Molecular recognition of nitrated fatty acids by PPAR[gamma

Peroxisome proliferator activated receptor-{gamma} (PPAR{gamma}) regulates metabolic homeostasis and adipocyte differentiation, and it is activated by oxidized and nitrated fatty acids. Here we report the crystal structure of the PPAR{gamma} ligand binding domain bound to nitrated linoleic acid, a potent endogenous ligand of PPAR{gamma}. Structural and functional studies of receptor-ligand interactions reveal the molecular basis of PPAR{gamma} discrimination of various naturally occurring fatty acid derivatives.

Cloning and expression of a human kidney cDNA for an /alpha//sub 2/-adrenergic receptor subtype

An /alpha//sub 2/-adrenergic receptor subtype has been cloned from a human kidney cDNA library using the gene for the human platelet /alpha//sub 2/-adrenergic receptor as a probe. The deduced amino acid sequence resembles the human platelet /alpha//sub 2/-adrenergic receptor and is consistent with the structure of other members of he family of guanine nucleotide-binding protein-coupled receptors. The cDNA was expressed in a mammalian cell line (COS-7), and the /alpha//sub 2/-adrenergic ligand (/sup 3/H)rauwolscine was bound. Competition curve analysis with a variety of adrenergic ligands suggests that this cDNA clone represents the /alpha//sub 2/B-adrenergic receptor. IGF-II receptor phosphorylation proceeded to a stoichiometry of approximately 0.5 mol of phosphate/IGF-II binding site after 10 min of incubation at 4 degrees C. A Km for ATP of 6 microM was calculated for this phosphorylation reaction. Full text: The maturation of chicken forebrain is protracted and occurs well after synapse formation providing a good model for studying mechanisms of brain maturation. Using microslices from immature (10 day) and adult chicken forebrain prepared after decapitation, we have examined functional properties of NMDA and AMPA receptors by measuring agonist-induced uptake of "4"5Ca"2"+ . The rate and extent of NMDA induced "4"5Ca"2"+ accumulation decreased during maturation with no change in EC_5_0. The rate and extent of the AMPA induced response also decreased with a 60-fold increase in EC_5_0. However, the total NMDA receptor content did not change as indicated by 3 H-MK801 binding and NR1 immunoreactivity in P2 fractions. We report the isolation of cDNA clones for the mouse {alpha}7 neuronal nicotinic acetylcholine receptor subunit (gene symbol Acra7), the only nicotinic receptor subunit known to bind a-bungarotoxin in mammalian brain. This gene may have relevance to nicotine sensitivity and to some electrophysiologic findings in schizophrenia. The mouse {alpha}7 subunit gene encodes a protein of 502 amino acids with substantial identity to the rat (99.6%), human (92.8%), and chicken (87.5%) amino acid sequences. The {alpha}7 gene was mapped to mouse chromosome 7 near the p locus with the following gene order from proximal to distal: Myod1-3.5 {+-}1.7 cM-Gas2-0.9 cM {+-} 0.9 cM-D7Mit70-1.8 {+-} 1.2 cM- Acra7-4.4 {+-}1.0 cM-Hras1-ps11/Igf1r/Snrp2a. The recently described retinoid X receptors (RXRs) respond to the novel retinoid 9-cis-retinoic acid and also serve as heterodimeric partners for the vitamin D, thyroid hormone, and retinoic acid receptors (VDR, TR, and RAR, respectively). In this work, the authors report high-resolution localization of the human RXR genes within cytogenetic bands and also within a standard reference map of cosmid DNA markers on human chromosomes. Extracellular ATP stimulates transmembrane ion fluxes in the mouse macrophage cell line J774. In the presence of Mg2+, nonhydrolyzable ATP analogs and other purine and pyrimidine nucleotides do not elicit this response, suggesting the presence of a specific receptor for ATP on the macrophage plasma membrane. One candidate for such a receptor is the ecto-ATPase expressed on these cells. We, therefore, investigated the role of this enzyme in ATP-induced /sup 86/Rb+ efflux in J774 cells. The ecto-ATPase had a broad nucleotide specificity and did not hydrolyze extracellular ATP in the absence of divalent cations. /sup 86/Rb+ efflux was not blocked by inhibition of the ecto-ATPase and did not require Ca2+ or Mg2+. The pharmacology of the alpha-2 adrenoceptor of the human adipocyte was improved by using some new alpha-2 antagonists from different chemical families (imidazolines, benzazepines and benzofuroquinolizines) in biological and binding assays. Moreover, investigations were also carried out to define the binding properties of a new imidazolinic antagonist, RX821002 (2-(2-methoxy-1,4-benzodioxan-2yl)-2-imidazoline), which could be a potential radioligand. We examined the effect of the trophic protein, nerve growth factor (NGF), on cultures of fetal rat neostriatum and basal forebrain-medial septal area (BF-MS) to define its role in brain development. Treatment of cultures with NGF resulted in an increase in the specific activity of the cholinergic enzyme choline acetyltransferase (CAT) in both brain areas. CAT was immunocytochemically localized to neurons. In the BF-MS, NGF treatment elicited a marked increase in staining intensity and an apparent increase in the number of CAT-positive neurons. Moreover, treatment of BF-MS cultures with NGF increased the activity of acetylcholinesterase, suggesting that the cholinergic neuron as a whole was affected. Peroxisome proliferator-activated receptor gamma coactivator-1 (PGC-1), a cold-inducible coactivator of nuclear receptors, stimulates mitochondrial biogenesis and respiration in muscle cells. In the present study, we first cloned a rat PGC-1 gene from a brown adipose tissue cDNA library which encodes a predicted 796-amino-acid protein and exhibits respectively 98% and 95% identity with the mouse and human homologues. Next, we examined the effect of swimming exercise training on the level of expression of the PGC-1 gene in rat epitrochlearis (Epi) muscle. PGC-1 mRNA level in Epi muscle in rats that swam 2 h a day for 3 and 7 days increased dramatically by 154% and 163%, respectively, compared to the non-exercised control group. Assessment of GABARAP self-association by its diffusion properties

Gamma-aminobutyric acid type A receptor-associated protein (GABARAP) belongs to a family of small ubiquitin-like adaptor proteins implicated in intracellular vesicle trafficking and autophagy. We have used diffusion-ordered nuclear magnetic resonance spectroscopy to study the temperature and concentration dependence of the diffusion properties of GABARAP. Our data suggest the presence of distinct conformational states and provide support for self-association of GABARAP molecules. Assuming a monomer-dimer equilibrium, a temperature-dependent dissociation constant could be derived. Based on a temperature series of {sup 1}H{sup 15}N heteronuclear single quantum coherence nuclear magnetic resonance spectra, we propose residues potentially involved in GABARAP self-interaction. The possible biological significance of these observations is discussed with respect to alternative scenarios of oligomerization.

The epileptogenic and neurodegenerative effects of gamma-dendrotoxin, from Dendroaspis angusticeps, a specific blocker of a non-inactivating, voltage-sensitive K+ channel, were studied after focal injection into one dorsal hippocampus in rats pretreated with CGP040116, a N-methyl-D-aspartate (NMDA) receptor antagonist, and in rats bearing a monolateral surgical lesion of the Schaffer collaterals whose terminals originate from CA3 pyramids and release glutamate in the CA1 hippocampal area. Administration of 35 pmol gamma-dendrotoxin elicited in all of the treated animals (n=8) bilateral EEG discharges and damage to the hippocampal formation. We studied the effect of extracellular Ca"2"+ concentration ([Ca"2"+]_e) on adipocyte differentiation. Preadipocytes exposed to continuous [Ca"2"+]_e higher than 2.5 mmol/l accumulated little or no cytoplasmic lipid compared to controls in 1.8 mmol/l [Ca"2"+]_e. Differentiation was monitored by Oil Red O staining of cytoplasmic lipid and triglyceride assay of accumulated lipid, by RT-PCR analysis of adipogenic markers, and by the activity of glycerol-3-phosphate dehydrogenase (GPDH). The authors have used limited proteolysis of affinity-labeled estrogen receptors (ER), coupled with antireceptor antibody immunoreactivity, to assess structural features of ER and the relatedness of ER from MCF-7 human breast cancer and rat uterine cells. MCF-7 ER preparations covalently labeled with ["3H]tamoxifen aziridine (["3H]TAZ) were treated with trypsin (T), #alpha#-chymotrypsin (C), or Staphylococcus aureus V8 protease prior to electrophoresis on sodium dodecyl sulfate gels. We show that #gamma##gamma# colliders have important advantages for the observation of excited leptons and quarks in comparison with ee, ep, and pp colliders discussed in previous papers. Although the radioreceptor method is widely used for estrogen receptor assay in human tissue, it has several limitations and a number of alternative methods are being explored. An immunohistochemical method of estrogen receptor assay using a specific antibody to estradiol has been proposed as a suitable alternative. The present study was designed to evaluate the validity of this method in detecting true estrogen receptors in human tumor tissue. Using radioiodinated antibody to estrogen, we have demonstrated that the estrogen antibody can detect the estrogen when it is bound to 4S type receptor but is unable to bind to estrogen when the hormone is bound to 8S type receptor. The authors study the processes {gamma}e {r{underscore}arrow} {gamma}e and {gamma}{gamma} {r{underscore}arrow} {gamma}{gamma}, in the context of the proposal for Weak Scale Quantum Gravity (WSQG) with large extra dimensions. With an ultraviolet cutoff M{sub S} {approximately} 1 TeV for the effective gravity theory, the cross sections obtained for these processes at the Next Linear Collider (NLC), with the e{gamma} an {gamma}{gamma} options, deviate from the predictions of the Standard Model significantly. The results suggest that, for typical proposed NLC energies and luminosities, the predictions of WSQG can be tested in the range 1 TeV {approx{underscore}lt} M{sub S} {approx{underscore}lt} 10 TeV, making e{gamma} an {gamma}{gamma} colliders important tools for probing WSQG.

Insulin receptors on the surface of isolated rat adipocytes were photoaffinity labeled at 12 degrees C with the iodinated photoreactive insulin analogue, 125I-B2 (2-nitro-4-azidophenylacetyl)-des-PheB1-insulin, and the pathways in the intracellular processing of the labeled receptors were studied at 37 degrees C. During 37 degrees C incubations, the labeled 440-kDa insulin receptors were continuously internalized (as assessed by trypsin inaccessibility) and degraded such that up to 50% of the initially labeled receptors were lost by 120 min. We summarize the predictions of different models for total #gamma##gamma# cross-sections. The experimentally observed rise of #sigma#_#gamma#_#gamma# with s radical _#gamma#_#gamma#, faster than that for #sigma#_p_-_b_a_r_p, #sigma#_#gamma#_p is in agreement with the predictions of the Eikonalized Minijet Models as opposed to those of the Regge-Pomeron models. The in vivo detection of insulitis could be of relevance for early prediction of diabetes. Medical imaging using single gamma-ray-emitting radionuclides typically makes use of parallel hole collimators or pinholes in order to achieve good spatial resolution. However, a tradeoff in sensitivity is inherent in the use of a collimator, and modern preclinical single photon emission computed tomography (SPECT) systems detect a very small fraction of emitted gamma rays, often less than 0.1%. A system for small animal SPECT imaging which uses no collimators could potentially achieve very high sensitivity-several tens of percent-with reasonably sized detectors. Using 347.5 fb-1 of data recorded by the BABAR detector at the PEP-II electron-positron collider, 244*10^3 signal events for the D+ --> K- pi+ e+ nu_e decay channel are analyzed. This decay mode is dominated by the \\bar{K}^*(892)^0 contribution. We determine the \\bar{K}^*(892)^0 parameters: m_{K^*(892)^0}=(895.4 +- 0.2 +- 0.2) MeV/c^{2}, \\Gamma^0_{K^*(892)^0}=(46.5 +- 0.3 +- 0.2) MeV/c^{2} and the Blatt-Weisskopf parameter $r_{BW}=2.1 +- 0.5 +- 0.5 (GeV/c)^{-1} where the first uncertainty comes from statistics and the second from systematic uncertainties. Cloning of the cDNA and gene for a human D sub 2 dopamine receptor

A clone encoding a human D{sub 2} dopamine receptor was isolated from a pituitary cDNA library and sequenced. The deduced protein sequence is 96% identical with that of the cloned rat receptor with one major difference: the human receptor contains an additional 29 amino acids in its putative third cytoplasmic loop. Southern blotting demonstrated the presence of only one human D{sub 2} receptor gene. Two overlapping phage containing the gene were isolated and characterized. DNA sequence analysis of these clones showed that the coding sequence is interrupted by six introns and that the additional amino acids present in the human pituitary receptor are encoded by a single exon of 87 base pairs. The involvement of this sequence in alternative splicing and its biological significance are discussed.

A range of 6-substituted imidazo[1,2-a]pyridines were synthesized using a multicomponent coupling reaction. Most of these compounds were found to exhibit excellent activity against the colon cancer cell lines HT-29 and Caco-2, whilst not showing significant toxicity against white blood cells. Our studies have shown that the proteolytic phase of apoptosis was initiated 2 h after treatment with these imidazo-[1,2-a]pyridines. The data suggests that the imidazo[1,2-a]pyridine-induced cell death in HT-29 and Caco-2 cells is mediated via pathway(s) that include the release of cytochrome c from the mitochondria to the cytosol and the activation of caspase 3 and caspase 8.

We present methods to measure the beam polarizations and the luminosity of [gamma][gamma] colliders at TeV energy scale. The beam polarizations of a [gamma][gamma] collider can easily be monitored by comparing the numbers of events of the processes [gamma][gamma] [yields] l[sup +]l[sup -] and [gamma][gamma] [yields] W[sup +] W[sup -], where l means e or [mu]. The luminosity of a [gamma][gamma] collider is also measurable by the event rate of W boson pair productions and the light lepton pair productions. (orig.)

We discuss the search of excited leptons and quarks with spin 1/2 at {gamma}{gamma} and at {gamma}e colliders. We show that {gamma}{gamma} colliders have important advantages for the observation of excited leptons and quarks in comparison with ee, ep, and pp colliders discussed in previous papers. These collisions give a simple test for the chirality of the l{sup *}l{gamma} transition. The anomalous magnetic moment of excited leptons can be observed when its value is not too small. (orig.).

Characterization of an oligopeptide chemoattractant receptor on human blood monocytes using a new radioligand

The study of chemoattractant receptors on human monocytes had been limited by the lack of a radioligand suitable for use with the small numbers of cells routinely available from human donors. A new synthetic oligopeptide radioligand f(/sup 35/S)Met-leu-phe, with a higher specific radioactivity than was available with the tritiated compound, was used to characterize a chemoattractant receptor on freshly isolated human blood monocytes. These cells bind f(/sup 35/)met-leu-phe with a dissociation constant (KD) of 30.2 +/- 5.6 nM and contain 84,000 +/- 11,300 receptors per cell. f(/sup 35/)met-leu-phe does not bind specifically to blood lymphocytes. We previously reported that the peroxisome proliferator-activated receptor gamma coactivator-1 (PGC-1) mRNA in rat epitrochlearis muscle was increased after swimming exercise training. In the present study, we demonstrated further that PGC-1 mRNA expression in the epitrochlearis muscle of 4-5-week-old male Sprague-Dawley rats was increased after a 6-h acute bout of low-intensity swimming exercise. With this increase, the expression level was approximately 8-fold of control and immersion group rats that stayed for 6-h in warm water, maintained at the identical temperature of the swimming barrel (35 degrees C) (p<0.01). Second, PGC-1 mRNA expression in the muscle was found to have increased 6-h after 30 10-s tetani contractions were induced by in vitro electrical stimulation. They recently demonstrated that sigma-1 receptors are expressed in human melanoma cells and that sigma receptors could be used as external markers for non-invasive imaging. In order to determine the presence of sigma receptors in other human tumors they have screened numerous non-small cell lung carcinoma, melanoma and breast cancer cell lines. Somatogenic hormones play an important role in regulation of receptors for prolactin (PRL) and estrogen. Plerocercoids of the tapeworm, S. mansonoides produce a factor which mimics some, but not all of the actions reported for GH. Intact female rats were subjected to a constant infusion of plerocercoid growth factor (PGF) via a subcutaneous infection for two weeks to determine if PGF influences receptors for PRL, GH or estradiol. The rate of weight gain in the PGF-treated rats was accelerated in spite of a marked reduction in serum GH. Partially-purified PGF specifically displaced [125I]hGH from rat liver receptors but microsomes prepared from rats treated with PGF specifically bound significantly less [125I]hGH than microsomes from control rats. A study was made of the kinetics of L-(/sup 3/H)quinuclidinyl benzilate binding with membrane-bound muscarine cholinoreceptors from the rat brain. Determination was made of the rate constants of this process, and constants as a function of ligand concentration was investigated, indicating that there is a complex mechanism of interaction of antagonists with muscarine receptor. With up to 3.5 nM concentrations of L-(/sup 3/H)quinuclidinyl benzilate, the binding reaction occurs in two stages: rapid equilibrated binding is followed by a slow process of conformational isomerization of the receptor-ligand complex. Triple-vector-boson processes in #gamma##gamma# colliders

We study the production of three gauge bosons (W"+W"-Z"0 and W"+W"-#gamma#) at the next generation of linear e"+e"- colliders operating in the #gamma##gamma# mode. We analyze the total cross sections as well as several kinematical distributions of the final state particles. We find out that a linear e"+e"- machine operating in the #gamma##gamma# mode will produce 5--10 times more three-gauge-boson states compared to the standard e"+e"- mode at high energies.

Singlet scalars as Higgs imposters at the Large Hadron Collider

An electroweak singlet scalar can couple to pairs of vector bosons through loop-induced dimension five operators. Compared to a Standard Model Higgs boson, the singlet decay widths in the diphotons and Z gamma channels are generically enhanced, while decays into massive final states like WW and ZZ are kinematically disfavored. The overall event rates into gamma gamma and Z gamma can exceed the Standard Model expectations by orders of magnitude. Such a singlet may appear as a resonant signal in the gamma gamma and Z gamma channels, even with a mass above the WW kinematic threshold.

Probing anomalous top-quark couplings induced by dim.6 operators at photon colliders

Possible anomalous top-quark couplings induced by SU(2)xU(1) gauge-invariant dimension-6 effective operators were studied in the process of ttbar productions and decays at polarized #gamma##gamma# colliders. Two CP-violating asymmetries, a linear-polarization asymmetry and a circular-polarization asymmetry, were computed including both non-standard ttbar#gamma# and #gamma##gamma#H couplings. An optimal-observable analysis for the process #gamma##gamma##->#ttbar#->#l"#+-# was performed in order to estimate the precision for determination of all relevant non-standard couplings, including the anomalous tbW coupling.

The decay modes $\\jpsi\\ar\\gamma\\pi^0, \\gamma\\eta$ and $\\gamma\\etap$ are analyzed using a data sample of 58 million $\\jpsi$ decays collected with the BESII detector at BEPC. The branching fractions are determined to be: $Br(\\jpsi\\ar\\gamma\\pi^0)=(3.13^{+0.65}_{-0.44})\\times10^{-5}$, $Br(\\jpsi\\ar\\gamma\\eta)=(11.23\\pm0.89)\\times10^{-4}$, and $Br(\\jpsi\\ar\\gamma\\etap)=(5.55\\pm0.44)\\times10^{-3}$, where the errors are combined statistical and systematic errors. The ratio of partial widths $\\Gamma(\\jpsi\\ar\\gamma\\etap)/\\Gamma(\\jpsi\\ar\\gamma\\eta)$ is measured to be $4.94\\pm0.40$, and the singlet-octet pseudoscalar mixing angle of $\\eta-\\etap$ system is determined to be $\\theta_{P}=(-22.08\\pm0.81)^{\\circ}$.

Single dilepton production at e"+e"-, e#gamma#, and #gamma##gamma# colliders

We calculate the cross sections for the single production of doubly charged dileptons, both scalar and vector, at e"+e"-, e#gamma#, and #gamma##gamma# colliders at #sq root#s =500 GeV and 1 TeV. The e#gamma# mode is by far the most promising---dileptons whose coupling is as weak as #approx#10"-"4#alpha#_e_m can be observed, for masses virtually up to the kinematic limit. Dileptons of mass up to #sq root#s can also be seen in e"+e"- and #gamma##gamma# colliders, for couplings of order #alpha#_e_m. In all three colliders, most of the cross section comes from events in which the only particles detected are e"-e"- (or #mu#"-#mu#"- or #tau#"-#tau#"-), the decay products of the dilepton, yielding an unmistakable experimental signature.

Single charged top-pion production at next generation #gamma##gamma# colliders

The future e"+e"- linear colliders can also operate in the e#gamma# or #gamma##gamma# modes. In the context of the top-color assisted technicolor (TC2) model, we study the single charged top-pion production process #gamma##gamma# #-># tb-bar#PI#_t"-. The results show that the production rates can reach the level of tens fb with reasonable parameter values. So one can expect that enough signals could be produced in future high-energy linear collider experiments. Furthermore, the flavor-changing (FC) decay mode #PI#_t"- #-># bc-bar is the best channel to detect the charged top pion due to the clean standard model background. With a large number of events and the clean background, the charged top pion should be observable at future linear colliders operating in #gamma##gamma# mode at the TeV energy scale

Atom-probe field-ion microscopy investigation of CMSX-4 Ni-base superalloy laser beam welds

CMSX-4 superalloy laser beam welds were investigated by transmission electron microscopy and atom probe field-ion microscopy (APFIM). The weld microstructure consisted of fine (10- to 50-nm) irregularly shaped {gamma}` precipitates (0.65 to 0.75 volume fraction) within the {gamma} matrix. APFIM compositions of the {gamma} and {gamma}` phases were found to be different from those in the base metal. Concentration profiles across the {gamma} and {gamma}` phases showed extensive variations of Cr, Co and Al concentrations as a function of distance within the {gamma} phase. Calculated lattice misfits near the {gamma}/{gamma}` interface in the welds are positive values compared to the negative values for base metal. (orig.).

Anomalous g_5"Z coupling at #gamma##gamma# colliders

We study the constraints on the anomalous coupling g_5"Z that can be obtained from the analysis of the reaction #gamma##gamma##->#W"+W"-Z at future linear e"+e"- colliders. We find out that a 0.5 (1) TeV e"+e"- collider operating in the #gamma##gamma# mode can probe values of g_5"Z of the order of 0.15 (4.5x10"-"2) for an integrated luminosity of 10 fb"-"1. This shows that the ability to search for this anomalous interaction of the #gamma##gamma# mode is better than the one of the usual e"+e"- mode, and it is similar to the ability of the e#gamma# mode.

["3H]QNB binding and contraction of rabbit colonic smooth muscle cells

The authors used radioligand binding and studies of cell contraction to characterize muscarinic receptors on dispersed smooth muscle cells from rabbit proximal and distal colon. Cells obtained after serial incubations in collagenase were used to measure binding of tritiated quinuclidinyl benzilate (["3H]QNB). At 37 degree C, specific ["3H]QNB binding was saturable and linearly related to cell number. Nonlinear regression analysis was used to determine the affinity of ["3H]QNB for its receptor. The IC_5_0 for the muscarinic agonists bethanechol and oxotremorine were 80 and 0.57 #mu#M, respectively. Hill coefficients were 0.67 for both, suggesting more complex interaction involving receptors of different affinities. In studies of cell contraction, bethanechol stimulated a dose-dependent decrease in cell length with half the maximal contraction occurring at 100 pM. The temporal development of the effect of THIP (4,5,6,7-tetrahydroisoxazolo[5,4-c]pyridin-3-ol) on the ultrastructure composition and GABA receptor expression in cerebellar granule cells was investigated by quantitative electron microscopy (morphometric analysis) and GABA binding assays. It was found that the cytoplasmic density of smooth endoplasmic reticulum was decreased, while the cytoplasmic density of rough endoplasmic reticulum, Golgi apparatus, vesicles and coated vesicles was greatly enhanced after exposure of the cells to THIP (150 microM) for only 1 hr. In cerebellar granule cells exposed to THIP (150 microM) for 3 hr low affinity GABA receptors were induced. The effects of muscarinic agonists and diisopropylfluorophosphate (DFP) on muscarinic receptor density and muscarinic receptor-mediated responses was assessed in mouse brain and mouse neuroblastoma cells (clone N1E-115). Utilizing the antagonist ({sup 3}H)quinuclidinyl benzilate (({sup 3}H)QNB), there was no difference in the maximal binding capacity (B{sub max}) or equilibrium dissociation constant (K{sub d}) between untreated and 24 hour DFP-treated mice. By improving the currently used lactoperoxidase method, we were able to obtain radioiodinated chicken luteinizing hormone (LH) that shows high specific binding and low nonspecific binding to a crude plasma membrane fraction of testicular cells of the domestic fowl and the Japanese quail, and to the ovarian granulosa cells of the Japanese quail. The change we made from the original method consisted of (1) using chicken LH for radioiodination that was not only highly purified but also retained a high receptor binding potency; (2) controlling the level of incorporation of radioiodine into chicken LH molecules by employing a short reaction time and low temperature; and (3) fractionating radioiodinated chicken LH further by gel filtration using high-performance liquid chromatography. Pair production of charged top-pions in the $\\gamma\\gamma$ collisions at the ILC

The top-color assisted technicolor (TC2) mode predicts the existence of a pair of charged top-pions $\\pi^{\\pm}_t$. In this paper, we study the production of the charged top-pions pair $\\pi^{\\pm}_t$ at next generation $\\gamma\\gamma$ colliders. The results show that the production rates can reach the level of $10^2$ fb with reasonable parameter space. With a large number of events and the clean background, the charged top-pion should be observable at the $\\gamma\\gamma$ colliders. Therefore, our studies can help us to search for charged top-pion, and furthermore, to test the TC2 model.

Compton backscattering of laser photons near the interaction point of an e"+e"- or e"-e"- collider can be used to produce a #gamma#-#gamma# or #gamma#-e"- collider. This paper describes the laser requirements, including pulse duration, intensity, energy, and wavelength, for such a collider. For most of the proposed, next generation, e"+e"- colliders, the laser wavelength should be in the near-infrared, with a pulse duration of 1 ps or less and an energy of similar 1 J per pulse. Current chirped pulse amplification laser systems in solid state lasing materials are well suited to meet these requirements. These systems are described. ((orig.)).

Heavy quark production in $\\gamma\\gamma$ collisions is analyzed within the approach to hard collisions of photons recently proposed by the author. In this approach evaluating the cross section $\\sigma(\\gamma\\gamma\\to Q\\bar{Q})$ in the ``next-to-leading order of QCD'' requires the inclusion of direct photon contributions up to the order $\\alpha^2\\alpha_s^2$, whereas in the standard approach direct photon terms only up to the order $\\alpha^2\\alpha_s$ are taken into account. We investigate the semihard production of neutral pseudoscalar and tensor mesons in high-energy [gamma][gamma] collisions (M=P=[pi][sup 0], [eta], [eta]' or M=T=a[sub 2], f[sub 2], f[sub 2]'). We deal with the exclusive [gamma][gamma][yields]MM' or semi-exclusive [gamma][gamma][yields]MX reactions (X is the hadron jet with not too large mass). The considered transfer momenta are small in comparison with the photon energies and they are large in comparison with the confinement scale. The amplitudes of these processes are determined by the odderon exchange, i.e. three-gluon exchange in the lowest order of perturbative QCD. The cross sections are calculated in this approximation. The production of pairs of doubly charged vector bileptons is studied at future #gamma##gamma# colliders. The unpolarized cross section for the #gamma##gamma##->#Y"-"-Y"+"+ subprocess is analytically calculated and convoluted to predict the number of events in the complete e"+e"-#->##gamma##gamma##->#Y"-"-Y"+"+ process. The gauge or nongauge character of the vector bilepton Y"#+-#"#+-# is discussed. It is found that, as a consequence of its spectacular signature, as it decays dominantly into two identical charged leptons, and also due to its charge contents, which significantly enhance the cross section, the detection of this class of particles with mass in the sub-TeV region can be at the reach of these colliders. The coupled neutron-gamma cross section libraries BUGLE-80 and SAILOR are compared, with focus on gamma production and transport and its sensitivity to the thermal component of the neutron flux. In addition, the importance of the prompt versus delayed components of the gamma source is addressed. Although BUGLE-80 shows some discrepancies in the core region, the two libraries demonstrate similar capability in the vicinity of the pressure vessel and beyond. The delayed fission product gammas from the core have little impact on the local gamma fluxes beyond the pressure vessel. Nitromethane (CH(3)NO(2)) and its chlorinated analogue, chloropicrin (CCl(3)NO(2)), were photolyzed at 193, 248, and 266 nm, and the products were observed by time-dependent Fourier transform infrared emission spectroscopy. At 193 and 248 nm, the primary photodissociation pathway for nitromethane was cleavage of the C-N bond to produce CH(3) + NO(2)(A (2)B(2)). At 266 nm, weak emission was observed following photodissociation of nitromethane, but an infrared spectrum could not be obtained. The photodissociation of chloropicrin at 193 nm produced the analogous product channel CCl(3) + NO(2)(A (2)B(2)) in addition to several other product channels. At 248 and 266 nm, only CCl(3) + NO(2)(A (2)B(2)) was observed. The production of phosgene (CCl(2)O) from chloropicrin photodissociation was not observed in this study. PMID:16571044

The plerocercoid stage of the tapeworm Spirometra mansonoides produces a functional analog of human growth hormone (hGH). Among the similarities between plerocercoid growth factor (PGF) and hGH is competition for the same receptors on rabbit liver membranes. To take advantage of this characteristic in a purification scheme for PGF, rabbit liver microsomes were solubilized in Triton X-100 and the hGH receptors were purified over an hGH affinity column. The purified receptors from six rabbit livers were coupled to Affi-Gel-10 to create a receptor affinity column which was used to purify PGF. Chromatography of crude PGF over the receptor column resulted in a 1044 fold increase in specific activity. Somatostatin receptor scintigraphy (SRS) is a valuable method for the detection of somatostatin receptor-positive lesions. Most gastrinomas (over-)express the somatostatin receptor subtype 2 which can be targeted by In-111 labeled Octreotide. Different studies show a high sensitivity of SRS for the localization and staging of gastrinomas. SRS seems to be superior to other non-invasive imaging modalities and has been proven to significantly contribute to patient management. However, the sensitivity depends on the size and exact localization of the tumors. Smaller lesions and lesions located in the duodenum show a significantly lower sensitivity. In any case, SRS belongs to the routine imaging procedure for gastrinomas for localization and staging and can also be used for evaluation of the tumor progression. (author)

Irradiation induced dislocation loop and its influence on the hardening behavior of Fe-Cr alloys by an Fe ion irradiation

Nano indentation analysis and transmission electron microscopy observation were performed to investigate a microstructural evolution and its influence on the hardening behavior in Fe-Cr alloys after an irradiation with 8 MeV Fe4+ ions at room temperature. Nano indentation analysis shows that an irradiation induced hardening is generated more considerably in the Fe-15Cr alloy than in the Fe-5Cr alloy by the ion irradiation. TEM observation reveals a significant population of the a0 dislocation loops in the Fe-15Cr alloy and an agglomeration of the 1/2a0 dislocation loops in the Fe-5Cr alloy. The results indicate that the a0 dislocation loops will act as stronger obstacles to a dislocation motion than 1/2a0 dislocation loops.

AFM Imaging of SWI/SNF action: mapping the nucleosome remodeling and sliding

We propose a combined experimental (Atomic Force Microscopy) and theoretical study of the structural and dynamical properties of nucleosomes. In contrast to biochemical approaches, this method allows to determine simultaneously the DNA complexed length distribution and nucleosome position in various contexts. First, we show that differences in the nucleo-proteic structure observed between conventional H2A and H2A.Bbd variant nucleosomes induce quantitative changes in the in the length distribution of DNA complexed with histones. Then, the sliding action of remodeling complex SWI/SNF is characterized through the evolution of the nucleosome position and wrapped DNA length mapping. Using a linear energetic model for the distribution of DNA complexed length, we extract the net wrapping energy of DNA onto the histone octamer, and compare it to previous studies.

It is found that the decomposition of primary MC carbides, which is accompanied by the precipitation of M23C6 particles, may lead to void nucleation and growth. Somatostatin receptor expression has been demonstrated on a number of plasma cell lines. Therefore, we questioned whether somatostatin receptor scintigraphy (SRS) can be used to demonstrate in vivo multiple myeloma (MM) activity. SRS was performed in newly diagnosed (n = 9) or relapsing (n = 18) MM patients or in patients with localized plasmacytoma (n = 2). The results were compared with radiographic findings. A positive SRS was demonstrated in 44% of the newly diagnosed patients, in 83% of the relapsed patients and in both patients with plasmacytoma. The SRS findings corresponded with radiographic abnormalities in 40% of the patients. However, in relapsed patients 60% demonstrated increased SRS uptake in areas without new radiographic abnormalities. First, CAAs (0.1 mM) caused a surface activity-dependent depression on ATPase activity in the cell membrane extract, but it had no effects on other cellular extracts, suggesting an indispensable role of the membrane environment for pharmacological activity. Recent reports indicate that ETB is localized to nuclei in cardiac ventricular myocytes, although it has been thought that ETB is localized mainly on the plasma membrane. It remains unknown, however, whether this unique distribution of ETB occurs in other tissues. To elucidate the subcellular distribution of ETB in the intestine, we performed immunofluorescence of ETB in mouse intestine using a specific antibody. The specific receptor sites and the endocytosis of transferrin (Tf) are evidenced in a cell line (HT-29) derived from a human colic adenocarcinoma by means of "1"2"5I radiolabeled Tf. The receptor density is studied in undifferentiated (UD) or differentiated (D) cells with respective doubling times of about 24 hours and 46 hours. The number of binding sites for Tf is 162,000 (K_d = 7.8 nmol/l in ND cells and 68,000 (K_d = 7.40 nmol/l) in D cells. The distribution between the Tf bound to the cell surface and the internalized Tf is investigated by elimination of Tf bound to the surface by an acid wash method. The intracellular cycle of Tf seems to be characterized by a slower kinetics in UD cells. The high density of Tf receptor sites in HT-29 UD cells should allow the detection or the treatment of highly evolutive colic adenocarcinoma by means of Tf.

The main goal of this conversion is to obtain high-energy intense #gamma# beams for #gamma#e and #gamma##gamma# colliders. This conversion has important by-products: (i) one can observe here effect of nonlinear QED (in a strong external field)-production of e"+e"- pairs by a high-energy photon and emission of very high-energy photons by an electron; (ii) one can produce and observe here the invisible axion in colliding #gamma#_0e beams. The NQED effects also result in variation of the spectra of high-energy photons and an additional background. 12 refs., 12 figs., 1 tab.

Low-lying states of {sup 109}Sn from the {sup 106}Cd({alpha},n{gamma}) reaction

Measurements of {gamma}-ray, {gamma}{gamma}-coincidence and internal conversion electron spectra from the {sup 106}Cd({alpha},n{gamma}){sup 109}Sn reaction were carried out at 15-20 MeV {alpha}-particle bombarding energies with Ge(HP) {gamma}-ray and superconducting magnetic lens plus Si(Li) electron spectrometers. The energies, relative intensities, internal conversion coefficients and coincidence relations of the {sup 109}Sn transitions were determined, and a more complete, consistent level scheme has been deduced. Spin and parity values have been determined from the internal conversion coefficients, the bombarding-energy dependence of the side-feeding intensities of the states and the available {gamma}-ray angular distribution data. The level scheme was interpreted in the framework of the quasi-particle shell model.

W, Z and H bosons in the three particle final states production at TeV energy #gamma#e and #gamma##gamma#colliders

The short review of complete tree level calculations for three particle final states production at the future e"+e"-, #gamma#e and #gamma##gamma# colliders is presented. The results obtained with the help of CompHEP system for total cross sections and other characteristics of processes in the energy range 0.1-2 TeV are summarized and their comparison with the results of different approaches is discussed. In particular we are interested in the processes of W, Z and H boson production. The reactions under consideration are especially interesting in connection with probing of new couplings, searching for new particle signals and as an important backgrounds to these experiments. Single leptoquark production at e"+e- and #gamma##gamma# colliders

We consider single production of leptoquarks (LQ's) at e"+e- and #gamma##gamma# colliders, for two values of the center-of-mass energy: #sq root#s =500 GeV and 1 TeV. We find that LQ's which couple within the first generation are observable for LQ masses almost up to the kinematic limit, both at e"+e- and #gamma##gamma# colliders, for an LQ coupling strength equal to #alpha#_e_m. The cross sections for single production of second- and third-generation LQ's at e"+e- colliders are too small to be observable. In #gamma##gamma# collisions, on the other hand, second-generation LQ's with masses much larger than #sq root#s /2 can be detected. However, third-generation LQ's can be seen at #gamma##gamma# colliders only for masses at most #approx# #sq root#s /2, making their observation more probable via the pair production mechanism.

GAMMA GLOBULIN METABOLISM IN RABBITS DURING THE ANAMNESTIC RESPONSE.

1. Gamma globulin metabolism and distribution were studied employing rabbit gamma globulin (RGG)I(131) 24 times in 13 control rabbits. Similar studies were performed before and during the ananmestic response in 4 rabbits previously sensitized with a polyvalent pneumococcal vaccine. 2. During the ananmestic response, gamma-globulin levels increased from 1.0 to 6.0 gm/100 ml, and the gamma-globulin pool increased from 0.7 to 4.7 gm/kg. There was no change in the intravascular-extravascular partition of gamma globulin. 3. Gamma globulin degradation increased from 0.06 to 0.33 gm/kg/day during the 28 days of the immunization period while gamma globulin synthesis increased even further to average 0.47 gm/kg/day. Detection of the heavy Higgs boson at #gamma##gamma# colliders

We consider the possibility of detecting a heavy Higgs boson (m_H>2m_Z) in proposed #gamma##gamma# colliders through the semileptonic mode #gamma##gamma##->#H#->#ZZ#->#q bar ql"+l-. We show that due to the nonmonochromatic nature of the photon beams produced by the laser-backscattering method, the resultant cross section for Higgs production is much smaller than the on-resonance cross section, and generally decreases with increasing collider energy. Although continuum ZZ production is expected to be negligible, we demonstrate the presence of, and calculate sizable backgrounds from, #gamma##gamma##->#l"+l-Z,q bar qZ, with Z#->#q bar q,l"+l-, respectively, and #gamma##gamma##->#t bar t#->#b bar bl"+l-#nu# bar #nu#. This channel may be used to detect a Higgs boson of mass m_H up to around 350 GeV at a 0.5 TeV e"+e- collider, assuming a nominal yearly luminosity of 10--20 fb"-"1.

A recent study has elucidated the significant differences in the metabolomes of adipocytes, serum and an adipocyte cell line after activation of two nuclear receptors, peroxisome proliferator activated receptor ?/? Resistance and tolerance are two types of host defense mechanisms that increase fitness in response to fungi. Several genetic polymorphisms in pattern recognition receptors, most remarkably Toll-like receptors (TLRs), have been described to influence resistance and tolerance to aspergillosis in distinct clinical settings. TLRs on dendritic cells pivotally contribute in determining the balance between immunopathology and protective immunity to the fungus. Epithelial cells also contribute to this balance via selected TLRs converging on indoleamine-2,3-dioxygenase (IDO). A case study of trastuzumab treatment for metastatic breast cancer in pregnancy: fetal risks and management of cerebral metastases

Trastuzumab increases survival amongst women with human epidermal growth factor receptor (HER)-2 receptor positive metastatic breast cancer, but maternal and fetal risks are associated with advanced disease and its treatment in pregnancy. We present a case of a primigravid with HER-2 positive metastatic breast cancer who received trastuzumab throughout pregnancy. She presented with cerebral metastases, requiring surgical decompression and resection. Reversible oligohydramnios developed during pregnancy. Fetal safety data on trastuzumab in pregnancy is limited, but case reports suggest a recurring pattern of (mostly reversible) oligohydramnios.

In-beam #gamma#-ray spectroscopy of fast beams at the NSCL

With the development of an array of highly-segmented germanium detectors, it now becomes possible to perform in-flight #gamma#-ray spectroscopy experiments on intermediate energy beams with unprecedented #gamma#-ray energy resolution. Presented in this report are examples of two techniques in which SeGA, the most highly-segmented operational germanium array for in-flight spectroscopy with fast beams, was used for the detection of #gamma# rays. SeGA used in conjunction with a high-resolution magnetic spectrograph (S800) to detect the reaction residues in coincidence represents a powerful combination for in-beam #gamma#-ray studies.
