In this study, apoptosis was identified by TUNEL staining and then related to cell proliferation, as determined by Ki-67 staining. The apoptotic index was at its highest at 8 days of involution, whereas the proliferation index was at its highest during lactation. Morusin is a pure compound isolated from root bark of Morusaustralis (Moraceae). In this study, we demonstrated that morusin significantly inhibited the growth and clonogenicity of human colorectal cancer HT-29 cells. Apoptosis induced by morusin was characterized by accumulation of cells at the sub-G_1 phase, fragmentation of DNA, and condensation of chromatin. Chicken B cell population in the bursa of Fabricius are very susceptible to PCD. Our present studies concern the development of radiation damage of chicken defence mechanisms. In 6 experiments pathogen free chicken were irradiated by gamma rays with the total doses of 0.25, 0.5, 1.0, 2.0 and 4.0 Gy. The induction of apoptosis was checked by Flow-cyto-meter 12 h after irradiation in bursa cell suspension. There is some increase in the number of induced apoptotic cells 12 h after irradiation at the dose 0.5-.4.0 Gy. A range of 6-substituted imidazo[1,2-a]pyridines were synthesized using a multicomponent coupling reaction. Most of these compounds were found to exhibit excellent activity against the colon cancer cell lines HT-29 and Caco-2, whilst not showing significant toxicity against white blood cells. Our studies have shown that the proteolytic phase of apoptosis was initiated 2 h after treatment with these imidazo-[1,2-a]pyridines. The data suggests that the imidazo[1,2-a]pyridine-induced cell death in HT-29 and Caco-2 cells is mediated via pathway(s) that include the release of cytochrome c from the mitochondria to the cytosol and the activation of caspase 3 and caspase 8.

Radiation therapy and Ewing's sarcoma in childhood

A novel glioblastoma cancer gene therapy using AAV-mediated long-term expression of human TERT C-terminal polypeptide.

Despite high local control rate in Ewing's sarcoma the exact indication of radiation therapy is still controversial as well as the choice of the target volume and the optimal dose of radiation. The importance of the quality of radiotherapy has been stressed in recently published data and has shown a significant impact on long term local control with adequate radiation therapy. The dramatic improvement of precision allowed by the conformal therapy and three dimensional dosimetry allow to expect a decrease of late effects expected for second malignancies. However, late sequelae and radio-induced osteosarcoma still remain the major side effects after radiotherapy. The authors discuss the results of the main trials on Ewing's sarcoma on the choice of dose and target volume. Surgery is still the preferred choice for small tumors if the foreseen outcome is identical. (authors). 28 refs.

The COOH-terminus of telomerase reverse transcriptase (hTERT) has been shown to participatein the nuclear translocation of TERT. Here, we constructed plasmids expressing the COOH-terminal M(r) 27,000 polypeptide of hTERT (hTERTC27) withthe telomerase RNA-binding domains and the reverse transcriptase domains deleted. We showed that ectopic overexpression of this polypeptide caused a defect in telomere maintenance in hTERT-positive HeLa cells, which led to senescence-like growth arrest and apoptosis. Abstract Epstein-Barr virus (EBV), which infects not only B cells, but also T cells and natural killer (NK) cells, is associated with multiple lymphoid malignancies. Recently, the proteasome inhibitor bortezomib was reported to induce apoptosis of EBV-transformed B cells. We evaluated the killing effect of this proteasome inhibitor on EBV-associated T lymphoma cells and NK lymphoma cells. A novel single-chain immunotoxin was constructed by combining a CD33-specific single chain Fv (scFv) antibody fragment with an engineered variant of Pseudomonas exotoxin A (ETA). Colon cancer is one of the leading causes of death and a major public health problem in western countries. We examined the isoflavone content of 70% ethanol extract of soy germ (SG) and its apoptotic effect in HT-29 colon cancer cells. Our results showed that the major isoflavones of the SG extract were daidzein and genistein, and it effectively induced apoptosis in HT-29 cancer cells. In addition, nuclear factor-kappa B (NF-kB) expression was reduced in cells treated with the SG extract, which reduced cyclooxygenase-2 (COX-2) and nitric oxide synthase (iNOS) expression. Pro-inflammatory cytokines may directly influence the viability and metabolic function of colonic epithelial cells (CEC) as an early event in the development of inflammatory bowel disease. We report here that TNF-alpha+IFN-gamma induced a synergistic, concentration-dependent decline in butyrate oxidation, an essential energy supply, in HT-29 and DLD-1 cells. TNF-alpha+IFN-gamma induced a parallel profound decline in cell viability in HT-29 cells, but not in DLD-1 cells, where impairment of butyrate oxidation seemed to precede later occurrence of cell damage. TNF-alpha+INF-gamma induced CEC damage was independent on NO formation and involved the IFN-gamma signalling pathway as well as induction of apoptosis. If cytokines have similar effects in vivo, these may lead to energy deficiency and thus contribute to CEC damage and disturbance of the epithelial integrity.

Role of nuclear medicine and magnetic resonance imaging in cardiac diagnostics; Rolle der nuklearmedizinischen Verfahren und der Magnetresonanztomographie in der kardialen Diagnostik

In this article, we report anticancer activity of 14 anthracenedione derivatives separated from the secondary metabolites of the mangrove endophytic fungi Halorosellinia sp. (No. 1403) and Guignardia sp. (No. 4382). Some of them inhibited potently the growth of KB and KBv200 cells, among which compound 6 displayed strong cytotoxicity with IC(50) values of 3.17 and 3.21 microM to KB and KBv200 cells, respectively. Furthermore, we demonstrate that the mechanism involved in the apoptosis induced by compound 6 is probably related to mitochondrial dysfunction. Punica granatum peel extract protects against ionizing radiation-induced enteritis and leukocyte apoptosis in rats

Nuclear medicine and magnetic resonance imaging (MRI) are widely used in cardiac diagnostics. Both techniques have been carefully evaluated and are equivalent for the assessment of regional and global wall motion as well as the evaluation of ejection fraction and cardiac volumes, though MRI is herein currently seen as gold standard. For evaluation of myocardial perfusion, extensive data exist for nuclear imaging, that, next to high diagnostic accuracy for evaluation of coronary artery disease, show incremental prognostic information and allow for independent risk stratification. Because of rapid technical advance, myocardial perfusion imaging has also become feasible with cardiac MRI. Results of recent studies are promising, but the data are few compared to perfusion scintigraphy. Transforming growth factor-b induces nerve growth factor expression in pancreatic stellate cells by activation of the ALK-5 pathway

Radiation-induced enteritis is a well-recognized sequel of therapeutic irradiation. Therefore we examined the radioprotective properties of Punica granatum peel extract (PPE) on the oxidative damage in the ileum. Rats were exposed to a single whole-body X-ray irradiation of 800 cGy. Irradiated rats were pretreated orally with saline or PPE (50 mg/kg/day) for 10 days before irradiation and the following 10 days, while control rats received saline or PPE but no irradiation. Then plasma and ileum samples were obtained. Irradiation caused a decrease in glutathione and total antioxidant capacity, which was accompanied by increases in malondialdehyde levels, myeloperoxidase activity, collagen content of the tissue with a concomitant increase 8-hydroxy-2'-deoxyguanosine (an index of oxidative DNA damage). Also in the pancreas, NGF was associated with pain and it can stimulate the proliferation of pancreatic cancer cells. Hepatic stellate cells (HSC) respond to NGF with apoptosis. Abstract in english Cell death is preceded by severe disruption of inorganic ion homeostasis. Seconds to minutes after an injury, calcium, protons, sodium, potassium and chloride are exchanged between the cell and its environment. Simultaneously, ions are shifted between membrane compartments inside the cell, whereby mitochondria and endoplasmic reticulum play a crucial role. Depending of the type and severity of injury, two mutually exclusive metastable states can be reached, which predict (more) the final outcome. Cells characterized by large increases in cytosolic [Ca2+], [Na+]; and [Mg2+] swell and die by necrosis; alternatively, cells characterized by high [H+]and low [K+], with normal [Na+] and normal to moderate [Ca2+] increases die by apoptosis. Involvement of the NF-?B pathway in multidrug resistance induced by HBx in a hepatoma cell line.

Hsp22 (HspB8/H11) knockdown induces sam68 expression and stimulates proliferation of glioblastoma cells

Summary.? It is widely believed that hepatocellular cancer (HCC), especially HBV associated HCC, is highly resistant to chemotherapy. To investigate the molecular influence of HBx protein on multidrug resistance (MDR) in HCC and the potential role of the NF-?B pathway in this process. We established HBx-expressing cells by liposome-mediated transfection of the HBx into the HepG2 cell line. We found that HBx expression in HCC cells induces drug resistance against multiple drugs, a significantly lower apoptosis ratio in HepG2-HBx and HepG2.2.15 cells, compared with HepG2 and HepG2-3.1 cells (P?HCC cells were downregulated by IMD-0354 treatment, which is the NF-?B pathway inhibitor. Abstract Sam68 (Src-associated protein in mitosis 68-kDa) is a multifunctional protein, known to govern cellular signal transduction, transcription, RNA metabolism, proliferation, apoptosis, and HIV-1 replication. Although intrinsic mechanisms that modulate Sam68 function are beginning to emerge, the regulatory events contributing to its expression remain elusive. We previously reported that heat shock protein-22 (Hsp22) antagonizes Sam68 function in rev-response element (RRE)-mediated gene expression. Biochemical and molecular genetic approaches on the radioprotective mechanism of natural products

To identify radio-protective agent candidate among medicinal plants and to elucidate the mechanism of action of the candidate material by using modern biochemical and molecular biological methods, we screened radio-protective activity among 48 medicinal plants. Seven samples showed above 20% protective activities against oxidative cell damage: Euryale ferox, Glycyrrhiza uralensis, Salvia miltiorrhiza, Eucomia ulmoides, Paeonia suffruticosa, Spirodela polyrrhiza, and Nelumbo nucifera. We also screened for oxidative stress sensitizing activity among other 51 medicinal plants. Among those samples, 11 samples showed good sensitizing effect; Melia azedarach, Agastache rugosa, Catalpa ovata, Prunus persica, Sinomenium acutum, Pulsatilla koreana, Oldenlandia diffusa, Anthriscus sylvestris, Schizandra chinensis, Gleditsia sinensis, and Cridium officinale. Propolis derivatives inhibit the systemic inflammatory response and protect hepatic and neuronal cells in acute septic shock

We reported the radio-protective effect of DTT. The treatment of DTT increased cell survival after gamma-irradiation, decreased in the frequencies of micronucleus, and reduction in DNA fragmentation and apoptotic cells. Induction of apoptosis after UV-C irradiation was revealed by the changes in the relative cell death, increase in the relative amount of apoptotic cells, and the induction of DNA fragmentation. About 100 medicinal plants were purchased and methanol extract samples were prepared. Radio-protective and radio-sensitizing activities of these samples were screened. OBJECTIVES: To investigate the protective effect of caffeic acid phenethye ester (CAPE) against lipopolysaccharide (LPS) induced endotoxemia, hepatic and neuronal damage and the associated systemic inflammatory response (SIR). METHODS: Fifty male Wister rats were divided into: control, LPS, and CAPE+LPS groups. Functions of mammalian Cdc7 kinase in initiation/monitoring of DNA replication and development

ERK-dependent and -independent pathways trigger human neural progenitor cell migration

Cdc7 kinase plays an essential role in firing of replication origins by phosphorylating components of the replication complexes. Cdc7 kinase has also been implicated in S phase checkpoint signaling downstream of the ATR and Chk1 kinases. Inactivation of Cdc7 in yeast results in arrest of cell growth with 1C DNA content after completion of the ongoing DNA replication. In contrast, conditional inactivation of Cdc7 in undifferentiated mouse embryonic stem (ES) cells leads to growth arrest with rapid cessation of DNA synthesis, suggesting requirement of Cdc7 functions for continuation of ongoing DNA synthesis. Furthermore, loss of Cdc7 function induces recombinational repair (nuclear Rad51 foci) and G2/M checkpoint responses (inhibition of Cdc2 kinase). Besides differentiation and apoptosis, cell migration is a basic process in brain development in which neural cells migrate several centimeters within the developing brain before reaching their proper positions and forming the right connections. For identifying signaling events that control neural migration and are therefore potential targets of chemicals to disturb normal brain development, we developed a human neurosphere-based migration assay based on normal human neural progenitor (NHNP) cells, in which the distance is measured that cells wander over time. Applying this assay, we investigated the role of the extracellular signal-regulated kinases 1 and 2 (ERK1/2) in the regulation of NHNP cell migration. Dimensional variability studies and published dimensional variability standards have been used by the foundry industry for years as an indicator of the casting process` ability to produce uniform parts. These studies are an extremely useful tool in the continuous ``dimensional dialogue`` between foundries and customers. The nature of these studies, and of the current tolerancing systems used by casting designers, leaves room for some misinterpretation and misuse of these study results. This paper contains two important discussions. The first part explains exactly what these studies represent. Following this is a brief explanation on dimensional and geometric tolerances and how they communicate dimensional requirements.

This paper gives a summary of a risk study assuming unprotected loss of flow (ULOF) in the SNR 300. This study was initiated in 1979/80 by the Karlsruhe Nuclear Research Center and performed in close cooperation with Science Applications Inc., Palo Alto, USA, and Interatom Company. Part of the results also was integrated in the 'Risk Related Analysis for the SNR 300' carried out by the Gesellschaft fuer Reactorsicherheit. The character of the study described here is similar to other risk studies like the Reactor Safety Study and the German Risk Study for Nuclear Power Plants. The objectives and the methodology of the analyses are described and its results are discussed. (orig./RW).

The focus of the Chernobyl Studies Project has now turned to the issue of health effects from the Chernobyl accident. Currently, we are involved in and making progress on the case-control and co-hort studies of thyroid diseases among Belarussian children. Dosimetric aspects are a fundamental part of these studies. We are working to implement similar studies in Ukraine. A major part of the effort of these projects is supporting these studies, both by providing methods and applications of dose reconstruction and by providing support and equipment for the medical teams.

Terror management among Taiwanese: Worldview defence or resigning to fate?

Experimental investigation data are systematized of free radical states and processes in irradiated proteins. The investigation is performed by the radiospectral methods. Results are discussed in detail of the study of free radicals electronic structure of amino acids, peptides and proteins formed by the action of ionizing radiation. The specificity is stressed of the study of monocrystalls of these compounds by the method of electronic paramagnetic resonance. The nature is also studied of primary centres formed under the effect of radiation on biologically important compounds and their subsequent reactions in solid and liquid solutions. Ion-radical states of different functional groups of the protein molecule are studied. Prospects of the study and the role of anion-radicals in biological processes are discusses.

Terror management theory (TMT) proposes that people who are reminded of their mortality should be motivated to defend their cultural worldview. Studies 1 and 2 examined whether the TMT worldview defence-buffering effect found in Western cultures could be generalized to Asians in Taiwan. No such effect was found in the present studies. This non-significant result was robust when either a stronger distraction task was used (study 1) or when a subliminal manipulation of mortality salience was utilized (study 2). A meta-analysis, including 24 TMT experiments in East Asia, was also conducted (study 3). Feasibility Study-II examined a high-performance Neutrino Factory providing 1 x 10{sup 20} neutrinos per year aimed at a long-baseline detector. The Study was sponsored jointly by BNL and the Neutrino Factory and Muon Collider Collaboration ration (MC) and is based on a 1 MW proton driver operating at 24 GeV, i.e., an upgraded version of the AGS accelerator. Compared with the earlier FNAL-sponsored study (Feasibility Study-I), there is a sixfold improvement in performance. Here we describe details of the implementation of Study-II concepts and discuss their efficacy. Alternative approaches that will be pursued in follow-on R and D activities are also described briefly.

The objective of this study was to analyse one of the goals of the latest National Waste Plan of Finland: the relative reduction of hazardous waste (HW) generation by 15% over the period 1992 to 2005.Official statistics and a national database of the annual reports of HW generators were used as one basis for this study. A case study was focused on a selection of HW classes that were considered to be typical to the metal workshop industry. These HW classes were combined to form functional HW groups for the purpose of forming time series. This report presents the data and results of a site specific case study comparing costs of an overhead line with an underground circuit traversing the same route. The costs used for the overhead line are actual numbers for a line scheduled to be built by a utility. The underground circuit costs were developed using the same criteria of line rating, contingencies, and location. Often cost comparisons between overhead and underground lines are made using figures taken from studies with widely varying assumptions. This project attempts to provide a comparison by studying the costs of overhead and underground lines following the same route and using the same design assumptions. Caution must be used, however, in extending the results of this study beyond its original intent. Reverse Engineering of Gene Regulatory Networks: A Comparative Study

Accurate engineering cost estimates are critical in developing bids for new work, and for effective resource allocation and project control. This study reports on design estimating methods found in the literature, and on the results of two empirical studies of how estimating techniques are used in professional practice and their accuracy. The study found great reliance on a classic {open_quotes}activity analysis{close_quotes} approach to estimating design resources, and significant hazards in commonly used parametric techniques. The study found that project managers expect their estimates to be accurate (with 80% confidence interval) of between -10% to +25%.
