1. Choline, and the choline analogues monoethylcholine (MEC) and N-aminodeanol (NAD) were examined for prophylactic activity in acute acetylcholinesterase inhibitor toxicity in mice. The rank order of potency of the compounds was MEC greater than NAD greater than choline. 2. Simultaneous administration of MEC (60 mg kg-1) or NAD (200 mg kg-1) with physostigmine reduced lethality to 17 and 13% respectively. MEC (60 mg kg-1) completely protected against disopropylfluorophosphate (DFP) and diazinon toxicity, and NAD reduced lethality to 17% for both agents. A range of artefacts have been investigated by our laboratory, in part for the purposes of history of art and in part as a basis for restoration works - medieval frescoes in some Czech castles and churches, metal sculptures and objects of applied art, paints and inks of old manuscripts, paintings. Some of these are among the most valuable monuments in the Czech cultural heritage. The contribution of the results of the tests to the investigation of their 'life story' and, in some cases, to their conservation, is not negligible. Later additions and repairs can be recognised, and materials and technologies that are close to their historic counterparts can be used in restoration work.

Cholera toxin catalyzes transfer of radiolabel from ["3"2P]NAD"+ to several peptides in particulate preparations of human foreskin fibroblasts. Resolution of these peptides by two-dimensional gel electrophoresis allowed identification of two peptides of M/sub r/ = 42,000 and 52,000 as peptide subunits of a regulatory component of adenylate cyclase. The radiolabeling of another group of peptides (M/sub r/ = 50,000 to 65,000) suggested that cholera toxin could catalyze ADP-ribosylation of cytoskeletal proteins. Bituminization has to be selected as the process for conditioning radioactive liquid wastes arising from the two VVER V-230 reactors being built at Temelin in the Czech Republic. In the process, a thin-film evaporator, operating at a waste-product temperature of 160"oC, evaporates all free water from the waste effluents. Remaining solids are homogeneously dispersed in a bitumen matrix which solidifies through natural cooling of the binder. The relative simplicity of the process reduces construction costs for on-line waste facilities and operating costs are less given the cheap basic material and simple maintenance. The reliability of the process has been demonstrated at Western reactors and reprocessing plants though adaptations have had to be made to accept VVER effluents. (UK).
After giving a brief history of preparation plants in this area, the thinking is described behind the design of the Chabarovice plant which is to open in 1988. Its capacity is 1120 t/h of 0-200 mm brown coal and its annual through-put is 3 Mt using two-shift operation with 30 men per shift. It occupies a total volume of 38,500 m/sup 3/ and cost 1556 Czech Crowns per m/sup 3/ to construct, 380 Czech Crowns of this was taken up by the provision of pile foundations which are 30 m long and embedded in the underlying coal seam. The preparation plant building is made of reinforced concrete and is situated 8 m below ground level.
Although the first references are from the 15th Century when both Hussites and crusaders are said to have used rockets during the Hussite Wars (also known as the Bohemian Wars) there is no strong evidence that rockets were actually used at that time. It is worth noting that Konrad Kyeser, who described several rockets in his Bellifortis manuscript written 1402-1405, served as advisor to Bohemian King Wenceslas IV. In this study, the nearly complete sequence (14,519 bp) of the mitochondrial DNA (mtDNA) of the entomophagous endoparasite Xenos vesparum (Insecta: Strepsiptera) is described. All protein coding genes (PCGs) are in the arrangement known to be ancestral for insects, but three tRNA genes (trnA, trnS(gcu), and trnL(uag)) have transposed to derived positions and there are three tandem copies of trnH, each of which is potentially functional. All of these rearrangements except for that of trnL(uag) is within the short span between nad3 and nad4 and there are numerous blocks of unassignable sequence in this region, perhaps as remnants of larger scale predisposing rearrangements. Cultures of 3T3-L1 cells were incubated with either 10 ng/ml cholera toxin or 10 ng/ml pertussis toxin from 4 days prior to the initiation of differentiation and throughout the subsequent incubation. Toxin concentrations were sufficient to completely prevent the labelling of alpha-subunits with ["3"2P]NAD"+ and pertussis toxin and to prevent by more than 90% the labelling with ["3"2P]NAD"+ and cholera toxin in membranes prepared from these cells. Neither toxin prevented the differentiation to the adipocyte phenotype. Neither toxin prevented the increases in the relative amounts of G-proteins which occur upon differentiation. Both toxins dramatically decreased the amount of beta-subunits. Treatment of cultures of 3T3-L1 cells with methylisobutyl-xanthine and dexamethasone has been shown to result in accumulation of lipid and conversion to the morphology of adipocytes in more than 90% of the cells. The status of the stimulatory (Gs), inhibitory (Gi) and Go-proteins during the course of 3T3-L1 differentiation was examined. Discusses methods for determining ash content in coal using backscattering in X-ray fluorescence. The following aspects are evaluated: principle of ash content determination, energy sources, factors that influence measurement accuracy, reliability. Types of X-ray fluorescence analyzers developed in Czechoslovakia and manufactured in the Czech Republic are comparatively evaluated. Operation of the RPM 113.1 radionuclide X-ray fluorescence analyzer developed by the PAR company and used for measuring ash content in coal and coal calorific value is discussed. 7 refs.
Testing of barrier properties of insulating materials against radon is usually based on the measurement of the radon diffusion coefficient. Presented report summarizes results of radon diffusion coefficients measurements in more than 120 insulating materials obtained throughout Europe. All measurements were performed by the Czech Technical University, Faculty of Civil Engineering in cooperation with the Radiation Protection Institute. We have found out that great differences exist in diffusion properties, because the diffusion coefficients vary within eight orders from 10"-"1"5 m"2/s to 10"-"8 m"2/s. For each material category of different chemical composition statistical evaluation of results is presented. Possibilities of usage of the radon diffusion coefficient for the design of radon resisting membranes are discussed. We have determined the actual RNA sequence of four mitochondrial genes: pcf, nad3, rps12, and coxII. Because plant mitochondrial proteins cannot be predicted from DNA sequences, the actual primary structures of the encoded proteins were unknown. We have gained information concerning the process of RNA editing. Editing can occur before splicing. Our data is consistent with the hypothesis that RNA editing is not simultaneous with transcription. Unlike other systems, the process of editing in plant does not exhibit a discernable direction. As a result of our RNA editing studies, we have produced a number of clones of the proper coding regions tube used for incorporating these mitochondrial genes into the nucleus. 6 refs., 4 figs.
DOE installations possessing sufficient quantities of fissile material to potentially constitute a critical mass, such that the excessive exposure of personnel to radiation from a nuclear accident is possible, are required to provide nuclear accident dosimetry services. This document describes the personal nuclear accident dosimeter (PNAD) used by SNL and prescribes methodologies to initially screen, and to process PNAD results. In addition, this report describes PNAD dosimetry results obtained during the Nuclear Accident Dosimeter Intercomparison Study (NAD23), held during 12-16 June 1995, at Los Alamos National Laboratories. Biases for reported neutron doses ranged from -6% to +36% with an average bias of +12%.
Indoleamine 2,3-dioxygenase (IDO) and tryptophan 2,3-dioxygenase (TDO) are tryptophan-degrading enzymes. Mammalian IDO expression is induced by cytokines and has antimicrobial and immunomodulatory effects. A major role of mammalian TDO is to supply nicotinamide adenine dinucleotide (NAD(+)). In fungi, the IDO homologue is thought to be expressed constitutively and supply NAD(+), as TDO is absent from their genomes. Here, we reveal the distribution of IDO genes among fungal species and characterize their enzymatic activity. The yeast, Saccharomyces cerevisiae has only one IDO gene, whereas the koji-mold, Aspergillus oryzae has two genes, IDO? and IDO?. The A. oryzae IDO? showed more similar enzymatic properties to those of S. cerevisiae IDO than IDO?, suggesting that the A. oryzae IDO? is a functional homologue of the S. cerevisiae IDO. Which flow routing algorithms are preferable for application in vegetation ecology? Location: Forests in three different regions of the Czech Republic. Monthly sampling of slightly alkaline arsenic-rich stream in the Mokrsko gold deposit revealed seasonal variations in dissolved Zn, Cu, As and Mo. Concentrations of trace metal cations (Zn, Cu) increased as much as 330 and 178%, respectively, from minimum mean values at autumn to maximum mean values at spring. In contrast, concentrations of trace element oxyanions (As, Mo) revealed opposite seasonal pattern with increase to 189% (As) and 123% (Mo) during summer?autumn, indicating that in-stream biogeochemical process(es) played the main role in controlling the seasonal variations of these trace elements. The workshop was divided into three task groups to analyze the self-assessments conducted at Krsko NPP, Slovenia; Dukovany NPP, Czech Republic; and Balakovo NPP, Russian Federation. In addition, they were asked to comment on the recommendations on the ASSET service made by the Consultants' Meeting of 26-30 August 1996 on ''Performance of the IAEA programme on Operational Safety Services''. Ground water is treated in the Czech Republic so that small water tanks are built above each water source to serve the primary ground water treatment; water so pretreated is then concentrated in large basins for subsequent treatment. Some water tanks where the first contact of the ground water with air takes place were selected as sites predisposed to radon accumulation. The examination was carried out near the town of Jihlava, where the bedrock contains slightly elevated radium concentrations. The average radon concentrations lay within the region of 2-4 kBq/m"3; the instantaneous values, however, exhibited appreciable periodical variations during the day. The relatively high radon concentrations will not pose a marked hazard for the personnel because the employees only reside at the sites in question for 10 to 15 minutes within 2 days, not for the whole working day

Pathway analyses are regarded by National Plant Protection Organizations as a very efficient way to address the risks posed by invasive alien species. Data on import of aquatic plants was obtained from 10 EPPO countries (Austria, Czech Republic, Estonia, France, Germany, Hungary, the Netherlands, Latvia, Switzerland and Turkey) and aggregated in order to consider whether invasive or potentially invasive alien plants could be introduced in the EPPO region through this pathway. This study highlights that this pathway mainly consists of the import of tropical plants for use in aquaria, and which do not represent a risk due to their climatic requirements. Mechanical properties of P-LDHA and V-ROL N steels of the Czech production are compared with other ultra-high strength steels produced in the world. The most important results can be summarized into following points: 1. Optimally heat treated P-LDHA steel exhibits substantially better strength characteristics then the V-ROL N steel. 2. Optimal tempering temperature of the P-LDHA steel is 300 C. 3. Basic mechanical properties of P-LDHA steel belong to the top of the UHS steels ranking and are well comparable with those of American 300 M steel. 4. In the low-cycle fatigue region, the P-LDHA steel exhibits slightly better fatigue resistance then the 300 M steel. On the other hand, the high-cycle fatigue resistance of 300 M steel is slightly better then that of the P-LDHA steel. (orig.)
Future climate change will likely influence the frequency and intensity of weather extremes. As such events are by definition rare, long records are required to understand their characteristics, drivers, and consequences on ecology and society. Herein we provide a unique perspective on regional-scale temperature extremes over the past millennium, using three tree-ring maximum latewood density (MXD) chronologies from higher elevations in the European Alps. We verify the tree-ring-based extremes using documentary evidences from Switzerland, the Czech Republic, and Central Europe that allowed the identification of 44 summer extremes over the 1550-2003 period. The introduction of live insects into human food is rare in developed countries. However, we report, for the first time, an emerging risk that exists from dried fruit in Central Europe. Recently, massive and frequent infestation of dried fruit imported from the Mediterranean region by the mite, Carpoglpyhus lactis L. (Acarina: Carpoglyphidae), has been found. In 180 samples taken from supermarkets, 13% were contaminated; the contamination levels ranged from 0 to 660 mites per g of dried fruit. Whether or not a current account deficit sustainable has important implications for policy. If the current account deficits of a nation is sustainable, then it implies that the government should have no incentive to default on its international debt. In this article we examine whether or not the current account deficits for the OECD countries can be characterized by a unit root process with regime switching. The econometric methodology allows us to distinguish periods that are associated with unsustainable outcomes from those in which the intertemporal national long-run budget constraint (LRBC) holds. Different technological procedures used for the chemical mining of uranium differ for one thing with a level of the deposit exploitation and for another with the expenditures. It is necessary to find out the highest acceptable value of the costs per unit of production - limit unit costs. This criterion serves both for selection of the minable parts of deposit, and for determination of the exploitation level of the deposit parts. The paper describes a calculation of the costs and their decomposition into the fixed and variable costs. An evaluation of the different variants of the deposit exploitation is based on the variable costs, expressed in the dependence on the parameters of technological procedure. These parameters are either primary, e.g. density of the wells or dosage of reagents, or secondary, e.g. the uranium production. In this study, the complete sequence of the mitochondrial DNA (mtDNA) of Atelura formicaria (Hexapoda: Zygentoma) is described. The molecule is 15.205 bp in length and it is the third complete mt genome sequenced from the Zygentoma. The genome organization conforms with the putative ancestral insect gene arrangement. All protein coding genes use standard initiation codons (methionine and isoleucine). The exception is nad4 that starts with GTG, a codon used for this purpose in other insect species. A peculiar strand skew bias is observed, given that the PCGs encoded on the J-strand contain more thymines than adenines and more cytosines than guanines. This trend in nucleotide composition has been observed also in the "firebrat" Thermobia domestica (Zygentoma, Lepismatidae), but differs from that of the majority of hexapod species, including Tricholepidion gertschi (Zygentoma, Lepidotrichidae), where adenines and cytosines outnumber thymines and guanines. A calorimetric microliter plate assay for ethanol amplified by aldehyde dehydrogenase (ALDH) was developed. In the assay ethanol from a sample took part in a chain-reaction catalyzed by alcohol dehydrogenase (ADH) and amplified by ALDH in the presence of NAD{sup +}, diaphorase, and p-ibdonitrotetrazolium-violet (INT-violet)(a precursor of red product). The resultant reaction gave a red color, the intensity of which was proportional to the amount of ethanol present. Using the technique, the content of activity from peanuts of differing maturity and curing stages were determined respectively. Data showed that immature peanuts had a higher level of ethanol and a lower ADH activity than mature peanuts, and that the level of ethanol and ADH activity decreased with the curing time. By increasing the price of crude oil and natural gas and the decline in their stock a possibility of more intense use of coal reserves as a feedstock not only for energy purposes but also as a feedstock source for other industries, mainly chemical and steel industry, is opening up. At present brown coal exploited in the Czech Republic is explicitly burned in the power sources of various outputs. Coal pyrolysis under the defined conditions may be an outstanding way of non-energetic brown coal processing. The target of our work was to determine the behaviour of various types of brown coal from the North Bohemian Basin under the pyrolysis up to the temperature 750{sup o}C. At the same time, qualitative parameters of the pyrolysis products in dependence on the features of the input brown coal were observed. This paper discussed issues related to the storage of electricity produced from renewable energy sources. Storage systems currently used by distribution systems were reviewed. The study showed that successful storage systems ensure high charging and discharging rates for shorter periods of time. Systems with high storage capacities must also be able to store energy for longer periods of time than traditional energy storage systems. Systems include pumped storage plants; electro-chemical accumulators; compressed air accumulators; and redox flow batteries. Hydrogen technologies for the storage of electric energy from renewable sources were reviewed. The paper also outlined energy storage and fuel cell research currently being conducted by researchers in the Czech Republic. The study concluded by stating that new energy storage technologies are required to ensure the reliability of electricity produced from renewable energy sources. 7 refs., 9 figs.

A reactor-neutrino experiment, Daya Bay, has been proposed to determine the least-known neutrino mixing angle theta_13 using electron antineutrinos produced at the Daya Bay nuclear power complex in China. Daya Bay is an international collaboration with institutions from China, the United States, the Czech Republic, Hong Kong, Russia, and Taiwan. The experiment will use eight identical detectors deployed at three different locations optimized for monitoring the antineutrino rates from the six reactors and for detecting any rate deficit and spectral distortion near the first oscillation maximum. The overburden of the under ground experimental halls, connected with tunnels, ranges from about 250 to 900 meters-water-equivalent so that the cosmogenic background is small compared to the number of observed antineutrino events. The role of the HCR system in the repair of prelethal lesions induced by UV light, #gamma# radiation and alkylating agents was studied in the Bacillus subtilis SPP1 phage, its heat sensitive mutants (N3, N73 nad ts_1) and corresponding infectious DNA. The survival of phages and their transfecting DNA after treatment with UV light is substantially higher in hcr"+ cells than in hcr cells, the differences being more striking in intact phages than in their transfecting DNA's. Repair inhibitors reduce survival in hcr"+ cells: caffeine lowers the survival of UV-irradiated phage SPP1 in exponentially growing hcr"+ cells but has no effect on its survival in competent hcr"+ cells; acriflavin and ethidium bromide decrease the survival of the UV-irradiated SPP1 phage in both exponentially growing and competent hcr"+ cells to the level of survival observed in hcr cells; moreover, ethidium bromide lowers the number of infective centres in hcr"+ cells of the UV-irradiated DNA of ...
Effect of helium and hydrogen production on radiation-hardening of F82H irradiated by dual or triple beam condition were investigated. The specimens used were four types of ferritic martensitic steels of F82H-std (Fe-8Cr-2W-0.2V-0.04Ta-0.1C) steels tempered at 750degC for 60 minutes, 20% cold worked F82H steel, F82H tempered for 10 minutes and non-tempered F82H steels. The irradiation was performed at 450degC to 50 dpa under simultaneous dual beams of 10.5 MeV Fe"3"+ and 1.05 MeV He"+ or triple beams of those and 380 keV H"+ ions. The ratios of He (appm)/dpa and H(appm)/dpa were 15 nad 15 (or 150) for dual and triple ion beams. The hardness of the irradiated specimens measured at room temperature using a micro indentation after the irradiations. The hardness in these F82H steels irradiated at 450degC to 18 dpa under triple beam irradiation was harder than that under dual beam irradiation. 1-Furan-2-yl-3-pyridin-2-yl-propenone (FPP-3) is an anti-inflammatory agent with a propenone moiety and chemically synthesized recently. In this study, we examined the chemopreventive effect of FPP-3 on 7,12-dimethylbenz[a]anthracene (DMBA)-induced genotoxicity in MCF-7 cells. FPP-3 reduced the formation of the DMBA-DNA adduct. DMBA-induced CYP1A1 and CYP1B1 gene expression and enzyme activity were inhibited by FPP-3. It inhibited DMBA-induced aryl hydrocarbon receptor (AhR) transactivation and DMBA-inducible nuclear localization of the AhR. Induction of detoxifying phase II genes by chemopreventive agents represents a coordinated protective response against oxidative stress and neoplastic effects of carcinogens. Metabolic profile is an important biological marker of neoplastic processes not only in the tumor itself but also in the host organism. The neuro-hormone melatonin has been implicated in the experiments as an oncostatic agent. Female Wistar:Han SPF rats (Velaz, Prague, Czech Republic) were irradiated continuously for 15 days using a daily gamma rays dose of 96 mGy. At the end of exposure one group of rats was administered 5 mg/kg b.w. of dimethylbenz/a/anthracene (DMBA) intragastrically. During the period of exposure to ionizing radiation a part of the animals was supplied with melatonin (M) at a concentration of 20 #mu#l/ml in drinking water. Selected parameters of lipid and carbohydrate metabolism and levels of selected hormones were determined 2, 30 and 100 days post-irradiation. Naturally-occurring wetlands perform such functions as flood control, pollution filtration, nutrient recycling, sediment accretion, groundwater recharge and water supply, erosion control, and plant and wildlife preservation. A large concentration of wetlands is located in Eastern Europe. A significant amount of Eastern European wetlands has been converted to agricultural use in the past, and remaining wetlands are subject to agricultural drainage. Drained wetlands are used as prime agriculture lands for a variety of food crops. Other agricultural uses of wetlands range from growing Phragmites australis (common reed) for thatch and livestock feed, to collecting peat for heating and cooking fuel.
