In three calves experimentally infected with Trypanosoma congolense the amounts of IgG1 and IgG2 were little changed and similar to those of normal animals. IgM increased in amount early in the infection and the amount of the increase appeared related to the parasite burden. The amounts of IgA and IgE were both much decreased and this also appeared related to the numbers of parasites in the blood. There was a decrease in the amounts of total haemolytic complement and complement components C1, C1q and C3 in the infected calves. Furthermore the amounts of properdin fluctuated with the cyclical changes in numbers of T. congolense parasites in the individual calves. No significant change in the amount of C8 was observed. It is considered that activation of both the alternative and the classical complement pathways occurs in trypanosome infected animals but that neither pathway goes to its terminal stages.

Background: Recent studies have developed a murine model of IgE-mediated food allergy based on oral coadministration of antigen and cholera toxin (CT) to establish a maximal response for studying immunopathogenic mechanisms and immunotherapeutic strategies. However, for studying subtle immunomodulating factors or factors effective during response initiation, this maximal response-based model is less suitable due to a lack of sensitivity. Therefore, in attempts to identify essential parameters to fine-tune the immune response towards a submaximal level, potentially more sensitive, we were interested in characterizing the individual effects of the parameters in the CT-based model: CT dose, antigen type and dose, and number of immunizations. Methods: BALB/c mice were orally sensitized weekly for 3 or 7 weeks with graded doses of CT and various food antigens (soy-trypsin inhibitor, ovalbumin or ovomucoid). Antigen-specific lgG1, IgG2a, IgA and ...

We evaluated the use of a novel gene porter (Den123-a nontoxic self-assembled dendritic spheroidal nanoparticle made of biodegradable monomers), aiming to enhance and improve the desired immune response in protection from allergy. Footpad DNA immunization in Balb/c mice was done three times using the Bet v 1a gene with or without Den123 with 2-week intervals followed by sensitization with rBetv1 (5mg) in alum twice in a weekly interval. Different doses of pCMV-Betv1 were used (10mg and 100mg). The protective role of different formulations was evaluated by measuring the IgG1, IgG2a and IgE antibody production, cytokine release of isolated splenocytes and b-hexosaminidase release from the RBL cells. Higher and increasing ratios of IgG2a/IgG1 were seen in mice which received plasmids in combi...

Summary The presence of human heterophilic antibodies against horse immunoglobulins (HHA-HI) was determined by ELISA in sera from healthy volunteers and from patients who received equine antivenom for therapy of snake bite envenoming. These patients were selected from two independent clinical studies: one in Colombia in which patients received antivenom constituted by whole IgG (n=25); and the other in Brazil where an antivenom constituted by F(abprime)2 fragments was administered (n=31). Results show that healthy volunteers have antibodies, mainly of the IgG class, able to react with whole equine IgG. Additionally, patients have IgG antibodies that react both with whole equine IgG and F(abprime)2 fragments. In both clinical studies, no significant differences were observed in the HHA-HI t...

Repassivation behavior and IGA resistance of nickel base alloys containing 0#approx#30 wt% chromium was investigated in high temperature acid sulfate solution. (1) The repassivation rate was increased with increasing chromium content. And so the amounts of charge caused by the metal dissolution were decreased with increasing chromium content. (2) Mill-annealed Alloy 600 suffered IGA at low pH environment below about 3.5 at the fixed potentials above the corrosion potential in 10%Na_2SO_4+H_2SO_4 solution at 598K. On the other hand, thermally-treated Alloy 690 was hard to occur IGA at low pH environments which mill-annealed Alloy 600 occurred IGA. (3) It was considered that the reason, why nickel base alloys containing high chromium content such as Alloy 690 (60%Ni-30%Cr-10%Fe) had high IGA/SCC resistance in high temperature acidic solution containing sulfate ion, is due to both the ...
Summary Background The incidence of food allergy to wheat is increasing. Its diagnosis depends on the purity of major allergens and their inclusion in tests. Isolation and characterization of wheat allergens are therefore of utmost importance. Objective To purify and identify wheat flour allergens most frequently recognized by patients' IgE antibodies and to study their allergenicity. Methods Water/salt-soluble extracts from wheat flour were prepared and separated using a combination of ultrafiltration, isoelectric focusing and liquid chromatography. Purified proteins were analysed by immunoblotting using pooled sera from patients with atopic dermatitis who possessed IgE specific to wheat. Wheat proteins found to bind IgE were subsequently identified by matrix-assisted laser desorption/ion...

Faulting the feedwater for a 19-tube, model steam generator with 10 ppM of caustic once a week produced widespread shallow 25 to 75 micrometers (1 to 3 mils) intergranular attack (IGA) on alloy 600 tubes and an axial tube rupture at the steam-water interface after 4.8 years. An extensive investigation of the IGA damage found little correlation with major test variables beyond the indication that mill-annealed tubing was more susceptible to attack than stress-relieved tubing. The most likely cause of the tube rupture was caustic that concentrated to high levels in a porous scale on the tube at the liquid-vapor interface where there was a high available superheat. Nondestructive examination (NDE) eddy current probes underestimated the depth of IGA and were not sensitive to circumferential cracks less than 152 micrometers (6 mils) deep that were above and below the roll transition zones of tubes in the tubesheet.
Urticaria is one of the most common dermatoses during the childhood, but it is very rare in the neonates. A 17-day-old infant with a generalized urticaria was admitted to our pediatric emergency unit. The mother of the infant reported having applied water boiled with stinging nettle (Urtica dioica) onto her nipples twice a day (before and after each breastfeeding) for 2?days in order to heal her nipple cracks. Serum total immunoglobulin E (IgE) and specific IgE levels for stinging nettle were high in the infant and the mother. The rashes began to regress within the first day of the hospitalization and disappeared completely on the second day without treatment. The skin prick test with the water boiled with stinging nettle was positive for the infant with significant induration, but not for...

Enzyme-linked Immunosorbent assay systems for the identification of irradiated egg, pork and chicken was developed. Eggs were irradiated in their shells to 0.5{approx}7kGy. Pork was irradiated to 0.5{approx}3kGy and chicken irradiated to 0.5kGy{approx}5kGy. The most sensitive proteins to irradiation were screened by SDS-PAGE and purified. Ovalbumin from egg, salt soluble protein(p) from pork, and salt soluble protein(c) from chicken showed the most sensitivity to irradiation. To investigate for a practical use in identifying of irradiated egg, pork and chicken, competitive ELISA was performed. The binding activity of ovalbumin to anti-ovalbumin IgG was reduced in a dose-dependent manner by irradiating up to 7kGy, and considerably lowered after irradiating at 7kGy. The concentration of 50% inhibition of ovalbumin to IgG was increased to 1.5(0.5kGy){approx}3.7(7kGy) times in an dose-dependent relationship. The binding activity of salt soluble ...

PWR steam generators have suffered from a variety of degradation phenomena. This paper identifies the corrosion-related defects and their probable causes and suggests approaches to correct and prevent corrosive activity. In the attempt to solve the degradation problems, research programs have concentrated on modifying materials, stresses, and the chemical environment in both new and operating steam generators. The following corrosion-related defects have been studied: tube wastage, denting, primary side (ID) intergranular stress corrosion cracking (IGSCC), OD-initiated intergranular attack (IGA), pitting, and corrosion fatigue. Plants affected by wastage have greatly reduced their problem by adopting an all volatile treatment (AVT). In the case of denting, a less aggressive chemical environment is recommended. The detection, in European steam generators, of intergranular stress corrosion cracking initiating from the primary side (PWSCC) and the somewhat similar detection of intergranular stress corrosion (IGSCC) and intergranular corrosion (IGA) initiated from the secondary side in Japanese steam generators has led to a growing awareness of the potential for such corrosion forms in United States PWR steam generators. What had been a minor occurrence at several units is now a cause of concern and repair measures at more than 30 sites in the United States, and in some cases, is the major cause of steam generator unavailability. The United States nuclear utilities and EPRI formed the first Steam Generator Owners' Group (SGOG) in 1977 in response to the prospect of continued denting corrosion. The occasional appearance of PWSCC, IGSCC and IGA in domestic steam generators was not a sufficient cause for initiating an extensive research program in SGOG I. ...
Since 1991, secondary side cracking has become a major concern in EDF steam generators. At 350 C, IGSCC of Alloy 600 was found in pure NaOH solutions from 4 to 500 g/l, with a maximum at 100 g/l. From the present results and available data, IGSCC of Alloy 690 occurred in pure NaOH solutions from 40 to 500 g/l, but the greatest sensitivity was observed at 100 g/l. TG/IGSCC of Alloy 800 became more severe with increasing NaOH solutions, from 10 to 500 g/l. On the whole, the ranking for SCC-resistance was Alloy 600 < Alloy 800 < Alloy 690. A significant improvement was still obtained with 690TT with intergranular carbides (industrial tubes) and perhaps with Alloy 800 NG (nuclear grade), but both alloys cracked at the highest stresses. IGSCC susceptibility of Alloy 600 MA was higher for the tubes with the lowest values of the yield stress. The depth of IGA in the case of Alloy 600 was higher than for Alloy 690; for this alloy, it appeared to be general oxidation. ...
Current bioartificial extra-corporeal systems are bioreactors where cells are separated from the surrounding media by porous polymeric membranes. The present work focuses on the design of membranes that allow the differential diffusion of plasma metabolites and proteins such as immunoglobulin (IgG). This design will improve catabolites removal and reduce possible immune response and virus infection. We demonstrate the feasibility to synthesize the hydrogels confined to the macroporous structure of membranes by radiation-induced in situ polymerization. The hollow-fiber membranes were soaked in aqueous monomeric solution, rinsed and irradiated while submerged in oil. This procedure confined the hydrogel to the void internal volume of the pores of the membrane. Hydrogels of polyacrylamide and polyHEMA were synthesized this way by irradiation at 10 kGy. Hydraulic permeability and diffusion of glucose, albumin and IgG were measured in these ...
Functionalization of carbon nanotubes is very challenging for their applications. The paper here describes a new method to functionalize multi-walled carbon nanotubes (MWCNTs) as specific affinity adsorbents. MWCNTs were acid purified and pretreated with (3-aminopropyl)-triethoxysilane (APTES) in order to introduce abundant amino groups on the surface of MWCNTs. After the conversion of amino groups to carboxyl groups by succinic acid anhydride, MWCNTs were attached to protein A or aminodextran using 1-ethyl-3,3' (dimethylamion)-propylcarbodiimide as a biofunctional crosslinker. The incorporation of aminodextran as a spacer arm noticeably increased the binding capacity of the APTES-modified MWCNTs for protein A. The application of affinity MWCNTs for purification of immunoglobulin G was then evaluated. The affinity of MWCNTs with AMD spacer exhibited a high adsorption capacity of {approx}361 {mu}g IgG/mg MWCNT (wet basis). About 75% of bound IgG was eluted ...
Building on the concepts of MIRD methods described by Webber, Watson and others at this Symposium on the Dosimetry of Administered Radionuclides, we will now consider how to apply these principles to several experimentally based open-quote macroclose quotes and multicellular models. Specifically, methods to calculate absorbed dose using different types of particulate emission (alpha, beta) with a variety of antibody carriers (IgG, F(ab')_2, Fab) for radioimmunotherapy (RIT) will be reviewed. Additionally, a discussion of direct measurement methods at the multicellular level will reveal the problems of tumor absorbed dose heterogeneity when applied to animal and clinical studies.
The long-term survival of serologically incompatible red cell units was measured in five patients with antibodies to high-frequency antigens. Initially, the survival of 1 ml of "5"1Cr-labeled incompatible red cells was measured over 1 hour. After demonstrating that the 1-hour survival times were successful (greater than 70%), each patient then received 5 ml of the same "5"1Cr-labeled red cells followed by the transfusion of the remainder of the red cell unit. The long-term T 1/2Cr survival for each case was patient 1 (anti-McCa), 15 days; patient 2 (anti-JMH), 12 days; patient 3 (anti-Kna), 31 days; patient 4 (anti-McCa), 12 days; and patient 5 (anti-Hya), 14 days. Each antibody tested in an in vitro homologous macrophage assay showed less than 5 percent phagocytosis. Anti-JMH was the only antibody to react with IgG subclass antisera and was determined to be IgG4. The macrophage assay, IgG subclass testing, and short-term ...
The authors have cloned and determined the entire nucleotide sequence of cDNAs corresponding to the putative {alpha} subunits of the human and rat mast cell high-affinity IgE receptors. Both human and rat cDNAs encode an NH{sub 2}-terminal signal peptide, two immunoglobulin-like extracellular domains (encoded by discrete exons), a hydrophobic transmembrane region, and a positively charged cytoplasmic tail. The human and rat {alpha} subunits share an overall homology with one another and the immunoglobulin gene family, suggesting that they arose from a common ancestral gene and continue to share structural homology with their ligands. In addition, the rat gene is transcribed into at least three distinct forms, each of which yields a somewhat different coding sequence.
The effects of dietary plant and yeast cerebroside (glucosylceramide), a major sphingolipid in plants and yeast, on atopic dermatitis (AD) like symptoms were investigated in a mouse model. After 7 wk of feeding with a diet containing maize glucosylceramide, plasma IgE levels became significantly lower and in contrast, the levels of interleukin (IL)-12, which induces cellular immunity, became significantly higher in the AD mice than in the controls. However, the sphingolipid constituents of the skin fraction in the maize glucosylceramide fed group did not contain sphingoid bases of plant origin, such as 8-unsaturated sphingoid bases. The results of the present study indicated that dietary plant glucosylceramide prevented AD-like symptoms in AD model mice via regulation of Th1/Th2 balance. P...
The steam generator availability is one of the important problems encountered during the pressurized water nuclear plant operation. Various kinds of corrosion phenomena were observed in the past. These phenomena result from the concentration of impurities mainly in three locations in the steam generators: the tubesheet crevices, the tube support plate crevices, and the sludge pile. Corrections were made in the design and the materials used but a number of steam generators suffer or will suffer from corrosion processes inducing in many cases forcing their replacement. In order to prevent or to retard the corrosions several laboratories have performed experiments to reproduce and to study the corrosion processes. The first step of the degradation is the concentration of chemical species. A method using /sup 24/Na as a radioactive tracer was used to establish the concentration kinetics of caustic which was identified as the major cause of the intergranular attack ...
Two plasmids which express either nearly intact or truncated filamentous hemagglutinin (FHA) from Bordetella pertussis and which are marked with a tetracycline resistance (Tcr) gene were transformed into Salmonella dublin SL1438, an aroA deletion mutant intended for use as an attenuated oral vaccine against salmonellosis. These S. dublin recombinants, when fed to mice, induced serum immunoglobulin, immunoglobulin M (IgM), and sometimes IgA antibody responses to FHA and S. dublin. In addition, IgA antibodies against FHA were found in gut wash fluids. S. dublin carrying pDB2300, a multicopy plasmid encoding truncated FHA protein, induced a better antibody response than did S. dublin carrying pDB2000, a low-copy-number plasmid encoding full-sized FHA. Administration of tetracycline to mice enhanced the stability of recombinant plasmids, and tetracycline-treated mice developed higher anti-FHA titers. Although neither strain examined is suitable for ...
The significant and rapid reduction of greenhouse gas emissions is recognized as necessary to mitigate the potential climate effects from global warming. The postcombustion capture (PCC) and storage of carbon dioxide (CO{sub 2}) produced from the use of fossil fuels for electricity generation is a key technology needed to achieve these reductions. The most mature technology for CO{sub 2} capture is reversible chemical absorption into an aqueous amine solution. In this study the results from measurements of the CO{sub 2} absorption capacity of aqueous amine solutions for 76 different amines are presented. Measurements were made using both a novel isothermal gravimetric analysis (IGA) method and a traditional absorption apparatus. Seven amines, consisting of one primary, three secondary, and three tertiary amines, were identified as exhibiting outstanding absorption capacities. Most have a number of structural features in common including steric hindrance and ...
The human articular cartilagineous proteoglycans (PG) R.I.A. is highly specific. The PG used as the standard and the /sup 125/I labelled molecule appear to be pure. Under these conditions, all the potential interfering substances which have been tested show no cross reaction. For instance, the Ag-Ab equilibrium is not affected by adding human IgG, human albumin, hyaluronic acid, chondroitin sulfate, rat type II collagen or total human serum proteins. This R.I.A. also exhibits a species spcificity since there is no cross reaction with rat PG and negligible cross section with dog PG. The results obtained after addition of enzymes to the antigen demonstrate that the antigenic sites are localized on the protein region and not on the glycosaminoglycan region of the molecule.

Phage display is effective in screening peptides that mimic venom's neutralizing epitopes. A phage display cyclized heptapeptide library (C7C library) was panned with purified divalent antivenin IgG, which neutralizes Naja naja atra venom (NAV) and Bungarus multicinctus venom (BMV). The selected heptapeptide sequences were aligned with known protein sequences of NAV and BMV in GenBank. One of the four consensus sequences, L/PKSSLL, mimicked the crucial epitope on Loop III of Taiwan cobra cardiotoxin that is associated with the venom's lethal potency. In dot blot analysis, several clones showed varying reactivities for NAV monovalent antivenin and lesser cross-reactions with BMV monovalent antivenin. The KSSLLRN-carrying phage occurred four times in selected clones and showed the strongest ...
PWR plants have made efforts to maintain the long-term integrity of the steam generators (SG) by reducing the amount of corrosion products entering the secondary side of the SG. Iron entered the SG can cause several problems: degraded heat conductivity of the SG tubes in locations where iron is deposited, water level oscillations in the SG due to tube support plate hole blockage, and initiation and propagation of inter-granular attacks (IGA) and stress corrosion cracking (SCC). One of the most effective measures, high all-volatile treatment (AVT) chemistry has been applied to actual plants to reduce the flow-accelerated corrosion (FAC) coming from the carbon steel piping. The secondary water chemistry at Genkai NPS 1 and 2 changed, from the Low AVT chemistry to the High AVT chemistry, in November 2006. In this paper, we will describe the results of experiments in applying the use of High pH water in the secondary water system at Genkai NPS. (author)
Computer programs have been written to allow the analysis of different types of eddy-current probes and their performance under different steam generator test conditions. The probe types include the differential bobbin probe, the absolute bobbin probe, the pancake probe and the reflection probe. The generator test conditions include tube supports, copper deposits, magnetite deposits, denting, wastage, pitting, cracking and IGA. These studies are based mostly on computed values, with the limited number of test specimens available used to verify the computed results. The instrument readings were computed for a complete matrix of the different test conditions, and then the test conditions determined as a function of the readings by a least-squares technique. A comparison was made of the errors in fit and instrument drift for the different probe types. The computations of the change in instrument reading due to the defects have led to an inversion'' ...
Isocapnic hyperventilation with cold air (IHCA) is a reliable technique for assessing indirect bronchial hyperresponsiveness in patients with asthma. Impedance measurement of the respiratory system by the forced pseudorandom noise oscillation technique is a sensitive technique to assess changes in bronchial tone after IHCA. The aim of this study was to evaluate the effect of 6 weeks of treatment with beclomethasone dipropionate, 1,000 microg x day-1, on IHCA in asthmatic patients, measured with both forced oscillation technique and flow-volume recordings. Forty patients with mild asthma were included in this double-blind, placebo-controlled parallel-group study. Stratification on the basis of sex was performed to overcome differences in airway diameter. At entry and every 2 weeks during the treatment period, IHCA was performed and patient diaries were evaluated. Characteristic changes in forced oscillation parameters after IHCA were observed in all patients. After 6 weeks of treatment, ...
During the Bruce A Nuclear Generating Station (BNGS-A) Unit 1 1997 planned outage, a dew point search method identified a leak in one steam generator(SG) tube. Subsequently, the tube was inspected with all available eddy current probes and removed for examination. The initial inspection results and metallurgical examination of the removed tube confirmed that the leak was due to intergranular attack/stress corrosion cracking (IGA/SCC) emanating from the secondary side of the tube at the top of the tubesheet location. Subsequently, eddy current and ultrasonic indications were found at the top of the tubesheet of other Alloy 600 SG tubes. To investigate the source of the indications and to validate the inspection probes, sections of 40 tubes with various levels of damage were removed. The metallurgical examination of the removed sections showed that both secondary side and primary side initiated, circumferential, stress corrosion cracking and intergranular attack ...
Steam generators (SGs) in PWRs and CANDUs are designed for at least a 30-year operating life. However, in the 15-25 years that SGs tubed with Alloy 600 have operated commercially, they have experienced reduced reliability, mainly due to SG tubing degradation. One of the degradation mechanisms that Alloy 600 SG tubing has suffered from is lead-induced stress corrosion cracking (PbSCC) in AVT and near-neutral SG environments. In contrast to Alloy 600 tubing, test data obtained in high temperature water indicate that Alloy 800 is resistant to cracking by lead and lead compounds. For Alloy 800 (cold worked, shot peened), the most aggressive environment is reported to be alkaline with minor concentration of PbO (80 ppm). Work by Max Helie et al. also concluded that Alloy 800 is not sensitive to lead assisted SCC for pH values close to neutrality, whereas it could be affected in high alkaline conditions. The work reported here investigated the influence of lead (Pb), deformation, temperature ...

Since 1975 All Volatile Treatment (AVT) has been the preferred method for controlling the secondary system operating environment in Westinghouse PWR plants. However, since AVT provides no buffering action against corrodent species present in the water as trace contaminants, utilities have initiated programs which combine control of contaminant ingress with total steam generator sludge management. The earliest applications of boric acid, to control denting type corrosion, began in 1978 and have continued up to the present time. Boric Acid has also been added as an inhibitor for SCC/IGA type corrosion since 1985. There are now approximately 30 plants operating with boric acid or boric acid/morpholine chemistry in the secondary system, and a growing number of plants where morpholine is being used as an additive to maintain pH and reduce iron transport attributable to erosion/corrosion. The impact of these modifications in the operating chemistry environment are ...
To overcome the limitations of conventional bobbin style eddy-current probes, it has become common practice to perform some portion of a steam generator tube inspection program with array coil probes. Typically, eight surface-riding pancake coils are utilized in a probe head, either as eight independent coils (8 x 1) or in a parallel combination of four serially connected coils (4 x 4). The advantages of the array coils are improved sensitivity to intergranular attack (IGA), reduced interference from tube denting, and improved sensitivity to circumferential cracking. The current status of commercially available probes is such that several weaknesses in array coils have limited the widespread application of this technology. Task S 301-6 was conceived to address each of the weaknesses in order to produce a more reliable and cost-effective probe. The emphasis was on reliability, since the cost associated with downtime for a probe change far exceeds the price of a ...
Focused ion beam (FIB) microscopes have become well-established in the semiconductor industry during the past decade, and are rapidly gaining attention in the field of materials science, both as a tool for producing site specific, parallel sided transmission electron microscope (TEM) specimens and as stand alone specimen preparation and imaging systems. FIB secondary electron imaging (SEI) of nickel-based alloys, such as commercially produced Alloy 600 (approximately Ni 15Cr 10Fe 0.5C), has been demonstrated to show a high degree of sensitivity to the presence of deformation in the alloy, and FIB secondary ion imaging (SII) is particularly useful for identifying the presence of grain boundary corrosion, as secondary ion yields from metallic specimens can increase by three orders of magnitude in the presence of oxygen. This 'oxygen enhanced yield', makes FIB SII ideal for detection of corrosion at grain boundaries down to thicknesses of only a few tens of nanometers. Historically, while ...
These studies were undertaken to assess the effects of increased galactose (v increased glucose) metabolism via the polyol pathway on vascular filtration function in the kidneys, eyes, nerves, and aorta. Quantitative radiolabeled tracer techniques were used to assess glomerular filtration rate (GFR) and regional tissue vascular clearance of plasma 131I-bovine serum albumin (BSA) in five groups of male Sprague-Dawley rats: nondiabetic controls, streptozotocin-diabetic rats, nondiabetic rats fed a 50% galactose diet, diabetic rats treated with sorbinil (an aldose reductase inhibitor), and galactose-fed rats treated with sorbinil. Sorbinil was added to the diet to provide a daily dose of approximately .2 mmol/kg body weight. After 2 months of diabetes or galactose ingestion, albumin clearance was increased twofold to fourfold in the eye (anterior uvea, choroid, and retina), sciatic nerve, aorta, and kidney; GFR was increased approximately twofold and urinary excretion of endogenous ...
Assessment of the immune system's capability to respond to antigens with the generation of specific antibodies, whilst under the influence of a test article, is required in toxicity tests according to the European guideline for repeated dose toxicity testing of medicinal products. The purpose of this study in rats was to validate methodology for the determination of Keyhole Limpet Haemocyanin (KLH)-specific antibodies under the influence of an immunologically active compound. The immunosuppressant FK506, commercially available as Prograf, was administered orally (gavage) to five rats per sex per group at dosages of 0.5mg/kg per day or 3mg/kg per day, for a period of 4 weeks. On days 14 and 22, KLH was administered subcutaneously, with an adjuvant (AluGel), to the two treated groups and a control (i.e. without FK506 treatment) approximately 1h following administration of FK506. Terminal investigations included haematology parameters, titration of KLH-specific antibodies in serum ...
The influence of reconstituting a murine monoclonal IgG_1 antibody kit with pertechnetate Tc99m on antibody distribution in the liver, spleen and sternal bone marrow of patients was examined. The "9"9"mTc-labelled antibody used is directed against non-specific cross-reacting antigen (NCA-95) and carcinoembryonic antigen (CEA) and has been successfully applied for imaging tissue inflammation and bone marrow scanning. Radioactivity uptake was determined in the liver, spleen, bone marrow and a precordial background region in a consecutive series of 25 patients, examined with an antibody preparation, routinely radiolabelled according to the manufacturer's recommendations and in 14 patients, in whom the antibody was reconstituted with special care, avoiding bubble formation and dropping of buffer into the antibody-containing vial. Gentle compared with routine antibody reconstitution caused a highly significant reduction of the antibody uptake in the liver, as determined ...
The spike (S) protein of severe acute respiratory syndrome coronavirus (SARS-CoV) has two major functions: interacting with the receptor to mediate virus entry and inducing protective immunity. Coincidently, the receptor-binding domain (RBD, residues 318-510) of SAR-CoV S protein is a major antigenic site to induce neutralizing antibodies. Here, we used RBD-Fc, a fusion protein containing the RBD and human IgG1 Fc, as a model in the studies and found that a single amino acid substitution in the RBD (R441A) could abolish the immunogenicity of RBD to induce neutralizing antibodies in immunized mice and rabbits. With a panel of anti-RBD mAbs as probes, we observed that R441A substitution was able to disrupt the majority of neutralizing epitopes in the RBD, suggesting that this residue is critical for the antigenic structure responsible for inducing protective immune responses. We also demonstrated that the RBD-Fc bearing R441A mutation could not bind to soluble and ...
Thirty-seven patients with primary nonresectable intrahepatic cholangiocarcinoma (57% with prior treatment and/or metastasis) were prospectively treated with external radiation, chemotherapy, and "1"3"1I labelled anti-CEA. Therapy began in all trials with whole liver irradiation (21.0 Gy, 3.0 Gy/Fx, 4 days/week, 10 MV photons) with alternate treatment day chemotherapy (Adriamycin, 15 mg + 5-FU, 500 mg). One month after external beam therapy, chemotherapy was given (Adriamycin, 15 mg + 5-FU, 500 mg) followed the next day by the first administration of "1"3"1I anti-CEA. The treatment schedule used was 20 mCi day 0; 10 mCi day 5 as an outpatient. This schedule was derived from tumor dose estimates which indicated that 20 mCi (8-10 mCi/mg IgG) was sufficient to achieve tumor saturation with a tumor effective half-life of 3 to 5 days, depending upon the species of animal from which the antibody was obtained. The median tumor dose for the 20 mCi + 10 mCi regimen was 6.2 ...
