  1. Antioxidant capacities of ten edible North American plants.

    The EtOAc extract obtained from ten edible North American plants, Acorus calamus, Clintonia borealis, Gaultheria shallon, Juniperus osteosperma, Opuntia polyacantha, Prunus americana, Prunus virginiana, Sambucus cerulea, Sorbus americana and Vaccinium parvifolium, were tested in the 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical assay. High antioxidant activity was obtained from the extracts of three fruits, Gaultheria shallon, Sambucus cerulea and Prunus americana and one extracted rhizome, Acorus calamus. Catechin and epicatechin, potent polyphenolic antioxidants, were identified in the EtOAc extracts of Gaultheria shallon and Sambucus cerulea by reversed-phase thin-layer chromatography (TLC) and reversed-phase high-performance liquid chromatography (HPLC).     Full Text Available The genus Gaultheria, comprised of approximately 134 species, is mostly used in ethnic drugs to cure rheumatism and relieve pain. Phytochemical investigations of the genus Gaultheria have revealed the presence of methyl salicylate derivatives, C6-C3 constituents, organic acids, terpenoids, steroids, and other compounds. Methyl salicylate glycoside is considered as a characteristic ingredient in this genus, whose anti-rheumatic effects may have a new mechanism of action. In this review, comprehensive information on the phytochemistry, volatile components and the pharmacology of the genus Gaultheria is provided to explore its potential and advance research.

    Gaultheria yunnanensis (Franch.) Rehder is a kind of traditional Chinese herbal medicine used for the treatments of rheumatoid arthritis, swelling and pain. Two methyl salicylate glycosides, namely methyl benzoate-2-O-β-D-xylopyranosyl(1-6)-O-β-D-gluco-pyranoside (J12122) and methyl benzoate-2-O-β-D-xylopyranosyl(1-2)[O-β-D-xylopyranosyl(1-6)]-O-β-D-glucopyranoside (J12123), are natural salicylic derivatives isolated from Gaultheria yunnanensis. In this study, we investigated the anti-inflammatory activity of J12122 and J12123 on LPS-induced RAW264.7 macrophage cells by measuring the production of pro-inflammatory cytokines, accumulation of nitric oxide (NO), and level of reactive oxygen species (ROS). The results showed that both methyl salicylate glycosides dose-dependently inhibited the production of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and IL-6, respectively. Consistent with these observations, J12122 and J12123 significantly suppressed the accumulation of NO, with an inhibitory rate of 56.20% and 51.72% at 3.0 μg/mL concentration, respectively. Furthermore, the two methyl salicylate glycosides reduced the level of ROS induced by LPS. These results showed that the isolated compounds possess anti-inflammatory properties through inhibition the production pro-inflammatory cytokines, NO, and ROS.

  6. Pharmacognostic standardization and preliminary phytochemical studies of Gaultheria trichophylla.

    Gaultheria trichophylla Royle (Ericaceae) has long been used for various ailments in traditional systems of medicines; most importantly it is used against pain and inflammation. This study determines various pharmacognostic and phytochemical standards helpful to ensure the purity, safety, and efficacy of medicinal plant G. trichophylla. Intact aerial parts, powdered materials, and extracts were examined macro- and microscopically and pharmacognostic standardization parameters were determined in accordance with the guidelines given by the World Health Organization (WHO). Parameters including extractive values, ash values, and loss on drying were determined. Preliminary phytochemical tests, fluorescence analysis, and chromatographic profiling were performed for the identification and standardization of G. trichophylla. The shape, size, color, odor, and surface characteristics were noted for intact drug and powdered drug material of G. trichophylla. Light and scanning electron microscope images of cross section of leaf and powdered microscopy revealed useful diagnostic features. Histochemical, phytochemical, physicochemical, and fluorescence analysis proved useful tools to differentiate the powdered drug material. High-performance liquid chromatography (HPLC) analysis showed the presence of important phytoconstituents such as gallic acid, rutin, and quercetin. The data generated from the present study help to authenticate the medicinally important plant G. trichophylla. Qualitative and quantitative microscopic features may be helpful for establishing the pharmacopeia standards. Morphology as well as various pharmacognostic aspects of different parts of the plant were studied and described along with phytochemical and physicochemical parameters, which could be helpful in further isolation and purification of medicinally important compounds.

    Gaultheria series Trichophyllae Airy Shaw is an angiosperm clade of high-alpine shrublets endemic to the Himalaya-Hengduan Mountains and characterized by recent species divergence and convergent character evolution that has until recently caused much confusion in species circumscription. Although multiple DNA sequence regions have been employed previously, phylogenetic relationships among species in the group have remained largely unresolved. Here we examined the effectiveness of the plastid genome for improving phylogenetic resolution within the G. series Trichophyllae clade. Plastid genomes of 31 samples representing all 19 recognized species of the series and three outgroup species were sequenced with Illumina Sequencing technology. Maximum likelihood (ML), maximum parsimony (MP) and Bayesian inference (BI) phylogenetic analyses were performed with various datasets, i.e., that from the whole plastid genome, coding regions, noncoding regions, large single-copy region (LSC) and inverted-repeat region a (IRa). The partitioned whole plastid genome with inverted-repeat region b (IRb) excluded was also analyzed with ML and BI. Tree topologies based on the whole plastid genome, noncoding regions, and LSC region datasets across all analyses, and that based on the partitioned dataset with ML and BI analyses, are identical and generally strongly supported. Gaultheria series Trichophyllae form a clade with three species and one variety that is sister to a clade of the remaining 16 species; the latter comprises seven main subclades. Interspecific relationships within the series are strongly supported except for those based on the coding-region and IRa-region datasets. Eight divergence hotspot regions, each possessing >5% percent variable sites, were screened across the whole plastid genome of the 28 individuals sampled in the series.   8. Effect of crude saponins from Gaultheria trichophylla extract on growth inhibition in human colorectal cancer cells

    Full Text Available The genus Gaultheria also comprised of species with reported cytotoxic activities. Current research work was carried out to evaluate G. trichophylla crude extract and respective saponins fraction against human colorectal cancer cell line (Caco-2 based on cell viability assays. Caco-2 cells treated with the crude extract showed significant growth inhibition (p< 0.001 in a dose dependent manner with apparent IC50 value of 200 μg/mL and 100 μg/mL in MTT and NRU assays respectively. The fractioned crude saponins showed an enhanced response and inhibited the growth of Caco-2 by 93.6 and 97.4% in MTT and NRU assays respectively, with compared to actinomycin-D (65%. The DAPI staining of cell treated with crude saponins observed under confocal microscope showed shrunken nuclei with apparent nuclear fragmentation and chromatin condensation indicating apoptosis mode of cell death. The study exhibited that the G. Trichophylla saponins induced apoptosis of Caco-2 cell lines. This study provides new evidences to further explore this plant for the novel targets in anticancer drug development.

    Full Text Available Objective: To evaluate antifeedant, insecticidal and insect growth inhibitory activities of eucalyptus oil (Eucalyptus globules and gaultheria oil (Gaultheria procumbens L. against black cutworm, Agrotis ipsilon. Methods: Antifeedant, insecticidal and growth inhibitory activities of eucalyptus oil and gaultheria oil were tested against black cutworm, A. ipsilon. Results: Significant antifeedant activity was found in eucalyptus oil (96.24% where as the highest insecticidal activity was noticed in gaultheria oil (86.92%. Percentages of deformities were highest on gaultheria oil treated larvae and percentage of adult emergence was deteriorated also by gaultheria oil. Conclusions: These plants oil has potential to serve as an alternative eco-friendly control of insect pest.

  10. Morphometric and phytochemical characterization of chaura fruits (Gaultheria pumila: a native Chilean berry with commercial potential

    Full Text Available BACKGROUND: For the first time, a morphometric characterization of chaura (Gaultheria pumila fruits has been conducted between natural populations growing in the Villarrica National Park, Araucania Region, Chile. Chaura is a native Ericaceae from Chile that produces aromatic and tasty fruits which could be of agricultural interest. RESULTS: To influence the decision for a further domestication of G. pumila, both the fruit sizes (indicator of productivity and the nutritional properties of the fruits have been determined from different subpopulations. Samples were a total of 74 plants and 15 fruits per plant which were randomly harvested following its natural distribution around the Villarrica volcano. Altogether, fresh weight, shape, color, diameter in the pole and the equatorial dimensions were determined as phenotypic traits of the G. pumila fruits. Meanwhile the total soluble solids, anthocyanin and pectin contents were calculated as nutritional traits of the Chaura fruits. Results showed a high phenotypic diversity between the sampled population with three main fruit shapes and three predominant colors. The round shapes were the most abundant, whereas a significant correlation was found among fruit size with weight and color. The highest fresh weight (597.3 mg, pole diameter (7.1 mm and equatorial diameter (6.5 mm were estimated in the pink color fruits. CONCLUSIONS: The total amount of anthocyanin was higher in red fruits, while the maximum pectin content was obtained in the round white fruits. Overall results must pave the way for a further domestication and introduction of the Chaura species in the agro-productive system in Chile.

  11. Metabolite Profiling of Eastern Teaberry (Gaultheria procumbens L. Lipophilic Leaf Extracts with Hyaluronidase and Lipoxygenase Inhibitory Activity

    Full Text Available The phytochemical profile and anti-inflammatory activity of Gaultheria procumbens dry lipophilic leaf extracts were evaluated. Forty compounds were identified by GC-MS, representing 86.36% and 81.97% of the petroleum ether (PE and chloroform (CHE extracts, respectively, with ursolic acid (28.82%, oleanolic acid (10.11%, methyl benzoate (10.03%, and methyl salicylate (6.88% dominating in CHE, and methyl benzoate (21.59%, docosane (18.86%, and octacosane (11.72% prevailing in PE. Three components of CHE were fully identified after flash chromatography isolation and spectroscopic studies as (6S,9R-vomifoliol (4.35%, 8-demethyl-latifolin (1.13%, and 8-demethylsideroxylin (2.25%. Hyaluronidase and lipoxygenase inhibitory activity was tested for CHE (IC50 = 282.15 ± 10.38 μg/mL and 899.97 ± 31.17 μg/mL, respectively, PE (IC50 = 401.82 ± 16.12 μg/mL and 738.49 ± 15.92 μg/mL, and nine of the main constituents versus heparin (IC50 = 366.24 ± 14.72 μg/mL and indomethacin (IC50 = 92.60 ± 3.71 μg/mL as positive controls. With the best activity/concentration relationships, ursolic and oleanolic acids were recommended as analytical markers for the extracts and plant material. Seasonal variation of both markers following foliar development was investigated by UHPLC-PDA. The highest levels of ursolic (5.36–5.87 mg/g DW of the leaves and oleanolic (1.14–1.26 mg/g DW acids were observed between August and October, indicating the optimal season for harvesting.

    Full Text Available Murta (Ugni molinae Turcz. and common chaura (Gaultheria phillyreifolia (Pers. Sleumer are native species of Chile. Plants of both species have shown over-branching like witches' broom. The causal agents of these symptoms in many plants are phytoplasma. To verify the presence of these microorganisms, DAPI (4',6-diamidino-2-phenylindole staining analysis and polymerase chain reaction (PCR were performed in symptomatic and asymptomatic plants. Positive PCR samples were sequenced to identify the pathogens involved. In individuals of both species with witches’ broom symptoms, DAPI staining showed fluorescent bodies in the phloem tissues, but not in asymptomatic plants. Verification by nested-PCR, phytoplasmatic DNA was amplified from diseased murta and chaura, but not in apparently healthy plants. Sequencing of amplified products allowed locating phytoplasma within the ash yellows group (16SrVII and related to Candidatus phytoplasma fraxini. This is the first report of phytoplasma in Chilean native species. Considering the diversity of plant species infected by the ash yellows group suggests that G. phillyreifolia and U. molinae could be a phytoplasma reservoir for other economically important agricultural crops.La murta (Ugni molinae Turcz. y la chaura común (Gaultheria phillyreifolia (Pers.     Full Text Available Essential oil from Gaultheria procumbens is mainly composed of methylsalicylate (>96%, a compound which can be metabolized in plant tissues to salicylic acid, a phytohormone inducing plant immunity against microbial pathogens. The potential use of G. procumbens essential oil as a biocontrol agent was evaluated on the model plant Arabidopsis thaliana. Expression of a selection of defence genes was detected 1, 6 and 24 hours after essential oil treatment (0.1 ml/L using a high-throughput qPCR-based microfluidic technology. Control treatments included methyl jasmonate and a commercialized salicylic acid analog, benzo(1,2,3-thiadiazole-7carbothiolic acid (BTH. Strong induction of defence markers known to be regulated by the salicylic acid pathway was observed after the treatment with G. procumbens essential oil. Treatment induced the accumulation of total salicylic acid in the wild -type Arabidopsis line Col-0 and analysis of the Arabidopsis line sid2, mutated in a salicylic acid biosynthetic gene, revealed that approximately 30% of methylsalicylate sprayed on the leaves penetrated inside plant tissues and was demethylated by endogenous esterases. Induction of plant resistance by G. procumbens essential oil was tested following inoculation with a GFP-expressing strain of the Arabidopsis fungal pathogen Colletotrichum higginsianum. Flurorescence measurement of infected tissues revealed that treatments led to a strong reduction (60% of pathogen development and that the efficacy of the G. procumbens essential oil was similar to the commercial product BION®. Together, these results show that the G. procubens essential oil is a natural source of methylsalicylate which can be formulated to develop new biocontrol products.

    Methyl salicylate-2-O-β-D-lactoside (MSL), a natural salicylate derivative of Gaultheria yunnanensis (Franch.) Rehder (G. yunnanensis), has been shown to provide a beneficial anti-inflammatory effect in animal models. Studies on the pharmacokinetics and bioavailability of MSL can provide both a substantial foundation for understanding its mechanism and empirical evidence to support its use in clinical practice. A simple and sensitive high-performance liquid chromatography (HPLC) method, coupled with ultraviolet analyte detection, was developed for determining the concentration of MSL and its metabolite in beagle plasma. Chromatographic separation was achieved on a Agilent Zorbax SB-C18 column (5 μM,4.6 × 250 mm). The mobile phase consisted of aqueous solution containing 0.1% phosphoric acid and acetonitrile (82:90, v/v), at a flow rate of 1 mL/min. Validation of the assay demonstrated that the developed HPLC method was sensitive, accurate and selective for the determination of MSL and its metabolite in dog plasma. After orally administering three doses of MSL, it could no longer be detected in dog plasma and its metabolite, salicylic acid, was detected. Salicylic acid showed a single peak in the plasma concentration-time curves and linear pharmacokinetics following the three oral doses (r(2) > 0.99). In contrast, only MSL was detected in plasma following intravenous administration. These results will aid in understanding the pharmacological significance of MSL. Methods In 2003 we conducted semi-structured interviews with 60 participants obtained using a purposive sample. Medicinal plants are used to treat a range of conditions. A draft manual prepared from the data was then evaluated by participants at a participatory workshop. Results There are 128 plants used for ruminant health and diets, representing several plant families. The following plants are used for abscesses: Berberis aquifolium/Mahonia aquifolium Echinacea purpurea, Symphytum officinale, Bovista pila, Bovista plumbea, Achillea millefolium and Usnea longissima. Curcuma longa L., Salix scouleriana and Salix lucida are used for caprine arthritis and caprine arthritis encephalitis.Euphrasia officinalis and Matricaria chamomilla are used for eye problems. Wounds and injuries are treated with Bovista spp., Usnea longissima, Calendula officinalis, Arnica sp., Malva sp., Prunella vulgaris, Echinacea purpurea, Berberis aquifolium/Mahonia aquifolium, Achillea millefolium, Capsella bursa-pastoris, Hypericum perforatum, Lavandula officinalis, Symphytum officinale and Curcuma longa. Wounds and injuries are treated with Bovista spp., Usnea longissima, Calendula officinalis, Arnica sp., Malva sp., Prunella vulgaris, Echinacea purpurea, Berberis aquifolium/Mahonia aquifolium, Achillea millefolium, Capsella bursa-pastoris, Hypericum perforatum, Lavandula officinalis, Symphytum officinale and Curcuma longa. Syzygium aromaticum and Pseudotsuga menziesii are used for coccidiosis. The following plants are used for diarrhea and scours: Plantago major, Calendula officinalis, Urtica dioica, Symphytum officinale, Pinus ponderosa, Potentilla pacifica, Althaea officinalis, Anethum graveolens, Salix alba and Ulmus fulva. Gaultherin has some characterictics which make it potential to become a natural aspirin anti-cancer antiinflamatory dan cardiopulmonary. Currently aspirin acetylsalicylic acid is a medicine which is used by most of the people in this world because of its function as antipiretic antiinflamatory and analgesic. Approximately the need of pharmacy industry towards gaultherin will be increased in the following year. However at the time being there is still no any effective methods to produce gaultherin from gandapura. This difficulty in the process of taking gaultherin is based on the process of its extraction where the tissue is broken so gaultherin will be hydrolyzed change to be its individual components like methyl salicylate and disaccharides. The hydrolysis process is believed to be catalyzed by the enzyme gaultherase inside. This research is aimed to analyze the production of gaultherin form gandapura using the gaultherase enzyme inactivation process through extraction with alcoholic solvent and determine the correct condition to get the highest production of gaultherin. The result of the calculation shows that the bioextraction process variables of gaultherase enzyme inactivation which is mostly influential are pH and alcohol concentration. The more pH extraction will increase the outcome of gaultherin active compounds The optimum condition of bioextraction enzyme inactivation is in pH 8 with 1446 gaultherin active compounds and regression equation in . The bigger solvent concentration the more gaultherin be extracted. The production of gaultherin will optimally reached in the 90 concentration of ethanol with the result of 1310 active compounds

    Methyl salicylate-2-O-β-d-lactoside (MSL) is a natural salicylate derivative from the traditional Chinese medicine of Gaultheria yunnanensis (Franch.) Rehder (G. yunnanensis). As a non-steroidal anti-inflammatory drug (NSAID), MSL exerts a significant anti-arthritis effect but hardly has any gastrointestinal toxicity. In this paper, the pharmacokinetics, distribution, excretion and identification of MSL and its metabolites are described following rat oral and intravenous administration. The biological samples were quantified by UPLC-MS/MS and the metabolites in urine and feces were identified by using Q-TOF-MS.     Full Text Available Los coprolitos son heces fósiles deshidratadas o mineralizadas que contienen inclusiones orgánicas e inorgánicas. El estudio de las inclusiones de origen vegetal de los coprolitos permite conocer los ítems vegetales consumidos por los organismos e inferir la estacionalidad en el uso del sitio. El objetivo de este trabajo es estudiar el contenido de polen y de microrrestos vegetales de un coprolito humano del sitio CCP7 relacionado a una capa arqueológica fechada en 8920±200 C14 (UGA 7383 años AP. El sitio CCP7 está ubicado en la provincia de Santa Cruz (Argentina en el ecotono bosque-estepa. Se realizaron los análisis polínicos y microhistológicos de los restos vegetales del coprolito. Se identificaron 13 tipos polínicos, tres de ellos pertenecientes a la vegetación del bosque y el resto a la estepa. Mediante el análisis microhistológico se identificaron las especies Azorella monanthos, Empetrum rubrum y Gaultheria mucronata. El análisis simultáneo de la información extraída de los datos palinológicos y de los microhistológicos mostró coincidencias parciales entre el contenido de polen y de restos vegetales, permitiendo reconstruir la porción vegetal de la paleodieta e inferir la época del año en que el hombre que dio origen al coprolito estuvo ocupando el sitio CCP7.   Abstract Coprolites are dehydrated or mineralized fossil feces containing organic and inorganic inclusions.     Vegetation associated with lacustrine systems in Northern Patagonia was studied for heavy metal and trace element contents, regarding their elemental contribution to these aquatic ecosystems. The research focused on native species and exotic vascular plant Salix spp. potential for absorbing heavy metals and trace elements. The native species studied were riparian Amomyrtus luma, Austrocedrus chilensis, Chusquea culeou, Desfontainia fulgens, Escallonia rubra, Gaultheria mucronata, Lomatia hirsuta, Luma apiculata, Maytenus boaria, Myrceugenia exsucca, Nothofagus antarctica, Nothofagus dombeyi, Schinus patagonicus, and Weinmannia trichosperma, and macrophytes Hydrocotyle chamaemorus, Isöetes chubutiana, Galium sp., Myriophyllum quitense, Nitella sp. (algae), Potamogeton linguatus, Ranunculus sp., and Schoenoplectus californicus. Fresh leaves were analyzed as well as leaves decomposing within the aquatic bodies, collected from lakes Futalaufquen and Rivadavia (Los Alerces National Park), and lakes Moreno and Nahuel Huapi (Nahuel Huapi National Park). The elements studied were heavy metals Ag, As, Cd, Hg, and U, major elements Ca, K, and Fe, and trace elements Ba, Br, Co, Cr, Cs, Hf, Na, Rb, Se, Sr, and Zn. Geochemical tracers La and Sm were also determined to evaluate contamination of the biological tissues by geological particulate (sediment, soil, dust) and to implement concentration corrections.

    Full Text Available Objective: The present investigation was aimed to assess the impact of five plants oil for their insecticidal and repellent activity against Tribolium castaneum (Herbst, a stored grain pest and they were tested in the laboratory. Method: Five plants oil Citrus autantium, Cinnamomum zeylanicum, Gaultheria fragrantissima, Lavandula officinalis, and Ocimum sanctum were evaluated for their insecticidal and repellent activities against T. castaneum by adapting the standard protocol in vitro. Results: In Tulsi oil showed powerful repellent against T. castaneum beetles at both the concentration and this property can be clearly seen from the values at 5毺 1 (-0.60 and -0.73 in 1h and 6hr respectively and 10毺 1 (-0.56 and -0.81 in 1h and 6h respectively. Tulsi oil had more repelling property than other oil tested here against T. castaneum. Maximum percentage of mortality (76 and 92% at 48h and 72 hours after treatment respectively in Tulsi oil. Wintergreen oil showed 86% mortality at 72 hours after treatment. Conclusions: The present work for botanical products to control the insect pest of stored grain T. castaneum .These results suggest the presence of actives principles in the plant oils. Further exploration of active principles and their structural elucidations are underway.

    Methyl salicylate 2-O-β-D-lactoside (MSL) is a derivative of natural salicylate isolated from Gaultheria yunnanensis (Franch.) Rehder, which is widely used for treating rheumatoid arthritis (RA), swelling and pain. The aim of the present study was to investigate the effect of MSL on the progression of adjuvant-induced arthritis (AIA) in rat in vivo and explore the anti-inflammatory effects and mechanism of MSL in lipopolysaccharide (LPS)-treated murine macrophages RAW264.7 cells in vitro. Our results showed that MSL significantly inhibited the arthritis progression in AIA rats, decreasing the right hind paw swelling and ankle diameter, attenuating histopathological changes and suppressing the plasma levels of TNF-α and IL-1β in AIA rats. Besides, MSL had potent anti-inflammatory effects on the LPS-activated RAW264.7. MSL dose-dependently inhibited the activity of COX-1, and COX-2. Moreover, MSL prominently inhibited LPS-induced activation of MAPK in RAW264.7 cells by blocking phosphorylation of p38 and ERK. Our study suggests that MSL may be effective in the treatment of inflammatory diseases by inhibiting the pro-inflammatory cytokine production and regulating the MAPK signal pathway. Copyright © 2015.     ASBTRACT Toxicity of twenty-two essential oils to three bacterial pathogens in different horticultural systems: Xanthomonas campestris pv. phaseoli (causing blight of bean), Clavibacter michiganensis subsp. michiganensis (bacterial wilt and canker of tomato), and Pseudomonas tolaasii (causal agent of bacterial brown blotch on cultivated mushrooms) was tested. Control of bacterial diseases is very difficult due to antibiotic resistance and ineffectiveness of chemical products, to that essential oils offer a promising alternative. Minimal inhibitory and bactericidal concentrations are determined by applying a single drop of oil onto the inner side of each plate cover in macrodilution assays. Among all tested substances, the strongest and broadest activity was shown by the oils of wintergreen (Gaultheria procumbens), oregano (Origanum vulgare), and lemongrass (Cymbopogon flexuosus. Carvacrol (64.0-75.8%) was the dominant component of oregano oils, while geranial (40.7%) and neral (26.7%) were the major constituents of lemongrass oil. Xanthomonas campestris pv. phaseoli was the most sensitive to plant essential oils, being susceptible to 19 oils, while 11 oils were bactericidal to the pathogen. Sixteen oils inhibited the growth of Clavibacter michiganensis subsp. michiganensis and seven oils showed bactericidal effects to the pathogen. The least sensitive species was Pseudomonas tolaasii as five oils inhibited bacterial growth and two oils were bactericidal. Wintergreen, oregano, and lemongrass oils should be formulated as potential biochemical bactericides against different horticultural pathogens.

    Palynological, palaeoparasitological and paleobotanical studies of coprolites found in archaeological sites from Perito Moreno National Park (47°57‧S72°05‧W) yielded information on diet, palaeoenvironment and health. These studies allowed adding evidence to the reconstruction of life history of the hunter-gatherers that inhabited Patagonia during the Holocene. We examined the season of the year when camelid Lama guanicoe coprolites (5400 ± 64 yr 14C BP to 9640 ± 190 yr 14C BP) were deposited at Cerro Casa de Piedra 7 (site CCP7). The study used palynological evidence and comparison with pollen spectra of modern feces collected during summer, fall, winter and spring of 2010. The dominant types were: pollen of Nothofagus, Empetrum rubrum, Asteraceae subfam. Asteroideae, Nassauvia, Caryophyllaceae and Poaceae; fern spores; remains of Eimeria macusaniensis; and plant remains of Poaceae, Festuca pallescens, Stipa speciosa, Armeria maritima, Gaultheria mucronata and E. rubrum. Pollen spectra of modern and fossil feces were used for multivariate analysis. Coprolites associated to fall and winter modern feces. These results and those obtained from pollen concentration values and the presence of pollen types indicators of seasonality, allowed the determination of summer, fall and winter coprolites. However, caution must be taken with the seasonality results of coprolites dated earlier than 9000 years BP since the environmental conditions differed from now. The site was probably a camelid shelter during the unfavorable seasons.

    Full Text Available A fast and sensitive ultra-high performance liquid chromatography-photodiode array (UHPLC-PDA method for simultaneous quantification of oleanolic acid (OA and ursolic acid (UA in plant materials was developed. A central composite design combined with a response surface methodology was utilized to establish optimal separation conditions. The final separation was accomplished on a Zorbax Eclipse XDB-C18 column (1.8 μm, 100 mm × 3 mm I.D., Agilent, Santa Clara, CA, USA using a mixture 90:10 (v/v of methanol and 1% (w/v aqueous orthophosporic acid as a mobile phase at a flow rate of 0.44 mL/min and temperature of 18 °C. The analysis was completed in 6.2 min with satisfactory resolution of 1.5 between the target analytes. The developed method proved to be precise (relative standard deviations below 3.2%, accurate (recoveries in the range of 95.27%–98.60%, and sensitive (limits of detection (LODs in the range of 0.047–0.051 mg/mL. The method was then successfully applied to evaluate OA and UA content in real samples of selected Ericaceae plant materials (leaves of Arctostaphylos uva ursi, Vaccinium myrtillus, Vaccinium vitis idaea, Gaultheria procumbens. The content of OA and UA in investigated samples varied in the range of 0.74–4.47 mg/g dry weight (dw and 1.30–18.61 mg/g dw, respectively.

    Acute lung injury (ALI), characterized by pulmonary edema and inflammatory cell infiltration, is a common syndrome of acute hypoxemic respiratory failure. Methyl salicylate 2-O-β-d-lactoside (MSL), a natural derivative of salicylate extracted from Gaultheria yunnanensis (Franch.) Rehder, was reported to have potent anti-inflammatory effects on the progression of collagen or adjuvant-induced arthritis in vivo and in vitro. The aim of this study is to investigate the therapeutic effect of MSL on lipopolysaccharide (LPS)-induced acute lung injury and reveal underlying molecular mechanisms. Our results showed that MSL significantly ameliorated pulmonary edema and histological severities, and inhibited IL-6 and IL-1β production in LPS-induced ALI mice. MSL also reduced MPO activity in lung tissues and the number of inflammatory cells in BALF. Moreover, we found that MSL significantly inhibited LPS-induced TAK1 and NF-κB p65 phosphorylation, as well as the expression of NLRP3 protein in lung tissues. Furthermore, MSL significantly inhibited LPS-induced TAK1 and NF-κB p65 phosphorylation in Raw264.7 cells. In addition, MSL significantly inhibited nuclear translocation of NF-κB p65 in cells treated with LPS in vitro. Taken together, our results suggested that MSL exhibited a therapeutic effect on LPS-induced ALI by inhibiting TAK1/NF-κB phosphorylation and NLRP3 expression. Copyright © 2016 Elsevier B.V.     Neisseria gonorrhoeae (Ng) has developed resistance to most antimicrobial agents and the antibiotics recommended for therapy are restricted, for the most part, to third generation cephalosporins. In order to investigate new potential sources of antimicrobial agents, the antibacterial properties of 14 Canadian plants used in traditional First Nations' medicine were tested against Ng isolates having differing antimicrobial susceptibility profiles. Ethanolic extracts of 14 Canadian botanicals, analyzed by high-performance liquid chromatography, were tested for their antimicrobial activity (disc diffusion and/or agar dilution assays) against susceptible Ng reference strains and a panel of 28 Ng isolates with various antimicrobial resistance profiles. Extracts of Arctostaphylos uva ursi (kinnikinnick or bearberry), Hydrastis canadensis (goldenseal), Prunus serotina (black cherry), and Rhodiola rosea (roseroot) inhibited the growth of all Ng isolates with minimum inhibitory concentrations of 32 μg/mL, 4 to 32 μg/mL, 16 to >32 μg/mL, and 32 to 64 μg/mL, respectively. Extracts of Acorus americanus (sweet flag), Berberis vulgaris (barberry), Cimicifuga racemosa (black cohosh), Equisetum arvense (field horsetail), Gaultheria procumbens (wintergreen), Ledum groenlandicum (Labrador tea), Ledum palustre (marsh Labrador tea), Oenothera biennis (common evening primrose), Sambucus nigra (elderberry), and Zanthoxylum americanum (prickly ash) had weak or no antimicrobial activity against the Ng isolates with minimum inhibitory concentrations ≥256 μg/mL. The phytochemical berberine from H. canadensis inhibited the growth of all Ng isolates. The phytochemicals, salidroside and rosavin, present in R. rosea, also showed inhibitory activity against Ng strains. Canadian botanicals represent a potential source of novel compounds which inhibit Ng, including isolates resistant to antibiotics.

  19. Fumigant toxicity of plant essential oils against Camptomyia corticalis (Diptera: Cecidomyiidae).

    The toxicity of 98 plant essential oils against third instars of cecidomyiid gall midge Camptomyia corticalis (Loew) (Diptera: Cecidomyiidae) was examined using a vapor-phase mortality bioassay. Results were compared with that of a conventional insecticide dichlorvos. Based on 24-h LC50 values, all essential oils were less toxic than dichlorvos (LC50, 0.027 mg/cm3). The LC50 of caraway (Carum carvi L.) seed, armoise (Artemisia vulgaris L.), clary sage (Salvia sclarea L.), oregano (Origanum vulgare L.), lemongrass [Cymbopogon citratus (DC.) Stapf], niaouli (Melaleuca viridiflora Gaertner), spearmint (Mentha spicata L.), cassia especial (Cinnamomum cassia Nees ex Blume), Dalmatian sage (Salvia offcinalis L.), red thyme (Thymus vulgaris L.), bay [Pimenta racemosa (P. Mill.) J.W. Moore], garlic (Allium sativum L.), and pennyroyal (Mentha pulegium L.) oils is between 0.55 and 0.60 mg/cm3. The LC50 of cassia (C. cassia, pure and redistilled), white thyme (T. vulgaris), star anise (Illicium verum Hook.f.), peppermint (Mentha X piperita L.), wintergreen (Gaultheria procumbens L.), cinnamon (Cinnamomum zeylanicum Blume) bark, sweet marjoram (Origanum majorana L.), Roman chamomile [Chamaemelum nobile (L.) All.], eucalyptus (Eucalyptus globulus Labill.), rosemary (Rosmarinus officinalis L.),Virginian cedarwood (Juniperus virginiana L.), pimento berry [Pimenta dioica (L.) Merr.], summer savory (Satureja hortensis L.), lavender (Lavandula angustifolia Mill.), and coriander (Coriandrum sativum L.) oils is between 0.61 and 0.99 mg/cm3. All other essential oils tested exhibited low toxicity to the cecidomyiid larvae (LC50, >0.99 mg/cm3). Global efforts to reduce the level of highly toxic synthetic insecticides in the agricultural environment justify further studies on the active essential oils as potential larvicides for the control of C. corticalis populations as fumigants with contact action.

  20. Chemical analysis of incense smokes used in Shaxi, Southwest China: a novel methodological approach in ethnobotany.

    Characterization and comparative analysis of the main VOCs (volatile organic compounds) present in the smoke of 11 experimentally combusted plant species used as incense in Shaxi, Southwest China. Substances which may be responsible for the pleasant smell of the smokes as well as substances with a potential pharmacological activity are discussed. We adopt the dynamic headspace sorption method for the collection of smoke samples as a novel methodological approach in ethnobotany. The VOCs were identified using gas chromatography-mass spectrometry (GC-MS). Principal component analysis and canonical discriminant analysis were performed using PASW statistics (Version 18.0.2). Among the identified compounds were 10 monoterpenoids, 7 sesquiterpenoids, 6 linear hydrocarbons, 6 methoxy phenolics, 2 benzenoids, 2 polycyclic aromatic hydrocarbons, and 2 fatty acids. Based on their volatile profiles, the species are well clustered intraspecifically and separated interspecifically. The most abundant among the compounds potentially responsible for the pleasant smells of the smokes are methyl salicylate (12.28±3.90%) for Gaultheria fragrantissima leaves, δ-cadinene (15.58±2.29%) for Juniperus squamata wood, and α-Pinene for Cupressus funebris branches (9.16±7.73%) and Pistacia weinmanniifolia branches (19.52±8.66%). A couple of substances found are known for pharmacological activity, such as methylsalycilate, beta-caryophyllene and cedrol. The species used by the local people in Shaxi for incense differ clearly with respect to the chemical compounds of their smoke. Further, incense contains substances, which are of pharmacological interest and might support medicinal uses of smoke. Cedrol with its pleasant smell and sedative properties may be an important factor why specific plants are chosen as incense. Our findings support the idea that the effects of the use of incense as well as medicinal smoke depend on both, the cultural as well as the pharmacological context

  1. Estimating contribution of anthocyanin pigments to osmotic adjustment during winter leaf reddening.

    The association between plant water stress and synthesis of red, anthocyanin pigments in leaves has led some plant biologists to propose an osmotic function of leaf reddening. According to this hypothesis, anthocyanins function as a solute in osmotic adjustment (OA), contributing to depression of osmotic potential (Ψ(π)) and maintenance of turgor pressure during drought-stressed conditions. Here we calculate the percent contribution of anthocyanin to leaf Ψ(π) during OA in two angiosperm evergreen species, Galax urceolata and Gaultheria procumbens. Both species exhibit dramatic leaf reddening under high light during winter, concomitant with declines in leaf water potential and accumulation of solutes. Data previously published by the authors on osmotic potential at full turgor (Ψ(π,100)) of G. urceolata and G. procumbens leaves before and after leaf reddening were used to estimate OA. In vivo molar concentrations of anthocyanin, glucose, fructose, and sucrose measured from the same individuals were converted to pressure equivalents using the Ideal Gas Law, and percent contribution to OA was estimated. Estimated mean OA during winter was -0.7MPa for G. urceolata and -0.8MPa for G. procumbens. In vivo concentrations of anthocyanin (3-10mM) were estimated to account for ∼2% of OA during winter, and comprised <0.7% of Ψ(π,100) in both species. Glucose, fructose, and sucrose combined accounted for roughly 50 and 80% of OA for G. urceolata and G. procumbens, respectively, and comprised ∼20% of Ψ(π,100). We observed that a co-occurring, acyanic species (Vinca minor) achieved similar OA without synthesizing anthocyanin. We conclude that anthocyanins represent a measurable, albeit meager, component of OA in red-leafed evergreen species during winter. However, due to their low concentrations, metabolic costliness relative to other osmolytes, and striking red color (unnecessary for an osmotic function), it is unlikely that they are synthesized solely for an

    Methyl salicylate-2-O-β-d-lactoside (MSL) is one of the main active components isolated from Gaultheria yunnanensis, which is a traditional Chinese medicine used to treat arthritis and various aches and pains. Pharmacological researches showed that MSL had various effective activities in both in vivo and in vitro experiments. However, the pharmacokinetics features and oral bioavailability of MSL in primates were not studied up to now. To study the pharmacokinetics of different doses of MSL in rhesus monkeys and investigate the absolute bioavailability of MSL after oral administration. Male and female rhesus monkeys were either orally administrated with MSL 200, 400 and 800 mg/kg or received an intravenous dose of 20mg/kg randomly. The levels of MSL and salicylic acid (SA) in plasma were simultaneous measured by a simple, sensitive and reproducible high performance liquid chromatography method. Mean peak plasma concentration values for groups treated with 200, 400 and 800 mg/kg doses ranged from 48.79 to 171.83 μg/mL after single-dose oral administration of MSL, and mean area under the concentration-time curve values ranged from 195.16 to 1107.76 μg/mL h. Poor linearity of the kinetics of SA after oral administration of MSL was observed in the regression analysis of the Cmax-dose plot (r(2)=0.812), CL-dose plot (r(2)=0.225) and AUC(0-t)-dose plot (r(2)=0.938). Absolute bioavailability of MSL was assessed to be 118.89 ± 57.50, 213.54 ± 58.98 and 168.72 ± 76.58%, respectively. Bioavailability of MSL after oral administration in rhesus monkeys was measured for the first time. Pharmacokinetics parameters did not appear to be dose proportional among the three oral doses of treatments, and MSL showed an apparent absolute bioavailability in excess of 100% in rhesus monkeys based on the present study. In addition, a rapid, sensitive and reliable HPLC method was established and demonstrated for the research of traditional Chinese medicine in this study. Copyright
