Myostatin is a member of the transforming growth factor-{beta} (TGF-{beta}) family and a strong negative regulator of muscle growth. Here, we present the crystal structure of myostatin in complex with the antagonist follistatin 288 (Fst288). We find that the prehelix region of myostatin very closely resembles that of TGF-{beta} class members and that this region alone can be swapped into activin A to confer signalling through the non-canonical type I receptor Alk5. Normal butylamine compounds are studied as salt-free wash reagents for degraded solvent used in PUREX process in spent fuel reprocessing. The solvent wash tests were carried out with two types of butylamine compounds, n-butylamine oxalate and n-butylamine bicarbonate, by counter-current mode using a small size mixer-settler composed of two 4-stage wash steps. Di-n-butyl phosphoric acid (HDBP), the main degradation product from TBP, was removed from real degraded solvent with decontamination factor of 2.5 {approx} 7.9. The plerocercoid stage of the tapeworm Spirometra mansonoides produces a functional analog of human growth hormone (hGH). Among the similarities between plerocercoid growth factor (PGF) and hGH is competition for the same receptors on rabbit liver membranes. To take advantage of this characteristic in a purification scheme for PGF, rabbit liver microsomes were solubilized in Triton X-100 and the hGH receptors were purified over an hGH affinity column. The purified receptors from six rabbit livers were coupled to Affi-Gel-10 to create a receptor affinity column which was used to purify PGF. Chromatography of crude PGF over the receptor column resulted in a 1044 fold increase in specific activity. Trastuzumab increases survival amongst women with human epidermal growth factorreceptor (HER)-2 receptor positive metastatic breast cancer, but maternal and fetal risks are associated with advanced disease and its treatment in pregnancy. We present a case of a primigravid with HER-2 positive metastatic breast cancer who received trastuzumab throughout pregnancy. She presented with cerebral metastases, requiring surgical decompression and resection. Reversible oligohydramnios developed during pregnancy. Fetal safety data on trastuzumab in pregnancy is limited, but case reports suggest a recurring pattern of (mostly reversible) oligohydramnios.

Structural instability of wild-type fibroblast growth factor (FGF)-1 and its dependence on exogenous heparin for optimal activity diminishes its potential utility as a therapeutic agent. Here we evaluated FGFC, an FGF1:FGF2 chimeric protein, for its receptor affinity, absolute heparin-dependence, stability and potential clinical applicability. Chlorinated organics are frequently found as harmful soil contaminants and persisted for extended periods of time. A novel approach, named pulsed corona discharge plasma (PCDP), was employed for the degradation of pentachlorophenol (PCP) in soil. Experimental results showed that 87% of PCP could be smoothly removed in 60 min. Increasing pulse voltage, enhancing soil pH, lowering humic acid (HA) in soil and reducing granular size of the soil were found to be favorable for PCP degradation efficiency. Oxidation and physical processes simultaneously contributed to PCP removal in soil and ozone was the main factor in PCDP treatment. Explicit analytical formulas are derived for the stress intensity factors at the tips of a main crack and of a microcrack for the two-dimensional case of a collinear microcrack. This configuration is used to derive an estimate of the toughness degradation due to microcracks linking up with an advancing main crack. The implications of this estimate for theoretical predictions of the toughening due to stress-induced microcracking are discussed.

The Steam Generator (SG) tubing degradation caused by corrosion and other age-related mechanisms continues to be a significant safety and cost concern for many Nuclear Power Plants (NPP). The understanding of the steam generator ageing mechanisms is the key to effective management of steam generator ageing and consists of the knowledge of steam generator materials and these one properties, stressors and operating conditions, like degradation sites and wear mechanisms. The principal types of corrosion are presented which can occur in CANDU steam generator. There are also presented the operation conditions, the specifications referring to the water chemistry and the construction materials of Steam Generator, the factors that have a great influence on the corrosion behaviour during the whole exploitation period of this equipment. A systematic study of the effects of environmental factors on nylon 66 and Kevlar 29 strength degradation in parachute components is being conducted at Sandia National Laboratories. It includes: (1) accelerated aging studies in air, inert environments, humidity, ozone, and smog; (2) a 25-year surveillance program of parachutes in a variety of natural climatic environments; (3) moisture absorption as a function of humidity; (4) effects of surface coatings normally applied to parachutes; and (5) development of nondestructive evaluation techniques which can be used to map mechanical properties over the entire parachute surface. The accelerated aging and moisture absorption studies show that air, humidity, and smog contribute to degradation. Chemiluminescence, gas chromatographic pyrograms, and uv spectroscopy show promise as nondestructive evaluation techniques.

Corrosion is said to be the most dominant factor which affects the physical life of ships and offshore structures. In order to secure the structural safety of ships and offshore structures, it is necessary to take measures to corrosion based on the prediction of condition of progressive corrosion. But such prediction is difficult because the behavior of corrosion`s progress is probabilistic. In this paper, probabilistic model, which could evaluate the degradation of coatings and the successive corrosion of steel members, was proposed in order to explain the tendency of the actual corrosion conditions of existing ships. Then, statistical inferences on the proposed model were made by applying the actual data on plate thickness measurements of Bulk Carriers` Cargo Hold. And validity of the proposed model was verified.
We examined the effect of the trophic protein, nerve growth factor (NGF), on cultures of fetal rat neostriatum and basal forebrain-medial septal area (BF-MS) to define its role in brain development. Treatment of cultures with NGF resulted in an increase in the specific activity of the cholinergic enzyme choline acetyltransferase (CAT) in both brain areas. CAT was immunocytochemically localized to neurons. In the BF-MS, NGF treatment elicited a marked increase in staining intensity and an apparent increase in the number of CAT-positive neurons. Moreover, treatment of BF-MS cultures with NGF increased the activity of acetylcholinesterase, suggesting that the cholinergic neuron as a whole was affected. The kinetics of benzoate degradation by the anaerobic syntrophic bacterium, Syntrophus buswellii, was studied in coculture with Desulfovibrio strain G11. The threshold value for benzoate degradation was dependent on the acetate concentration with benzoate threshold values ranging from 2.4 [mu]M at 20 mM acetate to 30.0 [mu]M at 65 mM acetate. Increasing acetate concentrations also inhibited the rate of benzoate degradation with a apparent K[sub i] for acetate inhibition of 7.0 mM. Lower threshold values were obtained when nitrate rather than sulfate was the terminal electron acceptor. These data are consistent with a thermodynamic explanation for the threshold, and suggest that there is a minimum Gibbs free energy value required for the degradation of benzoate. In canine and human studies, the designer natriuretic peptide CD-NP demonstrated more desirable therapeutic potential than recombinant B-type NP (BNP), which is known as nesiritide and is approved for treatment of acute decompensated heart failure. The performance of two-cell planar solid oxide fuel cell stacks using coal syngas, with and without hydrogen sulfide (H{sub 2}S), was studied. All cells were tested at 850 C with a constant current load of 15.2 A (current density of 0.22 A cm{sup -2} per cell) and 30% fuel utilization. The H{sub 2}S injection immediately and significantly affected the power degradation of the stack system regardless of the carrier fuel. Results for the test with only H{sub 2} and N{sub 2} in the presence of H{sub 2}S (119-120 ppm) indicated that the power decay and area-specific resistance (ASR) degradation values were lower than those for the tests where simulated syngas containing CO and increased water content was used. Neuron-restrictive silencer factor (NRSF) binds its consensus element to repress the transcription of various genes. The dominant-negative form (dnNRSF) has a hypertrophic effect on cardiogenesis through an unidentified mechanism. We examined the involvement of transient receptor potential (TRP) channel proteins, using transgenic mice overexpressing dnNRSF (dnNRSF mice). However, materials and components have been seriously degraded due to improper storage, protection, or layup, at facilities under construction as well as those with operating licenses. A C{sub 60} {sup +} primary ion source has been coupled to an ion microscope secondary ion mass spectrometry (SIMS) instrument to examine sputtering of silicon with an emphasis on possible application of C{sub 60} {sup +} depth profiling for high depth resolution SIMS analysis of silicon semiconductor materials. Unexpectedly, C{sub 60} {sup +} SIMS depth profiling of silicon was found to be complicated by the deposition of an amorphous carbon layer which buries the silicon substrate. Sputtering of the silicon was observed only at the highest accessible beam energies (14.5 keV impact) or by using oxygen backfilling. Bisaminoethanethiol (BAT) ligands with various gem-dimethyl and amide groups were prepared, and the corresponding neutral Tc-99m complexes were prepared and evaluated for their relative stabilities by ligand-exchange reactions. It was demonstrated that technetium complexes containing gem-dimethyl substituents have higher lipophilicities, whereas those with an amide group possess greater stability, which enhances ligand-exchange reaction. The most interesting observation was that the brain uptake in rats is not determined only by lipophilicity. Apparently, Tc-99m complexes with an amide functional group display lower brain uptakes in rats compared to those without an amide group. The brain uptake was strongly influenced by substituents on the BAT ligand. These factors are critically important and should be taken into consideration when designing Tc-99m-labeled agents for CNS receptor imaging.
The glass gene is required for proper photo-receptor differentiation during development of the Drosophila eye glass codes for a DNA-binding protein containing five zinc fingers that we show is a transcriptional activator. A comparison of the sequences of the glass genes from two species of Drosophila and a detailed functional domain analysis of the Drosophila melanogaster glass gene reveal that both the DNA-binding domain and the transcriptional-activation domain are highly conserved between the two species. Analysis of the DNA-binding domain of glass indicates that the three carboxyl-terminal zinc fingers alone are necessary and sufficient for DNA binding. We also show that a deletion mutant of glass containing only the DNA-binding domain can behave in a dominant-negative manner both in vivo and in a cell culture assay that measures transcriptional activation. One of the significant factors of neutron radiographic image degradation is scattering blur from the object. A practical method is described to enhance image quality by eliminating the overlapping of scattered thermal neutrons component from the objects in ETRR-2 neutron radiography facility, using aluminum Gd-coated antiscatter grid. The MCNP code was used to determine the optimum grid dimensions that will reduce the scattered thermal neutrons from the object. An experiment was performed to determine the optimum grid height and irradiation time that gives the best image with acceptable geometric unsharpness. Using the MCNP code it was found that 97% of the scattered neutrons were removed by the grid. The wall dimensions and Gd coating are so small that the facility resolution cannot detect the image pattern superposition on the film.
Ribbon parachute fabrics were tested during development and production of a 24 ft parachute, and after 2.5 years ambient exposure at several storage sites. The effects of sewing, packing, and parachute deployment on material strength were obtained in the 1000 lb Kevlar ribbon after moisture exposure during packing, and when packed parachutes absorbed moisture or were thermal cycled. Individual yarn samples from the low strength ribbons exhibited little change in strength indicating that the material itself had not degraded. Factors leading to the change in ribbon strength are discussed. Considering all materials, there have been no indications to date of any uncontrollable changes in material strength that would limit the use of Kevlar in parachute applications.

The tritium distribution ratio in the system of U - 30% TBP/n-dodecane(nDD) - HNO[sub 3] [minus]H[sub 2]O was measured in batch-wise experiments. The tritium decontamination factor in a tritium scrubbing step was measured in chemical flow sheet experiments using a 6 stage mixer-settler having an internal recycle system of aqueous solution in each stage. A clone encoding a human D{sub 2} dopamine receptor was isolated from a pituitary cDNA library and sequenced. The deduced protein sequence is 96% identical with that of the cloned rat receptor with one major difference: the human receptor contains an additional 29 amino acids in its putative third cytoplasmic loop. Southern blotting demonstrated the presence of only one human D{sub 2} receptor gene. Two overlapping phage containing the gene were isolated and characterized. DNA sequence analysis of these clones showed that the coding sequence is interrupted by six introns and that the additional amino acids present in the human pituitary receptor are encoded by a single exon of 87 base pairs. Bisaminoethanethiol (BAT) ligands with various gem-dimethyl and amide groups were prepared, and the corresponding neutral Tc-99m complexes were prepared and evaluated for their relative stabilities by ligand-exchange reactions. It was demonstrated that technetium complexes containing gem-dimethyl substituents have higher lipophilicities, whereas those with an amide group possess greater stability, which enhances ligand-exchange reaction. The most interesting observation was that the brain uptake in rats is not determined only by lipophilicity. Apparently, Tc-99m complexes with an amide functional group display lower brain uptakes in rats compared to those without an amide group. The brain uptake was strongly influenced by substituents on the BAT ligand. These factors are critically important and should be taken into consideration when designing Tc-99m-labeled agents for CNS receptor imaging.

The potential for energy generation from biogas through anaerobic digestion is approximately 30 MW. On-farm manure-based systems represent the most likely scenario for the development of anaerobic digestion in British Columbia in the near future. Certain materials, especially Sn, passivate the rare earth-exchanged Y zeolite (REY) used in petrochemical fluid-cracking catalysts against vanadium degradation caused by V impurities in the feed oil. The mechanism of passivation was investigated here from the standpoint of high-temperature oxide acid-base reaction; i.e., where the controlling factors were considered to be Lewis acid-base reactions between V{sub 2}O{sub 5}, the RE oxides, SnO{sub 2}, etc. Molten salt tests at 680{degree}C showed SnO{sub 2}, presumably because of its acidic nature, to be essentially nonreactive with V{sub 2}O{sub 5} or Na{sub 2}O-V{sub 2}O{sub 5} compounds. A hypothesis was developed to explain how the passivation effect by Sn might result from the unique resistivity of SnO{sub 2} to reaction with V{sub 2}O{sub 5}.

Platelets from dogs with Basset Hound Hereditary Thrombopathy (BHT) display a thrombasthenia-like aggregation defect but have been shown to have normal amounts of platelet membrane glycoproteins IIb and IIIa (GP IIb-IIIa). In order to investigate the possibility of a functionally abnormal GPIIb-IIIa complex, which might be unable to bind fibrinogen after stimulation, fibrinogen binding in BHT was evaluated. Two canine fibrinogen preparations were used, one from BHT dogs and one from normal control dogs, as well as a human fibrinogen preparation. Platelets from BHT and normal dogs were activated with 1 x 10/sup -5/M ADP in the presence of /sup 125/I-labeled fibrinogen and the surface bound radioactivity quantitated. For all fibrinogen preparations, the amount of fibrinogen bound by BHT platelets was not significantly different than that bound by normal dog platelets. However, since Trastuzumab (Herceptin) is a widely used monoclonal antibody as cancer therapy agent for tumors over-expressing HER-2, there is a need for an efficient way to detect host-generated antibodies against HER-2 without the confounding effect of Herceptin. Streptococcus pneumoniae colonizes the mucosal surface of the human upper respiratory tract. A colonization event is gradually cleared through phagocytosis by monocytes/macrophages that are recruited to the airway lumen. Here, we sought to define the bacterial and host factors that promote monocyte/macrophage influx and S. pneumoniae clearance using intranasal bacterial challenge in mice. We found that the recruitment of monocytes/macrophages required their expression of the chemokine receptor CCR2 and correlated with expression of the CCR2 ligand CCL2. The resistance of six tropical hardwoods to fungal degradation by two wild-type strains of Phanerochaete chrysosporium Burdsall was investigated using vermiculite burial and wood-block weight loss techniques. Radiation-induced wood-polymer composites (WPC), based on two hardwoods Ramin and Rubberwood with methyl methacrylate, were prepared, and samples were also exposed to the wood-rotting fungus. A significant improvement in resistance to fungal decay was observed in the WPC. Scanning-electron micrographs of the two woods and their composites after fungal degradation are presented and discussed.
The authors used radioligand binding and studies of cell contraction to characterize muscarinic receptors on dispersed smooth muscle cells from rabbit proximal and distal colon. Cells obtained after serial incubations in collagenase were used to measure binding of tritiated quinuclidinyl benzilate (["3H]QNB). The performance of anaerobic biological process is heavily process conditions dependent. In this study, an attempt has been made to investigate the influence of process conditions like temperature, sludge age and hydraulic retention time (HRT) on the efficiency of an upflow anaerobic sludge blanket (UASB) reactor and upflow anaerobic sludge filter (UASF) to treat combined industrial wastewater. Reactors were operated at easing ambient temperatures (38, 30, 20 and 14 deg. C) and correspondingly increasing sludge ages (60, 90, 120 and 150 days). At temperature 38 deg. During the Bruce A Nuclear Generating Station (BNGS-A) Unit 1 1997 planned outage, a dew point search method identified a leak in one steam generator(SG) tube. Subsequently, the tube was inspected with all available eddy current probes and removed for examination. The initial inspection results and metallurgical examination of the removed tube confirmed that the leak was due to intergranular attack/stress corrosion cracking (IGA/SCC) emanating from the secondary side of the tube at the top of the tubesheet location. Subsequently, eddy current and ultrasonic indications were found at the top of the tubesheet of other Alloy 600 SG tubes. To investigate the source of the indications and to validate the inspection probes, sections of 40 tubes with various levels of damage were removed. Since the integrity of the containment penetrations was confirmed under accident management (AM) conditions in the former test, failure criteria tests and aerosol trapping tests were carried out using low-voltage modules and flange gaskets of an actual plant under severe accident (SA) conditions, without AM. The safety margin for failure temperature of the penetrations and the credit for fission product (FP) aerosol trapping effect along the leakage paths of the degraded penetrations were evaluated in the present tests. Failure temperature of the penetrations ranged from 270 to 300degC for low-voltage modules and 300 to 350degC for flange gaskets under 0.4 to 1.0 MPa conditions. In this study, we focus on the effects of the cultivation of Italian ryegrass on the soil microbes collected at oil showings, which were expected to have a variety of crude oil degradable microorganisms. We evaluated the number of crude oil degradable microorganism, microbial activity, microflora using the PCR-DGGE method and the change in the concentration of crude oil in the soil. Using microslices from immature (10 day) and adult chicken forebrain prepared after decapitation, we have examined functional properties of NMDA and AMPA receptors by measuring agonist-induced uptake of "4"5Ca"2"+ . The rate and extent of NMDA induced "4"5Ca"2"+ accumulation decreased during maturation with no change in EC_5_0. Somatostatin receptor scintigraphy (SRS) is a valuable method for the detection of somatostatin receptor-positive lesions. Most gastrinomas (over-)express the somatostatin receptor subtype 2 which can be targeted by In-111 labeled Octreotide. Different studies show a high sensitivity of SRS for the localization and staging of gastrinomas. SRS seems to be superior to other non-invasive imaging modalities and has been proven to significantly contribute to patient management. However, the sensitivity depends on the size and exact localization of the tumors. Smaller lesions and lesions located in the duodenum show a significantly lower sensitivity. In any case, SRS belongs to the routine imaging procedure for gastrinomas for localization and staging and can also be used for evaluation of the tumor progression. (author)

Somatostatin receptor expression has been demonstrated on a number of plasma cell lines. Therefore, we questioned whether somatostatin receptor scintigraphy (SRS) can be used to demonstrate in vivo multiple myeloma (MM) activity. SRS was performed in newly diagnosed (n = 9) or relapsing (n = 18) MM patients or in patients with localized plasmacytoma (n = 2). The results were compared with radiographic findings. A positive SRS was demonstrated in 44% of the newly diagnosed patients, in 83% of the relapsed patients and in both patients with plasmacytoma. The endothelin-B (ETB) receptor is a G-protein-coupled receptor that binds endothelin ligands and is essential for the development of epidermal melanocytes and enteric neurons. The specific receptor sites and the endocytosis of transferrin (Tf) are evidenced in a cell line (HT-29) derived from a human colic adenocarcinoma by means of "1"2"5I radiolabeled Tf. The receptor density is studied in undifferentiated (UD) or differentiated (D) cells with respective doubling times of about 24 hours and 46 hours. The number of binding sites for Tf is 162,000 (K_d = 7.8 nmol/l in ND cells and 68,000 (K_d = 7.40 nmol/l) in D cells. The distribution between the Tf bound to the cell surface and the internalized Tf is investigated by elimination of Tf bound to the surface by an acid wash method. The intracellular cycle of Tf seems to be characterized by a slower kinetics in UD cells. The high density of Tf receptor sites in HT-29 UD cells should allow the detection or the treatment of highly evolutive colic adenocarcinoma by means of Tf.
