Monitoring brine chemistry to determine the extent of potential adverse reactions has been simplified by the development of a field-brine test kit and a series of nomographs. Results of the kit analyses serve as input to the nomographs, which provide a graphic means of determining the scaling tendency (Saturation Index value) of a brine. Brines that do not tend to form scale may be corrosive. Saturation Index values were correlated with various processes using data from geopressured wells in the Gulf Coast area. Control of scale in surface equipment with chemical inhibitors has been successful. DNA methylation is an important event in epigenetic changes in cells, and a fundamental regulator of gene transcription. Bisulfite genomic sequencing is a powerful technique used in studies of DNA methylation. However, the established procedures often require relatively large amounts of DNA. In everyday practice, samples submitted for analysis might contain very small amounts of poor quality material, as is often the case with forensic stain samples. In this study, we assess a modified, more efficient method of bisulfite genomic sequencing. Genomic DNA extracted from 3-mm dried blood spots using QIAamp micro kit was treated with sodium bisulfite (using EpiTect kit). Due to a leakage from a 12-inch pressurized diesel steel pipe four years ago, an area of approximately 30,000 square meters was contaminated. A pilot study applying the technology of in-situ enhanced bioremediation was conducted. In the study, a field test kit and on-site monitoring equipment were applied for site characterization and treatment verification. Physically, the enhanced bioremediation study consisted of an air extraction and air supply system, and a nutrition supply network. Certain consistent sampling methodology was employed. The Seismic Risk Explorer, an extension for ArcGIS Desktop developed by cedim AG, is a tool for computing the seismic risk, i.e. damage on residential buildings and thereby caused monetary losses for earthquake scenarios. It is based on the research results of the CEDIM (Center for Disaster Management and Risk Reduction Technology) research center, located at the Karlsruhe Institute of Technology (KIT) and the GeoForschungsZentrum (GFZ) in Potsdam. With this software tool seismic scenarios can be simulated for a given magnitude, depths and location. The seismic intensity, the resulting mean damage ratio for residential buildings and the seismic risk, expressed as the monetary loss caused by an earthquake can be computed downscaled to communities, postcode areas or even single buildings. Interested end-users are especially insurance companies, but also public authorities which may use the tool for risk management issues.
A case (female, 39?years of) of thyroid-like nasopharyngeal low-grade papillary adenocarcinoma with a significant spindle cell component is presented. The tumor was located on the posterior nasal septum. The spindle cells displayed nuclear features very much similar to the epithelial component and the two cell types merged imperceptibly. Immunohistochemically, the neoplastic cells (including the spindle cell component) were strongly and diffusely positive for TTF-1, cytokeratins (AE1-3), cytokeratin 19 and vimentin. The iodinated oil lipiodol is commonly used as a carrier for in situ delivery of drugs or radioactivity to hepatic tumors. Recently, we reported a new kit formulation for high-activity labeling of lipiodol with the {beta}-emitting radionuclide Re-188. Since the whole preparation involves different steps and complex manipulations of high-activity samples, we describe here an automated synthesis module that allows the easy preparation of sterile and pyrogen-free samples of Re-188 lipiodol ready to be administered to the patient. Important advantages include the possibility to incorporate high Re-188 activity into the lipiodol hydrophobic phase and a sharp reduction of radiation exposure of the operator assisting the labelling procedure. Application of this modular reaction system could be also extended to the preparation of other Re-188 radiopharmaceuticals and to compound labelled with different {beta}-emitting therapeutic radionuclides.
PCR detection of genetic material of the parasites present in faeces may be an alternative for microscopic and serological tests routinely used for diagnosing parasitic enteral infections. Current work presents the results of more than 100 measurements of 222Rn activity in drinking water collected at artesian bores at Curitiba region during the period of 2008 - 2009. The measurements were performed at the Laboratory of Applied Nuclear Physics of the Federal University of Technology in cooperation with the Nuclear Technology Development Center (CDTN) of Brazilian Nuclear Energy Committee (CNEN). Experimental setup was based on the Professional Radon Monitor (ALPHA GUARD) connected to specific kit of glass vessels Aqua KIT through the air pump. Objective: To compare the efficiency of dual energy subtraction and conventional direct digital radiography in signal detecting ability. Methods: 200 plastic balls which were 4 mm in diameter were taken as signals and were placed on a 10 cm thick plastic board and the right upper thorax of a healthy volunteer. The signals were examined by DES and CDR separately and were estimated by 3 radiologists with 5-value-differentiation method. ROC curve analysis was made by ROC kit software. Results: when signals on plastic board were detected by CDR and DES, ROC curve analysis indicated that there was no significant difference (P=0.4851 > 0.05, two-tailed) between the area under the ROC curves of the two methods (Az=0.9931 for CDR and Az=0.9879 for DES). The role of intracellular Ca2+ (Ca2+i) on hematopoiesis was investigated in long term bone marrow cultures using cytokines and agonists of P2 receptors. Cytokines interleukin 3 and granulocyte/macrophage colony stimulator factor promoted a modest increase in Ca2+i concentration ([Ca2+]i) with activation of phospholipase Cgamma, MEK1/2, and Ca2+/calmodulin kinase II. Involvement of protein kinase C was restricted to stimulation with interleukin 3. In addition, these cytokines promoted proliferation (20 times) and an increase in the Gr-1(-)Mac-1+ population with participation of gap junctions (GJ). Twenty-six unclassified Campylobacter-like strains previously isolated from 15 chicken carcasses and caecal contents, together with two more strains isolated from chicken faeces on a different occasion, were identified as Helicobacter pullorum using various phenotypic identification methods. API Campy identification kits and a 16-test identification scheme developed for campylobacters failed to identify these bacteria, or identified them as Campylobacter spp. Eighteen strains (including the two isolated on a different occasion) were chosen for examination using a more comprehensive probabilistic identification scheme. In mice, coat pigmentation requires a stem cell (SC) system in which the survival, proliferation, and differentiation of melanocytes (MCs) are regulated by microenvironments in hair follicles (HFs). In vitro systems are required to analyze the behavior of single melanocyte stem cells (MCSCs) and their potential to form SC systems in vivo. We describe here an experimental system for the isolation, self-renewal, and differentiation of MCSCs, as well as an in vivo reconstitution assay for assessing their potential. This study presents the data of an evaluation of the automated Nuclisens easyMAG and EasyQ systems versus the Roche AmpliPrep-AMPLICOR combination for testing of high-volume human immunodeficiency virus (HIV) load. This represents a follow-up of a previous study investigating the performance of the real-time Nuclisens assay using the semiautomated NucliSENS miniMAG extraction procedure. Three hundred eighteen patient samples were analyzed using both methods. The easyMAG-EasyQ HIV type 1 system has a higher sensitivity and broader dynamic range than the Cobas AmpliPrep-AMPLICOR system when the standard Roche assay is used alone, 25 to 3,000,000 IU/ml versus 400 to 750,000 HIV RNA copies/ml, respectively. From January 2008 to December 2009, 433 Streptococcus pneumoniae strains were examined to determine the serotype distribution and susceptibility to selected antibiotics. About 50% of them were invasive isolates. The strains were isolated from patients of all age groups and 33.55% were isolated from children below 5 years. The majority was isolated from blood (48.53%) and other sterile specimens (6.30%). Community acquired pneumonia (41.70%) is the most common diagnosis followed by sepsis (9.54%). However, the application of these materials for the production of complex shaped or relatively large pieces is limited, because of the difficulty and cost of making the necessary tool kit for the job. Full text: Immunoassays for prostate-specific antigen (PSA) are used to detect early-stage prostate cancer, monitor disease progress, and evaluate therapeutic response. At least two forms of PSA, free PSA (F-PSA) and PSA complexed to alpha-1 anti-chymotrypsin (PSA-ACT) are detected by commercial PSA assays. The fraction of F-PSA is shown to be smaller in patients with untreated prostate cancer than in patients with benign prostate hyperplasia (BPH). Thus, combined measurements of both total and free PSA are used for a better discrimination between BPH and prostate cancer. Detection of PSA for screening of prostate cancer has been a subject of debate for many years. The reason of this debate is mainly because screening for prostate cancer is not cost-effective, as was shown by studies undertaken in Europe and United States.
