Efflux of dopamine through the dopamine transporter (DAT) is critical for the psychostimulatory properties of amphetamines, but the underlying mechanism is unclear. Here we show that Ca(2+)/calmodulin-dependent protein kinase II (CaMKII) plays a key role in this efflux. CaMKIIalpha bound to the distal C terminus of DAT and colocalized with DAT in dopaminergic neurons. CaMKIIalpha stimulated dopamine efflux via DAT in response to amphetamine in heterologous cells and in dopaminergic neurons. CaMKIIalpha phosphorylated serines in the distal N terminus of DAT in vitro, and mutation of these serines eliminated the stimulatory effects of CaMKIIalpha. The ubiquitous presence of mobile communication devices and the continuous development of mo- bile data applications, which results in high level of mobile devices' activity and exchanged data, often transparent to the user, makes privacy preservation an important feature of mobile telephony systems. We present a formal analysis of the UMTS Authentication and Key Agreement protocol, using the applied pi-calculus and the ProVerif tool. We formally verify the model with respect to privacy properties. We show a linkability attack which makes it possible, for individuals with low-cost equipment, to trace UMTS subscribers. The aim of the present study was to describe the distribution and remobilization of plutonium (Pu) in the sediments off the Rhone river mouth. For the reuse of a waste salt from an electrorefining process of a spent oxide fuel, a separation of rare earth elements by an oxidative precipitation in a LiCl-KCl molten salt was tested without using precipitate agents. In recent years, cobia has become an emerging farmed species in Asia due to its quick growth and high economic value. This study collects biological and economic data affecting the economic performance of cobia farming in three countries, namely Taiwan, China, and Vietnam. The data are collected by questionnaire sampling and analyzed by multivariate statistical analysis in order to compare the key factors affecting the production and economy of cobia farming in these three countries. The control and regulation of immune reactions is an area of intense investigation that has important implications for allogeneic hematopoietic cell transplantation. Immune reactions are regulated in a number of important ways. Compartmentalization of immune responses and the production of both pro-inflammatory and anti-inflammatory cytokines play a major role. More recently several populations of T cells that regulate immune responses termed regulatory T cells have been identified. We have investigated the mechanism by which resveratrol, a stilbene that is pro-apoptotic in many tumor cell lines, causes apoptosis in human head and neck squamous cell carcinoma UMSCC-22B cells by a mechanism involving cellular COX-2. Pro-inflammatory cytokines may directly influence the viability and metabolic function of colonic epithelial cells (CEC) as an early event in the development of inflammatory bowel disease. We report here that TNF-alpha+IFN-gamma induced a synergistic, concentration-dependent decline in butyrate oxidation, an essential energy supply, in HT-29 and DLD-1 cells. TNF-alpha+IFN-gamma induced a parallel profound decline in cell viability in HT-29 cells, but not in DLD-1 cells, where impairment of butyrate oxidation seemed to precede later occurrence of cell damage. TNF-alpha+INF-gamma induced CEC damage was independent on NO formation and involved the IFN-gamma signalling pathway as well as induction of apoptosis. If cytokines have similar effects in vivo, these may lead to energy deficiency and thus contribute to CEC damage and disturbance of the epithelial integrity.
The paper considers research carried out on the topic of life cycle (LC) modelling of industrial processes with emphasis upon energy utilisation and gaseous emissions. The aim of the work is to investigate existing LC assessment codes, in particular for their outputs and decision making potential, and to develop the characteristics of new software as appropriate. The paper focuses on a comparison of two methods for LC modelling: use of ''SimaPro 5'' with ECO-indicators and logical-information modelling. The considered approaches require attention to dynamic modelling having features for decision making; for example, in areas of costs, sensitivity analysis and optimisation of LC process parameters to economically reduce the total environmental load. Eight phospholipases A2 (PLAs) and four three-finger-toxins (3FTx) from the pooled venom of Bungarus fasciatus (Bf) were previously studied and sequenced, but their expression pattern in individual Bf venom and possible geographic variations remained to be investigated. We herein analyze the individual venom of two Bf specimens from Kolkata (designated as KBf) to address this question. Seven PLAs and five 3FTx were purified from the KBf venoms, and respective cDNAs were cloned from venom glands of one of the snakes. Comparison of their mass and N-terminal sequence revealed that all the PLAs were conserved in both KBf venoms, but that two of their 3FTx isoforms were variable. When comparing the sequences of these KBf-PLAs with those published, only one was found to be identical to that of Bf Vb-2, and the other five were 94-98% identical to those of Bf II, III, Va, VI and XI-2, respectively. Intercrosses of heterozygous pro-opiomelanocortin (POMC) mice result in homozygous null progeny at lower frequencies than expected. Genotyping offspring at pre-, peri-, and postnatal stages revealed that over half of homozygous null mutants die in the early postnatal stages. To investigate the reasons for this early postnatal lethality, we analyzed in detail different parameters in the initial hours after birth. POMC null mutants born to heterozygous dams presented at birth with corticosterone levels no different from wildtype littermates, were euglycemic, and had normal liver glycogen stores. However, already 30 min after birth corticosterone levels dropped by 80% and were undetectable thereafter, while corticosterone levels in wildtype animals increased during postnatal hours. The presence of endothelin (ET)-like immunoreactivity and the cardiovascular effects of mammalian ET-1 in fish have been reported. To identify ET-related peptides in fish, we screened the cDNA library of the salmon (Oncorhynchus keta) stomach by means of rapid amplification of cDNA ends, and we cloned cDNAs encoding an ET-related peptide. The salmon ET-related sequence of 21 amino acids is identical to the trout ET-1 peptide recently purified from kidney specimens of Oncorhynchus mykiss. The deduced amino acid sequence of salmon pre-proET-1 (PPET-1) comprises 244 amino acids, including a putative signal sequence and mature ET-1, as well as big ET-1 and ET-1-like sequences. The purpose of this study was to assess the agreement between rectal temperature and infrared emission detection (IRED). The specificity and sensitivity, the positive predictive value and the negative predictive value were calculated. It was a prospective, observational and unblinded study. Thermoscan-pro-1 was used to collect pairs of data in 41 patients, all hospitalised on an acute geriatric ward during a 4-month period. 41% of the patients were febrile. The 'bias' between rectal and tympanic measurement (left ear) was -0.64 degree C (d) and the standard deviation 0.58 degree C (s). The '95% Confidence Interval' for the bias was -0.82 degree C and -0.46 degree C. The '95% limits of agreement' between the rectal measurement and the tympanic measurement (left ear) was -1.78 degrees C and 0.50 degree C. In this paper the structural and functional changes of gamma irradiated bovine hemoglobin are presented. Aqueous solutions/1%/of HbO_2 were irradiated in air with doses ranging from 1 to 4 Mrad. Isoelectric focusing indicated change of the charge of irradiated hemoglobin. The isoelectric point of hemoglobin was displaced towards more acid values with increasing doses, up from 1 Mrad. Fingerprint analysis and peptide column chromatography of irradiated hemoglobin demonstrated disturbances increasing with the dose. These changes were confirmed by amino acid analysis which showed that Cys, Met, Trp, His, Pro and Tyr residues were destroyed or modified following irradiation. At doses exceeding 1 Mrad the irradiated solutions of hemoglobin showed a decrease of heme-heme interaction and an increase of affinity for oxygen. Connective tissue in biopsy specimens taken from the lower part of the uterine cervix in 40 pregnant women at various gestational ages was compared to that in similar biopsy specimens from 15 nonpregnant women. The concentrations of collagen, sulfated glycosaminoglycans, and hyaluronic acid decreased during pregnancy. At the gestational age of 10 weeks, the collagen concentration was 70%, and at term 30%, of that in the nonpregnant cervix. After delivery, no further decrease was observed. The extractability of collagen increased during pregnancy, as well as during labor. Also, the water concentration increased. An increase in the collagenolytic activity was observed with advancing gestational age. We present a new photonic technology and demonstrate that it allows for precise immobilisation of biomolecules to sensor surfaces. The technology secures spatially controlled molecular immobilisation since immobilisation of each molecule to a support surface can be limited to the focal point of the ultraviolet (UV) beam, as small as a few micrometers. We can immobilise molecules according to any pattern, from classical microarrays to diffraction patterns creating unique watermarking safety patterns. Given that suitable protein markers exists for all relevant diseases it is entirely feasible to test for a range of disease indicators (antigens and other markers) in a single test. Mining patterns from multi-relational data is a problem attracting increasing interest within the data mining community. Traditional data mining approaches are typically developed for highly simplified types of data, such as an attribute-value table or a binary database, such that those methods are not directly applicable to multi-relational data. Nevertheless, multi-relational data is a more truthful and therefore often also a more powerful representation of reality. Mining patterns of a suitably expressive syntax directly from this representation, is thus a research problem of great importance. In this paper we introduce a novel approach to mining patterns in multi-relational data. While levels of weight bias vary among individuals, it is not clear why one person possesses stronger anti-fat attitudes than another person. This investigation examined whether individual differences commonly associated with greater anti-fat bias are also associated with a greater preference for thinness among people of varying levels of weight. Young adults (62% women; 84% Caucasian) recruited from psychology classes (N=308) rated four male and female figures with approximate BMIs of 18.5, 25, 30, and 40, on measures of dislike and personality characteristics and completed measures assessing weight controllability, attitudes toward the obese, and perceptual reliance. We present the results of a study on the 1.4 GHz kpc-scale radio emission in the complete flux density limited MOJAVE sample, comprising 135 radio-loud AGNs. While extended emission is detected in the majority of the sources, about 7% of the sources exhibit only radio core emission. Many BL Lacs exhibit extended radio power and kpc-scale morphology typical of powerful FRII jets, while a substantial number of quasars possess radio powers intermediate between FRIs and FRIIs. This poses challenges to the simple radio-loud unified scheme, which links BL Lacs to FRIs and quasars to FRIIs. We find a significant correlation between extended radio emission and pc-scale jet speeds: the more radio powerful sources possess faster jets. This indicates that the 1.4 GHz (or low frequency) radio emission is indeed related to jet kinetic power. Implementation of Noble Metal Chemical Application (NMCA) in a Boiling Water Reactor (BWR) requires injection of a noble metal compound while the reactor is idle at hot shutdown conditions. In order to minimize outage time, utilities are very pro-actively finding ways to reduce the number of critical path tasks. One of these tasks is to remove some containment vessel drywell head bolts during the NMCA idle time. This, thereby, saves the utilities outage time, or they can perform other tasks as desired. Maintenance of power stations so far operated on the concept of using or extending the expected service life of a machine or plant as far as possible in order to avoid early and costly replacement. This was done by means of time-oriented maintenance, inspection and repair, otherwise called preventive maintenance. A European study on maintenance methods totals the cost of these measures at between 140 to 200 billion DM. One way of reducing these costs is 'predictive maintenance'. In the present study, the immunomodulatory effect of {beta}-endorphin ({beta}-E) and shorter pro-opiomelancortin (POMC) fragments was evaluated by assessing their influence on respiratory burst in human polymorphonuclear leukocytes (PMN). The effect of the peptides on phorbol myristate acetate (PMA)-stimulated production of reactive oxygen metabolites was measured in a lucigenin-enhanced chemiluminescence (CL) assay. Both POMC peptides with opiate-like activity and their non-opioid derivatives were tested. With the exception of {alpha}-E, PMA-stimulated respiratory burst was suppressed by all POMC fragments tested. A U-shaped dose-response relation was observed. The Savannah River Site (SRS) is a 310-square-mile United States Department of Energy nuclear facility located along the Savannah River near Aiken, South Carolina. During operations, which started in 1951, hazardous substances (chemicals and radionuclides) were released to the environment. The releases occurred as a result of inadvertent spills and waste disposal in unlined pits and basins which was common practice before environmental regulations existed. The hazardous substances have migrated to the vadose zone and groundwater in many areas of the SRS, resulting in 515 waste units that are required by environmental regulations, to undergo characterization and, if needed, remediation. Trihydroxynaphthalene reductase catalyzes two intermediate steps in the fungal melanin biosynthetic pathway. The enzyme, a typical short-chain dehydrogenase, is the biochemical target of three commercial fungicides. The fungicides bind preferentially to the NADPH form of the enzyme. Three X-ray structures of the Magnaporthe grisea enzyme complexed with NADPH and two commercial and one experimental fungicide were determined at 1.7 {angstrom} (pyroquilon), 2.0 {angstrom} (2,3-dihydro-4-nitro-1H-inden-1-one, 1), and 2.1 {angstrom} (phthalide) resolutions. It is demonstrated that highly dispersed molybdenum sulfide catalysts are prepared by using Mo(CO){sub 6} adsorbed on the support. The structure of ultradispersed molybdenum sulfide is suggested by EXAFS techniques to be different from that of MoS{sub 2}. The TOF of the thiophene HDS is almost invariant with the dispersion of molybdenum sulfide at a high molybdenum dispersion, whereas the TOF of the hydrogenation greatly increases with increasing dispersion. On the basis of the HDS activity and optimum Co/Mo ratio of Co-Mo/Al{sub 2}O{sub 3} catalysts having a variety of molybdenum dispersion fabricated by using carbonyl complexes, it is demonstrated that highly active HDS catalysts are prepared by highly dispersing molybdenum sulfide phases. A portion of the active site of rabbit skeletal myosin near the ribose ring of ATP can be labeled by the photoaffinity analogue 3{prime}(2{prime})-O-(4-benzoylbenzoyl)adenosine triphosphate (Bz{sub 2}ATP). The specificity of the photolabeling was assured by first trapping ({sup 14}C)Bz{sub 2}ATP at the active site by use of thiol cross-linking agents. Radiation-induced enteritis is a well-recognized sequel of therapeutic irradiation. Therefore we examined the radioprotective properties of Punica granatum peel extract (PPE) on the oxidative damage in the ileum. Rats were exposed to a single whole-body X-ray irradiation of 800 cGy. Irradiated rats were pretreated orally with saline or PPE (50 mg/kg/day) for 10 days before irradiation and the following 10 days, while control rats received saline or PPE but no irradiation. Then plasma and ileum samples were obtained. Irradiation caused a decrease in glutathione and total antioxidant capacity, which was accompanied by increases in malondialdehyde levels, myeloperoxidase activity, collagen content of the tissue with a concomitant increase 8-hydroxy-2'-deoxyguanosine (an index of oxidative DNA damage). One main symptom of this condition is anhedonia. There is evidence that exercise training can be used as a rehabilitative intervention in the treatment of depressive disorders. In this scenario, the aim of the present study was to assess the effect of an aerobic exercise training protocol on the depressive-like behavior, anhedonia, induced by repeated dexamethasone administration. Accurate automated alignment of laser beams in the National Ignition Facility (NIF) is essential for achieving extreme temperature and pressure required for inertial confinement fusion. The alignment achieved by the integrated control systems relies on algorithms processing video images to determine the position of the laser beam images in real-time. Alignment images that exhibit wide variations in beam quality require a matched-filter algorithm for position detection. One challenge in designing a matched-filter based algorithm is to construct a filter template that is resilient to variations in imaging conditions while guaranteeing accurate position determination. The development of leukemia and thyroid cancer is characterized by activation of the abl oncogene and ret oncogene, respectively. In order to clarify the relationship between these gene aberrations and radiation, the pro-myelogenous leukemia-derived cell line HL60 and the thyroid cancer-derived cell line 8505C, were irradiated in vitro with 100Gy of X-rays. RNA was then extracted from 10"8 cells of the respective cell lines and examined by the reverse transcription PCR method for rearrangements of abl and ret genes. The present study was designed to assess the radioprotective effects of amifostine on ovarian follicles. Three week-old female mice with or without pretreatment of amifostine were irradiated with 6.42 Gy of #gamma# -ray. Ovaries were collected 0 and 6h after irradiation. DNA fragmentation pattern and expression of genes and activity of proteins related with apoptosis were investigated by means of RT-PCR and Western blot. Proliferation of granulosa cells was reduced and incidence rate of follicular atresia was increased in ovarian follicles in #gamma# -ray irradiated mice compared to those in control or amifostine-treated group. Nutritionist Pro was used to analyze lunches for calories, total fat, saturated fat, protein, fiber, vitamin A, vitamin C, calcium, iron, fiber, and sodium content. Under certain growth conditions, the human colon adenocarcinoma cell line HT-29 exhibits intestinal enterocyte-like properties. The differentiated cells possess a brush border with the enzyme markers (aminopeptidase and sucrase) normally associated with the intestine. To aid in the characterization of the transport properties of these cells, the uptake of a non-metabolizable amino acid analog, "1"4C-#alpha#-methylaminoisobutyric acid (MeAIB) as examined in the HT-29-Al subclone which possesses a brush border. One part of the RWE measure catalogue in order to guarantee an environmental friendly and competitive electric power generation based on brown coal is to carry out measures improving the efficiency at existing steam turbines in RWE brown coal power plants. Since 1993 a total of 21 steam turbines with a gross capacity of 7,100 MW or net capacity of 6540 MW - this corresponds to approximately 70 % of the installed RWE brown coal power plant capacity - have been improved with an investment of approximately DM 500 million. The net block efficiency has been increased by 1.3 % on average. The planned net capacity increase of approximately 250 MW is still going to be achieved taking into account the pending enhancements. Nuclear power plant turbines have been successfully improved in the same way. Full text: As one of the Top 5 utilities in Europe, Rheinisch-Westfaelisches Elektrizitaetswerk AG, FWB: RW, RWE, operates a diversified generating capacity of about 45 GW; RWE Power alone generates over 30 GW. Besides a share of approximately 20% nuclear power, a large proportion of the generating base is in lignite and hard coal. RWE has an ambitious, Euro10 bn newbuild program of about 10 GW modern fossil fired plants and further expansion plans across Europe. By 2012, RWE intends to invest between Euro 15 and 20 bn outside Germany.The European Union has a challenging policy to increase the share of renewables, improve energy efficiency, and reduce greenhouse gases by 2020. At the same time, ageing power plants need to be replaced. This requires an enormous investment program across Europe. Materials and methods: 12 patients with 22 coronary bypass grafts underwent intraoperative flow quantification using the transit time ultrasound method. These values were compared to postoperative MR phase shift measurements. Flow measurements were performed preoperatively in 28 coronary arteries of 20 patients. For flow measurement, we used a velocity-encoded k-space segmented gradient echo sequence with a temporal resolution of 110 or 125 ms, respectively. 6-8 pase shift images could be acquired during one cardiac cycle. Results: There was a significant correlation between intraoperative and flow measurements using velocity-encoded MR imaging (r=0.76, p<0.0001, t-test). Flow volumes determined by MR imaging were systematically larger than those determined by the transit time ultrasound method. Purpose: The aim of this study was to test the feasibility of iliac artery stent placement under MR guidance with real-time MR radial scanning in an animal model. Materials and methods: The experiments were performed on three pigs in a 1.5 T scanner. Radial scanning with a gradient echo technique (T{sub R} 8.4 ms, T{sub E} 3.6 ms, flip angle 10 ) was used. A dedicated backprojector performed the reconstruction of the raw data in real-time. The resulting MR-images were displayed on LCD screens beside the magnet. The sliding window reconstruction technique allowed image acquistion at a frame rate of 16 images per second. MR-compatible self-expanding stents with a diameter of 8 mm and a length of 3 cm were placed into the left iliac artery. Their positions were verified by digital subtraction angiography (DSA) and compared to MRI. Results: All stents were successfully placed. Unlike the majority of existing diffusion research practices, the dissertation examines the differences between mobile and PC video diffusion patterns through the lenses of user-oriented influences in the user-technology relationship. Built upon the established adoption user group classification, the research is informed by the Uses and Gratification theory, the Social Technical theory, and the Technical Affordance perspective. These synergistic theoretical arguments share the recognition of the role of user in the dynamic, usually socially intertwined user-technology interactions.
