We have been investigating radiation-induced neoplastic transformants of C3H 10T1/2 mouse fibroblasts for evidence of heritable changes. C3H 10T1/2 cells were treated with 8 Gy X rays. After approximately 8 weeks of culture, type II/III foci were isolated from the monolayer using cloning rings. Cell lines developed from these foci, and clones established from these cell lines, were examined for DNA content. The isolated focus-derived populations and derived clones often display aneuploidy and/or polyploidization. In one instance a clone (derived from a single cell) displayed multiple polyploidies. During passage the ploidy of many of the anomalous populations gradually reverted to the ploidy of the non-neoplastically transformed state. Poncirin, flavanone glycoside, isolated from the fruit of Poncirus trifoliata, has anti-bacterial and anti-inflammatory activities. In this study, the effects of poncirin on the differentiation of mesenchymal stem cells were investigated. The C3H10T1/2 mesenchymal stem cells and primary bone marrow mesenchymal stem cells were studied. In the C3H10T1/2 cells, poncirin prevented adipocyte differentiation, as demonstrated by inhibition of cytoplasm lipid droplet accumulation and peroxisome proliferator-activating receptor-? The half-life of "1"0"9Cd was measured by following the decay of sources from a radiochemically pure solution with two different measuring systems: an ionisation chamber and a high-purity germanium (HPGe) detector. The measurements were performed over a period of 3.6 years, i.e. about 2.8 half-lives of "1"0"9Cd. The resulting half-life values and detailed uncertainty budgets (k=1) are presented for both systems. The result obtained with the ionisation chamber, 462.36 (33) days, and the one obtained with the HPGe detector, 461.92 (76) days, are mutually consistent. The weighted mean of our measured values, T_1_/_2("1"0"9Cd)=462.29 (30) days, is consistent with the currently recommended values of 461.4 (12) days and 462.0 (3) days. Radioisotopes of Pb(II) have been of some interest in radioimmunotherapy and radioimmunoimaging (RII). However, the absence of a kinetically stable bifunctional chelating agent for Pb(II) has hampered its use for these applications. Studies on the separation of Ag(I) during the recovery of Pu from analytical waste generated during potentiometric determination of Pu using AgO as oxidant, by precipitation of Pu as ammonium plutonium (III) oxalate have shown that most of the Ag(I) is separated during the reduction of Pu to Pu(III) state by ascorbic acid. A decontamination factor of 54 was obtained. Additional a decontamination factor of 5.8 was obtained during the precipitation of Pu as ammonium plutonium (III) oxalate. The Ag content was determined by gamma spectrometry, using "1"1"1Ag as a tracer (T_1_/_2 7.45 d, #gamma# 342.1 keV) and HPGe as detector. The studies indicate that Ag is preferentially separated almost quantitatively during the recovery of plutonium, when acidity adjustment is done with NaOH instead of NH_3. (author)
In this work, the half-life of the "1"5"5Sm #beta#"- decay was determined using enriched "1"5"4Sm samples submitted to irradiation in the IEA-R1 reactor of IPEN; the activity of the samples were followed for 4-5 consecutive half lives using a 198 cm"3 HPGe detector. The data was corrected using a non paralizable dead time correction and fitted to an exponential decay function using a non linear fitting procedure developed on the MatLab platform. The resulting value--T_1_/_2 = 22.180(26) min--was compatible to the one found in the literature, with a lower uncertainty.

The objective of this research is to determine the role of DNA repair in mutagenesis and carcinogenesis in mammalian cells. Use of the host-cell reactivation viral suicide enrichment procedure was initiated in the isolation of repair-deficient mutants. Lightly mutagenized BHK cells were infected with irradiated Herpes simplex virus (HSV); several radiation-sensitive strains were isolated among the survivors of the infection. The characterization of these strains is progressing and the enrichments are continuing. That alterations in the frequency of mutation of C3H/10T1/2 cells, occurring as a result of holding the cells in a confluent state following treatment with ethylmethane sulfonate, parallel the alterations in the frequency of neoplastic transformation was found. The high-spin structure of the Z=77 nucleus "1"8"7Ir has been studied using the fusion-evaporation reaction "1"8"6W("7Li6n) at a beam energy of 59 MeV. The excitation scheme of this nucleus has been extended by more than 110 new states, including extensions of all previously established rotational bands. The band crossing region of the h_9_/_2 negative-parity yrast band has been revised and new intrinsic high-K states have been identified. In particular, a 29/2"- isomeric state [T_1_/_2=1.8(5)#mu#s] at an excitation energy of 2487 keV has been observed for the first time, and on top of it, a rich level scheme reaching up to spin (59/2"-) and excitation energies around 7 MeV has been established.

Full text: HMPAO, a butane monoxime derivative of propylene amine and ECD, ethyl cysteinate dimer, are currently being used as brain imaging agents. In an effort to develop new radiopharmaceuticals for imaging brain, we have synthesized amine and imine derivatives of ethyl cysteinate with butane monoxime, optimized labelling procedure with "9"9"mTc and studied the in vivo behaviour of their "9"9"mTc complexes. Imine derivative of ethyl cysteinate with butane monoxime was synthesized by refluxing equimolar amounts of butane monoxime and ethyl cystinate in dry benzene for 3 hours. The product was purified over silica with 10% ethyl acetate in chloroform as eluent. Amine derivative was prepared by reducing the imine derivative with sodium borohydride. According to the calculations, intensity of the KL_1L_2("3P_0) Auger transition should drastically increase with increasing atomic number Z due to the relativistic effects. However, this behavior was experimentally proved only for very few elements. A lack of enough precise experimental data in the atomic number region Z<45 does not enable one to distinguish between relativistic and non-relativistic approaches in this region. Thus for Z=38 the KL_1L_2("3P_0/"1P_1) intensity ratio was determined with relative uncertainty of 63 % in the only measurement with external excitation. The distribution of carrier-free "2"0"3Pb-acetate, "2"0"3HgCl_2, "5"7Cocl_2, "1"3"7CsCl and "2"0"1TlCl was investigated in rats bearing thigh-implanted Morris 7777 hepatomas. Viable and nonviable tumor tissue was collected in order to determine the relative affinities of the radiopharmaceutical for these tissues. The animals were sacrificed at 4, 24, 48, 72 and 96 hrs following intravenous injection. Washout of the radioisotope from the viable tumor tissue was rapid, the maximum concentration being reached on or before 4 hrs following injection. In contrast, residual activity within the nonviable tumor tissue decreased much more slowly and in some cases even increased with time. Designer natriuretic peptides (NPs) represent an active area of drug development. In canine and human studies, the designer natriuretic peptide CD-NP demonstrated more desirable therapeutic potential than recombinant B-type NP (BNP), which is known as nesiritide and is approved for treatment of acute decompensated heart failure. However, why CD-NP is more effective than BNP is not known. We previously reported that CD-NP is a poorer activator of human guanylyl cyclase-A (GC-A) and a better activator of human guanylyl cyclase-B than BNP. The formation and properties of Pd_2Si formed by focused ion beam implantation of Pd ions into Si is presented in this thesis. An extensive microstructural study using transmission electron microscopy was undertaken and the as-implanted as well as annealed microstructure is shown. Results of other analysis techniques such as Rutherford back scattering and secondary ion mass spectrometry etc. are also presented. Kinetic information on the growth of Pd_2Si obtained by both microstructural and resistance measurements indicates that the activation energy for growth of the silicide is around 0.36 to 0.39 eV. This can be compared with the normally reported value of 1.5 eV for Pd_2Si formed by annealing thin film Pd on Si. The growth of the silicide was found to follow t"1"/"2 kinetics. Purpose: To assess the prognostic value of biologic (p53, Ki-67) and clinical factors in squamous cell carcinoma of the oropharynx after radical surgery and postoperative radiotherapy (RT). Methods and Materials: Between 1985 and 1995, a total of 102 patients with 104 tumor sites were entered onto the study. Fifty-five primary tumors (53%) involved the tonsils, 26 (25%) the soft palate, and 23 (22%) the base of the tongue. Median age was 53 years (range 36-80 years). The clinical T- and N-categories (UICC 1997) were: T1 (30), T2 (47), T3 (22), T4 (5), N0 (33), N1 (28), N2 (42), and N3 (1). Histologically-clear margins were achieved in all patients by initial surgery.
