An oral nervous necrosis virus vaccine using Vibrio anguillarum as an expression host provides early protection

Oral vaccination is the most desirable immunization method by which to prevent fish disease occurring at the early larval stage. We developed an Escherichia coli-based oral nervous necrosis virus (NNV) subunit vaccine and demonstrated its effectiveness in orange spotted grouper (Epinephelus coioides) fry. However, the incubation period was determined to be over 2weeks. To enhance the efficacy and shorten the incubation period, a safe and digestible biological adjuvant should be found. Vibrio anguillarum, a common marine bacterium with immune-stimulatory capability was selected as an expression host. Kokanee Stock Status and Contribution of Cabinet Gorge Hatchery, Lake Pend Oreille, Idaho, 1986 Annual Progress Report.

Estimated kokanee (Oncorhynchus nerka) abundance in Lake Pend Oreille was 4.3 million during September 1986. This estimate was similar to 1985 and indicates continued suppression of the kokanee population since initial decline in the late 1960s. Atypically high survival of wild fry resulted in similar fry recruitment in 1986 as 1985, whereas hatchery-reared fry contributed only 8% to total fry recruitment as a result of low post-release survival (3%). Fry released into the Clark Fork River from Cabinet Gorge Hatchery had very low survival during emigration to Lake Pend Oreille, resulting from poor flow conditions and potentially high predation. Multielement determination of major-to-ultratrace elements in salmon egg by ICP-AES and ICP-MS

The multielement determination of major-to-ultratrace elements in salmon egg was carried out by ICP-AES and ICP-MS. First, salmon eggs (30 eggs used) were digested with conc. nitric acid and 60% perchloric acid by heating almost to dryness repeatedly. Finally, the residues of salmon eggs were dissolved in 1 M nitric acid and the sample solutions were analyzed by ICP-AES and ICP-MS. About 40 elements could be determined over the concentration range in 9 orders of magnitude. The concentrations of bio-essential elements, such as Zn, Fe, and Cu, in salmon egg were 10-100 times higher than those in human blood serum. In addition, the major-to-ultratrace elements in cell cytoplasm and cell membrane were separately determined in a similar manner to those in whole egg cell. The area of interest for this study focuses on the lower four miles of the Clearwater River and its confluence with the Snake River because this is an area where many fish delay their seaward migration. The goal of the project is to increase our understanding of the environmental and biological factors that affect juvenile life history of fall Chinook salmon in the Clearwater River. Depending on the raw material, bio diesel can contain more or less unsaturated fatty acids in its composition, which are susceptible to oxidation reactions accelerated by exposition to oxygen and high temperatures, being able to change into polymerized compounds. The objective of this work was to determine the oxidative stability of bio diesel produced by ethanolysis of neutralized, refined, soybean frying oil waste, and partially hydrogenated soybean frying oil waste. A numerical simulation of the evolution and fate of a FRI jet. The case of 3C 31

The evolution of FRI jets has been long studied in the framework of the FRI-FRII dichotomy. In this paper, we test the present theoretical and observational models via a relativistic numerical simulation of the jets in the radio galaxy 3C 31. We use the parameters derived from the modelling presented by \\cite{lb02a,lb02b} as input parameters for the simulation of the evolution of the source, thus assuming that they have not varied over the lifetime of the source. We simulate about 10 % of the total lifetime of the jets in 3C 31. Realistic density and pressure gradients for the atmosphere are used. The simulation includes an equation of state for a two-component relativistic gas that allows a separate treatment of leptonic and baryonic matter. We compare our results with the modelling of the observational data of the source. Purpose - The objective of this study is to examine the effects of pan-frying, boiling and cooking in a microwave on the amount of L-carnitine in meat and to look at its distribution in the surrounding fluid after food processing. Design/methodology/approach - Total carnitine, free carnitine and acylcarnitines were determined in meat samples from beef, pork and poultry (including ostrich) and in a liver sample from beef. The measurements were carried out before and after the specimens were subjected to different heat treatments. A radio-enzymatic assay was used for measurement of L-carnitine. The authors examined the effects of large escapements of sockeye salmon in to the Kenai River and into Red and Akalura Lakes on Kodiak Island. Estimated smolt numbers outmigrating from the Kenai River decreased from 30 million in 1989 to under 500,000 in 1992. Decreased overwintering survival of age-0 juvenile sockeye salmon rearing in Skilak and Kenai lakes is the major contributor to this decline. Preliminary examination of limnological data from Skilak and Kenai Lakes provided no obvious explanation for this major decline in winter survival of juvenile sockeye. Due to differences in diel vertical migration of zooplankton among glacial lakes on the Kenai Peninsula with varying planktivores levels, the authors hypothesize that behavioral changes of zooplankton induced by predation may be a major contributor to the juvenile sockeye decline.

The presence of endothelin (ET)-like immunoreactivity and the cardiovascular effects of mammalian ET-1 in fish have been reported. To identify ET-related peptides in fish, we screened the cDNA library of the salmon (Oncorhynchus keta) stomach by means of rapid amplification of cDNA ends, and we cloned cDNAs encoding an ET-related peptide. The salmon ET-related sequence of 21 amino acids is identical to the trout ET-1 peptide recently purified from kidney specimens of Oncorhynchus mykiss. The deduced amino acid sequence of salmon pre-proET-1 (PPET-1) comprises 244 amino acids, including a putative signal sequence and mature ET-1, as well as big ET-1 and ET-1-like sequences. This precursor, the first reported PPET-1 sequence for Salmoniformes, Teleostei, has low homology with the sequences of human, mouse, frog (Xenopus laevis), and zebrafish (Danio rerio) PPET-1 (26%, 29%, 24%, and 39%, respectively). The characteristics of diesel fuel jet atomization in carburetors depend on dynamic viscosity (v) density (d) and surface tension (st) of fuel, grouped in the combined functions v/d; st/v and st/d. Values of these combined functions for binary mixtures of a farm diesel fuel with a crude vegetable oil (sunflower, soybean, cotton and foot-cake olive oils) as well as with a used frying olive oil, are calculated. Three different diesel fuels have been studied. Combined function values of these mixtures are influenced by the characteristics of their diesel fuel. Mixtures containing less than 24% of any studied crude oils show kinematic viscosity values at 40 degrees C not exceeding 5.8 squared millimeters/s. This value is the higher limit for typical United States farm diesel fuels, according to specifications of the Cummins Engine Company. Mixture with 21% of the used frying olive oil shows at 40 degrees C this kinematic viscosity.

On The Parent Population of Radio Galaxies and the FRI - FRII Dichotomy

We test the hypothesis that radio galaxies are a random subset of otherwise normal elliptical galaxies. Starting with the observed optical luminosity functions for elliptical galaxies, we show that the probability of an elliptical forming a radio source is a continuous, increasing function of optical luminosity, proportional to L squared. With this probability function and the luminosity function of normal elliptical galaxies as input to Monte Carlo simulations, we reproduce the observed distribution of radio galaxies in the radio-optical luminosity plane. Our results show that radio galaxies are a luminosity-biased but otherwise random sample of elliptical galaxies. This unified view of radio-loud and radio-quiet ellipticals also explains the well known difference of ~0.5 mag in average optical luminosity between FRI and FRII radio galaxies as a simple selection effect. Cold gas & mergers: fundamental difference in HI properties of different types of radio galaxies?

We present results of a study of large-scale neutral hydrogen (HI) gas in nearby radio galaxies. We find that the early-type host galaxies of different types of radio sources (compact, FR-I and FR-II) appear to contain fundamentally different large-scale HI properties: enormous regular rotating disks and rings are present around the host galaxies of a significant fraction of low power compact radio sources, while no large-scale HI is detected in low power, edge-darkened FR-I radio galaxies. Preliminary results of a study of nearby powerful, edge-brightened FR-II radio galaxies show that these systems generally contain significant amounts of large-scale HI, often distributed in tail- or bridge-like structures, indicative of a recent galaxy merger or collision. Our results suggest that different types of radio galaxies may have a different formation history, which could be related to a difference in the triggering mechanism of the radio source. ...

A relativistic mixing-layer model for jets in low-luminosity radio galaxies

We present an analytical model for jets in Fanaroff & Riley Class I (FRI) radio galaxies, in which an initially laminar, relativistic flow is surrounded by a shear layer. We apply the appropriate conservation laws to constrain the jet parameters, starting the model where the radio emission is observed to brighten abruptly. We assume that the laminar flow fills the jet there and that pressure balance with the surroundings is maintained from that point outwards. Entrainment continuously injects new material into the jet and forms a shear layer, which contains material from both the environment and the laminar core. The shear layer expands rapidly with distance until finally the core disappears, and all of the material is mixed into the shear layer. Chemicals under the trade names Nalco 537-DA, Nalco 625, Nalco 7607, Nalco 5165, Ivamin, and technical monoethanolamine are used extensively in the oil refinery industry. Aquatic toxicity tests were conducted using zebra fish fry (Brachydanio rerio) and the unicellular algae Isochrysis galbana (a flagellate) and Chaetoceros gracilis (a diatom). Inhibition of cell division, chlorophyll content, and "1"4CO_2 uptake in the algae were sensitive end points. The effective concentrations (EC50s) of growth inhibition were 0.1 mg/L (Ivamin; I. galbana), 0.8 mg/L (Nalco 7607; I. galbana), 6 mg/L (Nalco 625; I. galbana), 10 mg/L (Nalco 5165; C. gracilis), and 15 mg/L (Nalco 537-DA; C. gracilis). The authors determined migration timing and abundance of juvenile spring chinook salmon Oncorhynchus tshawytscha and juvenile steelhead/rainbow trout O. mykiss from three populations in the Grande Ronde River basin. Based on migration timing and abundance, two distinct life-history strategies of juvenile spring chinook and O.mykiss could be distinguished. An early migrant group left upper rearing areas from July through January with a peak in the fall. A late migrant group descended from upper rearing areas from February through June with a peak in the spring.

What can we learn about extragalactic radio jets from X-ray data?

We review the current status of resolved X-ray emission associated with extragalactic radio jets and hotspots. The primary question for any particular jet is to decide if the X-rays come from the synchrotron process or from inverse Compton scattering. There is considerable evidence supporting synchrotron emission for knots in the jets of FRI galaxies. For FRII terminal hotspots detected in the X-ray band, synchrotron self-Compton emission continues to provide viable models with one possible exception (so far). Inverse Compton scattering on photons of the cosmic microwave background is indicated for a few powerful jets, and is expected to be an important contributor if not the dominating mechanism for higher redshift objects. The application of a model generally yields physical parameters and in many cases, these include the Doppler boosting factor.

Waste cooking oil can be recovered and processed into animal feed additives after purification. Unfortunately, the traditional purification processes are insufficient for the removal of the harmful compounds formed during frying, mainly about malondialdehyde and other 2-thiobarbituric acid reactive substances. In the present paper, firstly, a simple and reliable HPLC method was developed to measure the content of malondialdehyde in purified waste cooking oil. Based on data collected, a white sturgeon adaptive management plan will be developed. This report presents a summary of results from the 1997-2002 Phase II data collection and represents the end of phase II. From 1997 to 2001 white sturgeon were captured, marked, and population data were collected in the Snake and Salmon. A total of 1,785 white sturgeon were captured and tagged in the Snake River and 77 in the Salmon River. Since 1997, 25.8 percent of the tagged white sturgeon have been recaptured. Bio-inspired silver nanoparticles were synthesized with the aid of a novel, non-toxic, eco-friendly biological material namely, banana peel extract (BPE). Boiled, crushed, acetone precipitated, air-dried peel powder was used for reducing silver nitrate. Silver nanoparticles were formed when the reaction conditions were altered with respect to pH, BPE content, concentration of silver nitrate and incubation temperature. The colorless reaction mixtures turned brown and displayed UV-visible spectra characteristic of silver nanoparticles. The inhibitory activity of chicken and crocodilian ovomacroglobulins against trypsin was measured before and after their incubation with methylamine. The result for crocodilian ovomacroglobulin showed that methylamine treatment destroyed half of its activity, in unique contrast to human alpha 2-macroglobulin and chicken ovomacroglobulin for which methylamine either destroys the inhibitory activity of the former completely or does not affect that of the latter at all. Free sulfhydryl groups of chicken and crocodilian ovomacroglobulins were titrated with 5,5'-dithiobis(2-nitrobenzoic acid) before and after incubation with trypsin. The exhaust emissions of a Diesel direct injection Perkins engine fueled with waste olive oil methyl ester were studied at several steady-state operating conditions. Emissions were characterized with neat biodiesel from used olive oil and conventional Diesel fuel. Results revealed that the use of biodiesel resulted in lower emissions of CO (up to 58.9%), CO{sub 2} (up to 8.6%, excepting a case which presented a 7.4% increase), NO (up to 37.5%), and SO{sub 2} (up to 57.7%), with increase in emissions of NO{sub 2} (up to 81%, excepting a case which presented a slight reduction). A dialyzable factor(s) in human serum is known to stimulate gonococcal oxygen consumption. Its effect on other human pathogens was investigated. A 10% serum solution increased peak O2 consumption for Escherichia coli and Staphylococcus aureus to 157% (P less than 0.05) and 199% (P less than 0.02), respectively, of their O2 consumption when suspended in Hanks balanced salt solution, compared with a 356% increase for Neisseria gonorrhoeae with serum. Dialyzed serum lacked stimulatory capacity. Bacteria, serum, and neutrophils are often incubated to evaluate neutrophil bactericidal activity. Using quantitative electron microscopic (EM) autoradiography we find that, when /sup 125/I-insulin is incubated with 3T3-L1 adipocytes, the ligand at early times of incubation localizes to the plasma membrane of the cell preferentially to microvilli and coated pits. Gel filtration of human erythrocyte (RBC) lysate incubated with labeled thyroxine (Tu) or triiodothyronine (Tt) revealed co-elution of a major iodothyronine-binding fraction (R-2) and hemoglobin. Solutions of purified human hemoglobin and Tt also showed co-elution of hormone and hemoglobin. Because hematin and protoporphyrin were shown to bind labeled Tt, the oxygen-binding site on hemoglobin was excluded as the site of iodothyronine-hemoglobin interaction. Analysis of hormone binding by heme and globin moieties showed Tt binding to be limited to the heme fraction. Addition of excess unlabeled Tt to hemoglobin or heme incubated with labeled Tt indicated 75% to 90% of hormone binding was poorly dissociable. Actively migrating smolts were collected near the head of Lower Granite Reservoir and at Lower Granite Dam, tagged with passive integrated transponder (PIT) tags, and released to continue their downstream migration. When labeled cAMP is incubated with isolated mitochondria from rat liver, it penetrates not only through the outer membrane of the mitochondria but also into the mitoplasts, where it is accumulated chiefly in the matrix. Damage to the mitochondrial membrane by freezing-thawing promotes not an influx but an efflux of cAMP out of the mitoplasts. A substantial part of the labeled nucleotide in all the investigated submitochondrial fractions was identified as intact cAMP by the method of thin-layer chromatography. The transport of cAMP into the mitochondria can explain the activation of their functions by the hormones that activate cytoplasmic adenylate cyclase and by extramitochondrial (cytosol) cAMP, as well as the presence of cAMP-dependent protein kinase and cAMP-phosphodiesterase in the mitochondria.

A widely used veterinary antibiotic, tetracycline (TC), has been incorporated in Langmuir-Blodgett (LB) films of dipalmitoylphosphatidic acid (DPPA) by means of two different procedures: co-transfer and incubation in solution. The resulting structures were characterized by means of contact angle and ellipsometric measurements. The presence of the antibiotic in the phospholipid film was evidenced by means of UV-Vis electronic absorption and infrared vibrational spectroscopy. The two sets of measurements unambiguously indicated the presence of the drug in the LB layer films obtained with both methods, although incubation led to a smaller content of immobilized tetracycline. In both cases, the drug was found to reside in the hydrophilic portion of the layers due to specific interactions of the dimethylamino group of the molecule with the polar head groups of the phospholipid.

Titanium base 'alpha'-alloys (Pt-7M, RK-20 and Ti-5Al-2Zr-1.5V) are considered for their behaviour under neutron irradiation. The role of alloying elements in radiation hardening is discussed depending of neutron fluence and irradiation temperature. For PK-20 alloy three stages of change in mechanical properties are revealed which are following: incubation period and weak hardening, intense radiation hardening, radiation hardening attenuation. Irradiation temperature rise results in an increase of incubation period and threshold neutron fluence. A special attention is paid to hydrogen absorption in #alpha#-titanium alloys under irradiation. In this work, a metal biosorbent was prepared by drying biomass of the yeast Rhodotorula glutinis in an oven at 70C for 24-h followed by grinding. Two forms of the prepared biosorbent particles, washed and unwashed, were examined for their ability to remove lead from solution. It was found that the unwashed biosorbent exhibited higher lead uptake than the washed biosorbent. Cultures of 3T3-L1 cells were incubated with either 10 ng/ml cholera toxin or 10 ng/ml pertussis toxin from 4 days prior to the initiation of differentiation and throughout the subsequent incubation. Toxin concentrations were sufficient to completely prevent the labelling of alpha-subunits with ["3"2P]NAD"+ and pertussis toxin and to prevent by more than 90% the labelling with ["3"2P]NAD"+ and cholera toxin in membranes prepared from these cells. Neither toxin prevented the differentiation to the adipocyte phenotype. Neither toxin prevented the increases in the relative amounts of G-proteins which occur upon differentiation. A simple method for regeneration of filaments (clones) from spheroplasts of the cyanobacterium (blue-green alga), Anabaena variabilis, was developed and used to quantify cell growth in the presence of two antibiotics. Cells from exponential phase cultures of ATCC 29413 and M3 were harvested and incubated with lysozyme (0.12% in 0.03M K-phosphate, pH 6.8, 0.55M sorbitol; 37 C) to produce spheroplasts. The spheroplasts were washed with buffer, plated onto soft agar and incubated (18 h light: 6 hr dark, 27 C). Colonies became visible at 7 - 9 days and were monitored for times up to 21 days. The concentration of chloramphenicol which inhibited cell growth by 50% was approximately 1.8 mg ml/sup -1/ medium and the concentrations of ampicillin which inhibited cell growth by 50% were approximately 4 and 15 pg ml/sup -1/ medium for ATCC 29413 and M3, respectively. Three parameters - initial moisture content, incubation temperature, and initial pH - were optimized in culture flasks using response surface methodology. Parameter optimization was carried out with respect to filter paper activity and b-glucosidase activity in the culture. The K0.5 for intracellular sodium of the two forms of (Na ,K )-ATPase which exist in rat adipocytes has been determined by incubating the cells in the absence of potassium in buffers of varying sodium concentration; these conditions shut off the Na pump and allow sodium to equilibrate into the cell. The activity of (Na ,K )-ATPase was then monitored with YWRb /K pumping which was initiated by adding isotope and KCl to 5 mM, followed by a 3-min uptake period. Atomic absorption and SSNa tracer equilibration were used to determine the actual intracellular (Na ) under the different conditions. The K0.5 values thus obtained were 17 mM for alpha and 52 mM for alpha(+). Insulin treatment of rat adipocytes had no effect on the intracellular (Na+) nor on the Vmax of YWRb /K pumping, but did produce a shift in the sodium ion K0.5 values to 14 mM for alpha and 33 mM for alpha(+). Oxygenated sterol compounds (OSC), when incubated for 1 hr with human erythrocytes in lipoprotein-depleted medium at concentrations of 0.625-5 x 10/sup -5/M, are inserted into the cell membrane and remain there despite subsequent washing of the cells. The insertion results in expansion of the surface area of the red cell ghost membrane, an increase in critical hemolytic volume, and as a consequence, in dimished osmotic fragility of the erythrocytes. A hypotheses that mitoxantrone is a competitive antagonist at muscarinic cholinergic receptors was examined in guinea-pig hearts. In isolated left atrial muscle preparations, electrically paced at 2 Hz, the muscarinic agonist, carbachol, caused a concentration-dependent decrease in developed tension. Mitoxantrone caused a parallel right-ward shift of the concentration-response curve for carbachol. Schild plots for the effect of mitoxantrone on the carbachol concentration-response relationship were linear with a slope of 0.88 which was not significantly different from the unity. Carbon tetrachloride-induced release of calcium from isolated hepatocytes

Studies have shown that CCl_4 administration to rats inhibits endoplasmic reticulum calcium pump activity and reduces the amount of calcium associated with subsequently isolated microsomal subcellular fractions. This report confirms that exposure of isolated hepatocytes to CCl_4 rapidly produces these effects in isolated parenchymal cells and demonstrates that when isolated hepacytes are exposed to CCl_4 calcium is rapidly released from cells. This release can be detected with a calcium ion-selective electrode when cells are incubated in a medium with low extracellular calcium. The effect of oxygen and paraquat on the "1"4C-glucose oxidation of rabbit alveolar macrophages and lung slices

In this study, we measured the effects of different concentrations of paraquat (0,01 mM and 1,0 mM) on the 1-"1"4CO_2 and 6-"1"4CO_2 production of rabbit lung slices and isolated alveolar macrophages, in 20% and 95% oxygen phases respectively. A 95% oxygen phase induced an increase in the 6-"1"4C-glucose oxidation of control lung slices over a 3-hour period, while the increased activity of the pentose pathway over the first 2 hours started to decline during the third hour of incubation. Mesocarp tissue from sucrose-accumulating sweet melon (Cucumis melo cv. Galia) showed sucrose synthase activity (ca 1 nkat/gfw) while soluble acid invertase and sucrose phosphate synthase activities were not observed. Sucrose uptake into mesocarp discs was linear with sucrose concentration (1-500 mM) and unaffected by PCMBS and CCCP. Sucrose compartmentation into the vacuole also increased linearly with sucrose concentration as indicated by compartmental efflux kinetics. In this Letter, we demonstrate the application of time-resolved fluorescence anisotropy measurements to detect solution state hybridization of streptavidin conjugate (CdSe)ZnS quantum dots (QD). The study was performed on samples containing 10nM QD incubated with 800nM DNA. We show that the rotational correlation time of QD-DNA constructs increases significantly upon hybridization with values of 330ns (QD-ssDNA) and 1.3ms (QD-dsDNA), corresponding to a diameter of 14nm and 23nm respectively. Radiosensitization of human lung fibroblasts by chemicals that decrease ATP levels

Radiosensitization by lactate, pyruvate, nalidixic acid and novobiocin was studied in exponentially growing SH-18L human lung fibroblasts. All the chemicals had a slight radiosensitizing effect at a low concentration and a definite effect at a higher one. Decreases in the D_0 and/or Dq values were present in each dose survival curve. Fibroblasts incubated with the low concentration of each chemical for 24 hrs after X irradiation showed no reduction in intracellular ATP content, whereas, the higher concentration produced a significant decrease. These observations suggest that the decrease in the ATP content may be involved in the radiosensitization of human fibroblasts at high concentrations of these chemicals. In contrast, radiosensitization at a low concentration is not explained by a relationship to ATP content. Different mechanisms may be involved in radiosensitization at low and high concentrations of these chemicals. (author).

Resolution of these peptides by two-dimensional gel electrophoresis allowed identification of two peptides of M/sub r/ = 42,000 and 52,000 as peptide subunits of a regulatory component of adenylate cyclase. The radiolabeling of another group of peptides (M/sub r/ = 50,000 to 65,000) suggested that cholera toxin could catalyze ADP-ribosylation of cytoskeletal proteins. The first response of the host plant to the contact with Rhizobium lupini was the enhancement of nuclear DNA endoreplication in root cortex cells. A conversion of the initially mixoploid nodule meristem into a diploid one during root nodule formation was found. Differentiation of bacteroid-containing tissue both from diploid and polyploid cells was observed. The aim was to identify mechanisms and the effects of important microstructural parameters on damage of polished Al{sub 2}O{sub 3}, Al{sub 2}O{sub 3}-ZrO{sub 2}, ZrO{sub 2} and SiC ceramics. Results showed that surface damage of brittle ceramics was mainly dominated by intergranular fracture, followed by detachment of single grains or fragments of them. Gold nanoparticles were synthesized by using banana peel extract (BPE) as a simple, non-toxic, eco-friendly 'green material'. The boiled, crushed, acetone precipitated, air-dried peel powder was used to reduce chloroauric acid. A variety of nanoparticles were formed when the reaction conditions were altered with respect to pH, BPE content, chloroauric acid concentration and temperature of incubation. The activity of the components separated by gaschromatography is written down by a printing indicator and the impulses/min are registered and printed out by a ratemeter. Thus the aldehydes which are produced by the enzymatic oxydation with lipoxygenase from the molecule of the linoleic acid can be determined. The composition of the mixture of aldehydes is calculated in mol-% from the measured impulses for peak. A radioassay for the rapid determination of Gsub(M1) ganglioside concentration in small volumes of CSF from individual patients is described. The assay utilizes the high-affinity interaction between cholera enterotoxin ("1"2"5I-labelled) and Gsub(M1) ganglioside. The lower detection limit of Gsub(M1) ganglioside by this radioassay under the described incubation conditions is 2.5 ng/ml. The radioassay-determined lumbar CSF Gsub(M1) ganglioside concentrations in a small group of patients with diverse neurologic disorders are presented. Since DMSO has been shown to be a non-specific stimulator of tyrosine protein kinases they have examined the effect of DMSO on both activation and tyrosine phosphorylation in human T cells. Rates of sucrose, fructose, and D- and L-glucose uptake by slices were similar, whereas uptake rates for these sugars differed greatly in suspension cultures. Concentration dependence of sucrose, fructose, and D-glucose uptake was biphasic (consisting of linear plus saturable components) with suspension cultures but linear with slices. This study was undertaken to evaluate the potential of liver ZnMT to mediate the interaction of Zn and Pb with the Zn metalloenzyme, ALAD. Pretreatment of rats with Zn activated liver ALAD and attenuated the inhibition of ALAD by Pb in vitro and in vivo. Liver cytosol from Zn-pretreated rats injected with /sup 203/Pb was fractionated via gel filtration and anion-exchange chromatography and disclosed that both Zn and /sup 203/Pb co-eluted in the MT fractions. Cultured 3T3-L1 cells have been used as a model system to investigate the mechanism of fatty acid uptake by adipose tissue. Using a 1:1 molar ratio of /sup 14/C-oleate and defatted bovine serum albumin (BSA), fatty acid (FA) uptake was quantitated at 4/sup 0/ and 37/sup 0/ as cell associated radioactivity. The profile of FA uptake in preadipocytes and adipocytes was biphasic; an initial rapid phase (1-20s) followed by a second slower phase (60-480s). The effect of experimental diabetes on the phosphorylation of proteins in the rat sciatic nerve was studied. Nerves from animals made diabetic with streptozocin were incubated in vitro with (/sup 32/P)orthophosphate and divided into segments from the proximal to the distal end, and proteins from each segment were then separated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The principal labeled species were the major myelin proteins, P0, and the basic proteins. After 6 wk of diabetes, the incorporation of isotope into these proteins rose as a function of distance along the nerve in a proximal to distal direction and was significantly higher at the distal end compared with incorporation into nerves from age-matched controls. We have previously demonstrated that areas of injured aorta that have been re-endothelialized accumulate increased amounts of lipid and GAG when compared to areas remaining de-endothelialized. We have now examined the net incorporation of labeled precursors into the individual GAG present in both re-endothelialized and de-endothelialized areas of rabbit aorta. Aortic tissue was examined at 2-3 and 10-14 weeks after a denuding injury by incubating tissue minces with (/sup 3/H)glucosamine and sodium (/sup 35/S)sulfate for 24 hr. Dams cause direct mortality when fish pass through turbines, but dams may also cause indirect mortality by altering migration conditions in rivers. Forebays immediately upstream of dams have decreased the water velocity of rivers and may contribute substantially to the total migration delay of juvenile salmonids. The authors used radioligand binding and studies of cell contraction to characterize muscarinic receptors on dispersed smooth muscle cells from rabbit proximal and distal colon. Cells obtained after serial incubations in collagenase were used to measure binding of tritiated quinuclidinyl benzilate (["3H]QNB). At 37 degree C, specific ["3H]QNB binding was saturable and linearly related to cell number. Nonlinear regression analysis was used to determine the affinity of ["3H]QNB for its receptor. The IC_5_0 for the muscarinic agonists bethanechol and oxotremorine were 80 and 0.57 #mu#M, respectively. Hill coefficients were 0.67 for both, suggesting more complex interaction involving receptors of different affinities. The mechanisms involved in amino acid and sugar uptake by developing maize kernels were investigated. In the pedicel region of maize kernel, the site of nutrient unloading from phloem terminals, amino acids are accumulated in considerable amounts and undergo significant interconversion. A wide spectrum of enzymatic activities involved in the metabolism of amino acids is observed in these tissues. Subsequently, amino acids are taken up by the endosperm tissue in processes which require energy and the presence of carrier proteins. Conversely, no evidence was found that energy and carriers are involved in sugar uptake. This process of sugar uptake is not inhibited by metabolic inhibitors and shows nonsaturable kinetics, but the uptake is pH-dependent. The effect of low dose gamma irradiation on the activity of blood serum lysozyme and complement in broiler chickens

Samples from 314 broiler chickens at 2 months age were analyzed for blood serum level of lysozyme and 311 of them were analyzed also for activity of alternative path-way of complement activity (APCA). Chickens were hatched from eggs, irradiated immediately before incubation with different doses of gamma-rays using Rocus M device - group 1 - control (non irradiated), and groups 2, 3 and 4 - irradiated with doses of 0.15, 0.30 and 0.60 Gy, respectively. Lysozyme concentration was highest for the control group, and for the experimental ones it was lower (p<0.001) which is an indication of the strongly negative effect of gamma-rays on this indicator. Probiotic agents, live microorganisms with beneficial effects for the host, may offer an alternative to conventional antimicrobials in the treatment and prevention of enteric infections. The probiotic agents Lactobacillus plantarum 299v and Lactobacillus rhamnosus GG quantitatively inhibited the adherence of an attaching and effacing pathogenic Escherichia coli to HT-29 intestinal epithelial cells but did not inhibit adherence to nonintestinal HEp-2 cells. HT-29 cells were grown under conditions that induced high levels of either MUC2 or MUC3 mRNA, but HEp-2 cells expressed only minimal levels of MUC2 and no MUC3 mRNA. Media enriched for MUC2 and MUC3 mucin were added exogenously to binding assays and were shown to be capable of inhibiting enteropathogen adherence to HEp-2 cells. Qualitative and quantitative methods have been developed to relate phosphate/UO{sub 2} interactions and U dissolution, in vitro, in a medium containing 0 or 10 mM phosphate concentrations at pH 5. This was performed by Energy Dispersive X ray Spectrometry (EDS) at 200 kV on entire particles and by fluorimetric measurement. The U dissolution rate was larger in a medium without phosphate than in a medium containing 10 mM phosphate. During the first day, the initial U dissolution involved 80.55% and 91.13% of the powder in media with and without phosphate respectively. From 5 to 15 d after beginning the assay, the dissolution half-times were longer than 92 d and 22 d for media with and without phosphate. EDS analyses have shown a constant P/U X ray intensity ratio for reference U{sub 2}O(PO{sub 4}){sub 2} for the same range of particle diameter. Persistence of paraquat in the soil and observations with other herbicides relevant to the theme of bound residues

Results from three separate experiments that have some relevance to bound residues are reported. In the first, "1"4C-labelled paraquat was lost when applied to soil in the field, about 26% of the radioactivity disappearing in 15 months, whereas in laboratory incubation studies there was no loss of radioactivity in one year. Two possible explanations are (i) that there was photolytic decomposition in the field, (ii) the preparation of the soil for the laboratory study upset the microbial ecology of the soil to the detriment of organisms that can degrade paraquat. Peeled Avena sativa coleoptile sections have been used to study the control of solute uptake under conditions where the uptake is not limited by the cuticular barrier. In the presence of 2% sucrose, auxin enhances the rate at which the total osmotic solutes increase, but this appears to be a response to the increased growth rate, inasmuch as the auxin effect is eliminated when growth is inhibited osmotically. When sections are incubated in sucrose or in 10 millimolar NaCl, the osmotic concentration increases until a plateau is reached after 8 to 24 hours. Auxin has no effect on the initial rate of increase in osmotic concentration. This difference in steady-state osmotic concentration is, in part, a response to auxin itself, as it persists when auxin-induced growth is inhibited osmotically. Nonenzymatic glycosylation of human hemoglobin at multiple sites

The most abundant minor hemoglobin component of human hemolysate is Hb A1c, which has glucose bound to the N-terminus of the beta chain by a ketoamine linkage. Hb A1c is formed slowly and continuously throughout the 120 day lifespan of the red cell. It can be synthesized in vitro by incubating purified hemoglobin with 14C-glucose. Other minor components, Hb A1a1 and Hb A1a2 are adducts of sugar phosphates at the N-terminus of the beta chain. Hb A1b contains an unidentified nonphosphorylated sugar at the beta N-terminus. The acute effects of injections of the human growth hormone-like factor purified from plerocercoids of the tapeworm Spirometra mansonoides on carbohydrate, lipid, and protein metabolisms were determined in intact rats. Male rats were injected ip with saline, insulin, or various doses of partially purified PGF. The rats injected with insulin had significantly reduced serum glucose concentrations but no dose of PGF caused a change in serum glucose levels. Insulin and PGF stimulated [14C]glucose and [14C]leucine oxidation to 14CO2 in adipose tissue and muscle and increased incorporation of both [14C]glucose carbons into lipids and [14C]leucine into protein in fat and muscle. The responses to PGF were dose-dependent and persisted after 3 hr of incubation in vitro. Human complement protein C9 is shown to be a metalloprotein that binds 1 mol of Ca"2"+/mol of C9 with a dissociation constant of 3 #mu#m as measured by equilibrium dialysis. Incubation with EDTA removes the bound calcium, resulting in a apoprotein with decreased thermal stability. This loss in stability leads to aggregation and, therefore, to loss of hemolytic activity upon heating to a few degrees above the physiological temperature. Heat-induced aggregation of apoC9 can be prevented by salts that stabilize proteins according to the Hofmeister series of lyotropic ions, suggesting that the ion in native C9 may ligand with more than one structural element of domain of the protein. Ligand blotting indicates that the calcium binding site is located in the amino-terminal half of the protein. Fatigue crack growth in steam turbine rotor steels in near realistic media

In this study, growth measurements of fatigue cracks in air and various simulated media for steam turbines were made by means of rupture-mechanical methods. In this connection, it was detected that even pure water leads to a clear increase in the fatigue crack rates of low-pressure rotor steels. In this connection, it was striking to observe that the crack propagation velocities at 100"0C were higher than at 160"0C. It is probable that the kinetics of the film formation at the crack tip was of importance. The inhibiting effect of the alkaline solutions is explained by the increasing stability of a film of Fe(OH)_2 at an increased pH value. Escherichia coli B and K-12 are equally susceptible to the bacteriostatic effects of aerobic paraquat, but they differed strikingly when the lethality of paraquat was evaluated. E. coli B suffered an apparent loss of viability when briefly exposed to paraquat, whereas E. coli K-12 did not. This difference depended on the ability of the B-strain, but not the K-12 strain, to retain internalized paraquat; the B strain was killed on aerobic tryptic soy-yeast extract plates during the incubation which preceded the counting of colonies. This difference in retention of paraquat between strains was demonstrated by delayed loss of viability, by growth inhibition, and by cyanide-resistant respiration after brief exposure to paraquat, washing, and testing in fresh medium. Clove (Caryophyllus aromaticus L.) has been used for clinical procedures. Blood constituents labeled with technetium-99m (99mTc) are used in nuclear medicine. The aim of this work was to evaluate the effects of clove extract on the labeling blood constituents with 99mTc and on the morphology of red blood cells. Blood samples were incubated with clove, stannous chloride and 99mTc. Plasma, blood cells, insoluble fractions of plasma and blood cells were separated. The radioactivity was counted and percentage of radioactivity (%ATI) to each blood fraction was calculated. The shape and morphometric parameter (perimeter/area ratio) were evaluated. Chaoborus and chironomid larvae were studied to determine how they affect the flux of matter across the sediment/water interface. This was done by incubating undisturbed cores in situ and relating changes in water chemistry to larval density. The log-transformed flux rates of iron, manganese, and phosphorus were linearly related to larval biomass. Functional groups may be successfully used to predict rates of material flow. An assay for the microsomal hydroxylation of lauric acid (LA), based on HPLC with flow-through radiochemical detection, has been developed. Conditions were optimized for resolution and quantitation of three microsomal metabolites of "1"4C-LA, one of which has not been reported previously as a metabolite of LA in mammalian microsomal incubations. These products, 12-(omega)-hydroxy-LA, 11-(omega-1)-hydroxy-LA, and a novel metabolite, 10-(omega-2)-hydroxy-LA, were isolated by HPLC and identified by gas chromatography/mass spectrometry. In the presence of NADPH, the formation of all three metabolites was linear with time and microsomal protein concentration. Characteristics of sugar uptake by immature maize embryos were determined in vitro utilizing a /sup 14/C-sugar solution incubation method. Hexose uptake rates were greater than those for sucrose, however, all showed biphasic kinetics. Glucose and fructose saturable components were evidence at <50 mM and sucrose at <5 mM. Chemical inhibitors (CCCP, DNP, NaCN, and PCMBS) and low temperature reduced sugar uptake. Sucrose influx was pH dependent while glucose was not. Embryos maintained a high sucrose to hexose ratio throughout development. At 25 days after pollination sucrose levels exceeded 200 mM while hexose levels remained below 5 mM. Glucose was rapidly converted to sucrose upon transport into the embryo. Summary Potential predictors of egg quality were assessed in whiting Merlangius merlangus L. permitted to spawn in a tank from which eggs were collected. These included fertilisation rate, the proportion of viable buoyant eggs, egg diameter, and egg wet and dry weights; all were influenced by temporal effects and were negatively correlated with days from start of spawning. Addition of size-fractionated serum components to SAPI medium indicated that transferrin was required for norepinephrine stimulation of growth of Escherichia coli. Since bacteriostasis by serum is primarily due to the iron-withholding capacity of transferrin, we considered the possibility that norepinephrine can overcome this effect by supplying transferrin-bound iron for growth. Nitroaromatic radiosensitizers become metabolically bound preferentially to hypoxic cells and at least 10/sup 9/ adducts/cell can be tolerated as non-toxic. EMT-6 tumor cells have been incubated in hypoxia in the presence of /sup 3/H-Misonidazole and /sup 125/I-Azomycin Riboside for various times and the amount of /sup 3/H or /sup 125/I bound/cell was determined. Dicyclohexylcarbodiimide (DCCD) inhibits K/sup +/ influx. In the present studies, unidirectional fluxes of K/sup +/ or Mg/sup + +/ into respiring rat liver mitochondria were measured by means of /sup 42/K or /sup 28/Mg, with samples separated from incubation media by centrifugation through silicon. Multicellular tumor spheroids (MCTS) are three dimensional cell culture systems induced by suspension culture. MCTS are widely used in cancer research because of their similarity to solid tumors. CaSki cells are derived from a metastatic cervical cancer containing human papillomavirus 16 (HPV16). Cell death of CaSki cells in MCTS has been previously reported, and our model is used to better characterize the mechanisms of cell death of HPV16-positive keratinocytes. In this study, we found that apoptosis of CaSki cells was induced by suspension culture along with the formation of MCTS after 24 h of incubation. Microbial dechlorination of polychlorinated biphenyls (PCBs) often stops although a significant number of removable chlorines remain. To determine the reason for the cessation, we investigated the limitation of organic carbon, PCB bioavailability, and inhibition by metabolic products. Enrichment with carbon sources did not induce additional chlorination, indicating the plateau was not due to depletion of organic carbon. The bioavailability was not limiting, since a subcritical micelle concentration of the surfactant, which enhanced desorption without inhibiting dechlorinating microorganisms, failed to lower the plateau. This problem can be circumvented by covalent attachment of a bifunctional chelating group to the Mab followed by chelation with a suitable cationic radionuclide such as In-111. Competition of trace metal contaminants with In-111 for chelating sites, however, is known to lower the labeling yield. At a glucose concentration of 0.55 mmol/L, basal glucose utilization ranged from 0.5 to 1.0 nmol/min/10(6) cells. Perifused adipocytes showed a maximal response to insulin of a threefold to fourfold increase in the conversion of (5-"3H) glucose to "3HOH with a half-maximal response at an insulin concentration of 20 microU/mL. Studies were conducted to quantitate the evolution of nitrogen oxides (NO/sub (x)/) from soybean (Glycine max (L.) Merr.) leaves during in vivo nitrate reductase (NR) assays with aerobic and anaerobic gas purging. Anaerobic gas purging (N/sub 2/ and argon) consistently resulted in greater NO/sub (x)/ evolution than did aerobic gas purging (air and O/sub 2/). The evolution of NO/sub (x)/ was dependent on gas flow rate and on NO/sub 2//sup -/ formation in the assay medium; although a threshold level of NO/sub 2//sup -/ appeared to exist beyond which the rate of NO/sub (x)/ evolution did not increase further. We previously reported that the peroxisome proliferator-activated receptor gamma coactivator-1 (PGC-1) mRNA in rat epitrochlearis muscle was increased after swimming exercise training. In the present study, we demonstrated further that PGC-1 mRNA expression in the epitrochlearis muscle of 4-5-week-old male Sprague-Dawley rats was increased after a 6-h acute bout of low-intensity swimming exercise. With this increase, the expression level was approximately 8-fold of control and immersion group rats that stayed for 6-h in warm water, maintained at the identical temperature of the swimming barrel (35 degrees C) (p<0.01). Phlorizin is a competitive inhibitor of Na/sup +//glucose symport in animal cells and of H/sup +//3-O-methylglucose (3-O-MeG) symport in liverworts. The effects of this compound on the uptake of (U-/sup 14/C) sucrose and of (/sup 14/C) 3-O-MeG by broadbean (Vicia faba L.) leaf discs were studied. Leaf discs without lower epidermis were preincubated in the dark for 30 min (fresh discs) or for 12 h (aged discs) on a medium buffered at pH 5.0 containing mannitol as an osmoticum. They were then incubated for 30 min in the light on a similar solution added with 1 mM (/sup 14/C) sucrose or (/sup 14/C) 3-O-MeG and 5 mM phlorizin. Assimilation and metabolism of /sup 14/CO/sub 2/ was compared to that of (U-/sup 14/C) sucrose in young grapefruit (ca 25 mm diameter) to determine their respective roles in fruit growth. Fixation of /sup 14/CO/sub 2/ by isolated fruit tissues during 10 min in light exceeded that in dark by 2- to 30-fold depending on tissue content of chlorophyll. Greatest apparent photosynthesis occurred in outer green peel, but green juice tissues assimilated more than did adjoining inner peel tissue. One mitigation goal of the Hungry Horse Dam fisheries mitigation program, funded by the Bonneville Power Administration, is to replace lost production of 100,000 adult kokanee in Flathead Lake. The mitigation program calls for a five-year test to determine if kokanee can be reestablished in Flathead Lake. The test consists. of annual stocking of one million hatchery-raised yearling kokanee. There are three benchmarks for judging the success of the kokanee reintroduction effort: (1) Post-stocking survival of 30 percent of planted kokanee one year after stocking; (2) Yearling to adult survival of 10 percent (100,000 adult salmon); (3) Annual kokanee harvest of 50,000 or more fish per year by 1998, with an average length of 11 inches or longer for harvested fish, and fishing pressure of 100,000 angler hours or more. A comparison between chicken and rat lung was made for microsomal prostaglandin (PG) synthesis from 1-/sup 14/C-arachidonic acid. Microsomal protein (2.0 mg) from chicken or rat lung was incubated in the presence of 20 ..mu..g of 1-/sup 14/C-arachidonic acid (specific activity = 3 x 10/sup 6/ dpm/..mu..mol for chicken; 6 x 10/sup 6/ dpm/..mu..mol for rat), 0.05 M Tris-HCl buffer (pH = 8.0), 0.5 mM epinephrine, and 1 mM reduced glutathione in a total volume of 0.5 ml in a 37/sup 0/C water bath with shaking for 15 min. After acidification with 1 M HCl to pH 3, prostaglandins were extracted with ethyl acetate. The products of the reactions were separated by reversed phase chromatography, and the radioactivity of each prostanoid fraction was determined. Sliding wear tests were run on the self-mated ceramics and ceramic/steel pairs under conditions of boundary lubrication using a laboratory tribometer with cylinder-on-plate geometry. The C isotopic composition of CH4 emissions are strongly influenced by the pathway of CH4 formation. Contrary to data from other freshwater systems, soil gas and surface flux measurements made in the tropical rain forests of Puerto Rico strongly suggest that CH4 produced in these environments was derived from CO2 reduction, rather than from acetate consumption. This study explored the effects of bacterial competition for acetate, pH, and soil structure on the pathways of CH4 formation in tropical rain forest soils. Our goal was to test two principal hypotheses: (1) ferric iron-reducing bacteria out-competed methanogens for acetate, resulting in greater CO2 reduction rather than aceticlastic methanogenesis, and (2) the low pH of tropical rain forest soils favors CO2 reduction rather than aceticlastic methanogenesis. Since chlordecone (Kepone, CD) interferes with cardiac Na{sup +} ion translocases, we have studied CD effects on cardiac SR calcium pump activity. SR was isolated from heart ventricles of male Sprague-Dawley rats. Cardiac SR Ca{sup 2+}-ATPase, {sup 45}Ca-uptake and cAMP as well as calmodulin (CaM) dependent protein phosphorylation were measured. Ca{sup 2+}-ATPase was differentiated into low affinity and high affinity forms by measuring the activity using 50 and 0.7 {mu}M free Ca{sup 2+} respectively. CD in vitro inhibited {sup 45}Ca-uptake by SR in a concentration dependent manner with an IC50 value of 7 {mu}M and SR {sup 45}Ca-uptake was totally inhibited at 20-30 {mu}M CD. In agreement with this, both high affinity and low affinity Ca{sup 2+}-ATPases, which are involved in Ca{sup 2+} transport across membranes, were also inhibited by CD in a concentration dependent manner with IC50 values of 0.7 and 3.2 {mu}M respectively.
