Based on the enhanced cooperation between Republic of Korea (ROK) and International Atomic Energy Agency (IAEA), IAEA installed new safeguards equipment (Unattended Monitoring System, UMS) at Wolsung unit 2 on January, 2005. The UMS was utilized in parallel with traditional human surveillance during the transfer campaign of Wolsung unit 2 (7 March - 22 July). Installation of UMS at all Wolsung units will be decided on the basis of the result of this test. The transfer campaign with UMS at Wolsung units 2 and 4 was started from September 1, 2006 at the same time. Implementation of UMS to the transfer campaign in Wolsung site will be considerably reduced the IAEA's inspection effort and burden of operator.

It was agreed that both of the Republic Of Korea (ROK) and the International Atomic Energy Agency (IAEA) cooperate with the new approach using the Unattended Monitoring Systems (UMS), the mailbox declaration and the short notice inspections, which can decrease the agency inspection efforts and operator' s burden. As a result of the positive cooperation of ROK for applying new approach, the IAEA could accomplish a successful implementation of new safeguards approach during last transfer campaigns at Wolsung site. Preliminary experiments have been performed to investigate the effects of radiative cooling on plasma jets. Thin (3 um - 5 um) conical shells were irradiated with an intense laser, driving jets with velocities > 100 km/s. Through use of different target materials - aluminium, copper and gold - the degree of radiative losses was altered, and their importance for jet collimation investigated. A number of temporally resoved optical diagnostics was used, providing information about the jet evolution. Gold jets were seen to be narrower than those from copper targets, while aluminium targets produced the least collimated flows.

A thermohydraulic study of MTR fuel element is presented as a basic requirement for the development of an irradiation facility for testing fuel elements. A computer code named 'Thermo' has been developed for this purpose, which can stimulate different working conditions, such as, cooling, power elements and neutron flux, performing all pertinent thermohydraulic calculations. Thermocouples were used to measure the temperature gradients of the cooling fluid throughout the IEAR-1 reactor core. All experimental data are in good agreement with the theoretical model applied in this work. Finally, a draft of the proposed facility and its safety system is presented. (author)

Bacteriocins are an abundant class of antimicrobial molecules that appear to mediate population dynamics within species. The bacteriocins of Escherichia coli have served as a model for exploring the ecological role of these potent toxins. Studies suggest that colicins provide a competitive edge in nutrient-poor environments and that there might be a trade-off between the costs and benefits of colicin production. PMID:10203843

The CYP1A (EROD)-inducing potencies of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), 3,3minutes or feet,4,4minutes or feet,5-pentachlorobiphenyl (PCB126) and benzo(k)fluoranthene (B(k)F) were studied in avian embryo livers. TCDD and PCB126 proved to be much more potent as inducers in the chicken than in the other species examined. This finding is consistent with a considerably higher sensitivity of the chicken compared with a number of other avian species to the embryotoxic effects of these compounds. Furthermore, the relative potencies of the tested Ah receptor agonists as CYP1A inducers differed substantially between species. B(k)F and PCB126 showed similar induction potencies in domestic duck embryos, whereas PCB126 is much more potent than B(k)F in the chicken. Cell-penetrating peptides (CPPs) are useful tools for the delivery of hydrophilic bioactive molecules, such as peptides, proteins, and oligonucleotides, across the cell membrane. To realize the delivery of therapeutic macromolecules by CPPs, the CPPs are required to show resistance to protease and no cytotoxicity. In order to produce potent non-toxic and protease-resistant CPPs with high cellular uptake, we designed an amphipathic helix peptide using ?-aminoisobutyric acid (Aib, U) and named it MAP(Aib). In the MAP(Aib) molecule, five Aib residues are aligned on the hydrophobic face of the helix and five lysine (K) residues are aligned on the hydrophilic face. MAP(Aib) showed potent resistance to trypsin and pronase compared with MAP, an amphipathic helix peptide formed by usual amino acids. Several compounds were evaluated in nonhuman primates for their potential to induce neoplasms, especially hepatocellular carcinoma (HCC). The compounds can be classified into three groups: food contaminants, model rodent carcinogens, and nitrosamines. All three compounds in the food contaminants group, namely, aflatoxin B1, sterigmatocystin, and methylazoxymethanol acetate, induced HCC. None of the model rodent carcinogens tested consistently induced HCC in rhesus and cynomolgus monkeys. Three of four nitrosamines evaluated induced HCC in rhesus and cynomolgus monkeys. One nitrosamine, diethylnitrosamine, is a predictable and potent inducer of HCC and is useful for establishment of a nonhuman primate model for numerous oncologic studies.

A series of pyrrolo[2,3-d]pyrimidine-2,4-diones and intermediates was tested for anticonvulsant activity in mice. Eleven of the 15 compounds possessed anticonvulsant activity against pentylenetetrazol-induced convulsions. One compound afforded more anticonvulsant protection against pentylenetetrazol than did trimethadione (67 and 50%, respectively). A suspension of this compound was found to be as effective as a solution in producing anticonvulsant activity. The results suggest that the nitrile analogs were more potent compared to the carbamyl analog due to higher lipid solubility. PMID:7373552

There are 4 CANDU reactors at Wolsung site together with a spent fuel dry storage associated with unit 1. These CANDU reactors, classified as On-Load Reactor (OLR) for Safeguards application, change 16- 24 fuel bundles with fresh fuel in everyday. Especially, the spent fuel bundles are transferred from spent fuel bays to dry storage throughout a year because of the insufficient capacity of spent fuel pond. Safeguards inspectors verify the spent fuel transfer to meet safeguards purposes according to the safeguards criteria by means of inspector's presence during the transfer campaign. For the verification, 60-80 person-days of inspection (PDIs) are needed during approximately 3 months for each unit. 3407 events were registrated up to a distance of 300 m during the period from 12.10.1989 to 31.12.1990, of which 385 could be precisely located. Activities were determined in areas surrounding the dam construction traversal drifts and boreholes at the 875 m level, near the large cavern at 975 m depth, in drifts and caverns above the 875 m level and in the vicinity of the rock salt/carnallitite transition zone to the south-east of the 875 m level. Energies and source parameters were calculated by means of spectres. Energy variations range from 10 J to 10{sup -4} J, the source radii from 3.5 m to 0.2 m, the source dislocations from 10{sup -6} m to 10{sup -8} m, and the stress drops from 10{sup 5} Pa to 0.06 10{sup 5} Pa. Two ent-rosane- (cuzcol, 1 and 6-dehydroxycuzcol, 2) and a abietatriene- (salvadoriol, 3) type diterpenoids have been isolated from Maytenus cuzcoina and Crossopetalum uragoga, respectively, along with five known diterpene compounds (4-8). Their stereostructures have been elucidated on the basis of spectroscopic analysis, including 1D and 2D NMR techniques, and computational data. The absolute configuration of cuzcol was determined by application of Riguera ester procedure. This is the first instance of isolation of ent-rosane diterpenoids from species of the Celastraceae. The isolated diterpenes were found to be potent anti-tumour-promoter agents, and carnosol (7) also showed a remarkable chemopreventive effect in an in vivo two-stage carcinogenesis model.

Reactive blue 2 (RB-2) had been characterized as a relatively potent ectonucleoside triphosphate diphosphohydrolase (E-NTPDase) inhibitor with some selectivity for NTPDase3. Three analogs of lidocaine (benzyl carbamyl, benzyl nitrile and methyl nitrile) were synthesized and examined for cardiovascular and central activity. The benzyl carbamyl analog was more potent than lidocaine in lowering blood pressure but possessed only slight local anesthetic, antiarrhythmic and CNS-depressant activity. At 40 mg/kg the benzyl nitrile derivative was superior to lidocaine in protecting against chloroform-induced arrhythmias. The methyl nitrile analog was less active than the benzyl nitrile analog in most parameters examined. The benzyl nitrile derivative and lidocaine had similar potencies on blood pressure depression, local anesthetic activity and ability to protect against calcium chloride-induced arrhythmias. Unlike the benzyl carbamyl derivative both lidocaine and the benzyl nitrile compounds appear to depress the cardiovascular system via a common mechanism. PMID:515167

Abstract: The ability of melatonin as a potent antioxidant was used as a rationale for testing its antiapoptotic ability in normal cells. Recently, melatonin was shown to possess proapoptotic action by increasing reactive oxygen species in certain cancer cells. The modification of radiation-induced apoptosis by melatonin and the expression of apoptosis-associated upstream regulators were studied in normal mice splenocytes and Jurkat T leukemia cells. Glycyrrhizin is a triterpene glycoside, a major active constituent of licorice (Glycyrrhiza glabra) root and numerous pharmacological effects like anti-inflammatory, anti-viral, anti-tumour and hepatoprotective activities has been attributed to it. In this study we evaluated the anti-inflammatory activities of glycyrrhizin in mice model of acute inflammation, carrageenan-induced pleurisy. A nanosuspension of magnetically tagged metronidazole was developed by the solvent displacement method coupled with ultrasonication and was evaluated for its physicochemical properties. The drug release from metronidazole magnetic nanosuspension at pH 1.2 and 7.0 shows maximum correlation coefficient for zero order and Higuchi model, respectively. The anthelmintic activity of the formulated metronidazole magnetic nanosuspension was evaluated on Indian earthworms (Pheretima poi). Metronidazole magnetic nanosuspension at a dose of 10 and 50 mg/ml shortened by 31% and 34%, respectively, the mean time to death of the earthworms when compared against a non-magnetic metronidazole suspension. Thus, the developed metronidazole magnetic nanosuspension showed potent, controlled and targeted drug action and might be a good therapeutic avenue in combating infectious GI disorders.

In this article, we report anticancer activity of 14 anthracenedione derivatives separated from the secondary metabolites of the mangrove endophytic fungi Halorosellinia sp. (No. 1403) and Guignardia sp. (No. 4382). Some of them inhibited potently the growth of KB and KBv200 cells, among which compound 6 displayed strong cytotoxicity with IC(50) values of 3.17 and 3.21 microM to KB and KBv200 cells, respectively. Furthermore, we demonstrate that the mechanism involved in the apoptosis induced by compound 6 is probably related to mitochondrial dysfunction. Additionally, the structure-activity relationships of these compounds are discussed. PMID:20479985

The University of Michigan (UM) is a member of a team of researchers, including the universities of Florida, Texas, and Tennessee, along with Oak Ridge National Laboratory, developing robotic for hazardous environments. The goal of this research is to develop the intelligent and capable robots which can perform useful functions in the new generation of nuclear reactors currently under development. By augmenting human capabilities through remote robotics, increased safety, functionality, and reliability can be achieved. In accordance with the established lines of research responsibilities, our primary efforts during 1991 will continue to focus on the following areas: radiation imaging; mobile robot navigation; three-dimensional vision capabilities for navigation; and machine-intelligence. This report discuss work that has been and will be done in these areas.

An effect of laser-cooling of water was observed for the first time with a temperature decrease dT = -2.2 K after irradiation of liquid water surface by a powerful Ar-Xe pulse laser with a pulse energy of about 1 J and wavelength L = 1.73, 2.63 and 2.65 um. The discovered effect can apparently be ascribed to the optical excitation of vibrational states of H2O molecules followed by an endothermic consolidation of chemically active excited molecules into a quasi-stable cluster-like structure. The measured time dependences of the cooling effect show that a typical life time of the new state of water amounts to hours. It has also been shown that the life time of the excited vibrational molecular states due to a radiation trapping effect can be estimated to at least hundreds of seconds.

The total amount of uranium dissolved in seawater at a uniform concentration of 3 mg U/m3 in the world's oceans is 4.5 billion tons. An adsorption method using polymeric adsorbents capable of specifically recovering uranium from seawater is reported to be economically feasible. A uranium-specific nonwoven fabric was used as the adsorbent packed in an adsorption cage 16 m2 in cross-sectional area and 16 cm in height. We submerged three adsorption cages in the Pacific Ocean at a depth of 20 m at 7 km offshore of Japan. The three adsorption cages consisted of stacks of 52 000 sheets of the uranium-specific non-woven fabric with a total mass of 350 kg. The total amount of uranium recovered by the nonwoven fabric was >1 kg in terms of yellow cake during a total submersion time of 240 days in the ocean.

Embryos of chicken (Gallus domesticus), domestic duck (Anas platyrhynchos), and common eider duck (Somateria mollissima) were exposed in ovo to PCBs and to polycyclic aromatic hydrocarbons. Two coplanar PCBs, 3,3{prime},4,4{prime}-tetrachlorobiphenyl (PCB {number_sign}77) and 3,3{prime},4,4{prime},5-pentachlorobiphenyl (PCB {number_sign}126), were considerably more lethal and potent as inducers of 7-ethoxyresorufin O-deethylase (EROD) in chicken embryos (Gallus domesticus) than in embryos of the other two species. In chicken embryos, these compounds caused edema and eye and beak deformities. An artificial mixture of 18 PAHs which all have been detected in environmental samples, was slightly more toxic to embryos of the domestic duck and the common eider duck than to chicken embryos. The rapid growth in the use of in vitro methods for nanoparticle toxicity assessment has proceeded with limited consideration of the unique kinetics of these materials in solution. Particles in general and nanoparticles specifically, diffuse, settle and agglomerate in cell culture media as a function of several systemic and particle factors: media density and viscosity, particle size, shape and density. Cellular dose then is also a function of these factors as they determine the rate of transport of nanoparticles to cells in culture. Here we develop and apply the principles of particokinetics and dosimetry in vitro and outline an approach for simulation of nanoparticle particokinetics in cell culture systems. Borehole stability is affected to a significant extent by the 3D local and regional stress field. Preliminary information on underground stress fields is provided by the World Stress Map, which shows N-NW stress at a depth of about 5000 m for the Voelkersen field. Earlier data analyses showed various directions in the range of 310-360 degrees. This was assumed to be a methodical error. Compared to the reference year of 1990, CO{sub 2} emissions of the Federal Republic of Germany have been curbed by about 12%, primarily as a result of industrial backfitting and pollution abatement measures accomplished in the industrial infrastructure of the new Federal Laender of Germany. Federal Government policy sticks to the previous commitment, to reduce carbon dioxide emissions by 25% by the year 2005. Current forecasts and scenarios of future greenhouse gas emissions show that only immediate adherence to a very stringent reduction policy might ensure that this ambitious goal will be achieved. Expression of IGF-2 and changes in its expression in response to systemic endocrine alterations have not been demonstrated for adipocytes. Adipocytes were induced to develop within cultures of 3T3-L1 cells using a medium containing 0.5mM isobutylmethylxanthine, 1uM insulin and 100ng hydrocortisone/ml for 48 hours of exposure. Cultures containing developing adipocytes were incubated with 10% pig serum and 1 uM insulin for several days. The resultant adipocyte cultures were then treated with either 10% pig serum, diabetic pig serum or hypophysectomized pig serum in DMEM for 48 hours. Adipocytes within the cultures were separated from undifferentiated cells using percoll density gradient centrifugation. Total RNA was isolated from adipocytes and dot blotted. We present an alternative, Bayesian method for large-scale reconstruction from observed peculiar velocity data. The method stresses a rigorous treatment of the random errors and it allows extrapolation into poorly sampled regions in real space or in k-space. A likelihood analysis is used to determine the fluctuation power spectrum, followed by a Wiener Filter (WF) analysis to obtain the minimum-variance mean fields of velocity and mass density. Constrained Realizations (CR) are then used to sample the statistical scatter about the WF mean field. The WF/CR method is applied as a demonstration to the Mark III data with 1200 km/s, 900 km/s, and 500 km/s resolutions. The main reconstructed structures are consistent with those extracted by the POTENT method. A comparison with the structures in the distribution of IRAS 1.2Jy galaxies yields a general agreement. In the 80's and 90's our group and others discovered that the genes encoding various cytokines and their receptors are e (more) xpressed in vascular, glial, and neuronal structures of the adult brain. Most cytokines act through cell surface receptors that have one transmembrane domain and which transduce a signal through the JAK/STAT pathway. New tetradentate N(2)O(2) donor Schiff bases and their mononuclear Co(II), Ni(II), Cu(II), and Pd(II) complexes were synthesized and characterized extensively by IR, (1)H-, (13)C-NMR, mass, ESR, conductivity measurements, elemental and thermal analysis. Specifically the magnetic and electronic spectral measurements demonstrate the octahedral structures of cobalt(II), nickel(II) complexes and square planar geometries of copper(II), palladium(II) complexes. All the ligands and complexes were screened for their in vitro antibacterial activity against two gram-positive bacteria (Bacillus subtilis, Staphylococcus aureus) and two gram-negative bacteria (Escherichia coli, Klebsiella pneumonia). In this study, Pd(II) complexes exhibited potent antibacterial activity against B. subtilis, S. aureus whereas other metal complexes also exerted good activity towards all tested strains even than standard drugs streptomycin and ampicillin. Bioassay-guided fractionation of the EtOH extract of licorice (Glycyrrhiza glabra roots), using a GAL-4-PPAR-g chimera assay method, resulted in the isolation of 39 phenolics, including 10 new compounds (1-10). The structures of the new compounds were determined by analysis of their spectroscopic data. Among the isolated compounds, 5prime-formylglabridin (5), (2R,3R)-3,4prime,7-trihydroxy-3prime-prenylflavane (7), echinatin, (3R)-2prime,3prime,7-trihydroxy-4prime-methoxyisoflavan, kanzonol X, kanzonol W, shinpterocarpin, licoflavanone A, glabrol, shinflavanone, gancaonin L, and glabrone all exhibited significant PPAR-g ligand-binding activity. The activity of these compounds at a sample concentration of 10mg/mL was three times more potent than that of 0.5mM troglitazone.

Examination of a chloroform extract of Dracaena loureiri Gagnep (Agavaceae), a Thia medicinal plant possessing antibacterial activity, has led to the isolation of fifteen flavenoids. The biogenic relationships among these flavenoids isolated were briefly discussed. Definition of the skeleton and the unambiguous assignment of all of the protons of the isolates was achieved through extensive 2D-homonuclear chemical shift correlation, nuclear Overhauser effect (NOE) difference spectroscopy and 2D-NOE experiments. The effects of the dopants, Mg"2"+, Sr"2"+, Sc"3"+, Yb"3"+, Gd"3"+, La"3"+, Ti"4"+, Zr"4"+, Ce"4"+, and Nb"5"+, on the grain boundary mobility of dense Y_2O_3 have been investigated from 1,500 to 1,650 C. Parabolic grain growth has been observed in all cases over a grain size from 0.31 to 12.5 microm. Together with atmospheric effects, the results suggest that interstitial transport is the rate-limiting step for diffusive processes in Y_2O_3, which is also the case in CeO_2. The effect of solute drag cannot be ascertained but the anomalous effect of undersized dopants (Ti and Nb) on diffusion enhancement, previously reported in CeO_2, is again confirmed. Indications of very large binding energies between aliovalent dopants and oxygen defects are also observed. Overall, the most effective grain growth inhibitor is Zr"4"+, while the most potent grain growth promoter is Sr"2"+, both at 1.0% concentration.

When total mesorectal excision (TME) is accurately performed, dysfunction, theoretically, does not occur. However, there are differences among individuals in the running patterns and the volumes of nerve fibers, and if obesity or a narrow pelvis is present, nerve identification is difficult. Currently, the rate of urinary dysfunction after rectal surgery ranges from 33% to 70%. Many factors other than nerve preservation play a role in minor incontinence. Male sexual function shows impotence rates ranging from 20% to 46%, while 20%-60% of potent patients are unable to ejaculate. In women, information on sexual function is not easily obtained, and there are more unknown aspects than in men. Oxygenated sterol compounds (OSC), when incubated for 1 hr with human erythrocytes in lipoprotein-depleted medium at concentrations of 0.625-5 x 10/sup -5/M, are inserted into the cell membrane and remain there despite subsequent washing of the cells. The insertion results in expansion of the surface area of the red cell ghost membrane, an increase in critical hemolytic volume, and as a consequence, in dimished osmotic fragility of the erythrocytes. This effect is seen with echinocyte-forming as well as with non-echinocyte-forming OSC. Erythrocytes treated with OSC do not differ from control cells with respect to their mean cell volume (MCV) in isotonic solution, water content, ion fluxes, and filterability through polycarbonate filters. Methylxanthines and their derivatives are antagonists at cell surface adenosine receptors. They report here a systematic study of xanthine structure-activity relationships which compares potency at two adenosine receptor subtypes, R/sub a/ and R/sub i/. Adenylate cyclase stimulation (R/sub a/ in platelet membranes) and inhibition (R/sub i/ in adipocyte membranes) were used as models of receptor activation. K/sub i/ values were obtained by Schild analysis. The orders of potency of the xanthines to attenuate the effects of adenosine analogues were similar to those previously reported. Earlier work utilizing radioligand binding (R/sub i/ and (/sup 3/H) cAMP formation (R/sub a/) claimed that IIX and PACPX are at least 10 and 400 fold, respectively, more potent at R/sub i/ than at R/sub a/. However, in their assays which utilize modulation of receptor-coupled adenylate cyclase, the xanthines show little specificity for either adenosine receptor subtype.

The imaging should be adapted to the pathology looked for and either CT or MRI should be used alone or in combination.CT, especially high resolution CT (HRCT), provides an excellent bone contrast, while MRI has a much superior soft tissue contrast. Acute inflammatory changes of the inner ear are solely depicted by contrast-enhanced MRI. HRCT excellently depicts osseous changes of the inner ear and cerebellopontine angle such as chronic ossifying labyrinthitis occurring after acute labyrinthitis, otosclerotic or traumatic changes. Tumorous changes not yielding to bony changes are best delineated by MRI. Posttraumatic hemorrhage and chronic fibrotic changes within the labyrinth are depicted by MRI, only. Biodiesel can be produced from numerous materials, including by-products of animal chain. In this scenario, the chicken's oil, that is located and an environment not explored for biodiesel production, can be a good alternative in South and Southeast Brazilian Region, where they are slaughtered around 9.100.000 chickens per day. In order to determine the specific consumption from a generator set with single cylinder 6.6 kW of power and generation capacity of 5 kW, using chicken oil biodiesel (B-100), were performed the bench test using a auxiliary tank, precision scale and an energy analyzer. Were conducted four treatments (25%, 50%, 75% and 100% of generating capacity). The worst SC was the engine at 25% of generation capacity (1.5 kW) and better with 75% of maximum load (3.75 kW) with a mean of 479.34 g kWh{sup -1}. Much of the current technology on 3D-prestack depth migration is based on the Kirchhoff method and the traveltime information needed for this purpose is obtained by the ray-theory approximation. The main motivation driving these options is to reduce computations but it can also have an adverse effect on the quality of results. Experience with real data indicates that the corresponding poststack data migrated by the reverse-time method often yields superior results. Considerable computational efficiency can be obtained by employing nonuniform coarse grid spacing during the reverse-time migration. We followed this strategy for developing a parallel scheme for doing 3D- prestack depth migration. However, we worked only on a small subset of shot gathers whose determined by the Quasi-Monte Carlo method. The results were indistinguishable from those obtained by using a much larger population of uniformly spaced shot gathers. We present examples of migration of 3-D data. (author)

To study the biogeochemical behaviour, mobility and transport of trace metals in seawater (Se, Mo, Cd, Pb, Hg, Cu, Co, Fe, Ni, Zn), in suspended particulate matter (Mo, Cd, Pb, Cu, Co, Fe, Ni, Zn, Al, Mn) and in sediments of the inner and outer coastal regions, samples were collected on 3 separate cruises during a one year period (Mai 1993 - April 1994) at 20 stations in the coastal area of Mecklenburg-Vorpommern. Standard hydrographic parameters, nutrients and dissolved oxygen were also measured. Priority was taken to establish the methods for the determination of molybdenum and selenium. To study the behaviour of these two elements in detail, longterm measurements were carried out weekly between January 1993 and June 1994 at the `Messsteg/Warnemuende`. The study examines the isothermal fatigue properties of novel ODS iron-base or nickel-base superalloys at temperatures of 850 C and 1050 C. The materials are different in grain structure and texture, so that particularly the effects of grain structures and textures can be studied. In order to also perform examinations of creep-fatigue interactions, various load cycles are applied. It is shown that the LCF-affected life of the grain structures of both alloy types is determined by the elastic modulus. The cyclic lifetimes determined by the conventional Manson-Coffin method for both alloy types exhibit differences of up to two orders of magnitude. Using the product of the total strain range and the elastic modulus, however, will yield precise life predictions. In the present study, the immunomodulatory effect of {beta}-endorphin ({beta}-E) and shorter pro-opiomelancortin (POMC) fragments was evaluated by assessing their influence on respiratory burst in human polymorphonuclear leukocytes (PMN). The effect of the peptides on phorbol myristate acetate (PMA)-stimulated production of reactive oxygen metabolites was measured in a lucigenin-enhanced chemiluminescence (CL) assay. Both POMC peptides with opiate-like activity and their non-opioid derivatives were tested. With the exception of {alpha}-E, PMA-stimulated respiratory burst was suppressed by all POMC fragments tested. A U-shaped dose-response relation was observed. An assay system for the stem cell that colonizes the thymus and differentiates into T cells was developed, and by using this assay system the existence of two subpopulations of stem cells for T cell lineage was clarified. Part-body-shielded and 900-R-irradiated C57BL/6 (H-2b, Thy-1.2) recipient mice, which do not require the transfer of pluripotent stem cells for their survival, were transferred with cells from B10 X Thy-1.1 (H-2b, Thy-1.1) donor mice. The reconstitution of the recipient's thymus lymphocytes was accomplished by stem cells in the donor cells and those spared in the shielded portion of the recipient that competitively colonize the thymus. The topographical distribution of this decrement was analyzed by quantitative receptor autoradiography. After 4 weeks of soman, three times a week, quinuclidinyl benzilate binding decreased to 67 to 80% of control in frontal and parietal cortex, caudate-putamen, lateral septum, hippocampal body, dentate gyrus, superior colliculus, nucleus of the fifth nerve, and central grey. Minor or no decreases were observed in thalamic or hypothalamic nuclei, reticular formation, pontine nuclei, inferior colliculus, nucleus of the seventh nerve, and cerebellum. The functional interaction between the peroxisome proliferator-activated receptor {gamma} (PPAR{gamma}) and its coactivator PGC-1{alpha} is crucial for the normal physiology of PPAR{gamma} and its pharmacological response to antidiabetic treatment with rosiglitazone. Here we report the crystal structure of the PPAR{gamma} ligand-binding domain bound to rosiglitazone and to a large PGC-1{alpha} fragment that contains two LXXLL-related motifs. The structure reveals critical contacts mediated through the first LXXLL motif of PGC-1{alpha} and the PPAR{gamma} coactivator binding site. Through a combination of biochemical and structural studies, we demonstrate that the first LXXLL motif is the most potent among all nuclear receptor coactivator motifs tested, and only this motif of the two LXXLL-related motifs in PGC-1{alpha} is capable of binding to PPAR{gamma}. The kainoid amino acids are biologically important compounds because they show remarkable neuroexcitatory and excitotoxic activities. For exhibiting potent activity, the stereochemical relationship of the substituents on the pyrrolidine ring is crucial. We found simple methods for determining the relative stereochemistry of these compounds on the basis of the (1)H NMR chemical shifts of H-2 and H-4 in D(2)O solution. The signals of H-2 appear at fields higher than 4.2 ppm when the compounds have 2,3-trans stereochemistry whereas, in the 2,3-cis compounds, they appear lower than 4.2 ppm, irrespective of the C-4 substituent. This criterion holds when the solution is in the range of pD 3-8. Moreover, when an epimeric pair at C-2 is available and the spectra are recorded at the same or nearly equal pD, the H-2 chemical shift of the 2,3-trans isomer is higher than that of the corresponding 2,3-cis isomer. Radishes (Raphanus sativus L.) are members of the cruciferous vegetable family that contain many classes of biologically active phytochemicals. This study determined the phytochemical composition of the sprouts and mature taproots of 8 radish varieties. Radish sprouts contained significantly greater concentrations of glucosinolates (3.8-fold) and isothiocyanates (8.2-fold) than the mature radish taproot and also contained significantly greater concentrations of phenolics (on average 6.9-fold). The anthocyanin concentrations of the mature radish taproot were significantly greater than in the sprouts of red, pink, and purple varieties. The primary anthocyanidins present in the red and pink radish varieties were pelargonidin and delphinidin, while the primary anthocyanidin in the purple radish variety was cyanidin. Synthetic atrial natriuretic peptide (carperitide) and B-type natriuretic peptide (BNP; nesiritide) are used to treat congestive heart failure. However, despite beneficial cardiac unloading properties, reductions in renal perfusion pressures limit their clinical effectiveness. Recently, CD-NP, a chimeric peptide composed of C-type natriuretic peptide (CNP) fused to the C-terminal tail of Dendroaspis natriuretic peptide (DNP), was shown to be more glomerular filtration rate-enhancing than BNP in dogs. However, the molecular basis for the increased responsiveness was not determined. Here, we show that the DNP tail has a striking effect on CNP, converting it from a non-agonist to a partial agonist of natriuretic peptide receptor (NPR)-A while maintaining the ability to activate NPR-B. Spike-timing-dependent plasticity (STDP), a form of Hebbian plasticity, is inherently stabilizing. Whether and how GABAergic inhibition influences STDP is not well understood. Using a model neuron driven by converging inputs modifiable by STDP, we determined that a sufficient level of inhibition was critical to ensure that temporal coherence (correlation among presynaptic spike times) of synaptic inputs, rather than initial strength or number of inputs within a pathway, controlled postsynaptic spike timing. Inhibition exerted this effect by preferentially reducing synaptic efficacy, the ability of inputs to evoke postsynaptic action potentials, of the less coherent inputs. In visual cortical slices, inhibition potently reduced synaptic efficacy at ages during but not before the critical period of ocular dominance (OD) plasticity. Cigarette smoking is the major environmental risk factor for bladder cancer in humans. Aromatic amines, potent DNA-reactive bladder carcinogens present in cigarette smoke, contribute significantly. However, increased cell proliferation, caused by direct mitogenesis or in response to cytotoxicity, may also play a role since urothelial hyperplasia has been observed in human cigarette smokers. We examined the urothelial effects of cigarette smoke (whole body inhalation exposure (Teague) system) in female C57BL/6 mice at various times in two studies, including reversibility evaluations. In both studies, no urothelial hyperplasia was observed by light microscopy in any group. However, in study 1, the Ki-67 labeling index (LI) of the urothelium was significantly increased in the smoke exposed group compared to controls through 3 months, but was not present at 6, 9 or 12 months even with continued exposures. The effects of the dopants, Mg{sup 2+}, Ca{sup 2+}, Sr{sup 2+}, Sc{sup 3+}, Yb{sup 3+}, Y{sup 3+}, Gd{sup 3+}, La{sup 3+}, Ti{sup 4+}, Zr{sup 4+}, and Nb{sup 5+}, on the grain boundary mobility of dense CeO{sub 2} have been investigated from 1,270 to 1,420 C. Parabolic grain growth has been observed in all instances. Together with atmospheric effects, the results support the mechanism of cation interstitial transport being the rate-limiting step. A strong solute drag effect has been demonstrated for diffusion-enhancing dopants such as Mg{sup 2+} and Ca{sup 2+}, which, at high concentrations, can nevertheless suppress grain boundary mobility. Severely undersized dopants (Mg, Sc, Ti, and Nb) have a tendency to markedly enhance grain boundary mobility, probably due to the large distortion of the surrounding lattice that apparently facilitates defect migration. In this study, we examined the chemopreventive effect of FPP-3 on 7,12-dimethylbenz[a]anthracene (DMBA)-induced genotoxicity in MCF-7 cells. FPP-3 reduced the formation of the DMBA-DNA adduct. DMBA-induced CYP1A1 and CYP1B1 gene expression and enzyme activity were inhibited by FPP-3. It inhibited DMBA-induced aryl hydrocarbon receptor (AhR) transactivation and DMBA-inducible nuclear localization of the AhR. Induction of detoxifying phase II genes by chemopreventive agents represents a coordinated protective response against oxidative stress and neoplastic effects of carcinogens. Oxidative stress is demonstrated to be involved in the pathophysiological mechanism of erectile dysfunction (ED). Quercetin, a potent bioflavonoid, has been reported to have the antioxidant role. In the present study, we examined the effect of quercetin on ED and oxidative stress in streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in Sprague-Dawley rats with a single intravenous injection of STZ. The diabetic rats were then randomized to diabetic group and quercetin therapy groups which were treated with quercetin at different doses of 5, 20 and 50mg/kg per day respectively. At the end of the 8th week, erectile function was assessed by measuring the rise in intracavernous pressure (ICP) following cavernous nerve electrostimulation. CNG vehicles suffer from an uncontrolled temperature rise of the gas inside the tank during the filling process and heat exchange with the environment leads to variable filling levels. A partly filled tank reduces the range of CNG vehicles and works as a impediment to the spreading of environmentaly more friendly CNG vehicles. The increase of the pressure inside the tank combined with a prolongation of the filling time beyond three minutes can reduce the deficit of the filling process. Moreover its radionuclides may represent an incorporation risk for members of the public and/or radiation workers after a nuclear accident or a release of radioactive materials. However, only few reliable data on Mo biokinetics in humans were available. The results of recent tracer kinetic investigations with stable isotopes have shown several differences from the ICRP data with regard to the processes of intestinal absorption and of excretion. As a consequence, the dose coefficients calculated with a revised biokinetic model deviate from the ICRP estimates. The distribution of graphite particles in graphite containing copper alloy was further improved very significantly using several procedures and technological modifications. The developed techniques attacked the graphite distribution problem in two ways. Realizing that clustering of very fine (5um) graphite particles is one of the two major problems, a pretreatment process has been developed using aluminum powders to deagglomerate graphite particles. Along with this, a two-stage stirring technique was used to first incorporate and then to distribute uniformly the deagglomerated particles in the melt. During this year, based on these developments, several components were cast to evaluate the castability of Cu alloy-graphite melts. The results of the creep performance studies at temperatures from 700-1000 C with welds of alloy 800 HT, the manual shielded metal arc welds made with Githerm 85 and Grini 209 filler metals, as well as the MIG pulsed arc welds made with the Nicrofer S 206 and Nicrofer S 5520 filler metals, proved to be in very good agreement with the calculated characteristic values published on VdTUeV sheet 434 in comparative assessments based on creep values valid for a service period of 10 000 hours. The MIG pulsed arc weld made with the Nicrofer S 5520 filler metal achieved by far the best results and strength values comparable to those of the base material, whereas the values of the other welds were corresponding to calculated strength values of the base material reduced by 20%. The effect of the heavy particle irradiation on cell survival is different from that of X-ray irradiation. In Japan, heavy particles will be applied to cancer patients in the near future. This study was performed to determine the effect that carbon particle irradiation exerts to human monolayer cells in vitro, in comparison with the effect of 200 kVp X-ray. Two human cell lines, HeLaS3 and RMUG, were used. The irradiation was carried out by the RIKEN ring cyclotron and 200 kVp X-ray. The experimental procedure is explained. Also split dose experiment was carried out by applying equally divided doses at intervals of 2 - 4 hours. The Bragg curve of 135 MeV/n carbon beam through the lucite plates with different thickness is given. Thus the different LETs were obtained. Normally, the alloy NiMo28 is used in the solution annealed and quenched condition in which it has a face centred cubic matrix structure. The matrix of the material is supersaturated with molybdenum and, at temperatures above 800 C, has a tendency to form intermetallic precipitates such as the Ni{sub 4}Mo-ordered phase leading to the embrittlement of the NiMo28-ordered phase leading to the embrittlement of the NiMo28 alloy during hot working. For this reason, the aim of these investigations was to optimise the alloy by reducing its tendency to embrittle. The TEM-analyses carried out served to investigate the precipitation of the material as a function of its iron content after ageing at 700 C. The change in hardness and in the lattice constants of the NiMo28-specimens with iron content were also determined. The practical experiences that have been gathered with these classification criteria and values were evaluated in a questionnaire-based survey. One part of the RWE measure catalogue in order to guarantee an environmental friendly and competitive electric power generation based on brown coal is to carry out measures improving the efficiency at existing steam turbines in RWE brown coal power plants. Since 1993 a total of 21 steam turbines with a gross capacity of 7,100 MW or net capacity of 6540 MW - this corresponds to approximately 70 % of the installed RWE brown coal power plant capacity - have been improved with an investment of approximately DM 500 million. The net block efficiency has been increased by 1.3 % on average. The planned net capacity increase of approximately 250 MW is still going to be achieved taking into account the pending enhancements. The transversus perinei profundus muscle is a well known structure in found anatomical textbook. Fibres of this muscle are believed to form the so-called external urethral sphincter (musculus sphincter urethrae). Recently histomorphologic investigations have shown that there is no muscular connection between the musculus sphincter urethrae (external sphincter) and the muscle system of the pelvic floor. Furthermore, the external sphincter was found to be divided into parts: The transversely striated part (musculus sphincter urethrae transversostriatus) and the smooth part (musculus sphincter urethrae glaber). In order to reduce the work required for the present combination of parameters, the following substances were selected: Hexachloroethane, hexachlorocyclohexane, hexachlorobenzene and hexachlorophene. The model substances were pyrolyzed or combusted in varying conditions, and the combustion or pyrolysis products were analyzed. Only the main products were of interest; trace components are important only as far as they may throw light on the reaction mechanisms. The furnace design resulted in incomplete combustion and pyrolysis reactions, so that products of all stages of combusition or pyrolysis were available for analysis. Reaction processes can be derived from the products. As an alternative to thermal treatment of refuse, biological degradation of chlorinated hydrocarbons was gone into. On the basis of cylindrical roller thrust bearings it was systematically examined to what extent PVD-coatings are able to take over the function of EP/AW-additives. The bearings were tested under heavy-duty conditions in order to distinguish very fast the efficiency of different coating-substrate-systems. Several coatings were tested for their ability to protect an un-coated counterpart. So just the washers of the roller bearings were coated, the rollers stayed un-coated. Four Me-C:H-coatings showed the best performance and fulfilled the required criterion for roller bearings in the boundary friction: low loss of mass and hardly surface deviation. Material carryover from the carbonaceous coating to the 100Cr6 steel surface was developed by the Me-C:H-coatings during the tests. This mechanism was able to protect the un-coated rollers. For a wider application of GaAs single crystals for semiconductor devices a cheaper process to grow crystals with high purity and low dislocation density is required. According to todays knowledge the Czochralski method with a hot wall vessel should be the choice. The temperature of the vessel has to be 700deg C to avoid condensation of arsenic which vapourizes from the 1240deg C hot melt. Arsenic vapour is very agressive against all metals. Therefore the corrosion resistance of metals and alloys with a high melting point and also ceramics has been tested. All the alloys under investigation are corroded severely so that they have to be excluded as a material for the vessel. The metals tantalum, molybdenum and tungsten are more resistent, titanium forms initially a thick protection layer and further corrosion is greatly reduced. The influence of a decarburisation process during austenitising of the through-hardenable rolling bearing steel 100Cr6 (SAE 52100) on the rim zone properties of the martensitic through-hardened material was investigated by means of material analysis and diffusion calculations. For this purpose, two specimens were prepared under defined heat treatment conditions, and the near-surface carbon distribution was determined micro-chemically by using secondary ion mass spectrometry (SIMS). The studies carried out by Rosalyn Yalow, Solomon Berson and Roger Ekins in the 1960s gave a breakthrough in the development of this novel analytical method. This thesis deals with the analysis of dominant loss mechanisms in direct methanol fuel cells (DMFC) and hydrogen fed polymer electrolyte membrane fuel cells (PEFC) by means of experimental characterization and modeling work. Due to different fuels used in these two fuel cell types, the dominant loss mechanisms are different in their nature. All in-situ characterization techniques that are used in this work are based on a novel test fuel cell with embedded reference electrodes. The first part of this work presents a new concept for realizing a reference electrode configuration in a polymer electrolyte membrane fuel cell by means of laser ablation. The laser beam is used to evaporate a small gap into the electrode surface of a catalyst coated membrane (CCM) to isolate the reference electrode from the working/counter electrode (WE/CE). To evaluate the current situation and implementation of embolization of uterine leiomyomas into the treatment concept in women with symptomatic uterine leiomyomas in Germany. A questionnaire addressing the clinical background of uterine myomas, recommended treatment concepts, preclinical evaluation, technical approach and complications was sent to 164 departments of gynecology and radiology in Germany. 33 radiological departments and 19 gynecological departments submitted a completed questionnaire. Only 7 departments of radiology reported to have own experience with embolization of uterine leiomyomas, while only 2 departments of gynecology considered embolization as an alternative treatment option in patients with symptomatic leiomyomas. 18/33 radiological departments offer this treatment option but get no patient referrals. The experimental effect investigation programme on Zircaloy-4/air oxidation was pursued and expanded to isothermal specimen exposure and the comparison of the oxidation between the atmospheres Ar/O{sub 2} and air. In close connection with the EC project OPSA, which is meanwhile terminated, the investigation concerned specimen exposure in a thermobalance to flowing atmospheres, namely dry Ar/O{sub 2} of composition 80/20 or synthetic air, respectively. As test parameters the linear heat-up rate was varied in the range 5 to 40 K/min and the ramp or holding temperature between 800 and 1500 C. Mass increase and reaction rate were continuously recorded or evaluated in temperature/time dependence. The oxide scale growth was found to be accompanied by colour changes, crack formation and spalling of layers, as well as by dimensional substrate growth, which are the reasons for the observed kinetic results. Low energy buildings promise significant energy savings and help climate preservation by cutting down on annual heating requirements to values below 70 kWh per m{sup 2} dwelling space. It is expected that low energy constructions will become the common standard by the year 2000 as a result of the forthcoming energy saving ordinance. For making the introduction and implementation of this standard successful, practical experience will be essential.
