    Full Text Available O presente estudo teve como objetivo avaliar o efeito da aplicação de um análogo sintético da PGF2 (cloprostenol sodico, associado à ocitocina ou carbetocina, sobre a eficiência da indução ao parto em suínos. O experimento I foi realizado com 284 fêmeas, distribuídas em quatro tratamentos: cloprostenol; cloprostenol e 0,100mg de carbetocina; cloprostenol e 10UI de ocitocina; solução salina (NaCl 0,9%. No experimento II, foram utilizadas 276 fêmeas, distribuídas em quatro tratamentos: cloprostenol; cloprostenol e 0,10mg de carbetocina; cloprostenol e 0,05mg de carbetocina; cloprostenol e 10UI de ocitocina. A indução do parto foi realizada aos 113 dias de gestação, pela aplicação de 0,175mg de cloprostenol, via submucosa vulvar. A carbetocina ou a ocitocina foram aplicadas 24h após a aplicação de cloprostenol, pela via intramuscular. O intervalo indução-parto foi menor (P0,05 entre os tratamentos. A utilização de ocitócitos, em associação com cloprostenol, resulta em partos antecipados e mais sincronizados. O uso associado de cloprostenol e carbetocina reduz o tempo de parto e 99% ou mais dos partos ocorrem em até quatro horas após a aplicação de carbetocina, independentemente da dose utilizada.The aim of the present study was to evaluate the effect of a synthetic analogue of PGF2 (sodium cloprostenol associated to carbetocin or oxytocin on the efficiency of farrowing induction in swine. In Experiment I, 284 females were distributed in four treatments: - cloprostenol; - cloprostenol and 0.10mg of carbetocin; - cloprostenol and 10UI of oxytocin; and saline solution. By transrectal ultrasonography on days 1, 4, 10, 16, 22, 28, 34, 40 and 46 after birth, were evaluated uterus and ovaries.     The objective of the experiment was to compare the use of a PGF2α analogue (Cloprostenol) IM, with an intravaginal progestagen sponge, flurogestone acetate (FGA), and equine chorionic gonadotropin (eCG) IM application protocol. A total of 30 cyclical hair ewes (54.07 ± 0.5 kg live weight, body condition score 3.5 ± 0.5, and age 3 ± 1 years) were used. For the control group ewes (n = 15), intravaginal sponges (IS) impregnated with 20 mg of FGA were inserted for 12 days with 500 IU of eCG IM at sponges withdrawal. For the PG group ewes (Treatment group n = 15), two injections of Cloprostenol (75 mcg) were given 12 days apart. The presence of estrus was detected using two rams with 8 h interval beginning at the end of the treatment. Progesterone concentrations in blood were measured by solid phase radioimmunoassay. A student's t test was performed to analyze the duration of estrus and the interval between the ends of the treatment and the onset of estrus (ET-OE) presentation. Progesterone levels were compared with two-way ANOVA, with treatment, and day of menstrual cycle as fixed factors. Treatment costs ratio was calculated by dividing the total costs of FGA IS application between total costs of Cloprostenol application. Significant differences (P Progesterone levels in both treatments. Treatment costs of Cloprostenol protocol were 2.63 cheaper than FGA including disposable material, biological products, and labor. It was concluded that Cloprostenol could be an effective tool in estrus synchronization in hair sheep in tropical areas.

    Full Text Available Abstract Background The present study describes the clinical features and plasma profiles of bovine pregnancy-associated glycoprotein 1 (bPAG1, the main metabolite of prostaglandin F2α (PG metabolite and progesterone (P4 in heifers in which early abortions were induced. Methods Early abortions were induced in four heifers with cloprostenol and monitored by ultrasonography. Blood samples were collected and the plasma were analyzed for bPAG 1, P4 and PG metabolite. Results The foetal heartbeat rates varied from 170–186 beats per minute for all foetuses up to the date of cloprostenol treatment. Foetal death was confirmed within two days after cloprostenol treatment. Prior to cloprostenol injection, blood plasma concentrations of bPAG1, PG metabolite and P4 varied from 8.4 – 40.0 ng/mL, 158 – 275 pmol/L and 20.7 – 46.9 nmol/L, respectively. After the foetus expelled, the plasma level of bPAG1 began to decrease but the decrease was small and gradual. The estimated half-life of bPAG1 was 1.8 – 6.6 days. The plasma level of the PG metabolite started to have short lasting peaks (above 300 pmol/L within three hours after cloprostenol treatment. The plasma concentrations of P4 dropped sharply to less than 4 nmol/L after 24 hours of cloprostenol injection. Conclusion The current findings indicated that after early closprostenol-induced foetal death, the plasma concentration of bPAG1 decreased gradually and showed a tendency of variation with the stages of pregnancy.

  6. D-Cloprostenol enhances estrus synchronization in tropical hair sheep.

    To compare the effects of PGF2α (dinoprost tromethamine) and D-cloprostenol in a two-dose protocol for estrus synchronization in hair sheep during breeding season in Yucatán, México, two experiments were conducted. In experiment 1, 61 cyclic hair sheep were divided into two groups: G1 (control n = 30), two doses of 50 μg of dinoprost tromethamine IM with 12 days between applications, and G2 (n = 31), two doses of 50 μg of D-cloprostenol IM at the same time interval. For determination of progesterone levels, 16 ewes from each group were randomly selected. In experiment 2, 70 cyclic hair sheep were assigned at the same treatments (G1 and G2, n = 35) and 48 h after the second application, the ewes in estrus were detected by two vasectomized rams. Sheep with detected estrus were inseminated, and 45 days after, pregnant animals were identified by ultrasonography. An exact Fisher's test was performed for the analysis of ewes in estrus (experiments 1 and 2) and number of pregnant ewes (experiment 2); for the comparison of time between end of treatment-estrus presentation, a survival analysis was used. Duration of estrus in hours was analyzed using a generalized mixed model (GLM) ANOVA whereas plasma progesterone concentrations were analyzed by non-linear regression. There were significant differences (P  0.05) in duration of estrus (G1, 42 + 6.1 h, vs G2, 41.1 + 11.2 h, experiment 1) and pregnancy in the ewes that presented estrus, and were inseminated (G1, 38.4% vs 52.1%, experiment 2). With regard to concentrations of progesterone, significant differences (P < 0.01) were found between treatments, and progesterone levels before the second application of D-cloprostenol were higher. In consideration of the results, it can be concluded that in a two-dose protocol of a luteolytic agent, more ewes presented estrus in response to D-cloprostenol compared to dinoprost tromethamine with similar pregnancy rates.

  7. Ovarian and endocrine responses in tropical sheep treated with reduced doses of cloprostenol.

    The present study aimed to assess the efficacy of reduced doses of cloprostenol for synchronizing estrus and ovulation in hair sheep. With the aim to evaluate the luteolytic activity of reduced cloprostenol doses, a first experiment was performed using a relatively large (group H: 126 microg; n=8), medium (group M: 68.25 microg; n=6) and small (group L: 38.5 microg; n=6) cloprostenol dose. Luteolysis was assessed at Days 3 and 6 after injection (Day 0) by progesterone concentrations (P(4)) and transrectal ultrasonography (US). In Experiment 2, sheep were randomly assigned to the same three doses to evaluate a protocol for estrous synchronization using two injections administered 9 days apart. A third trial was performed with ewes treated (9 days apart) with the large dose (H=126 microg; n=12) and with a small dose adjusted for facilitating volume management (LA=43.75 microg; n=12). Presence of estrous cycling was determined in all the ewes by US and P(4) assay, at Days -9, -6, -2, 0 (Day of second cloprostenol injection), 8 and 11. Bleeding and US were done every 4h from 16 h of the beginning of the estrus during the third trial to assess the preovulatory LH surge and timing of ovulation. Additionally, blood samples were drawn at Days 0, 1, 2 and 3 to assess estradiol (Experiments 2 and 3) and P(4) (Experiment 2) concentrations during the ovarian follicular phase. In all experiments, percentage of animals showing luteolysis, preovulatory follicular dynamics and function and percentage of ewes showing behavioral estrus in response to treatment was similar among groups. Timing of estrus for group H was earlier than group L (28.6+/-1.8h compared with 37.1+/-2.4h; Psheep, with the additional advantage of reducing treatment costs.

    Full Text Available The aim of this study was to compare the reproductive efficiency in dairy cows with retained placenta (RP, treated with Florfenicol associated or not with Cloprostenol. One hundred and eight cows with RP from dairy herds in southern Minas Gerais state and northern São Paulo state, were included for this purpose. The experiment was designed in factorial 2X2 . The four treatments encompassed two levels of each factor : 1 Florfenicol - 4 0mg/kg ( IM divided in two injections or 40mg / kg ( SC in one injection after calving; 2 Cloprostenol (0.530mg - two injections associated with florfenicol treatment or without cloprostenol. The analysis of discrete variables used the Kruskal - Wallis (fou r groups or Wilcoxon (two groups statistic to compare groups. The Student "T" test was used to compare means of continuous variables. The overall occurrence of RP was 20.7%. The florfenicol groups had no effects on postpartum uterine health or subsequent reproductive efficiency of the cows. The cloprostenol injections of RP hastened the uterine involution and decreased the occurrence of uterine infection, and increased the percentage of animals with corpus luteum between 25 and 40 days after parturition. The intervals from parturition to first artificial insemination (AI and to conception, as well as the number of AI per conception were reduced in the cloprostenol treatment.

    This study evaluated the effect of two doses of prostaglandin at different intervals on reproductive parameters of crossbred ewes. In Experiment 1, 30 ewes received two doses of 120 μg cloprostenol at 7 (G 7 days ), 9 (G 9 days ), or 11.5 (G 11.5 days ) days apart. Ultrasound assessments were performed from the first and second cloprostenol administration for 5 days or ovulation detection. Estrus signs were checked by a teaser male. Plasma progesterone concentration was measured before each cloprostenol dose. In Experiment 2, 95 ewes were allocated into the same treatments and after the second dose, ewes in estrus were mated. At 30 days after breeding, pregnancy diagnosis was conducted and prolificacy was evaluated at lambing. In Experiment 1, at the first cloprostenol administration, 50% of ewes had an active CL and all showed estrus. At the second administration, 66.7% of ewes had an active CL and one did not present estrus. There was no difference (P > 0.05) after the second dose for as follows: overall estrous response (90%), interval from cloprostenol administration to estrous onset (42.0 ± 4.9 h), estrus duration (31.5 ± 2.1 h), ovulation rate (100.0%), and number of ovulations (1.5 ± 0.3). In Experiment 2, both pregnancy and prolificacy rates were similar (P > 0.05) for G 7 days (73.3; 145%), G 9 days (75.9; 125%), or G 11.5 days (75.9; 145%), leading to an overall pregnancy rate of 75.0% (66/88) and prolificacy rate of 137% . Therefore, the three treatments proposed were able to promote high pregnancy and prolificacy rates in crossbred ewes.

    Full Text Available O objetivo deste trabalho foi avaliar o efeito da dupla aplicação de d-cloprostenol associada ou não à gonadotrofina coriônica equina (eCG sobre o desempenho reprodutivo de cabras ½ Boer/SRD exploradas na região semiárida do Nordeste do Brasil. Utilizaram-se quarenta cabras distribuídas em dois protocolos de sincronização do estro, como a seguir: T1 (n = 20, duas aplicações de 75 µg de d-cloprostenol intervaladas de sete dias; T2 (n = 20, semelhante ao protocolo anterior, diferindo apenas pela adição de 300 UI de eCG na última aplicação de d-cloprostenol. A porcentagem de animais em estro (65% vs. 85%, bem como o intervalo entre o fim do tratamento e o início do estro (22,8 ± 13,4 h vs. 17,2 ± 7,6 h não diferiram (P>0,05 entre T1 e T2, respectivamente. Todavia, o T2 apresentou maior duração do estro que o T1 (P<0,05, sendo de 47,3 ± 12,3 h e 32,9 ± 11,9 h, respectivamente. Observaram-se  fertilidade e prolificidade, respectivamente, de 61,5% e 1,4 para o T1 e de 41,2% e 1,8 para o T2, não havendo diferença significativa entre os tratamentos. As aplicações de d-closprotenol em duas doses intercaladas de sete dias mostraram-se tecnicamente viáveis na sincronização do estro. Nas condições deste experimento, a adição de 300 UI de eCG não melhorou as taxas de fertilidade nem de prolificidade.

    Full Text Available El objetivo del presente trabajo fue evaluar la respuesta de la aplicación doble o triple de prostaglandina sobre la regresión de CL, ovulación y retorno al celo después de un tratamiento superovulatorio en vacas donantes de embriones. Para este experimento se tomaron 44 vacas asignadas al azar a dos grupos de tratamiento. Al primero (n = 23; CLO 0 & 7 se le aplicó una dosis de 500 μg de cloprostenol los días 0 (denominado día de la colecta y 7; al segundo grupo (n = 21; CLO 0, 1 & 7 se le aplicó una inyección adicional el día 1. Los animales fueron examinados por ultrasonografía los días 0, 4, 7, 10 y 13 para evaluar la regresión de cuerpos lúteos y la ovulación. Se realizó observación del celo dos veces al día (mañana y tarde. El 60,8 y 52,3% de los grupos CLO 0 & 7 y CLO 0, 1 & 7, respectivamente, presentaron signos de celo. No se presentaron diferencias en el día del celo entre los grupos (7,2 ± 0,3, 7,3 ± 0,5 días. Tampoco hubo diferencia para la variable ovulación (8,5 ± 0,9 y 9,8± 0,9 días. Todas las vacas tratadas regresaron los cuerpos lúteos antes de finalizar el experimento. Cuando se analizó el tiempo de ovulación, se encontró que las vacas con 4 o menos cuerpos lúteos ovulan en un tiempo menor (P < 0,01 que las vacas con 5 o más cuerpos lúteos. Se concluye que no hay diferencia entre la doble o triple aplicación de cloprostenol para las variables. Sería innecesaria una aplicación adicional de cloprostenol el día 7.

    Full Text Available The studies made by us are based on fundamental biological role of prostaglandin modulator of hormonal actions, influxs nerve transmission and cellular ionic exchange. One of the main actions of PGF2α is luteolyse, a process that includes the secretion of progesterone and regression yellow body. PGF2α is a luteolitic for almost species: cow, buffalo, sow, mare, sheep, rabbits, guinea pigs, hamsters and rats, except primates.     Full Text Available Avaliou-se a eficácia da dose de 0,10 ou de 0,25mg de gonadorelina nas taxas de ovulação e de sincronização no protocolo Ovsynch e de 150mcg de D-cloprostenol ou 0,530mg de cloprostenol sódico na regressão do corpo lúteo (CL de sete dias e de todos os CL. Foram utilizadas 136 vacas lactantes da raça Holandesa, com média de produção de leite de 23,75,8kg/dia, com 138,4±72,0 dias em lactação. As vacas foram distribuídas aleatoriamente em quatro grupos, de acordo com a dose de gonadorelina e o tipo da prostaglandina. As taxas de ovulação e de sincronização foram de 52,9% e 80,9% para 0,10mg de gonadorelina e de 57,4% e 80,9% para 0,25mg de gonadorelina, respectivamente. A taxa de regressão do CL de sete dias foi de 97,1% para o D-cloprostenol e de 97,5% para o cloprostenol sódico. Lactating Holstein cows (n=136, producing 23.75.8kg milk/d and 138.4±72.0 days in milk were randomly assigned to four groups, according to dose of gonadorelin and prostaglandin type.     The aim of the current study was to determine the possible effects of progestagen oestrous synchronization on vascular endothelial growth factor (VEGF) expression during sheep luteogenesis and the peri-implantation period and the relationship with luteal function. At days 9, 11, 13, 15, 17 and 21 of pregnancy, the ovaries from 30 progestagen treated and 30 ewes cycling after cloprostenol injection were evaluated by ultrasonography and, thereafter, collected and processed for immunohistochemical evaluation of VEGF; blood samples were drawn for evaluating plasma progesterone. The progestagen-treated group showed smaller corpora lutea than cloprostenol-treated and lower progesterone secretion. The expression of VEGF in the luteal cells increased with time in the cloprostenol group, but not in the progestagen-treated group, which even showed a decrease between days 11 and 13. In progestagen-treated sheep, VEGF expression in granulosa-derived parenchymal lobule capillaries was correlated with the size of the luteal tissue, larger corpora lutea had higher expression, and tended to have a higher progesterone secretion. In conclusion, the current study indicates the existence of deleterious effects from exogenous progestagen treatments on progesterone secretion from induced corpora lutea, which correlate with alterations in the expression of VEGF in the luteal tissue and, this, presumably in the processes of neoangiogenesis and luteogenesis. © 2010 Blackwell Verlag GmbH.

    Current study assessed differences in the response of sheep to estrus synchronization either by the administration of two doses of prostaglandin or by the insertion of an intravaginal progestagen sponge. The preovulatory follicular dynamics and estradiol secretion, the ovulatory response and progesterone secretion and the number and quality of embryos were studied in 27 ewes treated with two doses of 100 microg of cloprostenol, 10 days apart, and in 29 sheep treated with progestagen sponges for 14 days. Percentage of sheep responding to the synchronization treatments with signs of estrus behaviour was similar between both groups (81.5% versus 72.4%, respectively). The use of progestagen resulted in a higher diameter of the largest follicle (6.6+/-0.2 versus 5.9+/-0.2, Psheep (Pprogesterone concentration during the early luteal phase was again higher in sheep treated with cloprostenol (P<0.05). The mean number of retrieved oocytes/embryos was very similar in both treatments (1.2+/-0.2 versus 1.4+/-0.2) and showed similar fertilization rates (70.6% versus 66.7%), but, although differences did not reach statistical significance, final viability rate was higher in cloprostenol than in progestagen treated ewes (58.9% versus 46.1%, P=0.07). Current results give new evidences supporting the negative effects of progestagens on the functionality of ovulatory follicles and support the development of new protocols for assisted reproduction including the use of prostaglandin analogues.

    Full Text Available El objetivo del presente trabajo fue evaluar la respuesta de la aplicación doble o triple de prostaglandina sobre la regresión de CL, ovulación y retorno al celo después de un tratamiento superovulatorio en vacas donantes de embriones. Para este experimento se tomaron 44 vacas asignadas al azar a dos grupos de tratamiento. Al primero (n = 23; CLO 0 & 7 se le aplicó una dosis de 500 μg de cloprostenol los días 0 (denominado día de la colecta y 7; al segundo grupo (n = 21; CLO 0, 1 & 7 se le aplicó una inyección adicional el día 1. Los animales fueron examinados por ultrasonografía los días 0, 4, 7, 10 y 13 para evaluar la regresión de cuerpos lúteos y la ovulación. Se realizó observación del celo dos veces al día (mañana y tarde. El 0,8 y 52,3% de los grupos CLO 0 & 7 y CLO 0, 1 & 7, respectivamente, presentaron signos de celo. No se presentaron diferencias en el día del celo entre los grupos (7,2 ± 0,3, 7,3 ± 0,5 días. Tampoco hubodiferencia para la variable ovulación (8,5 ± 0,9 y 9,8± 0,9 días.     Full Text Available The aim of this study was to verify if the administration of a luteolytic dose of PGF2α at the time of AI is able to anticipate ovulation and, hence, to increase fertility rate. Furthermore, the presence of a “farm effect” was also evaluated. The trial was performed in two commercial farms (A and B located in Caserta on 584 buffaloes (Farm A: n=389; Farm B: n=195, synchronized by the Ovsynch-TAI Program. In each farm buffaloes were divided in two groups: treated group (n=181 and n=95, respectively in farms A and B received an intramuscular injection of 0.524 mg cloprostenol (Estrumate®, Schering-Plough Animal health, S.p.A., Milan, Italy at the time of AI while control group (n=208 and n=100 received an intramuscular injection of physiologic saline (0.9% NaCl. No differences were present with regard to the number of AI/subject, in terms of pregnancy rate and ovulation rate between treated and control groups. It is worth pointing out that the control group in farm A showed a higher (P<0.01 conception rate compared to the control group in farm B (46.15 vs. 30.00. Furthermore, within farm B buffaloes treated by cloprostenol on the day of AI showed higher (P<0.01 pregnancy rate vs. animals that received saline (51.58% vs. 30.00%. These results suggest that if fertility in control group is high, treatment by cloprostenol does not affect pregnancy rate and ovulation rate.

    Natural lambing in sheep in Ethiopia occurs throughout the year in a scattered manner negatively affecting survival and growth rates of the lambs born during the unfavorable season of the year. Thus, controlling the time of mating artificially using exogenous source of hormones is considered as one of the ways to mitigated problems related to haphazard lambing. To this end, an experiment was conducted to evaluate efficacy of prostaglandin-based estrus synchronization protocol in local and crossbred ewes. A total of 160 ewes (80 local and 80 crossbreds) which lambed at least once and aged 3-5 years were used. Lutalyse® (dinoprost tromethamine sterile solution equivalent to 5 mg dinoprost per ml) and its analog, Synchromate® (cloprostenol sodium equivalent to 0.250 mg cloprostenol per ml), were tested at different doses. The treatments used were intramuscular injection of (1) 2.50 ml of Lutalyse® (12.5 mg dinoprost tromethamine), (2) 2 ml of Lutalyse® (10.0 mg dinoprost tromethamine), (3) 1 ml of Synchromate® (0.25 mg of cloprostenol Sodium), and (4) 0.8 ml of Synchromate® (0.20 mg of cloprostenol Sodium). Forty ewes (20 local and 20 crossbreds) were allocated per treatment. Following injection of the respective hormones, rams of known fertility were introduced into the flock for the duration of 96 h at the ratio of one ram to 10 ewes. All estrus synchronization protocols except treatment 4 (0.8 ml of Synchromate®) induced estrus (heat) in majority (55-65%) of local and crossbred ewes within 96 h post-hormone injection. The time interval from hormone administration to onset of estrus was also more or less similar for all treatment groups except for treatment group 4 which showed heat quicker. The highest lambing rate was recorded in local ewes (84.62% (11/13) treated with 2.5 ml of Lutalyse®, whereas the least was obtained in crossbreds (33.33% (3/9) treated with 0.8 ml Synchromate®.     Full Text Available he use of estrogens in artificial insemination protocols for cattle is the least expensive and most efficient method currently available. However, the trend to prohibit the use of estrogens for this purpose has made it necessary to find alternatives that replace estrogens without compromising the reproductive performance of the animals. The objective of this study was to evaluate conception rates in Bos indicus beef heifers treated with a progesterone device (P4 combined with GnRH or an estradiol ester. On day 0, pubertal Nellore heifers (n = 100 received an intravaginal device containing 1 g P4 and were randomly divided into two groups. The GnRH group (n = 49 received an intramuscular injection of 100 µg GnRH, while the E2 group (n = 51 received 2 mg estradiol benzoate (EB. The P4 device was removed after 5 (GnRH group or 8 days (E2 group, followed by an injection of 125 µg of the PGF2α, analog cloprostenol. On that occasion, the E2 group received an additional injection of 300 IU eCG. Twenty-four hours later, the GnRH group received a second injection of 125 µg cloprostenol, while the E2 group received 1 mg EB. The heifers were inseminated 72 (GnRH group or 54 hours (E2 group after removal of the P4 device. At the time of insemination, the GnRH group received additionally an injection of 100 µg GnRH. Estrus was monitored during the period of cloprostenol injection until the time of artificial insemination and pregnancy was diagnosed 40 days after insemination by transrectal ultrasonography. The data were analyzed by Fisher’s exact test. The pregnancy rate was 38.8% and 31.4% in the GnRH and E2 groups, respectively (P>0.05. The ovarian condition of the heifers (estrus or anestrus tended to influence (P=0.07 pregnancy rates in the GnRH group, but not in the E2 group.     Negative regulators of white adipose tissue (WAT) expansion are poorly documented in vivo. Prostaglandin F(2α) (PGF(2α)) is a potent antiadipogenic factor in cultured preadipocytes, but evidence for its involvement in physiological context is lacking. We previously reported that Akr1b7, an aldo-keto reductase enriched in adipose stromal vascular fraction but absent from mature adipocytes, has antiadipogenic properties possibly supported by PGF(2α) synthase activity. To test whether lack of Akr1b7 could influence WAT homeostasis in vivo, we generated Akr1b7(-/-) mice in 129/Sv background. Akr1b7(-/-) mice displayed excessive basal adiposity resulting from adipocyte hyperplasia/hypertrophy and exhibited greater sensitivity to diet-induced obesity. Following adipose enlargement and irrespective of the diet, they developed liver steatosis and progressive insulin resistance. Akr1b7 loss was associated with decreased PGF(2α) WAT contents. Cloprostenol (PGF(2α) agonist) administration to Akr1b7(-/-) mice normalized WAT expansion by affecting both de novo adipocyte differentiation and size. Treatment of 3T3-L1 adipocytes and Akr1b7(-/-) mice with cloprostenol suggested that decreased adipocyte size resulted from inhibition of lipogenic gene expression. Hence, Akr1b7 is a major regulator of WAT development through at least two PGF(2α)-dependent mechanisms: inhibition of adipogenesis and lipogenesis. These findings provide molecular rationale to explore the status of aldo-keto reductases in dysregulations of adipose tissue homeostasis.

    Full Text Available The aim of this study was to compare ovarian response and embryo production of superovulated Bos indicus and Bos taurus cows adapted to the environmental conditions from São Paulo State, Brazil. Ninety non-lactating cows from Caracu ( Bos taurus, n=40 and Nelore (Bos indicus, n=50 were treated with an intravaginal device containing progesterone (1.38 mg; CIDRB ®, Pfizer Animal Health, Montreal, Québec, Canada and 2.5 mg, intramuscularly (IM, of estradiol benzoate (Estrogin®, Farmavet, São Paulo, Brazil. Four days later, all animals were treated with multiple IM injections of 400 IU of FSH (Pluset®, Calier, Spain in decreasing doses (75–75; 75–50; 50–25, and 25–25 IU at 12-h intervals over 4 days. On the seventh day, CIDR-B device was removed and cows received, IM, 150 ìg of cloprostenol (Veteglan®, Calier, Spain. Cows were then inseminated 48 and 62 h after cloprostenol treatment and embryos were recovered non-surgically seven days after first insemination. Differences in the number of corpora lutea (CL number, total number of structures (ova/embryos, and number of transferable embryos were analyzed by Student t test. There was no difference (P > 0.05 in the average number of CL, total ova/embryos and transferable embryos of Caracu (11.4 ± 3.3; 8.6 ± 2.6 e 6.0 ± 2.4 and Nelore (12.0 ± 4.1; 9.0 ± 4.3 e 5.1 ± 2.9 cows, respectively. These results suggest that Caracu and Nelore cows superovulated in tropical climate had similar ovarian responses and embryo production.

    The aim of this study was to evaluate the effect of (i) the duration of hormone treatment with progestogen sponges during the seasonal anestrus and (ii) the administration of two doses of prostaglandin at 7 days apart during the breeding season on reproductive parameters of Santa Inês ewes. In experiment 1, 32 ewes received intravaginal MAP sponges for 6 (G6 days), 9 (G9 days), or 12 (G12days) days and 75 μg D-cloprostenol i.m. and 300 IU eCG i.m. 1 day before sponge removal. In experiment 2, 23 ewes received two doses of 0.48-mg sodium cloprostenol i.m. 7 days apart. Ovarian follicular dynamic was assessed through transrectal ultrasonography. Blood samples were collected daily to determine progesterone concentrations. In experiment 1, estrus and ovulation rates did not differ (P > 0.05) among protocols and between cyclic and acyclic ewes at the beginning of the experiment. The G9 days treatment showed a lower dispersion of ovulations in relation to onset of estrus when compared to G6 days and G12 days. In experiment 2, all ewes exhibit estrus and ovulated after the second dose of prostaglandin, although ewes that were in diestrus at D0 showed subluteal concentrations of progesterone during the follicle development stage of the treatment. In conclusion, the use of progestogen device during 9 days promotes lower dispersion of ovulation when compared to its use for 6 or 12 days, and the protocol of two doses of prostaglandin 7 days apart synchronizes estrus efficiently but results in follicular development under low progesterone concentrations.

    A screening among bacterial strains isolated from water-brine interface of the deep hypersaline anoxic basins (DHABs) of the Eastern Mediterranean was carried out for the biocatalytical resolution of racemic propyl ester of anti-2-oxotricyclo[2.2.1.0]heptan-7-carboxylic acid (R,S)-1, a key intermediate for the synthesis of d-cloprostenol. Bacillus horneckiae 15A gave highly stereoselective reduction of (R,S)-1, whereas Halomonas aquamarina 9B enantioselectively hydrolysed (R,S)-1; in both cases, enantiomerically pure unreacted (R)-1 could be easily recovered and purified at molar conversion below 57–58 %, showing the potential of DHAB extremophile microbiome and marine-derived enzymes in stereoselective biocatalysis.

Bacillus horneckiae 15A gave highly stereoselective reduction of (R,S)-1, whereas Halomonas aquamarina 9B enantioselectively hydrolysed (R,S)-1; in both cases, enantiomerically pure unreacted (R)-1 could be easily recovered and purified at molar conversion below 57–58 %, showing the potential of DHAB extremophile microbiome and marine-derived enzymes in stereoselective biocatalysis.

    Full Text Available The substitution of equine chorionic gonadotropin (eCG by follicle-stimulating hormone (FSH in protocols for synchronization of ovulation in Santa Inês ewes was assessed. Ten females were submitted to the insertion of intravaginal sponges containing 60 mg medroxyprogesterone acetate for 10 days; after this period sponges were withdrawn and the animals were randomly divided into two groups. Group 1 (n = 5: intramuscular injection of 0.5 mL d-cloprostenol and 300 UI eCG; Group 2 (n = 5: intramuscular injection of 0.5 mL d-cloprostenol and 20 mg FSH. Interval between sponge withdrawal and estrus beginning was 27.7 h and 35.9 h for eCG and FSH, respectively. Interval between sponge withdrawal and the end of estrus was 55.8 h for eCG treatment and 55.6 h for FSH treatment. Estrus length was 29.3 h and 19.6 h, for eCG and FSH treatments, respectively. The biggest follicle and the second in size measured 0.74 cm and 0.54 cm for eCG treatment, whereas for the FSH treatment they measured 0.73 and 0.50 cm. The interval between the beginning of estrus and ovulation was similar within all groups: 21.0 h for eCG treated ewes and 25.2 h for the ones treated with FSH. Ewes treated with eCG presented an interval of 47.5 h between sponge withdrawal and ovulation, while the ones treated with FSH presented a 61.1 h interval. Ovulation occurred 8.3 h before the end of estrus in the eCG group. On the other hand, ewes treated with FSH ovulated 5.5 h after the end of estrus. Estrus and ovulation were efficiently synchronized in Santa Inês ewes by using long-term progestogen protocol associated to the administration of 20 mg FSH, along with Prostaglandin F2α (PGF2α at the moment of sponge withdrawal, thus substituting the use of eCG.

    Full Text Available This study evaluated the pregnancy rate in Nelore cows (Bos indicus that were subjected to fixed-time artificial insemination (FTAI using different protocols consisting of injectable progesterone (P4 or an intravaginal device (impregnated with P4. Multiparous cows 72-84 months in age, 30-45 days postpartum, were selected on the basis of the absence of a corpus luteum (CL and follicles < 8 mm after transrectal palpation and ultrasound examinations. On a random day of the estrus cycle (D0, the selected animals (n = 135 were randomly assigned to one of three experimental groups (n = 45 each. Group I (injectable P4/FTAI 36 hours received 250 mg of injectable P4 and 2 mg EB on D0; on D7, they received 500 µg of cloprostenol; on D8, 300 IU of eCG and 1 mg of EB were administered; and finally, FTAI was performed 36 hours after the application of EB. Group II (injectable P4/FTAI 48 hours received the same protocol as Group I, except that the FTAI was performed 48 hours after ovulation induction. The animals of Group III (Control/CIDR received a conventional protocol for FTAI using an intravaginal device (D0: P4 and 2 mg EB; D8: device removal, 500 µg cloprostenol, 300 IU eCG, 1 mg EB; and FTAI performed 48 hours after removal of the device. The results showed that cows synchronized with the conventional protocol for FTAI (Control/CIDR had a higher pregnancy rate (60 %, 27/45 than those synchronized with an injectable P4/FTAI 36 hours (33.33 %; 15/45, P = 0.010. However, the group receiving injectable P4 group/FTAI 48 hours had a similar pregnancy rate (48.9 %; 22/45; P = 0.290 when compared to both the group receiving the conventional protocol and that receiving injectable P4/FTAI 36 hours (P = 0.134. Although the injectable P4 may affect pregnancy rate with the FTAI performed in 36 hours, we found similar pregnancy rates from cows inseminated 48 hours after induction ovulation, considering injectable or intravaginal P4.     Lactating dairy cows (n = 667) at random stages of the oestrous cycle were assigned to either ovsynch (O, n = 228), heatsynch (H, n = 252) or control (C, n = 187) groups. Cows in O and H groups received 100 microg of GnRH agonist, i.m. (day 0) starting at 44 +/- 3 days in milk (DIM), and 500 microg of cloprostenol, i.m. (day 7). In O group, cows received 100 microg of GnRH (day 9) and were artificially inseminated without oestrus detection 16-20 h later. In H group, cows received 1 mg oestradiol benzoate (EB) i.m., 24 h after the cloprostenol injection and were artificially inseminated without oestrus detection 48-52 h after the EB injection. Cows in C group were inseminated at natural oestrus. On the day of artificial insemination (AI), cows in all groups were assigned to subgroups as follows: human Chorionic Gonadotrophin (O-hCG) (n = 112), O-saline (n = 116), H-hCG (n = 123), H-saline (n = 129), C-hCG (n = 94) and C-saline (n = 93) subgroups. Cows in hCG and saline subgroups received 3000 IU hCG i.m. and or 10 ml saline at day 5 post-AI (day 15), respectively. Pregnancy status was assessed by palpation per rectum at days 40 to 45 after AI. The logistic regression model using just main effects of season (summer and winter), parity (primiparous and pluriparous), method(1) (O, H and C) and method(2) (hCG and saline) showed that all factors, except method(1), were significant. Significant effects of season (p days after AI significantly improved pregnancy rates in those cows that were treated with the H protocol compared with saline treatments (41.5% vs 24.8%; p < 0.01). O and H were more effective in primiparous than in pluriparous cows (46.1% vs 29.9%; p < 0.1 and 43.6% vs 24.6%; p < 0.01). First service pregnancy rates were higher in primiparous hCG-treated than in pluriparous hCG-treated cows (57.9% vs 32.3%; p < 0.01). The pregnancy rate was higher for the hCG-treated cows compared with saline-treated cows during warm period (37.9% vs 23.6%; p < 0.001).

    Full Text Available The work was conducted in Acauã site, in Caucaia - Ceará State, in a "semi-árida" coast region. The objective was to evaluate thermal stress occurrence and his possible impact on ovulation induction treatment efficiency, onset of estrous behavior of dairy Goats from different breeds. Twenty two adult goats were utilized: 10 Saanen, 4 American Alpines and 8 ½ Murciana x undefined breed, raised in installations with high floor, side windows and sunny areas. The estrous induction treatment was conducted using vaginal sponges with MAP, intramuscularly Cloprostenol and eCG. In the environment were registered air temperature and humidity, Radiant Heat Load and Black-Globe Humidity Index. Rectal temperature was similar between breeds and partum order. Respiratory rate was higher in Alpine goats. The induction treatment and estrous behavior interval was similar in ½ Murciana x undefined breed goats and Saanen goats and higher in Alpines and multiparous goats. The fertility rate at 1st AI was lower in ½ Murciana x undefined breed goats and multiparous goats.

    Although various progestagens are often used to induce and synchronize estrus and ovulation in ruminants, concerns regarding residues are the impetus to develop alternative approaches, including reduced doses of progestagens. Therefore, the objective was to determine whether ovarian function was affected by halving the dose of fluorogestone acetate in intravaginal sponges for synchronizing ovulation in sheep during the physiologic breeding season. Twenty Manchega ewes, 4-6-year-old, were randomly allocated to receive an intravaginal sponge containing either 20mg (P20, n=10) or 40 mg of fluorogestone acetate (P40, n=10). Cloprostenol (125 microg) was given at sponge insertion, and all sponges were removed after 6d. Ovarian follicular dynamics (monitored by daily ultrasonography) and other aspects of ovarian function did not differ significantly between the two groups. Ovulatory follicles (OF) grew at a similar growth rate (r=0.62; Pprogesterone concentrations (3.8+/-0.35 ng/mL for P20 and 3.9+/-0.38 ng/mL for P40) were similar. In conclusion, reducing the dose of fluorogestone acetate from 40 to 20mg did not affect significantly ovarian follicular dynamics or other aspects of ovarian function.

  2. Effect of estradiol and cloprostenol combination therapy on expulsion of mummified fetus and subsequent fertility in four crossbred cows

    Full Text Available Four crossbred cows with mummified fetus were utilized for the study. The cows weresubjected togynecologicalexamination and based on the findings the cases were diagnosed asmummified fetus. The cows were treated with 2 mg estradiol valerate and 500 μg cloprostenoland were examined every 12 hr after 24 hr of the treatment for cervical dilatation and othersigns related to fetal expulsion. The time duration between treatment and starting of cervicaldilatation ranged from 48 to 58h (53.00 ± 2.08 hr. Complete dilatation of cervix was observedafter 70.00 ± 2.94 hr post treatment (Range = 64-76 hr. The mean fetalcrown-rump length(CRL was 31.5 cm, which ranged from 27.5 to 38 cm. The number of cotyledons in pregnanthorn also showed wide variation (Range 24-38 numbers with mean ± SE of 30.3 ± 3.07numbers. In the placenta of three animals irregular shaped large adventitious cotyledons wereobserved in the inter-cotyledonary areas. Out of the four animals treated, three animals wereconceived within three estrous cycles and one animal had cystic ovary in the next cycle and wasnot conceived even after four cycles. It was concluded that the estradiolandprostaglandin F2α(PGF2αcombinationtherapy was effective for expulsion of mummified fetus in crossbred cowswithout affecting much on future fertility.

    Full Text Available Avaliaram-se os efeitos de dois protocolos de punção folicular na quantidade/qualidade dos oócitos e na produção in vitro de embriões, em vacas da raça Gir, não-lactantes. O ciclo estral foi sincronizado com cloprostenol e ao longo do experimento os animais receberam implantes auriculares de norgestomet, renovados a cada 14 dias. Os animais foram submetidos aos protocolos I (sem estimulação hormonal, com punção folicular duas vezes/semana e II (pré-tratamento com 250 UI de FSH em doses decrescentes, três dias antes da punção folicular. Os oócitos recuperados foram levados ao laboratório em meio TALP-Hepes e submetidos à maturação. Na fecundação in vitro utilizou-se sêmen de touro Gir, previamente capacitado. Após 22 horas de fecundação, os prováveis zigotos foram co-cultivados com células da granulosa em CR2aa acrescido de 10% de soro fetal bovino. A taxa de clivagem foi avaliada 72 horas pós-fecundação e a de blastocisto 192 horas pós-fecundação. O número total de folículos foi maior (P0,05.     Full Text Available The objective of this study was to evaluate the influence of the diameter of the largest follicle at the time of TAI (DFOL on conception rate of Nellore cows submitted to TAI protocol. Therefore, 348 females were synchronized on day 0 (D0 with an intravaginal progesterone device (P4 associated with 2.0 mg of estradiol benzoate intramuscularly (im. On day 8 (D8, after P4 removal, 500 μg Sodium cloprostenol (im, 0.6mg of estradiol cypionate (im and 300 UI equine chorionic gonadotropin (eCG (im were administered. On day 10 (D10, immediately before each TAI, DFOL was measured in all animals by transrectal ultrasonography. Pregnancy diagnosis was performed 30 days after TAI by transrectal ultrasonography. Data were analyzed by SPSS (version 19 and DFOL means were compared by Student's t-test. The overall conception rate was 57.47% and DFOL overall average was 12.43±2.84mm. There were differences (P=0.0001 in the comparison of DFOL of pregnant (13.33±2.40mm and nonpregnant (11.27±2.75mm cows. The results showed a positive effect of DFOL on fertility of cows submitted to synchronization protocol.

    The aim of this study was to evaluate the use of a prostaglandin F2α analogue (PGF2α and oestradiol benzo-ate (ODB in estrus synchronization programs. A hundred and eighty five non-lactating females divided in 4 groups were used. G1 (44 and G2 (46 received two applications of PGF2α (D0 and D11 and G3 (26 and G4 (37 received one application after 5 days of estrous detection. G1 and G3 also received a 1 mg ODB application 48 hours after the PGF2α. The pregnancy rate at AI was 42,2% for G1, 47,8% for G2, 32,1% for G3 e 57,6% for G4 (p>0,05 and of 38,4% and 51,4% for animals submitted to FTAI and estrous detection, respectively (p>0,05. This results show that pregnancy rates of females fixed time inseminated didn’t differ of that inseminated after estrus detection.

    O objetivo deste estudo foi avaliar o emprego de um análogo da prostaglandina F2α (PGF2α em associação ao benzoato de estradiol (BE em programas de sincronização de cios. Utilizaram-se 185 fêmeas não-lactantes divididas em quatro grupos. O G1 (44 e o G2 (46 receberam duas aplicações de PGF2α (D0 e D11 e os animais do G3 (26 e G4 (37 uma aplicação de PGF2α, após cinco dias de detecção de cio. O G1 e o G3 receberam 1mg de BE 48 horas após a PGF2α. A taxa de prenhez na IA foi de 42,2% no G1, 47,8% no G2, 32,1% no G3 e 57,6% no G4 (p>0,05 e de 38,4% e 51,4% para animais submetidos à IATF e à detecção de cio, respectivamente (p>0,05. Notou-se que a taxa de prenhez nas fêmeas inseminadas em tempo-fixo não diferiu daquelas que foram inseminadas após a detecção de cio.

    Full Text Available The objective of this study was to evaluate the efficiency of progestagen intravaginal devices (IVDs in preventing parturition in sows by determining the effect of delaying parturition on the alive/total born piglets ratio. Evaluations of IVDs containing 0.5, 1.0 or 1.5g progesterone (P4 showed they were not effective in delaying parturition at any dosage tested. In a second experiment, seventy-five sows at day 112 of pregnancy were equally distributed (n=15 per group in the following treatments: prostaglandin (PGF2α; 250µg sodium cloprostenol; control group or PGF2α and simultaneous insertion of an IVD containing medroxyprogesterone acetate (MPA for 48h. Control sows initiated labor 27.7±1.6h after PGF2α injection. The mean time (±SEM between PGF2α administration and parturition was 72.1±8.8h, 72.7±3.8h, 82.7±7.1h and 81.8±3.5h for MPA 100, 200, 400 and 800mg, respectively, differing from control group (P0.05.     Full Text Available The objective of this study was to evaluate the effectiveness of reusing a controlled internal drug release (CIDR device for up to three times in the reproductive performance of dairy goats raised in the semi-arid zone of northeastern Brazil. Forty-five goats were allocated into three hormone treatments, as follows: CIDR1x, treated with new CIDR during nine days. Two days prior to device removal, injections of 75 µg d-cloprostenol and 300 IU equine chorionic gonadotropin (eCG were administrated. For the other treatments, the same hormone protocol was used, differing only by the use of the same CIDR for a second time in CIDR2x and for a third time in CIDR3x. The interval from device removal to the onset of estrus (13.3 ± 1.1h vs. 13.8 ± 2.6h vs. 13.3 ± 1.4h, as well as estrus duration (33.6 ± 7.3h vs. 29.6 ± 3.2h vs. 32.8 ± 4.5h, did not differ (p > 0.05 among groups CIDR1x, CIDR2x and CIDR3x, respectively. All synchronized females were found to be in estrus. The overall fertility and prolificacy after artificial insemination were 82.2% and 1.9 kids, respectively, without significant difference (p > 0.05 among treatments.     Full Text Available The objective of this study was to evaluate the effectiveness of reusing a controlled internal drug release (CIDR device for up to three times in the reproductive performance of dairy goats raised in the semi-arid zone of northeastern Brazil. Forty-five goats were allocated into three hormone treatments, as follows: CIDR1x, treated with new CIDR during nine days. Two days prior to device removal, injections of 75 ƒÝg d-cloprostenol and 300 IU equine chorionic gonadotropin (eCG were administrated. For the other treatments, thesame hormone protocol was used, differing only by the use of the same CIDR for a second time in CIDR2x and for a third time in CIDR3x.   10. Prostanoid Receptors Involved in Regulation of the Beating Rate of Neonatal Rat Cardiomyocytes

    Although prostanoids are known to be involved in regulation of the spontaneous beating rate of cultured neonatal rat cardiomyocytes, the various subtypes of prostanoid receptors have not been investigated in detail. In our experiments, prostaglandin (PG)F2α and prostanoid FP receptor agonists (fluprostenol, latanoprost and cloprostenol) produced a decrease in the beating rate. Two prostanoid IP receptor agonists (iloprost and beraprost) induced first a marked drop in the beating rate and then definitive abrogation of beating. In contrast, the prostanoid DP receptor agonists (PGD2 and BW245C) and TP receptor agonists (U-46619) produced increases in the beating rate. Sulprostone (a prostanoid EP1 and EP3 receptor agonist) induced marked increases in the beating rate, which were suppressed by SC-19220 (a selective prostanoid EP1 antagonist). Butaprost (a selective prostanoid EP2 receptor agonist), misoprostol (a prostanoid EP2 and EP3 receptor agonist), 11-deoxy-PGE1 (a prostanoid EP2, EP3 and EP4 receptor agonist) did not alter the beating rate. Our results strongly suggest that prostanoid EP1 receptors are involved in positive regulation of the beating rate. Prostanoid EP1 receptor expression was confirmed by western blotting with a selective antibody. Hence, neonatal rat cardiomyocytes express both prostanoid IP and FP receptors (which negatively regulate the spontaneous beating rate) and prostanoid TP, DP1 and EP1 receptors (which positively regulate the spontaneous beating rate).     Full Text Available A pilot project was initiated to introduce artificial insemination (AI in goats at farmer level with chilled semen. Does (n=18 were synchronized with progesterone impregnated vaginal sponges (60 mg Medroxyprogesterone acetate; MAP for 11 days. At 48 hrs prior to removal of the sponges, intramuscular injection of 400 IU equine chorionic gonadotropin (eCG and cloprostenol (0.075 mg was given. Fixed time vaginal insemination (43-45 hrs after sponge removal was done twice (at 12 hrs interval in 17 does with chilled Beetal buck semen (4°C extended with Tris-citric acid (TCA or skimmed milk (SM based extender (75 x 106 sperm/ml. Pregnancy test was performed at 45 days post insemination through ultrasonography. An overall 94.5% (17/18 of does showed heat signs and 78% of them were detected in heat between 12 - 24 hrs after sponge removal. An overall 29.4% (5/17 pregnancy rate was recorded. Higher pregnancy rate (44.4% was obtained in does inseminated with SM extended semen as compared to 12.5% for TCA extended semen. Results were encouraging in the sense that to the best of our knowledge it was the first report of kidding through AI in heat induced does in Pakistan. Moreover, it indicated the feasibility of using synchronization and fixed time AI during low breeding season to enhance the reproductive efficiency in local goats.

    This study compares the fertility after short- and long-term synchronization using a progesterone intravaginal device in Lacaune dairy sheep outside the breeding season. For the experiment 108 Lacaune sheep were treated with Eazi-breed™ CIDR® G intravaginal devices (Pfizer Animal Health, Zürich) for 12 days (Group L, n = 60) or 6 days (Group K, n = 48) in combination with eCG (Group L) or with eCG and 125 μg Cloprostenol (Group K) at device removal. Thereafter the ewes were kept together with rams for 60 days, ewes in estrus were recorded and the fertility was assessed after lambing. Blood progesterone concentration was measured at device application, withdrawal and 14 days later. Results show that neither treatment nor parity had an influence on estrus rate (Group L 91.7 %, Group K 93.8 %, nulli- and pluriparous animals 96.9 % and 90.8 %, respectively). Group L showed a tendency towards a better first cycle lambing rate and a significantly (P sheep of Group K (71.7 % vs. 60.4 % and 83.3 % vs. 72.9 %). Pluriparous ewes had higher (P synchronized ewe than nulliparous sheep for the first cycle (75.0 % vs. 46.9 % and 1.4 ± 1.0 vs. 0.9 ± 1.1) as well as for the overall service period (92.1 % vs. 46.9 % and 1.7 ± 0.8 vs. 0.9 ± 1.1). Fourteen days after insert withdrawal progesterone concentrations were higher (P progesterone treatment and nulliparous ewes were less suitable for estrus induction outside the breeding season.

    Full Text Available The purpose of this study was to perform the induction and estrous synchronization of Saanen goats in the non-breeding season. The study was conducted from July 2014 - April 2015 on 77 Saanen goats, aged between 1.5 to 5 years. The goats were divided into three experimental groups: in group I (n =37 goats intravaginal sponge (Ovigest containing 60 mg of MPA was left in the vagina for 14 days. Immediately after sponge removal the animals received two injections: prostaglandin (Prosolvin at the dose of 0.75 mg (1 ml/animal and PMSG hormone (Folligon at the dose of 500 IU/animal. In group II (n=20 induction and estrous synchronization was performed using synthetic prostaglandin (Proliz - 1 ml containing 0.2 mg isopropyl ester of cloprostenol and 9 mg benzyl alcohol at the dose of 0.5 ml/animal. The group III (n=20 was considered the control group and estrous synchronization was performed using fertile bucks. In group I the results showed that all goats expressed signs of estrous. The average value of the estrous interval was 33 hours and the average of estrous duration was 20 hours. The number of kids obtained in group I was 94, and prolificacy was P (% = 2.54. In group II and III was not observed occurrence of estrous. Induction and estrous synchronization show economic benefits by shortening the time needed for pregnancy installation, possibility of births grouping, preparation and organization of mating, only if we use a hormone associated protocol and we rigorous respect therapy steps.

    Full Text Available Pregnancy rate, estrous cycle lenght, serum progesterone and luteinizing hormone (LH concentrations were determined in gonadotropin releasing hormone (GnRH; 10.5 μg synthetic gonadotrophin releasing hormone agonist, receptal administered cows on day 12 post-mating (n=9 compared to control cows (n=8. Their oestrous cycles were synchronised by intramuscular administration of prostaglandin F2 alpha (its analog, cloprostenol twice at 11 days interval. Estrous exhibited cows were mated naturally. Blood samples were collected every two days from all animals. Serum progesterone and LH concentrations were measured by ELISA method. GnRH administration significantly increased serum LH concentration which reached peak levels 2-3 h after treatment. However, serum progesterone concentration was not affected. There were no differences in mean progesterone concentrations on days 12 to 24 post-mating between GnRH administrated and control pregnant cows. However, in non pregnant animals, progesterone concentrations on days 16 in the treated group were lower than control group (P<0.01. Pregnancy diagnosis in animals made by B-mode ultrasonography between the 30th and 35th day showed that 77.7% of treated cows were pregnant compared to 50% in control group. Duration of the estrous cycle in the non-pregnant animals was not affected by the treatment (control, 21.3 ± 0.8 days; treated, 22.5 ± 0.5 days. In conclusion, this study supports the use of GnRH on day 12 post-mating as a method for enhancing pregnancy rates in lactating dairy cattle.

    The objective of this study was to determine the effect on pregnancy outcome of either inseminating heifers twice (at 48 and 72 hours after withdrawal of a controlled internal drug release insert (CIDR) containing progesterone) or once (56 hours after CIDR withdrawal) following a seven-day CIDR synchronisation protocol. Dairy heifers (n=267) from five farms, with an age range of 388-736 days, were randomly assigned to one of two treatment groups (group A heifers were inseminated twice; group B heifers were inseminated once). Both groups received a CIDR on day (D) 0 and an intramuscular injection of d-cloprostenol on D6; the CIDR was withdrawn on D7. Measurements of withers height, body condition score and hearth girth (used to estimate weight) were taken on D0. The diameter of the largest follicles and corpora lutea was recorded on both D0 and D6. Data were analysed with the use of multivariable logistic regression modelling. Treatment group and farm were not statistically significantly associated with pregnancy per treatment (P/T). Age and dominant follicle size on D6 were significantly associated with P/T. Heifers with the largest dominant follicle sizes (16-22 mm) were 5.54 times less likely to be pregnant than those heifers with the smallest dominant follicles (8-10 mm) on D6.   16. Factors associated with fertility of nulliparous dairy heifers following a 10-day fixed-time artificial insemination program with sex-sorted and conventional semen.

    To examine factors associated with fertility on dairy farms that used a common fixed-time artificial insemination (FTAI) program in yearling heifers. Records were analysed from 954 yearling heifers on 10 south-west Victorian dairy farms that used a common FTAI program, involving the insertion of a 1.9-g progesterone-releasing device for 10 days; 2 mg oestradiol benzoate at insertion; 500 µg cloprostenol on day 7; and FTAI 48 h after device removal. Weight, age, expression of oestrus, sire, semen type (frozen sex-sorted or frozen conventional) and timing of insemination were examined for their relationship with first-service conception rates. Heifers over 300 kg body weight were 1.18-fold more likely to express oestrus during the FTAI program. For every extra 1 kg, there was a 1.5% increase in the likelihood of expressing oestrus. First-service conception rates were 40.3% and 56.0% for sex-sorted and conventional semen, respectively, and were significantly higher when oestrus was expressed. The difference was greater for sex-sorted semen (3.4-fold) compared with conventional semen (1.5-fold). The interval from device removal to insemination varied between 47 and 51.4 h and had no significant effect on conception rates. However, there was a trend towards a higher conception rate for sex-sorted semen when inseminations were performed >50 h after device removal. Increased fertility was associated with larger heifers and heifers that expressed oestrus, particularly when sexed-sorted semen was used. Variation in the timing of AI with respect to device removal between 47 and 51.4 h did not adversely affect conception rates. © 2016 Australian Veterinary Association.

  17. Applied biotechnology for improving fertility of herds in Argentina

    In an attempt to find methods to shorten the calving to conception interval of dairy cattle in Argentina, three research projects were carried out: (1) use of hormonal treatments in the early post-partum period to increase fertility in non-cycling cows; (2) treatments to increase the fertility of cycling cows; and (3) management systems to improve the efficiency of oestrus detection. In non-cycling cows, fewer services per conception and earlier pregnancies were obtained when a progesterone suppository was placed intravaginally (PRID treatment) 24-31 days post-partum. No statistical reduction in days open was observed, however, because of the great variability in response within groups. GnRH injection given 24 and 25 days post-partum did not improve reproductive indices. In cycling cows, clitoral massage after AI did not enhance fertility, while GnRH 14 days post-service resulted in pregnancy rates of 68.4% compared with 25% in the control group. In an attempt to improve the efficiency of oestrus detection successive periods of either 10 or 14 days of 'heat' observation followed by similar periods of no observation, combined with the systematic use of an oestrus synchronizer (cloprostenol), resulted in the same reproductive performance as in control cows observed continuously but with a saving of 50% in time spent in oestrus detection. In a seasonally bred herd, a reduction of 12 days in interval to conception and subsequent improvement in milk produced in the following season were obtained by synchronizing oestrus at the start of the breeding season with a norgestomet implant followed by a prostaglandin injection one day before implant removal. El día pre sincronización todas las vaquillonas fueron examinadas por ecógrafo transrectal Aquila Vet 7.5 MHz, donde se determinó el diámetro uterino (cm, área ovárica (cm², área de estructuras ováricas (cm². Día cero se colocó dispositivo intravaginal 0.5 mg progesterona, conjunto con una inyección intramuscular de 1 mg de Benzoato Estradiol; día 8 retiro del dispositivo más administración intramuscular de 500 µg de D (+ Cloprostenol; día 9 aplicación intramuscular de 0.75 mg de benzoato estradiol. Inseminando 54-57 h de retirado el dispositivo intravaginal. Se diagnosticó gestación 60 días post IATF. No se registraron diferencias significativas (P>0.05, entre peso corporal, útero, ovario izquierdo, y estructuras ováricas, solo encontrándose diferencias significativas en el ovario derecho. Se concluye que el área del ovario derecho influye positivamente sobre la tasa de preñez, en cuanto al mejor umbral en pronóstico de la preñez para seleccionar una vaquillona mestiza en función del ovario derecho el estándar 4.80 cm², es el apropiado.

  19. Effect of postpartum suppression of ovulation on uterine involution in dairy cows.

    The objective of this study was to investigate the effect of time of first postpartum ovulation after calving on uterine involution in dairy cows with and without uterine puerperal disease. Transvaginal follicular puncture (FP) of follicles >6 mm suppressed ovulation and development of a CL until Day 42 after calving. Fifty-three lactating Holstein Friesian cows (3.4 ± 1.2 years old, parity 2.5 ± 1.0 [median ± mean absolute deviation]) were divided into groups on the basis of the presence (UD+) or absence (UD-) of uterine disease and whether FP was carried out (FP+) or not (FP-). Uterine disease was defined as the occurrence of retained fetal membranes and/or metritis. This resulted in the following groups: UD-FP- (n = 15), UD-FP+ (n = 13), UD+FP- (n = 13), and UD+FP+ (n = 12). A general examination, vaginoscopy, transrectal palpation, and transrectal B-mode sonography of the reproductive organs were conducted on Days 8, 11, 18, and 25 and then every 10 days until Day 65 after calving. After hormonal synchronization of ovulation (cloprostenol between Days 55 and 60 postpartum and GnRH 2 days later), cows were inseminated in the next spontaneous estrus. On average, the cows ovulated on Day 21.0 ± 6.0 (UD-FP-), 50.0 ± 4.0 (UD-FP+), 16.0 ± 3.0 (UD+FP-), and 48.0 ± 2.0 (UD+FP+) postpartum. Calving-to-conception interval and first-service conception rates were not affected by FP (P > 0.05). Healthy cows with FP had smaller (P uterine horn and cervical diameters assessed sonographically than cows without FP. FP reduced the prevalence of purulent vaginal discharge and uterine size assessed transrectally in UD+ cows (P uterine involution in cows with and without uterine disease. Santa Inês and crossbred Santa Inês/Dorper ewes received 60 mg MAP sponges during 6 days plus 300 IU eCG and 30 µg d-cloprostenol 24 h prior to sponge withdrawal (SW. Ewes were assigned to receive 0.9% NaCl solution (Tcontrol; n = 32 or 25 µg GnRH (licerelin, T GnRH; n = 34 24 hours after SW.     Full Text Available Os objetivos neste experimento foram avaliar o efeito do tempo de exposição a um progestágeno (P4 na taxa de prenhez de novilhas púberes e pré-púberes inseminadas em tempo-fixo (IATF e estudar o retorno econômico dos protocolos utilizados, por meio da comparação desses protocolos ao sistema tradicional de acasalamento natural aos 26 meses de idade. Foram utilizadas 114 novilhas da raça Bradford com 16 meses de idade, classificadas em quatro grupos de acordo com o tratamento e a maturidade sexual: GPPLP - pré-puberes, longo tratamento com P4 (n=21; e GPPCP - pré-puberes, curto tratamento com (n=19; GPLP - púberes, longo tratamento com P4 (n=34; e GPCP - púberes, curto tratamento com P4 (n=40. As novilhas do GPPLP e GPLP receberam no dia -8 um pessário intravaginal impregnado com acetato de medroxiprogesterona. No dia 0, o pessário foi removido e outro foi inserido, juntamente com uma injeção de 2,5 mg de benzoato de estradiol (BE. No dia 8, o pessário foi retirado e as novilhas receberam 0,5 mg de BE e 250 µg de cloprostenol sódico. A inseminação artificial em tempo fixo (IATF foi realizada 54 horas depois. As novilhas do GPPCP e GPCP receberam o mesmo tratamento de sincronização dos grupos GPPLP e GPLP, com exceção do AMP do dia -8 ao dia 0. A taxa de prenhez foi maior nas novilhas púberes (60,3% que nas pré-puberes (35,0%. No entanto, não foi detectado efeito do tempo de tratamento na taxa de prenhez nas novilhas púberes e pré-puberes. A resposta econômica na utilização de protocolos com P4 foi maior em novilhas aos 16 meses que no sistema de acasalamento aos 26 meses.     Full Text Available The objective of this study was to compare the number of follicles, oocytes and the recovery rate in sheep submitted to the one-shot protocol with or without ovarian priming with estradiol benzoate (EB. Pluriparous non-lactating sheep (n=33 with an average age of five years (range 4-6 and a body condition score of 3.0±0.3 were divided into three groups. The one-shot group (n=10 was treated with a subcutaneous implant containing 1.5 mg of norgestomet from D0 to D10. The animals in this group were administered 0.04 mg of D-cloprostenol, 200 IU of follicle stimulating hormone (FSH and 300 IU of equine chorionic gonadotropin (eCG on D8. Animals in the EB group (n=11 received the same treatment as one-shot plus the administration of 0.6 mg of EB on D0. In the untreated group (n=12, the animals received no hormone stimulation. The collection of the oocytes was performed by laparotomy 36 h after the administration of gonadotropins (D10. Oocytes were searched and classified based on morphology. An increase was observed (p<0.05 in the number of follicles aspirated in the one-shot vs. the EB and untreated groups (16.3±5.6 vs. 9.5±2.4 and 12.1±4.1, respectively. The average number of oocytes and the recovery rate were higher (p<0.05 in the one-shot and EB groups compared to the untreated group, resulting in 14.2±9.0 and 87.1% (142/163, 11.0±6.2 and 91.4% (122/134 vs. 6.8±3.5 and 71.9% (82/114, respectively.     The objective was to investigate the ovarian response of Brahman heifers to two modified ovulation synchronisation protocols developed to increase the proportion of normal synchronous ovulations. Experiment 1 characterised the growth of the ovulatory follicle in heifers (n=19) treated with an intravaginal progesterone releasing device (IPRD) and oestradiol benzoate (ODB), to determine the optimal time to induce ovulation. Using the findings from Experiment 1, Experiment 2 investigated the effect of reducing the duration of IPRD insertion and increasing the interval from IPRD removal to ODB treatment (modified protocol 1 - OPO-6; n=20), and omitting ODB treatment at the time of IPRD insertion (modified protocol 2 - PO-6; n=20). An IPRD (0.78 g progesterone) was inserted at Day 0 (OPO-8) or Day 2 (OPO-6 and PO-6) and all heifers also received 1 mg ODB i.m. Day 8: IPRD removed + 500 μg cloprostenol i.m. At 24 h (OPO-8) and 36 h (OPO-6 and PO-6) post IPRD removal: 1 mg ODB i.m. Fixed-time AI (FTAI) occurred at 54 h for OPO-8 and 72 h for OPO-6 and PO-6, post IPRD removal. After IPRD treatment all OPO-6 and OPO-8 heifers initiated a new follicular wave whereas 25% of PO-6 heifers failed. Diameter of the dominant follicle was larger at FTAI in the PO-6 (11.34 ± 0.50 mm) compared to the OPO-8 protocol (9.74 ± 0.51 mm; P<0.05), but similar to the OPO-6 protocol (10.52 ± 0.51 mm). Proportion of ovulations occurring 12 h prior and 24 h post FTAI was similar for the PO-6 (80%) and OPO-6 (75%) protocols but numerically lower in the OPO-8 heifers (60%). The apparent improvement in ovarian response in heifers treated with the modified protocols needs to be confirmed in larger field studies. Copyright © 2014 Elsevier B.V. All rights reserved.

  4. Stress effect on conception rate in Nellore cows submmited to fixed time artificial insemination. Preliminary results

    Full Text Available In beef cattle, fixed time artificial insemination (FTAI provides a method to inseminate large numbers of females in a specific time, which result in economical gains due, among others, to a more uniform calf crop. However, FTAI requires frequent manipulation of animals in order to inject hormones and for clinical examination. Consequently, animals seemed stressed in less or higher extent at the time of insemination. This can be a problem because it has been demonstrated that application of an acute stress treatment (electric shock, confinement, restraint and rotation twice a day during the follicular phase of the oestrous cycle prevents the pre-ovulatory LH surge. This study aimed to evaluate if FTAI efficiency of Nellore cows is affected by the degree of stress observed at time of AI. Nellore cows (n=92 were treated (Day 0 with a progesterone intravaginal devise (Primer®, Tecnopec, São Paulo, Brazil containing 1 g of progesterone and injected with estradiol benzoate (2mg EB, Estrogin, AUSA, Brazil. Primer was removed on Day 8 (08:00 AM and administered one injection of cloprostenol (125 mcg, Prolise®, Tecnopec, São Paulo, Brazil. Twenty-four hours later, cows received 2 mg EB and insemination (semen from one sire was done on the afternoon (14:00 to 16:00 PM of day 10.   5. Effects of various procedures for synchronisation of parturition in sows. Assessment of sows with a complication-free birth.

    The effects of various parturition controlling measures in sows on the relevant parameters for parturition were investigated. A total of 1975 study animals were divided into five groups: control group: A (n = 710); trial groups: B (n = 719), C (n = 180), D (n = 175), E (n = 191). The control group (A) included sows that spontaneously farrowed until day 114 of gestation. The remaining sows were administered a PGF2α-analogue (175 µg cloprostenol) on day 114. All sows that farrowed within 24 hours after prostaglandin-medication were summarised in trial group B. Dams that did not farrow until day 115 were either administered oxytocin (20 IU i. m., group C) or carbetocin in two different doses (70 µg, group D or 35 µg, group E). The sows were monitored during the peripartal period over 24 hours. 83.8% of the farrowings were without any complications. The percentage of dystocia varied between the individual groups. Given a complication-free farrowing the expulsion stage began 13.0 hours (group B), 2.4 hours (group C), 1.6 hours (group D) and 1.4 hours (group E) after medication. The expulsion interval of the farrowing of two piglets decreased from 21.1 minutes with spontaneous farrowings (group A) to 18.6 minutes (group E) as a result of the administration of 35 µg carbetocin. The rate of stillborn piglets as well as the incidence of puerperal disorders and the losses of the suckling piglets also gradually, partly even significantly (p ≤ 0.05) decreased from group A to E. The results of this study show that the use of the long-acting oxytocin carbetocin has advantages compared to oxytocin (e.   g. shortened duration of birth, reduction of stillbirths). The tested dosages of carbetocin (35 µg or 70 µg) proved to be equally effective. Administration of the lower dose is recommended, since this reduces use of the active ingredient.

  6. Do high progesterone concentrations decrease pregnancy rates in embryo recipients synchronized with PGF2alpha and eCG?

    The objective of this study was to evaluate the effects of equine chorionic gonadotropin (eCG) treatment on the number of induced accessory corpora lutea (CL), plasma progesterone concentrations and pregnancy rate in cross-bred heifers after transfer of frozen-thawed (1.5M ethylene glycol) embryos. All recipients received 500 microg PGF2alpha (dl-cloprostenol, i.m.) at random stages of the estrous cycle (Day 0) and were observed for estrus for 7 days. On Day 14, heifers detected in estrus between 2 and 7 days after PGF2alpha treatment were randomly allocated to four groups ( n=83 per group) and given 0 (control), 200, 400, or 600 IU of eCG. Two days later (Day 16), these recipients were given PGF2alpha and observed for estrus. Six to eight days after detection of estrus, plasma samples were collected to determine progesterone concentration and ultrasonography was performed to observe ovarian structures. Heifers with multiple CL or a single CL >15 mm in diameter received an embryo by direct transfer. Embryos of excellent and good quality were thawed and transferred to the recipients by the same veterinarian. Pregnancy was diagnosed by ultrasonography and confirmed by transrectal palpation 21 and 83 days after embryo transfer (ET), respectively. Plasma progesterone concentrations on the day of transfer (Day 7 of the estrous cycle) were 3.9+/-0.7, 4.2+/-0.4,6.0+/-0.4 and 7.8+/-0.6 ng/ml for groups Control, 200, 400, and 600, respectively (Control versus treated groups P=0.009; 200 versus 400 and 600 groups P=0.0001; and 400 versus 600 P=0.012 ). Conception rates 83 days after ET were 41.9, 50.0, 25.0, and 20.9% for groups Control, 200, 400, and 600, respectively (200 versus 400 and 600 groups P=0.0036 ). In conclusion, an increase in progesterone concentration, induced by eCG treatment, did not improve pregnancy rates in ET recipients. Conversely, there was a decline in conception rates in the animals with the highest plasma progesterone concentrations.

  7. Resynchronising returns to service in anoestrous dairy cows in the South Island of New Zealand.

    To investigate the effect of targeted resynchronisation of cows treated for non-observed oestrus before the planned start of mating (PSM), that were not detected in oestrus or pregnant 23 days after treatment (phantom cows), on the proportion pregnant at 42 days after PSM and the end of mating. Farm staff from eight herds in two regions of the South Island of New Zealand identified 1,819 cows not showing oestrus by 10 days before PSM. These cows were treated with intravaginal progesterone for 7 days, and I/M gonadorelin 10 days and 1 day before PSM. Three days before PSM they were injected with cloprostenol and equine chorionic gonadotrophin, with fixed time artificial insemination (FTAI) at PSM. By 23 days after PSM, 1,218 cows had not returned to oestrus. Of these, 161 cows confirmed not pregnant by transrectal ultrasonography were randomly assigned to no treatment (control group; n=74) or were resynchronised 25 days after PSM using the same treatment programme as above, with FTAI 35 days after PSM (n=87). All cows that returned to oestrus were artificially inseminated until 42 days after PSM, when natural mating was used. All cows were examined using transrectal ultrasonography 80 to 90 days after PSM to confirm conception dates. Of the 1,819 anoestrous cows treated before PSM, 526 (29 (95% CI=23.1-34.0)%) had not been observed in oestrus by 23 days after PSM and had not conceived, so were diagnosed as phantoms cows. For resynchronised cows, 42/87 (48 (95% CI=37.8-58.8)%) were pregnant by 42 days after PSM compared to 21/74 (28 (95% CI=18.1-38.7)%) control cows (p=0.009). At the end of mating 58/87 (67 (95% CI=56.6-76.7)%) cows in the resynchronised group were pregnant and 46/74 (62 (95% CI=50.9-73.2)%) in the control group (p=0.554). The hazard of conception from 21 to 42 days after PSM was 1.9 (95% CI=1.07-3.12) times greater for resynchronised than control cows (p=0.026).     Full Text Available This study aimed to evaluate the effects of gonadorelin (GnRH, used both at the outset of a timed artificial insemination (TAI protocol to synchronize follicular wave recruitment and at the end to synchronize ovulation, on the conception rate (CR in Nellore cows. The experiment was conducted on two beef cattle farms in the Vale do Araguaia, MT. The body condition score (BCS evaluation and ultrasound examination to determine ovarian status [follicle diameter < 10 mm, ? 10 mm or presence of corpus luteum (CL] were performed at the beginning (Day 0 of the TAI protocol. Multiparous cows (n = 494 were subjected to the following protocol: Day 0 – insertion of intravaginal progesterone device (P4 DIB®, MSD previously used for 8 or 16 days and IM administration of 2.0 mg of estradiol benzoate (EB, Gonadiol®, MSD; Day 8 – P4 device withdrawal, IM injection of 300 IU of equine chorionic gonadotropin (eCG, Folligon®, MSD, 1.0 mg of estradiol cypionate (ECP®, Zoetis and 0.265 mg of Sodium cloprostenol (PGF2?, Ciosin®, MSD; Day 10 – TAI, performed by a single inseminator. Animals were randomly assigned to one of four groups: control (n = 126, GnRH D0 (n = 123, GnRH D10 (n = 123 and GnRH D0 + D10 (n = 122. Animals treated with GnRH received 50 mcg of gonadorelin (GnRH, Cystorelin®, Merial. Pregnancy was diagnosed by ultrasound 39 ± 10 days after TAI. Data were analyzed using the GLIMMIX procedure of SAS program (P < 0.05. The overall CR was 42.71%. Effect of treatment was not detected (P = 0.2482 on CR, which was 36.89% ± 0.19 (47/126 for the control group, 46.08% ± 0.19 (56/123 for GnRH D0, 48.30% ± 0.18 (61/123 for GnRH D10 and 38.48% ± 0.19 (47/122 for GnRH D0 + D10.     The objectives of this study were to assess the association of a 4-point scale of vaginal discharge score (VDS) with time to pregnancy to define criteria for a practical case of purulent vaginal discharge (PVD) in dairy cows, to test the risk factors for PVD, and, finally, the effect of a dose of PGF 2α on cure and reproductive performance. In experiment 1, grazing Holstein cows (n = 2,414) had their vaginal discharge scored at ∼32 d in milk (DIM) on a 4-point scale, the effect of VDS on the hazard of pregnancy by 300 DIM was then assessed to derive a case definition of PVD. Risk factors for PVD and self-cure were also assessed. In experiment 2, grazing Holstein cows (n = 6,326) from 5 herds were checked for PVD at ∼30 DIM. Cows with PVD were assigned to receive one dose of 500 μg of PGF 2α analog (Cloprostenol; Ciclase, Syntex SA, Buenos Aires, Argentina) per cow (odd ear tag number) or to remain untreated (even tag number). Cure was declared if cows presented clear normal vaginal discharge (VDS-0) at visit 2 (∼62 DIM). Data were analyzed with Cox's regression and mixed logistic models. In experiment 1, cows with VDS ≥1 had lower hazard of pregnancy and longer calving to pregnancy interval than cows with VDS-0. This finding was not affected by the time at which the diagnosis was performed. Therefore, a cow ≥21 DIM and having VDS ≥1 was used to define a case of PVD. The odds of PVD were greater in primiparous cows compared with multiparous, in cows with abnormal calving compared with those with normal calving, and in those losing BCS peripartum. In experiment 2, PGF 2α treatment tended to slightly increase the hazard of pregnancy (adjusted hazard ratio = 1.13). Conversely, PGF 2α had no effect on the odds of cure of PVD [adjusted odds ratio (AOR) = 1.19], pregnancy at first service (AOR = 1.03), or pregnancy by 100 DIM (AOR = 0.89) or 200 DIM (AOR = 1.27).     Full Text Available The objective of this experiment was to determine the reproductive performance of suckled beef cows in programs of artificial insemination (AI submitted to definitive early or temporary weaning (EW or TW and estrous synchronization protocol to fixed-time artificial insemination (FTAI. Two-hundred and five Angus x Nelore cows body with condition score 2.6±0.4 and post-partum period between 54 and 122 days were used in the trial. After EW, one-hundred, fifty three cows were separated three groups according to the AI or FTAI protocol. A group of cows was artificial inseminated according estrous detection after 10 days of definitive weaning during a period of 30 days (EW-AI, n= 53. Estrous synchronization programs to FTAI were carried out in two groups in different moments after EW. In the group EW-FTAI (n= 50, cows were treated at the moment calves were removed while in EW-FTAI 10 (n= 50 the hormonal treatment began 10 days later.  The hormonal treatment consisted of an intravaginal implant containing 1,9g of Progesterone, for 8 days, and an injection of  Estradiol benzoate (EB, 2mg, im. When the dispositives were removed, 75mcg of Cloprostenol were injected, im, and after 24 hours, 1mg of EB, im. Cows were fixed-time artificial inseminated 52 to 56 hours after implants removal. In TW-FTAI group (n= 52, cows were treated with the same hormonal therapy, but a temporary weaning of 60 hours was done after the implant removal and the FTAI moment. The breeding season was 60 days in EW-FTAI and TW-FTAI groups, 50 days in EW-FTAI10 group and 30 days in EW-AI group. In a sample of 20% of cows ovaries were scanned by ultrasound and it was determined that 55% of the cows were in anestrous. Follicular diameters were determined at beginning of hormonal treatment and at FTAI moment in cows submitted to estrous synchronization (EW-FTAI= 10, EW-FTAI10= 10 and TW-FTAI= 10 by ultrasound.     The combination of ram effect with two injections of PGF 2α 10-days apart and the same protocol plus an additional injection of GnRH prior to the first injection of PGF 2α were examined in Karakul ewes during breeding and non-breeding seasons, respectively. Plasma progesterone (P 4 ) concentrations (to detect the presence of active corpus luteum), twin lambing, litter size and synchronization of lambing were evaluated. In each study 70 ewes (2-4 years old) were divided to a treatment (n = 40) and a control (n = 30) group. During the breeding season, on days -10 and 0 before ram release, the treatment group was injected intramuscularly with PGF 2α (D-Cloprostenol; 0.15 mg). During the non-breeding season, on day -15 before ram release the treatment group was injected with GnRH (buserelin; 4.2 μg) intramuscularly followed by two injections of PGF 2α on days -10 and 0. In both studies, the rams were released into the ewe flock after the second prostaglandin injection (day 0). Blood samples of ewes were collected on days -10, 0, 20 and 70 of the study in breeding season and on days -15, -10, 0, 20 and 70 during non-breeding season. The treatment group had higher P 4 concentrations compared to the control ewes on day 0 in the breeding season (5.80 ± 0.61 vs. 5.0 ± 0.93 ng/mL) and day -10 in the non-breeding season (3.50 ± 0.33 vs. 2.70 ± 0.35 ng/mL) though the differences were not significant (P > 0.05). Based on plasma P 4 concentrations (>1 ng/mL) on day 70, in the breeding season all control ewes (100%) and 91.9% of the treatment ewes were detected to have active corpus luteum (P = 0.09). An almost inverse result (90% vs. 97.5%; P = 0.2) was detected in the non-breeding season. The lambing rate was higher (P = 0.03) in the treatment group compared to the control ewes during the non-breeding season (90% vs. 70%), but tended to be lower (P = 0.07) in the breeding season (73% vs. 90%).
