The herpes simplex virus type 1 thymidine kinase (HSV1-TK) reporter system is being used to directly and indirectly monitor therapeutic gene expression, immune cell trafficking and protein-protein interactions in various living animals. However, the issues of HSV1-TK enzyme stability in living cells and whether this reporter system is optimal for dynamic studies of gene expression events in genetic imaging have not be addressed. The purpose of the present study was to evaluate the application of this reporter system in dynamic studies of transcriptional gene regulation. We have observed significant differences in the response to ionizing radiation of two closely related human cell lines, and now compare the effects on these lines of both low and intermediate LET radiation. Compared to TK6, WTK1 has an enhanced X-ray survival, and is also more resistant to cell killing by {alpha}-particles. The hprt locus is more mutable in WTK1 than in TK6 by both X-rays and {alpha}-particles. WTK1 is also more mutable by {alpha}-particles than by X-rays at the hprt locus. X-ray-induced mutation at the heterozygous tk locus in WTK1 is about 25 fold higher than in TK6, while {alpha}-particle-induced mutation is nearly 50 fold higher at this locus. Also, the slowly growing tk- mutants, which comprise the majority of spontaneous and X-ray-induced tk- mutants of TK6, were not induced significantly by {alpha}-particles. ...

A tk{sup +/-} mouse embryonic stem (ES) cell line, designated 1G2, has been created in which one allele of the thymidine kinase (tk) gene was inactivated by targeted homologous recombination. This line is an analog of the mouse lymphoma tk{sup +/-} L5178Y cell line, which is used widely to assess the mutagenicity of chemical agents. Treatment of 1G2 cells with the alkylating agent N-ethyl-N-nitrosourea (ENU) resulted in a dose-related increase in tribluorothymidine-resistant colonies. Mutant frequencies of 152 and 296 per 10{sup 6} cells were determined for 0.1 and 0.3 mg/ml doses of ENU, compared with a spontaneous mutant frequency of 15 per 10{sup 6} cells. The data indicate that tk{sup +/-} 1G2 ES cells may be useful for the creation of a transgenic mouse model for assessing in vivo mutation using an endogenous autosomal gene. 45 refs., 2 figs., 1 tab.

Johnston Atoll (JA) is a small (270-ha), but strategic, US possession in the Pacific Ocean, which was previously used in nuclear weapons testing. Nuclear devices were launched by missile for detonations at very high altitudes. In 1962, one missile failed on the launch pad and two failed overhead. The devices were destructed without nuclear yield, but transuranium (TRU) elements were dispersed. Cleanup was swift and incomplete. A 2-ha area was placed under radiological controls and restricted from use due to residual contamination. Planning was begun in 1983 for a total JA cleanup to provide additional (unrestricted) land to meet future requirements. A TRUe soil cleanup is programmed to begin at JA in 1988 utilizing a full-scale mining plant. The plant should be able to process all contaminated soil by 1992 and produce less than #approx# 2000 m"3 of concentrated waste. This cleanup will increase the amount of land available ...

We present a drawing package optimised for Feynman diagrams. These can be constructed interactively with a mouse-driven graphical interface or from a script file, more suitable to work with a diagram generator. It provides most features encountered in Feynman diagrams and allows to modify every part of a diagram after its creation. Special attention has been paid to obtain a high quality printout as easily as possible. This package is written in Tcl/Tk and in C. (orig.).

A broad range of persistent organic pollutants (POPs) and selected heavy metals has been analysed in sediment cores of North-East Germany. The pollutants analysed include polychlorinated dioxins and furans (PCDD/Fs), polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs), several pesticides (DDT, HCH, CBz) and its metabolites as well as selected heavy metals. The sediment cores were sampled at five locations, reflecting a range of anthropogenic influences and background contamination: Arkona Basin (AK) representing a remote marine site, Lake Bugsin (BS) a background location only with atmospheric deposition, Lake Quenz (QS) close to the industrial city of Brandenburg, Teltowkanal (TK) in the suburban-industrial zone of Berlin and Lake White (WS) in the centre of Berlin. The objective of the inspection qualification is to ensure that applied non-destructive methods (NDT) have the required performance. The approach of the qualification process is systematic and carefully documented and therefore some new requirements can be seen concerning the composition of the documentation. There are also new document types, that only application of the inspection qualification requires. The most important of these is the technical justification, that includes the documented evidences concerning the performance of the inspection system. The aim of this case study is to analyse the socio-economic effects of forest fuel supply and use chains. Two of the four cases are rural district heating plants. In Perho, the co-operative organisation of forest owners maintains the fuel supply and operates the plant. In Ruukki, the local forestry association supplies forest chips to the plant. These medium-scale plants use small-sized trees from young thinning stands as their main fuel. The trees are felled manually. The other two cases are large power plants. Alholmens Kraft's new power plant in Pietarsaari aim to use 250 000 solid-m3 of forest chips annually. The logging residues are hauled to the plant as bales using traditional timber harvesting equipment or as bulk material. At young thinning stands the trees are harvested using an accumulating feller buncher. The Toppila I and II power plants in the city of Oulu used about 36 000 solid-m3 of forest chips. Isobaric specific heat capacities were measured for (2-methyl-2-butanol + heptane) mixtures and cyclopentanol within the temperature range from (284 to 353) K, and for 2-methyl-2-butanol in the (284 to 368) K temperature interval by means of a differential scanning calorimeter. The excess molar heat capacities were calculated from the experimental results. For the temperature range from (284 to 287) K, the excess molar heat capacity is S-shaped with negative values in the 2-methyl-2-butanol rich region and with small negative values at low alcohol concentrations at temperatures from (295 to 353) K. The excess molar heat capacities are positive for all compositions under test at temperatures from (288 to 294) K. The results are explained in terms of the influence of the molecular size and configuration of the alkanols on their self-association capability and of the change in molecular structure of the (2-methyl-2-butanol + heptane) mixtures. Formation of particles from heavy duty diesel and CNG vehicles was studied in laboratory conditions. PM, nano and soot mode particle emissions formed from buses of Euro 2-5 -emission levels were studied in relation to age and type approval characteristics of the vehicle. Characteristic particle emission factors from city driving were obtained by driving the buses on VTT's heavy duty chassis dynamometer applying the transient Braunschweig city cycle. The vehicles were a representative set of Euro 2 to 3 emission categories, supplemented with Euro 4-5 (EEV) -targeted CRT, CNG and DPF cars. Particle number emissions from Euro 4 to 5 buses were 1/100-1/1000 and mass emissions 1/10-1/100 of those of the current fleet. Particle emission from natural gas buses and after an advanced CRT trap contained almost entirely liquid < 60 nm nano particles, which will make filter mass based PM control very challenging in the near future.
