Study of Tumor Tissue Samples From Patients With Stage I, Stage II, or Stage III Malignant Melanoma

Aim: The purpose of this prospective study was to evaluate the value of immunoscintigraphy (ISG) with anti-225.28S in clinically suspected ocular melanoma. Methods: For this purpose standardized ISG was performed in 36 patients using both planar acquisition and emission computed tomography (ECT). Ocular melanoma was present in 31 patients. In 21 patients therapy was enucleation of the eye. These specimens were evaluated by histology and immunohistochemistry in 11 of 21 patients. Results: Regarding the clinical diagnosis, ISG was positive only in 15 of 31 patients with ocular melanoma, regarding histology in 11 of 21 and regarding immunohistochemistry in 5 of 6 patients with a positive immunoreaction. 5 patients showed no immunoreactivity, their ISG was negative. Intermittent sun exposure and sunburn are strong predictors of cutaneous malignant melanoma (CMM). On the other hand, melanomas may arise also in non-sun-exposed areas such as the vulva. However, little is known about a possible relationship between sun exposure and vulvar melanoma. Temporal and latitudinal dependencies of the incidence rates of vulvar melanoma were studied in comparison with those of CMM among Caucasians in Sweden, East Germany, USA and Victoria (Australia). The ratios of vulvar melanoma incidence rates to those of CMM tend to decrease with increasing CMM rates. Prevalence of Ocular Fundus Pathology in Patients with Chronic Kidney Disease

Objective: To analyze the early response of uveal melanomas in patients treated with ruthenium-106 brachytherapy. Materials And Methods: In the period between April 2002 and July 2003, 20 patients diagnosed with uveal melanoma were submitted to ruthenium-106 brachytherapy. The calculated dose delivered at the apex of the tumor ranged between 55 Gy and 100 Gy. Patients with lesions greater than 5 mm were submitted to transpupillary thermotherapy concomitantly with ophthalmic plaque insertion. Results: As regards the lesions site, 75% of the lesions were located in the choroid, 15% in the iris, and the remainder 10% in the ciliary body. In a median 19-month-follow-up, the progression-free survival for brachytherapy was 69%, and 87% for associated brachytherapy and transpupillary thermotherapy. A genetic model of melanoma tumorigenesis based on allelic losses

We irradiated proton beams on the ears of rabbits and the Harding-Passey mouse melanoma and observed their morphological change. We used 52 MeV proton beams from the INS-FM cyclotron. We adjusted the energy of the proton beams to be at the plateau part of the Bragg curve, at the half-way point of the Bragg peak, and at the Bragg peak. The amount of radiation was 5000rad in each case. The Harding-Passey mouse melanoma was transplanted into the subcutan of a three week old mouse. Previous karyotypic studies have indicated a possible series of non-random chromosomal events involved in the progression of melanoma. We sought to define a model of melanocyte tumorigenesis by studying allelic deletions of polymorphic simple tandem repeat markers mapping to chromosome 1, 6q, 7, 9p, 10, 11, 17, and 21 in thirty matched pairs of melanoma and constitutional DNAs. The most frequent and earliest deletions were found on 9p (57%) and 10q (32%) and with the exception of one case, no sample has loss of markers on another chromosome without concomitant loss of markers on 9p and/or 10q. Losses on 6q were also a frequent (32%) event that sometimes occurred in primary melanomas, whereas losses of loci on distal 1p (26%) or 11q (26%) occurred only in metastic melanomas. We investigated the efficacy of combined FDG-PET/CT imaging for the diagnosis of small-size uveal melanomas and the feasibility of combining separate, high-resolution (HR) FDG-PET with MRI for its improved localization and detection. Patients, methods: 3 patients with small-size uveal melanomas (0.2-1.5 ml) were imaged on a combined whole-body PET/CT, a HR brain-PET, and a 1.5 T MRI. Evaluation of Ocular Risk Factors Related to Asymmetric Visual Field Defects in Normal Tension Glaucoma

We compared the efficacy of timolol maleate ophthalmic gel-forming solution 0.5% QD with that of levobunolol hydrochloride 0.5% BID, as measured by change in intraocular pressure (IOP), effect on heart rate, and ocular tolerability. The study had a positive-controlled, double-masked, randomized, multicenter, 12-week, two-period (6 weeks each), crossover design. One hundred fifty-two patients with open-angle glaucoma or ocular hypertension were randomized to receive either timolol maleate gel-forming solution QD or levobunolol BID for 6 weeks, followed by a crossover to the alternate treatment. The authors have been involved for the last few years in developing high affinity sigma-receptor-specific radiopharmaceuticals for use in nuclear medicine. They recently demonstrated that sigma-1 receptors are expressed in human melanoma cells and that sigma receptors could be used as external markers for non-invasive imaging. In order to determine the presence of sigma receptors in other human tumors they have screened numerous non-small cell lung carcinoma, melanoma and breast cancer cell lines. All of these cell lines expressed high sigma receptor densities using commercially available ["3H]-(+)-pentazocine or ["3H]DTG tritiated sigma ligands. BACKGROUND: Controversy exists as to whether patients with thick (Breslow depth >4 mm), clinically lymph node-negative melanoma require sentinel lymph node (SLN) biopsy. The authors examined the impact of SLN biopsy on prognosis and outcome in this patient population. METHODS: A review of the authors institutional review board-approved melanoma database identified 293 patients with T4 melanoma who underwent surgical excision between 1998 and 2007. Patient demographics, histologic features, and outcome were recorded and analyzed. RESULTS: Of 227 T4 patients who had an SLN biopsy, 107 (47%) were positive. One of the research challenges in oncology is to develop new biochemical methods for noninvasive tumor therapy evaluation to determine whether the chemotherapeutics is effective. Vascular endothelial growth factor (VEGF) was labeled with radioiodine and evaluated in vitro as well as in vivo, using A2058, a melanoma cell line overexpressing VEGFR-1 and -2. Saturation binding analysis with [{sup 125}I]-VEGF resulted in a K {sub d} of 0.1 nM. Internalization assays indicate the preserved ligand induced internalization and metabolization of the tracer. Biodistribution studies with [{sup 123}I]-VEGF in wild type and A2058 tumor-bearing athymic mice showed low background activity and a tumor to reference tissue ratio of maximum 6.12. Purpose: The determination of tumor volume in ocular tumors is very important for the planning and success of radiation therapy. This study uses an animal model to evaluate the accuracy of MR-based volumetry of ocular tumors. Materials and methods: In a total of 25 porcine eyes obtained from the slaughterhouse, ocular tumors were produced by injecting a mixture of hand creme and Gd-DTPA under ophthalmoscopic guidance. The injected volume varied between 0.05 ml and 2.7 ml. The eyes were examined with a 1.5 Tesla scanner and a 4 cm circular surface coil especially developed for ocular MRI. The role of computed tomography in complex facial trauma

An analysis of the mortality of workers employed at the Commissariat a l'Energie Atomique (CEA) between 1946 and 1994 is presented. Standardized Mortality Ratios (SMR) are computed with reference to the French national population for the period 1968-1994. 51 286 workers are included in the study. 3 784 deaths occurred between 1968 and 1994. A healthy worker effect is observed for men (SMR=0.53 Cl{sub 90%}=[0.52; 0.55]) and for women (SMR=0.70 Cl{sub 90%}=[0.64; 0.76]). An excess is observed for male pleural cancers (SMR=1.54, Cl{sub 90%}=[1.03; 2.21]). An excess of breast cancer is observed among women, statistically significant for the 1980-1994 period (SMR=1.30, Cl{sub 90%}=[1.04; 1.61]). Tridimensional localization of bone and soft tissue injuries in complex facial trauma is fundamental for treatment. The face can be conceptualized as 3 groups of interconnected osseous struts that are oriented in the horizontal, sagittal and coronal planes. Each group of struts is closely related to specific soft tissue structures that are susceptible to injury. Any single CT section in the coronal plane or in the axial plane contains parts of one or more these struts. Facial CT in biplane were performed in 32 patients of facial trauma. The most common site of facial fractures is found in middle part of horizontal struts, geometrically in anteromedial portion of the inferior orbital fissure of the face. Exposure of Finnish population to solar UV radiation and consequent carcinogenic effects

We aim to examine the spectrum of bacteria causing corneal infections and their antibiotic susceptibility patterns. This will serve as a guideline for empiric therapy of corneal infections. We conducted the study over a period of 18 months from March 2001 through December 2002 in King Abdul-Aziz University Hospital, Riyadh, Kingdom of Saudi Arabia. Corneal specimens taken from 200 patients were inoculated directly onto different types of media. The isolates were identified and then tested against the appropriate topical or systemic antibiotics. Sixty-seven (33.5%) of the total specimens were culture positive and 133 (66.5%) were culture negative. Depletion of stratospheric ozone increases irradiance of terrestrial ultraviolet (UV) radiation at short wavelengths, which may be harmful to the human health. To understand quantitatively the risks caused by increasing UV radiation to the Finnish population, the actual UV exposure of the population has to be assessed. It was shown that the snow reflection increases the UV exposure to the face and eyes particularly in the northern Finland. In 1993 exceptionally low ozone levels persisted up to the end of May, which resulted in a theoretical increase in the annual UV dose ranging from 8 % to 13 % in Finland. This study was a retrospective evaluation of the efficacy of stereotactic radiosurgery (SRS) in patients with >4 metastases to the brain. Materials and Methods: Between January 2004 and December 2006, 68 patients with ?4 multiple brain metastases were included and reviewed retrospectively. Twenty-nine patients received SRS and 39 patients received whole brain radiotherapy (WBRT). Patients with small cell lung cancers and melanomas were excluded. To compare the accuracy in the detection and staging of various malignant tumors with high resolution whole-body MRI using parallel imaging with whole-body dual-modality PET-CT. Preliminary results of an interim analysis from a prospective, blinded study are presented, in which 20 patients (mean age 59 years, range 27-77 years) with different oncological diseases underwent whole-body dual modality FDG-PET-CT screening for tumor search or staging in case of confirmed or suspected metastatic disease. The interest of irradiation by carbon ions is in the fact that the carbon ions leave all their energy to a determined depth. ( phenomenon known under the name of Bragg peak)This high diffusion in tissue gives an RBE particularly high. The indications of this therapy are chordomas, and chondrosarcomas of the skull base, some cyst adenoid carcinomas, pulmonary cancer, sarcomas, hepato carcinomas, melanomas. Molecular phylogeography of the microturbellarian Monocelis lineata (Platyhelminthes: Proseriata) in the North-East Atlantic

The effects of a timolol maleate gel-forming solution (TMGS) on intraocular pressure (IOP), blood pressure (BP), and pupil size (PS) were evaluated in normotensive dogs. TMGS was administered once daily to six normotensive beagle dogs. TMGS administration reduced IOP and PS. The hypotensive effect persisted for 24 hr after the administration. The mean reduction in IOP was 5.3 mm Hg (P<0.01). The changes in BP and PS were not significant. These results suggest that TMGS can potentially be used in the treatments of glaucoma and ocular hypertension in dogs. PMID:16820724

Customized computer models of eyes with intraocular lenses

BackgroundMetastases to the stomach from an extra-gastric neoplasm are an unusual event, identified in less than 2% of cancer patients at autopsy. The stomach may be involved by hematogenous spread from a distant primary (most commonly breast, melanoma or lung), or by contiguous spread from an adjacent malignancy, such as the pancreas, esophagus and gallbladder. These latter sites may also involve the stomach via lymphatic or haematogenous spread. We present three cases of secondary gastric malignancy.Methods/ResultsThe first is a 19-year-old male who received a diagnosis of testicular choriocarcinoma in September 2004. Biological wound dressings sterilized with gamma radiation: Mexican clinical experience

We compared experimental wave aberrations in pseudophakic eyes with aspheric intraocular lenses (IOLs) to simulate aberrations from numerical ray tracing on customized computer eye models using corneal topography, angle ?, ocular biometry, IOL geometry, and IOL tilt and decentration measured on the same eyes. We found high correlations between real and simulated aberrations even for the eye with only the cornea, and these increased on average when the IOL geometry and position were included. Relevant individual aberrations were well predicted by the complete eye model. Corneal spherical aberration and horizontal coma were compensated by the IOL, and in 58.3% of the cases IOL tilt and decentration contributed to compensation of horizontal coma. We conclude that customized computer eye models are a good representation of real eyes with IOLs and allow understanding of the relative contribution of optical, geometrical and surgically-related factors to image quality. ...

Biological wound dressings sterilized with gamma radiation, such as amnion and pig skin, are a reality in Mexico. These tissues are currently processed in the tissue bank and sterilized in the Gamma Industrial Irradiation Plant; both facilities belong to the Instituto Nacional de Investigaciones Nucleares (ININ) (National Institute of Nuclear Research). With the strong support of the International Atomic Energy Agency, the bank was established at the ININ and the Mexican Ministry of Health issued its sanitary license on July 7, 1999. The Quality Management System of the bank was certified by ISO 9001:2000 on August 1, 2003; the scope of the system is 'Research, Development and Processing of Biological Tissues Sterilized with Gamma Radiation'. Maturation of GABAergic Inhibition Promotes Strengthening of Temporally Coherent Inputs among Convergent Pathways

Scrotal skin of black Long-Evans rats and human thigh skin were maintained in vitro as organ cultures for as long as 14 days, and examined histologically using the combined skin splitting and Dopa techniques. Selected rat skin cultures received testosterone in the culture medium and/or were irradiated with ultraviolet light (290 to 320 nm uvl). With increased time in culture, scrotal melanocytes round up and there is an increase in epidermal pigmentation. Human skin behaves similarly; after eight days in vitro human melanocytes also become rounded, but remain strongly Dopa-positive. Spike-timing-dependent plasticity (STDP), a form of Hebbian plasticity, is inherently stabilizing. Whether and how GABAergic inhibition influences STDP is not well understood. Using a model neuron driven by converging inputs modifiable by STDP, we determined that a sufficient level of inhibition was critical to ensure that temporal coherence (correlation among presynaptic spike times) of synaptic inputs, rather than initial strength or number of inputs within a pathway, controlled postsynaptic spike timing. Inhibition exerted this effect by preferentially reducing synaptic efficacy, the ability of inputs to evoke postsynaptic action potentials, of the less coherent inputs. In visual cortical slices, inhibition potently reduced synaptic efficacy at ages during but not before the critical period of ocular dominance (OD) plasticity. Dimensional variability studies and published dimensional variability standards have been used by the foundry industry for years as an indicator of the casting process` ability to produce uniform parts. These studies are an extremely useful tool in the continuous ``dimensional dialogue`` between foundries and customers. The nature of these studies, and of the current tolerancing systems used by casting designers, leaves room for some misinterpretation and misuse of these study results. This paper contains two important discussions. The first part explains exactly what these studies represent. Following this is a brief explanation on dimensional and geometric tolerances and how they communicate dimensional requirements.

This paper gives a summary of a risk study assuming unprotected loss of flow (ULOF) in the SNR 300. This study was initiated in 1979/80 by the Karlsruhe Nuclear Research Center and performed in close cooperation with Science Applications Inc., Palo Alto, USA, and Interatom Company. Part of the results also was integrated in the 'Risk Related Analysis for the SNR 300' carried out by the Gesellschaft fuer Reactorsicherheit. The character of the study described here is similar to other risk studies like the Reactor Safety Study and the German Risk Study for Nuclear Power Plants. The objectives and the methodology of the analyses are described and its results are discussed. (orig./RW).

The focus of the Chernobyl Studies Project has now turned to the issue of health effects from the Chernobyl accident. Currently, we are involved in and making progress on the case-control and co-hort studies of thyroid diseases among Belarussian children. Dosimetric aspects are a fundamental part of these studies. We are working to implement similar studies in Ukraine. A major part of the effort of these projects is supporting these studies, both by providing methods and applications of dose reconstruction and by providing support and equipment for the medical teams.

Terror management among Taiwanese: Worldview defence or resigning to fate?

Experimental investigation data are systematized of free radical states and processes in irradiated proteins. The investigation is performed by the radiospectral methods. Results are discussed in detail of the study of free radicals electronic structure of amino acids, peptides and proteins formed by the action of ionizing radiation. The specificity is stressed of the study of monocrystalls of these compounds by the method of electronic paramagnetic resonance. The nature is also studied of primary centres formed under the effect of radiation on biologically important compounds and their subsequent reactions in solid and liquid solutions. Ion-radical states of different functional groups of the protein molecule are studied. Prospects of the study and the role of anion-radicals in biological processes are discusses.

Terror management theory (TMT) proposes that people who are reminded of their mortality should be motivated to defend their cultural worldview. Studies 1 and 2 examined whether the TMT worldview defence-buffering effect found in Western cultures could be generalized to Asians in Taiwan. No such effect was found in the present studies. This non-significant result was robust when either a stronger distraction task was used (study 1) or when a subliminal manipulation of mortality salience was utilized (study 2). A meta-analysis, including 24 TMT experiments in East Asia, was also conducted (study 3). Feasibility Study-II examined a high-performance Neutrino Factory providing 1 x 10{sup 20} neutrinos per year aimed at a long-baseline detector. The Study was sponsored jointly by BNL and the Neutrino Factory and Muon Collider Collaboration ration (MC) and is based on a 1 MW proton driver operating at 24 GeV, i.e., an upgraded version of the AGS accelerator. Compared with the earlier FNAL-sponsored study (Feasibility Study-I), there is a sixfold improvement in performance. Here we describe details of the implementation of Study-II concepts and discuss their efficacy. Alternative approaches that will be pursued in follow-on R and D activities are also described briefly.

The objective of this study was to analyse one of the goals of the latest National Waste Plan of Finland: the relative reduction of hazardous waste (HW) generation by 15% over the period 1992 to 2005.Official statistics and a national database of the annual reports of HW generators were used as one basis for this study. A case study was focused on a selection of HW classes that were considered to be typical to the metal workshop industry. These HW classes were combined to form functional HW groups for the purpose of forming time series. This report presents the data and results of a site specific case study comparing costs of an overhead line with an underground circuit traversing the same route. The costs used for the overhead line are actual numbers for a line scheduled to be built by a utility. The underground circuit costs were developed using the same criteria of line rating, contingencies, and location. Often cost comparisons between overhead and underground lines are made using figures taken from studies with widely varying assumptions. This project attempts to provide a comparison by studying the costs of overhead and underground lines following the same route and using the same design assumptions. Caution must be used, however, in extending the results of this study beyond its original intent. Accurate engineering cost estimates are critical in developing bids for new work, and for effective resource allocation and project control. This study reports on design estimating methods found in the literature, and on the results of two empirical studies of how estimating techniques are used in professional practice and their accuracy. The study found great reliance on a classic {open_quotes}activity analysis{close_quotes} approach to estimating design resources, and significant hazards in commonly used parametric techniques. The study found that project managers expect their estimates to be accurate (with 80% confidence interval) of between -10% to +25%. The study also found that actual bids between engineering firms had a much greater range (-40% to +45%) than can be explained by accuracy.
