Total synthesis of cis-solamin A, a mono-tetrahydrofuran acetogenin isolated from Annona muricata

Total synthesis of cis-solamin A was accomplished without using protecting groups starting from (-)-muricatacin in 11 steps with an overall yield of 4.5%. The backbone of cis-solamin A was constructed by olefin cross-metathesis between the tetrahydrofuran moiety and g-lactone moiety. An enzymatic kinetic transesterification procedure was successfully applied to the synthesis of an optically pure g-lactone moiety.

1,3-dipolar cyclo addition of benzyl aside to alpha-beta-unsaturated five-membered lactones

The 1,3-dipolar cyclo addition reactions between benzyl azide and the alpha,beta-unsaturated lactones crotonolactone, alpha-methylenebutyrolactone and Beta-angelica lactone have been performed. The best yields of cycloadducts were obtained by working without solvent, at 60 degree centigree in a sealed tube under nitrogen atmosphere. (3aRS,6aSR)-1-Benzyl-3 a, 4,6,6a-tetrahydro-1H-furo[3,4-d]-1,2,3-triazol-4-one and 3-benzyl-7-oxa-1,2,3-triazaspiro[4.4] non-1-en-6-one have been isolated in 36% and 78% yield from the reactions of the two first lactones respectively. The second product is the first example of this heterocyclic system. New Lactone and Xanthone Derivatives Produced by a Mangrove Endophytic Fungus Phoma sp. SK3RW1M from the South China Sea

A new lactone, 1,8-dihydroxy-10-methoxy-3-methyldibenzo[b,e]oxepine-6,11-dione (1), and two new xanthones, 1-hydroxy-8-(hydroxymethyl)-6-methoxy-3-methyl-9H-xanthen-9-one (2) and 1-hydroxy-8-(hydroxymethyl)-3-methoxy-6-methyl-9H-xanthen-9-one (3), were isolated from a mangrove endophytic fungus Phoma sp. SK3RW1M collected from the South China Sea. This is the first report on xanthone derivatives isolated as secondary metabolites from Phoma species. Their structures were elucidated by spectroscopic methods, mainly 1D- and 2D-NMR techniques, and the structure of compound 2 was confirmed by X-ray crystallography. Cytotoxicity assays showed that compounds 1-3 were inactive against KB and KBv200 cells.

Control of human filarial infections currently depends on chemotherapeutic strategies predominantly directed at microfilariae. Doxycycline therapy in an extended daily dose regimen sterilizes and kills adult stages, but the utility of this drug for routine field use remains an issue of concern. No macrofilaricidal drugs with efficacy after one or two doses are available for use, delaying the achievement of the elimination or eradication of onchocerciasis and lymphatic filariasis. Moxidectin, a macrocyclic lactone, is currently in clinical trials for onchocerciasis. Methods were developed for the separation of radioactively labeled carbohydrate components of proteoglycans by isocratic ion-moderated partition HPLC. Neutral sugars were separated after hydrolysis in trifluoroacetic acid with baseline separation between glucose, xylose, galactose, fucose, and mannose. N-Acetylneuraminic acid, N-acetylated hexosamines, glucose, galactose, and xylitol were likewise well separated from each other under isocratic elution conditions. Glucuronic acid, iduronic acid, and their lactones were separated after hydrolysis in formic acid and sulfuric acid. Glucosamine, galactosamine, galactosaminitol, and glucosaminitol were separated by HPLC on a cation exchanger with neutral buffer after hydrolysis in hydrochloric acid. Easy access to 6-membered iminoalditols - important glycosidase inhibitors

Stereoselective methods for synthesising iminoalditols are still needed, as this class of compounds is known to contain strong glycosidase inhibitors. We have for some time investigated (1) the possibilities for using 2-amino-6-bromo-2,6-dideoxy-aldonolactones as starting materials for such compounds, since a substitution at C-6 by the 2-amino group should lead to the target molecules. We have found that treatment of mentioned 2-aminolacones with aqueous base gave 5- or 6-membered iminoalditols depending of the stereochemistry of the lactone, while under non aqueous conditions only formation of 6-membered iminoalditols were observed. The use of triethylamine in methanol thus gave methylesters of 6-membered iminouronic acids. Reduction of the ester group with sodium borohydride gave the target compounds. Confirmatory analysis method for zeranol, its metabolites and related mycotoxins in urine by liquid chromatography-negative ion electrospray tandem mass spectrometry

The determination of the banned anabolic substance zeranol and the metabolites taleranol and zearalanone in bovine urine is complicated by the occurrence of the structurally-related mycotoxin zearalenone and the corresponding {alpha}- and {beta}-zearalenol metabolites which possess similar estrogenic properties. A liquid chromatography-negative ion electrospray tandem mass spectrometric method is presented for the confirmatory analysis of all six resorcylic acid lactones ('zeranols') in urine samples using deuterium-labelled internal standards. The method was validated as a confirmatory method for bovine urine samples according to new draft EU guidelines and showed good precision and linearity, and CC{alpha} and CC{beta} values of 0.02-0.30 and <1.0 ng ml{sup -1}, respectively. Properties of dust particles sampled from windboxes of an iron ore sintering plant: Surface structures of unburned carbon

Aiming to understand the formation mechanism of dioxins in the iron ore sintering process, dust samples obtained from some windboxes of a commercial iron ore sintering plant have been characterized with a powder X-ray diffraction (XRD), by the transmission electron microscope (TEM) equipped with an electron energy loss spectroscopy (EELS), and by the temperature-programmed desorption (TPD) and temperature-programmed oxidation (TPO) techniques. The elemental and XRD analyses reveal that the content of the Cl present in the samples ranges from 0.075 mass%-dry to 5.1 mass%-dry and tends to be higher at smaller dust particles, and that some of the Cl exists as KCl with the average crystalline size between 40 nm and 50 nm. Dust samples also contain a significant amount of unburned carbon, and the smallest dust particles, < 500 {mu}m, show the highest C contents in many cases and consist partly of C, K, and Cl elements. Enrichment of trace cadmium by soybean protein for the analysis by atomic absorption method

A method for enrichment of the ppb level of cadmium in water by using the coagulation of soybean protein by adding acids or its complex-forming character with heavy metal ions was investigated. After adding fixed amounts of soybean milk and 2% sodium diethyldithiocarbamate(DDTC) aqueous solution and a suitable amount of delta-gluconic lactone (delta-GL) to a sample solution, the mixture was heated to boiling in order to coagulate the protein. The coagulum(soybean curd) was separated from the suspension by centrifugation and burned to ashes with a low temperature plasma asher. Then the cadmium enriched in it was determined by atomic absorption spectrometry. Culex mosquitoes introduce the pathogens responsible for filariasis, West Nile virus, St. Louis encephalitis, and other diseases into humans. Currently, traps baited with oviposition semiochemicals play an important role in detection efforts and could provide an environmentally friendly approach to controlling their populations. The odorant binding proteins (OBPs) in the female's antenna play a crucial, if yet imperfectly understood, role in sensing oviposition cues. Here, we report the X-ray crystallography and NMR 3D structures of OBP1 for Culex quinquefasciatus (CquiOBP1) bound to an oviposition pheromone (5R,6S)-6-acetoxy-5-hexadecanolide (MOP). Human senescence marker protein 30 (SMP30), which functions enzymatically as a lactonase, hydrolyzes various carbohydrate lactones. The penultimate step in vitamin-C biosynthesis is catalyzed by this enzyme in nonprimate mammals. It has also been implicated as an organophosphate hydrolase, with the ability to hydrolyze diisopropyl phosphofluoridate and other nerve agents. SMP30 was originally identified as an aging marker protein, whose expression decreased androgen independently in aging cells.
