We examined whether proton magnetic resonance spectroscopy (MRS) could provide accurate information on histological grade and cell proliferation in astrocytomas. We studied 23 patients with astrocytomas: five grade II, 10 grade III and eight with grade IV (glioblastoma multiforme). We performed proton MRS and determined the Ki-67 labeling index (LI), a tumour proliferation marker, in the same areas of the astrocytomas, and examined the statistical relationship between proton MRS and Ki-67 LI. The N-acetylaspartate (NAA)/creatine-phosphocreatine (Cr) and NAA/choline (Cho)-containing compound ratios were always significantly lower and the Cho/Cr ratios significantly higher than those for normal brain. Since microwaves are used in human cancer therapy, information on specific biological effects of microwaves at elevated temperatures is important. To help supply this information, we exposed mammalian cells (CHO) and bacteria (Serratia marcescens) to hyperthermal temperatures (43, 44, and 45/sup o/C for CHO and 48, 49, and 50/sup o/C for the bacteria) with and without microwave irradiation. Temperature control was maintained by a refrigeration-reheat system and high-velocity water recirculation. The 2450-MHz microwave source was operated in a pulsed mode with power density up to 500 mW/cm/sup 2/. As expected, the survival curve slopes for both cell types increased rapidly with temperature, doubling for each degree Celsius. Abstract This study reports the effects of varying concentrations of copper sulfate on the metabolic and gene transcriptional profile of a recombinant Chinese hamster ovary (CHO) cell line producing an immunoglobulin G (IgG)-fusion protein (B0). Addition of 50 M copper sulfate significantly decreased lactate accumulation in the cultures while increasing viable cell density and protein titer. The state of chromatin during the cell cycle was examined using synchronized cultures of CHO hamster cells. Results support Mazia's dynamic chromosome cycle model and indicate that DNA-interactive chemotherapeutic agents elicit different types of kinetic responses in treated cells, suggesting a degree of specificity of interaction between various alkylating and intercalating agents and the genome. Coward et al. reported the use of a single daily urine sample. The adsorption of benzyl alcohol and 1-phenylethanol on graphitized thermal carbon black at 300 K is investigated. Thermodynamic characteristics of the adsorption of conformers stable in the gas phase and in the adsorbed state are determined by a molecular statistics method. The geometry of the molecules is optimized using the MP2 and B3LYP methods with a 6-311++G(d,p) basis set. Using the PBE0/6-31G(d) method, it is established that, in the adsorption of benzyl alcohol, two types of interactions, CH?O and OH??GTCB, occur. It is shown that, in 1-phenylethanol, intramolecular interactions are maintained.
Asynchronous Chinese hamster ovary (CHO) cells treated with conditioning hyperthermia (10 min at 45/sup 0/C) followed immediately by x irradiation with 4 Gy at room temperature display a decreased radiation response when either irradiated, or heated and irradiated again 24 h later. In addition, conditioning by 4 Gy of x radiation alone followed at 24 h by the combined heat and radiation treatment als resulted in a larger radiation D/sub 0/. Ionizing radiations that differ in number, size, and distribution of energy deposition events might be expected to cause different effects for the same absorbed dose. Furthermore, microdosimetry calculations suggest that large variations in biological effectiveness might be expected for internally deposited alpha-emitting radionuclides for the same absorbed dose, depending upon the specific activity and spatial distribution of the sources in tissue. The study described in this article was designed to demonstrate these phenomena in vitro. Cultured Chinese hamster ovary (CHO-K1 BH4) cells were exposed in vitro to inert, insoluble ceramic microspheres in zirconium dioxide labeled with "2"3"9Pu. The average exit energy of the 5.15 MeV alpha particle was calculated to be 4.3 MeV. Exposure times varied from four to seven hours to achieve the desired dose level.
Large scale production of monoclonal antibodies has been accomplished using bioreactors with different length to diameter ratios, and diverse impeller and sparger designs. The differences in these physical attributes often result in dissimilar mass transfer, mechanical stresses due to turbulence and mixing inside the bioreactor that may lead to disparities in cell growth and antibody production. The deflagration-to-detonation transition of low density (0.88 g/cc) PETN during initiation by both an exploding bridgewire and laser driven source is being studied using both laser interferometry and streak photography. Cutback experiments using VISAR have confirmed a 1.0 mm run-distance to detonation in low density PETN powder. In a detonation system using a combination of low and high density powders, an apparent center of initiation (COI) analysis of streak data has yielded a surprisingly similar result. This data suggests that a compaction of low density powder to near theoretical maximum density (TMD) occurs before the onset of detonation, which is consistent with work done previously.^1 Additionally, data analysis shows that although function time increases significantly with decreasing firing voltage, the apparent COI changes very little. The human smooth muscle myosin heavy chain locus (MYH11) was mapped by fluorescence in situ hybridization to the middle of the p arm of chromosome 16 using a genomic cosmid clone containing coding sequences of the gene as probe. Probe from coding sequence, when applied to Southern blots of a panel of hybrids containing different portions of human chromosome 16, localized the gene to 16p13.13-13.12. Coding sequence PCR primers, when used on the DNA from a CHO-mouse hybrid clone mapping panel informative for mouse chromosomes, showed that the gene was located on mouse chromosome 16. Platinum complexes play an important role in the development of anticancer drugs. Their cytotoxicity can be influenced by the nature of the leaving ligands, due to the hydrolysis reaction that occurs prior to the binding of the platinum complex to DNA. Also, non-leaving groups such as lipophilic diamines may affect cellular uptake. In this work, we describe the synthesis of platinum(II) complexes having oxalato and long chain aliphatic N-alkyl ethylenediamines as ligands. The products were characterized by elemental analyses, infrared spectroscopy and {sup 1}H, {sup 13}C and {sup 195}Pt NMR spectroscopy. Biological activity was assessed against tumor cell lines (A{sub 549}, B16-F1, B16-F10, MDA-MB-231) and non-tumor cell lines (BHK-21 and CHO). Although UCP has nor been detected in mitochondria from any other tissue, it shares structural and catalytic properties with several other mitochondrial carrier proteins. Although UCP was discovered only recently it is one of the most extensively studied mitochondrial carrier proteins.More recently, the mouse, rat, and human genes encoding for UCP have been isolated and sequenced. Antitumor platinum(II) complexes derived from N-alkyl-propanediamine differing in the length of their carbon chain (C8, C10, C12 and C14) were incorporated in liposomes and the cytotoxic activity of these formulations was evaluated against tumor (A{sub 549}, MDA-MB-231, B16-F1 and B16-F10) and non-tumor (BHK-21 and CHO) cell lines. Natural sunlight induces a genotoxic response in cultured CHO cells pre-treated with shale oil retort process water. Near ultraviolet light (NUV) component of the solar spectrum is the apparent radiation responsible for photoactivation. Cultured human skin fibroblasts are acutely sensitive to the genotoxic effects of photoactivated process water. The mutagenic potential of photoactivated process water in human cells is the same as that witnessed for an equivalent killing dose of the potent skin carcinogen FUV. DNA repair processes are involved in modulating genotoxic effects of this photo-induced process. Here, we detected the heterodimerization of Mac-1 subunits in living cells by means of two fluorescence resonance energy transfer (FRET) techniques (fluorescence microscopy and fluorescence spectroscopy) and our results demonstrated that there is constitutive heterodimerization of the Mac-1 subunits and this constitutive heterodimerization of the Mac-1 subunits is cell-type independent. The presented paper describes the formulation of the fractional mass balances which are used to calculate the concentration and size distribution of char in the balance cells of a circulating fluidized bed boiler. In case of using the shrinking core model additionally the distribution of conversion of the char particles is calculated. Basically two reaction models (a shrinking particle and a shrinking core model) are available to describe the combustion process. The model considers all essential physico/chemical phenomena (primary fragmentation, attrition, mass transfer to gas phase) which have to be taken into account for the description of the combustion process. The precipitation of heavy metals as hydroxides is the standard technique for the decontamination of waste water streams polluted by these elements. On the other side, progress in research has been made concerning the biosorption onto dead biomass and bioprecipitation supported by physiologically active bacteria. As the aim of this study, a flexible strategy has been envisaged cleaning a waste water with definite heavy metal load underlying the process mentioned above. Suitable bacteria were enriched and the process was tested in a technical plant. As result, a very high efficiency of heavy metal elimination has been found. The field of application covered by the acquired process is identical with the whole range of the waste water streams polluted by heavy metals. In the bacterial reversion assay with S. typhimurium TA98, TA100, TA1535 and TA1537, gamma irradiated hyaluronic acid (10 and 50 kGy) did not induce a significant increase in the number of revertant colonies in the presence of S9 metabolic activation system. In chromosomal aberration tests with CHO cells, gamma irradiated hyaluronic acid (10 and 50 kGy) did not result in an increase in the frequency of chromosomal aberrations. In vivo mouse micronucleus assay, gamma irradiated hyaluronic acid (10 and 50 kGy) did not show an increase in the frequency of polychromatic erythrocytes with micronuclei. The nuclear lamina proteins, prelamin A, lamin B, and a 70-kD lamina-associated protein, are posttranslationally modified by a metabolite derived from mevalonate. This modification can be inhibited by treatment with (3-R,S)-3-fluoromevalonate, demonstrating that it is isoprenoid in nature. We have examined the association between isoprenoid metabolism and processing of the lamin A precursor in human and hamster cells. Inhibition of 3-hydroxy-3-methylglutaryl coenzyme A reductase by mevinolin (lovastatin) specifically depletes endogenous isoprenoid pools and inhibits the conversion of prelamin A to lamin A. Prelamin A processing is also blocked by mevalonate starvation of Mev-1, a CHO cell line auxotrophic for mevalonate. Moreover, inhibition of prelamin A processing by mevinolin treatment is rapidly reversed by the addition of exogenous mevalonate. This effort aims at the elucidation of P-wave and S-wave velocity structures, high-precision identification of data obtained by the surface reflection method, and collection of basic data for active faults investigation in the future. Among the velocity structures obtained for various layers, the S-wave velocity structures in particular agree with the stratigraphy excellently and may be utilized in seismic analyses to be conducted in the future. After 3 years of construction the trial run of the heating power station HKW 2 ended in November 1997 on time. No extraordinary damage occured during construction, assembly or commissioning. The plant has been operating according to schedule since November 1997. A brief inspection carried out in the summer of 1998 in order to assess warranty claims showed no damage to any part except one valve. Experience shows that all aforementioned activities were necessary. The paper reports about success and negative experience. Certification according to DIN EN ISO 9000ff., however, affords to guarantee of high-quality construction. Builders of such plants therefore need to set up their own quality assurance centre and ensure its technical competence. The basis of assessment for particulate exhaust emissions is the gravimetric analysis of the particle concentration of diluted exhaust gases at temperatures below 52 C. This is very time consuming and takes sufficient sensitivity at low particulate emission rates like downstream of particle filters. For this reason the C/S-method, based on the thermographic analysis of carbon and sulfur, has been developed as an alternative to the gravimetric anallysis. This method yields particle mass as well as the main particle compounds. Comparisons between both methods have been carried out for heavy duty and automobile diesel engines as well as road vehicles and large diesel engines. The tests were conducted with commercial diesel fuel and also with other fuels, such as heavy oils. Several examples of agonist-directed trafficking of receptor signalling at 5-HT2A and 5-HT2C receptors have been reported that involve independent downstream transduction pathways. We now report the functional selectivity of a series of chemically diverse agonists at human (h)5-HT2A, h5-HT2B and h5-HT2C-VSV by examining two related responses, the upstream activation of Gq/11 proteins in comparison with its associated cascade of calcium mobilisation. At the h5-HT2A receptor, d-lysergic acid diethylamide (LSD) and the antiparkinsonian agents lisuride, bromocriptine and pergolide exhibit a higher potency for Gq/11 activation than calcium release in contrast with all the other tested ligands such as 5-HT, mCPP and BW723C86, that show an opposite preference of signalling pathway. Purpose: An animal model is used to investigate whether MR angiography combined with super-paramagnetic particles of iron oxide (SPIO) is suitable for detecting thromboses. Methods: 42 rats in groups of 7 each were examined on days 1, 3, 5, 7, 9 and 11, respectively, after mechanical/chemical thrombus induction in a 1.5 Tesla magnet with a FISP sequence (TR/TE/FA 50 ms/6 ms/40 ). Imaging was performed before and up to 90 minutes after intravenous injection of 30 {mu}mol FE/kg BW of the experimental SPIO (hydrodynamic diameter, 34{+-}17 nm LLS; R1 and R2 relaxivity at 0.47 T, 31 and 57 L/(mmol*s)). MIP reconstructions of MR angiographies were submitted to consensus assessment by two examiners using histology as the gold standard. CT and MRI of acquired abnormalities of the inner ear and cerebello-pontine angle present themselves with very typical findings. The imaging should be adapted to the pathology looked for and either CT or MRI should be used alone or in combination.CT, especially high resolution CT (HRCT), provides an excellent bone contrast, while MRI has a much superior soft tissue contrast. Acute inflammatory changes of the inner ear are solely depicted by contrast-enhanced MRI. HRCT excellently depicts osseous changes of the inner ear and cerebellopontine angle such as chronic ossifying labyrinthitis occurring after acute labyrinthitis, otosclerotic or traumatic changes. Tumorous changes not yielding to bony changes are best delineated by MRI. Posttraumatic hemorrhage and chronic fibrotic changes within the labyrinth are depicted by MRI, only. Aim: Today, MRI is an integral part of the presurgical evaluation of patients suffering from partial epilepsy. Purpose: Measurement of the oscillating CSF flow in the spinal canal (SC) of healthy volunteers and in patients with post-traumatic syringomyelia (PTS) using an optimized MRI protocol as well as to determine whether stenosis induced velocity changes are detectable using MRI. Methods: In 68 healthy volunteers quantitative studies of CSF flow in the cervical, thoracic, and lumbar regions were performed. First, an optimized sequence was developed and tested in 19 volunteers using four different flow-encoding velocities (4, 8, 12, 16 cm/s).
