  4. Spatial Heterogeneity of the Forest Canopy Scales with the Heterogeneity of an Understory Shrub Based on Fractal Analysis

    Full Text Available Spatial heterogeneity of vegetation is an important landscape characteristic, but is difficult to assess due to scale-dependence. Here we examine how spatial patterns in the forest canopy affect those of understory plants, using the shrub Canada buffaloberry (Shepherdia canadensis (L. Nutt. as a focal species. Evergreen and deciduous forest canopy and buffaloberry shrub presence were measured with line-intercept sampling along ten 2-km transects in the Rocky Mountain foothills of west-central Alberta, Canada. Relationships between overstory canopy and understory buffaloberry presence were assessed for scales ranging from 2 m to 502 m. Fractal dimensions of both canopy and buffaloberry were estimated and then related using box-counting methods to evaluate spatial heterogeneity based on patch distribution and abundance. Effects of canopy presence on buffaloberry were scale-dependent, with shrub presence negatively related to evergreen canopy cover and positively related to deciduous cover. The effect of evergreen canopy was significant at a local scale between 2 m and 42 m, while that of deciduous canopy was significant at a meso-scale between 150 m and 358 m. Fractal analysis indicated that buffaloberry heterogeneity positively scaled with evergreen canopy heterogeneity, but was unrelated to that of deciduous canopy. This study demonstrates that evergreen canopy cover is a determinant of buffaloberry heterogeneity, highlighting the importance of spatial scale and canopy composition in understanding canopy-understory relationships.

    The ability of actinorhizal shrubs to tolerate inhospitable conditions while improving soil fertility and organic matter status has led to increased usage of these plants for land reclamation and amenity planting purposes. Silver-berry and buffalo-berry are two such shrubs being tested as potential candidates for the revegetation of the oil sands tailings in northeastern Alberta. Associated with the roots of silver-berry and buffalo-berry are two symbiants, the N/sub 2/-fixing actimomycete Frankia and the vesicular-arbuscular mycorrhizal (VAM) fungi. Numerous studies have demonstrated that, particularly in nutrient-limited conditions, mycorrhization and nodulation can result in significantly better plant performance as a consequence of improved N and P nutrition. It was found in this study that in Alberta, silver-berry and buffalo-berry are strictly VA mycorrhizal; that they are highly dependent on their symbiants for optimum growth; and that the VAM inoculum potential of both stockpiled and undisturbed muskeg peak is negligible, due to the absence of VAM hosts. Means to increase the inoculum potential of peat have been studied. The efficacy of inoculating seedlings grown in greenhouses with VAM and Frankia has been demonstrated. Overwinter mortality was higher for inoculated shrubs, but after one growing season, shoot-weights of silver-berry were 3 to 7 times greater than for uninoculated shrubs, and shoot weights of buffalo-berry were 3 to 5 times greater. 122 refs., 12 figs., 31 tabs.

    Full Text Available Round-leaf sundew (Drosera rotundifolia L., family Droseraceae, genus Drosera, is one of a few plant species with a strong antifungal potential. Chitinases of carnivorous plants play an important role in decomposition of chitin-containing cell structures of insect prey. The cell wall of many phytopathogenic fungi also contains chitin, which can be utilized by chitinases, thus round-leaf sundew represents an interesting gene source for plant biotechnology. The purpose of this study was to compare the suitability of 3 different E. coli expression strains (E. coli BL21- CodonPlus® (DE3-RIPL, E. coli ArcticExpress (DE3RIL and E. coli SHuffle® T7 for production and isolation of heterologous round-leaf sundew chitinase (DrChit. Results showed that the recombinant protein was successfully expressed in all three strains, but occurred in insoluble protein fraction. To get the DrChit protein into soluble protein fraction some modifications concerning to induction temperatures and concentration of the IPTG inductor were tested. In addition, composition of lysis buffer has been modified with supplementation of strong non-ionic detergents, Triton® X100 and Tween® 20, respectively. As these modifications didn’t increase the amount of the DrChit protein in soluble fraction, therefore, its isolation under denaturing conditions and subsequent refolding for activity assays is recommended.

    Induction of plant-derived chitinases in the leaves of a carnivorous plant was demonstrated using aseptically grown round-leaf sundew (Drosera rotundifolia L.). The presence of insect prey was mimicked by placing the chemical inducers gelatine, salicylic acid and crustacean chitin on leaves. In addition, mechanical stirring of tentacles was performed. Chitinase activity was markedly increased in leaf exudates upon application of notably chitin. Application of gelatine increased the proteolytic activity of leaf exudates, indicating that the reaction of sundew leaves depends on the molecular nature of the inducer applied. In situ hybridization of sundew leaves with a Drosera chitinase probe showed chitinase gene expression in different cell types of non-treated leaves, but not in the secretory cells of the glandular heads. Upon induction, chitinase mRNA was also present in the secretory cells of the sundew leaf. The combined results indicate that chitinase is likely to be involved in the decomposition of insect prey by carnivorous plants. This adds a novel role to the already broad function of chitinases in the plant kingdom and may contribute to our understanding of the molecular mechanisms behind the ecological success of carnivorous plants in nutritionally poor environments.

    An overwhelming body of research has now firmly established that the dietary intake of berry fruits has a positive and profound impact on human health, performance, and disease. Berry fruits, which are commercially cultivated and commonly consumed in fresh and processed forms in North America, include blackberry ( Rubus spp.), black raspberry ( Rubus occidentalis), blueberry ( Vaccinium corymbosum), cranberry (i.e., the American cranberry, Vaccinium macrocarpon, distinct from the European cranberry, V. oxycoccus), red raspberry ( Rubus idaeus) and strawberry ( Fragaria x ananassa). Other berry fruits, which are lesser known but consumed in the traditional diets of North American tribal communities, include chokecherry ( Prunus virginiana), highbush cranberry ( Viburnum trilobum), serviceberry ( Amelanchier alnifolia), and silver buffaloberry ( Shepherdia argentea). In addition, berry fruits such as arctic bramble ( Rubus articus), bilberries ( Vaccinuim myrtillus; also known as bog whortleberries), black currant ( Ribes nigrum), boysenberries ( Rubus spp.), cloudberries ( Rubus chamaemorus), crowberries ( Empetrum nigrum, E. hermaphroditum), elderberries ( Sambucus spp.), gooseberry ( Ribes uva-crispa), lingonberries ( Vaccinium vitis-idaea), loganberry ( Rubus loganobaccus), marionberries ( Rubus spp.), Rowan berries ( Sorbus spp.), and sea buckthorn ( Hippophae rhamnoides), are also popularly consumed in other parts of the world. Recently, there has also been a surge in the consumption of exotic "berry-type" fruits such as the pomegranate ( Punica granatum), goji berries ( Lycium barbarum; also known as wolfberry), mangosteen ( Garcinia mangostana), the Brazilian açaí berry ( Euterpe oleraceae), and the Chilean maqui berry ( Aristotelia chilensis).     Full Text Available Round-leaf sundew (Drosera rotundifolia L. from Droseraceae family belongs among a few plant species with strong antifungal potential. It was previously shown that chitinases of carnivorous plant species may play role during the insect prey digestion, when hard chitin skeleton is being decomposed. As many phytopathogenic fungi contain chitin in their cell wall our attention in this work was focused on isolation and in silico characterization of genomic DNA sequence of sundew chitinase gene. Subsequently this gene was fused to strong constitutive CaMV35S promoter and cloned into the plant binary vector pBinPlus and tested in A. tumefaciens LBA 4404 for its stability. Next, when transgenic tobacco plants are obtained, increasing of their antifungal potential will be tested.

    Four wild berry species, Amelanchier alnifolia, Viburnum trilobum, Prunus virginiana, and Shepherdia argentea, all integral to the traditional subsistence diet of Native American tribal communities, were evaluated to elucidate phytochemical composition and bioactive properties related to performance and human health. Biological activity was screened using a range of bioassays that assessed the potential for these little-known dietary berries to affect diabetic microvascular complications, hyperglycemia, pro-inflammatory gene expression, and metabolic syndrome symptoms. Nonpolar constituents from berries, including carotenoids, were potent inhibitors of aldose reductase (an enzyme involved in the etiology of diabetic microvascular complications), whereas the polar constituents, mainly phenolic acids, anthocyanins, and proanthocyanidins, were hypoglycemic agents and strong inhibitors of IL-1beta and COX-2 gene expression. Berry samples also showed the ability to modulate lipid metabolism and energy expenditure in a manner consistent with improving metabolic syndrome. The results demonstrate that these berries traditionally consumed by tribal cultures contain a rich array of phytochemicals that have the capacity to promote health and protect against chronic diseases, such as diabetes.

  17. The present flora and vegetation of the moraines of the Klutlan Glacier, Yukon Territory, Canada: A study in plant succession*1

    The flora and vegetation of six ice-cored moraines of the Klutlan Glacier were analyzed in 65 plots by European plant-sociological techniques. The age of each plot was estimated from annual growth rings of shrubs or trees in the plots. Nine major vegetation types are distinguished: Crepis nana, Dryas drummondii, Hedysarum mackenzii, Hedysarum-Salix, Salix-Shepherdia canadensis, Picea-Salix, Picea-Arctostaphylos, Picea-Ledum, and Picea-Rhytidium. These contain plants aged 2-6, 9-23, 10-20, 24-30, 32-58, 58-80, 96-178, 177-240, and >163- >339 yr, respectively. Six other vegetation types are described from windthrow areas, drainage channels, volcanic tephra slopes, lake margins, fens, and drained lakes. The major vegetation types reflect a vegetational succession related to moraine age and stability, with the Crepis nana type as the pioneer vegetation developing through the other vegetation types to the Picea-Rhytidium type on the oldest moraines. Changes in species diversity and soil development, particularly humus accumulation, parallel the vegetational succession. This succession differs from patterns of revegetation of deglaciated landscapes in Alaska and British Columbia today and in Minnesota in late-Wisconsin times because of differences in climate, plant migration, and local ecology.

    Prospects for a comprehensive inventory of global biodiversity would be greatly improved by automating methods of species delimitation. The general mixed Yule-coalescent (GMYC) was recently proposed as a potential means of increasing the rate of biodiversity exploration. We tested this method with simulated data and applied it to a group of poorly known bats (Hipposideros) from the Philippines. We then used echolocation call characteristics to evaluate the plausibility of species boundaries suggested by GMYC. In our simulations, GMYC performed relatively well (errors in estimated species diversity less than 25%) when the product of the haploid effective population size (N(e)) and speciation rate (SR; per lineage per million years) was less than or equal to 10(5), while interspecific variation in N(e) was twofold or less. However, at higher but also biologically relevant values of N(e) × SR and when N(e) varied tenfold among species, performance was very poor. GMYC analyses of mitochondrial DNA sequences from Philippine Hipposideros suggest actual diversity may be approximately twice the current estimate, and available echolocation call data are mostly consistent with GMYC delimitations. In conclusion, we consider the GMYC model useful under some conditions, but additional information on N(e), SR and/or corroboration from independent character data are needed to allow meaningful interpretation of results.
