The Geometry of Stable Quotients in Genus One

CERN Document Server

Stable quotient spaces provide an alternative to stable maps for compactifying spaces of maps. When the target is projective space and the domain curve has genus 1, these are smooth proper Deligne-Mumford stacks. In this paper we study the associated coarse moduli schemes. We show these schemes are projective, rationally connected and have Picard number 2. Then we give generators for the Picard group, compute the canonical divisor, and the cones of ample and effective divisors. In certain cases, we also give a closed formula for the Poincar\\'{e} polynomial.

Therapeutic efficacy evaluation of "1"1"1in-VNB-liposome on human colorectal adenocarcinoma HT-29/luc mouse xenografts

International Nuclear Information System (INIS)

The purpose of this study is to evaluate the therapeutic efficacy of the liposome encaged with vinorelbine (VNB) and "1"1"1In-oxine on human colorectal adenocarcinoma (HT-29) using HT-29/luc mouse xenografts. HT-29 cells stably transfected with plasmid vectors containing luciferase gene (luc) were transplanted subcutaneously into the male NOD/SCID mice. Biodistribution of the drug was performed when tumor size reached 500-600 mm"3. The uptakes of "1"1"1In-VNB-liposome in tumor and normal tissues/organs at various time points postinjection were assayed. Multimodalities, including gamma scintigraphy, bioluminescence imaging (BLI) and whole-body autoradiography (WBAR), were applied for evaluating the therapeutic efficacy when tumor size was about 100 mm"3. The tumor/blood ratios of "1"1"1In-VNB-liposome were 0.044, 0.058, 2.690, 20.628 and 24.327, respectively, at 1, 4, 24, 48 and 72 h postinjection. Gamma scinitigraphy showed that the ...

Diagnostic and therapeutic evaluation of "1"1"1In-vinorelbine-liposomes in a human colorectal carcinoma HT-29/luc-bearing animal model

International Nuclear Information System (INIS)

Colorectal carcinoma is a highly prevalent and common cause of cancer in Taiwan. There is still no available cure for this malignant disease. To address this issue, we applied the multimodality of molecular imaging to explore the efficacy of diagnostic and therapeutic nanoradiopharmaceuticals in an animal model of human colorectal adenocarcinoma [colorectal cancer (CRC)] that stably expresses luciferase (luc) as a reporter. In this study, an in vivo therapeutic efficacy evaluation of dual-nanoliposome (100 nm in diameter) encaged vinorelbine (VNB) and "1"1"1In-oxine on HT-29/luc mouse xenografts was carried out. HT-29/luc tumor cells were transplanted subcutaneously into male SCID mice. Multimodality of molecular imaging approaches including bioluminescence imaging (BLI), gamma scintigraphy, whole-body autoradiography (WBAR) and in vivo tumor growth tracing, histopathology and biochemistry/hematology analyses were applied ...

Development of recombinant adeno-associated virus and adenovirus cocktail system for efficient hTERTC27 polypeptide-mediated cancer gene therapy.

Science.gov (United States)

The low in vivo transduction efficiency of recombinant adeno-associated virus (rAAV) and the undesirably strong immunogenicity of adenovirus (rAdv) have limited their clinical utilization in cancer gene therapy. We have previously demonstrated that intratumoral injection of rAAV expressing a C-terminal polypeptide of human telomerase reverse transcriptase (rAAV-hTERTC27) effectively inhibits the growth of glioblastoma xenografts in nude mice. To further improve its efficacy, we combined rAAV-hTERTC27 with rAdv and investigated the efficiency of the cocktail vectors in vivo. At a nontherapeutic dose (1 x 10(8) plaque-forming units (PFUs)), rAdv-null and rAdv-hTERTC27 were equipotent in enhancing the therapeutic efficacy of rAAV-hTERTC27 (1.5 x 10(11) v.g.), and complete tumor regression was achieved in 25% of the treated animals. Importantly, the combination of rAAV-hTERTC27 and a therapeutic dose (2.5 x 10(9) PFU) of rAdv-hTERTC27 significantly augmented the therapeutic effects and led ...

Affine cones over Fano threefolds and additive group actions

CERN Document Server

We address the following question: When an affine cone over a smooth Fano threefold admits an effective action of the additive group? In this paper we deal with Fano threefolds of index 1 and Picard number 1. Our approach is based on a geometric criterion from our previous paper, which relates the existence of an additive group action on the cone over a smooth projective variety X with the existence of an open polar cylinder in X. Non-trivial families of Fano threefolds carrying a cylinder were found in loc. cit. Here we provide new such examples.

Auxiliary analyses in support of performance assessment of a hypothetical low-level waste facility: Two-phase flow and contaminant transport in unsaturated soils with application to low-level radioactive waste disposal. Volume 2

Energy Technology Data Exchange (ETDEWEB)

A numerical model of multiphase air-water flow and contaminant transport in the unsaturated zone is presented. The multiphase flow equations are solved using the two-pressure, mixed form of the equations with a modified Picard linearization of the equations and a finite element spatial approximation. A volatile contaminant is assumed to be transported in either phase, or in both phases simultaneously. The contaminant partitions between phases with an equilibrium distribution given by Henry`s Law or via kinetic mass transfer. The transport equations are solved using a Galerkin finite element method with reduced integration to lump the resultant matrices. The numerical model is applied to published experimental studies to examine the behavior of the air phase and associated contaminant movement under water infiltration. The model is also used to evaluate a hypothetical design for a low-level radioactive waste disposal facility. The model has been developed in both ...

In vitro evaluation of herpes simplex virus type 1 thymidine kinase reporter system in dynamic studies of transcriptional gene regulation

Energy Technology Data Exchange (ETDEWEB)

The herpes simplex virus type 1 thymidine kinase (HSV1-TK) reporter system is being used to directly and indirectly monitor therapeutic gene expression, immune cell trafficking and protein-protein interactions in various living animals. However, the issues of HSV1-TK enzyme stability in living cells and whether this reporter system is optimal for dynamic studies of gene expression events in genetic imaging have not be addressed. The purpose of the present study was to evaluate the application of this reporter system in dynamic studies of transcriptional gene regulation. To achieve this purpose, we established two tetracycline-inducible murine sarcoma cell lines, tetracycline-turn-off HSV1-tk-expressing cell line (NG4TL4/tet-off-HSV1-tk) and tetracycline-turn-off Luc-expressing cell line (NG4TL4/tet-off-Luc), to create an artificially regulated gene expression model in vitro. The dynamic transcriptional events mediating a series of doxycycline ...

UNSAT-H Version 3.0: Unsaturated Soil Water and Heat Flow Model Theory, User Manual, and Examples

Energy Technology Data Exchange (ETDEWEB)

The UNSAT-H model was developed at Pacific Northwest National Laboratory (PNNL) to assess the water dynamics of arid sites and, in particular, estimate recharge fluxes for scenarios pertinent to waste disposal facilities. During the last 4 years, the UNSAT-H model received support from the Immobilized Waste Program (IWP) of the Hanford Site's River Protection Project. This program is designing and assessing the performance of on-site disposal facilities to receive radioactive wastes that are currently stored in single- and double-shell tanks at the Hanford Site (LMHC 1999). The IWP is interested in estimates of recharge rates for current conditions and long-term scenarios involving the vadose zone disposal of tank wastes. Simulation modeling with UNSAT-H is one of the methods being used to provide those estimates (e.g., Rockhold et al. 1995; Fayer et al. 1999). To achieve the above goals for assessing water dynamics and estimating recharge rates, the UNSAT-H model addresses ...
