A phytase gene (phyA), isolated from Aspergillus ficuum (AF537344), was introduced into cotton (Gossypium hirsutum L.) by Agrobacterium-mediated transformation to increase the phosphorus (P) acquisition efficiency of cotton. Southern and Northern blot analyses showed that the phyA was successfully incorporated into the cotton genome and expressed in transgenic lines. After growing for 45?days with phytate (Po) as the only P source, the shoot and root dry weights of the transgenic plants all increased by nearly 2.0-fold relative to those of wild-type plants, but were similar to those of transgenic plants supplied with inorganic phosphorus. Homozygous transgenic cotton (Gossypium hirsutum L.) plants that accumulated glycinebetaine (GB) in larger quantities were more tolerant to salt than wild-type (WT) plants. Four transgenic lines, namely 1, 3, 4, and 5, accumulated significantly higher levels of GB than WT plants did both before and after salt stress. At 175 and 275?mM NaCl, seeds of all the transgenic lines germinated earlier and recorded a higher final germination percentage, and the seedlings grew better, than those of the WT. Under salt stress, all the lines showed some characteristic features of salt tolerance, such as higher leaf relative water content (RWC), higher photosynthesis, better osmotic adjustment (OA), lower percentage of ion leakage, and lower peroxidation of the lipid membrane. An intact basement membrane (BM) is essential for the proper function, differentiation and morphology of many epithelial cells. The disruption or loss of this BM occurs during normal development as well as in the disease state. To examine the importance of BM during mammary gland development in vivo, we generated transgenic mice that inappropriately express autoactivating isoforms of the matrix metalloproteinase stromelysin-1. The mammary glands from these mice are both functionally and morphologically altered throughout development. We have now documented a dramatic incidence of breast tumors in several independent lines of these mice. These data suggest that overexpression of stromelysin-1 and disruption of the BM may be a key step in the multi-step process of breast cancer.

We have focused this phytoremediation research on soil and water-borne ionic and methylmercury. Mercury pollution is a serious world-wide problem affecting the health of human and wild-life populations. Methylmercury, produced by native bacteria at mercury-contaminated wetland sites, is a particularly serious problem due to its extreme toxicity and efficient biomagnification in the food chain. A new area-wide pest control strategy using the pink bollworm, Pectinophora gossypiella (Saunders) (Lepidoptera: Gelechiidae), genetically transformed with a conditionally lethal gene, is under development. Conditional lethality of several transgenic pink bollworm strains was demonstrated in a series of laboratory rearing experiments. Pink bollworms were transformed with genetic constructs using the RIDL technology (Release of Insects with a Dominant Lethal gene) for development of an autocidal biological control system for possible supplement or replacement of radiation based sterile insect release. Neuron-restrictive silencer factor (NRSF) binds its consensus element to repress the transcription of various genes. The dominant-negative form (dnNRSF) has a hypertrophic effect on cardiogenesis through an unidentified mechanism. We examined the involvement of transient receptor potential (TRP) channel proteins, using transgenic mice overexpressing dnNRSF (dnNRSF mice). Electrophoretic mobility-shift assays revealed an interaction between NRSF and a neuron-restrictive silencer element-like sequence in intron 4 of TRPC1 genomic DNA. According to RT-PCR and Western analyses, TRPC1 was up-regulated in dnNRSF mouse heart. Transient overexpression of TRPC1 in HEK 293T cells increased the activity of the nuclear factor in activated T cells (NFAT) promoter and stimulated store-operated Ca"2"+ channel (SOCC)-mediated Ca"2"+ entry. A tk{sup +/-} mouse embryonic stem (ES) cell line, designated 1G2, has been created in which one allele of the thymidine kinase (tk) gene was inactivated by targeted homologous recombination. This line is an analog of the mouse lymphoma tk{sup +/-} L5178Y cell line, which is used widely to assess the mutagenicity of chemical agents. Treatment of 1G2 cells with the alkylating agent N-ethyl-N-nitrosourea (ENU) resulted in a dose-related increase in tribluorothymidine-resistant colonies. Mutant frequencies of 152 and 296 per 10{sup 6} cells were determined for 0.1 and 0.3 mg/ml doses of ENU, compared with a spontaneous mutant frequency of 15 per 10{sup 6} cells. The data indicate that tk{sup +/-} 1G2 ES cells may be useful for the creation of a transgenic mouse model for assessing in vivo mutation using an endogenous autosomal gene. 45 refs., 2 figs., 1 tab.

Carbon supported catalysts can lose their activity over a period of time due to the sintering of the nanometer-sized catalyst particles. The sintering of metal clusters on carbon supports can occur due to the weak interaction between the metal and the support and also due to the corrosion of carbon, especially in fuel cell electrocatalysts. The sintering may be reduced by increasing the interaction between the metal and the support and also by increasing the corrosion resistance of carbon supports. One of the most complicated tasks during the assembly of the Wendelstein W7-X is the installation of the superconductive coil system. The entire magnet system is enclosed between the Outer Vessel and the plasma vessel in high vacuum at 4 K. The coils are supported by a support structure, the Central Support Structure (CSS). The CSS carries all 70 coils, and it is designed as a closed ring made of 5 modules (10 half modules). The whole structure is supported by 10 Cryolegs which rest on the machine base. Vascular endothelial growth factor (VEGF) is a mitogen that is critically involved in vasculogenesis, angiogenesis, and hematopoiesis. However, what and how transcription factors participate in the regulation of vegf gene expression are not fully understood. Here we report the cloning and sequencing of the zebrafish vegf promoter which revealed that the promoter contains a number of bone morphogenetic protein (BMP)-activated Smad binding elements (SBE), implicating Smad1 and Smad5 in the regulation of BMP-induced expression of vegf. Electrophoretic mobility shift assays of adding recombinant Smad proteins to the SBE-containing DNA oligonucleotides that represent portions of zebrafish vegf promoter resulted in mobility shift of the oligonucleotides. These changes demonstrate potential interactions between Smad1/5 and the vegf promoter. Background The identification of sequences that control transcription in metazoans is a major goal of genome analysis. In a previous study, we demonstrated that searching for clusters of predicted transcription factor binding sites could discover active regulatory sequences, and identified 37 regions of the Drosophila melanogaster genome with high densities of predicted binding sites for five transcription factors involved in anterior-posterior embryonic patterning. Nine of these clusters overlapped known enhancers. Here, we report the results of in vivo functional analysis of 27 remaining clusters. Results We generated transgenic flies carrying each cluster attached to a basal promoter and reporter gene, and assayed embryos for reporter gene expression. Six clusters are enhancers of adjacent genes: giant, fushi tarazu, odd-skipped, nubbin, squeeze and pdm2; three drive expression in patterns unrelated to those of neighboring genes; the remaining 18 do not appear ...

ABSTRACT Extending the unified theory of acceptance and use of technology (UTAUT) model, this study postulates a model of online social support. The model is empirically tested using data from undergraduates in Taiwan regarding their usage of instant messaging (IM). The test results indicate that all model paths are significant, except that the path between online social support and facilitating conditions is insignificant. This study offers limitations and implications.

This study validates a research model that examines usage of instant messaging (IM) from the aspect of online social support. Drawing on the social capital theory, this study postulates that IM usage is indirectly affected by social support via the mediation of the following six dimensions of social capital: commitment, reciprocity, shared codes and language, shared narratives, centrality, and network ties. The model tests data obtained from business organizations in Taiwan, and the results suggest that the indirect influence of social support on IM usage through shared codes and language is significant, and the indirect influence of social support on IM usage through centrality is also significant. Managerial implications and limitations of the empirical findings are provided.

This paper describes examples of the damage resulting from the impact of a whipping pipe on a nearby pressurised pipe. The work is a by-product of a study of the motion of a whipping pipe. The tests were conducted with small-diameter pipes mounted in rigid supports and hence the results are not directly applicable to large-scale plant applications where flexible support mountings are employed. The results illustrate the influence of whipping pipe energy, impact position and support type on the damage sustained by the target pipe.

The present paper is a comparative study of the cerium-containing nickel catalysts supported on x- and y-zeolites. In general, the addition of cerium ions caused an increase in the catalytic activity for CO hydrogenation and shifted the product selectivity to high molecular weight hydrocarbons. The degree of the effect of cerium additive depends on the ratio of cerium to nickel contents, the reduction temperature, and the nature of the supporting materials. Catalyst characterization, including volumetric hydrogen chemisorption, temperature programmed reduction/desorption, x-ray diffraction, surface area measurements, in-situ infrared spectroscopy and x-ray photoelectron spectroscopy, was performed in order to interpret the phenomena due to the effects of cerium additives and the support effect on Ni/zeolite catalysts.

The Buried Waste Integrated Demonstration (BWID) is a program of the Department of Energy (DOE) Office of Technology Development whose mission is to evaluate different new and existing technologies and determine how well they address DOE community waste remediation problems. Twenty-three Technical Task Plans (TTPs) have been identified to support this mission during FY-92; 10 of these have identified some support requirements when demonstrations take place. Section 1 of this report describes the tasks supported by BWID, determines if a technical demonstration is proposed, and if so, identifies the support requirements requested by the TTP Principal Investigators. Section 2 of this report is an evaluation identifying facility characteristics of existing Idaho National Engineering Laboratory (INEL) facilities that may be considered for use in BWID technology demonstration activities.

This paper discusses the experience at Darlington in building and maintaining station teamwork among the station workgroups in support of day-to-day plant operations. In the WWER-440 reactor the primary piping consists of six horizontal loops going radially from the pressure vessel, each loop having a horizontal steam generator. In this reactor type the relatively long primary piping with many curved sections requires special attention in order to successfully eliminate the consequences of the design basis accident. Emergency supports are located in appropriate places to restrict the movements of the pipe in 1, 2, 3 or 4 directions depending on the geometry of the pipe near the support. Under normal conditions there is a gap of some centimeters between the pipe and a support so that the pipe can be deformed freely under changing loads. The overall objective of this research project is to develop a technical basis for flexible piping designs which will improve piping reliability and minimize the use of pipe supports, snubbers, and pipe whip restraints. The current study was conducted to establish the necessary groundwork based on the piping reliability analysis. A confirmatory piping reliability assessment indicated that removing rigid supports and snubbers tends to either improve or affect very little the piping reliability. We then investigated a couple of changes to be implemented in Regulatory Guide (RG) 1.61 and RG 1.122 aimed at more flexible piping design. We concluded that these changes substantially reduce calculated piping responses and allows piping redesigns with significant reduction in number of supports and snubbers without violating ASME code requirements. Results from a heavily fouled 19 tube C-E model boiler test to investigate the potential for egg crate corrosion in aggressively fouled AVT chemistry are reported. Substantial support plate and egg crate corrosion was produced in this test. Carbon steel drilled support plates exhibited extensive denting which resulted in flow hole ligament cracking. Corrosion of the carbon steel egg crate, through-wall at areas of tube contact, resulted in denting of the Alloy 600 heat transfer tubes. Corrosion performance of the 409 stainless steel egg crate was improved compared to the carbon steel egg crate although localized through-wall corrosion was noted. The overall objective of this research project is to develop a technical basis for flexible piping designs which will improve piping reliability and minimize the use of pipe supports, snubbers, and pipe whip restraints. The current study was conducted to establish the necessary groundwork based on the piping reliability analysis. A confirmatory piping reliability assessment indicated that removing rigid supports and snubbers tends to either improve or affect very little the piping reliability. A couple of changes to be implemented in Regulatory Guide (RG) 1.61 and RG 1.122 aimed at more flexible piping design were investigated. It was concluded that these changes substantially reduce calculated piping responses and allows piping redesigns with significant reduction in number of supports and snubbers without violating ASME code requirements.

At atmospheric pressure and room temperature, dielectric barrier discharge induced plasma oxidation for achieving supported TiO2 photocatalysts derived from TiCl4 adsorbed onto ?-Al2O3 pellets was studied. The supported TiO2/?-Al2O3photocatalysts prepared by a cyclic 'adsorption-discharge' approach, without requirement of heat treatment, exhibit high activity in the photocatalytic degradation reaction of formaldehyde. The mass spectra and optical emission spectra during O2/Ar discharge for oxidizing the adsorbed-state TiCl4 were measured. The mechanism for the TiO2 formation from adsorbed-state TiCl4 by plasma oxidation was discussed.

Personal health information management (PHIM) refers to activities that support consumers' access, integration, organization, and use of their personal health information. We investigated PHIM in the health consumer population using a focus group and participatory design. In collaboration with health consumers, we identified PHIM activities and explored the design of new supportive technology. Our findings describe prominent PHIM activities such as monitoring and assessing health, as well as health-related decision making, planning, and action. We describe design principles our participants used during the participatory design of a PHIM tool. These include individual control, sharing, integration, security and flexibility. These findings provide new insights into emerging ideas in consumer health informatics research and technology design. This very extensive estimating system is capable of supporting cost estimators from many different and varied fields. QUEST does not dictate any fixed method for estimating. QUEST supports many styles and levels of detail estimating. QUEST can be used with or without data bases. This system allows the estimator to provide reports based on levels of detail defined by combining work breakdown structures. QUEST provides a set of tools for doing any type of estimate without forcing the estimator to use any given method. The level of detail in the estimate can be mixed based on the amount of information known about different parts of the project. The system can support many different data bases simultaneously. Estimators can modify any cost in any data base.

In order to improve operating reliability of the device during unloading on the cart during drilling and placement of the elevator and movement of the foundation within the area of the cluster of boreholes, it is equipped with controllable rods for connecting the carts to one another with the power component. This article presents the essential features of programs for students with EBD. Program elements include (a) qualified and committed professionals, (b) utilitarian environmental supports, (c) effective behavior management plans, (d) valid social skill and social interpretation training and social interaction programs, (e) proven academic support systems, (f) strong parent- and family-involvement programs, (g) coordinated community support mechanisms, and (h) ongoing evaluation of essential program components and student outcomes and progress. This article examines reported willingness to pay for electric utility investments in renewable energy and energy efficiency resources using the results of Deliberative Polls conducted by electric utilities in Texas. Age, education, and salary tend to affect reported willingness to pay for utility investments in these resources. Intensive exposure to information about energy resource issues led to an increase in the number of respondents interested in paying a modest premium to support investments in both renewable energy resources and energy efficiency. However, average reported premiums actually declined following the Polls, as very high outlier responses moved to more reasonable values following the Polls. These results support the contention that informed dialogue about energy alternatives will result in broader interest in providing a modest level of support for these resources. (author)

Hydrogenation of carbon monoxide at 101.3 kPa has been studied over a series of ion-exchanged Ru catalysts supported on NaX, NaY, KL, Na mordenite, and HY zeolites. The type of zeolite had pronounced effects on the activity and selectivity of the Ru. The specific activity would appear to be related to the dispersion of reduced ruthenium in the zeolite. Methane selectivity, however, seems to be strongly influenced by the type and concentration of alkali cations remaining in the ion-exchanged zeolite. These cations appear to promote chain growth much as traditional alkali promoters would, though perhaps more indirectly. Due to bifunctional properties of the zeolite-supported catalysts, a significant fraction of C/sub 4/ was in the form of isobutane. Formation of isobutane seems to be related to either the Si/Al ratio in the zeolites or the concentration of the remaining alkali cations, but not to the OH concentration. In addition, H/sub 2/ ...

A test methodology for measuring temperature, impedance, pH, and electrochemical potential distributions within a sludge-packed tube support plate crevice in a laboratory test is described. The method successfully showed that there were large concentration gradients between the tube and tube support plate sides of the crevice. The testing also showed that strong bases concentrated more effectively than strong acids, and that the crevice pH, when exposed to seawater-based solutions, increased with increasing superheat and decreasing bulk concentration. The large variations in the crevice chemistry observed under heat transfer were eliminated upon shutdown. These new test data suggest that it might be beneficial to evaluate the variation in the extent of stress corrosion cracking with tube support plate elevation found in some steam generators in light of local chemistry changes, as well as the variation in tubing ...
