The preS2 antigens of hepatitis B virus (HBV), which causes a serious health problem in the world, have been implicated in hepatocyte cell binding and viral penetration. Therefore, the importance of antibody production against preS2 antigen for early diagnosis of HBV has been well established. In this study, the recombinant HBV preS2 single chain variable fragment (scFv) antibody was successfully expressed in E. coli with the novel cold shock vector (pCold) under the cspA promoter, and its expression level was compared with the pET vector under the T7 promoter. A CD33-specific single-chain immunotoxin mediates potent apoptosis of cultured human myeloid leukaemia cells.

A novel single-chain immunotoxin was constructed by combining a CD33-specific single chain Fv (scFv) antibody fragment with an engineered variant of Pseudomonas exotoxin A (ETA). The variant toxin carries the KDEL peptide at its C-terminus, a cellular peptide mediating improved retrograde transport to the endoplasmic reticulum. The purified recombinant fusion protein induced potent apoptosis of the human myeloid cell lines U937, HL-60 and THP-1. Up to 98% of U937 cells were eliminated after treatment for 72 h with a single dose of 500 ng/ml (c. 7 nmol/l). Killing was antigen-specific and occurred by apoptosis. A control protein, consisting of a CD19-specific scFv antibody fragment fused to the ETA-KDEL toxin, failed to induce death of the CD19-negative cell lines U937, HL-60 and THP-1. Useful vacancies: Positron beam interrogation of fluorine-vacancy complexes in semiconductor device structures

The formation, migration and agglomeration in silicon of fluorine-vacancy complexes have been monitored by single-detector Doppler broadening spectroscopy. After electronics engineers found that fluorine ion implantation effectively eliminated the transient-enhanced diffusion of dopants in the creation of ultra-shallow junctions, a vital step in the further miniaturization of device structures, positron beams have played a pivotal role in providing an insight into the mechanisms underlying this phenomenon, being able to detect FV complexes in implanted and annealed samples. Secondary Ion Mass Spectrometry has provided complementary information on fluorine concentrations so that the nature of the F{sub m}V{sub n} complexes can be further assessed. New results on Si and SiGe structures are presented.

Somatic effects in nuclear medicine and radiology

Probable risks for the occurrence of somatic effects due to diagnostic radiology and nuclear medicine are summarised. The biological background of radiation carcinogenesis and epidemiological results are discussed. At the Leiden University Medical Centre the average effective dose per examination due to diagnostic radiology and nuclear medicine amount to 0.95 and 4.4 mSv, respectively. These values correspond well with the average values of 0.82 and 3.0 mSv reported for The Netherlands as a whole. Since radiological examinations are performed at a much larger frequency than nuclear medicine the relative collective dose for the first type of examinations is higher than the latter. Risk for occurency of malignancies are at least one order of magnitude lower than the hypothetical risk due to the background radiation typical of The Netherlands.

Numerical study of steady/unsteady flow and heat transfer in porous media using a characteristics-based matrix-free implicit FV method on unstructured grids

In this study, a high-resolution characteristic-based finite-volume (FV) method on unstructured grids [Int. J. Numer. Method Eng. 50 (2001) 11; Int. J. Heat Fluid Flow 21 (2000) 432] is extended by a matrix-free implicit dual-time stepping scheme for the numerical simulation of steady and unsteady flow and heat transfer with porous media. The method has been used to study the characteristics of a complex problem: flow and heat transfer in a channel with multiple discrete porous blocks, which was originally proposed by Huang and Vafai [J. Thermophys. Heat Transfer 8 (3) (1994) 563]. In addition, flow and heat transfer in a channel partially or fully filled with porous layers and containing solid protruding blocks with constant heat flux on its lower surface are also investigated in details. Modern commercial alloys are not suited for future steam turbines of operating temperatures beyond 700 C. {gamma}'-hardened alloys like Waspaloy have good mechanical characteristics but have defects when large batches are melted. {gamma}'/{gamma}''-hardened alloys like Inconel 706, on the other hand, are easy to produce but have insufficient microstructural long-term stability and insufficient creep strength. Mixed crystal solidified alloys like Inconel 617 have excessive yield stress at forging temperatures. Two new alloys were therefore developed. Results so far show that they have good production characteristics but insufficient mechanical characteristics. The reasons for this are currently under investigation. (orig.) Pulmonary infections and tumors are a major cause of death in patients with AIDS. The combination of clinical, radiological, laboratory, and pathohistological data helps to narrow the spectrum of differential diagnoses or even allows a specific diagnosis in many patients. Nevertheless, an accurate diagnosis should be obtained as soon as possible during the clinical course of the illness to initiate treatment in time. Computed tomography (CT) has proven to provide promising results in the diagnosis of AIDS-related throacic diseases. The aim of this paper was to demonstrate the diagnostic capacities of CT in the context of particular AIDS-related thoracic pathologies. This action plan describes the working group's ideas on how the grid operators should prepare themselves for the implementation of decentralized applications in the next four years. The action plan contains three main lines that reflect the main actions expected from the grid operators by the government and society: knowledge development of grid operators; definition of standard solutions; and realization through investment. Installing smart meters in dwellings in the Netherlands is being prepared and will take several years. The working group sketches the options of the smart meter cupboard and the smart meter to support the developments under the Energy Transition (ET) and assigns actions.
