    Genetic (DNA) inactivation of fish sperm with ultraviolet irradiation is generally accompanied by a paradoxical effect on survival rates (Hertwig effect). In the present study, sperm samples from ten males bullseye puffer fish (Sphoeroides annulatus) were diluted 1:50 using Cortland's extender solution and used to test the effect of nine ultraviolet doses (0.2-1.0 J cm{sup -}2 ) on motility time in seconds, motility index, and embryo survival rate after fertilizing eggs from five bullseye puffer females. Motility time of sperm irradiated with 0.2-0.9 J cm{sup -}2 were not statistically different from the controls, but sperm irradiated with a dosage of 1.0 J cm{sup -}2 dosage had significant lower motility time. Motility indices (MI) allowed for the statistical differentiation of four groups in relation to their response to different radiation doses: the first had high MI, and included the controls and 0.2-0.3 J cm{sup -}2 treatments; the second had lower MI and included the 0.4-0.7 J cm{sup -}2 treatments; the third showed recovery of MI and included the 0.8-0.9 J cm{sup -}2 treatments; and the fourth showed the lowest MI with the 1.0 J cm{sup -}2 treatment. Embryo survival was highest for the controls and 0.2 J cm{sup -}2 treatment, decreasing in the 0.3-0.4 J cm{sup -}2 treatments, increasing again in the 0.5-0.8 J cm{sup -}2 treatments, until reaching lowest survival in the 0.9-1.0 J cm{sup -}2 treatments. These results indicate that the best ultraviolet dosage to achieve genetic inactivation of sperm of this species is close to 0.7 J cm{sup -}2, a dosage in which fish fry showed typical haploid syndrome characteristics.     Individual variation in the endocrine stress response has been linked to survival and performance in a variety of species. Here, we evaluate the relationship between the endocrine stress response and anti-predator behaviors in wild checkered puffers (Sphoeroides testudineus) captured at Eleuthera Island, Bahamas. The checkered puffer has a unique and easily measurable predator avoidance strategy, which is to inflate or 'puff' to deter potential predators. In this study, we measured baseline and stress-induced circulating glucocorticoid levels, as well as bite force, a performance measure that is relevant to both feeding and predator defence, and 'puff' performance. We found that puff performance and bite force were consistent within individuals, but generally decreased following a standardized stressor. Larger puffers were able to generate a higher bite force, and larger puffers were able to maintain a more robust puff performance following a standardized stressor relative to smaller puffers. In terms of the relationship between the glucocorticoid stress response and performance metrics, we found no relationship between post-stress glucocorticoid levels and either puff performance or bite force. However, we did find that baseline glucocorticoid levels predicted the ability of a puffer to maintain a robust puff response following a repeated stressor, and this relationship was more pronounced in larger individuals. Our work provides a novel example of how baseline glucocorticoids can predict a fitness-related anti-predator behavior. Copyright © 2015 Elsevier Inc. All rights reserved.

    Full Text Available In Brazil, where puffer fish are considered poisonous, there are few documented cases on human consumption and consequent poisoning. In this study, toxicity of two puffer fish species from the Brazilian coast was examined. Specimens of Sphoeroides spengleri and Lagocephalus laevigatus were caught in São Sebastião Channel (North coast of São Paulo State, Brazil between January 1996 and May 1997. Acidic ethanol extracts from muscle and skin plus viscera were tested for mice acute toxicity using the standard method of Kawabata. Polar extracts of S. spengleri showed high toxicity up to 946 MU/g. Extracts from L. laevigatus showed very low levels of toxicity, never exceeding 1.7 MU/g. All extracts from both species blocked amielinic nerve fiber evoked impulses of crustacean legs; this effect reverted on washing similar to the standard tetrodotoxin TTX. The aqueous extract solutions were partially purified using an ionic exchange column (Amberlit GC-50 followed by treatment with activated charcoal (Norit-A. The presence of TTX and their analogs in the semi-purified extracts were confirmed by HPLC and mass spectrometry (MALDI-TOF.

    Full Text Available Brasacanthus sphoeroides gen. n.. sp. n. is described from the marine fish, Sphoeroides greeleyi (Steindachner, taken in Paranaguá Bay, Paraná State, Brazil. The new genus differs from Acanthocephalus Koelreuter, 1771, the nearest genus in the family Echinorhynchidae, by having very flat and variable lemnisci in both sexes and a uterine egg reservoir in the female. The species is characterized by its spherical body form and in having parallel or diagonal testes.

  9. Puffer fish poisoning: summary of case reports from Thailand

    Full Text Available Puffer fish poisoning is a common seafood poisoning. The problem is due to tetradotoxin in puffy fish meat. It can be seen in many countries with seacoasts. The problem is resulted from tetradotoxin (TTX in puffer fish meat. This toxin is an important natural toxin. Here, the authors report summary of case reports from Thailand. The authors use standard search engines (PubMed and Thai Index Medicus for searching on the reports on puffer fish poisoning from Thailand. According to this work, there are at least 3 reports on 55 cases of puffer fish poisoning. All cases visit to the physician within 30 min. Focusing on severity, stage 1, 2, 3 and 4 can be seen in 16%, 8%, 4% and 72%, respectively. In the present report, the summary of the cases presenting to the physician at hospital is shown. Of interest, most patients have severe intoxication and the respirator failure is an important problem to be managed. It is clearly shown in the present report that if good respiratory support is done, the full recovery without problem can be derived. It is no doubt that there is no death case in the present series since the present report focuses on the cases that are successfully delivered to the hospital for management.

  10. K-space polarimetry of bullseye plasmon antennas.

    Surface plasmon resonators can drastically redistribute incident light over different output wave vectors and polarizations. This can lead for instance to sub-diffraction sized nanoapertures in metal films that beam and to nanoparticle antennas that enable efficient conversion of photons between spatial modes, or helicity channels. We present a polarimetric Fourier microscope as a new experimental tool to completely characterize the angle-dependent polarization-resolved scattering of single nanostructures. Polarimetry allows determining the full Stokes parameters from just six Fourier images. The degree of polarization and the polarization ellipse are measured for each scattering direction collected by a high NA objective. We showcase the method on plasmonic bullseye antennas in a metal film, which are known to beam light efficiently. We find rich results for the polarization state of the beamed light, including complete conversion of input polarization from linear to circular and from one helicity to another. In addition to uncovering new physics for plasmonic groove antennas, the described technique projects to have a large impact in nanophotonics, in particular towards the investigation of a broad range of phenomena ranging from photon spin Hall effects, polarization to orbital angular momentum transfer and design of plasmon antennas.

    Full Text Available The Research Institute for Freshwater Ornamental Fish Culture has been conducting domestication research since 2004 on adult freshwater puffer fish or buntal broodstock in which the fish were reared in aquaria and fed with small feed fish teri (anchovy and earthworm (Lumbricus sp.. The domestication of freshwater puffer fish or buntal has resulted in successful environmental adaptation and reproductive ability of the fish and further rearing of its fry. The adaptive ability was indicated by its high survival rate, good growth and development, and the reproductive success in spawning, egg production, hatching as well as growing of the larvae to fry stages. The mature gonad stage of the fish was reached at an average body weight of 150 g with a reproductive period of 5 months. The spawning occurred naturally and the eggs were gradually released at interval periods of 14—18 days with egg production rates of 1,000—1,900 eggs per spawning. The highest egg fertilization rate was 96%, with the highest egg hatching rate of 78.6%. Hatching process took place on the 6th to 13th days after spawning process. The surviving larvae began feeding filtered natural foods Moina sp. on the 3rd day up to the 6th day, whereas unfiltered Moina sp. was given from the 7th to 14th days. The fish were fed the live feed Culex sp. from the 15th to 30th days. In the following period the fish was fed earthworm and small feed fish teri (anchovy.

    The Integrated Data Collection Analysis (IDCA) program is conducting a proficiency study for Small- Scale Safety and Thermal (SSST) testing of homemade explosives (HMEs). Described here are the results for impact, friction, electrostatic discharge, and differential scanning calorimetry analysis of Bullseye® smokeless powder (Gunpowder). The participants found the Gunpowder: 1) to have a range of sensitivity to impact, from less than RDX to almost as sensitive as PETN, 2) to be moderately sensitive to BAM and ABL friction, 3) have a range for ESD, from insensitive to more sensitive than PETN, and 4) to have thermal sensitivity about the same as PETN and RDX.

    Several on-going, low level volcanic explosions exhibit background phenomena commonly known as puffing, or in some cases chugging. Recently these events have been scrutinized because of the initiation of infrasonic monitoring, whereas earlier the events may have gone undetected. The activity associated with a puffer at a volcanic vent is generally small in magnitude and is often not observed audibly. The low frequency signals are readily observed on sensitive acoustic instrumentation and they provide a new dimension for our understanding of volcanic processes at volcanoes like Stromboli and Etna that have constant puffing signals. At other volcanoes, like Karymsky volcano in Kamchatka and Sangay Volcano in Ecuador, chugging signals associated with Strombolian style eruptions also provides new insights into the physics of the conduit systems. Here we present a statistical method of event detection, and event cluster association. When multiple vents work in unison it may be difficult to separate out chugging and puffing signals between spatially separated vents. The cluster analysis automatically differentiates between the vents based on waveform characteristics in the acoustic and seismic wavefields. Data examples from May, 2001, at Stromboli and Etna, show extensive periods of puffing (1-5 second frequency) superimposed on a background of vigorous, small-scale explosive activity. At Karymsky and Sangay non-linear, dynamic models explain the fluid flow through vents which gives rise to chugging. Furthermore, the frequency of chugging events appears to be associated with the intensity of lava flows and eruption rate.

  15. Toxic Marine Puffer Fish in Thailand Seas and Tetrodotoxin They Contained

    Full Text Available A total of 155 puffers caught from two of Thailand’s seas, the Gulf of Siam and the Andaman seas, during April to July 2010 were included in this study. Among 125 puffers from the Gulf of Siam, 18 were Lagocephalus lunaris and 107 were L. spadiceus which were the same two species found previously in 2000–2001. Thirty puffers were collected from the Andaman seas, 28 Tetraodon nigroviridis and two juvenile Arothron reticularis; the two new species totally replaced the nine species found previously in 1992–1993. Conventional mouse bioassay was used to determine the toxicity in all fish tissue extracts, i.e., liver, reproductive tissue, digestive tissue and muscle. One of each of the species L. lunaris and L. spadiceus (5.56 and 0.93%, respectively were toxic. All 28 T. nigroviridis and 2 A. reticularis (100% from the Andaman seas were toxic. The toxicity scores in T. nigroviridis tissues were much higher than in the respective tissues of the other three fish species. Liquid chromatography/tandem mass spectrometry (LC-MS/MS revealed that the main toxic principle was tetrodotoxin (TTX. This study is the first to report TTX in L. spadiceus. Our findings raised a concern for people, not only Thais but also inhabitants of other countries situated on the Andaman coast; consuming puffers of the Andaman seas is risky due to potential TTX intoxication.

  16. Commercial production of tiger puffer ( Takifugu rubripes) in winter using a recirculating aquaculture system

    Tiger puffer ( Takifugu rubripes) is a promising species for aquaculture production because of its high value and limited supply. However, in the north of China, using sea cages to culture this species in winter is hampered by the fact that the seawater temperature is extremely low. Here, a large scale commercial production of tiger puffer has been successfully realized using a recirculating aquaculture system (RAS) from 3 October 2012 to 31 May 2013. The RAS was comprised of nine culture tanks (total water volume 200 m3) and stocked with approximately a total of 14400 fish (initial mean weight 160 g). The tiger puffer was hand-fed at a rate of 0.7% of total body weight per day, and the feed conversion rate was (1.21 ± 0.3) kg kg-1. The recycle water in RAS was treated by a sieve bend screen, a foam fractionator, a submerged biofilter, an UV sterilizer and a submersible aerator. During the whole culture period, an excellent water quality control was achieved in RAS. At the end of this experiment, the survival rate of tiger puffer was more than 98%. The final tank densities averaged 31.2 kg m-3, and the final individual mean weight was 440 g.

    PURPOSE: A particular problem in hypertrophic cardiomyopathy (HCM) is the need to distinguish between true and apparent ischemia in otherwise normal areas of muscle when these are compared with adjacent hypertrophic muscle. The authors of this paper studied patients with proved HCM to define patterns of perfusion. T1-201 single photon emission CT (SPECT) was performed in 83 HCM patients immediately after stress (dipyridamole, 0.5 mg/kg) and 3 hours later for the redistribution image. The data were analyzed by a normalized quantitative analysis using a local bulls-eye technique. In all patients, the pattern of tracer distribution was different from expected uptake in a normal population.     The development of high quality factor solid-state microcavities with low mode volumes has paved the way towards on-chip cavity quantum electrodynamics experiments and the development of high-performance nanophotonic devices. Here, we report on the implementation of a new kind of solid-state vertical microcavity, which allows for confinement of the electromagnetic field in the lateral direction without deep etching. The confinement originates from a local elongation of the cavity layer imprinted in a shallow etch and epitaxial overgrowth technique. We show that it is possible to improve the quality factor of such microcavities by a specific in-plane bullseye geometry consisting of a set of concentric rings with subwavelength dimensions. This design results in a smooth effective lateral photonic potential and therefore in a reduction of lateral scattering losses, which makes it highly appealing for experiments in the framework of exciton-polariton physics demanding tight spatial confinement.

Described here are the results for impact, friction, electrostatic discharge, and differential scanning calorimetry analysis of ammonium nitrate (AN) mixed with Bullseye® smokeless powder (Gunpowder). The participants found the AN/Gunpowder to: 1) have a range of sensitivity to impact, comparable to or less than RDX, 2) be fairly insensitive to friction as measured by BAM and ABL, 3) have a range for ESD, from insensitive to more sensitive than PETN, and 4) have thermal sensitivity about the same as PETN and Gunpowder. This effort, funded by the Department of Homeland Security (DHS), is putting the issues of safe handling of these materials in perspective with standard military explosives. The study is adding SSST testing results for a broad suite of different HMEs to the literature. Ultimately the study has the potential to suggest new guidelines and methods and possibly establish the SSST testing accuracies needed when developing safe handling practices for HMEs. Each participating testing laboratory uses identical test materials and preparation methods. Note, however, the test procedures differ among the laboratories. The testing performers involved are Lawrence Livermore National Laboratory (LLNL), Los Alamos National Laboratory (LANL), Indian Head Division, Naval Surface Warfare Center, (NSWC IHD), Sandia National Laboratories (SNL), and Air Force Research Laboratory (AFRL/RXQL).     Acetabular introducers have a built-in inclination of 45 degrees to the handle shaft. With patients in the lateral position, surgeons aim to align the introducer shaft vertical to the floor to implant the acetabulum at 45 degrees. We aimed to determine if a bulls-eye spirit level attached to an introducer improved the accuracy of implantation. A small circular bulls-eye spirit level was attached to the handle of an acetabular introducer. A saw bone hemipelvis was fixed to a horizontal, flat surface. A cement substitute was placed in the acetabulum and subjects were asked to implant a polyethylene cup, aiming to obtain an angle of inclination of 45 degrees. Two attempts were made with the spirit level masked and two with it unmasked. The distance of the air bubble from the spirit level's center was recorded by a single assessor. The angle of inclination of the acetabular component was then calculated. Subjects included both orthopedic consultants and trainees. Twenty-five subjects completed the study. Accuracy of acetabular implantation when using the unmasked spirit level improved significantly in all grades of surgeon. With the spirit level masked, 12 out of 50 attempts were accurate at 45 degrees inclination; 11 out of 50 attempts were "open," with greater than 45 degrees of inclination, and 27 were "closed," with less than 45 degrees. With the spirit level visible, all subjects achieved an inclination angle of exactly 45 degrees. A simple device attached to the handle of an acetabular introducer can significantly improve the accuracy of implantation of a cemented cup into a saw bone pelvis in the lateral position.

    Epizootic ulcerative syndrome (EUS) caused by the oomycete Aphanomyces invadans is an invasive, opportunistic disease of both freshwater and estuarine fishes. Originally documented as the cause of mycotic granulomatosis of ornamental fishes in Japan and as the cause of EUS of fishes in southeast Asia and Australia, this pathogen is also present in estuaries and freshwater bodies of the Atlantic and gulf coasts of the USA. We describe a mass mortality event of 343 captive juvenile bullseye snakehead Channa marulius collected from freshwater canals in Miami-Dade County, Florida. Clinical signs appeared within the first 2 d of captivity and included petechiae, ulceration, erratic swimming, and inappetence. Histological examination revealed hyphae invading from the skin lesions deep into the musculature and internal organs. Species identification was confirmed using a species-specific PCR assay. Despite therapeutic attempts, 100% mortality occurred. This represents the first documented case of EUS in bullseye snakehead fish collected from waters in the USA. Future investigation of the distribution and prevalence of A. invadans within the bullseye snakehead range in south Florida may give insight into this pathogen-host system.

Los Sphoeroides spp. basaron su alimentación en los organismos bentónicos, sobresaliendo por su consumo el mejillón (Brachidontes sp., la lapa (Balanus sp. y gasterópodos (Crepidula sp.. Previo al impacto del huracán, las tres especies utilizaron de forma diferencial los recursos alimenticios disponibles (bivalvos, gasterópodos, cirrípedos y decápodos recurriendo a diferentes estrategias que les permitieron minimizar la competencia interespecífica y coexistir.     Sphoeroides testudineus is a dominant species in the coastal systems of Yucatán. Because of its wide distribution, occurrence and abundance performs an important functional role in coastal ecosystems. We assessed the trophic preferences and trophic-level variation in space and time for this species in Ria Lagartos lagoon, an hyperhaline ecosystem located Northwest of Yucatan Peninsula. The specimens were collected bimonthly during two annual periods (2004-2005 and 2007-2008) in 23 sites distributed along the system into four zones (marine, channel, Coloradas West and Coloradas East). Spatial and seasonal trophic variations were evaluated using canonical correspondence analysis (ACC). In a total of 382 individuals, 68 food resources included in 20 trophic groups were obtained. Higher relative importance index values (IIR) were obtained for bivalves, gastropods and macrophytes. Seasonal trophic variation showed that bivalves and gastropods were consumed along the year, while macrophytes were a preferential food during rains and windy seasons. Spatial variation indicates lower gastropods consumption at the inner zone of the system, and the opposite ocurred with bivalves. The consumption ofmacrophytes may be due to their greater abundance during rains and in the channel zone. The ACC showed that spatial trophic variation was due because of the abundance of the preferential preys (bivalves, gastropods), while seasonal differences by the secondary (amphipods, eggs, nematodes, brachyuran, detritus, nematodes). The results contribute to the biological knowledge of the species and highlight the importance and trophic function of S testudineus for the energy transfer from the benthos to higher trophic levels of the food chain.

    The roasted fish fillet sample at the retail collected in Beijing and Qingdao were detected for TTX, and the TTX positive samples was analyzed for fish species identification. TTX was tested by EUSA method and the cytochrome c oxidase I (COI) genome of TTX-positive samples was extracted and identified by DNA barcode. Totally, 90 samples were tested by EUSA and 58 (64.4%) samples were positive for TTX with the levels ranging from 0.10 mg/kg to 63.81 mg/kg. Among the TTX positive samples, 24 (41.3%) were identified containing toxic puffer fish and 21 (87.5%) were Lagocephalus lunaris, the highly toxic puffer fish. Some roasted fish fillet samples obtained from the retail in two cities were positive for TTX and contained toxic puffer fish. Based on these results, we suggest that roasted fish fillet producers should prevent toxic puffer fish from mixing in the raw material and the I regulators should strengthen the TTX surveillance and product labeling supervision of roasted fish fillet.

    Full Text Available Objective: To evaluate the antimicrobial activity and brine shrimp lethality activity of six different species of puffer fishes, including Cyclichthys orbicularis, Diodon holocanthus, Canthigaster solandri, Arthron hispidus, A. inermis and Lagocephalua inermis (L. inermis. Methodology: The puffer fishes were collected from Annangkovil Fish Landing Centre (Lattitude 11°30.47' N; Longitude 79°47.02' E, Parangipettai, Southeast Coast of India during summer season because of availability. Fresh tissue samples were collected from the clearly washed specimens, extracted with methanol at 37 °C for 3 days and filtered through Whatman No. 1 filter paper. The solvents such as methanol and ethanol were concentrated by using rotary evaporator under reduced pressure. The dark brown gummy mass was stored at 4 °C for further analysis. Prepared crude samples were analysed with human pathogens to assess the antibacterial activity and this was carried out by using standard disc diffusion method. The brine shrimp lethality was calculated as the percentage of mortality which was firstly calculated by dividing the number of dead larvae by the total number and then multiplied to 100%. Results: The antibacterial activity of crude extract of puffer fishes were exhibited against 10 different human bacterial pathogens. Among the ten human pathogens, Arthron hispidus showed maximum zone of inhibition (8 mm against Staphylococcus aureus while L. inermis showed minimum activity (1 mm against Proteus mirabilis and no zone of inhibition was observed against Staphylococcus aureus. Brine shrimp lethality was examined with six puffer fish extracts. Cyclichthys orbicularis showed maximum mortalities as 100% and L. inermis showed minimum mortalities as 70% at a concentration of 500 µg/mL.     The study investigated physiochemical and functional properties of gelatins from tiger puffer skin (GTPS) extracted with warm-water method at 45, 55 and 65°C for 3, 6 and 12h. GTPS45 exhibited strong γ-, β-, α1- and α2-chains, but they faded with the presence of fragments (<80KDa) in other GTPS. As the increase of temperature and time, amide I and A were shifted to higher wavenumber and their amplitudes in GTPS65-12 decreased. Lightness declined while redness and yellowness were enhanced. Gel strength was in a range of 144.5-79.8g, where it of GTPS45-3 was the highest while it of GTPS65-12 was the lowest (p<0.05), coincided with the longer relaxation time of T 22 for immobilized water. Decline in gelling and melting temperature was observed when temperature and time increased. EAI and ESI increased as time prolonged or temperature climbed (p<0.05), in agreement with the form of emulsion droplet by microscopy. Both category and level of volatile compounds increased GTPS65-12 and 55-12 compared with those in GTPS45-12. Results suggest that temperature and time greatly affect the properties of GTPS. To ensure good properties, GTPS should be extracted at a temperature of 45-55°C for less than 12h. Copyright © 2017 Elsevier B.V. All rights reserved.

  9. Biological Activities of Tetrodotoxin-Producing Enterococcus faecium AD1 Isolated from Puffer Fishes

    Full Text Available Puffer fishes were collected from the central sea in Vietnam from spring to summer season. The eggs were incubated in MRS broth that was used to test the toxicity in mice and isolate the lactic acid bacteria community that could produce tetrodotoxin (TTX. Thin layer chromatography (TLC and high performance lipid chromatography (HPLC were used to detect and quantify TTX. As a result, Enterococcus faecium AD1 which was identified by biochemical test and 16S rRNA analysis could produce TTX 0.3 mg/mL when cultured in MRS broth. The bacterium was optimized for TTX production and gave 0.18 mg/mL, 0.07 mg/mL, and 0.15 mg/mL in media prepared from the meat-washing water of freshwater fishes (Pangasius bocourti, Oreochromis sp. and sea fish (Auxis thazard, respectively, that are also hopeful to answer some poisoning cases related to eating fishes. Enterococcus faecium also showed the wide antimicrobial activities on yeast, Gram-negative and -positive bacteria. Extracted exopolysaccharide (EPS that reacted with 2,2-diphenyl-1-picrylhydrazyl to give IC50 at 5 mg/mL equaled 11 mg/mL ascorbic acid which could show effects on Hela-6 and Hep G2 using sulforhodamine B test. Enterococcus faecium can be claimed as a promising source in tetrodotoxin and biological compounds.

  10. Differential expression patterns of PQRFamide peptide and its two receptor genes in the brain and pituitary of grass puffer during the reproductive cycle.

    Pain-modulatory neuropeptides, PQRFamide (PQRFa) peptides, have recently been implicated in the regulation of reproduction in fish. As a first step toward investigating the role of PQRFa peptides on reproductive function in the grass puffer Takifugu niphobles, which is a semilunar spawner, we cloned genes encoding PQRFa peptide precursor (pqrfa) and its two types of receptors (pqrfa-r1 and pqrfa-r2), and examined changes in their expression levels in the brain and pituitary over several months during the reproductive cycle. The grass puffer PQRFa peptide precursor of 126 amino acid residues contains two putative PQRFa peptides, PQRFa-1 and PQRFa-2, which correspond to NPFF and NPAF in other vertebrates, respectively. The grass puffer PQRFa-R1 and PQRFa-R2 consist of 426 and 453 amino acid residues, respectively, and contain distinct characteristics of G-protein coupled receptors. These three genes were exclusively expressed in the brain and pituitary. The expression levels of pqrfa and pqrfa-r1 were significantly increased during the late stage of sexual maturation, but low in the spawning fish just after releasing sperms and eggs. Therefore, the grass puffer PQRFa peptide may have a role in the late stage of sexual maturation before spawning via PQRFa-R1. In contrast, the pqrfa-r2 expression showed maximum levels in the spawning fish and in the post-spawning period. The present results provide fundamental data suggesting that the grass puffer PQRFa peptide may have multiple roles in the control of reproduction that are dependent on the reproductive stages.     This study aims to develop a puffer fish skin gelatin (PSG) film that contains Moringa oleifera Lam. leaf extract (ME) as a new biodegradable film. With the increase in ME concentration, the tensile strength and elongation at break of the PSG film increased, whereas the oxygen permeability and water vapor permeability decreased. In addition, the PSG film with ME exhibited antimicrobial activity against Listeria monocytogenes and antioxidant activity. To apply the ME-containing PSG film to food packaging, Gouda cheese was wrapped with the ME-containing PSG film. During storage, the cheese packaging with the ME-containing PSG film effectively inhibited the microbial growth and retarded the lipid oxidation of cheese compared with the control sample. Thus, the ME-containing PSG film can be used as an antimicrobial and antioxidative packaging material to improve the quality of food products.

  13. Contribution to the feeding ecology of the banded puffer fish Colomesus psittacus (Tetraodontidae in north Brazilian mangrove creeks

    Full Text Available Stomach contents were examined from 102 banded puffer, Colomesus psittacus (Tetraodontidae, caught from intertidal mangrove creeks at diurnal neap tides between June and September, 1997 (early dry season near Bragança (north Brazil. The study found that C. psittacus were specialized predators of Cirripedia (Balanus spp. and Brachyuran crabs (Uca spp., Pachygrapsus gracilis (mean: 58 and 38% by dry weight, respectively, emphasizing a short food chain in the mangrove system. Cirripedia and Brachyura dominated the diet in all size classes, however, the prey spectrum narrowed with fish size. The mean daily consumption of Cirripedia and Brachyura was 6.2% body weight of C. psittacus. On average C. psittacus consumed 100.3 g.ha-1.d-1 of Cirripedia and 178.7 g.ha-1.d-1 of Brachyura (wet weight. The predation on Brachyuran crabs - a significant driver of fluxes of organic matter and energy in the system - provides C. psittacus with an important ecological function in the mangrove food web. A plant-animal interaction is proposed where C. psittacus exerts a mutually beneficial cleaning function on the Aufwuchs (Cirripedia and associated epibiota of Rhizophora mangle stilt roots. Our results and those of other studies suggest that C. psittacus encounter optimum foraging conditions in the mangrove at high inundations at daylight (spring tide-day whereas darkness and low inundations are linked to poor foraging conditions (neap tide-night. The C. psittacus resource could be used as an alternative income in the region in terms of i sustainable catch and filet processing for exports to East Asia, ii developing certified aquaculture methods for breeding puffers for the aquarium trade.

    We examined whether infection by the monogenean Heterobothrium okamotoi induces production of specific antibodies against oncomiracidia and their cilia, larvae on the gills, and adults on the branchial cavity wall of tiger puffer Takifugu rubripes. We also investigated whether specific antibody production participates in acquired protection against H. okamotoi. Sera from persistently infected fish immobilized H. okamotoi oncomiracidia 89 days after exposure and antibody levels (measured by enzyme-linked immunosorbent assays) in the sera against oncomiracidia and their cilia increased compared with sera from control (naïve) fish. Antibody levels in these sera against the larvae and adult stages did not increase. The number of H. okamotoi on persistently infected fish was significantly lower than for control fish (Ptank. Thus tiger puffer produced specific antibodies against oncomiracidia and their cilia, and acquired partial protection against H. okamotoi. Intraperitoneal injection of proteins of sonicated oncomiracidia or their cilia with an adjuvant also produced oncomiracidium agglutination antibodies in sera from tiger puffer; the antibody levels in these sera against oncomiracidia and their cilia increased compared with sera from control fish (injection of BSA with an adjuvant) at 14, 44, and 75 days after the booster immunization. However, in a parasite challenge at 54-58 days after the booster immunization, the infection levels of fish immunized with parasites of sonicated oncomiracidia or their cilia were the same as the control fish. Western blot showed that sera from persistently infected fish and fish immunized with sonicated oncomiracidia or their cilia recognized similar antigenic bands, suggesting that tiger puffer tends to react against these antigens compared with other antigens. These results indicated that specific antibodies against these cilia and oncomiracidia induced by i.p. injection do not prevent H. okamotoi infection.

    Full Text Available The volatile and odor-active compounds in cooked meat of farmed obscure puffer (Takifugu obscurus were analyzed by gas chromatography–mass spectrometry–olfactometry (GC–MS–O. The volatile compounds were extracted by the simultaneous distillation–extraction (SDE method, then separated and identified by GC–MS. Odor-active compounds in the SDE extract were characterized by GC–MS–O. A total of 68 volatile compounds were found, including 23 aldehydes, 10 alcohols, nine ketones, 17 N- or S-containing compounds and aromatics, three acids, three alkanes, and three esters. Of these, 31 odor-active compounds were detected and identified.     The effect of dietary amylose/amylopectin (AM/AP) ratio on growth, feed utilization, digestive enzyme activities, plasma parameters, and postprandial blood glucose responses was evaluated in juvenile obscure puffer, Takifugu obscurus. Five isonitrogenous (430 g kg(-1) crude protein) and isolipidic (90 g kg(-1) crude lipid) diets containing an equal starch level (250 g kg(-1) starch) with different AM/AP ratio diets of 0/25, 3/22, 6/19, 9/16 and 12/13 were formulated. Each experimental diet was fed to triplicate groups (25 fish per tank), twice daily during a period of 60 days. After the growth trial, a postprandial blood response test was carried out. Fish fed diet 6/19 showed best growth, feed efficiency and protein efficiency ratio. Hepatosomatic index, plasma total cholesterol concentration, liver glycogen and lipid content, and gluconokinase, pyruvate kinase and fructose-1,6-bisphosphatase activities were lower in fish fed highest AM/AP diet (12/13) than in fish fed the low-amylose diets. Activities of liver and intestinal trypsin in fish fed diet 3/22 and diet 6/19 were higher than in fish fed diet 9/16 and diet 12/13. Activities of liver and intestinal amylase and intestinal lipase, and starch digestibility were negatively correlated with dietary AM/AP ratio. Fish fed diet 3/22 and diet 6/19 showed higher plasma total amino acid concentration than fish fed the other diets, while plasma urea nitrogen concentration and activities of alanine aminotransferase and aspartate aminotransferase showed the opposite trend. Equal values were found for viscerosomatic index and condition factor, whole body and muscle composition, plasma high-density and low-density lipoprotein cholesterol concentrations, and activities of lipase and hexokinase and glucose-6-phosphatase in liver.     The seasonal, daily and lunar control of reproduction involves photoperiodic, circadian and lunar changes in the activity of kisspeptin, gonadotropin-inhibitory hormone (GnIH) and gonadotropin-releasing hormone (GnRH) neurons. These changes are brought through complex networks of light-, time- and non-photic signal-dependent control mechanisms, which are mostly unknown at present. The grass puffer, Takifugu alboplumbeus, a semilunar spawner, provides a unique and excellent animal model to assess this question because its spawning is synchronized with seasonal, daily and lunar cycles. In the diencephalon, the genes for kisspeptin, GnIH and their receptors showed similar expression patterns with clear seasonal and daily oscillations, suggesting that they are regulated by common mechanisms involving melatonin, circadian clock and water temperature. For implications in semilunar-synchronized spawning rhythm, melatonin receptor genes showed ultradian oscillations in expression with the period of 14.0-15.4 h in the pineal gland. This unique ultradian rhythm might be driven by circatidal clock. The possible circatidal clock and circadian clock in the pineal gland may cooperate to drive circasemilunar rhythm to regulate the expression of the kisspeptin, GnIH and their receptor genes. On the other hand, high temperature (over 28 °C) conditions, under which the expression of the kisspeptin and its receptor genes is markedly suppressed, may provide an environmental signal that terminates reproduction at the end of breeding period. Taken together, the periodic regulation of the kisspeptin, GnIH and their receptor genes by melatonin, circadian clock and water temperature may be important in the precisely-timed spawning of the grass puffer.     Full Text Available Abstract The effects of daily feeding frequency (Exp I, water temperature (Exp II, and stocking density (Exp III on the growth of river puffer, Takifugu obscurus, juvenile fish of 10 and 40 g in body weight were examined to develop effective techniques to produce river puffer in a non-exchange water system. In Exp I, fish were fed commercial floating feed with 45 % protein one to five times per day to apparent satiation each by hand daily for 8 weeks at 25 °C. In both the 10- and 40-g size groups, the final body weight, daily feed consumption, and weight gain of fish fed one meal per day were significantly lower than those of fish fed five meals per day (P < 0.05. However, there were no significant differences in the final body weight, daily feed consumption, and weight gain among fish fed two, three, and five meals per day. Feed efficiency showed decreasing tendency with increasing size of fish. In Exp II, fish of 10 and 40 g in initial body weight were reared with the commercial feed at 15–30 °C for 8 weeks. The weight gain of fish increased with raising water temperature up to 25 °C and decreased drastically at 30 °C for both sizes. The Q10 of specific growth rate was decreased with raising water temperature from 5.04 (temperature interval, 15–20 °C to 0.66 (25–30 °C for the 10-g fish and from 4.98 to 0.31 for the 40-g fish. In Exp III, the effect of stocking density on growth was examined with fish of 10 and 40 g in initial body weight. The final body weight for initial stocking densities of 4, 8, and 12 kg/m3 was significantly higher than that of 20 kg/m3 for the 10-g fish, and the final stocking density reached 10.1, 19.2, 28.7, and 39.9 kg/m3, respectively. The authors report 27 human poisonings from ingestion of puffer fish in patients treated at Toxicology Centers in the states of Santa Catarina and Bahia, Brazil, between 1984 and January 2009. Poisonings were classified as moderate (52% and severe (33%, two deaths were observed.     The aim of the present study was to investigate the long-term effects of 17β-estradiol (E2) exposure on gonadal development in the tiger puff er ( Takifugu rubripes), which has a genetic sex determination system of male homogametic XY-XX. Tiger puff er larvae were exposed to 1, 10 and 100 μg/L E2 from 15 to 100 days post-hatch (dph) and then maintained in clean seawater until 400 dph. Changes in sex ratio, gonadal structure and gonadosomatic index (GSI) were monitored at 100, 160, 270 and 400 dph. Sex-associated single nucleotide polymorphism (SNP) markers were used to analyze the genetic sex of samples, except those at 100 dph. Exposure had a positive effect on the conversion of genetically male gonads into phenotypically female gonads at 100 dph. However, gonads from 60% of genetic XY males in the 1-μg/L E2 group and 100% in the 10-μg/L E2 group developed intersexual gonads at 160 dph; gonads of all genetic XY males in the two treatment groups reverted to testis by 270 dph. While 38%, 57% and 44% of gonads of XY fish in the 100-μg/L E2 group reverted to intersexual gonads at 160, 270 and 400 dph, respectively, none reverted to testis after E2 treatment. In addition, E2 exposure inhibited gonadal growth of both genetic sexes, as indicated by the clear dose-dependent decrease in GSI at 270 and 400 dph. The results showed that exposure to E2 during the early life stages of tiger puff er disrupted gonadal development, but that fish recovered after migration to clean seawater. The study suggests the potential use of tiger puff er as a valuable indicator species to evaluate the effects of environmental estrogens on marine fish, thereby protecting valuable fishery resources.

    There is increasing evidence for frequent turnover in sex chromosomes in vertebrates. Yet experimental systems suitable for tracing the detailed process of turnover are rare. In theory, homologous turnover is possible if the new sex-determining locus is established on the existing sex-chromosome. However, there is no empirical evidence for such an event. The genus Takifugu includes fugu (Takifugu rubripes) and its two closely-related species whose sex is most likely determined by a SNP at the Amhr2 locus. In these species, males are heterozygous, with G and C alleles at the SNP site, while females are homozygous for the C allele. To determine if a shift in the sex-determining locus occurred in another member of this genus, we used genetic mapping to characterize the sex-chromosome systems of Takifugu niphobles. We found that the G allele of Amhr2 is absent in T. niphobles. Nevertheless, our initial mapping suggests a linkage between the phenotypic sex and the chromosome 19, which harbors the Amhr2 locus. Subsequent high-resolution analysis using a sex-reversed fish demonstrated that the sex-determining locus maps to the proximal end of chromosome 19, far from the Amhr2 locus. Thus, it is likely that homologous turnover involving these species has occurred. The data also showed that there is a male-specific reduction of recombination around the sex-determining locus. Nevertheless, no evidence for sex-chromosome differentiation was detected: the reduced recombination depended on phenotypic sex rather than genotypic sex; no X- or Y-specific maker was obtained; the YY individual was viable. Furthermore, fine-scale mapping narrowed down the new sex-determining locus to the interval corresponding to approximately 300-kb of sequence in the fugu genome. Thus, T. niphobles is determined to have a young and small sex-determining region that is suitable for studying an early phase of sex-chromosome evolution and the mechanisms underlying turnover of sex chromosome.     Although fishers' knowledge has been recently considered into management programmes, there is still the need to establish a better understanding of fishers' perceptions and cognition. Fishers can provide novel information on the biology and ecology of species, which can potentially be used in the management of fisheries. The knowledge fishers have and how they classify nature is empirically based. It is common, for example, to observe that fishers' taxonomy is often represented by the generic level, one of the hierarchical categories of folk classification that is somewhat analogous to the Linnean genus, as it groups organisms of a higher rank than the folk species.In this study we compiled the knowledge fishers have on local fish, such as their folk names, diet and habitat. Five coastal communities widely distributed along the Brazilian coast were studied: two from the northeast (Porto Sauípe and Itacimirim, in Bahia State, n of interviewees = 34), two from the southeast (Itaipu at Niterói and Copacabana at Rio de Janeiro, Rio de Janeiro State, n = 35) and one from the south coast (Pântano do Sul, in Santa Catarina State, n = 23). Fish pictures were randomly ordered and the same order was presented to all interviewees (n = 92), when they were then asked about the species name and classification and its habitat and diet preferences. Fishers make clusters of fish species, usually hierarchically; fishers of the coast of Brazil use mostly primary lexemes (generic names) to name fish; and fishers did not differentiate between scientific species, since the same folk generic name included two different scientific species. The current article presents 51 geographically distinct records for 21 taxa belonging to 6 Phyla, extending from the western Mediterranean to the Levantine. The new records, per country, are as follows: Spain: the cryptogenic calcareous sponge Paraleucilla magna is reported from a new location in the Alicante region. Algeria: the rare Atlanto-Mediterranean bivalve Cardium indicum is reported from Annaba. Tunisia: new distribution records for the Indo-Pacific lionfish Pterois miles from Zembra Island and Cape Bon. Italy: the ark clam Anadara transversa is reported from mussel cultures in the Gulf of Naples, while the amphipod Caprella scaura and the isopods Paracerceis sculpta and Paranthura japonica are reported as associated to the –also allochthonous–bryozoan Amathia verticillata in the Adriatic Sea; in the latter region, the cosmopolitan Atlantic tripletail Lobotes surinamensisis also reported, a rare finding for the Mediterranean.   14. Effects of mating disruption treatments on navel orangeworm (Lepidoptera: Pyralidae) sexual communication and damage in almonds and pistachios.

    Two experiments in 2003 examined the effects of different ways of dispensing the principal sex pheromone component on sexual communication among and crop damage by the navel orangeworm, Amyelois transitella (Walker) (Lepidoptera: Pyralidae) in Nonpareil almonds and pistachios. A third experiment in 2004 compared the effect on navel orangeworm damage to several almond varieties using one of these dispensing systems by itself or with phosmet, phosmet alone, and an untreated control. Additional data are presented estimating release rates from timed aerosol release devices (PuffersNOW, Suterra LLC, Bend, OR) and hand-applied membrane dispensers. In 2003, puffers placed peripherally around 16-ha blocks, evenly spaced Puffers, and hand-applied dispensers reduced males captured in virgin-baited traps by > or = 95% and mating in sentinel females by > or = 69%, with evenly placed Puffers showing greater reduction of males captured and females mated compared with the other dispensing systems. Mating disruption with gridded Puffers or hand-applied devices in almonds resulted in an approximately 37% reduction of navel orangeworm damage (not significant), whereas peripheral Puffers resulted in a 16% reduction of navel orangeworm damage to almonds. In pistachios neither peripheral nor gridded Puffers reduced navel orangeworm damage, whereas insecticide reduced damage by 56%. In 2004, Puffers alone, insecticide alone, and both in combination significantly reduced navel orangeworm damage in Nonpareil almonds. In other, later harvested varieties, the insecticide treatments reduced damage, whereas the mating disruption treatment alone did not. We discuss application of these findings to management of navel orangeworm in these two crops.

    Full Text Available We were trying to study the validity of Puffer fish's poison(Tetrodotoxin- TTX to make a traditional Korean Medical treatment. The following conclusions were made after literary studies. 1. The first record of the puffer fish dates back 2000 years ago in the Chinese text Book of Mountain and Sea and other texts from the similar period. 2. Puffer fish's poison IS known as tetrodotoxin which is an amino perhydroquinazoline compound. It has a chemical formula of C11H17N3O8 in the hemiacetal structure and has the molecular weight of 319. 3. Tetrodotoxin (TTX plays a role as potent neurotransmitter blocker by blocking the Na+-gate channel which hinders the influx of Na+ ion into the cell. 4. Symptoms of the puffer fish poisoning ranges from blunted sense in the lips and tongue, occasional vomiting in the first degree to sudden descending of the blood pressure, apnea, and other critical conditions in the fourth degree. Intoxication of the puffer fish poison progresses at a rapid pace as death may occur after an hour and half up to eight hours in maximum. Typical death occurs after four to six hours. 5. Ways to treat the puffer fish poisoning include gastric irrigation, induce vomiting, purgation, intravenous fluid injection, and correcting electrolytic imbalance and acidosis. In cases of dyspnea, apply oxygen inhalation and conduct artificial respiration. 6. Tetrodotoxin (TTX may be applied in treating brain disorders, ocular pain, excess pain in the large intestine and ileum, and relieving tension of the skeletal museles, neuralgia, rheumatism, arthritis, and etc. 7. In terms of Oriental medicine, the puffer fish poison has characteristics of sweet, warm, and poisonous. It's known efficacies are to tonify weakness, dispel damp, benefit the lower back, relieve hemorrhoid, kills parasites, remove edema, and so forth.     Full Text Available In fish catching activity, the fishermen in Banyuanyar district use mini tools for catching fish (purse seine and use light assist tools for an operation in the night day. The light, that is used, is kerosene pressure lantern and mercury lights or lamp that is used on water surface (surface lamp. This kind of light is used to collect “pelagic” fish that has positive phototaxis characteristic. Puffer fish’s response towards different sourse of light, that’s kerosene pressure lantern and mercury light is needed to be known. Therefore, we can know the source of light that is more effective in order to collect the fish. It is hoped that the productivity of puffer fish will be developed for the fishermen. During the research with ten times repeating, the total account of puffer fish’s haul that is resulted with two different treatment  by using kerosene pressure lantern and mercury light is that 810 kg and 1.460 kg. The data of puffer fish’s haul that is gotten after using Mann-Whitney test shows that the kerosene pressure lantern and mercury light not to really different, especially in the haul of puffer fish in significant standart (0,05 is 2,262, while in Mann-Whitney test account is 0,171.   18. Quantitative analysis of normal thallium-201 tomographic studies

    To determine the normal (nl) distribution of Tl-201 uptake post exercise (EX) and at redistribution (RD) and nl washout, Tl-201 rotational tomographic (tomo) studies were performed in 40 subjects: 16 angiographic (angio) nls and 24 nl volunteers (12 from Emory and 12 from Yale). Oblique angle short axis slices were subjected to maximal count circumferential profile analysis. Data were displayed as a ''bullseye'' functional map with the apex at the center and base at the periphery. The bullseye was not uniform in all regions because of the variable effects of attenuation and resolution at different view angles. In all studies, the septum: lateral wall ratio was 1.0 in males and approximately equal to 1.0 in females. This occurred predominantly because of anterior defects due to breast soft tissue attenuation. EX and RD bullseyes were similar. Using a bi-exponential model for Tl kinetics, 4 hour normalized washout ranged 49-54% in each group and showed minimal variation between walls throughout the bullseye. Thus, there are well defined variations in Tl-201 uptake in the nl myocardium which must be taken into consideration when analyzing pt data. Because of these defects and the lack of adequate methods for attenuation correction, quantitative analysis of Tl-201 studies must include direct comparison with gender-matched nl data sets

  19. The genetic architecture of UV floral patterning in sunflower.

    The patterning of floral ultraviolet (UV) pigmentation varies both intra- and interspecifically in sunflowers and many other plant species, impacts pollinator attraction, and can be critical to reproductive success and crop yields. However, the genetic basis for variation in UV patterning is largely unknown. This study examines the genetic architecture for proportional and absolute size of the UV bullseye in Helianthus argophyllus , a close relative of the domesticated sunflower. A camera modified to capture UV light (320-380 nm) was used to phenotype floral UV patterning in an F 2 mapping population, then quantitative trait loci (QTL) were identified using genotyping-by-sequencing and linkage mapping. The ability of these QTL to predict the UV patterning of natural population individuals was also assessed. Proportional UV pigmentation is additively controlled by six moderate effect QTL that are predictive of this phenotype in natural populations. In contrast, UV bullseye size is controlled by a single large effect QTL that also controls flowerhead size and co-localizes with a major flowering time QTL in Helianthus . The co-localization of the UV bullseye size QTL, flowerhead size QTL and a previously known flowering time QTL may indicate a single highly pleiotropic locus or several closely linked loci, which could inhibit UV bullseye size from responding to selection without change in correlated characters. The genetic architecture of proportional UV pigmentation is relatively simple and different from that of UV bullseye size, and so should be able to respond to natural or artificial selection independently. © The Author 2017. A tiny magnet is mounted at that spot to catch wind-borne magnetic particles. The particles may have been tossed into the atmosphere surrounding the spacecraft at the time of landing and during the digging and delivery of the Mars soil sample by the surface sampler scoop.   4. Identification of tetrodotoxin and fish species in a dried dressed fish fillet implicated in food poisoning.

    There were five victims of neurotoxic food poisoning from a dried dressed fish fillet in Changhua County, Taiwan, in February 2000. The toxicity of the dried dressed fish fillets was 243 mouse units per g according to a tetrodotoxin bioassay. The partially purified toxin was identified as tetrodotoxin and anhydrotetrodotoxin. The sequence of the 376-nucleotide region in the cytochrome b gene of the mitochondrial DNA exhibited the same genotype as that of the toxic puffer fish Lagocephalus lunaris. The same single restriction site for Hinfl was found in the polymerase chain reaction (PCR) products from the dried dressed fish fillet and the muscle of L. lunaris, yielding two DNA fragments of 170 and 206 bp. However, no restriction site for Hinfl was found in the PCR products from other toxic puffer fishes, including Takifugu niphobles, Takifugu oblongus, and Takifugu rubripes. Therefore, the species of the dried dressed fish fillet was identified as L. lunaris and its causative agent was identified as tetrodotoxin.

  6. Trunk motion visual feedback during walking improves dynamic balance in older adults: Assessor blinded randomized controlled trial.

    Virtual reality and augmented feedback have become more prevalent as training methods to improve balance. Few reports exist on the benefits of providing trunk motion visual feedback (VFB) during treadmill walking, and most of those reports only describe within session changes. To determine whether trunk motion VFB treadmill walking would improve over-ground balance for older adults with self-reported balance problems. 40 adults (75.8 years (SD 6.5)) with self-reported balance difficulties or a history of falling were randomized to a control or experimental group. Everyone walked on a treadmill at a comfortable speed 3×/week for 4 weeks in 2 min bouts separated by a seated rest. The control group was instructed to look at a stationary bulls-eye target while the experimental group also saw a moving cursor superimposed on the stationary bulls-eye that represented VFB of their walking trunk motion. The experimental group was instructed to keep the cursor in the center of the bulls-eye. Somatosensory (monofilaments and joint position testing) and vestibular function (canal specific clinical head impulses) was evaluated prior to intervention. Balance and mobility were tested before and after the intervention using Berg Balance Test, BESTest, mini-BESTest, and Six Minute Walk. There were no significant differences between groups before the intervention. The experimental group significantly improved on the BESTest (p = 0.031) and the mini-BEST (p = 0.019). The control group did not improve significantly on any measure. Individuals with more profound sensory impairments had a larger improvement on dynamic balance subtests of the BESTest. Older adults with self-reported balance problems improve their dynamic balance after training using trunk motion VFB treadmill walking. Individuals with worse sensory function may benefit more from trunk motion VFB during walking than individuals with intact sensory function. Copyright © 2018 Elsevier B.V. All rights reserved.

    This paper reports on an expert system environment which automatically assigned certainty factors to abnormal regions in stress and delayed myocardial thallium-201 polar bulls-eye plots. MYCIN-type algorithms propagated certainty factors for the presence, location, and character of each coronary lesion. Ninety-four previously validated rules that considered only stress perfusion defects spawned 91 new rules considering tracer redistribution. Fifteen new rules assessed vascular territories for the presence and location of fixed or reversible defects. This artificial intelligence tool can provide novice readers of cardiac T1-201 studies automatic, consistent, objective, and justified interpretations that consider artifacts, coronary territory overlap, and multiple defects

  8. Peritoneal fluid causing inferior attenuation on SPECT thallium-201 myocardial imaging in women

    On SPECT thallium images, myocardial left ventricular (LV) anterior wall attenuation due to breast tissue is common in women. In contrast, in men, inferior wall counts are normally decreased compared to anterior counts. The purpose of this report is to describe cases of inferior wall attenuation of counts in women caused by peritoneal fluid, not myocardial disease. Twelve consecutive SPECT thallium myocardial studies performed in women on peritoneal dialysis, being evaluated for kidney transplant, were included in this study. For all studies, 3.5 mCi 201Tl were injected intravenously. Thirty-two images were acquired over 180 degrees (45 degrees RAO progressing to 45 degrees LPO) at 40 sec per stop. SPECT images were reviewed in short axis, horizontal long and vertical long axes. Data were also displayed in bullseye format with quantitative comparison to gender-matched normal files. Ten of 12 female patients studied had inferior wall defects on images, confirmed by bullseye display. All patients had approximately 2 liters of peritoneal fluid. Review of planar rotational views showed diaphragm elevation and fluid margin attenuations affecting left ventricular inferior wall.   9. Two types of SDR recognised in pre-stack velocity analysis of ultra-long-offset seismic reflection data in the South Atlantic

    Our understanding of continental breakup at volcanic margins has lagged behind that of non-volcanic margins in recent years. This is largely due to seismic imaging problems caused by the presence of thick packages of Seaward-Dipping Reflectors (SDRs) in the continent-ocean transition zone. These packages consist of interbedded tholeiitic lava flows, volcanic tuffs and terrestrial sediment that results in scattering, peg-leg multiples and defocusing of seismic energy. Here we analyse three ultra-long-offset (10.2 km), wide-bandwidth (5-100 Hz) seismic reflection profiles acquired by ION-GXT offshore South America during 2009-12 to gain new insights into the velocity structure of the SDRs and hence pattern of magmatism during continental breakup. We observe two seismic velocity patterns within the SDRs. The most landward packages show high velocity anomaly "bulls-eyes" of up to 1 km s-1. These highs occur where the stacked section shows them to thicken at the down-dip end of individual packages that are bounded by faults. All lines show 5-6 velocity highs spaced approximately 10 km apart. We interpret the velocity bulls-eyes as depleted mafic or ultramafic bodies that fed the sub-aerial tholeiitic lava flows during continental stretching. Similar relationships have been observed in outcrop onshore but have not been previously demonstrated in seismic data. The bulls-eye packages pass laterally into SDR packages that show no velocity highs. These packages are not associated with faulting and become more extensive going north towards the impact point of the Tristan da Cunha hotspot. This second type of SDR coincides with linear magnetic anomalies. We interpret these SDRs as the products of sub-aerial oceanic spreading similar to those seen on Iceland and described in the classic "Hinz model" and marine geophysical literature. Our work demonstrates that these SDRs are preceded by ones generated during an earlier phase of mechanical thinning of the continental crust. The

    A class of artificial intelligence (Al) programs known as neural networks are well suited to pattern recognition. A neural network is trained rather than programmed to recognize patterns. This differs from "expert system" Al programs in that it is not following an extensive set of rules determined by the programmer, but rather bases its decision on a gestalt interpretation of the image. The "bullseye" images from cardiac stress thallium tests performed on 50 male patients, as well as several simulated images were used to train the network. The network was able to accurately classify all patients in the training set. The network was then tested against 50 unknown patients and was able to correctly categorize 77% of the areas of ischemia and 92% of the areas of infarction. While not yet matching the ability of a trained physician, the neural network shows great promise in this area and has potential application in other areas of medical imaging.

    Full Text Available This data article provides further detailed information related to our research article titled “Microfabricated Blood Vessels Undergo Neovascularization” (DiVito et al., 2017 [1], in which we report fabrication of human blood vessels using hydrodynamic focusing (HDF. Hydrodynamic focusing with advection inducing chevrons were used in concert to encase one fluid stream within another, shaping the inner core fluid into ‘bullseye-like” cross-sections that were preserved through click photochemistry producing streams of cellularized hollow 3-dimensional assemblies, such as human blood vessels (Daniele et al., 2015a, 2015b, 2014, 2016; Roberts et al., 2016 [2–6]. Applications for fabricated blood vessels span general tissue engineering to organ-on-chip technologies, with specific utility in in vitro drug delivery and pharmacodynamics studies. Here, we report data regarding the construction of blood vessels including cellular composition and cell positioning within the engineered vascular construct as well as functional aspects of the tissues.

    Full Text Available A large number of Red Sea species are colonizing the eastern Mediterranean Sea via the Suez Canal, mainly following the Anatolian coasts and spreading westwards. Portunus pelagicus is one of the most common Red Sea swimming crabs, first recorded in the Levantine Basin in 1898. Four specimens of P. pelagicus were collected in different marine areas of Rhodes Island from 1991 to 2000, while three specimens of the lessepsian fish Apogon pharaonis, first recorded in the Mediterranean in 1947, were caught during 2002 in the NW coast of Rhodes. The sub-tropical character of the marine area around Rhodes seems to facilitate the propagation of lessepsian species. These migrants have reached the island at different velocity and degree of establishment of their populations. The occurrence of the blue swimmer crab P. pelagicus and of the bullseye cardinal fish A. pharaonis increases the number of the decapod Crustacea and fish species of Red Sea origin observed in Greek waters.

    Full Text Available Se describe el comportamiento de los parámetros de temperatura, salinidad, transparencia del agua y temperatura ambiente, y de la abundancia y diversidad de la comunidad del peces del sistema Candelaria-Panlau en escalas temporal y espacial. Los trabajos de campo se realizaron de manera mensual en cinco estaciones entre abril de 1993 y abril de 1994. La comunidad de peces quedó representada por 50 especies con 7 926 organismos y 144.7 kg de peso. Los valores de abundancia y diversidad de la comunidad fueron: 0.09 ind/m², 1.61 g/m² 18.25 g/ind, H’n = 1.879, J = 0.475 y D = 5.681. Las especies con dominio ecológico fueron: Cathorops melanopus, Diapterus rhombeus, Anchoa mitchilli, Sphoeroides testudineus, Bairdiella chrysura, B. ronchus, Cynoscion arenarius, y C. nebulosus, que en conjunto representan el 90.9 % en número y el 78.9 % en peso de la captura total. Las matrices de valores de los parámetros ambientales y de abundancia numérica de la comunidad de peces, fueron sometidas a un análisis de factores con el método de extracción de componentes principales, su representación gráfica confirma la definición de especies dominantes y demuestra que la salinidad, estación cinco y mes de febrero son los componentes que caracterizan al sistema y a la comunidad de peces.The environmental and ecological patterns of the fish community in Candelaria-Panlau system (Mexico are described.     Full Text Available The fish assemblage in a coastal lagoon with mangroves known as "Rancho Bueno" was determined and associated with environmental parameters. We used an experimental otter trawl net to catch the fish, and 62 fish species were identified from 48 genera and 30 families. The most abundant species were: Etropus crossotus, Eucinostomus gracilis, Paralabrax maculatofasciatus, Sphoeroides annulatus, and Eucinostomus dowii. The water temperature changed seasonally, being warm from July through December and cold from January through June. We found more fish species during the warm season than during the cold season. The southern area of the coastal lagoon had the highest diversity and species richness. The small size of the fishes registered confirms the ecological role of coastal lagoons as nursery areas that offer protection and feeding to commercially important fish near Bahía Magdalena, Mexico.Se determinó la estructura de peces asociada a factores ambientales en una laguna costera con manglar denominada "Rancho Bueno". Se utilizó una red de arrastre experimental para la captura y se identificaron 62 especies de peces de 48 géneros y 30 familias. Las especies más importantes fueron Etropus crossotus, Eucinostomus gracilis, Paralabrax maculatofasciatus, Sphoeroides annulatus y Eucinostomus dowii. La temperatura del agua varió estacionalmente, siendo cálida de julio a diciembre y fría de enero a junio. Se registró un mayor número de especies de peces durante la época cálida comparada con la época fría. La zona sur de la laguna costera presentó una mayor diversidad y riqueza específica.     There is still a significant amount of patients who do not sufficiently respond to cardiac resynchronization therapy (CRT). Previous studies demonstrated that the amount of dyssynchronous myocardium was predictive of response to CRT. Otherwise, non-response is frequently associated with high amounts of scar tissue. The combination of these parameters might yield a more accurate prediction of response. We hypothesized that the probability of a CRT response increases with the presence of high amounts of ''viable and dyssynchronous'' myocardium. A total of 19 patients (17 male, 61 {+-} 10 years) underwent ECG-gated [{sup 18}F]fluorodeoxyglucose (FDG) myocardial positron emission tomography (PET) before CRT device implantation and were followed for 6 months. Response to CRT was defined as clinical improvement of at least one New York Heart Association (NYHA) class in combination with left ventricular (LV) ejection fraction (EF) improvement of >5 %. Twelve responders (71 %) and seven non-responders (29 %) were identified. For each patient bullseye maps of FDG uptake and phase analysis were calculated (QPS/QGS 2012, Cedars-Sinai, Los Angeles, CA, USA) and fused. Amounts of myocardium representing ''viable and synchronous'', ''scar and synchronous'', viable and dyssynchronous or ''scar and dyssynchronous'' myocardium were quantified by planimetric measurements of the fused bullseye maps. Responders by definition showed significant decrease in NYHA class and significant increase of LVEF. Furthermore, a significantly higher amount of viable and dyssynchronous myocardium was found as compared to non-responders (21 {+-} 13 % vs 6 {+-} 5 %; p < 0.05). Combined assessment of myocardial viability and LV dyssynchrony is feasible using multiparametric [{sup 18}F]FDG PET and could improve conventional response prediction criteria for CRT. (orig.)

    The article by Puffer et al in this month's JABFM confirms a high burnout rate (25%) among family physicians renewing their credentials, with a higher rate among young and female doctors. Recent reports confirm high burnout rates among general internists. Thus, mechanisms to monitor and improve worklife in primary care are urgently needed. We describe the Mini Z (for "zero burnout program") measure, designed for these purposes, and suggest interventions that might improve satisfaction and sustainability in primary care, including longer visits, clinician control of work schedules, scribe support for electronic medical record work, team-based care, and an explicit emphasis on work-home balance. © Copyright 2017 by the American Board of Family Medicine.

    Spatial and temporal profiles of the space potential in the central-cell midplane of TMX have been obtained with a heavy-ion-beam probe. The absolute accuracy of measurements is +- 25 volts (with respect to the machine vacuum walls) with a resolution of approx. 2 volts. During moderate fueling with the gas boxes (i/sub gas/ approx. = 1200 Atom-Amperes D/sub 2/), the plasma potential is parabolic to at least 25 cm radius, with phi/sub e/ approx. = phi/sub max/(1-(r/32)/sup 2/) and 300 < phi/sub max/ <450 volts. With puffer-valve fueling, the space potential is relatively flat to at least 27 cm radius, with 250 < phi/sub e/ < 350 volts.

  1. Trophic transference of microplastics under a low exposure scenario: Insights on the likelihood of particle cascading along marine food-webs.

    Microplastics are emergent pollutants in marine environments, whose risks along food-web still need to be understood. Within this knowledge gap, MPs transference and persistence along trophic levels are key processes. We assessed the potential occurrence of these processes considering a less extreme scenario of exposure than used previously, with microplastics present only in the hemolymph of prey (the mussel Perna perna) and absent in the gut cavity. Predators were the crab Callinectes ornatus and the puffer fish Spheoeroides greeleyi. Transference of microplastics occurred from prey to predators but without evidences of particle persistence in their tissues after 10days of exposure. This suggests a reduced likelihood of trophic cascading of particles and, consequently, a reduced risk of direct impacts of microplastics on higher trophic levels. However, the contact with microplastics along food-webs is still concerning, modulated by the concentration of particles in prey and predators' depuration capacity and rate. Copyright © 2017.     Results of longitudinal studies in developmental psychology have shown the importance of protective factors especially within high risk samples for adaptive development as they serve as puffers against many risks. An outstanding protective factor is a secure parent-child attachment relationship. This article outlines the different ways how attachment relationships can serve as protective mechanisms and especially how disorganized attachment relationships are leading to maladaptation and hostile attitudes and behaviors. In doing so it underscores the special importance of attachment experiences in early infancy. The concept of attachment relationship is described as opposed to the bonding concept with regard to its implications for practice. One of the first is to intervene early. In the final part the evidence based early intervention program "STEEP" is described as an application of attachment research on a poverty sample and as a response to violence.

    Full Text Available Tetrodotoxin (TTX is a naturally occurring toxin that has been responsible for human intoxications and fatalities. Its usual route of toxicity is via the ingestion of contaminated puffer fish which are a culinary delicacy, especially in Japan. TTX was believed to be confined to regions of South East Asia, but recent studies have demonstrated that the toxin has spread to regions in the Pacific and the Mediterranean. There is no known antidote to TTX which is a powerful sodium channel inhibitor. This review aims to collect pertinent information available to date on TTX and its analogues with a special emphasis on the structure, aetiology, distribution, effects and the analytical methods employed for its detection.

    This paper describes the study of of uranium (VI) (2.5 10{sup -4}M) extraction from acetate puffer solutions with 0.01 M solution of tenoil trifluoroacetonate in benzene, chloroform, carbon tetrachloride, amyl acetate, cyclohexanon, n-butyl alcohol with the additions of 0.036 M TBP and 2.7 M acetone solution with 0.01 M solution of TTA. It was verified that the extraction depends on the property of the organic phase. Distribution coefficients for uranium when extracted by solvents containing oxygen is higher compared to the distribution coefficients obtained by solvents without oxygen (benzene, chloroform and carbon tetrachloride). TBP cyclohexanon, and acetone additions in the CCl{sub 4} cause more significant increase of distribution coefficients.

    Full Text Available The aim of this study was to determine the effect of the skin preservation type against shrinkage temperature of leather. The material used in this study was the skin of pufferfish (Arothon reticularis that have been preserved by salting, formaldehyde and pickling and also raw skin as a reference. The method used to measure the shrinkage temperature was thermal analysis using Differential Scanning Calorimeter (DSC that operated from 4°C up to 440°C with nitrogen stream. DSC measurement results showed that shrinkage temperature of puffer fish preserved with formaldehyde was higher than salting and pickling, which is 63.64°C; 47.95°C; 57.37oC respectively. The advantage of using formaldehyde compared to others preservation technique was not only can protect the skin from damage by microorganisms, but also can create a bond with the collagen .

    Full Text Available This paper seeks to contribute to the study of the vernacularisation process in late Middle English by measuring up to what an extent concrete and abstract noun suffixes (in line with Dalton-Puffer 1996 attach to either Germanic or Romance bases in the medical texts extracted from the MEMT (Middle English Medical Texts corpus. The findings obtained have been further described according to text type or genre and to target audience/readership. The description of these suffixes in relation to all the parameters already mentioned has confirmed the predominance of abstract suffixes of Romance origin although Germanic abstract suffixes are also abundant. More hybrid formations have been found with Germanic noun suffixes than with Romance ones which might be indicative of their versatility towards vernacularisation.

    Full Text Available Tetrodotoxin (TTX is a naturally occurring toxin in food, especially in puffer fish. TTX poisoning is observed frequently in South East Asian regions. In TTX-derived food poisoning outbreaks, the amount of TTX recovered from suspicious fish samples or leftovers, and residual levels from biological fluids of victims are typically trace. However, liquid chromatography–mass spectrometry and liquid chromatography–tandem mass spectrometry methods have been demonstrated to qualitatively and quantitatively determine TTX in clinical samples from victims. Identification and validation of the TTX-originating seafood species responsible for a food poisoning incident is needed. A polymerase chain reaction-based method on mitochondrial DNA analysis is useful for identification of fish species. This review aims to collect pertinent information available on TTX-borne food poisoning incidents with a special emphasis on the analytical methods employed for TTX detection in clinical laboratories as well as for the identification of TTX-bearing species.

    Strong magnetic field shear around the divertor x-point is numerically predicted to lead to strong spatial asymmetries in turbulence driven particle fluxes. To visualize the turbulence and associated impurity line emission near the lower x-point region, a new tangential observation port has been recently installed on NSTX. A reentrant sapphire window with a moveable in-vessel mirror images the divertor region from the center stack out to R 80 cm and views the x-point for most plasma configurations. A coherent fiber optic bundle transmits the image through a remotely selected filter to a fast camera, for example a 40500 frames/sec Photron CCD camera. A gas puffer located in the lower inboard divertor will localize the turbulence in the region near the x-point. Edge fluid and turbulent codes UEDGE and BOUT will be used to interpret impurity and deuterium emission fluctuation measurements in the divertor

    By 2020, more than one million electric-powered vehicles are envisaged on German roads. But when will they be commercially available? How much power do they really consume? Will there be batteries with sufficient capacity? And wouldn't it be more sensible to make conventional cars more efficient? These and other questions about electric-powered cars and electromobility are answered by the authors, competently and in a manner that also addresses the non-expert public. It is shown that electric-powered cars will be operated in fleets, serving not only as vehicles but also as buffers for renewables-derived power, which is known to be volatile. Controlled charging will offer a solution. Professionally managed fleets of electric-powered vehicles can store excess wind power, or they could serve as buffers at noon when more solar power is generated than is required in factories and kitchens.     A decrease in the arterial blood saturation by oxygen in patients with POChP is a frequent phenomenon. It is more serious in patients type blue boaters and less frequent among patients type pink puffers. The aim of the paper was to compare the arterial blood saturation by oxygen in the groups examined during two nights: during the first night the patients breathed atmospheric air whereas during the second night they were given oxygen. The author also studied the influence of oxygenation of an organism on the frequency of cardiac rhythm disorders (ZRS). The author examined a group of 20 patients with the predominance of chronic bronchitis--blue boaters (average VC was 1.95 l, FEV1--0.81 l, PaO2 while breathing atmospheric air 52 mm Hg and 68 mm Hg after giving oxygen, PaCO2 47 and 51 mm Hg respectively) and 20 patients with the predominance of emphysema--pink puffers (average VC--2.30 l, FEV1--0.86 l, PaO2 while breathing atmospheric air 60 mm Hg and 70 mm Hg after giving oxygen, PaCO2 39 and 40 mm respectively). It was found that the patients with heavy hypoxaemia and hypercapnia had worse arterial blood saturation by oxygen during the two nights of investigation in comparison with the other group. The author also found more frequent cardiac rhythm disorders in this group of patients. Giving oxygen improved blood oxygenation in the two groups and lowered the frequency of cardiac rhythm disorders. The results obtained indicate to the need of oxygen therapy in patients with advanced POChP, especially during the night so as to avoid nocturnal hypoxaemia of an organism.

  13. Transcriptome analysis of artificial hybrid pufferfish Jiyan-1 and its parental species: implications for pufferfish heterosis.

    Full Text Available Jiyan-1 puffer, the F1 hybrid of Takifugu rubripes and Takifugu flavidus, displays obvious heterosis in the growth performance, flavor and stress tolerance. In the present study, comparative analysis for the transcriptomes of T. rubripes, T. flavidus and Jiyan-1 was performed aiming to reveal the possible mechanisms of heterosis in pufferfish. Whole transcriptomes were sequenced using the SOLiD4 platform, and a total of 44,305 transcripts corresponding to 18,164 genes were identified collectively. A total of 14,148 transcripts were differentially expressed. By comparing the gene expression patterns of the three samples, the coexistence of overdominance, dominance, underdominance and additivity was observed in the gene action modes of Jiyan-1. There were 2,237 transcripts in the intersection of the differentially expressed transcripts from Jiyan-1 versus T. rubripes and Jiyan-1 versus T. flavidus, among which 213 transcripts were also in the T. rubripes versus T. flavidus. The potential functions of the remaining 2,024 transcripts were mainly associated with metabolic process, nucleotide binding and catalytic activity. The enrichment results indicated metabolism was the most activated biological function in the heterosis. In addition, 35 KEGG pathways were retrieved as affiliated with more than three differentially expressed transcripts and 8,579 potentially novel transcript isoforms were identified for Jiyan-1. The present study revealed the coexistence of multiple gene actions in the hybrid puffer, indicated the importance of metabolism, ion binding function and kinase activity, as well as provided a list of candidate genes and pathways for heterosis. It could be helpful for the better understanding of the determination and regulation mechanisms of heterosis.

    We describe three new species of blood flukes (Aporocotylidae) and propose their classification within the genus Psettarium Goto & Ozaki, 1929. All three species were collected from the circulatory systems of pufferfishes caught off Bali, central Indonesia. Psettarium pulchellum n. sp. was found in the gills of both the narrow-lined puffer (Arothron manilensis de Procé) and the spiny blaasop (Tylerius spinosissimus Regan), while P. ogawai n. sp. and P. jimbaranense n. sp. were found in the gills of the reticulated puffer (Arothron reticularis Bloch & Schneider). The morphological characteristics of these taxa necessitated emendation of the diagnosis for the genus Psettarium, to accommodate the presence of an oral sucker, multiple or entirely post-caecal testes and a degenerate posterior testis. Features such as proportion of body length occupied by the oesophagus, and posterior caeca being ≥7× the length of anterior caeca, are no longer regarded as useful genus-level characters. Additionally, Sasala nolani is reassigned to this genus as Psettarium nolani n. comb. In phylogenetic analyses of the 28S and ITS2 rDNA regions, all three new taxa form a well-supported clade, together with Psettarium sinense and Psettarium nolani n. comb., the two other species of tetraodontid-infecting aporocotylids for which comparative rDNA data were available. The short branch lengths within this clade, despite dramatic morphological differences between the five species, suggest that rapid morphological diversification has occurred among the tetraodontid-infecting aporocotylids. The genus Psettarium has long been considered problematic. Further commentary is given on the history of this genus and how the issues presented might be resolved.     Full Text Available An updated inventory of alien marine species from coastal and offshore waters of Cyprus is presented. Records were compiled based on the existing scientific and grey literature, including HCMR database of Mediterranean alien species, technical reports, scientific congresses, academic dissertations, and websites, as well as on unpublished/personal observations. The listed species were classified in one of five categories: established, invasive, casual, cryptogenic, and questionable. The mode of introduction and the year of first sighting were also reported for each species. Eight new records based on personal observations of the authors were reported (Chondria coerulescens, Neosiphonia sphaerocarpa, Enchelycore anatina, Lagocephalus spadiceus, Lagocephalus suezensis, Scomberomorus commerson, Sillago sihama, and Sphoeroides pachygaster. Nine species, previously reported as aliens in Cypriot waters, were excluded from the inventory for various reasons. Ten established species were characterized as invasive (Caulerpa racemosa var. cylindracea, Cerithium scabridum, Strombus persicus, Trochus erythraeus, Brachidontes pharaonis, Pinctada radiata, Fistularia commersonii, Lagocephalus sceleratus, Siganus luridus, and Siganus rivulatus as they have a substantial impact on biodiversity and/or local economy. The impact of alien marine species in Cyprus is expected to grow in the close future, and further effort directed towards recording alien invasions and their impact will be needed.

    Steroid hormones produce adverse effects on biota as well as bioaccumulation in fish and seafood, making it necessary to develop methodologies to evaluate these compounds in samples related to the food chain. This work presents an analytical method for evaluating 15 steroid hormones in fish tissue. It is based on microwave-assisted extraction and solid-phase extraction coupled to ultra-high-performance liquid chromatography tandem mass spectrometry (MAE-SPE-UHPLC-MS/MS). The proposed method shows appropriate detection limits (0.14-49.0ngg -1 ), recoveries in the range of 50% and good repeatability. After optimization, the method was applied to different tissues from two small fishes of the Canary Islands that constitute an important level of the food web (Boops boops and Sphoeroides marmoratus) and were exposed to the outfall of the Las Palmas de Gran Canaria wastewater treatment plant.   17. A critical review of records of alien marine species from the Maltese Islands and surrounding waters (Central Mediterranean

    Full Text Available An updated list of alien marine species recorded from the Maltese Islands and surrounding waters, compiled from scientific and ‘grey’ literature and from authenticated unpublished reports to the authors, is presented. The listed species are classified in one of four categories as regards establishment status: established, casual, invasive and questionable. Doubtful records are listed as ‘?’. A total of 48 species, including nine dubious ones, are included in the list. Of the accepted records, 64% are established, of which 15.4% are invasive, 18% are casual and 18% are questionable. The most represented groups are molluscs (14 species, fish (13 species and macrophytes (10 species. Six species are classified as invasive in Maltese waters: Lophocladia lallemandii, Womersleyella setacea, Caulerpa racemosa var. cylindracea, Percnon gibbesi, Fistularia commersonii and Sphoeroides pachygaster; impacts of some of these species on local ecosystems are discussed. Since the early 1900s, there has been an increasing trend in the number of alien marine species reported from the Maltese Islands. Transportation via shipping and in connection with aquaculture, as well as the range expansion of Lessepsian immigrants, appear to be the most common vectors for entry, accounting for 20%, 11% and 32% respectively of the alien species included in this review. The general warming trend of Mediterranean waters and increasing marine traffic may be facilitating the spread of warm-water Atlantic and Indo-Pacific species to the central Mediterranean, including the Maltese Islands.

  18. Fish community structure and dynamics in a coastal hypersaline lagoon: Rio Lagartos, Yucatan, Mexico

    Rio Lagartos, a tropical coastal lagoon in northern Yucatan Peninsula of Mexico, is characterized by high salinity during most of the year (55 psu annual average). Even though the area has been designated as a wetland of international importance because of its great biodiversity, fish species composition and distribution are unknown. To determine whether the salinity gradient was influencing fish assemblages or not, fish populations were sampled seasonally by seine and trawl from 1992 to 1993 and bimonthly during 1997. We identified 81 fish species, eight of which accounted for 53.1% considering the Importance Value Index ( Floridichthys polyommus, Sphoeroides testudineus, Eucinostomus argenteus, Eucinostomus gula, Fundulus majalis, Strongylura notata, Cyprinodon artifrons and Elops saurus). Species richness and density declined from the mouth to the inner zone where extreme salinity conditions are prominent (>80) and competitive interactions decreased. However, in Coloradas basin (53 average sanity) and in the inlet of the lagoon, the highest fish density and number of species were observed. Greater habitat heterogeneity and fish immigration were considered as the best explanation. Multivariate analysis found three zones distinguished by fish occurrence, abundance and distribution. Ichthyofaunal spatial differences were attributed to selective recruitment from the Gulf of Mexico due to salinity gradient and to changing climatic periods. Estuarine and euryhaline marine species are abundant, with estuarine dependent ones entering the system according to environmental preferences. This knowledge will contribute to the management of the Special Biosphere Reserve through baseline data to evaluate environmental and anthropogenic changes.

    Full Text Available Although pufferfish of the family Tetraodontidae contain high levels of tetrodotoxin (TTX mainly in the liver, some species of pufferfish, boxfish of the family Ostraciidae, and porcupinefish of the family Diodontidae do not. To clarify the mechanisms, uptake of TTX and saxitoxins (STXs into liver tissue slices of pufferfish, boxfish and porcupinefish was examined. Liver tissue slices of the pufferfish (toxic species Takifugu rubripes and non-toxic species Lagocephalus spadiceus, L. cheesemanii and Sphoeroides pachygaster incubated with 50 µM TTX accumulated TTX (0.99–1.55 µg TTX/mg protein after 8 h, regardless of the toxicity of the species. In contrast, in liver tissue slices of boxfish (Ostracion immaculatus and porcupinefish (Diodon holocanthus, D. liturosus, D. hystrix and Chilomycterus reticulatus, TTX content did not increase with incubation time, and was about 0.1 µg TTX/mg protein. When liver tissue slices were incubated with 50 µM STXs for 8 h, the STXs content was <0.1 µg STXs/mg protein, irrespective of the fish species. These findings indicate that, like the toxic species of pufferfish T. rubripes, non-toxic species such as L. spadiceus, L. cheesemanii and S. pachygaster, potentially take up TTX into the liver, while non-toxic boxfish and porcupinefish do not take up either TTX or STXs.

A total of 11,187 individuals distributed in 32 families and 63 species were collected. The most numerically abundant species were Floridichthys polyommus and Cyprinodon artifrons, while Sphoeroides testudineus contributed to the greatest biomass. Species composition consisted mainly of estuarine and euryhaline marine species. Spatially, a saline gradient was observed with marine conditions in the mouth, and increasing to over 100 in the inner zone of the system. Species richness, diversity and biomass declined from the mouth to the inner zone, while density showed an inverse tendency, with the highest values in the inner zone. Thus the salinity was the variable that best explained the spatial fish assemblages" structure. The ichthyofauna composition did not change over time, but the dominant species varied with the years. The abundance of juvenile specimens, suggest that the different habitats are used as feeding and breeding zones; hence it is proposed that protection strategies be pursued not only for the lagoon system but also for the northern zone of the Yucatan Peninsula.

    A passive plasmonics based chemical sensing system to be used in harsh operating environments was investigated and developed within this program. The initial proposed technology was based on combining technologies developed at the SUNY Polytechnic Institute Colleges of Nanoscale Science and Engineering (CNSE) and at the University of Minnesota (UM). Specifically, a passive wireless technique developed at UM was to utilize a heat-activated plasmonic design to passively harvest the thermal energy from within a combustion emission stream and convert this into a narrowly focused light source. This plasmonic device was based on a bullseye design patterned into a gold film using focused ion beam methods (FIB). Critical to the design was the use of thermal stabilizing under and overlayers surrounding the gold film. These stabilizing layers were based on both atomic layer deposited films as well as metal laminate layers developed by United Technologies Aerospace Systems (UTAS). While the bullseye design was never able to be thermally stabilized for operating temperatures of 500oC or higher, an alternative energy harvesting design was developed by CNSE within this program. With this new development, plasmonic sensing results are presented where thermal energy is harvested using lithographically patterned Au nanorods, replacing the need for an external incident light source. Gas sensing results using the harvested thermal energy are in good agreement with sensing experiments, which used an external incident light source. Principal Component Analysis (PCA) was used to reduce the wavelength parameter space from 665 variables down to 4 variables with similar levels of demonstrated selectivity. The method was further improved by patterning rods which harvested energy in the near infrared, which led to a factor of 10 decrease in data acquisition times as well as demonstrated selectivity with a reduced wavelength data set.     Purpose: Recent advances in preclinical radiotherapy systems have provided the foundation for scaling many of the elements of clinical radiation therapy practice to the dimensions and energy demanded in small animal studies. Such systems support the technical capabilities to accurately deliver highly complex dose distributions, but methods to optimize and deliver such distributions remain in their infancy. This study developed an optimization method based on empirically measured two-dimensional dose kernel measurements to deliver arbitrary planar dose distributions on a recently developed small animal radiotherapy platform.Methods: A two-dimensional dose kernel was measured with repeated radiochromic film measurements for the circular 1 mm diameter fixed collimator of the small animal radiotherapy system at 1 cm depth in a solid water phantom. This kernel was utilized in a sequential quadratic programming optimization framework to determine optimal beam positions and weights to deliver an arbitrary desired dose distribution. The positions and weights were then translated to a set of stage motions to automatically deliver the optimized dose distribution. End-to-end efficacy of the framework was quantified through five repeated deliveries of two dosimetric challenges: (1) a 5 mm radius bullseye distribution, and (2) a “sock” distribution contained within a 9 × 13 mm bounding box incorporating rectangular, semicircular, and exponentially decaying geometric constructs and a rectangular linear dose gradient region. These two challenges were designed to gauge targeting, geometric, and dosimetric fidelity.Results: Optimization of the bullseye and sock distributions required 2.1 and 5.9 min and utilized 50 and 77 individual beams for delivery, respectively.     Full Text Available Coastal lagoons are important systems for freshwater, estuarine and marine organisms; they are considered important zones of reproduction, nursery and feeding for many fish species. The present study investigates the fish assemblages of the natural reserve of Dzilam and their relationship with the hydrologic variables. A total of 6 474 individuals (81 species were collected, contributing with more than 50% considering the Importance Value Index (IVI, Sphoeroides testudineus, Fundulus persimilis, Anchoa mitchilli, Eucinostomus gula, Eucinostomus argenteus and Mugil trichodon. Differences in species composition were found between seasons the highest during the cold fronts. Spatially, differences were related with the presence of freshwater seeps, the highest in the ecological characterized eastern part and the lowest with higher difference in specific composition located in the western part of the internal zone, due to a higher abundance and dominance of L. rhomboides. Salinity and temperature were the variables that presented a higher influence in the distribution of some pelagic species such as A. mitchilli and A. hepsetus. Because of the abundant freshwater seeps characteristic of the coastal lagoons of Yucatan Peninsula their community structure and fish assemblage display spatial and temporal differences in specific composition.     Acanthocephalus amini n. sp. (Palaeacanthocephala: Echinorhynchidae) is described from the intestine of Cichlasoma urophthalmus (Günther) (Pisces: Cichlidae) collected in the Río Champotón, a river in Campeche State, Mexico. It is the fourth species of Acanthocephalus Koelreuther, 1771 described from North American freshwater fishes, although two other species are known from South America. The new species is distinguished from other members of Acanthocephalus by features of its trunk, which is small, clavate, slightly expanded medially and bluntly pointed posteriorly. It is further distinguished by having a cylindrical proboscis armed with 13-14 longitudinal rows of 11-12 stout hooks; the apical and medial proboscis hooks are almost uniform in size and shape, decreasing in size towards the base; the posteriormost hooks are smaller, straighter and more slender than the anterior and middle hooks; and the lateral rows of hooks are more widely spaced, forming a conspicuous longitudinal area devoid of hooks. Furthermore, the lemnisci are saccate and shorter than the proboscis receptacle; and the neck is very short with a thick collar of trunk tegument, which encircles the base of proboscis. In males, the testes are in the middle third of trunk, diagonal, spherical and small relative to the body size, and there are six clavate cement glands. In females, the uterus forms a conspicuous, elongate, cylindrical egg reservoir. The new species is most similar to A. alabamensis Amin & Williams, 1983, but can be distinguished by its swollen, clavate trunk, the largest proboscis hooks being present apically and medially, smaller testes, a shorter male reproductive system relative to body size and females with a prominent uterus. They have different hosts and geographical distribution. The new species can be differentiated from Brasacanthus sphoeroides Thatcher, 2001, a similar species in a monotypic echinorhynchid genus, because the latter is larger, has smaller proboscis hooks

    Full Text Available An updated review of marine alien species and other ‘newcomers’ recorded from the Maltese Islands is presented on account of new records and amendments to a previous review in 2007. Species were classified according to their establishment status (‘Questionable’, ‘Casual’, ‘Established’, ‘Invasive’ and origin (‘Alien’, ‘Range expansion’, ‘Cryptogenic’. A total of 31 species were added to the inventory, while 6 species have been removed, bringing the total number of species to 73. Of these, 66 are considered to be aliens (or putative aliens but with uncertain origin with the remaining 7 resulting from range expansion. Six records are considered to be questionable and hence unverified. For verified records, the dominant taxonomic groups are Mollusca (represented by 21 species and Actinopterygii (15 species, followed by Crustacea (8 species and Rhodophyta (7 species. Eight of these species (aliens: Caulerpa cylindracea, Lophocladia lallemandi, Womersleyella setacea, Brachidontes pharaonis, Percnon gibbesi, Fistularia commersonii, Siganus luridus; range extender: Sphoeroides pachygaster are considered to be invasive. The introduction pathway for 30 species is unknown. Amongst the alien species, ‘Shipping’ is the most common introduction pathway, followed by ‘Secondary dispersal’ from elsewhere in the Mediterranean Sea. An increasing trend in the number of alien marine species reported from the Maltese Islands is evident, with a peak of 22 species recorded during the last decade (2001–2010. A discussion on the rationale for including range-expanding species in national inventories of recent arrivals, and in the analysis of trends in records from the Maltese Islands, is included.     Human activities have strong impacts on coastal ecosystems functioning through their effect on primary organic sources distributions and resulting biodiversity. Hence, it appears to be of utmost importance to quantify contribution of primary producers to sediment organic matter (SOM) spatial variability and its associated ichthyofauna. The Terminos lagoon (Gulf of Mexico) is a tropical estuary severely impacted by human activities even though of primary concern for its biodiversity, its habitats, and its resource supply. Stable isotope data (d13C, d15N) from mangrove, seaweed, seagrass, phytoplankton, ichthyofauna and SOM were sampled in four zones of the lagoon and the continental shelf through windy (November to February), dry (March to June) and rainy (July to October) seasons. Stable Isotope Analysis in R (SIAR) mixing model were used to determine relative contributions of the autotrophic sources to the ichthyofauna and SOM. Analysis of variance of ichthyofauna isotopic values showed significant differences (P < 0.001) in the four zones of lagoon despite the variability introduced by the windy, dry and rainy seasons. In lagoons rivers discharge zone, the mangrove contribution to ichthyofauna was 40% and 84% to SOM. Alternative use of habitat by ichthyofauna was evidenced since in the deep area of the lagoon (4 m), the contribution of mangrove to fish is 50%, and meanwhile contribution to SOM is only 77%. Although phytoplankton (43%) and seaweed (41%) contributions to the adjacent continental shelf ichthyofauna were the main organic sources, there was 37% mangrove contribution to SOM, demonstrating conspicuous terrigenous influence from lagoon ecosystem. Our results point toward organic sources spatial variations that regulate fish distribution. In Terminos lagoon, significant correlation (p-value = 0.2141 and r=0.79) of Ariopsis felis and Sphoeroides testudineus abundances and seaweed and seagrasses contributions (30-35%) during both dry and rainy seasons

    Involvement of the myocardium in non-infectious myopathies presents in most cases as systolic dysfunction or a disturbed cardiac rhythm. We are interested in exploring how often cardiac involvement can be evaluated with various diagnostic techniques in patients with proven myopathy. We investigated 41 patients with myopathies of various etiology, including mitochondrial and congenital myopathies, Curshmann-Steinert disease, muscular dystrophy, and others. Myopathy was proven by muscular biopsy usually from the bicep. Fatty acid imaging was performed with 15-(p-[I-123]iodophenyl)pentadecanoic acid (IP-PA) and sequential SPECT-scintigraphy with a 180 deg. rotation starting at the 45 deg. RAO position. 190 MBq were injected at the maximal stage of a submaximal exercise. Filtered backprojection and reorientation of the slices were achieved by standard techniques. The quantitative comparison of the oblique slices (bulls-eye technique) of the SPECT-studies revealed turnover-rates as a qualitative measure of {beta}-oxidation. Serum levels of lactate (L), pyruvate (P), glucose (G) and triglycerides (TG) were measured at rest and stress. Ventricular function was investigated by radionuclide ventriculography (MUGA) at rest and under stress with Tc-99m labeled red blood cells. In addition, ECG, 24 hour-ECG, and echocardiography were also performed with standard techniques.

    Isobaric mass tagging (IMT) methods enable the analysis of thousands of proteins simultaneously. We used tandem mass tagging reagents (TMT™) to monitor the relative changes in the proteome of the mouse macrophage cell line RAW264.7 at the same six time points after no stimulation (baseline phenotype), stimulation with interferon gamma (pro-inflammatory phenotype) or stimulation with interleukin-4 (anti-inflammatory phenotype). The combined TMT datasets yielded nearly 12,000 protein profiles for comparison. To facilitate this large analysis, we developed a novel method that combines or multiplexes the separate IMT (mIMT) datasets into a single super dataset for subsequent model-based clustering and co-regulation analysis. Specially designed visual High Throughput Screening (visHTS) software screened co-regulated proteins. visHTS generates an interactive and visually intuitive color-coded bullseye plot that enables users to browse the cluster outputs and identify co-regulated proteins.     We present a comprehensive analysis of spatially resolved moderate spectral resolution near-infrared spectra obtained with the adaptive optics system at the Keck Observatory. We identify three compositionally distinct end member regions: the trailing hemisphere bullseye, the leading hemisphere upper latitudes, and a third component associated with leading hemisphere chaos units. We interpret the composition of the three end member regions to be dominated by irradiation products, water ice, and evaporite deposits or salt brines, respectively. The third component is associated with geological features and distinct from the geography of irradiation, suggesting an endogenous identity. Identifying the endogenous composition is of particular interest for revealing the subsurface composition. However, its spectrum is not consistent with linear mixtures of the salt minerals previously considered relevant to Europa. The spectrum of this component is distinguished by distorted hydration features rather than distinct spectral features, indicating hydrated minerals but making unique identification difficult. In particular, it lacks features common to hydrated sulfate minerals, challenging the traditional view of an endogenous salty component dominated by Mg-sulfates.   12. Diagnosis of myocardial involvement in patients with systemic myopathies with 15-(p-[I-123]iodophenyl) pentadecanoic acid (IPPA) SPECT

    Involvement of the myocardium in non-infectious myopathies presents in most cases as systolic dysfunction or a disturbed cardiac rhythm. We are interested in exploring how often cardiac involvement can be evaluated with various diagnostic techniques in patients with proven myopathy. We investigated 41 patients with myopathies of various etiology, including mitochondrial and congenital myopathies, Curshmann-Steinert disease, muscular dystrophy, and others. Myopathy was proven by muscular biopsy usually from the bicep. Fatty acid imaging was performed with 15-(p-[I-123]iodophenyl)pentadecanoic acid (IP-PA) and sequential SPECT-scintigraphy with a 180 deg. rotation starting at the 45 deg. RAO position. 190 MBq were injected at the maximal stage of a submaximal exercise. Filtered backprojection and reorientation of the slices were achieved by standard techniques. The quantitative comparison of the oblique slices (bulls-eye technique) of the SPECT-studies revealed turnover-rates as a qualitative measure of β-oxidation. Serum levels of lactate (L), pyruvate (P), glucose (G) and triglycerides (TG) were measured at rest and stress. Ventricular function was investigated by radionuclide ventriculography (MUGA) at rest and under stress with Tc-99m labeled red blood cells.   13. Current applications of high-resolution mass spectrometry for the analysis of new psychoactive substances: a critical review.

    The proliferation of new psychoactive substances (NPS) in recent years has resulted in the development of numerous analytical methods for the detection and identification of known and unknown NPS derivatives. High-resolution mass spectrometry (HRMS) has been identified as the method of choice for broad screening of NPS in a wide range of analytical contexts because of its ability to measure accurate masses using data-independent acquisition (DIA) techniques. Additionally, it has shown promise for non-targeted screening strategies that have been developed in order to detect and identify novel analogues without the need for certified reference materials (CRMs) or comprehensive mass spectral libraries. This paper reviews the applications of HRMS for the analysis of NPS in forensic drug chemistry and analytical toxicology. It provides an overview of the sample preparation procedures in addition to data acquisition, instrumental analysis, and data processing techniques. Furthermore, it gives an overview of the current state of non-targeted screening strategies with discussion on future directions and perspectives of this technique.   14. Integrating remote sensing techniques at Cuprite, Nevada: AVIRIS, Thematic Mapper, and field spectroscopy

    The Cuprite mining district in southwestern Nevada has become a test site for remote sensing studies with numerous airborne scanners and ground sensor data sets collected over the past fifteen years. Structurally, the Cuprite region can be divided into two areas with slightly different alteration and mineralogy. These zones lie on either side of a postulated low-angle structural discontinuity that strikes nearly parallel to US Route 95. Hydrothermal alternation at Cuprite was classified into three major zones: silicified, opalized, and argillized. These alteration types form a bulls-eye pattern east of the highway and are more linear on the west side of the highway making a striking contrast from the air and the imagery. Cuprite is therefore an ideal location for remote sensing research as it exhibits easily identified hydrothermal zoning, is relatively devoid of vegetation, and contains a distinctive spectrally diagnostic mineral suite including the ammonium feldspar buddingtonite, several types of alunite, different jarosites, illite, kaolinite, smectite, dickite, and opal. This present study brings a new dimension to these previous remote sensing and ground data sets compiled for Cuprite. The development of a higher resolution field spectrometer now provides the capability to combine extensive in-situ mineralogical data with a new geologic field survey and detailed Airborne Visible/Infrared Imaging Spectrometers (AVIRIS) images. The various data collection methods and the refinement of the integrated techniques are discussed.

  15. Patterns of species richness and the center of diversity in modern Indo-Pacific larger foraminifera.

    Symbiont-bearing Larger Benthic Foraminifera (LBF) are ubiquitous components of shallow tropical and subtropical environments and contribute substantially to carbonaceous reef and shelf sediments. Climate change is dramatically affecting carbonate producing organisms and threatens the diversity and structural integrity of coral reef ecosystems. Recent invertebrate and vertebrate surveys have identified the Coral Triangle as the planet's richest center of marine life delineating the region as a top priority for conservation. We compiled and analyzed extensive occurrence records for 68 validly recognized species of LBF from the Indian and Pacific Ocean, established individual range maps and applied Minimum Convex Polygon (MCP) and Species Distribution Model (SDM) methodologies to create the first ocean-wide species richness maps. SDM output was further used for visualizing latitudinal and longitudinal diversity gradients. Our findings provide strong support for assigning the tropical Central Indo-Pacific as the world's species-richest marine region with the Central Philippines emerging as the bullseye of LBF diversity. Sea surface temperature and nutrient content were identified as the most influential environmental constraints exerting control over the distribution of LBF. Our findings contribute to the completion of worldwide research on tropical marine biodiversity patterns and the identification of targeting centers for conservation efforts.

  16. Direct and Conceptual Replications of Burgmer & Englich (2012: Power May Have Little to No Effect on Motor Performance.

    Full Text Available Burgmer and Englich (2012 have reported that manipulating feelings of power can substantially improve performance on two motor tasks: golf and darts. We conducted two high-powered direct replications of the effects of power on golf, two online conceptual replications using mirror-tracing as a performance measure, and an additional conceptual replication using a cognitive performance measure (word-search. Overall, we found little to no effect of power on motor skill (d = 0.09, 95% CI[-0.07, 0.22], n = 603. We varied task difficulty, re-analyzed data without participants showing weak responses on manipulation checks, and tried adjusting performance scores for age, gender, and initial task skill. None of these secondary analyses revealed a strong effect of power on performance. A meta-analysis integrating our data with Burgmer & Englich leaves open the possibility that manipulating power could provide a modest boost in motor skill (d = 0.19, 95% CI [0.001, 0.38], n = 685. Unfortunately, the pattern of performance changes we observed was unrelated to group differences in perceived and rated power, suggesting that what motor effects do occur with this protocol may not be directly related to the construct of power.   18. Proteus mirabilis and Urinary Tract Infections.

    Proteus mirabilis is a Gram-negative bacterium and is well known for its ability to robustly swarm across surfaces in a striking bulls'-eye pattern. Clinically, this organism is most frequently a pathogen of the urinary tract, particularly in patients undergoing long-term catheterization. This review covers P. mirabilis with a focus on urinary tract infections (UTI), including disease models, vaccine development efforts, and clinical perspectives. Flagella-mediated motility, both swimming and swarming, is a central facet of this organism. The regulation of this complex process and its contribution to virulence is discussed, along with the type VI-secretion system-dependent intra-strain competition, which occurs during swarming. P. mirabilis uses a diverse set of virulence factors to access and colonize the host urinary tract, including urease and stone formation, fimbriae and other adhesins, iron and zinc acquisition, proteases and toxins, biofilm formation, and regulation of pathogenesis. While significant advances in this field have been made, challenges remain to combatting complicated UTI and deciphering P. mirabilis pathogenesis.

    Based on light and scanning electron microscopical studies, the following five species of the Philometridae (Nematoda: Dracunculoidea) are described from female specimens collected from marine fishes off the southwestern coast of Java, Indonesia: Philometra lobotidis sp. n. from the abdominal cavity of the Atlantic tripletail Lobotes surinamensis (Bloch) (Lobotidae, Perciformes); Philometra javaensis sp. n. from the abdominal cavity of the immaculate puffer Arothron immaculatus (Bloch et Schneider) (Tetraodontidae, Tetraodontiformes); Philometra psettoditis sp. n. from the musculature of the Indian spiny turbot Psettodes erumei (Bloch et Schneider) (Psettodidae, Pleuronectiformes); Philometroides indonesiensis sp. n. from the musculature of the hound needlefish Tylosurus crocodilus crocodilus (Péron et Lesueur) (Belonidae, Beloniformes); and Philometroides trichiuri sp. n. from the dorsal fin of the largehead hairtail Trichiurus lepturus Linnaeus (type host) and the savalai hairtail Lepturacanthus savala (Cuvier) (both Trichiuridae, Perciformes). All these new species are distinguished from their congeners parasitizing marine fishes by morphological (mainly the shape and structure of the cephalic and caudal ends and of the oesophagus) and biometrical features. Besides previously known Philometra pellucida (Jägerskiöld, 1893) and Philometra ocularis Moravec, Ogawa, Suzuki, Miyazaki et Donai, 2002, they are the only nominal philometrid species recorded from Indonesian waters.

    Full Text Available Abstract Background In mouse the cytokine interleukin-7 (IL-7 is required for generation of B lymphocytes, but human IL-7 does not appear to have this function. A bioinformatics approach was therefore used to identify IL-7 receptor related genes in the hope of identifying the elusive human cytokine. Results Our database search identified a family of nine gene candidates, which we have provisionally named fibronectin immunoglobulin leucine-rich repeat (FIGLER. The FIGLER 1–9 genes are predicted to encode type I transmembrane glycoproteins with 6–12 leucine-rich repeats (LRR, a C2 type Ig domain, a fibronectin type III domain, a hydrophobic transmembrane domain, and a cytoplasmic domain containing one to four tyrosine residues. Members of this multichromosomal gene family possess 20–47% overall amino acid identity and are differentially expressed in cell lines and primary hematopoietic lineage cells. Genes for FIGLER homologs were identified in macaque, orangutan, chimpanzee, mouse, rat, dog, chicken, toad, and puffer fish databases. The non-human FIGLER homologs share 38–99% overall amino acid identity with their human counterpart. Conclusion The extracellular domain structure and absence of recognizable cytoplasmic signaling motifs in members of the highly conserved FIGLER gene family suggest a trophic or cell adhesion function for these molecules.

    Some think that using derivatives of snake venom for medical purposes is the modern version of snake oil but they are seriously misjudging the research potentials of some of these toxins in medicines of the 2000's. Medical trials, using some of the compounds has proven their usefulness. Several venoms have shown the possibilities that could lead to anticoagulants, helpful in heart disease. The blood clotting protein from the taipan snake has been shown to rapidly stop excessive bleeding. The venom from the copperhead may hold an answer to breast cancer. The Malaysian pit viper shows promise in breaking blood clots. Cobra venom may hold keys to finding cures for Parkinson's disease and Alzheimer's. Rattlesnake proteins from certain species have produced blood pressure medicines. Besides snake venoms, venom from the South American dart frog, mollusks (i.e. Cone Shell Snail), lizards (i.e. Gila Monster & Komodo Dragon), some species of spiders and tarantulas, Cephalopods, mammals (i.e. Platypus & Shrews), fish (i.e. sting rays, stone fish, puffer fish, blue bottle fish & box jelly fish), intertidal marine animals (echinoderms)(i.e. Crown of Thorn Star Fish & Flower Urchin) and the Honeybee are being investigated for potential medical benefits.

  2. Reconsidering sex-based stereotypes of COPD.

    Chronic obstructive pulmonary disease (COPD) has historically been considered a disease of older, white, male smokers, as illustrated in Frank Netter's classic images of the 'pink puffer' and 'blue bloater'. However, women may be more susceptible to COPD than men, and the disease course may be reflective of that increased susceptibility. From a review of epidemiological data of COPD, we found differences in the way men and women present with COPD symptoms, a bias in the way COPD symptoms are treated in men and women, and differences in susceptibility to airway obstruction based on age, sex, and smoking history. These data show that classic stereotypes of COPD - including male predominance - should be abandoned, and that there are not two but multiple COPD phenotypes, which are characterised by differences between women and men in susceptibility, symptoms, and disease progression. These differences impact on physician perception. Although further research into this concept is needed, the differences we found should prompt, in the short term, changes in the way (and in whom) COPD is evaluated, diagnosed, and treated; in the long term, these differences should prompt research into the prognosis of COPD based on sex differences.

    Full Text Available The tactical need for fixed wing aircraft with hovering capably has long been recognized bythe military for two reasons: increased safety when landing on aircraft carriers and higher velocitiesthat the ones obtainable with rotary wing aircraft.Thus far, the only concept governing the field of vertical flight was to use thrust either from a liftfan-F35, puffer ducts –Harrier or smaller jet engines-D0 31 or Yak-141, i.e. direct lift thrust.In this paper we will look at the prospect of using a combination of the Coanda effect with theVenturi effect to generate lift by so- called “supercirculation”. This novel approach can yield manyadvantages to conventional vertical lifting by providing a more stable platform and requiring lowerpower settings – and thus lower fuel consumption.The aircraft has a fixed, negatively sweped wing that uses circulation control to achieve lift atzero air speed. The fluid used for supercirculation will come from the fan thrust reversers – which, ifcorrectly managed, can give a sufficient flow for lifting the craft and also a negative thrust componentto compensate for the positive thrust of the primary flow (not diverted.

    Full Text Available The Tetraodontidae are an Acantomorpha fish family with circumglobal distribution composed of 189 species grouped in 19 genera, occurring in seas, estuaries, and rivers between the tropical and temperate regions. Of these, the genus Colomesus is confined to South America, with what have been up to now considered only two species. C. asellus is spread over the entire Amazon, Tocantins-Araguaia drainages, and coastal environments from the Amazon mouth to Venezuela, and is the only freshwater puffers on that continent. C. psittacus is found in coastal marine and brackish water environments from Cuba to the northern coast of South America as far south as to Sergipe in Brazil. In the present contribution we used morphological data along with molecular systematics techniques to investigate the phylogeny and phylogeography of the freshwater pufferfishes of the genus Colomesus. The molecular part is based on a cytochrome C oxidase subunit I dataset constructed from both previously published and newly determined sequences, obtained from specimens collected from three distinct localities in South America. Our results from both molecular and morphological approaches enable us to identify and describe a new Colomesus species from the Tocantins River. We also discuss aspects of the historical biogeography and phylogeography of the South American freshwater pufferfishes, suggesting that it could be more recent than previously expected.

  5. Ecological conversion efficiency and its influencers in twelve species of fish in the Yellow Sea Ecosystem

    The ecological conversion efficiencies in twelve species of fish in the Yellow Sea Ecosystem, i.e., anchovy ( Engraulis japonicus), rednose anchovy ( Thrissa kammalensis), chub mackerel ( Scomber japonicus), halfbeak ( Hyporhamphus sajori), gizzard shad ( Konosirus punctatus), sand lance ( Ammodytes personatus), red seabream ( Pagrus major), black porgy ( Acanthopagrus schlegeli), black rockfish ( Sebastes schlegeli), finespot goby ( Chaeturichthys stigmatias), tiger puffer ( Takifugu rubripes), and fat greenling ( Hexagrammos otakii), were estimated through experiments conducted either in situ or in a laboratory. The ecological conversion efficiencies were significantly different among these species. As indicated, the food conversion efficiencies and the energy conversion efficiencies varied from 12.9% to 42.1% and from 12.7% to 43.0%, respectively. Water temperature and ration level are the main factors influencing the ecological conversion efficiencies of marine fish. The higher conversion efficiency of a given species in a natural ecosystem is acquired only under the moderate environment conditions. A negative relationship between ecological conversion efficiency and trophic level among ten species was observed. Such a relationship indicates that the ecological efficiency in the upper trophic levels would increase after fishing down marine food web in the Yellow Sea ecosystem.

  6. A Novel Approach to Power Circuit Breaker Design for Replacement of SF6

    Full Text Available This contribution explores the role of PTFE ablation in enhancing current interruption for various background gases in high voltage circuit breakers. An assessment of the current interruption capability has been made in terms of the arcing duration and the contact gap length at which critical arc extinction is achieved. These observations are supported by measurements of the magnitude of extinction and re-ignition voltage peaks. Most previous and other current experimental work on gas filled circuit breaker design follows conventional wisdom in investigating arcing behaviours at elevated gas pressures (usually up to 6 bar. But in this work we concentrate on the effects of using low gas pressures (less than 1 bar in the presence of a close-fitting shield of ablatant polymer material (PTFE that surrounds the electrode assembly of an experimental high power circuit breaker. We demonstrate that for several different gases, arc extinction capability compares well under these conditions with SF6, suggesting that SF6 could be replaced entirely in this novel system by more environmentally friendly gases. Moreover, the critical contact gap lengths at extinction are only slightly greater than when using SF6 at 6 bar. Weight loss measurements from the ablatant shield suggest that a chemical puffer action is the most likely mechanism for achieving the observed arc extinctions in this system.

  7. Pufferfish mortality associated with novel polar marine toxins in Hawaii

    Fish die-offs are important signals in tropical marine ecosystems. In 2010, a mass mortality of pufferfish in Hawaii (USA) was dominated by Arothron hispidus showing aberrant neurological behaviors. Using pathology, toxinology, and field surveys, we implicated a series of novel, polar, marine toxins as a likely cause of this mass mortality. Our findings are striking in that (1) a marine toxin was associated with a kill of a fish species that is itself toxic; (2) we provide a plausible mechanism to explain clinical signs of affected fish; and (3) this epizootic likely depleted puffer populations. Whilst our data are compelling, we did not synthesize the toxin de novo, and we were unable to categorically prove that the polar toxins caused mortality or that they were metabolites of an undefined parent compound. However, our approach does provide a template for marine fish kill investigations associated with marine toxins and inherent limitations of existing methods. Our study also highlights the need for more rapid and cost-effective tools to identify new marine toxins, particularly small, highly polar molecules.

  8. Plasmonic enhancement of ultraviolet fluorescence

    Plasmonics relates to the interaction between electromagnetic radiation and conduction electrons at metallic interfaces or in metallic nanostructures. Surface plasmons are collective electron oscillations at a metal surface, which can be manipulated by shape, texture and material composition. Plasmonic applications cover a broad spectrum from visible to near infrared, including biosensing, nanolithography, spectroscopy, optoelectronics, photovoltaics and so on. However, there remains a gap in this activity in the ultraviolet (UV, research. Motivating factors in the study of UV Plasmonics are the direct access to biomolecular resonances and native fluorescence, resonant Raman scattering interactions, and the potential for exerting control over photochemical reactions. This dissertation aims to fill in the gap of Plasmonics in the UV with efforts of design, fabrication and characterization of aluminium (Al) and magnesium (Mg) nanostructures for the application of label-free bimolecular detection via native UV fluorescence. The first contribution of this dissertation addresses the design of Al nanostructures in the context of UV fluorescence enhancement. A design method that combines analytical analysis with numerical simulation has been developed. Performance of three canonical plasmonic structures---the dipole antenna, bullseye nanoaperture and nanoaperture array---has been compared. The optimal geometrical parameters have been determined. A novel design of a compound bullseye structure has been proposed and numerically analyzed for the purpose of compensating for the large Stokes shift typical of UV fluorescence. Second, UV lifetime modification of diffusing molecules by Al nanoapertures has been experimentally demonstrated for the first time. Lifetime reductions of ~3.5x have been observed for the high quantum yield (QY) laser dye p-terphenyl in a 60 nm diameter aperture with 50 nm undercut.     Although consistent individual-level differences in behaviour are widespread and potentially important in evolutionary and ecological processes, relatively few studies focus on the physiological mechanisms that might underlie and regulate these individual-level differences in wild populations. We conducted experiments to determine whether checkered pufferfish (Sphoeroides testudineus), which were collected from a dynamic (in terms of depth and water temperature) tidal mangrove creek environment in The Bahamas, have consistent individual-level differences in locomotor activity and the response to a simulated predator threat, as well as swimming performance and puffing in response to stressors. The relationships between personality and performance traits were evaluated to determine whether they represented stress-coping styles or syndromes. Subsequently, a displacement study was conducted to determine how personality and performance in the laboratory compared to movements in the field. In addition, we tested whether a physiological dose of the stress hormone cortisol would alter individual consistency in behavioural and performance traits. We found that pufferfish exhibited consistent individual differences in personality traits over time (e.g., activity and the duration of a response to a threat) and that performance was consistent between the lab and the natural enclosure. Locomotor activity and the duration of startled behaviour were not associated with swimming and puffing performance. Locomotor activity, puffing performance, and swimming performance were not related to whether fish returned to the tidal creek of capture after displacement. Similarly, a cortisol treatment did not modify behaviour or performance in the laboratory. The results reveal that consistent individual-level differences in behaviour and performance were present in a population from a fluctuating and physiologically challenging environment but that such traits are not necessarily correlated

  10. Stomatal closure of Pelargonium × hortorum in response to soil water deficit is associated with decreased leaf water potential only under rapid soil drying.

    Soil water deficits applied at different rates and for different durations can decrease both stomatal conductance (gs ) and leaf water potential (Ψleaf ). Understanding the physiological mechanisms regulating these responses is important in sustainable irrigation scheduling. Glasshouse-grown, containerized Pelargonium × hortorum BullsEye plants were irrigated either daily at various fractions of plant evapotranspiration (100, 75 and 50% ET) for 20 days or irrigation was withheld for 4 days. Xylem sap was collected and gs and Ψleaf were measured on days 15 and 20, and on days 16-19 for the respective treatments. Xylem sap pH and NO3 (-) and Ca(2+) concentrations did not differ between irrigation treatments. Xylem abscisic acid (ABA) concentrations ([ABA]xyl ) increased within 24 h of irrigation being withheld whilst gs and Ψleaf decreased. Supplying irrigation at a fraction of daily ET produced a similar relationship between [ABA]xyl and gs , but did not change Ψleaf . Treatment differences occurred independently of whether Ψleaf was measured in whole leaves with a pressure chamber, or in the lamina with a thermocouple psychrometer. Plants that were irrigated daily showed lower [ABA]xyl than plants from which irrigation was withheld, even at comparable soil moisture content. This implies that regular re-watering attenuates ABA signaling due to maintenance of soil moisture in the upper soil levels. Crucially, detached leaves supplied with synthetic ABA showed a similar relationship between [ABA]xyl and gs as intact plants, suggesting that stomatal closure of P. hortorum in response to soil water deficit is primarily an ABA-induced response, independent of changes in Ψleaf . © 2015 Scandinavian Plant Physiology Society.

  11. Mars Global Digital Dune Database (MGD3): Global dune distribution and wind pattern observations

    The Mars Global Digital Dune Database (MGD3) is complete and now extends from 90°N to 90°S latitude. The recently released south pole (SP) portion (MC-30) of MGD3 adds ∼60,000 km2 of medium to large-size dark dune fields and ∼15,000 km2 of sand deposits and smaller dune fields to the previously released equatorial (EQ, ∼70,000 km2), and north pole (NP, ∼845,000 km2) portions of the database, bringing the global total to ∼975,000 km2. Nearly all NP dunes are part of large sand seas, while the majority of EQ and SP dune fields are individual dune fields located in craters. Despite the differences between Mars and Earth, their dune and dune field morphologies are strikingly similar. Bullseye dune fields, named for their concentric ring pattern, are the exception, possibly owing their distinctive appearance to winds that are unique to the crater environment. Ground-based wind directions are derived from slipface (SF) orientation and dune centroid azimuth (DCA), a measure of the relative location of a dune field inside a crater. SF and DCA often preserve evidence of different wind directions, suggesting the importance of local, topographically influenced winds. In general however, ground-based wind directions are broadly consistent with expected global patterns, such as polar easterlies. Intriguingly, between 40°S and 80°S latitude both SF and DCA preserve their strongest, though different, dominant wind direction, with transport toward the west and east for SF-derived winds and toward the north and west for DCA-derived winds.

    With the alarming rise in the deaths due to cardiovascular diseases (CVD), present medical research scenario places notable importance on techniques and methods to detect CVDs. As adduced by world health organization, technological proceeds in the field of cardiac function assessment have become the nucleus and heart of all leading research studies in CVDs in which electrocardiogram (ECG) analysis is the most functional and convenient tool used to test the range of heart-related irregularities. Most of the approaches present in the literature of ECG signal analysis consider noise removal, rhythm-based analysis, and heartbeat detection to improve the performance of a cardiac pacemaker. Advancements achieved in the field of ECG segments detection and beat classification have a limited evaluation and still require clinical approvals. In this paper, approaches on techniques to implement on-chip ECG detector for a cardiac pacemaker system are discussed. Moreover, different challenges regarding the ECG signal morphology analysis deriving from medical literature is extensively reviewed. It is found that robustness to noise, wavelet parameter choice, numerical efficiency, and detection performance are essential performance indicators required by a state-of-the-art ECG detector. Furthermore, many algorithms described in the existing literature are not verified using ECG data from the standard databases. Some ECG detection algorithms show very high detection performance with the total number of detected QRS complexes. However, the high detection performance of the algorithm is verified using only a few datasets. Finally, gaps in current advancements and testing are identified, and the primary challenge remains to be implementing bullseye test for morphology analysis evaluation.

    The AI-PLBR breeder plant design prepared for ERDA and EPRI is of 1000 MWe size, utilizing a loop-type sodium system configuration and producing 2200 psig/850 0 F steam. A 'bullseye' core geometry type sodium system configuration and is employed with Pu0 2 -UO 2 fuel and UO 2 fertile material. The reactor outlet coolant temperature is 930 0 F. A modified 'A'-frame refueling system is employed, which is capable of handling 1/3 of a core loading in 12 days. An inducer-type mechanical pump is used in the primary circuit because of its excellent NPSH characteristics. Hockey sticks steam generators are used to produce near-fossil steam conditions at the turbine throttle. The balance of plant (BOP) design was developed by the architectural-engineering firm of Burns and Roe. It includes Allis-Chalmers - KWU 1800-rpm tandem-compound turbine, selected with high-,intermediate-, and two double-flow low-pressure cylinders equipped with two moisture separator cyclones. A design feature to enhance the licensability of the reference design is the three-level Decay Heat Removal System (DHRS), which consists of normal, backup, and diverse decay heat removal paths. The Atomics International PLBR design illustrated the technical soundness of the LMFBR system for meeting the world's long-term electrical energy supply needs. The design includes design features to assure licensability and innovative engineering features to enhance reliability, constructability, economics, and U.S. utility grid compatibility. The design concept provides a sound basis for the future detailed design of a prototype plant and subsequent development of larger LMFBR plants. (author)

  14. Analysis of the RPE sheet in the rd10 retinal degeneration model

    The normal RPE sheet in the C57Bl/6J mouse is subclassified into two major tiling patterns: A regular generally hexagonal array covering most of the surface and a 'soft network' near the ciliary body made of irregularly shaped cells. Physics models predict these two patterns based on contractility and elasticity of the RPE cell, and strength of cellular adhesion between cells. We hypothesized and identified major changes in RPE regular hexagonal tiling pattern in rdl0 compared to C57BL/6J mice. RPE sheet damage was extensive but occurred in rd10 later than expected, after most retinal degeneration. RPE sheet changes occur in zones with a bullseye pattern. In the posterior zone around the optic nerve RPE cells take on larger irregular and varied shapes to form an intact monolayer. In mid periphery, there is a higher than normal density of cells that progress into involuted layers of RPE under the retina. The periphery remains mostly normal until late stages of degeneration. The number of neighboring cells varies widely depending on zone and progression. RPE morphology continues to deteriorate long after the photoreceptors have degenerated. The RPE cells are bystanders to the rd10 degeneration within photo receptors, and the collateral damage to the RPE sheet resembles stimulation of migration or chemotaxis. Quantitative measures of the tiling patterns and histopathology detected here, scripted in a pipeline written in Perl and Cell Profiler (an open source Matlab plugin), are directly applicable to RPE sheet images from noninvasive fundus autofluorescence (FAF), adaptive optics confocal scanning laser ophthalmoscope (AO-cSLO), and spectral domain optical coherence tomography (SD-OCT) of patients with early stage AMD or RP.

    • Ultraviolet (UV) floral patterns are common in angiosperms and mediate pollinator attraction, efficiency, and constancy. UV patterns may vary within species, yet are cryptic to human observers. Thus, few studies have explicitly described the distribution or ecological significance of intraspecific variation in UV floral patterning. Here, we describe the geographic distribution and pattern of inheritance of a UV polymorphism in the model plant species Mimulus guttatus (Phrymaceae). We then test whether naturally occurring UV phenotypes influence pollinator interactions within M. guttatus.• We document UV patterns in 18 annual and 19 perennial populations and test whether UV pattern is associated with life history. To examine the pattern of inheritance, we conducted crosses within and between UV phenotypes. Finally, we tested whether bee pollinators discriminate among naturally occurring UV phenotypes in two settings: wild bee communities and captive Bombus impatiens.• Within M. guttatus, perennial populations exhibit a small bulls-eye pattern, whereas a bilaterally symmetric runway pattern occurs mainly in annual populations. Inheritance of UV patterning is consistent with a single-locus Mendelian model in which the runway phenotype is dominant. Bee pollinators discriminate against unfamiliar UV patterns in both natural and controlled settings.• We describe a widespread UV polymorphism associated with life history divergence within Mimulus guttatus. UV pattern influences pollinator visitation and should be considered when estimating reproductive barriers between life history ecotypes. This work develops a new system to investigate the ecology and evolution of UV floral patterning in a species with extensive genomic resources. © 2015 Botanical Society of America, Inc.

    Full Text Available OBJETIVO: Avaliar a qualidade da certificação médica dos óbitos de mulheres de 10 a 49 anos na região Sul do Município de São Paulo. MÉTODOS: Utilizou-se a metodologia Puffer para investigar as causas de óbito de todas as mulheres de 10 a 49 anos residentes na região e falecidas no ano de 1989 (num total de 664 óbitos. Foram comparadas as causas de morte relatadas nos atestados de óbitos originais com as refeitas. Calculou-se o grau de sensibilidade das causas e o índice de kappa. RESULTADOS: Obteve-se alta sensibilidade entre as declarações de óbito originais e corrigidas nos grupos de causas (CID-9 das "Doenças do Aparelho Circulatório" (91,9%, "Neoplasias" (89,7% e "Causas Externas" (84,1%. Houve outros com sensibilidades muito baixas. Desses, chama a atenção o capítulo "Transtornos Mentais" (34,3% que, de 11, passa a ter 32 óbitos em que, na maioria dos casos, não havia sido feito o diagnóstico de alcoolismo. Também o capítulo "Complicações da Gravidez, Parto e Puerpério" apresentou uma sensibilidade baixa (44,9%. O índice de kappa foi de 0,63, o que indica uma concordância total regular. DISCUSSÃO: Ainda é insatisfatória a qualidade da certificação médica dos óbitos na área estudada. Essa má qualidade pode afetar negativamente as propostas de intervenção, tornando pouco visíveis problemas tão sérios para a atenção à saúde da mulher.OBJECTIVE: To evaluate the quality of the medical certification of deaths of 10-49 year-old women, resident in the Southern region of the city of S.     Full Text Available Abstract Background The mosaic sperm protein zonadhesin (ZAN has been characterized in mammals and is implicated in species-specific egg-sperm binding interactions. The genomic structure and testes-specific expression of zonadhesin is known for many mammalian species. All zonadhesin genes characterized to date consist of meprin A5 antigen receptor tyrosine phosphatase mu (MAM domains, mucin tandem repeats, and von Willebrand (VWD adhesion domains. Here we investigate the genomic structure and expression of zonadhesin-like genes in three species of fish. Results The cDNA and corresponding genomic locus of a zonadhesin-like gene (zlg in Atlantic salmon (Salmo salar were sequenced. Zlg is similar in adhesion domain content to mammalian zonadhesin; however, the domain order is altered. Analysis of puffer fish (Takifugu rubripes and zebrafish (Danio rerio sequence data identified zonadhesin (zan genes that share the same domain order, content, and a conserved syntenic relationship with mammalian zonadhesin. A zonadhesin-like gene in D. rerio was also identified. Unlike mammalian zonadhesin, D. rerio zan and S. salar zlg were expressed in the gut and not in the testes. Conclusion We characterized likely orthologs of zonadhesin in both T. rubripes and D. rerio and uncovered zonadhesin-like genes in S. salar and D. rerio. Each of these genes contains MAM, mucin, and VWD domains. While these domains are associated with several proteins that show prominent gut expression, their combination is unique to zonadhesin and zonadhesin-like genes in vertebrates. The expression patterns of fish zonadhesin and zonadhesin-like genes suggest that the reproductive role of zonadhesin evolved later in the mammalian lineage.

  18. Occurrence of tetrodotoxin-binding high molecular weight substances in the body fluid of shore crab (Hemigrapsus sanguineus).

    The shore crab (Hemigrapsus sanguineus) is highly resistant to tetrodotoxin (TTX) although it contains no detectable amount of TTX (less than 5 MU/g, where 1 MU is defined as the amount of TTX killing a 20 g mouse in 30 min). Its body fluid was examined for neutralizing effects against the lethal activity of TTX. When the mixture of the body fluid and TTX was injected i.p. into mice, the lethal activity of TTX was significantly reduced; 1 ml of the body fluid was evaluated to neutralize 3.6-4.0 MU of TTX. Higher neutralizing activity (7.2-12.5 MU/ml of the body fluid) was exhibited by i.v. administration of the body fluid into mice before or after i.p. challenge of TTX. The lethal effect of paralytic shellfish poisons was not counteracted by the body fluid. Analysis by gel filtration on Sepharose 6B revealed that the body fluid contained TTX-binding high mol. wt substances (> 2,000,000) responsible for the neutralizing activity of the body fluid against TTX, which accounts for the high resistibility of the crab to TTX. When the crude toxin extracted from the liver of puffer (Takifugu niphobles) was mixed with the body fluid and chromatographed on Sepharose 6B, almost pure TTX was obtained from the fractions containing the TTX-binding high mol. wt substances, suggesting that the TTX-binding high mol. wt substances could be useful in purification of TTX from biological samples.

  19. Protein A from orange-spotted nervous necrosis virus triggers type I interferon production in fish cell.

    Family Nodaviridae consists of two genera: Alphanodavirus and Betanodavirus, and the latter is classified into four genotypes, including red-spotted grouper nervous necrosis virus, tiger puffer nervous necrosis virus, striped jack nervous necrosis virus, and barfin flounder nervous necrosis virus. Type I interferons (IFNs) play a central role in the innate immune system and antiviral responses, and the interactions between IFN and NNV have been investigated in this study. We have found that the RNA-dependent RNA polymerase (RdRp) from orange-spotted nervous necrosis virus (OGNNV), named protein A, was capable of activating IFN promoter in fathead minnow (FHM) cells. Transient expression of protein A was found to induce IFN expression and secretion, endowing FHM cells with anti-tiger frog virus ability. Protein A from SJNNV can also induce IFN expression in FHM cells but that from Flock House virus (FHV), a well-studied representative species of genus Alphanodavirus, cannot. RdRp activity and mitochondrial localization were shown to be required for protein A to induce IFN expression by means of activating IRF3 but not NFκB. Furthermore, DsRNA synthesized in vitro transcription and poly I:C activated IFN promoter activity when transfected into FHM cells, and dsRNA were also detected in NNV-infected cells. We postulated that dsRNA, a PAMP, was produced by protein A, leading to activation of innate immune response. These results suggest that protein As from NNV are the agonists of innate immune response. This is the first work to demonstrate the interaction between NNV protein A and innate immune system, and may help to understand pathogenesis of NNV. Copyright © 2018. Published by Elsevier Ltd.

    Full Text Available Background: Marine neurotoxins as of Marine biotoxins are natural toxins that produced mainly by dinoflagellates, diatoms and several species of invertebrates and fish. Marine poisoning results from the ingestion of marine animals contain these toxins and causes considerable adverse effects. Materials and methods: This review provides some facts about the structures of marine neurotoxins, their molecular target and pharmacology, analytical methods for their detection and quantitation, diagnosis and laboratory testing, clinical manifestations, as well as prevention and treatment, if were obtainable. Furthermore, we focus on marine poisoning and various associated neurological syndromes like ciguatera, tetrodotoxin poisoning, and paralytic shellfish poisoning, after ingestion of the common marine toxins. Results: A number of neurotoxins that prescribed according to their potency (LD50 are: Maitotoxin, Ciguatoxins and Palytoxin, Tetrodotoxin and Saxitoxin, Brevetoxins, Azaspiracid, Yessotoxin, Cooliatoxin, Domoic acid and Conotoxins, Respectively. The primary target of most marine neurotoxins is voltage gated sodium channels and the resulting block of ion conductance through these channels. Moreover, these compounds interact with voltage-gated potassium and calcium channels and modulate the flux of stated ions into many cell types. As well, the target recognized for palytoxin is the Na+- K+ /ATPase. Conclusion: Results of reviewed studies revealed that, the Ciguatera is the commonest syndrome of marine poisoning, but is rarely lethal. Puffer fish poisoning results from the ingestion of fish containing tetrodotoxin and paralytic shellfish poisoning are less common, but have a higher fatality rate than ciguatera. Despite their high toxicity, no much research has been done on some of the toxins, like maitotoxin.     Women's participation in the work force shifted markedly throughout the twentieth century, from a low of 21 percent in 1900 to 59 percent in 1998. The influx of women into market work, particularly married women with children, put pressure on the ideology of domesticity: an ideal male worker in the outside market married to a woman taking care of children and home (Williams, 2000). Here, we examine some moments in the early-to-mid-twentieth century when female psychologists contested established norms of life-work balance premised on domesticity. In the 1920s, Ethel Puffer Howes, one of the first generation of American women psychologists studied by Scarborough and Furumoto (1987), challenged the waste of women's higher education represented by the denial of their interests outside of the confines of domesticity with pioneering applied research on communitarian solutions to life-work balance. Prominent second-generation psychologists, such as Leta Hollingworth, Lillian Gilbreth, and Florence Goodenough, sounded notes of dissent in a variety of forums in the interwar period. At mid-century, the exclusion of women psychologists from war work galvanized more organized efforts to address their status and life-work balance. Examination of the ensuing uneasy collaboration between psychologist and library scholar Alice Bryan and the influential male gatekeeper E. G. Boring documents gendered disparities in life-work balance and illuminates how the entrenched ideology of domesticity was sustained. We conclude with Jane Loevinger's mid-century challenge to domesticity and mother-blaming through her questioning of Boring's persistent focus on the need for job concentration in professional psychologists and development of a novel research focus on mothering. © 2017 Wiley Periodicals, Inc.

  2. Guanidinium Toxins and Their Interactions with Voltage-Gated Sodium Ion Channels

    Full Text Available Guanidinium toxins, such as saxitoxin (STX, tetrodotoxin (TTX and their analogs, are naturally occurring alkaloids with divergent evolutionary origins and biogeographical distribution, but which share the common chemical feature of guanidinium moieties. These guanidinium groups confer high biological activity with high affinity and ion flux blockage capacity for voltage-gated sodium channels (NaV. Members of the STX group, known collectively as paralytic shellfish toxins (PSTs, are produced among three genera of marine dinoflagellates and about a dozen genera of primarily freshwater or brackish water cyanobacteria. In contrast, toxins of the TTX group occur mainly in macrozoa, particularly among puffer fish, several species of marine invertebrates and a few terrestrial amphibians. In the case of TTX and analogs, most evidence suggests that symbiotic bacteria are the origin of the toxins, although endogenous biosynthesis independent from bacteria has not been excluded. The evolutionary origin of the biosynthetic genes for STX and analogs in dinoflagellates and cyanobacteria remains elusive. These highly potent molecules have been the subject of intensive research since the latter half of the past century; first to study the mode of action of their toxigenicity, and later as tools to characterize the role and structure of NaV channels, and finally as therapeutics. Their pharmacological activities have provided encouragement for their use as therapeutants for ion channel-related pathologies, such as pain control. The functional role in aquatic and terrestrial ecosystems for both groups of toxins is unproven, although plausible mechanisms of ion channel regulation and chemical defense are often invoked. Molecular approaches and the development of improved detection methods will yield deeper understanding of their physiological and ecological roles.     Purpose: Recent progress in small animal radiotherapy systems has provided the foundation for delivering the heterogeneous, millimeter scale dose distributions demanded by preclinical radiobiology investigations. Despite advances in preclinical dose planning, delivery of highly heterogeneous dose distributions is constrained by the fixed collimation systems and large x-ray focal spot common in small animal radiotherapy systems. This work proposes a dual focal spot dose optimization and delivery method with a large x-ray focal spot used to deliver homogeneous dose regions and a small focal spot to paint spatially heterogeneous dose regions. Methods: Two-dimensional dose kernels were measured for a 1 mm circular collimator with radiochromic film at 10 mm depth in a solid water phantom for the small and large x-ray focal spots on a recently developed small animal microirradiator. These kernels were used in an optimization framework which segmented a desired dose distribution into low- and high-spatial frequency regions for delivery by the large and small focal spot, respectively. For each region, the method determined an optimal set of stage positions and beam-on times. The method was demonstrated by optimizing a bullseye pattern consisting of 0.75 mm radius circular target and 0.5 and 1.0 mm wide rings alternating between 0 and 2 Gy. Results: Compared to a large focal spot technique, the dual focal spot technique improved the optimized dose distribution: 69.2% of the optimized dose was within 0.5 Gy of the intended dose for the large focal spot, compared to 80.6% for the dual focal spot method. The dual focal spot design required 14.0 minutes of optimization, and will require 178.3 minutes for automated delivery. Conclusion: The dual focal spot optimization and delivery framework is a novel option for delivering conformal and heterogeneous dose distributions at the preclinical level and provides a new experimental option for unique radiobiological investigations

  4. Surface composition of Europa based on VLT observations

    Jupiter's moon Europa may harbor a global salty ocean under an 80-170 km thick outer layer consisting of an icy crust (Anderson et al. 1998). Meanwhile, the 10-50 My old surface, dated by cratering rates (Pappalardo et al. 1999) implies rapid surface recycling and reprocessing that could result in tectonic activity (Kattenhorn et al. 2014) and plumes (Roth et al. 2014). The surface could thus exhibit fingerprints of chemical species, as minerals characteristics of an ocean-mantle interaction and/or organics of exobiological interest, directly originating from the subglacial ocean. In order to re-investigate the composition of Europa's surface, a global mapping campaign of the satellite was performed with the near-infrared integral field spectrograph SINFONI on the Very Large Telescope (VLT) in Chile. The high spectral binning of this instrument (0.5 nm) and large signal noise ratio in comparison to previous observations are adequate to detect sharp absorptions in the wavelength range 1.45-2.45 μm. In addition, the spatially resolved spectra we obtained over five epochs nearly cover the entire surface of Europa with a pixel scale of 12.5 by 25 m.a.s ( 35 by 70 km on Europa's surface), thus permitting a global scale study. Several icy and non-icy compounds were detected and mapped at <100 km resolution. They are unevenly distributed on the moon's surface. Amorphous and crystalline water ice are both present and, in spite of a particularly strong amorphization process likely engendered by the Io plasma torus, the crystalline form is found to be approximately twice as abundant as the amorphous ice based on the analysis of the 1.65 μm band. If the surface is dominated by small and mid-sized water ice grains (25-200 μm), crystalline water-ice grains exhibit spatial inhomogeneities in their distribution.   5. Simultaneous emission transmission tomography using technetium-99m for both emission and transmission

    This phantom study investigates whether attenuation maps from transmission data degraded by increased noise from subtraction of emission counts can still provide useful attenuation correction in the regular and obese chest. Technetium-99m was used for both emission and transmission on a triple head simultaneous emission transmission tomography (Tc-Tc SETT) system. Fanbeam transmission counts were computed by subtracting emission counts estimated from the two parallel collimator heads. Radioactive decay was used to simulate organ counts from injections of 900 and 400 MBq sestamibi for regular and obese chest sizes. Line source activity was 350 MBq. Control attenuation maps were obtained with no emission activity. Noise control included catering for negative and zero transmission counts, pre-filtering and segmentation of mu maps. Pre-filtering was tried before and after subtraction and before and after setting negative pixels to zero. Mean±SD count/pixel at the heart in anterior transmission projections was typically 33±18 for the regular and 1±7 for the obese chest. For the obese chest, pre-filtering before resetting negative counts best preserved mean mu in soft tissue and lung. Tc-Tc SETT mu mean±SD for the regular chest were 0.144±0.012 and 0.058±0.004 for soft tissue and lung and for the obese chest, 0.152±0.075 and 0.059±0.017. The accuracy of the Tc-Tc SETT bullseye plots for the regular chest was the same as with control map attenuation correction and 3 times better than with no correction. For the obese chest it was as good as with control map correction only if mu map segmentation was applied. Tc-Tc SETT soft tissue and lung mu in 28 patient studies indicated that segmentation is practical for a wide range of chest sizes. Tc-Tc SETT on a triple-head system offers an accurate, inexpensive method of attenuation correction for the majority of chest sizes. (orig.)

    Purpose: Recent progress in small animal radiotherapy systems has provided the foundation for delivering the heterogeneous, millimeter scale dose distributions demanded by preclinical radiobiology investigations. Despite advances in preclinical dose planning, delivery of highly heterogeneous dose distributions is constrained by the fixed collimation systems and large x-ray focal spot common in small animal radiotherapy systems. This work proposes a dual focal spot dose optimization and delivery method with a large x-ray focal spot used to deliver homogeneous dose regions and a small focal spot to paint spatially heterogeneous dose regions. Methods: Two-dimensional dose kernels were measured for a 1 mm circular collimator with radiochromic film at 10 mm depth in a solid water phantom for the small and large x-ray focal spots on a recently developed small animal microirradiator. These kernels were used in an optimization framework which segmented a desired dose distribution into low- and high-spatial frequency regions for delivery by the large and small focal spot, respectively.     Modified and/or new keys to the four subfamilies now recognized within the Megaperidae Manter, 1934 n. comb. (Syn. Apocreadiidae Skrjabin, 1942) as well as the genera within each subfamily are presented. Two new genera, Paraschistorchis n. gen. and Plesioschistorchis n. gen., both within the Schistorchiinae Yamaguti, 1942, are erected and keys are provided to the species considered in both new genera-distinguished by possessing caeca that end either in separate ani or blindly. Plesioschistorchis callyodontis (Yamaguti, 1942) n. comb. and Plesioschistorchis haridis (Nagaty, 1957) n. comb. are re-described from new material collected from the common parrotfish, Scarus psittacus Forsskål (Perciformes: Scaridae), inhabiting the Red Sea off Egypt; S. psittacus represents a new host record for both species. The taxonomic status of Schistorchis sensu stricto Lühe, 1906 is examined and revised, a key to the four species we consider in this genus offered, and the monotypic genus Megacreadium Nagaty, 1956 declared a junior synonym of Schistorchis. Members of Schistorchis sensu stricto possess a unique "complex" (i.e. highly cellular/glandular) instead of "simple" (i.e. entirely muscular) type of oral sucker that is quite large in relation to body size; an elongate, somewhat sub-rectangular-shaped body; 5+ testes arranged in at least two rows; caeca that open via separate ani; a long post-testicular region; a median genital pore either at the anterior margin of or just anterior to the ventral sucker; and species of Schistorchis sensu stricto parasitize the intestine of marine fish within the Order Tetraodontiformes Berg. With the revision of this genus, we re-describe Schistorchis carneus Lühe, 1906 from the lower and mid-intestine of the white-spotted puffer, Arothron hispidus (Linnaeus) (Tetraodontiformes: Tetraodontidae), collected in the Red Sea off Egypt.     The concept of the sea as a source of noxious agents is perhaps not a familiar one to clinical neurologists, judging by the lack of reference to these agents in standard textbooks. Chemical, physiologic, and pharmacologic laboratories are increasingly investigating the properties of marine toxins, finding in them compounds with interesting and novel structures or unusual physiologic effects. Such substances are seen as possible agents for biologic and, more particularly, physiologic research, and as possible sources of new pharmaceuticals. These include hormone-like substances and antiviral or antitumor agents. Despite these specialized developments, which are in large measure a consequence of the technological advances of the present century, the clinician is at times directly concerned with the effects of marine toxic substances. For example, in Japan, puffer fish or tetrodotoxic poisoning is one of the major causes of deaths from food poisoning. Another marine toxin that has caused many explosive outbreaks of food poisoning. with many deaths in various parts of the world, comes from clams or mussels. This toxin, saxitoxin, is produced by species of marine protozoa including Gonyaulax, and is concentrated in filter-feeding molluscs. These two examples were of significant interest in medicine long before the technologic developments of the twentieth century. In the last few decades, entirely new problems of marine intoxication have arisen as a result of marine pollution from the disposal of industrial wastes in the sea. The most striking example of a man-made marine intoxication has been the outbreak of Minamata disease. In Minamata, Japan, the disposal of mercury-contaminated industrial wastes from a plastics factory into an enclosed bay, followed by human consumption of the contaminated fishes, crabs, or shellfish, led to many instances of acute cerebral degeneration.     Discovery of gonadotropin-inhibitory hormone (GnIH) in the Japanese quail in 2000 was the first to demonstrate the existence of a hypothalamic neuropeptide inhibiting gonadotropin release. We now know that GnIH regulates reproduction by inhibiting gonadotropin synthesis and release via action on the gonadotropin-releasing hormone (GnRH) system and the gonadotrope in various vertebrates. GnIH peptides identified in birds and mammals have a common LPXRF-amide (X = L or Q) motif at the C-terminus and inhibit pituitary gonadotropin secretion. However, the function and structure of GnIH peptides are diverse in fish. Goldfish GnIHs possessing a C-terminal LPXRF-amide motif have both stimulatory and inhibitory effects on gonadotropin synthesis or release. The C-terminal sequence of grass puffer and medaka GnIHs are MPQRF-amide. To investigate the evolutionary origin of GnIH and its ancestral structure and function, we searched for GnIH in agnathans, the most ancient lineage of vertebrates. We identified GnIH precursor gene and mature GnIH peptides with C-terminal QPQRF-amide or RPQRF-amide from the brain of sea lamprey. Lamprey GnIH fibers were in close proximity to GnRH-III neurons. Further, one of lamprey GnIHs stimulated the expression of lamprey GnRH-III peptide in the hypothalamus and gonadotropic hormone β mRNA expression in the pituitary. We further identified the ancestral form of GnIH, which had a C-terminal RPQRF-amide, and its receptors in amphioxus, the most basal chordate species. The amphioxus GnIH inhibited cAMP signaling in vitro. In sum, the original forms of GnIH may date back to the time of the emergence of early chordates. GnIH peptides may have had various C-terminal structures slightly different from LPXRF-amide in basal chordates, which had stimulatory and/or inhibitory functions on reproduction. The C-terminal LPXRF-amide structure and its inhibitory function on reproduction may be selected in later-evolved vertebrates, such as birds and mammals.

Dazu wurden in den Staedten Berlin, Eckernfoerde, Essen, Halle, Kiel, Rostock und Stuttgart repraesentative Boeden aus technogenen (Bau- bzw. Truemmerschutt, Hausmuell, Aschen, Schlacken, Schlaemme) sowie umgelagerten natuerlichen alkalischen (Hafenschlick) bzw. sauren (Kipplehm, Kokereilehm) Substraten bzw. Substratmischungen ausgewaehlt.     Profiles of alpha-melanocyte-stimulating hormone (alpha-MSH) in the Japanese flounder were examined by a newly developed time-resolved fluoroimmunoassay (TR-FIA) and immunohistochemistry. A TR-FIA for alpha-MSH was newly developed, and its levels in the pituitary gland and plasma of Japanese flounder reared in a white or black tank for 5 months were compared. A competitive assay using two antibodies was performed among secondary antibodies in the solid phase, alpha-MSH antibodies, samples, and europium-labeled Des-Ac-alpha-MSH. The sensitivity of the assay, defined as twice the standard deviation at a zero dose, was 0.98 ng/ml (49 pg/well). The intra- and interassay coefficients of variation of the assay were 8.8% (n=8) and 17.3% (n=5), respectively, at about 50% binding. Cross-reactivities of Des-Ac-alpha-MSH and Di-Ac-alpha-MSH were about 100%. Cross-reactivities of adrenocorticotropic hormone, salmon gonadotropin-releasing hormone (sGnRH), and chicken GnRH-II were less than 0.2%, and that of melanin-concentrating hormone was less than 2.0% at 50% binding. Displacement curves of serially twofold-diluted hypothalamus extract, pituitary gland extract, and plasma extract of Japanese flounder with the assay buffer were parallel to the alpha-MSH standard curve. Moreover, displacement curves of serially twofold-diluted hypothalamus and/or pituitary gland extract of masu salmon, goldfish, red seabream, Japanese eel, tiger puffer, and barfin flounder with the assay buffer were also parallel to the alpha-MSH standard. In Japanese flounder, total immunoreactive (ir)-alpha-MSH levels in the pituitary gland were lower in the black tank, whereas those in the plasma tended to be higher in the black tank, suggesting that the synthesis and release of alpha-MSH are higher in the black tank. alpha-MSH-ir cells were detected in the pars intermedia and a small part of the pars distalis of the pituitary gland. alpha-MSH-ir cell bodies were located in the basal hypothalamus and alpha

  13. The Geologic Signature of Anaerobic Oxidation of Methane (Invited)

    Anaerobic oxidation of methane (AOM) is an enormous sink in anoxic marine sediments for methane produced in situ or ascending through the sediment column towards the seafloor. Existing estimates indicate that between 75 and 382 Tg of sedimentary methane are oxidized each year before reaching the sediment-water interface making AOM a diagenetic process of global significance. This methane is derived from a variety of sources including microbial production, thermocatalytic cracking of complex organic matter, decomposing gas hydrates, and possibly abiogenic processes. Stables isotopes of membrane lipid biomarkers and authigenic carbonates associated with zones of AOM, fluorescence in situ hybridization, and anaerobic methane incubations have substantiated the role Archaea and sulfate-reducing bacteria have in driving AOM. The products of AOM are dissolved inorganic carbon (predominantly HCO3-) and bisulfide (HS-). Stable isotope measurements of authigenic carbonates associated with zones of AOM are consistent with the diagenetic carbon being primarily methane derived. These methane-derived carbonates occur in a variety of forms including sedimentary nodules and thin lenses within and below zones of contemporary AOM; outcrops of slabs, ledges, and jagged authigenic carbonates exhumed on the seafloor; and authigenic carbonate mounds associated with near-subsurface methane gas accumulations. Examples of exhumed authigenic carbonates include rugged outcrops along the Guaymas Transform in the Gulf of California, extensive slabs and ledges in the Eel River Basin, and mounds in various stages of development near Bullseye Vent, off Vancouver Island and in the Santa Monica Basin. It is clear from basic microbial biogeochemistry and the occurrences of massive authigenic carbonate which span a large range in size that DIC produced by AOM is preserved as authigenic carbonate within the seafloor and not on the seafloor.     The accretionary prism of the northern Cascadia margin is a classic gas hydrate research area. Ocean Drilling Program Leg 146 and Integrated Ocean Drilling Program (IODP) Expedition 311 documented that gas hydrate is widely distributed across the margin. In recent years an increased research focus has been on cold vents, where methane gas is actively released. Two recent expeditions funded by the Monterey Bay Aquarium Research Institute (MBARI) were conducted in the area of IODP Sites U1327 and U1328. An autonomous underwater vehicle (AUV) was used to map the seafloor bathymetry followed by dives with the ROV Doc Ricketts for ground truth information of various seafloor morphological features identified. The two cruises revealed many new seafloor features indicative of methane venting that were previously unknown. Bullseye Vent (BV) has been extensively studied using seismic imaging, piston coring, heat-flow, controlled-source EM, and deep drilling. BV is seismically defined by a circular wipe-out zone but the new AUV data show that BV is rather an elongated depression. BV is associated with a shoaling in the BSR, but lacks evidence for the existence of an underlying fault in the previous data. Although a massive gas-hydrate plug was encountered within the top 40 mbsf in the IODP holes, the ROV observations only revealed some platy methane derived carbonate outcrops at the outer-most rim of the depressions, a few beds of Vesicomya clams, and no observed gas vents, which together do not indicate that BV is especially active now. Further northeast of BV, but along the same trend, active gas venting was found associated with seafloor blistering and bacterial mats suggesting that there is an underlying fault system providing a fluid flow conduit. The newly discovered vent area has few seismic line crossings; however the available seismic data surprisingly are not associated with wipe-out zones.
