. Results Early growth response factor 1 (EGR-1) bound with higher affinity to a site comprising the minor A-allele of SNP -347C/A. TPH1 genotype frequencies did not differ between IBS patients and controls overall. The CC genotype was more prevalent in the IBS-D subtype (47%) than in the IBS-C (25%) and IBS-M (37%) subtypes (P=0.039) after adjusting for race and other covariates. Colonic biopsies from a small cohort of IBS patients from one center were tested for higher TPH1 mRNA expression in samples with CC compared to CA genotype, but the results did not reach statistical significance. Conclusions The TPH1 promoter SNP -347C/A differentially binds EGR1, correlates with IBS bowel habit subtypes and possibly colonic TPH1 expression consistent with its role in modulating intestinal 5-HT signaling.   18. Cell apoptosis and lipid content of in vitro-produced, vitrified bovine embryos treated with forskolin.

    The presence of fetal calf serum in culture medium influences embryo quality, causing a reduction in postcryopreservation survival. Forskolin has been used to induce lipolysis and increase cryotolerance, functioning as an activator of adenylate cyclase and elevating cAMP levels. In the present experiment, bovine zygotes were cultured in synthetic oviduct fluid with amino acid plus 2.5% fetal calf serum for 6 days, when forskolin was added in three concentrations: 2.5, 5, and 10 μM. Treatment with forskolin lasted for 24 hours. Blastocyst formation rate, quantification of lipid granules, total cell numbers, and apoptosis rate were evaluated. In a second assessment, embryos were vitrified, and warming, re-expansion rate, total cell numbers, and apoptosis rate were also evaluated. There was no difference due to forskolin in blastocyst formation or re-expansion rates after vitrification. However, lipid measurements were lower (control: 136.8 and F 2.5 μM: 128.5; P < 0.05), and number of cells per embryo higher (control: 140.1 and F 2.5 μM: 173.5; P < 0.05) than controls for 2.5 μM forskolin but not for higher forskolin concentrations. The number of intact cells per embryo was higher, and the rate of apoptosis was lower in fresh than in vitrified embryos (number of cells of warmed embryos, control: 104.1, F 2.5 μM: 101.3, F 5 μM: 115.4, F 10 μM: 95.1; apoptotic of fresh cells, control: 12.1%, F 2.5 μM: 16.7%, F 5 μM: 11.1%, F 10 μM: 14.2%; and apoptotic warmed embryos, control: 22.3%, F 2.5 μM: 37.3%, F 5 μM: 33.2%, F 10 μM: 30.3%; P < 0.05). It was concluded that forskolin is an effective lipolytic agent even at low concentrations, leading to formation of blastocysts with a comparatively larger number of cells.   19. Corneal endothelial autocrine trophic factor VIP in a mechanism-based strategy to enhance human donor cornea preservation for transplantation.

    Vasoactive intestinal peptide (VIP) and ciliary neurotrophic factor (CNTF) are identified as autocrines of human corneal endothelial (CE) cells working in concert to maintain the differentiated state and promote the survival of the corneal endothelium. From VIP gene knockdown study, endogenous VIP is shown to maintain the level of the differentiation marker, the adhesion molecule N-cadherin, CE cell size, shape, and retention, in situ in the human donor corneoscleral explants. Exogenous VIP protects the corneal endothelium against the killing effect of oxidative stress, in part by upholding ATP levels in CE cells dying of oxidative stress-induced injury, allowing them to die of an apoptotic death instead of an acute necrotic one. The switch from the acute necrosis to the programmed cell death (apoptosis) may have allowed the injured CE cell to be rescued by the VIP-upregulated pathways, including those of Bcl-2 and N-cadherin, and resulted in long-term CE cell survival. The endogenous VIP in CE cells is upregulated by CNTF, which is released by CE cells surviving the oxidative stress. The CNTF receptor (CNTFRα) is expressed in CE cells in human donor corneoscleral explant and gradually becomes lost during corneal storage. VIP treatment (10(-8) M, 37 °C, 30 min) prior to storage of freshly dissected human donor corneoscleral explants increases their CE cell CNTFRα level and responsiveness to CNTF in upregulating the gap junctional protein connexin-43 expression. VIP treatment of both fresh and preserved corneoscleral explants reduces CE damage in the corneoscleral explants and in the corneal buttons trephined from them. CE cell loss is a critical risk factor in corneal graft failure at any time in the life of the graft, which can be as late as 5-10 years after an initially successful transplant. Through the use of a panel of genetically diverse mouse inbred strains, we developed a drug screening platform aimed at examining interstrain differences in viability on normal, noncancerous immune cells following chemotherapeutic cytotoxic insult. Drug effects were investigated by comparing selective chemotherapeutic agents, such as BEZ-235 and selumetinib, against conventional cytotoxic agents targeting multiple pathways, including doxorubicin and idarubicin. Methods: Splenocytes were isolated from 36 isogenic strains of mice using standard procedures. Of note, the splenocytes were not stimulated to avoid attributing responses to pathways involved with cellular stimulation rather than toxicity. Cells were incubated with compounds on a nine-point logarithmic dosing scale ranging from 15 nM to 100 µM (37°C, 5% CO2. The objectives of this study were as follows: to analyze the influence of age and education level on cognitive domains; to verify whether being parents and living at home with parents affect both parenting style and cognitive domains; to investigate how the type of the maternal and paternal parenting independently affects cognitive styles; to measure the predictive variables for the use of cognitive dysfunctional patterns and to investigate age as a moderating variable of the relation between parenting styles and cognitive domains in a group of adult men and women. The research involved 209 adults (118 males and 91 females living in Sicily (Italy aged between 20 and 60 years (M = 37.52; SD = 11.42. The research lasted for 1 year.     Full Text Available In this study, we present summertime concentrations and fluxes of biogenic volatile organic compounds (BVOCs measured at a sub-arctic wetland in northern Sweden using a disjunct eddy-covariance (DEC technique based on a proton transfer reaction mass spectrometer (PTR-MS. The vegetation at the site was dominated by Sphagnum, Carex and extit{Eriophorum} spp. The measurements reported here cover a period of 50 days (1 August to 19 September 2006, approximately one half of the growing season at the site, and allowed to investigate the effect of day-to-day variation in weather as well as of vegetation senescence on daily BVOC fluxes, and on their temperature and light responses. The sensitivity drift of the DEC system was assessed by comparing H3O+-ion cluster formed with water molecules (H3O+(H2O at m37 with water vapour concentration measurements made using an adjacent humidity sensor, and the applicability of the DEC method was analysed by a comparison of sensible heat fluxes for high frequency and DEC data obtained from the sonic anemometer. These analyses showed no significant PTR-MS sensor drift over a period of several weeks and only a small flux-loss due to high-frequency spectrum omissions. This loss was within the range expected from other studies and the theoretical considerations.

    The nature of the Jurassic Quiet Zone (JQZ) has been a long-standing debate in understanding Earth's geomagnetic field history and behavior. We present a coherent and likely globally significant marine magnetic reversal record for the JQZ by constructing a correlation of new and previously acquired magnetic anomaly profiles in the western Pacific. We obtained a high-resolution marine magnetic anomaly record using sea surface, mid-water (3-km level deep-towed), and near-bottom (Autonomous Underwater Vehicle (AUV)) profiles that targeted a spreading corridor in the Hawaiian lineation in 2011 (TN272 on R/V Thompson) and 2014 (SKQ2014S2 on R/V Sikuliaq). To extract crustal magnetic signals, the sea surface and mid-water magnetic data were corrected for ship-to-sensor offset, the diurnal effect, and the present-day ambient geomagnetic field. Mid-water data were upward continued to a constant 3 km level plane and to the sea surface. Near-bottom data were calibrated to remove the induced magnetic field by AUV Sentry, then corrected for IGRF and diurnal variations. We used these near-source data as an anchor for correlations with the sea surface and mid-water level data because of the AUV's superb inertial navigation and hydrodynamically stable, quiet platform environment. Our sea surface anomaly correlation with the previously established Japanese lineation sequence shows (i) an excellent correlation of anomaly shapes from M29 to M42; (ii) a remarkable similarity in anomaly amplitude envelope, which decreases back in time from M19 to M38, with a minimum at M41, then increases back in time from M42; and (iii) refined locations of pre-M25 lineations in the Hawaiian lineation set. Moreover, short-wavelength anomalies from the mid-water and near-bottom profiles show a strong similarity in the M37/M38 polarity attributes found both in the magnetostratigraphic and marine magnetic records, implying that rapid magnetic reversals were occurring at that time.     The present work was carried out at the Faculty of Agriculture and Animal Science, UNESP-Jaboticabal. Three ¾ Holstein x Zebu bovines fistulated on rumen, abomasum and intestine using a fixed cannula type T were used. Four types of feed: corn (M, 1% EE corn gluten meal (G1, 7% EE corn gluten meal and 10% EE corn gluten meal (G10 were tested.  All feed were also extruded: CornEx, G1Ex, G7Ex and G10EX. Ruminal incubation was done using the nylon bags technique, and degradability of dry matter, crude protein and disponibilization of amino acids were analyzed. Basic diet was composed of corn gluten meal, soybean meal and hay at roughage (70: concentrate (30 ratio. Ruminal degrading of dry matter feed M, G1, G7 and G10 presented the following values: 37.5%, 56.7%, 56.8% and 55.1% respectively.  Extruded feed: 52.3%, 68.9%; 69.0% and 61.6% for Mex, G1Ex, G7Ex and G10Ex. The corn was significantly inferior to others and extruded feed increased ruminal degradedly for all feed. The degradability of crude protein was 27.0%, 60.9%, 56.8% and 35.1% for M, G1, G7 and G10; 50.8, 52.2, 66.4 and 59.6 for MEx; G1Ex; G7Ex and G10Ex, respectively. Statistically the G1 feed was equal to M and both were inferior to G7 and G10. The extrusion process did not increase the degradability of crude protein of G1. The lysine presented high ruminal degradability for tested feed, showing values above 99%. The metionin presented values of degradability inferior for M (37.4% related to G1 (57.1%.  The treonine did not present differences between feed and extrusion. It can be concluded that the processing of corn to obtain by-products can increase the ruminal degradability of dry matter, protein and metionine, but depending on intensity of processing, the degradability of crude protein may not be altered when compared to corn. The extrusion increased the ruminal degradability of dry matter of corn and by-products.

    Full Text Available One hundred five (54M, 37F, 13L, 1 adult of unknown sex recently dead Cyprus mouflon were collected from November 1985 to January 1998 from Pafos forest in Cyprus. Most carcasses (37.2% were recovered in the fall (18M, 20F, 1L followed by winter (31.4%; 24M, 8F, 0L, Spring (22.9%; 10M, 3F, 11L, and finally Summer (8.6%; 2M, 6F, 1L. The highest mortality of rams (13 rams occurred in January, followed by December (10 rams, November (8 (during the rutting season, and September (6. Seventeen (45.9% females were found dead in late fall, 6 (16.2% in early winter, 6 (16.2% in summer, 3 (8.1% in spring and 3 (8.1% in early fall. During the long dry summer of Cyprus, forage is reduced in quality and quantity. The high energy requirements of the rut, poor nutrition and health problems, in combination with the first winter cold, likely leads to the increased mortality observed in late fall and early winter.

    Full Text Available From a total of 187 fecal samples from children with ages between 0 and 5 years, collected in the Hospital Universitário -USP, Brazil, from 1994 to 1996, 54 (28.9% were positive for rotavirus. Positive samples were characterized by electropherotyping, subgrouping, G serotype and genotype and P genotype. Rotavirus electropherotypes were characterized in four different long genome patterns (38.9%, one short genome pattern (34.0% and 18.0% were characterized as an unusual pattern. Subgroup I was found in 38.9% strains, subgroup II in 50.0% and 7.7% was subgroup nonI-nonII. For G serotypes, G2 was found in 59.3%, G1 was identified in 33.3% of strains, two samples showed mixtures of G1+G2 and one sample was G1+G3. Ten samples characterized as serotype G2 showed a long eletropherotype. Genotype G2 was the most frequently and was found in 37 (44.0% samples (23 samples as a single genotype and 14 as mixtures of genotypes. G1 was found in 15 samples. G3 and G4 was detected mainly in mixtures of genotypes and G5, G6 and G9 were identified only in mixtures. A total of 20 (38.5% samples were characterized as G genotype mixtures and P mixtures were found in 16 (29.6% samples. P[4] was found in 55.6% of samples, P[8] in 51.9% and P[6-M37 like] in 22.3% of cases. P[6-Gottfried like] and P[11] were detected only in mixtures.     Full Text Available O estudo objetivou descrever o comportamento de adesão ao tratamento anti-retroviral em pessoas vivendo com HIV/AIDS e investigar preditores da adesão entre as variáveis escolaridade, presença de efeitos colaterais, interrupção anterior da terapia anti-retroviral (TARV por conta própria, auto-estima, expectativa de auto-eficácia, estratégias de enfrentamento, suporte social e satisfação com a relação profissional de saúde-usuário. Adesão foi medida pelo auto-relato da perda do número de comprimidos/cápsulas dos medicamentos anti-retrovirais na última semana e mês, sendo considerada satisfatória na ocorrência de omissão inferior a 5% do total prescrito. Participaram 101 pessoas, 60,4% homens, idades entre 20 a 71 anos (M = 37,9 anos, 73,3% sintomáticos. A coleta de dados incluiu entrevista e instrumentos auto-aplicáveis. A maioria (n = 73; 72,3% relatou adesão igual ou superior a 95%. Nos resultados da regressão logística, interrupção anterior da TARV e expectativa de auto-eficácia foram preditores significativos da adesão. Faz-se necessária a qualificação da assistência pela constituição de equipes interdisciplinares, para o desenvolvimento de abordagens adequadas às dificuldades médicas e psicossociais de adesão das pessoas vivendo com HIV/AIDS.This study aimed to describe the adherence of persons living with HIV/AIDS to antiretroviral therapy (ART and to investigate adherence predictors among the following: level of schooling, presence of side effects, current or previous interruption of ART by the persons themselves, self-esteem, self-efficacy expectation, coping strategies, social support, and satisfaction with the health professional-patient relationship.
