Numerical results on the translocation of long biopolymers through mid-sized and wide pores are presented. The simulations are based on a novel methodology which couples molecular motion to a mesoscopic fluid solvent. Thousands of events of long polymers (up to 8000 monomers) are monitored as they pass through nanopores. Comparison between the different pore sizes shows that wide pores can host a larger number of multiple biopolymer segments, as compared to smaller pores. The simulations provide clear evidence of folding quantization in the translocation process as the biopolymers undertake multi-folded configurations, characterized by a well-defined integer number of folds. It is anticipated that early in the next century manned missions into outer space will occur, with a mission to Mars scheduled between 2015 and 2020. However, before such missions can be undertaken, a realistic estimation of the potential risks to the flight crews is required. One of the uncertainties remaining in this risk estimation is that posed by the effects of exposure to the radiation environment of outer space. Although the composition of this environment is fairly well understood, the biological effects arising from exposure to it are not. Events detected in a shielded plastic scintillation counter occurring in the 26 microsec preceding the arrival of an extensive air shower at ground level with local electron density or = 20 m to the -2 power and the 240 microsec after its arrival have been studied. No significant excess of events (tachyons) arriving in the early time domain have been observed in a sample of 11,585 air shower triggers.

Maintenance of genomic integrity in embryonic cells is pivotal to proper embryogenesis, organogenesis and to the continuity of species. Cultured mouse embryonic stem cells (mESCs), a model for early embryonic cells, differ from cultured somatic cells in their capacity to remodel chromatin, in their repertoire of DNA repair enzymes, and in the regulation of cell cycle checkpoints. Using 129XC3HF1 mESCs heterozygous for Aprt, we characterized loss of Aprt heterozygosity after exposure to ionizing radiation. We report here that the frequency of loss of heterozygosity mutants in mESCs can be induced several hundred-fold by exposure to 5-10Gy of X-rays. Abstract Ultrastructural studies have shown that the formation of pigment glands in Gossypium hirsutum L. leaves is a lysigenous process, originating from a cluster of cells in the ground meristem. Various techniques were used here to investigate whether programmed cell death (PCD) plays a critical role in this developmental process. Nuclei of internal cells in the pigment gland-forming tissue were TUNEL-positive and DAPI-negative, suggesting that DNA cleavage is an early event and complete DNA degradation is a late event. Smeared bands and a lack of laddering after gel electrophoresis indicate that DNA cleavage is random. The effect of a complexon therapy scheme including early (in 1 h) administration of Na/sub 3/Ca DTPA and subsequent administration of Na/sub 3/Zn DTPA or Na/sub 3/Ca DTPA at a dose of 25 ..mu..Cimol/kg/day by three 2-week courses (5 times a week) with 2-week interruptions was studied in experiments on 541 male rats after intraperitoneal administration of /sup 239/Pu citrate complex (95 kBq/kg). The treatment resulted in a 3-fold lessening of the content of /sup 239/Pu and absorbed doses in the skeleton, a significant prolongation of the mean survival time (MST) from 452 to 593 days (Na/sub 3/Zn DTPA) and 643 days (Na/sub 3/Ca DTPA), and in a decrease of the osteosarcoma incidence from 76.4 to 32.6-41.2%. The ratio of osteosarcomas per 1 Gy retained in rats (0.076-0.083 %) did not differ from that in untreated animals.

Patients with acute myocardial infarction (AMI) and diabetes mellitus, as well as patients admitted with elevated blood glucose without known diabetes, have impaired outcome. Therefore intensive glucose-lowering therapy with insulin (IGL) has been proposed in diabetic or hyperglycaemic patients and has been shown to improve survival and reduce incidence of adverse events. The primary aims of the BESS-Polar program are precise measurements of the low-energy antiproton spectrum and search for cosmologically significant antimatter, which would provide new clues to understand the early Universe. The second flight (BESS-Polar II) over Antarctica was successfully carried out in December 2007 - January 2008. We performed 24.5 days scientific observation just at the solar minimum. The payload worked well during the flight and 4.7 billion cosmic-ray events were collected, which corresponds to 10-20 times statistics of the BESS data taken in the previous solar minimum period (1995 and 1997). Based on the BESS-Polar II data, we will present recent preliminary results of cosmic-ray antiproton measurements and sensitive search for antimatter. (authors)

The carbonate rocks of the Lower Ordovician Majiagou Formation represent important hydrocarbon reservoirs in the Ordos basin in the western part of the North China plate. Cores and thin sections from more than 60 wells were examined. The main reservoir in the Majiagou Formation consists of fine crystalline dolomite, which was altered by meteoric water during an extended subaerial exposure. Three phreatic zones have been identified in the Majiagou Formation in the northern part of the basin. Oxygen isotopes and fluid inclusion microthermometry data indicates that the porosity developed during the early karst event was considerably reduced by the late-stage, coarse crystalline calcite cements that appear to be related to hydrothermal fluids during deep burial. It was concluded that the main factors that determine the porosity and permeability of these carbonate reservoirs are dolomitization, karstification, and calcite cementation.

However, in underground coal mines, the CO and CO2 concentrations and their related fire ratios may be affected by mine ventilation. In this study, CO and CO2 emissions from spontaneous heating of a U.S. coal sample were evaluated in an isothermal oven under different airflow ventilation rates ranging from 100 to 500cm^3/min. The Plutonium-Uranium Extraction (PUREX) Plant, conceived during the early Cold War years, was a vehicle to increase significantly US nuclear weapons production capacity. The original PUREX Plant was a concrete rectangle 1,005 feet long and 61.5 feet wide. The shielding capacity of the concrete was designed so that personnel in non-regulated service areas would not receive radiation in excess of 0.1 millirem per hour. This report discusses the design of the PUREX Plant, the production chronology, projects and equipment changes, equipment decontamination and reuse, waste management, and contamination events that have occurred during the operation of the plant. We revisit our original papers on the burst mode of accretion by incorporating a detailed energy balance equation into a thin-disk model for the formation and evolution of circumstellar disks around low-mass protostars. Our model includes the effect of radiative cooling, viscous and shock heating, and heating due to stellar and background irradiation. Following the collapse from the prestellar phase allows us to model the early embedded phase of disk formation and evolution. During this time, the disk is susceptible to fragmentation, depending upon the properties of the initial prestellar core. Globally, we find that higher initial core angular momentum and mass content favors more fragmentation, but higher levels of background radiation can moderate the tendency to fragment. Quantitative radiolabeled tracer techniques were used to assess glomerular filtration rate (GFR) and regional tissue vascular clearance of plasma 131I-bovine serum albumin (BSA) in five groups of male Sprague-Dawley rats: nondiabetic controls, streptozotocin-diabetic rats, nondiabetic rats fed a 50% galactose diet, diabetic rats treated with sorbinil (an aldose reductase inhibitor), and galactose-fed rats treated with sorbinil. Sorbinil was added to the diet to provide a daily dose of approximately .2 mmol/kg body weight. Early white matter (WM) injury affects brain maturation in preterm infants as revealed by diffusion tensor imaging and volumetric magnetic resonance (MR) imaging at term postmenstrual age (PMA). The aim of the study was to assess quantitatively brain maturation in preterm infants with and without milder forms of WM damage (punctate WM lesions, PWML) using conventional MRI. Brain development was quantitatively assessed using a previously validated scoring system (total maturation score, TMS) which utilizes four parameters (progressive myelination and cortical infolding, progressive involution of glial cell migration bands and germinal matrix tissue). PWML were defined as foci of increased signal on T1-weighted images and decreased signal on T2-weighted images with no evidence of cystic degeneration. Early effects of 100 Kilorads of X-rays on muscle cell membrane properties have been measured in sartorius muscles from Leptodactylus ocellatus. Threshold strength for rectangular current pulses increased 10% after irradiation, and action potential propagation velocity decreased 10%. Passive membrane parameters were calculated from potential responses to sub-threshold current pulses, assuming conventional cable theory. Specific membrane conductance increased to 18% after irradiation, membrane capacitance increased 14%, and length constant decreased 10% but membrane time constant was unchanged. Cell diameter decreased 5%, and resting membrane potential decreased 8%. Membrane parameters during an action potential were also evaluated by the phase-plane and current-voltage plot techniques. However, some analyses of accident initiators during low power, shutdown, and other modes of plant operation other than full power have been performed. These studies as well as the Chernobyl accident and recent operating experience at U.S. pressurized water reactors suggested that risks during low power and shutdown could be significant. As such, the analysis of the frequencies, consequences, and risks of these accidents was identified as one task in the Nuclear Regulatory Commission staff's study of the implications of the Chernobyl accident to U.S. commercial nuclear power plants. The Simplified Boiling Water Reactor (SBWR) designed by General Electric is a natural circulation reactor with enhanced safety features for potential accidents. It has a strong coupling between power and flow in the reactor core, hence the neutronic coupling with thermal-hydraulics is specially important. The potential geysering instability during the early part of a SBWR startup at low flow, low power and low pressure is of particular concern. The RAMONA-4B computer code developed at Brookhaven National Laboratory (BNL) for the SBWR has been used to simulate a SBWR startup transient and evaluate its stability, using a simplified four-channel representation of the reactor core for the thermal-hydraulics. The murine 3T3-L1 fibroblast under appropriate incubation conditions differentiates into an adipocyte phenotype. This 3T3-L1 adipocyte exhibits many of the morphologic, biochemical, and insulin-responsive features of the normal rodent adipocyte. Using quantitative electron microscopic (EM) autoradiography we find that, when /sup 125/I-insulin is incubated with 3T3-L1 adipocytes, the ligand at early times of incubation localizes to the plasma membrane of the cell preferentially to microvilli and coated pits. When the incubation is continued at 37 degrees C, /sup 125/I-insulin is internalized by the cells and preferential binding to the villous surface is lost. With the internalization of the ligand, two intracellular structures become labeled, as determined by the method of hypothetical grain analysis. Twenty undergraduate students participated in an elaborative learning test to evaluate the relationship between electrical brain activity and subsequently recalled and not-recalled words. Data collected from the midline (Fz, Cz, Pz) and lateral scalp sites (F3, F4, C3, C4, P3, P4) were analysed. The difference between event-related potentials (ERPs) elicited by subsequently recalled and not-recalled words, the ERP memory effect, was evaluated for each portion (primacy, plateau and recency) of the serial-position curve (SPC). We compared peak amplitudes for the P1, N1, P2, N400, P3 and frontal positive slow wave (FPSW) components. The electrophysiological data support the hypothesis that different mechanisms underlie primacy and recency effects during free recall paradigms. The Ordos Basin is a typical lapped basin, including three sequences of strata: early Paleozoic, late Paleozoic and Mesozoic, with a total thickness of 4000-6000m. Impermeable sealing beds are well developed at the top and base of the upper Paleozoic sequence, separating it from the Mesozoic and the lower Paleozoic strata to form an independent petroleum system. In this petroleum system, the source rocks are widely distributed coals and dark mudstones occurring in the Carboniferous-lower Permian coal measures, with a thickness of 10-15 and 40-60m, respectively. The reservoirs are mainly early Permian tight sandstones, mostly with a porosity of 4-8% and a permeability of 0.1-1.0x10{sup -3}{mu}m{sup 2}. The regional cap rock is a 100-150m thick mudstone in the upper Permian strata. A study is made of the metamorphism of coals in Kuznetsk Basin. To interpret the stages of coalification, measurements were used of the indicators of vitrinite along the hinges of folds, graphic plottings of isometamorphism of coals, analysis of interrelationships of the isolines of coal metamorphism with isopachites covering their deposits. The elevation of the coal mass of Kuzbass in the post-inversion time was accompanied by the formation of steep linear folds and longitudinal bending of their hinges. As a consequence of this, the hinges of the folds in beds of the same name occurred at different depths from the modern surface, however coal metamorphism along the hinges has not changed. In the Kuznetsk Basin there are no traces of post-inversion coalification.

Apatite fission track analysis, vitrinite reflectance data, and K-Ar dating of Permian-Carboniferous and Mesozoic core samples have been successfully integrated to reconstruct the thermal and tectonic history of the Ordos basin, China. Apatite fission track ages of Carboniferous-Jurassic sedimentary rocks range between 3 and 137 Ma, and are significantly younger than the stratigraphic ages. Confined fission track lengths demonstrate exclusively mixed length distribution, indicating complex thermal history. The data suggest that the samples must have all experienced higher paleotemperatures in the past. Stargardt`s disease (MIM 248200), originally described in 1909, is an autosomal recessive condition of childhood, characterized by a sudden and bilateral loss of central vision. Typically, it has an early onset (7 to 12 years), a rapidly progressive course and a poor final outcome. The central area of the retina (macula) displays pigmentary changes in a ring form with depigmentation and atrophy of the retinal pigmentary epithelium (RPE). Perimacular yellowish spots, termed fundus flavimaculatus, are observed in a high percentage of patients. We have recently reported the genetic mapping of Stargardt`s disease to chromosome 1p13. The burning characteristics of fuel droplets containing nano and micron-sized aluminum particles were investigated. Particle size, surfactant concentration, and the type of base fluid were varied. In general, nanosuspensions can last much longer than micron suspensions, and ethanol-based fuels were found to achieve much better suspension than n-decane-based fuels. Five distinctive stages (preheating and ignition, classical combustion, microexplosion, surfactant flame, and aluminum droplet flame) were identified for an n-decane/nano-Al droplet, while only the first three stages occurred for an n-decane/micron-Al droplet. For the same solid loading rate and surfactant concentration, the disruption and microexplosion behavior of the micron suspension occurred later with much stronger intensity. The Lake Superior region (Wisconsin, the Upper Peninsula of Michigan, and Minnesota) contains 41 Precambrian crystalline rock complexes comprising 64 individual but related rock bodies with known surface exposures. Each complex has a map area greater than 78 km/sup 2/. About 54% of the rock complexes have areas of up to 500 km/sup 2/, 15% fall between 500 km/sup 2/ and 1000 km/sup 2/, 19% lie between 1000 km/sup 2/ and 2500 km/sup 2/, and 12% are over 2500 km/sup 2/. Crystalline rocks of the region vary widely in composition, but they are predominantly granitic. Daya Bay Nuclear Power Plant is put into commercial operation in 1994 Until 1996, there are 368 human error events in operating and maintenance area, occupying 39% of total events. These events occurred mainly in the processes of maintenance, test equipment isolation and system on-line, in particular in refuelling and maintenance. The author analyses root causes for human errorievents, which are mainly operator omission or error procedure deficiency; procedure not followed; lack of training; communication failures; work management inadequacy. The protective measures and treatment principle for human error events are also discussed, and several examples applying them are given. Finally, it is put forward that key to prevent human error event lies in the coordination and management, person in charge of work, and good work habits of staffs.

The effect of insulin on protein biosynthesis was examined in differentiated 3T3-L1 and 3T3-F442A adipocytes. Insulin altered the relative rate of synthesis of specific proteins independent of its ability to hasten conversion of the fibroblast (preadipocyte) phenotype to the adipocyte phenotype. Although more than one pattern of response to insulin was observed, the authors focused on the induction of a Mr 33,000 protein which was identified as the glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase (GAPDH). Exposure of 3T3 adipocytes to insulin throughout differentiation specifically increased GAPDH activity and protein content by 2- to 3-fold as compared to 3T3 adipocytes differentiated in the absence of insulin. These changes in enzyme activity and content could be accounted for by a 4-fold increase in the relative rate of synthesis of GAPDH and a 9-fold increase in hybridizable mRNA levels. In this study we examine the effect of climate, geology, land use, flood types and initial soil moisture conditions on the distribution functions of the event runoff coefficients for a set of 14 mountainous catchments located in the eastern Italian Alps, ranging in size from 7.3 to 608.4km2. The GntR-like protein NorG has been shown to affect Staphylococcus aureus genes involved in the resistance to quinolones and ?-lactams, such as those encoding the NorB and AbcA transporters. To identify the target genes regulated by NorG, we carried out transcriptional profiling assays using S. aureus RN6390 and its isogenic norG::cat mutant. Our data showed that NorG positively affected the transcription of global regulators mgrA, arlS, and sarZ. The three putative drug efflux pump genes most positively affected by NorG were the NorB efflux pump (5.1-fold), the MmpL-like protein SACOL2566 (5.2-fold), and the BcrA-like drug transporter SACOL2525 (5.7-fold). The addition of paraquat (methyl viologen) to a growing culture of Escherichia coli K-12 led within 1 hr to a 10- to 20-fold increase in the level of endonuclease IV, a DNase for apurinic/apyrimidinic sites. The induction was blocked by chloramphenicol. Increases of 3-fold or more were also seen with plumbagin, menadione, and phenazine methosulfate. H_2O_2 produced no more than a 2-fold increase in endonuclease IV activity. The following agents had no significant effect: streptonigrin, nitrofurantoin, tert-butyl hydroperoxide, #gamma# rays, 260-nm UV radiation, methyl methanesulfonate, mitomycin C, and ascorbate. Paraquat, plumbagin, menadione, and phenazine methosulfate are known to generate superoxide radical anions via redox cycling in vivo. The presence of washed or unwashed cellophane alone or together with a bleomycin, mitomycin C or hydrochlorothiazide, ('Esidrex') showed no appreciable effect on survival of either unirradiated or irradiated conidia. Irradiation for a period of 20min reduced the survival of conidia to 20%. The growth of irradiated conidia in the presence of bleomycin, mitomycin C or Esidrex is associated with a 2- to 3-fold increase in the frequency of gene convertants, but was not accompanied by an increase in point mutants. When conidia were grown on cellophane but otherwise treated as before the frequency of gene convertants was increased 8-fold, but induction of point mutants was negligible. This effect was the same for irradiated and unirradiated conidia. The environment created by the cellophane in contract with the medium appears to affect the action of each of the three compounds synergistically. (author).

A multiphase origin of the Cu-Co ores in the western part of the Lufilian fold-and-thrust belt in Central Africa is proposed based on literature, satellite image interpretations and petrographic and fluid inclusion analyses on samples from the stratiform mineralization of Kamoto and Musonoi (DR Congo). The various mineral occurrences in the Katanga Copperbelt can be classified in distinct categories: stratiform, supergene enrichment and vein-type. The stratiform mineralization form the largest group and can be found mainly in Lower Roan (R-2) rocks, which can be identified as ridges on satellite imagery. By collecting all diagrams up to O(N{sup -2}) of the imaginary part of the self-energy of the conduction electrons, the sum of those is shown to be given by a form proportional to {omega}{sup 2} + {pi}{sup 2}T{sup 2} up to O(N{sup -2}) in the single-channel model. On the other hand, the imaginary part of the self-energy of O(N{sup -1}) in the multichannel model has more singular frequency-/temperature-dependence, so the system is regarded as non-Fermi liquid. Iodo(/sup 14/C)antipyrine autoradiography was used to measure gastric mucosal blood flow in anesthetized rats and to study regional distribution. Blood flows of 61 +/- 8 ml.100 g-1.min-1 (means +/- SE) in corpus and 84 +/- 9 ml.100 g-1.min-1 in antral mucosa compared well with previously reported measurements by hydrogen clearance. Blood flow in the crests of corpus mucosal folds was significantly higher than in the valleys between folds, indicating that the greater susceptibility of the former areas to acute injury, documented in several studies, is not associated with a perfusion defect in the resting stomach. Corpus mucosal blood flow was also higher in the side walls of the stomach than in the greater curvature region, and in distal than in proximal locations. No systematic regional variations within antral mucosa were demonstrated.

The technqiues include a new approach to empirical travel-time calibration that simultaneously fits data from multiple stations and events, using a generalization of the kriging method, and predicts travel-time corrections for arbitrary event-station paths. Combined sewer overflows (CSOs) have been recognised as one of the serious sources of pollution to the water environment during rain events, although field surveys to investigate the effect of their magnitude and duration on receiving waters have been very limited. The fates of enteric viruses (norovirus G1, G2, enteroviruses) and coliforms were determined in the wastewater treatment plant on a fine day and on a rainy day. Not all microorganisms were reduced in the primary treatment, but were reduced in the secondary treatment. Occurrences of enteric viruses and levels of coliforms were surveyed in the receiving coastal area after a CSO event, with the profiles of the enteric viruses in the coastal seawater being almost at the same positive ratio for 4 d after the CSO event. A program of tests were conducted in July 1986 at the Fast Flux Test Facility (FFTF) to demonstrate that the reactor could withstand a prototypic loss of flow (LOF) without scram without sustaining fuel damage. The reactor was taken to powers up to 50%, and the main primary coolant pump motors were tripped without scramming the control rods. This paper summarizes the analyses performed to demonstrate the maintenance of redundant protection for all design events as well as potential new events introduced by the test. The analyses focused on the following consequences: (1) unexpected test behavior; (2) transient overpower event during the test; and (3) LOF event during the test.

A position-sensitive event-counting electronic readout system for microchannel plates (MCPs) is described that offers the advantages of high spatial resolution and fast time resolution. The technique relies upon a four-quadrant electron-collecting anode located behind the output face of the microchannel plate, so that the electron cloud from each detected event is partly intercepted by each of the four quadrants. The relative amounts of charge collected by each quadrant depend on event position, permitting each event to be localized with two ratio circuits. A prototype quadrant anode system for ion, electron, and extreme ultraviolet imaging is described. Hyaluronate (HA) distribution patterns were examined in the cranial mesenchyme underlying the mesencephalic neural folds of mouse embryos maintained in roller tube culture. Using standard image-processing techniques, the digitized images of Alcian blue-stained or 3H-glucosamine-labeled sections digested with an enzyme specific for HA, were subtracted from adjacent, undigested sections. The resultant difference picture images (DPI) accurately depicted the distribution of stained or labeled HA within the cranial mesenchyme. 3H-glucosamine-labeled HA was distributed uniformly throughout the cranial mesenchyme as 12, 18, and 24 hr of culture. By contrast, the mesenchyme was uniformly stained with Alcian blue at 12 hr, but stain intensity decreased in the central regions of the mesenchyme at 18 and 24 hr. The optimal concentrations of nutrient medium components, aeration conditions, and pH providing for maximum biomass yields, as well as fumarase and L-aspartase activities, during submerged cultivation of Erwinia sp. were determined. The data showed that different concentrations of carbon source (molasses) and pH of the nutrient medium were required to reach the maximum fumarase and L-aspartase activities. Calculations performed by application of the additive lattice model suggested that the combination of these optimized factors would result in 3.2-, 3.4-, and 3.8-fold increases as compared to the experimental means in Erwinia sp. biomass, and L-aspartase and fumarase activities, respectively. The authors have shown previously that the domain recognizing receptors on activated human platelets is located on the human fibrinogen {gamma} chain between residues 400 and 411. To study the correlation between the structure of this segment of the {gamma} chain and its reactivity toward receptors on ADP-activated human platelets, they designed a series of analogues containing replacements at 9 out of 12 positions. A double substitution of the normal His{sup 400}-His{sup 401} sequence by Ala-Ala reduced the inhibitory potency of the dodecapeptide 3-fold. When Lys{sup 406} was replaced by Arg, the inhibitory potency of the dodecapeptide decreased 15 times. For the first time a segmented n-type HPGe detector was operated directly submerged in liquid nitrogen over a long period. As this kind of detector is envisioned to be used in GERDA phase II, it was operated with a low mass signal cable with snap-contacts and mounted in a low-mass copper holder. The detector performance was stable over 146 days, indicating that neither detector nor contacts deteriorated.

Large rigid-body domain movements are critical to GroEL-mediated protein folding, especially apical domain elevation and twist associated with the formation of a folding chamber upon binding ATP and co-chaperonin GroES. Here, we have modeled the anisotropic displacements of GroEL domains from various crystallized states, unliganded GroEL, ATP?S-bound, ADP-AlFx/GroES-bound, and ADP/GroES bound, using translation-libration-screw (TLS) analysis. Remarkably, the TLS results show that the inherent motions of unliganded GroEL, a polypeptide-accepting state, are biased along the transition pathway that leads to the folding-active state. In the ADP-AlFx/GroES-bound folding-active state the dynamic modes of the apical domains become reoriented and coupled to the motions of bound GroES. We present a measurement of the t{bar t} production cross section using events with one charged lepton and jets from p{bar p} collisions at a center-of-mass energy of 1.96TeV. In these events, heavy flavor quarks from top quark decay are identified with a secondary vertex tagging algorithm. From 162 pb{sup -1} of data collected by the Collider Detector at Fermilab, a total of 48 candidate events are selected, where 13.5 {+-} 1.8 events are expected from background contributions. Total body potassium (TBK), measured at different institutions, has no reference standard for comparison. A useful reference formula, however, based on body size and age, has been derived at the Brookhaven National Laboratory. This formula was used to help calibrate a simple whole body counter for measuring total potassium and applied to nutrition and hypertension studies. A sodium iodide detector was used for counting gamma rays emitted by potassium-40 to estimate TBK. The mean ratio (+/- S.D.) of adjusted TBK measurements to predicted values was 1.002 +/- 0.047. The ratio of lean body mass estimated by the TBK measurement to that derived from skin fold thickness was 1.069 +/- 0.056. In hypertensives on low dose thiazide, the ratio of TBK measurements to predicted values was 0.994 +/- 0.052. Thus, our TBK measurements were adjusted in close agreement with a predictor formula, provided higher estimates of lean body mass than did skin fold ...

The reaction of the retinylidene Schiff base in bacteriorhodopsin (bR) to the water-soluble reagent hydroxylamine is enhanced by greater than 2 orders of magnitude under illumination. We have used this reaction as a probe for changes in Schiff base reactivity during the photocycle of wild-type bR and mutants defective in proton transport. We report here that under illumination at pH 6, the D85N mutant has a 20-fold lower rate and the D212N mutant has a greater than 4-fold higher rate for the light-dependent reaction with hydroxylamine compared with wild-type bR. In contrast, the reactivities of wild-type bR and the D96N and T46V mutants are similar. It has been previously shown that the D96N and T46V replacements have no significant effect on the kinetics of M formation but have dramatic effects on rate of the decay of M. Escherichia coli spheroplast protein y (EcSpy) is a small periplasmic protein that is homologous with CpxP, an inhibitor of the extracytoplasmic stress response. Stress conditions such as spheroplast formation induce the expression of Spy via the Cpx or the Bae two-component systems in E. coli, though the function of Spy is unknown. Here, we report the crystal structure of EcSpy, which reveals a long kinked hairpin-like structure of four ?-helices that form an antiparallel dimer. The dimer contains a curved oval shape with a highly positively charged concave surface that may function as a ligand binding site. Sequence analysis reveals that Spy is highly conserved over the Enterobacteriaceae family. Notably, three conserved regions that contain identical residues and two LTxxQ motifs are placed at the horizontal end of the dimer structure, stabilizing the overall fold. Fish retinal ganglion cells (RGCs) can survive and regrow their axons after optic nerve injury. Injured RGCs express anti-apoptotic proteins, such as Bcl-2, after nerve injury; however, upstream effectors of this anti-apoptotic protein are not yet fully understood. Heat shock proteins (HSPs) play a crucial role in cell survival against various stress conditions. In this study, we focused on HSP70 expression in the zebrafish retina after optic nerve injury. HSP70 mRNA and protein levels increased rapidly 2.3-fold in RGCs by 1-6 h after injury and returned to control levels by 1-3 days. HSP70 transcription is regulated by heat shock factor 1 (HSF1). To study the plasma evolution and spatial structure at the L/H transition, the double hysteresis is examined by use of the 1-dimensional transport model equations. Three mechanisms for the bipolar losses, i.e., the loss cone loss, collisional bulk viscosity loss of ions and the anomalous loss are simultaneously retained. Five-fold multiple bifurcations are found to exist at the plasma edge, similar to the previous 0-dimensional study. Double hysteresis causes a self-generated oscillation, which is attributed to the compound dither, a kind of ELMs. Spatio-temporal evolution of the compound dither is analyzed. (author)

E. coli HB101 cells transformed to tetracycline resistance with the plasmids pMB9 or pBR322 display a 10/sup 5/-10/sup 6/-fold lower plating efficiency on agar containing 440 ..mu..M CdCl/sub 2/ than nontransformed cells. When DNA is inserted into the BamH1 site of the plasmid tet gene, or when DNA spanning the BamH1 site is deleted, tetracycline resistance and cadmium hypersensitivity are both lost. In contrast, insertion of DNA into the ampicillin resistance gene does not affect cadmium hypersensitivity.

The polymerization of vinyl pyrrolidinone on nylon 66, with the assistance of aqueous phenol and formic acid, was investigated by mutual and post irradiation procedures. Both solvents were found to greatly facilitate the polymerization with the post irradiation procedure, whereas neither presented any advantage with the mutual irradiation procedure. Tensile properties of the modified fibers were affected in similar ways by both the irradiation procedures and solvents in that the only notable changes were the somewhat higher extensions at yield and at break. The moisture regain values of the treated yarns and fabrics were increased up to three-fold. Surface morphology of the modified fibers was revealed by SEM.

Differential cross sections have been measured for the elastic and inelastic scattering of 104 MeV alpha-particles from /sup 90/,/sup 92/Zr. The experimental data are analyzed in terms of coupled channels on the basis of a flexible anharmonic vibrator model and using different parametrizations of the radial shape of the extended optical potential. The results favour the squared Saxon-Woods type for the real part. Additionally to a radial momentum analysis of the real potentials a semimicroscopic folding model has been invoked for extracting isoscalar quadrupole and hexadecapole transition rates.

Real-time neutron radiography (RTNR) is now proving to be a valuable research tool in the study of hydrogenous fluid flow. One of the most significant advantages of neutron radiography is the ability to image hydrogenous substances (such as lubricants, coolants, and fuels) inside metallic materials such as aluminum and titanium engines. By using RTNR, one can then study dynamic events such as the movement of liquids inside these solids. The Phoenix Memorial Laboratory (PML) at the University of Michigan has recently developed and installed a facility dedicated to RTNR. The work at PML has shown that RTNR of dynamic events can provide information enabling the researcher to follow dynamic events that were previously impossible or impractical. This paper will show the variety of uses of RTNR presently being pursued at PML.

The ATLAS High Level Trigger (HLT) is a distributed real-time software system that performs the final online selection of events produced during proton-proton collisions at the Large Hadron Collider (LHC). It is designed as a two-stage trigger and event filter running on a farm of commodity PC hardware. Currently the system consists of about 850 processing nodes and will be extended incrementally following the expected increase in luminosity of the LHC to about 2000 nodes. Elution profiles of total lipoproteins, apolipoprotein B (apoB) concentrations in lipoproteins, and plasma triglyceride (TG) levels were examined in early-, late-, and non-lactating cows. Additionally, arteriovenous (A-V) differences were also measured to elucidate the uptake of TG and apoB-containing lipoproteins in mammary gland. The distribution of i.v. administered lead (/sup 203/Pb-acetate; 50 nmol/kg b.w.) was studied by means of autoradiography and impulse counting in pregnant C57BL mice (day 18) treated orally with dithiocarbamates. Diethyldithiocarbamate (DEDTC), disulfuram or thiram (2 X 1) mmol/kg b.w.) or vehicle (gelatine) alone, was given by gavage 2 h before and immediately after the injection of lead. All three dithiocarbamates, especially thiram, changed the distribution pattern of lead. Thiram and DEDTC had the greatest effect at 4 h after lead administration, disulfiram at 24 h. In the mother, most notably the brain concentration increased (70-fold for thiram at 4 h) while that of erythrocytes and skeleton decreased (50- and 4-fold, respectively). We previously reported that the peroxisome proliferator-activated receptor gamma coactivator-1 (PGC-1) mRNA in rat epitrochlearis muscle was increased after swimming exercise training. In the present study, we demonstrated further that PGC-1 mRNA expression in the epitrochlearis muscle of 4-5-week-old male Sprague-Dawley rats was increased after a 6-h acute bout of low-intensity swimming exercise. With this increase, the expression level was approximately 8-fold of control and immersion group rats that stayed for 6-h in warm water, maintained at the identical temperature of the swimming barrel (35 degrees C) (p<0.01). Second, PGC-1 mRNA expression in the muscle was found to have increased 6-h after 30 10-s tetani contractions were induced by in vitro electrical stimulation. A unique feature of cyanobacteria genomes is the abundance of genes that code for hypothetical proteins containing tandem pentapeptide repeats approximately described by the consensus motif A(N/D)LXX. To date, the structures of two pentapeptide-repeat proteins (PRPs) have been determined, with the tandem pentapeptide-repeat sequences observed to adopt a novel type of right-handed quadrilateral ?-helix, or Rfr-fold, in both structures. One structure, Mycobacterium tuberculosis MfpA, is a 183-residue protein that contains 30 consecutive pentapeptide repeats and appears to offer antibiotic resistance by acting as a DNA mimic. The other structure, Cyanothece 51142 Rfr32, is a 167-residue protein that contains 21 consecutive pentapeptide repeats. Understanding the role of fractures during such events will, in part, depend on our ability to make geophysical measurements of perturbations in the moisture distribution in the vicinity of fractures.
