The authors discuss radioimmunoassays (RIAs) for eicosanoids widely used in the field of experimental, clinical and pharmacological research. The main drawback of RIAs is the possibility of low specificity. This was clearly demonstrated during the controversy on plasma levels of 6-keto-PGF_1 #alpha#, the stable hydrolysis product of PGI_2. When extensive purification including reverse phase high pressure liquid chromatography (rp-HPLC) was performed prior to the RIA for 6-keto-PGF_1 #alpha# the levels were at least one order of magnitude less than previously claimed, and did not support the so-called circulating hormone hypothesis.
Objective: To develop a method for RIA of transferrin without dilution of the serum sample. Methods: The measurable range was broadened through: Lowering of the specific radioactivity of the labelled antigen, increasing the concentration of the anti-serum and improve the curve fitting method. Results: The antiserum affinity was 3.82 x 10"-"1"0 mol/L, the specific radioactivity of the labelled antigen was 15.3 mg/mic, inter-assay error CV 5.38%, intra-assay error CV = 3.5% and the shape of the curve was good. For 30 normal control specimens, the mean TF content was 2.81 +- 0.972 mg/ml. Conclusion: Development of a method for RIA of TF without previous dilution of the test serum was of clinical value

Gel filtration of human erythrocyte (RBC) lysate incubated with labeled thyroxine (Tu) or triiodothyronine (Tt) revealed co-elution of a major iodothyronine-binding fraction (R-2) and hemoglobin. Solutions of purified human hemoglobin and Tt also showed co-elution of hormone and hemoglobin. Because hematin and protoporphyrin were shown to bind labeled Tt, the oxygen-binding site on hemoglobin was excluded as the site of iodothyronine-hemoglobin interaction. Analysis of hormone binding by heme and globin moieties showed Tt binding to be limited to the heme fraction. Addition of excess unlabeled Tt to hemoglobin or heme incubated with labeled Tt indicated 75% to 90% of hormone binding was poorly dissociable. These observations suggested that the presence of hemoglobin in RBC lysate or in serum could influence the measurement of Tu and Tt by specific radioimmunoassay (RIA). The human articular cartilagineous proteoglycans (PG) R.I.A. is highly specific. The PG used as the standard and the /sup 125/I labelled molecule appear to be pure. Under these conditions, all the potential interfering substances which have been tested show no cross reaction. For instance, the Ag-Ab equilibrium is not affected by adding human IgG, human albumin, hyaluronic acid, chondroitin sulfate, rat type II collagen or total human serum proteins. This R.I.A. also exhibits a species spcificity since there is no cross reaction with rat PG and negligible cross section with dog PG. The results obtained after addition of enzymes to the antigen demonstrate that the antigenic sites are localized on the protein region and not on the glycosaminoglycan region of the molecule.
Serum heat-stable alkaline phosphatase (HSAP) level in 649 cases of normal pregnancy and 164 cases of high-risk pregnancy is measured by radioimmunoassay (RIA). The results indicate that the HSAP level in normal pregnancy increased proportionally with gestation weeks (r = 0.9843). In 33 cases of pregnancy induced hypertension and 21 cases of intrauterine fetal growth retardation, the HSAP level is significantly low. In 7 cases of neonatal asphyxia and 26 cases of fetal distress, the HSAP level in the mother's serum is also low. In 53 cases of intrahepatic cholestasis of pregnancy, the HSAP level is similar to those of normal pregnancy. This study illustrates that HSAP RIA can play an important role in the evaluation of placental function and fetal prognosis for cases of high-risk pregnancy.
A case report of falsely elevated serum hormone values measured by radio-immunoassay (RIA) is described. The radio-immunoassays concerned have a first antibody raised in rabbits and mostly a separation technique based on a second antibody-solid phase system. The presence of heterophyle (anti-rabbit) antibodies in patients' serum is proved.
We present an empirical model for periodic variations of diurnal and sub-diurnal Earth rotation parameters (ERPs) that was derived based on the transformation of normal equation (NEQ) systems of Very Long Baseline Interferometry (VLBI) observing sessions. NEQ systems that contain highly resolved polar motion and UT1-TAI with a temporal resolution of 15?min were generated and then transformed to the coefficients of the tidal ERP model to be solved for. To investigate the quality of this model, comparisons with empirical models from the Global Positioning System (GPS), another VLBI model and the model adopted by the conventions of the International Earth Rotation and Reference Systems Service (IERS) were performed. The integrity of the RPV head and reactor internals was assessed by means of fluid-structural analyses using a coupled method to evaluate the water hammer phenomenon arising from high burnup fuel failure under RIA conditions. The fluid viscosity effect on the water column burst as well as the complex three-dimensional flow paths caused by a core shroud and standpipes were considered in this study. The three analysis scenarios were designed to investigate the above mentioned influential factors separately. In the first scenario, a two-dimensional axisymmetric reactor vessel model without any reactor internals was modeled to assess the influence of the fluid dynamics in the NSC RIA regulatory evaluation. To make the surface composites with hard ceramics particles is an effective means for satisfying the request for the wear-resistance of the casting products. Covering casting is a general method for forming the composites by adding the adhesive into ceramics particles. However, due to the using of adhesive, pore and intermediate are easy to generate, and it is one of the reasons making the wear-resistance unstable. In the present study, it is attempted to form the wear-resistant composite layer by dispersing cast tungsten carbide (CTC) particles on the surface of wear-resistant 7 mass% Mn steel without using adhesives. Subsequently, the experiments on the wear-resistance of the obtained surface composites under several wear conditions are carried out. In order to clarify the alkali-resistance of fibers to be utilized as reinforcing materials for concrete, such as glass fiber, Aramid fiber, and carbon fiber, accelerated aging tests in alkaline solution were conducted. In detail, the degree of degradation of the fracture strength and Young's modulus were investigated through static tensile strength tests after soaking in alkaline solution of these fibers and then the mechanism of degradation was examined by observation using a scanning electron microscope. The conclusion obtained was as follows. Glass fiber was easily degraded in the alkaline solution and the progress of degradation was simulated quantitatively by the method proposed in this paper, which was based on a diffusion theory. Both Aramid fiber and carbon fiber showed a significant alkali-resistance without degrading. The effect of atmospheric pressure on tracking resistance of organic insulating materials were investigated in the range of 500 to 1007 hPa. The tracking resistance for Polycarbonate (PC), Modified Polyphenylene Oxide (M-PPO) and Paper Base Phenolic Resin Laminate (PL) increase with the decrease in atmospheric pressure. The scintillating discharges on these samples surface lead to the ignition. The tracking failure on these results from the ignition on the surface. For Polybutylene Terephthalate (PBT) and Polyethylene Terephthalate (PET), the tracking resistance decrease with the decrease in atmospheric pressure. The discharges observed on the these surface are the glow-like. The discharge area spreads on the surface between the electrodes at the lower pressure. The failure on this group results from the carbonization by the discharges on the surface. The difficulty and unstability of the parameter estimation of mass action model for radioimmunoassay are the major obstacles to its application in routine work. A unique robust curve fitting method for the mass action model was introduced. large numbers of simulated data were generated with Monte Carlo method to compare the accuracy and robustness of the robust fitting and the least squre fitting (Marquardt method) of the mass action model. The results demonstrated that the new robusst method significantly increased the accuracy and reliability of data processing with the mass action model. It also markedly reduced the influence of outliers in the fitting of RIA data, and it was always 'convergent'. This robust method could also be applied to other mathematical models which can be transformed into high degree equations and no initial estimates are needed for curve fitting.
Full text.National programs of neonatal screening were introduced in most countries and have proved their effectiveness against different types of retardation in children. Congenital hypothyroidism is one of the most common causes of physical and mental retardation in children. Programs of screening have shown that the incidence of this disorder is ethnic-background: it is low in Africans: 1-32000 whereas it is 1-3500 and 1-5000 in Hispanic and European newborns respectively. No previous mass screening programs were done in Lebanon. For this purpose, this program was introduced in collaboration with the IAEA. Cord blood is spotted into circles on filter paper just after delivery. These samples are air-dried at room temperature and kept at 4 degree C. They are then collected from hospitals to be screened. The principal object of this trial was to evaluate the influence of three alternatives of feeding suplementation in dairy cows in the post-partum period in ecuadorian highlands. Thirty sic animals in fist lactation were used in this experiment and were divided in three groups according to the feed intake: Group A diet was 5 Kg. of a commercial concentrate mixture with 12 per cent of crude protein plus pasture ad libitum; Group B diet was green banans (Musa paradisiaca) and pasture and Group C diet was the control only pasture. Objective: To study the changes of serum leptin, insulin and adiponectin levels and their relationship with BMI in females with simple obesity. Methods: Serum leptin, adiponectin and insulin levels were measured with RIA in 48 pre-obese females (BMI=23-24.9 kg/m"2), 40 females with simple obesity, (BMI#>=#25 kg/m"2) and 42 female controls (BMI 18-22.9 kg/m"2). Correlations among these variables were studied. Results: Serum leptin, insulin levels were significantly higher and serum adiponectin levels were significantly lower in both the pre-obese and obese females than those in controls. Serum leptin, insulin levels were positively correlated to BMI; Serum adiponectin levels were negatively correlated to BMI. Conclusion: Within normal range of BMI, the leptin-insulin feedback mechanism provided satisfactory self-regulation. However, with excessive BMI, this dynamic equilibrium would be disrupted. The intracellular A chromogranin (ACg) is specific in neuroendocrine cells. This work is a contribution to the study of clinical interest of serumal dosage of this protein in a series of patients carrying neuroendocrine tumors (NET). From January `94 to November `96, 143 patients were subjected to OctreoScan scintigraphy and/or to MIBG. In 104 of them a dosage of ACg by RIA method was effectuated. A study of a statistical relation was performed on the rate of ACg and histopathological, clinical and scintigraphic criteria. Analysis of results excluded the patients with abnormal creatininemy; it referred exclusively to the patients the histopathology of whose it was proved. The global results concerning the sensitivity and specificity of ACg are: 68% and 92%, respectively, for a threshold of normality at 100 ng/ml; 65% and/or 100% for a threshold at 110 ng/ml. An experiment was conducted to evaluate the circadian rhythms of cortisol, thyroxine (T/sub 4/) and triiodothyronine (T/sub 3/) in pigs under two temperature and feeding regimes. Twenty-eight barrows were randomly assigned to one of the following: 1) ad-libitum fed at 5/sup 0/C(AL-5); 2) ad-libitum fed at 20/sup 0/C(AL-20); 3) meal fed at 5/sup 0/C(M-5); and 4) meal fed at 20/sup 0/C(M-20). M-5 and M-20 animals were fed at 0730 and 1400 hrs. Lights were on from 0600 to 2000 hrs. After 5 wks, blood samples were collected for 27 hrs. Serum cortisol, T/sub 4/ and T/sub 3/ concentrations were determined by RIA. No significant differences were found in the mesors, amplitudes or acrophases for cortisol. The mesors for T/sub 4/ (p<.01) were 60.6 +/- 5.6, 40.2 +/- 5.6, 61.2 +/- 5.6 and 49.1 +/- 5.0 ng/ml for AL-5, AL-20, M-5, and M-20, respectively. A factor produced by the plerocercoid stage of S. mansonoides mimics some, but not all, of the actions reported for hGH. The biological actions of plerocercoid growth factor (PGF) suggest structural similarity to human GH (hGH). Plerocercoid membranes were solubilized, and PGF was purified more than 1000-fold by hGH receptor affinity chromatography. The ability of purified PGF to displace [125I]hGH from monoclonal antibodies specific for four distinct nonoverlapping antigenic determinants of hGH and from an anti-hGH polyclonal antibody was tested in liquid phase RIA.
